    Beschreibung: Sarcomere shortening in striated muscle appears to follow a regionally synchronized staircase-like time course not anticipated in some cross-bridge models. The visualization method used has been criticized as subject to Bragg diffraction effects.     Beschreibung: A new technique has been developed for identifying, in humans, dynamic spatiotemporal electrical patterns of the brain during purposive behaviors. In this method, single-trial time-series correlations between brain macropotentials recorded from different scalp sites are analyzed by distribution-independent mathematical pattern recognition. Dynamic patterns of correlation clearly distinguished two brief visuomotor tasks differing only in type of mental judgement required (spatial or numeric). These complex patterns shifted in the anterior-posterior and left-right axes between successive 175-millisecond intervals, indicating that many areas in both cerebral hemispheres were involved even in these simple judgements.     Beschreibung: Squirrel monkeys pressed a level at high rates under a second-order schedule of reinforcement in which level pressing produced a brief visual stimulus that was occasionally contiguous with an intravenous injection of nicotine. The rate of lever pressing could be markedly reduced either by substituting saline for nicotine injections or by blocking the effects of nicotine with mecamylamine. The rate of level pressing could be reduced by eliminating the brief visual stimulus.     Beschreibung: The effects of long- and short-term administration of the tricyclic antidepressant desipramine on intracranial self-stimulation in rats were studied with electrodes in the A10 region of the dopamine-containing cell bodies of the ventromedial tegmentum. Long-term desipramine administration resulted in a significant shift to the left in the ascending portion of the rate--current intensity function, indicating that the activity of the mesolimbic dopamine system was enhanced.     Beschreibung: Subjects with type III hyperlipoproteinemia develop premature atherosclerosis and have hyperlipidemia due to an increase in cholesterol-rich very low density lipoproteins (VLDL) of abnormal electrophoretic mobility. Apolipoprotein E is a major protein constituent of VLDL and appears to be important for the hepatic uptake of triglyceride-rich lipoproteins.     Beschreibung: Human malignant cancer tumors grafted into nude mice produce tumors containing both human cancer cells and the host's stromal cells. After short-term propagation of these tumors in vitro, the murine mesenchymal cells appear transformed and are tumorigenic in nude mice. However, established human cancer cell lines fail to similarly after adjacent murine stromal cells when used to produce tumors in nude mice.     Beschreibung: An electron microscopic and electrophysiological investigation was made of Merkel cell-neurite complexes in the sinus hair follicles of the cat. These mechanoreceptors respond with very precise phase locking to heavy-frequency vibratory stimuli as well as to static hair displacements. The mechanoelectric transduction process is faster than that known for any other somatic mechanoreceptor.     Beschreibung: Plasma membranes isolated from the protozoan parasite Leishmania donovani were enriched in acid phosphatase (E.C. 3.1.3.2) activity. Cytochemically, the enzyme was distributed uniformly on the surface of intact cells and was localized on the external face of isolated membranes.     Beschreibung: A single injection of the melanotropin analog [4-norleucine, 7-D-phenylalanine]-alpha-melanotropin into frogs (Rana pipiens) caused near maximum darkening of the skins of the frogs for at least 6 weeks. Injections of the natural hormone alpha-melanotropin or of the analog [Nle4]-alpha-melanotropin also caused darkening, but this effect lasted only a few days. Morphological examination of the skins of frogs injected with [Nle4, D-Phe7]-alpha-melanotropin revealed that both dermal and epidermal melanophores were dispersed during the entire 6-week period. In vitro [Nle4, D-Phe7]-alpha-melanotropin also causes prolonged darkening of the skin of the lizard Anolis carolinensis. In the absence of the melanotropin, skins previously darkened with the analog could be lightened by removal of calcium from the incubation medium but could then be redarkened by adding calcium. The cycle could be repeated indefinitely without addition of melanotropin.     Beschreibung: Natural killer cells are a recently discovered subpopulation of lymphoid cells that are present in most normal individuals of a range of mammalian and avian species. Natural killer cells have spontaneous cytolytic activity against a variety of tumor cells and some normal cells, and their reactivity can be rapidly augmented by interferon. They have characteristics distinct from other types of lymphoid cells and are closely associated with large granular lymphocytes, which comprise about 5 percent of blood or splenic leukocytes.     Beschreibung: The perception of emotion in verbal and facial expression, and the spontaneous production of conversational speech were studied in patients with unilateral focal excisions of frontal, temporal, or parieto-occipital cortex. Lesions of the left hemisphere impaired the matching of verbal descriptions to appropriate verbal categories of emotional states, whereas with lesions of the right hemisphere, the matching of different faces displaying similar emotional states was impaired. The effects of lesions of both left and right hemisphere occurred regardless of the locus of the lesion. On the other hand, frontal-lobe lesions had differential effects upon unsolicited talking; lesions of the left frontal lobe virtually abolished this behavior, whereas lesions of the right frontal lobe produced excessive talking.     Beschreibung: Human keratinocytes, derived from the skin of newborns and of adults, were grown in the complete absence of serum, in a hormone-supplemented medium on fibronectin-coated cell culture dishes at low seed density. The cell culture medium consisted of Medium 199 containing epidermal growth factor, triiodothyronine, hydrocortisone, Cohn fraction IV, insulin, transferrin, bovine brain extract, and trace elements. Removal of the brain extract from the hormone supplement had a greater negative impact on proliferation of the keratinocyte cultures than did the removal of epidermal growth factor, hydrocortisone, and triiodothyronine or Cohn fraction IV.     Beschreibung: Ten patients with multiple sclerosis who were treated with human fibroblast interferon (IFN-B) for 6 months showed a significant reduction in their exacerbation rates compared with their rates before treatment (P 〈 .01). The IFN-B was administered intrathecally by serial lumbar punctures. There was no significant change in the exacerbation rates of ten multiple sclerosis control patients before and during the period of observation. The IFN-B recipients have now been on the study a mean of 1.5 years, the controls, 1.2 years. The clinical condition of five of the IFN-B recipients and one of the control patients has improved, whereas the condition of five of the controls and one of the IFN-B recipients has deteriorated (P 〈 .036).     Beschreibung: Data from the New York Cancer Registry show no evidence for higher cancer rates associated with residence near the Love Canal toxic waste burial site in comparison with the entire state outside of New York City. Rates of liver cancer, lymphoma, and leukemia, which were selected for special attention, were not consistently elevated. Among the other cancers studied, a higher rate was noted only for respiratory cancer, but it was not consistent across age groups and appeared to be related to a high rate for the entire city of Niagara Falls.     Beschreibung: A single application of electroconvulsive shock produced a rapid but short-lasting increase in tyrosine hydroxylase activity above control values in the rat adrenal medulla and striatum. After repeated electroconvulsive shock treatment (once per day for 7 days), tyrosine hydroxylase activity increased significantly in the locus ceruleus, nucleus of the tractus solitarius, hippocampus, cerebellum, and frontal cortex and remained elevated for 4 to 8 days. Adrenal tyrosine hydroxylase activity increased 1 day after the termination of repeated electroconvulsive shock treatments and remained elevated for at least 24 days, possibly reflecting the establishment of a new and higher steady-state level of catecholamine biosynthesis in the adrenal.     Beschreibung: A substance related to luteinizing hormone-releasing hormone was demonstrated, by immunohistochemical procedures, in the cytoplasm of interstitial cells within the rat testes. In many seminiferous tubules, nuclei of spermatogonial cells were also immunopositive. Both cytoplasmic and nuclear fractions of testicular homogenates contain immunoreactive compounds, and this report identifies which cell types contain this substance.     Beschreibung: Raji cells, a human B lymphoblastoid cell line of Burkitt lymphoma origin, formed lupus inclusions when grown in a medium conditioned by the growth of Raji cells whose DNA thymidine residues had been unifilarly (single-strandedly) substituted with bromodeoxyuridine. Ultracentrifugation of this medium in excess of that required to remove Epstein-Barr virus and all other known mammalian viruses did not prevent the formation of the inclusions, and treatment of the conditioned medium with pronase destroyed the activity. These results demonstrate the presence of a protein that is secreted from bromodeoxyuridine-substituted Raji cells and is capable of inducing nonbromodeoxyuridine-substituted cells to form lupus inclusions. Interferon (100 units per milliliter) was found in the conditioned medium.     Beschreibung: During the rod-isolated phase of dark adaption, b-wave implicit time of the human cone electroretinogram increased exponentially with a time constant corresponding to that for the regeneration of rhodopsin. In the presence of different photopically equated short-wave backgrounds, cone b-wave implicit time varied inversely with the scotopic brightness of the background.     Beschreibung: The insertion of oxygenated sterol compounds into human red blood cell membranes as well as the consequent transformation of the red cells to an echinocyte shape and the expansion of the membranes are impeded by the presence of serum lipoproteins in the incubation medium. All density classes of human serum lipoproteins bind oxygenated sterol compounds, and lipoproteins can act as acceptors of oxygenated sterols previously inserted into red cells.     Beschreibung: The proteins of the three major rate components of axonal transport in guinea pig retinal ganglion cells were analyzed by one- and two-dimensional gel electrophoresis. Each rate component consisted of a different set of proteins that remained associated with each other during transport. This suggests that each rate component represents a distinct macromolecular complex and that these complexes may be definable organelles such as microtubules, microfilaments, and smooth endoplasmic reticulum.     Beschreibung: Erythrocytes infected with the late stages of the human malarial parasite Plasmodium falciparum became attached to a subpopulation of cultured human endothelial cells by knoblike protrusions on the surface of the infected erythrocytes. Infected erythrocytes did not bind to cultured fibroblasts; uninfected erythrocytes did not bind to either endothelial cells or fibroblasts. But they still have higher unemployment rates and lower salaries than men in all fields of science and engineering, at all degree levels, and at all levels of experience; and the disparities between men and women widen with higher degree levels and with years of experience. Reversals of preference are demonstrated in choices regarding monetary outcomes, both hypothetical and real, and in questions pertaining to the loss of human lives. The effects of frames on preferences are compared to the effects of perspectives on perceptual appearance.     Beschreibung: Aversive classical conditioning of Aplysia californica, a gastropod mollusk suited for neurobiological study, produces a learned reaction to the chemosensory conditioned stimulus that is expressed as a marked facilitation of four defensive responses: two graded reflexes (head and siphon withdrawal), an all-or-none fixed act (inking), and a complex fixed action pattern (escape locomotion). In addition, the conditioned stimulus produces a concomitant depression of at least one appetitive response, feeding.     Beschreibung: Arginine vasopressin and a number of its synthetic analogs augment memory functions in experimental animals. One of these analogs, 1-desamino-8-D-arginine vasopressin (DDAVP), influences human learning and memory. Cognitively unimpaired, as well as cognitively impaired adults, treated with DDAVP for a period of several days, learn information more effectively, as measured by the completeness, organization, and consistency (reliability) of recall.     Beschreibung: An intrinsic birefringence signal with two components occurring before sarcomere shortening was measured in mammalian cardiac muscle. The second component was sensitive to the inotropic state of the muscle as affected by external calcium concentration and epinephrine but not by changes of resting length. More of these bacteria reverted to histidine independence than controls not incubated with cells.     Beschreibung: The cumulative effects of a repetitive stress induced by anticipation of pain (noxious foot shock) were studied on the threshold of a nociceptive flexion reflex of the lower limb. The threshold of the nociceptive reflex progressively increased with the repetition of the stress. This effect was reversed by naloxone, which even produced hyperalgesia, since a rapid and significant decrease in this threshold, below the initial values, was noted.     Beschreibung: Southern blot hybridization was used to identify human and other vertebrate DNA sequences that were homologous to cloned DNA fragments containing the oncogenic nucleic acid sequences of three different type C mammalian retroviruses (simian sarcoma virus, the Snyder-Theilen strain of feline sarcoma virus, and the Harvey strain of murine sarcoma virus). Each onc gene counterpart has a single genetic locus, which probably contains non-onc intervening sequences. The human DNA sequences may represent genes important to cell growth or cell differentiation, or both.     Beschreibung: The amount of red chromatic valence of the red-green opponent colors channel of the human visual system has been reported to be greatly reduced in short wavelengths when the hue-matching method is used to measure red valence instead of the more typical cancellation method. Receptive fields with a silent surround were postulated to explain the reduction, and it was emphasized that the reduced valence curve represented the true chromatic valence curve of the visual system. In the present studies the previous results are interpreted to be a direct consequence of the method and the particular matching stimuli used.     Beschreibung: Golden Syrian hamsters were placed individually in cages with three drinking bottles--one empty, one containing water, and the third containing water and ethanol. Control hamsters received water only. After 1 year the experimental hamsters showed a significantly lower concentration of leucine-enkephalin-like immunoreactive substance in the basal ganglia than the control hamsters.     Beschreibung: Mice will replace the tip of a foretoe when it is amputated distal to the last interphalangeal joint. Amputation of the digit more proximal to the joint does not result in regrowth of the foretoe. Though this growth shares certain similarities with the epimorphic regeneration of amphibian limbs, the two processes are not the same.     Beschreibung: Brief tetanic stimulation of the preganglionic nerves to the superior cervical ganglion enhances the postganglionic response to single preganglionic stimuli for 1 to 3 hours. This long-term potentiation of transmission through the ganglion is apparently not attributable to a persistent muscarinic action of the preganglionic neurotransmitter, acetylcholine, since neither the magnitude nor the time course of the phenomenon is reduced by atropine. The decay of long-term potentiation can be described by a first-order kinetic process with a mean time constant of 80 minutes. We conclude that long-term potentiation, once considered a unique property of the hippocampus, is in fact a more general feature of synaptic function.
