    Notizen: Abstract The five synthetic pheromone components of the West Indian sugarcane borer (WISB), Metamasius hemipterus (L.) (Coleoptera, Curculionidae) were tested in the field. The combination of sugarcane (SC) and the major pheromone compound, 4-methyl-5-nonanol (1) was attractive. However, the addition of 2-methyl-4-heptanol (2) or 2-methyl-4-octanol (3) was required to reach high catch levels while 5-nonanol (4) or 3-hydroxy-4-methyl-5-nonanone (5) did not enhance WISB attraction. The redundancy phenomenon, here reported for the first time in rhynchophorinous species, was observed between compounds 2 and 3. SC +1 +3 was more attractive than living male baits, however, the sex-ratio of the catches was equivalent between both treatments. The sex-ratio of catches was affected by the qualitative composition of the pheromone formulation. Compound 3 had a sexual role, attracting more females while 5 seemed to play an aggregation role, luring both sexes in the same proportion.

    Notizen: Abstract Promising diets were screened and the most successful modified with additives used in artificial diets for other entomophagous insects. Two suitable diets were obtained, one for adults and one for larvae ofChilocorus nigritus (Fabricius) (Coleoptera: Coccinellidae). They were still inferior to natural prey and not adequate as the sole food source for rearing consecutive generations. They are valuable as substitute food in the insectary during shortages of natural prey. Oleander scaleAspidiotus nerii Bouché andAsterolecanium miliaris (Boisduval) were evaluated as natural prey forC. nigritus and two other potential biocontrol agents in southern Africa,C. bipustulatus (Linnaeus) andC. infernalis Mulsant.A. nerii andA. miliaris were suitable for all life stages ofC. nigritus and adults ofC. bipustulatus andC. infernalis. A. miliaris was inadequate for larvae ofC. bipustulatus andC. infernalis.

    Notizen: Abstract We studied preference for willows along a pollution gradient on the Kola Peninsula, Russia, by the leaf beetle, Melasoma lapponica. Multiple tests with leaf disks demonstrated low preference for Salix borealis, S. caprea and S. phylicifolia from the plot situated 14 km from the smelter, in comparison with conspecific plants from plots situated at 1 and 29 km distances. This pattern was observed when testing beetles orginating from any plot both in 1993 and 1994, using both young and mature leaves of S. borealis. Although fumigation of S. borealis with realistic SO2 concentration (100 μg/m3) increased plant palatability, preference for plants from our study plots did not correlate with plot-specific mean SO2 concentrations. Furthermore, no correlation with foliar concentrations of the main metal pollutants (Ni and Cu) was found. Palatability of plants was negatively correlated with population density of M. lapponica, which peaked in the moderately polluted plot 14 km from the smelter. Within this plot, beetles clearly preferred non-damaged bushes of S. borealis to previously damaged bushes. We therefore conclude that low preference of S. borealis from the moderately polluted area was caused by plant resistance induced by severe damage from M. lapponica in previous years rather than by pollution impact. However, S. caprea and S. phylicifolia had little damage from M. lapponica, and low palatability of these species in the moderately polluted plot suggests changes in plant quality similar to changes in heavily damaged bushes of S. borealis.

The behaviour of different beetle species in arenas containing millet was monitored over 24 h and the frequency of encounters with the trap rim resulting in capture recorded. The capture efficiency of four types of pitfall traps (i.e. polystyrene, polythene, glass and tin-plated steel can) with rims exposed or submerged below the millet surface level were compared. Capture was related to beetle size, locomotory rate, and beetle behaviour at the trap rim as well as trap design and placement. The lighter and smaller species were least captured. Glass jars were more effective than plastic and metal containers. Traps placed with their rims submerged below the grain surface level were more efficient than those with rims exposed. Capture rate was unrelated to trap size. The frequency of encounters with trap rims was not correlated with capture rate. Three types of avoidance behaviours at the trap rims i.e. probing, skirting and spontaneous retreat, were related to capture rate, spontaneous retreat being the most effective escape mechanism and probing least.

    Notizen: Abstract We used artificial selection experiments to study genetic allocation costs and physiological mechanisms of resistance to herbivory and fungal disease. Genetic costs to resistance were present in some instances and absent in others. Genetic resistance to the fungal pathogen, Leptosphaeria maculans was cost-free, while resistance to Peronospora parasitica showed a negative genetic correlation between disease resistance and growth rate. Leptosphaeria resistant genotypes had 13% higher chitinase activity. Genetic increases in myrosinase activity were correlated with increased resistance to flea beetles (Phyllotreta cruciferae), but resulted in lower plant fecundity, presumably due to production costs of myrosinase. Genetic costs of resistance may maintain genetic variation in natural plant populations. These studies demonstrate the predictive and explanatory power of a functional approach to plant-herbivore and plant-pathogen interactions.

    Notizen: Abstract The effects of host plant and dietary cucurbitacin on the growth of larval southern corn rootworm (SCR), Diabrotica undecimpunctata howardi Barber (Chrysomelidae: Luperini), were investigated. SCR were reared on four hosts: corn, Zea mays; peanuts, Arachis hypogaea; and two squash varieties, Cucurbita pepo cv. Ambassador (containing cucurbitacin D (0.08 mg g−1 fr.wt.) = bitter), and C. pepo cv. Early Yellow Crookneck (lacking cucurbitacin = non-bitter). Larval growth was significantly greater on corn and peanuts than on either squash variety. After four weeks, adults had emerged from corn and peanut plants, while squash-reared larvae had not yet entered the pupal stage. There was no difference in larval growth on the two varieties of squash. Primary metabolite measurements showed no nutritional differences between the two squash varieties. Artificial diet experiments were used to test the effect of three concentrations of cucurbitacin D (0.0, 0.1, and 0.6 mg g−1 diet) on growth of larval SCR. Larvae reared on diet containing 0.6 mg g−1 cucurbitacin weighed significantly less than larvae reared on diet containing 0.1 mg g−1 or no cucurbitacin after 10 d. No significant difference in growth was measured between the 0.1 mg g−1 diet and the 0.0 mg g−1 diet. Results are discussed relative to theories about the relationship between diabroticites and cucurbitacins.

    Notizen: Abstract Individual pairs of overwintered adult apple blossom weevils, Anthonomus pomorum (L.), confined with apple twigs under different ambient temperatures in the laboratory and on apple trees in the field, were observed through day and night for their spring activities. Flight behavior in relation to ambient temperature was also investigated under laboratory conditions using flight stands. Both sexes displayed predominantly nocturnal behavior patterns in both the laboratory and the field. Feeding, crawling, and mating activities increased following sunset in the field or onset of scotophase in the laboratory while resting occurred most frequently during daylight hours. Results of the laboratory experiments showed that temperature affected significantly the activity patterns. The diel pattern of activities became less distinctive at higher temperatures (above 15°C), and total activities in crawling, feeding, and mating were suppressed significantly at lower temperatures (below 5°C). Over 97% of the test weevils initiated take-off response from flight stands at 20°C within the 30 min trial period; however, flight initiation rarely occurred at temperatures 12°C or below. Overall, results of the laboratory and field experiments indicate that A. pomorum is a remarkably cold-adapted insect with ability to crawl, feed, and mate at a few degrees above freezing, a physiological attribute necessary for the exploitation of early stages of apple bud development in the cold early spring.

    Notizen: Abstract Snowdrop and wheatgerm lectins were found to be insecticidal and growth inhibiting dietary proteins for larvae of the sugarcane whitegrub Antitrogus parvulus. At concentrations as low as 0.5 mg of snowdrop lectin per gram of semi-artificial diet, growth was inhibited by 21 days of feeding and significant mortality was apparent by 28 days. Wheatgerm lectin was active at similar concentrations, although expression of the effects was slower. Avidin was found to be a growth inhibiting dietary protein for larvae of Antitrogus consanguineus. At levels as low as 0.01 mg g-1 of diet, growth was inhibited by 28 days of feeding. Avidin caused no significant mortality after 35 days of feeding. Snowdrop and wheatgerm lectins and avidin are insect growth-inhibiting proteins whose genes potentially could be manipulated into sugarcane and improve host-plant resistance to whitegrubs.

    Notizen: Abstract Higher flight activity has been observed in aged, high-density cultures ofProstephanus truncatus (Horn) (Coleoptera: Bostrichidae), but adults in new, lowdensity culture jars showed less flight activity. In order to understand this change in behavior, the effects of population density, age, resource quality, and sex on the flight ofP. truncatus were studied in a wind tunnel. While an immediate density on the release platform had no significant effect on flight, beetles from high-density cultures were more inclined to fly than those from low-density cultures. Resource quality exerted a major influence on flight; insects in food suitable for boring and oviposition seldomly exhibited flight, however, when food was absent or of inferior quality for boring and oviposition, the dominant behavior was flight. Also, insects maintained for a week in food suitable for boring and oviposition were less ready to fly than those maintained in food unsuitable for boring and oviposition. The optimum age range for flight activity was before the peak of reproduction and insects rarely flew before 4 days or after 32 days of emergence. There were no significant differences between the flight activity of males and that of females. Based on these results, we conclude that age and resource quality are major influences on the flight activity ofP. truncatus and a hypothesis is proposed in which reproductively active male and female beetles disperse from habitats of low resource quality to those that support their reproductive behavior. The practical implications of these results and the possible role of the male-produced aggregation pheromone are discussed.

    Notizen: Abstract The behavioural responses ofCryptolestes pusillus Oryzaephilus surinamensis andProstephanus truncatus to different doses of carob extract were assessed in a two-choice pitfall bioassay and in plastic grain probe traps. The extract evoked a quick directional response and induced high beetle attraction to treated pitfall arenas compared with the controls. Overall, the extract improved probe trap efficiency by about 50% compared to unbaited probes. Beetle response was dose-dependent with the medium dose of 10 μl probably eliciting optimum response.

    Notizen: Abstract Cantharidin, a potent defensive chemical, is present in all ten life stages of the blister beetleEpicauta funebris. The first five larval stages accumulate cantharidin as they feed and grow in size. When disturbed, they exude cantharidin in a milky oral fluid, not in hemolymph which adult beetles reflexively discharge from leg joints. Two subsequent larval stages and the pupa do not feed, grow, regurgitate, or change in their defensive reserves (110 μg cantharidin/insect, regardless of sex). Adult beetles kept in isolation for 60–90 d exhibit a pronounced sexual dimorphism in cantharidin production: the male biosynthesizes about 17 mg of the toxin, representing 10% of his live weight, whereas the female actually loses most of her defensive reserves. But in the wild a female beetle repeatedly acquires cantharidin as copulatory gifts from her mates.

    Notizen: Abstract A pot experiment was conducted to compare the uptake and dry matter production potential of NH+ 4 and NO– 3 and to study the effect of Baythroid, a contact poison for several insect pests of agricultural crops, on growth and N uptake of maize (Zea mays L.). Nitrogen was applied as (15NH4)2SO4, K15NO3, or 15NH4NO3 and in one treatment Baythroid was combined with 15NH4NO3. Source of N had, in general, a nonsignificant effect on dry matter and N yield, but uptake of NO– 3 was significantly higher than that of NH+ 4 when both N sources were applied together. Substantial loss of N occurred from both the sources, with NH+ 4 showing greater losses. Baythroid was found to have a significant positive effect on dry matter yield of both root and shoot; N yield also increased significantly. Uptake of N from both the applied and native sources increased significantly in the presence of Baythroid and a substantial added nitrogen interaction (ANI) was determined. The positive effect of Baythroid was attributed to: (1) a prolonged availability of NH+ 4 due to inhibition of nitrification, (2) an increased availability of native soil N through enhanced mineralization, and (3) an enhanced root proliferation.

Rotations were: IA, cotton (Gossypium hirsutum L.) grown every year without a winter legume and without N fertilization; IB, cotton grown every year with a winter legume and without N fertilization; IC, cotton grown every year without a winter legume and with 134 kg N as NH4NO3 ha-1 year-1; IIA, 2-year cotton-corn (Zea mays L.) rotation with a winter legume and without N fertilization; IIB, 2-year cotton-corn rotation with a winter legume and with 134 kg N ha-1 year-1 as NH4NO3; and III, 3-year cotton-corn- alternating soybean [Glycine max (L.) Merr.] or rye (Secale cereale L.) rotation with a winter legume and with 134 g N as NH4NO3 ha-1 year-1. Crimson clover (Trifolium incarnatum L.) was the winter legume cover crop. The 2-year cotton-corn rotation with a winter legume and with 134 kg N ha-1 year-1 (IIB) and the 3-year cotton-corn soybean/rye rotation with a winter legume and with 134 kg N ha-1 year-1 (III) had higher amounts of soil organic matter, soil microbial biomass C and crop yield than the other four treatments. The cotton grown every year without a winter legume or N fertilizer (IA) had a lower amount of soil organic matter, soil microbial biomass C and N and cotton seed yield than all other rotations. In 1988 and 1992 cotton seed and legume yield were correlated in positive, curvilinear relationships with soil organic matter (r 2 ranged from 0.72 to 0.87). In most months, soil microbial biomass C and N was lower in the cotton grown every year without winter legumes or fertilizer (IA) than the other five rotations. In 1994, microbial biomass C and the Cmic:Corg ratio correlated in positive, curvilinear relationships with seed cotton yield (r 2=0.87 and 0.98, respectively). After 99 years of management the “Old Rotation” cotton experiment indicates that winter legumes increase amounts of both C and N in soil, which ultimately contribute to higher cotton yields. Microbial biomass C and the Cmic:Corg ratio are poor predictors of annual crop yield but may be an accurate indicator of soil health and a good predictor of long-term crop yield.

    Notizen: Abstract A pot experiment was conducted to compare the uptake and dry matter production potential of NH inf4 sup+ and NO inf3 sup- and to study the effect of Baythroid, a contact poison for several insect pests of agricultural crops, on growth and N uptake of maize (Zea mays L.). Nitrogen was applied as (15NH4)2SO4, K15NO3, or 15NH4NO3 and in one treatment Baythroid was combined with 15NH4NO3. Source of N had, in general, a nonsignificant effect on dry matter and N yield, but uptake of NO inf3 sup- was significantly higher than that of NH inf4 sup+ when both N sources were applied together. Substantial loss of N occurred from both the sources, with NH inf4 sup+ showing greater losses. Baythroid was found to have a significant positive effect on dry matter yield of both root and shoot; N yield also increased significantly. Uptake of N from both the applied and native sources increased significantly in the presence of Baythroid and a substantial added nitrogen interaction (ANI) was determined. The positive effect of Baythroid was attributed to: (1) a prolonged availability of NH inf4 sup+ due to inhibition of nitrification, (2) an increased availability of native soil N through enhanced mineralization, and (3) an enhanced root proliferation.

    Notizen: Summary In bagged and unbagged shoot experiments, we investigated the survival and growth rate of first instar larvae ofChrysophtharta bimaculata on 9 families of a natural host,Eucalyptus regnans. Families used had been previously assessed as being either of low or high susceptibility toC. bimaculata damage. In conjunction with larval experiments, we measured 24 tree and leaf characteristics (including foliar elemental concentrations, foliar terpenes, leaf toughness and tree growth rates) and attempted to correlate the plant characters measured with differences in larval performance and previous scorings ofE. regnans family susceptibility. First instar larval growth and survival did not differ significantly across families or between low and high susceptibility family groups (=susceptibility classes), although survival was significantly greater in bagged than unbagged treatments. As predators were in low abundance at the study site, we attribute higher survival rates of larvae in bagged treatments to increased protection from adverse weather conditions. Only one plant character measured, an unidentified foliar phlorglucinol, was significantly negatively correlated with larval survival. Of the 24 plant characters measured, 11 were significantly different between families and 10 were signficantly different between susceptibility classes. Only 4 plant characters were significantly different at both the family and susceptibility class levels;viz. proportion red leaves, tree height at end of season, trunk volume at end of season and relative growth rate based on tree height. Principle Component Analysis using all plant characters measured, or subsets of them, could not separate individual families or susceptibility classes. Our results suggest that herbivore resistance mechanisms inE. regnans do not affectC. bimaculata larvae, but may influence adult feeding and/or oviposition.

The ratio of these solvents is species-specific. Application experiments using some of the natural insect predators (L. riparia, C. bicolor, Pogonus, Di. gustavii, Dyschirius) revealed that these solvent ratios provided a more effective deterrent than other possible ratios. Thus by combining the solvents in certain ratios, the capability of cuticular penetration and therefore the effectiveness of the defensive secretions are adapted to their natural targets.

They have paired pygidial glands with a secretion that contains high molecular weight substances rendering them toxic for predatory fish. In this paper we report on a laboratory study on volatile components released by three different gyrinid species when irritated. The volatile pattern had a clear difference between the different species.Gyrinus substriatus andG. aeratus, both produced 3-methyl-1-butanal and 3-methyl-1-butanol when irritated, but in quite different amounts. The third tested species,G. minutus, did not produce any substances above the detection level.     Notizen: Abstract Mounting evidence suggests that heat-shock proteins (HSPs) play a vital role in enhancing survival at high temperature. There is, however, considerable variation in patterns of HSP production among species, and even among and within individuals of a species. It is not known why this variation exists and to what extent variation in HSPs among organisms might be related to differences in thermotolerance. One possibility is that production of HSPs confers costs and natural selection has worked towards optimizing the cost-to-benefits of HSP synthesis and accumulation. However, the costs of this production have not been determined. If HSP production confers significant nitrogen (N) costs, then we reasoned that plants grown under low-N conditions might accumulate less HSP than high-N plants. Furthermore, if HSPs are related to thermotolerance, then variation in HSPs induced by N (or other factors) might correlate with variation in thermotolerance, here measured as short-term effects of heat stress on net CO2 assimilation and photosystem II (PSII) function. To test these predictions, we grew individuals of a single variety of corn (Zea mays L.) under different N levels and then exposed the plants to acute heat stress. We found that: (1) high-N plants produced greater amounts of mitochondrial Hsp60 and chloroplastic Hsp24 per unit protein than their low-N counterparts; and (2) patterns of HSP production were related to PSII efficiency, as measured by F v/F m. Thus, our results indicate that N availability influences HSP production in higher plants suggesting that HSP production might be resource-limited, and that among other benefits, chloroplast HSPs (e.g., Hsp24) may in some way limit damage to PSII function during heat stress.

    Notizen: Abstract The natural host of Ophraella notulata is Iva frutescens (Asteraceae); its close relative feeds on a related plant, Ambrosia artemisiifolia. We reared beetles on both plants, obtained progeny from the four possible crosses (two sexes X two parental hosts), and reared the progeny on both plant species. Survival to the imaginal stage of progeny reared on Iva varied with both maternal and paternal host. Hatchling feeding response to both plants showed a maternal host X paternal host interaction. Consumption of Ambrosia by adult beetles was, counter to expectation, higher for progeny of Iva-reared males than Ambrosia-reared males. Oviposition response, although based on too few data to be definitive, was peculiar: parental host did not affect oviposition on Ambrosia; on Iva daughters of Iva-reared males laid significantly more eggs than did daughters of Ambrosia-reared males, but only if they had been reared on Iva; those reared on Ambrosia displayed the reverse pattern. We discuss the possibility that nongenetic paternal transmission of host plant effects may explain these results, but offer a somewhat uncomfortable hypothesis of selection as a preferable explanation. An important outcome of the experiment is that it provided no evidence of maternal effects of host plant on offspring feeding or oviposition.

    Notizen: Summary Sperm cells are thought to be quiescent in pollen and activated upon pollen germination. To test this hypothesis, protein, RNA and DNA synthesis were assessed in Zea mays sperm cells at different times after isolation from pollen. Protein synthesis changed with time; while some proteins were found to be constitutive in both 0 and 24 h cells, others were synthesized and some disappeared. Overall, the number of proteins detected at 24 h doubled compared with freshly isolated cells. Incorporation of [3H]leucine in 24 h cells was about 50 times that in freshly isolated cells, and that of [5, 6-3H]uridine, about 7 times. Very low incorporation of [6-3H]thymidine into the cells was detected; there was no difference between freshly isolated and 24 h cells. It is possible that the differences in synthetic activity between freshly isolated and 24-h-old cells might correspond to sperm cell activation during pollen tube growth. If so, these metabolic changes may play an important role in fertilization.

    Notizen: Summary An in vitro method to simulate pollen development was developed in maize (Zea mays L.). Microspores at the late uninucleate to early binucleate stage were isolated and cultured under various conditions. Cell viability, starch content and the formation of the three nuclei as found in normal mature pollen were monitored during the course of the culture. Media composition was modified in order to promote starch accumulation and frequency of mitosis, while maintaining the viability of the microspores. Under the best conditions, up to 12% of the microspores matured in vitro into trinucleate, starch-filled viable pollen grains which were unable to germinate or produce seeds. At different stages during in vitro maturation, proteins patterns were analyzed and compared with their in vivo equivalent and the patterns were only partially similar.

Bombarded tassels were also cultured in vitro and GUS activity was detected in the vascular tissue of mature anthers that developed within 4 weeks. The ploidy level of the calli was then evaluated using flow cytometry, at different times after the treatment. Untreated haploid calli did not change in ploidy level for 97 days but by 466 days, there were up to 50% diploid or higher ploidy cells thus showing that spontaneous doubling may occur during corn calli subculture with this genotype. Pronamide treatment did increase the percentage of diploid and tetraploid cells and by 466 days, all of the lines showed an additional change toward higher ploidy levels. This change may be due to spontaneous chromosome doubling or to differential cell cycle times of cells with different ploidy levels. The ploidy level of plants regenerated from the cultures was determined by counting the guard cell chloroplast numbers and the correlation with the ploidy level of the cultures was r2=0.84. These studies show that pronamide treatments can increase haploid maize callus chromosome numbers and that spontaneous chromosome doubling can occur with time in maize callus.

    Notizen: Summary Vesicular-arbuscular mycorrhizal (VAM) fungi affect diverse aspects of plant form and function. Since mycorrhiza-mediated changes in host-plant responses to root colonization by different VAM fungi vary widely, it is important to assess each endophyte for each specific effect it can elicit from its host as part of the screening process for effectiveness. Three species of VAM fungi and a mixture of species were compared with non-VAM controls for their effects on soil organic matter contents and on nutrition and morphology in two varieties (native and hybrid) of corn (Zea mays L.) and one of sunflower (Helianthus annuus L.) in P-sufficient and N-deficient soil in pot cultures. Differences in soil organic matter due to the fungal applications were highly significant with all host plants. Native corn responded more to VAM colonization than the hybrid did; differences in treatments were significant in leaf area, plant biomass, and root: shoot ratio in the former, but not in the latter. Responses in the sunflower were similar to those in the native corn. Significant VAM treatment-related differences in shoot N and P contents were not reflected in shoot biomass, which was invariant. Correlations between plant or soil parameters and the intensity of VAM colonization were found only in soil organic matter with the native corn, in specific leaf area in the hybrid corn, and in plant biomass in the sunflower. The presence of the different endophytes and not the intensity of colonization apparently elicited different host responses.

    Notizen: Summary Vesicular-arbuscular mycorrhizal (VAM) fungi improve plant growth in marginal soils. This study was conducted to determine the effects of three species of VAM fungi on plant nutrition in two cultivars of corn (Zea mays L.) and one of sunflower (Helianthus annus L.). Plants were grown in pot cultures under controlled (greenhouse) conditions in a soil high in K, Mg, and P, and low in Ca and N, and were supplied with amounts of VAM-fungal inocula in which equal numbers of infective propagules had previously been determined. Analysis of variance showed highly significant main effects and interactions due to both factors (plant and fungus) for N, P, Ca, and Mg. For K, only plant effects were significant (P〈0.043). The uptake of nutrients was selectively enhanced or inhibited by one or the other VAM fungus relative to non-VAM control plants. In sunflower, N concentration was markedly enhanced (73%) by the mixed inoculum of the three fungi, even though individual effects were not significant. Evaluation of leaf nutrient analyses by the Diagnosis and Recommendation Integrated System (DRIS) revealed the utility of this system to rank nutritional effects by VAM fungi in an order of relative nutrient deficiency. The DRIS therefore is seen as a useful tool in evaluating and selecting VAM fungi for the alleviation of specific nutrient disorders.

    Notizen: Summary The application of NH inf4 su+ -based fertilizers to soils slowly lowers soil pH, which in turn decreases nitrification rates. Under these conditions nitrification and N mineralization may be reduced. We therefore investigated the impact of liming fertilizer-acidified soils on nitrification and N mineralization. Soil samples were collected in the spring of 1987 from a field experiment, initiated in 1980, investigating N, tillage, and residue management under continuous corn (Zea mays L.). The pH values (CaCl2) in the surface soil originally ranged from 6.0 to 6.5. After 6 years the N fertilizer and tillage treatments had reduced the soil pH to values that ranged between 3.7 and 6.2. Incubation treatments included two liming rates (unlimed or SMP-determined lime requirement), two 15N-labeled fertilizer rates (0 or 20 g N m-2), and three replicates. Field-moist soil was mixed with lime and packed by original depth into columns. Labeled-15N ammonium sulfate in solution was surface-applied and columns were leached with 1.5 pore volumes of deionized water every 7 days over a 70-day period. Nitrification occurred in all pH treatments, suggesting that a ferilizer-acidified soil must contain a low-pH tolerant nitrifier population. Liming increased soil pH values (CaCl2) from 3.7 to 6.2, and increased by 10% (1.5 g N m-2) the amount of soil-derived NO3 --N that moved through the columns. This increase was the result of enhanced movement of soil-derived NO3 --N through the columns during the first 14 days of incubation. After the initial 14-day period, the limed and unlimed treatments had similar amounts of soil N leaching through the soil columns. Lime increased the nitrification rates and stimulated the early movement of fertilizer-derived NO3 --N through the soil.

    Notizen: Abstract The defensive secretion of the alpine chrysomelidOreina gloriosa is a complex mixture of mainly cardenolides and tyrosine betaine. Individually sampled secretions of adult laboratory-reared and field-collected beetles were analyzed by reverse-phase HPLC; 16 secretion components were quantified. Quantities and concentrations of different components were significantly affected by the age, sex, and reproductive status of individual beetles. Aging was correlated with marked increases (up to 4.4-fold) and decreases (up to 2.7-fold) of quantities and concentrations of several components. Differences between the sexes were smaller, but quantities of all components and concentrations of several components were larger in laboratory-reared females than in males. There was less of one component of the secretion in mated than unmated females, but the concentrations of four secretion components were higher (up to 1.6-fold) in mated females.

    Notizen: Abstract Acute toxicity of soil amended with allyl isothiocyanate (AITC) to three size classes ofLimonius californicus (Mann.) wireworms was determined in the laboratory. Wireworms were exposed to AITC at initial concentrations of 120–300 nmol/g soil for one day. During this time, extractable AITC concentrations decreased by 66 to 93 %. Probit analysis estimated LC50 values of 238 and 226 nmol/g soil at one day posttreatment for medium and large wireworms, respectively. For small wireworms, LC50 values decreased from 211 to 157 nmol/g soil during 1–137 days posttreatment. Sublethal concentrations of AITC significantly reduced feeding activity of treated wireworms at three posttreatment times and over the entire 137 days. Wireworm weight was not significantly affected by AITC. The potential exists to use glucosinolate-containing plant tissue as an isothiocyanate (ITC) source to reduce crop damage caused byL. californicus wireworms.

We analyzed the behavioral responses of four species of entomopathogenic nematodes during contact with feces of natural or experimental hosts. Host recognition by nematodes was manifested in alterations in the frequency and/or duration of one or more search parameters including forward crawling, headwaving, body-waving, stopping, backward crawling, head-rubbing, and headthrusting.Heterorhabditis bacteriophora andSteinernema glaseri showed behavioral responses to contact with feces of their natural hosts,Spodoptera exigua (Lepidoptera) andPopillia japonica (Coleoptera), and to the experimental hosts,Acheata domesticus (Orthoptera) andBlatella germanica (Blatteria).Steinernema carpocapsae responded only toB. germanica feces, whereas5. scapterisci did not significantly respond to any of the insect species. During contact with cockroach feces, all nematodes, exceptS. scapterisci, showed avoidance behavior. We suggest that ammonia present in cockroach feces is inhibitory to nematodes. Specific host recognition by entomopathogenic nematodes may be an important mechanism to maintain host affinities.

    Notizen: Abstract (R)-(−)-Linalool was identified as a minor component sex pheromone of the scarab beetleHolotrichia parallela (Coleoptera: Scarabaeidae). Field evaluations revealed that, although not attractive per se, (R)-(−)-linalool enhances the attractiveness of the major sex pheromone,L-isoleucine methyl ester (LIME). Analyses of the pheromone titers in the glands of field-collected females demonstrated the occurrence of peak levels of 48-hr (“circabidian”) periodicity. The levels of LIME in the glands of 45-day-old virgin females increased over three times from the scototo the photophase of a calling day, but the amounts of (R)-(−)-linalool did not significantly change. Virgin females had in average two times more LIME and 3.6 times more (R)-(−)-linalool than the average amount found in the field-captured beetles throughout the season.

    Notizen: Abstract Synthetic 2-(E)-nonenol, previously identified as the sex pheromone ofAnomala schonfeldti (Coleoptera: Scarabaeidae), is demonstrated to be very attractive to males in the field. Nevertheless, no significant differences were found between treatments with 1, 5, 10, and 20 mg dosages. Males ofA. schonfeldti were more significantly attracted to traps at 30 cm high than at 90 cm. Although the observed behavior seemed to indicate a trend of more attraction to buried traps than those placed at 30 cm, there was no statistical difference between the two treatments. Pheromone-baited traps caught significantly more beetles than traps containing three virgin females. Over 70% of released beetles were recaptured in six traps surrounding the point of release and separated from each other by 50 m, suggesting a possible use of the pheromone (in combination with floral compounds) in mass trapping.

    Notizen: Abstract Coupled GC-EAG techniques have been applied to the study of volatiles from the grain weevil,Sitophilus granarius. for the first time. The size of EAG response was independent of the sex of the responding insect but was consistently larger to extracts of males than those of females. This difference was reflected in a behavioral preference for the male extracts by mated adults of both sexes tested together and virgin adults of both sexes tested separately. The GC-EAG results provide evidence for two materials that are released specifically by the males. Using circular dichroism. one has been found to be identical stereochemically with the (2S,3R)-sitophilate reported by others as the aggregation pheromone in a different strain. This enhances the prospects for the development of a single pheromone lure that would be generally applicable whatever the origin of the strain. The small amount of sitophilate found in the males suggests that it is not stored in large amounts. The other material, present in such a small amount that it has yet to be fully characterized, elicits a higher antennal activity than sitophilate and may have a significant role to play in enhancing the trap catch of this economically important pest.

    Notizen: Abstract The abdominal glands of three bark-inhabiting larvae of generaBolitochara, Leptusa (Staphylinidae), andHypophloeus (Tenebrionidae) were studied chemically and morphologically. Behavior of the larvae indicated that secretion is emitted only after severe disturbance of the larvae. These mechanical contacts may also occur incidentally with coinhabiting nonpredatory arthropods when the beetle larvae move within small interstices under bark. Depending on the species, the secretions contained 1,4-benzoquinone derivatives and three 6-alkyl-naphthoquinones dissolved within various alkanes, alkenes, ethyl-, isopropyl-, and isoamylesters. More erratically distributed gland constituents also detected were acetophenone, benzyl propionate, and methyl hydroxybenzoate. In the laboratory, synthetic quinone-containing solutions simulating those found inLeptusa andBolitochara larvae acted as strong topical irritants and caused further damage to last-stageCalliphora vomitoria larvae if hydrocarbons or esters were used as solvents. The natural secretions ofHypophloeus versipellis elicited considerable mortality in two subcortical sciarid larvae cooccurring with tenebrionid larvae. Bioassay and secretion chemistry of the Staphylinidae/Tenebrionidae larval secretions indicated that they are typical defensive secretions that act topically. Morphological data characterized Bolitocharini larvae as possessing protuberant abdominal tergites supplied with an interiorly situated gland reservoir. After mechanical contact, the defensive secretion is topically applied to other arthropods by dabbing this tergal protuberance on targets. The defensive gland ofHypophloeus versipellis is unusual in possessing a movable reservoir opening situated at the anterior border of tergite IX. By this peculiar gland morphologyHypophloeus larvae are capable of shooting secretion droplets frontally from their slightly depressed dorsal abdominal surface without bending their abdominal tips dorsally. This seems an adaptation to the interstitial habitat of the larvae. The types of defensive glands and their phylogenetic value in Aleocharinae/Tenebrionidae larvae are discussed.

    Notizen: Abstract Responses of threeHylastes species,Dryocoetes autographus, and twoHylobius species to terpenes and ethanol were studied in field experiments on clear-cut forest sites in Sweden using baited ground traps.α-Pinene alone did not attract any of the six species. A terpene blend (spruce turpentine consisting mainly ofα-pinene,β-pinene, and 3-carene) attractedHylastes cunicularius, H. brunneus, andHylobius abietis in some experiments, but not in others. The attractiveness of ethanol also varied; the only species consistently attracted wasH. abietis. Baits containing both terpenes and ethanol, particularly the combination of spruce turpentine and ethanol, were attractive to all species exceptHylobius pinastri. InH. abietis, the terpene plus ethanol/ ethanol catch ratios increased during early summer. Seasonal differences in catch levels were observed inH. cunicularius andH. abietis. The addition ofα-pinene reduced the attractiveness of the combination of spruce turpentine and ethanol toH. cunicularius, H. opacus, andD. autographus. The differences in response to the volatiles between species are probably related to differences in reproductive behavior and host preferences.

    Notizen: Abstract The maize genome has been shown to contain six glutamine synthetase (GS) genes with at least four different expression patterns. Noncoding 3′ gene-specific probes were constructed from all six GS cDNA clones and used to examine transcript levels in selected organs by RNA gel blot hybridization experiments. The transcript of the single putative chloroplastic GS2 gene was found to accumulate primarily in green tissues, whereas the transcripts of the five putative GS1 genes were shown to accumulate preferentially in roots. The specific patterns of transcript accumulation were quite distinct for the five GS1 genes, with the exception of two closely related genes.

    Notizen: Abstract A cDNA clone (Zm58.1) was isolated by differential screening from a cDNA library made to mature Zea mays pollen, and shown to be pollen-specific by RNA blot analysis. When this partial-length clone was used to probe a genomic library, a similar but distinct pollen-specific genomic clone (68% sequence identity) was isolated (Zm58.2). The putative proteins coded for by these two clones show sequence homology to several flower-expressed gene products from various plant species, including known pollen allergens from short ragweed (Ambrosia artemisiifolia), and to pectate lyases from the plant pathogenic bacteria Erwinia spp. The two genes map to different chromosomes.

    Notizen: Abstract The homodimeric alcohol dehydrogenase gene product of maize (Zea mays L.)Adh1-1S1108 mutation was purified and compared with the parentalAdh1-1S enzyme. The mutant alcohol dehydrogenase activity had pH optima and substrate specificity similar to those of the parental enzyme, but exhibited somewhat increased and decreasedK mvalues for acetaldehyde and NADH, respectively.     Notizen: Abstract The 4942 bp DNA sequence of Zea mays transposon MuA2 was determined. Previous evidence indicated MuA2 controls activity of the Mu1 transposon located in the mutable allele a1-mum2. MuA2 contains two large, ATG-initiated open reading frames (ORFs) of 612 and 232 codons, respectively, located on opposite strands. MuA2 produces two transcripts, each containing one of these ORFs. Four different tandem direct repeat sequences are located downstream of the 612 codon ORF. The restriction map of MuA2 is identical to that of transposon MuR1, which also is known to regulate mutability of a1-mum2. Furthermore, except for a single nucleotide, MuA2 is identical to the Mutator element Mu9.

    Notizen: Abstract Maize leaf phosphoenolpyruvate carboxylase was completely and irreversibly inactivated by treatment with micromolar concentrations of Woodward's reagentK (WRK) for about 1 min. The inactivation followed pseudo-first-order reaction kinetics. The order of reaction with respect to WRK showed that the reagent causes formation of reversible enzyme inhibitor complex before resulting in irreversible inactivation. The loss of activity was correlated to the modification of a single carboxyl group per subunit, even though the reagent reacted with 2 carboxyl groups per protomer. Substrate PEP and PEP + Mg2+ offered substantial protection against inactivation by WRK. The modified enzyme showed a characteristic absorbance at 346 nm due to carboxyl group modification. The modified enzyme exhibited altered surface charge as seen from the elution profile on FPLC Mono Q anion exchange column. The modified enzyme was desensitized to positive and negative effectors like glucose-6-phosphate and malate. Pretreatment of PEP carboxylase with diethylpyrocarbonate prevented WRK incorporation into the enzyme, suggesting that both histidine and carboxyl groups may be closely physically related. The carboxyl groups might be involved in metal binding during catalysis by the enzyme.

    Notizen: Abstract We have investigated the presence of higher-order chromatin structures in different maize tissues. Taking advantage of the pulsed-field gel electrophoresis technique to analyse large DNA fragments from intact nuclei and cells, we have determined the size distribution of the high-molecular-weight DNA fragments obtained from chromatin degradation by endogenous nucleases in isolated nuclei. Chromatin digestion leads to the appearance of stable DNA fragments of about 50 kb in all the tissues examined, suggesting the folding of DNA in higher-order chromatin domain structures. It has been reported that such chromatin domains are formed by loops of the 30 nm fibres anchored to the nuclear matrix by a complex set of proteins, including DNA topoisomerase II. Treatment of maize protoplasts with the calcium ionophore A23187 and the antitumour drug VM-26, which specifically inhibit the religation of the cleaved DNA in the topoisomerase II reaction, also produces the 50 kb structure. Analysis of the DNA contained in the 50 kb chromatin structure shows a higher degree of methylation than in bulk maize chromosomal DNA. The role of methylated DNA in the chromatin folding is discussed.

    Notizen: Abstract Chromosomal high-mobility-group (HMG) proteins occur ubiquitously in eukaryotes and their common structural and biochemical features indicate a critical role. In this context, we compared structural and functional aspects of HMG proteins from the monocotyledonous plant maize and the dicotyledonous plant Vicia faba. Besides biochemical similarities and immunological differences found between these proteins, the isolation and characterization of a cDNA encoding the V. faba homologue of the maize HMGa protein revealed great similarities between these two proteins, including the HMG-box DNA-binding motif and an acidic domain. Therefore, like the maize HMGa protein, the V. faba HMG protein belongs to the vertebrate HMG1 family, which consists of HMG proteins and transcription factors of various eukaryotes.

    Notizen: Abstract The reduction of zein synthesis in the maize (Zea mays L.) opaque-2 mutant is associated with an increased percentage of lysine in the endosperm protein. When expressed on an endosperm basis, we found that W64A opaque-2 contains 490 μg of lysine compared with 350 μg in W64A normal. SDS-PAGE analysis of endosperm proteins indicated that several non-zein proteins are more abundant in the mutant than in normal genotype. To determine the subcellular origin of these proteins, we separated an endosperm homogenate from developing kernels by sucrose density gradient centrifugation and used marker enzyme assays and immunoblot analyses to identify cellular components. Amino acid analysis of proteins in the gradient fractions showed that the majority of the lysine occurs in soluble proteins at the top of the gradient. To identify these proteins, we prepared a complex antiserum against the entire soluble protein fraction and used it to immunoscreen an endosperm cDNA expression library. Sequence analysis of clones identified mRNAs involved in carbohydrate metabolism, amino acid biosynthesis, and protein synthesis. RNA dot blot hybridization analysis with these clones revealed significant variation in the levels of transcripts between normal and opaque-2 endosperm, but we identified several mRNAs that are elevated in opaque-2 and that may encode proteins responsible for the enhanced lysine content.

    Notizen: Abstract Clones of two highly related genes, ZmHox2a and ZmHox2b (Zea mays homeobox), were isolated from maize embryo cDNA libraries by screening with the ZmHox1a homeobox sequence. The genes map to chromosomes 3 and 8, respectively, and encode mRNA transcripts of 6 kb. The encoded proteins, ZmHox2a and b, share 84% sequence identity and exhibit a modular structure with several novel plant-specific protein domains. Interestingly, each ZmHox2 gene product contains two complete homeodomains which, for Zmhox2a, were both shown to be functional DNA-binding motifs in vitro. Not only probes encoding the homeobox but also DNA fragments corresponding to other ZmHox2 domains hybridize to multiple bands in genomic Southern blots, indicating that related protein domains may be conserved in other maize genes. The ZmHox2a/b genes, therefore, are members of a novel and large class of maize genes, some of which can be expected to encode new transcription factors.

    Notizen: Abstract The maize Bronze2 (Bz2) gene encodes a glutathione S-transferase that is required for anthocyanin pigment accumulation. Two classes of regulatory proteins, R and C1, are required for transcriptional activation of Bz2 and several additional structural genes. Functional domains of the Bz2 promoter were identified using Bz2 promoterdriven luciferase reporter genes electroporated into maize protoplasts together with R and C1 expression plasmids. Complete regulation was conferred by 224 nt of the Bz2 promoter. Within this region at least two separable regions are independently capable of conferring regulation by R and C1. Predicted regulatory elements corresponding to two classes of sequence motifs, the Myb-box homologous ‘C1-motif’, TAACTG/CAGTTA, and the G-box and E-box homologous ‘R-motif’, CACGTG, were shown to be important for full R and C1 activation of the Bz2 promoter. Expression of reconstructed Bz2 genes with mutated promoters was quantified using RNase protection, and this analysis confirmed results obtained using reporter genes.

    Notizen: Abstract This study was conducted to compare maize quantitative trait loci (QTL) detection for grain yield and yield components in F2∶3 and F6∶7 recombinant inbred (RI) lines from the same population. One hundred and eighty-six F6∶7 RIs from a Mo17×H99 population were grown in a replicated field experiment and analyzed at 101 loci detected by restriction fragment length polymorphisms (RFLPs). Single-factor analysis of variance was conducted for each locus-trait combination to identify QTL. For grain yield, 6 QTL were detected accounting for 22% of the phenotypic variation. A total of 63 QTL were identified for the seven grain yield components with alleles from both parents contributing to increased trait values. Several genetic regions were associated with more than one trait, indicating possible linked and/or pleiotropic effects. In a comparison with 150 F2∶3 lines from the same population, the same genetic regions and parental effects were detected across generations despite being evaluated under diverse environmental conditions. Some of the QTL detected in the F2∶3 seem to be dissected into multiple, linked QTL in the F6∶7 generation, indicating better genetic resolution for QTL detection with RIs. Also, genetic effects at QTL are smaller in the F6∶7 generation for all traits.

    Notizen: Abstract Three molecular marker protocols, chemiluminescent restriction fragment length polymorphisms (c-RFLPs), radioactivity-based restriction fragment length polymorphisms (r-RFLPs), and randomly amplified DNA polymorphisms (RAPDs) were compared in terms of cost and time efficiency. Estimates of cost of supplies and time requirements were obtained from simulations of maize (Zea mays L.) genotyping experiments utilizing protocols currently in use. The increase in total cost with increasing numbers of individuals genotyped and markers analyzed is higher for RAPDs than for RFLPs. RAPDs were generally found to be more cost and time efficient for studies involving small sample sizes, while RFLPs have the advantage for larger sample sizes. Because of the shorter exposure times involved, c-RFLPs require less time than r-RFLPs to obtain a given amount of information. Variations in the protocols, such as number of re-uses of Southern blots or cost of Taq DNA polymerase per reaction of amplification, also affect the relative merits of RAPDs and RFLPs. Two examples were analyzed where molecular markers are used: a germ plasm survey and quantitative trait loci (QTL) mapping in a segregating population. No protocol was found to be the most cost and time efficient over the entire range of sample sizes and number of marker loci studied.

Also, uniparental genome elimination in karyotypically unstable hybrids has been utilized for cereal haploid production. We have crossed hexaploid oat (2n=6x=42, Avena sativa L.) and maize (2n=2x=20, Zea mays L.) and recovered 90 progenies through embryo rescue. Fifty-two plants (58%) produced from oatxmaize hybridization were oat haploids (2n=3x=21) following maize chromosome elimination. Twenty-eight plants (31%) were found to be stable partial hybrids with 1–4 maize chromosomes in addition to a haploid set of 21 oat chromosomes (2n=21+1 to 2n=21+4). Ten of the ninety plants produced were found to be apparent chromosomal chimeras, where some tissues in a given plant contained maize chromosomes while other tissues did not, or else different tissues contained a different number of maize chromosomes. DNA restriction fragment length polymorphisms (RFLPs) were used to identify the maize chromosome(s) present in the various oat-maize progenies. Maize chromosomes 2, 3, 4, 5, 6, 7, 8, and 9 were detected in partial hybrids and chromosomal chimeras. Maize chromosomes 1 and 10 were not detected in the plants analyzed to-date. Furthermore, partial self-fertility, which is common in oat haploids, was also observed in some oat-maize hybrids. Upon selfing, partial hybrids with one or two maize chromosomes showed nearly complete transmission of the maize chromosome to give self-fertile maize-chromosome-addition oat plants. Fertile lines were recovered that contained an added maize chromosome or chromosome pair representing six of the ten maize chromosomes. Four independently derived disomic maize chromosome addition lines contained chromosome 4, one line carried chromosome 7, two lines had chromosome 9, one had chromosome 2, and one had chromosome 3. One maize chromosome-8 monosomic addition line was also identified. We also identified a double disomic addition line containing both maize chromosomes 4 and 7. This constitutes the first report of the production of karyotypically stable partial hybrids involving highly unrelated species from two subfamilies of the Gramineae (Pooideae — oat, and Panicoideae — maize) and the subsequent recovery of fertile oat-maize chromosome addition lines. Using gel retardation and DNase I protection assays we identified maize proteins which bind to seven subterminal sites in both ends ofAc. Four DNase I-protected sites contain a GGTAAA sequence, the other three include either GATAAA or GTTAAA. The specificity of the maize protein binding toAc was verified by using a synthetic fragment containing four GGTAAA motifs as probe and competitor in gel retardation assays. All seven binding sites are located within regions requiredin cis for transposition. A maize protein binding site with the same sequence has previously been identified in the terminal inverted repeats of the maizeMutator element. Thus, the protein, that recognizes this sequence is a good candidate for a regulatory host factor forAc transposition.

To study the functional relevance of some of these introns for gene expression we have analysed the influence of three 5′ introns on transient gene expression of the anaerobically inducible maize GapC4 promoter in maize cells. Under aerobic conditions, reporter gene expression is increased in the presence of the first introns of the GapC4 and GapC1 genes, and the first intron of the nuclear encoded chloroplast-specific GapA1 gene. In contrast, the GapC4 intron increases anaerobic gene expression above the level obtained for the intronless construct, while anaerobic expression of constructs harboring the GapA1 and GapC1 introns was similar to the anaerobic expression level of the intronless construct. Splicing analysis revealed that the GapC4 intron is processed more efficiently under anaerobic conditions, while no change in splicing efficiency is observed for the GapC1 and the GapA1 introns when subjected to anaerobic conditions. These results suggest that an increase in splicing efficiency contributes to the anaerobic induction of the maize GapC4 gene.

In contrast, theGapC4 intron increases anaerobic gene expression above the level obtained for the intronless construct, while anaerobic expression of constructs harboring theGapA1 andGapC1 introns was similar to the anaerobic expression level of the intronless construct. Splicing analysis revealed that theGapC4 intron is processed more efficiently under anaerobic conditions, while no change in splicing efficiency is observed for theGapC1 and theGapA1 introns when subjected to anaerobic conditions. These results suggest that an increase in splicing efficiency contributes to the anaerobic induction of the maizeGapC4 gene.

    Notizen: Summary The effects of red and blue light on the orientation of cortical microtubules (MTs) underneath the outer epidermal wall of maize (Zea mays L.) coleoptiles were investigated with immunofluorescent techniques. The epidermal cells of dark-grown coleoptiles demonstrated an irregular pattern of regions of parallel MTs with a random distribution of orientations. This pattern could be changed into a uniformly transverse MT alignment with respect to the long cell axis by 1 h of irradiation with red light. This response was transient as the MTs spontaneously shifted into a longitudinal orientation after 1–2 h of continued irradiation. Induction/reversion experiments with short red and far-red light pulses demonstrated the involvement of phytochrome in this response. In contrast to red light, irradiation with blue light induced a stable longitudinal MT alignment which was established within 10 min. The blue-light response could not be affected by subsequent irradiations with red or far-red light indicating the involvement of a separate blue-light photoreceptor which antagonizes the effect of phytochrome. In mixed light treatments with red and blue light, the blue-light photoreceptor always dominated over phytochrome which exhibited an apparently less stable influence on MT orientation. Long-term irradiations with red or blue light up to 6 h did not reveal any rhythmic changes of MT orientation that could be related to the rhythmicity of helicoidal cell-wall structure. Subapical segments isolated from dark-grown coleoptiles maintained a longitudinal MT arrangement even in red light indicating that the responsiveness to phytochrome was lost upon isolation. Conversely auxin induced a transverse MT arrangement in isolated segments even in blue light, indicating that the responsiveness to blue-light photoreceptor was eliminated by the hormone. These complex interactions are discussed in the context of current hypotheses on the functional significance of MT reorientations for cell development.

    Notizen: Summary The seed storage proteins of maize (Zea mays L.) are synthesized during endosperm development on membrane-bound polyribosomes. Protein body formation in normal genotypes occurs via a sequential deposition of the various types of zeins, and leads to the formation of spherical structures with a diameter of about l μm. In the endosperm mutantopaque-2 the level of one zein class is reduced; these kernels exhibit an opaque phenotype instead of the vitreous phenotype displayed in normal genotypes, presumably due to the decrease in total zein protein at the time of desiccation. Previous microscopic examination ofopaque-2 protein bodies at 22 DAP (days after pollination) showed that the protein bodies were morphologically similar to those of normal genotypes. However, the endosperm ofopaque-2 maize at 14 DAP contains tubular arrays within the rough endoplasmic reticulum. These tubular arrays are tightly associated with the developing protein bodies. Long strands of tubules, sometimes 10 μm in length, are observed in the endosperm, and partially formed protein bodies often seem to be forming directly from these tubular arrays. No immunostaining is associated with this tubular material when any of the anti-zein antibodies are used.

    Notizen: Summary Microtubules (MTs) in cells of various tissues at different distances from the apex of the maize root exhibited different sensitivities to cold (5 °C), as judged by MT reorientation and tendency to depolymerization. Their responses seem to be related to their initial intracellular arrangements. Generally, MTs in cells which were ceasing elongation were the least sensitive during the early stages (6–24 h) of cold treatment, but during the later stages (5–7 d) MTs in most of these cells eventually depolymerized. Pericycle cells showed a unique cold response. Here the MTs were conspicuously cold-labile and quickly depolymerized near the root-tip. However, after 1 d many pericycle cells in more proximal regions had repolymerized their MTs as dense, randomly organized arrays. These persisted for the remainder of the cold treatment. A similar resistance to longterm chilling, by means of MT repolymerization, was found in cells of the root cap, quiescent centre and cells of the distal part of the former meristem. MT repolymerization in the cold may enable the apex to resume growth when more favourable (warmer) conditions return.

    Notizen: The diversity in different groups of obligate saproxylic beetles was related to ecological variables at three levels of spatial scale in mature spruce-dominated forest. The variables were connected to: (i) decaying wood, (ii) wood-inhabiting fungi, (iii) the level of disturbance, (iv) landscape ecology, and (v) vegetational structure. Several strong relationships were found at medium (1 km2) and large scales (4 km2), while only weak relationships were found at a small scale (0.16 ha; 1 ha=104 m2). This may be explained by the local variations in habitat parameters and the high mobilities of many beetle species. Factors connected to decaying wood and wood-inhabiting fungi were clearly the most important factors at all scale levels. In particular, the variables diversity of dead tree parts, number of dead trees of large diameter and number of polypore fungi species increased the species richness of many groups and increased the abundance of many species. Eight species were absent below a certain density of decaying wood per 1 or 4 km2. Former extensive cutting was a negative factor at large scale, probably because of decreasing recolonization with increasing distance to the source habitats. Thinning reduced the diversity of species associated with birch. The development of guidelines favouring the diversity of saproxylic beetles are discussed below.

    Notizen: Abstract The En/Spm-encoded TNPA protein binds to 12-bp DNA sequence motifs that are present in the sub-termini of the transposable element. DNA binding of TNPA to monomeric and dimeric forms of the binding motif was analyzed by gel retardation and cross-linking studies. A DNA binding domain at the N-terminal and a dimerization domain at the C-terminal portion of TNPA were localized using deletion derivatives of TNPA. These domains are novel since no apparent homology has been found in the data bases. The stoichiometry of the TNPA-DNA complexes was analyzed. A special complex is formed with a tail-to-tail dimeric DNA binding motif, most probably involving two DNA-bound TNPA molecules that interact via their dimerization domains. In redox reactions the requirement for one or two disulfide bonds for DNA binding of TNPA was shown. The implications of these findings for the excision mechanism of En/Spm are discussed.

    Notizen: Summary Two different factors control the mutability of an unstable allele (c2-m8810581) of the C2 gene of maize. Both an autonomous En/Spm element and an unrelated independent factor, named Mediator, are coordinately required for the excision of the insert in c2-m881058Y. According to genetic analysis, Mediator does not have the suppressor (S) function or mutator (M) function of En/Spm. Mediator has no effect on the timing or frequency of excision of Enl, En-low, or various I/dSpm elements. Hence, Mediator only mediates a specific interaction between En and the insert at c2m881058Y. Molecular analysis of c2-m881058Y has revealed a 3.3 kb, complex, En-related receptor element inserted into the second exon of the C2 gene. The ends of this element are homologous to the ends of En/Spm, but an internal l.7 kb region shows no En/Spm homology. A great degree (11–14%) of nucleotide changes, relative to Enl, occur within and between the 12 bp TNPA binding motifs. Alterations of these critical cis-determinants may account for the need for a “helper” factor for excision. This element is named Irma, for Inhibitor that requires Mediator also, and represents a unique, low copy number class of receptor element.

Inhibition of NRA by OKA and stimulation by H-7 indicated that NRA is increased by dephosphorylating the enzyme. We have identified a protein kinase (C type) that can phosphorylate the purified NR in vitro without the involvement of other accessory proteins. By in vivo labelling with 32P and immunoprecipitation of NR with NR antibodies it was found that in the presence of OKA most NR protein (NRP) was present in phosphorylated state, while with H-7 the reverse was seen. The red (R) and far-red (FR) light reversible experiments suggested that phytochrome (Pfr, an active form) stimulation of NRA is mediated by dephosphorylation of the enzyme, suggesting that Pfr regulates both NR transcription and NRA via phosphorylation/dephosphorylation steps controlled by separate signal transduction pathways.

Chloroplast development in some cells appears to be able to proceed in the absence of the Ij polypeptide.

    Notizen: Abstract In all eukaryotes α- and β-tubulins are encoded by small families of closely related genes and are highly conserved. In Zea mays, at least six different α-tubulin coding sequences are known. We describe the isolation from scutellar nodes of the maize inbred line W22 of a clone (CTM5) coding for an α-tubulin. On the basis of the 3′ end nucleotide sequence, this clone can be assigned to the already reported tua4 gene. Northern analysis demonstrates that CTM5 encodes a 1.5 kb transcript, which is expressed in different tissues of the seed and of the seedling. In order to define the spatial and temporal expression of α-tubulin genes, in situ hybridization experiments were performed on these tissues. Unexpectedly, a specific signal was detected with both antisense and sense RNA strands. Temporal and spatial distribution of the two RNAs, however, shows that high levels of the two transcripts are always discordant. In tissues where sense transcripts are highly abundant (embryos at various developmental stages, root tips, pollen grains), the antisense transcripts are expressed in relatively small amounts, while in pericarp, coleoptile, leaves, and scutellar node, where antisense transcripts accumulate, the sense transcript only reaches a very low level. Northern analysis using single-stranded DNA probes confirmed the presence of an antisense transcript of 1.5 kb, prompting speculation about the role of this transcript in the regulation of the expression of α-tubulin genes.

    Notizen: Abstract The aim of this research was to determine whether the structural homology between the O2 gene, a maize transcriptional activator, and the GCN4 gene, a yeast transcriptional factor, is reflected at the level of function. The O2 cDNA was cloned in the yeast expression vector pEMBLyex4 under the control of a hybrid, inducible promoter, and used to transform the yeast Saccharomyces cerevisiae. Transformed yeast cells produced O2 mRNA and a polypeptide immunoreactive with anti-O2 antibodies during growth in galactose. The heterologous protein was correctly translocated into the yeast nuclei, as demonstrated by immunofluorescence, indicating that the nuclear targeting sequences of maize are recognized by yeast cells. Further experiments demonstrated the ability of O2 to rescue a gcn4 mutant grown in the presence of aminotriazole, an inhibitor of the HIS3 gene product, suggesting that O2 activates the HIS3 gene, gene normally under control of GCN4. It was shown that the O2 protein is able to trans-activate the HIS4 promoter in yeast cells and binds to it in vitro. The sequence protected by O2, TGACTC, is also the binding site for GCN4. Finally, the expression of O2 protein in yeast did not produce alterations during batch growth at 30° C, while transformants expressing O2 protein showed a conditionally lethal phenotype when grown in galactose at 36° C; this phenotype mimics the behaviour of gcd mutants. The results support the idea that basic mechanisms of transcription control have been highly conserved in eukaryotes.

    Notizen: Abstract  Genetic data suggest that transposition of the maize element Activator (Ac) is modulated by host factors. Using gel retardation and DNase I protection assays we identified maize proteins which bind to seven subterminal sites in both ends of Ac. Four DNase I-protected sites contain a GGTAAA sequence, the other three include either GATAAA or GTTAAA. The specificity of the maize protein binding to Ac was verified by using a synthetic fragment containing four GGTAAA motifs as probe and competitor in gel retardation assays. All seven binding sites are located within regions required in cis for transposition. A maize protein binding site with the same sequence has previously been identified in the terminal inverted repeats of the maize Mutator element. Thus, the protein, that recognizes this sequence is a good candidate for a regulatory host factor for Ac transposition.

    Notizen: Abstract Plasma membrane fractions were isolated from maize (Zea mays L.) endosperms and etiolated kernels to investigate the possible membrane location of the sucrose synthase (SS) protein. Endosperms from seedlings at both 12 and 21 days after pollination (DAP), representing early and mid-developmental stages, were used, in addition to etiolated leaf and elongation zones from seedlings. Plasma membrane fractions were isolated from this material using differential centrifugation and aqueous two-phase partitioning. The plasma membrane-enriched fraction obtained was then analyzed for the presence of sucrose synthase using protein blots and activity measurements. Both isozymes SS1 and SS2, encoded by the lociSh1 andSus1, respectively, were detected in the plasma membrane-enriched fraction using polyclonal and monoclonal antisera to SS1 and SS2 isozymes. In addition, measurements of sucrose synthase activity in plasma membrane fractions of endosperm revealed high levels of specific activity. The sucrose synthase enzyme is tightly associated with the membrane, as shown by Triton X-100 treatment of the plasma membrane-enriched fraction. It is noteworthy that the gene products of bothSh1 andSus1 were detectable as both soluble and plasma membrane-associated forms.

    Notizen: Abstract Experiments with two maize (Zea mays L.) hybrids were conducted to determine (a) if the inhibition of nitrate uptake by aluminium involved a restriction in the induction (synthesis/assemblage) of nitrate transporters, and (b) if the magnitude of the inhibition was affected by the concurrent presence of ambient ammonium. At pH 4.5, the rate of nitrate uptake from 240 μM NH4NO3 was maximally inhibited by 100 μM aluminium, but there was little measurable effect on the rate of ammonium uptake. Presence of ambient aluminium did not eliminate the characteristic induction pattern of nitrate uptake upon first exposure of nitrogen-depleted seedlings to that ion. Removal of ambient aluminium after six hours of induction resulted in recovery within 30 minutes to rates of nitrate uptake that were similar to those of plants induced in absence of aluminium. Addition of aluminium to plants that had been induced in absence of aluminium rapidly restricted the rate of nitrate uptake to the level of plants that had been induced in the presence of aluminium. The data are interpreted as indicating that aluminium inhibited the activity of nitrate transporters to a greater extent than the induction of those transporters. When aluminium was added at initiation of induction, the effect of ambient ammonium on development of the inhibition by aluminium differed between the two hybrids.     Notizen: Abstract In a 2-year field experiment conducted on a Gleyic Luvisol in Stuttgart-Hohenheim one experimental and nine commercial maize cultivars were compared for their ability to utilize soil nitrate and to reduce related losses of nitrate through leaching. Soil nitrate was monitored periodically in CaCl2 extracts and in suction cup water. Nitrate concentrations in suction water were generally higher than in CaCl2 extracts. Both methods revealed that all cultivars examined were able to extract nitrate down to a soil depth of at least 120 cm (1988 season) or 150 cm (1987 season). Significant differences among the cultivars existed in nitrate depletion particularly in the subsoil. At harvest, residual nitrate in the upper 150 cm of the profile ranged from 73–110 kg N ha−1 in 1987 and from 59–119 kg N ha−1 in 1988. Residual nitrate was closely correlated with nitrate losses by leaching because water infiltration at 120 cm soil depth started 4 weeks after harvest (1987) or immediately after harvest (1988) and continued until early summer of the following year. The calculated amount of nitrate lost by leaching was strongly influenced by the method of calculation. During the winter of 1987/88 nitrate leaching ranged from 57–84 kg N ha−1 (suction cups) and 40–55 kg N ha−1 (CaCl2 extracts), respectively.     Notizen: Abstract The carbon and nitrogen partitioning characteristics of wheat (Triticum aestivum L.) and maize (Zea mays L.) grown hydroponically at a constant pH on either 4 mM or 12 mM NO3 - or NH4 + nutrition were investigated using either 14C or 15N techniques. Greater allocation of 14C to amino-N fractions occurred at the expense of allocation of 14C to carbohydrate fractions in NH4 +-compared to NO3 --fed plants. The [14C]carbohydrate:[14C]amino-N ratios were 1.5-fold and 2.0-fold greater in shoots and roots respectively of 12 mM NO3 --compared to 12 mM NH4 +-fed wheat. In both 4 mM and 12 mM N-fed maize the [14C]carbohydrate:[14C]amino-N ratios were approximately 1.7-fold and 2.0-fold greater in shoots and roots respectively of NO3 --compared to NH4 +-fed plants. Similar results were observed in roots of wheat and maize grown in split-root culture with one root-half in NO3 --and the other in NH4 +-containing nutrient media. Thus the allocation of carbon to the amino-N fractions occurred at the expense of carbohydrate fractions, particularly within the root. Allocation of 14N and 15N within separate sets of plants confirmed that NH4 --fed plants accumulated more amino-N compounds than NO3 --fed plants. Wheat roots supplied with 15NH4 + for 8 h were found to accumulate 15NH4 + (8.5 μg 15N g-1 h-1) whereas in maize roots very little 15NH4 + accumulated (1.5 μg 15N g-1 h-1) It is proposed that the observed accumulation of 15NH4 + in wheat roots in these experiments is the result of limited availability of carbon within the roots of the wheat plants for the detoxification of NH4 +, in contrast to the situation in maize. Higher photosynthetic capacity and lower shoot: root ratios of the C4 maize plants ensure greater carbon availability to the root than in the C3 wheat plants. These differences in carbon and nitrogen partitioning between NO3 --and NH4 +-fed wheat and maize could be responsible for different responses of wheat and maize root growth to NO3 - and NH4 + nutrition.

    Notizen: Abstract The effect of amino acids on nitrate transport was studied in Zea mays cell suspension cultures and in Zea mays excised roots. The inclusion of aspartic acid, arginine, glutamine and glycine (15mM total amino acids) in a complete cell-culture media containing 1.0 mM NO3 - strongly inhibited nitrate uptake and the induction of accelerated uptake rates. The nitrate uptake rate increased sharply once solution amino acid levels fell below detection limits. Glutamine alone inhibited induction in the cell suspension culture. Maize seedlings germinated and grown for 7 days in a 15 mM mixture of amino acids also had lower nitrate uptake rates than seedlings grown in 0.5 mM Ca(NO3)2 or 1 mM CaCl2. As amino acids are the end product of nitrate assimilation, the results suggest an end-product feed-back mechanism for the regulation of nitrate uptake.

    Notizen: Summary Crop improvement for tolerance to specific herbicides is an important breeding target, since molecules performing well with regard to environmental safety are frequently not completely selective for crops. The glutathione (GSH)/glutathione-S-transferase (GST) system is a general mechanism of detoxification that in higher plants may confer tolerance to some herbicides. GSH level and GST activity were measured in different maize inbred lines, in the absence or in the presence of EPTC (a thiocarbamate) and of Alachlor (a chloroacetanilide); a wide genetic variability was observed for these parameters, which appear to be involved in plant tolerance to herbicides. Isozyme analysis was performed on roots, leaves, scutellum, pollen, coleoptile, mesocotyl of the same inbreds: it revealed the presence of many GST forms in maize, showing high polymorphism; they are controlled by at least five genes, the expression of which is developmentally regulated in the different tissues analyzed.

In five of eight trials, heterogeneity due to environmental index was significant. Heterogeneity due to the other environmental factors was not significant in any trial. A comparison of δ i 2 (stability-variance statistic derived from total GE interaction) and s i 2 (stability-variance statistic derived from residual GE interaction following removal of heterogeneity due to encovariate) helped identify genotypes that performed stably or unstably because of a linear effect of environmental index. In three of the five trials showing significant heterogeneity due to environmental index, the YSi statistic selected a reduced number of unstable genotypes as compared with selection based solely on yield.     Notizen: Summary Maize pollen quality was investigated after long-term storage both in a refrigerator and in liquid nitrogen by a combination of viability tests and cytochemical methods. Determination of the activities of a number of enzymes involved in important metabolic pathways was carried out. Quinone formation was also studied, as some products of secondary metabolism affect pollen grain viability. One year of pollen storage in liquid nitrogen had little effect on the activities of oxidoreductases and hydrolases and had no significant effect on pollen grain viability evaluated by acetocarmine, neutral red and acridine organe. Only the FCR test showed slightly decreased viability. After one and two years of storage in a refrigerator, pollen grain viability, tested using acetocarmine, neutral red and acridine orange, did not change substantially. Simultaneously the FCR test showed a considerable decrease in pollen grain viability. Long-term storage in a refrigerator resulted in the loss of cytochrome oxidase activity and rise of alcohol dehydrogenase, lactate dehydrogenase, peroxidase and polyphenoloxidase activities as well as of quinone formation.

    Notizen: Summary This study was conducted to determine the relationships among various agronomic traits in inbreds, hybrids and between inbreds and their progeny of maize in short season areas. Phenotypic correlations of each measured trait between as well as among hybrids and inbreds were not the same. The highest correlation coefficient (r=0.78) was obtained between days to anthesis and yield. The high yielding inbreds did not necessarily give rise to high yielding progeny. Given that days to anthesis plays an important role in two major traits (yield and moisture) for hybrids, an emphasis on optimum anthesis dates during inbred development could be a useful criterion for the prediction of hybrid performance in short season areas.

    Notizen: Summary Grain yield in the maize (Zea mays L) plant is sensitive to drought in the period three weeks either side of flowering. Maize is well-adapted to the use of restriction fragment length polymorphisms (RFLPs) to identify a tight linkage between gene(s) controlling the quantitative trait and a molecular marker. We have determined the chromosomal locations of quantitative trait loci (QTLs) affecting grain yield under drought, anthesis-silking interval, and number of ears per plant. The F3 families derived from the cross SD34(tolerant) × SD35 (intolerant) were evaluated for these traits in a two replicated experiment. RFLP analysis of the maize genome included non-radioactive DNA-DNA hybridization detection using chemiluminescence. To identify QTLs underlying tolerance to drought, the mean phenotypic performances of F3 families were compared based on genotypic classification at each of 70 RFLP marker loci. The genetic linkage map assembled from these markers was in good agreement with previously published maps. The phenotypic correlations between yield and other traits were highly significant. In the combined analyses, genomic regions significantly affecting tolerance to drought were found on chromosomes 1,3,5,6, and 8. For yield, a total of 50% of the phenotypic variance could be explained by five putative QTLs. Different types of gene action were found for the putative QTLs for the three traits.

This paper addresses a set of questions that are usually presented to the breeders, in light of the evidence obtained from many trials conducted in Brazil and in the U.S.A.: Will the genotypes bred for sole crop conditions, perform well when grown in intercrop; How different should a genotype be, for cultivation in intercropping compared to genotypes developed for sole crop conditions; Is there a need for special breeding programs for intercropping and How could a breeding program focus the question of multiple (associated) cropping?

    Notizen: Summary World trend is to more maize hectarage grown to fewer, more widely-adapted hbrids. My purpose is to document research events leading to the most popular corn hybrids, Pioneer Brand 3780 and 3732. I provide background on maize adaptedness, on the seed maize business, on commercial maize hybrid development process, and on cultural practice change. I provide historical information on the cultivars: Leaming corn, Reid Yellow Dent, and Minn. 13 were widely-adapted open-pollinated varieties. Shift from open-pollinated to hybrid maize caused a shift in emphasis from local to wider adaptation. The first widely-adapted hybrids were Iowa 939 and U.S. 13. DEKALB 404A was the first popular and most popular double-cross hybrid. Pioneer 349 was the second-most popular double-cross hybrid. The first popular single cross hybrids were DEKALB 805 in medium and DEKALB XL45 in early U.S. Corn Belt in the early-1960's. Cultural practices (higher planting density, narrower rows, more nitrogen, better pesticides, and larger equipment allowing more timely operations) changed greatly in the 1960's and 1970's; allowing fewer, more widely-adapted genotypes to prevail. First hybrid 3780, then 3732 became the most popular hybrid; each exceeded one billion (1 × 109) dollars sales. They were the result of wide-area testing, of utilizing higher plant density stress for selecting inbred lines and for screening hybrids, and of modern information management.

    Notizen: Abstract Flooding effects on membrane permeability, lipid peroxidation and activated oxygen metabolism in corn (Zea mays L.) leaves were investigated to determine if activated oxygens are involved in corn flooding-injury. Potted corn plants were flooded at the 4-leaf stage in a controlled environment. A 7-day flooding treatment resulted in a significant increase in chlorophyll breakdown, lipid peroxidation (malondialdehye content), membrane permeability, and the production of superoxide (O 2 - ) and hydrogen peroxide (H2O2) in corn leaves. The effects were much greater in older leaves than in younger ones. Spraying leaves with 8-hydroxyquinoline (an O 2 - scavenger) and sodium benzoate (an .OH scavenger) reduced the oxidative damage and enhanced superoxide dismutase (SOD) activity. A short duration flooding treatment elevated the activities of SOD, catalase, ascorbate peroxidase (AP), and glutathione reductase (GR), while further flooding significantly reduced the enzyme activities but enhanced the concentrations of ascorbic acid and reduced form glutathione (GSH). It was noted that the decline in SOD activity was greater than that in H2O2 scavengers (AP and GR). The results suggested that O 2 - induced lipid peroxidation and membrane damage, and that excessive accumulation of O 2 - is due to the reduced activity of SOD under flooding stress.

    Notizen: Abstract Seeds of Zea mays L., germinating in soil, were exposed to very low doses of the sulfonylurea herbicides chlorsulfuron and metsulfuron methyl. At a concentration of 0.012 mg L−1, chlorsulfuron caused 72% and metsulfuron methyl 55% growth reduction of the young primary roots. Both herbicides also caused obvious injuries to the root tips. Scanning electron microscopic observations of the root tip surfaces indicated an inhibition of slime secretion at a herbicide concentration of 1.5 mg L−1. Transmission electron microscopy revealed obvious changes to the nuclei and deformation of radial cell walls in the primary root cortex at 0.012 and 1.5 mg L−1 for both herbicides. Moreover, the secretory cells of the root cap periphery showed partially irregular deposition of premature cell wall or slime material at a concentration of 0.012 mg L−1 of both herbicides. From the results of our electron microscopic observations we conclude that the primary roots of maize seedlings are seriously affected by extremely low concentrations of even those herbicides which (as chlorsulfuron and metsulfuron methyl) have been developed to inhibit the growth of dicotyledonous weeds. Moreover, we suggest that the frequently observed growth retardation of crop seedlings is a consequence of early root tip injuries caused by herbicide residues in the soil. ei]H Lambers

    Notizen: Abstract During a study of the diffusivity of sulphorhodamine G in the cortical apoplast of maize roots widely discrepant rates were found between different samples. In roots which had developed large aerenchyma spaces, the diffusion in some regions was very fast, indistinguishable from the rate in water. In other regions the rate was as much as 100 times slower. Examination of frozen intact roots with the cryo-scanning electron microscope showed the presence of liquid filling some of the aerenchyma spaces, while other spaces of the same root contained air. X-ray microanalysis of the liquid (for oxygen) showed that the liquid was water with few detectable ions. Similar liquid was present in small intercellular spaces within the spoke-like radial files of cells between the large spaces, or between remnants of collapsed cell walls at the edges of the large spaces. It is proposed that regions of roots with high diffusivity are those in which some of the aerenchyma spaces are filled with water. In seeking the origin of this liquid, the progress of aerenchyma formation could be followed in the frozen tissues. The first change observed in a group of contiguous cells was a loss of vacuolar solutes and of cell turgor. Next the walls broke apart and collapsed back onto the surrounding turgid cells leaving a volume of ion-poor liquid. The liquid was probably not that found in some aerenchyma spaces of the mature roots, because the final stage of space formation was a loss of the liquid, leaving an air filled cavity surrounded by a composite lining formed from the collapsed walls of the broken cells. It is likely that the liquid in the spaces of mature aerenchyma is exuded from the remaining living cortical cells at times when the root turgor is high. This would be consistent with several recent studies which have shown periodic exudation of water from the surface of turgid roots. The spasmodic occurrence of root cortex tissue with enhanced diffusivity would have important implications for the transport of nutrient ions across the root.

Using these methods, it was found that ethylene and carbon dioxide (CO2) concentrations of soils varied considerably depending on the presence or absence of a rhizosphere. Ethylene was much higher (31–375 nL L−1; mean: 207) in non-cropped areas (i.e., soils without rhizosphere) than in the rhizosphere region (8–136 nL L−1; mean: 38) of a field in which maize or soybean were grown. On the other hand, CO2 concentrations were higher in rhizosphere than in non-rhizosphere soil, especially in pot experiments. The rate of ethylene decomposition was, however, much greater in rhizosphere soil (55 nL g−1 day−1) than in non-rhizosphere soil (34 nL g−1 day−1). Higher microbial activity was presumed to result in the decrease of ethylene concentration and the increase in CO2 in rhizosphere regions. The implications of these results in relation to the influence of ethylene in rhizosphere on plant growth, and the role of soil microbes on decomposition of ethylene is discussed.

    Notizen: Summary The Rp1 locus of maize is a complex rust resistance locus where multiple resistance genes are clustered. Rare recombination events between Rp1 genes or alleles can produce two or more detectable genes linked in coupling phase. Such ‘compound’ genes can then be manipulated as a single gene in breeding programs. Several compound Rp1 genes, each carrying two or three tightly linked resistance genes, were constructed to test their utility in controlling common rust. While none of the lines carrying single Rp1 genes were resistant to all of the characterized North American P. sorghi biotypes, most of the two component and all of the three component Rp1 complexes were resistant. The potential for utilization of compound resistance genes in other crop species is discussed.

    Notizen: Summary Embryogenesis was analyzed in wheat × maize hybrids using paraffin sectioning. Embryogenesis in wheat × maize hybrids is different from that in self-pollinated wheat plants. Development of the embryo is not accompanied by the formation of an endosperm. The endosperm nuclei remain free in the cytoplasm, fail to advance into the cellular stage, and degenerate at a later time. The antipodal cells quickly degenerate in the fertilized ovaries of wheat × maize hybrids similar to self-pollinated ovaries. The antipodal cells remain normal in unpollinated ovaries. The pre-embryo will abort if it is allowed to develop on the plant, because of a nutritional shortage in the absence of an endosperm. Therefore, embryo rescue is necessary for haploid production from a wheat × maize hybrids. Haploid polyembryos were obtained from spikelet culture of wheat × maize hybrids. The formation of polyembryos is due to the cleavage of the pre-embryo and the effect of 2,4-D. The frequency of haploid embryo production and plant regeneration is affected significantly by maize genotypes, but not by wheat genotypes. The concentration of 2,4-D affects only the size of the embryo.
