    Description: Equilibration of 1,9- and 7,8-C(70)H(2) has allowed the relative free energy of these isomers to be measured. These "simplest hydrocarbon derivatives of C(70)" are formed by hydroboration of C(70) at room temperature. Analysis of the platinum-catalyzed equilibration of these isomers yielded a relative free energy at 295 kelvin of 1.4 +/- 0.2 kilocalories per mole, with the 1,9 isomer being more stable.     Description: Calcium-calmodulin-dependent protein kinase II (CaMKII) is a necessary component of the cellular machinery underlying learning and memory. Here, a constitutively active form of this enzyme, CaMKII(1-290), was introduced into neurons of hippocampal slices with a recombinant vaccinia virus to test the hypothesis that increased postsynaptic activity of this enzyme is sufficient to produce long-term synaptic potentiation (LTP), a prominent cellular model of learning and memory. Postsynaptic expression of CaMKII(1-290) increased CaMKII activity, enhanced synaptic transmission, and prevented more potentiation by an LTP-inducing protocol.     Description: For survival, embryonic motoneurons in vertebrates depend on as yet undefined neurotrophic factors present in the limb bud. Members of the neurotrophin family are currently the best candidates for such neurotrophic factors, but inactivation of their receptor genes leads to only partial loss of motoneurons, which suggests that other factors are involved. Glial cell line-derived neurotrophic factor (GDNF), originally identified as a trophic factor specific for dopaminergic neurons, was found to be 75-fold more potent than the neurotrophins in supporting the survival of purified embryonic rat motoneurons in culture. GDNF messenger RNA was found in the immediate vicinity of motoneurons during the period of cell death in development. In vivo, GDNF rescues and prevents the atrophy of facial motoneurons that have been deprived of target-derived survival factors by axotomy. GDNF may therefore be a physiological trophic factor for spinal motoneurons.     Description: Results from a global coupled ocean-atmosphere climate model and a model with specified tropical convective heating anomalies show that the South Asian monsoon was an active part of the tropical biennial oscillation (TBO). Convective heating anomalies over Africa and the western Pacific Ocean associated with the TBO altered the simulated pattern of atmospheric circulation for the Northern Hemisphere winter mid-latitude over Asia. This alteration in the mid-latitude circulation maintained temperature anomalies over South Asia through winter and helped set up the land-sea temperature contrast for subsequent monsoon development. Averaged across inventoried forests, turnover, as measured by tree mortality and recruitment, has increased since the 1950s, with an apparent pantropical acceleration since 1980.     Description: The association of lipid molecules into spherical vesicles in solution as a result of non-specific intermolecular forces constitutes a primary self-assembly process. Such vesicles can undergo a secondary self-assembly into higher order structures in a controlled and reversible manner by means of site-specific ligand-receptor (biotin-streptavidin) coupling. Cryoelectron microscopy shows these structures to be composed of tethered, rather than adhering, vesicles in their original, unstressed state. In contrast, vesicles aggregated by nonspecific, such as van der Waals, forces are deformed and stressed, producing unstable structures.     Description: An equilibrium phase belonging to the family of bilayer liposomes in ternary mixtures of dimyristoylphosphatidylcholine (DMPC), water, and geraniol (a biological alcohol derived from oil-soluble vitamins that acts as a cosurfactant) has been identified. Electron and optical microscopy reveal the phase, labeled Ltv, to be composed of highly entangled tubular vesicles. In situ x-ray diffraction confirms that the tubule walls are multilamellar with the lipids in the chain-melted state. Macroscopic observations show that the Ltv phase coexists with the well-known L4 phase of spherical vesicles and a bulk L alpha phase.     Description: The primate visual system is often divided into two channels, designated M and P, whose signals are relayed to the cerebral cortex by neurons in the magnocellular and parvicellular layers of the dorsal lateral geniculate nucleus. We have identified a third population of geniculocortical neurons in the dorsal lateral geniculate nucleus of macaques, which is immunoreactive for the alpha subunit of type II calmodulin-dependent protein kinase. This large third population occupies interlaminar regions (intercalated layers) ventral to each principal layer.     Description: A depth-age scale and an accumulation history for the Holocene have been established on the Greenland Ice Sheet Project 2 (GISP2) deep core, providing the most continuously dated record of annual layer accumulation currently available. The depth-age scale was obtained with the use of various independent techniques to count annual layers in the core. An annual record of surface accumulation during the Holocene was obtained by correcting the observed layer thicknesses for flow-thinning. Fluctuations in accumulation provide a continuous and detailed record of climate variability over central Greenland during the Holocene.     Description: In Drosophila and human cells, the TATA binding protein (TBP) of the transcription factor IID (TFIID) complex is tightly associated with multiple subunits termed TBP-associated factors (TAFs) that are essential for mediating regulation of RNA polymerase II transcription. The Drosophila TAFII150 has now been molecularly cloned and biochemically characterized. The deduced primary amino acid sequence of dTAFII150 reveals a striking similarity to the essential yeast gene, TSM-1. Furthermore, like dTAFII150, the TSM-1 protein is found associated with the TBP in vivo, thus identifying the first yeast homolog of a TAF associated with TFIID. Both the product of TSM-1 and dTAFII150 bind directly to TBP and dTAFII250, demonstrating a functional similarity between human and yeast TAFs. Surprisingly, DNA binding studies indicate that purified recombinant dTAFII150 binds specifically to DNA sequences overlapping the start site of transcription.     Description: Endogenous DNA adducts may contribute to the etiology of human genetic disease and cancer. One potential source of endogenous DNA adducts is lipid peroxidation, which generates mutagenic carbonyl compounds such as malondialdehyde. A sensitive mass spectrometric method permitted detection and quantitation of the major malondialdehyde-DNA adduct, a pyrimidopurinone derived from deoxyguanosine.     Description: As part of an ongoing research program to synthesize novel pillared layered materials, nickel and cobalt hydroxyacetates were inserted between the layers of amine intercalates of alpha-zirconium phosphate. The structure of the resultant nickel composite, derived from x-ray powder data, was found to consist of a three-tiered layer of nickel atoms bridged by hydroxo and acetato groups. Heating to 420 degrees C converted the hydroxyacetate layers to oxide and imparted ordered magnetic domains to the composite.     Description: A drop falling from a faucet is a common example of a mass fissioning into two or more pieces. The shape of the liquid in this situation has been investigated by both experiment and computer simulation. As the viscosity of the liquid is varied, the shape of the drop changes dramatically. Near the point of breakup, viscous drops develop long necks that then spawn a series of smaller necks with ever thinner diameters. Simulations indicate that this repeated formation of necks can proceed ad infinitum whenever a small but finite amount of noise is present in the experiment.     Description: Glycosyl-phosphatidylinositol (GPI)-anchored proteins have been reported to reside in clusters collected over small membrane invaginations called caveolae. The detection of different GPI-anchored proteins with fluorescently labeled monoclonal antibodies showed that these proteins are not constitutively concentrated in caveolae; they enter these structures independently after cross-linking with polyclonal secondary antibodies. Analysis of the cell surface distribution of the GPI-anchored folate receptor by electron microscopy confirms these observations.     Description: The El Nino-Southern Oscillation (ENSO) cycle is modeled as a low-order chaotic process driven by the seasonal cycle. A simple model suggests that the equatorial Pacific ocean-atmosphere oscillator can go into nonlinear resonance with the seasonal cycle and that with strong enough coupling between the ocean and the atmosphere, the system may become chaotic as a result of irregular jumping of the ocean-atmosphere system among different nonlinear resonances.     Description: Light absorption by rhodopsin generates metarhodopsin, which activates heterotrimeric guanine nucleotide-binding proteins (G proteins) in photoreceptor cells of vertebrates and invertebrates. In contrast to vertebrate metarhodopsins, most invertebrate metarhodopsins are thermally stable and regenerate rhodopsin by absorption of a second photon. In experiments with Rh1 Drosophila rhodopsin, the thermal stability of metarhodopsin was found not to be an intrinsic property of the visual pigment but a consequence of its interaction with arrestin (49 kilodaltons). The stabilization of metarhodopsin resulted in a large decrease in the efficiency of G protein activation. Light absorption by thermally stable metarhodopsin initially regenerated an inactive rhodopsin-like intermediate, which was subsequently converted in the dark to active rhodopsin.     Description: Representational difference analysis was used to isolate unique sequences present in more than 90 percent of Kaposi's sarcoma (KS) tissues obtained from patients with acquired immunodeficiency syndrome (AIDS). These sequences were not present in tissue DNA from non-AIDS patients, but were present in 15 percent of non-KS tissue DNA samples from AIDS patients. The sequences are homologous to, but distinct from, capsid and tegument protein genes of the Gammaherpesvirinae, herpesvirus saimiri and Epstein-Barr virus.     Description: Simple discrete-time ecological models for a species with alternating reproduction and dispersal are shown to have complex transient dynamics. If the density dependence (nonlinearity) is strong enough, then the time required to reach the final dynamics is usually very long, approaching thousands of generations, and there are typically very sudden changes in the form of the dynamics. Apparent chaos can change to cycles or vice versa.     Description: The Clementine mission has provided the first comprehensive set of high-resolution images of the south pole region of the moon. Within 5 degrees of latitude of the pole, an area of an estimated 30,000 square kilometers remained in shadow during a full lunar rotation and is a promising target for future exploration for ice deposits. The Schrodinger Basin (320 kilometers in diameter), centered at 75 degrees S, is one of the two youngest, least modified, great multiring impact basins on the moon.     Description: Signals mediated by the T cell receptor (TCR) are required for thymocyte maturation and selection. To examine the role of TCR zeta chain signals in development, TCR expression was restored in zeta-deficient mice with transgenic zeta chains that partially or completely lacked sequences required for signal transduction.     Description: Large, abrupt shifts in the (l8)O/(16)O ratio found in Greenland ice must reflect real features of the climate system variability. These isotopic shifts can be viewed as a result of air temperature fluctuations, but determination of the cause of the changes-the most crucial issue for future climate concerns-requires a detailed understanding of the controls on isotopes in precipitation. Altimetry profiles show the Aristarchus plateau dipping about 1 degrees to the north-northwest and rising about 2 kilometers above the surrounding lavas of Oceanus Procellarum to the south. Dark, reddish pyroclastic glass covers the plateau to average depths of 10 to 30 meters, as determined from the estimated excavation depths of 100- to 1000-meter-diameter craters that have exposed materials below the pyroclastics. These craters and the walls of sinuous rilles also show that mare basalts underlie the pyroclastics across much of the plateau. Near-infrared images of Aristarchus crater reveal olivine-rich materials and two kilometer-sized outcrops of anorthosite in the central peaks.     Description: The microinjection of messenger RNA encoding the eukaryotic translation initiation factor 4E (eIF-4E) into early embryos of Xenopus laevis leads to the induction of mesoderm in ectodermal explants. This induction occurs without a stimulation of overall protein synthesis and is blocked by the co-expression of a dominant negative mutant of the proto-oncogene ras or a truncated activin type II receptor. Although other translation factors have been studied in vertebrate and invertebrate embryos, none have been shown to play a direct role in development.     Description: The formation of specific connections in the developing central nervous system is thought to result from mechanisms that increase the strengths of synapses at which pre- and postsynaptic activity are correlated and decrease it otherwise. In the visual cortex, initially widespread inputs normally sort out into eye-specific patches during early life. If only one eye can see during this period, its patches are much larger than normal, and patches from the occluded eye become much smaller.     Description: Long-tailed manakins mate in leks and cooperate in multiyear male-male partnerships. An alpha male is responsible for virtually all mating, whereas a beta male assists in the courtship displays. Such altruism by the beta male poses a problem for evolutionary theory because most theoretical treatments and empirical examples of cooperative behavior involve kin selection or reciprocity. Here it is shown that alpha and beta partners are not relatives and that reciprocity is not involved. Instead, direct, though long-delayed benefits to beta males are demonstrated, which include rare copulations, ascension to alpha status, and female lek fidelity.     Description: The three-dimensional structure of a catalytic antibody (1F7) with chorismate mutase activity has been determined to 3.0 A resolution as a complex with a transition state analog. The structural data suggest that the antibody stabilizes the same conformationally restricted pericyclic transition state as occurs in the uncatalyzed reaction. Overall shape and charge complementarity between the combining site and the transition state analog dictate preferential binding of the correct substrate enantiomer in a conformation appropriate for reaction. Comparison with the structure of a chorismate mutase enzyme indicates an overall similarity between the catalytic mechanism employed by the two proteins. Differences in the number of specific interactions available for restricting the rotational degrees of freedom in the transition state, and the lack of multiple electrostatic interactions that might stabilize charge separation in this highly polarized metastable species, are likely to account for the observed 10(4) times lower activity of the antibody relative to that of the natural enzymes that catalyze this reaction.     Description: Measurements of stomatal density and delta(13)C of limber pine (Pinus flexilis) needles (leaves) preserved in pack rat middens from the Great Basin reveal shifts in plant physiology and leaf morphology during the last 30,000 years. Sites were selected so as to offset glacial to Holocene climatic differences and thus to isolate the effects of changing atmospheric CO(2) levels. Stomatal density decreased approximately 17 percent and delta(13)C decreased approximately 1.5 per mil during deglaciation from 15,000 to 12,000 years ago, concomitant with a 30 percent increase in atmospheric CO(2). Water-use efficiency increased approximately 15 percent during deglaciation, if temperature and humidity were held constant and the proxy values for CO(2) and delta(13)C of past atmospheres are accurate.     Description: Gating currents provide a direct record of the spatial rearrangement of charges occurring within the protein of voltage-sensitive ion channels. If the elementary charges move as very brief discrete pulses of current, they will produce fluctuations in the macroscopic gating current. The variance of such fluctuations in gating currents was measured in Shaker potassium channels expressed in Xenopus oocytes with a sufficiently high recording bandwidth to estimate the magnitude and time distribution of the elementary transition charge movements. Channel activation occurred in two sequential stages.     Description: The techniques that underpin modern molecular biology have been rapidly adopted by those interested in the major parasitic infections of humans. The parasitological literature is full of reports of genes and their amino acid sequences, of molecules, of cell membrane receptors and channels, and of the fine details of the immunological responses mounted by the host to combat infection. Much less enthusiasm has been shown for the mathematical techniques that facilitate the analysis and interpretation of dynamical processes such as transmission, evolution, and the interplay between parasite population growth and immunological responses within the host.     Description: The Chinese Early Cambrian Chengjiang fauna includes three different anomalocaridids, a globally spread, extinct marine group including the largest known Cambrian animals. Anomalocaridids were active predators, and their presence implies that a complex ecosystem appeared abruptly in the earliest Phanerozoic. Complete specimens display several sets of characters shared only with some other exclusively Cambrian forms. This evidence indicates that anomalocaridids, Opabinia, and Kerygmachela form a monophyletic clade.     Description: Single molecules of DNA, visualized in video fluorescence microscopy, were stretched to full extension in a flow, and their relaxation was measured when the flow stopped. The molecules, attached by one end to a 1-micrometer bead, were manipulated in an aqueous solution with optical tweezers. Inverse Laplace transformations of the relaxation data yielded spectra of decaying exponentials with distinct peaks, and the longest time component (tau) increased with length (L) as tau approximately L 1.68 +/- 0.10. A rescaling analysis showed that most of the relaxation curves had a universal shape and their characteristic times (lambda t) increased as lambda t approximately L 1.65 +/- 0.13.     Description: The transcription factor nuclear factor-kappa B (NF-kappa B) is critical for the inducible expression of multiple cellular and viral genes involved in inflammation and infection including interleukin-1 (IL-1), IL-6, and adhesion molecules. The anti-inflammatory drugs sodium salicylate and aspirin inhibited the activation of NF-kappa B, which further explains the mechanism of action of these drugs. This inhibition prevented the degradation of the NF-kappa B inhibitor, I kappa B, and therefore NF-kappa B was retained in the cytosol.     Description: Here it is shown, with the use of protein-protein photocrosslinking, that the carboxyl-terminal region of the alpha subunit of RNA polymerase (RNAP) is in direct physical proximity to the activating region of the catabolite gene activator protein (CAP) in the ternary complex of the lac promoter, RNAP, and CAP.
