    Notes: Abstract The antifungal activity of two drimane sesquiterpene dialdehydes, polygodial (1) and warburganal (2), alone and in combination with several other substances, was examined against three fungi,Candida albicans, Saccharomyces cerevisiae andPityrosporum ovale employing a broth dilution method. Anethole significantly synergized the activity of the two sesquiterpenoids againstC. albicans andS. cerevisiae however, it had only an, additive effect againstP. ovale. By contrast, two antioxidants, ascorbic acid (vitamin C) and BHA (butylated hydroxyanisole), noticeably enhanced the activity of the sesquiterpenoids againstP. ovale, but had no, effect againstC. albicans andS. cerevisiae.

    Notes: Summary The thiolase family is a widespread group of proteins present in prokaryotes and three cellular compartments of eukaryotes. This fact makes this family interesting in order to study the evolutionary process of eukaryotes. Using the sequence of peroxisomal thiolase from Saccharomyces cerevisiae recently obtained by us and the other known thiolase sequences, a phylogenetic analysis has been carried out. It shows that all these proteins derived from a primitive enzyme, present in the common ancestor of eubacteria and eukaryotes, which evolved into different specialized thiolases confined to various cell compartments. The evolutionary tree obtained is compatible with the endosymbiotic theory for the origin of peroxisomes.

    Notes: Abstract The occurrence of introns in nuclear precursor RNAs (pre-mRNAs) is widespread in eukaryotes, and the splicing process that removes them is basically the same in yeasts as it is in higher eukaryotes. Splicing takes place in a very large, multi-component complex, the spliceosome, and biochemical studies have been complicated by the large number of splicing factors involved. This review describes how genetic approaches used to study RNA splicing inSaccharomyces cerevisiae have complemented the biochemical studies and led to rapid advances in the field.

    Notes: Summary In order to obtain markers for the Y chromosome ofSilene latifolia, we pooled equal weights of leaf tissue from 18 female siblings into one sample and repeated the process with 18 male siblings. Pooling was intended to provide a common genetic background for each sample, leaving the absence or presence of the Y chromosome as the primary difference between the two samples. DNA was extracted from each sample and subjected to polymerase chain reaction (PCR) amplification with arbitrary 10 bp primers. Four of 60 primers used gave an amplification with the male DNA not found among those from the female DNA. Each of these was subsequently shown to provide a reliable marker for the Y chromosome.

    Notes: Summary IncQ-derived conjugative shuttle vectors, which carried the yeast gene URA3 and/or the yeast autonomously replicating sequence (ARS1), were constructed. Both the ars-plus plasmid pAY205 and the ars-less plasmid pAY201 were successfully transmitted from E. coli to S. cerevisiae by the action of mob and tra. In this trans-kingdom conjugation, plasmid pAY205 could replicate and be retained in transconjugants. Plasmid pAY201 caused the formation of “micro-colonies” of abortive transconjugants due to its transient expression and rapid disappearance. Nevertheless, one per about 103 colonies caused by transmitted pAY201 plasmids were uncurable by integration into the homologous region of a yeast chromosome. Analyses by restriction enzyme mapping and Southern hybridization indicate that this integration is primarily caused by a double crossover during conjugation and not by a single reciprocal recombination.

    Notes: Summary The PAR1/SNQ3 gene of S. cerevisiae, which increases resistance to iron chelators in multi-copy transformants, is identical to the YAP1 gene, a yeast activator protein isolated as a functional homologue of the human c-jun oncogene by binding specifically to the AP-1 consensus box. The observed H2O2-sensitivity of par1 mutants has been attributed to an increased sensitivity to reduced oxygen intermediates. Accordingly, par1 mutants did not survive an elevated oxygen pressure and were very sensitive to menadione and methylviologene, two chemicals enhancing the deleterious effects of oxygen. The specific activities of enzymes involved in oxygen detoxification, such as superoxide dismutase, glucose 6-phosphate dehydrogenase and glutathione reductase, were decreased in par1 mutants and increased after PAR1 over-expression. As in the case of oxygen detoxification enzymes, the cellular levels of glutathione were similarly affected. These observations indicate that PAR1/YAP1/SNQ3 is involved in the gene regulation of certain oxygen detoxification enzymes. The finding that H2O2 promotes DNA-binding of human c-jun is consistent with a similar function for PAR1/YAP1/SNQ3 and c-jun in cellular metabolism.

    Notes: Summary The conditional respiratory-deficient Saccharomyces cerevisiae mutant pet-ts2281 was complemented by an yeast genomic DNA library. The gene thus isolated was sequenced and proved to be identical to the known MSW1 sequence encoding mitochondrial tryptophanyl-tRNA synthetase (Myers and Tzagoloff 1985). Compared to the wild-type, the ts2281 mutant allele of MSW1 contained a single T→C transition leading to a Leu→Ser replacement at position 294 of the protein sequence. In addition to this mutational alteration, our sequence data for the wild-type gene differ from the originally published MSW1 sequence at five other DNA positions which affect two locally restricted regions of the polypeptide chain. As expected, at the non-permissive temperature ts2281 cells are specifically defective in mitochondrial trp-tRNA formation and, thus, in overall mitochondrial protein synthesis. In addition, the patterns of cytochrome b mRNA maturation intermediates were distinctly different in ts2281 and wild-type yeast cells. The mutational effect of the observed amino-acid substitution in ts2281 is discussed in terms of weakened hydrogen bonding in the C-terminal half of the MSW1-encoded protein.

    Notes: Summary The hts1.1 temperature-sensitive histidinyl-tRNA synthetase mutation enables Saccharomyces cerevisiae to be starved for His-tRNAHis by upshift to the non-permissive temperature of 38°C. If yeast behaves similarly to bacterial and mammalian cells, this lack of His-tRNAHis should greatly enhance misreading at histidine codons (CAU/CAC) by Gln-tRNAGln, resulting in substitution of the neutral amino acid glutamine in place of histidine, a basic amino acid. Such misreading causes the isoelectric point (pI) of proteins to shift to lower values, and is readily detectable as “stuttering” on two-dimensional (2D) protein gels. By gel analysis of pulse-labelled proteins of hts1.1 yeast cells that were overexpressing phosphoglycerate kinase (PGK), our study sought to detect this specific translational error in PGK protein. It was not detected by this relatively sensitive technique, indicating that missense errors due to glutamine insertion at histidine codons do not occur in yeast at the readily-detectable level found in bacterial and mammalian cells.

    Notes: Summary The map position of three 1,3-β-glucanase-encoding genes in S. cerevisiae has been determined following conventional meiotic and mitotic mapping combined with recombinant DNA techniques. EXG1, EXG2 and SSG1 were localized to chromosomes XII, IV and XV, respectively, by hybridizing the cloned genes to Southern blots of chromosomes sepaated by pulsed-field gel electrophoresis, in conjunction with the rad52-1-dependent chromosome-loss mapping technique. Meiotic tetrad analyses further localized the EXG1 gene 6.1 centimorgans centromere-proximal to CDC25 on the right arm of chromosome XII. EXG2 was positioned between LYS4 and GCN2 on the right arm of chromosome IV, at distances of 6.2 centimorgans from LYS4 and 4.9 centimorgans from GCN2. Finally, the SSG1 locus mapped on the right arm of chromosome XV, about 8.2 centimorgans to the centromere-proximal side of HIS3.

    Notes: Summary Hydrostatic pressure and a dye plate method were used to investigate the direct induction of tetraploids or homozygous diploids from the industrial diploid or haploid yeast Saccharomyces cerevisiae. Above 200 MPa, hydrostatic pressure greatly inactivated the strains HF399s1 (α haploid), P-540 (a/α diploid), and P-544 (a/α diploid). At the same time, when pressure-treated cells of these strains were spread on a dye plate, some of the visible colonies were stained red/blue or dark blue (variant colonies); the rest stained violet, similar to colonies originating from diploid cells or haploid cells that were not pressure-treated. In addition, above 100 MPa, the formation of variant colonies increased with increasing pressure, and maximized (1x10-1) at 200 and 250 MPa, respectively. The size of almost all variant cells from P-544, P-540, and HF399s1 was visibly increased compared with that of untreated cells and the measured cellular DNA content of P-540 and HF399s1 was double that of untreated cells. Furthermore, based on random spore analysis and mass-matings, induced variants in the diploid strains were found to be tetraploid with an a/a/α/α genotype at the mating-type locus or, in the haploid strains, homozygous diploid with an α/α genotype. From these results we conclude that pressure treatment in combination with a dye plate is a useful method for strain improvement by direct induction of tetraploids or homozygous diploids from industrial strains whether diploid or haploids.

    Notes: Summary Compared to wild type cells, strains bearing the pleiotropic regulatory mutations cgr4 or cas1 synthesize apocatalase T at a high rate when grown on high glucose. Like heme-deficient ole3 single mutants, ole3 cgr4 and ole3 cas1 double mutants accumulate no catalase T protein in vivo. This defect introduced by the ole3 mutation is cured by the addition of ALA. By use of the inhibitor actinomycin D we confirm previous findings that ole3 mutants lack catalase T mRNA and show that (i) the ole3 cgr4 and ole3 cas1 double mutants do accumulate catalase T mRNA or mRNA precursor, and (ii) the processing or translation of this RNA or the accumulation of apocatalase T depends on the presence of home.

    Notes: Abstract Growth inhibition and cell killing caused by sulphite were reduced in seven Saccharomyces cerevisiae sulphite-resistant independent mutants, compared to their parental strains. Genetic analysis showed that in the seven mutants resistance was inherited as a single-gene dominant mutation and that all the analyzed mutations were allelic, thus identifying a major gene responsible for sulphite resistance in S. cerevisiae. Two of the mutants, MBS20-9 and MBS30, were further characterized. 35S-sulphite uptake experiments showed that the ability to accumulate sulphite was markedly reduced in the two resistant strains. No difference between resistant and sensitive strains with respect to glyceraldehyde-3-phosphate dehydrogenase sensitivity to sulphite, or to intracellular glutathione content, were revealed. In contrast, the extracellular acetaldehyde concentration was higher in the resistant mutants, both in the presence and in the absence of sulphite.

    Notes: Abstract Double-mutant cells of the budding yeast Saccharomyces cerevisiae harboring the gcs1-1 and sed1-1 mutations are conditionally defective (cold-sensitive) only for reentry into the mitotic cycle from stationary phase. If already proliferating at the permissive temperature (29°C), these reentry-mutant cells continue to proliferate when transferred to the restrictive temperature of 14°C, but under these conditions reentry-mutant cells lose mitochondrial DNA (mtDNA). In addition, upon exhaustion of the nutrient supply at 14°C, these reentry-mutant cells entered stationary phase at a decreased cell concentration and did not accumulate the reserve carbohydrates trehalose and glycogen. Both of these deficiencies were due to the loss of mtDNA, as shown by the responses of wild-type cells also lacking mtDNA. Mitochondrial status did not affect other aspects of the reentry-mutant phenotype. Although mitochondrial activity and the accumulation of carbohydrate reserves are typical features of cells in stationary phase, the reentry-mutant phenotype reveals that neither entry into nor exit from stationary phase need involve mitochondrial function.

    Notes: Summary Purified double- and single-stranded DNAs of the autonomously replicating vector M13RK9-T were irradiated with ultraviolet light (UV) in vitro and introduced into competent whole cells of Saccharomyces cerevisiae. Incoming double-stranded DNA was more sensitive to UV in excision repair-deficient rad2-1 cells than in proficient repair RAD + cells, while single-stranded DNA exhibited high sensitivity in both host cells. The results indicate that in yeast there is no effective rescue of UV-incoming single-stranded DNA by excision repair or other constitutive dark repair processes.

    Notes: Summary Eighteen nuclear mutants of the yeast Saccharomyces cerevisiae, each disturbed in the biosynthesis of the mitochondrially encoded cytochrome c oxidase subunit 1 (cox 1) and each representing a distinct complementation group, have been examined to identify the level at which COX1 expression is affected. RNA blotting revealed that most have a defect in the processing of COX1 precursor-mRNA; only a few are defective in COX1 transcription and/or pre-mRNA stability. In most RNA-processing mutants, the absence of the COX1 messenger results from a defect in the splicing of one or more COX1 introns. In turn, this defect can be ascribed to a mutation in a nuclear gene which is either directly involved in splicing or else acts indirectly by impairing COX1 translation.

The cloned fragment hybridized to the transverse-alternating-field-electrophoresis band corresponding to chromosomes VII and XV. According to the 2 μm DNA chromosome-loss procedure, the cys2 and cys4 mutations, which are linked together and co-operatively confer cysteine dependence, were assigned to chromosome VII. By further mapping involving tetrad analysis, the cys2-cys4 pair was localized between SUP77 (SUP166) and ade3 on the right arm of chromosome VII.

    Notes: Summary Serine and glycine biosynthesis in yeast proceed by two pathways; a “glycolytic” pathway, using 3-phosphoglycerate, and a “gluconeogenic” pathway, using glyoxylate. We used a mutation in the cat1 gene to abolish the glucose-repressible “gluconeogenic” pathway and re-isolated two mutants, ser1 and ser2, in the “glycolytic” pathway. The ser1 mutation corresponded to phosphoserine transaminase and ser2 to that of phosphoserine phosphatase. Mutagenesis of a ser1 ser2 cat1 triple mutant facilitated the isolation of a mutation in a new gene, SER10. SER10 appears to be part of a pathway which, under normal growth conditions, is less important in serine biosynthesis. The ser1 ser2 ser10 triple mutants were totally serine auxotrophic on glucose media but serine prototrophic during growth on non-fermentable carbon sources. This phenotype was used to select for possible regulatory mutants that synthesize serine by the gluconeogenic pathway even in the presence of glucose, e.g., with a non-glucose repressible glyoxylate cycle. In an alternative approach to isolate such mutants URA3 and TRP1 expression were placed under the control of the glucose-repressible FBP1 (fructose-1,6-bisphosphatase) promoter. Although both systems resulted in strong selection pressure we could not isolate constitutively derepressed mutants. These results indicate that transcription of glucose-repressible gluconeogenic enzymes is mainly dependent on positive regulatory elements.

    Notes: Summary The autonomously replicating plasmid YEpSS1, containing the S. cerevisiae SOD1 and SRB1 genes, was highly unstable in a wild-type strain. When transformed into a fragile srb1-1 mutant host, the same plasmid displayed different characteristics depending on the growth medium used. Both batch and continuous culture experiments demonstrated that the plasmid was very unstable when the transformed strain SLU15 was grown in the presence of an osmotic stabiliser (10% w/v sorbitol). However, in the absence of the osmoticum, nearly 100% of the cells retained the plasmid and produced the Sod1 protein after 80 generations of growth.

    Notes: Summary Wine yeast strains are characterized by a high chromosomal DNA polymorphism. This can be explained partly by a size difference of different variants of specific chromosomes. This difference can reach up to 45% of the size of the chromosome in question. Two strains, SB1 and Eg8, have a very complex chromosomal pattern and show one band hybridizing with probes from two different chromosomes derived from a reference strain. This is an indication of the presence of “hybrid” chromosomes in these wine strains. The most astonishing result concerns chromosome VIII, frequently present in wine strains in two variant forms. The first normal form has a size of about 580 kb while the second is around 1000 kb. These two forms segregate at meiosis and recombine with a normal chromosome VIII from a laboratory strain. Wine yeasts are thus very different from haploid laboratory strains.

    Notes: Summary The addition of 6-azauracil to the growth medium causes a strong reduction of the GTP level in the nucleotide pool of Saccharomyces cerevisiae. In-vitro experiments show a strong inhibition of IMP dehydrogenase activity by 6-azaUMP explaining the preceeding effect. PPR2 mutants, previously characterized by an increased sensitivity to 6-azauracil compared to the wildtype, are specifically susceptible to the lowering of the GTP pool, and are able to grow in presence of 6-azauracil when guanine is added to the medium.

    Notes: Summary We have transformed Saccharomyces cerevisiae with a genomic library contained in the replicative vector pFL44. The resulting transformants were screened for resistance to fenpropidin, a specific inhibitor of sterol 14-reductase. A plasmid was isolated that transformed yeast both to resistance to fenpropidin and to an increased specific activity of sterol 14-reductase. Sterol analysis of transformed cells grown in the presence of increasing concentrations of the inhibitor confirmed that resistance was a consequence of over-production of sterol 14-reductase. By chromosomal gene disruption, we have, for the first time, constructed yeast strains defective in sterol 14-reductase. As expected, since yeast in unable to take up sterols in aerobiosis, the disrupted strains do not grow in the presence of oxygen, even if exogenous sterols are supplied. However, disrupted cells grow in anaerobiosis with exogenous oleic acid and ergosterol supplemens. They also grow in aerobiosis if they bear an additional mutation allowing sterol uptake. In this last growth condition the cells require a “sparking” ergosterol supplementation (25nM) and accumulate ignosterol (ergosta-8, 14-dienol) as the end-product of the sterol pathway. These results reveal that ignosterol is not obviously toxic to yeast membranes and strongly suggest that the molecular basis of the antifungal-activity morpholine and piperidine is directly related to the specific inhibition of ergosterol formation.

    Notes: Summary Fructose-1,6-bisphosphatase is a key enzyme in gluconeogenesis and the FBP1 gene is not transcribed during growth with glucose. Genetic analysis indicated a positive regulation of FBP1 expression after exhaustion of glucose. By linker-deletion analysis, two upstream activation sites (UAS1 and UAS2) were localized and the respective UAS-binding factors (DAP I and DAP II for derepression activating protein) were identified by gel retardation. UAS1 and UAS2 span about 30 bp each, and are separated by approximately 30 bp. Both UAS sites act synergistically. Although UAS1 showed some similarities to the DNA-binding consensus for the general yeast activator Rap1, competition experiments and DEAE-chromatography proved that DAP I and Rap1 correspond to different proteins. Gel retardation by DAP I depended on carbon sources and did not occur in cells growing logarithmically with glucose, whereas a strong retardation signal was obtained with ethanol-grown cells. The present results suggest that DAP I and DAP II are the final regulatory elements for glucose derepression.

    Notes: Summary Two vectors, termed pG63.11 (7.6 Kb) and pHCG3 (9.6 Kb), suitable for yeast transformation have been constructed. The pHCG3 vector has cosmid properties. Both vectors contain a single 3.3 Kb EcoRI-HindIII fragment of yeast origin which carries the yeast URA3 gene (1.1 Kb) and the origin of replication of the 2 µm plasmid (2.2 Kb). They confer ampicillin resistance and they contain 5 unique EcoRI,HpaI,HindIII,BamHI and SalI restriction sites. Cosmid pHCG3 was used to clone the nitrogen fixation (nif) gene cluster of Klebsiella pneumoniae carried by twoHindIII fragments of 17 and 26 Kb, respectively. The resulting cosmid, termed pGPC875 (53 Kb) which conferred a Nif+ phenotype to Escherichia coli, was introduced in yeast by transformation. No acetylene reduction activity was detectable in the transformants. However it was shown that the entire information for nitrogen fixation can be replicated and maintained intact in yeast for more than 50 generations of growth.

    Notes: Summary We have studied the organization and expression of a group of tRNA genes located on a 2,700 base pair portion of the yeast mitochondrial genome between the genetic markers cap (chloramphenicol resistance) and oxil (cytochrome oxidase subunit II). This region is spanned by mitochondrial DNA inserts of two recombinant plasmids, pYm162 and pYm267, which have been extensively mapped and sequenced. This tRNA group is composed of six tRNA genes, coding for tRNA AAR Lys , tRNA AGR Arg , tRNA GGN Gly , tRNA GAY Asp , tRNA AGY Ser , and tRNA CGN Arg . We report the sequence for the majority of the 2,700 base pair region including the genes for all six tRNAs. All six genes are oriented in the same direction and are, therefore, transcribed from the same DNA strand. Further, a comparison of the organization of this region with the analogous region of a related wild type strain shows that the tRNA gene order in the two strains is the same. Five of the six tRNA genes have corresponding transcripts in wild type RNA. Although a potential structural gene for tRNA CGN Arg is present, we do not detect a tRNA CGN Arg gene transcript.

    Notes: Abstract Borrelidin is a specific inhibitor of the threonyl-tRNA-synthethase. A class of dominant borrelidin resistant mutants (BOR1) of Saccharomyces cerevisiae was biochemically characterized. The mutants possess an altered aspartokinase which is insensitive to threonine inhibition. The threonine and homoserine pools in these mutants are 22 times larger than in the wild type. By feeding aspartate under a variety of conditions the homoserine pool is increased even 57 times whilst the threonine pool is reduced.

    Notes: Abstract Congo red was applied to growing yeast cells and regenerating protoplasts in order to study its effects on wall biogenesis and cell morphogenesis. In the presence of the dye, the whole yeast cells grew and divided to form chains of connected cells showing aberrant wall structures on both sides of the septum. The wall-less protoplasts in solid medium with the dye exhibited an abnormal increase in volume, regeneration of aberrant cell walls and inability to carry out cytokinesis or protoplast reversion to cells. In liquid medium, the protoplasts synthesized glucan nets composed mainly of thin fibrils orientated at random, whereas normally, in the absence of dye, the nets consist of rather thick fibrils, 10 to 20 nm in width, assembled into broad ribbons. These fibrils are known to consist of triple 6/1 helical strands of (1 » 3)-β-d-glucan aggregated laterally in crystalline packing. The thin fibrils (c. 4 to 8 nm wide) can contain only a few triple helical strands (c. 1.6 nm wide) and are supposed to be prevented from further aggregation and crystallization by complexing with Congo red on their surfaces. Some loose triple 6/1 helical strands (native elementary fibrils) are also discernible. They represent the first native (1 » 3)-β-d-glucan elementary fibrils depicted by electron microscopy. The effects of Congo red on growth and the wall structure in normal cells and regenerating protoplasts in solid medium can be explained by the presence of a complex which the dye forms with (helical) chain parts of the glucan network and which results in a loss of rigidity by a blocked lateral interaction between the helices.

    Notes: Abstract The effect of the tryptophan analogue 4-methyltryptophan in Saccharomyces cerevisiae has been investigated. 4-Methyltryptophan inhibits the aminoacylation of tryptophan specific transfer ribonucleic acid (tRNATrp). The mode of inhibition is competitive and the analogue is not charged onto tRNATrp. Thus 4-methyltryptophan application depletes the cells from charged tRNATrp. As a consequence cell growth and protein synthesis are strongly reduced. 4-Methyltryptophan is degraded efficiently in culture media inoculated with the wild type strain; the effects of 4-methyltryptophan were therefore found to be transient.

    Notes: Abstract The intracellular levels of adenosine triphosphate and several glycolytic intermediates were determined in Saccharomyces cerevisiae in relation to the presence of the metabolically antagonistic enzymes phosphofructokinase and fructose-1,6-bisphosphatase. Phosphofructokinase is synthesized constitutively in cells grown in the presence of glucose and fructose-1,6-bisphosphatase derepression occurs upon the exhaustion of glucose from the growth medium. Transcriptional regulation of fructose-1,6-bisphosphatase was suggested based on experiments with wild type cells using 8-hydroxyquinoline, a known inhibitor of nuclear transcription, and with the S. cerevisiae mutant strain A364A (ts-136) blocked in the transport of nuclear RNA at non-permissive temperature. The level of phosphofructokinase was reduced more than 25-fold under conditions of high citrate accumulation in an aconitaseless, glutamate requiring mutant strain, MO-1-9B. There was a rapid decrease in the levels of adenosine triphosphate and fructose-1,6-bisphosphate at the end of log-phase of culture growth when both fructose-1,6-bisphosphatase and phosphofructokinase were present in the cells simultaneously. The changes in the levels of key glycolytic intermediates, but not the changes in adenosine triphosphate, during the simultaneous presence of these two enzymes, can be explained without involving any futile cycling.

    Notes: Abstract The yeast, Hansenula wingei has two mating types designated 5 and 21. Cells of each mating type were found to produce mating type-specific sex pheromone which induces sexual agglutinability of the opposite mating type. Crude fractions of these pheromones were prepared by using an Amberlite CG 50 (H+ type) column. The agglutinability-inducing action of the pheromones required glucose as carbon source, but no external nitrogen source. The action of the pheromones was inhibited by 5 μg/ml cycloheximide. The optimum pH for the pheromone action was 4.0. α Pheromones of Saccharomyces cerevisiae and Saccharomyces kluyveri induced sexual agglutinability of 5 mating type cells but did not that of 21 mating type cells. a Pheromones of the Saccharomyces yeasts had no effect on both 5 and 21 mating type cells. The sex pheromones of H. wingei had no effect on the sexual agglutinability of inducible a cells of S. cerevisiae. From the experimental results obtained so far, we propose to call 5 and 21 mating types in H. wingei a and α mating types, respectively.

    Notes: Abstract Scar rings (SR) from scarless cells at the early stages of budding and mature bud scars from Saccharomyces cerevisiae isolated by both chemical and enzymic treatment of cell walls were observed by selected-area electron diffraction (SAED), X-ray diffraction and electron microscopy with simultaneous physico-chemical characterization (including molar mass, intrinsic viscosity and crystallite size) of α-chitin and glucan. The SR, composed of glucan with strong 0.608; 0.397 and 0.293 nm X-ray reflections, was formed at the start of budding. The SAED patterns of α-chitin both in the adjacent circular zone and in the parts of newly formed primary septum (PS), observed when the development of the PS started, did not differ from those of α-chitin in the single mature bud scar. The bud scar consisted of α-chitin, glucan and mannan and their content, as well as the crystallite size of chitin, depended on the mode of preparation.

    Notes: Abstract In fungi, cellular resistance to heavy metal cytotoxicity is mediated either by binding of metal ions to proteins of the metallothionein type or by chelation to phytochelatin-peptides of the general formula (γ-Glu-Cys)n-Gly. Hitherto, only one fungus, Candida glabrata has been shown to contain both metal inactivating systems. Here we show by unambiguous FAB-MS analysis that both a metallothionein-free mutant of Saccharomyces cerevisiae as well as a wildtype strain synthesize phytochelatin (PC2) upon exposure to 250 μM Cd2+ ions. The presence of Zn and/or Cu ions in the nutrient broth also induces PC2 synthesis in this organism. By 109Cd exchange and subsequent monobromobimane fluorescence HPLC, it could be shown that the presence of Cd2+ in the growth medium also induces phytochelatin synthesis in Neurospora crassa, which contains metallothioneins.

    Notes: Abstract Both insect and mammalian genes have previously been cloned by genetic complementation in yeast. In the present report, we show that the method can be applied also to plants. Thus, we have cloned a rape cDNA for 3-isopropylmalate dehydrogenase (IMDH) by complementation of a yeast leu2 mutation. The cDNA encodes a 52 kDA protein which has a putative chloroplast transit peptide. The in vitro made protein is imported into chloroplasts, concomitantly with a proteolytic cleavage. We conclude that the rape cDNA encodes a chloroplast IMDH. However, Southern analysis revealed that the corresponding gene is nuclear. In a comparison of IMDH sequences from various species, we found that the rape IMDH is more similar to bacterial than to eukaryotic proteins. This suggests that the rape gene could be of chloroplast origin, but has moved to the nucleus during evolution.

    Notes: Summary Oligonucleotides hybridizing to simple repetitive DNA patterns are highly informative as probes for DNA fingerprinting in all investigated animal species, including man. Here we demonstrate the applicability of this technique in higher plants. The oligonucleotide probes (GTG)5 and (GATA)4 were used to investigate the differences in DNA fingerprint patterns of the following angiosperm species: Triticum aestivum, Secale cereale, Hordeum vulgare, Beta vulgaris, Petunia hybrida, Brassica oleracea, and Nicotiana tabacum. Two species, Hordeum vulgare as a monocot and Beta vulgaris as a dicot, were analyzed in more detail. Their genomes differ considerably in both amount and organization of the simple repetitive sequences (GATA)n, (GACA)n, (GTG)n, and (CT)n due to the evolutionary distance of these two species. Furthermore, several lines and cultivars of Beta vulgaris and Hordeum vulgare can clearly be distinguished on the basis of their highly polymorphic patterns of these repetitive sequences.

    Notes: Abstract The genetic variability of natural populations ofCapsella bursapastoris in North- and Middle-Europe has been estimated by means of enzyme assays. Zymograms of 81 populations have been developed. 17 loci could be identified, and 8 of them can be heterozygous. Genetic variability is greater between populations than within. No correlation between actual population sizes and genetic heterogeneity could be detected. Some electromorphs shift their frequencies proportionally to increasing adversity of climatic conditions, some appear to be constant over the whole area, and others are characterized by an apparently irregular variation pattern. Marginal populations comprise a significantly higher proportion of heterozygous plants than central ones. Apart from this clinal variation pattern, a mosaic pattern, strongly related to habitat conditions, was observed: genetic heterogeneity is greater in more intensively disturbed sites. The pattern of genetic variation in natural populations ofCapsella bursa-pastoris is rather highly influenced by the breeding system.

Crossing experiments betweenR. seguieri and the species of theR. alpestris group were unsuccessful. All 5 species have a chromosome number of 2n = 16, the record forR. bilobus is new. There is no statistically significant difference between the karyotypes ofR. seguieri andR. alpestris s. str. Nevertheless, according to morphological evidence and crossing experiments,R. seguieri is not closely related to theR. alpestris group.

    Notes: Abstract Allozymic variation in proteins encoded by 22 loci was analyzed electrophoretically in 278 individual plants of wild barley,Hordeum spontaneum, the progenitor of cultivated barley, in four 100 meter transects, in Israel, each equally subdivided into basalt and terra rossa soil types. Significant differentiation according to soil was found in 9 alleles. Our results suggest that allozyme polymorphisms in wild barley are at least partly adaptive, and differentiate by edaphic natural selection rather than by stochastic processes, and/or neutrality of allozymic variants.

    Notes: Abstract Genetic variation—primarily in 19 genetic loci of seven enzyme systems—was analyzed in accessions from various parts of the geographic range ofSolanum pennellii, which according to all tested biosystematic criteria behaves like a species ofLycopersicon. In comparison with the largely sympatricL. hirsutum andL. pimpinellifolium, this species exhibits the same trends of reduced allogamy and decreased genetic variation toward the north and south margins of its distribution, though to a much lesser degree; it does not exhibit their trends toward smaller flower size in the same peripheral regions. All three species agree to a considerable extent in the ranking of their tested loci in respect to degree of variablility; however, overall polymorphy is highest inS. pennellii. Except for the appearance of self-compatibility at its southernmost margin,S. pennellii is exclusively and rigidly self-incompatible. Alleles are distributed much more uniformly over the range than in the previously mentioned species, marginal and internal endemic mutants being much less abundant. A marked geographic disagreement is evident in regions of high and low variation. These differences in patterns of genetic variability are reconciled in terms of observed differences in mating systems, probable age of distributions, and adaptive strategies.

    Notes: Abstract The ultrastructure of the canal cells and the canal filling substance ofCitrus limon have been studied. At maturity the canal cells are very rich in cytoplasm. Their inner tangential walls lining the canal are much thickened and formed by two layers: the outer corresponds to the original wall, the inner is formed by subsequent deposition of abundant materials of different origin. This thickening occurs at the same time as the filling of the stylar canal. Both events are paralleled by considerable dictyosomic activity, the formation of a large amount of rough endoplasmic reticulum, and the incorporation of small cytoplasmic masses into the cell wall, due to plasmalemma evaginations. — The material in the stylar canal has a heterogeneous ultrastructure aspect and consists of polysaccharides, proteins and lipids; it presumably provides nutrients for the growing pollen tubes.

    Notes: Abstract Fraction 1 protein (F-1-protein) (ribulose bisphosphate carboxy-lase-oxygenase) contained inLemnaceae has been evolving for at least 50 million years because fossils of these plants have been identified in strata belonging to the Upper Cretaceous. Electrofocusing F-1-protein resolves the large subunit polypeptides coded by extranuclear DNA and the small subunit polypeptides coded by nuclear DNA. Four differences affecting isoelectric points of the large subunit polypeptides and eight affecting the small subunit polypeptides are now present among eleven species representing the four genera comprising theLemnaceae. In comparison, four differences in the large and 13 in the small subunit polypeptides exist among 63 species ofNicotiana; four differences in the large and eight differences in the small subunit polypeptides exist among 19 species ofGossypium. The number of differences in F-1-protein composition being of the same order of magnitude for the generaNicotiana, Gossypium, and the familyLemnaceae, we infer that these Angiosperms are of similar antiquity. Nicotiana species indigenous to Australia and Africa contain F-1-proteins whose large subunit polypeptides are different but some of whose small subunit polypeptides are like those found in species from the Western Hemisphere. The same situation is found for the F-1-protein inGossypium. These results are in harmony with the view that species ofNicotiana andGossypium have arrived in Australia via former land connections between S. America, Antarctica, and Australia.

    Notes: Abstract The ChileanMicroseris pygmaea has a ten-part paleaceous pappus while the CalifornianM. bigelovii has five pappus parts on each achene. Hybrids between the two species have between five and ten pappus parts with averages below 7.5. Hybrid B 87 has an F 1 average value of 6.7 pappus parts. 140 F 2 plants were raised from this hybrid, and 12 F 3 families were obtained by selfing from F 2 plants. One larger F 4 family has been raised. Pappus part number in all of these is still canalized between 5 and 10. Variation within these limits is genetically determined by a quantitatively acting polygenic system. Modeling of this system suggests that a minimum of four, but probably not many more, genes are involved. This opens the possibility of a complete genetic analysis of the system.

Only some species are known to have a trilacunar nodal pattern with three traces. In the unilacunar type one pair of smaller bundles separates from the complex lacunary leaf trace laterally, each soon forking into two arms: One arm becomes a marginal vein of the petiole, the other, besides supplying the stipules, forms a ± distinct vascular “bridge” within the cortex of the nodal flanks. In theRubieae this flank bridge develops as a very distinct vascular “ring” commissure out of which the whorled leaf-like appendages are vascularized; only the opposite true leaves receive their complex trace out of the lacunes directly.     Notes: Abstract Several anatomical characters in leaves were described, and their distribution determined, for 153 species ofPolygonum, mostly from herbarium specimens. Structures surveyed were epidermal (glandular and nonglandular trichomes, nodules, specialized parenchyma, stomatal apparatus) and internal (cavities, crystals, laticifer-like cells, nodules, subepidermal fibers). Cleared leaves were supplemented by resin-embedded sections and SEM preparations of selected species. No feature defines any taxonomic section, but some features occur only within one section. Laticifer-like cells, epidermal and internal nodules, resin cups, and unique epidermal and subepidermal cavities seem to be unknown elsewhere; other features (invaginated epidermal cells; enlarged crystal cells confined to paraveinal layer) are rare among angiosperms.

Phenetic analyses of mainland taxa suggest thatP. fernandeziana belongs to subg.Sphaerocarpidium whereas the endemic taxa form their own subg.Tildenidium connecting to subg.Tildenia. Cladistic analyses indicate thatP. margaritifera is the most primitive species in the archipelago and thatP. berteroana is the most derived, especially patristically. Chromosomally, the four species are all n = 22, which may be tetraploid on a base of x = 11. Sulfated flavones occur only inP. berteroana andP. skottsbergii, which are otherwise unknown for the family. Dispersal of propagules to the islands from the continent and between islands is believed to have been accomplished by birds.

    Notes: Abstract The taxa in this study have in the past been treated together in sect.Tiniaria Meissner ofPolygonum L. s.l., and more recently the large erect rhizomatous herbaceous perennials have been separated from the twining annuals and perennials as the generaReynoutria andFallopia respectively. These taxa range in ploidy level from diploid to octoploid, with base numbers of both 10 and 11 present. The plants are primarily Asiatic in distribution, although many of the large erect perennials are now naturalized in many parts of Europe. Cytological examination has revealed the presence of a number of previously unknown hybrid taxa in the British Isles. The readiness with which hybridisation occurs between taxa of differing base number and ploidy level, and similarities revealed in chromosome morphology, meiotic pairing and in DNA C-value, suggest to the authors that these two genera are best amalgamated underFallopia.

    Notes: Abstract A taxonomically difficult purple-flowered group within the genusErysimum, restricted to the Iberian Peninsula, is analyzed by multivariate and cladistic analyses. 51 specimens have been scored for 14 characters. Both principal components and discriminant analyses provide support to the recognition of the five species considered by the author, namelyE. linifolium, E. lagascae, E. baeticum, E. popovii, andE. cazorlense. Cladistic analysis, using 7 characters resulted in a single most parsimonious cladogram containing no homoplasies. The pattern of morphologic divergence follows a clear NW.-SE. trend, which is congruent with the topology of the cladogram. This trend significantly affects growth-form as well as fruit characters, both providing the main grounds for species recognition. The different behavior and significance of several characters in both kinds of analysis is discussed. The co-occurrence of morphologically similar individuals differing in the flower color is discussed, too. Possible explanations for this phenomenon involve hybridization in a wide sense or, alternatively, rejecting the assumption of monophyly for the group.

The aim was to investigate whether the present sectional delimitation of the genus was in agreement with genomic data and if there was a correlation between genome constitution and morphology.     Notes: Abstract The paleotropical tree genusMacaranga (Euphorbiaceae) comprises all stages of interaction with ants, from facultative associations to obligate myrmecophytes. In SE.-Asia food availability does not seem to be the limiting factor for the development of a close relationship since all species provide food for ants in form of extrafloral nectar and/or food bodies. Only myrmecophyticMacaranga species offer nesting space for ants (domatia) inside internodes which become hollow due to degeneration of the pith. Non-myrmecophytic species have a solid stem with a compact and wet pith and many resin ducts. The stem interior of some transitional species remains solid, but the soft pith can be excavated. The role of different ant-attracting attributes for the development of obligate ant-plant interactions is discussed. In the genusMacaranga, the provision of nesting space seems to be the most important factor for the evolution of obligate myrmecophytism.

    Notes: Abstract The ancestral angiosperm is commonly interpreted as an arborescent to shrubby magnolialean with large, multiparted, complex flowers. We examined this hypothesis using a phylogenetic analysis of new and reevaluated characters polarizabled with outgroup comparison. Our cladistic analysis of basal angiosperms placed the nonmagnolialeanChloranthaceae andPiperaceae at the bottom of the tree. We further inferred the probable ancestral states of characters not polarizable with outgroup comparison by examining their distribution among taxa at the base of our cladogram. The sum of ancestral character states suggests that the protoangiosperm was a diminutive, rhizomatous to scrambling perennial herb, with small, simple flowers.

    Notes: Abstract The variation in two sets of morphological characters of the flowers of the highly inbreedingScleranthus annuus (Caryophyllaceae) was assessed using 15–20 plants from each of 20 natural populations from the southernmost region of Sweden. The stamen fertility data set consisted of 10 characters describing the degree of fertility of the ten stamens/staminoids, while the sepal/gynoecium data set comprised 10 sepal characters, together with style and stigma length. Substantial variation was found in both the degree of development and the fertility of the stamens, the degree of variability in stamen fertility being related to stamen position within the flower. Considerable variation was found in the characters of the sepal and the gynoecium. Hierarchical analyses of variance indicated that 29% of the variation in total male reproductive effort was distributed among populations, 28% among plants within populations and 43% represented within-plant variation. The corresponding averages for the characters from the sepal/gynoecium data set are 26, 38, and 35%: a greater proportion of the total variance in female reproductive characters is accounted for by among individual variation than is the case with the male reproductive characters. Significance tests of Mahalanobis’ distances derived by canonical variate analyses indicated that all populations were significantly separated using the sepal/gynoecium data set, while only 50% of the pairwise comparisons on the basis of the stamen fertility data set were significant.     Notes: Abstract Eight genotypes ofLemna minor, originating from four continents, were grown for 15 days in eight different environmental treatments. Fronds under each treatment were then transferred into each of the eight environmental conditions for 15 days. The rate of frond production (relative growth rate) and mean frond biomass were recorded for each pre- and post-transfer treatment and root length was measured for each pre-transfer treatment. For all the traits, the levels of response varied significantly between genotypes (G) and between environmental conditions (E). G × E interaction effect was significant for all traits under pre-transfer treatments and some post-transfer treatments. Both pattern and amount of plasticity were genotypically variable but the amount of variation depended on the trait. The trait representing the best estimate of fitness, growth rate, exhibited the least amount of plasticity and on average, showed the most conservative pattern of plasticity. In contrast, the trait least related to fitness, root length, was the most plastic and showed the most divergent pattern of plasticity. Under some post-transfer treatments, growth rate and mean frond biomass were affected by origin (initial treatment) effect. Pattern and amount of plasticity were also influenced by initial treatments. Since some genotypes may be more affected than others by environmental conditions, origin effect may accentuate G × E interaction and therefore, modify the pattern and amount of plasticity. Comparison between dendrograms based on genetic and phenotypic similarities suggested that there is no relationship between genetic and phenotypic divergence. This lack of relationship may be due to the fact that plasticity is not necessarily adaptive.

    Notes: Abstract The restricted perennial diploid herb,Villarsia lasiosperma, of southwestern Western Australia has distylous flowers with dimorphism in style and stamen length, stigma morphology, and pollen size. In order to assess the presence and nature of an incompatibility system in this species, a crossing program was carried out using 17 plants grown from seeds collected in two field populations. Pollen stainabilities of these plants mostly exceeded 95%. Mean seed-set of Longs following intermorph pollinations was c. 24 seeds per pollination and for Shorts was c. 16 seeds per pollination. Approximately three-quarters of the Longs and Shorts produced no seeds after self-pollination; the remainder produced very few seeds following such pollinations. No Shorts produced seeds after intramorph cross-pollinations. However, about half the Longs exhibited various levels of weakened intramorph incompatibility, with mean seed-sets following intramorph cross-pollinations up to half the mean seed-sets following intermorph pollinations. In the behavior of Longs, the incompatibility system ofV. lasiosperma differs from the idealized distylous breeding system, but resembles that of a purported close relative,V. exaltata of eastern and southeastern Australia.

Using 14 endonucleases, a total of 124 phylogenetically informative restriction sites was detected. Dollo parsimony, Wagner parsimony, and bootstrap methods were employed to construct phylogenetic trees and evaluate confidence intervals of monophyletic groups. With a few exceptions, groups circumscribed on the basis of morphological variation are in agreement with groupings based on restriction site variation. Relationships between subgeneric groupings, however, disagree substantially with those proposed in the past. Although conflicting hypotheses for some intersectional relationships are not presently resolvable, cpDNA restriction site analyses propose the following refinements of existing classification schemes.Ipomoea ser.Setosae is divided into distantly related groups, as is sect.Pharbitis. SeriesTyrianthinae, a proposed segregate of sect.Pharbitis, is associated with sect.Calonyction and the Tricolor complex (subg.Quamoclit).Ipomoea sect.Batatas is segregated from other herbaceous groups of the heterogeneous subg.Quamoclit sensu lato and aligned as a derivative ofI. setosa, subg.Eriospermum. To test for homology of key characters weighted in traditional schemes, morphological features were studied with respect to their distribution on lineages defined by restriction site data. Characters such as setose sepals, foliose-pubescent sepals, and erect growth habit, among others, are interpreted as having multiple origins, while 3-locular ovaries, 4-locular ovaries, and long-haired seeds have evolved only once.

    Notes: Abstract Chromosomal studies ofMedicago lesinsii (n = 8) and its close relativeM. murex (n = 7) have led to the competing hypotheses that the latter is derived directly from the former, or that both originated from a common ancestor. In contrast to the relatively variableM. murex, M. lesinsii proved to be almost uniform isozymically, except that most populations of Greece differed by one allele from plants of the remainder of the range. This Greek variant ofM. lesinsii was indistinguishable from one of the isozyme variants ofM. murex. The greater level of allozyme variation inM. murex was consistent with its greater ecological amplitude and competitive ability. Also, this suggests thatM. murex is unlikely to have originated directly from the less variableM. lesinsii. The data suggest that either both species originated from a common ancestor, or that the n = 8 species evolved from the n = 7 species, a mode of chromosome evolution not previously hypothesized for the genus.

Restriction fragment length polymorphisms in the nuclear 45s ribosomal DNA (rDNA) were identified among two of five accessions of each of six cultivated Alliums. Restriction enzyme sites forBamHI,DraI,EcoRI,EcoRV,SacI, andXbaI were mapped. Different lengths of the rDNA repeat unit among the cultivated Alliums were due to sizes of the intergenic spacer. Nineteen polymorphic restriction enzyme sites were discovered and used to estimate phylogenetic relationships. Cladistic analyses based on Wagner parsimony were completed without an outgroup and resulted in two equally most parsimonious trees of 22 steps. A combined analysis of differences at RE sites in the ribosomal (19 characters) and chloroplast (15 characters) DNA generated a single most parsimonious tree of 39 steps. Single trichotomies were observed at 40 and 41 steps. Strict consensus of the three trees of 41 or fewer steps consisted of a lineage forA. tuberosum, a second forA. ampeloprasum andA. sativum, and a third forA. cepa, A. fistulosum, andA. schoenoprasum. Estimates of phylogenetic relationships based on variability at restriction enzyme sites in the rDNA and chloroplast DNA agree with the classification scheme ofTraub. Because of the predominance of autapomorphies, restriction enzyme analysis of the nuclear 45s rDNA is of limited use in estimating phylogenies amongAllium sections. However it is useful in the establishment of interspecific hybridity.

Tests were undertaken to examine whether in vitro culture of pollen grains can serve as a useful tool for understanding the self- and cross-pollination mechanisms among species. Characteristics of pollen germination were interpreted in relation to mating system and floral biology. Four annual species (Campanula kremeri, C. dichotoma, C. afra, C. lusitanica), and two perennial species (C. rotundifolia andC. persicifolia) were investigated. In the annual species pollen germinability is controlled by (1) the age of pollen and/or (2) in what position pollen is deposited around the style. Correlations were found between pollen germinability and mating system in three of the four annual species. No correlations were found either between germinability and age of pollen or position on the style in the perennial species. Pollen germinability reached its maximum in the middle of the male phase in all species, except forC. dichotoma, which had a decreasing germinability throughout anthesis. The germinability was lowest at the time of stigma receptivity for all species except forC. persicifolia, where the stigma did not develop as long as pollen remained on the style. The pollen collecting hairs and pollen removal have been found to play an important role controlling the stigma development, thus affecting self-pollination. This was especially pronounced inC. persicifolia. Further studies will be undertaken to elucidate factors influencing pollination within the genusCampanula.

The survey over two main flowering periods indicated that the sexual expression of this plant is complex and labile. Beside the dioecious male and female states, five monoecious states were detected. The dioecious states are more abundant and less labile. There is a clear shift from monoecious towards dioecious states and in general favour of males from the first (late autumn) to the second (late winter) flowering period. These results are discussed in view of the adaptive theories and evolution.

Chloroplast DNA restriction site data was obtained from 23 populations and four 10–20 year old herbarium specimens ofD. glomerata and three populations ofD. cannabina from throughout their geographic ranges in western North America and southwest-central Asia, respectively. InD. glomerata, plastome diversity is partitioned geographically. All populations from southern California have a common plastome, while most populations north of this region share a relatively divergent plastome (0.49% sequence divergence). Likewise, these plastomes are highly divergent (0.87% mean sequence divergence) from those found inD. cannabina. Biogeographic processes dating to the Pleistocene and Late Miocene may be responsible for these intra- and interspecific patterns of chloroplast DNA divergence.

The breaking point pressures of most pollen grain walls are equivalent to those reported in the literature for other types of living cell walls such as bacterial spore coats, algal cell walls, mold sporophyte cells, and dicot suspension culture cells. The strongest pollen grain walls are two or three orders of magnitude stronger, however. Pollen grain walls are proportionately very thick in comparison to other types of cell walls.     Notes: Abstract Tiarella trifoliata comprises varietieslaciniata, trifoliata, andunifoliata, and is distributed from southeastern Alaska to northern California. We analyzed restriction site variation of chloroplast DNA (cpDNA) using 23 endonucleases in 76 populations representing the entire geographic range of the species and the three recognized varieties. We also employed comparative restriction site mapping of PCR-amplified chloroplast DNA fragments using 16 restriction endonucleases. This species exhibits low cpDNA restriction site variation. No differentiation is evident among varieties of this species based on cpDNA data; some plants of each variety were characterized by each of the two major cpDNA types detected. The two major cpDNA clades, which differ by only a single restriction site mutation, are geographically structured. A northern clade comprises populations from Alaska to central Oregon; most populations analyzed from southern Oregon and California form a southern clade. Populations that possess the typical northern cpDNA type also occur disjunctly to the south at high elevations in the Siskiyou—Klamath Mountain area of southern Oregon and northern California. Conversely, the southern cpDNA type is found disjunctly to the north in the Olympic Peninsula of Washington. Both geographic areas characterized by disjunct cytoplasms are considered glacial refugia.Tiarella trifoliata joins two other species,Tolmiea menziesii andTellima grandiflora, in having well-demarcated northern and southern cpDNA lineages. All three species have similar life-history traits and geographic distributions. We suggest that glaciation may have played a major role in the formation of the cpDNA discontinuities present in these three taxa. The pronounced relationship between cpDNA variation and geographic distribution suggests the potential applicability of “intraspecific phylogeography” to plants via the analysis of intraspecific cpDNA variation. These three examples also join a rapidly growing data base which indicates that cytoplasms are often geographically structured within species and species complexes.

    Notes: Abstract In melittophilous plants the colour pattern of the flowers, as perceived by bumblebees, is a gradient of centripetally increasing spectral purity. This pattern serves as a signal for innate flower recognition in naive bumblebees permitting orientation to flowers and landing on flowers. Structures which make up the total signal pattern can include the background (e.g., green leaves), corollas, and stamens or floral guides. How various colour parameters, such as dominant wavelength, intensity, and spectral purity influence the colour signal pattern of flowers is analyzed. The process of strong absorption of ultraviolet light is shown to be a mechanism for the enhancement of spectral purity in flower guides. The importance of other mechanisms is also demonstrated. The presence of a gradient of centripetally increasing spectral purity in floral colour patterns as perceived by a bumblebee's eyes is demonstrated by a comparison of the spectral reflectance in different parts of the flower and a representation of colour loci in the colour triangle.

    Notes: Abstract The DNA sequence properties of 5S DNA (5S RNA gene plus spacer) from a wide range of families of plants is reviewed with particular reference to the possibility of using the information for phylogenetic inference. Although the data-base is extremely limited, the available evidence suggests that within a subclass or tribe phylogenetic inference can be made, provided that a knowledge about the number of chromosomal locations of the gene loci (5S Dna loci) is available. The evidence suggests little, if any, exchange occurs between the 5S DNA units at different chromosomal loci and the available data favour a mechanism involving amplification/deletion processes for creating structural changes at the5S Dna loci. Sequences originating from species in the familiesRosaceae, Poaceae, andBrassicaceae tended to group together in cladistic analyses but with low confidence limits. Surprisingly little of the spacer region showed conservation of sequence that may relate to a function in the control of transcription by RNA polymerase III.

    Notes: Summary Intracellular localization of calmodulin was examined in the budding yeast,Saccharomyces cerevisiae. Distribution of calmodulin changes in a characteristic way during the cell cycle. Calmodulin localizes to a patch at the presumptive bud site of unbudded cells. It concentrates at the bud tip in small-budded cells, and later it diffuses throughout the entire bud. At cytokinesis, calmodulin is largely at the neck between the mother and daughter cells. Double staining experiments have shown that the location of some polarized actin dots is coincident with that of calmodulin dots. Polarized localization of actin dots is observed in cells depleted of calmodulin, suggesting that calmodulin is not required for localization of the actin dots. Thecdc24 mutant that has a defect in bud assembly at the restrictive temperature fails to exhibit polarized localization of calmodulin, indicating that theCDC24 gene product is responsible for controlling the polarity of calmodulin.

    Notes: Abstract Data are presented on genetic variation at 27 enzyme loci of the Green-Winged orchid,Orchis morio, in 18 population samples from Italy. The existence in Italy of two subspecies, i.e. subspp.morio andpicta, is not supported by allozyme data. No genetic heterogeneity was found betweenmorio-like andpicta-like samples and specimens. Moreover, morphological transition between the two forms was observed in different Italian populations. The parameters of genetic variability estimated forO. morio populations are consistent with those found among monocotyledon plants, and among those outcrossing, animal-pollinated and with wind-dispersed seeds. Genetic diversity of ItalianO. morio is mostly within populations. Correspondingly, low values of interpopulational genetic distance were found. This appears to be due to high levels of gene flow, which were estimated with different methods. The lack ofO. longicornu from Italian samples, as well as of any hybrid withO. morio (F1, backcrossed or recombinant individuals) is demonstrated on the basis of genetic data. It is concluded that recurrent reports ofO. longicornu from Italy are due to confusion withO. morio or with otherOrchis species.

    Notes: Abstract InMalaxis saprophyta, anther wall development corresponds to the Monocotyledonous type. The uninucleate tapetum is of secretory type and the endothecium develops U- and V-shaped thickenings on the inner tangential and radial walls. Cytokinesis is simultaneous; tetrahedral, isobilateral and T-shaped tetrads are formed which are compactly aggregated in pollinia. At anthesis the microspore tetrads are 2-celled. The ovule is anatropous, bitegmic and both integuments are dermal in origin. A single hypodermal cell develops directly into a megaspore mother cell. Embryo sac development is predominantly monosporic and less often bisporic. Irrespective of the type of development, the mature embryo sac is 6-nucleate. Although double fertilization occurs, the primary endosperm nucleus degenerates. Embryogeny is of the Onagrad type. The mature embryo lacks differentiation into cotyledon, plumule and radicle. The reticulate seed coat is formed entirely by the outer layer of outer integument. There are three sterile and three fertile valves in the ovary. Although initially parenchymatous, the entire three sterile valves in the ovary and the upper half of the three fertile valves become sclerified after fertilization. The embryological characters support the disputed systematic position ofMalaxis within subtribeMalaxidinae ofEpidendreae.

The composition of volatile compounds in the same plants has also been studied. The occurrence in Sardinia of two differentT. marum entities is evidenced. One of them, growing in the NW. part of the island, is very similar toT. marum from Minorca.T. subspinosum from Sardinia and Majorca differ for the presence in the last one of short capitate hairs and for a higher ratio of dolichodial/teucrein. While the degree of variability of all the characters is very high, there is a good homogeneity between all the taxa, due to the presence of monoterpene cyclopentanoids not found until now in other species.

Alpha-gliadins, endosperm proteins coded by multigenic families, have been analyzed in the D genome species cluster and in their putative progenitors.     Notes: Abstract Based on optical reassociation studies of total nuclear DNAs at 55°C, 62°C, 69°C, and 75°C, it is concluded that repeat families in great millet, little millet, barn yard millet and finger millet are heterogeneous while those of fox tail millet are homogeneous. In great millet, almost one third of the sequences that behave as single copy at standard conditions are actually fossil repeats. Such “fossil” repeats are not a prominent feature of the genomes of the other four millets. The ratios of sequence complexities of repeats isolated at 75°C to those isolated at 55°C are 2.2, 3.5, 81, and 0.3 in case of little millet, finger millet, fox tail millet, and great millet respectively. On the basis of the above three observations, it is suggested that among these millets, the rate of turnover of the genome of fox tail millet is the slowest while that of great millet is the fastest. Such comparative estimates of differences in the turnover rates of genomes of related species are expected to generate useful data about the evolution of genomes.

    Notes: Abstract An electrophoretic survey of isozyme variation was conducted to test the hypothesis thatSenecio vulgaris L. (2n = 40) is of autotetraploid origin fromS. vernalis Waldst. & Kit. (2n = 20). It was established thatS. vulgaris exhibited fixed heterozygosity at three loci examined, showed disomic inheritance at all polymorphic loci, and contained a gene (αEst-1) and an allele (Aat-3b) which were not present in the single population ofS. vernalis surveyed. From this it is concluded thatS. vulgaris is not of autotetraploid origin. Instead, the genetic evidence is in keeping with an allopolyploid origin ofS. vulgaris with the possibility thatS. vernalis acted as one of its two parents.

    Notes: Abstract Patterns of zinc tolerance were examined in eightDeschampsia cespitosa (L.)Beauv. populations from normal and zinc-contaminated soils, using solution culture methods. Zinc-tolerant populations have evolved beneath pylons, and their tolerance patterns (degree, variance, heritability) vary. Tolerance is genetically based in all of them. In contrast to the cases of other species previously reported, some within-population differences are considerable, and may be due to both heterogeneity of the soils zinc contents, various gene combinations and gene recombinations between genotypes. One control population contains many tolerant plants, an original result which is discussed.

    Notes: Abstract Floral nectaries in theLimnanthaceae are established as exoscopic basal bulges of the episepalous stamens. Their nectariferous tissues include the epidermis and hypodermal parenchyma and inLimnanthes are vascularized by phloematic branches of the staminal bundles. Secretion occurs mainly through anomocytic stomata but, in addition, probably through the outer cuticularized thin walls of the epidermal cells. The flower structure is comparatively simple. The nectar is often slightly concealed. A wide range of pollinators can be expected, but bees are observed to be the dominant ones. The systematic position of the family is still obscure. Taxonomic placement near to any other geranialian families or to theCaryophyllaceae is only weakly justified.

    Notes: Abstract The genetic relationships among 10 inbred lines representing 10 populations of the autogamous annualMicroseris elegans from throughout California has been determined using random amplified polymorphic DNAs (RAPDs). Seventeen arbitrary 10 base pair primers produced 134 amplification products; 81 of these were shared by two or more strains. The 3 genotypes from Northern California are closely related as are 3 genotypes from Middle Californian populations which are not nearest neighbors. DNA fingerprinting with the oligonucleotide (GATA)4 gave compatible results, but the comparison was limited to samples run on one gel. Isoenzyme patterns are compatible with the DNA results, but limited by the very low number of informative polymorphisms. The clustered relationship among genotypes within a species and their geographic distribution suggests very restricted genetic recombination and an origin of new populations from randomly dispersed achenes within the range of the species.
