    Beschreibung: The highly selective, enzyme-activated, irreversible inhibitor of L-ornithine decarboxylase, DL-alpha-difluoromethylornithine, suppresses the increase in uterine L-ornithine decarboxylase activity associated with early embryogenesis in the mouse and arrests embryonic development at that stage. Contragestational effects were confirmed in the rat and rabbit.     Beschreibung: Neurons in deep laminae of the rabbit cingulate cortex develop discriminative activity at an early stage of behavioral discrimination learning, whereas neurons in the anteroventral nucleus of thalamus and neurons in the superficial cortical laminae develop such activity in a late stage of behavioral learning. It is hypothesized that early-forming discriminative neuronal activity, relayed to anteroventral neurons via the corticothalamic pathway, contributes to the construction of changes underlying the late-forming neuronal discrimination in the anteroventral nucleus.     Beschreibung: A new N-methylpurine riboside (doridosine), probably N1-Methylisoguanosine, was isolated from the digestive glands of a nudibranch.     Beschreibung: In BALB/c female mice with melanoma transplants, the incidence of "takes" is decreased and survival is increased by hydroquinone, a melanocytolytic agent. The mechanism of drug action is suggested by via DNA.     Beschreibung: Amnesic patients acquired a mirror-reading skill at a rate equivalent to that of matched control subjects and retained it for at least 3 months. The results indicate that the class of preserved learning skills in amnesia is broader than previously reported. Amnesia seems to spare information that is based on rules or procedures, as contrasted with information that is data-based or declarative--"knowing how rather than "knowing that."     Beschreibung: In kidney proximal tubules of male mice the mitochondria are larger and more electron-lucent, autophagic vacuoles and lysosomes (predominantly myeloid bodies) more numerous and voluminous, and exocytosed intraluminal myeloid bodies more common than in females. Males also have higher kidney activities of mitochondrial cytochrome c oxidase and lysosomal hydrolases, and excrete larger quantities of hydrolases and protein in the urine. Orchiectomy evokes the feminine pattern whereas testosterone administration induces the male pattern.     Beschreibung: A spontaneous B cell leukemia (BCL1) grew progressively in normal BALB/c mice after injection of tumor cells but did not grow in splenectomized recipients. Despite the absence of progressive tumor growth, residual tumor cells with malignant potential were found in the peripheral blood of the splenectomized animals. Splenectomy performed after injection of tumor cells but before the development of marked leukocytosis also prevented progressive tumor growth and death of the host.     Beschreibung: Two types of immature B cells, namely fetal liver hybridomas and the leukemic cell line 70Z/3, both of which have cytoplasmic mu chains but no light chains, were examined for DNA rearrangements of their light chain and heavy chain immunoglobulin genes. In the fetal liver hybridomas, which were constructed from fetal liver cells and a tumor cell, no light chain gene rearrangement was observed, whereas in the 70Z/3 cell line a kappa light chain rearrangement probably occurred.     Beschreibung: A single DNA fragment containing both mu and delta immunoglobulin heavy chain genes has been cloned from normal BALB/c mouse liver DNA with a new lambda phage vector Charon 28. The physical distance between the membrane terminal exon of mu and the first domain of delta is 2466 base pairs, with delta on the 3' side of mu. A single transcript could contain a variable region and both mu and delta constant regions.     Beschreibung: Contrast thresholds for sine-wave gratings of spatial frequencies of 2, 4, 12, and 16 cycles per degree were determined for normal and disabled readers at a range of stimulus durations. Normal readers demonstrated monotonically decreasing sensitivity with increasing spatial frequency at exposure durations between 40 and 100 milliseconds. At exposure durations of 150 to 1000 milliseconds, they showed peak sensitivity at 4 cycles per degree. In comparison, disabled readers showed monotonically decreasing sensitivity with increasing spatial frequency at all stimulus durations.     Beschreibung: A 200-fold inhibition in the titer of infectious vesicular stomatitis virus (VSV) was produced in cultures of Ly cells treated with 30 reference units of interferon per milliliter. Virus particle production, as measured by VSV particle-associated transcriptase, or nucleocapsid protein was inhibited by a maximum of tenfold. The glycoprotein and membrane protein content was reduced in VSV derived from interferon-treated cells. Thus interferon-treated cells may have produced VSV particles with low infectivity, which may be related to the reduced amount of glycoprotein incorporated into such particles.     Beschreibung: When unilamellar vesicles were administered subcutaneously in mice, the half-time for the destruction of the vesicles varied from 12 to 600 hours, depending on their composition. The vesicles tested consisted of distearoyl phosphatidylcholine, cholesterol, and certain sugar and amino-sugar derivatives of cholesterol. Vesicle with amino-sugar derivatives showed the greatest longevity and became localized with high specificity in aggregates of polymorphonuclear leukocytes.     Beschreibung: DNA containing the herpes simplex virus thymidine kinase (HSVtk) gene was used to transform wild-type tk+ mouse L cells to a tk++ status in vitro using methotrexate as a selective agent. HSVtk DNA was also used to transform mouse bone marrow cells in vitro.     Beschreibung: A 15,8-kilobase pair fragment of BALB/c mouse liver DNA, cloned in the Charon 4A lambda phage vector system, was shown to contain the mu heavy chain constant region (CHmu) gene for the mouse immunoglobulin M. In addition, this fragment of DNA contains at least two J genes, used to code for the carboxyl terminal portion of heavy chain variable regions. These genes are located in genomic DNA about eight kilobase pairs to the 5' side of the CHmu gene.     Beschreibung: The effects of a vitamin D deficiency on insulin and glucagon release was determined in the isolated perfused rat pancreas by radioimmunoassay of the secreted proteins. During a 30-minute period of perfusion with glucose and arginine, pancreases from vitamin D-deficient rats exhibited a 48 percent reduction in insulin secretion compared to that for pancreases from vitamin D-deficient rats that had been replenished with vitamin D. Vitamin D status had no effect on pancreatic glucagon secretion.     Beschreibung: Noradrenergic neurons in the hypothalamus involved in feeding and satiety are activated by gastrointestinal receptors. In the unrestrained rat, sites were first identified at which norepinephrine injected in the medial hypothalamus caused spontaneous feeding, or in the lateral hypothalamus caused no response. The activity of in vivo norepinephrine at these two sites was characterized by localized push-pull perfusion. When a nutrient was infused directly into the rat's duodenum, the synaptic release of hypothalamic norepinephrine was enhanced at lateral sites insensitive to norepinephrine, but suppressed at medial sites reactive to norepinephrine.     Beschreibung: The activity of cyanide-sensitive, Cu-Zn superoxide dismutase (SOD) was studied in liver sytosols from H-2 congenic strains of mice. Higher SOD activity was found in livers of mice having H-2b/A.BY, B10, and C3H.SW/haplotypes than in those of H-2a, H-2k and H-2d haplotypes. Segregation studies supported these correlations.     Beschreibung: In the hot plate test, substance P given intravenously at doses of 5 x 10-5 and 5 x 10-4 gram per kilogram caused analgesia, while lower doses caused hyperalgesia. The influence of substance P on nociception depended on the individual mouse's sensitivity to pain (control response latency). Analgesia was produced by substance P administered to mice with high sensitivity to thermic stimulation, whereas hyperalgesia occurred in mice whose control latencies were longer than normal.     Beschreibung: Hybrid cells formed between human lymphocytes and mouse myeloma cells produce human immunoglobulin in culture. Stable antibody-producing cell lines can be isolated after multiple cycles of low-density passage, cloning, and continued selection for immunoglobulin production. The origin and characteristics of a hybrid of human and mouse cells is described. This hybrid produces high concentrations (8.3 micrograms per milliliter) of human immunoglobulin M reactive with the terminal disaccharide of the Forssman glycolipid.     Beschreibung: The interplay of insulin, cortisol, and prolactin induces synthesis of casein and alpha-lactalbumin in cultured mammary explants from mature virgin mice. A striking difference has been found between the optimal concentrations of cortisol required for maximal induction of the two milk proteins in vitro: 3 x 10(-8) molar for alpha-lactalbumin and 3 x 10(-6) molar for casein. Moreover, 10(-7) to 10(-5) molar cortisol caused progressive inhibition of alpha-lactalbumin accumulation.     Beschreibung: A 15.0-kilobase (kb) Eco RI DNA fragment from normal mouse Balb/c genomic DNA that contains sequences (sarc) homologous to the acquired cell sequences (src) of Moloney sarcoma virus (MSV) has been cloned in phage lambda. The sarc region (1.2 to 1.3 kb) of the 15.0-kb cell fragment is indistinguishable from the src region of two isolates of MSV as judged by heteroduplex and restriction endonuclease analyses. The cellular sequences flanking sarc show no homology to other MSV sequences.     Beschreibung: The human genes for growth hormone (GH), chorionic somatomammotropin (CSH), and a third growth hormone-like gene (GHL) have been located on chromosome 17 in humans. DNA fragments of 2.6, 2.8, and 9.5 kilobase pairs containing GH, CSH, and GHL, respectively, were identified in human genomic DNA, and a 7.5-kilobase DNA fragment related to growth hormone DNA sequences was found in mouse cells.     Beschreibung: Naloxazone, a hydrazone derivative of the opiate antagonist naloxone, has a high affinity for opiate receptor binding sites. Naloxazone injections reduce opiate receptor binding to extensively washed mouse brain membranes for more than 24 hours, suggesting that the effect is irreversible. High-affinity binding sites are abolished by this treatment, whereas low-affinity sites are unaffected. Naloxazone treatment blocks the analgesic effects of morphine for at least 24 hours but does not prevent death from high doses of morphine.     Beschreibung: The growth of the MCF-7 human breast cancer cell line is unresponsive to the presence of estrogen in culture media. Paradoxically, in nude mice, growth of these cells and formation of solid tumors are dependent on estrogen. Tumors fail to develop in ovariectomized mice, but do develop in intact mice and in ovariectomized mice given estrogen. Primary cultures derived from MCF-7 tumors revert to unresponsiveness to estrogen. However, when these cultures are again transplanted into nude mice, estrogen is required for tumor formation. The continuous culture, the solid tumor, and the primary cultures therefrom have similar estrogen-binding capacities and affinities.     Beschreibung: Forty children were given a diet free of artificial food dyes and other additives for 5 days. Twenty of the children had been classified as hyperactive by scores on the Conners Rating Scale and were reported to have favorable responses to stimulant medication. A diagnosis of hyperactivity had been rejected in the other 20 children. Oral challenges with large doses (100 or 150 milligrams) of a blend of FD & C approved food dyes or placebo were administered on days 4 and 5 of the experiment.     Beschreibung: Glucan, a macrophage stimulant, was evaluated for its ability to alter survival and phagocytic dysfunction in mice challenged with mouse hepatitis virus strain MHV-A59. Administration of glucan before the mice were challenged with the virus significantly prolonged median survival time but did not modify overall mortality compared with control mice given dextrose. Maximal effectiveness was achieved when glucan was administered both before and after the viral challenge. In contrast to the marked hepatic parenchymal cell necrosis observed in the control mice, glucan-treated mice exhibited reduced pathology. Intraperitoneal administration of MHV-A59 resulted in a significant depression of phagocytic activity compared with controls that were not exposed to the virus. The enhancement in phagocytic function in glucan-treated control mice was unaltered in virus-challenged, glucan-treated mice. Thus glucan is capable of increasing survival, inhibiting hepatic necrosis, and maintaining an activated state of phagocytic activity in mice challenged with MHV-A59.     Beschreibung: In rats, multiple daily amphetamine injections (2.5 milligrams per kilogram of body weight, injected subcutaneously every 4 hours for 5 days) resulted in a progressive augmentation in response, characterized by a more rapid onset and an increased magnitude of stereotypy. By contrast, offset times of both the stereotypy and the poststereotypy hyperactivity periods were markedly shortened. When the animals were retested with the same dose of amphetamine 8 days after the long-term treatment was discontinued, the time of offset of the stereotypy and hyperactivity phases had recovered to values found with short-term amphetamine treatment, whereas the more rapid onset of stereotypy persisted. Brain monoamine and amphetamine concentrations and tyrosine hydroxylase activity were determined in comparably treated rats at times corresponding to the behavioral observations.     Beschreibung: Mice inoculated with herpes simplex virus (type 1) by the lip or corneal route and then passively immunized with rabbit antibody to herpes simplex virus developed a latent infection in the trigeminal ganglia within 96 hours. Neutralizing antibody to herpes simplex virus was cleared from the circulation and could not be detected in most of these mice after 2 months. Examination of ganglia from the antibody-negative mice revealed latent virus in over 90 percent of the animals, indicating that serum neutralizing antibody is not necessary to maintain the latent state. When the lips or corneas of these mice were traumatized, viral reactivation occurred in up to 90 percent of the mice, as demonstrated by the appearance of neutralizing antibody. Testosterone rather than 5 alpha-dihydrotestosterone appears to be the major intracellular androgen in organs other than skin and reproductive tract, but other steroid metabolites and their receptors are required to produce the diverse tissue differences observed in males and females. There is also evidence that multiple hormones from several endocrine glands are required to act in concert with androgens to produce and maintain their effects.     Beschreibung: Voltage-clamp recordings from mouse spinal neurons grown in culture were used to study the membrane current fluctuations induced by 12 substances structurally similar to gamma-aminobutyric acid (GABA). Fluctuation analysis provided estimates of the electrical properties of the elementary events underlying these responses. Estimates of the mean conductance of channels activated by all of the substances except glycine did not differ significantly from that estimated for GABA, whereas mean durations of agonist-activated channels all differed significantly from that found for GABA.     Beschreibung: The synthetic peptide NH2-Tyr-Pro-Phe-Pro-CONH2 (morphiceptin), which is the amide of a fragment of the milk protein beta-casein, has morphinelike activities and is highly specific for morphine (mu) receptors but not for enkephalin (delta) receptors. It is as active as morphine in the guinea pig ileum but much less active in the mouse and rat vas deferens. The discovery of this specific morphine receptor ligand substantiates the hypothesis of multiple opiate receptors.     Beschreibung: Cells of the homogeneous hybrid line neuroblastoma x glioma (NG108-15) have many neuronal properties. Immunocytochemical tests show that they contain both immunoreactive renin and angiotensin; direct radioimmunoassays show that they are positive for renin, angiotensin I, and angiotensin II; enzymatic assays show that they contain angiotensinogen and converting enzyme as well. The renin appears to be present in an enzymatically inactive form that can be activated by trypsin and then blocked by antiserum to purified mouse submaxillary renin. Renin concentration and activity are increased by enhancing cellular differentiation with dibutyryl cyclic adenosine monophosphate or by serum withdrawal.     Beschreibung: The effects of long- and short-term administration of the tricyclic antidepressant desipramine on intracranial self-stimulation in rats were studied with electrodes in the A10 region of the dopamine-containing cell bodies of the ventromedial tegmentum. Long-term desipramine administration resulted in a significant shift to the left in the ascending portion of the rate--current intensity function, indicating that the activity of the mesolimbic dopamine system was enhanced.     Beschreibung: Human malignant cancer tumors grafted into nude mice produce tumors containing both human cancer cells and the host's stromal cells. After short-term propagation of these tumors in vitro, the murine mesenchymal cells appear transformed and are tumorigenic in nude mice. However, established human cancer cell lines fail to similarly after adjacent murine stromal cells when used to produce tumors in nude mice.     Beschreibung: An electron microscopic and electrophysiological investigation was made of Merkel cell-neurite complexes in the sinus hair follicles of the cat. These mechanoreceptors respond with very precise phase locking to heavy-frequency vibratory stimuli as well as to static hair displacements. The mechanoelectric transduction process is faster than that known for any other somatic mechanoreceptor.     Beschreibung: Voltage clamp studies of macrophages from cultures of mouse spleen macrophages produced N-shaped steady-state current-voltage curves containing a region of negative slope resistance. Some macrophages exhibit two stable states of membrane potential, having current-voltage relationships that cross the voltage axis at three points. Outward currents that turn on at voltages of +15 millivolts or greater were noted in several cells. The addition of barium chloride to the bathing medium abolished the negative slope resistance and reduced the inward currents in response to hyperpolarizing voltage steps.     Beschreibung: A single injection of the melanotropin analog [4-norleucine, 7-D-phenylalanine]-alpha-melanotropin into frogs (Rana pipiens) caused near maximum darkening of the skins of the frogs for at least 6 weeks. Injections of the natural hormone alpha-melanotropin or of the analog [Nle4]-alpha-melanotropin also caused darkening, but this effect lasted only a few days. Morphological examination of the skins of frogs injected with [Nle4, D-Phe7]-alpha-melanotropin revealed that both dermal and epidermal melanophores were dispersed during the entire 6-week period. In vitro [Nle4, D-Phe7]-alpha-melanotropin also causes prolonged darkening of the skin of the lizard Anolis carolinensis. In the absence of the melanotropin, skins previously darkened with the analog could be lightened by removal of calcium from the incubation medium but could then be redarkened by adding calcium. The cycle could be repeated indefinitely without addition of melanotropin.     Beschreibung: A loss in the number of functional, sodium ion-dependent, high-affinity choline transport sites was observed in the cortex and hippocampus of mice given an intracerebroventricular injection of 65 nanomoles of AF64A (ethylcholine mustard aziridinium ion) 3 days earlier. Such an effect was not observed in the striatum. This effect of AF64A represents a long-term neurochemical deficit at cholinergic nerve terminals in some brain regions which can lead to a persistent deficiency in central cholinergic transmission.     Beschreibung: Analogs of adenosine 3',5'-monophosphate (cyclic AMP) inhibit the growth of cultured cell lines. The effects of 8-bromo- and N6-butyryl-substituted analogs of cyclic and noncyclic AMP on six cell lines were examined and were equally inhibitory. Variant cell lines with altered cyclic AMP-dependent protein kinase were more resistant to both cyclic and noncyclic nucleotides.     Beschreibung: A single application of electroconvulsive shock produced a rapid but short-lasting increase in tyrosine hydroxylase activity above control values in the rat adrenal medulla and striatum. After repeated electroconvulsive shock treatment (once per day for 7 days), tyrosine hydroxylase activity increased significantly in the locus ceruleus, nucleus of the tractus solitarius, hippocampus, cerebellum, and frontal cortex and remained elevated for 4 to 8 days. Adrenal tyrosine hydroxylase activity increased 1 day after the termination of repeated electroconvulsive shock treatments and remained elevated for at least 24 days, possibly reflecting the establishment of a new and higher steady-state level of catecholamine biosynthesis in the adrenal.     Beschreibung: The proteins of the three major rate components of axonal transport in guinea pig retinal ganglion cells were analyzed by one- and two-dimensional gel electrophoresis. Each rate component consisted of a different set of proteins that remained associated with each other during transport. This suggests that each rate component represents a distinct macromolecular complex and that these complexes may be definable organelles such as microtubules, microfilaments, and smooth endoplasmic reticulum.     Beschreibung: An intrinsic birefringence signal with two components occurring before sarcomere shortening was measured in mammalian cardiac muscle. The second component was sensitive to the inotropic state of the muscle as affected by external calcium concentration and epinephrine but not by changes of resting length. The second component was absent in frog heart.     Beschreibung: Thirty minutes after inoculation of reovirus type 1 into the intestinal lumen of the mouse, viruses were found adhering to the surface of intestinal M cells but not other epithelial cells. Within 1 hour, viruses were seen in the M cell cytoplasm and were associated with mononuclear cells in the intercellular space adjacent to the M cell.     Beschreibung: Growth of mouse lymphoma L5178Y, which contains large quantitites of the gangliotriosylceramide (GgOs3Cer), in DBA/2 mice was suppressed by passive immunization with monoclonal immunoglobulin G3 antibodies to GgOS3Cer, but not by immunoglobulin M antibodies with or without added complement. Most groups of mice treated with monoclonal immunoglobulin G3 antibodies did not develop tumors, but the tumor that appeared in a treated animal had a much lower amount of the GgOS3Cer than the cells used for inoculation.     Beschreibung: Female athymic nude mice and their phenotypically normal littermates were exposed transplacentally to ethylnitrosourea. Skin tumors (papillomas and sebaceous adenomas) developed on the nude mice with an almost tenfold greater incidence than on their haired littermates. Skin tumors were also induced on nude mice but not haired controls by direct intraperitoneal treatment with ethylnitrosourea. Control hamsters received water only.     Beschreibung: Mice will replace the tip of a foretoe when it is amputated distal to the last interphalangeal joint. Amputation of the digit more proximal to the joint does not result in regrowth of the foretoe. Though this growth shares certain similarities with the epimorphic regeneration of amphibian limbs, the two processes are not the same.     Beschreibung: Brief tetanic stimulation of the preganglionic nerves to the superior cervical ganglion enhances the postganglionic response to single preganglionic stimuli for 1 to 3 hours. This long-term potentiation of transmission through the ganglion is apparently not attributable to a persistent muscarinic action of the preganglionic neurotransmitter, acetylcholine, since neither the magnitude nor the time course of the phenomenon is reduced by atropine. The decay of long-term potentiation can be described by a first-order kinetic process with a mean time constant of 80 minutes. We conclude that long-term potentiation, once considered a unique property of the hippocampus, is in fact a more general feature of synaptic function.     Beschreibung: The messenger RNA for a beta-crystallin polypeptide with a molecular size of 27 kilodaltons, first detected 5 to 10 days after birth in the normal mouse lens and the Nakano mouse cataract, was not detected in the Philly mouse cataract with translation in vitro. The heterozygous Philly lens had intermediate levels of the 27-kilodalton beta-crystallin polypeptide and exhibited delayed onset of the cataract. Mouse blastocysts grown in vitro for 10 days showed blood circulation in the yilk sac, forelimb buds, and the primordia of liver, pancreas, and lungs. These characteristics are indicative of a developmental stage equivalent to one-half of the total gestation period in utero.     Beschreibung: Receptors for maleylated or acetylated proteins as well as for alpha-2-macroglobulin-protease complexes on macrophages serve as scavengers by mediating the uptake of macromolecules from the extracellular compartment. Described in this report is a novel function of these receptors on macrophages: regulation of neutral protease secretion. The binding of maleylated bovine serum albumin to macrophages triggered secretion of three neutral proteases: neutral caseinases, plasminogen activator, and cytolytic proteinase. Release of acid phosphatase, however, was not induced. An important biological consequence of protease secretion by macrophages, tumor-cytolysis, was also triggered by engagement of the receptor for maleylated bovine serum albumin. By contrast, the binding of alpha-2-macroglobulin-protease complexes to the macrophages suppressed secretion of all three proteases.     Beschreibung: Alkyl-Substituted gamma-butyrolactones were synthesized and tested for their convulsant and anticonvulsant actions in mice and guinea pigs. The alpha-substituted compounds, alpha, alpha-dimethyl-, and alpha-ethyl-alpha-methyl-gamma-butyrolactone were anticonvulsant compounds with a spectrum of activity similar to that of ethosuximide. In contrast, beta-substituted compounds were convulsant agents similar to picrotoxinin.     Beschreibung: The 14S messenger RNA (1300 to 1500 nucleotides) for the alpha A chain of alpha-crystallin of the mammalian lens is nearly three times larger than required to code for the polypeptide that contains 173 amino acids. As a means of accounting for this anomaly, a complementary DNA clone for the mouse alpha A-crystallin messenger RNA was constructed in pBR322 and sequenced. Derivation of the protein sequence from the nucleic acid sequence showed that mouse alpha A-crystallin is similar to that of other organisms. The messenger RNA contains 536 nucleotides located on the 3' side of the coding region, excluding the polyadenylate stretch.     Beschreibung: Murine B16 melanoma cells were adapted for lung survival and growth by allowing them to attach to Bio-Carrier beads and injecting the beads intravenously into normal mice. The beads lodged mechanically in the microcirculation of the lung. When the melanoma cells had grown into visible tumors from the arrested beads, the tumors were removed and the cells were dispersed, cultured to remove normal cells, and reattached to new beads. The process was repeated nine times. Previously another B16 subline was injected intravenously as a suspension of separate tumor cells. Those cells that survived and colonized the lungs were harvested, cultured, and injected again. This selection process was also repeated nine times.     Beschreibung: Protein degradation within retinal ganglion cell axons in vitro is 50 to 110 percent faster than normal in mutant mice exhibiting deficiencies of myelin in the central nervous system. Proteolysis is increased proximally and distally within retinal ganglion cell axons of mice carrying the jumpy mutation or its allele, myelin synthesis deficiency, and is increased distally within those axons of quaking mice. The proteolytic defect is axon (neuron)-specific since the rate of protein degradation within glial cells is normal. Increased axonal proteolysis does not bear a simple relation to hypomyelination since shiverer, another mouse mutant deficient in central myelin, displayed normal rates of axonal protein degradation under the same conditions.     Beschreibung: The size of the gene pool potentially encoding antibodies to p-azophenyl arsonate has been examined. A heavy chain-specific full-length complementary DNA clone has been constructed with the use of messenger RNA from a hybridoma that produces antibodies to the arsonate hapten and bears nearly a full complement of the determinants comprising the cross-reactive idiotype (CRI). The sequences of both the complementary DNA clone and the corresponding immunoglobulin heavy chain have been independently determined. A probe for the variable region gene was prepared from the original heavy chain complementary DNA clone and used to analyze, by Southern filter hybridization, genomic DNA from both A/J (CRI positive) and BALB/c (CRI negative) mice.     Beschreibung: A cultured cell line of the K-1735 melanoma was x-irradiated to induce chromosome breakage and rearrangements and then was implanted into the footpads of syngenic C3H mice. Spontaneous lung metastases were isolated from different animals, established in culture as individual lines, and then karyotyped. Within certain metastases, the same chromosomal abnormality (or abnormalities) (recombinant chromosomes) was found in all the cells examined. Most metastases differed from one another in that they exhibited characteristic combinations of chromosomal markers.     Beschreibung: alpha-Difluoromethylornithine is an enzyme-activated irreversible inhibitor of ornithine decarboxylase that forms a covalent bond with the enzyme. When alpha-[5-14C]difluoromethylornithine was administered to androgen-treated mice, only ornithine decarboxylase became labeled. Autoradiographic examination of kidney, liver, and brain indicated much more extensive incorporation of labeled difluoromethylornithine into kidney protein than into the protein of the other tissues. Such incorporation was greatly reduced by prior treatment of the mice with cycloheximide. These results correlate with the presence of ornithine decarboxylase activity which is much higher in the kidney than in the other tissues and is lost rapidly when protein synthesis is inhibited. The binding of this drug in vivo, therefore, is useful for determining the distribution of ornithine decarboxylase.     Beschreibung: The ability of antiserum against murine L1210 leukemia to remove residual leukemia cells from murine bone marrow was investigated. Leukemic marrow was treated in vitro with antiserum and complement and used to hematologically reconstitute mice that had been irradiated with doses lethal to bone marrow. Following infusion of treated leukemic marrow, normal marrow returned without evidence of leukemia.     Beschreibung: Breast-feeding is important to infant nutrition, morbidity, and mortality, and to postpartum amenorrhea (hence to birth intervals). Evidence on breast-feeding patterns in low-income countries from nationally representative World Fertility Surveys and secondary sources shows that in all but a few such countries most children are breast-fed for at least a few months. The limited evidence available on trends seems to indicate a decline in the duration of breast-feeding, but in most of Asia and Africa breast-feeding is almost universal during at least the first 6 months.     Beschreibung: Large numbers of granulocytes can be collected repeatedly from the supernatant medium of long-term cultures of mouse bone marrow cells. A constant relationship was found between the number of adherent hematopoietic stem cells and the lifetime cell production per culture. The data indicate that there is a limit to the proliferative capacity of normal and of irradiated stem cells.
