    Description: Interferon derived from human leukocytes, human fibroblasts, and mouse fibroblasts was found to inhibit the motility of cultured cells. It inhibits the tumor-induced motility of capillary endothelial cells as well as the spontaneous migration of other cell types. The ability of a given preparation of interferon to inhibit the motility of a given cell type is proportional to its antiviral activity in that particular cell type.     Description: Mononuclear cells from peripheral blood of normal humans, unselected spleen cells from patients with Hodgkin's disease, and selected T and non-T lymphoid cells from normal peripheral blood and from the spleens of Hodgkin's disease patients were examined for de novo synthesis and secretion of ferritin. After precipitation of labeled lysates and supernatants from unseparated and selected T cells with antiserum to human liver ferritin, two bands were visible on sodium dodecyl sulfate-polyacrylimide gel analysis. The two bands were detected in molecular weight regions 19,000 and 21,000, which are thought to represent the L and H subunits of the ferritin molecule, respectively. The slower band (subunit H) was more radioactive than the faster band (subunit L). The H subunit is found in greater amounts in the serum of some tumor patients, but its cellular origin has not been established.     Description: Using chromosome heteromorphisms and blood cell types as genetic markers, we demonstrated chimerism in a chi46,XX/46,XY true hermaphrodite. The pattern of inheritance of the chromosome heteromorphisms indicates that this individual was probably conceived by the fertilization, by two different spermatozoa, of an ovum and the second meiotic division polar body derived from the ovum and subsequent fusion of the two zygotes. This conclusion is based on the identification of the same maternal chromosomes 13, 16, and 21 in both the 46,XX and 46,XY cells of the patient.     Description: The coagulant activity of partially purified and delipidated tissue factor (TF) (coagulation Factor III) has previously been recovered by dialysis of the apoprotein after addition of mixed brain lipids and deoxycholate. Inclusion of cadmium chloride in the relipidation mixture greatly increases the recovered activity of highly purified TF from human placenta by promoting incorporation of TF into phospholipid vesicles; TF that had not been incorporated into vesicles showed no coagulant activity. Thus, TF must be present in a lipid bilayer for expression of coagulant activity.     Description: Mutual coupling among oscillators of an ensemble has been proposed to explain the precision of some circadian rhythms. Reciprocal triggering is one of the most familiar forms of mutual coupling in nervous systems, but it can at best produce only modest improvement in temporal precision.     Description: Data from adoption studies on handedness indicate that the effects of shared biological heritage are more powerful determinants of hand preference than sociocultural factors. Biological offspring were found to show nonrandom distributions of right- and non-right-handedness as a function of parental handedness; these distributions were consistent with the results fo previous family studies.     Description: A classical conditioning paradigm was used to demonstrate that male rats can learn to secrete luteinizing hormone and testosterone in anticipation of sexual activity. Sexually naive males were exposed to a neutral stimulus and then to a sexually receptive female once daily. After exposure to the paired stimuli for 14 trials, the neutral stimulus was as effective as the female in triggering luteinizing hormone and testosterone secretion.     Description: A method devised for conjugating liposomes with protein resulted in the binding of up to 200 micrograms of immunoglobulin G per micromole of lipid.     Description: A two-interval forced-choice procedure was used to study monocular detection of a briefly presented low-contrast sine-wave grating pattern. Uncertainty about which eye was stimulated degraded detection performance for stereo-blind observers but not for normal ones.     Description: Vasoactive intestinal peptide, a smooth-muscle relaxant neuropeptide with neurotransmitter properties, was relaxed during electrical field stimulation of guinea pig trachea. The amount released correlated with the degree of relaxation, and the release was blocked by tetrodotoxin. Prior incubation of the trachea with antiserum to vasoactive intestinal peptide reduced the relaxation.     Description: At several points during the growth of a clone of human embryonic lung fibroblasts in vitro, 100 to 200 cells were removed at random and the proliferative potential of each cell was determined. At each sample point, a wide variation in remaining population doubling ability was observed among the individual cells and the distributions of doubling potentials were distinctly bimodal. Furthermore, the two cells arising from a single mitosis differed in their ability to proliferate by as many as eight population doublings (256-fold in the number of cells produced).     Description: The teratogenicity of alcohol has been demonstrated in humans through clinical studies, behavioral studies, and epidemiologic studies, and in animals through controlled laboratory experiments. In humans exposed to alcohol during gestation the effects can range from fetal alcohol syndrome in some offspring of chronic alcoholic women to reduced average birth weight in offspring of women reporting an average consumption of two to three drinks or more per day. The behavioral effects of such exposure may range from mental retardation in children with fetal alcohol syndrome to milder developmental and behavioral effects in infants born to social drinkers. In animals, exposure to alcohol in utero may result in death, malformation, and growth deficiency as well as behavioral and developmental abnormalities.     Description: An experimental and theoretical investigation is made into the range and nature of the exposure of the nonsmoking public to respirable suspended particulates from cigarette smoke. A model incorporating both physical and sociological parameters is shown to be useful in understanding particulate levels from cigarette smoke in indoor environments. Observed levels of particulates correlate with the predictions of the model. It is shown that nonsmokers are exposed to significant air pollution burdens from indoor smoking.     Description: Immunomicrospheres are specially designed microscopic particles that have antibodies or similar molecules chemically bound to their surfaces. The antibody-coated microspheres react in a highly specific way with target cells, viruses, or other antigenic agents. Immunomicropheres may be synthesized so that they incorporate compounds that are highly radioactive, intensely fluorescent, magnetic, electron opaque, highly colored, or pharmacologically active. These various types of microspheres may be coated with pure, highly specific monoclonal antibodies obtained by the new hybridoma cell cloning techniques or with conventional antibody preparations. Some of the many present and potential applications for these new reagents are (i) new types of radioimmune or immunofluorescent assays, (ii) improved fluorescence microscopy, (iii) separation of cells on the basis of the fluorescent, electrophoretic, or magnetic properties of bound immunomicrospheres, (iv) markers for use in several types of electron or standard light microscopy, and (v) delivery of lethal compounds to specific undesirable living cells.     Description: Administration of physostigmine to normal volunteers produced significant elevations in plasma cortisol and beta-endorphin immunoreactivity as well as alterations in mood, cognition, and behavior. These observations might be explained by a cholinergically mediated stress syndrome. However, peak elevations in plasma beta-endorphin immunoreactivity (but not in plasma cortisol) were significantly correlated with physostigmine-induced increases in depression ratings.     Description: Four recombinant lambda phages containing nucleotide sequences complementary to a cloned human preproinsulin DNA probe have been isolated from human DNA. Restriction analyses in conjunction with Southern hybridizations reveal two types of gene sequences. One isolate of each type was subjected to complete nucleotide sequence determination. The sequences contain the entire preproinsulin messenger RNA region, two intervening sequence. 260 nucleotides upstream from the messenger RNA capping site, and 35 nucleotides beyond the polyadenylate attachment site.     Description: When young adults detected auditory stimuli at split-second intervals, different components of the event-related brain potentials showed markedly different speeds of recovery. The P3 component (latency 300 to 350 milliseconds) was fully recovered at intervals of less than 1.0 second, while the N1--P2 components (latencies 100 to 180 milliseconds) were markedly attenuated with stimulus repetition even at longer interstimulus intervals.     Description: The affinity of Hb Zurich for carbon monoxide is approximately 65 times that of normal hemoglobin. The carboxyhemoglobin content in serum from individuals with Hb Zurich ranged from 3.9 to 6.7 percent in nine nonsmokers and from 9.8 to 19.7 percent in six smokers.     Description: Two coronaviruses were isolated from brain material obtained at autopsy from two multiple sclerosis patients. The viruses were neutralized by serum and spinal fluid from these patients. Although most of the population have antibody to these virus isolates, multiple sclerosis patients have slightly higher concentrations of serum antibody than controls.     Description: Infants, aged 2 to 4 months, discriminated synthetic speech patterns that varied in duration of the formant transitions; this variation provides information sufficient to signal the phonetic distinction between a stop consonant and a semivowel in adult listeners. In addition, the discriminability of a given difference in transition duration was a function of both the particular stimulus values and the total duration of the syllable. This contextual effect occurred even though the information for syllable duration came after the transition information.     Description: Repeated electroconvulsive shock, applied to rats, induces a subsensitivity of dopamine autoreceptors located in the substantia nigra as indexed by single-unit electrophysiological techniques. This reduced sensitivity is time-dependent, since effects similar to those seen with repeated treatment were also observed when single electroconvulsive shock was followed by an appropriate treatment-free interval.     Description: Acid extract of human placental tissue contain, by both radioimmunoassay and radioreceptor assay, beta-endrophin-like material. Half of this material will not go through a 5000-dalton filter and on Sephadex G-200 has a molecular size between 25,00 and 50,000 daltons. Of the material going through a 5000-dalton ultrafilter, 80 percent is excluded on Sephadex G-25 and held back, very slightly, on Bio-Rad P6, indicating a molecular size of approximately 4500 to 4800 daltons.     Description: In a patient with gyrate atrophy of the choroid and retina, an arginine-deficient diet has reduced plasma ornithine concentration fivefold during the past 20 months. Subjective improvement in her visual function was noted approximately 15 months after institution of her diet. This has been documented by improvements in the electroretinogram, dark-adaptation, and color vision. The improvement involves rod and, to a lesser extent, cone function.     Description: A polymorphic HpaI endonuclease recognition site on the 3' side of the beta-globin gene was used to analyze the evolution of the beta-globin gene mutants S and C. Study of the world wide distribution of the normal and variant HpaI sites showed that the mutation which resulted in the variant 13.0-kilobase fragment arose in a localized region in West Africa. It predated the hemoglobin S and C mutations, both of which arose separately from a chromosome with the variant 13.0-kilobase HpaI site.     Description: The synthesis of a placental luteinizing hormone-releasing factor (pLRF), which is immunologically, physiochemically, and biologically indistinguishable from synthetic LRF, was demonstrated. The incorporation of 3H-labeled leucine by human placental tissue in vitro into pLRF was determined by purification on carboxymethyl-cellulose and specific immunoprecipitation of the 3H-labeled pLRF. The specific activity of the pLRF released into the medium increased 100-fold from day 1 to day 2 of culture and attained a concentration of 2.84 microcuries per microgram.     Description: Mothers among !Kung hunter-gatherers nurse briefly and frequently, with brief intervals between nursing bouts (mean +/- standard error, 13.19 +/- 1.28 minutes). The low levels of 17 beta-estradiol and progesterone in the serum of the mother are correlated with infant's age and with interbout interval, but not with total nursing time. Maternal gonadal function is apparently suppressed by a timing-dependent, prolactin-mediated effect of breast stimulation. Interbout interval may be a key variable in lactation infertility.     Description: Concentrations of norepinephrine in cerebrospinal fluid are higher in schizophrenic patients, particularly in those with paranoid features, than in normal volunteer subjects of the same age. This observation supports recent reports of elevated concentrations of norepinephrine in specific brain areas adjacent to the cerebral ventricles of paranoid schizophrenic patients. The electrode exhibits a super-Nernstian behavior and gives a mean pH response of 71.4 millivolts per [pH] (standard deviation, 5.2 millivolts). Uncorrected zero current potential values can be used to determine the pH value of the medium to within 0.012 pH.     Description: Both hybrids of mouse and human microcells and whole cell hybrids generated by the fusion of primary mouse cells and SV40-transformed human fibroblasts were used to establish the syntenic association of the murine cytoplasmic superoxide dismutase and the interferon sensitivity genes on mouse chromosome 16. This assignment adds two new markers to chromosome 16 and provides another example of an evolutionarily conserved linkage. It also shares the following properties with natural leukocyte interferon (IFN).     Description: The alpha-like and beta-like subunits of human hemoglobin are encoded by a small family of genes that are differentially expressed during development. Through the use of molecular cloning procedures, each member of this gene family has been isolated and extensively characterized. Although the alpha-like and beta-like globin genes are located on different chromosomes, both sets of genes are arranged in closely linked clusters. In both clusters, each of the genes is transcribed from the same DNA strand, and the genes are arranged in the order of their expressions during development. Structural comparisons of immediately adjacent genes within each cluster have provided evidence for the occurrence of gene duplication and correction during evolution and have led to the discovery of pseudogenes, genes that have acquired numerous mutations that prevent their normal expression. Recently, in vivo and in vitro systems for studying the expression of cloned eukaryotic genes have been developed as a means of identifying DNA sequences that are necessary for normal gene function.     Description: The sequence of a human leukocyte-derived complementary DNA (cDNA), Hif-2h, which directs the formation in Escherichia coli of a polypeptide, IFN-alpha 1, with interferon (IFN) activity has been described. A second IFN cDNA, Hif-SN206, which also elicits synthesis of a biologically active IFN, IFN-alpha 2, is described in this article. Whereas IFN-alpha 2 is twice as active on human as on bovine cells, IFN-alpha 1 is 10 to 20 times more active on bovine than on human cells. As deduced from the cDNA's, the messenger RNA's for the two IFN's differ in length and in 20 percent of the nucleotides; the mature IFN polypeptides differ in 17 percent of the amino acids. Both IFN-alpha 1 and IFN-alpha 2 differ from the lymphoblastoid IFN described by others.     Description: Numerous peptides appear to be neurotransmitter candidates in the brain. Some, such as the opioid peptide enkephalins, neurotensin, and substance P, were first isolaterd from the brain. Peptides, such as cholecystokinin and vasoactive intestinal polypeptide, were known as intestinal hormones and later recognized as brain constituents. Certain hypothalamic-releasing hormones, pituitary peptides, and blood-derived peptides like angiotensin II and bradykinin, may also be central neurotransmitters.     Description: Two major functionally distinct T cell subsets in man have been defined with heteroantiserums and monoclonal antibodies directed against stable cell surface antigens that appear during thymic ontogeny. A monoclonal antibody to T4 antigen (anti-T4) is reactive with the peripheral inducer T cell population while a monoclonal antibody to T5 antigen (anti-T5) is reactive with the cytotoxic and suppressor population. Immunoprecipitation and electrophoresis on sodium dodecyl sulfate polyacrylamide gel were used to show that on human thymocytes or peripheral T cells the T4 antigen is a single 62,000-dalton glycoprotein while the T5 antigen is a complex of two glycoproteins, one being 30,000 daltons and the other 32,000 daltons. Similar glycoproteins have been isolated with antibodies to murine Lyt 1 and Lyt 2,3 antigens.
