Notizen: Summary Managanese has been proven to be an essential trace element in animals since 1931. Today, it is known that manganese deficiency results in a wide variety of structural and physiological defects in animals. For humans, the safe and adequate range of intakes for manganese by enteral nutrition is 2.0 to 5 mg per day; for parenteral nutrition a range of 0.15 to 0.8 mg per day is proposed. The manganese content of components of enteral nutrition (EN) and total parenteral nutrition (TPN) solutions is determined using electrothermal atomic absorption spectrometry. Comparison is made between calculated and measured values. The manganese present in supplementary nutrition and flavor stuffs is also measured. The intake of manganese by EN is calculated based on the daily administration, and is found to vary from 192 to 7373 µg per day. In prepared TPN-solutions, the manganese contribution of all components before adding the Oligo Complex is found to be less than 0.2% of the total manganese content. After addition of the Oligo Complex mixture, a mean daily intake of 5.02±0.16 mg per day is found.

Notizen: Abstract TheA-value method, involving the application of a higher15N rate to a reference non-N2-fixing plant, was used to assess the magnitude of N2 fixation in two bambara groundnut cultivars at four growth stages [vegetative, 0–47 days after planting (DAP); early pod-filling, 47–99 DAP; mid-pod-filling, 99–120 DAP; physiological maturity, 120–148 DAP). The cultivars were Ex-Ada, a bunchy type, and CS-88-11, a slightly spreading type. They were grown on a loamy sand. Uninoculated Ex-Ada and CS-88-11 were used as reference plants to measure the N2 fixed in the inoculated bambara groundnuts. In this greenhouse study, soil was the major source of N in bambara groundnuts during vegetative growth, and during this period it accounted for over 80% of the N accumulaed in the plants. However, N2 fixation became the major source of plant N during reproductive growth. There were significant differences between the two cultivars in the ability to fix N2, and at physiological maturity, almost 75% of the N in CS-88-11 was derived from the atmosphere compared to 55% in Ex-Ada. Also, the total N fixed in CS-88-11 at physiological maturity was almost double that in Ex-Ada. Our data indicate that the higher N2 fixation in CS-88-11 was due to two factors, a higher intensity of N2 fixation and a longer active period of N2 fixation. The results also suggest that bambara groundnut genotypes could be selected for higher N2 fixation in farining systems.

Notizen: Summary Two Australian Acacia species, A. mangium and A. auriculiformis were inoculated in vitro with eight strains of Bradyrhizobium spp. and two strains of Rhizobium spp. On the two plant species, only Bradyrhizobium spp. strains formed effective N2-fixing nodules. A. mangium, which nodulates effectively with a restricted range of Bradyrhizobium spp. strains, is a specific host compared to A. auriculiformis. A. auriculiformis is assumed to be a promiscuous host because it nodulates effectively with a wide range of Bradyrhizobium spp. strains. Nodule efficiency as expressed by the ratio of N2 fixed to nodule dry weight appeared to be higher in A. auriculiformis (0.44–0.81) than in A. mangium (0.23–0.55).

Notizen: Summary TwoAtriplex spp. growing in low-fertility saline sodic soils were assayed for root-associated nitrogenase activity. The excised washed and unwashed root of the two species.A. lentiformis andA. amnicola, showed high root-associated nitrogenase activity. Acetylene-reducing activity seemed to be directly influenced by moisture. The highest number of diazotrophs, enumerated using a most probable number technique was observed on the root surface. Most of the isolated diazotrophs were identified asEnterobacter agglomerans. Root-associated nitrogenase activity inAtriplex spp. may explain the high protein and biomass content of these plants growing in low-fertility saline sodic soils.

Notizen: Summary In a 35-year-old calcareous beech forest stand five beech trees (Fagus sylvatica L.) were felled every 2 weeks, and xylem sap was obtained by means of water displacement from the lowest trunk sections, each 100 cm in length. From mid-October 1988 to mid-October 1989 a total of 130 trees were investigated. The seasonal variations of the Ca, Mg, K and Mn contents, as well as those of pH, show four characteristic phases. Additionally, distribution of the mineral contents along the trunk was studied in four trees. The seasonal increase and decrease of xylem sap mineral contents along the trunk is shown for the characteristic phenophases. The Ca, Mg, K, and Mn contents of xylem saps were determined by means of atomicabsorptionspectrophotometry.

Notizen: Summary Hydrogen uptake is thought to increase the efficiency of nitrogen fixation by recycling H2 produced by nitrogenase that would otherwise be lost by diffusion. Here we demonstrate the capacity of eight Rhizobium strains to take up molecular hydrogen. Uptake by nodule homogenates from Robinia pseudoacacia was measured amperometrically under nitrogenase repression. Markedly lower activities were found than in soybean nodules. In addition hydrogenase activity was detected by the ability of bacteroids to reduce methylene blue in the presence of hydrogen. It was demonstrated that hydrogenase structural genes are present in the black locust symbiont, Rhizobium sp. strain R1, using hybridization with a plasmid, which contained hydrogenase genes from R. leguminosarum bv. viceae.

Notizen: Summary Pot-culture studies were carried out to examine the response of barley (Hordeum vulgare L.) to inoculation with Azospirillum brasilense and Glomus versiforme, singly and/or in combination, under varying levels of nitrogenous [(15NH4)2SO4] and soluble phosphatic (single superphosphate) fertilizers. The interaction between both the endophytes led to increased growth and nutrition of the barley plants. Roots from plants inoculated with Azospirillum brasilense and Glomus versiforme exhibited very low acetylene reduction activity. N2 fixation in the plants increased with the increase in plant growth but the mycorrhiza alone gave a low level of N2 fixation in the plants compared to combined inoculation with both the endophytes.

Notizen: Summary We studied the dominant diazotrophs associated with maize roots and rhizosphere soil originating from three different locations in France. An aseptically grown maize plantlet, the “spermosphere model”, was used to isolate N2-fixing (acetylene-reducing) bacteria. Bacillus circulans was the dominant N2-fixing bacterium in the rhizosphere of maize-growing soils from Ramonville and Trogny, but was not found in maize-growing sandy soil from Pissos. In the latter soil, Enterobacter cloacae, Klebsiella terrigena, and Pseudomonas sp. were the most abundant diazotrophs. Azospirillum sp., which has been frequently reported as an important diazotroph accociated with the maize rhizosphere, was not isolated from any of these soils. The strains were compared for their acetylene-reducing activity in the spermosphere model. The Bacillus circulans strains, which were more frequently isolated, also exhibited significantly greater acetylene-reducing activity (3100 nmol ethylene day-1 plant-1) than the Enterobacteriaceae strains (180 nmol ethylene day-1 plant-1). This work indicates for the first time that Bacillus circulans is an important maizerhizosphere-associated bacterium and a potential plant growth-promoting rhizobacterium.

Notizen: Summary Variation in nodulation and N2 fixation by the Gliricidia sepium/Rhizobium spp. symbiosis was studied in two greenhouse experiments. The first included 25 provenances of G. sepium inoculated with a mixture of three strains of Rhizobium spp. N2 fixation was measured using the 15N isotope dilution method 12 weeks after planting. On average, G. sepium derived 45% of its total N from atmospheric N2. Significant differences in fixation were observed between provenances. The percentage of N derived from atmospheric N2 ranged from 26 to 68% (equivalent to 18–62 mg N plant-1) and was correlated with total N in the plant (r=0.70; P=0.05). The second experiment included six strains of Rhizobium spp. and two methods of inoculation and the plants were harvested 14,35 and 53 weeks after planting. In the first harvest significant differences were found between the number of nodules and the percentage and amount of N2 fixed. There was also a significant correlation between the number of nodules and the amount of N2 fixed (r=0.92; P=0.05). In the final harvest no correlation was observed, although there were significant differences between the number of nodules and the percentage of N derived from the atmosphere. The amount of N2 fixed increased with time (from an average of 27% at the first harvest to 58% at the final harvest) and was influenced by the Rhizobium spp. strain and the method of inoculation. It ranged from 36% for Rhizobium sp. strain SP 14 to 71% for Rhizobium SP 44 at the last harvest. Values for the percentage of atmosphere derived N2 obtained by soil inoculation were slightly higher than those obtained by seed inoculation.

Notizen: Summary Virgin alkali (sodic) soils have a high pH and high exchangeable Na and are often barren. Blue-green algae, however, tolerate excess Na and grow extensively on the soil surface in wet seasons. Experiments using a highly degraded alkali soil (silt loam, pH 10.3, electrical conductivity 3.5 dS m-1, 90% exchangeable Na) were conducted in soil columns, with or without gypsum, in order to study the influence of waterlogging on the growth of indigenous and inoculated blue-green algae and hence, soil reclamation. The growth of indigenous blue-green algae was initially slow in alkali soil, due to the high pH and exchangeable Na, and depressed in gypsum-amended soil, due to excess Ca. Inoculation hastened the establishment of blue-green algae in both the unamended alkali soil and the gypsum-amended soil, overcoming the adverse influence of excess Na in the former and excess Ca in the latter. Gypsum was effective in amelioration (pH 9.05, electrical conductivity 1.2 dS m-1, 41% exchangeable Na after 11 weeks) but blue-green algae were ineffective even after 17 weeks. In combination with gypsum, blue-green algae had no additional effect, and the C and N increases due to the growth of indigenous or inoculated blue-green algae were insignificant. Alkali soil reclamation by biological methods requires mobilization of Ca from native soil calcite and the exchange of Ca for Na in the exchange complex. The ineffectiveness of blue-green algae was ascribed to their inability to mobilize Ca. It is argued that current theories favouring blue-green algae as a biological amendment to bring about alkali soil reclamation are untenable and are not comparable with an effective chemical amendment such as gypsum.

Notizen: Summary Acetylene reduction activity by Azospirillum brasilense, either free-living in soils or associated with wheat roots, was determined in a sterilised root environment at controlled levels of O2 tension and with different concentrations of mineral N. In an unplanted, inoculated soil nitrogenase activity remained low, at approximately 40 nmol C2H4 h-1 per 2kg fresh soil, increasing to 300 nmol C2H4 h-1 when malic acid was added as a C source via a dialyse tubing system. The N2 fixation by A. brasilense in the rhizosphere of an actively growing plant was much less sensitive to the repressing influence of free O2 than the free-living bacteria were. An optimum nitrogenase activity was observed at 10 kPa O2, with a relatively high level of activity remaining even at an O2 concentration of 20 kPa. Both NO inf3 sup- and NH inf4 sup+ repressed nitrogenase activity, which was less pronounced in the presence than in the absence of plants. The highest survival rates of inoculated A. brasilense and the highest rates of acetylene reduction were found in plants treated with azospirilli immediately after seedling emergence. Plants inoculated at a later stage of growth showed a lower bacterial density in the rhizosphere and, as a consequence, a lower N2-fixing potential. Subsequent inoculations with A. brasilense during plant development did not increase root colonisation and did not stimulate the associated acetylene reduction. By using the 15N dilution method, the affect of inoculation with A. brasilense in terms of plant N was calculated as 0.067 mg N2 fixed per plant, i.e., 3.3% of the N in the root and 1.6% in the plant shoot were of atmospheric origin. This 15N dilution was comparable to that seen in plants inoculated with non-N2-fixing Psudomonas fluorescens.

Notizen: Summary N2(C2H2) fixation by Ulex gallii Planchon (dwarf or autumn flowering gorse/furze) seedlings was determined following 8 months of growth (December-August) in the glasshouse in a very acid, N- and P-deficient forest soil. Application of Na2HPO4·12H2O or North African ground rock phosphate fertilizer was essential for growth, nodulation and C2H2 reduction activity. Overall, both the sodium phosphate and the rock phosphate were equally effective P sources and the maximum acetylene reduction by intact roots was measured as 4.09 and 4.69 μmol C2H4g-1 fresh weight nodule h-1, respectively. Applied NH4Cl severely inhibited nodulation and restricted acetylene reduction activity but not seedling growth. The results are discussed in relation to the spread of U. gallii in the south of Ireland and its potential as a leguminous nurse crop for Sitka spruce on the very impoverished forest soils of the region.

Notizen: Summary Barley-field pea intercrops have been shown to increase N yield when grown under cryoboreal subhumid conditions. In this study, we extended previous research by testing the hypotheses that (1) the intercropped field pea fixes a greater proportion of its shoot and root N than does sole-cropped field pea; (2) N is transferred from the annual legume to the cereal during the growing season; and (3) root production is greater under intercropped than sole-cropped conditions. Unconfined microplots seeded to barley, field peas, or a barley-field pea intercrop were fertilized with N at 10 kg ha-1 as (NH4)2SO4 (5.21 atom % 15N excess). Both the intercropped and sole-cropped barley derived more than 93% of their N from the soil. In contrast, 40% of N in the intercropped field pea was derived from soil. This study provided no evidence for transfer of N from the legume to the cereal. On average, the proportion of N derived from air by both pea intercrops was 39% higher than that derived by the sole-cropped pea. Root length determined by a grid intersection method following digitization using an image analyzer tended to be higher under intercropping than in sole crops. We conclude that even on fertile soils benefits may accrue from annual intercropping that includes a legume. The benefits arise from (1) increased N production, (2) greater N-fixation efficiency, and/or (3) more shoot and root residue-N mineralization for subsequent crops.

Notizen: Summary Wheat seedlings, treated with the auxine 2,4-dichlor-phenoxy acetic acid (2,4-D) during germination developed only a residual root system. Root elongation was extremely restricted and root tips were deformed to thick club-shaped tumours. When 2,4-D was added in a later stage of plant growth the plants developed additional nodule-like knots along primary roots. Root and shoot dry-matter production was slightly repressed in all 2,4-D treatments and N translocation from roots to shoots was repressed as well. When transferred to an auxine-free growth medium, the 2,4-D-affected roots were not capable of complete recovery. In plants inoculated gnotobiotically with Azospirillum brasilense, either with the wild type or with the NH 4 + -excreting mutant strain C3, a 2,4-D addition increased rhizosphere acetylene-reduction activity at pO2 1.5 kPa. The O2 sensitivity of root-associated nitrogenase activity tended to be reduced. The number of root-colonizing bacteria, at approximately 108 colony-forming units (cfu) per g dry root, was similar in the 2,4-D treatments and untreated controls. Plant treatment with high concentrations of the chemical isomer 3,5-dichlor-phenoxy acetic acid (3,5-D) did not have comparable effects, either on plant development or on rhizosphere-associated nitrogenase activity. Root-tumour tissue inhabited by A. brasilense showed purple staining when subjected to a tetrazolium chloride solution, which may indicate intensive local nitrogenase activity in this tissue. Exposed to an 15N2-enriched atmosphere, plants treated with 2,4-D and with A. brasilense incorporated significantly higher amounts of 15N than untreated controls. In all cases the highest values of 15N enrichment were found following inoculation with the NH 4 + -excreting mutant strain C3.

Notizen: Summary The effect of length of dry storage period and subsequent moist incubation on the availability of Mn was examined in a calcareous soil. Increasing the time of dry storage (for up to 4 years) generally increased the availability of Mn as determined by plant growth and Mn concentration in wheat and barley. Moist incubation of stored soil had variable effects on Mn availability depending on how long the soil had been stored before use and on the method used to assess Mn availability. When assessed by Mn concentration in plant tissues, increasing the moist incubation time (from 0 to 30 days) of soil stored dry for 4 years increased Mn availability in soil initially and thereafter decreased it. However, incubation time had little effect on Mn availability in soil stored for only 1 year or soil used fresh from the field. When Mn availability was assessed using a chemical extractant (DTPA; diethylenetriaminepentaacetic acid), both soils showed an initial increase in Mn availability immediately on wetting. However, Mn availability in the soil stored for only 1 year decreased rapidly and remained consistently below that of soil stored for 4 years. In the latter soil, Mn availability also decreased but only after a few days. Microbial studies indicated that there was a decrease in the ratio of Mn oxidising to Mn reducing microorganisms with increasing storage time. Inoculation of one soil with another suggested that the factor responsible for the low Mn availability in soils stored for a short period could be transferred to soils stored for a longer period. These results suggest that the change in Mn availability in a calcareous soil with dry storage is a result of changes in microbial populations that cannot be fully restored by moist incubation.

Notizen: Summary Tree species differ in the quantity and quality of litter produced, and these differences may significantly affect ecosystem structure and function. I examined the importance of tree species in determining earthworm densities in replicated stands of Eucalyptus saligna Sm. and Albizia falcataria (L.) Fosberg, and in mixed stands (25% albizia and 75% eucalyptus). Mean earthworm densities ranged from 92 m-2 in the pure eucalyptus, to 281 m-2 in the mixture, and a maximum of 469 m-2 in the pure albizia stands. Only two earthworm species were present, Pontoscolex corethrurus and Amynthas gracilis. Leaf biomass on the forest floor was highest in the pure eucalyptus and lowest in the pure albizia stands, whereas the annual fine litterfall production was lowest in the pure eucalyptus and highest in the albizia stands. The N content of fine litterfall was correlated positively with earthworm density, and the fine litterfall biomass: N ratio was correlated negatively with earthworm density. Greater leaf biomass on the forest floor under eucalyptus stands despite lower rates of litterfall suggests that litter quality, rather than litter quantity, was primarily responsible for the greater earthworm density in the albizia stands. Some biogeochemical effects of tree species in the tropics may be mediated through effects on earthworm populations.

Notizen: Summary In three field trials conducted during the summer season of 1986, 1987 and 1989 in an alkaline soil, 17 accessions of annual Sesbania spp. were evaluated for nodulation, N2 fixation (acetylene reduction assay), dry weight of roots and shoots, woody biomass production, and nutrient uptake. At 50 days after sowing all the accessions were effectively nodulated (average 36.4 root nodules plant-1) with a high nodule score (3.4). There was a lot of variation in nodule volume and mass and in acetylene reduction activity but not in N content (5.2%). N uptake in shoots, roots and nodules averaged 639, 31, and 13 mg plant-1, respectively, and much of the fixed N remained in shoots. Accessions of ‘S. cannabina’ complex performed better than others. S. rostrata had poor root nodulation but exhibited excellent stem nodulation (300 nodules plant-1) even though not inoculated with Azorhizobium sp. Average concentrations of N, P, K, S, Ca, and Mg in the shoots were high, at 3.2, 0.28, 1.5, 0.28, 1.5, and 0.4% respectively, and Na was low (0.15%), reflecting the usefulness of Sesbania spp. as an integrated biofertilizer source. Green matter production was 26.0 Mg ha-1 (5.9 Mg dry matter) and N uptake was 158 kg ha-1, 54 days after sowing. Average woody biomass of six accessions at maturity, 200 days after sowing, was high (19.9 Mg ha-1), showing its potential for shortterm firewood production. Total nutrient uptake for production of woody biomass (200 days of growth) was no more demanding than growing the plant to the green-manuring stage of 50–60 days' growth.

Notizen: Summary Dissimilarities in soil N uptake between N2-fixing and reference non-N2-fixing plants can lead to inaccurate N2 fixation estimates by N difference and 15N enrichment methods. The natural 15N abundance (δ 15N) method relies on a stabilized soil 15N pool and may provide reliable estimates of N2 fixation. Estimates based on the δ 15N and differences in N yield of nodulating and non-nodulating isolines of soybean were compared in this study. Five soybeans from maturity groups 00, IV, VI, and VIII and their respective non-nodulating isolines were grown at three elevations differing in ambient temperature and soil N availability. Despite large differences in phenological development and N yield between the non-nodulating isolines, the δ 15N values measured on seeds were relatively constant within a site. The δ 15N method consistently produced lower N2 fixation estimates than the N difference method, but only in three of the 15 observations did they differ significantly. The average crop N derived from N2 fixation across sites and maturity groups was 81% by N difference compared to 71% by δ 15N. The magnitude of difference between the two methods increased with increasing proportions of N derived from N2 fixation. These differences between the two methods were not related to differences in total N across sites or genotypes. The low N2 fixation estimates based on δ 15N might indicate that the nodulating isolines had assimilated more soil N than the non-nodulating ones. A lower variance indicated that the estimates by N difference using non-nodulating isolines were more precise than those by δ 15N. Since the differences between the estimates were large only at high N2 fixation levels (low soil N availability), either method may be used in most situations when a non-nodulating isoline is used as the reference plant. The δ 15N method may have a comparative advantage over N difference and 15N enrichment methods in the absence of a suitable non-N2-fixing reference plant such as a non-nodulating isoline.

Notizen: Summary The objective of this study was to assess the number and effectiveness of peanut rhizobia in soils of the major peanut-growing areas of Thailand. Three cropping areas, (1) continuously cropped with peanuts, (2) continuously cropped with non-legumes, and (3) non-cultivated fields, were chosen in each region. Peanut rhizobia were found in the soil at 38 to 55 sites sampled. Cultivated fields with a peanut cultivation history contained (as estimated by most probable numbers) an average of 1.6×103 cells g-1 of soil. The numbers of peanut rhizobia in most of the fallow fields and some of the noncultivated shrub or forest locations were much the same as at the sites where Arachis hypogaea was cultivated. In contrast, there were no or few (28–46 cells g-1 soil) peanut rhizobia in the majority of fields continuously cultivated with sugarcane, cassava, corn, and pineapple. It appears that in these areas the indigenous peanut rhizobial populations are not adequate in number for a maximal nodulation of peanuts. A total of 343 Bradyrhizobium isolates were tested for effectiveness and were found to vary widely in their ability to fix N2. In some areas the majority of rhizobia were quite effective while in others they were less effective than the inoculum strain THA 205 recommended in Thailand.

Notizen: Abstract A field experiment was conducted to obtain the N balance sheet for sole crops and intercrops of sorghum [Sorghum bicolor (L.) Moench] and pigeonpeas [Cajanus cajan (L.) Millsp.]. Intercropping gave a significant advantage over sole cropping in terms of dry matter production and grain yield, as calculated on the basis of the land equivalent ratio and area-time equivalent ratio. The N fertilizer use efficiency and atmospheric N2 fixation by pigeonpea were estimated using 15N-labeling and natural abundance methods. The N fertilizer use efficiency of sorghum was unaltered by the cropping system, while that of the pigeonpea was greatly reduced by intercropping. Although intercropping increased the fractional contribution of fixed N to the pigeonpeas, no significant difference was observed between the cropping systems in total symbiotically fixed N. There was no evidence of a significant transfer of N from the pigeonpea to the sorghum. This study showed that use of soil N and fertilizer N by pigeonpeas was almost the same as that by sorghum in sole cropping, indicating the potential competence of pigeonpeas to exploit soil N. However, when N was exhausted by a companion crop in intercropping, the pigeonpea crop increased its dependency on atmospheric N2 fixation. We conclude that knowledge of how N from different sources is shared by companion crops is a prerequisite to establishing strategies to increase N use, and consequently land productivity, in intercropping systems.

Notizen: Abstract Pot experiments were conducted with two soils, from Rottenhaus and Seibersdorf in Austria, to ascertain whether the rate of fertilizer N application and the test crop would influence the amount of N available in the soil as assessed by the A-value method. 15N-labelled fertilizer was applied at rates of 10, 25, 40, 60, and 100 mg N kg-1 soil, corresponding approximately to 20, 50, 80, 120 and 200 kg N ha-1 respectively, and two crop species, barley (Hordeum vulgareL.) and non-nodulating soybean (Glycine max L.) were used to determine the soil A N value under the various fertilizer regimes. The results showed that the Rottenhaus soil had a higher A N value than the Seibersdorf soil, suggesting that the former was more fertile than the latter. The A N values of both soils were significantly affected by the level of N application. When grown in the same soil, the two test crops showed significantly different fertilizer use efficiency and per cent N derived from fertilizer when the rate of N application exceeded 20 kg ha-1. Thus, the A N value as determined by the two test crops differed significantly for the same soil when the rate of N application was greater than 20 kg/ha. The difference was greater when the soil fertility level was high. The dependence of the A N value on the level of N application and the species of crop seriously compromises the suitability of this method for determining plant-associated N2 fixation. Hence, considerable caution is required when using this method to estimate plant-associated N2 fixation.

Notizen: Abstract Maize seedlings develop nodule-like tumour knots (para-nodules) along primary roots when treated with the auxin 2,4-dichlor-phenoxy-acetic acid (2,4-D). Inoculated NH 4 + -excreting Azospirillum brasilense cells were shown to colonize these tumours, mostly intracellularly, promoting a high level of N2 fixation when microaerophilic conditions were imposed. The nitrogenase activity inside the para-nodules was less sensitive to free O2 than in non-para-nodulating roots. Both light and electron microscopy showed a dense bacterial population inside intact tumour cells, with the major part of the cell infection along a central tumour tissue. The bacteria colonized the cytoplasm with a close attachment to inner cell membranes. In an auxin-free growth medium, young 2,4-D-induced para-nodules grew further to become mature differentiated root organs in which introduced bacteria survived with a stable population. These results provide evidence that gramineous plants are potentially able to create a symbiosis with diazotrophic bacteria in which the NH 4 + -excreting symbiont will colonize para-nodule tissue intracellularly, thus becoming well protected.

Notizen: Summary Bradyrhizobial strain BTAi 1 nodulates both stems and roots of Aeschynomene spp. Previous work has shown that it contains bacteriochlorophyll a and forms photosynthetic reaction centers, and has provided indirect evidence of photosynthesis by bacteroids within stem nodules. Here we report physiological and biochemical characteristics of BTAi 1 ex planta, which also suggest the presence of photosynthetic activity. Light-stimulated uptake of 14CO2 by BTAi 1 was detected at all stages of growth. Inhibitors of photosynthesis, 1,10-orthophenanthroline and 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), and the uncoupler NH4Cl, immediately suppressed light-driven 14CO2 uptake and increased O2 uptake. BTAi 1 is strictly aerobic and was unable to grow without organic C even in the light; also, it was unable to grow chemoautotrophically in an atmosphere enriched with H2 and CO2. In micro-aerobic conditions, strain BTAi 1 expressed acetylene reducing activity ex planta in an N-free medium. The highest rates of light-stimulated 14CO2 uptake and acetylene-reducing activity occurred during the exponential and early stationary phases of growth. Acetylene-reducing rates at a low glucose concentration were increased following a light-dark cycle in comparison with continuous dark conditions.

Notizen: Abstract A field experiment was conducted to investigate the effects of 1,2,3,4,5,6-hexachlorocyclohexane (BHC), phorate, carbofuran, and fenvalerate, at their recommended doses, on some chemical and microbiological properties of the rhizosphere soil in relation to rice yields. In general, the insecticides had a beneficial effect on rhizosphere soil properties. Carbofuran strongly stimulated the mineralization of organic C. BHC and phorate led to the retention of less total N in the soil. BHC released more NH inf4 sup+ -N than the other insecticides. Phorate, however, liberated the most NO inf3 sup- -N. Phorate and fenvalerate released more available P than BHC and carbofuran did. All the insecticides stimulated the proliferation of aerobic non-symbiotic N2-fixing and phosphate-solubilizing microorganisms, resulting in an overall increase in rice yield. BHC had the greatest effect on rice yields, followed by phorate.

Notizen: Summary Experiments with black locust (Robinia pseudoacacia L.) seedlings grown under strictly controlled laboratory conditions indicated that the availability of nitrate has a marked impact on nitrogen fixation. When nitrate concentrations were very low, both nodulation and seedling growth were impaired, whereas nitrate concentrations high enough to promote plant growth strongly inhibited symbiotic nitrogen fixation. When nitrate was added to the growth medium after infection, nodulation and nitrogen fixation of the seedlings decreased. This effect was even more marked when nitrate was applied before infection with rhizobia. Higher nitrogen concentrations also reduced nodule number and nodule mass when applied simultaneously with the infecting bacteria. The contribution of symbiotic nitrogen fixation to black locust shoot mass by far exceeded its effects on shoot length and root mass. When nitrate availability was very low, specific nitrogen fixation (i. e. nitrogenase activity per nodule wet weight) was improved with increasing nitrogen supply, but rapidly decreased with higher nitrogen concentrations.

Notizen: Summary Four varieties of each paddy and wheat crops were grown in pots to see their cation exchange capacities of roots and their relationships with the uptake of nutrients by shoot and grain at different physiological growth stages. The results are summarised, as follows: (i) The cation exchange capacity of roots was maximum at tillering stage which continued to decrease with an increase in the age of plants. At early stages of plant growth, significant differences in the root CEC of different varieties of paddy and wheat were recorded but the differences became almost narrow in later stages of plant life and ultimately the CEC of roots became almost similar, in all the varieties as the crops reached to maturity. Higher root CEC of paddy varieties were observed than wheat varieties throughout their physiological growth. (ii) Uptake of phosphorus, potassium, iron and manganese by shoot and grain was found significantly and positively correlated with the CEC of roots in most of the varieties of both, paddy and wheat crops throughout their plant life.

Notizen: Abstract It was observed in a long-term fertility study that P application results in better nodulation and seed yield. The N yield per plant increased significantly with P, K, and B application, but the promoting effect of K and B on N content per plant was not reflected in the final yield. Data on mean nutrient uptake indicate that about 63% of N, 68% of P, 23% of K, 4% of Ca and 24% of Mg were present in the kernels, the remainder being in the haulm and shells. Soil will be exhausted fast if the nutrient rich groundnut residues are completely exported from the field and not compensated by adequate fertilizer application. Judicious use of fertilizers on groundnut has been shown to yield more than 100 kg of biologically fixed N per hectare.

Notizen: Summary Studies under growth cabinet conditions investigated the effect of source and concentration of nitrogen and timing of nitrogen application on the growth and nitrogen fixation byLotus pedunculatus cv. Maku andTrifolium repens cv. S184. KNO3, NaNO3 and NH4NO3 were added at transplanting at the following rates: 3.33, 7.78 and 13.33 mg N/plant. KNO3 was added at 3.33 and 7.78 mg N/plant at 0, 6, 12, 18, 24 or 30 days after transplanting. Lotus shoot weight increased with all increasing nitrogen sources but clover only responded to KNO3 and NaNO3. The root weight of both species increased with increasing KNO3 and NH4NO3. The percentage increase in lotus and clover shoot growth was greater than that of root growth when KNO3 was added within a week of transplanting. Increases in growth by both species resulted from added nitrogen except with lotus when NaNO3 was applied where increased nitrogen fixation also contributed to increased growth. Weight and number of effective nodules on both species were increased with 3.33 mg N per plant as KNO3 but nitrogen fixation was not affected. Addition of 13.33 mg N as NaNO3 reduced weight and number of effective nodules in both species and also nitrogen fixation by lotus. KNO3 increased growth and nodulation of both species when applied within one week after transplanting. Nodulated lotus plants responded to KNO3 by increasing growth but not nodulation. KNO3 appeared to affect infection and development of nodules on lotus and may affect the growth of existing nodules on clover.

Notizen: Summary Experiments in the laboratory, glasshouse and field during 1975–78 tested manganous oxide as a seed-pellet additive for controlling deficiency in sugar-beet seedlings. There was no experimental evidence that manganous oxide in the seed pellet was ever harmful to seedling establishment. On the contrary, germination tests in the laboratory and experiments in the glasshouse indicated that, in certain conditions, manganous oxide may improve plant establishment even when plants are not likely to be deficient, probably by accelerating germination of seed and emergence of seedlings. In field experiments where sugar beet were severely deficient, the plants were heavier and contained more manganese on plots sown with seed pelleted with material containing 50% manganous oxide than on plots sown with ordinary pelleted seed. Applying a foliar spray of manganese sulphate during the third week of June in addition to pelleting the seed with material containing manganous oxide gave bigger yields than either the seed-pellet treatment or foliar spraying alone.

Greenleaf was studied in a pot experiment. After ten weeks growth, the highest moisture stress (20 per cent water holding capacity) significantly reduced only the top weight of both plants. Moisture stress progressively retarded top growth in the two legumes. Similar trends were also observed in defoliated plants. Moisture stress had little or no effect on the nodulation or nitrogenase activity of the plants.

Notizen: Summary The effect of acid irrigation on the growth of rooted cuttings ofPicea abies (L.) Karst, was investigated in a pot experiment lasting 3 years. It involved two clones of Norway spruce, H 253 Bogstad I and H 254 Bogstad II. Irrigation water of pH 5.4, 4.0, 3.0 and 2.5 was used. Liming was included in the experiment. After the experimental period, the plants of all treatments were growing reasonably well. However, those plants irrigated at pH 2.5 were slightly discoloured. The plant mortality was only 3% throughout the experiment, and was not connected to acid irrigation. The limiting growth factor was N. All other nutrient elements measured in the plants were close to optimal concentration. Plants irrigated at pH 2.5, and to some extent at pH 3, contained excessively high concentrations of Al, t-S and SO4. The total amount of Ca, Fe and Mn taken up by the plants decreased with increasing soil acidity. The increased growth of clone H 254 relative to H 253, produces a corresponding impression on soil characteristics. Soil acidity is governed by acid irrigation and CaCO3 application, but the clonal effects are also of importance. Norway spruce appears to be tolerant to Al concentrations as high as 50 mmol/kg in the needles.

Notizen: Summary A study has been made of the influence of waterlogging on the distribution of trace metals between the various reservoirs in which they are held in a soil. Selective extractants have been used to remove metals held in 4 ways: soil solution and exchangeable; specifically adsorbed by inorganic sites; adsorbed or chelated by organic sites; adsorbed onto oxide surfaces. Waterlogging over a period of 16 weeks resulted in the release of both manganese and iron from the organic — and oxide — bound reservoirs to the soluble, exchangeable and inorganic reservoirs. Addition of both 1% dried grass (as an actively decomposing organic material) and 1% CaCO3 to the soil resulted in an acceleration of the metal redistribution. For manganese, selective extraction methods accounted for the distribution of all the metal in the reservoirs studied. In the case of iron however, there appeared to be some release from a reservoir not being extracted.

Notizen: Summary An all glass tight growth chamber, entirely sterilizable, has been constructed to carry out axenic and gnotobiotic cultures of rice plants (Oryza sativa L.). When grown in liquid medium and in the absence of combined nitrogen but in the presence of the diazotrophsAzotobacter vinelandii andRhodopseudomonas capsulata, rice plants exhibited a complete biological cycle from germination up to ear stage, during a period of time similar to the one encountered in french paddy soil of Camargue. In one experiment, mannitol was given to rice culture medium together withAzotobacter vinelandii andRhodopseudomonas capsulata. In another experiment, mannitol was not given together with Rhodopseudomonas, and still positive nitrogen gain was obtained, although it was less than culture with mannitol. When15N labeled cells of Rhodopseudomonas were added in rice culture medium,15N was partly transferred to rice plant. Among the nitrogen substances excreted from the bacteria in the rhizosphere medium, large organic molecules were shown to be the most abundant in our experimental conditions. Moreover, the concentration of free ammonia or aminoacids present in the rice rhizosphere were always compatible with a bacterial nitrogenase activity.

Notizen: Summary The effect of CaCO3 and organic matter on the availability of iron and manganese was studied on sorghum (Sorghum bicolor). Addition of calcium carbonate caused significant decrease in dry matter and grain yield. But yields tended to increase with the addition of organic matter at the rate of 15 t/ha. The iron concentration decreased with the addition of CaCO3 at all the stages of crop growth. Like iron, concentration of manganese also decreased with the increase in added CaCO3. Organic matter increased the iron concentration both at seedling and flowering stage. The concentration of manganese was, however, dependent on the stage of the crop.

Notizen: Summary Acetylene reduction activity and nitrogen accumulation in the plant top per unit nodule mass were compared among peanut, cowpea and siratro plants nodulated by six different strains of Rhizobium. Peanut was found to have several fold higher values than cowpea and siratro for both parameters for all strains of Rhizobium which nodulated it effectively, but the bacteroid content of the peanut nodules was similar to those of cowpea and siratro.

Notizen: Summary Vertisols from field plots fertilized continuously with P, K, FYM alone and in combination at the rate of 60 kg P2O5, 30 kg K2O and 1.5 ton FYM per ha were tested for different forms of Fe and Mn as a consequence of continuous cropping with sorghum and wheat. The transformations that occurred in different forms of plant-available iron were greater than those of Mn. Exchangeable and easily reducible Fe contents were markedly influenced by treatments. In manganese, transformations of easily reducible to exchangeable forms were conspicuous.

Notizen: Summary Boron, copper, iron, manganese and zinc concentrations were measured in sunflower leaves from plants grown hydroponically in a closed continuous flow system and with boron concentrations ranging from deficient to toxic. Leaves were analyzed at the stage of flower development. There was a highly significant inverse correlation between B concentration in solution and Mn concentration in leaves. Cu, Fe and Zn concentrations in the leaves were not changed by the different B levels, although the B concentration in leaves increased with an increasing concentration in solution, showing a close correlation each other.

Notizen: Summary Ectomycorrhizae were found in root samples of the treesAfzelia quanzensis Welw. andBrachystegia spiciformis Benth. (Caesalpiniaceae), collected in the coastal miombo type woodland 50 km west of Dar-es-Salaam, Tanzania. Root nodules with a structure resembling that of nitrogen-fixing root nodules of other leguminous plants were observed in theA. quanzensis material. The climate of the locality is rather dry, and strongly seasonal. In the tropics, ectomycorrhizae have previously been found only in humid or rain forest climate zones.

Notizen: Summary The literature does not contain a field report of nodulation for the xerophytic tree legumes of the genus Prosopis despite their widespread occurrence in semi-arid regions of the world. A phraetophytically simulated greenhouse experiment was conducted with mesquite in a 3m deep soil column to determine if nodules would form in deper depths where moisture is more abundant. The upper 0.5m in the soil column was allowed to dry to 2200 kPa but the 3.2 m depth was maintained at soil moistures more positive than 70 kPa by water additions to the bottom of the soil column. Over 100 nodules and an acetylene reduction rate of 1.9 mg/h were observed at the 3.2m depth. Nodulation or acetylene reduction were not observed closer than 2.7m from the surface. Air temperatures during these assays exceeded 45°C. Leaf xylem water potentials were in the 2800–3500 kPa range.

Notizen: Summary Nitrogen fixing trees and shrubs may be useful in revegetation efforts. Speculation that insufficient endophyte populations in surface soils may limit non-leguminous symbiotic nitrogen fixation in marginal land was explored.Purshia tridentata andP. glandulosa seedlings were grown in greenhouse trials using ten soils from nativePurshia sites. Treatments include a control, an inoculated treatment, and six mmole nitrogen amendment. When inoculated with aP.tridentata crushed nodule inoculum, two of five non-nodulating soils and three sparsely nodulating soils produced well nodulated plants. Inoculation also increased nodule mass, total nitrogen, nitrogen content and shoot dry mass in plants from some of the soils. Of the three soils failing to produce nodulated plants when inoculated, one produced plants that responded well to nitrogen additions but failed to nodulate under low nitrogen conditions; another produced severely stunted plants indicating nutritional limitations on the host; and the third produced plants that were not nitrogen deficient. An application of nitrogen completely suppressed nodulation in all but one soil. The twoPurshia species were similar in nodulation, nitrogen fixation and growth, although important exceptions exist that indicate species may differ in adaptability to certain soil conditions.

Notizen: Summary Natural senescence of the root cortex was assessed by nuclear staining, for cultivars and chromosome substitution lines of spring wheat known to differ in (1) susceptibility to common root rot, (2) total rhizosphere populations and (3) ability to support growth of a free-living nitrogen-fixing bacterium. Together, three root rot susceptible wheat lines showed significantly more cortical senescence than did three resistant lines; the susceptible lines also support larger rhizosphere populations. The wheat line that supports growth of a nitrogen-fixing bacterium showed significantly less cortical death than did any other line. Substitution of chromosome pairs 5B or 5D between the parent cultivars Rescue and Cadet substantially altered the amount of root cortex death, which is thus genetically determined. It is suggested thatCochliobolus sativus and other weak parasites benefit from early natural senescence of the root cortex, and that the degree of susceptibility or resistance of wheat lines to common root rot is at least partly determined by differences in cortical senescence.

Notizen: Summary Effect of amendments, gypsum (12.5 tonnes/ha), farmyard manure (30 tonnes/ha), rice husk (30 tonnes/ha) and also no amendment (control) on the availability of native Fe, Mn and P and applied Zn in a highly sodic soil during the growth period of rice crop under submerged conditions was studied in a field experiment. Soil samples were collected at 0, 30, 60 and 90 days of crop growth. Results showed that extractable Fe (1N NH4OAC pH 3) and Mn (1N NH4OAC pH 7) increased with submergence upto 60 days of crop growth but thereafter remained either constant or declined slightly. Application of farmyard manure and rice husk resulted in marked improvement of these elements over gypsum and control. Increases in extractable Mn (water soluble plus exchangeable) as a result of submergence and crop growth under different amendments were accompanied by corresponding decreases in easily reducible Mn content of the soil. Application of 40 kg zinc sulphate per hectare to rice crop could substantially raise the available Zn status (DTPA extractable) of the soil in gypsum and farmyard manure treated plots while the increase was only marginal in rice husk and control plots indicating greater fixation of applied Zn. Available P (0.5M NaHCO3 pH 8.5) behaved quite differently and decreased in the following order with crop growth: gypsum〉rice husk〉farmyard manure〉control.

Notizen: Abstract Element and extractant-specific desorption of Mn, Fe, Cu, and Zn fromSpartina alterniflora detritus was observed. Desorption of a substantial fraction of the total detrital Mn, Fe, Cu and Zn occured rapidly when the detritus was treated with 0.1N HCl or 1 MgCl2. This treatment removed precipitated/coprecipitated and adsorbed trace elements, respectively, suggesting that a large fraction of detritus-bound trace elements are in readily exchangeable, surface reaction sites. The carboxylic acid functional group cation exchange capacity of the detritus also suggests an important role of surface exchange reactions in the dynamics of trace elements during decomposition. The rate and magnitude of changes in the trace element content of detritus has important implications for estuarine biogeochemical cycling of these elements including the potential for biological uptake of trace elements by detritus-consuming fauna.

Notizen: Abstract In pot experiments the relationships between rhizosphere pH, the extractable levels of Fe, Mn and Zn in the soil and their uptake into the roots and shoots of dwarf French beans have been studied. Variations in rhizosphere pH were induced by applying three different sources of nitrogencholine phosphate, ammonium phosphate and calcium nitrate-to an initially homogenised soil (preadjusted to either pH 7 or 8). The rhizosphere pH was found to be significantly lower following the application of either ammonium or choline phosphates and to be increased by calcium nitrate treatment. The Fe and Zn contents of both shoot and root were inversely proportional to rhizosphere pH. The Mn contents also increased with decreasing pH but a sharp increase was apparent below pH 5.5. The shoot Fe, Zn and Mn content were significantly correlated with the extractable levels determined in the rhizosphere and non-rhizosphere soil.

Notizen: Summary The uptake of trace metals by two plant species (French bean and maize) has been measured on two soils subjected to various waterlogging regimes. Uptake of both manganese and iron was increased due to soil waterlogging, although reoxidation of the soil affected iron more than manganese. Zinc and copper uptake was influenced by a species factor; French bean (Phaseolus vulgaris) showed preferential uptake of zinc, whereas maize (Zea mays) took up copper preferentially. Uptake of cobalt by both species was increased due to waterlogging, following the pattern of manganese. The abilities of these species to take up trace metals from soil followed the pattern predicted by selective extraction of soil for manganese, iron and cobalt, but not for zinc and copper.

Notizen: Summary It has been demonstrated by an agar film technique thatL. albus can cause the breakdown of colloids of iron/silicate, iron/phosphate, aluminium/silicate and aluminium phosphate and destabilise suspensions of manganese dioxide, calcium mono-hydrogen phosphate and ferric hydroxide. Dissolution of these compounds was most marked in areas adjacent to proteoid roots (dense clusters of secondary laterals of limited growth which develop on lateral roots) and parts of the tap root. Soil associated with these regions of the root system contained more reductants and chelating agents than the bulk soil. Soil from around the roots ofL. albus exhibited much greater reducing and chelating activity than that associated with the roots of rape and buckwheat.

Notizen: Summary The beneficial effect of spraying some highly active phyllosphere N2-fixing microorganisms on high and low yielding cultivars of rice plants as compared with that of urea applied at different doses are described. The dry weight, N-content, 1000 grain weight, and yield were remarkably increased in all cases with the application of phyllosphere microorganisms. The performance of two isolates KUP4 and KUPBR2 with IR-8 and IR-26 rice, was better than that of 52 kg urea-N per hectare. IR-579 rice leaves in association with some phyllosphere bacteria reduced acetylene at the rate of 664–816 nmoles/g leaf/h. In IR-26 rice the effect of application of bacterial suspension at three phases of plant growth corresponded very well with that of urea application in three split doses under identical conditions. Recommended fertilizer rates produced the same yield as the half dose plus bacterial spray in the cultivars Pankaj and Rupsail. Fertilizer application in Pankaj and Rupsail rice reduced nitrogenase activity and the beneficial effects of phyllosphere N2-fixation was reduced by 40–55%.

Notizen: Summary Laboratory incubation experiments were conducted with three clay loam lowland rice soils (Haplustalf) to study the effect of application of three different levels of Cu(0, 1.25, 2.5 ppm) and Mn (0, 10, 20 ppm) in all possible combinations on the changes in the DTPA extractable Zn, Cu, F, Mn and Brays 2 extractable P in soil. The results showed that application of Cu decreased the content of extractable Zn, Fe, Mn and P in soils, the rate of decrease gradually declining with the progress of incubation period. Application of Mn also depressed the content of extractable Cu, Fe and P but increased that of extractable Zn.

Notizen: Summary Seven isolates belonged toA. brasilense and 3 belonged toA. lipoferum. Isolates having more denitrifying capacity fixed less nitrogen in nitrogen free semi-solid malate medium. One strain ofA. lipoferum having high nitrogen fixing capacity with negative test for denitrification was tested as inoculant to supplement the nitrogen need of a wheat crop in field condition with different doses of N with and without the inoculant. While control without nitrogen yielded 1260 kg/ha the yield in inoculated treatment was 2070 kg/ha resulting in significant increase. In a treatment receiving 40 kg N/ha the grain yield was 2370 kg/ha as against yield of 3110 kg/ha in a similar treatment receiving fertiliser plus inoculant. Thus increase in yield was about 30%. Further the treatment receiving 80 kg N/ha yielded 2970 kg/ha as against yield of 4150 kg/ha in a treatment receiving inoculant alongwith the above dose of the fertiliser. Thus increase in yield due to application of inoculant was about 36%. Similarly, the uptake of N in different treatments was augmented due to inoculation of seeds with the culture.

Notizen: Summary Growth responses of Frankia isolates to decreasing water potential were monitored in systems where potentials were controlled by KCl, NaCl and Polyethylene glycol. The highest potential tested was −2 bar (basal medium). The general pattern emerging was that isolates fromAlnus glutinosa, A. viridis andComptonia peregrina showed declining growth at potentials below −2 to −5 bar. AMyrica gale isolate showed declining growth with decreasing potential. All isolates were more sensitive to decreases in potential in a matric controlled than an osmotic controlled system. They all showed approximately 50 percent growth reduction at −5 to −8 bar, and meagre growth at −16 bar after 35 days. The Comptonia isolate was the most vigorous at low potentials. Nitrogen fixation ability was monitored for two isolates. Highest specific activities were observed between −3 and −5 bar for the Myrica isolate and between −5 and −7.5 bar for theA. glutinosa isolate.

Notizen: Summary Aerobic and anaerobic N2-fixing bacteria developed in the rhizosphere of barley seedlings and exhibited N2ase activity when seedlings were grown in sterilized sand-nutrient cultures containing low levels of combined nitrogen. The source of the N2-fixing bacteria appeared to be the seed. Average daily rates up to 0.9 μmoles C2H4 h−1 g−1 dry root tissue were measured, but the intensity of the activity was affected by moisture levels and concentration of combined N in the rhizosphere. Removal and washing of the roots did not remove the activity, and roots remained active even after surface-sterilization. An unidentified aerobic N2-fixing bacterium was isolated from the rhizoplane of active barley roots. Inoculation of barley seedlings with the aerobic N2-fixing bacterium enhanced N2ase activity of excised roots 10-fold, with average rates of 0.9, 1.1 and 1.3 μmoles h−1 g−1 dry root assayed under pO2 of 0.01, 0.02 and 0.04 atm respectively. The aerobic N2-fixing bacterium also exhibited N2ase activity when inoculated into the rhizosphere of oat, rice and wheat seedlings. Microscopic observations of sterilized live and stained barley roots suggest that the aerobic N2-fixing bacterium is an endophyte which infects root tissue and metamorphoses into vesicle-like structures.

Notizen: Summary Mineral nitrogen did not increase grain yield and seed protein levels ofVicia faba L. andLupinus luteus L. in field trials and pot experiments. Fixed N2 was substituted by mineral nitrogen in these cases because of inhibition of N2 fixation by mineral nitrogen. Contrary to these results mineral nitrogen increased grain yields and seed protein amounts ofLupinus albus L.,Pisum sativum L., andGlycine max. (L.) Merr. The nitrogen effect was caused at an early stage by saving energy due to inhibition of N2 fixation (measurement of gas exchange by means of IRGA). In case of the N application after flowering grain, yields and seed protein levels increased because the mineral N was an additional nitrogen source for plants. At this stage the plants had ceased fixing atmospheric nitrogen. The high sink activity of growing fruits induced a lack of assimilates in nodules (determined by means of14CO2 application). The N effect was therefore the consequence of the lower assimilate pool for supplying root nodules in these plants in comparison withVicia faba L. andLupinus luteus L. Hence it follows that response to mineral nitrogen can be a criterion for discovering more effective Rhizobium-host combinations.

Notizen: Summary The three major soil series comprising the Gezira scheme (Sudan) are Hosh, Suleimi, and Laota. Surface soil samples from each soil series were employed to study the effect of soil moisture on the DTPA-extractable micronutrient cation under aerobic conditions. The study continued for 8 weeks using an incubation technique at two levels of soil moisture (continuously moist and moist/dry cycles). The DTPA-extractable Cu, Fe, Mn and Zn from air-dry soil samples were much higher compared to values from their incubated counterparts. For the three soils the CO2 production (microbial activity) reached the maximum in 5 weeks and then levelled off while the lowest values of micronutrient cation from the incubated soils were obtained between 2 to 8 weeks. Generally, the study suggests that the hot dry months preceding crop growth should increase clay surface acidity and hence availability of mironutrient cations.

Notizen: Summary Experimental assessment of differences between cultivars of crop species or ecotypes of wild species from different localities in their capacities for ion absorption and transport is made difficult by the problem of obtaining seed material of comparable ionic content. When young seedlings are used this problem is particularly acute if the seeds of the different cultivars have not been raised under identical soil conditions. Propagation of material from ecotypes under controlled conditions is one approach to the solution of this problem. Six maize cultivars have been selected for similarity of phosphate content and the capacity for phosphate absorption from 5 μM KH2PO4 has been shown to vary by threefold whereas the proportion of the accumulated phosphate that reaches the shoot differs by much less. This level of phosphate supply approached that likely to induce deficiency and when the concentration is reduced to 1 μM differences in transport capacity of up to fourfold were observed when the rate of arrival at the tip of the first leaf was continuously monitored. The rapidity with which the transport is shut off by adding 1 mM D(+) mannose to the root environment also varies significantly indicating that sizeable differences in either the accumulation of mannose or the activity of phosphomannoisomerase exist in these cultivars. Ecotypes ofArmeria maritima collected from three sites, inland serpentine, inland mine dumps and coastal salt marsh were maintained as stock plants on the same peat mixture. Samples taken from these stocks were raised on a standard culture solution to provide genetically different material grown under constant conditions. The capacities for ion uptake were shown to differ very considerably and these differences were accentuated when the plants were grown in a range of concentrations of MgSO4, NaCl and MnSO4. The absorption of phosphate and its incorporation into nucleic acids were increased temporarily in the presence of 50 mM MgSO4 but the pattern of these changes was different in the three ecotypes. The absorption of Na, Cl, and Rb was measured after treatment with a range of concentrations of NaCl and the effect of treatment with MnSO4 on subsequent absorption of Mn and SO4 was also measured. The coastal plants were significantly more efficient in their absorption of these ions when treated at the lower levels of NaCl (0.5 and 10.0 mM). The short term absorption rates were not reflected in the overall accumulation of sodium over periods of 10 weeks and the coastal plants appeared to reduce the root content of sodium by transfer to the shoot and by increased active pumping to the exterior.

Beschreibung / Inhaltsverzeichnis: Summary The kinetic of15N2 diffusion has been measured in a system similar to that for the estimation of N2 fixation in plant microorganism associations cultivated in soil. The15N2 enrichment of the soil atmosphere reached an homogenous value one hour after injection of15N2 and is identical to that obtained by calculation, indicating that no adsorption occurs in the soil particles.

Notizen: Summary Experiments were performed to investigate the causes of low relative efficiency, RE, in legume root nodules. Nitrogen fixing activity and RE varied with time of incubation of nodules and with different temperatures and oxygen concentrations. The effects of nitrogen concentration and carbon dioxide concentration were also examined. In each case the RE was inversely related to nitrogen fixing activity; measured by acetylene reduction. Increasing the nitrogen concentration had no effect on either nitrogen fixing activity or RE. Experiments with isolated bacteroids gave higher RE values than the whole nodules from which they were isolated. All the results were consistent with hydrogen inhibition of nitrogenase within the nodule being the cause of low RE.

Notizen: Summary The electro-chemical and chemical kinetics of six California rice soils were significantly influenced by the presence of salts up to an EC of 9 mmhos/cm in saturation extract (ECe). Subsamples of each soil salinity treatment were incubated for periods up to 10 weeks after flooding. Most of the changes in Eh and pH values took place in the first 3–4 weeks after submergence. Salinity decreased pH values, but slightly increased the redox-potential. Both ammonification and nitrate reduction were significantly decreased, by increasing soil salinity. Salinity up to 9 mmhos/cm did not affect levels of Bray and Kurtz extractable P, but increased the water extractable Ca, Mg, K and Mn. In DTPA extract, salinity in incubated soils had no effect on Zn in 4 soils, but it decreased Fe in acid and neutral soils. Possible explanations for the electro-chemical and chemical kinetic changes due to flooding and salinity are discussed.

Notizen: Summary Two different strains, An 1 and An 2, were obtained from root nodules ofAlnus nitida Endl., collected from one locality in the area of its natural habitat near Bahrin, District Swat, Pakistan. The light and electron microscopy of the isolates revealed the occurrence of septate and branched hyphae bearing sporangia and vesicles. The strains differed in their growth requirements, nitrogen-fixing ability and production of extracellular pigments, thus indicating the existence of more than oneFrankia strain in the same locality. In the absence of combined nitrogen in the medium strain An 1 formed vesicles and fixed N2 (up to 200 nmol C2H4. mg protein−1.h−1), while strain An 2 under the experimental conditions formed only few vesicles and fixed N2 at a very low rate (ca 10 nmol C2H4. mg protein−1 .h−1). The nitrogenase activity of strain An 1 was strongly affected by the O2 concentration.Frankia An 1 and An 2 were infective and effective onA. nitida andA. glutinosa but not onDatisca cannabina andElaeagnus umbellata. Both An 1 and An 2 strains were more infective and effective onA. glutinosa thanFrankia strains AvcIl and CpI1.

Notizen: Summary An investigation was conducted to study the levels of nitrogen fixation on the leaf or sheath surfaces of four cultivars of paddy plants by using acetylene reduction technique. Varying levels of positive nitrogenase activity were observed on all the leaf surfaces. Sheath of IET 1991 cultivar showed a higher rate of fixation than the leaf surface. All the nitrogen-fixing organisms on the leaf or sheath surfaces belonged to the genus Beijerinckia. There was no correlation between the bacterial density and the level of fixation. Scanning electron microscopic data revealed that the upper surface of IET 1991 leaf was highly silicified and the microflora was either scanty or nil while the lower surface appeared quite different and harboured more micro-organisms. Similarly, the inner surface of sheath was devoid of silicification and showed the presence of micro-organisms.

Notizen: Summary Frankia sp. LDAgpl, an isolate from spore positive nodules ofAlnus glutinosa, only slowly infects its host plant. Reisolates obtained from occasional nodules caused by infection with LDAgpl, are capable of infecting the alder much more rapidly. A variability analysis of LDAgpl has been performed to obtain more insight into the question whether these reisolates constitute a different genotype within LDAgpl and if the plant is exerting an influence during plant passage. High dilutions of mildly sonicatedFrankia suspensions were plated to obtain genetically homogeneous colonies. Clones thus generated showed differences in growth pattern, sporulation and C2H2-reduction on media containing propionic acid as sole C-source (P-medium). Differences in sporulation on P-medium indicate that LDAgpl was a highly heterogeneous strain. Comparisons of sporulation on several different media gave evidence that the differences in sporulation between LDAgpl clones are the result of differences in efficiency of propionic acid utilization. The differences observed between the reisolates and LDAgpl clones indicate that the reisolates constitute a different genotype, which could be selected for by the plant during the infection process. Comparison with similar changes in phenotype occuring in a spore negative type strain fromA. glutinosa is discussed.

Notizen: Summary The nodulation and the morphology and physiology of the nodules were studied onDatisca cannabina, a perennial herb from northern Pakistan andAlnus nitida, a nodulated tree in the same locality. Both species bear coralloid clusters of actinorhizal nodules. The main free amino acid inD. cannabina nodules was arginine while the predominant free amino acid inA. nitida nodules was citrulline. The infectivity of crushed nodules of both types of plants on their respective host was about 106 infective particles per gram of nodule fresh wt. In cross-inoculation experiments crushed nodule inoculum fromA. nitida failed to induce nodulation onD. cannabina seedlings but the crushed nodule inoculum fromD. cannabina caused low nodulation on seedlings ofA. nitida (103 infective particles. g. nodule fresh wt.). The activity of nitrogenase, hydrogenase and respiration (O2 uptake) were measured in detached nodules, nodule homogenates and the 20 μm residue and 20 μm filtrate preparations from the nodules of both species. Both species showed similar patterns of activities except that only the nodule homogenate and 20 μm residue preparations fromD. cannabina showed pronounced enhancement of the O2 uptake by succinate which was further stimulated by ADP. This has in part been explained by the presence of mitochondria in close connection with the endophyte.

Notizen: Summary Inoculation effect ofA. chroococcum, P. striata andA. awamorii on yield and nutrients uptake in rice was studied under green house conditions. The organisms appreciably increased the yield and uptake of nutrients with or without chemical fertilizers. Phosphorussolubilizing microorganisms and a mixture of the three showed better response than the rest of the treatments among single and mixed culture inoculations respectively. Chemical fertilizers further improved the yield and nutrients uptake. The yield response remained unaffected by replacing superphosphate with rock phosphate and microbial inoculations.

Notizen: Summary The relative nitrogen fixation efficiencies (RE 1-[H2 evolved÷C2H2 reduced]·100) of four mesquite (Prosopis glandulosa var.torreyana) rhizobia (Strains WR 1001, WR 1002, L5, L9) and a cowpea rhizobia (Strain 176A32) on mesquite were evaluated in a glasshouse experiment. Plant yield, shoot N accumulation, and the natural15N abundance (δ15N) of nodule tissue were determined. Strain WR 1002 failed to nodulate mesquite and strain L5 produced ineffective nodules. Among the three effective strains (WR 1001, L9, 176A32) the cowpea strain (176A32) and strain L9 had significantly higher RE than strain WR 1001. Differences in RE, however, were not accompanied by significantly higher plant yield and shoot N accumulation. The difference in15N abundance between foliar tissue and nodules (nodules minus leaves) was 0.47 δ15N for the ineffective L5 nodules, while for the effective WR 1001, L9, and 176A32 nodules, respectively, this difference was 8.35, 7.81, and 8.35 δ15N. This indicates a similar relationship between N2-fixing effectiveness and natural15N enrichment of nodules that was previously observed in soybeans (Glycine max, L. Merr.). Strains WR 1001 and L9 produced elongate, indeterminate nodules typical for mesquite. The ineffective L5 nodules had few infected cells and an abundance of cortical amyloplasts. Mesquite nodules produced by the cowpea strain were spherical and were somewhat more similar in internal morphology to determinate nodules typical of cowpea than indeterminate nodules normally associated with mesquite.

Notizen: Summary The fine structures of the microsymbiont inside the root nodules ofDatisca cannabina have been studied by light, by transmission- and by scanning-electron microscopy. The endophyte is prokaryotic and actinomycetal in nature. The hyphae are septate and branched, diameter 0.3–0.5 μm. The tips of hyphae are swollen to form electron-dense, clubshaped to filamentous vesicles, ranging in diameter: 0.4–1.4 μm. The endophyte penetrates through walls of the cortial cells. The infected zone is kidney shaped and confined to one side of the acentric stele. The orientation of infection is reversed from other actinorhizae exceptCoriaria. The hyphae are near the host cell wall and vesicles are directed towards the central vacuole. Vesicles are aseptate and no collapsing of the vesicle cell wall (void area) has been observed. Vesicle clusters structures are globular with an opening at one side of the cluster. The host cell is multinucleate or contains a lobed nucleus. Groups of mitochondria are located in between the hyphae, suggesting a strong association between the host and the endophyte for energy supply and amino acid production. The consequences of the inability to separate the mitochondria from the vesicle clusters in nodule homogenates in physiological studies have been discussed. Isolated vesicles clusters showed dehydrogenase activity, indicated by the presence of formazan crystals, after incubation with NADH and NBT. Strongest reducing activity was found within the vesicles. The possible role of filamentous vesicles in nitrogen fixation has been discussed.

Notizen: Summary An acid mine spoil in Southern Indiana was amended with lime (CaCO3) (0.0, 12.5, 25 and 39t/ha) and planted withElaegnus umbellata Thunb.,Alnus glutinosa Gaertn.,Robinia pseudoacacia L.,Robinia fertilis Ashe, ‘Arnot’,Myrica pensylvania Lois,Caragana arborescens L. andShepherdia argentea Nutt. Survival and soil data were collected periodically and plants were harvested 15 months after planting. Nodule and top dry weights were determined and acetylene reduction assays performed on the nodules. Addition of lime caused significant increases in pH, and 39 t/ha of lime were required to maintain a pH above 5.5. Survival of plant material was greatest at the highest lime addition, although response of individual species varied.Elaeagnus umbellata, R. pseudoacacia, R. fertilis ‘Arnot’, andA. glutinosa appeared more tolerant of the harsh conditions. OnlyC. arborescens showed a linear increase in top dry weight due to lime addition.Alnus glutinosa andS. argentea achieved statistically the same growth regardless of pH, andR. fertilis ‘Arnot’ andE. umbellata did not increase in top dry weight above an addition of 25 t/ha.Robinia pseudoacacia achieved maximum top dry weight at 25 t/ha, whereasM. pensylvanica growth declined with increasing pH. Nodule dry weights increased with increasing pH; however,S. argentea showed greater nodule dry weights at lower lime levels. Acetylene reduction rates increased with lime addition.Elaegnus umbellata did not respond above 25 t/ha lime, whereasA. glutinosa did not show an increase until this point.

Notizen: Summary The absorption of gamma-emitting fission products106Ru,125Sb,137Cs and144Ce and activation products59Fe,58Co.54Mn and65Zn by rice plants grown on two contrasting tropical soils, namely, a blak soil (pellustert) and a laterite (oxisol), and the effects of flooding were studied under controlled conditions. Results indicated greater uptake of106Ru and125Sb from the black soil than from the laterite. In contrast, the uptake of144Ce and137Cs was greater in the laterite than in the black soil. Flooding treatment enhanced the uptake of all these fission products by rice plants in the laterite soil whereas this effect was observed only for125Sb and137Cs in the black soil. The plant uptake of activation products from the two soil types showed maximum accumulation of65Zn followed by54Mn,59Fe and58Co in both soil types. Besides, uptake of these nuclides was greater from the laterite soil than from the black soil. Flooding treatment for rice while showing a reduction of59Fe uptake, showed an increase in plant uptake of58Co,54Mn and65Zn in both soil types.

Notizen: Summary Soils influenced by acid mine drainage (pH〈5.0) are characterized by low concentrations of essential nutrients and increased solubility of heavy metals. The conditions typically reduce plant establishment and growth. However, river birch (Betula nigra L.) is commonly found along low pH streams in southeastern Ohio. The objective of this study was to determine the concentration of Al, Mn, Ca and Mg inB. nigra tissues. The results indicate Al and Mn are accumulating inB. nigra when compared to other species. Within river birch, Al concentrations are highest in roots; Mn concentrations are highest in leaves. There is not a concomitant reduction in Ca and Mg concentrations as suggested by soil levels.

Notizen: Summary Using a factorial design with deficient to toxic levels of Mn and B, the absorption and distribution of Fe in tomato plants (Lycopersicum esculentum, var. Marglobe), grown hydroponically in a greenhouse was studied. B was added to disturb growth and hence nutrient demands. Deficient or normal Mn levels antagonize Fe absorption, but the reverse was true when Mn reached toxic values; nevertheless, Mn effect was always antagonistic on Fe transport. A hypothesis for explaining the above-related results, together with P and Ca absorption and distribution, is proposed. Mn/Fe in the shoot is not related at all with plant growth. B levels influence Fe absorption and translocation paralleling the dry matter production.

Notizen: Summary Eight ineffective mutant strains were isolated from N-methyl-N'-nitro-N-nitrosoguanidine mutagenized cultures of cowpea Rhizobium strain 32H1. Strains CR1, CR2, CR3, CR4, CR5 and CR6 induced more, but smaller, nodules than the wild type. With the exception of strain CR2, these mutant strains reduced less than 1% of the amount of acetylene reduced by the wild type, in both the free-living and symbiotic assays. Strain CR2 reduced acetylene in the free-living assay but not in the symbiotic assay. Strains CR7 and CR8 responded variably (5–20% of the wild type) in free-living and symbiotic acetylene reduction assays. Nodules also varied from small white to normal-sized pink nodules. The phenotypic characteristics of the mutant strains were consistant with all leguminous plants tested and were stable upon reisolation from nodules. Fully effective revertants were selected from 4 of the ineffective mutant strains by the use of the leguminous plant,Macroptilium lathyroides. Serology, patterns of resistance to anti-bacterial agents, phage-typing, and antibiotic resistance markers were used to confirm strain identification.

Notizen: Summary The effects of ammonium or nitrate-nitrogen on biological nitrogen fixation by an algal crust are compared. Nitrate-nitrogen up to 3.0 μmoles N g−1 sand/algal crust at 60% water holding capacity did not affect fixation, whereas an ammonium-nitrogen concentration of 0.2 μmoles N g−1 crust markedly depressed fixation. Consequences of these differential effects are considered.

Notizen: Summary An assessment of the status of available micronutrients has been made in a long term field experiment on a fixed rotation of soybean-potato-wheat at the Ranchi Agricultural College, Kanke on acid soil. Results of eight years fertilizer applications with lime has resulted in reduction of available micronutrients whereas the FYM application with balanced dose of chemical fertilizers increased the available micronutrients of soil. The maximum loss of 1.0 Zn and 13.2 Fe kg/ha respectively were from the lime treated plots with balanced dose of fertilizers and the highest gain of 0.26 Zn, 1.08 Cu, 13.2 Fe and 54.0 Mn kg/ha respectively were recorded from the FYM treated plots with balanced dose of chemical fertilizers. Organic carbon was significantly positive correlated with av. Zn & Mn, whereas av. Mn significantly negative correlated with pH. Other relationship were found to be non significant.

Notizen: Summary The nitrogen fixation rate in a 4-year-old stand of the woody legumeLeucaena leucocephala (Lam.) de Wit. was estimated in the field at a rather dry site in Tanzania by use of an acetylene reduction technique. The diurnal mean value during April–May was 35 nmol C2H4 mg−1 (dry weight) nodules h−1, with a variation between 22±8 and 48±12 nmol C2H4 mg−1 (dry weight) nodules h−1 in early morning and at midday, respectively. The nodule biomass was determined by auger sampling to be 51±16 kg (dry weight) ha−1. Most of the nodules were found at the 10–30 cm soil depth level. A rough calculation of the amount of nitrogen fixed annually arrived at 110±30 kg ha−1. The results give strong support for the use ofL. leucocephala for soil enrichment in less humid areas of tropical Africa.

Notizen: Summary In a factorial pot experiment on garden peas investigating the effect of growing medium, inoculation, nitrogen fertilisation and charcoal addition, shoot growth was significantly better in John Innes potting compost than in 50/50 peat/sand compost and when receiving nitrogen fertilisation but was depressed by the presence of wood charcoal. In the peat/sand medium, root systems were smaller but carried more and larger nodules. The number of nodules was increased by inoculation but the addition of carbon decreased root size, nodule number and nodule size.

Notizen: Summary The Thriasian Plain near Athens in Greece is a site of concentrated industrial development. Total concentrations of Pb, Cd and extractable SO4 2− in surface soil samples collected from this area, were found to be, respectively, 2–7, 6–34 and 2–20 times higher than those in similar soil samples from parts of the country remote from industrial activity. Total concentrations of Pb, Cd, Zn, Cr and S in samples of olive leaves from the same area were found to be, respectively, 4–40, 3–10, 3–9, 2–6 and 17–21 times higher than those found in samples of olive leaves from rural sites. Leaves of cabbages growing in the area contained between 0.82 and 40 μg/g (wet weight) of Pb. Addition of Cd, Ni and Cu to a calcareous potted soil at concentrations of 100, 200 and 200 ppm increased the concentrations of the metals in the needles of Aleppo pine (Pinus halepensis) seedlings to 4.5, 3.5 and 10 ppm, respectively, after 7 months growth in the nursey.

Notizen: Summary Effects of three solution aluminium concentrations (0, 25, and 100μM) on nitrogen fixation by well-nodulated plants ofStylosanthes hamata, Stylosanthes humilis andStylosanthes scabra are reported. Plants were inoculated with Rhizobium CB756 and grown for 21 days in an aluminium-free nutrient solution at pH 5.3 before imposition of the aluminium treatments. Nitrogen fixation was measured both by the increase in total nitrogen content of the plants and acetylene reduction in roots of plants harvested at 10 and 20 days after imposition of the aluminium treatments. Solution aluminium concentrations as high as 100μM, had no detrimental effect on nitrogen fixation in any species.

Notizen: Summary The effect of two granular nematicidesviz. oxamyl and fenamiphos, on the nodulation and growth of Rhizobium inoculatedArachis hypogaea L. was studied in glasshouse and field trials. In the glasshouse trial at the suggested rates of application shoot fresh weight was significantly reduced by oxamyl whilst root fresh weight was similarly affected by fenamiphos. In the field trial vegetative growth and plant emergence were significantly reduced by both nematicides. Nodulation at the higher rates of application was increased by both oxamyl and fenamiphos whilst oxamyl caused a significant increase in pod number at the highest rate of application.

Notizen: Summary The development of proteoid roots byLupinus albus was studied under glasshouse conditions in acid-washed sand with varying iron and phosphorus supply and in three contrasting soils (acid, neutral and alkaline) at a range of phosphorud levels. Aqueous extracts of soil from around the roots were compared with regard to their pH and their reducing and chelating activity. In both experiments, proteoid formation was predominantly affected by the phosphorus status of the plant. Manganese uptake in the second experiment was related to proteoid root development and decreased as phosphorus supply increased. The ability ofL. albus to utilise soil and added phosphorus decreased as pH of the soil increased.

Notizen: Summary This field study was undertaken to determine the effect of inoculation withGlomus mosseae on N2 fixation and P uptake by soybean. The inoculation withGlomus mosseae was achieved using a new type of inoculant, alginate-entrapped (AE) endomycorrhizal fungus. N2 fixation was assessed using the A value method. In P-fertilized plots, inoculation with AEGlomus mosseae increased the harvest index based on dry weight (+20%) and N content of seeds (+17%), the A value (+31%) and %N derived from fixation (+75%). Inoculation with AEGlomus mosseae decreased the coefficient of variation for the A value and for the dry weights of the different plant parts.

Notizen: Summary Transformation of iron and manganese under three different moisture regimes, viz continuous waterlogged (W1), continuous saturated (W2) and alternate waterlogged and saturated (W3) and three levels of organic matterviz 0, 0.5 and 1.0% in all possible combinations was studied in four soils. The results showed that under waterlogged moisture regime there was a sharp increase in the content of water soluble plus exchangeable manganese accompanied by significant decrease in the content of reducible manganese in all the soils excepting the acidic soil which was very poor in active manganese content. The increase in respect of iron in similar form was, however, very small. The increase in the content of water soluble plus exchangeable manganese as well as iron under the continuous saturated and alternate waterlogged and saturated moisture regimes was always much lower as compared to that under the continuous waterlogged condition. Application of organic matter brought about an increase in the content of water soluble plus exchangeable manganese in all the soils excepting the lateritic one irrespective of moisture regimes but did not cause any change in the content of iron and manganese in insoluble complex. The content of water soluble plus exchangeable iron and of insoluble ferrous iron although recorded some increase due to organic matter application, the increase was not so marked in any of the soils.

Notizen: Summary Field trials were conducted during theKharif seasons of 1979 and 1980 to examine the effect of the nitrogen-fixing bacteriumAzotobacter chroococcum (isolate M4) on yields of maize and N-economy. Different levels of nitrogen and farmyard manure (FYM) were supplied to assess their interaction with Azotobacter inoculation. Seed inoculation without fertilization raised grain and stover yields significantly. FYM coupled with inoculation gave higher yields than either could singly. The financial gain due to an increase in grain yield upon applying per ha 80 kg N, 10×103 kg FYM and Azotobacter over the treatment 40N + FYM + Azotobacter was offset by the cost of the additional N; thus, the higher N-application was not economical. Azotobacter inoculation was economically most efficient at lower doses of fertilizer nitrogen which not only increased yields but resulted in a saving of fertilizer N when applied in combination with FYM.

Notizen: Summary The economic feasibility of using cultures of nitrogen fixing microorganisms in programmes to increase crop production, as a selfgenerating source of nitrogen, has been proved beyond doubtviz. Legume-Rhizobium symbiosis and blue green algal ‘fertilizer’ for rice. The extent to which the free living, N-fixing, aerobic, heterotropicAzotobacter chroococcum could replace the application of nitrogenous fertilizer to maize was investigatedin vivo. Total nitrogen uptake (kg ha−1) by maize after inoculation with Azotobacter combined with moderate applications of nitrogen fertilizer and farmyard manure was influenced significantly and resulted in a higher nitrogen concentration in grain and stover along with a higher yield.

Notizen: Summary Organic components leaked fromSorghum bicolor seedlings (‘root exudates’) were examined by recovering14C labelled compounds from root solutions of seedlings inoculated withAzospirillum brasilense, Azotobacter vinelandii orKlebsiella pneumoniae nif-. Up to 3.5% of the total14C recovered from shoots, roots, and nutrient solutions was found in the root solutions. Inoculation with Azospirillum and Azotobacter increased the amounts of14C and decreased the amounts of carbohydrates in the root solutions. When sucrose was added as a carbon source for the bacteria, the increase of14C in the solutions did not occur. Quantities of14C found in the root solutions were proportional to amounts of mineral nitrogen supplied to the plants. Bacterial growth also was proportional to nitrogen levels. When sorghum plants were grown in soil and labelled with14CO2, about 15% of the total14C recovered within 48 hours exposure was found in soil leachates.

Notizen: Summary Phosphorus and Mn relationship was studied in chickpea at two stages of growth in pot culture using 0, 7.5, 15 and 30 ppm P and 0, 5, 10 and 15 ppm Mn. The dry matter yield increased with P at both stages of growth. Manganese improved the yield only in the first stage. Initial levels of Mn enhanced while higher levels had a depressing effect on tissue P. Addition of 7.5 ppm P enhanced Mn concentration at first stage and at higher levels a marked reduction in Mn content was observed at both the stages.

Notizen: Summary A pot experiment was conducted to assess the effects of different fertilizers and soil shading on nodulation and acetylene reduction ofInga jinicuil seedlings. Initially, seedlings produced maximum nodule biomass when grown with high levels of phosphorus but reduced the most acetylene under intermediate phosphorus fertilization. These response differences, however, gradually diminished with age, being negligible when the seedlings were a year old. Nitrogen fertilization inhibited nodulation and acetylene reduction throughout the experiment. Potassium did not significantly affect nodulation, but low levels of potassium stimulated, and high levels inhibited acetylene reduction activity relative to unfertilized control plants. Neither magnesium nor molybdenum affected nodulation or acetylene reduction. Soil shading resulted in decreased nodule biomass and less nitrogen-fixing activity during summer months. However, the data suggest that shading may favour nitrogen fixation in colder periods by moderating soil temperatures. These results confirm findings from an earlier field study and show that nodulation and nitrogen-fixing activity by leguminous trees is influenced by the types and amounts of nutrients supplied. This suggests that the quantity of nitrogen fixed by leguminous shade trees in coffee plantations may be amenable to manipulation through simple management techniques.

Notizen: Abstract In experiments with 29 cowpea genotypes considerable variation in Mn tolerance could be found. Ranking according to Mn tolerance was almost the same in sand and water culture. Mn tolerance is not related to greater vigour or exclusion of Mn from uptake and translocation, but depends mainly on the internal tolerance to excess Mn especially in the leaf tissue. Growth depression by Mn excess is characterized by local accumulatiòn of Mn, deposition of Mn oxides, and typical macro-symptoms on the older leaves (brown spots→clorosis→shedding of the leaves). Autoradiographic studies with54Mn and extraction of the leaves with methanol and H2O indicate a causal relationship between Mn tolerance and the more homogenous distribution of Mn in the tissue. In tolerant genotypes local accumulation and deposition of Mn is inhibited or retarded. Mn applied to the petioles of fully expanded leaves induces the same toxicity symptoms on the leaf blades as Mn absorbed by the roots. There is a good agreement between the rankings of the different genotypes for Mn tolerance according to the depression of shoot dry matter production by Mn excess in long term pot experiments and the appearance of toxicity symptoms after application of Mn to the petioles. The regulation of Mn tolerance at the leaf tissue level allows a quick and non-destructive screening of large numbers of genotypes for Mn tolerance.

Notizen: Abstract Plant genotypes differ in their uptake, translocation, accumulation, and use of mineral elements. Examples of genotype differences to iron, nitrogen, phosphorus, potassium, calcium, magnesium, manganese, boron, copper, zinc, and molybdenum are discussed. Current knowledge is sufficient to indicate that many crop plants can be improved for the efficient use of mineral elements and better adaptation to mineral stress conditions.

Notizen: Summary The rates of uptake of nutrients from solution by apple roots were measured (a) in a root laboratory, using intact roots of mature trees growing under field conditions and (b) in controlled environment using young trees. Maximum nitrate inflows into Discovery/M.9 roots under field conditions were only slightly lower than those into roots of the same genotype in controlled environment, but up to 80 times lower than those into roots of Worcester Pearmain seedlings. At any given external P concentration, P inflows into roots of field-grown trees were about 2.5-times lower than those into the roots of young trees in controlled environment. Nitrate inflows were constant above a solution concentration of 20 mmol m−3 in both field-grown and small trees. In both cases, phosphate inflows increased linearly with solution concentration up to 10 mmol m−3. Among the various plant and environmental factors influencing nutrient uptake characteristics of apple roots were: the scion genotype, tissue nutrient levels, root origin, the form in which N is supplied, level of irradiance of the shoot, root temperature and the season of the year. The effects of these factors are illustrated with examples.

Notizen: Summary Ten cultivars of subterranean clover were grown in pots on a poorly buffered, sandy soil of pH 4.9; the plants were dependent throughout upon symbiotic fixation for their supply of N. There were some marked increases in soil acidity which resulted in changes in pH of 0.76 to 1.08 (mean 0.94) units. Increasing soil acidity was associated with an increasing total content of excess cations,i.e. (Ca2+++Mg2++K++Na+)−(H2PO 4 t- +SO 4 2- +Cl−), which ranged from 15.8 to 21.9 meq pot−1. The differences between the cultivars in their effects on acidity were largely related to differences in growth rather than to marked differences in the concentration of total cations or inorganic anions. There was no obvious grouping amongst the ten cultivars in terms of either their effects on soil pH, or on their uptake of manganese.

Notizen: Summary Cross-inoculation experiments withParasponia parviflora plants and a large number of Rhizobium strains isolated from legumes, mainly of temperate origin, showed that strains ofRhizobium trifolii andR. lupini could produce root nodulation in Parasponia. Root nodulation was also obtained with some tropical Rhizobium isolates such as those from Arachis and Albizzia, but not withRhizobium japonicum strains. In addition, strains ofR. leguminosarum, R. phaseoli andR. meliloti produced abnormal root nodules or pseudo-root nodules in Parasponia seedlings. All root nodules induced in Parasponia by foreign Rhizobium species lacked, however, nitrogenase activity. Testing the reverse combination Parasponia-Rhizobium on legumes yielded with regard to root nodulation negative results, except withVigna sinensis and twoMacroptilium species (M. atropurpureum andM. lathyroides). The root nodules produced in the latter legumes showed nitrogenase activity.Trema cannabina seedlings inoculated with Parasponia-Rhizobium invariably did not produce root nodules. The feasibility of Parasponia to accept foreign rhizobia bacteria or to produce pseudo-nodulation is probably genetically determined. In a karyological study it was shown thatParasponia parviflora had the chromosome number 2n=20. Growth experiments revealed thatP. parviflora can be cultivated in water culture, perlite and in soil (pot) culture. It can also be grown in agar tubes, although growth under these conditions is sometimes rather variable. Nitrogenase (C2H2) tests showed that water-culture root nodules were 5–10 times less active than those from perlite culture, soil or agar tubes. On the whole, nitrogenase activity of root nodules grown on the various substrates and receiving different treatments, was rather variable.Vigna sinensis plants inoculated with Parasponia-Rhizobium showed on basis of fresh nodule weight a 3–4 times higher nitrogenase activity as compared with the same strain on Parasponia. However, as shown by acetylene reduction tests of nodule slices, the main activity of mature Parasponia root nodules is situated in the apical part of the root nodule. On fresh weight basis the nitrogenase activity of this nodule section is of the same order as that of leguminous root nodules. Intact Parasponia plants showed higher nitrogenase activities than excised root nodules. The maximal nitrogen fixation rate ofParasponia parviflora plants was 20–25 μmol. C2H4·g−1 fresh weight nodule tissue.h−1, which value is comparable to that of legumes and higher than estimates obtained by Parasponia in the field. No decrease of the nitrogen-fixation rate was observed in Parasponia plants tested in a diurnal cycle during the 12-hours dark period. Amino-acid analyses of Parasponia root nodules showed large quantities of aspartic acid, asparagine, glutamic acid and glutamine suggesting a role of these amino acids in the nitrogen fixation or transport processes. In contrast to actinorhizal root nodules Parasponia root nodules do not contain citrulline. The latter amino acid was, however, found—although in rather low concentration—inVigna sinensis andMacroptilium atropurpureum nodules obtained withParasponia-Rhizobium.

Notizen: Summary In pea cv. Afghanistan a recessive gene sym 6 prevents the full expression of nitrogenase activity in root nodules, induced byRhizobium leguminosarum strain F 13. In contrast, nitrogenase activity is fully expressed in pea cv. Iran. A comparison of the reciprocal hybrids of these two plants showed that the size of the plant was determined by the mother plant (maternal effect). Therefore the shoot weight and the total amount of nitrogen fixed are not suitable as parameters for a genetic analysis. The % nitrogen of the shoot and the specific activity of the nodules per gram of nodules are more reliable, but for practical purposes the specific activity of the nodules expressed per gram of shoot tissue can be used.

Notizen: Summary Physiological and symbiotic characteristics were identified in fast-growing (FG)Rhizobium japonicum. Carbon nutritional patterns linked these rhizobia to other FG rhizobia. They were able to use hexoses, pentoses, disaccharides, trioses, and organic acids for growth, but they were unable to use dulcitol or citrate. These rhizobia produced acid with all carbon sources except intermediates of the Krebs cycle. FGR. japonicum showed no vitamin requirements and were tolerant to 1% NaCl but not to 2%. They nodulated cowpea, pigeon pea, and mung bean but not peanut. Effective, nitrogen-fixing symbioses were observed only with cowpea and pigeon pea. In addition, FGR. japonicum formed effective symbioses with Asian-type soybeans. We concluded that although the physiological characteristics of FGR. japonicum were similar to other FG rhizobia, their symbiotic properties were similar to slow-growing rhizobia of the cowpea miscellany.

Notizen: Summary The isolation and pure culture of the symbiotic nitrogen-fixing frankiae has always been difficult. In the past the isolation of these actinomycetes directly from soil samples has proven impossible and isolations from root nodules of many genera has been only poorly successful. We report here a modified sucrose fractionation procedure which increased the success of isolations from root nodules and which permitted the isolation ofFrankia directly from soil samples. Crushed nodule suspensions or soil suspensions were incubated briefly in 0.7% phenol (carbolic acid) just before application to a sucrose density gradient. This phenol incubation decreased the number of contaminating eubacteria and fungi but more importantly increased the number ofFrankia developing on the isolation plates. If the phenol incubation was used solely without sucrose fractionation noFrankia were isolated, suggesting the death of the organisms due to phenol toxicity. The use of selective nitrogen-deficient media proved important for the isolation of frankiae from soils.

Notizen: Summary Root nodules ofDryas drummondii are of the coralloid type (Alnus type). The endophyte is present in the middle cortical cells of the root-nodule tissue. Transmission electron micrographs revealed an actinorhizal endophyte with septate hyphae and non-septate spherical or ovoid vesicles. Vesicles always possess at the base a septum; septa formation in the endophyte is always associated with the presence of mesosomes. Branching of the endophyte is not necessarily correlated with septum formation. Hyphal structures are more prominent in the apical part of the root nodule and vesicles are more numerous in a broad zone below this. In the middle and towards the base of the root nodule the endophytic structures appear in a stage of disintegration. Vesicles appear in a broad region near the periphery of the host cell and regularly show no strict orientation towards the host-cell wall. In the center of the host cells only hyphae occur. In the intercellular spaces between the host cells theFrankia endophyte produces spore-like structures although the outline of the sporangia is often faint. The coralloid root ofRubus ellipticus shows characteristically a basal rootlet initiated below the dichotomous branching of the nodular lobes, but extending beyond the root nodule. The endophyte is only present in the outer cortex of the root nodule in a 1–2 cell wide layer. This endophytic layer is bounded, internally as well as externally, with a 4–5 cell wide layer of tannin-filled host cells. The implications of this situation are discussed. Tannin-filled cells occur regularly inRubus species and their arrangement has been used for taxonomic purposes within the genus. TheRubus endophyte is aFrankia species with septate hyphae and distinctly septate spherical vesicles. The ultrastructure of the vesicles of theRubus endophyte is very similar to that of theAlnus endophyte.

Notizen: Summary Effects of factors associated with soil acidity (low pH, low calcium, high aluminium and high manganese) on theTrifolium repens-Rhizobium trifolii symbiosis were investigted under laboratory conditions using an axenic solution-culture technique. 200 μM manganese increased root elongation in the range pH 4.3–5.5, but had no effect on root hair formation, the number of Rhizobium in the rhizosphere, or nodule formation. Root elongation and root hair formation were unaffected at pH 4.3 when 500 or 1000μM calcium was supplied, whereas multiplication of Rhizobium in the rhizosphere and nodulation were inhibited at pH 4.3 and 4.7.50–1000μM calcium had no effect either on the multiplication of Rhizobium in the range pH 4.3–5.5, or on nodule formation in the absence of aluminium. 50 μM aluminium inhibited, root elongation and root hair formation at pH 4.3 and 4.7; the effect on root elongation was reduced by increasing the calcium concentration from 50 to 1000μM. 50μM aluminium also inhibited Rhizobium multiplication in the rhizosphere and reduced nodule formation at pH 5.5 (at which aluminium precipitated out of solution), but root elongation and root hair formation were unaffected. These, effects of aluminium at pH 5.5 may explain the poor response to inoculation by white clover in acid mineral soils after liming.

Notizen: Summary Rye grass (Lolium perenne, cv.S-23) was grown for 4 weeks in a non-calcareous Seaton loam soil with varying amounts of Ni as NiSO4. The purpose of this investigation was to study the Ni toxicity and the relationship of Ni with other essential elements. Nickel depressed shoot yield at all levels except at the lowest levelviz 30 μg Ni/g soil. Nickel concentration of 50 μg/g in shoots did not reduce the dry matter production in rye grass although slight chlorosis did appear at this level. The Ni and Fe concentration of the shoots increased and that of Mn and Zn decreased with increasing rates of Ni application. Uptake of Mn and Zn decreased at all level of Ni. But Fe uptake showed a slight increase at the first two levels and a profound depression at the subsequent levels. The pattern of Ni uptake is different, being highest at the middle level and decreasing on both sides which showed that the increase of Ni concentration of shoots is not proportional to the reduction in the yield. The Ni−Fe ratio rather than Ni and Fe concentration in plants has shown better relationship with the toxic effects of Ni. The implications of Ni phytotoxicity are discussed with particular reference to serpentine soils.
