    Notes: Summary Whole-cell sealed-on pipettes have been used to measure electrical properties of the plasmalemma surrounding protoplasts isolated from Black Mexican sweet corn shoot cells from suspension culture. In these protoplasts the membrane resting potential (V m ) was found to be −59±23 mV (n=23) in 1mm K o − . The meanV m became more negative as [K−] o decreased, but was more positive than the K+ equilibrium potential. There was no evidence of electrogenic pump activity. We describe four features of the current-voltage characteristic of the plasmalemma of these protoplasts which show voltagegated channel activity. Depolarization of the whole-cell membrane from the resting potential activates time- and voltage-dependent outward current through K+-selective channels. A local minimum in the outward current-voltage curve nearV m =150 mV suggests that these currents are mediated by two populations of K+-selective channels. The absence of this minimum in the presence of verapamil suggests that the activation of one channel population depends on the influx of Ca2+ into the cytoplasm. We identify unitary currents from two K+-selective channel populations (40 and 125 pS) which open when the membrane is depolarized; it is possible that these mediate the outward whole-cell current. Hyperpolarization of the membrane from the resting potential produces time- and voltage-dependent inward whole-cell current. Current activation is fast and follows an exponential time course. The current saturates and in some cases decreases at membrane potentials more negative than −175 mV. This current is conducted by poorly selective K+ channels, whereP Cl/P K=0.43±0.15. We describe a low conductance (20 pS) channel population of unknown selectivity which opens when the membrane is hyperpolarized. It is possible that these channels mediate inward whole-cell current. When the membrane is hyperpolarized to potentials more negative than −250 mV large, irregular inward current is activated. A third type of inward whole-cell current is briefly described. This activates slowly and with a U-shaped current-voltage curve over the range of membrane potentials −90〈V m 〈0 mV.

    Notes: Abstract Lactic acid bacteria increased from 3.2 × 106 and 1.6 × 107 c.f.u./g (wet wt) to 2 × 109 and 1.6 × 109 c.f.u./g after 12 to 24 h of fermentation of home-produced mawè (a dough produced from dehulled maize) and commercial mawè, respectively. In commercial mawè, the yeast count increased from 1.3 × 105 to 2.5 × 107 c.f.u./g after 48 h of fermentation before decreasing, whereas in the home-produced mawè it increased from 2.5 × 104 to 3.2 × 107 c.f.u./g after 72 h of fermentation; the dominant yeasts were mainly Candida krusei, although C. kefyr, C. glabrata and Saccharomyces cerevisiae were also present. Enterobacteriaceae counts increased slightly during the initial stage ofthe fermentation, but decreased below the detection level after 24 to 48 h. Enterobacter cloacae was mostly found in commercial mawè and Escherichia coli mostly in homeproduced mawè.

    Notes: Abstract The reaction of 15 varieties of maize to the growth and cyclopiazonic acid (CPA) production ofPenicillium griseofulvum was examined as a means of identifying varieties which would be resistant to this infestation. Only one variety, MMEH-25, was resistant to the fungus, seeds containing little CPA 30 days after infestation. This variety and Ganga-5, which was only moderately resistant, are popular commercial cultivars.

    Notes: Abstract Freshly prepared pozol, a traditional Mexican fermented maize dough, contained (c.f.u./g wet wt): lactic acid bacteria, 104 to 106; aerobic mesophiles, 104 to 105; Enterobacteriaceae, 102 to 103; yeasts, 102 to 104; and mould propagules, 〈103. After 30 h at 28°C the numbers were, respectively: 109, 7×106, 5×105, 106 and 104. Soaking alkali-treated grains overnight allowed lactic acid bacteria, aerobic mesophiles and Enterobacteriaceae to grow and these then constituted the primary microbial flora of the pozol dough. Grinding in a commercial mill inoculated the dough with lactic acid bacteria, aerobic mesophiles, Enterobacteriaceae and yeasts. Other processing stages, including the nature of the surface upon which the balls were made, handling of the dough, and air, contributed only minor numbers of microbes compared with the two major sources, soaking and grinding. The pH of pozol fell from an initial value of 7.3 to 4.6 after 30 h incubation at 28°C. The numbers of Enterobacteriaceae and other aerobic mesophilic bacteria remained constant between 11 and 30 h incubation and there was no evidence of the acidic conditions having any lethal effects on these organisms.

    Description / Table of Contents: Résumé Le parasitoïde scelionid,Platytelenomus busseolae, a été recueilli des ooplaques de la sésamie dans la région d'Istiaea, en Grèce Centrale. Le parasitisme naturel a été étudié pendant les années 1986 et 1987. Un nombre de 3 382 ooplaques avec un total de 205 227 œufs ont été recueillis. Les premiers œufs parasitisés ont été récoltés fin-juillet, début-août et les derniers à la mi-octobre. En 1986, 76,2% des ooplaques et 42,8% des œufs étaient parasitisés, mais en 1987 les valeurs respectives étaient 27,6% et 12,9%. L'activité moindre du parasitoïde en 1987 serait due aux conditions climatiques hivernales et estivales. En 1987, la sex ratio, femalle: mâle, a été 1,5: 1 et 26,2% des ooplaques seulement ont donné naissance à des ♂♂. Les ♀♀ fécondées ont produit 3 fois plus de ♀♀ que de ♂♂. Dans des conditions climatiques favorables, le parasitoïde peut jouer un rôle important dans la lutte biologique contre la sésamie.
    Notes: Abstract The egg parasitoid,Platytelenomus busseolae (Gahan) (Hymenoptera: Scelionidae) was recorded from egg masses of the corn stalk borer,Sesamia nonagrioides (Lefebvre) (Lepidoptera: Noctuidae) on maize in the area of Istiaea, Central Greece. Natural parasitism was studied during the years 1986, 1987. A total of 3,382 egg masses, containing 205,227 eggs, was collected from corn fields. Parasitized eggs were found from end July to mid-October. In 1986. 76.2% of the egg masses and 42.8% of all eggs were parasitized, while in 1987, respective values were 27.6% and 12.9%. The inferior performance of the parasitoid in 1987 may be due to the unfavorable winter and summer conditions. Parasitism decreased when the host population was at its peak, but it increased again within 1 to 3 weeks. A small percentage of egg masses was completely parasitized (9.5% and 4.7% for the 2 years respectively). In 1987, the sex ratio, ♀♀/♂♂ was 1.5: 1 and 26.2% of the egg masses produced only ♂♂. Mated ♀♀ produced 3 times more ♀♀ than ♂♂. It seems that the parasitoid, under favorable weather conditions, can play a significant role in the control of the corn stalk borer.

Eighty-seven percent of parasitism is expected on larvae with tunnel diameters of 0.9 to 1.8 mm and 6% on prepupae and pupae, given this host-size distribution. Preference was greatest for large host larvae (1.6 mm diameter), intermediate for other larvae in the range 0.9 to 1.8 mm, and least for pupae and small larvae.     Notes: Summary The genetic relationship between acetyl-coenzyme A carboxylase (ACCase; EC 6.4.1.2.) activity and herbicide tolerance was determined for five maize (Zea mays L.) mutants regenerated from tissue cultures selected for tolerance to the ACCase-inhibiting herbicides, sethoxydim and haloxyfop. Herbicide tolerance in each mutant was inherited as a partially dominant, nuclear mutation. Allelism tests indicated that the five mutations were allelic. Three distinguishable herbicide tolerance phenotypes were differentiated among the five mutants. Seedling tolerance to herbicide treatments cosegregated with reduced inhibition of seedling leaf ACCase activity by sethoxydim and haloxyfop demonstrating that alterations of ACCase conferred herbicide tolerance. Therefore, we propose that at least three, and possible five, new alleles of the maize ACCase structural gene (Acc1) were identified based on their differential response to sethoxydim and haloxyfop. The group represented by Acc1-S1, Acc1-S2 and Acc1-S3 alleles, which had similar phenotypes, exhibited tolerance to high rates of sethoxydim and haloxyfop. The Acc1-H1 allele lacked sethoxydim tolerance but was tolerant to haloxyfop, whereas the Acc1-H2 allele had intermediate tolerance to sethoxydim but was tolerant to haloxyfop. Differences in tolerance to the two herbicides among mutants homozygous for different Acc1 alleles suggested that sites on ACCase that interact with the different herbicides do not completely overlap. These mutations in maize ACCase should prove useful in characterization of the regulatory role of ACCase in fatty acid biosynthesis and in development of herbicide-tolerant maize germplasm.

    Notes: Abstract This paper presents a case study of the relationship between farmers' knowledge of maize varieties and their selection and management of these varieties under conditions of technological change. Research for this paper was done among Spanish-speaking small farmers in an ejidoof central Chiapas, Mexico. This ejido is well integrated into the market, and the use of modem technologies is widespread. This research demonstrates that farmers have an extensive and widely shared knowledge of their maize varieties. This knowledge reflects objective maize characteristics. Variation occurs in the farmers' selection and management of maize varieties, but on average the variation deviates from a random pattern in the direction predicted by the farmers' knowledge base. They have incorporated the technological changes brought about by development into their knowledge base. Farmers maintain maize varieties with contrasting traits, and their knowledge base provides important information about which traits and constraints are important to them.

    Notes: Summary Variation and covariation for agronomic and digestibility traits of silage maize are reported from a compilation of 22 years of experiments with standard sheep. Genotype effects of DOM and DCF were highly significant, even when genotypes were nested in earliness groups or brown-midrib hybrids discarded (Table 2). The genetic variance of crude fiber content was low, but the variance of the DCF was high. The genetic variance of DOM was about 4 times lower than genetic variance of DCF, but broad sense heritability of DOM was higher because of lower residual variance (Table 3). Genetic correlations between grain or crude fiber content and DOM had similar absolute values, 0.65, so each of these two traits was an important but not the unique determinant of silage maize quality. There was no correlation between DCF and grain or crude fiber content. Yield was not related to DOM or DCF within each group of earliness, allowing some quality improvement without agronomic drift (Table 4). Except for late hybrids, most of DOM differences between groups of earliness came from lowering of minimum value, while maximum values were similar. It was the contrary for DCF, with similar minimum values for all groups (Table 5). There was no obvious correlation between year of registration of hybrids and DOM or DCF, but extra new variation seemed obtained only for low values (Figs 1, 2; Table 6). IVDOM according to the APC process was a poor predictor of DOM, especially when brown-midrib hybrids and earliness effects were discarded; but because heritability of this trait was similar to DOM heritability, such enzymatic processes could probably be used to avoid drift towards poor DOM with hybrids bred for higher stalk strength.

    Notes: Summary Near infrared reflectance spectroscopy (NIRS) is emerging as a potentially useful tool in breeding plants for quality traits. Information is lacking, however, on its use in forage maize (Zea mays L.). The objectives of the present investigation were to evaluate the prediction of digestibility traits of maize stover using NIRS technique and to study the effect of laboratory (Lab) and NIRS assays on the estimates of variation and covariation. Twelve inbred lines, 66 diallel crosses among them and eight hybrid checks were evaluated at silage and grain harvests for 2 years at two agro-climatically diverse locations in the Federal Republic of Germany. Standard methods were used for Lab analysis of in vitro digestible organic matter (IVDOM), neutral detergent fibre (NDF), acid detergent fibre (ADF) and acid detergent lignin (ADL). In NIRS evaluation, calibration equations were developed by modified stepwise regression. The standard error of calibration was 2.5, 1.7, 1.4 and 0.4 for IVDOM, NDF, ADF and ADL, respectively. The coefficient of multiple determination was high (≥0.9) except for ADL. The validation statistics (standard error and correlation coefficient) were similar. In the diallel crosses, the estimates of variation (heritabilities in broad and narrow sense, genotypic and error coefficients of variation), generally, did not vary appreciably and consistently in the comparisons between Lab and NIRS methods particulary at silage harvest. Simple and rank correlations between Lab and NIRS analyses were positive and significant. These correlation coefficients based on the mean performance of the diallel crosses at silage harvest were 〉0.9 and at least 16 hybrids were common between the two analyses, among the upper one-third or lower one-third (22) hybrids. The study showed that NIRS analysis should be useful in maize breeding programmes wherein a large number of genotypes need to be evaluated.

    Notes: Summary White-endosperm (y y y) maize (Zea mays L.) is vital to the maize dry-milling industry. However, a substantially greater acreage in the U.S.A. is planted to yellow (Y Y Y) than white maize. Data on comparative performance of yellow and white maize is scanty, and results are inconclusive. This field study was conducted to provide information on dosage effects of y gene on grain yield, 500-kernel (K) weight, shelling percentage, plant and ear height, and cob weight. Yellow x yellow, yellow x white, and white x white endosperm crosses were made among five yellow and five white endosperm lines of Mo14W × Oh7B parentage. In the material studied, the genetic complement of the white-endosperm parent Mo14W varied from 12.50% to 87.50% across 15 treatments. The effect of Mo14W dosages among treatments was removed through analysis of covariance. If differences among treatment means existed, they were attributed to the y gene. A test of parallelism among regression coefficients (Ho. β1=β2=---β15=βc) indicated that these regressions coefficients for plant height, ear height, 500-K weight, and husk weight showed parallelism across yellow x yellow, yellow x white, and white x white crosses. Differences in plant height were significant in 3 out of 5 comparisons each between Y Y vs. y y, Y y vs. y y, and Y Y vs. Y y. Differences in ear height were significant in 1 of 5 Y Y vs. y y comparisons, in 3 of 5 Y y vs. y y comparisons, and in 1 of 5 Y Y vs. Y y comparisons. Differences in 500-K weight were significant in 4 of 5 Y Y vs. y y, in 2 of 5 Y Y vs. Y y and in 1 of 5 Y y vs. y y comparisons. Differences in husk weight were significant in all the Y Y vs. y y comparisons, in 4 of 5 Y y vs. y y comparisons, and in 1 of 5 Y Y vs. Y y comparisons. An increase in the y gene dosage caused, in general, a significant decrease in grain yield, 500-K weight, plant height, ear height, and husk weight. Shelling percentage was decreased but not appreciably. The Y y combination had a heterotic effect on plant height, ear height, and cob weight. An implication of the findings of this study is that it would be difficult to breed for high-yielding white maize if one used this y gene source. At this time, this is the only source (chromosome 6) available for white-endosperm maize. Non-parrallelism among regression coefficients was noted for shelling percentage, grain yield, and cob weight. Non-parallelism was observed in shelling percentage of the yellow x yellow crosses, cob weight of the yellow x white crosses and in grain yield of both yellow x yellow and yellow x white crosses. In general, an increase in mean Mo14W dosage resulted in increased grain yield. Mo14W dosages generally reduced cob weight in the yellow x white crosses. Shelling percentage increased as the mean Mo14W dosages increased for the yellow x yellow crosses. An increase in Mo14W dosages resulted in an increase in 500-K weight.

    Notes: Abstract Diethyl oxaloacetate was found to be a competitive inhibitor of maize leaf phosphoenolpyruvate carboxylase activity with respect to the substrate phosphoenolpyruvate. The Ki values, based on total diethyl oxaloacetate, decreased with increasing pH, while the Ki values, based on the enol tautomer (average of 4 μM), were similar and independent of pH. The results suggest that inhibition is dependent on the enol tautomer.     Notes: Abstract The monomeric chlorophyll-protein complexes, CP 29 and CP 26 seen in the Camm and Green (1980) and Dunahay and Staehelin (1986) green gels do not always migrate in the order of the apparent molecular weight of their apoproteins as determined by denaturing gel electrophoresis. In barley and corn they do, but in spinach they do not. In addition, in some higher plant species these chlorophyll-protein complexes comigrate on green gels causing confusion in the literature. To remedy this situation and circumvent future confusion, we propose that the CP 29 and CP 26 complexes be named according to the relative molecular weight of their apoproteins on denaturing gels. Our proposal is supported by the results obtained from four antibodies used on Western blot samples of whole thylakoids, grana membranes, and PS II preparations from different plants. The higher molecular weight proteins (proposed CP 29's) react strongly to one set of antibodies, and the lower molecular weight proteins (proposed CP 26's) react strongly to a different set. In spinach, CP 26 antibodies react also with CP 29, but the extent of the cross-reactivity depends critically on the gel electrophoresis system used. Accordingly, a lack of antibody reactivity under certain conditions may not indicate two proteins are unrelated, just simply that a particular epitope is no longer accessible following gel electrophoresis with a particular buffer system.

    Notes: Abstract The effect of growth temperatures on the photosynthetic performance of field grown maize (Zea mays cv. LG11) was examined for crops sown on 1 May and 28 June 1991. During the period of growth, 2 May to 10 August, the early-sown crop experienced temperatures below 10 °C on 33 occasions compared with only one for the crop sown on 28 June. The prolonged period of low temperatures throughout May and beginning of June were associated with a marked depression in CO2 assimilation rates at all light levels in the early-sown treatment. Chill-induced depression of the photosynthetic light-response curve reflected a sustained reduction in canopy leaf photosynthesis and crop radiation-use efficiency (RUE). During the early stages of growth, RUE was 65% lower in the early- than late-sown treatment, with no marked recovery observed in the former treatment until approximately three weeks after chilling conditions had ceased. Data show a close correlation between chill-induced depression of quantum yield (Φ) and RUE, with corresponding reductions in the light-saturated rates of CO2 assimilation (Pmax). The convexity of the light-response curve recovered most rapidly from chilling temperatures, and at least three weeks before any improvement in RUE. It is concluded that photosynthetic productivity of immature maize stands is less sensitive to changes in the convexity of the light response, than to changes in either Φ or Pmax.

Sur les 3 plantes, les mâles se développent en moyenne 1,2 j plus vite que les femelles. Les relations entre vitesse de développement et température ont été déterminées en utilisant à la fois un modèle linéaire et le modèle biophysique à 2 paramètres de Sharpe & DeMichele (1977). Les températures plus basses donnent des adultes des 2 sexes plus gros et plus lourds. Moins de G. nigrifrons se sont développés sur la graminée vivace que sur les 2 graminées annuelles à la température la plus élevée (30°C), tandis qu'à la température la plus basse (18°C) moins de cicadelles se sont développées sur les graminées annuelles. La température semble jouer un rôle significatif en déterminant l'adéquation des plantes comme hôtes convenant au développement de G. nigrifrons. Le potentiel de ponte de cette cicadelle avait été sous-estimé par les étudies précédentes.
    Notes: Abstract Population dynamics of the blackfaced leafhopper, Graminella nigrifrons (Forbes) (Homoptera: Cicadellidae), was studied at five temperatures (18, 21, 24, 27, & 30°C) in the laboratory on seedling maize (Zea mays L.), oats (Avena sativa L.), and the perennial johnsongrass (Sorghum halepense (L.) Pers.). Effects of temperature and host plant on egg to adult mean development time, adult size and weight, and fecundity were determined. Leafhoppers on all three hosts developed fastest at the highest temperature tested (21.3 days), and slowest at the lowest temperature tested (73.2 days). The duration from first to last adult eclosion was shortest at 30°C, (11.5 days) and longest at 18°C (43 days). The sex ratio of males to females did not differ from 1:1, but males developed an average of 1.2 days faster than females on all three hosts. Mean percent development/day ranged from 1.4% at 18°C to 4.7% at 30°C. The relationship of this development rate and temperature was determined using both a linear model and a variable parameter biophysical model. Based on these models, the developmental threshold is estimated at 12–15°C. The lowest temperature yielded larger and heavier adults (312 μg, dry weight) than did the highest temperature (225 μg). Fewer leafhoppers developed on the perennial than the annuals at 30°C and fewer on the annuals than the perennial at 18°C. Our results suggest that early in the season johnsongrass and perhaps other perennials are the superior developmental hosts for this leafhopper, whereas in midsummer when temperatures are highest, annuals are the better hosts.

    Notes: Abstract This study examined the impact of irrigation water on certain aspects of an insect-plant relationship in the field including the assessment of plant-mediated water effects on an herbivore's development, survival, and behavior, and plant damage parameters and host tissue water status. Maize (Zea mays L.) plants were arranged in a randomized complete block design in the field over two years in North Carolina (NC). Four blocks were subjected to three different irrigation treatments initiated ca. one week before anthesis: optimal, intermediate, deficit water supply. Each plant was infested with one (1986) or two (1987) black head stage, E-race European corn borer [Ostrinia nubilalis (Hübn.)] (ECB) egg masses at tasselling. ECB development, tunnelling site, and survival as well as plant tissue water status (tissue % water contents [θ] & leaf water potentials [Ψ]) were recorded through July. The irrigation effect on ECB parameters was slight and variable. Internal stalk temperatures of optimal plants were consistently cooler than their deficit counterparts (1 day-degree/day). With degree-days included as an explanatory variable in the analyses, there were no significant irrigation effects on the ECB parameters, except for total proportion of ECB's bored into maize plant parts. More ECB's bored into drier plants than in optimal plants; however, this trend was not significant in 1987. Plant water indices showed that though Ψ responded to irrigation, there were only minor changes in tissue θ, particularly in view of the larger diurnal tissue changes observed and the relatively high, sustained stalk θ levels seen over all treatments. Examination of ECB pupal θ confirmed that dietary water changes were minor or non-limiting to the insects' developmental physiology, because pupal θ was not sensitive to the irrigation treatments. Though water supply changes have drastic developmental and agronomic consequences for the maize plant, little or no changes were seen in the ECB feeding environment. Furthermore, a plant damage model was developed whereby the total % of ECB's tunnelled into maize was related to the mean larval age. The implications of this model on the understanding of ECB tunnelling behavior, damage potential, and pest management is noted.

    Notes: Abstract The diel flight periodicity of the nocturnal moth Chilo partellus (Swinhoe) (Lepidoptera;Pyralidae) was measured in the laboratory using an actograph and in the field with suction traps. Females showed almost no flight activity on the night of eclosion. Flight activity of mated females peaked before midnight, the period of peak oviposition activity. Male peak activity occurred after midnight coinciding with female eclosion. Presence or absence of females did not affect when or how long males were active.     Notes: Abstract Food production in the densely populated Rwandan highlands is impeded by soil erosion and loss in fertility. Alley cropping leguminous shrubs with food crops on contours is purported to minimize the problem and to provide wood and forage. This study reports the effect of Sesbania prunings plus moderate levels of N and P on bean (Phaseolus sp) and maize (Zea mays) yields in alley cropping. Experimental design was a randomized complete block with split-split plots. Main plots were alley width: 2, 4, 6 and 8 m. Phosphorus (P) at 0, 30 and 60 kg P2O5/ha occupied the subplot and nitrogen (N) at 0, 30 and 60 kg/ha were assigned at the sub-sub plot level. No P was applied to maize during the second cropping season. Crop yield in kg/ha included the land space taken by hedgerows. Bean yield in 6 m alleys (1100 kg/ha) was about twice that in 2 m alleys (500 kg/ha). Bean responded to N and P. Optimum alley width and N for bean yield were 6 m and 30 kg/ha, respectively. Cuttings from alley hedgerows provided stakes for climbing beans. Maize responded to N but not to residual P. The highest maize yield came from 8 m alleys with 40 kg/ha, but yields from 8 and 6 m alleys with the same N treatment were not significantly different. Maize plants in middle rows were significantly taller than plants in rows adjacent to hedgerows. Maize rust development showed significant alley width and row position effect. There were significantly fewer uredinia in the Sebania alleys relative to the control plots without shrub hedgerows. Rust development on maize in middle rows was significantly greater than development in border rows.

    Notes: Abstract A detailed study of the soil chemical and physical properties in seven-year-old alley cropping trial containingLeucaena leucocephala andFlemingia congesta in Northern Zambia is described. There was a strong correlation between the maize yield and the total amount of nitrogen applied, both from prunings and fertiliser, suggesting that a major reason for the observed benefit from alley cropping, particularly withLeucaena, was due to an improvement in nitrogen supply.Leucaena produced significantly more biomass, and its leaves had higher concentrations of nitrogen, phosphorous and potassium and lower C/N and C/P ratios than did those ofFlemingia. There was also evidence that the trees had a beneficial effect on other soil chemical properties; under the hedgerows, particularly those ofLeucaena, there were higher levels of organic carbon, Mg, K and ECEC, and pH values were also highest. It is suggested that higher levels of organic carbon in the alley crop treatments were responsible for the improvements observed in soil physical properties. Lower bulk density, lower penetration resistance, and a higher infiltration rate and pore volume fraction were measured in the alley crops, although there was no significant change in the soil water release parameters. A deteriorating effect of constant applications of nitrogen fertiliser on soil fertility was observed; as the level of urea application increased, there were significant decreases in Mg, K and pH, increases in Al and soil acidity, and higher penetrometer resistance. These results highlight the urgent need for further research on biological methods of maintaining soil fertility.

    Notes: Abstract Three multipurpose tree species (MPTS)-leucaena (Leucaena leucocephala), sesbania (Sesbania sesban var. nubica) and pigeonpea (Cajanus cajan) were pruned at a height of 60 cm above the ground every two months, and resulting plant biomass was incorporated into the soil as green manure. For comparison, maize (Zea mays) stover was also incorporated into some plots, while some other plots were left fallow. Varying quantities of plant biomass which were incorporated into the soil over a period of 12 months caused large changes in major soil plant nutrients, and it substantially improved soil fertility. To test for improved soil fertility, test crops of maize and beans (Phaseolus vulgaris) were grown on the plots after six biomass incorporations of 4806, 13603, 16659 and 7793 kg. ha−1yr−1 for pigeonpea, sesbania leucaena and maize, respectively. Responses of the test crops indicated that sesbania and leucaena green manures improved maize stover, cobs and grain yields; and bean haulms and grain yields by 77.6% when compared to fallow plots. Residual effects of green manures still resulted in significant (P 〈 0.05) yield differences in the test cropin the third testing season. Economic significance of green manures in increasing food crop yields to small scale farmers is discussed.

    Notes: Abstract Maize growing next toErythrina hedgerows had 44% lower biomass (p〈0.01) and 35% lower N content (p〈0.1) than maize growing in the middle of the alleys. Maize growing next toGliricidia hedgerows had the same biomass but 56% higher N content (p〈0.1) than maize growing in the middle of the alleys. However these differences did not develop until 2 months after sowing of the maize. Spatial variability in soil nitrogen mineralization and mulch nitrogen release did not explain any of the differences in growth or N uptake of the maize with respect to distance from the trees. It is hypothesized that the slower growth of the maize next to theErythrina trees after 2 months is due to increasing light and/or nutrient competition from the trees as the trees recover from pollarding. The apparent lack of competition fromGlirigidia may be due to different rates of regrowth or different shoot and root architecture. A theoretical model is described demonstrating that if a crop is to take advantage of the higher nutrient availability under alley cropping it must complete the major part of its growth before the trees recover significantly from pollarding, and start competing strongly with the crop.

    Notes: Abstract Measurements from trees and plants of a BrazilianInga/maize/beans alleycrop experiment were assessed for their distributional properties for three years of the study. Results for the tree data were similar to those found in an earlier study. Results for the crop data demonstrated similarities between the distributions for alleycrops and solecrops, nonnormality shown by positive skewness and peaked distributions. Changes in the distributional shapes were demonstrated from year to year as soil nutrient depletion increased and as climatic variables fluctuated. Variation across the rows in the alley was often of a similar size to variation from plant to plant within the rows. Initial recommendations for analysis of such data are given on the basis of these results.

In the laboratory test, growth of maize seedlings was significantly depressed by addition of leachate ofGliricidia prunings.     Notes: Abstract The effects of leucaena hedgerows, mulching with leucaena foliage (0,50 and 100% of harvested foliage), cowpea intercropping and adition of dairy cattle slurry (55 t ha−1 per maize crop) on the yield of maize from a sandy soil were assessed. The four-year results from five maize crops are reported. Except in the first year, yields of maize grain and stover were significantly reduced by 30% in the presence of leucaena hedgerows. Use of leucaena mulch eliminated this effect; application of all the harvested leucaena mulch (100%) increased the total maize grain yield of the five crops by 44% over sole maize. Hedgerow and mulching management required an additional 36 mandays labour ha−1 which was more than compensated by the increased maize yields. Furthermore leucaena hedgerows substantially depressed the growth of weeds between cropping seasons. Intercropping with cowpea significantly depressed yields of maize grain and stover when both crops were sown together, but not in later seasons when cowpea was sown four weeks after the maize. Application of slurry increased the total yields of maize grain and stover by 35 and 37%, respectively. The grain yield of maize in leucaena hedgerow treatments fertilized with slurry did not respond to application of more than 50% of leucaena foliage, which suggested that half of the foliage could be spared for feeding to livestock. The cumulative yield of maize grain from the highest yielding organic system was 85% of the yield from the fertilizer treatment. The study, which is continuing, demonstrates that large increases in agricultural productivity are possible through the intercropping of maize with woody forage and grain legumes and the integration of dairy cattle production into the system. It thus shows the importance of exploiting crop/livestock interactions.

    Notes: Abstract Theee trials to evaluat the potential of alley cropping in maize production on the low fertility, acidic soils in Northern Zambia are described. Leucaena leucocephala, Gliricidia sepium, Sesbania sesban, Albizia falcataria, Fleminga congesta, and Cassia spectabilis, were grown in alley crops with hybrid maize and soybean. All trials received recommended rates of P and K fertiliser; N fertiliser was applied at three rates as a subplot treatment. One trial received lime before establishment. Only in the limed trial was there a significant improvement in maize yields through alley cropping; when no N fertiliser was applied, incorporation of Leucaena leucocephala prunings resulted in an increase of up to 95% in yields, with a smaller improvement being produced by Flemingia congesta. There was a significant correlation between the quantity of prunings biomass applied and the proportional increase in maize yields over the control treatment. It is suggested that the lack of effect of most of the tree species on crop yields was due to low biomass production. An economic analysis showed that alley cropping with limed Leucaena was only profitable when fertiliser costs were high in relation to maize prices. However, lime is both expensive and difficult to obtain and transport for most small scale farmers in the region, and is therefore not a practical recommendation. It is suggested that future alley cropping research should focus on screening a wider range of tree species, including other species of Leucaena, for acid tolerance and higher biomass production.

    Notes: Abstract Long-term agroforestry demonstrations/trials using Acacia albida and other nitrogen fixing multipurpose trees/shrubs were initiated in mid-1982 to assess soil and crop productivity at a coastal lowland site characterized by low soil fertility, weed problems and consequent poor crop yields. Growth performance (height and diameter at breast height, dbh) of Acacia albida under eight densities rotationally intercropped with maize (Zea mays) and green gram (Phaseolus aureus), crop grain yields, soil fertility changes and weed control were assessed for a 5-year period (May 1982 to March 1987). A parallel-row systematic spacing field layout was used. Intercropped Acacia albida mean hight and dbh were 140 and 24% respectively higher than tree-only controls by the fifth year. Growth rate was low during the first year but increased in subsequent years to mean height and dbh of 9 m and 10 cm respectively by March 1987. While differences in dbh were significant, those between stand heights were not. Crop yields, especially under higher tree densities, declined considerably due to unexpected shade which also caused significant reductions in weed biomass. Soil fertility levels remained unchanged during the experimental period relative to the initial status, and differences between the intercropped Acacia albida plots and the tree — or crop — only control appeared not to be significant. We conclude that an understanding of the mechanism regulating leaf fall/retention phenomena of Acacia albida is crucial towards determining the intercropping potentials of the species.

    Notes: Abstract A maize-leguminous tree alley cropping system was studied on N-deficient soils in Hawaii to determine mulch effects on maize yields. Calliandra calothyrsus, Cajanus cajan, Cassia siamea, Gliciridia sepium, KX1 — Leucaena hybrid (L. pallida X L. diversifolia), L. leucocephala, L. pallida, L. salvadorensis, and Sesbania sesban were evaluated for green manure and yield of intercropped maize. S. sesban, G. sepium, L. pallida, and KX1 produced between 5 and 12 dry t/ha/yr green manure with nitrogen yields between 140 and 275 kg N/ha in 4 prunings. Maize yields responded linearly to nitrogen applied as green manure. Maize yield increased 12 kg for each kg of nitrogen applied. Additions of prunings from hedge rows were able to support maize grain yields at about 1800 kg/ha for two consecutive cropping seasons, while control plot yields averaged less than 600 kg/ha. Maize yields reflected the amount of nitrogen applied as green manure, regardless of tree species from which the nitrogen was derived. In March, maize yields decreased 34% in the row spaced 40 cm from the hedge, relative to the one spaced 110 cm away. In July, increasing the distance away from the hedge to 60 cm and coppicing the hedge earlier in maize growth, significantly improved grain yield. Grain yields decreased only 10% in the row closest to the trees.

    Notes: Abstract ‘Soil Changes Under Agroforestry’ (SCUAF), a computer model designed to predict changes in erosion, soil carbon and soil nitrogen over time within various agroforestry systems and climatic regimes, was assessed using input data from an undisturbed miombo woodland and an adjacent maize field in Zimbabwe. Predicted changes in soil carbon were in a realistic range for both miombo and maize treatments. However, the accuracy of the model may be a reflection of the detail required in its initialisation. Several problematic relationships were found within the model, in particular inconsistent patterns between nitrogen uptake and plant productivity under complex fertilisation simulations and a lack of attenuation of productivity in the simulation of miombo woodland as it approached maturity.

    Notes: Abstract The effect of feeding maize (Zea mays L.) husk and leucaena (Leucaena leucocephala) as a mixture or separately on the voluntary intake of maize husk was studied over a 40-day period. Ten male goats with an average weight of 15 kg were used. The maize husk intake and total dry matter intake for the two feeding methods was about the same. Thus, total dry matter intake was not affected (P〈0.05) by the feeding method. There was also no significant difference (P〈0.05) between the two feeding methods in the digestibility values. The data were considered to indicate that there were no significant differences in the intake of maize husk when fed in a mixture with leucaena or when offered separately but at the same time.

    Notes: Abstract The potential of allye cropping systems to sustain a high productivity with low external inputs and the reduction of maize/weed competition through weed suppression in different alley cropping and sole-cropped mulched systems was studied in Costa Rica at CATIE. Data were recorded eight years after establishment of the experiment. Plant residues ofErythrina poeppigiana trees (10 t/ha dry matter) planted at 6 by 3 m reduced weed biomass by 52%, whileGliricidia sepium trees (12 t/ha dry matter) planted at 6 by 0.5 m reduced weed biomass by 28%, in comparison to controls.Erythrina had a considerable impact on grass weeds, whileGliricidia reduced the incidence of some dicot weeds. Weed competition significantly reduced maize yield in all systems. Nevertheless weed suppression contributed to the higher maize grain yield underErythrina andGliricidia alley cropping of 3.8 t per hectare as opposed to the unmulched control yield of 2.0 t per hectare.

    Notes: Abstract Oviposition by the African migratory locust, Locusta migratoria migratorioides (Orthoptera: Acrididae), was studied in maize and wheat crops on the Orange Free State Highveld. Maize was shown to be the most important oviposition habitat with peak laying taking place in autumn and early winter when highest pod densities were recorded. Laying was mainly concentrated along the middle of the crop interrows in maize and within clearings in the wheat crop. Despite the uniform layout of these crops, the distribution of egg pods was found to be aggregated. Non-reproductive behaviour, such as locust aggregation, basking and feeding, as well as environmental factors appeared to influence the distribution of egg pods in these crops. Secondary selection for optinum soil moisture and compaction on the laying site enhanced the aggregation of pods.

    Notes: Abstract The toxicity and anticholinesterase activity of tremorgenic fungal metabolites, territrems, to the corn earworm, Helioverpa zea (Boddie) (Lepidoptera, Noctuidae) were examined. In oral toxicity studies, territrem A significantly inhibited growth by 40% at 25 ppm and by 89% at 250 ppm. Territrem B and an epoxy-derivative significantly inhibited growth by ca. 45% at 250 ppm. Piperonyl butoxide administered orally synergized the toxicity of the territrems tested. In topical toxicity studies, the epoxy-derivative caused 26% mortality and 25% growth retardation at 10 mg/gm insects. Territrem A and B were not significantly lethal, but did reduce growth by 15–20% at 10 mg/gm insect. Paraoxon tested in the same way caused 100% mortality at 25 ppm orally and 10 mg/gm topically. However, all territrems tested in vitro as inhibitors of H. zea head acetylcholinesterase were at least comparable to or were more active than paraoxon. Topically administered epoxy-territrem B also inhibited H. zea head acetylcholinesterase. The potential for development of new insecticides from a territrem lead structure is discussed.

    Notes: Abstract The relationship between second generation European corn borer (Ostrinia nubilalis Hübner) egg mass numbers and subsequent field corn damage, as measured by stalk cavity numbers, was studied in 79 fields in northeastern North Carolina over three years. A mean of 0.028 egg masses per plant (645 egg masses/23400 plants) was found over the course of the study. Significant differences in oviposition rate were detected between fields and years. Ca. 85% of egg masses were deposited in a five leaf zone surrounding the primary ear; of these, 89% were found on the lower four leaves in this zone. Egg masses appeared to be distributed randomly within fields but at low rates of incidence, and oviposition was relatively uniform between sampling areas within individual fields. Under moderate to high oviposition pressure (mean number of egg masses per plant over the duration of the oviposition period 〉ca. 0.02), eggs laid during the early phases of the oviposition period account for more subsequent stalk damage than eggs laid during the later phases of the oviposition period. Variations in second generation egg mass numbers accounted for ca. 70% of variation in stalk cavity numbers.

    Notes: Abstract Supplying both N forms (NH4 ++NO3 −) to the maize (Zea mays L.) plant can optimize productivity by enhancing reproductive development. However, the physiological factors responsible for this enhancement have not been elucidated, and may include the supply of cytokinin, a growth-regulating substance. Therefore, field and gravel hydroponic studies were conducted to examine the effect of N form (NH4 ++NO3 − versus predominantly NO3 −) and exogenous cytokinin treatment (six foliar applications of 22 μM 6-benzylaminopurine (BAP) during vegetative growth versus untreated) on productivity and yield of maize. For untreated plants, NH4 ++NO3 − nutrition increased grain yield by 11% and whole shoot N content by 6% compared with predominantly NO3 −. Cytokinin application to NO3 −-grown field plants increased grain yield to that of NH4 ++NO3 −-grown plants, which was the result of enhanced dry matter partitioning to the grain and decreased kernel abortion. Likewise, hydroponically grown maize supplied with NH4 ++NO3 − doubled anthesis earshoot weight, and enhanced the partitioning of dry matter to the shoot. NH4 ++NO3 − nutrition also increased earshoot N content by 200%, and whole shoot N accumulation by 25%. During vegetative growth, NH4 ++NO3 − plants had higher concentrations of endogenous cytokinins zeatin and zeatin riboside in root tips than NO3 −-grown plants. Based on these data, we suggest that the enhanced earshoot and grain production of plants supplied with NH4 ++NO3 − may be partly associated with an increased endogenous cytokinin supply.

Immature ears (5.1–10.0 mm long) of maize were cultured in kinetin medium to study microsporogenesis and pollen maturation in developing stamens. Male spikelets developed on ears cultured in kinetin medium. Meiosis-I began by 7 days of culture in the developing anthers and the mature tri-nucleate pollen grains were developed by 20 days of culture. Further, kinetin was required in the culture medium for at least initial 5 days to obtain the microspores in differentiated stamens. To observe the embryosac formation in developed ovaries, ears were cultured in control, kinetin (10.1–15.0 mm long ears) medium, and kinetin + gibberellic acid (5.1–10.0 mm long ears) medium. Formation of embryosacs was noticed in the developed ovaries which were sampled after 20 days of culture. This differential flower development using two growth regulators provides an opportunity to uncover the biochemistry and physiology of micro- and mega-gametophyte development in maize.

    Notes: Summary Contamination of maize (Zea mays L.) grain prior to harvest with aflatoxin frequently occurs in the southern U.S.A. and sources of resistance are needed. Maize from twelve genotypes (varieties) crossed to two testers was analyzed for aflatoxin B1 (AFB1) contamination at Baton Rouge, LA and Tifton, GA in 1987. Variety Philippines had the lowest amount of AFB1 at Baton Rouge and one of the lowest amount of AFB1 at Tifton. In the combined analysis of variance, the location x treatment x tester x variety interaction was significant. This interaction was analyzed and a stability variance (σ1 2) statistic calculated. Mean AFB1 accumulation and stability of AFB1 production were examined simultaneously, i.e., variety ranks for AFB1 and σ1 2 were summed. The most desirable (lowest rank sum) variety across locations, testers, and treatments (inoculation and control) was St. Charles White, followed by Delta Prolific White, and Hopi. The most inconsistent AFB1 accumulation, along with high AFB1, was on Fino, Atkinson, USAWI, Navajo, and Michoacan-21. The ranks of varieties between the inoculation and control treatments were different (rs=−0.51 for Baton Rouge and 0.29 for Tifton). The rs (rank correlation coefficient) for the control treatments between Baton Rouge and Tifton was 0.60 (P〈0.05) and that for the inoculation treatments was −0.20. The varieties identified in this study as having relatively low concentration of AFB1 should be useful to maize breeders in reducing AFB1 contamination in field maize.

    Notes: Summary The objective of this research was to obtain information about expression of genes controlling N concentration in the lower stalk internodes of maize at silking stages and in two other stages during the grain filling period. The inheritance of nitrate-N concentration, total N concentration, and the nitrate-N/total N ratio in the lower stalk internodes was measured in an eight-line diallel cross experiment and in an experiment involving segregating generations. The results show that the parameters examined, particularly nitrate-N concentration, are genetically controlled and that maize plants differ in this respect. Moreover, the magnitude of the general combining ability effects in relation to the size of specific combining ability obtained from the diallel analysis and the mean squares of the analysis of variance of generation means indicated additive heritability of nitrate-N concentration and other N-related traits in maize stalks. These findings suggest to us that a cyclic selection program in maize should be effective in modifying the level of N-related traits concentration in the stalks.

The leaf appearance rate and associated variance components are affected by a physiological stress attributable to the transition to autotrophic nutrition by the plant. At this stage, specific combining ability plays an important role in the total genetic variation. It is concluded that differing genetic controls exist in early and late material, and that this difference is also manifested in leaf initiation and elongation rates. The ground coverage rate, as a component of plant development, is genetically correlated to total dry matter yield. The genetic correlations vary according to the growth stage.

    Notes: Summary In this study, 31 elite inbred lines of maize (Zea mays L.) were analyzed with 149 clone-enzyme combinations for their respective RFLP profiles. Objectives were (1) to determine the utility of RFLPs for estimation of genetic similarties among 16 inbred lines from the Iowa Stiff Stalk Synthetic (BSSS) and among 15 inbred lines from the Lancaster Sure Crop (LSC) heterotic groups and (2) to compare genetic similarities based on molecular markers with those based on pedigree information. Coefficients of genetic similarity (GS) and coancestry (f) between pairs of lines from the same heterotic group were calculated from RFLP and pedigree data, respectively. For lines from the BSSS heterotic group, cluster analyses based on RFLP and pedigree data revealed similar associations. GS and f values were closely correlated (r=0.70) for related BSSS lines. For lines from the LSC heterotic group, considerable discrepancies existed between the GS and f values, especially for those pairs involving inbreds Va22 and Lo924. Effect of selection and/or erroneous pedigree records are discussed as possible explanations for the low correlation of GS and f values (r=0.07) for related LSC lines. RFLPs seem useful for investigation of relationships among maize inbreds, verification of pedigree records, and quantification of the degree of relatedness.

Mass honeycomb selection for yield was applied for 11 cycles from the F2 of the single cross maize hybrid F68×NE2 in a field B with silty-clay-loam soil texture. Response to selection compared to the original single cross hybrid was estimated both in absence of competition and under solid stand in the selection field B and in a nearby field A differing in soil texture (clay-loam). A strong crossover type of interaction occurred both under solid stand and in the absence of competition in the two tests the improved population outyielded the hybrid in field B in the two densities, but lagged behing the hybrid in field A. The results suggest that interaction between genotype and soil texture might affect efficiency of selection detrimentally unless provision is taken for parallel selection early in the crop improvement program in fields differing in soil texture.

Occasionally, A. parasiticus Speare is also used for this purpose. However, only limited data are available on whether one species is as effective as the other for identifying aflatoxin-resistant genotypes. Our objective was to determine relative aflatoxin accumulation on kernels of maize containing the leafy (Lfy) gene in response to A. flavus and A. parasiticus. Aflatoxin production by A. flavus and A. parasiticus on grain of seven maize synthetics containing the Lfy gene, viz., A619, A632, Mo17, B73, HY, Wf9, and 914, was examined in three environments in Louisiana. Ears were doubly inoculated at 14 and 21 days after mid-silk by atomizing over external silks a 2 ml suspension containing 2.0×107 spores ml-1 of either A. flavus or A. parasiticus. All genotypes responded similarly in the three environments to both the fungal species. Aflatoxin B1 and B2 production did not differ in the three environments. The seven genotypes did not differ in levels of aflatoxin accumulation in response to either A. flavus or A. parasiticus. Aflatoxin production by A. flavus was detected in maize samples from all three environments, but aflatoxin production by A. parasiticus was found only in samples from Winnsboro, where moisture stress occurred. Mean B1 and B2 production by A. flavus from the three environments was, respectively, four and one-half times and two times more than that by A. parasiticus.

    Notes: Summary Two multivariate techniques were used to characterize 30 maize accessions collected from three ecological zones of Bendel State, Nigeria. Differentiation of the 30 accessions into five distinct groups was achieved with the unweighted variable group method of the average linkage cluster analysis of 34 agronomic characters. Four of the taxonomic groups contained at least three accessions each, while a fifth group contained only one. The single accession contained in the fifth group was characterised by very early maturity, deeply pigmented leaves and ear husks and short statured plants. Clustering of the accessions into different phenetic groups followed substantially along geographical and traditionally trading routes. A few cases of overlapping of accessions from different geographical locations were obtained. Principal component analysis revealed that days to 50% tasseling and silking, number of nodes/plant, ear length, ear weight, leaf width, and kernel colour were the principal discriminatory characters that differentiated the accessions. Sixty-four percent of the total variation among the 34 characters were accounted for by the first five principal components while the first and second components accounted for 26 and 14 respectively.

    Notes: Summary Maize pollen quality was investigated after long-term storage both in a refrigerator and in liquid nitrogen by a combination of viability tests and cytochemical methods. Determination of the activities of a number of enzymes involved in important metabolic pathways was carried out. Quinone formation was also studied, as some products of secondary metabolism affect pollen grain viability. One year of pollen storage in liquid nitrogen had little effect on the activities of oxidoreductases and hydrolases and had no significant effect on pollen grain viability evaluated by acetocarmine, neutral red and acridine organe. Only the FCR test showed slightly decreased viability. After one and two years of storage in a refrigerator, pollen grain viability, tested using acetocarmine, neutral red and acridine orange, did not change substantially. Simultaneously the FCR test showed a considerable decrease in pollen grain viability. Long-term storage in a refrigerator resulted in the loss of cytochrome oxidase activity and rise of alcohol dehydrogenase, lactate dehydrogenase, peroxidase and polyphenoloxidase activities as well as of quinone formation.

    Notes: Abstract Success in breeding maize resistant to the European corn borer has been limited, with the exception of leaf feeding resistance. The inheritance of resistance to leaf, sheath-collar and ear damage in four maize germplasms and their six F1 crosses was evaluated by diallel analysis. Plants in a completely randomized design were artificially infested at the whorl, anthesis or full silk stage of plant development and were evaluated in the field for insect damage. A damage index based on size, number and location of lesions was calculated for each stage. Stowell's Evergreen (susceptible) had a mean damage index three to six times that of Maiz Amargo (resistant) at the whorl stage and the progeny plants were more resistant than the susceptible parent. Maiz Amargo and its crosses had significantly lower mean indices than Stowell's Evergreen for sheath-collar damage in Year 1 but not Year 2. Zapalote Chico, Maiz Amargo and their cross were significantly less damaged than other genotypes at the full silk stage. Heterosis values indicated an increase in resistance of crosses over the midparent average at all three stages of development. General combining ability (GCA) was highly significant for all types of damage, but specific combining ability was significant only for leaf damage. Based on estimates of GCA, Maiz Amargo was the best source of resistance to leaf and sheath-collar damage and both Zapalote Chico and Maiz Amargo would be good parents for ear damage resistance. Results suggest that resistance at different plant development stages can be combined.

    Notes: Summary Sixty-seven inbred lines of maize were evaluated for resistance to low-temperature photoinhibition of photosynthesis, using a pulse-modulated chlorophyll fluorescence technique. The evaluation procedure was based on leaf discs, which were exposed to a high irradiance (1000 µmol/m2/s) at 7°C. The efficiency of open PSII reaction centres as a reflection of overall photosynthesis was measured before and after a photoinhibition-inducing treatment. Exposure of leaf discs to photoinhibitory condition for 2, 4, and 8 hours resulted in an efficiency reduction of 30, 53 and 83%, respectively. Testing of inbred lines showed large differences for photoinhibition susceptibility. The difference in photosynthetic efficiency between the most extreme lines after a treatment of eight hours was 39%. Resistance to photoinhibition was shown to be relevant under cool field conditions. It proved to be a trait strongly amenable to selection.

    Notes: Abstract Although feedback regulation of photosynthesis by carbon metabolites has long been recognized and investigated, its underlying molecular mechanisms remain unclear. The recent discovery that glucose and acetate trigger global repression of maize photosynthetic gene transcription provides the first direct evidence that a fundamental mechanism is used for feedback regulation of photosynthesis in higher plants. The metabolic repression of photosynthetic genes has now been found in many higher plants and is likely universal. It overrides other regulation by light, tissue type and developmental stage, and serves potentially as the molecular basis of interactions between sink and source tissues. Using simplified and convenient cellular systems and transgenic plants, the study of metabolic regulation of gene expression offers an excellent opportunity for the understanding of global and coordinate gene control and metabolite-mediated signal transduction in higher plants.

    Notes: Summary The resistance to the European corn borer, Ostrinia nubilalis (Hubner), of thirty-seven indigenous landraces of Mexican maize was examined. The relationship of resistance and existing taxonomy of maize according to Wellhausen et al., (1952), was subjected to numerical analyses. Variables examined were: seedling DIM-BOA content, the extent of leaf feeding damage by early instar larvae both in the field and in the laboratory, the extent of plant breakage and stalk tunneling by late instar larvae, plant height, and the extent of fungal damage by Gibberella zeae and Ustilago maydis. Significant differences in resistance among the major taxonomic groupings were reflected in the existing taxonomy of maize (Wellhausen et al., 1952). The most resistant landrace grouping was Wellhausen et al.'s Prehistoric Mestizos. Eighty-five percent of a series of modern inbred lines, pools, and Argentine landraces were found to have affinities with one of the more susceptible groupings, the Ancient Indigenous Races, based upon analysis of the resistance data.

    Notes: Summary The capacity of the maize genotype 4c1 to regenerate microcalli and embryos from cultured microspores has been examined by comparing various cold pretreatments and culture media, using microspores and pollen at different stages of development. Viability of cultured cells was tested with FDA and their development was traced with light and fluorescence microscopy using DAPI as a nuclear dye. It was found that a pre-incubation of dissected flowers floating in a liquid nutrient medium at 8°C during 10–14 days was most successful for the induction of cell division. Among the developmental stages tested only the microspores appeared to regenerate. Subculture at 25°C in the same liquid medium, supplemented with 0.1 mg/l TIBA, gave highest rates of microspore division, i.e. up to 70% at 4 to 6 days of culture. All pathways described earlier for maize androgenic embryogenesis were observed within the 4c1 genotype. Symmetric divisions occurred in cultured microspores but most frequently asymmetric divisions lead to the formation of microcalli within 12 days of culture. In at least 60% of all dividing microspores cells were derived from the generative nucleus. Microcalli further developed either into loose or compact calli. Compact calli formed embryo-like structures.

First, to establish the phenetic relationships among the maize (Zea mays L.) landraces from Galicia (Northwestern Spain) maintained at the Misión Biológica de Galicia. Second, to assess the resemblance between a collection of Spanish populations (including the landraces from Galicia) and a set of US Corn Belt varieties. For the first objective 73 varieties from Galicia, along with 9 hybrid checks, were grown in 9×9 simple lattices at two locations for two years. For the second objective 131 populations from the US Corn Belt and Spain, along with 9 hybrid checks, were grown for three years in unreplicated experiments. Cluster analyses were carried out with the first principal components that accounted for a significant amount of the total variation. Four groups were found among the landraces from Galicia. The populations from Spain and America were classified as belonging to nine main groups. The replicated experiment was more accurate than the unreplicated one. However, it is concluded that an unreplicated test grown in several environments is accurate enough to detect the main groups, although some inaccuracies should be expected.

In response to this concern governments have set up biosafety regulations. In this paper a step-by-step scheme is described by which the safety of genetically modified organisms can be assessed. The first step is to determine the level of safety concern for the unmodified organism. Important aspects of the safety concern of the unmodified organism are the potential to hybridize with the wild flora and the ability of the crop to run wild. These aspects have been investigated by a desk study for four agricultural crops (potato, beet, oilseed rape and maize). Maize and potato are genetically isolated from the wild flora. Beet and oilseed rape on the contrary can potentially hybridize with wild relatives in the Netherlands. The risk assessment of the latter two species should focus entirely on the effects of the introduced genetic material.

    Notes: Abstract Analysis is made of the energetics of CO2 fixation, the photochemical quantum requirement per CO2 fixed, and sinks for utilising reductive power in the C4 plant maize. CO2 assimilation is the primary sink for energy derived from photochemistry, whereas photorespiration and nitrogen assimilation are relatively small sinks, particularly in developed leaves. Measurement of O2 exchange by mass spectrometry and CO2 exchange by infrared gas analysis under varying levels of CO2 indicate that there is a very close relationship between the true rate of O2 evolution from PS II and the net rate of CO2 fixation. Consideration is given to measurements of the quantum yields of PS II (φ PS II) from fluorescence analysis and of CO2 assimilation ( $$\phi _{CO_2 } $$ ) in maize over a wide range of conditions. The $${{\phi _{PSII} } \mathord{\left/ {\vphantom {{\phi _{PSII} } {\phi _{CO_2 } }}} \right. \kern-\nulldelimiterspace} {\phi _{CO_2 } }}$$ ratio was found to remain reasonably constant (ca. 12) over a range of physiological conditions in developed leaves, with varying temperature, CO2 concentrations, light intensities (from 5% to 100% of full sunlight), and following photoinhibition under high light and low temperature. A simple model for predicting CO2 assimilation from fluorescence parameters is presented and evaluated. It is concluded that under a wide range of conditions fluorescence parameters can be used to predict accurately and rapidly CO2 assimilation rates in maize.

    Notes: Summary This study analyzes the components of phenotypic variation for abscisic acid (ABA) content in maize (Zea mays L.) leaves and the correlations with drought sensitivity index (DSI) and silk delay (SD), involved in the reaction to water deficit. Eight early- and seven medium-maturity inbreds were examined in field trials: in 1990 with low irrigation volume and in 1991 with low and high irrigation volumes. ABA concentration and DSI were investigated at growth stages (S) corresponding to stem elongation (S3), appearance of the first husks (S4), and mid-end of silking (S5). The ABA concentration was significantly higher in conditions of water deficit and in the later growth stage. The genetic component for ABA concentration attained higher relative values than those shown by DSI in the same growth stages and by SD; moreover, it increased from growth stage 3 to stage 5. The genotype × year and genotype × irrigation volume interactions were smaller for ABA concentration than for DSI and SD. The broad sense heritability on a plant basis, estimated in drought conditions, for ABA concentration ranged from 21.4 to 55.1% according to maturity group and growth stage. A wide variation was observed among lines for ABA concentration: the medium-maturity group showed a three-fold range (from 219 to 605 ng ABA g−1 dry weight). No clear relationships between ABA concentration, DSI and SD were found. These results indicate the feasibility of a selection for ABA concentration within segregating populations derived from crosses between the inbred lines herein tested.

    Notes: Summary The variation of response to acetochlor was studied in a two-year experiment carried out by subjecting 18 maize (Zea mays L.) inbred lines to three herbicide rates (0, 2.5 and 5 l a.i./ha). In both years some inbred lines consistently exhibited an evident susceptibility, with symptoms consisting of the seedling curling up below the soil surface and causing impaired field emergence. The results were poor plant density and lower grain yield in comparison to control. In contrast, other lines showed a satisfactory level of tolerance. Then, to gather data on the inheritance of response to acetochlor, four tolerant inbreds (T) and four susceptible inbreds (S) were crossed to obtain four T×T, four S×S, four S×T and the corresponding four T×S two-way hybrids. These hybrids were studied together with parental lines by applying the same herbicide rates used in the previous trial. The S×S hybrids showed susceptibility to the herbicide and the T×T were tolerant, whereas the S×T and the T× S hybrids showed a tolerance very close to that of the T×T hybrids. No difference was found between S×T and the corresponding T×S hybrids as to herbicide response. On average, the 16 hybrids exhibited greater tolerance than the eight parental lines, with each hybrid group being more tolerant than its parental line group. These results indicate that tolerance to acetochlor is prevailingly dominant, that action of extranuclear genes should be ruled out, and that the level of plant vigour can affect herbicide reactiveness.

    Notes: Summary The identification of environments suitable for selection should enable plant breeders to test reliably a larger number of genotypes with given resources. This research was undertaken to evaluate discrimination ability (DA) and prediction ability (PA) of eight environments involving two locations. Eckartsweier (Eck) 1 and Voelkenrode (Voe); 2 yer. 1984 (84) and 1985 (85); and two harvests, silage harvest (I) and later (II). It was based on dry matter yield of plant (PDMY), ear (EDMY) and stover (SDMY) and on in vitro digestible organic matter (IVDOM), neutral detergent fibre (NDF), acid detergent fibre (ADF) and acid detergent lignin (ADL) of stover of 12 inbred lines and their 66 diallel crosses in maize (Zea mays L.). Linear regression coefficient of the performance of genotypes in an environment on that averaged across all environments and its degree of determination were used as measures of the DA and PA of environments, respectively. In hybrids, the DA of environments differed significantly for all traits except SDMY and ADL. Environment Eck851 showed better PA (≥0.63) than other environments for PDMY, EDMY, IVDOM, NDF, and ADF. Among the 12 two-environment combinations. EckI was a better predictor for PDMY, EDMY, IVDOM and NDF (PA≥0.80). The more productive environments showed better DA and PA for PDMY and FDMY than less productive ones. For IVDOM and NDF the first harvest provided better discrimination and prediction than the second harvest. In inbred lines the DA of environments differed significantly for EDMY only. In the present study, PA and DA seemed to be functions of the environments, PA was improved by combining two environments, and some environments seemed to be suitable for preliminary selection of genotypes for such diverse traits as yield and digestibility.

    Notes: Summary One of the primary avenues of improving the biological efficiency of crop plants is through the improvement of the leaf and canopy photosynthetic rates. However, the question whether the superiority of hybrids in respect of productivity potential could be traced, in retrospective fashion, to the photosynthetic parameters should be answered first. Once established and standardized, such indices could be streamlined in innovative breeding to predict the heterotic combinations for final yield formation. To answer this question, the photosynthetic rate and other components of photosynthetic efficiency were monitored among eight inbred stocks of maize (selected for variable photosynthetic rate from a previous study) and their all possible one-way crosses. The results demonstrated extensive heterosis in respect of photosynthetic rate and other photosynthetic indices which, in turn, was also realized in terms of higher biomass productivity and yield. Nonetheless, high leaf photosynthesis alone did not result in higher grain yield. On the contrary, component interaction among the photosynthetic indices like photosynthetic rate, leaf area/plant, number of leaves and chlorophyll content on one hand and the complementary gene action on the other, could be held responsible for higher yields in hybrids. Since the components of photosynthetic efficiency can predict heterosis for biomass and grain yield upto a reasonable extent, judicious incorporation of such indices in selection parameters for applied genetic protocols can add yet another dimension to the strategies for future yield improvements.

    Notes: Summary Presented are the results of a two-year study of haploid maize plants in the field. The haploids were produced with the aid of inducer line ZMS. In total, 604 and 1030 haploids were obtained and studied in the first and second years, respectively. Tassels of haploid plants were found to be almost completley sterile. Fertility of ears was studied by pollinating them with the pollen from diploid inbred lines, the cross resulting in almost all of the haploid ears carrying kernels. On average 27.4 kernels per ear of haploid plant were obtained in the first year of study and 26.3 in the second. These gave rise to normal diploid plants. This property allows genotypes selected at the level of haploid plants to be involved in breeding process. Unusual plants were found among haploids, phenotypically resembling homozygous lines. It was assumed that the plants had resulted from spontaneous chromosome doubling in haploids. The results of comparative studies of progenies of unusual plants and inbred lines derived from the same synthetic population are presented.

    Notes: Summary Accessions representing twenty eight landraces of maize were assessed for susceptibility to the maize weevil, Sitophilus zeamais in standardized resistance tests. Susceptibility parameters such as weight loss of grain, number of insect progeny produced, the Dobie index of susceptibility, and oviposition on grain were found to vary significantly by genotype, with exceptional resistance found in accessions representing the Naltel, Chapalote and Palomero landraces. As in improved genotypes, susceptibility was negatively correlated to phenolic and protein content of the variety tested but positively correlated to moisture content. A detailed analysis of the phenolics revealed the presence of diferulate which may contribute to mechanical resistance of the seed by cross-linking of cell wall hemicelluloses. A canonical discriminant analysis of the resistance data suggests that most of the five landrace groupings are significantly different. The ancient indigenous and prehistoric mestisos groupings are sources of resistant genotypes.

    Notes: Abstract Changes in the photosynthetic light-response curve during leaf development were determined for the fourth leaf of maize crops sown on 23 April and 10 June. Temperatures were unusually mild during late spring/early summer and neither crop experienced chilling damage. The concept of thermal time was used to take into account the effects of different temperature regimes on developmental stage, thereby enabling photosynthetic light-response data to be combined for both crops to describe the general response. Large variations in the upper asymptote (Asat) and convexity (Θ) of the light-response curve occurred during leaf development, but the maximum quantum yield of CO2 assimilation remained relatively constant throughout. Dark respiration rates showed a small but significant decrease with leaf age and generally ranged between 5 and 10% of Asat. A simple mathematical model was developed to assess the sensitivity of daily leaf photosynthesis (AL) to reductions in the Asat, Θ and the initial slope (Φ) of the light-response curve at different stages of leaf development. On bright sunny days, and at all developmental stages, AL was ca. twice as sensitive to reductions in Asat than to reductions in Φ and Θ. In overcast conditions, however, all three parameters contributed significantly to reductions in leaf photosynthesis, although the contribution of Φ was greatest during early leaf growth, while older leaves were most sensitive to depressions in Asat. The implications of these results for modelling the sensitivity of canopy photosynthesis to chill-induced photoinhibition of the light-response curve are discussed.

    Notes: Abstract The uptake of IAA into excised mesocotyls of non-irradiated maize seedlings was linear up to a concentration of about 4×M and in this range there was a tight coupling between the IAA in the stele and the cortex. Prior irradiation with white light of intact seedlings unbalanced this coupling. Lateral and longitudinal transport were affected differently. In the stele, the effect of prior irradiation on longitudinal transport was multiphasic, with an initial stimulatory effect followed by a negative effect at longer prior irradiation times. The lateral transport from the stele to the cortex showed no stimulatory effect and appeared to be inhibited within at least 15 min. The effect of the prior irradiation on longitudinal transport in the stele appeared to be a high intensity effect. In contrast, the effect of the prior irradiation on the lateral transport from the stele to the cortex was saturated at much lower intensities. The data suggest that the light induced change in the lateral transport of IAA between the two tissues may be due to changes either in the number of open lateral transport channels/carriers or in the conductivity of these channels/carriers.

    Notes: Abstract Heat-shocked maize seedlings (cv. Oh43) synthesize a characteristic set of heat-shock proteins (hsps) which include an 18 kDa family containing at least six major isoelectric variants. A cDNA library was constructed from poly(A)+ RNAs isolated from the radicles of heat-shocked maize seedlings and screened with a DNA fragment from the theoretical open reading frame of a putative Black Mexican Sweet maize hsp 18 genomic clone. Two clones, cMHSP18-3 and cMHSP18-9, were isolated, and the RNA transcripts generated from them were translated into proteins which immunoreact with antibodies directed against the maize 18 kDa hsps and exhibit the same electrophoretic characteristics as two different members of the 18 kDa hsp family. Nucleotide sequence analyses of the cDNAs in these clones reveal that their 5′ and 3′ untranslated regions exhibit 33–34% identity and that their protein encoding regions share 93% identity. The deduced amino acid sequences of these clones show 90% identity, and the apparent molecular masses and isoelectric points of these proteins agree with those established for two different 18 kDa hsps, numbered 3 and 6. This report substantiates that at least two of the 18 kDa hsps in maize are products of different but related genes. Moreover, it establishes that transcripts for these proteins accumulate during heat shock and that both their nucleotide and deduced amino acid sequences share extensive similarities with the class VI small hsps in soybean and with transcripts expressed during meiosis in Lilium.

    Notes: Abstract Chimeric genes containing the coding sequence for bacterial chloramphenicol acetyl transferase (CAT) have been introduced by electroporation into maize protoplasts (Black Mexican Sweet) and transient expression monitored by enzyme assays. Levels of CAT expression were enhanced 12-fold and 20-fold respectively by the inclusion of maize alcohol dehydrogenase-1 introns 2 and 6 in the chimeric construct. This enhancement was seen when the intron was placed within the 5′ translated region but not when it was located upstream of the promoter or within the 3′ untranslated region. Deletion of exon sequences adjacent to intron 2 abolished its ability to mediate enhancement of CAT gene expression. Northern analysis of protoplasts electroporated with intron constructs revealed elevated levels of CAT mRNA. However, this elevation was insufficient to account for the increased enzyme activity. One explanation of these results is that splicing affects both the quantity of mRNA.

    Notes: Abstract The 139 kb circular maize chloroplast genome was cloned in a linear form in a single yeast artificial chromosome (YAC), and several such clones were then analyzed to study the stability of large DNA sequences in YACs. The YAC clones were isolated from a partial maize DNA YAC library. The library was constructed byBal-31 nuclease digestion of high-molecular-weight total maize genomic DNA followed by blunt-end ligation into a modified YAC vector, pYAC11. All chloroplast genome-containing YACs (cpYACs) were characterized by comparing their restriction enzyme digestion patterns with that of purified maize chloroplast DNA (MCD). Comparison of the restriction patterns of four cpYACs, MY503, MY504, MY518 and MY526, with that of MCD did not show any change in the size of the restriction fragments except at the chloroplast DNA-vector junction fragments. These altered size fragments were confirmed as junction fragments by hybridization of the pYAC11 border fragments. The end-specific T3 and T7 transcripts synthesized from MY503 also identified the same junction fragments as did the pYAC11 border fragments. Most cpYACs were completely stable through the analyzed one hundred generations.

    Notes: Abstract The effect of Ac copy number on the frequency and timing of germinal transposition in tobacco was investigated using the streptomycin phosphotransferase gene (SPT) as an excision marker. The activity of one and two copies of the element was compared by selecting heterozygous and homozygous progeny of transformants carrying single SPT::Ac inserts. It was observed that increasing gene copy not only increases the transposition frequency, but also occasionally alters the timing of transposition such that earlier events are obtained. The result is that some homozygous plants generate multiple streptomycin resistant progeny carrying the same transposed Ac (trAc) element. We have also investigated the effect of modification of the sequence in the region around 82 bp downstream of the polyadenylation site and 177 bp from the 3′ end of the element on germinal excision frequencies. Alteration of three bases to create a BglII site at this location caused a minor decrease in germinal excision events, but insertion of four bases to create a Cla I site caused a 10-fold decrease in the transposition activity of the Ac element.

    Notes: Abstract In cereal root tissue, hypoxia induces the enzyme lactate dehydrogenase (LDH); (S)-lactate:NADH oxidoreductase, EC 1.1.1.27). In barley, both biochemical and genetic data indicate that five isozymes are induced under hypoxia. These isozymes are tetramers and arise from the random association of the products of two Ldh genes. The induction of LDH activity in root tissue has been shown to be correlated to an increase in LDH protein and Ldh mRNA. In order to more fully characterize the hypoxic induction of LDH, we have isolated a maize Ldh genemic clone which has strong homology at both the amino acid and nucleotide level to the barley LDH cDNA clones. The Ldh1 gene consists of two exons separated by a 296 bp intron, has the expected eukaryotic regulatory signals and a sequence that has strong homology to the maize anaerobic regulatory element.

    Notes: Abstract An Eco RI polymorphism, present in the 26S ribosomal RNA gene (rDNA) of the maize hybrid Sx19 (B73×Mo17), was utilized to correlate DNaseI sensitivity, undermethylation and expression in rDNA. We had previously shown that in double digest experiments with methylation-sensitive restriction enzymes and Eco RI, Sx19 rDNA fragments originating from repeat units with two Eco RI sites (8.0 kb) are undermethylated, whereas the fragments originating from repeat units with a single Eco RI site (9.1 kb) are completely methylated. In the present study, Sx19 rDNA chromatin structure was examined by purifying intact nuclei and digesting them briefly with increasing amounts of DNaseI. Analysis of this DNA with Eco RI showed that the 8.0 kb rDNA fragments are extremely sensitive to DNaseI digestion, while the 9.1 kb rDNA fragments are relatively resistant to digestion even at high levels of DNasel. Specific sites hypersensitive to DNaseI cleavage were mapped to a region in the intergenic spacer (IGS) near the major undermethylated site. Analysis of polymerase chain reaction (PCR) products synthesized using Sx19, B73, and Mo17 DNAs as templates indicated that the Eco RI polymorphism is due to a base change in the recognition site. Direct rRNA sequencing identified a single-base change in Mo17 rRNA relative to B73 rRNA. Allele-specific oligonucleotide probes containing the region surrounding and including the Eco RI polymorphic site were utilized to detect a nucleolar dominance effect by quantitating levels of rRNA transcripts in Sx19 and the reciprocal cross. Results from these single-base-pair mismatch hybridization experiments indicate that the majority of the rRNA transcripts in Sx19 orginate from the DNaseI-sensitive, undermethylated, Eco RI-polymorphic rDNA repeat units.

    Notes: Abstract Plant introns are generally short (〈200 nt) and AU-rich, and an elevated AU content is necessary for efficient splicing. Further, an intron in some plant genes enhances gene expression by a post-transcriptional mechanism that results in an increase of cytoplasmic mRNA. The specific intron features responsible for efficient splicing and enhancement are not well characterized in plants. Internal deletions of up to 80% of two maize introns, Adh1 intron 1 and maize actin intron 3, indicate that large segments of these introns are dispensable for normal function. However, extensive deletion (〉75%) of Adh1 intron 1 diminishes both intron enhancement and splicing efficiency. This finding suggests that there are internal sequence motifs required for intron function, and that these motifs are redundant. We attempted to repair a deletion-impaired Adh1 intron 1 variant by adding back either oligomers of defined sequence content or fragments of maize internal intron sequence. The addition of AU-rich oligomers improved splicing efficiency and in one example, a U-rich oligomer activated a cryptic 3′ splice acceptor. We also found that replacing the region proximal to the Adh1 intron 1 3′ acceptor with U-rich sequence improved splicing. We found that adding G- and C-rich oligomers did not improve intron function, but a C-rich oligomer activated a cryptic 3′ acceptor. The addition of internal intron sequence to an impaired intron improved splicing, and in one case, resulted in the activation of a cryptic 3′ acceptor. We present evidence that U-rich sequence immediately upstream of the 3′ splice junction increases splicing efficiency and contributes to, but does not uniquely specify, 3′ acceptor AG choice.

    Notes: Abstract Maize (Zea mays var. Caldera) plants were grown under sterile and not sterile conditions in soil in an atmosphere continuously enriched with 14CO2 for 36 days. At harvest the above ground parts of the maize were cut off and the roots were separated from the soil by washing with water. The soil was dispersed using ultrasonics and separated into soluble clay silt and sand fraction. Roots were included in the coarse sand fraction. 25% of the total label present in the soil ≡ 5.5% of that in the soil-plant system, was water soluble. Very little label was present in the clay and silt fractions (5% in each) and most (65%) was in the sand fraction as root material. Rapid extraction of soil after the removal of roots without ultrasonic treatment released soluble matter which amounted to 〈0.5% of the total activity in the soil-plant system. Isolated roots steeped in water released about 18% of their activity. Much of the soluble fraction may therefore be root lysate. The soil and roots accounted for 22% of the total activity in the soil-plant system. Glucose accounted for 89% of the sugars in the soluble fraction of the soil. 78% or more of the 14C present in glucose, arabinose and xylose constituents of the root-soil mixture occurred in the coarse and fine sand fractions, which also included root material. For mannose and galactose the value was 70% and for rhamnose, 50%. After reinoculation of the soil-root mixture and decomposition for 56 weeks, the water soluble material obtained on fractionation of the soil decreased to less than 1% of the total activity. A much greater proportion, 25%, was present in the clay fraction as a result of decomposition.

    Notes: Abstract The ability of several chemical soil N availability indexes to predict the N supplying capability (NSC) of soils to maize in the field was evaluated in 49 field experiments over 3 years in Pennsylvania. Two recently proposed indexes based on the amount of NH4 + released by treating soil with 2 M KCl at 100°C for 4 hr or with a pH 11.2 phosphate-borate buffer for 8 min were not good predictors of NSC (r=0.484 and 0.254, respectively). The absorbance of a 0.01 M NaHCO3 extract at 260 nm was also poorly correlated with field-measured NSC (r=0.412). The pre-sidedress soil NO3 − test (PSNT), the soil NO3 − concertration in the surface 20 cm of soil at planting, and the UV absorbance at 200 nm of a 0.01 M NaHCO3 extract of at planting soils were all moderately well correlated with NSC (r=0.672, 0.750, and 0.737, respectively). The latter two indexes are very simple, rapid, and inexpensive to perform and offer the possibility of improving the prediction of NSC in heavily manured fields.

    Notes: Abstract Maize (Zea mays) and sorghum (Sorghum bicolor) were inoculated with a range of VAM fungi and grown under water-stressed and unstressed conditions. There was considerable variation amongst the inocula in their effects on plant growth. Inoculation withGlomus clarum produced the biggest plants in each host, withGlomus monosporum andAcaulospora sp. giving the least growth overall. Root infection produced by the different inocula also varied, but levels were not correlated with effects on plant growth. Water-stress reduced plant growth, with the effects not being altered by mycorrhizal infection. VAM infection levels were not affected by water-stress. Spore production from most inocula was reduced by water-stress, both in total spore numbers and in terms of spores per gram plant weight. Sporulation ofG. clarum, G. epigeum andG. monosporum were affected less by stress than were the other inocula. Spore production was in general greater on sorghum than on maize, but the host effect varied amongst the inocula.

    Notes: Abstract An investigation was carried out to test whether the mechanism of increased zinc (Zn) uptake by mycorrhizal plants is similar to that of increased phosphorus (P) acquisition. Maize (Zea mays L.) was grown in pots containing sterilised calcareous soil either inoculated with a mycorrhizal fungus Glomus mosseae (Nicol. and Gerd.) Gerdemann and Trappe or with a mixture of mycorrhizal fungi, or remaining non-inoculated as non-mycorrhizal control. The pots had three compartments, a central one for root growth and two outer ones for hyphal growth. The compartmentalization was done using a 30-μm nylon net. The root compartment received low or high levels of P (50 or 100 mg kg−1 soil) in combination with low or high levels of P and micronutrients (2 or 10 mg kg−1 Fe, Zn and Cu) in the hyphal compartments. Mycorrhizal fungus inoculation did not influence shoot dry weight, but reduced root dry weight when low P levels were supplied to the root compartment. Irrespective of the P levels in the root compartment, shoots and roots of mycorrhizal plants had on average 95 and 115% higher P concentrations, and 164 and 22% higher Zn concentrations, respectively, compared to non-mycorrhizal plants. These higher concentrations could be attributed to a substantial translocation of P and Zn from hyphal compartments to the plant via the mycorrhizal hyphae. Mycorrhizal inoculation also enhanced copper concentration in roots (135%) but not in shoots. In contrast, manganese (Mn) concentrations in shoots and roots of mycorrhizal plants were distinctly lower, especially in plants inoculated with the mixture of mycorrhizal fungi. The results demonstrate that VA mycorrhizal hyphae uptake and translocation to the host is an important component of increased acquisition of P and Zn by mycorrhizal plants. The minimal hyphae contribution (delivery by the hyphae from the outer compartments) to the total plant acquisition ranged from 13 to 20% for P and from 16 to 25% for Zn.

    Notes: Abstract In 1985, 1986 and 1988, maize (Zea mays L.) was monocropped or intercropped with nodulating or nonnodulating soybean (Glycine max [L.] Merr.). In addition, nodulating soybean and nonnodulating soybean were each monocropped and grown as a mixture. In 1985 and 1986, treatments were grown at 0 and 60 kg N ha−1 and in 1988, the treatments were grown without N fertilizer, on N-depeted soil and on non-N-depleted soil. 15N enriched N was applied to soil in all the aforementioned treatments to test for N transfer from nodulating soybean to non-N2-fixing crops by the 15N dilution method. The 15N dilution method did not show the occurrence of N transfer in 1985 and 1986, but the N sparing effect was evident from the total N uptake of nonnodulating soybean, dwarf maize and tall maize, in 1986. In 1988, maize and nonnodulating soybean seed yields and seed N yields were higher on non-N-depleted soil than on N-depleted soil. On N-depleted soil, the 15N dilution method indicated N transfer from nodulating soybean to maize and to nonndulating soybean. At a population ratio of 67% nodulating soybean to 33% nonnodulating soybean, N transfer was also seen on non-N-depleted soil in 1988.

There was a 5-week lag period before any differences between experimental and control groups could be seen. After this period, the plants grown without phosphate devoted a higher percentage of their total mass to roots than did the controls. The roots of the phosphate-free plants were longer and less bushy than those of the control plants. Plants grown without phosphate showed an increase in the amount of acid phosphatase extractable from the external surfaces of the roots by a high salt solution. These phosphate stress responses were induced by 5 μM phosphate but not by 25 μM phosphate.

    Notes: Abstract Soluble root exudates were collected from three plants (Nicotiana tabacum L., Nicotiana rustica L. and Zea mays L.), grown under axenic and hydroponic conditions, in order to study their metal-solubilizing ability for Cd and other cations (Cu, Fe, Mn, Ni, Zn). Nicotiana spp. and Zea mays L. root exudates differed markedly in C/N ratio, sugars vs. amino acids ratio and organic acids content. Metals from two soils were extracted with either root exudate solutions, containing equal amounts of organic carbon, or distilled water as control. In the presence or absence of root exudates, the solubility of Fe and Mn was much higher than of the four other metals tested. Root exudates increased the solubilities of Mn and Cu, whereas those of Ni and Zn were not affected. Root exudates of Nicotiana spp. enhanced the solubility of Cd. The extent of Cd extraction by root exudates (N. tabacum L. N. rustica L. Zea mays L.) was similar to the order of Cd bioavailability to these three plants when grown on soil. An increase in Cd solubility in the rhizosphere of apical root zones due to root exudates is likely to be an important cause of the relatively high Cd accumulation in Nicotiana spp.

    Notes: Abstract The re-sorption of carbon compounds from the rhizosphere was investigated using 14C-labelled glucose, mannose and citric acid. Uptake in roots of 5-day-old, intact Zea mays plants in sterile solution culture was determined over a period of 48 hours. Under optimal growth conditions significant re-absorption of glucose and mannose occurred with the uptake rates being 70.5 and 40.2 μg compound g-1 root DW h-1, respectively. For glucose and mannose approximately 25% of the 14C label taken up by the root was recovered inside the plant as low-MW compounds and 33% polymerized into high MW compounds. 42% was respired as 14C-CO2. Citric acid by comparison showed little accumulation within plant tissues (11.4%) with most being respired and recovered as 14C-CO2 in KOH traps (88%). The uptake rate for citric acid was 34.8 μg g-1 root DW h-1. Over the 48-hour period a net efflux (i.e. exudation) of labelled plus unlabelled C was observed at a rate of 608 μg C g-1 root DW h-1 (equivalent to 1520 μg glucose/mannose). Of the C released as root exudates, a minimum estimate of the amount of C taken back into the plant was therefore 9.5%. The two main C fluxes within the rhizosphere, namely release of C by the root and uptake by the microorganisms, have been well documented in recent years. It is now apparent however that a third flux term, re-sorption of C by roots, can also be identified. This may play an important but previously overlooked role within the rhizosphere, and further work is needed to determine its significance. A comparison between exudate release in static (permitting accumulation of C) and flowing culture (C removed as it is released) was also made with the respective rates being 15.36 and 45.18 mg C g-1 root DW in 2 days. The relative important of re-sorption in natural environments and laboratory experiments is discussed.

    Notes: Abstract A field experiment was conducted on an Ultisol in Malaysia to assess changes in soil solution composition and their effects on maize and groundnut yields, resulting from limestone and gypsum application. The results showed that soil solution Ca in the lime treatment remained mainly in the zone of incorporation, but in the gypsum treatment some Ca moved into 15–30 cm zone. Al3+ and AlSO4 + were dominant Al species in the soil solution of nil treatment. Liming decreased Al3+ and AlSO4 +, but increased hydroxy-Al monomer activities. However, gypsum application resulted in an increase of AlSO4 + activity and in a decrease of Al3+ activity. Relative maize and groundnut yields were negatively correlated with Al3+, Al(OH)2+ and Alsum activities. Likewise, relative yields were negatively correlated with Al concentration and the Al concentration ratio and positively correlated with soil solution Mg concentration and Ca/Al ratio.

    Notes: Abstract Under high rainfall conditions on acid soils with shallow crop root systems the rate of N leaching is high. A simple model predicts nitrogen uptake efficiency as a function of the amount of rainfall in excess of evapotranspiration, rooting depth and degree to which N leaching is retarded in comparison with water transport. Field observations on acid soils in S.E. Nigeria and S. Sumatera (Indonesia) showed that this model should be amended to include the role of old tree root channels. Crop roots can follow these channels, which are coated with partly decayed organic matter, into the acid subsoil. Measurements of water infiltration with a Guelph permeameter and a methylene blue dye showed that such channels form the major infiltration sites during rainstorms. Implications for nitrogen use efficiency and cropping pattern are discussed.

    Notes: Abstract Due, in part, to the relative paucity of published comparisons based on field generated data, there is still poor agreement regarding the relative merits of lime requirement indices based on exchangeable Al and those based on pH. The objective of this study was to compare such indices using results obtained from long-term field experiments. Data were obtained over 22 site-years from lime trials conducted on clay (Typic Haplorthox) and sandy loam (Plinthic Paleudult) soils differing widely in organic carbon content. Relative maize (Zea mays L.) yields were used to compare the prognostic value of soil pH with indices obtained using exchangeable Al and exchangeable acidity (Al+H). Both within and across soils, pH proved to be markedly inferior to Al based indices. Exchangeable acid saturation of the effective cation exchange capacity, a readily obtained and popular index of lime requirement in some countries, proved as effective as less easily acquired indices based on exchangeable Al per se. The findings reported are consistent with those of many glasshouse studies and support the viewpoint that indices based on Al or acid saturation should replace pH as a measure of lime requirement.

    Notes: Abstract The restriction endonucleases Hpa II and Msp I were used to examine cytosine methylation in the ribosomal RNA genes (rDNA) of inbred lines of maize and species of teosinte. In all of the rDNAs examined, Msp I (not sensitive to mCpG) digestion yielded a distribution of lower molecular weight fragments indicative of multiple recognition sites. The majority of the rDNA arrays in an individual were inaccessible to Hpa II (sensitive to mCpG) cleavage, but a significant fraction (10–25%) was cleaved at least once by Hpa II into repeat unit length fragments (9.1 kbp). In some maize inbred lines, one or two additional fragment populations (less than 9.1 kbp in length) were also produced by Hpa II digestion. All of the unmethylated Hpa II sites mapped to the intergenic spacer (IGS), and the major unmethylated site was located approximately 800 bp 5′ to the start of the 18S RNA coding sequence. An Eco RI polymorphism, present in the 26S gene of certain inbred lines and hybrids, was utilized to investigate the organization of unmethylated repeat units in the rDNA array. In double digest experiments with Hpa II/Eco RI, the fragments from repeat units with two Eco RI sites were sensitive to Hpa II digestion, whereas, the fragments from repeat units with a single Eco RI site were almost completely resistant to Hpa II digestion. Similar digestion patterns were also observed in Eco RII (sensitive to mCNG)/Eco RI digests. These results suggest that unmethylated and Eco RI polymorphic sites occur in the same repeat units.

    Notes: Abstract Zeins from Zea mays L cv. Maya and coixins from Coix lacryma-jobi L. cv. Adlay were fractionated to obtain α-, β-, and γ-zein and α-, β-, and γ-coixin. The α-coixins were composed of 4 polypeptide classes of 27 kDa (C1), 25 kDa (C2), 17 kDa (C4) and 15 kDa (C5) with solubility properties very similar to those of the 22 kDa and 19 kDa α-zeins. Like the α-zeins, the C1 and C2 α-coixins corresponded to 80% of total Coix prolamins. The fraction corresponding to γ-coixin contained only one protein band of 22 kDa (C3). This coixin fraction has solubility properties similar to those of γ-zein and represents 15% of the total coixin. The β-zein fraction was composed of a major 17 kDa protein band, while the β-coixin fraction consisted of a mixture of α- and γ-coixins. Polyclonal antibodies raised against C1 recognized C1 and C2 and cross-reacted strongly with the 22 kDa α-zein, as did C4 and C5 antisera. The antiserum against γ-coixin showed strong cross-reaction with γ-zein. The homology between coixins and zeins was further investigated by using Southern hybridization analyses. The genomic DNA of maize and Coix were digested with several restriction enzymes and probed with cDNA clones representing 19 and 22 kDa α-zeins as well as the 28 and 16 kDa γ-zeins. The Coix genome showed complex cross-hybridization sequences with the 22 kDa α-zein cDNA, while no cross-hybridization was observed with the 19 kDa cDNA clone. The cDNA clone representing the 28 kDa γ-zein cross-hybridized with only one band of Coix genomic DNA, in contrast to the three bands observed in maize. This same Coix sequence also cross-hybridized with the cDNA clone representing the 16 kDa γ-zein. The relevance of these findings are discussed in the context of the origin of zein and coixin genes.

    Notes: Abstract Pathogenesis-related proteins (PRs) are plant proteins produced in leaves in response to infection by pathogens including viruses, viroids, fungi and bacteria. Information on the presence and/or expression of PRs in monocotyledonous plants is scarce. Here we report the identification of cDNA and genomic clones coding for a basic form of a protein from germinating maize seeds having a high homology with the group of PR-1 from tobacco. A cDNA library enriched in aleurone-specific sequences was prepared from maize seeds two days after germination. One clone was found to contain an open reading frame encoding a protein homologous to PR proteins from tomato (p14) and tobacco (PR-1 group). Sequence analysis of the corresponding genomic clone revealed that it was encoded by a single exon. Besides, DNA blot hybridization indicates that this PR-like protein is encoded by a single-copy gene in maize. The accumulation of its mRNA increases after rehydration of desiccated seeds. Furthermore, a relationship was found between its expression and infection by a natural pathogen of maize, the fungus Fusarium moniliforme. The possible role of this protein as a response mechanism following fungal infection in cereal seeds is discussed.

    Notes: Abstract A cDNA encoding maize enolase (2-phospho-D-glycerate hydrolase) was purified by functional genetic complementation using an enolase deficient mutant of Escherichia coli, DF261. This cDNA, pZM245, was characterized by restriction mapping and DNA sequence analysis. The cDNA contained an open reading frame encoding a protein of 446 amino acids with a high degree of similarity to enolase sequences from other organisms (72% identity to yeast enolase and 82% identity to human enolase). The pZM245 contains a correctly positioned consensus prokaryotic translation initiation sequence. The specific activity of enolase in maize increases to about twice its initial level after 48 hours of anaerobiosis. Northern-blot analysis showed a five-fold anaerobic induction in enolase mRNA, while heat shock or cold shock increased enolase mRNA levels only slightly. Southern-blot analysis of maize genomic DNA indicated that there is one copy of the pZM245 hybridizing sequence per haploid genome in maize.

    Notes: Abstract A search of compounds capable of inducing specific gene expression in plants without affecting growth and development led to the examination of changes in the pattern of gene expression in corn after treatment with substituted benzenesulfonamide herbicide safeners. Following hydroponic treatment of corn with the safener N-(aminocarbonyl)-2-chlorobenzenesulfonamide (2-CBSU), the specific induction of new translatable mRNA species was observed. Replicate copies of a cDNA library made using RNA from 2-CBSU-treated corn roots were differentially screened with cDNA probes made from either the same mRNA fraction used for library construction or mRNA isolated from roots treated with 2-chlorobenzenesulfonamide (2-CBSA), an inactive analog of the safener. Colonies showing hybridization only with the probe made using mRNA from 2-CBSU-treated roots were further characterized to assess the specificity of the induction and decay of the corresponding induced RNA species. RNA blot analyses showed two clones, designated In2-1 and In2-2, contained plasmids that hybridized to RNAs that were induced from an undetectable background in corn roots within 30 minutes after treatment with 2-CBSU. Leaf and meristem tissues showed similar inductions of the In2-1 and In2-2 RNA species after a delay of several hours. In addition, both RNA species were induced in corn by foliar application of 2-CBSU. In contrast, neither RNA species was induced following stress treatments of plants. These results indicate a substituted benzenesulfonamide safener might be used with the promoters from the In2-1 and In2-2 genes to develop a new inducible gene expression system for plants.

    Notes: Abstract We have isolated by differential hybridization a cDNA, termed Emb564, which is complementary to an 800 nt embryo-specific transcript in Zea mays. The expression of Emb564 can be enhanced by exogenous abscisic acid (ABA) and the effect of ABA on the accumulation of Emb564 transcript appears to be confined to embryos at the early and mid embryonic stages. In addition, Emb564 is expressed at low levels in ABA-deficient but not in ABA non-responsive embryos. Genomic analysis suggested that the Emb564 mRNA is encoded by a single gene. Sequence analysis showed that Emb564 exhibits extensive similarities with several known ABA-inducible genes.

    Notes: Abstract While a heat shock treatment of 40 °C or 45 °C induced the vegetative tissues of maize to produce the typical heat shock proteins (HSPs), germinating maize pollen exposed to the same temperatures did not synthesize these characteristic HSPs. Comparison of RNA accumulation in shoot and tassel tissue showed that mRNAs for HSP70 and HSP18 increased several-fold, reaching high levels within 1 or 2 hours. At the higher temperature of 45 °C these vegetative tissues were blocked in removal of an intron from the HSP70 mRNA precursor, which accumulated to a high level in tassel tissue. In germinating pollen exposed to heat shock, mRNAs for these HSPs were induced but accumulated only to low levels. The stressed pollen maintained high levels of RNA for α-tubulin, a representative normal transcript. It is likely that the defective heat shock response of maize pollen is due to inefficient induction of heat shock gene transcription.

    Notes: Abstract This study concerned the developmental regulation of wall-localized, hydroxyproline-containing proteins in maize tissues and organs. Silk and pericarp cell walls contained more peptidyl hydroxyproline than did walls of any vegetative tissue, although all tissues and organs accumulated these proteins as they matured. In many tissues, hydroxyproline-rich proteins are first associated with the wall in a soluble form before being insolubilized through covalent attachment to the matrix. Because hydroxyproline was more soluble earlier than later in development, it appears that insolubilization was occurring in maize tissues and organs as well. Tissue prints reacted with an anti-extensin antibody gave positive results, indicating the presence of a soluble form of this common hydroxyproline-rich glycoprotein (HRGP). Silk and pericarp cells actively synthesized this extensin from abundant transcripts. In vegetative tissues, extensin transcripts were somewhat more abundant in seedlings than in pre-anthesis or mature plants, but levels were much lower than in silk and pericarp. Southern blots of maize genomic DNA indicated that these extensin transcripts are encoded by a small multigene family. Potential roles for extensin in reproductive/protective tissues versus the embryo or vegetative tissues are suggested.

    Notes: Abstract As one component of a maize genome project, we report the analysis of a number of randomly selected cDNAs, by a combination of measuring mRNA expression, ‘single-pass’ sequencing (SPS), and genome mapping. Etiolated seedling (490) and membrane-free polysomal endosperm cDNA clones (576) were evaluated for their transcription levels by hybridizing with a probe prepared from total mRNA and categorized as corresponding to abundantly or rarely expressed mRNAs and as either constitutive or tissue-specific. A total of 313 clones from the two libraries were submitted to ‘single-pass’ sequencing from the presumed 5′ end of the mRNA and the nucleotide sequence compared with the GenBank database. About 61% of the clones showed no significant similarities within GenBank, 14% of the clones exhibited a high degree of similarity, while the remaining 25% exhibited a lesser degree of similarity. The chromosomal location of more than 300 clones was determined by RFLP mapping using standard populations. The results demonstrate that a combination of analyses provides synergistic information in eventually deducing the actual function of these types of clones.

    Notes: Abstract The mechanisms of intron recognition and processing have been well-studied in mammals and yeast, but in plants the biochemistry of splicing is not known and the rules for intron recognition are not clearly defined. To increase understanding of intron processing in plants, we have constructed new pairs of vectors, pSuccess and pFail, to assess the efficiency of splicing in maize cultured cells. In the pFail series we use translation of pre-mRNA to monitor the amount of unspliced RNA. We inserted an ATG codon in the Bz2 (Bronze-2) intron in frame with luciferase: this construct will express luciferase activity only when splicing fails. In the pSuccess series the spliced message is monitored by inserting an ATG upstream of the Bz2 intron in frame with luciferase: this construct will express luciferase activity only when splicing succeeds. We show here, using both the wild-type Bz2 intron and the same intron with splice site mutations, that the efficiency of splicing can be estimated by the ratio between the luciferase activities of the vector pairs. We also show that mutations in the unique U-rich motif inside the intron can modulate splicing. In addition, a GC-rich insertion in the first exon increases the efficiency of splicing, suggesting that exons also play an important role in intron recognition and/or processing.

    Notes: Abstract Four different β-tubulin coding sequences were isolated from a cDNA library prepared from RNA from maize seedling shoots. The four genes (designated tub4, tub6, tub7 and tub8) represented by these cDNA clones together with the tub1 and tub2 genes reported previously encode six β-tubulin isotypes with 90–97.5% amino acid sequence identity. Results from phylogenetic analysis of 17 β-tubulin genes from monocot and dicot plant species indicated that multiple extant lines of β-tubulin genes diverged from a single precursor after the appearance of the two major subfamilies of α-tubulin genes described previously. Hybridization probes from the 3′ non-coding regions of six β-tubulin clones were used to quantify the levels of corresponding tubulin transcripts in different maize tissues including developing anthers and pollen. The results from these dot blot hybridization experiments showed that all of the β-tubulin genes were expressed in most tissues examined, although each gene showed a unique pattern of differential transcript accumulation. The tub1 gene showed a high level of transcript accumulation in meristematic tissues and almost no accumulation in the late stages of anther development and in pollen. In contrast, the level of tub4 transcripts was very low during early stages of male flower development but increased markedly (more than 100 times) during the development of anthers and in pollen. Results from RNAse protection assays showed that this increased hybridization signal resulted from expression of transcripts from one or two genes closely related to tub4. The tub4-related transcripts were not present in shoot tissue. Transcripts from the tub2 gene accumulated to very low levels in all tissues examined, but reached the highest levels in young anthers containing microspore mother cells. RNAse protection assays were used to measure the absolute levels of α- and β-tubulin transcripts in seedling shoot and in pollen. The α-tubulin gene subfamily I genes (tua1, tua2, tua4) contributed the great majority of α-tubulin transcripts in both shoot and pollen. Transcripts from the β-tubulin genes tub4, tub6, tub7, and tub8 were predominant in shoot, but were much less significant than the tub4-related transcripts in pollen.

    Notes: Abstract Maize (Zea mays var. Caldera) plants were grown under sterile and not sterile conditions in soil in an atmosphere continuously enriched with14CO2 for 36 days. At harvest the above ground parts of the maize were cut off and the roots were separated from the soil by washing with water. The soil was dispersed using ultrasonics and separated into soluble clay silt and sand fraction. Roots were included in the coarse sand fraction. 25% of the total label present in the soil ≡ 5.5% of that in the soil-plant system, was water soluble. Very little label was present in the clay and silt fractions (5% in each) and most (65%) was in the sand fraction as root material. Rapid extraction of soil after the removal of roots without ultrasonic treatment released soluble matter which amounted to 〈0.5% of the total activity in the soil-plant system. Isolated roots steeped in water released about 18% of their activity. Much of the soluble fraction may therefore be root lysate. The soil and roots accounted for 22% of the total activity in the soil-plant system. Glucose accounted for 89% of the sugars in the soluble fraction of the soil. 78% or more of the14C present in glucose, arabinose and xylose constituents of the root-soil mixture occurred in the coarse and fine sand fractions, which also included root material. For mannose and galactose the value was 70% and for rhamnose, 50%. After reinoculation of the soil-root mixture and decomposition for 56 weeks, the water soluble material obtained on fractionation of the soil decreased to less than 1% of the total activity. A much greater proportion, 25%, was present in the clay fraction as a result of decomposition.

    Notes: Abstract Distribution of plant-parasitic nematode species at soil depths of 0–15 cm, 15–30 cm, and 30–45 cm was examined in two maize fields in Florida during 1987 and 1988. Soil type in both fields was an Arredondo fine sand, consisting of 94–96% sand at all three depths. On most sampling dates, more than 50% of the Belonolaimus longicaudatus collected occurred at the 0–15 cm depth. Only 20–30% of Criconemella sphaerocephala were present at this depth, and the depth distribution of this nematode differed in the two fields. The greatest densities of Pratylenchus brachyurus often occurred at 15–30 cm. Vertical distributions of Meloidogyne incognita and Paratrichodorus minor were erratic and showed marked seasonal variation. For all species examined except B. longicaudatus, a diagnostic soil sample collected 15 cm deep would recover only a minority of the nematode population present in the soil profile.

    Notes: Abstract To obtain plants of different P status, maize and soybean seedlings were grown for several weeks in flowing nutrient solution culture with P concentrations ranging from 0.03–100 µmol P L-1 kept constant within treatments. P uptake kinetics of the roots were then determined with intact plants in short-term experiments by monitoring P depletion of a 3.5 L volume of nutrient solution in contact with the roots. Results show maximum influx, Imax, 5-fold higher in plants which had been raised in solution of low compared with high P concentration. Because P concentrations in the plants were increased with increase in external P concentration, Imax was negatively related to % P in shoots. Michaelis constants, Km, were also increased with increased pretreatment P concentration, only slightly with soybean, but by a factor of 3 with maize. The minimum P concentration, Cmin, where net influx equals zero, was found between 0.06 and 0.3 µmol L-1 with a tendency to increase with pretreatment P concentration. Filtration of solutions at the end of the depletion experiment showed that part of the external P was associated with solid particles. It was concluded that plants markedly adapt P uptake kinetics to their P status, essentially by the increase of Imax, when internal P concentration decreases. Changes of Km and Cmin were of minor importance.

    Notes: Abstract Zinc deficiency decreased pollen viability in maize (Zea mays L. cv. G2) grown in sand culture. On restoring normal zinc supply to zinc-deficient plants before the pollen mother cell stage of anther development, the vegetative yield of plants and pollen fertility could be recovered to a large extent, but the recovery treatment was not effective when given after the release of microspores from the tetrads. If zinc deficiency was induced prior to microsporogenesis it did not significantly affect vegetative yield and ovule fertility, but decreased the fertility of pollen grains, even of those which visibly appeared normal. If the deficiency was induced after the release of microspores from the tetrads, not only vegetative yield and ovule fertility but pollen fertility also remained unaffected.

    Notes: Abstract Maize seedlings were grown for 10 to 20 days in either nutrient solution or in soils with or without fertilizer supply. Air temperature was kept uniform for all treatments, while root zone temperature (RZT) was varied between 12 and 24°C. In some treatments the basal part of the shoot (with apical shoot meristem and zone of leaf elongation) was lifted up to separate the indirect effects of root zone temperature on shoot growth from the direct effects of temperature on the shoot meristem. Shoot and root growth were decreased by low RZT to a similar extent irrespective of the growth medium (i.e. nutrient solution, fertilized or unfertilized soil). In all culture media Ca concentration was similar or even higher in plants grown at 12 as compared to 24°. At lower RZT concentrations of N, P and K in the shoot dry matter decreased in unfertilized soil, whereas in nutrient solution and fertilized soil only the K concentration decreased. When direct temperature effects on the shoot meristem were reduced by lifting the basal part of the shoot above the temperature-controlled root zone, shoot growth at low RZT was significantly increased in nutrient solution and fertilized soil, but not in unfertilized soil. In fertilized soil and nutrient solution at low RZT the uptake of K increased to a similar extent as plant growth, and thus shoot K concentration was not reduced by increasing shoot growth rates. In contrast, uptake of N and P was not increased, resulting in significantly decreased shoot concentrations. It is concluded that shoot growth at suboptimal RZT was limited both by a direct temperature effect on shoot activity and by a reduced nutrient supply through the roots. Nutrient concentrations in the shoot tissue at low RZT were not only influenced by availability in the substrate and dilution by growth, but also by the internal demand for growth.
