    Notes: Seed storage protein content of durum wheat (Triticum turgidum var. durum) has an important effect on nutritional value and pasta-making characteristics. The objective of this study was to determine by association with genetic markers the number, chromosomal location, and magnitude of effect of quantitative trait loci (QTLs) controlling protein concentration in kernels. A set of 65 recombinant inbred lines (RIs) was developed by single seed descent from a cross between cultivated durum wheat cv. ‘Messapia’ (low protein content) and accession MG4343 of the wild tetraploid wheat var. dicoccoides (high protein content). This population was characterized for eight morphological, six storage protein, one isozyme and 124 RFLP loci. Field trials were conducted in one location in 1993 and two locations in 1994. QTLs were mapped by regression analysis on each marker locus for each location and for the average across environments. A total of six putative QTLs were located on chromosome arms 4BS, SAL, 6AS, 6BS and 7BS. The number and size of QTLs detected varied across environments. The marker with the highest r2 value per QTL in each environment and across environments was chosen for a multiple linear regression analysis, which explained 49.2- 56.4% of the phenotypic variation for protein content. Only some of the markers were found to be negatively associated with plant grain yield and/or seed weight in one or two of the environments.

    Notes: The recessive dwarfing alleles of rye (Secale cereale L.), ct1 and ct2, caused a 35–55% reduction in the length of leaf 2 compared with corresponding tall lines grown at both 10°C and 20°C. The dwarf lines were 45–50% as responsive to applied GA3 as the tall lines at 20°C but the absolute GA-responsiveness of the dwarfs was greater at 10°C than at 20°C. There was no significant difference in the contents of GA19, GA20, GA29, GA1, GA3 and GA8 in the leaf extension zone of tall and dwarf seedlings grown at 20°C. It was concluded that the mechanism whereby GA homeostasis is maintained is functional in both tall and dwarf lines despite marked differences in leaf extension rate. The recessive rye mutations may cause loss of function late in the GA-cell elongation pathway or, alternatively, indirectly affect GA-responsiveness in vegetative tissues. The genetic and physiological evidence indicates that ct1 and ct2 are unrelated to the GA-insensitive Rht genes in hexaploid bread wheat.

    Notes: Abstract Wheat telomere-associated sequences (TASs) were cloned using a Vectorette approach and sequenced. Reverse primers specific to the TASs were combined with labelled degenerate telomere primers in PCR reactions containing total genomic DNA as template. Amplification products were separated on sequencing gels. In total, seventeen primer combinations provided 47 polymorphic fragments. Nine of these mapped beyond the most distal RFLP markers and defined the ends of seven chromosome arms. Seven of the nine terminal fragments were derived from a 118-bp tandem repeat, indicating that subtelomeric tandem repeat sequences provide an efficient means to target chromosome ends. A telomere cloning strategy and the terminal and interstitial location of TASs are discussed.

    Notes: Abstract Map-based DNA fingerprinting with AFLP markers provides a fast method for scanning the rice genome. Three hundred AFLP markers identified with ten primer combinations were mapped in two rice populations. The genetic maps were aligned and almost full coverage of the rice genome was obtained. The transferability of AFLP markers between indica × japonica and indica × indica crosses was tested. The chromosomes were divided into DNA Fingerprint Linkage Blocks (DFLBs) defined by specific AFLP markers. Using these blocks, the degree of similarity or divergence within specific chromosome regions was calculated for nine varieties. Applications of map-based fingerprinting for biodiversity studies and maker-assisted selection are discussed.

    Notes: Summary Analysis of grain esterase isozymes in Chinese Spring aneuploid genotypes by IEF confirmed that genes on the long arms of chromosomes 3A, 3B and 3D (Est-5) control the production of 19 isozymes. Allelic variants have been found for the isozyme pattern controlled by each chromosome. Segregational data involving null alleles and complex phenotypic differences indicate that the wheat grain esterases are encoded by three compound and probably homoeoallelic loci, each capable of producing at least six different isozymes.     Notes: Summary The effects of the Tom Thumb dwarfing gene, Rht3, on the quality and quantity of grain α-amylase produced during germination and by induction with exogenous gibberellic acid are described. In a season conducive to high sprouting damage the gene reduced α-amylase levels in the field by 77%. Selection among random Rht3 genotypes showed that other genetic factors can be combined with the dwarfing gene to further increase sprouting damage resistance.

    Notes: Summary An analysis of the α-amylase isozymes in GA-treated endosperm of wheat nullisomic-tetrasomics shows that there is more variation at the α-Amy-1 and α-Amy-2 homoeoallelic loci than was previously thought. Among the 16 isozymes produced by genes on the group 7 chromosomes, most could be definitely established as products of a single homoeoallele. Inter-varietal allelic differences would be expected at such loci and clear variation was found in isozymes produced by chromosomes 6B and 7B. The latter allele, α-Amy-B2b carried by the variety ‘Hope’, was used to locate the enzyme structural gene within chromosome 7B relative to the centromere and five other gene markers. The nature of the α-Amy-B2b phenotype and the rare non-parental isozyme patterns found among the recombinant lines indicates that the locus is large and compound, probably involving some degree of intra-locus gene duplication.

    Notes: Summary Thirty-three β-amylase isozymes were separated in ‘Chinese Spring’ by IEF and the structural genes encoding seventeen of these were located by nullisomic analysis. The locations of the previously reported β-Amy-1 loci on chromosome arms 4Aβ (β-Amy-A1) and 4DL (β-Amy-D1) were confirmed and another set, β-Amy-2, was found on the group 5 chromosomes. A locus on 5AL (β-Amy-A1) was identified by nullisomic analysis and another on chromosome 5B (β-Amy-B2) was identified by analysis of inter-varietal chromosome substitution lines. The loci are complex, each coding for several isozymes, and allelic variation occurs at all four. Two alleles were identified at β-Amy-A1, five at β-Amy-D1, five at β-Amy-A2 and two at β-Amy-B2. Eleven different β-AMY phenotypes were distinguished amongst the 46 wheat varieties screened. The β-Amy-A2 locus was mapped by employing chromosome 5A recombinant lines and found to be closely linked and proximal to the awn inhibitor, B1, on the long arm. An attempt to map the β-Amy-D1 locus on chromosome 4D showed the gene to be located in the distal region of the long arm.

    Notes: Summary The effects of the ‘Tom Thumb’ Rht3 dwarfing gene in wheat on plant height, yield components and grain quality in field trials of random inbred lines from two ‘Minister Dwarf’ (Rht3) x rht3 crosses are described. Limited genetic increases in Rht3 plant height, up to 70 cm, were achieved by incorporating genes from the extremely tall variety ‘April Bearded’. Amongst lines from the higher yielding cross, the dwarfing gene did not reduce grain yield but was associated with an increase in grain number of over 25%. Although Rht3 reduced grain protein content by up to 0.8%, breadmaking quality, as measured by SDS sedimentation values and baking tests, was not affected. A controlled environment experiment with near-isogenic Rht3 and rht3 lines indicated that reduced grain size in the dwarfs is a competitive response to the increase in fertility rather than a primary effect of the dwarfing gene.
