    Notes: Summary The genusPlebeia has a special significance for the study of social evolution of stingless bees: morphologically primitive, its species display a wealth of behavioural evolution, especially with respect to the oviposition process. We comparePlebeia remota with the few other members of the genus studied so far. Related to its subtropical geographical range, brood production is seasonal (there is no brood in the colony in colder months), and adult workers occur as summer and winter bees. The nest is in tree cavities, and the involucrum is absent or restricted to the winter period. Brood cells are arranged in horizontal combs, and new cells are built completely synchronously. Each series consists of up to 50 cells, their number being mainly dependent on colony size. Construction speed is remarkably constant, allowing 4–6 batches per 24 hours. Cell building and provisioning are activities of a small group of specialized workers. The oviposition cycle follows the classical subdivisions for stingless bees. During the patrolling phase a worker may “offer” a trophic egg in a most remarkable way: while retreating backward from the queen she bends the abdomen under thorax and head, and lays an egg on the comb. This egg is eaten by the queen or a worker. The provisioning, oviposition and operculation of all cells occur simultaneously, each cell is provisioned by 4–9 workers. Localization of a cell by the queen may be facilitated by its characteristic guard, which “defends” the cell against the approaching queen. The degree of synchronization within a batch is very high: the duration per cell lasts 420–950 sec, the batch of up till 50 cells needs only 557–1160 sec. Operculation is done by a worker that was not involved in the previous steps. Males are generally produced by the queen. Several male producing cycles per year occur. In orphan colonies laying workers give rise to males, and in queenright colonies workers may occasionally reproduce as well. Division of labour follows the general pattern for stingless bees; however, cell building and provisioning are activities of a specialized group of workers.

    Notes: Summary Queen rearing is suppressed in honey bees (Apis mellifera L.) by pheromones, particularly the queen's mandibular gland pheromone. In this study we compared this pheromonally-based inhibition between temperate and tropically-evolved honey bees. Colonies of European and Africanized bees were exposed to synthetic queen mandibular gland pheromone (QMP) for ten days following removal of resident queens, and their queen rearing responses were examined. Queen rearing was suppressed similarly in both European and Africanized honey bees with the addition of synthetic QMP, indicating that QMP acts on workers of both races in a comparable fashion. QMP completely suppressed queen cell production for two days, but by day six, cells containing queen larvae were present in all treated colonies, indicating that other signals play a role in the suppression of queen rearing. In queenless control colonies not treated with QMP, Africanized bees reared 30% fewer queens than Europeans, possibly due to racial differences in response to feedback from developing queens and/or their cells. Queen development rate was faster in Africanized colonies, or they selected older larvae to initiate cells, as only 1 % of queen cells were unsealed after 10 days compared with 12% unsealed cells in European colonies.

Furthermore, it shows that the adult castes of the ponerine andHypoponera sp. have different habits of trophallaxis. Workers display a soliciting behavior toward queens, males and workers, but receive regurgitated food only from workers. The workers are forced to stop soliciting for regurgitated food by the “whipping behavior” of the queen. Callow queens solicit regurgitated food from workers and ergatoid males and receive it, while mature queens do not solicit regurgitation from workers. Ergatoid males receive regurgitated food from workers and regurgitate it to queens. Alate males show no trophallactic food exchange with workers and queens. Trophallactic behavior was never observed between males of either winged or ergatoid types.

    Notes: Summary A comparison was made of some physicochemical characteristics of epigeous termitaries (nest walls and surrounding horizons) of four species of soil-feeding termites living in tropical rainforests. Our aim was to determine whether these species affect the different compounds involved in the structural stability of soil in a similar manner. Our data support the general finding that the structural stability of soil is correlated with organic matter, cations and the relative proportion of mineral elements. Of these parameters, the content of organic matter is the most significant factor effecting the stability of termite building materials. Analysis of humic compound distribution revealed that fulvic and humic acids, owing to their electrochemical properties, are highly involved. Also, the organic matter in termitaries is more polymerized than that of humiferous control horizons, leading to FA/HA ratios close to 1. The stability of nest walls and topsoils differs between the species. Generally, the speciesNoditermes lamanianus, Thoracotermes macrothorax andCubitermes fungifaber build nests that are enriched with organic matter and exchangeable cations, resulting in high structural stability. In contrast, materials worked byCrenetermes albotarsalis are not enriched with organic matter or cations and do not differ in stability from the control soils. It is concluded that any generalization on the overall influence of soil-feeding termites on soil fertility might be misleading. Only species which enrich their materials with organic matter, especially stabilised humic acids, contribute to soil conservation and hence fertility. Once the termitary is dead, its organic matter is again available to the soil ecosystem.

    Notes: Summary First descendants' founding ability was tested inP. dominulus colonies by experimentally removing the original nests and queens 21–24 days after the first emergence. Foundation tests were carried out after three different periods had elapsed after the removal and the results were analyzed with regards to the effects of participation in social tasks, duration of stay within the colony and seasonal factors. 1.Immediately after the removal, the foundations that occurred were performed almost entirely by workers, regardless of the duration of their stay within the colony. The number of foundations depended on the season in which the females had emerged. 2.Two months and four months after the removal, foundations were made by only a small number of females (workers and non-workers). These females, which were therefore able to found a nest the year they were born, were mostly among the very first-born individuals in the colonies. 3.The following year after hibernation: females (workers and non-workers) which had stayed for only a short time within the colony (1–15 days) had the highest survival rates. The foundation rates among the surviving females depended only on seasonal factors, since the foundation rates were higher among the non-workers which had emerged later in the season. The results throw some light on the first descendants' nest-founding potential in colonies developing under natural conditions, where many workers in fact stay for only a short time at the nest.

    Notes: Abstract A field survey of the 1992 Flores Island earthquake tsunami was conducted during December 29, 1992 to January 5, 1993 along the north coast of the eastern part of Flores Island. We visited over 40 villages, measured tsunami heights, and interviewed the inhabitants. It was clarified that the first wave attacked the coast within five minutes at most of the surveyed villages. The crust was uplifted west of the Cape of Batumanuk, and subsided east of it. In the residential area of Wuring, which is located on a sand spit with ground height of 2 meters, most wooden houses built on stilts collapsed and 87 people were killed even though the tsunami height reached only 3.2 meters. In the two villages on Babi Island, the tsunami swept away all wooden houses and killed 263 of 1,093 inhabitants. Tsunami height at Riang-Kroko village on the northeastern end of Flores Island reached 26.2 meters and 137 of the 406 inhabitants were killed by the tsumani. Evidence of landslides was detected at a few points on the coast of Hading Bay, and the huge tsunami was probably formed by earthquake-induced landslides. The relationship between tsunami height and mortality was checked for seven villages. The efficiencies of trees arranged in front of coastal villages, and coral reefs in dissipating the tsunami energy are discussed.

    Notes: Abstract Laboratory experiments of a 7.2-m-diameter conical island were conducted to study three-dimensional tsunami runup. The 62.5-cm tall island had 1 on 4 side slopes and was positioned in the center of a 30-m-wide by 25-m-long flat-bottom basin. Solitary waves with height-to-depth ratios ranging from 0.05 to 0.20 and “source” lengths ranging from 0.30 to 7.14 island diameters were tested in water depths of 32 and 42 cm. Twenty-seven capacitance wave gages were used to measure surface wave elevations at incident and four radial transects on the island slope. Maximum vertical runup measurements were made at 20 locations around the perimeter of the island using rod and transit. A new runup gage was located on the back or lee side of the island to record runup time series.

    Notes: Abstract Runup data in Hokkaido and in three prefectures in the Tohoku District are described with a few witnessed arrival times and with comments of tide records. The highest runup of 31.7 m was found at the bottom of a narrow valley on the west coast of Okushiri Island. In order to explain high runups of 20 m at Hamatsumae in the sheltered area, roles of edge waves, refraction of the Okushiri Spur and tsunami generation by causes other than the major fault motion should be understood. An early arrival of the tsunami on the west coast of Hokkaido suggests another tsunami generation mechanism in addition to the major fault motion.

    Notes: Abstract The values of seismic moment andS-wave corner frequency from 1575 seismic events induced in South African, Canadian, Polish, and German underground mines were collected to study their scaling relations. The values ofP-wave corner frequency from 649 events were also available. Seismic moments of these events range from 5*103 to 2*1015 N·m (moment magnitude is from −3.6 to 4.1), theS-wave corner frequency ranges from 0.7 to 4438 Hz, and theP-wave corner frequency is between 5 and 4010 Hz. The slope of a regression line between the logarithm ofS- andP-wave corner frequencies is equal to one, and the corner frequencies ofP waves are higher than those ofS waves on the average by about 25 percent. In studies of large and moderate earthquakes it has been found that stress drop is approximately independent of the seismic moment, which means that seismic moment is inversely proportional to the third power of corner frequency. Such a behavior was confirmed for most of the data considered here. A breakdown in the similarity betwen large and small events seems to occur for the events with moment magnitude below −2.5. The average values of seismic moment referred to the same range of corner frequency, however, are vastly different in various mining areas.

    Notes: Abstract From the events synthesized from the one-dimensional dynamical mass-spring model proposed byBurridge andKnopoff (1967), the relation between rupture length Δ and earthquake momentM is studied for various model parameters. The earthquake moment is defined to be the total displacement of a connected set of mass elements which slide during an event. A parameter stiffness ratios is defined as the ratio of the spring constant between the two mass elements to that between one mass element and the moving plate. The velocity-dependent friction law (including weakening and hardening processes) is taken to control the sliding of a mass element. The distribution of the breaking strengths over the system is considered to be a fractal function. The cases for severals values and different velocity-dependent friction laws with different decreasing ratesr w of the frictional force with sliding velocity are studied numerically. The weakening process of the frictional force from the static one to the dynamic one obviously affects theM−Δ relation. Meanwhile, a rapid weakening process rather than a slow weakening process can result in aM−Δ relation, which is comparable to the observed one. Although an increase in thes value can yield an increase in the upper bound of the Δ value and the number of events with largeM and Δ values, the scaling of theM−Δ relation is not affected by the change of thes value. For the cases in this study, the theoretical Δ−M relations for small events withM〈1 are almost in the form: Δ∼M 1/2, while those for large events withM〉1 have a scaling exponent less than but close to 1. In addition, the fractal dimension, the friction drop ratio and the roughness of the distribution of the breaking strengths over the fault surface are the minor parameters influencing the Δ−M relation. A comparison between the theoreticalM−Δ relation and the observed one for strike-slip earthquakes shows that for large events the theoreticalM−Δ relation is quite consistent with the observed one, while for small events there is a one-order difference in the two relations. For the one-dimensional model, the decreasing rate of the dynamic frictional force with velocity is the main factor in affecting the characteristic value of the earthquake moment, at which the scaling of theM−Δ relation changes.

    Notes: Abstract The Bayesian extreme-value distribution of earthquake occurrences has been adopted to estimate the seismic hazard in some seismogenic zones in Greece and surrounding regions. Seismic moment, slip rate, earthquake recurrence rate and magnitude were considered as basic parameters for computing the prior estimates of the seismicity. These estimates are then updated in terms of Bayes' theorem and historical estimates of seismicity associated with each zone. High probabilities for earthquakes withM≥6.0 have been obtained for the northwestern part of Greece as well as for the southwestern part of the Hellenic arc.

    Notes: Abstract Observations taken by aircraft and conventional platforms are used to investigate dynamical, physical, and radiative processes within a marine stratus cloud during the Canadian Atlantic Storms Program (CASP) II field project which took place over the east coast of Canada. Stratus which formed over the ocean on February 6, 1992 during the nighttime, is studied to analyze cloud top and base processes. The cloud was supercooled during the study period. Fluctuations and fluxes are calculated along constant flight altitude legs approximately 100 km long in space. The scales of structures larger than 5 km are removed from the analysis using a running average technique. Droplet spectra obtained by a forward scattering spectrometer probe (FSSP) were used in a 1-D radiative transfer model to calculate infrared (IR) fluxes and radiative heating rates. A heat conservation equation was used to estimate vertical air velocity (w a ) within the cloud. The results showed that, because of a warmer ocean surface, significant moisture and heat were transferred from the ocean surface to the boundary layer. The cloud base was at about 400 m height and the top was at about 1.4 km.w a at the cloud base was estimated about 5 cm s−1. Strong IR cooling rate at the cloud top was calculated to be 75°C day−1 for a 100 m thick layer. Negative skewness inw a , suggesting narrow downdrafts, was likely due to radiative cooling at the cloud top. The entrainment velocity was found to be about 1.5 cm s−1 at cloud top. Mean moisture and heat fluxes within the cloud were estimated to be comparable to those from the ocean surface. Vertical air velocity at the cloud top due to radiative cooling was found to be about −40 cm s−1.

    Notes: Abstract We study eight tsunamigenic earthquakes of 1992–1994 with data from single near-field 3-component long-period stations. The analysis is made from the standpoint of tsunami warning by an automatic process which estimates the epicentral location and the seismic moment through the variable-period mantle magnitudeM m . Simulations of early warning based on the real-time computation of the seismic moment are also tested with this system, which would give a justified warning in each region of tsunami potentiality. By exploiting the dependence of moment rate release with frequency, the system has the capability of recognizing both “tsunami earthquakes” such as the 1992 Nicaragua and 1994 Java events, as well as instances of the opposite case of low-frequency deficiency, interpreted as indicating a deeper than normal source (1993 Guam event). We report both the results of delayed-time processing of the near-field stations, and the actual real-time warnings at PPT, which confirm the former.

    Notes: Abstract Source parameter estimates based on the homogeneous and inhomogeneous source models have been examined for an anomalous sequence of seven mine-induced events located between 640 and 825 m depth at Strathcona mine, Ontario, and having magnitudes ranging betweenm N 0.8 and 2.7. The derived Brune static stress drops were found to be similar to those observed for natural earthquakes (∼30 bars), whereas dynamic stress drops were found to range up to 250–300 bars. Source radii derived from Madariaga's model better fit documented evidence of underground damage. These values of source radii were similar to those observed for the inhomogeneous model. The displacement at the source, based on the observed attenuation relationship, was about 60 mm for three magnitude 2.7 events. This is in agreement with slip values calculated using peak velocities and assuming the asperity as a Brune source within itself (72 mm). By using Madariaga's model for the asperity, the slip was over 3 times larger than observed. Peak velocity and acceleration scaling relations with magnitude were investigated by incorporating available South African data, appropriately reduced to Canadian geophysical conditions. The dynamic stress drop scaled as the square root of the seismic moment, similar to reported results in the literature for crustal earthquakes. This behavior suggests that the size of the asperities responsible for the peak ground motion, with respect to the overall source size, follow distributions that may be similar over a wide range of magnitudes. Measurements of source rupture complexity (ranging from 2 to 4) were found to agree with estimates of overall source to asperity radii, suggesting, together with the observed low rupture velocities (0.3 β to 0.6 β), that the sources were somewhat complex. Validation of source model appropriateness was achieved by direct comparison of the predicted ground motion level to observed underground damage in Creighton mine, located within the same regional stress and geological regime as Strathcona mine. Close to the source (〈100 m), corresponding to relatively higher damage levels, a good agreement was found between the predicted peak particle velocities for the inhomogeneous model and velocities derived based on established geomechanical relationships. The similarity between asperity radii and the regions of the highest observed damage provided additional support for the use of the inhomogeneous source model in the assessment of damage potential.

    Notes: Abstract In China, the earthquakes induced by water injection have occurred in four oil fields including the Renqiu oil field, and in two mines. Production of oil from the Renqiu oil field began in 1975 and the injection of water into the oil field commenced in July 1976. The induced earthquakes have been occurring in the area for the past 17 years, since December 1976. The controlled experiments of water injection showed the cause and effect relation between water injection and earthquakes. Source parameters such as source dimension, seismic moment and stress drop of a large number of the induced earthquakes, andQ factor for the area have been determined. The results indicate that the stress drop varies from 0.2 to 3.0 bar and theQ factor has an average value of 75.0. The low-stress drop and lowQ factor values imply that the earthquakes are caused by the brittle fracture of weak rocks under low ambient stresses, due to a decrease in their strength because of the injection of water. The induced earthquakes are unevenly distributed in the oil field. The northern part of the oil field, where the reservoir rocks are characterized by low porosity and low permeability, exhibits high seismic activity with the largest earthquake registering a magnitude of 4.5 and about 68% of the total number of induced earthquakes in this part. Whereas, the southern part of the oil field with higher porosity and higher permeability is characterized by low seismic activity with the largest earthquake registering a magnitude of 2.5 and only 4% of the total number of earthquakes which occurred in this part. These features of the focal region suggest that larger earthquakes may not occur in the Renqiu oil field area.

Assuming a fault model under the sea bottom, we estimated the focal parameters jointly, using synthesized tsunami source spectra as well as the tsunami numerical simulation. The fault proposed by this study consists of a plane sized 15×15 km, dipping N47°W with the dip angle of 42°, which is almost pure reverse fault (slip angle 87°) with a dislocation of 1 meter. The numerical simulation shows that the shallow sea in the source region caused a comparatively long recurring tsunami (the periods are 12–18 minutes) in spite of its small size. The model fault is corresponding to an aftershock area of this earthquake.

    Notes: Abstract Two earthquakes occurred in 1993 off southern Kamchatka. They have similar surface wave magnitudes, focal mechanisms, and depths, but have distinctly different characteristics. The November earthquake is a standard or “impulsive”M7 underthrusting event. The June earthquake is a tsunamigenic or “low-stress-drop” event with several unusual characteristics, including a large, diffuse aftershock zone, directivity, and a long source time function. The 1993 earthquakes ruptured a segment of the Kamchatka Arc which has not ruptured since 1904. The 1993 earthquakes seem to signal the midpoint in the southern Kamchatka seismic cycle.

    Notes: Abstract The southwest Hokkaido tsunami of July 12th, 1993, left continuous onshore sand deposits along the west coast of Oshima Peninsuka, Hokkaido, northern Japan. We investigated spatial distribution and lithofacies of the new tsunami deposits for its identification of ancient tsunami deposits. An eyewitness acount and bent plants helped our interpretation of the onshore tsunami behavior. We regard the following properties as typical of the coastal tsunami sand deposits: (1) The deposits cover the surface almost continuously on gentle topography. (2) Deposit thicknesses and mean grain sizes descrease with distance from the sea. (3) Deposit thicknesses and lithofacies vary greatly across local surface undulation. (4) Graded bedding reflecting tsunami runup and backwash is present in thick deposits. (5) The deposits are widely distributed along the coast and extend inland several tens of meters to 100 m. We examined a candidate for the paleo-tsunami deposits associated with the 1640 Komagatake eruption, and confirmed that the similar patterns are typical of ancient tsunami deposits.

    Notes: Abstract The geological and hydrological conditions near the reservoir site play an important role in the generation or absence of seismic activity. Near Bhatsa reservoir, along the west coast of India intense seismic activity occurred during August–September 1983, after a lag of six years of initial impounding. From July 1983 to September 1990, 15,388 earthquakes (mostlyM 1〈3.0) were recorded, the largest being of magnitude 4.9. The spatial distribution of well located 172 earthquakes suggest a strong correlation between the epicenters and the disposition of dykes and faults around the Bhatsa region. It is inferred that these dykes have acted as “barriers” for the diffusion of water from the reservoir, thereby becoming zones of instability due to increased pore pressure not only along them but also over the volume they bound.

    Notes: Abstract Nurek, Aswan and Koyna reservoirs were affected by moderate earthquakes with continuing seismic activity. Microearthquake data recorded through local networks have been used to determine the strange attractor dimensions, using deterministic chaos which were found as 7,2, 3.8 and 4.8, respectively. This would imply that while 8 parameters are needed to model earthquakes near Nurek reservoir, only 4 to 5 parameters are needed for the Aswan and Koyna regions. The differences in the strange attractor dimension suggest them to be a measure of seismotectonics around such reservoirs.

We have analysed mode-converted shear waves in a data set acquired with a hybrid marine/land geometry in Isfjorden, Svalbard. Through a kinematic 2D ray-tracing modellingV p/Vs ratios for part of the uppermost 5km of the crust are obtained. Low values (V p /V s =1.65) are tentatively associated with the section of Devonian sandstones which appears to attain a minimum thickness of 1.5km below 3 km depth about 10km west of Kapp Thorden.

    Notes: Abstract On December 12, 1992 a large earthquake (M s 7.5) occurred just north of Flores Island, Indonesia which, along with the tsunami it generated, killed more than 2,000 people. In this study, teleseismicP andSH waves, as well asPP waves from distances up to 123°, are inverted for the orientations and time histories of multiple point sources. By repeating the inversion for reasonable values of depth, time separation and spatial separation, a 2-fault model is developed. Next, the vertical deformation of the seafloor is estimated from this fault model. Using a detailed bathymetric model, linear and nonlinear tsunami propagation models are tested. The data consist of a single tide gauge record at Palopo (650 km to the north), as well as tsunami runup height measurements from Flores Island and nearby islands. Assuming a tsunami runup amplification factor of two, the two-fault model explains the tide gauge record and the tsunami runup heights on most of Flores Island. It cannot, however, explain the large tsunami runup heights observed near Leworahang (on Hading Bay) and Riangkroko (on the northeast peninsula). Massive coastal slumping was observed at both of these locations. A final model, which in addition to the two faults, includes point sources of large vertical displacement at these two locations explains the observations quite well.

    Notes: Abstract To analyse segmental differentiation processes in muscle development, we studied the embryogenesis of the ventral body wall muscles in thoracic and abdominal segments of the grasshopper Schistocerca gregaria at the identified cell level. We visualized differentiating muscle pioneer and muscle precursor cells by staining with a muscle-specific monoclonal antibody and with rhodamine-coupled phalloidin. Our results show that a similar pattern of serially reiterated early muscle pioneers is initially established in all segments. Subsequently, two major segmental differentiation processes occur. First, segment-specific sets of additional, later differentiating muscle pioneers are generated de novo. Second, segment-specific sets of existing early muscle precursors are eliminated through atrophy and eventual loss. These events have consequences for matching homonomy of muscles and their innervating motoneurons. Taken together, these processes in the embryo, in concert with postembryonic differentiation events, play critical roles in shaping the highly specialized muscular structures of the mature animal.

    Notes: Abstract The ascidian egg contains cytoplasmic determinants that specify the fate of larval muscle cells. In a previous study, we developed an experimental system to identify the molecular nature of muscle determinants, in which unfertilized Ciona savignyi eggs were fragmented into four pieces by centrifugation. When inseminated, only nucleated fragments (red fragments) develop into partial embryos that only show differentiation of epidermal cells. One type of enucleated fragment (black fragment) has the remarkable ability to promote muscle differentiation when fused with red fragments. In the present study, using this experimental system, we investigated the molecular nature of muscle determinants. UV irradiation of black fragments suppressed the ability to promote expression of the muscle-specific protein, myosin heavy chain. The wavelength of UV light responsible for the inactivation (250–275 nm) suggested that UV-sensitive targets are nucleic acids. Injection of poly(A)+ RNA isolated from an un-irradiated black-fragment-rich fraction into UV-irradiated black fragments partially recovered the ability to promote the expression of myosin heavy chain protein. Poly(A)+ RNA from a red-fragment-rich fraction did not rescue the suppression of UV-irradiated black fragments. These results suggest that maternal mRNAs enriched in black fragments are closely associated with muscle determinants in the ascidian egg.

    Notes: Abstract Several Drosophila receptor-linked protein tyrosine phosphatases (R-PTPs) are selectively expressed on axons of the developing embryonic central nervous system. The extracellular domains of these axonal R-PTPs are homologous to neural adhesion molecules. Thus, R-PTPs may directly couple cell recognition to signal transduction via control of tyrosine phosphorylation. To examine the function of these molecules during nervous system development, we wished to generate mutations in R-PTP genes. It was unclear whether a mutation in a single R-PTP gene would confer lethality, however, because the similarities in sequence and expression pattern between the axonal R-PTPs suggest that they may have partially redundant functions. To circumvent this problem, we developed a directed mutagenesis strategy based on local transposition of P elements, and used this approach to isolate a null mutation in the DPTP99A gene. This strategy, which we describe in detail here, should be applicable to any Drosophila gene within a lettered division of an appropriately marked P element. Flies lacking DPTP99A expression are viable and fertile, and we have been unable to detect any alterations in the embryonic nervous system of DPTP99A embryos using a variety of antibody markers.

    Notes: Abstract Our present detailed understanding of the genetic mechanisms controlling segmentation has been made possible, in large part, by comprehensive screens of cuticular morphology that identified genes involved in epidermal patterning. To systematically identify genes involved in internal morphogenesis, specifically development of the gut, we have screened mutant embryos produced by a collection of 53 embryonic lethal mutations affecting embryonic pattern formation or differentiation, and a collection of 161 deficiencies covering, in aggregate, approximately 70% of the genome. Staining with the anti-crumbs antibody was used to characterize the Malpighian tubules and hindgut, as well as other internal organs. The geneshuckebein, tailless andwingless, and two previously undescribed loci at 24C/D and 68D/E, are required to establish the primordia for the posterior midgut and hindgut/Malpighian tubules. A locus in region 30A/C is required for extension of the midgut epithelium to surround the yolk, and region 36E/37F is required for outbudding of the Malpighian tubule primordia. Several deficiencies were identified that uncover loci with specific effects on the morphogenesis (elongation, lumen formation) of the hindgut and Malpighian tubules and on the formation of constrictions in the midgut.

    Notes: Abstract The process of labyrinth determination has been studied in three urodelean and seven anuran species by means of homoplastic transplantation of ear region epidermis, defined as the piece of epidermal layer containing the material of the prospective ear vesicle (labyrinth rudiment). The ear region epidermis was grafted onto the abdominal wall of embryos of the same developmental stage. The earliest stage of operation resulting in ectopic ear vesicle formation was determined, suggesting the appearance of organ-specific properties in the ear ectoderm. These properties were enhanced in the further course of development, as indicated by the frequency of ear vesicle formation and by the volume and degree of complexity that the vesicles reached. The data obtained allowed us to arrange the species studied in a sequence, ranging from most Ranidae and Bufo viridis, in which organ-specific properties appear earlier and are most strongly expressed, to Triturus vulgaris in which their expression is least pronounced. Comparison of properties of the material giving rise to the ear vesicle or to several other ectodermal derivatives led to the conclusion that species-specific differences in determination of different ectodermal rudiments are due to species specific properties of the whole ectoderm. These differences appear to be determined by an evolutionary shift of the beginning of gastrulation towards later cleavage cycles.

    Notes: Abstract We have investigated the effects of the glucocorticoid, dexamethasone, and five structural analogs on Drosophila development in an effort to identify steroid ligands that may play a role in the embryogenesis of this organism. Embryos were exposed to glucocorticoids either by direct culture in supplemented medium, or by examining embryos from adult flies raised on supplemented fly food. After exposure, embryos were examined for developmental defects. At a morphological level, exposure to dexamethasone disrupts the dorsolateral folding of the amnioserosa during germ band extension. In addition, germ band retraction and dorsal closure is also disrupted. The phenocritical period of these effects is within the first 4 h of embryogenesis. This response is dosage sensitive, with embryos responding to concentrations of dexamethasone ranging from 10−6 to 10−3M. Furthermore, glucocorticoids which are closely related structural analogs of dexamethasone also disrupt germ band retraction and dorsal closure, while other tested steroids had no effect on embryonic development. At a molecular level, expression of the gene, Krüppel, is absent from the amnioserosa of dexamethasone-treated embryos. The cuticular phenocopy resulting from exposure to dexamethasone and related glucocorticoids is morphologically similar to the mutant phenotype associated with four genes required for germ band retraction, namely hindsight, serpent, tail-up and u-shaped. The results of this study represent the first association of a glucocorticoid with dose, stage and tissue specific effects on Drosophila development at both morphological and molecular levels.

    Notes: Abstract A carp caudal cDNA of 1.3 kb was cloned after screening an early segmentation stage cDNA library with a probe produced by PCR using conserved homeobox sequences as primers and genomic DNA as template. The homeobox gene was called carp-cdxl. The gene appears highly similar to other vertebrate caudal homologs, especially the zebrafish gene cdx(Zf-cad). The possible relationship to homeobox genes within the Hox-C gene complexes is discussed. A weak expression of the gene, detected by in situ hybridization, was found shortly before gastrulation (at 25% epiboly) in cells likely to have a posterior fate. During gastrulation expression became stronger. At the early segmentation stage, cells of the neural keel in the area of the prospective spinal cord expressed the gene. During the progression of segmentation, expression retracted in a caudal direction. The tailbud expressed the gene throughout, but the somites lost expression shortly after their formation. Only the most lateral mesoderm cells maintained expression in the trunk area. Carp-cdxl was also expressed in the endoderm. At 24 h after fertilization the gene was only expressed in the tailbud. At 48 h, no expression could be detected. The expression pattern suggests a function for carp-cdxl in gastrulation and patterning along the anterior-posterior axis of the embryo.

    Notes: Abstract Protein-carbohydrate interactions are supposed to play key roles in the mechanisms of cell adhesion, biosignalling and intracellular routing, warranting the analysis of the developmental course of expression of epitopes of this system. Thus, a panel of carrier-immobilized carbohydrate ligands was used as probes, namely lactose,N-acetylgalactosamine,N-acetylglucosamine, mannose, fucose and maltose. Additionally, an antibody to an endogenous β-galactoside-binding lectin (anti-galectin-1), the biotinylated lectin and two further human lectins, namely the macrophage migration inhibitory factor-binding sarcolectin and serum amyloid P component (SAP) that displays selectivity for sulphated sugars and mannose-6-phosphate, were included. They enabled us to assess the extent of the presence of respective binding sites in fixed sections from human lungs (pulmonary epithelial cells), livers (hepatocytes) and hearts (myocard cells) of 10–50 weeks gestation. Invariably, specific binding was detected in the three organ types, at least in certain stages. In most of the cases, the intensity of staining exhibited developmental regulation. The apparent patterns reveal similarities between the different cell types, as seen with immobilizedN-acetylglucosamine as well as with labelled galectin-1 and sarcolectin. However, drastic differences among such patterns with nearly opposite developmental courses do also occur, as detected for carrier-attached mannose and maltose residues. These results point to a potential importance for the detected glycohistochemical features in human development and substantiate the possibility of differential regulation of the presence of binding sites for distinct sugars within a certain organ and between the individual cell types of the monitored organs.

    Notes: Abstract Various portions of the splanchnopleural mesoderm lateral to the somites of 1.5-day chick embryos were marked in ovo by local injection of Dil, and the distribution of the labelled cells in the digestive-tract mesoderm formed after 3 days' reincubation was analysed. The presumptive area of the digestive organs was confined to bands of splanchnic mesoderm lying lateral to the somites, on both sides, with a width two or three times that between the midline of the embryo and the lateral edge of the somite. Each band generally contributed cells to its own side of the digestive-tract mesoderm, except for the region around the bile duct. The anterior and posterior portion of the pre-gut area contributed cells to the anterior and posterior region of the digestive tract, respectively, but label originating from the portion furthest from the somite took the more ventral and posterior position. Thus, the presumptive areas of the respective digestive organs were located anteroposteriorly in the same order as in the digestive tract with their boundaries lying oblique to the embryonic axis.

    Notes: Abstract We have isolated three classes of “enhancertrap” lines of Drosophila in which lacZ expression patterns in the imaginal discs are consistent with the idea of a polar (radial and angular) coordinate system of positional information. In the first class (HZ76), a circular pattern was expressed transiently during the mid-third instar larval stage when the radial components of the coordinate are probably generated. The expression patterns of the second class (HZ84) were sector-shaped and circular in the leg disc, suggesting a correlation with both radial and angular coordinate values. The expression patterns found in the third class (PZ63 and PZ22) were circular and appeared to reflect radial positional values. Expression in the latter two classes always began in the presumptive dorsal region of the leg disc and gradually spread to the ventral region. These developmental profiles of expression suggested the existence of a centre that initiates patterned gene expression in the presumptive dorsal region of the leg disc. The PZ22 line showed transient expression during tarsal segmentation, suggesting its involvement in tarsal segment formation. We have cloned the PZ22 gene and partially determined its sequence. The deduced amino acid sequence contained a zinc finger motif found in DNA/RNA binding proteins. By in situ hybridization, we determined that the PZ22 gene was transcribed in the leg disc in a pattern identical to that of the lacZ expression. In addition, it was expressed transiently in the embryonic mesoderm during mesoderm segmentation. The PZ22 gene, therefore, may function both in mesodermal segmentation in the embryo and in tarsal segmentation in the imaginal disc.

    Notes: Abstract Two closely related cDNA fragments, named pTC14-1 and pTC14-2, encoding C-type lectins were cloned from the budding ascidian Polyandrocarpa misakiensis by means of the polymerase chain reaction. The amino acid sequence deduced from pTC14-1 was identical to that of a 14-kDa calcium-dependent galactose-binding lectin, TC-14, that had been purified from this species. Between the two clones, nucleotide sequence similarity was 90%, whilst that of the deduced amino acid sequences was 82%. The cDNA inserts of these clones hybridized weakly with each other. Antisense RNA probes prepared from these clones gave intense hybridization signals on Northern blots of the W strain, but very weak signals on those of the other strains. Therefore, both clones were suggested to originate from the W strain, but from two separate genes, since the base substitution was scattered throughout the entire translated region. The amount of TC14-1 mRNA increased during bud development, and peaked at 36 h after separation of the bud from the parental body wall. At this stage, extracellular matrix containing TC-14 lectin developed in the mesenchymal space around the morphogenetic region of the bud. There was much less TC14-2, than TC14-1 mRNA at every stage of bud development. TC14-1 and TC14-2 mRNAs were detected on Northern blots of RNAs from adults and growing buds, suggesting that these genes can be used as the earliest markers of budding in this species.

    Notes: Abstract A maternal protein showing a unique distribution during early Cynops embryogenesis was screened by monoclonal antibody. The antigen protein, designated as ABP-25 (animal blastomere protein, molecular weight 25,000), was distributed uniformly in the uncleaved egg and concentrated into blastomeres of the animal half during cleavage. At the blastula stage, ABP-25 was definitely localized in cells of the animal half and a polarized distribution was observed within the cytoplasm. During gastrulation, immunohistochemical analysis indicated that the reactivity of the marginal zone (presumptive mesoderm) to the monoclonal antibody ABP-25 decreased after involution. At the end of gastrulation, a polarized distribution was still clearly observed in the ventral epidermis, but not in the neuroectoderm. Both Western and Northern blots indicated that the amount of antigen protein and the intensity of gene expresion were almost constant until the neurula stage. The deduced amino acid sequence of the ABP-25 cDNA showed a strong homology (84%) with that of the pag gene associated with cell proliferation.

    Notes: Abstract The outline of the adult wing of lepidopteran insects (butterflies and moths) emerges as a result of disappearance of a group of cells at the periphery of the pupal wing. Histological observation of the pupal wing of Pieris rapae showed that, just after apolysis of the wing epithelium from the pupal cuticle, there occurs a rapid and localized decrease of the number of cells at the periphery of the wing. This decrease occurs through cell death, which lasts 1–1.5 days at 20°C. Dying cells lose contact with the neighbouring cells and show condensation of chromatin and cytoplasm. They then appear to be phagocytosed by neighbouring epithelial cells or discharged through the basal surface of the epithelium into the lumen within the wing and taken up by phagocytes. Fragmentation of DNA in the nuclei was detected in the dead cells or their debris. These results indicate that programmed cell death in the lepidopteran wing proceeds through a mechanism closely similar to that of apoptosis in the vertebrate.

    Notes: Abstract Fibronectin-like proteins were purified from ovaries of the sea urchin species, Paracentrotus lividus (PI), Sphaerechinus granularis (Sg), Arbacia lixula (Al), Pseudocentrotus depressus (Pd), and Anthocidaris crassispina (Ac), by gelatin-Sepharose affinity chromatography. The major component had a molecular mass of 180 kDa and was eluted by 1 M NaCl or 8 M urea, depending on the species used. By substrate adhesion assay, we tested the biological activity of the 180 kDa protein purified from Paracentrotus lividus (P1-180K) and showed that it promotes the adhesion of homologous embryonic cells to the substrate. An antiserum, developed against Temnopleurus hardwickii fibronectin-like protein (Th-180K), was used in Western blots of the proteins purified from the five species. The antibody cross-reacted with Pl-180K, Pd-180K and Ac-180K. A peptide map of P1-180K, obtained by V8 protease partial digestion, was compared with those obtained from the other four proteins and showed an homology between 40 and 56%. This report confirms that fibronectin-like proteins can be purified from sea urchins on the basis of their binding to gelatin-Sepharose; the proteins differ for their binding affinity to gelatin and share different epitopes, suggesting that they are members of a sea urchin fibronectin super family.

    Notes: Abstract Generating developmental gradients by temperature gradients established within a developing organism is an easy, non-invasive technique to study physiological interdependencies between locally separated subsystems. A linear temperature gradient of about 10° C/mm was maintained up to 5 h in either direction along the long axis of a long-germ-type hymenopteran egg, which was simultaneously filmed by the 16 mm timelapse technique. The result was a dramatic desynchronization of development, which between the egg poles could reach up to 9.3 h relative to normal development. Within the same egg, up to seven mitotic waves (i.e. eight different nuclear generations) were observed at the same time, and the subsequent cellularization process was extremely asynchronous. The “initial regions” of the mitotic waves, the “fountain flow” of the ooplasm, and the gastrulation process were shifted towards the egg pole kept at higher temperatures. Developmental processes occurring successively in normal development now took place simultaneously, with either direction of the temperature gradient. For instance, while gastrulation had started in the warm region, midblastula transition and cellularization were in progress in the middle of the egg, and intravitelline nuclear multiplication occurred at the cold pole, by rapid and still biphasic cell cycles. In some respects, development resembled that of a short-germ-type insect egg. Nevertheless, the developmental processes were resynchronized after the temperature gradient was switched off. Surprisingly, the extreme desynchronization during early development did not affect the segment pattern of the resulting embryos. The technique of inducing well-defined developmental asynchronies might be applied in Drosophila to analyse the subtle interplay between maternal and zygotic gene activities described in this species.

    Notes: Abstract Based on results of transplantation experiments, it has long been believed that trunk neural crest cells are incapable of chondrogenesis. When pigmented trunk neural crest cells of Ambystoma mexicanum are transplanted to cranial levels of albino (a/a) embryos, the graft cells ultimately produce ectopic fins, but are incapable of following the chondrogenic cranial neural crest pathways. Therefore, heterotopic transplantation does not expose these cells to the same environment experienced by cranial neural crest cells, and is neither an adequate nor a sufficient test of chondrogenic potential. However, in vitro culture of trunk neural crest cells with pharyngeal endoderm does provide a direct test of chondrogenic ability. That cartilage does not form under these conditions demonstrates conclusively that trunk neural crest cells possess no chondrogenic potential.

    Notes: Abstract Extracellular matrices (ECMs) of phylogenetically very distant organisms were tested for their ability to support cell adhesion, spreading and DNA replication in reciprocal xenograft adhesion tests. Mechanically dissociated cells of the medusa Podocoryne carnea (Cnidaria, Hydrozoa) were seeded on ECMs of polyps and medusa, and on several ECM glycoproteins or entire ECMs from vertebrates. In reciprocal experiments, cells from different vertebrate cell-lines were seeded on ECMs of polyps, medusae and also on electrophoresed and blotted extracts of both types of ECMs. The results demonstrate that medusa cells adhere and spread on polyp and medusa ECMs but do not recognize vertebrate ECMs or purified ECM glycoproteins. Vertebrate cells in contrast adhere, spread and proliferate on ECMs of polyps and medusae. The number of attached cells depends on the cell type, the type of ECM and, in certain cases, on the stage of the cell cycle. Cell adhesion experiments with pretreated ECMs of polyps and medusae, e.g. oxidation of carbohydrate residues with sodium-metaperiodate, or blocking of certain carbohydrate moieties with the lectin wheat germ agglutinin or a carbohydrate-specific monoclonal antibody, demonstrate that ECM carbohydrates are more important for cell-ECM interactions of medusa cells than for vertebrate cells. Furthermore, the experiments indicate that polyp and medusa ECMs contain different components which strongly modulate adhesion, spreading and DNA replication of vertebrate cells.

    Notes: Abstract To asses the potential of androgenetic cells to participate in post-midgestation fetal development we have made use of an in situ detectable cell lineage marker in the analysis of chimeric mouse fetuses containing an androgenetic cell lineage. Our results show conclusively that androgenetic cells participate in the formation of derivatives of all lineages and in some tissues may contribute the majority of the total cell population. However, the allocation or persistence of androgenetic cells was non-random. High contribution of androgenetic cells was observed in brown adipose tissue, mesenchyme, smooth muscle, perichondrium, peripheral nerves and epithelia of the intestinal tract and the trachea. Thus, androgenetic cells were able to efficiently populate mesodermal, ectodermal and endodermal derivatives. In contrast, there was a clear prejudice against androgenetic cells in the brain.

    Notes: Abstract We have used antibodies directed against a unique portion of the Drosophila POU domain protein Cfla to localize its sites of expression in developing embryos. Cfla protein is first detected during germ band extension in the tracheal placodes and in the midline mesectoderm cells. Tracheal expression continues throughout embryonic development, especially in the main longitudinal tracheal trunks. Additional sites of high Cfla expression are in the anterior portion of the hindgut, the roof of the stomodeum, a subset of central nervous system cells, the oenocytes, and the ring gland. In addition, Cfla expression was localized in embryos mutant for several loci involved in determining fate along the midline of the CNS and the tracheal system. Cfla midline cell expression is dependent on proper single-minded gene function, and Cfla either regulates or acts in parallel to the genes pointed and rhomboid during midline CNS and tracheal development.

    Notes: Abstract The properties of androgenetic cells and their ability to proliferate and differentiate were examined in post-midgestation chimeras. In several tissues, namely the brain, cardiac muscle, skeletal muscle and intestinal epithelium, the rate of proliferation of androgenetic cells was higher than that of normal cells in day 13 embryos. This higher rate of proliferation was however less pronounced by day 17–18 of development. It is possible that IGF2, a major growth factor regulating fetal growth, could play a role in the increased proliferation of androgenetic cells. Igf2 is also an imprinted gene that is expressed only when inherited paternally. Indeed, in the smooth muscle, cartilage and intestinal epithelium, patches of androgenetic (ag) cells exhibited higher levels of IGF2 mRNA than neighbouring wild-type cells. Surprisingly, we also detected expression of Igf2 in ag cells of ectodermal origin; this gene is not normally expressed in this lineage. This expression was observed in the brain, epidermis and in the epithelium of the tongue. We attempted to confirm the identity and differentiation status of ag cells with the help of cell-type specific antibodies and lectins. Evidence for only one of the cell types analysed, i.e. the goblet cells of the gut, suggests a delay or aberrant differentiation of ag cells.

    Notes: Abstract Denervation of the amputated limb of newts stops the regeneration process by decreasing blastema cell proliferation. We investigated the effect of the denervation on each of the two compartments (epidermal cap, mesenchyme) in mid-bud blastemas on the level of sulphated glycosaminoglycans (GAGS). Denervation resulted in an increase of about threefold in the incorporation of [35S] sulphate into mesenchyme GAGs but had no effect on the epidermal cap. The increase of GAG synthesis in the mesenchymal part of the blastema involved both heparan sulphates and chondroitin-dermatan sulphates. Gel filtration showed no change in GAGs size after denervation. These results confirm that the mesenchymal part of the mid-bud blastema is the main target of nerves and, as heparan sulphates are known to store acidic fibroblast growth factor (aFGF), a polypeptide found in the blastema (Boilly et al.. 1991), this suggest that the nerves' effect on glycosaminoglycans turnover could be implicated in the control of bioavailability of this growth factor in the blastema.

    Notes: Abstract Migration of neurons over long distances occurs during the development of the adult central nervous system of the sphinx moth Manduca sexta, and the turnip moth Agrotis segetum. From each of the suboesophageal and three thoracic ganglia, bilaterally-paired clusters of immature neurons and associated glial cells migrate posteriorly along the interganglionic connectives, to enter the next posterior ganglion. The first sign of migration is observed at the onset of metamorphosis, when posterio-lateral cell clusters gradually separate from the cortex of neuronal cell bodies and enter the connectives. Cell clusters migrate posteriorly along the connective to reach the next ganglion over the first three days (approximately 15%) of pupal development. During migration, each cell cluster is completely enveloped by a single giant glial cell spanning the entire length of the connective between two adjacent ganglia. Intracellular cobalt staining reveals that each migrating neuron has an ovoid cell body and an extremely long leading process which extends as far as the next posterior ganglion; this is not a common morphology for migrating neurons that have been described in vertebrates. Once the cells arrive at the anterior cortex of the next ganglion, they rapidly intermingle with the surrounding neurons and so we were unable to determine the fate of the migrating neurons at their final location.

    Notes: Abstract We describe the development of 20 sensory organs in the embryonic Drosophila head, which give rise to 7 sensory nerves of the peripheral nervous system (PNS), and 4 ganglia of the stomatogastric nervous system (SNS). Using these neural elements and the optic lobes as well as expression domains of the segment polarity gene engrailed in the wild-type head of Drosophila embryos as markers we examined the phenotype of different mutants which lack various and distinct portions of the embryonic head. In the mutants, distinct neural elements and engrailed expression domains, serving as segmental markers, are deleted. These mutants also affect the optic lobes to various degrees. Our results suggest that the optic lobes are of segmental origin and that they derive from the ocular segment anteriorly adjacent to the antennal segment of the developing head.

    Notes: Abstract Using the conserved basic helix-loop-helix (bHLH) domain of AS-C and MASH, we amplified by polymerase chain reaction, the HLH region of a chick AS-C/MASH homologue from cDNA prepared from poly (A+) RNA isolated from a region rich in cardiac neural crest cells. A full-length cDNA clone was obtained from a stage 23 chick head cDNA library using this conserved sequence as a probe. Comparison of this clone with MASH1 revealed strong homologies in the bHLH domain and carboxyl terminal region of the conceptually translated protein. The clone was named chick achaete-scute homologue (CASH). This transcript has a single open reading frame that codes for a protein of 218 residues. The deduced amino acid sequence of CASH is 100% identical to MASH1 in the bHLH region and 83.9% identical over the entire protein. The bHLH domain of CASH is highly conserved within the AS-C family. Northern analysis indicates that CASH is expressed transiently in a defined spatial pattern. The highest expression is in the head between stages 23 and 26. Whole mount in situ hybridization suggests that CASH is expressed at stage 23 in the dorsal ventricular zone and central sensory pathways and later in the ventral ventricular zone and ventral horn. By stage 28, it is expressed ubiquitously in the peripheral nervous system including all of the neurons derived from the the neural crest and epipharyngeal placodes, as well as neural crest-derived chromaffin cells in the adrenal medulla. Amplification by polymerase chain reaction indicates that CASH is expressed in the cardiac neural fold and continues to be expressed in cultured cardiac neural crest cells.

    Notes: Abstract In the honey bee, Apis mellifera, the fifth larval instar is a critical period for caste differentiation. During this premetamorphic phase the hormonal milieu shows pronounced caste differences and several organs, particularly the ovaries, enter different developmental pathways leading to highly fertile queens and nearly sterile workers. Developmental profiles of total protein synthesis in larval ovaries showed marked caste differences starting with the early fifth instar. By two-dimensional electrophoresis, caste-specific patterns could be detected in the synthesis of a 29 kDa/pI 4.6 and two 24 kDa/pI 5.2–5.5. proteins (pI=isoelectric point). A marked decrease in the expression of these proteins was found to coincide with caste-specific differences in the haemolymph ecdysteroid titer. In vitro exposure of larval worker ovaries to physiological (10−7 m) concentrations of synthetic makisterone A elicited an identical response. Juvenile hormone did not affect protein synthesis patterns in larval ovaries, and also did not inhibit or reverse the ecdysteroid-induced effects. Heat shock experiments revealed that the 29 kDa/pI 4.6 ecdysteroid-regulated protein belongs to the class of small heat shock proteins.

    Notes: Abstract Tentacles excised from syphistoma polyps of Aurelia aurita undergo rapid regeneration to form whole polyps following exposure to an excess or absence of specific ions. It has been shown that a 12–18 h exposure of isolated tentacles to 58 mM excess of Cs+ results in a rapid firing of nematocysts, followed by an accelerated, synchronous polyp morphogenesis. Absence of Mgt2+ from the culture solution for 4–24 h also led to an accelerated, synchronous polyp regeneration. In either experimental set-up, incubation in 5–10 mM hydroxyurea effectively halted regeneration. Exposure to an excess of Li+ (50–200 mm) or K+ (10–50 mM) caused no firing of nematocysts and a percentage of polyp regeneration only slightly higher than control tentacles. Use of the K+ channel blocker tetraethylammonium (TEA; 100–300 mM) lead to similar levels of regeneration. A Ca2+ or K+-reduced artificial culture solution did not enhance regeneration. Ouabain (1 mM) dampened the Cs+ induced acceleration of polyp morphogenesis, and when given without Cs+, elicited a control level response.

    Notes: Abstract Sizes of yolk platelets were measured in sections of oocytes and embryos in Xenopus. It was found that the average size of the largest group of platelets in cells differed between germ layers of neurulae. It was small (3 to 5 μm) in the ectoderm, medium-sized (5 to 8 µm) in the mesoderm, and large (over 8 μm) in the endoderm. Platelets of these size classes formed layers in egg, the yolk gradient, by the end of oocyte maturation. The yolk gradient contained products of the mitochondrial cloud and a part of the germinal vesicle material at certain positions. The layers of small, medium and large platelets in the egg changed their locations to distribute to the ectoderm, mesoderm and endoderm of neurulae, respectively. The yolk layers in the egg thus represented different prospective fates, and a figure describing the locations of these layers could be regarded as a fate map for the one-cell stage. Most of the marginal blastomeres of embryos at cleavage stages consisted of a few parts with different prospective fates. Results were discussed with reference to available fate maps for cleavage stage embryos.

The data include video time-lapse recordings of living embryos and bisbenzimide stains of fixed embryos as well as morphometry on fixed, serially-sectioned embryos. We present the cell lineage and cell cycle durations up to the first indication of symmetrization within this embryo. We show that early cell cycles last approximately 1h. A dramatic extension of cell cycle duration and a concomitant asynchrony among the various cell lines was observed starting at the fifth cleavage. Short cell cycles, like those of early blastomeres, were a constant property of the largest descendants of the 2d-cell line only. In contrast to Meisenheimer's observations and our experiences with other spiralian embryos, the cleavage pattern proved to follow multiple alternatives. The embryonic quadrants A-D were arranged in either a clockwise or counter-clockwise fashion and the chirality of the third cleavage was either dextral or sinistral irrespective of the arrangement of the quadrants. As a consequence, four different blastomere configurations were encountered and the dorsoventral axis could take four different angles with respect to the plane of first cleavage. The dorsal side was most easily recognized by the position of the 2d-micromere at the 16-cell stage. The fact that all of such embryos could develop into normal, uniform larvae is interpreted as the result of cell-cell interactions in morphogenetic regulation.

    Notes: Abstract The late stages of stenotele development and the migration and installation of freshly matured stenoteles in Hydra have been studied by kinetic and immunofluorescence investigations with rhodamine-labelled polyps. It was found that the high concentration of osmotic pressure-generating poly(γ-glutamic acid)s is synthesised exclusively within the lumen of the immature nematocyst. Assembly of the polymers, which is completed after approximately 0.5 days, is accompanied by a swelling of the capsule and ends when the cyst is mature. Active migration of the stenoteles to the tentacles begins only about 1.0 day later, and the time required for installation of a stenotele on the outer surface of the tissue amounts to about another 1.5 days. Furthermore, the results obtained suggest that the disintegration of the clusters of growing stenoteles, which begins 0.5 to 1.0 days before maturation, is a passive process; the ability of a nematocyte to migrate actively to the tentacles is acquired after maturation and might be controlled directly by regulating factors contained in the tissue.

    Notes: Abstract The Drosophila melanogaster segmentation gene fushi tarazu (ftz) encodes a homeodomain-type transcription factor involved in the control of larval pattern formation. Loss of function mutations cause an embryonic lethal, pair-rule phenotype. The segmentation defects, but not the lethality, can be partially rescued by the ftz orthologue from Drosophila hydei. In this work, the primary structure, expression and regulation of the D. hydei ftz gene was characterized. Sequence comparisons classify ftz as a rather fast evolving gene. However, since the homeodomain of the D. hydei FTZ protein is highly similar to that of D. melanogaster, proper regulation of D. melanogaster ftz downstream genes would be expected. In D. melanogaster embryos, a D. hydei ftz transgene is expressed normally, independent of endogenous ftz gene activity, suggesting that D. hydei ftz regulatory sequences are correctly recognized by D. melanogaster transcription factors. Accordingly, lacZ fusion constructs driven by the D. hydei ftz upstream element are expressed normally in D. melanogaster embryos. Altogether, the similarities between the two ftz orthologues by far outweigh the differences. The limited success of the trans-species rescue might be, therefore, a consequence of the accumulation of too many subtle changes in gene function, exceeding the limits of developmental plasticity during fly embryogenesis.

    Notes: Abstract We have used the axolotl oocyte (Ambystoma mexicanum Shaw) to study the stability of exogenously injected Xenopus RNAs. Three different cellular developmental stages have been analysed: (1) the growing oocyte (stage III–IV of vitellogenesis), (2) the full-grown oocyte at the end of vitellogenesis (stage VI) and (3) the progesterone-matured stage VI oocyte. Three exogenous RNAs have been synthesized in vitro from a c-myc Xenopus cDNA clone. One transcript is 2.3 kb long (full length), the second is 1.5 kb long, with most of the 3′ untranslated region (3′UTR) removed, and the third corresponds to the 3′UTR (0.8 kb). After injection or coinjection of these exogenous Xenopus RNAs into axolotl oocytes, the stability of the molecules was studied after 5 min, 6 h and 21 h by extraction of total RNA and Northern blot analysis. Results show a difference in Xenopus RNA stability during axolotl oogenesis. In growing oocytes, the three synthetic transcripts are gradually degraded. The absence of the 3′UTR is not therefore sufficient to stabilize the transcript during early oogenesis. No degradation is observed in full-grown oocytes, suggesting the existence of stabilizing factors at the end of oogenesis. When stage VI oocytes are induced to mature by progesterone, only the 2.3 and 1.5 kb Xenopus RNAs disappear. This suggests a role for germinal vesicle breakdown in this degradation process as well as the existence of a factor present in the nucleus and involved in the specific destabilization of these RNAs after oocyte maturation. This degradation might implicate several destabilizing sequences localized in the coding or in the 3′UTR of the c-myc gene. In contrast, the 0.8 kb transcript (3′UTR) is not degraded during this period and remains very stable. Therefore, degradation appears distinct from one transcript to another and from one region to another within the same molecule. During maturation, the behaviour of the 2.3 and 1.5 kb transcripts is different when coinjected with the 3′UTR, suggesting a role in trans of this untranslated molecule in c-myc stability. Our approach allows us to analyse the role of the coding and 3′UTR regions of the c-myc RNA in the control of mRNA degradation in vivo.

    Notes: Abstract We have previously described an in vivo heterologous system which has allowed us to study the stability of different Xenopus c-myc RNA constructs injected into axolotl oocytes. In full-grown oocytes, degradation of c-myc RNA does not occur. In mature oocytes treated with progesterone, transcripts containing the coding sequence of the gene are degraded, whereas those corresponding to the 3'UTR (untranslated region) alone are stable. In order to study the role of nuclear or cytoplasmic components in this process, degradation of injected c-myc transcripts was analysed (i) after inhibition of germinal vesicle breakdown (GVBD) in progesterone treated oocytes (ii) after induced maturation of enucleated oocytes, (iii) injection of nuclear contents into immature oocytes and (iv) after direct injection into the germinal vesicle of full-grown oocytes. Our results demonstrate the role of a nuclear factor stockpiled in the germinal vesicle of full-grown oocytes and specifically involved in the degradation of c-myc transcripts containing the coding region. Further biochemical characterization of this new nuclear component should lead to a better understanding of the post-transcriptional control of c-myc expression during oogenesis and early development.

    Notes: Abstract XCsx2, a homeobox-containing gene, is expressed in cardiac muscle during Xenopus development, while the XMEF2A gene is expressed in both cardiac and skeletal muscle. Microinjection of either XCsx2 or XMEF2A mRNA into single blastomeres of two-cell stage Xenopus embryos induced precocious expression of the myosin heavy-chain alpha (XMHCα) gene at the neural plate stage (stage 14). Co-injection of both XCsx2 and XMEF2A mRNAs induced still earlier expression at the late gastrula stage (stage 12). These changes were evident in whole embryos but not in animal pole explants from injected embryos. Overexpression of XCsx2 or XMEF2A also caused an enlarged heart and abnormalities of notochord and tail in Xenopus embryos. These findings suggest that both XCsx2 and XMEF2A transcription factors have an important role in regulating the expression of the XMHCα gene and the morphogenesis of heart tissue in Xenopus development.

    Notes: Abstract Parthenogenetic cells are lost from fetal chimeras. This may be due to decreased proliferative potential. To address this question, we have made use of combined cell lineage and cell proliferation analysis. Thus, the incorporation of bromodeoxyuridine in S-phase was determined for both parthenogenetic and normal cells in several tissues of fetal day 13 and 17 chimeras. A pronounced reduction of bromodesoxyuridine incorporation by parthenogenetic cells at both developmental stages was only observed in cartilage. In brain, skeletal muscle, heart and intestinal epithelium, this reduction was either less pronounced or observed only at one of the developmental stages analysed. No difference between parthenogenetic and normal cells was observed in epidermis and ganglia. Our results show that a loss of proliferative potential of parthenogenetic cells during fetal development contributes to their rapid elimination in some tissues. The analysis of the fate of parthenogenetic cells in skeletal muscle and cartilage development demonstrated different selection mechanisms in these tissues. In skeletal muscle, parthenogenetic cells were largely excluded from the myogenic lineage proper by early post-midgestation. In primary hyaline cartilage, parthenogenetic cells persisted into adulthood but were lost from cartilages that undergo ossification during late fetal development.

    Notes: Abstract Differentiation tendency and the inducing ability of the presumptive ectoderm of newt early gastrulae were examined after treatment with activin A at a high concentration (100 ng/ml). The activin-treated ectoderm differentiated preferentially into yolk-rich endodermal cells. Combination explants consisting of three pieces of activin-treated ectoderm formed neural tissues and axial mesoderm along with endodermal cells. However, the neural tissue was poorly organized and never showed any central nervous system characteristics. When the activin-treated ectoderm was sandwiched between two sheets of untreated ectoderm, the sandwich explants differentiated into trunk-tail or head structures depending on the duration of preculture of activin-treated ectoderm in Holtfreter's solution. Short-term (0–5 h) precultured ectoderm induced trunk-tail structures accompanied by axial organs, alimentary canal and beating heart. The arrangement of the explant tissues and organs was similar to that of normal embryos. However, archencephalic structures, such as forebrain and eye, were lacking or deficient. On the other hand, long-term (10–25 h) precultured ectoderm induced archencephalic structures in addition to axial organs. Lineage analysis of the sandwich explants using fluorescent dyes revealed that the activin-treated ectoderm mainly differentiated into endodermal cells and induced axial mesoderm and central nervous system in the untreated ectoderm. These results suggest that activin A is one of the substances involved in triggering endodermal differentiation and that the presumptive ectoderm induced to form endoderm displays “trunk-tail organizer” or “head organizer” effects, depending on the duration of preculture.

    Notes: Abstract As the first five cleavages of the Patella vulgata embryo are synchronous, they are well suited to determine the mRNA level of cyclin A and B genes in an embryo. During the third and fourth cleavage cycle the quantity of A and B mRNA is regulated in a cell-cycle-dependent way, reaching a high level between cleavages and a lower level just after mitosis. This implies that transcription of the cyclin genes occurs before the overall transcription increases directly after the fifth cleavage. During the first cleavages cyclin A and B mRNA is localized in distinct parts of the cytoplasm. Between two successive cell devisions it is found as a crescent-shaped domain at the peripheral side of the nucleus. At cytokinesis it is present between two separating nuclei and at newly formed cell membranes. At the fifth cleavage this localization disappears. Changes in the expression pattern of cyclin A and B may be expected after the fifth cleavage, when the first cells become arrested in cell division and differentiate. The mechanism causing cell division arrest of these primary trochoblasts is still unknown. Cell division arrest caused by the absence of cyclin A and/or B mRNA could be conditional for further differentiation. However, a decrease in cyclin A and B mRNA level in the trochoblasts is not detectable until 4 h after their last division. Later in development no cyclin A and B mRNA can be detected in these cells, whereas cyclin A and B mRNA is present in other cells of the embryo. Thus, the absence of cyclin A and B mRNA in primary trochoblasts, and in the later differentiating secondary and accessory trochoblasts is not obligatory for cell division arrest or cell differentiation.

    Notes: Abstract An antiserum to transdifferentiated striated muscle cells from the medusa of Podocoryne carnea was prepared and used to screen a λ gt11-expression library prepared from gonozoids of P. carnea. We isolated a cDNA clone termed Pod-EPPT with at least 63 tandem repeats of the tetrapeptide-motive glu-pro-pro-thr, named Pod-EPPT. Using Pod-EPPT as a molecular marker for head quality the morphological relationship between the two metagenic life stages of this hydroid, the polyp and the medusa, was studied. In situ hybridization demonstrated that expression of the gene corresponding is restricted to secretory cells in the endoderm of the oral hypostome region of polyps and medusae and, presumably, to progenitor cells of this type. Cells expressing Pod-EPPT could not be observed in the larval stage. During head regeneration in polyps, Pod-EPPT expression is upregulated soon after head removal in previously non-expressing cells and in newly differentiating secretory cells. This activation of a head-specific gene precedes the morphologically obvious events of head regeneration. Pod-EPPT is one of the genes that are activated during manubrium (mouth) regeneration from experimentally combined subumbrellar plate endoderm and striated muscle of the medusa.

    Notes: Abstract Spatial distribution of mRNAs for activin receptors and follistatin was studied by Northern blot hybridization using RNAs from different parts of dissected Xenopus embryos. mRNAs of two activin receptors (type IIA and IIB) occurred uniformly in pre-gastrular embryos, but occurred in larger amounts in ectoderm (in gastrulae), neural plate (in neurulae) and anterior (head) regions (in tailbud embryos) than in other embryonic regions. By contrast, follistatin mRNA appeared almost exclusively in the dorsal mesoderm including invaginating organizer region at the gastrula stage, in notochord and in dorsal ectoderm at the neurula stage, then in anterior part at the tailbud stage. The localized patterns of the distribution of these mRNAs may be due to the regionally different zygotic expression of genes in embryos at later stages. From the relatively widespread pattern of distribution of their mRNAs, we assume that both type IIA and type IIB activin receptors have broad functions in ectodermal and neural differentiation. On the other hand, follistatin mRNA showed quite a restricted pattern of expression, and therefore, we assume that follistatin may have functions more specifically related to the sites of expression of its mRNA. Thus, follistatin may be involved in the differentiation of notochord itself and/or directly be responsible for organizer functions such as neural induction and subsequent differentiation of induced neural tissues at the gastrula and later stages.

    Notes: Abstract New antibody markers have allowed more refined examinations of embryogenesis. Features are being found that were overlooked in whole and sectioned embryos stained with traditional histochemical labels. Two monoclonal antibodies that recognize two different cell surface proteins in Manduca sexta label cells of the developing reproductive system. These specific immunolabels reveal that during a brief period of Manduca embryogenesis, rudiments of both male and female genital ducts are present in a single embryo. This transient phase of genital differentiation parallels the transient indifferent stage known to occur during development of reproductive systems in many vertebrate embryos. At the end of this indifferent stage, one of the two pairs of genital ducts retracts and degenerates. The dynamic expression of the two surface proteins on cells involved in morphogenesis of both the female and male reproductive systems also suggests that these proteins are important in orchestrating the specific cellular interactions that occur between mesodermal cells of the genital ducts and the nearby ventral ectoderm.

    Notes: Abstract We have studied a group of midline cells in the embryonic brain of the grasshopper by using immunocytochemical and intracellular dye injection techniques. This cluster of midline cells differentiates between the pars intercerebralis lobes of the protocerebrum during early embryogenesis, and is composed of putative midline progenitors as well as neuronal and glial cells. Annulin immunoreactive glial processes surround the borders of the midline cell cluster and also form a network of processes extending from there to the borders of proliferative clusters in the brain hemispheres. Among the cells that derive from the midline cluster are two bilaterally symmetrical pairs of identified primary commissure pioneer neurons. By navigating along the glial bound borders of the midline proliferative cluster, the axons of these pioneers establish an initial axonal bridge across the brain midline. This analysis identifies a glial-bound midline proliferative cluster in the brain and shows that neuronal and glial cells of this cluster are closely associated with neurons pioneering the primary brain commissure. Comparable features of midline cells in the ventral ganglia and similarities to other proliferative clusters in the brain hemispheres are discussed.

    Notes: Abstract The adult antennal lobe of Drosophila melanogaster emerges from a precursor, the larval antennal lobe. Pulse and pulse-chase labelling of dividing cells in larvae and pupae with bromodeoxyuridine confirmed previous data that some of the interneurons of the adult antennal lobe derive from a lateral neuroblast which starts to divide early in the first larval instar. However, the majority of these interneurons originate from neuroblasts that initiate mitosis at later stages, with a peak of about 10–12 pairs of dividing neuroblasts in the late third larval instar. No clustering of adult antennal lobe neurons according to their birthdates was observed. In contrast to neurons, terminal divisions of glia in the antennal lobe reach their maximum only 12 h after puparium formation.

    Notes: Abstract Oocytes of hymenopterans are equipped with peculiar organelles termed accessory nuclei. These organelles originate from the germinal vesicle (oocyte nucleus) and gather preferentially at the anterior pole. To gain insight into the mechanism of uneven (asymmetrical) distribution of accessory nuclei, the organization of the microtubule cytoskeleton in the oocytes of two hymenopterans Chrysis ignita and Cosmoconus meridionator has been studied. It is shown that during late previtellogenesis two networks of microtubules are present along the contact zone between the oocyte and enveloping follicular epithelium. The external one is associated with belt desmosomes connecting neighbouring follicular cells. The internal network is composed of randomly orientated microtubules and separates transparent, organelle-free periplasm from the endoplasm. All cellular organelles and the germinal vesicle are localized in the endoplasm. Accessory nuclei are accumulated in the anterior endoplasm; they always lie in direct contact with the subcortical network. Treatment with colchicine results in the disappearance of the periplasm as well as in the redistribution of cellular organelles including accessory nuclei. Presented findings suggest that subcortical microtubules play an important role in the positioning of accessory nuclei throughout the ooplasm.

    Notes: Abstract The effects of juvenile hormone (JH) and 20-hydroxyecdysone (20E) on the developmental expression of the two insecticyanin genes, ins-a and ins-b, were investigated with two gene-specific probes. Removal of the corpora allata (-CA, source of JH) clearly delayed and down-regulated the epidermal expression of these genes but enhanced their expression in the fat body during the early development of the fifth instar. Application of JH I to the -CA larvae at the time of head capsule slippage completely restored the normal epidermal expression pattern of the two genes in the early fifth instar, then INS-a mRNA declined prematurely whereas INS-b mRNA remained similar to that in the intact larvae. By contrast, in the fat body of -CA larvae, the exogenous JH had little effect on the levels of INS-a mRNA, but enhanced expression of INS-b mRNA relative to intact larvae. Culture of epidermis from day 1 fifth instar larvae with 40 ng/ml 20E for up to 24 h accelerated the loss of INS-a mRNA without affecting the levels of INS-b mRNA. Both mRNAs declined in isolated larval abdomens over a 24 h period, and this decline was slowed by 1 μg methoprene (a JH analog). Together these results indicate that JH controls the levels of the two mRNAs in both the epidermis and fat body, with additional factors involved in regulating these genes in the fat body during the molt and in the epidermis during the growth phase.

    Notes: Abstract The proctodeum of the Drosophila embryo originates from the posterior end of the blastoderm and forms the hindgut. By enhancer-trap mutagenesis, using a P-element-lacZ vector, we identified a mutation that caused degeneration of the proctodeum during shortening of the germ band and named it aproctous (apro). Expression of the lacZ reporter gene, which was assumed to represent expression of the apro gene, began at the cellular blastoderm stage in a ring that encompassed about 10–15% of the egg's length (EL) and included the future proctodeum, anal pads, and posterior-most part of the visceral mesoderm. In later stages, strong expression of lacZ was detected in the developing hindgut and anal pads. Expression continued in the anal pads and epithelium of the hindgut of larvae; the epithelium of the hindgut of the adult fly also expressed lacZ. The spatial patterns of the expression of lacZ in various mutants suggested that the embryonic expression of apro was regulated predominantly by two gap genes, tailless (tll) and huckebein (hkb): tll is necessary for the activation of apro, while hkb suppressed the expression of apro in the region posterior to 10% EL. Cloning and sequencing of the apro cDNA revealed that apro was identical to the T-related gene (Trg) that is a Drosophila homolog of the vertebrate Brachyury gene. apro appears to play a key role in the development of tissues derived from the proctodeum.

    Notes: Abstract Retinoic acid has been linked to pattern formation in the vertebrate anteroposterior axis. This report describes the spatial and temporal distributions of both endogenous retinoic acid and retinoic acid synthase activity along the anteroposterior axis of neurulating zebrafish embryos, as detected by a transient transgenic assay and by a zymography bioassay. Both retinoic acid levels and synthase activity were found to be highest in anterior regions of the trunk at all of the stages which were analysed. The drug disulfiram inhibited retinoic acid synthase activity in the zebrafish trunk both in vitro and in vivo, and reduced retinoic acid levels in vivo. Disulfiram treatment of neurulating embryos resulted in larvae with hypertrophic wavy notochords, shortened spinal cords and deformed pectoral fins. The results support the hypothesis that retinoic acid plays a role in the coordination of axial patterning at the developing node/zone of involution, as well as in the subsequent development of anterior trunk structures such as the fins.

    Notes: Abstract In the non-feeding larva of the marine gastropod, Haliotis rufescens, gut morphogenesis is initiated at metamorphosis. Intestine-specific chymotrypsin gene expression begins in amoebocytes located in the dorsoposterior region of the undifferentiated digestive gland prior to metamorphosis, 5 d post-fertilization. Transcript accumulates steadily in these cells over the next 6 d while the amoebocytes migrate slowly dorsally. Induction of metamorphosis dramatically accelerates the rates of chymotrypsin mRNA accumulation and amoebocyte migration, and is required for homing of the amoebocytes to the hindgut region. Induction of chymotrypsin gene expression occurs only in larvae that had developed competence to recognize an exogenous morphogenetic cue and initiate metamorphosis, with a more pronounced increase in chymotrypsin mRNA accumulation in occurring older larvae. Chymotrypsin mRNA accumulation patterns suggest that hindgut cell specification occurs prior to metamorphosis, but that completion of the morphogenetic program requires signaling events associated with metamorphosis.

    Notes: Abstract Developmental expression of transduced mini-white(w) gene of Drosophila is sensitive to its flanking genomic enhancers. Taking advantage of this phenomenon, we mobilized a P lacW transposon and screened for new transposant lines which showed patterned expression of the mini-w gene in adult eyes. From a screen of about 1,000 independent P lacW transposant lines on the second chromosome, we identified 7 lines which showed patterned w expression in adult eyes. These P insertions were assigned to engrailed, wingless and teashirt genes based on their chromosomal locations, developmental expression of the lacZ reporter gene, lethal embryonic mutant phenotypes and, finally, their failure to complement the lethal alleles of the respective genetic loci. Our results show that although only a small fraction of the total transposant lines displayed patterned w expression, the genetic loci thus identified are those which play essential roles in pattern formation. Scopes of screens for genetic loci based on w reporter gene expression in adult eyes are discussed.

    Notes: Abstract The development of the autonomic ganglia of Auerbach's plexus and gizzard smooth muscle was studied in chicken embryos. Nervous system and smooth-muscle-specific antibodies were employed in immunofluorescence stainings on tissue sections to investigate the temporal and spatial frame of neural and muscular differentiation in relation to each other. Subserosal clusters of neural cells were clearly demonstrable at embryonic day 5 (ED5), the earliest stage analysed, with the monoclonal antibody El (SGIII-1). Fine nerve fibres (ED6) and, later, large axon bundles projecting from subserosal neuron clusters towards the lumen were followed and found to reach the luminal border by ED11. Already in early development the area of the future laminar tendons on the ventral and dorsal surface of the gizzard was devoid of neuroblasts, and nerve fibres were not extending to the muscle-tendon borderline until ED16. Double stainings with antibodies to smooth muscle myosin (SMM) and El revealed that SMM expression, taken as an indicator for muscle differentiation, followed neural growth. It was first detectable in close apposition to the differentiating neuroblasts in the caudal and cranial portion of the gizzard at ED6. With further development, myosin expression proceeded inward towards the lumen in a wave which followed the ingrowth of E1-positive nerve fibres from the prospective Auerbach plexus. Neuromuscular differentiation deviated from this pattern in the lateral tendon area where nerve growth was delayed and myosin expression preceeded the arrival of E1-positive nerve fibres. The findings suggest that the gizzard could serve as a model system for the analysis of potential early nervous system imprints on smooth premuscle mesenchyme differentiation.

    Notes: Abstract A novel biologically active peptide (metamorphosin A, MMA, pEQPGLW.NH2) has recently been described. It was isolated from Anthopleura elegantissima and triggers metamorphosis in Hydractinia echinata. Antibodies directed against the C-terminal part of the molecule immunohistochemically stain neurosensory cells and processes in the anterior part of larvae of H. echinata. We assume that in metamorphosis MMA (or a closely related LW-amide) is an internal signal transmitted from the anterior to the posterior body parts. Immunoreactivity is also found in ectodermal nerve processes — but not cell bodies — in the tentacles and in the basal disk of the foot of Hydra magnipapillata. This is, to our knowledge, the first report of LW-amide(s) as (a) neuropeptide(s).

A GAL4 enhancer-trap screen for glial-specific expression was performed. Using UAS-lacZ and UAS-kinesin-lacZ as reporter constructs, we describe the distribution and morphology of the identified glial cells in the fully differentiated ventral nerve cord of first-instar larvae just after hatching. The three-dimensional structure of the glial network was reconstructed using a computer. Using the strains with consistent GAL4 expression during late embryogenesis, we traced back the development of the identified cells to provide a glial map at embryonic stage 16. We identify typically 60 (54–64) glial cells per abdominal neuromere both in embryos and early larvae. They are divided into six subtypes under three categories: surface-associated glia (16–18 subperineurial glial cells and 6–8 channel glial cells), cortex-associated glia (6–8 cell body glial cells), and neuropile-associated glia (8–10 nerve root glial cells, 14–16 interface glial cells, and 3–4 midline glial cells). The proposed glial classification system is discussed in comparison with previous insect glial classifications.

    Notes: Summary Vitellin from the pharaoh's ant,Monomorium pharaonis (L.), was found to contain a single apoprotein with aM r of 198.3 kDa. The protein is a glycoprotein exposing mannose containing carbohydrate groups. Antibodies to pharaoh's ant vitellin (v t), raised in rabbits, were used to determine the head-, thorax-, and abdominal contents of vitellogenin and vitellin (v g+vt) in queens in relation to the presence or absence of larvae and workers using an ELISA test. Thev g+vt contents were also compared to the egg-laying rate and the weight of the queens. The results confirm the existence of a positive correlation betweenv g+vt contents in the queens and their access to larvae, probably related to the queens' preferential feeding on larval secretions. In queens without larvae the abdominalv g+vt contents declined in concordance with a low oviposition rate of 6–8 eggs/day. In spite of cessation of egg-laying within 24 hours after removal of both larvae and workers, the queens maintained basal contents ofv g+vt. This may indicate that the presence of larvae is not only essential for the nutrition of the queens, but also for the uptake of vitellogenin in the growing oocytes. This additional stimultive factor may be based on the queens' response to primer pheromones liberated by the larvae. v g+vt could not be demonstrated in workers or larvae with the ELISA test. If anyv g+vt is present in larvae and workers the amount is lower than the detection limit (1–2 ng/individual). This seems to rule out the possibility of transfer of proteins of this kind to the queens.

While the secondary sex ratio inS. dumicola was female-biased, the overall operational sex ratio (numbers of breeding males to breeding females over the season) showed no strong female bias. Males matured before females and had a shorter lifespan than the females. Mating took place in the natal colony. Males fought over access to the few mature females available early in the reproductive season, but females appeared to control the duration of mating. Later in the season, some adults of both sexes dispersed alone to breed elsewhere. We conclude that different rates of maturation between the sexes within a colony provide the opportunity for females that mature early in the season to be choosy in selecting a mate and this forces males to compete. Early reproduction may be beneficial for both females and males, because the offspring of females that reproduce early may have a competitive advantage over later (and smaller) offspring in the colony.

    Notes: Summary Intraspecific interference competition in the harvester ant,Messor aciculatus, was studied. Colonies of this species were found not to have territories. Some nests were located very close to each other, and the foraging areas of the neighbors usually overlapped. Even though the frequency with which alien and resident ants met was very high in the vicinity of the nest entrances, aggressive interactions between them rarely occurred. However, when hostile workers encountered each other, they exhibited a kind of ritualized combat and the winner ejected, but did not injure the loser. If any aliens entered the nest, some of them were pulled out, mainly by the residents. Aliens roaming near a neighbor's nest entrance ferociously attacked the residents carrying seeds in their mandibles and robbed them. On other occasions, aliens entered the nest and stole the collected seed. Although seed robbing and stealing occurred among neighboring colonies, there were remarkable differences in the frequency of their occurrence. The results of field observations and experiments suggest the existence of a dominance order among the neighbors. In one instance, extermination of an inferior colony by its neighbor was observed. The raider colony transferred the stored seeds from the nest of the inferior colony to its own and deposited the larvae and workers some distance away from the nest. The influence of ritualized combat and food robbing on colony activities, and the ecological significance of this interference behavior in terms of spatial distribution and temporal persistence of the nest sites, is discussed.

    Notes: Abstract We analyzed the broadband body waves of the 1992 Nicaragua earthquake to determine the nature of rupture. The rupture propagation was represented by the distribution of point sources with moment-rate functions at 9 grid points with uniform spacing of 20 km along the fault strike. The moment-rate functions were then parameterized, and the parameters were determined with the least squares method with some constraints. The centroid times of the individual moment-rate functions indicate slow and smooth rupture propagation at a velocity of 1.5 km/s toward NW and 1.0 km/s toward SE. Including a small initial break which precedes the main rupture by about 10 s, we obtained a total source duration of 110 s. The total seismic moment isM o =3.4×1020 Nm, which is consistent with the value determined from long-period surface waves,M o =3.7×1020 Nm. The average rise time of dislocation is determined to be τ≈10 s. The major moment release occurred along a fault length of 160 km. With the assumption of a fault widthW=50 km, we obtained the dislocationD=1.3 m. From τ andD the dislocation velocity isD=D/τ≈0.1 m/s, significantly smaller than the typical value for ordinary earthquakes. The stress drop Δσ=1.1 MPa is also less than the typical value for subduction zone earthquakes by a factor of 2–3. On the other hand, the apparent stress defined by 2μE s /M o , where μ andE s are respectively the rigidity and the seismic wave energy, is 0.037 MPa, more than an order of magnitude smaller than Δσ. The Nicaragua tsunami earthquake is characterized by the following three properties: 1) slow rupture propagation; 2) smooth rupture; 3) slow dislocation motion.

    Notes: Abstract Heterogeneous fault motion of the 1993 Hokkaido Nansei-Oki earthquake is studied by using seismic, geodetic and tsunami data, and the tsunami generation from the fault model is examined. Seismological analyses indicate that the focal mechanism of the first 10 s, when about a third of the total moment was released, is different from the overall focal mechanism. A joint inversion of geodetic data on Okushiri Island and the tide gauge records in Japan and Korea indicates that the largest slip, about 6 m, occurred in a small area just south of the epicenter. This corresponds to the initial rupture on a fault plane dipping shallowly to the west. The slip on the northernmost subfault, which is dipping to the east, is about 2 m, while the slips on the southern subfaults, which are steeply dipping to the west, are more than 3 m. Tsunami heights around Okushiri Island are calculated from the heterogeneous fault model using different grid sizes. Computation on the smaller grids produces large tsunami height that are closer to the observed tsunami runup heights. Tsunami propagation in the nearly closed Japan Sea is examined as the free oscillation of the Japan Sea. The excitation of the free oscillation by this earthquake is smaller than that by the 1964 Niigata or 1983 Japan Sea earthquake.

    Notes: Abstract The fault parameters of the Guam earthquake of August 8, 1993 are estimated from seismological analyses, and the possibility of identifying the actual fault plane from tsunami waveforms is tested. The Centroid Moment Tensor solution of long-period surface waves shows one nodal plane shallowly dipping to the north and the other nodal plane steeply dipping to the south. The seismic moment is 3.5×1020 Nm and the corresponding moment magnitude is 7.7. The Moment Tensor Rate Function inversion ofP waves also yields a similar focal mechanism and seismic moment. The point source depth is estimated as 40–50 km. This earthquake generated tsunamis that propagated toward the Japanese coast along the Izu-Bonin-Mariana ridge system. The tsunamis are recorded on ocean bottom pressure gauges and tide gauges. Numerical computation of tsunamis shows that the computed waveforms from the two possible fault planes match well with the observed tsunami waveforms. The numerical computation also shows that the tsunami waveforms at Guam Island, just above the fault, should contain useful information regarding the identification of the actual fault plane. However, the current sampling rate of the tide gauges is so small that the records cannot help the identification.

    Notes: Abstract Onshore tsunami deposits resulting from the 1993 Southwest Hokkaido and 1983 Japan Sea earthquakes were described to evaluate the feasibility of tsunami deposits for inferring paleoseismic events along submarine faults. Tsunami deposits were divided into three types, based on their composition and aerial distribution: (A) deposits consisting only of floating materials, (B) locally distributed siliclastic deposits, and (C) widespread siliclastic deposits. The most widely distributed tsunami deposits consist of the first two types. Type C deposits are mostly limited to areas where the higher tsunami runup was observed. The scale of tsunami represented by vertical tsunami runup is an important factor controlling the volume of tsunami deposits. The thickest deposits, about 10 cm, occur behind coastal dunes. To produce thick siliclastic tsunami deposits, a suitable source area, such as sand bar or dune, must be available in addition to sufficient vertical tsunami runup. Estimation of the amounts of erosion and deposition indicates that tsunami deposits were derived from both onshore and shoreface regions. The composition and grain size of the tsunami deposits strongly reflect the nature of the sedimentary materials of their source area. Sedimentary structures of the tsunami deposits suggest both low and high flow régimes. Consequently, it seems very difficult to identify tsunami deposits based only on grain size distribution or sedimentary structure of a single site in ancient successions.

    Notes: Abstract Detailed field work at Okushiri Island and along the southwest coast of Hokkaido has revealed quantitatively (1) the advancing direction of tsunami on land, (2) the true tsunami height (i.e., height of tsunami, excluding its splashes, as measured from the ground) and (3) the flow velocity of tsunami on land, in heavily damaged areas. When a Japanese wooden house is swept away by tsunami, bolts that tie the house to its concrete foundation resist until the last moment and become bent towards the direction of the house being carried away. The orientations of more than 850 of those bent bolts and iron pipes (all that can be measured, mostly at Okushiri Island) and fell-down direction of about 400 trees clearly display how tsunami behaved on land and caused serious damage at various places. The true tsunami height was estimated by using several indicators, such as broken tree twigs and a window pane. The flow velocity of tsunami on land was determined by estimating the hydrodynamic force exerted on a bent handrail and a bent-down guardrail by the tsunami throughin situ strength tests. Contrary to the wide-spread recognition after the tsunami hazard, our results clearly indicate that only a few residential areas (i.e., Monai, eastern Hamatsumae, and a small portion at northern Aonae, all on Okushiri Island) were hit by a huge tsunami, with true heights reaching 10 m. Southern Aonae was completely swept away by tsunami that came directly from the focal region immediately to the west. The true tsunami height over the western sea wall of southern Aonae was estimated as 3 to 4 m. Northern Aonae also suffered severe damage due to tsunami that invaded from the corner zone of the sand dune (8 m high) and tide embankment at the northern end of the Aonae Harbor. This corner apparently acted as a tsunami amplifier, and tide embankment or breakwater can be quite dangerous when tsunami advances towards the corner it makes with the coast. The nearly complete devastation of Inaho at the northern end of Okushiri Island underscored the danger of tsunami whose propagation direction is parallel to the coast, since such tsunami waves tend to be amplified and tide embankment or breakwater is constructed low towards the coast at many harbors or fishing ports. Tsunami waves mostly of 2 to 4 m in true height swept away Hamatsumae on the southeast site of Okushiri Island where there were no coastal structures. Coastal structures were effective in reducing tsunami hazard at many sites. The maximum flow velocity at northern Aonae was estimated as 10 to 18 m/s (Tsutsumi et al., 1994), and such a high on-land velocity of tsunami near shore is probably due to the rapid shallowing of the deep sea near the epicentral region towards Okushiri Island. If the advancing direction, true height, and flow velocity of tsunami can be predicted by future analyses of tsunami generation and progagation, the analyses will be a powerful tool for future assessment of tsunami disasters, including the identification of blind spots in the tsunami hazard reduction.

    Notes: Abstract A fault plane model and a finite element hydrodynamic model are applied to the simulation of the Hokkaido Nansei-Oki tsunami of July 12, 1993. The joint performance of the models is assessed based on the overall ability to reproduce observed tsunami waveforms and to preserve mass and energy during tsunami propagation. While a number of observed characteristics of the waveforms are satisfactorily reproduced (in particular, amplitudes and arrival times at tidal gauges relatively close to the source, and general patterns of energy concentration), others are only marginally so (notably, wave periods at the same gauges, and wave heights along Okushiri); differences between observations and simulations are traceable to both the fault plane and the hydrodynamic models. Nonnegligible losses of energy occur throughout the simulated tsunami propagation. These losses seem to be due to a combination of factors, including numerical damping and possible deficiencies of the shallow water equations in preserving energy.

    Notes: Abstract The 1994 Shikotan earthquake was one of the greatest earthquakes in recent years with a magnitude ofM s 8.0. A tsunami survey was conducted by Russian and U.S. geophysicists from October 16–30, 1994, less than two weeks after the earthquake. The survey results and a numerical hindcast simulation are reported. Tsunami focusing effect at locations supposedly sheltered by the island chain is discussed. Based on the obtained data, tsunamis which attacked Shikotan Island are characterized as long waves (the order of 10–20 min wave period) with a positive leading wave. Possible consequences of the positive leading wave form are discussed in relation to the observed minimal destruction of beach vegetation and relatively small transport of marine sediment onto the shore. The high-quality tide-gage record in Malokurilskaya Bay indicates the occurrence of a 53 cm subsidence at the site.

    Notes: Abstract The topic of this paper is to review recent processes of increasing seismic activity in the Khibiny Massif in the Kcla Peninsula. It is a typical example of induced seismicity caused by rock deformation due to the extraction of more than 2·109 tons of rock mass since the mid-1960s. The dependence of seismic activity on the amount of extracted ore is demonstrated. Some of the induced earthquakes coincide with large mining explosions, thus indicating a trigger mechanism. The largest earthquake, which occurred on 16 April 1989 (M L= 4.1) could be traced along the surface for 1200 m and observed to a depth of at least 220 m. The maximum measured displacement was 15–20 cm.

    Notes: Abstract The effects of variable viscosity on flow dynamics within spherical shells are investigated using a finite-element thermal convection model, and preliminary result for cases with relatively low Rayleigh numbers and small viscosity contrasts are reported. These results demonstrate some general effects of viscosity variation on mantle dynamics, and, in particular, the generation of toroidal energy. Since lateral viscosity variations are necessary in the generation of toroidal motion in a thermally driven convective system, it is not surprising our results show that flows with greater viscosity contrasts produce greater amounts of toroidal energy. Our preliminary study further shows that solutions become more time-dependent as viscosity contrasts increase. Increasing the Rayleigh number is also found to increase the magnitude of toroidal energy. Internal heating, on the other hand, appears to lead to less toroidal energy compared wth bottom heating because it tends to produce a thermally more uniform interior and thus smaller viscosity variations.

    Notes: Abstract Shear-crack model with a cohesive zone (or breakdown zone) is appropriate for the analysis of a fault surface in which slip distribution is strongly nonuniform. As the slipped portion advances, slip-weakening occurs over the so-called cohesive zone, a distance behind the fault tip. For a prescribed strength vs. displacement constitutive relation, however, the zone structure is difficult to determine by an analytical method except for some simple cases, thus it often requires a certain numerical procedure. This work proposes a numerical procedure to obtain approximated solutions of the problem by combining a series of elastic solutions derived bySmith (1974). The series is linearly combined and the unknown coefficients are determined by a nonlinear least square method. This method can fit a wide range of prescribed strength vs. displacement relations which may be simple algebraic relations or curves obtained by laboratory tests. By examining the residual errors and in comparison with a derived result in which linear stress is assumed within the zone, it could be concluded that the results provide good accuracy. Moreover, because the results are written in formulae, they can be easily referred to or used. By fitting constitutive curves in many different shapes, it is found that the stress distribution within the zone is more sensitive to the constitutive curve shape than the displacement. The most interesting fact is that the zone size is not sensitive to the curve shape, i.e., the zone size can be estimated by $$R = 3\mu \zeta \upsilon _c /\{ 2(1 - \upsilon )(\tau _c - \tau _f )\}$$ with ζ=1±0.11 for most cases.

    Notes: Abstract Uniaxial compression tests on marble plates containing two prefabricateden echelon fractures were performed in this study. Photographs showing the typical characteristics of subcrack development were taken under direct scanning electron microscope (SEM) observation during the test. From these photographs, the effects of the lengthL of a single fracture, the separation distanced and overlapping proportiona/L between two prefabricateden echelon fractures on the development of subcracks were analyzed. The results show that the interaction betweenen echelon fractures strengthened with decreasingd. Ford≥L, there was little interaction and the development of subcracks near one of theen echelon fractures was unaffected by the existence of the other. Whereas ford〈L, the number of subcracks in the area intermediate betweenen echelon fractures tended to increase with increasinga and decreasingd. In order to ascertain whether the experiment can furnish some implications for research on earthquake prediction and neotectonic activity, the test results were compared with the spatial-temporal development of foreshocks and ground-water anomalies before the 1975 Haicheng earthquake and the structural framework of the Ganzi pull-apart basin. The results of the comparison are encouraging. According to the similarity between the test results in the laboratory and the natural phenomena in the field, the fault on which the Haicheng earthquake would occur, could be inferred immediately from the relative geometry of the two sub-parallel active faults in the area. Thus, it is considered that the test results would advance our understanding of the process of neotectonic activities and give us inspiration for earthquake prediction.

    Notes: Abstract We show analytically that addition of a quintic term to the positive Kerr-type nonlinearity offers a unique type of kink soliton-like solution with Fermi-Dirac profile. This type of optical kink allows, in contrast to other optical kinks discovered so far, stationary kink formation not only in the time domain but in the spatial domain. The latter could admit of a route for the first time to our knowledge to spatial kink solitons of intensified laser beams. The underlying principle of the optical kink propagation is described.

    Notes: Abstract Recently, we have discovered that—besides the all-optical photo-thermal bistability in transmission—the excitation of thin CdS films with the 514.5 nm line of an argon laser causes all-optical bistability in the luminescent radiation. The emitted radiation of the CdS layers lies far enough in the infrared region of the spectrum so that all-optical bistability in luminescence can be observed through GaAs wafers, i.e. an all-optical bistable loop remains in the direction of the propagation of the incident laser beam even though GaAs is strongly absorptive at the excitation wavelength. On the basis of this penetrating feature of all-optical bistability, we present a new concept for an architecture of future optical data processing devices.

    Notes: Abstract A new CGH (computer generated hologram) common path interferometer, which can be used for testing an aspherical mirror with large aperture, is proposed. This interferometer does not have a circle of least confusion, and a spatial filter can be used effectively to stop extraneous lights so that interferograms with good contrast can be obtained. The phase shifting technique can be easily applied to the interferometer. The method of designing and making the CGH zone-plate is described.

    Notes: Abstract In designing lenses with the damped least squares method, the solution obtained by optimization routine is a local minimum of the merit function. To get out of this and seek a different solution, we propose to use an ‘escape function’ as an additional operand of the lens system, to be controlled. Experiments were made on simple models of merit function and the advantage of this technique was ascertained. We also planted this algorithm into OSLO SIX (lens design software by Sinclair Optics) by means of CCL (C-compatible language) and applied it to actual lens design. Experiments convinced us that the method would be an effective tool for global optimization.
