Beschreibung: Slope dynamics in volcanic environments comprise a wide spectrum of phenomena, from large lateral collapse to shallow debris remobilization, which may represent a major threat for human communities and infrastructures. Many volcanos built up from the ocean floor and large portions of the volcano edifice are submerged. In these settings, only the edifice’s summit can be investigated by terrestrial remote sensing and in-situ approaches. Growth and destruction, including tectonics and gravitational phenomena, affect entire volcano flanks and are not limited to the physical boundary of the sea level but could comprise their subaqueous parts.

Notizen: Summary In order to obtain markers for the Y chromosome ofSilene latifolia, we pooled equal weights of leaf tissue from 18 female siblings into one sample and repeated the process with 18 male siblings. Pooling was intended to provide a common genetic background for each sample, leaving the absence or presence of the Y chromosome as the primary difference between the two samples. DNA was extracted from each sample and subjected to polymerase chain reaction (PCR) amplification with arbitrary 10 bp primers. Four of 60 primers used gave an amplification with the male DNA not found among those from the female DNA. Each of these was subsequently shown to provide a reliable marker for the Y chromosome.

Notizen: Summary Disagreement has arisen over the presence of actin-containing microfilaments (Mfs) in angiosperm generative cells and sperm (GSP). In order to address this issue, we subjected GSP of Tradescantia virginiana, Nicotiana tabacum and Rhododendron laetum to a series of localizations using different antiactins, rhodamine phalloidin and antimyosin. Coordinate staining with antitubulin and Hoechst 33258 defined the status of the microtubule (Mt) cytoskeleton and stages of generative cell division. Additional experiments utilized cytochalasin D (CD). In no instance could Mfs be detected in GSP of the three species. Instead, Mfs seen at the periphery of GSP appear to be continuous with vegetative Mfs and thus are in the vegetative cytoplasm. Mfs are not seen in the constriction zone of dividing T. virginiana generative cells, nor are they indicated in the phragmoplast of N. tabacum and R. laetum. Myosin localizations reveal punctate staining in the vegetative cytoplasm and a thin line of fluorescence around the the outside of the generative cell. While CD seems to delay generative cell division, cytokinesis still takes place. CD-induced Mf fragments are evident in the vegetative cytoplasm but not in GSP. The weight of evidence therefore indicates that GSP do not contain Mfs. The implications of this conclusion for the behavior of GSP and the mechanism of cytokinesis in dividing generative cells are considerable.

Notizen: Summary The activity of acid phosphatase produced in pure culture by the endomycorrhizal fungus Hymenoscyphus ericae (Read) Korf & Kernan (H. ericae LPA 2) was inhibited by high phosphorus levels, alkaline pH, fluoride, molybdate and mannosidase, and activated by concanavalin A. Over 80% of the enzyme activity was due to two wall-bound acid phosphatase isozymes with the characteristics of mannose-rich glycoproteins. Antiserum was raised against the major, low-molecular-weight wall isozyme and its activity tested by immunoblotting and ELISA. The antiserum cross reacted 100% with exocellular (excreted) and 28% with cytoplasmic cellular fractions of H. ericae (LPA 2) cultures, and showed high reactivity with other strains of H. ericae but not with fungal isolates from Erica hispidula L. or E. mauritanica L. Ultrastructural localization of acid phosphatase by cytoenzymology and indirect immunogold labelling confirmed its association with the fungal wall in pure culture and showed that the influence of a high phosphorus level, fluoride and molybdate is through inactivation of the enzyme. Intense acid phosphatase activity, sensitive to the latter inhibitors, was also present on external hyphae growing over a host or non-host root but it was weak or absent from intracellular hyphae where these developed within a host root. Indirect immunolabelling confirmed that this acid phosphatase was of fungal origin and that the specific inhibitory effect of host cells is due to inactivation of the enzyme rather than repression of its synthesis. Possible implications of fungal acid phosphatase in ericoid endomycorrhizal infection processes are discussed together with mechanisms that may be regulating the enzyme activity.

Notizen: Abstract The existence of positive equilibrium solutions of the McKendrick equations for the dynamics of an age-structured population is studied as a bifurcation phenomenon using the inherent net reproductive rate n as a bifurcation parameter. The local existence and uniqueness of a branch of positive equilibria which bifurcates from the trivial (identically zero) solution at the critical value n=1 are proved by implicit function techniques under very mild smoothness conditions on the death and fertility rates as functional of age and population density. This first requires the development of a suitable linear theory. The lowest order terms in the Liapunov-Schmidt expansions are also calculated. This local analysis supplements earlier global bifurcation results of the author. The stability of both the trivial and the positive branch equilibria is studied by means of the principle of linearized stability. It is shown that in general the trivial solution losses stability as n increases through one while the stability of the branch solution is stable if and only if the bifurcation is supercritical. Thus the McKendrick equations exhibit, in the latter case, a standard exchange of stability with regard to equilibrium states as they depend on the inherent net reproductive rate. The derived lower order terms in the Liapunov-Schmidt expansions yield formulas which explicitly relate the direction of bifurcation to properties of the age-specific death and fertility rates as functionals of population density. Analytical and numerical results for some examples are given which illustrate these results.

Notizen: Abstract The Gurtin and Levine model5 is studied in this paper under the assumption that the fecundity of prey depends on age as well as on the total population sizes of prey and predators. The purpose of this study is to see the effect of this density dependence on the stability criteria for the equilibria of the model equations. It is shown that there are cases when, due to density dependence, the model which is originally neutrally stable becomes stable.

Notizen: Abstract The deterministic dynamics of the classical single-locus multiple-allele model of gametophytic incompatibility is analyzed with the intention to prove the conjecture that the symmetric state (uniform distribution of genotypes) is the only polymorphic equilibrium and that this equilibrium is globally asymptotically stable in the interior of the frequency simplex. It is shown that the minimum allelic frequency increases strictly over the generations as long as a uniform allelic distribution is not realized. Hence, the minimum allelic frequency is a Ljapunov function for the invariant set of genotypic frequencies characterized by a uniform allelic distribution. Within this set, the uniform genotypic distribution is approached in an exponential fashion, which proves the assertion. An evolutionary optimization rule associated with the global convergence to the symmetric state is implied by the fact that at this state the overall amount of pollen elimination resulting from incompatible crosses is minimized.

Notizen: Abstract We address the question of the long term coexistence of three interacting species whose dynamics are governed by the ordinary differential equations x i = X i f i (i = 1, 2, 3). In order for any theory in this area to be useful in practice, it must utilize as little information as possible concerning the forms of the f i , in view of the great difficulty of determining these experimentally. Here we obtain, under rather general conditions on the equations, a criterion for judging whether the species will coexist in a biologically realistic manner. This criterion depends only on the behaviour near the one or two species equilibria of the two dimensional subsystems, the behaviour there being relatively easy to examine experimentally. We show that with the exception of one class of cases, which is a generalization of a classical example of May and Leonard [21], invasibility at each such equilibrium suitably interpreted is both necessary and sufficient for a strong form of coexistence to hold. In the exceptional case, a single additional condition at the equilibria is enough to ensure coexistence.

Notizen: Abstract The FitzHugh-Nagumo equation u t =u xx +f(u)-w, u t =b(u-dw), is a simplified mathematical description of a nerve axon. If the parameters b〉0 and d⩾0 are taken suitably, this equation has two travelling pulse solutions with different propagation speeds. We study the stability of the fast pulse solution when b〉0 is sufficiently small. It is proved analytically by eigenvalue analysis that the fast pulse solution is “exponentially stable” if d〉0, and is “marginally stable” but not exponentially stable if d=0.

Notizen: Abstract Aquaporin 2 is a collecting duct water channel that is located in apical vesicles and in the apical plasma membrane of collecting duct principal cells. It shares 42% identity with the proximal tubule/thin descending limb water channel, CHIP28. The present study was aimed at addressing three questions concerning the location and behavior of the AQP2 protein under different conditions. First, does the AQP2 channel relocate to the apical membrane after vasopressin treatment? Our results show that AQP2 is diffusely distributed in cytoplasmic vesicles in collecting duct principal cells of homozygous Brattleboro rats that lack vasopressin. In rats injected with exogenous vasopressin, however, AQP2 became concentrated in the apical plasma membrane of principal cells, as determined by immunofluorescence and immunogold electron microscopy. This behavior is consistent with the idea that AQP2 is the vasopressin-sensitive water channel. Second, is the cellular location of AQP2 modified by microtubule disruption? In normal rats, AQP2 has a mainly apical and subapical location in principal cells, but in colchicine-treated rats, it is distributed on vesicles that are scattered throughout the entire cytoplasm. This is consistent with the dependence on microtubules of apical protein targeting in many cell types, and explains the inhibitory effect of microtubule disruption on the hydroosmotic response to vasopressin in sensitive epithelia, including the collecting duct. Third, is AQP2 present in neonatal rat kidneys? We show that AQP2 is abundant in principal cells from neonatal rats at all days after birth. The detection of AQP2 in early neonatal kidneys indicates that a lack of this protein is not responsible for the relatively weak urinary concentrating response to vasopressin seen in neonatal rats.

Notizen: Summary We used physiological recordings, intracellular dye injections and immunocytochemistry to further identify and characterize neurons in the buccal ganglia of Aplysia calif ornica expressing Small Cardioactive Peptide-like immunoreactivity (SCP-LI). Neurons were identified based upon soma size and position, input from premotor cells B4 and B5, axonal projections, muscle innervation patterns, and neuromuscular synaptic properties. SCP-LI was observed in several large ventral neurons including B6, B7, B9, B10, and B11, groups of s1 and s2 cluster cells, at least one cell located at a branch point of buccal nerve n2, and the previously characterized neurons B1, B2 and B15. B6, B7, B9, B10 and B11 are motoneurons to intrinsic muscles of the buccal mass, each displaying a unique innervation pattern and neuromuscular plasticity. Combined, these motoneurons innervate all major intrinsic buccal muscles (I1/I3, I2, I4, I5, I6). Correspondingly, SCP-LI processes were observed on all of these muscles. Innervation of multiple nonhomologous buccal muscles by individual motoneurons having extremely plastic neuromuscular synapses, represents a unique form of neuromuscular organization which is prevalent in this system. Our results show numerous SCPergic buccal motoneurons with widespread ganglionic processes and buccal muscle innervation, and support extensive use of SCPs in the control of feeding musculature.

Notizen: Abstract The ultrastructure of the storage parenchyma cells of the cotyledons of developing bean (Phaseolus vulgaris L.) seeds was examined in ultrathin frozen sections of specimens fixed in a mixture of glutaraldehyde, formaldehyde and acrolein, infused with 1 M sucrose, and sectioned at-80° C. Ultrastructural preservation was excellent and the various subcellular organelles could readily be identified in sections which had been stained with uranyl acetate and embedded in Carbowax and methylcellulose. The cells contained large protein bodies, numerous long endoplasmic reticulum cisternae, mitochondria, dictyosomes, and electron-dense vesicles ranging in size from 0.2 to 1.0 μm. Indirect immunolabelling using rabbit immunoglobulin G against purified phaseolin (7S reserve protein), and ferritin-conjugated goat immunoglobulin G against rabbit immunoglobulin G was used to localize phaseolin. With a concentration of 0.1 mg/ml of anti-phaseolin immunoglobin G, heavy labeling with ferritin particles was observed ober the protein bodies, the cisternae of the endoplasmic reticulum, and the vesicles. The same structures were lightly labeled when the concentration of the primary antigen was 0.02 mg/ml. Ferritin particles were also found over the Golgi bodies. The absence of ferritin particles from other organelles such as mitochondria and from areas of cytoplasm devoid of organelles indicated the specificity of the staining, especially at the lower concentration of anti-phaseolin immunoglobulin G.

Notizen: Abstract. An antiserum raised against the purified 33-kDa β-1,3-glucanase of wheat (Triticum aestivum L.) was employed to investigate the ultrastructural localization of the enzyme in wheat leaves infected with Puccinia recondita Rob. ex Desm. f.sp. tritici Eriks. and Henn. using a post-embedding immunogold labelling technique. In both compatible and incompatible interactions, β-1,3-glucanase was detected in the host plasmalemma and in the domain of the host cell wall near the plasmalemma of the mesophyll cells, but higher concentrations of the enzyme were detected in infected resistant wheat leaves than in infected susceptible ones. β-1,3-Glucanase was also found in the secondary thickening of xylem vessels and in the walls of guard cells, epidermal cells and phloem elements, while no labelling was observed in host organelles, viz. vacuoles, mitochondria, endoplasmic reticulum, Golgi bodies, nuclei and chloroplasts. A low concentration of the enzyme was detected on the intercellular hyphal wall and in the hyphal cytoplasm. In the compatible interaction, β-1,3-glucanase was demonstrated to accumulate predominantly in the haustorial wall and extrahaustorial matrix. In the incompatible interaction, strong labelling for β-1,3-glucanase was found in host cell wall appositions, in the extracellular matrix in the intercellular space, and in electron-dense structures of host origin which occurred in the incompatible interaction only.

Notizen: Abstract Development of legume seeds is accompanied by the synthesis of storage proteins and lectins, and the deposition of these proteins in protein-storage vacuoles (protein bodies). We examined the subcellular distribution, in developing seeds of the common bean, Phaseolus vulgaris L., of the major storage protein (phaseolin) and the major lectin (phytohemagglutinin, PHA). The proteins were localized using an indirect immunocytochemical method in which ultrathin frozen sections were immunolabeled with rabbit antibodies specific for either PHA or phaseolin. Bound antibodies were then localized using goat-anti-rabbit immunoglobulin G adsorbed onto 4- to 5-nm colloidal gold particles. The sections were post-fixed with OsO4, dehydrated, and embedded in plastic on the grids. Both PHA and phaseolin exhibited a similar distribution in the storage-parenchyma cells, being found primarily in the developing protein bodies. Endoplasmic reticulum and Golgi complexes (cisternal stacks and associated vesicles) also were specifically labeled for both proteins, whereas the cytosol and other organelles, such as mitochondria, were not. We interpret these observations as supporting the hypothesis that the transport of storage proteins and lectins from their site of synthesis, the rough endoplasmic reticulum, to their site of deposition, the protein bodies, is mediated by the Golgi complex.

Tissue was fixed in buffered aldehyde and embedded in LR White resin which was polymerised by addition of catalyst. Sections were labelled by the indirect method of absorption of Protein A-gold to specifically bound antibodies. This method gives high levels of specific labelling on sections which retain good ultrastructural preservation and have high contrast after conventional staining. The albumin is located throughout the cytoplasm although no labelling was found associated with the endoplasmic reticulum, Golgi apparatus, vacuoles-protein bodies or other organelles.

Notizen: Abstract Xanthine dehydrogenase (XDH, EC 1.2.1.37) was purified from root nodules of soybean (Glycine max) and used to prepare a polyclonal rabbit antiserum. Monospecificity of this antiserum was ascertained by sodium dodecyl sulfate-polyacrylamide gel electrophoresis of the immunoprecipate. During root nodule development of soybean, only one form of XDH was detected on an immunological basis. Titration of XDH by immunoelectrophoresis showed that a remarkable increase in the amount of XDH occurred between two and four weeks after inoculation, in parallel with the increase in enzyme activity. Localization of XDH by immunofluorescence indicated that the enzyme was present exclusively in uninfected cells where it appeared to be associated with discrete organellels

Notizen: Summary Oligonucleotides hybridizing to simple repetitive DNA patterns are highly informative as probes for DNA fingerprinting in all investigated animal species, including man. Here we demonstrate the applicability of this technique in higher plants. The oligonucleotide probes (GTG)5 and (GATA)4 were used to investigate the differences in DNA fingerprint patterns of the following angiosperm species: Triticum aestivum, Secale cereale, Hordeum vulgare, Beta vulgaris, Petunia hybrida, Brassica oleracea, and Nicotiana tabacum. Two species, Hordeum vulgare as a monocot and Beta vulgaris as a dicot, were analyzed in more detail. Their genomes differ considerably in both amount and organization of the simple repetitive sequences (GATA)n, (GACA)n, (GTG)n, and (CT)n due to the evolutionary distance of these two species. Furthermore, several lines and cultivars of Beta vulgaris and Hordeum vulgare can clearly be distinguished on the basis of their highly polymorphic patterns of these repetitive sequences.

Notizen: Summary Complete or partial nucleotide sequences of five different rRNA species, coded by nuclear (18S, 5.8S, and 5S) or chloroplast genomes (5S, 4.5S) from a number of seed plants were determined. Based on the sequence data, the phylogenetic dendrograms were built by two methods, maximum parsimony and compatibility. The topologies of the trees for different rRNA species are not fully congruent, but they share some common features. It may be concluded that both gymnosperms and angiosperms are monophyletic groups. The data obtained suggest that the divergence of all the main groups of extant gymnosperms occurred after the branching off of the angiosperm lineage. As the time of divergence of at least some of these gymnosperm taxa is traceable back to the early Carboniferous, it may be concluded that the genealogical splitting of gymnosperm and angiosperm lineages occurred before this event, at least 360 million years ago, i.e., much earlier than the first angiosperm fossils were dated. Ancestral forms of angiosperms ought to be searched for among Progymnospermopsida. Genealogical relationships among gymnosperm taxa cannot be deduced unambiguously on the basis of rRNA data. The only inference may be that the taxon Gnetopsida is an artificial one, andGnetum andEphedra belong to quite different lineages of gymnosperms. As to the phylogenetic position of the two Angiospermae classes, extant monocotyledons seem to be a paraphyletic group located near the root of the angiosperm branch; it emerged at the earliest stages of angiosperm evolution. We may conclude that either monocotyledonous characters arose independently more than once in different groups of ancient Magnoliales or that monocotyledonous forms rather than dicotyledonous Magnoliales were the earliest angiosperms. Judging by the rRNA trees, Magnoliales are the most ancient group among dicotyledons. The most ancient lineage among monocotyledons leads to modern Liliaceae.

Notizen: Abstract Since the beginning of this century evidence has accumulated which demonstrates that non-mammalian vertebrates possess photoreceptors situated deep within the brain. While many attempts have been made to localize these sensory cells, studies have either failed or been inconclusive. In this report we have used several experimental approaches to localize the deep brain photoreceptors of the lizard Anolis carolinensis. Using 3 antibodies that bind vertebrate cone opsins, we have immunolabelled cerebrospinal fluid (CSF)-contacting neurons located at the ventricular border within the nucleus ventromedialis of the septum. Western blot analysis indicates that these antibodies recognized a single 40 kD protein in ocular, anterior brain, and pineal extracts. Immunoblots of rodent brain did not show a similar protein band. We have also identified specific retinoids associated with phototransduction (11-cis and all-trans-3,4-didehydroretinaldehyde) within anterior brain extracts. This combined data provides the most detailed analysis of deep brain photoreceptors in any vertebrate. Consequently, we feel Anolis provides an excellent model to study this unexplored sensory system of the vertebrates.

Notizen: Abstract A protein solubilized from a membrane preparation of the gonad of Aplysia californica has been isolated by affinity chromatography, using bag cell egg-laying hormone (ELH) as the bound ligand, and partially purified and characterized by gel electrophoresis. The protein has an apparent molecular weight of 52 kDa and consists of two disulfide-linked subunits of about 30 kDa each. The protein is glycosylated and has an acidic pI. Approximately 10–15 μg of this protein can be isolated from a single ovotestis, representing less than 1% of the total protein in the gonad; but the protein could not be detected in buccal mass or body wall, tissues which do not have apparent response to ELH. Antibodies generated against this ELH-binding protein (ELHBP) were used to localize sites in the ovotestis which might contain this molecule and thus represent targets for egg-laying hormone. Immunocytochemical results indicate that the oocytes are a rich source of this protein, since their cytoplasm was the only detectable site of immunoreactivity. Whether this binding protein represents an egg-laying hormone receptor is uncertain, but its prevalence in oocytes suggests that ELH plays a signaling role on these gametes.

Notizen: Abstract In this article, we consider a two-person game in which the first player picks a row representative matrixM from a nonempty set $$A$$ ofm ×n matrices and a probability distributionx on {1,2,...,m} while the second player picks a column representative matrixN from a nonempty set ℬ ofm ×n matrices and a probability distribution y on 1,2,...,n. This leads to the respective costs ofx t My andx t Ny for these players. We establish the existence of an ɛ-equilibrium for this game under the assumption that $$A$$ and ℬ are bounded. When the sets $$A$$ and ℬ are compact in ℝmxn, the result yields an equilibrium state at which stage no player can decrease his cost by unilaterally changing his row/column selection and probability distribution. The result, when further specialized to singleton sets, reduces to the famous theorem of Nash on bimatrix games.

Notizen: Abstract Nuclear protein coding sequences from gymnosperms are currently scarce. We have determined 4 kb of nuclear protein coding sequences from gymnosperms and have collected and analyzed 〉60 kb of nuclear sequences from gymnosperms and nonspermatophytes in order to better understand processes influencing genome evolution in plants. We show that conifers possess both biased and nonbiased genes with respect to GC content, as found in monocots, suggesting that the common ancestor of conifers and monocots may have possessed both biased and nonbiased genes. The lack of biased genes in dicots is suggested to be a derived character for this lineage. We present a simple but speculative model of land-plant genome evolution which considers changes in GC bias and CpG frequency, respectively, as independent processes and which can account for several puzzling aspects of observed nucleotide frequencies in plant genes.

Notizen: Abstract Analysis of legumin-encoding cDNAs fromDioscorea caucasica Lipsky (Dioscoreaceae) and fromAsarum europaeum L. (Aristolochiaceae) shows that there is an especially methionine-rich legumin subfamily present in the lower angiosperm clades including the Monocotyledoneae. It is characterized by a methionine content of 3–4 mol% which is roughly triple the methionine proportion of most other legumins. These “MetR” legumins, if present, still have to be detected in the higher angiosperms including the important seed crops. Evolutionary analysis suggests that the MetR legumins are the result of a gene duplication allowing the differentiation of legumin genes according to their sulfur content. The duplication event must have taken place before the split into mono- and dicotyledonous plants but probably after the separation of angiosperms and gymnosperms.

Notizen: Summary Expression of androgen receptor (AR) in mouse osteoclast-like multi-nucleated cells (OCs) was examined with immunocytochemical techniques. Murine OCs were obtained by co-culturing mouse osteoblastic cells and bone marrow cells. Three preparations of polyclonal anti-AR antibody which were raised in rabbit against different parts of the human AR were employed for the experiments. Specific staining for AR was demonstrated in the nuclei and the perinuclear area of mouse OCs. This is the first report demonstrating the presence of AR in osteoclast-like cells.

Notizen: Summary Growth hormone (GH) may regulate tooth formation and bone remodeling associated with tooth eruption. This study reports the distribution of growth hormone receptor/binding protein in developing rat molars and adjacent alveolar bone by immunocytochemistry using well-characterized anti-growth hormone receptor monoclonal antibodies. These tissues represent an excellent model for studying the ontogenic changes that occur in odontogenic and osteogenic cells, as these cells are found in linear arrays displaying the various stages of morphological and functional differention, and differentiated function. Immunoreactivity was first seen in precementoblasts in contact with the epithelial root sheath, and preodontoblasts. However, growth hormone receptor immunoreactivity was associated primarily with the cytoplasm of odontogenic and osteogenic cells forming their respective matrices. Thus, cementoblasts and odontoblasts at sites of new matrix formation showed intense immunoreactivity whereas cementocytes and mature odontoblasts at later stages of tooth development were nonreactive. Osteoblasts engaged in intramembranous ossification in the alveolar bone were positive, although osteocytes and endosteal cells were immunonegative. Osteoclasts at sites of alveolar bone remodeling resorption were also immunopositive. These patterns of receptor expression parallel the ontogenic sequences of odontogenic and osteogenic cells and suggest that GH promotes the functional state of these cells. Our results also imply that GH may influence differentiation or differentiated functions associated with odontogenesis, osteogenesis, and bone remodeling independent of systemic insulin-like GF-I.

Although unaffected by surgical parasympathectomy, the periarterial and periductal nerves were abolished by chemical sympathectomy, suggesting that NPY is partially costored with sympathetic transmitters in nerve fibers.

Notizen: Summary Calmodulin is a regulator of several calcium-dependent cellular processes. It has been suggested that it plays a role in the mechanism of secretion. Employing an indirect immunoperoxidase technique at the light microscope level, this study demonstrates the presence of calmodulin in several exocytotic cells (mast cells, thyroid follicular cells, neurohypophyseal neurosecretory terminals, pancreaticβ-cells and pancreatic acinus cells) in rat and man. The positive staining reaction for calmodulin was granular and at least in the case of rat mast cells it appeared to be associated with the granule membrane.

Notizen: Abstract A set of ordinary differential equations describing a mechanical system subject to forcing and dissipation is considered. A topological argument is employed to show that if all time-dependent solutions of the governing equations are bounded, the equations admitN steady solutions, whereN is a positive odd integer and where at least (N−1)/2 of the steady solutions are unstable. The results are discussed in the context of atmospheric flows, and it is shown that truncated forms of the quasigeostrophic equations of dynamic meteorology and of Budyko-Sellers climate models satisfy the hypotheses of the theorem.

Notizen: Summary Vitellin was purified from eggs of the silkworm,Bombyx mori, by a new method in which vitellin was extracted from isolated yolk granules. The purified vitellin had a molecular weight of 540,000. An antibody against purified vitellin was prepared in rabbits. It reacted with the hemolymph vitellogenin as well as with purified vitellin, but not with other proteins in the hemolymph or in the extract from yolk granules. The anti-vitellin IgG was used to immunocytochemically locate vitellin in theBombyx non-diapause egg during early developmental stages. In the egg, just after oviposition, vitellin was located in internal yolk granules and in small yolk granules of the periplasm. During the early developmental stages studied, vitellin was not metabolized uniformly throughout the egg. The vitellin of the internal yolk granules located at the posterior-dorsal part and of the small peripheral yolk granules was utilized in 16 h and 2 days, respectively, after oviposition. A thin, very vitellin-poor layer was located between the periplasm and the vitellin-rich interior in the newly laid egg. it was always in close contact with the periphery where blastoderm and germ-band cells developed.

Notizen: Summary The developmental profile of the major haemolymph proteins (ceratitins) inCeratitis capitata was studied. Ceratitin concentration in the haemolymph decreases dramatically during the last days of pupal life, while the amounts of ceratitins in whole organism extracts remain unchanged. By electrophoretic, immunological and immunofluorescence techniques it was revealed that ceratitins are reabsorbed by the fat body and a fraction of them is deposited in the cuticle. The possible role of ceratitins is discussed.

By introducing the solutions ofJ- integral in the completely yielding state for the ideal plastic material, the critical condition of losing stability for the crack propagation in the specimen is obtained, based on the cusp catastrophe theory. The process of the crack growth from geometrical sense is described.

Notizen: Summary The immunocytochemical localization of the egg-jelly macromolecule, a fucose sulphate glycoconjugate (FSG) that induces the acrosome reaction in spermatozoa, was investigated in ovaries of the sea urchin Hemicentrotus pulcherrimus by use of a polyclonal antibody. The polyclonal antibody reacted with the accessory cells and oocytes in the ovarian lumen. In the accessory cells, evidence of an intense immunohistochemical reaction was observed in many globules of variable density. Products of the specific immunohistochemical reaction were frequently observed in the surface region of oocytes, at a distance from the ovarian wall. At the ultrastructural level, the polyclonal antibody was found to react with the material present in the vacuole-like structures of the globules in the accessory cells. Many gold particles, demonstrating specific immunolabelling, were associated with well-developed microvilli on the vitellogenic oocytes. In the mature oocytes, intense labelling was observed in the jelly coat but not in the vitelline coat. By contrast, oogonia and early oocytes were barely labelled. Quantitative data indicated that the extent of immunolabellings in the surface region of oocytes was very high in the vitellogenic and mature oocytes. In all cases, neither the oocyte cytoplasm nor the subcellular organelles were labelled. These results suggest that FSG is produced by the accessory cells and is deposited initially on the surface of vitellogenic oocytes for the formation of jelly. These findings may provide a new insight into the role of the accessory cells in the reproductive process of the sea urchin.

Notizen: Abstract In the sea urchin Hemicentrotus pulcherrimus, the egg-jelly macromolecule, a fucose sulphate glycoconjugate (FSG) that induces the acrosome reaction in spermatozoa, originates from the accessory cells in the ovary. In the present study we examined the seasonal variations in the distribution of FSG in the ovary by immunocytochemistry with a polyclonal antibody. An enzyme-linked immunosorbent assay indicated that FSG was present in supernatants of extracts of ovaries throughout the development of the ovary. However, the immunohistochemical study showed that there are marked seasonal changes in the distribution of FSG in ovaries. The polyclonal antibody reacted strongly with globules of accessory cells before the beginning of the breeding season (August to December). During the breeding season (February to April), the immunohistochemical reaction was found on the surface of oocytes but was weak in the accessory cells. At the ultrastructural level, the antibody reacted with globules of variable density in accessory cells. Intense immunolabelling was observed in the vacuole-like structures of the globules. Sometimes, products of the specific immunocytochemical reaction were found in the Golgi apparatus in these globules. Quantitative examination indicated that FSG was actively produced by the accessory cells from the late non-breeding season to the pre-breeding season. These results suggest that there are marked seasonal variations in the production of FSG by the accessory cells in the sea urchin ovary. These findings also provide new evidence that accessory cells exhibit dynamic changes during the reproductive process in the sea urchin.

Notizen: Abstract  Trees represent a, probably the, major component of the biosphere and have a unique place in the history of Mankind. One of their most fascinating features is the process of secondary growth which is effected principally by the secondary vascular system, the developmental continuum of secondary phloem, vascular cambium, and secondary xylem. However, for too long assumptions about the developmental biology of trees have had to be based upon studies of primary growth systems within annual, herbaceous species because study of the secondary vascular system had been largely ignored. Even when attempts are made to understand some of the most fundamental features of the secondary vascular system, such as xylogenesis, the current model system, isolated Zinnia mesophyll cells, is not entirely appropriate to the situation in the intact tree. Some deficiencies of the Zinnia system are discussed, and the advantages of the genus Populus as a model for study of the hardwood secondary vascular system are considered. Some of the new approaches which are poised to lead to significant advances in our knowledge of the cell bio-logy of the secondary vascular system of trees – spe-cifically of the cell wall, the plasmalemma, and the cytoskeleton – are discussed. The value of one of these new techniques – immunocytochemistry – is demonstrated by a consideration of recent work on the role of the cytoskeleton in the hardwood secondary vascular system.

Notizen: Abstract Research on bias in machine learning algorithms has generally been concerned with the impact of bias on predictive accuracy. We believe that there are other factors that should also play a role in the evaluation of bias. One such factor is the stability of the algorithm; in other words, the repeatability of the results. If we obtain two sets of data from the same phenomenon, with the same underlying probability distribution, then we would like our learning algorithm to induce approximately the same concepts from both sets of data. This paper introduces a method for quantifying stability, based on a measure of the agreement between concepts. We also discuss the relationships among stability, predictive accuracy, and bias.

Notizen: Summary The continuously erupting rabbit incisor tooth is normally thought of as having an enamel covered “crown” on its labial surface and a cementum covered “root” on its lingual surface. We have examined both surfaces of continuously erupting rabbit incisor teeth taken from near term embryos by a variety of means, including transmission and scanning electron microscopy, biochemical fractionation, and immunohistochemistry. In all cases, we could detect no qualitative difference in the early extracellular matrices taken from the labial and lingual surfaces of the teeth. Both matrices were shown to be composed of dentin and enamel, although the thickness and geometry of the enamel matrix on the lingual surface was somewhat different from that on the labial surface.

Notizen: Abstract An extensive network of extracellular fibrils was revealed by negative staining in the greenish gold algal flagellate, Chrysochromulina breviturrita. These fibrils were of uniform diameter (4–5 nm), sometimes exceeding 5 μm in length. In addition there were short, narrower fibrils (2–3 nm) on the surface of the flagella. Six protein bands were isolated from spent culture medium by SDS-PAGE and one of 80,000 Da was found to polymerize after dialysis into 4–5 nm fibrils identical to those found on the cell surface. Two other proteins of 58,000 Da and 65,000 Da also formed 4–5 nm fibrils but these were either rare or of a shorter length and different appearance. An antiserum directed against the surface 7 nm fibrils (fimbriae) of fungi agglutinated cells of C. breviturrita and some other Prymnesiophyceae and Chrysophyceae, but did not agglutinate cells of algal species in other groups. Immunofluorescence and protein A gold labelling confirmed that antigens related to fungal fimbriae were present on the surface of cells of C. breviturrita. Only the 80,000 and 58,000 Da proteins labelled heavily following protein A gold labelling. Some individual 4–5 nm fibrils labelled with gold were observed in the material prepared from the 80,000 Da band. These results therefore establish that C. breviturrita produces a surface network of fibrils that are serologically related to the fimbriae of fungi, and suggest a previously unrecognized relationship between members of the Prymnesiophyceae, Chrysophyceae and fungal groups.

Notizen: Abstract Immunogoldlabelling on ultrathin cryosections of Frankia sp. Cc1.17 showed specific labelling of nitrogenase in the spherical cells called vesicles. No label was found in the hyphae in any cells grown on a medium with combined nitrogen, nor in those to which no specific antiserum was added. Similar results were obtained with cultures grown under high (20%) and low (2%) oxygen tension in the gas phase.

Notizen: Abstract We study thermomechanical deformations of a viscoplastic body deformed in simple shear. The strain gradients are taken as independent kinematic variables and the corresponding higher order stresses are included in the balance laws, and the equation for the yield surface. Three different functional relationships, the power law, and those proposed by Wright and Batra, and Johnson and Cook are used to relate the effective strain rate to the effective stress and temperature. Effects of strain hardening of the material and elastic deformations are neglected. The homogeneous solution of the problem is perturbed and the stability of the problem linear in the perturbation variables is studied. Following Wright and Ockendon's postulate that the wavelength whose initial growth rate is maximum determines the minimum spacing between adjacent shear bands, the shear band spacing is computed. It is found that the minimum shear band spacing is very sensitive to the thermal softening coefficient/exponent, the material characteristic length and the nominal strain-rate. Approximate analytical expressions for the critical wave length for heat conducting nonpolar materials and locally adiabatic deformations of gradient dependent materials are also derived.

Notizen: Abstract The subcellular localization of dihydroxyacetone synthase (DHAS) in the methylotrophic yeast Hansenula polymorpha was studied by various biochemical and immunocytochemical methods. After cell fractionation involving differential and sucrose gradient centrifugation of protoplast homogenates prepared from methanol-grown cells, DHAS cosedimented with the peroxisomal enzymes alcohol oxidase and catalase. Electron microscopy of this fraction showed that it contained mainly intact peroxisomes, whereas SDS-polyacrylamide gel electrophoresis revealed two major protein bands (75 and 78 kDa) which were identified as alcohol oxidase and DHAS, respectively. The localization of DHAS in peroxisomes was further established by immunocytochemistry. After immuno-gold staining carried out on ultrathin sections of methanol-grown H. polymorpha using DHAS-specific antibodies, labelling was confined to the peroxisomal matrix.

Notizen: Abstract Cells of Methanobacterium thermoautotrophicum were fixed with glutaraldehyde, sectioned and labeled with antibodies against the β subunit of component C (=methyl-CoM reductase) of methyl-CoM reductase system and with colloidal gold-labeled protein A. It was found that the gold particles were located predominantly in the vicinity of the cytoplasmic membrane, when the cells were grown under conditions where methyl-CoM reductase was not overproduced. This finding confirms the recent data obtained with Methanococcus voltae showing via the same immunocytochemical localization technique that in this organism methyl-CoM reductase is membrane associated.

Notizen: Abstract We have investigated the in situ location of glutaconyl-CoA decarboxylase and 2-htdroxyglutaryl-CoA dehydratase in Acidaminococcus fermentans using the antibody-gold and protein A-gold techniques carried out as a post-embedding immunoelectron microscopic procedure. Polyclonal antisera were raised in rabbits against homogeneous fractions of the enzymes. Anaerobically grown cells of A. fermentans of the late exponential growth phase were fixed with 0.2% glutaraldehyde and 0.3% formaldehyde (final concentrations) in the growth medium. Dehydration of the cells was achieved with methanol. The cells were embedded in the low temperature embedding resin Lowicryl K4M. The markers indicative for antigenic sites of the two enzymes unequivocally demonstrate that the sodium pump glutaconyl-CoA decarboxylase is located at the cell periphery being a membrane-bound enzyme as expected whereas 2-hydroxyglutaryl-CoA dehydratase is a soluble cytoplasmic enzyme.

Notizen: Abstract  The immunogold silver staining method (IGSS) is widely used as a sensitive and specific immunohistochemical visualisation technique. IGSS involves the specific deposition of metallic silver at the site of immunogold labelling and provides a means of visualisation at low magnification by light or electron microscopy. Silver developers for IGSS rapidly deposit metallic silver only at the site of heavy metals, including gold and silver, because of their catalytic activity. The developing solution contains the silver ions and reducing agent necessary for this reaction. Using different silver salts as ion donors and by selecting an appropriate temperature and pH, visible amounts of silver can be deposited in a few minutes at the site of colloidal gold labelling while little non-specific background deposition occurs. Inclusion of protective colloids in the solution can also be used to control the reaction. Although studies of the chemical basis of silver deposition around unlabelled colloidal gold date back to 1939, immunogold enhancement by silver was established in 1983. The IGSS method evolved from the combination of disparate photographic, histochemical and immunogold techniques which have been effectively combined and optimised over the last 10 years to provide a visualisation system which is well suited to many immunohistochemical studies.

Notizen: Summary We have examined the effects of light on circadian locomotor rhythms in retinally degenerate mice (C57BL/6J mice homozygous for the rd allele: rd/rd). The sensitivity of circadian photoreception in these mice was determined by varying the irradiance of a 15 min light pulse (515 nm) given at circadian time 16 and meauring the magnitude of the phase shift of the locomotor rhythm. Experiments were performed on animals 80 days of age. Despite the loss of visual photoreceptors in the rd/rd retina, animals showed circadian responses to light that were indistinguishable from mice with normal retinas (rd/+ and +/+). While no photoreceptor outersegments were identified in the retina of rd/rd animals (80–100 days of age), we did identify a small number of perikarya that were immunoreactive for cone opsins, and even fewer cells that contained rod opsin. Using HPLC, we demonstrated the presence and photoisomerization of the rhodopsin chromophore 11-cis retinaldehyde. The rd/rd retinas contained about 2% of 11-cis retinaldehyde found in +/+ retinas. We have yet to determine whether the opsin immunoreactive perikarya or some other unidentified cell type mediate circadian light detection in the rd/rd retina.

Notizen: Abstract Immunogold labelling techniques on ultrathin sections of low temperature embedded cells yielded evidence for the periplasmic location of the respiratory enzymes N2O reductase and nitrite reductase (cytochrome cd 1) in Pseudomonas stutzeri strain ZoBell. Cell fractionation by spheroplast preparation and two-dimensional electrophoresis showed the absence of a membrane association of these enzymes. Immunocytochemical localization of N2O reductase in a mutant strain deficient in the chromophore of N2O reductase showed the gold label at the cell periphery, indicating that the copper chromophore processing takes place after export of this protein's apoform.

Notizen: Abstract The effect of the August 1987 River Reuss flood event on the phytoplankton biocoenosis in Lake Uri (Urnersee, part of Lake Lucerne, Switzerland) was investigated firstly by comparing biological, chemical and physical data sampled before the event with equivalent data sampled after the event; and secondly by comparing the phytoplankton succession in 1987 with that occurring in the “floodfree” year 1989. As a consequence of the flood, the physical and chemical environment of the phytoplankton was found to have undergone a change which resulted in an alteration in the composition of the phytoplankton community. The phytoplankton community existing previous to the flood event, which had been dominated byTabellaria fenestrata sensu Husted 1930 (K-strategist), was replaced by a biocoenosis characterized mainly by various species of flagellates, which represent a typical spring successional stage (r-strategists). After the externally-imposed perturbation, the return to stable physical and chemical conditions was followed by the re-establishment of the successional stage which had existed before the flood (termed “reversion” by Reynolds, 1980).

Notizen: Abstract We present a new simulation code able to simulate the entire propagation of laser pulse, from the amplifiers level up to the focusing stage. This algorithm has some new characteristics that we intend to present. It computes the three-dimensional optical propagation equation using no approximation other than its picosecond expression. The stability has been carefully studied so that it can be applied to any geometry. This is a great improvement since, up to now only cylindrical geometry was accessible for accuracy. In this paper we also present a method using Fast Fourier Transform able to evaluate with a high accuracy, Fresnel’s distribution of a focused laser pulse. The advantages provided by our algorithm are its rapidity and its high physical understanding of the focusing phenomena.

Notizen: Abstract We establish the stability of axial motions (steady motions along the lengthwise direction) of nonlinearly elastic loops of string. A key observation here is that a linear combination of the total energy and the total circulation of the string, both of which are conserved quantities, yields an appropriate Liapunov function. From our previous work [5], we know that there are uncountably many shapes corresponding to a given axial speed. Accordingly, we establish “orbitai” stability (modulo this collection of relative equilibria). For a well-defined class of “soft” materials, there is an upper bound on the axial speed sufficient for stability; “stiff” materials are shown to be orbitally stable at any axial speed.

Notizen: Abstract We have examined the properties and subcellular localization of phytohemagglutinin (PHA), the major lectin of the common bean (Phaseolus vulgaris.), in the axis cells of nearly mature and imbibed mature seeds. On a protein basis the axis contained about 15% as much PHA as the cotyledons. Localization of PHA was done with an indirect immunolabeling method (rabbit antibodies against PHA, followed by colloidal gold particles coated with goat antibodies against rabbit immunoglobulins) on ultra-thin cryosections which were embedded in plastic on the grids after the immunolabeling procedure. The embedding greatly improved the visualization of the subcellular structures. The small (4 nm) collodial gold particles, localized with the electron microscope, were found exclusively over small vacuoles or protein bodies in all the cell types examined (cortical parenchyma cells, vascular-bundle cells, epidermal cells). The matrix of these vacuoles-protein bodies appears considerably less dense than that of the protein bodies in the cotyledons, but the results confirm that in all parts of the embryo PHA is localized in similar structures.

Comparison of phytochrome photoversibility measurements to the distribution of the pigment within the cell indicates that the spectral assay is not influenced by the observed compartmentalization of the chromoprotein. However, the observed compartmentalization of phytochrome is correlated with a loss in spectrophotometrically detectable Pr.

Notizen: Abstract Sperm cells are released from pollen tubes of tobacco as linked cells, associated with the vegetative nucleus in an assemblage known as the male germ unit (MGU). Using light microscopy, the MGU assemblage appears to be ensheathed by cytoplasmic material of the pollen tube, which may stabilize their association. Following their release, the shape of the sperm cells and vegetative nucleus changes from an ellipsoidal to a more spheroidal morphology. When most of the cytoplasmic material is dispersed, a boundary remains around the two sperm cells. Using scanning electron microscopy, the cytoplasmic material surrounding the MGU appears filamentous, sometimes twisted and rope-like. Based on these observations, the function of the MGU of tobacco is discussed.

Notizen: The distribution of assembly workstations enables assembly operations to be done in parallel, while the multiple routing of parts in flexible assembly systems allows the opportunistic scheduling of assembly operations. This paper presents an assembly planning system, called the Assembly Coplanner, which automatically constructs an assembly partial order and generates a set of assembly instructions from a liaison graph representation of an assembly based on the extraction of preferred subassemblies. Assembly planning in Coplanner is carried out by the co-operation of multiple planning agents, such as the geometric reasoner, the physical reasoner, the resource manager and the plan coordinator, under the constraints of finding a cost-effective assembly plan in a flexible assembly system. The Coplanner identifies spatial parallelism in assembly as a means of constructing temporal parallelism among assembly operations. This is achieved in the following way: (1) the selection of a set of tentative subassemblies by decomposing a liaison graph into a set of subgraphs based on feasibility and difficulty of disassembly; (2) the evaluation of each of the tentative subassemblies in terms of assembly cost represented by subassembly selection indices; and (3) the construction of a hierarchical partial order graph (HPOG) as an assembly plan. A case study applying the Coplanner to a mechanical assembly is illustrated in this paper.

Notizen: Abstract The stability of time-varying autoregressive (AR) models is an important issue in such applications as time-varying spectrum estimation and electroencephalography simulation and estimation. In some cases, such as time-varying spectrum estimation, the models that exhibit roots near unit moduli are difficult to use. Thus a tighter stability condition such as stability with a positive margin is needed. A time-varying AR model is stable with a positive margin if the moduli of the roots of the time-varying characteristic polynomial are somewhat less than unity for every time instant. Recently, a new method for the estimation of the time-varying AR models was introduced. This method is based on the interpretation of the underdetermined time-varying prediction equations as an ill-posed inverse problem that is solved by Tikhonov regularization. The method is referred to as the deterministic regression smoothness priors (DRSP) scheme. In this paper, a stabilization method in which the DRSP scheme is augmented with nonlinear stability constrainst is proposed. The problem is formulated so that stability with a positive margin can also be achieved. The problem is solved iteratively with an exterior point algorithm. The performance of the algorithm is studied with a simulation. It is shown that the proposed approach is well suited to stable modeling of signals containing narrowband transitions.

Notizen: Abstract This work presents a specific stock-effort dynamical model. The stocks correspond to two populations of fish moving and growing between two fishery zones. They are harvested by two different fleets. The effort represents the number of fishing boats of the two fleets that operate in the two fishing zones. The bioeconomical model is a set of four ODE's governing the fishing efforts and the stocks in the two fishing areas. Furthermore, the migration of the fish between the two patches is assumed to be faster than the growth of the harvested stock. The displacement of the fleets is also faster than the variation in the number of fishing boats resulting from the investment of the fishing income. So, there are two time scales: a fast one corresponding to the migration between the two patches, and a slow time scale corresponding to growth. We use aggregation methods that allow us to reduce the dimension of the model and to obtain an aggregated model for the total fishing effort and fish stock of the two fishing zones. The mathematical analysis of the model is shown. Under some conditions, we obtain a stable equilibrium, which is a desired situation, as it leads to a sustainable harvesting equilibrium, keeping the stock at exploitable densities.

Notizen: Abstract To address the possibility that proliferative disorders may originate from interactions between multiple populations of proliferating and maturing cells, we formulate a model for this process as a set of coupled nonlinear first order partial differential equations. Using recent results for the asymptotic behaviour of the solutions to this model, we demonstrate that there exists a region of coupling coefficients, maturation rates, and proliferation rates that will guarantee the stable coexistence of coupled cellular populations. The analysis shows that increases in the coupling between populations may ultimately lead to a loss of stability. Furthermore, the analysis indicates that increases (decreases) in the maturation and/or proliferation rates above (below) critical levels will lead either to instability in the populations or the destruction of one population and the persistence of the other.

Notizen: Abstract A compartmental model is described for the spread of Gambian sleeping sickness in a spatially heterogeneous environment in which vector and human populations migrate between two "patches": the village and the plantations. The number of equilibrium points depends on two "summary parameters": gr the proportion removed among human infectives, and R0, the basic reproduction number. The origin is stable for R0 〈1 and unstable for R0 〉1. Control strategies are assessed by studying the mix of vector control between the two patches that bring R0 below 1. The results demonstrate the importance of vector control in the plantations. For example if 20 percent of flies are in the village and the blood meal rate in the village is 10 percent, then a 20 percent added vector mortality in the village must be combined with a 9 percent added mortality in the plantations in order to bring R0 below 1. The results are quite insentive to the blood meal rate in the village. Optimal strategies (that minimize the total number of flies trapped in both patches) are briefly discussed.

Notizen: Abstract LetK be ad-dimensional star body (with respect to the origino). It is known that the (d−1)-dimensional volume of the intersections ofK with the hyperplanes througho does not uniquely determineK. Uniqueness can only be achieved under additional assumptions, such as central symmetry. Here it is pointed out that if one uses, instead of intersections by hyperplanes, intersections by half-planes that containo on the boundary, then, without any additional assumptions, the volume of these intersections determinesK uniquely. This assertion, and more general results of this kind, together with stability estimates, are obtained from uniqueness results and estimates concerning a particular spherical integral transformation.

Notizen: Abstract An immunocytochemical method was used to determine the proportion of cells in the DNA synthesis (S phase) of the mitotic cycle in suspension cultures of soybean (Glycine max (L.) Merr. cv. Acme) callus of cotyledonary origin, the stably cytokinin-dependent tissue used in the cytokinin bioassay devised by Carlos O. Miller. A standard cell synchronization protocol involving hydroxyurea was used to demonstrate the applicability of the immunocytochemical method to this cell culture. Cells were brought to mitotic arrest by cytokinin withdrawal, and the cell division cycle was restarted by the addition of cytokinin. We have followed the pattern of resumption of S phase after the readdition of cytokinin. This pattern reveals the existence of three subpopulations of cells in cytokinin-starved cultures, consistent with the occurrence of three cytokinin-requiring events in the cell cycle: one in mitosis, one in S phase, and one in the G1 phase.

Notizen: Abstract The structure of solutions to the nonlinear parametric programming problem with a one dimensional parameter is analyzed in terms of the bifurcation behavior of the curves of critical points and the persistence of minima along these curves. Changes in the structure of the solution occur at singularities of a nonlinear system of equations motivated by the Fritz John first-order necessary conditions. It has been shown that these singularities may be completely partitioned into seven distinct classes based upon the violation of one or more of the following: a complementarity condition, a constraint qualification, and the nonsingularity of the Hessian of the Lagrangian on a tangent space. To apply classical bifurcation techniques to these singularities, a further subdivision of each case is necessary. The structure of curves of critical points near singularities of lowest (zero) codimension within each case is analyzed, as well as the persistence of minima along curves emanating from these singularities. Bifurcation behavior is also investigated or discussed for many of the subcases giving rise to a codimension one singularity.

Notizen: Abstract We prove that theε-optimal solutions of convex optimization problems are Lipschitz continuous with respect to data perturbations when these are measured in terms of the epi-distance. A similar property is obtained for the distance between the level sets of extended real valued functions. We also show that these properties imply that theε-subgradient mapping is Lipschitz continuous.

Notizen: Abstract Box schemes (finite volume methods) are widely used in fluiddynamics, especially for the solution of conservation laws. In this paper two box-schemes for elliptic equations are analysed with respect to quadrilateral meshes. Using a variational formulation, we gain stability theorems for two different box methods, namely the so-called diagonal boxes and the centre boxes. The analysis is based on an elementwise eigenvalue problem. Stability can only be guaranteed under additional assumptions on the geometry of the quadrilaterals. For the diagonal boxes unsuitable elements can lead to global instabilities. The centre boxes are more robust and differ not so much from the finite element approach. In the stable case, convergence results up to second order are proved with well-known techniques.

Notizen: Abstract If the sequence of functions ϕj, k is a wavelet frame (Riesz basis) or Gabor frame (Riesz basis), we obtain its perturbation system ψj,k which is still a frame (Riesz basis) under very mild conditions. For example, we do not need to know that the support of ϕ or ψ $$(\hat \phi or\hat \psi )$$ is compact as in [14]. We also discuss the stability of irregular sampling problems. In order to arrive at some of our results, we set up a general multivariate version of Littlewood-Paley type inequality which was originally considered by Lemarié and Meyer [17], then by Chui and Shi [9], and Long [16].

Notizen: Abstract The thermal oxidative stability of various formulations based on emulsion-grade ABS was studied by the chemiluminescence technique. Emulsion products were found to be essentially less stable than ionic mass polymerization resins. Among the antioxidants studied, Santonox R is clearly more efficient than Irganox 1076 and Irganox 3114, and its superiority is reflected primarily in improved induction period values. The introduction of Tinuvin 770 and Tinuvin 328 UV stabilizers into emulsion resins does not change the durability of the products. In mixtures where both Irganox 1076 and UV stabilizers are present, a certain antagonistic effect was noted at high UV stabilizer concentrations.

Notizen: Abstract Interaction of nicotine with tetracyanoethylene and iodine was investigated spectrophotometrically and found to form strong CT complexes (n−π and n-σ*, respectively). The donor site involved in CT interaction is the pyrrolidine nitrogen. The nicotine-I2 complex exists as the ionic structure (nicotine) I+-I 3 − . Formation constants of the CT complexes in various solvents were determined from 10 to 25°C and are discussed in terms of the nature of the electron acceptor and solvent polarity. Solid CT complexes were synthesized and were characterized by microchemical analysis and infrared spectra techniques.

Notizen: Abstract The influence of lubricants, UV stabilizers, antioxidants, and metal deactivators on the resistance of ABS to thermal oxidation was studied by means of the chemiluminescence technique. Neither of the additives seems to affect significantly the induction period of oxidation. At the same time, the influence of various additives on the oxidation rate constant is remarkably different: the introduction of lubricants and UV stabilizers increases its value, while antioxidants and metal deactivators have the opposite effect. For the particular systems studied durability is decreased in samples containing the lubricant and UV stabilizers and increased in samples stabilized with the antioxidant and metal deactivator.

Notizen: Abstract In an attempt to increase the range of analytical techniques able to monitor ultimate degradation stages of degradable, biodegradable, and bioresorbable polymers, capillary zone electrophoresis (CZE) was used to analyze tentatively oligomers formed during thermal condensation of lactic, glycolic, anddl-3-hydroxybutyric acids. The influence of the buffer and of capillary coating are discussed in terms of electroosmotic flow. Typical analyses were first performed using a 0.1M borate buffer (pH 8.9) with anodic injection. In the case of lactic acid, seven peaks were well separated, while only three peaks were observed for glycolic acid. A more complex situation was found fordl-3-hydroxybutyric acid oligomers. The first five peaks were split. The major component of each doublet was attributed to hydroxy-terminated oligomers, whereas the satellite peaks were assigned to oligomers bearing a C=C double bond at the noncarboxylic terminus. CZE of pH-sensitive lactic acid oligomers was also performed in 0.05M phosphate buffer (pH 6.8) with cathodic injection after physical coating of the fused-silica capillary with DEAE-Dextran. The buffer-soluble fraction present in lactic acid oligomers was extracted from a dichloromethane solution. Extracts issued from different batches of lactic acid condensates gave a constant water-solubility pattern whose cutoff was at the level of the decamer. CZE was also used to monitor thein vitro aging of aqueous solutions of these water-soluble oligomers. The lactyllactic acid dimer appeared more stable than higher oligomers, thus showing that ultimate stages of the degradation did not proceed at random. These physicochemical characteristics were used to complement the degradation pathway based on diffusion of oligomers duringin vitro aging of large size lactic acid plates made by compression molding. CZE data showed that lactic acid was the only component which was released in the aqueous medium during degradation.

Notizen: Abstract The thermal properties and devitrification behaviour of substituted InF3 glasses were studied by means of differential thermal analysis. A comparison of various simple quantitative methods to assess the level of stability of multicomponent fluoride glass systems was also made. Most of these methods are based on critical temperatures. In this paper, a new parameter,k d(T), is introduced to the stability criteria. The stabilities of several substituted InF3 glasses were evaluated experimentally and correlated with the activation energies of crystallization via this new kinetic criterion and compared with those evaluated by other criteria.

Notizen: Abstract In this work we report a thermodynamic characterization of stability and melting behaviour of two 24-mer DNA triplexes. The third strand, that binds the Watson-Crick double helix with Hoogsteen hydrogen bonds, contains 3′-3′ phosphodiester junction that determines the polarity inversion. The target double helix is composed of adjacent and alternate fragments of oligopurine-oligopyrimidine tracts. The two helices differ from the substitution of the cytosine, involved in the junction, with the thymine. Calorimetric data reported here provide a quantitative measure of the influence of pH and base modification on the stability of a DNA triplex.

Notizen: Abstract Well‐characterized catalyst model compounds of KAlO2 and KFeO2 are investigated by thermal desorption of potassium from the material. The desorbing fluxes of ions, atoms and highly excited states (field ionizable Rydberg states) were studied with surface and field ionization detectors in a vacuum apparatus. From the Arrhenius plots the activation energies for desorption of K and K+ were determined. The chemical state of potassium at the surfaces is concluded to be: ionic on KAlO2 (with the K desorption barrier of 1.76 eV) and covalent on KFeO2 (barrier of 2.73 eV). These results agree with the data obtained earlier for industrial catalysts for ammonia and styrene production. They are interpreted in terms of the Schottky cycle, which is completed for KAlO2 and fails for KFeO2. This failure indicates a non‐equilibrium desorption process. K Rydberg states are only found to desorb from KFeO2, in agreement with the suggestion that such states in some way are responsible for the catalytic activity.

Hyers, who gave the first significant partial solution in 1941. In particular, during the last two decades, the notion of stability of functional equations has evolved into an area of continuing research from both pure and applied viewpoints. Both classical results and current research are presented in a unified and self-contained fashion. In addition, related problems are investigated. Some of the applications deal with nonlinear equations in Banach spaces and complementarity theory.

Notizen: Abstract We consider a problem of the dynamics of prey-predator populations suggested by the content of a letter of the biologist Umberto D'Ancona to Vito Volterra. The main feature of the problem is the special type of competition between predators of the same species as well as of different species. Two classes of cases are investigated: a first class in which the behaviour of the predator is ‘blind’ and the second one in which the behaviour is ‘intelligent’. A qualitative analysis of the dynamical systems under consideration is followed by a numerical analysis of the most significant cases.

Notizen: Abstract It is known that it is not possible to introduce C0 -structural stability for whole systems in topological dynamics. Using the methods of Nonstandard Analysis, we suggest four different purely topological stability concepts for dynamical systems on compact subsets of Rn. Classically these amount to considering the space of all systems on a given subset of Rn as the fundamental entity when deforming a continuous system (instead of the space of all continuous systems as is normally done in topological dynamics). For two of the introduced stability concepts, we will show that all minimal flows are stable in this sense. Besides this, we will show that one of our stability concepts is related to what is called the pseudo orbit tracing property in a recently published book by Aoki and Hiraide and compare some of our results to the theory of dynamical systems as presented there.

Notizen: Abstract For orthogonal wavelets, the discrete wavelet and wave packet transforms and their inverses are orthogonal operators with perfect numerical stability. For biorthogonal wavelets, numerical instabilities can occur. We derive bounds for the 2-norm and average 2-norm of these transforms, including efficient numerical estimates if the numberL of decomposition levels is small, as well as growth estimates forL → ∞. These estimates allow easy determination of numerical stability directly from the wavelet coefficients. Examples show that many biorthogonal wavelets are in fact numerically well behaved.

Notizen: Abstract The problem of stability of the Lagrangian pointL 4 in the circular restricted problem of three bodies is investigated close to the 1 : 2 commensurability of the long and short period libration. By stability we define boundedness of the solution for a given initial finite displacement from the equilibrium point as function of the mass parameter μ close to the commensurability. A rigorous treatment close to the resonance condition is possible using a transformation that diagonalizes the matrix related to the linear part of the equations of motion. The so obtained equations are further transformed to action angle type variables. Then using an isolated resonance approach, only the slowly varying terms are kept in the equations and two independent isolating first integrals can be found. These integrals finally enable us to solve the stability problem in an exact way. The so obtained results are compared to numeric integration of the equations of motion and are found to be in perfect agreement.

Notizen: Abstract Numerical simulations are made within the framework of the plane restricted three-body problem, in order to find out if stable orbits for planets around one of the two components in double stars can exist. For any given set of initial parameters (the mass ratio of the two stars and the eccentricity of their orbit around each other), the phase-space of initial positions and velocities is systematically explored. In previous works, systematic exploration of the circular model as well as studies of more realistic (elliptic) cases such as Sun-Jupiter and the nearby α Centauri and Sirius systems, large stable planetary orbits were found to exist around both components of the binary, up to distances from each star of the order or more than half the binary's periastron separation. The first results presented here for the η Coronae Borealis system confirm the previous studies.

Notizen: Abstract The non-linear stability of the libration pointL 4 in the restricted problem has been studied when there are perturbations in the potentials between the bodies. It is seen that the pointL 4 is stable for all mass ratios in the range of linear stability except for three mass ratios depending upon the perturbing functions. The theory is applied to the following four cases: (i) There are no perturbations in the potentials (classical problem). (ii) Only the bigger primary is an oblate spheroid whose axis of symmetry is perpendicular to the plane of relative motion (circular) of the primaries. (iii) Both the primaries are oblate spheroids whose axes of symmetry are perpendicular to the plane of relative motion (circular) of the primaries. (iv) The primaries are spherical in shape and the bigger is a source of radiation.

Notizen: Abstract A new class of linear ordinary differential equations with periodic coefficients is found which can be transformed to the Gauss hypergeometric equation, and therefore the monodromy matrices are computable explicitly. These equations appear as the variational equations around a straight-line solution in Hamiltonian systems of the form H = T(p) + V(q), where T(p) and V(q) are homogeneous functions of p and q, respectively.

Notizen: Abstract The paper presents an approach to sequence planning consisting in determining assembly sequences defined in terms of mating and non-mating operations and based on a dynamic expansion of the assembly tree obtained using a knowledge base management system. The planner considers the case of a single-robot assembly workcell. The use of stability and the detailed definition of sequences also by means of several non-mating operations are shown to be powerful instruments in the control of the tree expansion. Forward assembly planning has been chosen, in order to minimize the number of stability checks. Backtracking is avoided by combining precedence relations and stability analysis. Hard and soft constrains are introduced to drive the tree expansion. Hard constraints are precedence relations and stability analysis. All operations are associated to costs, which are used as soft constraints. The operation based approach enables one to manage even non-mating operations and to easily overcome the linearity constraint. Costs enable the planner to manage the association among tools and components. The first section of the paper concerns Stability Analysis that is subdivided into Static and Dynamic Stability Analysis. The former is mainly involved in analyzing gravity effects; the latter is mainly involved in evaluate inertia effects due to manipulation. Stability Analysis is implemented in a simplified form. Fundamental assumptions are: no rotational equilibrium condition is considered; for each reaction force only direction and versus, but not magnitude, are considered; friction is neglected. The second section discusses the structure of the planner and its implementation. The planner is a rule based system. Forward chaining and hypothetical reasoning are the inference strategies used. The knowledge base and the data base of the system are presented and the advantages obtained using a rule based system are discussed. The third section shows two planning examples, showing the performance of the system in a simple case and in an industrial test case, the assembly of a microwave branching filter composed of 26 components.

Notizen: Abstract Legged-climbing robot is considered. Each foot of the robot has an electromagnet system for robot"s holding on a metal surface. This surface can be both vertical and inclined, including negative slope. Analytical calculation of robot stability under turn over or sliding conditions has been made. Critical slopes have been determined. One of these slopes corresponds to minimal reserve of robot stability towards sliding and another to minimal reserve of robot stability towards turning-over. As total reserve of stability of a robot is always equal to the minimal one of these reserves. Additional support elements of elastic material with high coefficient of friction, along with electromagnet, allows to increase minimal reserve of robot stability towards sliding. The use of such support elements leads to redistributing force of normal support reaction between electromagnet (which surface has low coefficient of friction) and additional support element (which surface has high coefficient of friction). It is just what leads to increasing the total friction force and as a consequence to increasing of minimal reserve of robot stability towards sliding.

Notizen: Abstract Sensitivity of the returns to scale (RTS) classifications in data envelopment analysis is studied by means of linear programming problems. The stability region for an observation preserving its current RTS classification (constant, increasing or decreasing returns to scale) can be easily investigated by the optimal values to a set of particular DEA-type formulations. Necessary and sufficient conditions are determined for preserving the RTS classifications when input or output data perturbations are non-proportional. It is shown that the sensitivity analysis method under proportional data perturbations can also be used to estimate the RTS classifications and discover the identical RTS regions yielded by the input-based and the output-based DEA methods. Thus, our approach provides information on both the RTS classifications and the stability of the classifications. This sensitivity analysis method can easily be applied via existing DEA codes.

Notizen: Abstract A robust regulator for flexible-joint robots is proposed, which yields constant torque disturbance rejection acting on the links. The design uses the integrator backstepping technique [4,5] to cancel nonlinearities and disturbance not in the range space of the control. Stability of the closed loop system is shown using iterative Liapunov functions.

Notizen: Abstract A genetic manufacturing environment is considered. The emphasis is on small-lot, discrete, and asynchronous type of manufacturing systems rather than high volume and continuous type. Two classes of scheduling policies are proposed to render the machine stable. The policies are of feedback type. The decision is made in real-time and on-line.
