    Notes: Abstract  The ability of restriction fragment length polymorphism (RFLP) analysis of the rDNA internal transcribed spacer (ITS) region to discriminate 10 co-occurring Cortinarius and Dermocybe species at a southeastern Australian sclerophyll forest site was assessed. Using the basidiomycete-specific primers ITS1F and ITS4B, some taxa were separated on the basis of individual RFLP patterns derived using the restriction endonucleases Hae III or Hinf I. Combined data from both endonucleases were, however, required to separate all taxa [Dermocybe austro-veneta Clel. (Moser & Horak), C. rotundisporus Clel. & Cheel, C. archeri Berk., C. sinapicolor Clel., C. violaceus (L.: Fr.) S.F.Gray, C. radicatus Clel. and four morphologically-distinct, but unidentified Cortinarius spp.]. ITS sequence comparisons confirmed that D. austro-veneta belongs in Dermocybe, that C. rotundisporus is correctly placed in subgenus Phlegmacium, and suggest that Australian C. violaceus collections are not conspecific with northern hemisphere C. violaceus.

    Abstract: Zusammenfassung Diese Arbeit wurde durchgeführt um festzustellen, ob sich der Faktor für die Calcium-bindung, durch das calcifizierendeBacterionema matruchotii, in der Lipoidfraktion befindet. Die lyophiilisierten Zellen wurden mit Chloroform-Methanol extrahiert. Die Chloroform-Methanol-Fraktion, die extrahierten Zellen, sowie die nicht behandelten Zellen wurden auf eine Calciumbindung hin untersucht. Die Chloroform-Methanol-Fraktion und die nicht behandelten Zellen demonstrierten eine Calciumbindung. Die extrahierten Zellen hingegen nicht.
    Notes: Abstract This work was done to determine whether the factor responsible for calcium binding by a calcifiableBacterionema matruchotii is in the lipid fraction of the cell. Freeze-dried cells were extracted with chloroform-methanol. The chloroform-methanol fraction, the extracted cells and untreated cells were examined for calcium binding. The chloroform-methanol fraction and the untreated cells bound calcium. The extracted cells did not.

    Description / Table of Contents: Résumé Des coupes de cartilage épiphysaire frais de jeunes rats, effectuées à la main, sont colorées à pH=4,5 dans des solutions à 0,01% de divers colorants cationiques, appartenant aux groupes de la thiazine, oxazine, azine, triphénylméthane, acridine, et phthallocyanine. Les granules intracellulaires métachromatiques, mises en évidence antérieurement par le bleu de toluidine, sont également identifiées à l'aide de l'azur A, le bleu de méthylène et le bleu de crésyl. Les granules se colorent moins bien à la thionine, le rouge neutre, la safranine O, le bleu de toluylène et l'acridine orange. Dans les conditions utilisées, la matrice de la zone de réserve et la matrice de la zone hypertrophique inférieure (en voie de calcification) se colorent, alors que les matrices des zones prolifératives et hypertrophiques supérieures ne prennent pas les colorants. La gallocyanine, le violet cristal, la fuchsine basique, l'azocarmin B, le bleu de gallamine et la bleu alcian ne se colorent pas ou donnent des réactions colorées différentes de celles décrites ci-dessus. Il semble que le pK et le poids moléculaire des colorants jouent un rôle important, mais ils ne paraissent pas être les seuls facteurs intervenant dans la coloration des granules. Un changement, lié à la calcification, semble intervenir au niveau du matériel métachromatique (probablement des polysaccharides protéiques), aussi bien dans la matrice que les cellules cartilagineuses épiphysaires.
    Abstract: Zusammenfassung Handpräparierte Schnitte von frischem Epiphysenknorpel junger Ratten wurden bei einem pH von 4,5 in 0,01% igen Lösungen verschiedener kationischer Farbstoffe folgender Klassen gefärbt: Thiazin, Oxazin, Azin, Triphenylmethan, Acridin und Phthalocyanin. Die intracellulären β-und γ-metachromatischen Granula, erstmals mit Toluidinblau im frischen Gewebe nachgewiesen, konnten auch gut mit Azur A, Methylenblau und Brillantkresylblau dargestellt werden. Die Granula konnten ebenfalls, aber weniger gut, mit Thionin, Neutralrot, Safranin D, Toluylenblau und Acridinorange gefärbt werden. Unter diesen Färbungsbedingungen werden die inaktive Matrixzone und die untere hypertrophische (verkalkende) Matrixzone angefärbt, während die proliferative und die obere hypertrophische Matrixzone sich nicht färben. Gallocyanin, Kristallviolett, basisches Fuchsin, Azokarmin B, Gallaminblau und Alzianblau färbten entweder gar nicht, oder gaben ein anderes als das obenbeschriebene Färbemuster. Es wird vorgeschlagen, daß das pK und das Molekulargewicht der Farbstoffe wichtig aber nicht unbedingt die einzigen Faktoren sind, die die Färbung der Granula bestimmen. Die Resultate zeigen, daß eine Veränderung im metachromatischen Material (vermutlich Proteinpolysaccharide) vorliegt, und zwar sowohl in der Matrix als in den Zellen des Epiphysenknorpels; diese Veränderung scheint im Zusammenhang mit der Verkalkung zu stehen.
    Notes: Abstract Hand-cut sections of fresh epiphyseal cartilage from young rats were stained at pH 4.5 in 0.01% solutions of various cationic dyes of the thiazine, oxazine, azine, triphenylmethane, acridine, and phthallocyanin classes. The intracellular β-and γ-metachromatic granules, previously demonstrated in fresh tissues with toluidine blue, were also demonstrated well with azure A, methylene blue, and brilliant cresyl blue. The granules were also demonstrated, but not as well, by thionin, neutral red, safranin O, toluylene blue, and acridine orange. Under the conditions of staining, the reserve zone matrix and the lower hypertrophic (calcifying) zone matrix stained, whereas the proliferative and upper hypertrophic zone matrix did not stain. Gallocyanin, crystal violet, basic fuchsin, azocarmine B, gallamine blue, and alcian blue either did not stain, or gave a different pattern of staining from that described above. It is suggested that the pK and molecular weight of the dyes are important, but not necessarily the only factors in determining the staining of the granules. The results indicate that there is a change in the metachromatic material (presumably proteinpolysaccharide) in both the matrix and cells of epiphyseal cartilage, which appears to be related to calcification.

    Description / Table of Contents: Résumé Un procédé de dissection a été mis au point pour permettre l'analyse zonale du cartilage de l'épiphyse des os de la jambe d'un foetus bovin. Des échantillons de tissu complet et lavé venant des différentes zones ont été analysés pour déterminer leur contenu en électrolyte et en constituants organiques, ainsi que pour leur densité, cendres et humidité. Les résultats ont montré que lorsque la quantité de cendres et la densité augmentaient, l'eau contenu dans le tissu diminuait. Les quantités de cendres dans les zones de cartilage en voie de calcification étaient plus grandes qu'il avait été. Quand elles étaient exprimées comme un pourcentage du poids sec, elles étaient les plus importantes dans le cartilage lavé calcifié que dans le autre zones. Au début de la minéralisation du cartilage, la quantité de Na (m moles/l de tissu frais) diminuait tandis que celles du Ca et du P inorganique augmentaient. Les niveaux de Mg augmentaient pendant que la calcification se poursuivait, mais seulement à une faction du taux du Ca et du P. Les rapports Ca/P inorganique étaient les plus grands dans le cartilage au repos (Cartilage non-différentié hyalin), suggérant un lien initiale entre Ca et les chrondromucoprotéines. Cependant, au début de la calcification, pendant la prolifération du cartilage les rapports Ca/P étaient beaucoup plus petits (ca. 1.50) mais augmentaient graduellement avec l'advancement de la minéralisation. Des changements importants survenaient dans la composition de la phase organique, pendant la calcification endochondrale. Comme il a été déterminé par l'analyse de l'hydroxyproline la quantité de collagéne diminuait progressivement pendant la calcification du cartilage, mais augmentait rapidement pendant la formation d'os. Comme il a été déterminé par l'analyse de l'héxosamine et du sulfute les chrondromucoprotéines étaient aux niveaux les plus éléves pendant la prolifération du cartilage et diminuaient constamment au cours de la calcification. Cependant, bien que la calcification était déja très avancée dans le cartilage hypertrophique, de grandes quantites de mucopolysaccharides étaient encore présentes. Les rapports sulfure/hhéxosamine montraient un léger déclin pendant les premiéres étapes de la calcification, mais augmentaient beaucoup pendant le cours de la minéralisation. Les quantités d'acide sialique étaient plus grandes dans le cartilage de l'épiphyse que dans le cartilage au repos ou dans l'os. Les lipides augmentaient rapidement pendant la calcification du cartilage, mais étaient très réduites dans l'os complètement formé. La signification de ces résultats est discutée.
    Abstract: Zusammenfassung Eine Seziermethode, die eine Schichten-Analyse der Beinepiphysenplatte von Rinderfeten erlaubt, wurde entwickelt. Proben vor und nach Waschen des Gewebes der verschiedenen Schichten werden untersucht in bezug auf Elektrolyte und organische Bestandteile, als auch in bezug auf Dichte, Aschengehalt und Feuchtigkeit. Die Resultate zeigten eine Zunahme des Aschengehaltes und der Dichte, während der Wassergehalt abnahm. Unerwartet hoch waren die Aschenwerte im in Verkalkung begriffenen Knorpel. Ausgedrückt in Prozent Trockengewicht, ergab gewaschener, verkalkter Knorpel den höchsten Wert aller Zonen. In den Frühstadien der Knorpelmineralisation nahm der Natriumgehalt (m Mol/l Frischgewebe) ab, während Ca und anorganischer P zunahmen. Mit fortschreitender Verkalkung erhöhte sich auch der Magnesium-Spiegel, allerdings nur zu einem Bruchteil des Ausmaßes, in welchem Ca und P zunahmen. Die höchsten Ca/P anorg. Verhältnisse wurden im Ruheknorpel (undifferenzierter hyaliner Knorpel) gefunden, was auf eine initiale Bindung von Ca durch Chondromucoproteine hinweist. Die Ca/P-Verhältnisse proliferierenden Knorpels waren jedoch bei Verkalkungsbeginn viel tiefer (ca. 1.50). Diese nahmen allerdings mit fortschreitender Mineralisierung stetig zu. In der endochondralen Verkalkungsphase fanden markante Veränderungen in der Zusammensetzung des organischen Anteils statt. Basierend auf der Hydroxyprolinanalyse nahm der Collagengehalt in der knorpeligen Verkalkungsperiode fortschreitend ab, während er jedoch bei der Knochenbildung rasch zunahm. Die an Hand von Hexosamin- und Schwefelanalysen bestimmten Chondromucoproteingehalte ergaben Höchstwerte im proliferierenden Knorpel und fielen stetig ab mit zunehmender Verkalkung. Trotz der im hypertrophischen Knorpel schon weit fortgeschrittenen Verkalkung waren immer noch große Mengen an Mucopolysacchariden vorhanden. Die Schwefel/Hexosamin-Verhältnisse zeigten eine minimale Abnahme in den frühen Verkalkungsphasen, nahmen jedoch markant zu bei fortschreitender Mineralisation. Der Sialinsäurespiegel war im Epiphysenknorpel, verglichen mit demjenigen des Ruheknorpels oder Knochens, erhöht. In der knorpeligen Verkalkungsphase nahmen die Lipide rasch zu, während jedoch die Werte des vollständig ausgebildeten Knochens stark vermindert waren. Die Bedeutung dieser Ergebnisse wird besprochen.
    Notes: Abstract A dissection procedure has been devised to permit zonal analysis of the epiphyseal plate of fetal calf leg bones. Samples of whole and washed tissue from the various zones were analyzed for their content of electrolyte and organic constituents, as well as for density, ash and moisture. Results showed that as ash content and density increased, water content decreased. Ash levels in calcifying cartilage zones were unexpectedly high. When expressed as a percentage of dry weight, washed calcified cartilage had the highest content of any zone. In the early stages of the mineralization of cartilage, Na content (mmoles/l of fresh tissue) decreased as Ca and inorganic P increased. Magnesium levels increased as calcification proceeded, but only at a fraction of the rate of Ca and P. Ratios of Ca/inorganic P were highest in resting cartilage (non-differentiated hyaline cartilage), suggesting an initial binding of Ca to chondromucoproteins. However, at the onset of calcification in proliferating cartilage, Ca/P ratios were much lower (ca. 1.50), but gradually increased with advancing mineralization. Marked changes occurred in the composition of the organic phase during endochondral calcification. As determined by hydroxyproline analysis, collagen content progressively decreased during cartilaginous calcification, but increased rapidly during bone formation. As determined by hexosamine and sulfur analysis, chondromucoproteins were at highest levels in proliferating cartilage and decreased steadily as calcification increased. However, although calcification was already well advanced in hypertrophic cartilage, large amounts of mucopolysaccharide still were present. Sulfur/hexosamine ratios showed a slight decline during the early stages of calcification, but increased markedly with further mineralization. Sialic acid levels were elevated in epiphyseal cartilage over those in resting cartilage or bone. Lipids increased rapidly during cartilaginous calcification, but were greatly reduced in fully-formed bone. The significance of these findings is discussed.

    Notes: Abstract  The uptake of 45Ca and/or 14C by the skeleton of coral colonies has been commonly used to investigate the processes of calcification. This study reports the differential uptake of these tracers within different regions of the skeleton and tissues of individual corallites and polyps of the hermatypic coral Galaxea fascicularis. Incubation in 45Ca in the light resulted in 80 percent of the 45Ca taken up being deposited in the skeleton. Autoradiography of transverse and longitudinal slices of freeze-substituted polyps and corallites showed that in the light 45Ca was incorporated into the exsert septa, the outside of the thecal walls of the corallite and the inner edges of the septa. Incorporation did not occur in the costae. The radioactivity in the skeleton was considerably greater than in the tissues. In the dark, or in the presence of the photosynthetic inhibitor Diuron, 45Ca was taken up by the exsert septa and was patchily distributed in the corallite walls which suggests that it was not a result of isotopic exchange. The differential incorporation of 45Ca onto the exsert septa was confirmed by scintillation counting. Negligible radioactivity remained in the extrathecal coelenteron after a brief 5 min rinse in non-radioactive seawater. Only 0.1% of 14C taken up in the light was incorporated into the skeleton and this was confirmed by autoradiography. In the presence of Diuron or in the dark, very little 14C was incorporated into tissues or skeleton and in autoradiographs was either not evident in the skeleton or the distribution was similar to that seen in autoradiographs of 45Ca uptake. These results show that the deposition of 45Ca, and therefore calcium carbonate, occurs at specific loci on the skeleton of a corallite. In the dark, deposition occurs specifically at the growing points of the corallite. Differential deposition of calcium carbonate within individual corallites has not been previously reported.

    Notes: Abstract Analysis of the Escherichia coli population by multilocus enzyme electrophoresis (MLEE) has established its clonal organization, but there is increasing evidence that horizontal DNA transfer occurs in E. coli. We have assessed the genetic structure of the species E. coli and determined the extent to which recombination can affect the clonal structure of bacteria. A panel of 72 E. coli strains from the ECOR collection was characterized by random amplified polymorphic DNA (RAPD) and restriction-fragment-length polymorphism (RFLP) of the ribosomal RNA gene (rrn) regions. These strains have been characterized by MLEE and are assumed to reflect the range of genotypic variation in the species as a whole. Statistical analysis, including factorial analysis of correspondence (FAC) and hierarchical classifications, established that the data obtained with the three genetic markers are mutually corroborative, thus providing compelling evidence that horizontal transfer does not disrupt the clonal organization of the population. However, there is a gradient of correlation between the different classifications which ranges from the highly clonal structure of 132 group strains causing extraintestinal infections in humans to the less-stringent structure of B1 group strains that came mainly from nonprimate mammals. This group (B1) appears to be the framework from which the remaining non-A group strains have emerged. These results indicate that RAPD analysis is well suited to intraspecies characterization of E. coli. Lastly, treating the RAPD data by FAC allowed description of subgroup-specific DNA fragments which can be used, in a strategy comparable to positional cloning, to isolate virulence genes.

    Notes: Abstract Four DNA extraction protocols were compared for ability to produce DNA from the leaves or needles of several species: oak, elm, pine, fir, poplar and maize (fresh materials) and rhododendron (silica dried or frozen material). With the exception of maize and poplar, the species are known to be difficult for DNA extraction. Two protocols represented classical procedures for lysis and purification, and the other two were a combination of classical lysis followed by anion exchange chromatography. The DNA obtained from all procedures was quantified and tested by PCR and Southern hybridisation.Test results indicated superiority of one of the four protocols; a combination of CTAB lysis followed by anion exchange chromatography which enabled DNA extraction from all seven species. A second protocol also produced DNA from leaves or needles of all species investigated and was well suited for PCR applications but not Southern hybridisations.     Notes: Abstract A single-seed DNA extraction method was developed for rapid identification of plant genotype. The method was applied to 12 plant species, including the oil seeds sesame and soybean. The results were comparable to those obtained for oil-less seeds such as rice. This method will be useful for genotypic selection which requires rapid screening of large populations. It can also be used to identify varietal purity of seed stocks by PCR and RFLP analysis. The method includes two major steps, (i) treatment by proteinase K in an SDS extraction buffer, and (ii) grinding of a single half seed in the buffer after incubation. About 1.5–2 µg of DNA per half seed (the endosperm part) of rice was obtained and more than 200 half seed samples could be handled by one person in a day. The DNA could be used for fingerprinting and detection of target genes in a transgenic plant by PCR. The amplified PCR products from the half seed DNA exhibited the same banding patterns as those from leaf DNA. Yield and quality of DNA extracted from half seeds of rice was also sufficient for RFLP analysis. The remnant half seeds containing the embryo can be maintained for later germination of selected genotypes.

    Description / Table of Contents: Résumé On a étudié la formation de l'hydroxyapatite de calcium cristallin à partir de solutions d'ions de calcium et de phosphate et l'inhibition de la croissance de cristaux de l'hydroxyapatite de calcium au moyen de polyphosphonates et de polyphosphates. Les polyphosphonates, éthane hydroxy-1-diphosphonate-1,1 de disodium et dichlorométhanediphosphonate de disodium, sont inhibiteurs efficaces contre la croissance de cristaux de l'hydroxyapatite de calcium. Les polyphosphates sont aussi inhibiteurs efficaces contre la croissance de cristaux de l'hydroxyapatite de calcium tant que le niveau exigé de polyphosphate intact est présent dans le système. Cependant, à cause de leur instabilité hydrolytique, qui est soulignée par une température élevée, valeur de pH basse, et certaines enzymes, la concentration du polyphosphate diminue avec le tempsin vitro, et son activité comme inhibiteur est perdue. Au contraire aux polyphosphates, les polyphosphonates sont hydrolytiquement stables. Les polyphosphonates sont chimiosorbés sur la surface des microcristallites de l'hydroxyapatite de calcium, ainsi empêchant l'occurrence d'autre croissance de cristaux semblable à l'action d'autres poisons connus de croissance de cristaux. On propose l'extension de cette action sur la formation de l'apatite et cette stabilité des polyphosphonates aux applications médicales et dentaires concernant le metabolisme pathologique de calcium et de phosphate.
    Abstract: Zusammenfassung Die Bildung des kristallinen Calciumhydroxyapatit aus Lösungen, welche Calcium- und Phosphationen enthalten, und die Hemmung der Bildung von kristallinen Calciumhydroxyapatit durch Polyphosphonate und Polyphosphate wurden untersucht. Polyphosphonate, Dinatriumäthan-1-hydroxyl-1,1-diphosphonat und Dinatriumdichloromethandiphosphonate verhindern das Kristallwachstum des Calciumhydroxyapatits. Die Polyphosphate verhindern ebenfalls das Kristallwachstum des Calciumhydroxyapatits, solange die notwendige Konzentration des nicht hydrolysierten Polyphosphats vorhanden ist. Wegen ihrer hydrolytischen Unbeständigkeit, die durch hohe Temperatur, niedrige pH und bestimmte Enzyme erhöht wird, vermindert sich jedoch die Konzentration des Polyphosphats allmählichin vitro, und ihre Hemmungsaktivität geht verloren. Im Gegensatz zu den Polyphosphaten sind die Polyphosphonate hydrolytisch beständig. Die Polyphosphonate werden an der Oberfläche der Mikrokristallite des Calciumhydroxyapatits chemisorbiert und verhindern, wie andere bekannte Kristallwachstumsgifte, auf diese Weise weiteres Kristallwachstum. Die Beständigkeit der Polyphosphonate und ihre Chemisorption an dem Apatit empfehlen ihren Gebrauch in der ärztlichen und zahnärztlichen Praxis, soweit sie den pathologischen Calcium- und Phosphatstoffwechsel betreffen.
    Notes: Abstract The formation of crystalline calcium hydroxyapatite from solutions of calcium and phosphate ions and the inhibition of calcium hydroxyapatite crystal growth by polyphosphonates and polyphosphates have been studied. The polyphosphonates, disodium ethane-1-hydroxy-1,1-diphosphonate and disodium dichloromethane diphosphonate, are effective inhibitors of calcium hydroxyapatite crystal growth. The polyphosphates are also effective inhibitors of calcium hydroxyapatite crystal growth as long as the required level of intact polyphosphate is present in the system. However, because of their hydrolytic instability, which is enhanced by high temperature, low pH, and certain enzymes, the concentration of the polyphosphate decreases with timein vitro, and its activity as an inhibitor is lost. In contrast to the polyphosphates, the polyphosphonates are hydrolytically stable. The polyphosphonates are chemisorbed on the surface of the microcrystallites of calcium hydroxyapatite and, in the manner of other known crystal growth poisons, thus prevent further crystal growth. The stability of the polyphosphonates and their chemisorption on apatite suggest their use in medical and dental applications involving pathological calcium and phosphate metabolism.

    Description / Table of Contents: Résumé Des cultures d'épiphyse prélevées sur des rats et des souris ont été étudiées sous le microscope électronique. Un profile de granules mitochondriques de densité électronique a été trouvé. Les chondrocytes dans la zone proliférative avaint peu de granules, alors que ceux des zones successives ont montré une augmentation progressive de leur nombre et densité jusqu'à ce que la zone de calcification provisoire ait été atteinte. Cette zone a montré une distribution périphérique de mitochondries et une réduction du nombre et de la densité des granules mitochondriques. Du calcium isotopique 47 a été utilisé autoradiographiquement pour déterminer la location de calcium dans ces cellules. Des grains ont été trouvés sur les membranes R.E. et sur la plupart des mitochondries. La preuve d'un profile de ces granules et de leur rapport spatial avec la face de minéralisation indique une action éventuelle de mitochondries dès le début de la calcification de la matrice.
    Abstract: Zusammenfassung Epiphysekulturen von Ratten und Mäusen wurden unter dem Elektronenmikroskop untersucht. Ein Profil von mitochondrischen Körnchen mit elektronischer Dichte wurde gefunden. Chondrozyten in der Proliferationszone wiesen wenig Körnchen auf, während die der nachfolgenden Zonen allmählich an Zahl und Dichte zunahmen, bis die Zone der provisorischen Verkalkung erreicht wurde. Diese Zone zeigte eine periphere Verteilung der Mitochondrien und eine Abnahme in Zahl und Dichte der mitochondrischen Körnchen. Isotopes Kalzium 47 wurde autoradiographisch verwendet, um die Lage des Kalziums in diesen Zellen zu bestimmen. Körnchen wurden auf den E.R.-Membranen und auf einem Großteil der Mitochondrien gefunden. Der Nachweis eines Profils dieser Körnchen und ihres räumlichen Verhältnisses zur Mineralisierungsfläche weist auf einen möglich Einfluß der Mitochondrien mit Beginn der Matrixverkalkung hin.
    Notes: Abstract Rat and mice epiphyseal growth plates were studied with the electron microscope. A gradient of mitochondrial electron-dense granules was found. Chondrocytes in the proliferative zone had few granules, while those of the succeeding zones showed a gradual increase in number and density until the zone of provisional calcification was reached. This zone showed a peripheral distribution of mitochondria and a decrease in the number and density of mitochondrial granules. Isotopic47calcium was used autoradiographically to determine the location of calcium in these cells. Grains were found over the endoplasmic reticulum membranes and over most mitochondria. The demonstration of a gradient of these granules and their spatial relation to the mineralization front suggests a possible involvement of mitochondria in the onset of matrix calcification.

    Description / Table of Contents: Résumé Les effects d'une dose de 1 mg de vitamine D3 (40 000 Unités,) administrée par voie intra-veineuse, ont été étudiés sur l'histologie osseuse et le métabolisme du phosphore chez 19 sujets contrôles dont l'histologie était normale et 28 malades présentant les caractères histologiques de l'ostéomalacie par carence vitaminique D. L'administration de la vitamine D n'a entraîné aucune modification histologique ou biologique significative chez les sujets contrôles. Mais chez les ostéomalaciques, il est apparu en moins de sept jours une augmentation très significative du front de calcification à l'interface tissue-ostéoïde-tissue-calcifié. Cette modification s'accompagnait d'une ascension progressive de la phosphatémie et de la réabsorption tubulaire du phosphore atteignant dans le même délai des valeurs normales.
    Abstract: Zusammenfassung Bei 19 Patienten mit normaler Knochenhistologie und bei 28 Patienten mit histologisch gesicherter Osteomalacie wurde die Wirkung von 1 mg Vitamin D3 (40000 Iv) i.v. auf die Knochenhistologie und den Phosphatmetabolismus untersucht. Bei den Kontrollpatienten konnten keine signifikanten Änderungen nach Vitamin D festgestellt werden, wogegen die Osteomalacie-patienten innerhalb von 7 Tagen eine deutliche Zunahme der Verkalkungszone an der Grenze zwischen Osteoid- und Knochengewebe zeigten. Diese Änderung war von einer fortschreitenden Zunahme des Serum-Phosphates, verbunden mit einer gesteigerten renalen tubulären Rückabsorption des Phosphates begleitet; beide kehrten anschließend gleichzeitig wieder zur Norm zurück.
    Notes: Abstract The effect of a 1 mg dose of intravenous Vitamin D3 (40,000 i.u.) on bone histology and phosphate metabolism was investigated in 19 patients with normal bone histology and 28 patients with histological evidence of osteomalacia. No significant changes occurred in the control patients after Vitamin D but the patients with osteomalacia showed a marked increase, within seven days, in the proportion of osteoid having a calcification front. This was accompanied by a progressive rise in the serum phosphate, which was associated with an increase in the renal tubular reabsorption of phosphate to normal.

Cette propriété est exprimée par les concentrations de calcium (et de phosphate) de solutions synthétiques inductrices de calcification, de concentrations et d'activité connues. Un plasma humain typique a une activité inductrice correspondant à celle d'une solution contenant du calcium, 0,70 mM, Ca/P, 1,67 et fluorure, 0,05 mM. L'activité relativement peu élevée des sérums et des plasmas s'explique par la présence d'inhibiteurs ioniques. L'ion phosphate est l'un d'eux, mais ne peut rendre compte de l'inhibition totale. Des anions et des cations sont responsables de l'inhibition, avec un rôle majeur pour les cations. Les ions suivants, en concentration physiologique, jouent un rôle inhibiteur: P7O 7 4− , HCO 3 − , SiO 4 2− , CrO 4 2− , Mg2+, Zn2+. Un mélange de ces ions provoque une inhibition totale identique à celle du plasma.
    Abstract: Zusammenfassung Der Einfluß biologischer Flüssigkeiten auf den Verkalkungsvorgang kann in vitro und in vivo anhand ihrer Fähigkeit, enthärtete Blöcke von Zahnschmelz wieder zu härten, gemessen werden. Diese Aktivität wird ausgedrückt als Konzentration des Calciums (und Phosphates) synthetischer calcifizierender Lösungen mit bekannter Konzentration und Aktivität. Die Aktivität eines charakteristischen menschlichen Plasmas entsprach derjenigen einer Lösung folgender Zusammensetzung: Calcium 0,70 mM; Ca/P=1,67 und Fluorid 0,05 mM. Die relativ niedrige Aktivität von Serum und Plasma ist bedingt durch das Vorhandensein einer Anzahl ionischer Inhibitoren. Das Pyrophosphation ist ein solcher; er kann aber nicht für die gesamte Inhibition verantwortlich gemacht werden. Sowohl anionische als auch kationische Inhibitoren sind vorhanden, wobei die Kationen den Hauptanteil ausmachen. Folgende Ionen erwiesen sich in einer physiologischen Konzentration als Inhibitoren synthetischer Systeme: P7O 7 3− , HCO 3 − , SiO 4 1− , CrO 4 2− , Mg2+, Zn2+. Zusammengenommen verursachten diese Ionen eine Gesamtinhibition ähnlich derjenigen des Plasmas.
    Notes: Abstract The calcifying activities of biological fluids can be measured,in vitro andin vivo, by their ability to reharden softened blocks of tooth enamel. The activity is expressed in terms of the calcium (and phosphate) concentrations of synthetic calcifying solutions of known concentration and activity. A typical human plasma had an activity corresponding to that of a solution of the following concentration: calcium, 0.70 mM; Ca/P, 1.67 and fluoride, 0.05 mM. The relatively low activity of serums and plasmas was shown to arise from the presence of a number of ionic inhibitors. Pyrophosphate ion is one such inhibitor but cannot account for the major inhibition. Both anionic and cationic inhibitors were shown to be present, with the cations respresenting the major portion. The following ions at their reported physiological concentration were shown to be inhibitors in synthetic systems: P7O 7 4− , HCO 3 − , SiO 4 2− , CrO 4 2− , Mg2+, Zn2+. In combination, these ions caused a total inhibition similar to those for plasmas.

    Description / Table of Contents: Résumé Avec la modification d'une méthode de von Kossa nous avons macroscopiquement representé les dessins de calcification de la membrane élastique intérieure, ainsi que les grosses artères du bassin, des artères des extrémités inferieures, des artères du bras, de la rate et des reins. Dans les artères du type élastique, dans les A.a. ilicae communes et internae, se présentaient les dégénérations calcaires de la membrane élastique intérieure sous forme d'incrustations noires arrondies ou polygonales qui ont été formes en groupes. Elles étaient toujours présentes chez des enfants agés de plus de trois mois et chez des aux adultes. Dans les artères musculaires, se montraient les parties calcifiées de la membrane élastique intérieure en forme de «bandes de calcaire» noires et groupes en paires le long des bordures des fissures de la membrane. Dans le cas où existait une calcinose forcée, on pouvait montrer avec cette méthode tout le système de fissures de la mambrane élastique intérieure. Chez tous les morts âgés de 10 à 20 ans, nous avons trouvé des bandes de calcaire dans les artères musculaires des extrémités inferieures. Ces bandes de calcaire ont été constatées aussi dans des personnes plus âgées. Au point de la fusion des bandes de calcaire et des incrustations calcaire polygonales des dépôts calcaires en forme de feuille («feuilles de calcaire») se forment dans la membrane elastique. Les bandes de calcaire et les feuilles de calcaire représentent des points de cristallisation pour des dépôts calcaire granuleux. Ils se forment dans la plupart des cas sur le la côté externe tourné vers la media.
    Abstract: Zusammenfassung Mit einer modifizierten von Kossa-Methode wurden die Verkalkungsmuster der inneren elastischen Membran der großen Beckenarterien, der Arterien der unteren Extremität, der Oberarm-, Milz-und Nierenarterien makroskopisch dargestellt. In den Arterien vom elastischen Typ, i.e. in den Aa. ilicae communes et internae, erschienen die Verkalkungen der inneren elastischen Membran als schwarze rundliche oder polygonale Inkrustationen, die zu Gruppen angeordnet waren. Sie wurden häufig bereits bei Neugeborenen festgestellt. Bei Kindern, die älter waren als 3 Monate und bei Erwachsenen lagen sie stets vor. — In den muskulären Arterien traten die verkalkten Anteile der inneren elastischen Membran makroskopisch als schwarze paarweise angeordnete „Kalkbänder” entlang den Rändern vorgebildeter Membranspalten auf. Bei ausgeprägter Calcinose konnte mit der angewandten Methode das gesamte Spaltensystem der inneren elastischen Membran dargestellt werden. Kalkbänder wurden in den muskulären Arterien der unteren Extremität bei allen 10–20 Jahre alten Verstorbenen vorgefunden und waren auch in den nachfolgenden Altersstufen stets vorhanden. Beim Zusammenfluß von Kalkbändern und polygonalen Kalkinkrustationen entstehen in der inneren elastischen Membran folienartige Kalkablagerungen („Kalkfolien”). Die Kalkbänder und Kalkfolien stellen Kristallisationspunkte für körnige Kalkablagerungen dar, die zumeist an ihrer äußeren, der Media zugekehrten Seite entstehen.
    Notes: Abstract Calcification patterns of the internal elastic membrane of the main pelvic arteries, lower limb arteries, brachial, splenic and renal arteries were demonstrated grossly by a modified von Kossa technique. In the elastic segment in the common and internal iliac arteries, the membrane calcification appeared as groups of roundish or polygonal incrustations. They were found frequently in newborns, and were always present in infants of more than three months, as well as in adults. In the muscular arteries, the calcified parts of the internal elastic membrane appeared grossly as pairs of bands (“calcific bands”) along the edges of the pre-existing gaps in this membrane. When calcification was pronounced, the whole pattern of the membrane gaps could be demonstrated by the method used in this study. Calcific bands were found in the muscular arteries of the lower limbs in all 10–20 year-old subjects, and were always present in the older age groups. The confluence of calcific bands or polygonal membrane incrustations found in the iliac arteries lead to sheet-like membrane calcification. The calcific bands and sheets represent crystallizing points for grain-like calcific deposits, which appear later on the medial surface.

    Description / Table of Contents: Résumé Les rapports entre composants organiques et inorganiques du cartilage ont été étudiés au niveau des épiphyses tibiales chez des rats de 5 jours et au niveau des jonctions costo-chondrales chez des cobayes d'un mois. Voici les principaux résultats auxquels on est parvenu: (a) Dans les zones où la calcification s'amorce les cristallites d'apatite sont susceptibles d'être facilement éliminés des coupes en araldite et en glycol methacrylate (GMA) après traitement pendant une heure par une solution d'acide formique au 2%. (b) Les zones décalcifiées sont douées d'une faible densité aux électrons et vont exemptes d'une structure quelconque. Seulement la fixation à l'osmium peut révéler du matériel amorphe. (c) Indépendemment du fixateur et du moyen d'inclusion, les coupes décalcifiées, traitées par l'acétate d'uranyl et/ou par le citrate de plomb, montrent de fines structures allongées dont l'aspect est semblable à celui des cristallites. (d) Ces structures ainsi qu'un fond dense interposé prennent l'acide phosphotungstique (PTA) dans les coupes en GMA, une méthode celle-ci qui met en évidence les polysaccharides. (e) Le fond dense ne prend plus le PTA après traitement par la hyaluronidase et en même temps les structures semblables au cristallites apparaissent plus faibles. Après traitement par l'acétate d'uranyl et le citrate de plomb, l'attaque par la hyaluronidase ne produit aucune modification appréciable. Au contraire l'aspect des structures semblables aux cristallites apparait modifié par la digestion à la papaïne. (f) Quand la décalcification à l'EDTA précéde l'enrobage, des amas de matériel fibrillaire se mettent en évidence dans les zones où la calcification s'était amorcée. L'ensemble de ces résultats permet d'établir la nature organique des structures semblables au cristallites et porte aussi à admettre que celles-ci soient formées par de fins filaments cylindriques (digérés par la papaïne) entourés, et probablement engainés par del polysaccharides (digérés par la hyaluronidase et prennant le PTA dans les coupes en GMA).
    Abstract: Zusammenfassung Die Wechselbeziehungen zwischen den organischen und inorganischen Substanzen des verkalkenden Knorpels wurden in der tibialen und femuralen Epiphyse 5 Tage alter Ratten und in den Rippen 1 Monat alter Meerschweinchen untersucht. Die Hauptergebnisse waren: a) Die Hydroxyapatitkristalle der Frühverkalkungszonen können in Araldit- wie auch in Glycolmethylacrilatschnitten (GMA) mit einer lstündigen Behandlung durch 1% ige Ameisensäure leicht entfernt werden. b) Die entkalkten Zonen haben eine sehr niedrige Elektronendichte und enthalten keine sichtbaren Festkörper. Nach Fixierung mit Osmiumsäure kann man darin ein formloses Material erkennen. c) Eine Färbung der Schnitte mit Uranacetat und Bleicitrat, welche von den Fixier- und Einbettungsmitteln nicht beeinträchtigt wird, zeigt feine, längliche Körper (“Kristallgeister”) an, welche morphologisch den Kristallen von Hydroxyapatit gleichen. d) Die Färbung der GMA-Schnitte mit Phosphowolframsäure (PWS), eine Methode welche Polysaccharide anzeigt, hob diese “Kristallgeister” besonders hervor und demonstrierte auch eine dichte dazwischenliegende Grundsubstanz. e) Diese Substanz ist nach Behandlung mit Hyaluronidase nicht mehr färbbar mit PWS; die Färbbarkeit der “Kristallgeister” wird dabei jedoch nur verringert. Die Hyaluronidasebehandlung verändert die Uranacetat- und Bleicitratfärbung nicht. Die Feinstruktur der “Kristallgeister” wird durch Papainbehandlung sehr angegriffen. f) Falls die Schnitte vor dem Einbetten mit EDTA entkalkt werden, können Faserbündel in den Frühzonen der Verkalkung nachgewiesen werden. Diese Ergebnisse bestätigen die organische Natur der “Kristallgeister”. Sie beweisen auch, daß dieselben aus feinen Proteinstäbchen bestehen (verdaulich in Papain) und von Polysacchariden umgeben und vielleicht auch überzogen sind (verdaulich in Hyaluronidase und anfärbbar mit PWS).
    Notes: Abstract The organic/inorganic relationships in calcifying cartilage have been studied in tibialfemoral epiphyses of 5-day-old rats and in costochondral junctions of 1-month-old guinea pigs. The main results are: (a) Apatite crystallites in areas of early cartilage calcification are easily removed from araldite and glycol methacrylate (GMA) sections by 1-hour treatment with 2% formic acid. (b) The decalcified areas have a very low electron density and do not contain recognizable structures. Only after osmium fixation can an amorphous material be found in them. (c) Independently from the fixative and the embedding medium, staining decalcified areas with uranyl acetate and/or lead citrate reveals thin, elongated structures (crystallite ghosts) similar in shape to apatite crystallites. (d) These crystallite ghosts and a dense background are stained by phosphotungstic acid (PTA) in GMA sections, a method which reveals polysaccharides. (e) The dense background is no longer stained by PTA after hyaluronidase digestion and the stainability of the crystallite ghosts is reduced. After uranyl acetate/lead citrate staining there are no recognizable changes due to hyaluronidase. But, on the other hand, the fine structure of the crystallite ghosts is modified by papain digestion. (f) Clusters of fibrillar material are present in the areas of early calcification when EDTA decalcification is carried out before embedding the specimens. These results confirm the organic nature of the crystallite ghosts and suggest that they are formed by thin, rod-like protein/like structures (digested by papain) surrounded, and probably sheathed, by acid polysaccharides (digested by hyaluronidase and stained by PTA in GMA sections).

    Description / Table of Contents: Résumé L'activité des cellules osseuses et la composition du fémur de pondeuses furent examinées pendant sept jours de déficience calcique (diète contenant 0,13% de calcium) et sept jours de réplétion (diète contenant 3,2% de calcium). Du point de vue histologique, seul l'os cortical donnait des signes nets de résorption et d'activité ostéoclastique. Le nombre d'ostéoclastes dans l'os médullaire différait peu des valuers témoin pendant les périodes de déficience et de réplétion subséquente, sauf pour une augmentation significative au premier jour de déplétion. L'effect histologique le plus important dans l'os médullaire était une augmentation marquée en nombre d'ostéoblastes aux troisième, cinquième, et un peu moins au septième jours de déplétion. Le nombre d'ostéoblastes était en corrélation positive avec la teneur de l'os médullaire en ostéoide et négative avec son degré de calcification. L'activité de l'os médullaire en phosphatase alcaline augmentait avec la longueur de la déficience calcique. Un jour après le retour des pondeuses à une diète contenant 3,2% de calcium, la calcification de l'os médullaire avait augmenté de façon significative, le nombre d'ostéoblastes avait diminué au niveau ou au-dessous du niveau de contrôle et l'activité de la phosphatase alcaline avait baissé considérablement. L'importance de ces résultats est discutée par rapport au controle des populations des cellules dan l'os et au rôle de l'os médullaire.
    Abstract: Zusammenfassung Die Zahl der Knochenzellen und die Zusammensetzung des Femurs von Legehennen wurden während einer siebentägigen Calciumentzugsperiode (Calciumgehalt des Futters 0,13%) und einer siebentägigen Ersatzperiode (Calciumgehalt des Futters 3,2%) untersucht. Tag des Calciumentzuges, variierte die Zahl der Osteoklasten im Markknochen sowohl während der Entzugs- als auch während der nachfolgenden Ersatzperiode wenig. Die wichtigste histologische Änderung im Markknochen bestand in einer starken Zunahme in der Zahl der Osteoblasten am 3., 5. und etwas weniger am 7. Tag der Entzugsperiode. Die Zahl der Osteoblasten zeigte eine positive Korrelation mit dem Osteoidgehalt des Markknochens und eine negative mit dem Grade seiner Verkalkung. Die Aktivität der alkalischen Phosphatase im Markknochen war desto größer je länger den Hennen die calciumarme Ration verfüttert worden war. Die Wiederverabreichung der Ration, welche 3,2% Calcium enthielt, verursachte innerhalb eines Tages eine signifikante Zunahme in der Verkalkung des Markknochens, ein Absinken der Osteoblastzahl auf die Kontrollwerte oder unter sie und eine drastische Verringerung der alkalischen Phosphataseaktivität. Die Bedeutung dieser Ergebnisse in bezug auf die Kontrolle des Knochenzellenbestandes und auf die Funktion des Markknochens wird diskutiert.
    Notes: Abstract Bone cell activity and the composition of the femur of laying hens were studied during 7 days of calcium depletion on a 0.13% calcium diet and 7 days of calcium repletion on a 3.2% calcium diet. Histologically, only cortical bone showed clear signs of bone resorption and osteoclastic activity during the depletion period. The number of osteoclasts in medullary bone varied little from control values throughout both calcium depletion and repletion, except for a significant increase on the first day of depletion. The major histologicalchange in medullary bone was a marked increase in the number of osteoblasts on the third, fifth and, to a lesser extent, seventh, day of depletion. The number of osteoblasts in medullary bone was positively correlated with its osteoid content and negatively correlated with its degree of calcification. Alkaline phosphatase activity of medullary bone increased with the time the hens had been on the calcium-deficient diet. Returning the hens to the 3.2% calcium ration caused, within one day, a significant increase in medullary bone calcification, a decrease of osteoblast numbers to, or below, control levels, and a drastic reduction in alkaline phosphatase activity of medullary bone. The significance of these findings in relation to the control of bone cell populations and the functions of medullary bone is discussed.

Les cristaux se forment au niveau du front de calcification, à la surface interne du ligament. Ils sont entourés par une enveloppe. Ils se présentent comme des monocristaux d'aragonite, allongés et en forme d'aiguilles, dispersés dans le ligament. Bien que les constituants de la carapace et du ligament soient identiques, il existe des différences concernant l'augmentation quantitative de conchioline et une diminution en nombre, forme diverse, groupement et croissance des cristaux. Ces différences sont probablement liées à la fonction spécialisée du ligament.
    Abstract: Zusammenfassung Die Bildung und Struktur des inneren Ligamentes vonMytilus edulis undPinctada radiata wurden am Elektronenmikroskop untersucht. Dieser Teil des Ligamentes stammt von den Isthmuszellen ab, deren Form unregelmäßig säulenartig ist. Sie zeigen ein vorspringendes, rauhes endoplasmatisches Reticulum und einen Golgiapparat, welche sich mit der Bildung von Bläschen und Granula befassen, die schließlich in einem integralen Teil des Conchiolins eingebaut werden. Die Kristalle entstehen an der Calcifikationsgrenze an der inneren Oberfläche des Ligamentes und sind in Hüllen eingeschlossen. Sie bestehen aus langen, nadelförmigen, einzelnen Aragonit-Kristallen, die über das ganze Ligament verteilt sind. Obschon die Bestandteile der Muschel und des Ligamentes gleichartig sind, unterscheiden sich die beiden durch eine erhöhte conchiolinmenge, wie auch durch eine Abnahme der Anzahl der Kristalle, welche verschieden in der Form, in der Anordnung und im Wachstum sind. Dies alles ist vermutlich auf die spezielle Funktion des Ligamentes zurückzuführen.
    Notes: Abstract An electron microscope study was made of the formation and structure of the inner ligament ofMytilus edulis andPinctada radiata. This part of the ligament is derived from the isthmus cells which are irregular columnar in shape. They exhibit a prominent rough endoplasmic reticulum and a Golgi apparatus, which are concerned with the elaboration of vesicles and granules eventually incorporated into an integral part of the conchiolin. The crystals arise at the calcification front at the inner surface of the ligament and are enclosed in envelopes. They consist of long, needle-shaped, single aragonite crystals widely dispersed in the ligament. Although the components of the shell and ligament are similar, differences between them consist of an increased amount of conchiolin, as well as a decrease in the amount, diversity of form, arrangement and growth of the crystals; all probably related to the specialized function of the ligament.

Die Resultate lassen vermuten, daß unlösliche Phosphate, die mit diesen Ionen entstehen, eine Fällung von Apatit veranlassen. Die Bedeutung dieser Befunde bei pathologischen Verkalkungen und Calciphylaxis werden besprochen.
    Notes: Abstract The effect was studied of various polyvalent metal ions on the metastability of a calcification buffer. At concentration of 1 μM or less, Pb2+, Fe2+, Fe3+, Cr3+, Bi3+ and Al3+ ions were found to destabilize the buffer. The results suggest that insoluble phosphates formed by these ions initiate the precipitation of calcium apatite. The implications of these findings to pathological calcification and calciphylaxis are discussed.

Le degré de calcification au quatrième jour de culture measuré par la relation calcium/hydroxyproline, suit un fonction lineáire avec le logarithme des doses de sulfate de dehydroepiandrostérone employées (0.5, 1,0 et 2,0 mM). Les renseignements obtenus indiquent que les frontaux de 13 jours, cultivés “in vitro” constituent modeles experimentaux appropriés pour étudier l'effet des androgénes sur le tissue osseux.
    Abstract: Zusammenfassung Stirnbeine von Hühnerembryonen an ihrem 12. und 13. Entwicklungstag entnommen und in vitro kultiviert zeigen verschiedene Arten der Glucoseverwertung der Periostzellendichte, des Calcium- und Hydroxyprolingehaltes. Wird Dehydroepiandrosteronsulfat dem Medium in einer 1 mM-Konzentration zugegeben, so beteiligen sich die 12tägigen Stirnbeine vorwiegend an der Knochengewebesynthese, während die 13tägigen signifikant stärker verkalken als die Kontrollen. Gemessen an der Calcium/hydroxyprolin Ratio bildet die Verkalkung der 13tägigen Stirnbeine eine lineare Funktion mit den Logarithmen der verwendeten Dosen von Dehydroepiandrosteronsulfat (0,5, 1,0 und 2,0 mM). Das in vitro kultivierte 13tägige Stirnbein schein ein geeignetes Experimentiermodell zur Studie der Dehydroepiandrosteronsulfatwirkung auf das Knochengewebe zu sein, weil es das grundlegende Phänomen (erhöhte Verkalkung) wiedergibt, welches man auch bei mit Androgenen behandelten Menschen und Tieren beobachtet.
    Notes: Abstract Chick embryo frontal bones at 12 and 13 days of development cultivatedin vitro exhibit different patterns of glucose utilization, periosteal cellular density and calcium and hydroxyproline content. When dehydroepiandrosterone sulfate is added to the medium at a concentration 1 mM, 12-day frontals engage primarily in osteoid tissue synthesis while 13-day frontals calcify at a significantly greater rate than controls. Measured with the ratio calcium/hydroxyproline, the calcification of 13-day frontals follows a linear function with the logarithm of the doses of dehydroepiandrosterone sulfate employed (0.5, 1.0 and 2.0 mM). The 13-day frontal bone cultivatedin vitro seems to be an adequate experimental model for the study of the effects of dehydroepiandrosterone sulfate on bone tissue because it reproduces the basic phenomenon (increased calcification) observed in man and animals treated with androgens.

    Notes: Abstract Bone sialoprotein (BSP) has an affinity to collagen fibrils [25]. A role of carbohydrate chains in the affinity was examined by removing sialic acids of BSP. Neuraminidase treatment of the BSP increased the binding to collagen. Binding sites of BSP on collagen were examined by biochemical and electron-microscopic methods. Purified bovine BSP was labeled with biotin. Collagen α chains or CNBr peptides were separated by electrophoresis and transfered to nitrocellulose membranes. The membranes were incubated with the biotin-labeled BSP, and the bound BSP was visualized with avidin conjugated with alkaline phosphatase. The labeled BSP was preferentially bound to the α 2 chain, and peptides derived from α 2 chain. In another experiment, the labeled BSP was incubated with reconstituted native collagen fibrils. The mixture was put on a copper grid, reacted with avidin conjugated with gold particles, and observed with an electron microscope. The gold particles were seen mainly within hole zones of the fibrils. BSP bound to the α 2 chain within the hole zones may regulate the onset of calcification at hole zones and the cell binding to collagen fibrils.

    Description / Table of Contents: Résumé L'objet de ce travail était de comparer le phénomène de la maturation postéruptive dans les molaires des rats ordinaires et sans germes à qui on a donné à manger un régime «non-cariogène». On a étudié les effets de l'application topique du fluorure et de la nourriture de la penicilline sur la maturation. Les molaires des rats ordinaires (nourris d'un régime «chow» — aliment mixte pour les animaux préparé d'habitude sous la forme d'une farine ou en boulettes) traités topiquement avec une solution à 1% de NaF et les molaires des rats ordinaires nourris d'un régime «chow» complété avec la penicilline à 1% ont montré un degré de maturation significativement plus grand que les molaires des rats de la même portée nourris de «chow» et traités avec de l'eau. De l'autre côté, les molaires des rats sans germes traités topiquement avec une solution à 1% de NaF n'ont montré aucune différence significative dans le degré de maturation des molaires de rats de contrôle semblables traités topiqument avec l'eau. Ces résultats sont en accord avec l'hypothèse que dans un milieu normal le procédé de minéralisation (maturation) est opposé par un procédé de déminéralisation. Le procédé de déminéralisation est un résultat de la production d'acide par des bactéries qui métabolisent les aliments encastrés dans les «sulci» des molaires. On propose que dans les animaux ordinaires le fluorure et la penicilline peuvent influencer la maturation en empêchant le procédé de déminéralisation. Dans les animaux sans germes le procédé de déminéralisation est absent parce que la microflore orale est absente.
    Abstract: Zusammenfassung Der Zweck dieser Arbeit war ein Vergleich des nach dem Durchbruch auftretenden Reifungsphänomens der Backenzähne von gewöhnlich und von keimfrei gehaltenen Ratten, die mit einer nicht-cariogenen Diät ernährt wurden. Es wurden die Auswirkungen von topisch angewendetem Fluorid, verbunden mit Penicillin-Fütterung auf die Reifung untersucht. Die Backenzähne der gewöhnlich gehaltenen Ratten (mit “Chow”-Diät ernährt), die mit einer 1%igen Na-Fluoridlösung topisch behandelt wurden, und solchen deren “Chow”-Diät zu 1% mit Penicillin versetzt wurde, zeigten einen erheblich größeren Reifungsgrad als die Backenzähne von Tieren des gleichen Wurfes, die nur mit der “Chow”-Diät und Wasser ernährt wurden. Diese Resultate stimmen mit der Hypothese überein, daß in einer normalen Umgebung dem Mineralisationsprozeß (Reifung) ein Demineralisationsprozeß entgegenwirkt. Der Demineralisationsprozeß ist durch eine Säureproduktion von Bakterien bedingt, welche die in die Zahnfurchen eingepreßten Nahrungsbestandteile metabolisieren. Es wird die Annahme vorgeschlagen, daß Fluoride und Penicillin bei den gewöhnlich gehaltenen Tieren die Reifung durch eine Inhibition des Demineralisationsprozesses beeinflussen. Bei keimfrei gehaltenen Tieren findet dagegen kein Demineralisationsprozeß statt, da die orale Mikroflora fehlt.
    Notes: Abstract The purpose of this work was to compare the phenomenon of post-eruptive maturation in molars of conventional and germfree rats fed non-cariogenic diets. The effects of topical application of fluoride and feeding penicillin on maturation were studied. The molars of conventional rats (fed a chow diet) treated topically with a 1% NaF solution and the molars of conventional rats fed a chow diet supplemented with 1% penicillin showed a significantly greater degree of maturation than did the molars of littermate rats fed chow and treated with water. On the other hand, the molars of germfree rats treated topically with a 1% NaF solution showed no significant difference in degree of maturation from the molars of similar control rats topically treated with water. These data are consistent with an hypothesis that in a normal environment the mineralization (maturation) process is opposed by a demineralization process. The demineralization process is a result of production of acid by bacteria metabolizing the diet impacted in the sulci of molars. It is proposed that in conventional animals fluoride and penicillin may influence maturation by inhibiting the demineralization process. In the germfree animals the demineralization process is absent because the oral microflora is absent.

    Description / Table of Contents: Résumé Certaines régions du cortex des fibres de kératine contiennent une proportion relativement élevée de matériel non kératinisé. L'orientation microfibrillaire dans ces régions est biaxiale et on y trouve, en outre, des zones de matériel amorphe. Les sels calciques, et en particulier l'hydroxyleapatite, sont étudiés par diffraction électronique qui permet de les localiser et de les identifier. En outre, cette technique permet de déterminer la fréquence de cellules nonkératinisées dans la partie interne du cortex et de les comparer avec les propriétés histochimiques de cette région. Une comparison est effectuée entre la calcification de la kératine et celle intéressant le collagène et l'élastine.
    Abstract: Zusammenfassung Es wird gezeigt, daß Bereiche im Cortex von gewissen Keratinfasern einen relativ hohen Anteil an nichtkeratinösem Material enthalten. Ferner konnte nachgewiesen werden, daß die Orientierung der Mikrofibrillen in diesen Bereichen biaxialer Art ist, und daß Bezirke von amorphem Material vorhanden sind. Die Calciumsalze und insbesondere das Hydroxyapatit konnten mit Hilfe der Elektronendiffraktion lokalisiert und identifiziert werden. Diese Methode hat sich auch als wertvoll erwiesen, um die Wechselbeziehung zwischen dem Vorkommen von einigen nichtkeratinisierten Zellen im Cortexinnern und den histochemischen Eigenschaften dieser Regionen zu zeigen. Ein Vergleich wird gemacht zwischen der Umgebung der Verkalkungsstellen im Keratin mit derjenigen im Kollagen und im Elastin.
    Notes: Abstract Regions in the cortex of some keratin fibres are shown to contain a relatively high proportion of non-keratinous material. The microfibrillar orientation in these regions is shown to be biaxial in nature and, in addition, areas of amorphous material are reported. Calcium salts, in particular hydroxyapatite, have been located and identified using electron diffraction, which has also proved valuable in correlating the incidence of some non-keratinised cells in the inner part of the cortex with the histochemical properties of this region. A comparison is made between the environment of the sites of calcification in keratin with those in collagen and elastin.

    Description / Table of Contents: Résumé Les facteurs, influençant la vitesse et l'intensité du phénomène d'association des ions calcium et phosphates avec des fibres contenant du collagène, et préparés à partir du tendon de boeuf par deux méthodes d'extraction différentes, ont été étudiés. Les fibres, obtenues par ces deux méthodes, nécessitent spécifiquement du phosphate pour absorber du calcium et vice versa. L'absorption ionique des deux préparations est inhibée par du Mg++, du pyrophosphate et un peptide acidique, isolé du sérum humain. Alors que les fibres contenant du collagène, préparées selon les deux méthodes, présentent une absorption ionique à des vitesses sensiblement identiques, seule une des méthodes donne une matrice réagissant positivement à la technique de coloration au nitrate d'argent de von Kossa. Etant donné que les deux critères de calcification sont intéressés de façon identique par des conditions de réaction et par des inhibiteurs, il apparait que les deux facteurs sont des manifestations de différents stades de calcification et que des études d'absorption ionique fournissent une base quantitative d'appréciation de la calcification, pouvant être d'importance pour l'étude du mécanisme et de contrôle de la minéralisation tissulaire.
    Abstract: Zusammenfassung Überprüft wurden die Faktoren, welche Geschwindigkeit und Ausmaß der Erscheinung beeinflussen, wobei Calcium- und Phosphationen sich mit den kollagenhaltigen, durch zwei verschiedene Extraktionsmethoden aus Rindersehnen gewonnenen Fasern eng zusammenbinden. Die mit beiden Methoden zubereiteten Fasern benötigen spezifisch Phosphat für die Calciumaufnahme und Calcium für die Phosphataufnahme. Die Ionenaufnahme beider Arten wird durch Mg++, Pyrophosphat und saure, aus dem menschlichen Serum isolierte Peptide gehemmt. Während die nach beiden Methoden präparierten kollagenhaltigen Fasern eine Ionenaufnahme von beinahe gleicher Geschwindigkeit verursachen, ergibt nur eine dieser Methoden eine Matrix, die mit der Silbernitratfärbung nach vonKossa positiv reagiert. Da beide Calcifikationskriterien gleicherweise durch Reaktionsbedingungen und Inhibitoren beeinflußt werden, wird daraus geschlossen, daß beide Erscheinungen verschiedener Stadien des Gesamtcalcifikationsprozesses sind. Untersuchungen über die Ionenaufnahme ergeben eine quantitative Angabe der Verkalkung, welche für die Erforschung des Mechanismus und der Kontrolle der Mineralisation der Gewebe wichtig sein könnte.
    Notes: Abstract Factors that influence the rate and extent of the phenomenon in which calcium and phosphate ions become firmly associated with collagen-containing fibers prepared from beef tendon by two different extraction methods have been investigated. The fibers produced by both methods specifically require phosphate for calcium uptake and calcium is required for phosphate uptake. Ion uptake by both types is inhibited by Mg++, pyrophosphate, and an acidic peptide isolated from human serum. Whereas the collagen-containing fibers prepared by both methods induce ion uptake at nearly identical rates, only one of the methods produced a matrix that gives a positive response to the silver nitrate staining technique of von Kossa. Since both criteria of calcification are similarly influenced by reaction conditions and inhibitors, it is concluded that both are manifestations of different stages of the overall calcification process and that studies of ion uptake provide a quantitative assessment of calcification which could be of importance for investigating the mechanism and control of tissue mineralization.

    Notes: Abstract Bone sialoprotein (BSP) containing an Arg-Gly-Asp cell-binding sequence was purified from bovine bone 4 M guanidine-HCl extract after HCl demineralization by a series of chromatographic procedures. When this protein was coated on culture dishes in the presence of type I collagen, it increased both DNA content and alkaline phosphatase (ALP) activity in osteoblast-like MC3T3-E1 cells, and stimulated calcification in the cells, whereas fibronectin, another cell-binding protein, showed a marked increase in the DNA content but had little effect on the ALP activity. These findings suggest that BSP is mitogenic for preosteoblasts and differentiating the cells into osteoblasts, thereby stimulating bone calcification

    Notes: Abstract  The ultrastructure of egg cells in Abies alba was examined to elucidate the lack of maternal inheritance of plastids. Before fertilization, maternal plastids are absent in the perinuclar zone containing mainly mitochondria and smooth endoplasmic reticulum. During egg cell development the maternal plastids are transformed into large inclusions which are situated mostly towards the periphery of the egg cell, and finally disintegrate. As a consequence, they do not participate in zygote formation. RFLP analysis of cpDNA of parental trees and their F1 interspecific hybrids (A. alba×A. numidica, A. alba×A. nordmanniana, A. nordmanniana×A. Alba) using HindIII and BamHI showed a paternal mode of cpDNA inheritance. Paternal inheritance has also been found with PCR/RFLP analysis of cpDNA from parental trees and their hybrids (A. alba×A. pinsapo, A. pinsapo×A. alba, A. pinsapo×A. numidica) using ApaI and HaeIII digests, as well as in the crosses of A. cephalonica×A. nordmanniana, A. nordmanniana×A. cephalonica, A. cephalonica×A. numidica using TagI digests.

    Notes: Abstract The inheritance of DNA markers was investigated in 27 F2 progeny from a single F1 hybrid derived from a wide cross inUromyces appendiculatus. This cross was unusual because asexual spores were used to fertilize sexual fruiting structures. Sixty percent of the DNA markers failed to segregate according to simple Mendelian ratios. Segregation bias was evident, in that F2 progeny inherited on average 91 % of maternal bands and 52% of paternal bands, which deviates significantly from the expected value for each of 75% for dominant markers. Because of these distortions, linkage mapping was not possible with this population. Evaluation of two F1s from a second wide cross, reciprocals obtained by normal fertilization, also showed non-Mendelian inheritance of one of three co-dominant RFLPs and five of six isozyme markers, indicating that the method of crossing was probably not responsible for the abnormal segregation patterns in the first cross. Either genetic incompatibility, similar to that of an interspecific cross, or selection of particular genotypes could explain the genetic anomalies reported here.

    Notes: Abstract Planktonic, filamentous cyanobacterial strains from different genera, both toxic and nontoxic strains, were characterized by SDS-PAGE of whole-cell proteins and PCR/RFLP of the 16S rRNA gene. Total protein pattern analysis revealed the mutual relationships at the genus level. Restriction fragment length polymorphism (RFLP) of the 16S rRNA gene with reference strains proved to be a good method for the cyanobacterial taxonomy. The nonheterocystous strains outgrouped from the nitrogen-fixing ones. With both methods, Aphanizomenon clustered with Anabaena, and Nodularia with Nostoc. In the RFLP study of Anabaena, the neurotoxic strains were identical, but the hepatotoxic ones formed a heterogeneous group. Genetic distances found in the RFLP study were short, confirming that close genotypic relationships underlie considerable diversity among cyanobacterial genera.

    Notes: Abstract Megaplasmid DNA was detected in ten isolates belonging to the recently described thermophilic eubacterial species Thermus oshimai and isolated from hot springs in Portugal (eight isolates) and Iceland (two isolates). The estimated size of the large plasmids purified from T. oshimai SPS-18 from S. Pedro do Sul, Portugal, and from isolate JK-91 from Hveragerdhi-Hengill, Iceland, was 214 and 275 kb, respectively. No sequence homologous to isolate SPS-18 megaplasmid is present in chromosomal DNA as indicated by Southern hybridization analysis. Overall examination of the HindIII fragment profiles of megaplasmid DNAs purified from isolates from the same geographical area gave similar but not always identical restriction profiles on agarose gels. Restriction fragment length polymorphism (RFLP) was higher for megaplasmids present in isolates purified from the Portuguese and Icelandic isolates than for megaplasmids from the same hot spring. Megaplasmid RFLP correlated with previous results obtained on the polymorphism of macrorestriction patterns of whole genomic DNA and with the RFLP of co-resident small plasmid DNA that was found in one half of the isolates examined. The 16-kb HindIII–HindIII fragment from isolate SPS-18 megaplasmid showed DNA–DNA homology with restriction fragments of similar size generated by the large plasmids present in all the other isolates, even in those from hot springs of widely separated geographical areas. This suggests a high degree of sequence conservation in T. oshimai megaplasmids.

    Notes: Abstract The chalcone synthase is a key enzyme that catalyses the first dedicated reaction of the flavonoid pathway in higher plants. The chs gene and its protein product in rice has been investigated. The presence of a chalcone synthase (CHS) protein in rice seedlings and its developmental stage-specific expression has been demonstrated by western analysis. The chalcone synthase of rice was found to be immunologically similar to that of maize. A rice cDNA clone, Os-chs cDNA, encoding chalcone synthase, isolated from a leaf cDNA library of an indica rice variety Purpleputtu has been mapped to the centromeric region of chromosome 11 of rice. It was mapped between RFLP markers RG2 and RG103. RG2 is the nearest RFLP marker located at a genetic distance of 3.3 cM. Some segments of chromosome 11 of rice including chs locus are conserved on chromosome 4 of maize. The markers, including chs locus on chromosome 11 of rice are located, though not in the same order, on chromosome 4 of maize. Genetic analysis of purple pigmentation in two rice lines, Abhaya and Shyamala, used in the present mapping studies, indicated the involvement of three genes, one of which has been identified as a dominant inhibitor of leaf pigmentation. The Os-chs cDNA shows extensive sequence homology, both for DNA and protein (deduced), to that of maize, barley and also to different monocots and dicots.

    Notes: Abstract The A genome of the Triticeae is carried by three diploid species and subspecies of the genus Triticum: T. monococcum ssp. monococcum, T. monococcum ssp. boeoticum, and T. urartu, the A-genome donor of bread wheat. These species carry many genes of agronomic interest, including disease resistances, and may also be used for the genetic mapping of the A genome. The aim of this study was to evaluate the variability present in a sample of 25 accessions representative of this group using RFLP markers. Twenty probes, consisting of genomic DNA or cDNA from wheat, were used in combination with four restriction enzymes. A high level of polymorphism was found, especially at the interspecific level. Selecting the most informative enzymes appeared to be of great importance in order to obtain a stable structure for the diversity observed with only 20 probes. The results are largely consistent with taxonomy and data relating to geographical origins. The probes were also tested on 14 wheat cutivars. A good correlation coefficient was found for their informative values on wheat cultivars and diploid lines. Whether the group of species studied here would be useful for genetic mapping remains to be determined. Nevertheless, RFLP markers will be useful to follow genes that can possibly be introgressed from these species into cultivated wheat.

    Notes: Abstract Polyploid plants in the genus Tripsacum, a wild relative of maize, reproduce through gametophytic apomixis of the diplosporous type, an asexual mode of reproduction through seed. Moving gene(s) responsible for the apomictic trait into crop plants would open new areas in plant breeding and agriculture. Efforts to transfer apomixis from Tripsacum into maize at CIMMYT resulted in numerou intergeneric F1 hybrids obtained from various Tripsacum species. A bulk-segregant analysis was carried out to identify molecular markers linked to diplospory in T. dactyloides. This was possible because of numerous genome similarities among related species in the Andropogoneae. On the basis of maize RFLP probes, three restriction fragments co-segregating with diplospory were identified in one maize-Tripsacum dactyloides F1 population that segregated 1∶1 for the mode of reproduction. The markers were also found to be linked in the maize RFLP map, on the distal end of the long arm of chromosome 6. These results support a simple inheritance of diplospory in Tripsacum. Manipulation of the mode of reproduction in maize-Tripsacum backcross generations, and implications for the transfer of apomixis into maize, are discussed.

    Notes: Abstract Quantitative trait loci (QTLs) for resistance to pathogen populations of Scelerospora graminicola from India, Nigeria, Niger and Senegal were mapped using a resistant x susceptible pearl millet cross. An RFLP map constructed using F2 plants was used to map QTLs for traits scored on F4 families. QTL analysis was carried out using the interval mapping programme Mapmaker/QTL. Independent inheritance of resistance to pathogen populations from India, Senegal, and populations from Niger and Nigeria was shown. These results demonstrate the existence of differing virulences in the pathogen populations from within Africa and between Africa and India. QTLs of large effect, contributing towards a large porportion of the variation in resistance, were consistently detected in repeated screens. QTLs of smaller and more variable effect were also detected. There was no QTLs that were effective against all four pathogen populations, demonstrating that pathotype-specific resistance is a major mechanism of downy mildew resistance in this cross. For all but one of the QTLs, resistance was inherited from the resistant parent and the inheritance of resistance tended to be the result of dominance or over-dominance. The implications of this research for pearl millet breeding are discussed.

    Notes: Abstract F2 progeny (105 individuals) from the cross Jo4002 x Sv3402 were used to identify DNA markers associated with palmitic-acid content in spring turnip rape (Brassica rapa ssp. oleifera). QTL mapping and ANOVA analysis of 140 markers exposed one linkage group with a locus controlling palmitic-acid content (LOD score 27), and one RAPD (random amplified polymorphic DNA) marker, OPB-11a, closely linked (1.4 cM) to this locus. Palmitic-acid content in the 62 F2 plants with the visible allele of marker OPB-11a was 8.45 ±3.15%, while that in the 24 plants without it was 4.59 ±0.97%. As oleic-acid concentration is affected by a locus on the same linkage group as the palmitic-acid locus, this locus probably controls the chain elongation from palmitic acid to oleic acid (through stearic acid). Marker OPB-11a may be used in future breeding programs of spring turnip rape to simplify and hasten the selection for palmitic-acid content.

    Notes: Abstract Restriction fragment length polymorphisms of cytoplasmic DNAs and nuclear rDNA were analyzed in several Cichorium intybus genotypes, comprising four white inbred lines, eight red witloof experimental lines, and a number of F1 hybrids derived from two white parents. Chloroplast and mitochondrial restriction patterns led to the distinction between two different cytoplasms, called I and II. Southern hybridization using a nuclear rDNA probe revealed that all the lines possessed two types of rDNA repeat units. The shortest unit was 10 kb and was common to all lines. The largest rDNA repeat unit was 10.5 kb in lines I and 10.4 kb in lines II. In addition, a sequence heterogeneity between the 10.5 and 10.4-kb rDNA repeat units was revealed by Sac I digestion. A 10-kb rDNA unit was successively cloned, mapped, and used as a probe to check the genetic purity of F1 hybrid seeds between line I and II white parents. We found a 30% average percentage of impurities, originating both from selfing and full-sib crossing, in different open-pollinated hybrid samples.

    Notes: Abstract Resistance to the soybean cyst nematode (SCN) (Heterodera glycines Ichinohe) is difficult to evaluate in soybean [Glycine max (L.) Merr.] breeding. PI 437.654 has resistance to more SCN race isolates than any other known soybean. We screened 298 F6∶7 recombinant-inbred lines from a cross between PI 437.654 and ‘BSR101’ for SCN race-3 resistance, genetically mapped 355 RFLP markers and the I locus, and tested these markers for association with resistance loci. The Rhg 4 resistance locus was within 1 cM of the I locus on linkage group A. Two additional QTLs associated with SCN resistance were located within 3cM of markers on groups G and M. These two loci were not independent because 91 of 96 lines that had a resistant-parent marker type on group G also had a resistant-parent marker type on group M. Rhg 4 and the QTL on G showed a significant interaction by together providing complete resistance to SCN race-3. Individually, the QTL on G had greater effect on resistance than did Rhg 4, but neither locus alone provided a degree of resistance much different from the susceptible parent. The nearest markers to the mapped QTLs on groups A and G had allele frequencies from the resistant parent indicating 52 resistant lines in this population, a number not significantly different from the 55 resistant lines found. Therefore, no QTLs from PI 437.654 other than those mapped here are expected to be required for resistance to SCN race-3. All 50 lines that had the PI 437.654 marker type at the nearest marker to each of the QTLs on groups A and G were resistant to SCN race-3. We believe markers near to these QTLs can be used effectively to select for SCN race-3 resistance, thereby improving the ability to breed SCN-resistant soybean varieties.

    Notes: Abstract Several gene linkage maps have been produced for cultivated barley. We have produced a new linkage map for barley, based on a cross between Hordeum vulgare subsp. spontaneum and Hordeum vulgare subsp. vulgare (Hvs x Hvv), having a higher level of polymorphism than most of the previous barley crosses used for RFLP mapping. Of 133 markers mapped in the Hvs x Hvv F2 population, 69 were previously mapped on other barley maps, and 26 were mapped in rice, maize, or wheat. Two known gene clones were mapped as well as two morphological markers. The distributions of previously mapped markers were compared with their respective barley maps to align the different maps into one consensus map. The distributions of common markers among barley, wheat, rice and maize were also compared, indicating colinear linkage groups among these species.

    Notes: Abstract Forty-seven recombinant inbred (RI) lines derived from a cross between two indica rices, cv ‘Phalguna’ and the Assam land race ARC 6650, were subjected to restriction fragment length polymorphism (RFLP) analysis using cloned probes defining 150 single-copy loci uniformly dispersed on the 12 chromosomes of rice. Of the probes tested, 47 detected polymorphism between the parents. Heterozygosity was calculated for each line and for each of the polymorphic loci. Average heterozygosity per line was 9.6% but was excessive (〉20%) in the 5 lines that seemed to have undergone outcrossing immediately prior to harvest. Average heterozygosity detected by each probe across the 47 RI lines was 9.7%. The majority of probes revealed the low level of heterozygosity (〈8%) expected for F5-F6 lines in a species showing about 5% outbreeding. On the other hand, 7 probes exhibited heterozygosity in excess of 15%, while with a eighth probe (RG2 from chromosome 11) heterozygosity varied according to the restriction enzyme employed, ranging from 2% with SaII to 72% with EcoRV. The presence of 34 recombination sites in a segment of the genome as short as 24 kb indicates a strong selection for recombination between two neighbouring loci, one required as homozygous for the ‘Phalguna’ allele, and the other heterozygous. Since selection was principally for yield advantage over that of the high-yielding parent, ‘Phalguna’, one or both of these loci may be important for heterosis in this cross. The results also indicate that heterozygosity as measured by RFLP can depend on the particular restriction endonuclease employed.

    Notes: Abstract Improving seedling vigor is an important objective of modern rice (Oryza saliva L.) breeding programs. The purpose of this study was to identify and map quantitative trait loci (QTL) underlying seedling vigor-related traits using restriction fragment length polymorphisms (RFLPs). An F2 population of 204 plants was developed from a cross between a low-vigor japonica cultivar ‘Labelle’ (LBL) and a high-vigor indica cultivar ‘Black Gora’ (BG). A linkage map was constructed of 117 markers spanning 1496 Haldane cM and encompassing the 12 rice chromosomes with an average marker spacing of 14 cM. The length of the shoots, roots, coleoptile and mesocotyl were measured on F3 families in slantboard tests conducted at two temperatures (18° and 25°C). By means of interval analysis, 13 QTLs, each accounting for 7% to 38% of the phenotypic variance, were identified and mapped in the two temperature regimes at a log-likelihood (LOD) threshold of 2.5. Four QTLs controlled shoot length, 2 each controlled root and coleoptile lengths and 5 influenced mesocotyl length. Single-point analysis confirmed the presence of these QTLs and detected additional loci for shoot, root and coleoptile lengths, these latter usually accounting for less than 5% of the phenotypic variation. Only 3 QTLs detected both by interval and singlepoint analyses were expressed under both temperature regimes. Additive, dominant and overdominant modes of gene action were observed. Contrary to what was predicted from parental phenotype, the low-vigor LBL contributed 46% of the positive alleles for shoot, root and coleoptile lengths. Positive alleles from the high-vigor parent BG were identified for increased root, coleoptile and mesocotyl lengths. However, BG contributed alleles with only minor effects for shoot length, the most important determinant of seedling vigor in water-seeded rice, suggesting that it would not be an ideal donor parent for introducing faster shoot growth alleles into temperate japonica cultivars.

    Notes: Abstract The objective of this study was to identify molecular markers linked to the wheat leaf rust resistance gene Lr24 derived from Agropyron elongatum (3DL/3Ag translocation). Two near isogenic lines (NILs), ‘Arina’ and Lr24/7 * “Arina”, were screened for polymorphism at the DNA level with 115 RFLP probes. Twenty-one of these probes map to the homoeologous group 3. In addition, 360 RAPD primers were tested on the NILs. Six RFLP probes showed polymorphism between the NILs, and 11 RAPD primers detected one additional band in the resistant NIL. The genetic linkage of the polymorphic markers with Lr24 was tested on a segregating F2 population (150 plants) derived from a cross between the leaf rust resistant Lr24/7 * “Arina” and the susceptible spelt (Triticum spelta) variety ‘Oberkulmer’. All 6 RFLP markers were completely linked to Lr24: one was inherited as a codominant marker (PSR1205), one was in coupling phase (PSR1203) and 4 were in repulsion phase (PSR388, PSR904, PSR931, PSR1067) with Lr24. The localization of these probes on chromosome 3D was confirmed by nulli-tetrasomic analysis. Distorted genotypic segregation was found for the Codominant RFLP marker PSR1205. This distortion can be explained by the occurrence of hemizygous plants. One of the 11 RAPD markers (OPJ-09) also showed complete linkage to theLr24 resistance gene. The polymorphic RAPD fragment was cloned and sequenced. Specific primers were synthesized, and they produced an amplification product only in the resistant plants. This specific marker allows a reliable and rapid screening of a large number of genotypes in practical breeding. Analysis of 6 additional lines containing Lr24 revealed that 3 lines have a smaller chromosomal segment of A. elongatum than lines derived from ‘Agent’, a commonly used gene donor for the Lr24 resistance gene.

    Notes: Abstract  Sugar beet (Beta vulgaris L.) is highly susceptible to the beet cyst nematode (Heterodera schachtii Schm.). Three resistance genes originating from the wild beets B. procumbens and B. webbiana have been transferred to sugar beet via species hybridization. We describe the genetic localization of the nematode resistance genes in four different sugar beet lines using segregating populations and RFLP markers from our current sugar beet linkage map. The mapping studies yielded a surprising result. Although the four parental lines carrying the wild beet translocations were not related to each other, the four genes mapped to the same locus in sugar beet independent of the original translocation event. Close linkage (0–4.6 cM) was found with marker loci at one end of linkage group IV. In two populations, RFLP loci showed segregation distortion due to gametic selection. For the first time, the non-randomness of the translocation process promoting gene transfer from the wild beet to the sugar beet is demonstrated. The data suggest that the resistance genes were incorporated into the sugar beet chromosomes by non-allelic homologous recombination. The finding that the different resistance genes are allelic will have major implications on future attempts to breed sugar beet combining the different resistance genes.

    Notes: Abstract  Comparative mapping within maize, sorghum and sugarcane has previously revealed the existence of syntenic regions between the crops. In the present study, mapping on the sorghum genome of a set of probes previously located on the maize and sugarcane maps allow a detailed analysis of the relationship between maize chromosomes 3 and 8 and sorghum and sugarcane homoeologous regions. Of 49 loci revealed by 46 (4 sugarcane and 42 maize) polymorphic probes in sorghum, 42 were linked and were assigned to linkage groups G (28), E (10) and I (4). On the basis of common probes, a complete co-linearity is observed between sorghum linkage group G and the two sugarcane linkage groups II and III. The comparison between the consensus sorghum/sugarcane map (G/II/III) and the maps of maize chromosomes 3 and 8 reveals a series of linkage blocks within which gene orders are conserved. These blocks are interspersed with non-homoeologous regions corresponding to the central part of the two maize chromosomes and have been reshuffled, resulting in several inversions in maize compared to sorghum and sugarcane. The results emphasize the fact that duplication will considerably complicate precise comparative mapping at the whole genome scale between maize and other Poaceae.

    Notes: Abstract  Quantitative triat loci (QTLs) for yield and related traits in rice were mapped based on RFLP maps from two indica/indica populations, Tesanai 2/CB and Waiyin 2/CB. In Tesanai 2/CB, 14 intervals carrying QTLs for eight traits were detected, including 3 for grain weight per plant (GWT), 2 for number of panicles per plant (NP), 2 for number of grains per panicle (NG), 1 for total number of spikelets per panicle (TNS), 1 for spikelet fertility (SF), 3 for 1000-grain weight (TGWT), 1 for spikelet density (SD), and 1 for number of first branches per main panicle. The 3 QTLs for GWT were located on chromosomes 1, 2, and 4, with 1 in each chromosome. The additive effect of the single locus ranged from 2.0 g to 9.1 g. A major gene (np4) for NP was detected on chromosome 4 within the interval of RG143-RG214, about 4 cM for RG143, and this locus explained 26.1% of the observed phenotypic variance for NP. The paternal allele of this locus was responsible for reduced panicles per plant (3 panicles per plant). In another population, Waiyin 2/CB, 12 intervals containing QTLs for six of the above-mentioned traits were detected, including 3 for GWT, 2 for each of NP, TNS, TGWT and SD, 1 for SF. Three QTLs for GWT were located on chromosome 1, 4, and 5, respectively. The additive effect of the single locus for GWT ranged from 6.7 g to 8.8 g, while the dominance effect was 1.7–11.5 g. QTL mapping in two populations with a common male parent is compared and discussed.

    Notes: Abstract  We mapped and characterized quantitative trait loci (QTLs) for resistance to Ustilago maydis and investigated their consistency across different flint-maize populations. Four independent populations, comprising 280 F3 lines (A×BI), 120 F5 lines (A×BII), 131 F4 lines (A×C) and 133 F4 lines (C×D), were produced from four European elite flint inbreds (A, B, C, D) and genotyped at 89, 151, 104, and 122 RFLP marker loci, respectively. All Fn lines were evaluated in field trials with two replications in five German environments. Genotypic variances were highly significant for the percentage of U. maydis infected plants (UST) in all populations, and heritabilities exceeded 0.69. Between five and ten QTLs were detected in individual populations by composite interval mapping, explaining between 39% and 58% of the phenotypic variance. These 19 different QTLs were distributed over all ten chromosomes without any clustering on certain chromosomes. In most cases, gene action was dominant or overdominant. Fourteen pairs of the detected QTLs for UST displayed significant digenic epistatic interactions, but only two of them did so after arcsin √UST/100 transformation. Significant QTL× environment interactions occurred frequently. Between two to four QTLs were common between pairs of populations. Population C×D was also grown in Chartres, a location with a high U. maydis incidence. Two out of six QTLs identified for Chartres were in common with QTLs detected across five German environments for C×D. Consequently, marker-assisted or phenotypic selection based on results from natural infection seem to be suitable breeding strategies for improving the resistance of maize to U. maydis.

    Notes: Abstract In order to determine the homologous regions shared by the cultivated Brassica genomes, linkage maps of the diploid cultivated B. rapa (A genome, n = 10), B. nigra (B genome, n = 8) and B. oleracea (C genome, n = 9), were compared. We found intergenomic conserved regions but with extensitve reordering among the genomes. Eighteen linkage groups from all three species could be associated on the basis of homologous segments based on at least three common markers. Intragenomic homologous conservation was also observed for some of the chromosomes of the A, B and C genomes. A possible chromosome phylogenetic pathway based on an ancestral genome of at least five, and no more than seven chromosomes, was drawn from the chromosomal inter-relationships observed. These results demonstrate that extensive duplication and rearrangement have been involved in the formation of the Brassica genomes from a smaller ancestral genome.

    Notes: Abstract  Restriction fragment length polymorphism (RFLP) and the structure of ribosomal RNA genes (rDNA) were investigated in 117 landraces of foxtail millet, Setaria italica (L.) P. Beauv. Five RFLP phenotypes were found when the genomic DNA was digested with BamHI; these were named types I–V. Of these types I, II and III were the most frequent. Type I was mainly distributed in the temperature zone, type II in the Taiwan-Philippines Islands and type III in South Asia. Restriction mapping of the cloned rDNA and comparison with RFLP phenotypes showed that the different types originated from a polymorphism in the length within the intergenic spacer (IGS) and BamHI site changes within the IGS.

    Notes: Abstract  Chloroplast DNA of 22 species of Acacia (Tourn.) Miller was digested with ten restriction endonucleases, Southern-blotted and probed with cloned fragments covering the chloroplast genome of tobacco (Nicotiana tabacum L.). Phyletic and phenetic analyses of the resulting 176 polymorphic bands recorded among the 22 species were performed. The phylogram was reconstructed using heuristic search and Wagner parsimony. The resulting most parsimonious consensus phylogram displayed three major phyletic lineages, consistent with the previously established three subgenera of Acacia. The 10 species of subgenus Acacia and the 6 species of subgenus Heterophyllum formed two monophyletic sister clades. The 5 species of subgenus Aculeiferum studied and Acacia albida (Syn. Faidherbia albida) grouped together and were basal to the clades of subgenera Acacia and Heterophyllum. The phylogram indicated that subgenus Heterophyllum diverged earlier from subgenus Aculeiferum than did subgenus Acacia; however, the phenogram indicated the reverse. The study indicated that A. nilotica and A. farnesiana are sister species, though A. nilotica is Afro-Asiatic and A. farnesiana is American. The phenogram separated the three subgenera in agreement with the phylogram, but the two dendrograms differed regarding the topologies of the species and the distance of evolution between subgenera Acacia and Heterophyllum.

    Notes: Abstract  The soil-borne fungi Verticillium spp. cause vascular wilt disease in a wide range of crop plants. In tomato, resistance to Verticillium dahliae race 1 is conferred by a single dominant gene, Ve. Previous efforts to map Ve in tomato have yielded confusing results, locating it on different chromosomes, which subsequently raised the possibility that Verticillium resistance may be controlled by a number of loci. We used three different mapping populations to obtain an unambiguous map location of Ve: a recombinant inbred (RI) line population; an F2 population segregating for Verticillium resistance; and a population of 50 introgression lines (IL). In all of the mapping populations Ve was positioned on the short arm of chromosome 9 tightly linked to the RFLP marker GP39. This linkage was confirmed by screening for GP39 in different breeding lines with known resistance or susceptibility to Verticillium. A perfect match was found between GP39 and the Verticillium response of the lines, indicating the potential of GP39 in the rapid detection of Verticillium resistance and as a starting point for map-based cloning of Ve. This approach is particularly relevant for Verticillium dahliae race 1, since in the present work we also show that the isolate that infects tomato is responsible for wilt disease in other important crop plants.

    Notes: Abstract  We have constructed a genetic linkage map within the cultivated gene pool of cowpea (2n=2x=22) from an F8 recombinant inbred population (94 individuals) derived from a cross between the inbreds IT84S-2049 and 524B. These breeding lines, developed in Nigeria and California, show contrasting reactions against several pests and diseases and differ in several morphological traits. Parental lines were screened with 332 random RAPD decamers, 74 RFLP probes (bean, cowpea and mung bean genomic DNA clones), and 17 AFLP primer combinations. RAPD primers were twice as efficient as AFLP primers and RFLP probes in detecting polymorphisms in this cross. The map consists of 181 loci, comprising 133 RAPDs, 19 RFLPs, 25 AFLPs, three morphological/classical markers, and a biochemical marker (dehydrin). These markers identified 12 linkage groups spanning 972 cM with an average distance of 6.4 cM between markers. Linkage groups ranged from 3 to 257 cM in length and included from 2 to 41 markers, respectively. A gene for earliness was mapped on linkage group 2. Seed weight showed a significant association with a RAPD marker on linkage group 5. This map should facilitate the identification of markers that “tag” genes for pest and disease resistance and other traits in the cultivated gene pool of cowpea.

    Notes: Abstract  We constructed a genetic map of a cross between the Swiss winter wheat (Triticum aestivum L.) variety Forno and the Swiss winter spelt (Triticum spelta L.) variety Oberkulmer. For the linkage analysis,176 polymorphic RFLP probes and nine microsatellites were tested on 204 F5 recombinant inbred lines (RILs) of Forno×Oberkulmer revealing 242 segregating marker loci. Thirty five percent of these loci showed significant (P〉0.05) deviation from a 1 : 1 segregation, and the percentage of Forno alleles ranged from 21% to 83% for individual marker loci. Linkage analysis was performed with the program MAPMAKER using the Haldane mapping function. Using a LOD threshold of 10, we obtained 37 linkage groups. After finding the best order of marker loci within linkage groups by multi-point analysis we assembled the linkage groups into 23 larger units by lowering the LOD threshold. All except one of the 23 new linkage groups could be assigned to physical chromosomes or chromosome arms according to hybridisation patterns of nulli-tetrasomic lines of Chinese Spring and published wheat maps. This resulted in a genetic map comprising 230 marker loci and spanning 2469 cM. Since the analysed population is segregating for a wide range of agronomically important traits, this genetic map is an ideal basis for the identification of quantitative trait loci (QTLs) for these traits.

    Notes: Abstract  An RFLP-based map consisting of 160 loci was constructed in an intervarietal cross of foxtail millet [Setaria italica (L.) P. Beauv.], Longgu 25×Pagoda Flower Green. The map comprises nine linkage groups, which were aligned with the nine foxtail millet chromosomes using trisomic lines, and spans 964 cM. The intraspecific map was compared to an interspecific map, constructed in a S. italica×S. viridis cross. Both the order of the markers and the genetic distances between the loci were highly conserved. Deviations from the expected 1 : 2 : 1 Mendelian segregation ratios were observed in both the intra- and inter-specific populations. The segregation data indicate that chromosome VIII in the Longgu 25×Pagoda Flower Green cross carries a gene that strongly affects gamete fertility.

    Notes: Abstract  Genetic and physical maps, consisting of a large number of DNA markers for Arabidopsis thaliana chromosomes, represent excellent tools to determine the organization of related genomes such as those of Brassica. In this paper we report the chromosomal localization and physical analysis by pulsed-field gel electrophoresis (PFGE) of a well-defined gene complex of A. thaliana in the Brassica nigra genome (B genome n=8). This complex is approximately 30 kb in length in A. thaliana and contains a cluster of six genes including ABI1 (ABA-responsive), RPS2 (resistance against Pseudomonas syringae, a bacterial disease), CK1 (casein kinase I), NAP (nucleosome-assembly protein), X9 and X14 (both of unknown function). The Arabidopsis chromosomal complex was found to be duplicated and conserved in gene number at different levels in the Brassica genome. Linkage group B1 had the most-conserved arrangement carrying all six genes tightly linked. Group B4 had an almost complete complex except for the absence of RPS2. Other partial complexes of fewer members were found on three other chromosomes. Our studies demonstrate that by this approach it is possible to identify ancestrally related chromosome segments in a complex and duplicated genome, such as the genome of B. nigra, permitting one to draw conclusions as to its origin and evolution.

    Notes: Abstract  We mapped and characterized quantitative trait loci (QTL) for resistance to Sporisorium reiliana. A population of 220 F3 families produced from the cross of two European elite inbreds (D32, D145) was evaluated with two replications at a French location with high natural incidence of S. reiliana and at a Chinese location employing artificial inoculation. The 220 F3 families were genotyped with 87 RFLP and seven SSR markers. Using composite interval mapping, we identified two different sets of 3 and 8 QTL for the French and the Chinese locations explaining 13% and 44% of respectively. Individual QTL explained up to 14% of σ^2 p. The 11 QTL mapped to eight maize chromosomes and displayed mostly additive or partial dominant gene action. Significant digenic epistatic interactions were detected for one pair of these QTL. Only a few QTL for S. reiliana were in common with QTL for resistance to Ustilago maydis and Puccinia sorghi, identified at a German location for the same population. Consequently, in our materials resistance to these three fungal pathogens of maize seems to be inherited independently.

    Notes: Abstract  The rym3 (formerly designated ym3) gene conferring resistance to barley yellow mosaic virus (BaYMV) is effective against all strains of the virus but up to now has not been mapped to any chromosome. We performed a linkage analysis, using DNA extracted from individually harvested mature leaves of 153 F2 plants derived from a cross between BaYMV-resistant cv ’Ishuku Shirazu’ carrying rym3 and susceptible cv ’Ko A’. Additionally, the F3 lines derived from F2 plants were grown in the BaYMV-infested field and examined for their reaction to BaYMV. Our results indicated that rym3 is located on the short arm of chromosome 5H and flanked by RFLP markers MWG28and ABG705A at distances of 7.2 and 11.7 cM, respectively. The chromosomal configuration estimated by DNA markers around rym3 and the utilization of these molecular markers for pyramiding with the BaYMV resistance genes in barley breeding programs are discussed.

    Notes: Abstract  DNA from pooled leaf samples of 11 true major mangrove, three true minor mangrove, two mangrove associate, two mangrove parasite, three terrestrial and one cultivated species were isolated for the present study. In total, 198 random amplified polymorphic DNAs (RAPDs) and 180 restriction fragment length polymorphism (RFLP) loci were scored by using ten primers and 14 enzyme-probe combinations respectively. The polymorphism observed for these markers revealed a high degree of genetic diversity in mangroves at both inter-specific or inter-generic levels. A dendrogram, constructed after pooling both RAPD and RFLP data, using a similarity index was analysed for genome relationships among these species. The dendrogram showed clustering of all the major mangroves, except for Nypa fruticans (Arecaceae), into one group. All species under the tribe Rhizophorae formed a sub-cluster, to which Xylocarpus granatum was found to be the most closesly related species. The clustering pattern implied that Excoecaria agallocha and Acanthus ilicifolius should be considered as true minor mangroves. The present study also provided molecular data favouring the separation of Avicennia spp. from the Verbenaceae to create a monotypic family the Avicenniaceae. The separation of Viscum orientale into the Viscaceae was also favoured.

    Notes: Abstract  A genetic linkage map of tetraploid wheat [Triticum turgidum (L.) Thell.] was constructed using segregation data from a population of 65 recombinant inbred lines (RILs) derived from a cross between the durum wheat cultivar Messapia and accession MG4343 of T. turgidum (L.) Thell. ssp dicoccoides (Korn.) Thell. A total of 259 loci were analysed, including 244 restriction fragment length polymorphisms (RFLPs), one PCR (polymerase chain reaction) marker (a sequence coding for a LMW (low-molecular-weight) glutenin subunit gene located at the Glu-B3 locus), seven biochemical (six seed-storage protein loci and one isozyme locus) and seven morphological markers. A total of 213 loci were mapped at a LOD≥3 on all 14 chromosomes of the A and B genomes. The total length of the map is 1352 cM and the average distance between adjacent markers is 6.3 cM. Forty six loci could not be mapped at a LOD≥3. A fraction (18.6%) of the markers deviated significantly from the expected Mendelian ratios; clusters of loci showing distorted segregation were found on chromosomes 1B, 3AL, 4AL, 6AL and 7AL. The durum wheat map was compared with the published maps of bread wheat using several common RFLP markers and general features are discussed. The markers detected the known structural rearrangements involving chromosomes 4A, 5A and 7B as well as the translocation between 2B-6B, but not the deletion on 2BS. This map provides a useful tool for analysing and breeding economically important quantitative traits and for marker-assisted selection, as well as for studies of genome organisation in small grain cereal species.

    Notes: Abstract  A genetic map of the long arm of chromosome 6R of rye was constructed using eight homoeologous group-6 RFLP clones and five PCR markers derived from the rye-specific dispersed repetitive DNA family, R173. The map was developed using a novel test-cross F1 (TC-F1) population segregating for resistance to the cereal cyst nematode. Comparisons were made between the map generated with other rye and wheat group-6 chromosome maps by the inclusion of RFLP clones previously mapped in those species. Co-linearity was observed for common loci. This comparison confirmed a dramatic reduction in recombination for chromosome 6R in the TC-F1 population. The CreR locus was included in the linkage map via progeny testing of informative TC-F1 individuals. CreR mapped 3.7 cM distal from the RFLP locus, XksuF37. Comparative mapping should allow the identification of additional RFLP markers more closely linked to the CreR locus.

The incidence of polymorphism was 36% for RFLP probes, 20% for RAPD primers, and 42% for AFLP primers. The overall incidence of disturbed segregation was 18%. Linkage relationships at a LOD score of 4.0 and θ=0.25 indicated 11 linkage groups. The total map length was 534.4 cM and the map was clearly unsaturated with markers spaced at 4.9 cM. AFLP P6B15 was 1.7 cM from P 1, AFLP P1B34 was 2.2 cM from Y 2, and AFLP P3B30XA was 8.1 cM from Rs.

    Notes: Abstract Sugar beet (Beta vulgaris L.) is highly susceptible to the beet cyst nematode (Heterodera schachtii Schm.). Three resistance genes originating from the wild beets B. procumbens (Hs1 pro-1) and B. webbiana (Hs1 web-1, Hs2 web-7) have been transferred to sugar beet via species hybridization. We describe the genetic localization of the nematode resistance genes in four different sugar beet lines using segregating F2 populations and RFLP markers from our current sugar beet linkage map. The mapping studies yielded a surprising result. Although the four parental lines carrying the wild beet translocations were not related to each other, the four genes mapped to the same locus in sugar beet independent of the original translocation event. Close linkage (0–4.6 cM) was found with marker loci at one end of linkage group IV. In two populations, RFLP loci showed segregation distortion due to gametic selection. For the first time, the non-randomness of the translocation process promoting gene transfer from the wild beet to the sugar beet is demonstrated. The data suggest that the resistance genes were incorporated into the sugar beet chromosomes by non-allelic homologous recombination. The finding that the different resistance genes are allelic will have major implications on future attempts to breed sugar beet combining the different resistance genes.

    Notes: Abstract Inheritance of resistance to rust was investigated in the self progeny of the sugarcane cultivar ‘R570’ also used to build a RFLP genetic map. Resistance was evaluated through both field and controlled greenhouse trials. A clear-cut 3 (resistant) ∶ 1 (susceptible) segregation indicative of a probable dominant resistant gene was observed. This is the first documented report of a monogenic inheritance for disease resistance in sugarcane. This gene was found linked at 10 cM with an RFLP marker revealed by probe CDSR29. Other minor factors involved in the resistance were also detected.

    Notes: Abstract An integrated genetic linkage map for E. nitens was constructed in an outbred three-generation pedigree. Analysis of 210 RFLP, 125 RAPD and 4 isozyme loci resulted in 330 markers linked in 12 linkage groups covering 1462 cM (n=11 in eucalypts). The 12th linkage group is comprised of only 5 markers and will probably coalesce with another linkage group when further linked loci are located. Co-dominant RFLP loci segregating in both parents were used to integrate linkages identified in the male and female parents. Differences in recombination frequencies in the two parents were observed for a number of pairs of loci, and duplication of sequences was identified both within and between linkage groups. The markers were distributed randomly across the genome except for the RFLPs in linkage group 10 and for some loci showing segregation distortion, which were clustered into three regions of the map. The use of a large number of co-dominant RFLP loci in this map enables it to be used in other pedigrees of E. nitens and forms a basis for the detection and location of QTL in E. nitens and other eucalypt species.

    Notes: Abstract A DNA isolation method suitable for genomic library construction and RFLP analyses of the forage legume Stylosanthes was developed. Probes isolated using this method were used to investigate the feasibility of constructing RFLP-based genetic maps in this genus. Two hundred and seventy-one PstI genomic DNA and 134 cDNA clones were analysed against four Stylosanthes accessions, including two tetraploids and two diploids, with the use of two restriction enzymes, DraI and HindIII. The proportion of clones which detected single-copy sequences from the PstI genomic library was higher than that from the cDNA library, but the percentage of clones which detected low-copy sequences was doubled in the latter. There was no significant difference in the level of RFLPs detected by gDNA and cDNA probes, although the level of polymorphism was lower in the diploids. A large proportion of RFLPs seemed to have resulted from mutation/base substitution events, and this was especially the case in diploids.

    Notes: Abstract   Improving seedling vigor is an important objective of modern rice (Oryza sativa L.) breeding programs. The purpose of this study was to identify and map quantitative trait loci (QTL) underlying seedling vigor-related traits using restriction fragment length polymorphisms (RFLPs). An F2 population of 204 plants was developed from a cross between a low-vigor japonica cultivar `Labelle' (LBL) and a high-vigor indica cultivar `Black Gora' (BG). A linkage map was constructed of 117 markers spanning 1496 Haldane cM and encompassing the 12 rice chromosomes with an average marker spacing of 14 cM. The length of the shoots, roots, coleoptile and mesocotyl were measured on F3 families in slantboard tests conducted at two temperatures (18° and 25°C). By means of interval analysis, 13 QTLs, each accounting for 7% to 38% of the phenotypic variance, were identified and mapped in the two temperature regimes at a log-likelihood (LOD) threshold of 2.5. Four QTLs controlled shoot length, 2 each controlled root and coleoptile lengths and 5 influenced mesocotyl length. Single-point analysis confirmed the presence of these QTLs and detected additional loci for shoot, root and coleoptile lengths, these latter usually accounting for less than 5% of the phenotypic variation. Only 3 QTLs detected both by interval and single-point analyses were expressed under both temperature regimes. Additive, dominant and overdominant modes of gene action were observed. Contrary to what was predicted from parental phenotype, the low-vigor LBL contributed 46% of the positive alleles for shoot, root and coleoptile lengths. Positive alleles from the high-vigor parent BG were identified for increased root, coleoptile and mesocotyl lengths. However, BG contributed alleles with only minor effects for shoot length, the most important determinant of seedling vigor in water-seeded rice, suggesting that it would not be an ideal donor parent for introducing faster shoot growth alleles into temperate japonica cultivars.

    Notes: Abstract   In saline environments, bread wheat, Triticum aestivum L. (genomes AABBDD), accumulates less Na+ and more K+ in expanding and young leaves than durum wheat, T. turgidum L. (genomes AABB). Higher K+/Na+ ratios in leaves of bread wheat correlate with its higher salt tolerance. Chromosome 4D from bread wheat was shown in previous work to play an important role in the control of this trait and was recombined with chromosome 4B in the absence of the Ph1 locus. A population of plants disomic for 4D/4B recombined chromosomes in the genetic background of T. turgidum was developed to investigate the genetic control of K+/Na+ discrimination by chromosome 4D. Evidence was obtained that the trait is controlled by a single locus, designated Kna1, in the long arm of chromosome 4D. In the present work, K+/Na+ discrimination was determined for additional families with 4D/4B chromosomes. The concentrations of Na+ and K+/Na+ ratios in the youngest leaf blades clustered in two nonoverlapping classes, and all recombinant families could be unequivocally assigned to Kna1 and kna1 classes. The Kna1 locus scored this way was mapped on a short region in the 4DL arm and was completely linked to Xwg199, Xabc305, Xbcd402, Xpsr567, and Xpsr375; it was also mapped as a quantitative trait. The results of the QTL analysis, based on the K+/Na+ ratios in the young leaves of greenhouse-grown plants and flag leaves of field-grown plants, agreed with the position of Kna1 determined as a qualitative trait. Several aspects of gene introgression by manipulation of the Ph1 locus are discussed.

    Notes: Abstract A rare phenomenon of the occurrence of novel non-parental chloroplast DNA (cpDNA) variants in natural sexual interspecific hybrids between Populus deltoides var deltoides and P. nigra, P. x canadensis is described. Restriction fragment variation of cpDNA in 17 P. x canadensis cultivars was examined and compared with that of representative samples of P. deltoides and P. nigra using 83 combinations of 16 restriction enzymes and six Petunia hybrida cpDNA probes. Twelve cultivars had one to five novel non-parental cpDNA fragments in the chloroplast genome region homologous to the 9.0-kb PstI cpDNA fragment of Petunia from the large single-copy region.

    Notes: Abstract A genetic linkage map for radiata pine (Pinus radiata D. Don) has been constructed using segregation data from a three-generation outbred pedigree. A total of 208 loci were analyzed including 165 restriction fragment length polymorphism (RFLP), 41 random amplified polymorphic DNA (RAPD) and 2 microsatellite markers. The markers were assembled into 22 linkage groups of 2 or more loci and covered a total distance of 1382 cM. Thirteen loci were unlinked to any other marker. Of the RFLP loci that were mapped, 93 were detected by loblolly pine (P. taeda L.) cDNA probes that had been previously mapped or evaluated in that species. The remaining 72 RFLP loci were detected by radiata pine probes from a PstI genomic DNA library. Two hundred and eighty RAPD primers were evaluated, and 41 loci which were segregating in a 1∶1 ratio were mapped. Two microsatellite markers were also placed on the map. This map and the markers derived from it will have wide applicability to genetic studies in P. radiata and other pine species.

    Notes: Abstract The genomic clone RG64, which is tightly linked to the blast resistance gene Pi-2(t) in rice, provides means to perform marker-aided selection in a rice breeding program. The objective of this study was to investigate the possibility of generating a polymerase chain reaction (PCR)-based polymorphic marker that can distinguish the blast resistant gene, Pi-2(t), and susceptible genotypes within cultivated rice. RG64 was sequenced, and the sequence data was used to design pairs of specific primers for (PCR) amplification of genomic DNA from rice varieties differing in their blast disease responsiveness. The amplified products, known as sequenced-tagged-sites (STSs), were not polymorphic between the three varieties examined. However, cleavage of the amplified products with the restriction enzyme HaeIII generated a polymorphic fragment, known as specific amplicon polymorphism (SAP), between the resistant and the susceptible genotypes. To examine the power of the identified SAP marker in predicting the genotype of the Pi-2 (t) locus, we determined the genotypes of the F2 individuals at this locus by performing progeny testing for the disease response in the F3 generation. The results indicated an accuracy of more than 95% in identifying the resistant plants, which was similar to that using RG64 as the hybridization probe. The identification of the resistant homozygous plants increased to 100% when the markers flanking the genes were considered simultaneously. These results demonstrate the utility of SAP markers as simple and yet reliable landmarks for use in marker-assisted selection and breeding within cultivated rice.

    Notes: Abstract Mitochondrial variability was investigated in natural populations of wild carrot (Daucus carota ssp carota) in different regions: South of France, Greece, and various sites in the Mediterranean Basin and Asia. Total DNA was digested with two restriction endonucleases (EcoRV and HindIII) and probed with three mitochondrial DMA-specific genes (coxI, atp6, and coxII). Twenty-five different mitochondrial types were found in 80 analyzed individuals. Thirteen mitotypes were found among the 7 French populations studied. On average, 4.4 different mitotypes were observed per population, and these mitotypes were well-distributed among the populations. All of the mitochondrial types were specific to a single region. However, the proportion of shared restriction fragments between 2 mitotypes from different regions was not particularly lower than that which occurred among mitotypes from a single region. On the basis of the sexual phenotype [male-sterile (MS) or hermaphrodite] of the plants studied in situ and that of their progeny, 2 mitotypes were found to be highly associated with male sterility. Eighty percent of the plants bearing these mitotypes were MS in situ, and all of these plants produced more than 30% MS plants in their progeny. This association with male sterility was consistent in several populations, suggesting an association with a cytoplasmic male-sterility system. Moreover, these two mitotypes had very similar mitochondrial DNA restriction patterns and were well-differentiated from the other mitotypes observed in wild plants and also from those observed in the two CMS types already known in the cultivated carrot. This suggests that they correspond to a third cytoplasmic sterility.

    Notes: Abstract Drought is an important climatic phenomenon which, after soil infertility, ranks as the second most severe limitation to maize production in developing countries. When drought stress occurs just before or during the flowering period, a delay in silking is observed, resulting in an increase in the length of the anthesis-silking interval (ASI) and in a decrease in grain yield. Selection for reduced ASI in tropical open-pollinated varieties has been shown to be correlated with improved yields under drought stress. Since efficient selection for drought tolerance requires carefully managed experimental conditions, molecular markers were used to identify the genomic segments responsible for the expression of ASI, with the final aim of developing marker-assisted selection (MAS) strategies. An F2population of 234 individuals was genotyped at 142 loci and F3 families were evaluated in the field under several water regimes for male flowering (MFLW), male sterility (STER), female flowering (FFLW) and ASI. The genetic variance of ASI increased as a function of the stress intensity, and the broad-sense heritabilites of MFLW, FFLW and ASI were high under stress conditions, being 86%, 82% and 78%, respectively. Putative quantitative trait loci (QTLs) involved in the expression of MFLW and/or FFLW under drought were detected on chromosomes 1, 2, 4, 5, 8, 9 and 10, accounting for around 48% of the phenotypic variance for both traits. For ASI, six putative QTLs were identified under drought on chromosomes 1, 2, 5, 6, 8 and 10, and together accounted for approximately 47% of the phenotypic variance. Under water stress conditions, four QTLs were common for the expression of MFLW and FFLW, one for the expression of ASI and MFLW, and four for the expression of ASI and FFLW. The number of common QTLs for two traits was related to the level of linear correlation between these two traits. Segregation for ASI was found to be transgressive with the drought-susceptible parent contributing alleles for reduced ASI (4 days) at two QTL positions. Alleles contributed by the resistant line at the other four QTLs were responsible for a 7-day reduction of ASI. These four QTLs represented around 9% of the linkage map, and were stable over years and stress levels. It is argued that MAS based on ASI QTLs should be a powerful tool for improving drought tolerance of tropical maize inbred lines.

    Notes: Abstract  An population was established for mapping the two dominant genes for dwarfness (Ddw1) and hairy peduncle (Hp) on chromosome 5R. The location of both genes was shown to be on the segment of chromosome 5RL which was ancestrally translocated and is homoeologous to Triticeae 4L. Hp co-segregated with the wheat gDNA probe WG199, localised in wheat on chromosomes 5AL, 4BL and 4DL. No segregation was observed between the traits hairy peduncle and hairy leaf sheath. The locus for Ddw1 was found to map distally to Hp/Xwg199 but proximal to the isozyme marker β-amy-R1. The genetical distances were 5.6 cM between Hp/Xwg199 and Ddw1 and 11.5 cM between Ddw1 and β-amy-R1, respectively. The map position of Ddw1 suggests that it is homoeologous to the wheat dominant dwarfing gene Rht12, present on chromosome 5AL and linked to β-amy-A1.

    Notes: Abstract Studies on the nature of restriction fragment length polymorphisms (RFLPs) were undertaken to characterize the Citrus genome. This type of analysis has not been carried out with any other perennial crop. Citrus reticulata Blanco cv Clementine, C. xparadisi Macf. cv Duncan, and an F1 hybrid (LB 1–21) were used to determine what probe/enzyme combinations revealed polymorphisms in Southern analysis, and a backcross family (LB 1–21x‘Clementine’) of 65 randomly selected hybrid seedlings was used for some analyses. A majority (73%) of the clones examined from a PstI genomic library appeared to detect single-copy sequences based on RFLP banding patterns, while clones from a cDNA library revealed a lower percentage of single copy sequences. When hybridization stringencies were lowered, 21% of the genomic clones examined revealed greater copy numbers. PstI digestion of ‘Duncan’ DNA indicated abundant methylation, so the relatively high frequency of multiple-copy sequences observed at moderate stringency cannot be attributed to a lack of methylation of the Citrus DNA. The polymorphisms in banding patterns observed primarily resulted from insertions and/or deletions rather than from base substitutions, and a model is presented to account for the varying patterns obtained from individual probes with different restriction enzymes. Finally, a model for transposon activity in Citrus is proposed, based on observations made during the course of these studies.

    Notes: Abstract   Genetic maps of the homoeologous group-6 chromosomes of bread wheat, Triticum aestivum, have been constructed spanning 103 cM on 6A, 90 cM on 6B and 124 cM on 6D. These maps were transferred to a Chinese Spring (CS)×line #31 cross to locate a dominant powdery mildew resistance gene, Pm12, introgressed into line #31 from Aegilops speltoides. Pm12 was shown to lie on the short arm of translocation chromosome 6BS-6SS.6SL in line #31, but could not be mapped more precisely due to the lack of recombination between the 6S Ae. speltoides segment and chromosome 6B. Possible strategies to reduce the size of the alien segment, which probably encompasses the complete long arm and more than 82% of the short arm of chromosome 6B, are discussed.

    Notes: Abstract Morphology, the extent of elimination of donor chromosomes and the organelle composition of highly asymmetric somatic hybrid plants between a interspecific tomato hybrid Lycopersicon esculentum x L. pennellii (EP) as donor and a Solarium melongena, eggplant (E), recipient, were studied. Morphologically, the somatic hybrids most resemble eggplant but, due to polyploidy, growth is slower relative to both fusion parents. The somatic hybrids produce flowers that are characterized by abnormal styles, stigmas and by anthers which do not produce pollen. Limited amounts of donor EP genomic DNA were found in the three somatic hybrid plants (H18-1, H18-2 and H18-3), by dot-blot hybridization with probe pTHG2, equivalent to 6.23,5.41, and 5.95% EP, respectively. These percentages translated to the presence of 3.59, 2.90 and 3.19 average-size EP chromosomes in plants H1 8-1,-2 and-3, respectively. RFLP determination of L. esculentum- and L. pennellii-specific chromosomes revealed that only fragments of eight to ten out of the 24 EP chromosomes (EP has 12 L. esculentum and 12 L. pennellii chromosomes) are present in the asymmetric somatic hybrid plants. Loci of L. esculentum and L. pennellii were evenly represented in plants H18-1, -2, and -3: four to five from L. esculentum and four to five from L. pennellii. All somatic hybrid plants retained locus TG22, chromosome 4, from both EP species. Although the regeneration of plants, H18-1, -2 and-3 was from one callus, loci TG31 and TG79 of L. esculentum chromosome 2 and L. pennellii chromosome 9, respectively, were missing in hybrid plant H18-1. The three somatic hybrid plants all had chloroplast DNA fragments specific for S. melongena. The mitochondrial genome (mtDNA) in the asymmetric somatic hybrids showed predominantly the pattern of eggplant; however, some eggplant-specific polymorphic bands were not present in the three plants.

Linkage maps for these crosses consisted of 199 and 235 markers, respectively. QTLs for seed weight in the ‘Primo’ x ‘OSU442-15’ cross were identified by interval mapping, bulked segregant analysis, and selective genotyping. Four QTLs were identified in this cross, demonstrating linkage to four intervals on three linkage groups. QTLs for seed weight in the ‘JI1794’ x ‘Slow’ cross were identified by single-marker analyses. Linkage were demonstrated to four intervals on three linkage groups plus three unlinked loci. In the two crosses, only one common genomic region was identified as containing seed-weight QTLs. Seed-weight QTLs mapped to the same region of linkage group III in both crosses. Conserved linkage relationships were demonstrated for pea, mungbean (Vigna radiata L.), and cowpea (V. unguiculata L.) genomic regions containing seed-weight QTLs by mapping RFLP loci from the Vigna maps in the ‘Primo’ x ‘OSU442-15’ and ‘JI1794’ x ‘Slow’ crosses.

    Notes: Abstract Cross-compatibility behaviour of doubled haploid (DH) and hybrid genotypes of Coffea camphora was established using both phenotypic bioassay and in situ seed-set examination. The availability of DHs provided the opportunity of working with genetically homogenous pollen and female parents. The aniline blue fluorescence (ABF) method was applied to detect callose accumulation in pollen and pistil. Clear cross-compatibility/incompatibility situations were observed and confirmed by in situ seed-set analysis. Cross-compatibility analysis of hybrid combinations involving different DHs corroborated the crossing behaviour observed at the DH level. Expression of the self-incompatibility system did not appear to be affected by the low vigour of the DH. The crossing-behaviour distribution observed within DHs derived from clone IF200 confirmed that self-incompatibility in C. canephora is a gametophytic self-incompatibility system controlled by a single locus (S-locus). Reduced seed-set developments following incompatible crosses may indicate the occurrence of pseudo-incompatibility. Molecular marker linkage analysis showed that the S-locus is associated with an RFLP marker on linkage group 9. The availability of a linked DNA marker should facilitate the genetic analysis of self-incompatibility in relation to coffee breeding programmes.

    Notes: Abstract Molecular markers provide the opportunity to identify marker-quantitative trait locus (QTL) associations in different environments and populations. Two soybean [Glycine max (L.) Merr.] populations, ‘Young’ x PI 416 937 and PI 97100 x ‘Coker 237’, were evaluated with restriction fragment length polymorphism (RFLP) markers to identify additional QTLs related to seed protein and oil. For the Young x PI 416937 population, 120 F4-derived lines were secored for segregation at 155 RFLP loci. The F4-derived lines and two parents were grown at Plains, G.a., and Windblow and Plymouth, N.C. in 1994, and evaluated for seed protein and oil. For the PI 97100 x Coker 237 population, 111 F2-derived lines were evaluated for segregation at 153 RFLP loci. Phenotypic data for seed protein and oil were obtained in two different locations (Athens, G.a., and Blackville, S.C.) in 1994. Based on single-factor analysis of variance (ANOVA) for the Young x PI 416937 population, five of seven independent markers associated with seed protein, and all four independent markers associated with seed oil in the combined analysis over locations were detected at all three locations. For the PI 97 100 x Coker 237 population, both single-factor ANOVA and interval mapping were used to detect QTLs. Using single-factor ANOVA, three of four independent markers for seed protein and two of three independent markers for seed oil were detected at both locations. In both populations, singlefactor ANOVA, revealed the consistency of QTLs across locations, which might be due to the high heritability and the relatively few QTLs with large effects conditioning these traits. However, interval mapping of the PI 97100 x Coker 237 population indicated that QTLs identified at Athens for seed protein and oil were different from those at Blackville. This might result from the power of QTL mapping being dependent on the level of saturation of the genetic map. Increased seed protein was associated with decreased seed oil in the PI 97100 x Coker 237 population (r = −0.61). There were various common markers (P⩽0.05) on linkage groups (LG) E, G,H,K, and UNK2 identified for both seed protein and oil. One QTL on LG E was associated with seed protein in both populations. The other QTLs for protein and oil were population specific.

    Notes: Abstract  Self-incompatibility (SI) in Brassica is a sporophytic system, genetically determined by alleles at the S-locus, which prevents self-fertilization and encourages outbreeding. This system occurs naturally in diploid Brassica species but is introduced into amphidiploid Brassica species by interspecific breeding, so that in both cases there is a potential for yield increase due to heterosis and the combination of desirable characteristics from both parental lines. Using a polymerase chain reaction (PCR) based analysis specific for the alleles of the SLG (S-locus glycoprotein gene) located on the S-locus, we genetically mapped the S-locus of B. oleracea for SI using a F2 population from a cross between a rapid-cycling B. oleracea line (CrGC-85) and a cabbage line (86-16-5). The linkage map contained both RFLP (restriction fragment length polymorphism) and RAPD (random amplified polymorphic DNA) markers. Similarly, the S-loci were mapped in B. napus using two different crosses (91-SN-5263×87-DHS-002; 90-DHW-1855-4×87-DHS-002) where the common male parent was self-compatible, while the S-alleles introgressed in the two different SI female parents had not been characterized. The linkage group with the S-locus in B. oleracea showed remarkable homology to the corresponding linkage group in B. napus except that in the latter there was an additional locus present, which might have been introgressed from B. rapa. The S-allele in the rapid-cycling Brassica was identified as the S29 allele, the S-allele of the cabbage was the S 5 allele. These same alleles were present in our two B. napus SI lines, but there was evidence that it might not be the active or major SI allele that caused self-incompatibility in these two B. napus crosses.

    Notes: Abstract  Three mutant loci of rye determining absence of ligules (al), waxless plant (wa1) and waxy endosperm (Wx) characters were mapped in a single F2 population, comprising 84 individual plants. The three loci could be clearly tagged in relation to 7 (al on chromosome 2R), 4 (wa1 on chromosome 7R) or 6 (Wx on chromosome 4R) RFLP markers. The mapping data are compared with existing data for homoeologous regions containing equivalent mutants of wheat, barley, rice and maize. It is shown that the loci analysed are highly conserved across the cereal species, including rye.

    Notes: Abstract  In soybean [Glycine max (L.) Merr.] heterosis has been reported for seed yield. Molecular markers may be useful to select diverse parents for the expression of heterosis and yield improvement. The objective of this study was to determine if molecular markers could be used to predict yield heterosis in soybean. From each Maturity Group (MG) II and III, 21 genotypes were selected on the basis of high yield (HY), different geographic origin (GO), and isozyme loci (ISO) and for diversity in restriction fragment length polymorphisms (RFLP), and crosses were made within MGs and selection criteria groups to obtain 6 F1 hybrids per group. The 21 parents and the 24 F1 hybrids of each MG were evaluated for yield in replicated tests at two locations in 2 years, and midparent heterosis (MPH) and high-parent heterosis (HPH) estimates were calculated. On the basis of hybrid performance during the first year, 12 parents (3 per selection criteria group) were chosen in each MG to conduct a second RFLP analysis using 129 probes. Genetic distances (GDM) for pairs of the 12 genotypes were calculated with this RFLP information and correlated with MPH and HPH estimates. Significant MPH averages for seed yield were observed in the combined analysis of variance in each of the four selection criteria groups of MG II, and in the HY, ISO, and GO of MG III. Significant HPH averages were observed only in the ISO and GO groups of MG II. The greatest frequency of F1 hybrids with significant MPH was observed in the ISO and GO groups of both MGs. For HPH, the greatest frequency was observed in the ISO group of both MGs. In both MGs, the ISO group had the largest absolute MPH value; the RFLP group had generally the smallest. The observations indicated that the expression of heterosis in seed yield might be associated with diversity in the isozyme loci present in the parents. For the genotypes included in the second RFLP analysis, correlations of GDMs with MPH and HPH values on an entry-mean basis were low and not significant, indicating that heterosis in yield may not be associated with genetic diversity at the molecular level as determined by RFLPs. The results suggest that in soybean, parent selection on the basis of RFLPs and isozyme loci to exploit heterosis in seed yield may not be feasible. There was no association between genetic distance estimated by the RFLP analysis and seed yield heterosis, and in spite of the observed relationship between isozyme loci and heterosis for yield, the practicality of using the isozyme markers to select parents may be limited because of the reduced number of assayable isozyme loci in soybean.

    Notes: Abstract Targeted homoeologous recombination mediated by the absence of the Ph1 locus is currently the most efficient technique by which foreign genes can be introgressed into polyploid wheat species. Because intra-arm homoeologous double cross-overs are rare, introgressed foreign genes are usually on terminal foreign chromosome segments. Since the minimum length of such a segment is determined by the position of a gene in the chromosome, large chromosome segments with undesirable genetic effects are often introgressed. Introgression of foreign genes on short interstitial segments based on two cycles of homoeologous recombination is described here. The utility of the technique is demonstrated by the introgression of the Kna1 locus, which controls K+/Na+ selectivity in T. aestivum L., on short interstitial segments of chromosome 4D into chromosome 4B of Triticum turgidum L. The level of recombination in a homoeologous segment is not significantly affected by a juxtaposed proximal homologous segment in the absence of the Ph1 locus.

For Pop1, the phenotypic data were collected from Plains, GA., Windblow, N.C., and Plymouth, N.C., in 1994. For Pop2, data were collected from Athens, GA., in 1994 and 1995, and Blackville, S.C., in 1995. Based on single-factor analysis of variance (ANOVA), seven and nine independent loci were associated with SW in Pop1 and Pop2, respectively. Together the loci explained 73% of the variability in SW in Pop1 and 74% in Pop2. Transgressive segregation occurred among the progeny in both populations. The marker loci associated with SW were highly consistent across environments and years. Two QTLs on linkage group (LG) F and K were located at similar genomic regions in both populations. The high consistency of QTLs across environments indicates that effective marker-assisted selection is feasible for soybean SW.

    Notes: Abstract A genetic map of six chromosomes of rye, (all of the rye chromosomes except for 2R), was constructed using 77 RFLP and 12 RAPD markers. The map was developed using an F2 population of 54 plants from a cross between two inbred lines. A rye genomic library was constructed as a source of clones for RFLP mapping. Comparisons were made between the rye map and other rye and wheat maps by including additional probes previously mapped in those species. These comparisons allowed (1) chromosome arm orientation to the linkage groups to be given, (2) the corroboration of several evolutionary translocations between rye chromosomes and homoeologous chromosomes of wheat; (3) an increase in the number of available markers for target regions of rye that show colinearity with wheat. Inconsistencies in the location of markers between the wheat and rye maps were mostly detected by multi-copy probes.

    Notes: Abstract  The Yr15 gene of wheat confers resistance to the stripe rust pathogen Puccinia striiformis West., which is one of the most devastating diseases of wheat throughout the world. In the present study, molecular markers flanking the Yr15 gene of wheat have been identified using the near-isogenic-lines approach. RFLP screening of 76 probe-enzyme combinations revealed one polymorphic marker (Nor/TaqI) between the susceptible and the resistant lines. In addition, out of 340 RAPD primers tested, six produced polymorphic RAPD bands between the susceptible and the resistant lines. The genetic linkage of the polymorphic markers was tested on segregating F2 population (123 plants) derived from crosses between stripe rust-susceptible Triticum durum wheat, cv D447, and a BC3F9 resistant line carrying Yr15 in a D447 background. A 2.8-kb fragment produced by the Nor RFLP probe and a 1420-bp PCR product generated by the RAPD primer OPB13 showed linkage, in coupling, with the Yr15 gene. Employing the standard maximum-likelihood technique it was found that the order OPB13 1420 –Yr15–Nor1 on chromosome 1B appeared to be no less than 1000-times more probable than the closest alternative. The map distances between OPB13 1420 –Yr15–Nor1 are 27.1 cM and 11.0 cM for the first and second intervals, respectively. The application of marker-assisted selection for the breeding of new wheat cultivars with the stripe rust resistance gene is discussed.

    Notes: Abstract Targeted homoeologous recombination mediated by the absence of the Ph1 locus is currently the most efficient technique by which foreign genes can be introgressed into polyploid wheat species. Because intra-arm homoeologous double cross-overs are rare, introgressed foreign genes are usually on terminal foreign chromosome segments. Since the minimum length of such a segment is determined by the position of a gene in the chromosome, large chromosome segments with undesirable genetic effects are often introgressed. Introgression of foreign genes on short interstitial segments based on two cycles of homoeologous recombination is described here. The utility of the technique is demonstrated by the introgression of the Kna1 locus, which controls K+/Na+ selectivity in T. aesivum L., on short interstitial segments of chromosome 4D into chromosome 4B of Triticum turgidum L. The level of recombination in a homoeologous segment is not significantly affected by a juxtaposed proximal homologous segment in the absence of the Ph1 locus.

    Notes: Abstract Seed weight (SW) is a component of soybean, Glycine max (L.) Merr., seed yield, as well as an important trait for food-type soybeans. Two soybean populations, 120 F4-derived lines of `Young'×PI416937 (Pop1) and 111 F2-derived lines of PI97100×`Coker 237' (Pop2), were mapped with RFLP makers to identify quantitative trait loci (QTLs) conditioning SW across environments and populations.     Notes: Abstract  The identification of diagnostic cytoplasmic molecular markers is of prime interest to pearl millet breeders wishing to identify sources of cytoplasmic-nuclear male sterility (CMS) which can be used as an alternative to the single source currently used in the production of F1 hybrid seed. Here, we report the classification of five pearl millet CMS sources based on RFLP analysis of isonuclear lines carried out using mitochondrial gene-specific DNA probes in combination with eight restriction endonucleases. On the basis of RFLP data, the five CMS cytoplasms can be distinguished from each other and from the isonuclear fertile cytoplasm. In addition, based on cox1, cox3, atp6 and atp9 polymorphisms, these lines can be classified into two major groups: one corresponds to A5, Aegp, Av and A1 cytoplasms, and the other consists of the A4 cytoplasm. Our results suggest that a rearrangement involving the cox1 gene might be related to CMS in the first group (A5, Aegp, Av and A1), whereas a rearrangement within the atp6/cox3 cluster region might be related to CMS in the second group (A4).

    Notes: Abstract  Genetic diversity in a set of 11 red and 11 white wheat lines from the Eastern U.S. soft wheat germplasm pool was measured using restriction fragment length polymorphism (RFLP) assay and coefficients of parentage (COP) analysis. On average, 78% of all bands revealed by three enzymes with 48 RFLP clones were monomorphic. Average pairwise genetic similarity (GS) was 0.97 when data from all enzymes were pooled. Probe Polymorphic Information Content (PIC) indexes ranged from 0 to 0.73 with a mean of 0.2. Fewer than 55% of the probes revealed any polymorphism. The frequency of polymorphism in the Eastern U.S. soft white winter (SWW) wheat gene pool was much lower than that observed in the Eastern U.S. soft red winter (SRW) wheat gene pool. SWW lines formed a single group on a dendrogram based on cluster analysis of RFLP-derived GS estimates, while SRW lines did not form a single group. COP values for all pairs of the Eastern U.S. soft wheat lines ranged from 0.02 to 0.9 with a mean of 0.21. SWW wheat lines traced to 53 ancestral lines and had an average COP of 0.51. The SRW wheat gene pool had more complex parentages (mean COP=0.15 and a total of 65 ancestral lines). COPs were correlated with RFLP-based GS for all line pairs (r=0.73, P〈0.01). However, correlations between the two similarity measures were substantially lower when the SRW and SWW wheat gene pools were considered individually (r values of 0.23 and 0.28, respectively). The actual GS among unrelated lines in the U.S. Eastern soft wheat gene pool appears to be higher than that observed for unrelated landraces from Southwest Asia (0.96 vs. 0.905), suggesting that the ancestral landrace parents of this gene pool were themselves drawn from a base population where inbreeding, i.e., F, was greater than zero.

    Notes: Abstract  We have generated 66 sequence-tagged-site (STS) markers from cDNA clones of Cryptomeria japonica, and 60% of them have already been mapped into C. japonica linkage groups. All of the STS markers showed a single fragment following polymerase chain reaction (PCR) amplification. We investigated by polymorphism of these STS markers in a mapped F2 population and 15 plus trees by means of a restriction endonuclease analysis. Polymorphism levels were 10.6% and 22.7% in the F2 population and the 15 plus trees, respectively. PCR amplification levels of the 66 STS markers in 14 conifer species varied depending on their genetic relationship with C. japonica. Taxodium, which is closely related to C. japonica, had the most amplifications (31.82%), followed by Sequoiadendron giganteum, which is of the same family. The average proportion of PCR amplifications in each family gradually declined in the following order: from Taxodiaceae to Cuppresaceae, Sciadopityaceae, Pinaceae, and Taxaceae. These results are in general agreement with a molecular phylogenetic relationship based on chloroplast DNA. The 66 STS markers will be useful as on anchor point for genome mapping and population genetics, and some of them will also be useful when studying other conifers.
