Notizen: Abstract Although thermal influences on the physiology of rainbow trout (Oncorhynchus mykiss) have been widely studied, there is little information about the responses of different genetic strains to temperatures. The effects of water temperature (10, 14, 19, 22, and 25 °C) on food consumption rate, growth rate, gross conversion efficiency, resting routine oxygen consumption rate, upper critical thermal tolerance and critical swimming velocity were investigated in juvenile rainbow trout of the Eagle Lake (O. m. aquilarum) subspecies and the Mt. Shasta strain. No strain-related differences in conversion efficiency, oxygen consumption rates, thermal tolerance or swimming performance were observed in 1995 (19, 22, and 25 °C) or 1996 (10, 14, and 19 °C), but Mt. Shasta strain trout grew faster at highest temperatures than did Eagle Lake trout. Food consumption rates, growth rates, conversion efficiency, and oxygen consumption rates declined at the extreme temperatures (10 and 25 °C) in both Eagle Lake and Mt. Shasta trout. Swimming performance was temperature-independent between 10 and 19 °C (overall mean: 5.43 body lengths s−1).

Notizen: Abstract The structural stability of fish myosin depends upon species and temperatures of water in which fish live. Primary, secondary, and quaternary structures of myosin heavy chain (MyHC) from three species of fish living at different temperature ranges have been compared with those of rabbit MyHC in order to investigate the differences in stability. Primary structure of MyHC, although being accessible for warm-water and cold-water fish (carp and walleye pollack), was not available in previous for tropical-water fish literature; so in this study primary structure of MyHC of the tropical-water fish amberjack has been determined by cloning and sequencing its cDNA. The MyHC has 1938 amino acid residues (AA), which are almost as much as as those of carp and walleye pollack. The amberjack MyHC is 91–95% homologous with other fish and rabbit MyHCs. There is a discernible difference between animal species with stable myosin rod (amberjack, carp, and rabbit) and walleye pollack with unstable rod. Stable rod species have a high probability of forming coiled-coil around the COOH-terminal end of the rod, while the pollack has a low coiled-coil formation probability. In addition, the average scores of the coiled-coil for myosin rod were rabbit (1.738) 〉 amberjack (1.691) 〉 carp (1.680) 〉 walleye pollack (1.674) which correlated exactly with the observed stability. The results suggest that coiled-coil forming ability, particularly around the COOH-terminal end, directs structural stability of fish myosin rod.

Notizen: Abstract The mitochondrial ATP synthase is a molecular motor that drives the phosphorylation ofADP to ATP. The yeast mitochondrial ATP synthase is composed of at least 19 differentpeptides, which comprise the F1 catalytic domain, the F0 proton pore, and two stalks, oneof which is thought to act as a stator to link and hold F1 to F0, and the other as a rotor.Genetic studies using yeast Saccharomyces cerevisiae have suggested the hypothesis thatthe yeast mitochondrial ATP synthase can be assembled in the absence of 1, and even 2, ofthe polypeptides that are thought to comprise the rotor. However, the enzyme complexassembled in the absence of the rotor is thought to be uncoupled, allowing protons to freelyflow through F0 into the mitochondrial matrix. Left uncontrolled, this is a lethal process andthe cell must eliminate this leak if it is to survive. In yeast, the cell is thought to lose ordelete its mitochondrial DNA (the petite mutation) thereby eliminating the genes encodingessential components of F0. Recent biochemical studies in yeast, and prior studies in E. coli, have provided support for the assembly of a partial ATP synthase in which the ATP synthaseis no longer coupled to proton translocation.

Notizen: Abstract Evidence from a broad panel of countries shows little overall relation between income inequality and rates of growth and investment. For growth, higher inequality tends to retard growth in poor countries and encourage growth in richer places. The Kuznets curve—whereby inequality first increases and later decreases during the process of economic development—emerges as a clear empirical regularity. However, this relation does not explain the bulk of variations in inequality across countries or over time.

Notizen: Abstract This article analyzes the impactof inequality on growth when consumers have hierarchic preferencesand technical progress is driven by innovations. With hierarchicpreferences, the poor consume predominantly basic goods, whereasthe rich consume also luxury goods. Inequality has an impacton growth because it affects the level and the dynamics of aninnovator's demand. It is shown that redistribution from veryrich to very poor consumers can be beneficial for growth. Ingeneral, the growth effect depends on the nature of redistribution.Due to a demand externality from R&D activities, multipleequilibria are possible.

Notizen: Abstract Thisarticle decomposes the well-documented relationship between financialdevelopment and growth. We examine whether financial developmentaffects growth solely through its contribution to growth in ``primitives'' or factor accumulation rates or whether it alsohas a positive impact on total factor productivity growth. Ourresults suggest that indicators of financial development arecorrelated with both total factor productivity growth and investment. However, the indicators that are correlated with total factorproductivity growth differ from those that encourage investment. In addition, many of the results are sensitive to the inclusionof country fixed effects, which may indicate that the financialdevelopment indicators are proxying for broader country characteristics.

Notizen: Abstract The cost of public investment is not the increment to the value ofpublic capital. Unlike with private investors, there is no plausiblebehavioral model in which every dollar that the public sectorspends as ``investment'' creates economically valuable ``capital.''While this simple analytic point is obvious, it has so far beenuniformly ignored in the empirical literature on economic growth, which uses—at best—cumulated, depreciated, investmenteffort (CUDIE) as a proxy for capital stocks. However, particularlyfor developing countries the difference between investment costand capital value is of first-order empirical importance: governmentinvestment is half of more of total investment, and calculationspresented here suggest that in many countries government investmentspending has created little useful capital. This has implicationsin three broad areas. First, none of the existing empirical estimatesof the impact of public spending has identified the productivityof public capital. Even where public capital has a potentiallylarge contribution to production, public-investment spendingmay have had a low impact. Second, it implies that all estimatesof total factor productivity in developing countries are deeplysuspect as there is no way to empirically distinguish betweenlow growth because of investments that create no factors andlow growth due to slow productivity growth. Third, multivariateregressions to date have not adequately controlled for capitalstock growth, which leads to erroneous interpretations of regressioncoefficients.

Notizen: Abstract This articlepresents a model of knowledge, seen as a set of ideas definedin a multidimensional idea space. Knowledge is created throughconvex combinations of older ideas and through paradigm shifts. When normal science has made the knowledge set convex, scientificopportunity is exhausted. Individual countries are endowed withdifferent knowledge sets, which gives rise to idea gaps. Thegrowth of a country's knowledge depends on diffusion from othercountries, on own production, and on the state of its human capitaland institutions. In the long run, economic growth will dependon knowledge growth, but only paradigm shifts can save R&Dfrom diminishing returns

Notizen: Abstract This paper relates recent empiricalresearch on the growth of U.K. companies to the maineconomic theories of firms' growth and to empiricalresults for the U.S.A. Smaller and younger firms havebeen growing more quickly than larger and older firms, thus generating proportionately more new jobs. Theseresults do not support the various theories of staticand dynamic economies of scale. Serial correlation ofgrowth is very low, so success does not persist. Thesystematic tendency for small and younger firms togrow more quickly is the main reason why firm growthis not entirely stochastic.

Notizen: Seagrass beds in South-east Asia sometimes consist of a mosaic of different species in monospecific patches. We examined whether the magnitude of within-patch variation in the seagrass Halophila ovalis is affected by the presence or absence of surrounding vegetation consisting of another seagrass species Thalassia hemprichii in an intertidal flat in Thailand waters. We measured biomass and growth rates of H. ovalis at the edges and centers of two different types of patches: (i) H. ovalis patches adjoining T. hemprichii vegetation (HT patches), and (ii) H. ovalis patches adjoining unvegetated sand flats (HS patches). Furthermore, we examined the possible effects of interspecific interactions on the growth of H. ovalis by experimentally removing adjoining T. hemprichii at the edges of HT patches. The biomass of H. ovalis was greater at the patch centre than the patch edge in both types of patches. For the growth rate of H. ovalis, significant interactions were detected between patch types and positions in patches. The difference in growth was significant and more than 4-fold between edges and centers of the HS patches, whereas the growth was not significantly different between edges and centers of the HT patches. The removal of T. hemprichii did not significantly affect the growth rate of H. ovalis at the edge of the HT patches. These findings demonstrate that the magnitude of within-patch variation in H. ovalis growth is affected by the conditions of adjoining habitats. However, any effects of local competition with T. hemprichii on H. ovalis growth were not evident in this short-term manipulative experiment.

Notizen: Abstract Five anaerobic proteolytic bacteria were isolated from water bodies of Leh, India, where the ambient temperature varies from −25 to 25 °C. Isolates showed growth at all temperatures ranging from 5 to 37 °C except SPL-4 and SPL-5 which showed no growth at 5 °C. The cultures could grow and produce proteases on various protein substrates and the yield varied with the substrates. Two of the cultures showed the presence of spores. Acetate was the dominant VFA during hydrolysis of protein substrates.

Notizen: Abstract The Environmental Kuznets Curve (EKC) is a hypothesis which implies that it is possible to “grow out of environmental degradation”. Most theoretical models of the EKC relation have not accounted for transboundary and intergenerational externalities nor have empirical studies provided evidence that validates an inverted U shaped relation between environmental degradation and economic growth for pollution problems where the effects are far-displaced or are long-delayed. This paper integrates the theory of transboundary externalities into the most common theoretical framework applied to the EKC hypothesis. It shows that where a significant proportion of the environmental impacts of economic activity occurs outside the territories in which those activities take place, the de-linking of growth and environmental degradation is less likely to happen. This proposition is demonstrated by assuming that decisionmakers have a Nash-type non cooperative strategic behavior.

Notizen: Abstract We present the first evidence for a fitness cost of an inducible response that is detectable in a nitrogen (N) currency. Nicotine is an induced defense in Nicotiana sylvestris that can utilize 5–8% of the plant's total N, an investment that apparently cannot be recouped by metabolism. Induced nicotine production is endogenously regulated by jasmonic acid (JA), and we treated leaves with the methyl ester of this wound hormone (MeJA) in quantities (0, 25, 250 μg) known to elicit changes in endogenous JA and subsequent nicotine responses comparable to those elicited by mechanical wounding and herbivory in this species. We grew plants in competition chambers (CCs) in which three same-sized plants could compete for a communal but fixed pool of 15NO3 to quantify the outcome of competition for this fitness-limiting resource that is used both in defense and seed production. Competition profoundly increased all measures of growth and reproductive performance measured per milligram of N acquired. While plants acquired all the N supplied to them in the hydroponic solution, plants grown in CCs (as compared those grown in individual chambers—ICs) retained more of this N and produced more biomass, had larger nicotine contents, allocated less of their N to nicotine, produced larger floral stalks with more flowers, aborted fewer flowers, matured more capsules, and produced a greater mass of seed. Plants grown in ICs produced heavier seed, but this difference did not translate into a difference in seed viability. MeJA treatment increased nicotine concentrations in proportion to the amount applied and significantly reduced growth (13–23%) and reproductive (31–44%) performance for plants grown with uninduced competitors, reflecting a large opportunity cost of induction. The effects of MeJA treatment on growth and reproduction were significantly less pronounced for plants grown in ICs. MeJA treatment significantly reduced the ability of plants to compete for [15N]KNO3 (reducing uptake by 9.5% and 23.7% for 25- and 250-μg MeJA-treated plants, respectively); no reductions in N acquisition were found in IC grown plants treated with MeJA. This impairment of competitive ability could account for 41–47% of the jasmonate-induced reductions in biomass by the day 15 harvest and 12–20% of the reductions in seed set and, in addition, created by "opportunity benefit" for neighboring uninduced plants, which grew larger, aborted fewer flowers, and matured more seed (a 28% increase) than did uninduced plants competing with similarly uninduced plants. Competition dramatically increased plant growth and reproductive performance, and MeJA treatment of these high-performing plants significantly reduced their competitive ability, which translated into opportunity costs for induced plants and opportunity benefits for neighboring uninduced plants. Induced plants minimized these fitness costs by reducing their use of recently acquired N for nicotine biosynthesis when growing with competitors. MeJA treatments also altered stalk length, flower production, flower abortion, and allocation to seed mass. In spite of all this plasticity, induced responses incur large fitness costs, costs that could be in part attributed to reductions in competitive ability for N. We conclude that inducibility functions to minimize these costs.

Notizen: Abstract Because many optimal control problems require solution using iterative procedures they fall naturally in the realm of 2-D systems where the two dimensions are response time horizon and iteration index, respectively. The paper uses this observation to employ 2-D systems theory, in the form of unit memory repetitive process techniques, to analyse local stability and convergence behaviour of a continuous optimal control algorithm based on dynamic system optimisation and parameter estimation. Existing work is extended to incorporate unmatched terminal constraints. Necessary and sufficient conditions for stability are obtained whose evaluation require the solution of a difficult eigenvalue problem. The paper shows how solutions can be achieved using numerical and graphical facilities of MATLAB.

Notizen: Abstract In this paper, we discussthe jump behavior and stability problems for 2-D linear shift-invariantsingular systems under the standard boundary conditions. It isshown that once a boundary condition or the input is inadmissiblein the classical sense, a group of non-causal or backward jumpsof the system states will be incited. This interpretation releasesthe conventional admissibility constraints on the boundary conditionsand inputs. Based on this observation, a systematic stabilitytheory is developed for 2-D singular systems. The well-knownbasic stability theorem for the 1-D singular systems or 2-D regularsystems is thus extended to the 2-D singular case.

The lethal effect of the K2 mycoviral toxin was confirmed by methylene blue staining. Scanning electron microscopy of cells from challenge experiments revealed rippled cell surfaces, accompanied by cracks and pores, while those unaffected by the toxin, as in the control experiments, showed a smooth surface. Transmission electron microscopy revealed that the toxin damaged the cell wall structure and perturbed cytoplasmic membranes to a limited extent.

Notizen: Abstract In Saccharomyces cerevisiae pseudohyphae formation may be triggered by nitrogen deprivation and is stimulated by cAMP. It was observed that even in a medium with an adequate nitrogen supply, cAMP can induce pseudohyphal growth when S. cerevisiae uses ethanol as carbon source. This led us to investigate the effects of the carbon source and of a variety of stresses on yeast morphology. Pseudohyphae formation and invasive growth were observed in a rich medium (YP) with poor carbon sources such as lactate or ethanol. External cAMP was required for the morphogenetic transition in one genetic background, but was dispensable in strain Σ1278b which has been shown to have an overactive Ras2/cAMP pathway. Pseudohyphal growth and invasiveness also took place in YPD plates when the yeast was subjected to different stresses: a mild heat-stress (37 °C), an osmotic stress (1 m NACl), or addition of compounds which affect the lipid bilayer organization of the cell membrane (aliphatic alcohols at 2%) or alter the glucan structure of the cell wall (Congo red). We conclude that pseudohyphal growth is a physiological response not only to starvation but also to a stressful environment; it appears to require the coordinate action of a MAP kinase cascade and a cAMP-dependent pathway.

Notizen: Abstract Wild-caught specimens of the lacertid lizard Gallotia galloti eisentrauti from the Canary Island of Tenerife were checked for ectoparasites. The parasitic gamasid mite Ophionyssus galloticolus Fain and Bannert (2000) was very abundant on these lizards. Additionally, parasitism by larvae of two species of Trombiculidae (Prostigmata: Parasitengona) was observed. O. galloticolus was reared in the laboratory on its natural host in order to investigate its life cycle, reproductive biology, and development. The life history of O. galloticolus is documented in detail and compared to literature data of other Ophionyssus species. O. galloticolus was found to be similar to other species of the same genus with respect to the duration of development, the precopulatory association of protonymphs, and the arrhenotokous development of eggs. However, it seems to be more tolerant towards low relative humidity and longer starvation periods than other Ophionyssus species. Evolutionary transformations of the life-history pattern of this genus and other parasitic mites in comparison to its predatory precursors involve a reduction or partial suppression of ontogenetic instars in order to decrease mortality during host-seeking phases, and a compensating increase in growth capacity of the remaining feeding instars facilitated by replacement of sclerites through elastic cuticle or by growth of new cuticle unrelated to a moult (neosomy).

Notizen: Abstract The accumulation of compatible solutes is one of the strategies that plants have developed to tolerate salt stress. Proline and betaine are the main metabolites that accumulate in various species of higher plants in response to salt stress. In Helianthus annuus L., pre-treatment of seeds with ethanolamine led to enhanced seedling tolerance to conditions of saline stress during germination, as evidenced by the greater growth of pretreated seedlings (EAS group) versus untreated seedlings (S group), evaluated through such parameters as length, water and chlorophyll content. During the germination period, a considerable increase was observed in proline levels (up to 300%) in seedlings subjected to saline stress, whereas in the EAS group, the proline increment was much smaller (20%). Starting from the fourth day of germination, betaine levels in seedlings pretreated with ethanolamine and then with water (EAW group) and in EAS showed a significant increase versus C (control) and S seedlings, possibly because such a precursor promotes betaine biosynthesis. This could be responsible for the enhanced growth observed in EAS versus S seedlings, as well as for preventing the decrease in chlorophyll content in the EAS group. The accumulation of betaine seems to correlate with the greater tolerance of these seedlings against stress induced by sodium chloride.

Notizen: Abstract The response, in terms of capsaicinoid content, of chile (Capsicum annuum L.) genotypes to different environments was studied. Double haploidlines, an F1 hybrid, and an open-pollinated cultivar estimated the genotype, environment, and genotype-by-environment interaction effect on the total capsaicinoids and on individual capsaicinoids. Significant differences were observed among the genotypes and among genotype-by-environment interactions over the environments. Among the genotypes in an environment, the within-genotype variances were also significantly different. The double haploid line, HDA 207, had low within-genotype variance for individual and total capsaicinoids, with the exception of the isomer of dihydrocapsaicin. Also for HDA 270, the genotype-by-environment interaction was negligible for individual and total capsaicinoids, Indicating stability across environments.

Notizen: Abstract The effects of salinity on leaf growth, initiation and senescence, on transpiration rates, on leaf water potential and on uptake and distribution of several ions were studied in two sugarcane cultivars differing in salinity sensitivity. Plants, growing in a growing mixture in pots, were exposed to salinized irrigation water for 68 days, starting 60 days after planting. EC values of the irrigation water were 1.0, 2.0, 4.0, 8.0 and 12 dS/m, obtained by using a mixture of NaCl and CaCl2. Plants were also grown in nutrient solution and were at a similar age when exposed to a salinity level of 3 dS/m for 30 days followed by 6.0 dS/m for an additional 30 days. Two Na:Ca ratios of 18:1 and 1:2 were used for salinization of the nutrient solution. Both leaf dry weight and area decreased with increasing salinity, but in the more salinity tolerant cultivar H69-8235, the decrease was moderate. Salinity hardly reduced average area per leaf in H69-8235, while the number of leaves declined sharply. This decline was caused by enhanced senescence of mature leaves and not by a decreased rate of leaf initiation. In the more sensitive cultivar, H65-7052, leaf area and initiation of new leaves were sharply reduced by salinity while leaf senescence was less affected. Leaf water potential decreased during the early stages of salinity exposure, and the reduction in water potential was larger in H69-8235. Salinity also decreased the rate of transpiration rate but to a lesser extent than leaf development and growth. The accumulation of Cl and Na in the TVD (top visible dewlap) leaf of the tolerant cultivar H69-8235 was greater than in the sensitive cultivar H65-7052. The concentration of Cl in the TVD leaf was more than 10 times that of Na in both cultivars. The concentration of both ions, but not of K, increased during the early stages of salinity exposure and then remained constant. A gradient in concentration of Cl and Na over the plant was found in both cultivars at all salinity levels, and was steepest between the TVD and younger leaves. No specific Na effect on leaf growth or transpiration could be detected. The accumulation of Cl and Na but not of K occurred primarily in the roots rather than in the leaves and stalks.

Notizen: Abstract Using in vitro culture of isolated small berries of Vitis vinifera L. cv. Sultana, it was possible to study the effect of different carbon sources and sucrose concentration on fruit growth, hexose accumulation and soluble invertase activity during the first stage of berry development by eliminating the source tissue. Berries cultured in vitro lack stage III of berry development which is characterised by massive accumulation of water and sugars, and thereby berries reached only 30% of the weight of those grown in the plant. Sucrose and glucose were both good carbon sources for berry growth, while fructose was not as good. Berry growth, hexose accumulation and invertase activity increased as sucrose concentration increased up to 15% in the medium. Furthermore, the onset of hexose accumulation in cultured berries depended on the concentration of sucrose in the medium, starting earlier at higher concentrations.

Notizen: Abstract A full-length (LeHT2) and two partial (LeHT1 and LeHT3) cDNA clones, encoding hexose transporters, were isolated from tomato (Lycopersicon esculentum) fruit and flower cDNA libraries. Southern blot analysis confirmed the presence of a gene family of hexose transporters in tomato consisting of at least three members. The full-length cDNA (LeHT2) encodes a protein of 523 amino acids, with a calculated molecular mass of 57.6 kDa. The predicted protein has 12 putative membrane-spanning domains and belongs to the Major Facilitator Superfamily of membrane carriers. The three clones encode polypeptides that are homologous to other plant monosaccharide transporters and contain conserved amino acid motifs characteristic of this superfamily. Expression of the three genes in different organs of tomato was investigated by quantitative PCR. LeHT1 and LeHT3 are expressed predominantly in sink tissues, with both genes showing highest expression in young fruit and root tips. LeHT2 is expressed at relatively high levels in source leaves and certain sink tissues such as flowers. LeHT2 was functionally expressed in a hexose transport-deficient mutant (RE700A) of Saccharomyces cerevisiae. LeHT2-dependent transport of glucose in RE700A exhibited properties consistent with the operation of an energy-coupled transporter and probably a H+/hexose symporter. The K m of the symporter for glucose is 45 μM.

Notizen: Abstract The Svalbard Islands are influenced by warm Atlantic water in the south and west, and cold Arctic water in the east. Ice cover, and hence the location of the highly productive marginal ice zone, varies both intra and interannually. Part of the primary production accumulates on the bottom and is utilized by the benthos. In this study, the annual growth of the cockle Clinocardium ciliatum (Fabricius, 1780) from three sites in Svalbard waters is reported. Moffen, the site in the north (80° 01′ N, 13° 48′ E) is located in the northernmost areas influenced by Atlantic water. The Storfjorden site (77° 10′ N, 20° 09′ E) is situated in cold Arctic water masses, and the Bear Island site (74° 50′ N, 18° 54′ E) is in the Polar front area where Atlantic and Arctic water masses meet. Annual growth of cockles was analysed retrospectively by measuring external growth increments, which gave annual growth records from the 1970s to 1996. Shell height for age for different year classes was highest at the Storfjorden site, and lowest at Bear Island. Periods of high growth occurred at Storfjorden and Bear Island during the 1980s while the beginning of 1990s was characterized by low growth. At Moffen, growth was more variable between single years. Several factors are influencing the growth of C. ciliatum in the Svalbard area and growth cannot be coupled to only one environmental factor like ice cover.

Notizen: Abstract The long-term stability of high-level expression is the mostimportant factor to consider when choosing cell lines for the expression of recombinant proteins. Declining volumetricyields in large-scale fermentation can be caused by changes affecting the cell population as a whole such as loss in viability, depletion of nutrients or accumulation of metabolites affecting cell growth. Alternatively, geneticinstability may lead to the outgrowth of a less productive, metabolically favored sub-population. Currently a variety ofparameters are measured to monitor the condition of cells infermenters including glucose uptake, lactate accumulation andoxygen consumption; in addition, periodic viable cell countsallow the determination of the growth rate and viability of the population. All of these methods measure the condition ofthe cell population as a whole and changes must involve a significantly large proportion of the total culture in orderto be detectable. Here we report on a method that allows theevaluation of the productivity of individual cells. Using the gel microdrop secretion assay, we detected the appearance ofa sub-population of cells with lower productivity. Subsequentanalysis of the culture confirmed the existence of lower productivity cells with a lower vector copy number. Therefore, the single cell secretion assay proved to be a rapid method todetect and isolate a low productivity variant of the producer cell line.

Notizen: Abstract The dynamical stability of a homogeneous, simple supported column, subjected to a periodic axial force, is investigated. The viscoelastic material is assumed to obey the Leaderman nonlinear constitutive relation. The equation of motion was derived as a nonlinear integro-partial-differential equation, and was simplified into a nonlinear integro-differential equation by the Galerkin method. The averaging method was employed to carry out the stability analysis. Numerical results are presented to compare with the analytical ones. Numerical results also indicate that chaotic motion appears.

Notizen: Abstract The governing equation of solid-liquid couple vibration of pipe conveying fluid on the elastic foundation was derived. The critical velocity and complex frequency of pipe conveying fluid on Winkler elastic foundation and two-parameter foundation were calculated by power series method. Compared with pipe without considering elastic foundation, the numerical results show that elastic foundation can increase the critical flow velocity of static instability and dynamic instability of pipe. And the increase of foundation parameters may increase the critical flow velocity of static instability and dynamic instability of pipe, thereby delays the occurrence of divergence and flutter instability of pipe. For higher mass ratio β, in the combination of certain foundation parameters, pipe behaves the phenomenon of restabilization and redivergence after the occurrence of static instability, and then coupled-mode flutter takes place.

Notizen: Abstract The spatial stability equation of moving jet containing dense suspended solid particles was derived out by means of the continuum phase-coupled model. The stability curves of moving jet for different downstream distances, Reynolds number of flow-field, particle properties and velocities of jetting device are got by the finite difference method based on the asymptotic method and the Eulerian conservative difference scheme. Founded on the analysis of the obtained stability curves it is found that the positive velocity of jetting device widens the unstable frequency range of flow-field but the effect of the negative one is contrary. In addition, particles existing in the flow-field curb the instability of flow-field and the effect enhances with the decrease of Reynolds number of flow-field. These conclusions benefit learning the development of moving two-phase jet.

Notizen: Abstract By adopting the energy method, a new method to calculate the stability of the composite shell of revolution is presented. This method takes the influence of nonlinear prebuckling deformations and stresses on the buckling of the shell into account. The relationships between the prebuckling deformations and strains are calculated by nonlinear Kármán equations. The numerical method is used to calculate the energy of the total system. The nonlinear equations are solved by combining gradient method and amendatory Newton iterative method. The computer program is also developed. An example is given to demonstrate the accuracy of the method presented.

Notizen: Abstract The recursive least square is widely used in parameter identification. But it is easy to bring about the phenomena of parameters burst-off. A convergence analysis of a more stable identification algorithm-recursive damped least square is proposed. This is done by normalizing the measurement vector entering into the identification algorithm. It is shown that the parametric distance converges to a zero mean random variable. It is also shown that under persistent excitation condition, the condition number of the adaptation gain matrix is bounded, and the variance of the parametric distance is bounded.

Notizen: Abstract In the mitochondrion, essential genetic elements for replication and transcription are mostly housed within a shortsegment of its DNA located between tRNAPhe and tRNAPro genes, which is called mitochondrial regulatoryregion (mrr). RNAs are known to be transcribed from mrr, thestructures and the functions of which are yet to be fullycharacterized. We detected ca. 1.3 kb H-strand transcripts of mrr (mrrH-RNAs),and 0.2 kb L-strand transcripts of mrr (mrrL-RNAs) in varioushuman cultured cells and tissues using double stranded mrrDNAprobes. The steady state levels of mrrL-RNAs were generally highin cultured cells, while they varied among tissues. On the otherhand, the levels of mrrH-RNAs varied among tissues and amongcultured cells. A tendency was observed in these cells andtissues that a high level of mrrL-RNA is associated with cellproliferation, and a high level of mrrH-RNA withdifferentiation. Several cDNA clones to 1.3 kb mrrH-RNA were obtained from humanskeletal muscle polyadenylated RNAs. The 5′ terminus of the 1.3 kb RNA was determined to be at nucleotide position 15953 whichis immediately downstream of tRNAThr sequence.Polyadenylation site for most of the clones was demonstrated tobe at nucleotide position 576 which is immediately upstream oftRNAPhe sequence. The longest cDNA insert obtained was 1177 bps long spanning from nucleotide positions 15969 to 576 which could code for a peptide of 76 amino acids. The cDNAs isolatedhere are the first cDNA clones reported to human mrrH-RNAs.These results, together with previous results, furthersubstantiate that polyadenylated mrrH- and mrrL-RNAs are commonly present at varying levels among human tissues andcells. The 3′ end sequences of the cloned mrrH-cDNA provideswith insights into the mechanisms of transcription termination. The cDNA clones will provide tools to further the study of thefunction of mrr RNAs.

Notizen: Abstract Selective catalytic reduction of NOx by propene has been investigated on Pt-USY and compared to other Pt-catalysts. The catalyst was characterized by XRD, Ar adsorption at 87 K, TEM, and CO chemisorption, and tested in a gas mixture system in excess oxygen. Pt-USY shows an excellent activity in the reaction, with a molar NOx conversion of 90% at 475 K. Stability during time-on-stream and resistance to SO2 and H2O in the feed stream has also been investigated. Pt-USY performs better under lean-burn conditions than other Pt-catalysts on ZSM-5, Al2O3, or SiO2. The selectivity to N2 was similar for all the catalysts (∼30%), the other major product being N2O.

Notizen: Abstract While dopamine is likely to modulate hippocampal synaptic plasticity, there has been little information about how dopamine affects synaptic transmission in the hippocampus. The expression of IEGs including c-fos has been associated with late phase LTP in the CA1 region of the hippocampus. The induction of c-fos by dopaminergic receptor activation in the rat hippocampus was investigated by using semiquantitative RT-PCR and immuno-cytochemistry. The hippocampal slices which were not treated with dopamine showed little expression of c-fos mRNA. However, the induction of c-fos mRNA was detected as early as 5 min after dopamine treatment, peaked at 60 min, and remained elevated 5 h after treatment. Temporal profiles of increases in c-fos mRNA by R(+)-SKF-38393 (50 μM) and forskolin (50 μM) were similar to that of dopamine. An increase in [cAMP] was observed in dopamine-, SKF-, or forskolin-treated hippocampal slices. By immunocytochemical studies, control hippocampal cells showed little expression of c-Fos immunoreactivity. However, when cells were treated with dopamine, an increase in the expression of c-Fos immunoreactivity was observed after treatment for 2 h. The treatment of hippocampal neurons with R(+)-SKF38393 (50 μM) or forskolin (50 μM) also induced a significant increase in c-Fos expression. These results indicate that the dopamine D1 receptor-mediated cAMP dependant pathway is associated with the expression of c-Fos in the hippocampal neurons. These data are consistent with the possible role of endogenous dopamine on synaptic plasticity via the regulation of gene expression. Furthermore, these results imply that dopamine might control the process of memory storage in the hippocampus through gene expression.

Notizen: Abstract Purpose. Paclitaxel is currently administered i.v. as a slow infusion of asolution of the drug in an ethanol:surfactant:saline admixture. However, poor solubilization and toxicity are associated with this drug therapy.Alternative drug delivery systems, including parenteral emulsions, areunder development in recent years to reduce drug toxicity, improveefficacy and eliminate premedication. Methods. Paclitaxel emulsions were prepared by high-shearhomogenization. The particle size of the emulsions was measured by dynamiclight scattering. Drug concentration was quantified by HPLC and invitro drug release was monitored by membrane dialysis. The physicalstability of emulsions was monitored by particle size changes in boththe mean droplet diameter and 99% cumulative distribution. Paclitaxelpotency and changes in the concentration of known degradants wereused as chemical stability indicators. Single dose acute toxicity studieswere conducted in healthy mice and efficacy studies in B16 melanomatumor-bearing mice. Results. QW8184, a physically and chemically stable sub-micronoil-in-water (o/w) emulsion of paclitaxel, can be prepared at high drugloading (8-10 mg/mL) having a mean droplet diameter of 〈100 nmand 99% cumulative particle size distribution of 〈200 nm. In vitro release studies demonstrated low and sustained drug release both inthe presence and absence of human serum albumin. Based on singledose acute toxicity studies, QW8184 is well tolerated both in miceand rats with about a 3-fold increase in the maximum-tolerated-dose(MTD) over the current marketed drug formulation. Using the B16mouse melanoma model, a significant improvement in drug efficacywas observed with QW8184 over Taxol®. Conclusions. QW8184, a stable sub-micron o/w emulsion of paclitaxelhas been developed that can be filter-sterilized and administered i.v.as a bolus dose. When compared to Taxol®, this emulsion exhibitedreduced toxicity and improved efficacy most likely due to thecomposition and dependent physicochemical characteristics of the emulsion.

Notizen: Abstract Purpose. To investigate the effect of atomization conditions on particle size and stability of spray-freeze dried protein. Methods. Atomization variables were explored for excipient-free (no zinc added) and zinc-complexed bovine serum albumin (BSA). Particle size was measured by laser diffraction light scattering following sonication in organic solvent containing poly(lactide-co-glycolide) (PLG). Powder surface area was determined from the N2 vapor sorption isotherm. Size-exclusion chromatography (SEC) was used to assess decrease in percent protein monomer. Fourier-transform infrared (FTIR) spectroscopy was employed to estimate protein secondary structure. PLG microspheres were made using a non-aqueous, cryogenic process and release of spray-freeze dried BSA was assessed in vitro. Results. The most significant atomization parameter affecting particle size was the mass flow ratio (mass of atomization N2 relative to that for liquid feed). Particle size was inversely related to specific surface area and the amount of protein aggregates formed. Zinc-complexation reduced the specific surface area and stabilized the protein against aggregation. FTIR data indicated perturbations in secondary structure upon spray-freeze drying for both excipient-free and zinc-complexed protein. Conclusions. Upon sonication, spray-freeze dried protein powders exhibited friability, or susceptibility towards disintegration. For excipient-free protein, conditions where the mass flow ratio was 〉 ∼0.3 yielded sub-micron powders with relatively large specific surface areas. Reduced particle size was also linked to a decrease in the percentage of protein monomer upon drying. This effect was ameliorated by zinc-complexation, via a mechanism involving reduction in specific surface area of the powder rather than stabilization of secondary structure. Reduction of protein particle size was beneficial in reducing the initial release (burst) of the protein encapsulated in PLG microspheres.

Notizen: Abstract We are concerned with ∈-mixed solutions for weak Stackelberg problems corresponding to two-player nonzero-sum noncooperative games. Two cases are considered: (i) mixed strategies for only the second player; (ii) mixed strategies for both players. After giving basic results relating convergence of functions and weak convergence of probability measures, we establish existence and stability results for ∈-mixed solutions under general assumptions of minimal character without any convexity assumption. Our results improve previous work of Mallozzi and Morgan (Refs. 1–2).

Notizen: Abstract The paper investigates the problem of the robust stability of Schur polynomials. Recently, a new approach based on the Rouche theorem of classical complex analysis has been adopted for the solution of this problem. In this paper, an improvement of the previous solution is presented. This is the optimum solution of the robust stability problem for Schur polynomials, which is obtained by solving a minimization problem and is better than other methods in robust stability literature. Three numerical examples are given to illustrate the proposed method.

Notizen: Abstract In this paper, the concept of extended well-posedness of scalar optimization problems introduced by Zolezzi is generalized to vector optimization problems in three ways: weakly extended well-posedness, extended well-posedness, and strongly extended well-posedness. Criteria and characterizations of the three types of extended well-posedness are established, generalizing most of the results obtained by Zolezzi for scalar optimization problems. Finally, a stronger vector variational principle and Palais-Smale type conditions are used to derive sufficient conditions for the three types of extended well-posedness.

A new approach based on the Rouché theorem is adopted. The extension to the robust stability for multidimensional (multivariable) polynomials is also provided. Interesting sufficient conditions for such robust stability are derived. A two-dimensional example is included to support the theoretical result.

Notizen: Abstract In vector optimization, several authors have studied the upper and lower semicontinuity for mappings involving constraints in topological vector spaces partially ordered through a cone with nonempty interior. In this paper, we give conditions about the upper and lower semicontinuity in the case that the ordering cone in the parameter space has possibly empty interior, as it happens in many function spaces and seqence spaces.

Notizen: Abstract Cadmium toxicity to the diatom Phaeodactylum tricornutum Bohlin has been studied on the basis of the effect of this metal on growth, ATP content, 14C uptake and cellular ultrastructure. The microalga was exposed to 1, 5, 10, 25, 50, 75 and 100 mg l-1 of cadmium and showed a concentration-dependent inhibition in those physiological parameters. At cadmium concentrations of 5 mg l-1 or higher, a significant effect on growth of P. tricornutum was observed. After 8 hours of exposure to the different cadmium concentrations, the decrease in the ATP content was significant only at cadmium concentrations of 25 mg l-1 or higher. 14C uptake was more susceptible to cadmium than the decrease in the ATP content, as 5 mg l-1 of cadmium caused a severe decrease in this parameter. Cadmium also caused ultrastructural changes in P. tricornutum cells: deposition of cadmium on the surface of cell, increase in the chloroplast size, appearance of electrodense granulations, and reduction in lipid inclusions.

Notizen: Abstract The constitutive functions of soft biological tissues during growth are studied. A growth, treated as addition (often non-uniform) of material points, results in deformation, residual stresses, and evolution of the constitutive functions. A theory based on the concept of equivalent material points is developed with the current configuration taken as the reference. The residual stresses developed in a spherical shell undergoing spherical growths are studied.

Notizen: Abstract A mathematical model is presented for four-wheel-steeringvehicles, with the time delay in driver's response and the nonlinearityin lateral tyre forces taken into account. It is proved that thevehicle-driver system has a trivial steady state motion, as well aseight non-trivial steady state motions due to the nonlinearity of tyreforces. The asymptotic stability and Hopf bifurcation of the trivialsteady state are analyzed for two control strategies ofrear-wheel-steering. It is shown through the numerical simulations thatthe four-wheel-steering technique based on the bilinear control strategyworks better when the driver's response involves time delay.

Notizen: Abstract Intracranial saccular aneurysms remain an enigma; it is not known why they form, why they enlarge, or why only some of them rupture. Nonetheless, there is general agreement that mechanics plays an essential role in each aspect of the natural history of these potentially deadly lesions. In this paper, we review recent findings that discount limit point instabilities under quasi-static increases in pressure and resonance under dynamic loading as possible mechanisms of enlargement of saccular aneurysms. Indeed, recent histopathological data suggest that aneurysms enlarge due to a stress-mediated process of growth and remodeling of collagen, the primary load-bearing constituent within the wall. We submit that advanced theoretical, experimental, and numerical studies of this process are essential to further progress in treating this class of pathologies. The purpose of this review is to provide background and direction that encourages elasticians to contribute to this important area of research.

Notizen: Abstract The effects of different concentrations of lead nitrate (10−5 to 10−3 M) on root, hypocotyl, and shoot growth of Indian mustard (Brassica juncea L. var. megarrhiza), and the uptake and accumulation of Pb2+ by its roots, hypocotyls, and shoots were investigated. Lead had no significant inhibitory effect on the root growth at concentrations of 10−5 to 10−4 M during the entire treatment, while at 10−3 M, Pb slightly inhibited the root and shoot growth. B. juncea has ability to take up Pb from solutions and accumulate it in its roots, and transport and concentrate it. The Pb contents in the parts of plants treated with 10−3 M Pb were greater than those of untreated plants, by factors of 230 in the roots, 170 in the hypocotyls, and 3 in the shoots.

Notizen: Abstract The effect of lead and cadmium on biomass accumulation of cucumber seedlings (Cucumis sativus L.) as well as the contents of abscisic acid (ABA), free proline and soluble proteins in leaves were studied. Seedlings were subjected to lead nitrate or cadmium bromide in low concentrations (1 – 5 µM) for 1, 4 or 7 d, and then to the action of the same substances in high concentrations (500 – 1000 µM). The pretreatments of the seedlings with heavy metals in low concentrations enabled them to tolerate the subsequent high concentrations of cadmium and lead without injury. The plant responses to heavy metal treatment were accompanied by the accumulation of ABA, free proline and soluble proteins in leaf tissues.

Notizen: Abstract This study is aimed at detecting gastrointestinal sounds (GIS) and correlating their characteristics with gastrointestinal (GI) conditions. The central hypotheses are that GIS generation depends on the motility patterns and the mechanical properties of the gut, and that changes in those result in measurable differences in GIS. An animal model which included both healthy rats and those with small bowel obstruction (SBO) was developed. The acoustic bursts, of GIS were detected by amplitude thresholding the signal envelope. Three methods of envelope estimation were proposed and evaluated. Envelope estimation using a Hilbert transform was found to produce the best results in the current application. The duration and dominant frequency of each detected GIS event was estimated and clear differences between healthy and diseased rats were discovered. In the control state, GIS events were found to consistently be of relatively short duration (3–65ms). Although the majority of events in the SBO state had similar short duration, infrequent longer events were also detected and appeared to be pathognomonic. Long duration events (〉100 ms) occurred in each of seven obstructed, but in none of 14 non-obstructed, cases (p〈0.001). It is concluded that GIS analysis may prove useful in the non-invasive, rapid, and accurate diagnosis of SBO.

Notizen: Abstract If the sequence of functions ϕj, k is a wavelet frame (Riesz basis) or Gabor frame (Riesz basis), we obtain its perturbation system ψj,k which is still a frame (Riesz basis) under very mild conditions. For example, we do not need to know that the support of ϕ or ψ $$(\hat \phi or\hat \psi )$$ is compact as in [14]. We also discuss the stability of irregular sampling problems. In order to arrive at some of our results, we set up a general multivariate version of Littlewood-Paley type inequality which was originally considered by Lemarié and Meyer [17], then by Chui and Shi [9], and Long [16].

Notizen: Abstract. Augmented Dickey Fuller (ADF) and Kalman filter convergence tests are applied to annual GDPs per head to 16 industrialised countries from 1890 to 1989. Results favour convergence towards the US with a structural break following the Second World War. Estimates suggest that steady-states were higher after the war and that speeds of convergence are different across countries. The Kalman filter method dismissed the no convergence hypothesis more often than its ADF counterpart. This could explain the apparent contradiction in earlier empirical work on similar data sets (cross-section methods tended to favour convergence while time series methods were unable to dismiss the no convergence hypothesis.)

Notizen: Abstract Transport proteins of microorganisms may either belong to the ATP-binding cassette (ABC) superfamily or to the major facilitator (MFS)-superfamily. MFS transporters are single-polypeptide membrane transporters that transport small molecules via uniport, symport or antiport mechanisms in response to a chemiosmotic gradient. Although Saccharomyces cerevisiae is a non-siderophore producer, various bacterial and fungal siderophores can be utilized as an iron source. From yeast genome sequencing data six genes of the unknown major facilitator (UMF) family were known of which YEL065w Sce was recently identified as a transporter for the bacterial siderophore ferrioxamine B (Sit1p). The present investigation shows that another UMF gene, YHL047c Sce, encodes a transporter for the fungal siderophore triacetylfusarinine C. The gene YHL047c Sce (designated TAF1) was disrupted using the kanMX disruption module in a fet3 background (strain DEY 1394 Δfet3), possessing a defect in the high affinity ferrous iron transport. Growth promotion assays and transport experiments with 55Fe-labelled triacetylfusarinine C showed a complete loss of iron utilization and uptake in the disrupted strain, indicating that TAF1 is the gene for the fungal triacetylfusarinine transport in Saccharomyces cerevisiae and possibly in other siderophore producing fungi.

Notizen: Abstract Relative equilibria occur in a wide variety of physical applications, including celestial mechanics, particle accelerators, plasma physics, and atomic physics. We derive sufficient conditions for Lyapunov stability of circular orbits in arbitrary axisymmetric gravitational (electrostatic) and magnetic fields, including the effects of local mass (charge) and current density. Particularly simple stability conditions are derived for source‐free regions, where the gravitational field is harmonic (∇2U = 0) or the magnetic field irrotational (∇ × B = 0). In either case the resulting stability conditions can be expressed geometrically (coordinate‐free) in terms of dimensionless stability indices. Stability bounds are calculated for several examples, including the problem of two fixed centers, the J2 planetary model, galactic disks, and a toroidal quadrupole magnetic field.

Notizen: Abstract We present an analysis of the model introduced by Kokoriev and Kirpichnikov (1988) for the study of unrestricted planar motion of a point mass and a symmetric rigid body whose gravity field is approximated by two point masses (a dumb-bell model). To show possible generalization of the model, we give a systematic derivation of equations of motion for a more general unrestricted problem of a point and a rigid body possessing a plane of dynamical symmetry. We give a simple description of bifurcation of triangular libration points, and we perform an analysis of their linear stability. We propose to extend the model of Kokoriev and Kirpichnikov (1988) to a case when the symmetric body is oblate. In the proposed model the gravity field of moving and rotating body is approximated by two complex masses at complex distance (a complex dumb-bell model). An analysis of bifurcation of the triangular libration points in this model is also presented.

Notizen: Abstract Self-organized rule-following systems are increasingly relevant objects of study in organization theory due to such systems&2018; capacity to maintain control while enabling decentralization of authority. This paper proposes a network model for such systems and examines the stability of the networks&2018; repetitive behavior. The networks examined are Ashby nets, a fundamental class of binary systems: connected aggregates of nodes that individually compute an interaction rule, a binary function of their three inputs. The nodes, which we interpret as workers in a work team, have two network inputs and one self-input. All workers in a given team follow the same interaction rule. We operationalize the notion of stability of the team&2018;s work routine and determine stability under small perturbations for all possible rules these teams can follow. To study the organizational concomitants of stability, we characterize the rules by their memory, fluency, homogeneity, and autonomy. We relate these measures to work routine stability, and find that stability in ten member teams is enhanced by rules that have low memory, high homogeneity, and low autonomy.

Notizen: Abstract This paper studies the stability of a three‐station fluid network. We show that, unlike the two‐station networks in Dai and Vande Vate [18], the global stability region of our three‐station network is not the intersection of its stability regions under static buffer priority disciplines. Thus, the “worst” or extremal disciplines are not static buffer priority disciplines. We also prove that the global stability region of our three‐station network is not monotone in the service times and so, we may move a service time vector out of the global stability region by reducing the service time for a class. We introduce the monotone global stability region and show that a linear program (LP) related to a piecewise linear Lyapunov function characterizes this largest monotone subset of the global stability region for our three‐station network. We also show that the LP proposed by Bertsimas et al. [1] does not characterize either the global stability region or even the monotone global stability region of our three‐station network. Further, we demonstrate that the LP related to the linear Lyapunov function proposed by Chen and Zhang [11] does not characterize the stability region of our three‐station network under a static buffer priority discipline.

Notizen: Abstract We prove a general implicit function theorem for multifunctions with a metric estimate on the implicit multifunction and a characterization of its coderivative. Traditional open covering theorems, stability results, and sufficient conditions for a multifunction to be metrically regular or pseudo-Lipschitzian can be deduced from this implicit function theorem. We prove this implicit multifunction theorem by reducing it to an implicit function/solvability theorem for functions. This approach can also be used to prove the Robinson–Ursescu open mapping theorem. As a tool for this alternative proof of the Robinson–Ursescu theorem, we also establish a refined version of the multidirectional mean value inequality which is of independent interest.

Notizen: Abstract This paper describes a family of discrete-review policies for scheduling open multiclass queueing networks. Each of the policies in the family is derived from what we call a dynamic reward function: such a function associates with each queue length vector q and each job class k a positive value r k (q), which is treated as a reward rate for time devoted to processing class k jobs. Assuming that each station has a traffic intensity parameter less than one, all policies in the family considered are shown to be stable. In such a policy, system status is reviewed at discrete points in time, and at each such point the controller formulates a processing plan for the next review period, based on the queue length vector observed. Stability is proved by combining elementary large deviations theory with an analysis of an associated fluid control problem. These results are extended to systems with class dependent setup times as well as systems with alternate routing and admission control capabilities.

Notizen: Abstract The subject of the paper is the stability analysis of some neural networks consisting of a finite number of interacting neurons. Following the approach of Dai [5] we use the fluid limit model of the network to derive a sufficient condition for positive Harris-recurrence of the associated Markov process. This improves the main result in Karpelevich et al. [11] and, at the same time, sheds some new light on it. We further derive two different conditions that are sufficient for transience of the state process and illustrate our results by classifying some examples according to positive recurrence or transience.

Notizen: Abstract We focus on window flow control as used in packet-switched communication networks. The approach consists in studying the stability of a system where each node on the path followed by the packets of the controlled connection is modeled by a FIFO (First-In-First-Out) queue of infinite capacity which receives in addition some cross traffic represented by an exogenous flow. Under general stochastic assumptions, namely for stationary and ergodic input processes, we show the existence of a maximum throughput allowed by the flow control. Then we establish bounds on the value of this maximum throughput. These bounds, which do not coincide in general, are reached by time-space scalings of the exogenous flows. Therefore, the performance of the window flow control depends not only on the traffic intensity of the cross flows, but also on fine statistical characteristics such as the burstiness of these flows. These results are illustrated by several examples, including the case of a nonmonotone, nonconvex and fractal stability region.

Notizen: Abstract The characteristic equation of a system of delay differential equations (DDEs) is a nonlinear equation with infinitely many zeros. The stability of a steady state solution of such a DDE system is determined by the number of zeros of this equation with positive real part. We present a numerical algorithm to compute the rightmost, i.e., stability determining, zeros of the characteristic equation. The algorithm is based on the application of subspace iteration on the time integration operator of the system or its variational equations. The computed zeros provide insight into the system’s behaviour, can be used for robust bifurcation detection and for efficient indirect calculation of bifurcation points.

Notizen: Abstract This paper describes the construction of block predictor–corrector methods based on Runge–Kutta–Nyström correctors. Our approach is to apply the predictor–corrector method not only with stepsize h, but, in addition (and simultaneously) with stepsizes a i h, i = 1 ...,r. In this way, at each step, a whole block of approximations to the exact solution at off‐step points is computed. In the next step, these approximations are used to obtain a high‐order predictor formula using Lagrange or Hermite interpolation. Since the block approximations at the off‐step points can be computed in parallel, the sequential costs of these block predictor–corrector methods are comparable with those of a conventional predictor–corrector method. Furthermore, by using Runge–Kutta–Nyström corrector methods, the computation of the approximation at each off‐step point is also highly parallel. Numerical comparisons on a shared memory computer show the efficiency of the methods for problems with expensive function evaluations.

Notizen: Abstract We study time integration methods for equations of mixed quantum-classical molecular dynamics in which Newtonian equations of motion and Schrödinger equations are nonlinearly coupled. Such systems exhibit different time scales in the classical and the quantum evolution, and the solutions are typically highly oscillatory. The numerical methods use the exponential of the quantum Hamiltonian whose product with a state vector is approximated using Lanczos' method. This allows time steps that are much larger than the inverse of the highest frequencies. We describe various integration schemes and analyze their error behaviour, without assuming smoothness of the solution. As preparation and as a problem of independent interest, we study also integration methods for Schrödinger equations with time-dependent Hamiltonian.

Notizen: Abstract We present a new simulation code able to simulate the entire propagation of laser pulse, from the amplifiers level up to the focusing stage. This algorithm has some new characteristics that we intend to present. It computes the three-dimensional optical propagation equation using no approximation other than its picosecond expression. The stability has been carefully studied so that it can be applied to any geometry. This is a great improvement since, up to now only cylindrical geometry was accessible for accuracy. In this paper we also present a method using Fast Fourier Transform able to evaluate with a high accuracy, Fresnel’s distribution of a focused laser pulse. The advantages provided by our algorithm are its rapidity and its high physical understanding of the focusing phenomena.

Notizen: Abstract This paper examines the effects of ionic gold on Saccharomyces cerevisiae, as determined by long-term (growth in gold-containing media) and short-term interactions (H+ efflux activity). An increasing gold concentration inhibited growth and at 〈0.2 mM Au, growth was not observed. Transmission electron microscopy revealed no differences in ultrastructure but fine electron dense particles were observed in unstained preparations from gold-containing medium. After glucose addition (to 10mM) to starved suspensions of S. cerevisiae, glucose-dependent reduction of external pH occurred as the cells extruded protons. In the presence of increasing gold concentrations, the lag time before proton extrusion did not change but the rate and duration decreased significantly with a marked influence on proton efflux rate being observed at ≤ 10 μM. Extension of preincubation time of yeast cells in gold-containing medium resulted in a decreasing proton efflux rate and colloidal phase formation in the cell suspensions, the time between gold addition and the beginning of colloidal phase formation depending on the gold concentration used. Both Ca and Mg enhanced the inhibitory effect of gold on the yeast cells with Ca showing a stronger inhibitory effect than Mg.

Notizen: Summary In this paper, the global behavior of relative equilibrium states of a three-body satellite with flexible connection under the action of the gravitational torque is studied. With geometric method, the conditions of existence of nontrivial solutions to the relative equilibrium equations are determined. By using reduction method and singularity theory, the conditions of occurrence of bifurcation from trivial solutions are derived, which agree with the existence conditions of nontrivial solutions, and the bifurcation is proved to be pitchfork-bifurcation. The Liapunov stability of each equilibrium state is considered, and a stability diagram in terms of system parameters is presented.

Notizen: Abstract Sensitivity of the returns to scale (RTS) classifications in data envelopment analysis is studied by means of linear programming problems. The stability region for an observation preserving its current RTS classification (constant, increasing or decreasing returns to scale) can be easily investigated by the optimal values to a set of particular DEA-type formulations. Necessary and sufficient conditions are determined for preserving the RTS classifications when input or output data perturbations are non-proportional. It is shown that the sensitivity analysis method under proportional data perturbations can also be used to estimate the RTS classifications and discover the identical RTS regions yielded by the input-based and the output-based DEA methods. Thus, our approach provides information on both the RTS classifications and the stability of the classifications. This sensitivity analysis method can easily be applied via existing DEA codes.

Notizen: Abstract The first ecosystem mimic hypothesis suggests clear advantages if man-made land use systems do not deviate greatly in their resource use patterns from natural ecosystems typical of a given climatic zone. The second hypothesis claims that additional advantages will accrue if agroecosystems also maintain a substantial part of the diversity of natural systems. We test these hypotheses for the savannah zone of sub-Saharan Africa, with its low soil fertility and variable rainfall. Where annual food crops replace the natural grass understorey of savannah systems, water use will decrease and stream and groundwater flow change, unless tree density increases relative to the natural situation. Increasing tree density, however, will decrease crop yields, unless the trees meet specific criteria. Food crop production in the parkland systems may benefit from lower temperatures under tree canopies, but water use by trees providing this shade will prevent crops from benefiting. In old parkland trees that farmers have traditionally retained when opening fields for crops, water use per unit shade is less than in most fast growing trees introduced for agroforestry trials. Strong competition between plants adapted to years with different rainfall patterns may stabilise total system productivity -- but this will be appreciated by a farmer only if the components are of comparable value. The best precondition for farmers to maintain diversity in their agroecosystem hinges on the availability of a broad basket of choices, without clear winners or 'best bets'.

Notizen: Abstract One of the fundamental problems of quasicrystals is to understand their occurrence in microscopic models of interacting particles. We review here recent attempts to construct stable quasicrystalline phases. In particular, we compare two recently constructed classical lattice-gas models with translation-invariant interactions and without periodic ground-state configurations. The models are based on nonperiodic tilings of the plane by square-like tiles. In the first model, all interactions can be minimized simultaneously. The second model is frustrated; its nonperiodic ground state can arise only by the minimization of the energy of competing interactions. We put forward some hypotheses concerning stabilities of nonperiodic ground states. In particular, we introduce two criteria, the so-called strict boundary conditions, and prove their equivalence to the zero-temperature stability of ground states against small perturbations of potentials of interacting particles. We discuss the relevance of these conditions for the low-temperature stability, i.e., for the existence of thermodynamically stable nonperiodic equilibrium states.

Notizen: Abstract The classical Lifshitz–Slyozov–Wagner theory of domain coarsening predicts asymptotically self-similar behavior for the size distribution of a dilute system of particles that evolve by diffusional mass transfer with a common mean field. Here we consider the long-time behavior of measure-valued solutions for systems in which particle size is uniformly bounded, i.e., for initial measures of compact support. We prove that the long-time behavior of the size distribution depends sensitively on the initial distribution of the largest particles in the system. Convergence to the classically predicted smooth similarity solution is impossible if the initial distribution function is comparable to any finite power of distance to the end of the support. We give a necessary criterion for convergence to other self-similar solutions, and conditional stability theorems for some such solutions. For a dense set of initial data, convergence to any self-similar solution is impossible.

Notizen: Abstract The occurrence of periodic changes in the length of the internodes of the seedlings of SE Asian mangrove species (Rhizophora apiculata, Avicennia alba, and Sonneratia caseolaris) growing in the Pak Phanang estuary (Nakhon Si Thammarat, SE Thailand) was confirmed. The annual nature of these periodic changes was verified through comparisons with the observed average number of internodes produced by plants tagged for one year. The cycles in the sequence of internodal length of the seedlings had an average (± SE) period of 16.0 ± 0.78 internodes for Avicennia alba, 25.0 ± 1.2 internodes for Sonneratia caseolaris, and 8.29 ± 0.40 internodes for Rhizophora apiculata. These values are remarkably close to, and not significantly different (t‐test, P 〉 0.05) from the average (± SE) annual number of internodes of 17.6 ± 0.8 for Avicennia alba, 28.8 ± 2.1 for Sonneratia caseolaris, and 8.03 ± 0.36 for Rhizophora apiculata produced in a year by tagged plants. In addition, the average (± SE) growth of the tagged plants was 0.39 ± 0.018 cm day-1 for Avicennia alba, 0.24 ± 0.015 cm day-1 for Sonneratia caseolaris, and 0.091 ± 0.0036 cm day-1 for Rhizophora apiculata. These estimates were very close to those (0.45 ± 0.06 cm day-1 for Avicennia alba, 0.32 ± 0.01 cm day-1 for Sonneratia caseolaris, and 0.13 ± 0.005 cm day-1 for Rhizophora apiculata) derived as the product of the mean number of internodes produced per day, calculated from the inferred number of internodes produced per year, and the mean length of the internodes of the plants. These findings indicate that the use of regular, annual changes in the length of the internodes of mangrove seedlings can be used reliably to determine their age and to estimate their growth rate.

Notizen: Abstract An indirect adaptive control approach is developed in this paper for robots with unknown nonlinear dynamics using neural networks (NNs). A key property of the proposed approach is that the actual joint angle values in the control law are replaced by the desired joint angles, angle velocities and accelerators, and the bound on the NN reconstruction errors is assumed to be unknown. Main theoretical results for designing such a neuro-controller are given, and the control performance of the proposed controller is verified with simulation studies.

Notizen: Abstract Uptake by Saccharomyces cerevisiae of the sulphur-containing amino acid L-cysteine was found to be non-saturable under various conditions, and uptake kinetics suggested the existence of two or more transport systems in addition to the general amino-acid permease, Gap1p. Overexpression studies identified BAP2, BAP3, AGP1 and GNP1 as genes encoding transporters of cysteine. Uptake studies with disruption mutants confirmed this, and identified two additional genes for transporters of cysteine, TAT1 and TAT2, both very homologous to BAP2, BAP3, AGP1 and GNP1. While Gap1p and Agp1p appear to be the main cysteine transporters on the non-repressing nitrogen source proline, Bap2p, Bap3p, Tat1p, Tat2p, Agp1p and Gnp1p are all important for cysteine uptake on ammonium-based medium. Furthermore, whereas Bap2p, Bap3p, Tat1p and Tat2p seem most important under amino acid-rich conditions, Agp1p contributes significantly when only ammonium is present, and Gnp1p only contributes under the latter condition.

Notizen: Abstract The frequency of reversion in a histidine-requiring mutant of Saccharomyces cerevisiae increases about ten-fold in stationary cells during histidine starvation. Histidine starvation enhances a similar frequency of reversion in a tryptophan-requiring mutant. Starvation, therefore, enhances mutation frequencies in a non-adaptive manner. The base analogue 6-N-hydroxylaminopurine (HAP) added prior to plating on medium with limited histidine strongly increases reversion of the histidine mutant. HAP-induced reversion increases further in stationary starving cells with the same kinetics as that which increases spontaneous reversion. Adding HAP to the stationary starving cells does not produce any effect.

Notizen: Abstract Site-directed mutagenesis was used to construct yeast centromere plasmids in which a strand nick or gap could be placed 5′ or 3′, on either strand, to a reporter gene (SUP4-o) carrying defined base mismatches. The plasmids were then transformed into yeast cells and the direction and efficiency of mismatch repair were assayed by scoring colouring of the transformant colonies. Strands that were nicked were consistently corrected more often than intact strands, but the effect was very small. However, placement of a small gap at the same positions as the nicks resulted in a marked increase in selection for the gapped strand and an enhanced efficiency of mismatch repair. Both the preference for the gapped strand and correction of the mismatch were offset by deletion of the mismatch repair gene PMS1. Together, the results suggest that strand interruptions can direct intracellular mismatch correction of plasmid-borne base mispairs in yeast.

Notizen: Abstract It is thought that the FLO8 gene encodes a transcriptional activator of the dominant flocculation gene FLO1 in Saccharomycescerevisiae. To determine other genes which are regulated by FLO8, a detailed comparison of the transcripts from the FLO8 and Δflo8 strains was carried out. In addition to the FLO1 gene, it was found that transcription of the FLO11 and STA1 genes is positively regulated by FLO8. In flo8 strains, not only transcripts of the FLO11, STA1, and FLO1 genes but also invasive growth, extracellular glucoamylase production, and flocculation were undetected. From these results, it is suggested that FLO8 regulates these characteristics via the transcriptional regulation of the FLO11, STA1, and FLO1 genes.

Notizen: Abstract. While learning and development are well characterized in feedforward networks, these features are more difficult to analyze in recurrent networks due to the increased complexity of dual dynamics – the rapid dynamics arising from activation states and the slow dynamics arising from learning or developmental plasticity. We present analytical and numerical results that consider dual dynamics in a recurrent network undergoing Hebbian learning with either constant weight decay or weight normalization. Starting from initially random connections, the recurrent network develops symmetric or near-symmetric connections through Hebbian learning. Reciprocity and modularity arise naturally through correlations in the activation states. Additionally, weight normalization may be better than constant weight decay for the development of multiple attractor states that allow a diverse representation of the inputs. These results suggest a natural mechanism by which synaptic plasticity in recurrent networks such as cortical and brainstem premotor circuits could enhance neural computation and the generation of motor programs.

Notizen: Abstract We study thermomechanical deformations of a viscoplastic body deformed in simple shear. The strain gradients are taken as independent kinematic variables and the corresponding higher order stresses are included in the balance laws, and the equation for the yield surface. Three different functional relationships, the power law, and those proposed by Wright and Batra, and Johnson and Cook are used to relate the effective strain rate to the effective stress and temperature. Effects of strain hardening of the material and elastic deformations are neglected. The homogeneous solution of the problem is perturbed and the stability of the problem linear in the perturbation variables is studied. Following Wright and Ockendon's postulate that the wavelength whose initial growth rate is maximum determines the minimum spacing between adjacent shear bands, the shear band spacing is computed. It is found that the minimum shear band spacing is very sensitive to the thermal softening coefficient/exponent, the material characteristic length and the nominal strain-rate. Approximate analytical expressions for the critical wave length for heat conducting nonpolar materials and locally adiabatic deformations of gradient dependent materials are also derived.

Notizen: Abstract The yeast gene PSO7 was cloned from a genomic library by complementation of the pso7-1 mutant's sensitivity phenotype to 4-nitroquinoline-1-oxide (4NQO). Sequence analysis revealed that PSO7 is allelic to the 1.1-kb ORF of the yeast gene COX11 which is located on chromosome XVI and encodes a protein of 28-kDa localized in the inner mitochondrial membrane. Allelism of PSO7/COX11 was verified by non-complementation of 4NQO-sensitivity in diploids homo- and hetero-allelic for the pso7-1 and cox11::TRP1 mutant alleles. Sensitivity to 4NQO was the same in exponentially growing cells of the pso7-1 mutant and the cox11::TRP1 disruptant. Allelism of COX11 and PSO7 indicates that the pso7 mutant's sensitivity to photoactivated 3-carbethoxypsoralen and to 4NQO is not caused by defective DNA repair, but rather is due to an altered metabolism of the pro-mutagen 4NQO in the absence of cytochrome oxidase (Cox) in pso7-1/cox11::TRP1 mutants/disruptants. Lack of Cox might also lead to a higher reactivity of the active oxygen species produced by photoactivated 3-carbethoxypsoralen. The metabolic state of the cells is important for their sensitivity phenotype since the largest enhancement of sensitivity to 4NQO between wild-type (WT) and the pso7 mutant occurs in exponentially growing cells, while cells in stationary phase or growing cells in phosphate buffer have the same 4NQO resistance, irrespective of their WT/mutant status. Strains containing the pso7-1 or cox11::TRP1 mutant allele were also sensitive to the oxidative stress-generating agents H2O2 and paraquat. Mutant pso7-1, as well as disruptant cox11::TRP1, harboured mitochondria that in comparison to WT contained less than 5% and no detectable Cox activity, respectively.

Notizen: Abstract The major yeast plasma membrane H+-ATPase is encoded by the essential PMA 1 gene. The PMA 2 gene encodes an H+-ATPase that is functionally interchangeable with the one encoded by PMA 1 , but it is expressed at a much lower level than the PMA 1 gene and it is not essential. Using genetically manipulated strains of Saccharomyces cerevisiae that exclusively synthesize PMA1 ATPase or PMA2 ATPase under control of the PMA1 promoter, we found that yeast cultivation under mild copper stress leads to a similar activation of PMA2 and PMA1 isoforms. At high inhibitory copper concentrations (close to the maximum that allowed growth), ATPase activity was reduced from maximal levels; this decrease in activity was less important for PMA2 ATPase than for PMA1 ATPase. The higher tolerance to high copper stress of the artificial strain synthesizing PMA2 ATPase exclusively, as compared to that synthesizing solely PMA1 ATPase, correlated both with the lower sensitivity of PMA2 ATPase to the deleterious effects of copper in vivo and with its higher apparent affinity for MgATP, and suggests that plasma membrane H+-ATPase activity plays a role in yeast tolerance to copper.

Notizen: Abstract Modern thermal analysis, microcalorimetry and new emerging combined techniques which deliver calorimetric, microscopic and spectroscopic data offer a powerful analytical battery for the study of pharmaceuticals. These techniques are very useful in all steps of development of new drug products as well as methods for quality control in production. The characterization of raw materials enables to understand the relationships between polymorphs, solvates and hydrates and to choose the proper development of new drug products with very small amount of material in a very short time. Information on stability, purity is valuable for new entities as well as for marketed drug substances from different suppliers. Excipients which vary from single organic or inorganic entity to complexes matrixes or polymers need to be characterized and properly controlled. The thermodynamic phase-diagrams are the basis of the studies of drug-excipients interactions. They are very useful for the development of new delivery systems. A great number of new formulations need proper knowledge of the behaviour of the glass transition temperature of the components. Semi-liquid systems, interactions in aqueous media are also successfully studied by these techniques.

It was found that the model could be used for the verification of the ignitability method developed by Miller et al. [1]. Different steady states of combustion were found when using the model proposed. There is a characteristic steady state for normal flaming combustion, another for non-flaming combustion, and there are also unstable steady states that have no real physical meaning.

Notizen: Abstract In this work we report a thermodynamic characterization of stability and melting behaviour of two 24-mer DNA triplexes. The third strand, that binds the Watson-Crick double helix with Hoogsteen hydrogen bonds, contains 3′-3′ phosphodiester junction that determines the polarity inversion. The target double helix is composed of adjacent and alternate fragments of oligopurine-oligopyrimidine tracts. The two helices differ from the substitution of the cytosine, involved in the junction, with the thymine. Calorimetric data reported here provide a quantitative measure of the influence of pH and base modification on the stability of a DNA triplex.

Notizen: Abstract Bighead carp (Aristichthys nobilis Oshima) fry of various ages (11, 18, and 35 days post-hatch) were exposed to the low salinities encountered during the annual intrusion of seawater in Laguna Lake, Philippines. Practical indices of salinity tolerance assessed the effect of a 96 h direct exposure to low salinities (0–16‰). Mean (MST) and median survival times (MST50) of fry decreased as salinity of rearing medium increased. Younger fry were less able to tolerate exposure to these salinities than their older cohorts. Median lethal salinity after 96 h (MLS) revealed higher tolerance among 35–day old fry (7.6‰) than 11 (2.3‰) and 18–day old fry (6.0‰), demonstrating that survival in saline water depends on their age at initial exposure to low salinities. Mean body weight of 18–day old fry reared in 0 and 2‰ for 3 and 4 weeks was higher than for those reared in 4 and 6‰ for the same period. Growth over these periods was inversely related with the range of salinities tested. These results demonstrate that, despite their known stenohalinity, bighead carp fry possess some degree of osmoregulatory capability, allowing them to survive and grow in lakes subjected periodically to saltwater inflow.

Notizen: Abstract Juvenile tiger puffer (Takifugu rubripes) were fed semi-purified diets with graded levels (0–0.40%) of calcium (Ca) supplements for 8 weeks at a temperature of 23.0–25.5 °C. At the end of the feeding experiment, average final body weight increased with increasing dietary Ca levels. A diet without Ca supplement resulted in poor average final body weight and 0.05% dietary Ca supplement could not significantly improve the growth. However, supplemental Ca levels of 0.10–0.40% significantly increased growth. Dietary Ca supplements of 0.10% or more also improved the feed efficiency in tiger puffer. Ca and phosphorus contents of the bone were independent of dietary Ca supplements. However, 0.40% Ca in the diet decreased the zinc and manganese contents of bone. Results indicated that supplemental Ca levels of 0.10–0.20% in a semi-purified diet were required to maintain normal growth, feed utilization and bone mineralization of tiger puffer.

Notizen: Abstract Calmodulin of Saccharomyces cerevisiae has different Ca2+ binding properties from other calmodulins. We previously reported that the maximum number of Ca2+ binding was 3 mol/mol and the fourth binding site was defective, which was different from 4 mol/mol for others. Their macroscopic dissociation constants suggested the cooperative three Ca2+ bindings rather than a pair of cooperative two Ca2+ bindings of ordinary calmodulin. Here we present evidence for yeast calmodulin showing the intramolecular close interaction between the N-terminal half domain and the C-terminal half domain, while the two domains of ordinary calmodulin are independent of each other. We will discuss the relationship of the shape and the shape change caused by the Ca2+ binding to the enzyme activation in yeast. The functional feature of calmodulin in yeast will also be considered, which might be different from the one of vertebrate calmodulin.

Notizen: Abstract Dipyridyl-dithio substrates were cleaved by isolated vesicles of plasma membranes prepared from etiolated hypocotyls of soybean. The cleavage was stimulated by auxins at physiological concentrations. The substrates utilized were principally 2,2′-dithiodippyrine (DTP) and 6,6′-dithiodinicotinic acid (DTNA). The DTP generated 2 moles of 2-pyridinethione whereas the 6,6′-dithiodinicotinic acid generated 2 moles of 6-nicotinylthionine. Both products absorbed at 340 nm. The auxin herbicide, 2,4-dichlorophenoxyacetic acid (2,4-D) stimulated the activity approximately 2-fold to a maximum at about 10 μM. Concentrations of 2,4-D greater than 100 μM inhibited the activity. Indole-3-acetic acid stimulated the activity as well. The growth-inactive auxin, 2,3-dichlorophenoxyacetic acid (2,3-D), was without effect. DTNA cleavage correlated with oxidation of NADH and reduction of protein disulfide bonds reported earlier in terms of location at the external plasma membrane surface, absolute specific activity, pH dependence and auxin specificity. The dipyridyl-dithio substrates provide, for the first time, a direct measure of the disulfide-thiol interchange activity of the protein previously measured only indirectly as an auxin-dependent ability of isolated plasma membrane vesicles to restore activity to scrambled and inactive RNase.

Notizen: Abstract The 2818 amino acids of neurofibromin, the product of the human NF1 gene, include a 230 amino acid Ras-GAP related domain (GRD). Functions which may be associated with the rest of the protein remain unknown. However, many NF1 mutations in neurofibromatosis 1 patients are found downstream of the GRD, suggesting that the C-terminal region of the protein is also functionally important. Since the C-terminal region of neurofibromin encompassing these mutations is homologous with the corresponding regions in the two Saccharomyces cerevisiae Ras-GAPs, Ira1p and Ira2p, we chose yeast as a model system for functional exploration of this region (Ira-C region). Three missense mutations that affect the Ira-C region of NF1 were used as a model for the mutagenesis of IRA1. The yeast phenotypes of heat shock sensitivity, iodine staining, sporulation efficiency, pseudohyphae formation, and GAP activity were scored. Even though none of the mutations directly affected the Ira1p-GRD, mutations at two of the three sites resulted in a decrease in the GAP activity present in ira1 cells. The third mutation appeared to disassociate the phenotypes of sporulation ability and GAP activity. This and other evidence suggest an effector function for Ira1p.

Notizen: Abstract We imaged pores on the surface of the cell wall of three different industrial strains of Saccharomyces cerevisiae using atomic force microscopy. The pores could be enlarged using 10 mM diamide, an SH residue oxidant that attacks surface proteins. We found that two strains showed signs of oxidative damage via changes in density and diameter of the surface pores. We found that the German strain was resistant to diamide induced oxidative damage, even when the concentration of the oxidant was increased to 50 mM. The normal pore size found on the cell walls of American strains had diameters of about 200nm. Under conditions of oxidative stress the diameters changed to 400nm. This method may prove to be a useful rapid screening process (45-60 min) to determine which strains are oxidative resistant, as well as being able to screen for groups of yeast that are sensitive to oxidative stress. This rapid screening tool may have direct applications in molecular biology (transference of the genes to inside of living cells) and biotechnology (biotransformations reactions to produce chiral synthons in organic chemistry.

Notizen: Abstract This paper studies the expected total cost (ETC) criterion for discrete-time Markov control processes on Borel spaces, and possibly unbounded cost-per-stage functions. It presents optimality results which include conditions for a control policy to be ETC-optimal and for the ETC-value function to be a solution of the dynamic programming equation. Conditions are also given for the ETC-value function to be the limit of the α-discounted cost value function as α ↑ 1, and for the Markov control process to be `stable" in the sense of Lagrange and almost surely. In addition, transient control models are fully analized. The paper thus provides a fairly complete, up-dated, survey-like presentation of the ETC criterion for Markov control processes on Borel spaces.

Notizen: Abstract We examine the effects of the cyanogenesis polymorphism in Turnera ulmifolia on larvae, pupae, and adults of Euptoieta hegesia, the most damaging herbivore of T. ulmifolia in terms of tissue loss per unit time. We provide evidence that female E. hegesia do not show preference for host plants on the basis of their cyanogenesis level but do prefer T. ulmifolia over equally cyanogenic, closely related secondary host-plant species (Passiflora sp.). Similarly, cyanogenesis in T. ulmifolia has little effect on the food preference, growth, or development of the larvae. The potential host range of E. hegesia is limited, even within the genus Turnera, but this does not appear to be due to host-plant cyanogenesis. Pupae suffer very high mortality levels in the wild that are not associated with host-plant cyanogenesis, although our studies indicate that larvae are capable of sequestering cyanogenic glycosides from their host plants and possibly of synthesizing these or similar compounds. We provide evidence that the presence of sequestered cyanogenic compounds in the larvae protects them from terrestrial-based predators such as Anolis lizards

Notizen: Abstract Toxicity assessment based on sediment chronic bioassays with the aquatic worm Tubifex tubifex was performed at ten contaminated sites in the industrial area of Bilbao (Northern Spain). One control and three reference sites were also included. Tubifex bioassay measures both survival and reproduction impairment. These endpoints have been contrasted and discussed in relation to somatic growth and both individual and total biomass of cocoons. Survival was only affected at one site which was heavily contaminated by organic compounds, mainly PAHs. A group of four severely ecotoxic sediments was characterised by a drastic reduction in number and size of cocoons, and adult somatic growth. In other group of sediments, some significant increases were found for these variables. It is suggested that these increases represent an effect of hormesis. An index of reproductive effort was used to integrate the relationship between somatic growth and reproduction. Values of reproductive effort at the test sediments were lower than those at the control sediment, suggesting a conservative strategy of oligochaete worms which consisted in an investment into somatic line (growth) at the expenses of offspring. Rates of food consumption which were estimated from egestion rates, were low at the contaminated sites. This fact could be related to the low production levels found at these sites and may reflect avoidance feeding behaviour of the oligochaete worms within the sediments. At some reference sites, high production could have resulted from high nutritional quality of sediments, or to an hormetic effect due to low concentration of some chemicals. Results are discussed in relation to toxicity data from sediment three-brood bioassay with Daphnia magna Straus performed separately on the same sediments.

Notizen: Abstract Two members of the ATP-dependent class of phosphoenolpyruvate carboxykinases (PEPCKs) (Saccharomyces cerevisiae and Anaerobiospirillum succiniciproducens) have been comparatively studied with regard to their oxaloacetate (OAA) decarboxylase and pyruvate kinase-like activities. The pyruvate kinase-like activities were dependent on the presence of Mn2+; at the same concentrations Mg2+ was not effective. These activities were synergistically activated by a combination of both metal ions. V max for these activities in A. succiniciproducens and S. cerevisiae PEPCKs was 0.13% and 1.2% that of the principal reaction, respectively. The OAA decarboxylase activity was nucleotide independent and, with decreasing order of effectiveness, these activities were supported by Mn2+ and Mg2+. AMP is an activator of these reactions. V max for the OAA decarboxylase activities in A. succiniciproducens and S. cerevisiae PEPCKs was 4% and 0.2% that of the PEP-forming reaction, respectively.

Notizen: Abstract It is demonstrated that mixtures of ditetradecyl- phosphatidylcholine or didodecyl-phoshatidylcholine and dihexyl- phosphatidylcholine in water form lyotropic liquid crystalline phases under similar conditions as previously reported for bicelles consisting of dimyristoyl-phosphatidylcholine (DMPC) and dihexanoyl- phosphatidylcholine (DHPC). The carboxy-ester bonds present in DMPC and DHPC are replaced by ether linkages in their alkyl analogs, which prevents acid- or base-catalyzed hydrolysis of these compounds. 15N-1H dipolar couplings measured for ubiquitin over the 2.3–10.4pH range indicate that this protein retains a backbone conformation which is very similar to its structure at pH 6.5 over this entire range.

Notizen: Abstract We have previously shown that a combination of the cytokines interleukin (IL)-1, IL-11, leukemia inhibitory factor (LIF), and glial cell line-derived neurotrophic factor (GDNF) can convert rat fetal (E14.5) mesencephalic progenitor cells into tyrosine hydroxylase (TH)-immunoreactive (ir) neurons in vitro. The experiments described here characterize the mesencephalic progenitor cells and their cytokine-induced conversion into dopamine (DA) neurons. For all experiments, we used bromodeoxyuridine (BrdU)-ir cultures of (E14.5) mesencephalic progenitor cells that had been expanded at least 21 days. We first demonstrated that IL-1 induced DA neuron conversion in mesencephalic progenitors, but not in striatal progenitors (P〈0.001). Thus, these cells should be classified as lineage-restricted progenitors, and not omnipotent stem cells. To further characterize cell populations in these cultures, we used monoclonal antibodies against Hu (an early marker for neurons), growth-associated protein (GAP)-43 (a marker for neuronal process extension), TH (a marker for DA neurons), and glial fibrillary acidic protein (GFAP, a marker for astrocytes). We assessed (E14.5) mesencephalic progenitor cell cultures (plated at 125,000 cells/cm2) incubated in the cytokine mixture (described above) or in complete media (CM, negative control). Following 7 days incubation, GFAP-positive cells formed a nearly confluent carpet in both types of cultures. However, numbers of Hu-ir and GAP-43-ir cells in the cytokine-incubated cultures far exceeded those in CM-incubated controls (P=0.0003, P=0.0001, respectively), while numbers of TH-ir cells were 58-fold greater in the cytokine-incubated cultures versus CM-incubated controls. The TH phenotype persisted for 7 days following withdrawal of the differentiation media. Numerous double-labeled cells that were BrdU-ir and also TH-ir, or Hu-ir and also TH-ir, were observed in the cytokine-incubated cultures. These data suggest that cytokines ”drive” the conversion of progenitor cells into DA neurons.
