    Beschreibung: At Santorini, active normal faulting controls the emission of volcanic products. Such geometry has implication on seismic activity around the plumbing system during unrest. Static Coulomb stress changes induced by the 2011–2012 inflation within a preexisting NW-SE extensional regional stress field, compatible with fault geometry, increased by more than 0.5MPa in an ellipsoid-shaped zone beneath the Minoan caldera where almost all earthquakes (96%) have occurred since beginning of unrest. Magmatic processes perturb the regional stress in the caldera where strike-slip rather than normal faulting along NE-SW striking planes are expected. The inflation may have also promoted more distant moderate earthquakes on neighboring faults as the M〉5 January 2012, south of Christiania. Santorini belongs to a set of en echelon NE-SW striking rifts (Milos, Nysiros) oblique to the Aegean arc that may have initiated in the Quaternary due to propagation of the North Anatolian fault into the Southern Aegean Sea.

    Beschreibung: Deep-water ecosystems are characterized by relatively low carbonate concentration values and, due to ocean acidification (OA), these habitats might be among the first to be exposed to undersaturated conditions in the forthcoming years. However, until now, very few studies have been conducted to test how cold-water coral (CWC) species react to such changes in the seawater chemistry. The present work aims to investigate the mid-term effect of decreased pH on calcification of the two branching CWC species most widely distributed in the Mediterranean, Lophelia pertusa and Madrepora oculata. No significant effects were observed in the skeletal growth rate, microdensity and porosity of both species after 6 months of exposure. However, while the calcification rate of M. oculata was similar for all colony fragments, a heterogeneous skeletal growth pattern was observed in L. pertusa, the younger nubbins showing higher growth rates than the older ones. A higher energy demand is expected in these young, fast-growing fragments and, therefore, a reduction in calcification might be noticed earlier during long-term exposure to acidified conditions.

    Beschreibung: The rate of reaction of OH with CS2 to form OCS by reaction (1) has been measured through observation of O14CS following 254 nm equation image photolysis of mixtures of H2O2 with 14CS2. The OH concentrations have been monitored through simultaneous measurement in the same cell of either (a) the oxidation of CO to CO2, or (b) the removal of a hydrocarbon such as C3H8 or iso-C4H10. The upper limit for the formation of OCS based on (a) corresponds to a rate constant k1 〈 0.3 × 10−14 cm³ molecule−1 sec−1. Other chemical reactions in the system have led to the formation of both 14CO and 14CO2, indicating the existence of a complex combination of reactions such that the observed O14CS need not have been formed by (1). The rate of reaction (1) is sufficiently slow that it is neither an important atmospheric sink for CS2 nor an important source for atmospheric OCS. The reaction of OH with OCS has not been measured in these experiments, but by analogy with k1 it is probably not an important atmospheric sink for OCS nor an important source of SO2.

    Beschreibung: We reconstructed a high‐resolution, alkenone‐based sea surface temperature (SST) record spanning the last ca. 150 years, from a sediment core retrieved within the main upwelling zone off Peru. A conspicuous SST decline is evidenced since the 1950s despite interdecadal SST variability. Instrumental SST data and reanalysis of ECMWF ERA 40 winds suggest that the recent coastal cooling corresponds mainly to an intensification of alongshore winds and associated increase of upwelling in spring. Consistently, both proxy and instrumental data evidence increased productivity in phase with the SST cooling. Our data expand on previous reports on recent SST cooling in other Eastern Boundary upwelling systems and support scenarios that relate coastal upwelling intensification to global warming. Yet, further investigations are needed to assess the role of different mechanisms and forcings (enhanced local winds vs. spin‐up of the South Pacific High Pressure cell).

    Beschreibung: A major term in the global carbon cycle is the ocean's biological carbon pump which is dominated by sinking of small organic particles from the surface ocean to its interior. Several different approaches to estimating the magnitude of the pump have been used, yielding a large range of estimates. Here, we use an alternative methodology, a thorium isotope tracer, that provides direct estimates of particulate organic carbon export. A large database of thorium-derived export measurements was compiled and extrapolated to the global scale by correlation with satellite sea surface temperature fields. Our estimates of export efficiency are significantly lower than those derived from the f-ratio, and we estimate global integrated carbon export as ∼5 GtC yr−1, lower than most current estimates. The lack of consensus amongst different methodologies on the strength of the biological carbon pump emphasises that our knowledge of a major planetary carbon flux remains incomplete.

    Beschreibung: Terrestrial actinomycetes are noteworthy producers of a multitude of antibiotics, however the marine representatives are much less studied in this regard. In this study, 90 actinomycetes were isolated from 11 different species of marine sponges that had been collected from offshore Ras Mohamed (Egypt) and from Rovinj (Croatia). Phylogenetic characterization of the isolates based on 16S rRNA gene sequencing supported their assignment to 18 different actinomycete genera representing seven different suborders. Fourteen putatively novel species were identified based on sequence similarity values below 98.2% to other strains in the NCBI database. A putative new genus related to Rubrobacter was isolated on M1 agar that had been amended with sponge extract, thus highlighting the need for innovative cultivation protocols. Testing for anti-infective activities was performed against clinically relevant, Gram-positive (Enterococcus faecalis, Staphylococcus aureus) and Gram-negative (Escherichia coli, Pseudomonas aeruginosa) bacteria, fungi (Candida albicans) and human parasites (Leishmania major, Trypanosoma brucei). Bioactivities against these pathogens were documented for 10 actinomycete isolates. These results show a high diversity of actinomycetes associated with marine sponges as well as highlight their potential to produce anti-infective agents.

    Beschreibung: Actinomycetes are prolific producers of pharmacologically important compounds accounting for about 70% of the naturally derived antibiotics that are currently in clinical use. In this study, we report on the isolation of Streptomyces sp. strains from Mediterranean sponges, on their secondary metabolite production and on their screening for anti-infective activities. Bioassay-guided isolation and purification yielded three previously known compounds namely, cyclic depsipeptide valinomycin, indolocarbazole alkaloid staurosporine and butenolide. This is the first report of the isolation of valinomycin from a marine source. These compounds exhibited novel anti-parasitic activities specifically against Leishmania major (valinomycin IC50 〈 0.11 μM; staurosporine IC50 5.30 μM) and Trypanosoma brucei brucei (valinomycin IC50 0.0032 μM; staurosporine IC50 0.022 μM; butenolide IC50 31.77 μM). These results underscore the potential of marine actinomycetes to produce bioactive compounds as well as the re-evaluation of previously known compounds for novel anti-infective activities.

    Beschreibung: In this paper, we report new protease inhibitory activity of plakortide E towards cathepsins and cathepsin-like parasitic proteases. We further report on its anti-parasitic activity against Trypanosoma brucei with an IC₅₀ value of 5 μM and without cytotoxic effects against J774.1 macrophages at 100 μM concentration. Plakortide E was isolated from the sponge Plakortis halichondroides using enzyme assay-guided fractionation and identified by NMR spectroscopy and mass spectrometry.     Beschreibung: High resolution Fourier transform mass spectrometry (HRFTMS) and nuclear magnetic resonance (NMR) spectroscopy were employed as complementary metabolomic tools to dereplicate the chemical profile of the new and antitrypanosomally active sponge-associated bacterium Actinokineospora sp. EG49 extract. Principal Component (PCA), hierarchical clustering (HCA), and orthogonal partial least square-discriminant analysis (OPLS-DA) were used to evaluate the HRFTMS and NMR data of crude extracts from four different fermentation approaches. Statistical analysis identified the best culture one-strain-many-compounds (OSMAC) condition and extraction procedure, which was used for the isolation of novel bioactive metabolites. As a result, two new O-glycosylated angucyclines, named actinosporins A (1) and B (2), were isolated from the broth culture of Actinokineospora sp. strain EG49, which was cultivated from the Red Sea sponge Spheciospongia vagabunda. The structures of actinosporins A and B were determined by 1D- and 2D-NMR techniques, as well as high resolution tandem mass spectrometry. Testing for antiparasitic properties showed that actinosporin A exhibited activity against Trypanosoma brucei brucei with an IC₅₀ value of 15 µM; however no activity was detected against Leishmania major and Plasmodium falciparum, therefore suggesting its selectivity against the parasite Trypanosoma brucei brucei; the causative agent of sleeping sickness.

    Beschreibung: The diversity of actinomycetes associated with marine sponges collected off Fsar Reef (Saudi Arabia) was investigated in the present study. Forty-seven actinomycetes were cultivated and phylogenetically identified based on 16S rRNA gene sequencing and were assigned to 10 different actinomycete genera. Eight putatively novel species belonging to genera Kocuria, Mycobacterium, Nocardia, and Rhodococcus were identified based on sequence similarity values below 98.2% to other 16S rRNA gene sequences available in the NCBI database. PCR-based screening for biosynthetic genes including type I and type II polyketide synthases (PKS-I, PKS-II) as well as nonribosomal peptide synthetases (NRPS) showed that 20 actinomycete isolates encoded each at least one type of biosynthetic gene. The organic extracts of nine isolates displayed bioactivity against at least one of the test pathogens, which were Gram-positive and Gram-negative bacteria, fungi, human parasites, as well as in a West Nile Virus protease enzymatic assay. These results emphasize that marine sponges are a prolific resource for novel bioactive actinomycetes with potential for drug discovery.

    Beschreibung: Two sponge-derived actinomycetes, Actinokineospora sp. EG49 and Nocardiopsis sp. RV163, were grown in co-culture and the presence of induced metabolites monitored by ¹H NMR. Ten known compounds, including angucycline, diketopiperazine and β-carboline derivatives 1-10, were isolated from the EtOAc extracts of Actinokineospora sp. EG49 and Nocardiopsis sp. RV163. Co-cultivation of Actinokineospora sp. EG49 and Nocardiopsis sp. RV163 induced the biosynthesis of three natural products that were not detected in the single culture of either microorganism, namely N-(2-hydroxyphenyl)-acetamide (11), 1,6-dihydroxyphenazine (12) and 5a,6,11a,12-tetrahydro-5a,11a-dimethyl1,4benzoxazino3,2-b1,4benzoxazine (13a). When tested for biological activity against a range of bacteria and parasites, only the phenazine 12 was active against Bacillus sp. P25, Trypanosoma brucei and interestingly, against Actinokineospora sp. EG49. These findings highlight the co-cultivation approach as an effective strategy to access the bioactive secondary metabolites hidden in the genomes of marine actinomycetes

    Beschreibung: Marine invertebrate-associated symbiotic bacteria produce a plethora of novel secondary metabolites which may be structurally unique with interesting pharmacological properties. Selection of strains usually relies on literature searching, genetic screening and bioactivity results, often without considering the chemical novelty and abundance of secondary metabolites being produced by the microorganism until the time-consuming bioassay-guided isolation stages. To fast track the selection process, metabolomic tools were used to aid strain selection by investigating differences in the chemical profiles of 77 bacterial extracts isolated from cold water marine invertebrates from Orkney, Scotland using liquid chromatography-high resolution mass spectrometry (LC-HRMS) and nuclear magnetic resonance (NMR) spectroscopy. Following mass spectrometric analysis and dereplication using an Excel macro developed in-house, principal component analysis (PCA) was employed to differentiate the bacterial strains based on their chemical profiles. NMR 1H and correlation spectroscopy (COSY) were also employed to obtain a chemical fingerprint of each bacterial strain and to confirm the presence of functional groups and spin systems. These results were then combined with taxonomic identification and bioassay screening data to identify three bacterial strains, namely Bacillus sp. 4117, Rhodococcus sp. ZS402 and Vibrio splendidus strain LGP32, to prioritize for scale-up based on their chemically interesting secondary metabolomes, established through dereplication and interesting bioactivities, determined from bioassay screening.

    Beschreibung: Diazepinomicin is a dibenzodiazepine alkaloid with an unusual structure among the known microbial metabolites discovered so far. Diazepinomicin was isolated from the marine sponge-associated strain Micromonospora sp. RV115 and was identified by spectroscopic analysis and by comparison to literature data. In addition to its interesting preclinical broad-spectrum antitumor potential, we report here new antioxidant and anti-protease activities for this compound. Using the ferric reducing antioxidant power (FRAP) assay, a strong antioxidant potential of diazepinomicin was demonstrated. Moreover, diazepinomicin showed a significant antioxidant and protective capacity from genomic damage induced by the reactive oxygen species hydrogen peroxide in human kidney (HK-2) and human promyelocytic (HL-60) cell lines. Additionally, diazepinomicin inhibited the proteases rhodesain and cathepsin L at an IC50 of 70–90 µM. It also showed antiparasitic activity against trypomastigote forms of Trypanosoma brucei with an IC50 of 13.5 µM. These results showed unprecedented antioxidant and anti-protease activities of diazepinomicin, thus further highlighting its potential as a future drug candidate.

    Beschreibung: Genomic mining revealed one major nonribosomal peptide synthetase (NRPS) phylogenetic cluster in 12 marine sponge species, one ascidian, an actinobacterial isolate and seawater. Phylogenetic analysis predicts its taxonomic affiliation to the actinomycetes and hydroxy-phenyl-glycine as a likely substrate. Additionally, a phylogenetically distinct NRPS gene cluster was discovered in the microbial metagenome of the sponge Aplysina aerophoba, which shows highest similarities to NRPS genes that were previously assigned, by ways of single cell genomics, to a Chloroflexi sponge symbiont. Genomic mining studies such as the one presented here for NRPS genes, contribute to on-going efforts to characterize the genomic potential of sponge-associated microbiota for secondary metabolite biosynthesis.

    Beschreibung: Four new tetromycin derivatives, tetromycins 1–4 and a previously known one, tetromycin B (5) were isolated from Streptomyces axinellae Pol001T cultivated from the Mediterranean sponge Axinella polypoides. Structures were assigned using extensive 1D and 2D NMR spectroscopy as well as HRESIMS analysis. The compounds were tested for antiparasitic activities against Leishmania major and Trypanosoma brucei, and for protease inhibition against several cysteine proteases such as falcipain, rhodesain, cathepsin L, cathepsin B, and viral proteases SARS-CoV Mpro, and PLpro. The compounds showed antiparasitic activities against T. brucei and time-dependent inhibition of cathepsin L-like proteases with Ki values in the low micromolar range.

    Beschreibung: Two new 20-membered macrolides, levantilide A and B, were isolated from the Micromonospora strain M71-A77. Strain M71-A77 was recovered from an Eastern Mediterranean deep-sea sediment sample and revealed to produce the levantilides under in situ salinity of 38.6‰. The chemical structures of the levantilides were elucidated on the basis of different one- and two- dimensional NMR experiments. Levantilide A exhibits a moderate antiproliferative activity against several tumor cell lines.

    Beschreibung: Climate change with increasing temperature and ocean acidification (OA) poses risks for marine ecosystems. According to Portner and Farrell [1], synergistic effects of elevated temperature and CO(2)-induced OA on energy metabolism will narrow the thermal tolerance window of marine ectothermal animals. To test this hypothesis, we investigated the effect of an acute temperature rise on energy metabolism of the oyster, Crassostrea gigas chronically exposed to elevated CO(2) levels (partial pressure of CO(2) in the seawater similar to 0.15 kPa, seawater pH similar to 7.7). Within one month of incubation at elevated PCO(2) and 15 degrees C hemolymph pH fell (pH(e) = 7.1 +/- 0.2 (CO(2)-group) vs. 7.6 +/- 0.1 (control)) and P(e)CO(2) values in hemolymph increased (0.5 +/- 0.2 kPa (CO(2)-group) vs. 0.2 +/- 0.04 kPa (control)). Slightly but significantly elevated bicarbonate concentrations in the hemolymph of CO(2)-incubated oysters ([HCO(3)(-)](e) = 1.8 +/- 0.3 mM (CO(2)-group) vs. 1.3 +/- 0.1 mM (control)) indicate only minimal regulation of extracellular acid-base status. At the acclimation temperature of 15 degrees C the OA-induced decrease in pHe did not lead to metabolic depression in oysters as standard metabolism rates (SMR) of CO(2)-exposed oysters were similar to controls. Upon acute warming SMR rose in both groups, but displayed a stronger increase in the CO(2)-incubated group. Investigation in isolated gill cells revealed a similar temperature-dependence of respiration between groups. Furthermore, the fraction of cellular energy demand for ion regulation via Na(+)/K(+)-ATPase was not affected by chronic hypercapnia or temperature. Metabolic profiling using (1)H-NMR spectroscopy revealed substantial changes in some tissues following OA exposure at 15 degrees C. In mantle tissue alanine and ATP levels decreased significantly whereas an increase in succinate levels was observed in gill tissue. These findings suggest shifts in metabolic pathways following OA-exposure. Our study confirms that OA affects energy metabolism in oysters and suggests that climate change may affect populations of sessile coastal invertebrates such as mollusks.

    Beschreibung: A fast and efficient approach was established to identify bacteria possessing the potential to biosynthesize phenazines, which are of special interest regarding their antimicrobial activities. Sequences of phzE genes, which are part of the phenazine biosynthetic pathway, were used to design one universal primer system and to analyze the ability of bacteria to produce phenazine. Diverse bacteria from different marine habitats and belonging to six major phylogenetic lines were investigated. Bacteria exhibiting phzE gene fragments affiliated to Firmicutes, Alpha- and Gammaproteobacteria, and Actinobacteria. Thus, these are the first primers for amplifying gene fragments from Firmicutes and Alphaproteobacteria. The genetic potential for phenazine production was shown for four type strains belonging to the genera Streptomyces and Pseudomonas as well as for 13 environmental isolates from marine habitats. For the first time, the genetic ability of phenazine biosynthesis was verified by analyzing the metabolite pattern of all PCR-positive strains via HPLC-UV/MS. Phenazine production was demonstrated for the type strains known to produce endophenazines, 2-hydroxy-phenazine, phenazine-1-carboxylic acid, phenazine-1,6-dicarboxylic acid, and chlororaphin as well as for members of marine Actinobacteria. Interestingly, a number of unidentified phenazines possibly represent new phenazine structures.

    Beschreibung: Fungi associated with the marine sponge Tethya aurantium were isolated and identified by morphological criteria and phylogenetic analyses based on internal transcribed spacer (ITS) regions. They were evaluated with regard to their secondary metabolite profiles. Among the 81 isolates which were characterized, members of 21 genera were identified. Some genera like Acremonium, Aspergillus, Fusarium, Penicillium, Phoma, and Trichoderma are quite common, but we also isolated strains belonging to genera like Botryosphaeria, Epicoccum, Parasphaeosphaeria, and Tritirachium which have rarely been reported from sponges. Members affiliated to the genera Bartalinia and Volutella as well as to a presumably new Phoma species were first isolated from a sponge in this study. On the basis of their classification, strains were selected for analysis of their ability to produce natural products. In addition to a number of known compounds, several new natural products were identified. The scopularides and sorbifuranones have been described elsewhere. We have isolated four additional substances which have not been described so far. The new metabolite cillifuranone (1) was isolated from Penicillium chrysogenum strain LF066. The structure of cillifuranone (1) was elucidated based on 1D and 2D NMR analysis and turned out to be a previously postulated intermediate in sorbifuranone biosynthesis. Only minor antibiotic bioactivities of this compound were found so far.

    Beschreibung: The recovery of natural gas from CH4-hydrate deposits in sub-marine and sub-permafrost environments through injection of CO2 is considered a suitable strategy towards emission-neutral energy production. This study shows that the injection of hot, supercritical CO2 is particularly promising. The addition of heat triggers the dissociation of CH4-hydrate while the CO2, once thermally equilibrated, reacts with the pore water and is retained in the reservoir as immobile CO2-hydrate. Furthermore, optimal reservoir conditions of pressure and temperature are constrained. Experiments were conducted in a high-pressure flow-through reactor at different sediment temperatures (2 °C, 8 °C, 10 °C) and hydrostatic pressures (8 MPa, 13 MPa). The efficiency of both, CH4 production and CO2 retention is best at 8 °C, 13 MPa. Here, both CO2- and CH4-hydrate as well as mixed hydrates can form. At 2 °C, the production process was less effective due to congestion of transport pathways through the sediment by rapidly forming CO2-hydrate. In contrast, at 10 °C CH4 production suffered from local increases in permeability and fast breakthrough of the injection fluid, thereby confining the accessibility to the CH4 pool to only the most prominent fluid channels. Mass and volume balancing of the collected gas and fluid stream identified gas mobilization as equally important process parameter in addition to the rates of methane hydrate dissociation and hydrate conversion. Thus, the combination of heat supply and CO2 injection in one supercritical phase helps to overcome the mass transfer limitations usually observed in experiments with cold liquid or gaseous CO2.

    Beschreibung: Superparamagnetic iron-oxide particles (SPIO) are used in different ways as contrast agents for magnetic resonance imaging (MRI): Particles with high nonspecific uptake are required for unspecific labeling of phagocytic cells whereas those that target specific molecules need to have very low unspecific cellular uptake. We compared iron-oxide particles with different core materials (magnetite, maghemite), different coatings (none, dextran, carboxydextran, polystyrene) and different hydrodynamic diameters (20-850 nm) for internalization kinetics, release of internalized particles, toxicity, localization of particles and ability to generate contrast in MRI. Particle uptake was investigated with U118 glioma cells und human umbilical vein endothelial cells (HUVEC), which exhibit different phagocytic properties. In both cell types, the contrast agents Resovist, B102, non-coated Fe(3)O(4) particles and microspheres were better internalized than dextran-coated Nanomag particles. SPIO uptake into the cells increased with particle/iron concentrations. Maximum intracellular accumulation of iron particles was observed between 24 h to 36 h of exposure. Most particles were retained in the cells for at least two weeks, were deeply internalized, and only few remained adsorbed at the cell surface. Internalized particles clustered in the cytosol of the cells. Furthermore, all particles showed a low toxicity. By MRI, monolayers consisting of 5000 Resovist-labeled cells could easily be visualized. Thus, for unspecific cell labeling, Resovist and microspheres show the highest potential, whereas Nanomag particles are promising contrast agents for target-specific labeling.

    Beschreibung: Microbial studies of the Mediterranean sponge Tethya aurantium led to the isolation of the fungus Bartalinia robillardoides strain LF550. The strain produced a number of secondary metabolites belonging to the chloroazaphilones. This is the first report on the isolation of chloroazaphilones of a fungal strain belonging to the genus Bartalinia. Besides some known compounds (helicusin A (1) and deacetylsclerotiorin (2)), three new chloroazaphilones (helicusin E (3); isochromophilone X (4) and isochromophilone XI (5)) and one new pentaketide (bartanolide (6)) were isolated. The structure elucidations were based on spectroscopic analyses. All isolated compounds revealed different biological activity spectra against a test panel of four bacteria: three fungi; two tumor cell lines and two enzymes.

    Beschreibung: Heating rate calculations with the FUBRad shortwave (SW) radiation parameterization have been performed to examine the effect of prescribed spectral solar fluxes from the NRLSSI, MPS and IUP data sets on SW heating rates over the 11 year solar cycle 22. The corresponding temperature response is derived from perpetual January General Circulation Model (GCM) simulations with prescribed ozone concentrations. The different solar flux input data sets induce clear differences in SW heating rates at solar minimum, with the established NRLSSI data set showing the smallest solar heating rates. The stronger SW heating in the middle and upper stratosphere in the MPS data warms the summer upper stratosphere by 2 K. Over the solar cycle, SW heating rate differences vary up to 40% between the irradiance data sets, but do not result in a significant change of the solar temperature signal. Lower solar fluxes in the newer SIM data lead to a significantly cooler stratosphere and mesosphere when compared to NRLSSI data for 2007. Changes in SW heating from 2004 to 2007 are however up to six times stronger than for the NRLSSI data.     Beschreibung: The accumulation of methane hydrate in marine sediments is controlled by a number of physical and biogeochemical parameters including the thickness of the gas hydrate stability zone (GHSZ), the solubility of methane in pore fluids, the accumulation of particulate organic carbon at the seafloor, the kinetics of microbial organic matter degradation and methane generation in marine sediments, sediment compaction and the ascent of deep-seated pore fluids and methane gas into the GHSZ. Our present knowledge on these controlling factors is discussed and new estimates of global sediment and methane fluxes are provided applying a transport-reaction model at global scale. The modeling and the data evaluation yield improved and better constrained estimates of the global pore volume within the modern GHSZ ( ≥ 44 × 1015 m3), the Holocene POC accumulation rate at the seabed (~1.4 × 1014 g yr−1), the global rate of microbial methane production in the deep biosphere (4−25 × 1012 g C yr−1) and the inventory of methane hydrates in marine sediments ( ≥ 455 Gt of methane-bound carbon).

    Beschreibung: The marine-derived filamentous fungus Asteromyces cruciatus 763, obtained off the coast of La Jolla, San Diego, USA, yielded the new pentapeptide lajollamide A (1), along with the known compounds regiolone (2), hyalodendrin (3), gliovictin (4), 1N-norgliovicitin (5), and bis-N-norgliovictin (6). The planar structure of lajollamide A (1) was determined by Nuclear Magnetic Resonance (NMR) spectroscopy in combination with mass spectrometry. The absolute configuration of lajollamide A (1) was unambiguously solved by total synthesis which provided three additional diastereomers of 1 and also revealed that an unexpected acid-mediated partial racemization (2:1) of the l-leucine and l-N-Me-leucine residues occurred during the chemical degradation process. The biological activities of the isolated metabolites, in particular their antimicrobial properties, were investigated in a series of assay systems.

    Beschreibung: Photosynthesis by phytoplankton in sunlit surface waters transforms inorganic carbon and nutrients into organic matter, a portion of which is subsequently transported vertically through the water column by the process known as the biological carbon pump (BCP). The BCP sustains the steep vertical gradient in total dissolved carbon, thereby contributing to net carbon sequestration. Any changes in the vertical transportation of the organic matter as a result of future climate variations will directly affect surface ocean carbon dioxide (CO 2) concentrations, and subsequently influence oceanic uptake of atmospheric CO 2 and climate. Here we present results of experiments designed to investigate the potential effects of ocean acidification and warming on the BCP. These perturbation experiments were carried out in enclosures (3,000 L volume) in a controlled mesocosm facility that mimicked future pCO 2 (∼900 ppmv) and temperature (3°C higher than ambient) conditions. The elevated CO 2 and temperature treatments disproportionately enhanced the ratio of dissolved organic carbon (DOC) production to particulate organic carbon (POC) production, whereas the total organic carbon (TOC) production remained relatively constant under all conditions tested. A greater partitioning of organic carbon into the DOC pool indicated a shift in the organic carbon flow from the particulate to dissolved forms, which may affect the major pathways involved in organic carbon export and sequestration under future ocean conditions.

    Beschreibung: Chemical analysis of the Indonesian sponge Plakortis cfr. lita afforded two new analogues of the potent trypanocidal agent manadoperoxide B (1), namely 12-isomanadoperoxide B (2) and manadoperoxidic acid B (3). These compounds were isolated along with a new short chain dicarboxylate monoester (4), bearing some interesting relationships with the polyketide endoperoxides found in this sponge. Some semi-synthetic analogues of manadoperoxide B (6-8) were prepared and evaluated for antitrypanosomal activity and cytotoxicity. These studies revealed crucial structure-activity relationships that should be taken into account in the design of optimized and simplified endoperoxyketal trypanocidal agents.

    Beschreibung: In this study, thirteen sponge-derived terpenoids, including five linear furanoterpenes: furospinulosin-1 (1), furospinulosin-2 (2), furospongin-1 (3), furospongin-4 (4), and demethylfurospongin-4 (5); four linear meroterpenes: 2-(hexaprenylmethyl)-2-methylchromenol (6), 4-hydroxy-3-octaprenylbenzoic acid (7), 4-hydroxy-3-tetraprenyl-phenylacetic acid (8), and heptaprenyl-p-quinol (9); a linear triterpene, squalene (10); two spongian-type diterpenes dorisenone D (11) and 11β-acetoxyspongi-12-en-16-one (12); a scalarane-type sesterterpene; 12-epi-deoxoscalarin (13), as well as an indole alkaloid, tryptophol (14) were screened for their in vitro activity against four parasitic protozoa; Trypanosoma brucei rhodesiense, Trypanosoma cruzi, Leishmania donovani and Plasmodium falciparum. Cytotoxic potential of the compounds on mammalian cells was also assessed. All compounds were active against T. brucei rhodesiense, with compound 8 being the most potent (IC50 0.60 μg/mL), whereas 9 and 12 were the most active compounds against T. cruzi, with IC50 values around 4 μg/mL. Compound 12 showed the strongest leishmanicidal activity (IC50 0.75 µg/mL), which was comparable to that of miltefosine (IC50 0.20 µg/mL). The best antiplasmodial effect was exerted by compound 11 (IC50 0.43 µg/mL), followed by compounds 7, 10, and 12 with IC50 values around 1 µg/mL. Compounds 9, 11 and 12 exhibited, besides their antiprotozoal activity, also some cytotoxicity, whereas all other compounds had low or no cytotoxicity towards the mammalian cell line. This is the first report of antiprotozoal activity of marine metabolites 1–14, and points out the potential of marine sponges in discovery of new antiprotozoal lead compounds.

    Beschreibung: In the present study,13 bromopyrrole alkaloids, including the oroidin analogs hymenidin (2), dispacamide B (3) and dispacamide D (4), stevensine (5) and spongiacidin B (6), their derivatives lacking the imidazole ring bromoaldisin (7), longamide B (8) and longamide A (9), the dimeric oroidin derivatives sceptrin (10) and dibromopalau’amine (11), and the non-oroidin bromopyrrolohomoarginin (12), manzacidin A (13), and agelongine (14), obtained from marine sponges belonging to Axinella and Agelas generahave been screened in vitro against four parasitic protozoa, i.e., two Trypanosoma species (T. brucei rhodesiense and T. cruzi), Leishmania donovani and Plasmodium falciparum (K1 strain, a chloroquine resistant strain), responsible of human diseases with high morbidity and, in the case of malaria, high mortality. Our results indicate longamide B (8) and dibromopalau’amine (11) to be promising trypanocidal and antileishmanial agents, while dispacamide B (3) and spongiacidin B (6) emerge as antimalarial lead compounds.In addition,evaluation of the activity of the test alkaloids (2–14) against three different enzymes (PfFabI, PfFabG, PfFabZ) involved in the de novo fatty acid biosynthesis pathway of P. falciparum (PfFAS-II) identified bromopyrrolohomoarginin (12) as a potent inhibitor of PfFabZ. The structural similarity within the series of tested molecules allowed us to draw some preliminary structure-activity relationships. Tests against the mammalian L6 cells revealed important clues on therapeutic index of the metabolites. This is the first detailed study on the antiprotozoal potential of marine bromopyrrole alkaloids.

    Beschreibung: A novel spirocyclic drimane coupled by two drimane fragment building blocks 2 and a new drimane 1 were identified in mycelia and culture broth of Stachybotrys sp. MF347. Their structures were established by spectroscopic means. This is the first example of spirocyclic drimane coupled by a spirodihydrobenzofuranlactam unit and a spirodihydroisobenzofuran unit; and the connecting position being N-C instead of an N and N connecting unit. Strain MF347 produced also the known spirocyclic drimanes stachybocin A (12) and stachybocin B (11) featured by two sesquiterpene-spirobenzofuran structural units connected by a lysine residue; the known spirocyclic drimanes chartarlactam O (5); chartarlactam K (6); F1839A (7); stachybotrylactam (8); stachybotramide (9); and 2α-acetoxystachybotrylactam acetate (10); as well as ilicicolin B (13), a known sesquiterpene. The relative configuration of two known spirobenzofuranlactams (3 and 4) was determined. All compounds were subjected to biological activity tests. The spirocyclic drimane 2, 11, and 12, as well as the sesquiterpene 13, exhibited antibacterial activity against the clinically relevant methicillin-resistant Staphylococcus aureus (MRSA).

    Beschreibung: Two unusual pyridones, trichodin A (1) and trichodin B (2), together with the known compound, pyridoxatin (3), were extracted from mycelia and culture broth of the marine fungus, Trichoderma sp. strain MF106 isolated from the Greenland Seas. The structures of the new compounds were characterized as an intramolecular cyclization of a pyridine basic backbone with a phenyl group. The structure and relative configuration of the new compounds were established by spectroscopic means. The new compound 1 and the known compound 3 showed antibiotic activities against the clinically relevant microorganism, Staphylococcus epidermidis, with IC50 values of 24 μM and 4 μM, respectively.

    Beschreibung: Eight streptophenazines (A–H) have been identified so far as products of Streptomyces strain HB202, which was isolated from the sponge Halichondria panicea from the Baltic Sea. The variation of bioactivities based on small structural changes initiated further studies on new derivatives. Three new streptophenazines (I–K) were identified after fermentation in the present study. In addition, revised molecular structures of streptophenazines C, D, F and H are proposed. Streptophenazines G and K exhibited moderate antibacterial activity against the facultative pathogenic bacterium Staphylococcus epidermidis and against Bacillus subtilis. All tested compounds (streptophenazines G, I–K) also showed moderate activities against PDE 4B.

    Beschreibung: Bioactive compounds were detected in crude extracts of the fungus, Calcarisporium sp. KF525, which was isolated from German Wadden Sea water samples. Purification of the metabolites from the extracts yielded the five known polyesters, 15G256α, α-2, β, β-2 and π (1–5), and five new derivatives thereof, named calcarides A–E (6–10). The chemical structures of the isolated compounds were elucidated on the basis of one- and two-dimensional NMR spectroscopy supported by UV and HRESIMS data. The compounds exhibited inhibitory activities against Staphylococcus epidermidis, Xanthomonas campestris and Propionibacterium acnes. As the antibacterial activities were highly specific with regard to compound and test strain, a tight structure-activity relationship is assumed.

    Beschreibung: New isolates of Streptomyces champavatii were isolated from marine sediments of the Gotland Deep (Baltic Sea), from the Urania Basin (Eastern Mediterranean), and from the Kiel Bight (Baltic Sea). The isolates produced several oligopeptidic secondary metabolites, including the new octapeptide champacyclin (1a) present in all three strains. Herein, we report on the isolation, structure elucidation and determination of the absolute stereochemistry of this isoleucine/leucine (Ile/Leu = Xle) rich cyclic octapeptide champacyclin (1a). As 2D nuclear magnetic resonance (NMR) spectroscopy could not fully resolve the structure of (1a), additional information on sequence and configuration of stereocenters were obtained by a combination of multi stage mass spectrometry (MSn) studies, amino acid analysis, partial hydrolysis and subsequent enantiomer analytics with gas chromatography positive chmical ionization/electron impact mass spectrometry (GC-PCI/EI-MS) supported by comparison to reference dipeptides. Proof of the head-to-tail cyclization of (1a) was accomplished by solid phase peptide synthesis (SPPS) compared to an alternatively side chain cyclized derivative (2). Champacyclin (1a) is likely synthesized by a non-ribosomal peptide synthetase (NRPS), because of its high content of (d)-amino acids. The compound (1a) showed antimicrobial activity against the phytopathogen Erwinia amylovora causing the fire blight disease of certain plants.

    Beschreibung: Research to date has suggested that both individual marine species and ecological processes are expected to exhibit diverse responses to the environmental effects of climate change. Evolutionary responses can occur on rapid (ecological) timescales, and yet studies typically do not consider the role that adaptive evolution will play in modulating biological responses to climate change. Investigations into such responses have typically been focused at particular biological levels (e.g., cellular, population, community), often lacking interactions among levels. Since all levels of biological organisation are sensitive to global climate change, there is a need to elucidate how different processes and hierarchical interactions will influence species fitness. Therefore, predicting the responses of communities and populations to global change will require multidisciplinary efforts across multiple levels of hierarchy, from the genetic and cellular to communities and ecosystems. Eventually, this may allow us to establish the role that acclimatisation and adaptation will play in determining marine community structures in future scenarios.

    Beschreibung: Terrestrial plants have proven to be a prolific producer of clinically effective antimalarial drugs, but the antimalarial potential of seaweeds has been little explored. The main aim of this study was to assess the in vitro chemotherapeutical and prophylactic potential of the extracts of twenty-three seaweeds collected from the south coast of England against blood stage (BS) and liver stage (LS) Plasmodium parasites. The majority (14) of the extracts were active against BS of P. falciparum, with brown seaweeds Cystoseira tamariscifolia, C. baccata and the green seaweed Ulva lactuca being the most active (IC(50)s around 3 g/mL). The extracts generally had high selectivity indices (〉10). Eight seaweed extracts inhibited the growth of LS parasites of P. berghei without any obvious effect on the viability of the human hepatoma (Huh7) cells, and the highest potential was exerted by U. lactuca and red seaweeds Ceramium virgatum and Halopitys incurvus (IC50 values 14.9 to 28.8 g/mL). The LS-active extracts inhibited one or more key enzymes of the malarial type-II fatty acid biosynthesis (FAS-II) pathway, a drug target specific for LS. Except for the red seaweed Halopitys incurvus, all LS-active extracts showed dual activity versus both malarial intracellular stage parasites. This is the first report of LS antiplasmodial activity and dual stage inhibitory potential of seaweeds.

    Beschreibung: The parameterization of ocean/sea-ice/atmosphere interaction processes is a challenge for regional climate models (RCMs) of the Arctic, particularly for wintertime conditions, when small fractions of thin ice or open water cause strong modifications of the boundary layer. Thus, the treatment of sea ice and sub-grid flux parameterizations in RCMs is of crucial importance. However, verification data sets over sea ice for wintertime conditions are rare. In the present paper, data of the ship-based experiment Transarktika 2019 during the end of the Arctic winter for thick one-year ice conditions are presented. The data are used for the verification of the regional climate model COSMO-CLM (CCLM). In addition, Moderate Resolution Imaging Spectroradiometer (MODIS) data are used for the comparison of ice surface temperature (IST) simulations of the CCLM sea ice model. CCLM is used in a forecast mode (nested in ERA5) for the Norwegian and Barents Seas with 5 km resolution and is run with different configurations of the sea ice model and sub-grid flux parameterizations. The use of a new set of parameterizations yields improved results for the comparisons with in-situ data. Comparisons with MODIS IST allow for a verification over large areas and show also a good performance of CCLM. The comparison with twice-daily radiosonde ascents during Transarktika 2019, hourly microwave water vapor measurements of first 5 km in the atmosphere and hourly temperature profiler data show a very good representation of the temperature, humidity and wind structure of the whole troposphere for CCLM.

Herein, we performed an alternative approach for targeted selection of the best co‐cultivation pair. Eight marine sediment‐derived fungi were classified as strong or weak, based on their anti‐phytopathogenic potency. The fungi were co‐cultured systematically and analyzed for their chemical profiles and anti-phytopathogenic activity. Based on enhanced bioactivity and a significantly different metabolite profile including the appearance of a co‐culture specific cluster, the co‐culture of Plenodomus influorescens (strong) and Pyrenochaeta nobilis (weak) was prioritized for chemical investigation. Large‐scale co‐cultivation resulted in isolation of five polyketide type compounds: two 12‐membered macrolides, dendrodolide E (1) and its new analog dendrodolide N (2), as well as two rare azaphilones spiciferinone (3) and its new analog 8a-hydroxy-spiciferinone (4). A well‐known bis‐naphtho‐γ‐pyrone type mycotoxin, cephalochromin (5), whose production was specifically enhanced in the co-culture, was also isolated. Chemical structures of compounds 1–5 were elucidated by NMR, HRMS and [] 20/D analyses. Compound 5 showed the strongest anti‐phytopathogenic activity against Xanthomonas campestris and Phytophthora infestans with IC50 values of 0.9 and 1.7 µg/mL, respectively.

    Beschreibung: The fungal genus Pyrenochaetopsis is commonly found in soil, terrestrial, and marine environments, however, has received little attention as a source of bioactive secondary metabolites so far. In a recent work, we reported the isolation and characterization of three new anticancer decalinoyltetramic acid derivatives, pyrenosetins A-C, from the Baltic Fucus vesiculosus-derived endophytic fungus Pyrenochaetopsis sp. FVE-001. Herein we report a new pentacyclic decalinoylspirotetramic acid derivative, pyrenosetin D (1), along with two known decalin derivatives wakodecalines A (2) and B (3) from another endophytic strain Pyrenochaetopsis FVE-087 isolated from the same seaweed and showed anticancer activity in initial screenings. The chemical structures of the purified compounds were elucidated by comprehensive analysis of HR-ESIMS, FT-IR, [a]D, 1D and 2D NMR data coupled with DFT calculations of NMR parameters and optical rotation. Compounds 1-3 were evaluated for their anticancer and toxic potentials against the human malignant melanoma cell line (A-375) and the non-cancerous keratinocyte cell line (HaCaT). Pyrenosetin D (1) showed toxicity towards both A-375 and HaCaT cells with IC50 values of 77.5 and 39.3 μM, respectively, while 2 and 3 were inactive. This is the third chemical study performed on the fungal genus Pyrenochaetopsis and the first report of a pentacyclic decalin ring system from the fungal genus Pyrenochaetopsis.

    Beschreibung: There is growing evidence that passive margin sediments in offshore settings host large volumes of fresh and brackish water of meteoric origin in submarine sub-surface reservoirs. Marine geophysical methods, in particular seismic reflection data, can help characterize offshore hydrogeological systems and yet the existing global database of industrial basin wide surveys remains untapped in this context. In this paper we highlight the importance of these data in groundwater exploration, by reviewing existing studies that apply physical stratigraphy and morpho-structural interpretation techniques to provide important information on—reservoir (aquifer) properties and architecture, permeability barriers, paleo-continental environments, sea-level changes and shift of coastal facies through time and conduits for water flow. We then evaluate the scientific and applied relevance of such methodology within a holistic workflow for offshore groundwater research.

    Beschreibung: The study of wetlands is particularly important as these systems act as natural water purifiers and thus can act as sinks for contaminated particles. Wetland sediments are important as they provide an indication of potential contamination across temporal and spatial scales. The current study aimed to investigate the distributions of selected metals and nutrients in different sites in relation to sediment depth, and identify relationships among sediment metals. Significant differences in nutrient (i.e., N, P) and metal (i.e., K, Mg, Na, Fe, Cu, B) concentrations were found across study sites, whereas nutrients (i.e., N, P) and metals (i.e., Ca, Mg, Fe, Cu, Zn) were significantly different with sediment depths. When compared against Canadian sediment standards, most of the assessed metals were within the “no effect” level across the different sites and depths. The K, Ca, and Mg concentration showed extreme contamination across all sites and depths. The enrichment factor values for K, Ca, and Mg showed extremely high enrichment levels for all sites and sediment depths. The Na, Mn, Fe, Cu, Zn, and B concentration showed mostly background enrichment levels. All sediments across the different sites and sediment depths indicated deterioration of sediment quality. Pearson correlations suggest that most metals might have originated in a similar source as that of Mn and B, owing to a lack of significant differences. These results provide baseline information for the general management of the Nylsvley Wetland in relation to sediment metal pollution. The specific sources of metal contaminants also require further elucidation to further inform management efforts.

    Beschreibung: Detailed multi-beam bathymetry, sub-bottom acoustic, and side-scan sonar observations of submerged canyons with tufa barriers were used to characterize the Zrmanja River karst estuary on the eastern Adriatic coast, Croatia. This unique karst environment consists of two submerged karst basins (Novigrad Sea and Karin Sea) that are connected with river canyons named Novsko Ždrilo and Karinsko Ždrilo. The combined use of high-resolution geophysical data with legacy topography and bathymetry data in a GIS environment allowed for the description and interpretation of this geomorphological setting in relation to the Holocene sea-level rise. The tufa barriers had a predominant influence on the Holocene flooding dynamics of the canyons and karst basins. Here, we describe the possible river pathways from the basins during the lowstand and the formation of a lengthening estuary during the Holocene sea-level rise. Based on the analyzed morphologies and the relative sea-level curve for the Adriatic Sea, the flooding of the Novsko Ždrilo occurred 9200 years before present (BP) and Karinsko Ždrilo was flooded after 8400 years BP. The combination of high-resolution geophysical methods gave an accurate representation of the karst estuarine seafloor and the flooding of semi-isolated basins due to sea-level rise.

    Beschreibung: Many filamentous vibriophages encode virulence genes that lead to the emergence of pathogenic bacteria. Most genomes of filamentous vibriophages characterized up until today were isolated from human pathogens. Despite genome-based predictions that environmental Vibrios also contain filamentous phages that contribute to bacterial virulence, empirical evidence is scarce. This study aimed to characterize the bacteriophages of a marine pathogen, Vibrio alginolyticus (Kiel-alginolyticus ecotype) and to determine their role in bacterial virulence. To do so, we sequenced the phage-containing supernatant of eight different V. alginolyticus strains, characterized the phages therein and performed infection experiments on juvenile pipefish to assess their contribution to bacterial virulence. We were able to identify two actively replicating filamentous phages. Unique to this study was that all eight bacteria of the Kiel-alginolyticus ecotype have identical bacteriophages, supporting our previously established theory of a clonal expansion of the Kiel-alginolyticus ecotype. We further found that in one of the two filamentous phages, two phage-morphogenesis proteins (Zot and Ace) share high sequence similarity with putative toxins encoded on the Vibrio cholerae phage CTXΦ. The coverage of this filamentous phage correlated positively with virulence (measured in controlled infection experiments on the eukaryotic host), suggesting that this phage contributes to bacterial virulence.

    Beschreibung: The migration of methane through the gas hydrate stability zone (GHSZ) in the marine subsurface is characterized by highly dynamic reactive transport processes coupled to thermodynamic phase transitions between solid gas hydrates, free methane gas, and dissolved methane in the aqueous phase. The marine subsurface is essentially a water-saturated porous medium where the thermodynamic instability of the hydrate phase can cause free gas pockets to appear and disappear locally, causing the model to degenerate. This poses serious convergence issues for the general-purpose nonlinear solvers (e.g., standard Newton), and often leads to extremely small time-step sizes. The convergence problem is particularly severe when the rate of hydrate phase change is much lower than the rate of gas dissolution. In order to overcome this numerical challenge, we have developed an all-at-once Newton scheme tailored to our gas hydrate model, which can handle rate-based hydrate phase change coupled with equilibrium gas dissolution in a mathematically consistent and robust manner.

    Beschreibung: Latrunculia sponges represent a rich source of discorhabdin-type pyrroloiminoquinone alkaloids, a few of which comprise a dimeric structure. The anticancer-activity-guided isolation of the n-hexane subextract of the Antarctic deep-sea sponge Latrunculia biformis yielded the known compound (−)-(1R,2R,6R,8S,6'S)-discorhabdin B dimer (1) and two new derivatives, (−)-(1S,2R,6R,8S,6'S)-discorhabdin B dimer (2) and (−)-(1R,2R,6R,8S,6'S)-16',17'-dehydrodiscorhabdin B dimer (3). The chemical structures of compounds 1–3 were elucidated by means of HR-ESIMS, NMR, [], ECD spectroscopy, and a comparison with the previously reported discorhabdin analogs. Compounds 1 and 2 showed significant in vitro anticancer activity against the human colon cancer cell line (HCT-116), with IC50 values of 0.16 and 2.01 µM, respectively. Compared to monomeric discorhabdins, dimeric discorhabdins are very rare in Nature. This study adds two new discorhabdin dimers (2 and 3) to this small pyrroloiminoquinone subfamily. This is also the first report of compound 1 as a natural product and the first assessment of its in vitro anticancer activity.

    Beschreibung: Sea foam harbors a diverse range of fungal spores with biological and ecological relevance in marine environments. Fungi are known as the producers of secondary metabolites that are used in health and agricultural sectors, however the potentials of sea foam-derived fungi have remained unexplored. In this study, organic extracts of six foam-derived fungal isolates belonging to the genera Penicillium, Cladosporium, Emericellopsis and Plectosphaerella were investigated for their antimicrobial activity against plant and human pathogens and anticancer activity. In parallel, an untargeted metabolomics study using UPLC-QToF–MS/MS-based molecular networking (MN) was performed to unlock their chemical inventory. Penicillium strains were identified as the most prolific producers of compounds with an average of 165 parent ions per strain. In total, 49 known mycotoxins and functional metabolites were annotated to specific and ubiquitous parent ions, revealing considerable chemical diversity. This allowed the identification of putative new derivatives, such as a new analog of the antimicrobial tetrapeptide, fungisporin. Regarding bioactivity, the Penicillium sp. isolate 31.68F1B showed a strong and broad-spectrum activity against seven plant and human pathogens, with the phytopathogen Magnaporthe oryzae and the human pathogen Candida albicans being the most susceptible (IC50 values 2.2 and 6.3 µg/mL, respectively). This is the first study mining the metabolome of the sea foam-derived fungi by MS/MS-based molecular networking, and assessing their biological activities against phytopathogens

    Beschreibung: This paper presents the technological developments and the policy contexts for the project “Autonomous Robotic Sea-Floor Infrastructure for Bentho-Pelagic Monitoring” (ARIM). The development is based on the national experience with robotic component technologies that are combined and merged into a new product for autonomous and integrated ecological deep-sea monitoring. Traditional monitoring is often vessel-based and thus resource demanding. It is economically unviable to fulfill the current policy for ecosystem monitoring with traditional approaches. Thus, this project developed platforms for bentho-pelagic monitoring using an arrangement of crawler and stationary platforms at the Lofoten-Vesterålen (LoVe) observatory network (Norway). Visual and acoustic imaging along with standard oceanographic sensors have been combined to support advanced and continuous spatial-temporal monitoring near cold water coral mounds. Just as important is the automatic processing techniques under development that have been implemented to allow species (or categories of species) quantification (i.e., tracking and classification). At the same time, real-time outboard processed three-dimensional (3D) laser scanning has been implemented to increase mission autonomy capability, delivering quantifiable information on habitat features (i.e., for seascape approaches). The first version of platform autonomy has already been tested under controlled conditions with a tethered crawler exploring the vicinity of a cabled stationary instrumented garage. Our vision is that elimination of the tether in combination with inductive battery recharge trough fuel cell technology will facilitate self-sustained long-term autonomous operations over large areas, serving not only the needs of science, but also sub-sea industries like subsea oil and gas, and mining.

    Beschreibung: The Thessaloniki Bay is a eutrophic coastal area which has been characterized in recent years by frequent and intense phytoplankton blooms and red tides. The aim of the study was to investigate the underexplored diversity of marine unicellular eukaryotes in four different sampling sites in Thessaloniki Bay during a year of plankton blooms, red tides, and mucilage aggregates. High-Throughput Sequencing (HTS) was applied in extracted DNA from weekly water samples targeting the 18S rRNA gene. In almost all samples, phytoplankton blooms and/or red tides and mucilage aggregates were observed. The metabarcoding analysis has detected the known unicellular eukaryotic groups frequently observed in the Bay, dominated by Bacillariophyta and Dinoflagellata, and revealed taxonomic groups previously undetected in the study area (MALVs, MAST, and Cercozoa). The dominant OTUs were closely related to species known to participate in red tides, harmful blooms, and mucilage aggregates. Other OTUs, present also during the blooms in low abundance (number of reads), were closely related to known harmful species, suggesting the occurrence of rare taxa with potential negative impacts on human health not detectable with classical microscopy. Overall, the unicellular eukaryote assemblages showed temporal patterns rather than small-scale spatial separation responding to the variability of physical and chemical factors.

    Beschreibung: Metal pollution is pervasive across terrestrial and aquatic ecosystems owing to anthropogenic activities. Sediments can accrue high concentrations of metals and act as secondary sources, and thus may be valuable indicators of metal contamination across spatiotemporal scales. In aquatic systems, the extent of metal pollution may be further mediated by transference among sediments and living organisms, with plant metal contaminants potentially predictive of underlying sediment concentrations. The present study thus quantifies the extent of metal pollutants (Na, K, Ca, Mg, Cu, Zn, Mn, B, Fe) across multiple study sites and seasons (cool-dry, hot-wet, hot-dry) in a subtropical river system. Furthermore, uptake by a key macrophyte species, Phragmites australis, was examined and correlated with sediment pollution levels among different plant parts. Overall, sediment pollution load indices differed seasonally, being significantly highest during the cool-dry season irrespective of sampling location, suggesting that periods with reduced water flows can exacerbate metal pollution levels in riverine sediments. Also, metal concentrations were highest in upstream wetland sites, indicating a capacity for metal sink effects in these areas. Overall, macrophytes contained high concentrations of select metals, however composition and concentrations differed across plant parts, with roots containing particularly high concentrations of Fe and B. Correlations between sediment and macrophyte concentrations were mostly non-significant, whilst stem Mn and Fe concentrations correlated significantly negatively and positively to sediment concentrations, respectively. The present study identifies key spatiotemporal differences in multiple metal contaminants in an understudied subtropical aquatic system that align with hydrological regime differences. Whilst macrophytes were not found to be major accumulators, or predictors, of metal contaminants in this study, they may collectively play a central role in concentration regulation in aquatic systems.

    Beschreibung: Cyanobacterial blooms are the most important and best studied type of harmful algal blooms in fresh waters and brackish coastal seas. We here review how and to which extent they resist grazing by zooplankton, how zooplankton responds to cyanobacterial blooms and how these effects are further transmitted to fish. Size, toxicity and poor nutritional value are widespread mechanisms of grazing defense by cyanobacteria. In some cases, defenses are inducible, in some they are obligate. However, to some extent zooplankton overcome grazing resistance, partly after evolutionary adaptation. Cyanotoxins are also harmful to fish and may cause fish kills. However, some fish species feed on Cyanobacteria, are able to reduce their abundance, and grow on a cyanobacterial diet. While reduced edibility for crustacean zooplankton tends to elongate the food chain from primary producers to fish, direct feeding by fish tends to shorten it. The few available comparative studies relating fish yield to nutrients or phytoplankton provide no indication that cyanobacteria should reduce the ratio fish production: primary production.

    Beschreibung: Brown alga Bifurcaria bifurcata is a prolific source of bioactive acyclic (linear) diterpenes with high structural diversity. In the continuation of our investigations on Irish brown algae, we undertook an in-depth chemical study on the n-hexanes and chloroform subextracts of B. bifurcata that led to isolation of six new (1–6) and two known (7–8) acyclic diterpenes. Chemical structures of the compounds were elucidated by a combination of 1D and 2D NMR, HRMS, FT-IR, [α]D and vibrational circular dichroism (VCD) spectroscopy. Compounds 1–8, as well as three additional linear diterpenes (9–11), which we isolated from the same seaweed before, were tested against the human breast cancer cell line (MDA-MB-231). Several compounds moderately inhibited the growth of the MDA-MB-231 cell line with IC50 values ranging from 11.6 to 32.0 μg/mL. The present study carried out on the lipophilic extracts of the Irish B. bifurcata shows the enormous capacity of this seaweed to produce a large palette of acyclic diterpenes with diverse oxygenation and substitution patterns and promising bioactivities.

    Beschreibung: In the last few decades, numerous studies have investigated the impacts of simulated ocean acidification on marine species and communities, particularly those inhabiting dynamic coastal systems. Despite these research efforts, there are many gaps in our understanding, particularly with respect to physiological mechanisms that lead to pathologies. In this review, we trace how carbonate system disturbances propagate from the coastal environment into marine invertebrates and highlight mechanistic links between these disturbances and organism function. We also point toward several processes related to basic invertebrate biology that are severely understudied and prevent an accurate understanding of how carbonate system dynamics influence organismic homeostasis and fitness-related traits. We recommend that significant research effort be directed to studying cellular phenotypes of invertebrates acclimated or adapted to elevated seawater pCO2 using biochemical and physiological methods.

    Beschreibung: The on-board identification of ore minerals during a cruise is often postponed until long after the cruise is over. During the M127 cruise, 21 cores with deep-seafloor sediments were recovered in the Trans-Atlantic Geotraverse (TAG) field along the Mid Atlantic Ridge (MAR). Sediments were analyzed on-board for physicochemical properties such as lightness (L*), pH and Eh. Selected samples were studied for mineral composition by X-ray powder diffraction (XRD). Based on XRD data, sediment samples were separated into high-, low- and non-carbonated. Removal of carbonates is a common technique in mineralogical studies in which HCl is used as the extraction agent. In the present study, sequential extraction was performed with sodium acetate buffer (pH 5.0) to remove carbonates. The ratio between the highest calcite XRD reflection in the original samples (Iorig) vs its XRD-reflection in samples after their treatment with the buffer (Itreat) was used as a quantitative parameter of calcite removal, as well as to identify minor minerals in carbonated samples (when Iorig/Itreat 〉 24). It was found that the lightness parameter (L*) showed a positive correlation with calcite XRD reflection in selected TAG samples, and this could be applied to the preliminary on-board determination of extraction steps with acetate buffer (pH 5.0) in carbonated sediment samples. The most abundant minerals detected in carbonated samples were quartz and Al- and Fe-rich clays. Other silicates were also detected (e.g., calcic plagioclase, montmorillonite, nontronite). In non-carbonated samples, Fe oxides and hydroxides (goethite and hematite, respectively) were detected. Pyrite was the dominant hydrothermal mineral and Cu sulfides (chalcopyrite, covellite) and hydrothermal Mn oxides (birnessite and todorokite) were mineral phases identified in few samples, whereas paratacamite was detected in the top 20 cm of the core. The present study demonstrates that portable XRD analysis makes it possible to characterize mineralogy at cored sites, in particular in both low- and high-carbonated samples, before the end of most cruises, thus enabling the quick modification of exploration strategies in light of new information as it becomes available in near-real time.

    Beschreibung: The brown alga Fucus vesiculosus is common to the intertidal zones of the Baltic Sea, where it is exposed to high fouling pressures by microorganisms. Our previous studies showed, repeatedly, the consistent antimicrobial activity of F. vesiculosus crude extracts against human pathogens, while untargeted metabolomics analyses have revealed a variety of metabolites. In this study, we applied the UPLC-QToF-MS/MS-based “bioactive molecular networking” (BMN) concept on the most bioactive n-hexane and n-butanol subextracts of Baltic F. vesiculosus coupled with in silico dereplication tools to identify the compounds responsible for antimicrobial activity. The first antimicrobial cluster identified by BMN was galactolipids. Our targeted isolation efforts for this class led to the isolation of six monogalactosyldiacylglycerol (MGDG) derivatives (1–6) and one digalactosyldiacylglycerol (DGDG, 7). The MGDGs 5 and 6 and the DGDG 7 exhibited activity against Staphylococcus aureus. The second compound class with high bioactivity was phlorotannins. In particular, phlorethol-type phlorotannins showed high correlations with antimicrobial activity based on the BMN approach, and two phlorotannins (8–9) were isolated. This study shows that antimicrobial components of F. vesiculosus reside in the algal cell walls and membranes and that BMN provides a complementary tool for the targeted isolation of bioactive metabolites.

    Beschreibung: Plastic pollution is a major global issue and its impacts on ecosystems and socioeconomic sectors lack comprehensive understanding. The integration of plastics issues into the educational system of both primary and secondary schools has often been overlooked, especially in Africa, presenting a major challenge to environmental awareness. Owing to the importance of early age awareness, this study aims to investigate whether plastic pollution issues are being integrated into South African primary and secondary education school curriculums. Using face-to-face interviews with senior educators, we address this research problem by investigating (i) the extent to which teachers cover components of plastic pollution, and (ii) educator understandings of plastic pollution within terrestrial and aquatic environments. The results indicate that plastic pollution has been integrated into the school curriculum in technology, natural science, geography, life science, life skills and life orientation subjects. However, there was a lack of integration of management practices for plastics littering, especially in secondary schools, and understanding of dangers among different habitat types. This highlights the need for better educational awareness on the plastic pollution problem at both primary and secondary school level, with increased environmental programs needed to educate schools on management practices and impacts

    Beschreibung: In this work, we present MorphoCluster, a software tool for data-driven, fast, and accurate annotation of large image data sets. While already having surpassed the annotation rate of human experts, volume and complexity of marine data will continue to increase in the coming years. Still, this data requires interpretation. MorphoCluster augments the human ability to discover patterns and perform object classification in large amounts of data by embedding unsupervised clustering in an interactive process. By aggregating similar images into clusters, our novel approach to image annotation increases consistency, multiplies the throughput of an annotator, and allows experts to adapt the granularity of their sorting scheme to the structure in the data. By sorting a set of 1.2 M objects into 280 data-driven classes in 71 h (16 k objects per hour), with 90% of these classes having a precision of 0.889 or higher. This shows that MorphoCluster is at the same time fast, accurate, and consistent; provides a fine-grained and data-driven classification; and enables novelty detection.

    Beschreibung: Imaging technologies are being deployed on cabled observatory networks worldwide. They allow for the monitoring of the biological activity of deep-sea organisms on temporal scales that were never attained before. In this paper, we customized Convolutional Neural Network image processing to track behavioral activities in an iconic conservation deep-sea species—the bubblegum coral Paragorgia arborea—in response to ambient oceanographic conditions at the Lofoten-Vesterålen observatory. Images and concomitant oceanographic data were taken hourly from February to June 2018. We considered coral activity in terms of bloated, semi-bloated and non-bloated surfaces, as proxy for polyp filtering, retraction and transient activity, respectively. A test accuracy of 90.47% was obtained. Chronobiology-oriented statistics and advanced Artificial Neural Network (ANN) multivariate regression modeling proved that a daily coral filtering rhythm occurs within one major dusk phase, being independent from tides. Polyp activity, in particular extrusion, increased from March to June, and was able to cope with an increase in chlorophyll concentration, indicating the existence of seasonality. Our study shows that it is possible to establish a model for the development of automated pipelines that are able to extract biological information from times series of images. These are helpful to obtain multidisciplinary information from cabled observatory infrastructures.

    Beschreibung: Marine algae represent a prolific source of filamentous fungi for bioprospecting. In continuation of our search for new anticancer leads from fungi derived from the brown alga Fucus vesiculosus, an endophytic Pyrenochaetopsis sp. FVE-001 was selected for an in-depth chemical analysis. The crude fungal extract inhibited several cancer cell lines in vitro, and the highest anticancer activity was tracked to its CHCl3–soluble portion. A bioactivity-based molecular networking approach was applied to C18-SPE fractions of the CHCl3 subextract to predict the bioactivity scores of metabolites in the fractions and to aid targeted purification of anticancer metabolites. This approach led to a rapid isolation of three new decalinoylspirotetramic acid derivatives, pyrenosetins A–C (1–3) and the known decalin tetramic acid phomasetin (4). The structures of the compounds were elucidated by extensive NMR, HR-ESIMS, FT-IR spectroscopy, [α]D and Mosher’s ester method. Compounds 1 and 2 showed high anticancer activity against malignant melanoma cell line A-375 (IC50 values 2.8 and 6.3 μM, respectively), in line with the bioactivity predictions. This is the first study focusing on secondary metabolites of a marine-derived Pyrenochaetopsis sp. and the second investigation performed on the member of the genus Pyrenochaetopsis.

    Beschreibung: Electromagnetic (EM) geophysical methods are well equipped to distinguish electrical resistivity contrasts between freshwater-saturated and seawater-saturated formations. Beneath the semi-arid, rapidly urbanizing island of Malta, offshore groundwater is an important potential resource but it is not known whether the regional mean sea-level aquifer (MSLA) extends offshore. To address this uncertainty, land-based alongshore and across-shore time-domain electromagnetic (TDEM) responses were acquired with the G-TEM instrument (Geonics Ltd., Mississauga, ON, Canada) and used to map the onshore structure of the aquifer. 1-D inversion results suggest that the onshore freshwater aquifer resides at 4–24 m depth, underlain by seawater-saturated formations. The freshwater aquifer thickens with distance from the coastline. We present 2D and 3D electromagnetic forward modeling based on finite-element (FE) analysis to further constrain the subsurface geometry of the onshore freshwater body. We interpret the high resistivity zones that as brackish water-saturated bodies are associated with the mean sea-level aquifer. Generally, time-domain electromagnetic (TDEM) results provide valuable onshore hydrogeological information, which can be augmented with marine and coastal transition-zone measurements to assess potential hydraulic continuity of terrestrial aquifers extending offshore.

    Beschreibung: The geographic position, highly fluctuating sea temperatures and hypersalinity make Persian Gulf an extreme environment. Although this unique environment has high biodiversity dominated by invertebrates, its potential in marine biodiscovery has largely remained untapped. Herein, we aimed at a detailed analysis of the metabolome and bioactivity profiles of the marine sponge Axinella sinoxea collected from the northeast coast of the Persian Gulf in Iran. The crude extract and its Kupchan subextracts were tested in multiple in-house bioassays, and the crude extract and its CHCl3-soluble portion showed in vitro antibacterial activity against Methicillin-resistant Staphylococcus aureus (MRSA) and Enterococcus faecium (Efm). A molecular networking (MN)-based dereplication strategy by UPLC-MS/MS revealed the presence of phospholipids and steroids, while 1H NMR spectroscopy indicated the presence of additional metabolites, such as diketopiperazines (DKPs). Integrated MN and 1H NMR analyses on both the crude and CHCl3 extracts combined with an antibacterial activity-guided isolation approach afforded eight metabolites: a new diketopiperazine, (-)-cyclo(L-trans-Hyp-L-Ile) (8); a known diketopiperazine, cyclo(L-trans-Hyp-L-Phe) (7); two known phospholipids, 1-O-hexadecyl-sn-glycero-3-phosphocholine (1) and 1-O-octadecanoyl-sn-glycero-3-phosphocholine (2); two known steroids, 3β-hydroxycholest-5-ene-7,24-dione (3) and (22E)-3β-hydroxycholesta-5,22-diene-7,24-dione (4); two known monoterpenes, loliolide (5) and 5-epi-loliolide (6). The chemical structures of the isolates were elucidated by a combination of NMR spectroscopy, HRMS and [α]D analyses. All compounds were tested against MRSA and Efm, and compound 3 showed moderate antibacterial activity against MRSA (IC50 value 70 μg/mL). This is the first study that has dealt with chemical and bioactivity profiling of A. sinoxea leading to isolation and characterization of pure sponge metabolites.

    Beschreibung: Leaf litter contributes to the functioning of aquatic ecosystems through allochthonous inputs of carbon, nitrogen, and other elements. Here, we examine leaf litter nutrient inputs and decomposition associated with four plant species using a mesocosm approach. Native sycamore fig Ficus sycomorus L., and silver cluster–leaf Terminalia sericea Burch. ex DC. decomposition dynamics were compared to invasive tickberry Lantana camara L. and guava Psidium guajava L., whereby phosphate, nitrate, nitrite, silicate, and ammonium releases were quantified over time. Leaf inputs significantly reduced pH, with reductions most marked by invasive L. camara. Conductivity was heightened by all leaf input treatments, except native T. sericea. Leaf inputs significantly affected all nutrient levels monitored in the water over time, except for silicate. In particular, leaf litter from invasive L. camara drove significantly increased nutrient concentrations compared to other native plant species, whilst effects of invasive P. guajava were less statistically clear. The end weights of the leaf litter demonstrated decomposition differences among the species types, following a decreasing order of P. guajava 〉 T. sericea 〉 F. sycomorus 〉 L. camara, further suggesting high organic inputs from invasive L. camara. The study results highlight that differential leaf litter decomposition rates of four plant species can play a significant role in nutrient release, in turn altering aquatic ecosystem productivity. However, these effects likely depend on species-specific differences, rather than between invasive–native species generally. Shifting terrestrial plant communities may alter aquatic community composition, but specific effects are likely associated with leaf traits

    Beschreibung: Terrestrial plants are known to “garden” the microbiota of their rhizosphere via released metabolites (that can attract beneficial microbes and deter pathogenic microbes). Such a “gardening” capacity is also known to be dynamic in plants. Although microbial “gardening” has been recently demonstrated for seaweeds, we do not know whether this capacity is a dynamic property in any aquatic flora like in terrestrial plants. Here, we tested the dynamic microbial “gardening” capacity of seaweeds using the model invasive red seaweed Agarophyton vermiculophyllum. Following an initial extraction of surface-associated metabolites (immediately after field collection), we conducted a long-term mesocosm experiment for 5 months to test the effect of two different salinities (low = 8.5 and medium = 16.5) on the microbial “gardening” capacity of the alga over time. We tested “gardening” capacity of A. vermiculophyllum originating from two different salinity levels (after 5 months treatments) in settlement assays against three disease causing pathogenic bacteria and seven protective bacteria. We also compared the capacity of the alga with field-collected samples. Abiotic factors like low salinity significantly increased the capacity of the alga to deter colonization by pathogenic bacteria while medium salinity significantly decreased the capacity of the alga over time when compared to field-collected samples. However, capacity to attract beneficial bacteria significantly decreased at both tested salinity levels when compared to field-collected samples. Dynamic microbial “gardening” capacity of a seaweed to attract beneficial bacteria and deter pathogenic bacteria is demonstrated for the first time. Such a dynamic capacity as found in the current study could also be applicable to other aquatic host–microbe interactions. Our results may provide an attractive direction of research towards manipulation of salinity and other abiotic factors leading to better defended A. vermiculophyllum towards pathogenic bacteria thereby enhancing sustained production of healthy A. vermiculophyllum in farms

    Beschreibung: Ascidians and their associated microbiota are prolific producers of bioactive marine natural products. Recent culture-independent studies have revealed that the tunic of the solitary ascidian Ciona intestinalis (sea vase) is colonized by a diverse bacterial community, however, the biotechnological potential of this community has remained largely unexplored. In this study, we aimed at isolating the culturable microbiota associated with the tunic of C. intestinalis collected from the North and Baltic Seas, to investigate their antimicrobial and anticancer activities, and to gain first insights into their metabolite repertoire. The tunic of the sea vase was found to harbor a rich microbial community, from which 89 bacterial and 22 fungal strains were isolated. The diversity of the tunic-associated microbiota differed from that of the ambient seawater samples, but also between sampling sites. Fungi were isolated for the first time from the tunic of Ciona. The proportion of bioactive extracts was high, since 45% of the microbial extracts inhibited the growth of human pathogenic bacteria, fungi or cancer cell lines. In a subsequent bioactivity-and metabolite profiling-based approach, seven microbial extracts were prioritized for in-depth chemical investigations. Untargeted metabolomics analyses of the selected extracts by a UPLC-MS/MS-based molecular networking approach revealed a vast chemical diversity with compounds assigned to 22 natural product families, plus many metabolites that remained unidentified. This initial study indicates that bacteria and fungi associated with the tunic of C. intestinalis represent an untapped source of putatively new marine natural products with pharmacological relevance.

    Beschreibung: Here we present the first radiometric age and geochemical (major and trace element and isotope) data for samples from the Hydrographer Ridge, a back arc volcano of the Kurile Island Arc, and a newly discovered chain of volcanoes (“Sonne Volcanoes”) on the northwestern continental slope of the Kurile Basin on the opposite side of the arc. The 40Ar/39Ar age and geochemical data show that Hydrographer Ridge (3.2–3.3 Ma) and the “Sonne Volcanoes” (25.3–25.9 Ma) have very similar trace element and isotope characteristics to those of the Kurile Island Arc, indicating derivation from a common magma source. We conclude that the age of the “Sonne Volcanoes” marks the time of opening of the Kurile Basin, implying slow back arc spreading rates of 1.3–1.8 cm/y. Combined with published data from the Kurile fore arc, our data suggest that the processes of subduction, Kurile Basin opening and frontal arc extension occurred synchronously and that extension in the rear part and in the frontal part of the Kurile Island Arc must have been triggered by the same mechanism.

    Beschreibung: Massive fouling by the invasive ascidian Ciona intestinalis in Prince Edward Island (PEI, Canada) has been causing devastating losses to the local blue mussel farms. In order to gain first insights into so far unexplored factors that may contribute to the invasiveness of C. intestinalis in PEI, we undertook comparative microbiome and metabolome studies on specific tissues from C. intestinalis populations collected in invaded (PEI) and native regions (Helgoland and Kiel, Germany). Microbial community analyses and untargeted metabolomics revealed clear location- and tissue-specific patterns showing that biogeography and the sampled tissue shape the microbiome and metabolome of C. intestinalis. Moreover, we observed higher microbial and chemical diversity in C. intestinalis from PEI than in the native populations. Bacterial OTUs specific to C. intestinalis from PEI included Cyanobacteria (e.g., Leptolyngbya sp.) and Rhodobacteraceae (e.g., Roseobacter sp.), while populations from native sampling sites showed higher abundances of e.g., Firmicutes (Helgoland) and Epsilonproteobacteria (Kiel). Altogether 121 abundant metabolites were putatively annotated in the global ascidian metabolome, of which 18 were only detected in the invasive PEI population (e.g., polyketides and terpenoids), while six (e.g., sphingolipids) or none were exclusive to the native specimens from Helgoland and Kiel, respectively. Some identified bacteria and metabolites reportedly possess bioactive properties (e.g., antifouling and antibiotic) that may contribute to the overall fitness of C. intestinalis. Hence, this first study provides a basis for future studies on factors underlying the global invasiveness of Ciona species

    Beschreibung: The Atlantic Multidecadal Oscillation (AMO) is coherently linked to climate variations over many parts of the globe. Despite recent achievements, the mechanism by which the AMO influences regional precipitation over South America is not well understood. In this study, we isolate the atmospheric response to the AMO using a water isotope-enabled version of the Community Earth System Model version 1.2 (iCESM1.2) and determine its influence on (sub)tropical South American regional precipitation and atmospheric circulation. The results suggest an interhemispheric seesaw in Hadley circulation strength and that the section of the Atlantic Hadley cell is marked by a stronger upward air component south of the equator during the cold AMO phase. We also find that the precipitation anomalies over (sub)tropical South America during AMO phases are mainly related to changes in the Atlantic Intertropical Convergence Zone (ITCZ) core strength, where in the cold (warm) AMO phase the core region strengthens (weakens) from February to July, while from July to November the core region weakens (strengthens). Our results stress the importance of acknowledging the dynamics of season- and regional-dependent ITCZ responses, as they are sufficient to produce observed AMO-related signals even in the absence of marked changes in the ITCZ position.

    Beschreibung: Military munitions from World War I and II dumped at the seafloor are a threat to the marine environment and its users. Decades of saltwater exposure make the explosives fragile and difficult to dispose of. If required, the munition is blast-in-place. In August 2019, 42 ground mines were detonated in a controlled manner underwater during a NATO maneuver in the German Natura2000 Special Area of Conservation Fehmarn Belt, the Baltic Sea. In June 2020, four detonation craters were investigated with a multibeam echosounder for the first time. This dataset is represented here as maps of bathymetry, slope angle, and height difference to the surrounding. The circular craters were still clearly visible a year after the detonation. The diameter and depth of the structures were between 7.5–12.6 m and 0.7–2.2 m, respectively. In total, about 321 m2 of the seafloor was destroyed along the track line.

    Beschreibung: It is widely accepted that the commensal gut microbiota contributes to the health and well-being of its host. The solitary tunicate Ciona intestinalis emerges as a model organism for studying host–microbe interactions taking place in the gut, however, the potential of its gut-associated microbiota for marine biodiscovery remains unexploited. In this study, we set out to investigate the diversity, chemical space, and pharmacological potential of the gut-associated microbiota of C. intestinalis collected from the Baltic and North Seas. In a culture-based approach, we isolated 61 bacterial and 40 fungal strains affiliated to 33 different microbial genera, indicating a rich and diverse gut microbiota dominated by Gammaproteobacteria. In vitro screening of the crude microbial extracts indicated their antibacterial (64% of extracts), anticancer (22%), and/or antifungal (11%) potential. Nine microbial crude extracts were prioritized for in-depth metabolome mining by a bioactivity- and chemical diversity-based selection procedure. UPLC-MS/MS-based metabolomics combining automated (feature-based molecular networking and in silico dereplication) and manual approaches significantly improved the annotation rates. A high chemical diversity was detected where peptides and polyketides were the predominant classes. Many compounds remained unknown, including two putatively novel lipopeptides produced by a Trichoderma sp. strain.     Beschreibung: The main objective of the present study was to assess the effects of sulfur (S) nutrition on plant growth, overall quality, secondary metabolites, and antibacterial and radical scavenging activities of hydroponically grown lettuce cultivars. Three lettuce cultivars, namely, Pazmanea RZ (green butterhead, V1), Hawking RZ (green multi-leaf lettuce, V2), and Barlach RZ (red multi-leaf, V3) were subjected to two S-treatments in the form of magnesium sulfate (+S) or magnesium chloride (−S). Significant differences were observed under −S treatments, especially among V1 and V2 lettuce cultivars. These responses were reflected in the yield, levels of macro- and micro-nutrients, water-soluble sugars, and free inorganic anions. In comparison with the green cultivars (V1 and V2), the red-V3 cultivar revealed a greater acclimation to S starvation, as evidenced by relative higher plant growth. In contrast, the green cultivars showed higher capabilities in production and superior quality attributes under +S condition. As for secondary metabolites, sixteen compounds (e.g., sesquiterpene lactones, caffeoyl derivatives, caffeic acid hexose, 5-caffeoylquinic acid (5-OCQA), quercetin and luteolin glucoside derivatives) were annotated in all three cultivars with the aid of HPLC-DAD-MS-based untargeted metabolomics. Sesquiterpene lactone lactucin and anthocyanin cyanidin 3-O-galactoside were only detected in V1 and V3 cultivars, respectively. Based on the analyses, the V3 cultivar was the most potent radical scavenger, while V1 and V2 cultivars exhibited antibacterial activity against Staphylococcus aureus in response to S provision. Our study emphasizes the critical role of S nutrition in plant growth, acclimation, and nutritional quality. The judicious-S application can be adopted as a promising antimicrobial prototype for medical applications.

    Beschreibung: Six-degree-of-freedom (6DoF) sensors measure translation along three axes and rotation around three axes. These collocated measurements make it possible to fully describe building motion without the need for an external reference point. This is an advantage for building health monitoring, which uses interstory drift and building eigenfrequencies to monitor stability. In this paper, IMU50 6DoF sensors are characterized to determine their suitability for building health monitoring. The sensors are calibrated using step table methods and by comparison with earth’s rotation and gravity. These methods are found to be comparable. The sensor’s self-noise is examined through the power spectral density and the Allan deviation of data recorded in a quiet environment. The effect of temperature variation is tested between 14 and 50 ∘C. It appears that the self-noise of the rotation components increases while the self-noise of the acceleration components decreases with temperature. The comparison of the sensor self-noise with ambient building signal and higher amplitude shaking shows that these sensors are in general not sensitive enough for ambient signal building health monitoring in the frequency domain, but could be useful for monitoring interstory drift and building motion during, for example, strong earthquake shaking in buildings similar to those examined here.

    Beschreibung: The Persian Gulf is a unique and biologically diverse marine environment dominated by invertebrates. In continuation of our research interest in the chemistry and biological activity of marine sponges from the Persian Gulf, we selected the excavating sponge Cliona celata for detailed metabolome analyses, in vitro bioactivity screening, and chemical isolation studies. A UPLC-MS/MS (MS2) molecular-networking-based dereplication strategy allowed annotation and structural prediction of various diketopiperazines (DKPs) and etzionin-type diketopiperazine hydroxamates (DKPHs) in the crude sponge extract. The molecular-networking-guided isolation approach applied to the crude extract afforded the DKPH etzionin (1) and its two new derivatives, clioetzionin A (2) and clioetzionin B (3). Another new modified DKP (4) was identified by MS/MS analyses but could not be isolated in sufficient quantities to confirm its structure. The chemical characterization of the purified DKPHs 1–3 was performed by a combination of 1D and 2D NMR spectroscopy, HRMS, HRMS/MS, and [α]D analyses. Compounds 1 and 2 exhibited broad antibacterial, antifungal, and anticancer activities, with IC50 values ranging from 19.6 to 159.1 µM. This is the first study investigating the chemical constituents of a C. celata specimen from the Persian Gulf. It is also the first report of full spectroscopic data of etzionin based on extensive spectroscopic analyses

    Beschreibung: Ocean warming and acidification pose serious threats to cold-water corals (CWCs) and the surrounding habitat. Yet, little is known about the role of natural short-term and seasonal environmental variability, which could be pivotal to determine the resilience of CWCs in a changing environment. Here, we provide continuous observational data of the hydrodynamic regime (recorded using two benthic landers) and point measurements of the carbonate and nutrient systems from five Lophelia pertusa reefs in the Langenuen Fjord, southwestern Norway, from 2016 to 2017. In this fjord setting, we found that over a tidal (〈24 h) cycle during winter storms, the variability of measured parameters at CWC depths was comparable to the intra-annual variability, demonstrating that single point measurements are not sufficient for documenting (and monitoring) the biogeochemical conditions at CWC sites. Due to seasonal and diurnal forcing, parts of the reefs experienced temperatures up to 4 °C warmer (i.e., 〉12 °C) than the mean conditions and high CT concentrations of 20 µmol kg−1 over the suggested threshold for healthy CWC reefs (i.e., 〉2170 µmol kg−1). Combined with hindcast measurements, our findings indicate that these shallow fjord reefs may act as an early hotspot for ocean warming and acidification. We predict that corals in Langenuen will face seasonally high temperatures (〉18 °C) and hypoxic and corrosive conditions within this century. Therefore, these fjord coral communities could forewarn us of the coming consequences of climate change on CWC diversity and function.

    Beschreibung: Organic matter (OM) plays a significant role in the formation of oxygen minimum zones (OMZs) and associated biogeochemical cycling. OM supply processes to the OMZ include physical transport, particle formation, and sinking as well as active transport by migrating zooplankton and nekton. In addition to the availability of oxygen and other electron acceptors, the remineralization rate of OM is controlled by its biochemical quality. Enhanced microbial respiration of OM can induce anoxic microzones in an otherwise oxygenated water column. Reduced OM degradation under low-oxygen conditions, on the other hand, may increase the CO2 storage time in the ocean. Understanding the interdependencies between OM and oxygen cycling is of high relevance for an ocean facing deoxygenation as a consequence of global warming. In this review, we describe OM fluxes into and cycling within two large OMZs associated with eastern boundary upwelling systems that differ greatly in the extent of oxygen loss: the highly oxygen-depleted OMZ in the tropical South Pacific and the moderately hypoxic OMZ in the tropical North Atlantic. We summarize new findings from a large German collaborative research project, Collaborative Research Center 754 (SFB 754), and identify knowledge gaps and future research priorities.

    Beschreibung: Previous studies with Baltic Sea phytoplankton combining elevated seawater temperature with CO2 revealed the importance of size trait‐based analyses, in particular dividing the plankton into edible (〉5 and 〈100 μm) and inedible ([removed]100 μm) size classes for mesozoopankton grazers. While the edible phytoplankton responded predominantly negative to warming and the inedible group stayed unaffected or increased, independent from edibility most phytoplankton groups gained from CO2. Because the ratio between edible and inedible taxa changes profoundly over seasons, we investigated if community responses can be predicted according to the prevailing composition of edible and inedible groups. We experimentally explored the combined effects of elevated temperatures and CO2 concentrations on a late‐summer Baltic Sea community. Total phytoplankton significantly increased in response to elevated CO2 in particular in combination with temperature, driven by a significant gain of the inedible 〈5 μm fraction and large filamentous cyanobacteria. Large flagellates disappeared. The edible group was low as usual in summer and decreased with both factors due to enhanced copepod grazing and overall decline of small flagellates. Our results emphasize that the responses of summer communities are complex, but can be predicted by the composition and dominance of size classes and groups.

    Beschreibung: Current trends in the global climate facilitate the displacement of numerous marine species from their native distribution ranges to higher latitudes when facing warming conditions. In this work, we analyzed occurrences of a circumtropical reef fish, the spotfin burrfish, Chilomycterus reticulatus (Linnaeus, 1958), in the Madeira Archipelago (NE Atlantic) between 1898 and 2021. In addition to available data sources, we performed an online survey to assess the distribution and presence of this species in the Madeira Archipelago, along with other relevant information, such as size class and year of the first sighting. In total, 28 valid participants responded to the online survey, georeferencing 119 C. reticulatus sightings and confirming its presence in all archipelago islands. The invasiveness of the species was screened using the Aquatic Species Invasiveness Screening Kit. Five assessments rated the fish as being of medium risk of establishing a local population and becoming invasive. Current temperature trends might have facilitated multiple sightings of this thermophilic species in the Madeira Archipelago. The present study indicates an increase in C. reticulatus sightings in the region. This underlines the need for updated comprehensive information on species diversity and distribution to support informed management and decisions. The spread of yet another thermophilic species in Madeiran waters provides further evidence of an ongoing tropicalization, emphasizing the need for monitoring programs and the potential of citizen science in complementing such programs.

    Beschreibung: Ocean science and hydroacoustic seafloor mapping rely on accurate navigation underwater. By exploiting terrain information provided by a multibeam echosounder system, it is possible to significantly improve map quality. This article presents an algorithm capable of improving map quality and accuracy by aligning consecutive pings to tiles that are matched pairwise. A globally consistent solution is calculated from these matches. The proposed method has the potential to be used online in addition to other navigation solutions, but is mainly targeted for post processing. The algorithm was tested using different parameter settings on an AUV and a ship-based dataset. The ship-based dataset is publicly available as a benchmark. The original accurate navigation serving as a ground truth, alongside trajectories that include an artificial drift, are available. This allows quantitative comparisons between algorithms and parameter settings.

    Beschreibung: Understanding trophic interactions is essential for the prediction and measurement of structure and function in aquatic environments. Communities in these ecosystems may be shaped by variables such as predator diversity, prey density and emergent multiple predator effects (MPEs), which are likely to influence trophic dynamics. In this study, we examined the effect of key predatory fish in floodplain wetlands, namely Oreochromis mossambicus and Enteromius paludinosus, towards Chironomidae prey, using a comparative functional response (FR) approach. We used single predator species as well as intra- and interspecific paired species to contrast FRs under multiple predator scenarios. Attack rate and handling time estimates from single predator FRs were used to predict multiple predators’ feeding rates, which were compared to observe multiple predators’ feeding rates to quantify potential MPEs. From single fish trials, each species displayed a significant Type II FR, characterized by high feeding rates at low prey densities. Oreochromis mossambicus had a steeper (initial slope, i.e., higher attack rate) and higher (asymptote of curve, i.e., shorter handling time and higher maximum feeding rate) FR, whereas E. paludinosus exhibited lower-magnitude FRs (i.e., lower attack rate, longer handling time and lower feeding rate). In multiple predator scenarios, feeding rates were well-predicted by those of single predators, both in conspecific and interspecific pairs, and thus we did not find evidence for antagonistic or synergistic MPEs. Predator–prey interactions in wetland systems can have significant consequences on the structure and dynamics of ecological communities. In turn, this could have destabilizing effects on resources in tropical wetlands. These results, although experimental, help us understand how trophic interaction among conspecific or interspecific fish species in Austral tropical wetlands might influence their aquatic prey species. This will help us to understand food web dynamics better.

    Beschreibung: Seaweed endophytic (algicolous) fungi are talented producers of bioactive natural products. We have previously isolated two strains of the endophytic fungus, Pyrenochaetopsis sp. FVE-001 and FVE-087, from the thalli of the brown alga Fucus vesiculosus. Initial chemical studies yielded four new decalinoylspirotetramic acid derivatives with antimelanoma activity, namely pyrenosetins A–C (1–3) from Pyrenochaetopsis sp. strain FVE-001, and pyrenosetin D (4) from strain FVE-087. In this study, we applied a comparative metabolomics study employing HRMS/MS based feature-based molecular networking (FB MN) on both Pyrenochaetopsis strains. A higher chemical capacity in production of decalin derivatives was observed in Pyrenochaetopsis sp. FVE-087. Notably, several decalins showed different retention times despite the same MS data and MS/MS fragmentation pattern with the previously isolated pyrenosetins, indicating they may be their stereoisomers. FB MN-based targeted isolation studies coupled with antimelanoma activity testing on the strain FVE-087 afforded two new stereoisomers, pyrenosetins E (5) and F (6). Extensive NMR spectroscopy including DFT computational studies, HR-ESIMS, and Mosher’s ester method were used in the structure elucidation of compounds 5 and 6. The 3′R,5′R stereochemistry determined for compound 6 was identical to that previously reported for pyrenosetin C (3), whose stereochemistry was revised as 3′S,5′R in this study. Pyrenosetin E (5) inhibited the growth of human malignant melanoma cells (A-375) with an IC50 value of 40.9 μM, while 6 was inactive. This study points out significant variations in the chemical repertoire of two closely related fungal strains and the versatility of FB MN in identification and targeted isolation of stereoisomers. It also confirms that the little-known fungal genus Pyrenochaetopsis is a prolific source of complex decalinoylspirotetramic acid derivatives.

    Beschreibung: Machine learning spatial modeling is used for mapping the distribution of deep-sea polymetallic nodules (PMN). However, the presence and influence of spatial autocorrelation (SAC) have not been extensively studied. SAC can provide information regarding the variable selection before modeling, and it results in erroneous validation performance when ignored. ML models are also problematic when applied in areas far away from the initial training locations, especially if the (new) area to be predicted covers another feature space. Here, we study the spatial distribution of PMN in a geomorphologically heterogeneous area of the Peru Basin, where SAC of PMN exists. The local Moran’s I analysis showed that there are areas with a significantly higher or lower number of PMN, associated with different backscatter values, aspect orientation, and seafloor geomorphological characteristics. A quantile regression forests (QRF) model is used using three cross-validation (CV) techniques (random-, spatial-, and cluster-blocking). We used the recently proposed “Area of Applicability” method to quantify the geographical areas where feature space extrapolation occurs. The results show that QRF predicts well in morphologically similar areas, with spatial block cross-validation being the least unbiased method. Conversely, random-CV overestimates the prediction performance. Under new conditions, the model transferability is reduced even on local scales, highlighting the need for spatial model-based dissimilarity analysis and transferability assessment in new areas.

    Beschreibung: Serum calcium isotopes (δ44/42Ca) have been suggested as a non-invasive and sensitive Ca balance marker. Quantitative δ44/42Ca changes associated with Ca flux across body compartment barriers relative to the dietary Ca and the correlation of δ44/42CaSerum with bone histology are unknown. We analyzed Ca and δ44/42Ca by mass-spectrometry in rats after two weeks of standard-Ca-diet (0.5%) and after four subsequent weeks of standard- and of low-Ca-diet (0.25%). In animals on a low-Ca-diet net Ca gain was 61 ± 3% and femur Ca content 68 ± 41% of standard-Ca-diet, bone mineralized area per section area was 68 ± 15% compared to standard-Ca-diet. δ44/42Ca was similar in the diets, and decreased in feces and urine and increased in serum in animals on low-Ca-diet. δ44/42CaBone was higher in animals on low-Ca-diet, lower in the diaphysis than the metaphysis and epiphysis, and unaffected by gender. Independent of diet, δ44/42CaBone was similar in the femora and ribs. At the time of sacrifice, δ44/42CaSerum inversely correlated with intestinal Ca uptake and histological bone mineralization markers, but not with Ca content and bone mineral density by µCT. In conclusion, δ44/42CaBone was bone site specific, but mechanical stress and gender independent. Low-Ca-diet induced marked changes in feces, serum and urine δ44/42Ca in growing rats. δ44/42CaSerum inversely correlated with markers of bone mineralization.

    Beschreibung: Corals are globally important calcifiers that exhibit complex responses to anthropogenic warming and acidification. Although coral calcification is supported by high seawater pH, photosynthesis by the algal symbionts of zooxanthellate corals can be promoted by elevated pCO2. To investigate the mechanisms underlying corals’ complex responses to global change, three species of tropical zooxanthellate corals (Stylophora pistillata, Pocillopora damicornis, and Seriatopora hystrix) and one species of asymbiotic cold-water coral (Desmophyllum pertusum, syn. Lophelia pertusa) were cultured under a range of ocean acidification and warming scenarios. Under control temperatures, all tropical species exhibited increased calcification rates in response to increasing pCO2. However, the tropical species’ response to increasing pCO2 flattened when they lost symbionts (i.e., bleached) under the high-temperature treatments—suggesting that the loss of symbionts neutralized the benefit of increased pCO2 on calcification rate. Notably, the cold-water species that lacks symbionts exhibited a negative calcification response to increasing pCO2, although this negative response was partially ameliorated under elevated temperature. All four species elevated their calcifying fluid pH relative to seawater pH under all pCO2 treatments, and the magnitude of this offset (Δ[H+]) increased with increasing pCO2. Furthermore, calcifying fluid pH decreased along with symbiont abundance under thermal stress for the one species in which calcifying fluid pH was measured under both temperature treatments. This observation suggests a mechanistic link between photosymbiont loss (‘bleaching’) and impairment of zooxanthellate corals’ ability to elevate calcifying fluid pH in support of calcification under heat stress. This study supports the assertion that thermally induced loss of photosymbionts impairs tropical zooxanthellate corals’ ability to cope with CO2-induced ocean acidification.

    Beschreibung: Microbial co-cultivation is a promising approach for the activation of biosynthetic gene clusters (BGCs) that remain transcriptionally silent under artificial culture conditions. As part of our project aiming at the discovery of marine-derived fungal agrochemicals, we previously used four phytopathogens as model competitors in the co-cultivation of 21 marine fungal strains. Based on comparative untargeted metabolomics analyses and anti-phytopathogenic activities of the co-cultures, we selected the co-culture of marine Cosmospora sp. with the phytopathogen Magnaporthe oryzae for in-depth chemical studies. UPLC-MS/MS-based molecular networking (MN) of the co-culture extract revealed an enhanced diversity of compounds in several molecular families, including isochromanones, specifically induced in the co-culture. Large scale co-cultivation of Cosmospora sp. and M. oryzae resulted in the isolation of five isochromanones from the whole co-culture extract, namely the known soudanones A, E, D (1-3) and their two new derivatives, soudanones H-I (4-5), the known isochromans, pseudoanguillosporins A and B (6, 7), naphtho-γ-pyrones, cephalochromin and ustilaginoidin G (8, 9), and ergosterol (10). Their structures were established by NMR, HR-ESIMS, FT-IR, electronic circular dichroism (ECD) spectroscopy, polarimetry ([α]D), and Mosher’s ester reaction. Bioactivity assays revealed antimicrobial activity of compounds 2 and 3 against the phytopathogens M. oryzae and Phytophthora infestans, while pseudoanguillosporin A (6) showed the broadest and strongest anti-phytopathogenic activity against Pseudomonas syringae, Xanthomonas campestris, M. oryzae and P. infestans. This is the first study assessing the anti-phytopathogenic activities of soudanones.

    Beschreibung: Two linear proline-rich peptides (1–2), bearing an N-terminal pyroglutamate, were isolated from the marine bacterium Microbacterium sp. V1, associated with the marine sponge Petrosia ficiformis, collected in the volcanic CO2 vents in Ischia Island (South Italy). Peptide production was triggered at low temperature following the one strain many compounds (OSMAC) method. Both peptides were detected together with other peptides (3–8) via an integrated, untargeted MS/MS-based molecular networking and cheminformatic approach. The planar structure of the peptides was determined by extensive 1D and 2D NMR and HR-MS analysis, and the stereochemistry of the aminoacyl residues was inferred by Marfey’s analysis. Peptides 1–8 are likely to arise from Microbacterium V1 tailor-made proteolysis of tryptone. Peptides 1 and 2 were shown to display antioxidant properties in the ferric-reducing antioxidant power (FRAP) assay.

    Beschreibung: Several strains of a Gram-negative, anaerobic photoautotrophic, motile, rod-shaped bacterium, designated as B14B, A-7R, and A-7Y were isolated from biofilms of low-mineralized soda lakes in central Mongolia and Russia (southeast Siberia). They had lamellar stacks as photosynthetic structures and bacteriochlorophyll a as the major photosynthetic pigment. The strains were found to grow at 25–35 °C, pH 7.5–10.2 (optimum, pH 9.0), and with 0–8% (w/v) NaCl (optimum, 0%). In the presence of sulfide and bicarbonate, acetate, butyrate, yeast extract, lactate, malate, pyruvate, succinate, and fumarate promoted growth. The DNA G + C content was 62.9–63.0 mol%. While the 16S rRNA gene sequences confirmed that the new strains belonged to the genus Ectothiorhodospira of the Ectothiorhodospiraceae, comparison of the genome nucleotide sequences of strains B14B, A-7R, and A-7Y revealed that the new isolates were remote from all described Ectothiorhodospira species both in dDDH (19.7–38.8%) and in ANI (75.0–89.4%). The new strains are also genetically differentiated by the presence of a nitric oxide reduction pathway that is lacking from all other Ectiothiorhodospiraceae. We propose to assign the isolates to the new species, Ectothiorhodospira lacustris sp. nov., with the type strain B14BT (=DSM 116064T = KCTC 25542T = UQM 41491T).

    Beschreibung: Marine heatwaves (MHWs) are extreme events characterized by abnormally high sea surface temperatures, and they have significant impacts on marine ecosystems and human society. The rapid and accurate forecasting of MHWs is crucial for preventing and responding to the impacts they can lead to. However, the research on relevant forecasting methods is limited, and a dedicated forecasting system specifically tailored for the South China Sea (SCS) region has yet to be reported. This study proposes a novel forecasting system utilizing U-Net and ConvLSTM models to predict MHWs in the SCS. Specifically, the U-Net model is used to forecast the intensity of MHWs, while the ConvLSTM model is employed to predict the probability of their occurrence. The indication of an MHW relies on both the intensity forecasted by the U-Net model exceeding threshold T and the occurrence probability predicted by the ConvLSTM model surpassing threshold P. Incorporating sensitivity analysis, optimal thresholds for T are determined as 0.9 °C, 0.8 °C, 1.0 °C, and 1.0 °C for 1-, 3-, 5-, and 7-day forecast lead times, respectively. Similarly, optimal thresholds for P are identified as 0.29, 0.30, 0.20, and 0.28. Employing these thresholds yields the highest forecast accuracy rates of 0.92, 0.89, 0.88, and 0.87 for the corresponding forecast lead times. This innovative approach gives better predictions of MHWs in the SCS, providing invaluable reference information for marine management authorities to make well-informed decisions and issue timely MHW warnings.

    Beschreibung: Mesoscale eddies are ubiquitous in the ocean, yet our understanding of their evolutions, particularly eddy merging processes, remains enigmatic. In this study, the merging processes of two cyclonic–cyclonic and two anticyclonic–anticyclonic eddies are analyzed in the Subtropical Northwest Pacific using satellite remote sensing altimetry data. The results reveal that, as eddies approach each other, their contours become connected, leading to the formation of multi-core eddies. Simultaneously, the merging process prompts substantial exchanges of energy and vorticity, resulting in the dissipation of one eddy and the emergence of a more extensive merged eddy. Throughout the merging process, the eddy contours elongate upwards along the centerline (the line connecting eddy centers) and there are distinct changes in both the horizontal and vertical morphology of the eddies. Notably, after the merging, the eddies distinctly exhibit intensified signals of sea surface temperature and vertical temperature anomaly, an outcome of their transformative fusion. The findings of this study significantly enhance our understanding of mesoscale eddy dynamics, particularly in the intricate eddy merging process. However, it is important to note that, due to limitations in vertical observational data, this study does not provide a quantitative portrayal of the vertical mechanisms of eddy merging, which also underscores a pivotal avenue for future research in the field.

    Beschreibung: In this work, water extracts from different bio-based products of plant origin were studied to evaluate their antioxidant capacity and their potential to form metal nanoparticles from aqueous solutions. Two traditional tests, the Folin–Ciocalteu assay and the DPPH radical scavenging capacity method were compared with a more recent one, SNPAC, based on the formation of silver nanoparticles. The silver nanoparticle antioxidant capacity method (SNPAC) was optimized for its application in the characterization of the extracts selected in this work; kinetic studies and extract concentration were also evaluated. The extracts were obtained from leaves of oak, eucalyptus, green tea, white and common thyme, white cedar, mint, rosemary, bay, lemon, and the seaweed Sargassum muticum. The results demonstrate that any of these three methods can be used as a quick test to identify an extract to be employed for nanoparticle formation. Additionally, we studied the synthesis of Cu, Fe, Pb, Ni, and Ag nanoparticles using eucalyptus extracts demonstrating the efficiency of this plant extract to form metallic nanoparticles from aqueous metal salt solutions. Metal nanoparticles were characterized by transmission electron microscopy and dynamic light scattering techniques.

    Beschreibung: The Lagrangian dispersion statistics of the Black Sea are estimated using satellite-tracked drifters, satellite altimeter data and a high-resolution ocean model. Comparison between the in-situ measurements and the model reveals good agreement in terms of the surface dispersion. The mean sub-basin coherent structures and currents of the Black Sea are well reproduced by the model. Seasonal variability of the dispersion in the upper (15 m), intermediate (150 m) and deep (750 m) layers are discussed with a special focus of the role of sub-basin scale structures and currents on the turbulent dispersion regimes. In terms of the surface relative dispersion, the results show the presence of the three known turbulent exponential, Richardson and diffusive-like regimes. The non-local exponential regime is only detected by the model for scales 〈10 km, while the local Richardson regime occurs between 10 and 100 km in all cases due to the presence of an inverse energy cascade range, and the diffusive-like regime is well detected for the largest distance by drifters (100–300 km) in winter/spring. Regarding the surface absolute dispersion, it reflects the occurrence of both quasi-ballistic and random-walk regimes at small and large times, respectively, while the two anomalous hyperbolic (5/4) and elliptic (5/3) regimes, which are related to the topology of the Black Sea, are detected at intermediate times. At depth, the signatures of the relative and absolute dispersion regimes shown in the surface layer are still valid in most cases. The absolute dispersion is anisotropic; the zonal component grows faster than the meridional component in any scenario.

    Beschreibung: Phytoplankton cell size is important for a multitude of functional traits such as growth rates, storage capabilities, and resistance to grazing. Because these response traits are correlated, selective effects on mean community cell size of one environmental factor should impact the ability of phytoplankton to cope with other factors. Here, we experimentally apply expectations on the functional importance of phytoplankton cell size to the community level. We used a natural marine plankton community, and first altered the community’s cell size structure by exposing it to six different grazer densities. The size-shifted communities were then treated with a saturated nutrient pulse to test how the changes in community size structure influenced the mean community growth rate in the short-term (day 1–3) and nutrient storage capacity in the postbloom phase. Copepod grazing reduced the medium-sized phytoplankton and increased the share of the smallest (〈10 µm3 ) and the largest (〉100,000 µm3 ). Communities composed of on average small cells grew faster in response to the nutrient pulse, and thus confirmed the previously suggested growth advantage of small cells for the community level. In contrast, larger phytoplankton showed better storage capabilities, reflected in a slower post-bloom decline of communities that were on average composed of larger cells. Our findings underline that the easily measurable mean cell size of a taxonomically complex phytoplankton community can be used as an indicator trait to predict phytoplankton responses to sequential environmental changes.

    Beschreibung: Accurate, on-site determinations of macronutrients (phosphate (PO43−), nitrate (NO3−), and silicic acid (H4SiO4)) in seawater in real time are essential to obtain information on their distribution, flux, and role in marine biogeochemical cycles. The development of robust sensors for long-term on-site analysis of macronutrients in seawater is a great challenge. Here, we present improvements of a commercial automated sensor for nutrients (including PO43−, H4SiO4, and NO2− plus NO3−), suitable for a variety of aquatic environments. The sensor uses the phosphomolybdate blue method for PO43−, the silicomolybdate blue method for H4SiO4 and the Griess reagent method for NO2−, modified with vanadium chloride as reducing agent for the determination of NO3−. Here, we report the optimization of analytical conditions, including reaction time for PO43− analysis, complexation time for H4SiO4 analysis, and analyte to reagent ratio for NO3− analysis. The instrument showed wide linear ranges, from 0.2 to 100 μM PO43−, between 0.2 and 100 μM H4SiO4, from 0.5 to 100 μM NO3−, and between 0.4 and 100 μM NO2−, with detection limits of 0.18 μM, 0.15 μM, 0.45 μM, and 0.35 μM for PO43−, H4SiO4, NO3−, and NO2−, respectively. The analyzer showed good precision with a relative standard deviation of 8.9% for PO43−, 4.8% for H4SiO4, and 7.4% for NO2− plus NO3− during routine analysis of certified reference materials (KANSO, Japan). The analyzer performed well in the field during a 46-day deployment on a pontoon in the Kiel Fjord (located in the southwestern Baltic Sea), with a water supply from a depth of 1 m. The system successfully collected 443, 440, and 409 on-site data points for PO43−, Σ(NO3− + NO2−), and H4SiO4, respectively. Time series data agreed well with data obtained from the analysis of discretely collected samples using standard reference laboratory procedures and showed clear correlations with key hydrographic parameters throughout the deployment period.

    Beschreibung: Critical questions exist regarding the abundance and, especially, the export of picophytoplankton (≤2 µm diameter) in the Arctic. These organisms can dominate chlorophyll concentrations in Arctic regions, which are subject to rapid change. The picoeukaryotic prasinophyte Micromonas grows in polar environments and appears to constitute a large, but variable, proportion of the phytoplankton in these waters. Here, we analyze 81 samples from the upper 100 m of the water column from the Fram Strait collected over multiple years (2009–2015). We also analyze sediment trap samples to examine picophytoplankton contributions to export, using both 18S rRNA gene qPCR and V1-V2 16S rRNA Illumina amplicon sequencing to assess the Micromonas abundance within the broader diversity of photosynthetic eukaryotes based on the phylogenetic placement of plastid-derived 16S amplicons. The material sequenced from the sediment traps in July and September 2010 showed that 11.2 ± 12.4% of plastid-derived amplicons are from picoplanktonic prasinophyte algae and other green lineage (Viridiplantae) members. In the traps, Micromonas dominated (83.6% ± 21.3%) in terms of the overall relative abundance of Viridiplantae amplicons, specifically the species Micromonas polaris. Temporal variations in Micromonas abundances quantified by qPCR were also observed, with higher abundances in the late-July traps and deeper traps. In the photic zone samples, four prasinophyte classes were detected in the amplicon data, with Micromonas again being the dominant prasinophyte, based on the relative abundance (89.4% ± 8.0%), but with two species (M. polaris and M. commoda-like) present. The quantitative PCR assessments showed that the photic zone samples with higher Micromonas abundances (〉1000 gene copies per mL) had significantly lower standing stocks of phosphate and nitrate, and a shallower average depth (20 m) than those with fewer Micromonas. This study shows that despite their size, prasinophyte picophytoplankton are exported to the deep sea, and that Micromonas is particularly important within this size fraction in Arctic marine ecosystems.

    Beschreibung: The Arctic is warming 2–3 times faster than the global average, leading to a decrease in Arctic sea ice extent, thickness, and associated changes in sea ice structure. These changes impact sea ice habitat properties and the ice-associated ecosystems. Sea-ice algal blooms provide various algal-derived carbon sources for the bacterial and archaeal communities within the sea ice. Here, we detail the transition of these communities from winter through spring to early summer during the Norwegian young sea ICE (N-ICE2015) expedition. The winter community was dominated by the archaeon Candidatus Nitrosopumilus and bacteria belonging to the Gammaproteobacteria (Colwellia, Kangiellaceae, and Nitrinocolaceae), indicating that nitrogen-based metabolisms, particularly ammonia oxidation to nitrite by Cand. Nitrosopumilus was prevalent. At the onset of the vernal sea-ice algae bloom, the community shifted to the dominance of Gammaproteobacteria (Kangiellaceae, Nitrinocolaceae) and Bacteroidia (Polaribacter), while Cand. Nitrosopumilus almost disappeared. The bioinformatically predicted carbohydrate-active enzymes increased during spring and summer, indicating that sea-ice algae-derived carbon sources are a strong driver of bacterial and archaeal community succession in Arctic sea ice during the change of seasons. This implies a succession from a nitrogen metabolism-based winter community to an algal-derived carbon metabolism-based spring/ summer community.

    Beschreibung: A vision for the establishment of a Geopark in Jordan is given in this work, with a subsequent application to the UNESCO Global Geopark programme. The Dead Sea area and its surroundings have suffered strong changes in the last decades, accompanied by a variety of natural hazards related to enhanced erosional processes. The aspiring Geopark will thematically encompass the influence that these changes and related natural hazards, including flash floods and subsidence, have had on the local population, from geological, over historical up to recent times. The hydrogeology and geomorphology, i.e., the connection between erosion by water, dissolution of minerals, and landscape evolution, will be the main guiding theme that connects the Eastern Rim Highlands with the Dead Sea rift valley through ephemeral wadis, vegetated springs areas, and traditionally communities. The creation of the Geopark is aimed at holistic, sustainable development and management of the area by eco-tourism, and includes education on water resource management, hazard awareness and resilience, as well as international research. We here present the conceptual approach to the initial development of a Geopark network in Jordan. In a narrative discourse, we highlight realised and further implementation steps, with an evaluation of the expected timeline, potential partner institutions, regional involvement and the chances for realisation.

    Beschreibung: Deep learning has been successfully applied to many classification problems including underwater challenges. However, a long-standing issue with deep learning is the need for large and consistently labeled datasets. Although current approaches in semi-supervised learning can decrease the required amount of annotated data by a factor of 10 or even more, this line of research still uses distinct classes. For underwater classification, and uncurated real-world datasets in general, clean class boundaries can often not be given due to a limited information content in the images and transitional stages of the depicted objects. This leads to different experts having different opinions and thus producing fuzzy labels which could also be considered ambiguous or divergent. We propose a novel framework for handling semi-supervised classifications of such fuzzy labels. It is based on the idea of overclustering to detect substructures in these fuzzy labels. We propose a novel loss to improve the overclustering capability of our framework and show the benefit of overclustering for fuzzy labels. We show that our framework is superior to previous state-of-the-art semi-supervised methods when applied to real-world plankton data with fuzzy labels. Moreover, we acquire 5 to 10% more consistent predictions of substructures.

    Beschreibung: Image annotation is a time-consuming and costly task. Previously, we published MorphoCluster as a novel image annotation tool to address problems of conventional, classifier-based image annotation approaches: their limited efficiency, training set bias and lack of novelty detection. MorphoCluster uses clustering and similarity search to enable efficient, computer-assisted image annotation. In this work, we provide a deeper analysis of this approach. We simulate the actions of a MorphoCluster user to avoid extensive manual annotation runs. This simulation is used to test supervised, unsupervised and transfer representation learning approaches. Furthermore, shrunken k-means and partially labeled k-means, two new clustering algorithms that are tailored specifically for the MorphoCluster approach, are compared to the previously used HDBSCAN*. We find that labeled training data improve the image representations, that unsupervised learning beats transfer learning and that all three clustering algorithms are viable options, depending on whether completeness, efficiency or runtime is the priority. The simulation results support our earlier finding that MorphoCluster is very efficient and precise. Within the simulation, more than five objects per simulated click are being annotated with 95% precision.
