    Description: Total sleep deprivation leads to decrements in neurobehavioral performance and changes in electroencephalographic (EEG) oscillations as well as the incidence of slow eye movements ad detected in the electro-oculogram (EOG) during wakefulness. Although total sleep deprivation is a powerful tool to investigate the association of EEG/EOG and neurobehavioral decrements, sleep loss during space flight is usual only partial. Furthermore exposure to the microgravity environment leads to changes in sodium and volume homeostasis and associated renal and cardio-endocrine responses. Some of these changes can be induced in head down tilt bedrest studies. We integrate research tools and research projects to enhance the fidelity of the simulated conditions of space flight which are characterized by complexity and mutual interactions. The effectiveness of countermeasures and physiologic mechanisms underlying neurobehavioral changes and renal-cardio endocrine changes are investigated in Project 3 of the Human Performance Team and Project 3 of the Cardiovascular Alterations Team respectively. Although the. specific aims of these two projects are very different, they employ very similar research protocols. Thus, both projects investigate the effects of posture/bedrest and sleep deprivation (total or partial) on outcome measures relevant to their specific aims. The main aim of this enhancement grant is to exploit the similarities in research protocols by including the assessment of outcome variables relevant to the Renal-Cardio project in the research protocol of Project 3 of the Human Performance Team and by including the assessment of outcome variables relevant to the Quantitative EEG and Sleep Deprivation Project in the research protocols of Project 3 of the Cardiovascular Alterations team. In particular we will assess Neurobehavioral Function and Waking EEG in the research protocols of the renal-cardio endocrine project and renin-angiotensin and cardiac function in the research protocol of the Quantitative EEG and Waking Neurobehavioral Function project. This will allow us to investigate two additional specific aims: 1) Test the hypothesis that chronic partial sleep deprivation during a 17 day bed rest experiment results in deterioration of neurobehavioral function during waking and increases in EEG power density in the theta frequencies, especially in frontal areas of the brain, as well as the nonREM- REM cycle dependent modulation of heart-rate variability. 2) Test the hypothesis that acute total sleep deprivation modifies the circadian rhythm of the renin-angiotensin system, changes the acute responsiveness of this system to posture beyond what a microgravity environment alone does and affects the nonREM-REM cycle dependent modulation of heart-rate variability.

    Description: The risks to personnel in space from the naturally occurring radiations are generally considered to be one of the most serious limitations to human space missions, as noted in two recent reports of the National Research Council/National Academy of Sciences. The Core Project of the Radiation Effects Team for the National Space Biomedical Research Institute is the consequences of radiations in space in order to develop countermeasure, both physical and pharmaceutical, to reduce the risks of cancer and other diseases associated with such exposures. During interplanetary missions, personnel in space will be exposed to galactic cosmic rays, including high-energy protons and energetic ions with atomic masses of iron or higher. In addition, solar events will produce radiation fields of high intensity for short but irregular durations. The level of intensity of these radiations is considerably higher than that on Earth's surface, and the biological risks to astronauts is consequently increased, including increased risks of carcinogenesis and other diseases. This group is examining the risk of cancers resulting from low-dose, low-dose rate exposures of model systems to photons, protons, and iron by using ground-based accelerators which are capable of producing beams of protons, iron, and other heavy ions at energies comparable to those encountered in space. They have begun the first series of experiments using a 1-GeV iron beam at the Brookhaven National Laboratory and 250-MeV protons at Loma Linda University Medical Center's proton synchrotron facility. As part of these studies, this group will be investigating the potential for the pharmaceutical, Tamoxifen, to reduce the risk of breast cancer in astronauts exposed to the level of doses and particle types expected in space. Theoretical studies are being carried out in a collaboration between scientists at NASA's Johnson Space Center and Johns Hopkins University in parallel with the experimental program have provided methods and predictions which are being used to assess the levels of risks to be encountered and to evaluate appropriate strategies for countermeasures. Although the work in this project is primarily directed toward problems associated with space travel, the problem of protracted exposures to low-levels of radiation is one of national interest in our energy and defense programs, and the results may suggest new paradigms for addressing such risks.

    Description: A major concern associated with long-duration space flight is the possibility of infectious diseases posing an unacceptable medical risk to crew members. One major hypothesis addressed in this project is that space flight will cause alterations in the immune system that will allow latent viruses that are endogenous in the human population to reactivate and shed to higher levels than normal, which may affect the health of crew members. The second major hypothesis being examined is that the effects of space flight will alter the mucosal immune system, the first line of defense against many microbial infections, including herpesviruses, polyomaviruses, and gastroenteritis viruses, rendering crew members more susceptible to virus infections across the mucosa. We are focusing the virus studies on the human herpesviruses and polyomaviruses, important pathogens known to establish latent infections in most of the human population. Both primary infection and reactivation from latent infection with these groups of viruses (especially certain herpesviruses) can cause a variety of illnesses that result in morbidity and, occasionally, mortality. Both herpesviruses and polyomaviruses have been associated with human cancer, as well. Effective vaccines exist for only one of the eight known human herpesviruses and available antivirals are of limited use. Whereas normal individuals display minimal consequences from latent viral infections, events which alter immune function (such as immunosuppressive therapy following solid organ transplantation) are known to increase the risk of complications as a result of viral reactivations.

    Description: The National Aeronautics and Space Administration (NASA) has had sufficient concern for the well-being of astronauts traveling in space to create the National Space Biomedical Research Institute (NSBRI), which is investigating several areas of biomedical research including those of immunology. As part of the Immunology, Infection, and Hematology Team, the co-investigators of the Space Flight Immunodeficiency Project began their research projects on April 1, 1998 and are now just into the second year of work. Two areas of research have been targeted: 1) specific immune (especially antibody) responses and 2) non-specific inflammation and adhesion. More precise knowledge of these two areas of research will help elucidate the potential harmful effects of space travel on the immune system, possibly sufficient to create a secondary state of immunodeficiency in astronauts. The results of these experiments are likely to lead to the delineation of functional alterations in antigen presentation, specific immune memory, cytokine regulation of immune responses, cell to cell interactions, and cell to endothelium interactions.

    Description: This project is concerned with identifying ways to prevent neurobehavioral and physical deterioration due to inadequate sleep in astronauts during long-duration manned space flight. The performance capability of astronauts during extended-duration space flight depends heavily on achieving recovery through adequate sleep. Even with appropriate circadian alignment, sleep loss can erode fundamental elements of human performance capability including vigilance, cognitive speed and accuracy, working memory, reaction time, and physiological alertness. Adequate sleep is essential during manned space flight not only to ensure high levels of safe and effective human performance, but also as a basic regulatory biology critical to healthy human functioning. There is now extensive objective evidence that astronaut sleep is frequently restricted in space flight to averages between 4 hr and 6.5 hr/day. Chronic sleep restriction during manned space flight can occur in response to endogenous disturbances of sleep (motion sickness, stress, circadian rhythms), environmental disruptions of sleep (noise, temperature, light), and curtailment of sleep due to the work demands and other activities that accompany extended space flight operations. The mechanism through which this risk emerges is the development of cumulative homeostatic pressure for sleep across consecutive days of inadequate sleep. Research has shown that the physiological sleepiness and performance deficits engendered by sleep debt can progressively worsen (i.e., accumulate) over consecutive days of sleep restriction, and that sleep limited to levels commonly experienced by astronauts (i.e., 4 - 6 hr per night) for as little as 1 week, can result in increased lapses of attention, degradation of response times, deficits in complex problem solving, reduced learning, mood disturbance, disruption of essential neuroendocrine, metabolic, and neuroimmune responses, and in some vulnerable persons, the emergence of uncontrolled sleep attacks. The prevention of cumulative performance deficits and neuroendocrine disruption from sleep restriction during extended duration space flight involves finding the most effective ways to obtain sleep in order to maintain the high-level cognitive and physical performance functions required for manned space flight. There is currently a critical deficiency in knowledge of the effects of how variations in sleep duration and timing relate to the most efficient return of performance per unit time invested in sleep during long-duration missions, and how the nature of sleep physiology (i.e., sleep stages, sleep electroencephalographic [EEG] power spectral analyses) change as a function of sleep restriction and performance degradation. The primary aim of this project is to meet these critical deficiencies through utilization of a response surface experimental paradigm, testing in a dose-response manner, varying combinations of sleep duration and timing, for the purpose of establishing how to most effectively limit the cumulative adverse effects on human performance and physiology of chronic sleep restriction in space operations.

    Description: Alterations in cardiovascular regulation and function that occur during and after space flight have been reported. These alterations are manifested, for example, by reduced orthostatic tolerance upon reentry to the earth's gravity from space. However, the precise physiologic mechanisms responsible for these alterations remain to be fully elucidated. Perhaps, as a result, effective countermeasures have yet to be developed. In this project we apply a powerful, new method - cardiovascular system identification (CSI) - for the study of the effects of space flight on the cardiovascular system so that effective countermeasures can be developed. CSI involves the mathematical analysis of second-to-second fluctuations in non-invasively measured heart rate, arterial blood pressure (ABP), and instantaneous lung volume (ILV - respiratory activity) in order to characterize quantitatively the physiologic mechanisms responsible for the couplings between these signals. Through the characterization of all the physiologic mechanisms coupling these signals, CSI provides a model of the closed-loop cardiovascular regulatory state in an individual subject. The model includes quantitative descriptions of the heart rate baroreflex, autonomic function, as well as other important physiologic mechanisms. We are in the process of incorporating beat-to-beat fluctuations of stroke volume into the CSI technique in order to quantify additional physiologic mechanisms such as those involved in control of peripheral vascular resistance and alterations in cardiac contractility. We apply CSI in conjunction with the two general protocols of the Human Studies Core project. The first protocol involves ground-based, human head down tilt bed rest to simulate microgravity and acute stressors - upright tilt, standing and bicycle exercise - to provide orthostatic and exercise challenges. The second protocol is intended to be the same as the first but with the addition of sleep deprivation to determine whether this contributes to cardiovascular alterations. In these studies, we focus on the basic physiologic mechanisms responsible for the alterations in cardiovascular regulation and function during the simulated microgravity in order to formulate hypotheses regarding what countermeasures are likely to be most effective. Compared to our original proposal, the protocol we are using has been slightly modified to lengthen the bed rest period to 16 days and streamline the data collection. These modifications provide us data on a longer bed rest period and have enabled us to increase our subject throughput. Based on review of our preliminary data we have decided to test a countermeasure which is applied the very end of the bed rest period. We will use the same bed rest protocol to test this countermeasure. We anticipate completing the baseline data collection in our first protocol plus testing of the countermeasure in an additional eight subjects, at which time we plan to initiate the second protocol which includes sleep deprivation. In future studies, we plan to apply CSI to test other potential countermeasures in conjunction with the same bed rest, sleep deprivation and acute stressor models. We also anticipate applying CSI for studying astronauts before and after space flight and ultimately, during space flight. The application of CSI is providing information relevant to the development and evaluation of effective countermeasures allowing humans to adapt appropriately upon re-exposure to a gravity field, and to live and work for longer periods of time in microgravity.

    Description: The presence of ice in permanently shadowed depressions near the lunar poles and determination of its properties will significantly influence both the near- and long-term prospects for lunar exploration and development. Since data from the Lunar Prospector spacecraft indicate that water ice is likely present (the instrument measures hydrogen strongly suggests the presence of water), it is important to understand how to extract it for beneficial use, as well as how to preserve it for scientific analysis. Two types of processes can be considered for the extraction of water ice from the lunar poles. In the first case, energy is transported into the shadowed regions, ice is constrain models of impacts on the lunar surface and processed in-situ, and water is transported out of the cold trap. In the second case, ice-containing regolith can be mined in the cold trap, transported outside the cold trap, and the ice extracted in a location with abundant solar energy. A series of conceptual implementations has been examined and criteria have been developed for the selection of systems and subsystems for further study.

    Description: As new personnel join the Medical Operations Branch, it is critical that they understand the effects of microgravity on medical procedures, hardware, and supplies. The familiarization flight provided new personnel with a better understanding of the effects of microgravity on (1) medical procedures, (2) patient and rescuer restraint, (3) medical fluids, and (4) medical training for space flight. The flight process also provided experience in flight proposal preparation, flight test plan preparation and execution, and final report preparation. In addition, first time flyers gained insight on their performance level in microgravity for future flights.

    Description: This paper reports on the model, test, and results from the Langley Supersonic Aftbody Closure wind tunnel test. This project is an experimental evaluation of the 1.5% Technology Concept Aircraft (TCA) aftbody closure model (Model 23) in the Langley Unitary Plan Wind Tunnel. The baseline TCA design is the result of a multidisciplinary, multipoint optimization process and was developed using linear design and analysis methods, supplemented with Euler and Navier-Stokes numerical methods. After a thorough design review, it was decided to use an upswept blade attached to the forebody as the mounting system. Structural concerns dictated that a wingtip support system would not be feasible. Only the aftbody part of the model is metric. The metric break was chosen to be at the fuselage station where prior aft-sting supported models had been truncated. Model 23 is thus a modified version of Model 20. The wing strongback, flap parts, and nacelles from Model 20 were used, whereas new aftbodies, a common forebody, and some new tails were fabricated. In summary, significant differences in longitudinal and direction stability and control characteristics between the ABF and ABB aftbody geometries were measured. Correcting the experimental data obtained for the TCA configuration with the flared aftbody to the representative of the baseline TCA closed aftbody will result in a significant reduction in longitudinal stability, a moderate reduction in stabilizer effectiveness and directional stability, and a moderate to significant reduction in rudder effectiveness. These reductions in the stability and control effectiveness levels of the baseline TCA closed aftbody are attributed to the reduction in carry-over area.

    Description: The first microscopic sedimentological studies of the Martian surface will commence with the landing of the Mars Polar Lander (MPL) December 3, 1999. The Robotic Arm Camera (RAC) has a resolution of 25 um/p which will permit detailed micromorphological analysis of surface and subsurface materials. The Robotic Ann will be able to dig up to 50 cm below the surface. The walls of the trench will also be inspected by RAC to look for evidence of stratigraphic and / or sedimentological relationships. The 2001 Mars Lander will build upon and expand the sedimentological research begun by the RAC on MPL. This will be accomplished by: (1) Macroscopic (dm to cm): Descent Imager, Pancam, RAC; (2) Microscopic (mm to um RAC, MECA Optical Microscope (Figure 2), AFM This paper will focus on investigations that can be conducted by the RAC and MECA Optical Microscope.

    Description: There were two objectives for this test. First, was to assess the reasons why there is approximately 1.5 drag counts (cts) discrepancy between measured and computed drag improvement of the Non-linear Cruise Validation (NCV) over the Technology Concept Airplane (TCA) wing body (WB) configurations. The Navier-Stokes (N-S) pre-test predictions from Boeing Commercial Airplane Group (BCAG) show 4.5 drag cts of improvement for NCV over TCA at a lift coefficient (CL) of 0. I at Mach 2.4. The pre-test predictions from Boeing Phantom Works - Long Beach, BPW-LB, show 3.75 drag cts of improvement. BCAG used OVERFLOW and BPW-LB used CFL3D. The first test entry to validate the improvement was held at the NASA Langley Research Center (LARC) UPV;T, test number 1687. The experimental results showed that the drag improvement was only 2.6 cts, not accounting for laminar run and trip drag. This is approximately 1.5 cts less than predicted computationally. In addition to the low Reynolds Number (RN) test, there was a high RN test in the Boeing Supersonic Wind Tunnel (BSWT) of NCV and TCA. BSV@T test 647 showed that the drag improvement of NCV over TCA was also 2.6 cts, but this did account for laminar run and trip drag. Every effort needed to be done to assess if the improvement measured in LaRC UPWT and BSWT was correct. The second objective, once the first objective was met, was to assess the performance increment of NCV over TCA accounting for the associated laminar run and trip drag corrections in LaRC UPWT. We know that the configurations tested have laminar flow on portions of the wing and have trip drag due to the mechanisms used to force the flow to go from laminar to turbulent aft of the transition location.

    Description: An improved laminar run and trip drag correction methodology for supersonic cruise performance testing was derived. This method required more careful analysis of the flow visualization images which revealed delayed transition particularly on the inboard upper surface, even for the largest trip disks. In addition, a new code was developed to estimate the laminar run correction. Once the data were corrected for laminar run, the correct approach to the analysis of the trip drag became evident. Although the data originally appeared confusing, the corrected data are consistent with previous results. Furthermore, the modified approach, which was described in this presentation, extends prior historical work by taking into account the delayed transition caused by the blunt leading edges.

The Athena payload has been adopted for the analysis and selection of samples, and the will include many of the same measurements to be performed during the '01 mission. In addition, the missions will include yet to be determined experiments to be done on or from the lander. Several groups are now competing for instruments and science to be done on the lander for both '03 and '05.

    Description: The DART ("Dust Accumulation and Removal Test") package is an experiment which will fly as part of the MIP experiment on the Mars-2001 Surveyor Lander. Dust deposition could be a significant problem for photovoltaic array operation for long duration emissions on the surface of Mars. Measurements made by Pathfinder showed 0.3% loss of solar array performance per day due to dust obscuration. The DART experiment is designed to quantify dust deposition from the Mars atmosphere, measure the properties of settled dust, measure the effect of dust deposition on the array performance, and test several methods of mitigating the effect of settled dust on a solar array. Although the purpose of DART (along with its sister experiment, MATE) is to gather information critical to the design of future power systems on the surface of Mars, the dust characterization instrumentation on DART will also provide significant scientific data on the properties of settled atmospheric dust.

    Description: The Mars Environmental Compatibility Assessment (MECA) is an instrument suite that will fly on the Mars Surveyor 2001 Lander Spacecraft. MECA is sponsored by the Human Exploration and Development of Space (HEDS) program and will evaluate potential hazards that the dust and soil of Mars might present to astronauts and their equipment on a future human mission to Mars. Four elements constitute the integrated MECA payload: a microscopy station, patch plates, an electrometer, and the wet chemistry laboratory (WCL). The WCL consists of four identical cells, each of which will evaluate a sample of Martian soil in water to determine conductivity, pH, redox potential, dissolved C02 and 02 levels, and concentrations of many soluble ions including sodium, potassium, magnesium, calcium and the halides. In addition, cyclic voltammetry will be used to evaluate reversible and irreversible oxidants present in the water/soil solution. Anodic stripping voltammetry will be used to measure concentrations of trace metals including lead, copper, and cadmium at ppb levels. Voltammetry is a general electrochemical technique that involves controlling the potential of an electrode while simultaneously measuring the current flowing at that electrode. The WCL experiments will provide information on the corrosivity and reactivity of the Martian soil, as well as on soluble components of the soil which might be toxic to human explorers. They will also guide HEDS scientists in the development of high fidelity Martian soil simulants. In the process of acquiring information relevant to HEDS, the WCL will assess the chemical composition and properties of the salts present in the Martian soil.

    Description: Martian surface composition and processes are under study through analysis of spectral, magnetic and chemical data from Mars and analysis of laboratory analog materials. The focus of this study is on potential lander/rover measurements of weathered volcanic tephra and hydrothermal rocks because these samples resulted from processes that may have occurred on Mars. Fine-grained particles from these sources may be responsible for origination of the dust/soil on Mars that is shaping the planet's surface character. Alteration on the surface of Mars likely includes both chemical and physical interactions of soil particles and rock surfaces. Many of the minerals present in hydrothermal samples may be associated with organisms and may be useful as indicators of life or environments supportive of life on Mars. Characterization of the spectroscopic properties in the visible/near-infrared (VIS/NIR) and mid-infrared (IR) regions using reflectance, emittance and Raman, as well as the thermal properties of minerals thought to be present on Mars are being performed in order to identify them remotely. Particular interest is directed toward locating minerals, and hence landing sites, important to Astrobiology.

    Description: The Mars Surveyor 2001 mission to Mars was initially a key element in the Mars sample return sequence of missions. A capable rover, carrying the Cornell Athena instruments would be placed on Mars to roam over several kilometers, select samples, and place them in a cache for return by a subsequent mission. Inevitably, budget constraints forced descopes. At one critical point, the landed payload consisted only of the HEDS (Human Exploration and Development of Space) payloads selected for testing environmental properties of the surface for future human exploration. Then Congress intervened and put back some of the funding that had been deleted. NASA next redefined the payload to include as many of the Athena instruments as possible, to be distributed between the lander deck and a Sojourner class rover. This payload would then be placed on a modified version of the Mars Polar Lander rather than on the much larger, and more expensive, lander that had been originally designed for the mission. With this functionality restored the '01 mission remains an important and pivotal element of the Mars Surveyor program. It completes the Mars Observer objectives with the gamma ray spectrometer mapping. This mission will largely complete the global characterization phase of Mars exploration and mark the beginning of focused surface exploration leading to return of the first samples and the search for evidence of past Martian life. MSP'01 also is the first mission in the combined Mars exploration strategy of the HEDS and Space Science Enterprises of NASA. This mission, and those to follow, will demonstrate technologies and collect environmental data that will provide the basis for a decision to send humans to Mars. The NASA exploration strategy for Mars includes orbiters, landers and rovers launched in 2001 and 2003 and a sample return mission to be launched in 2005, returning a sample by 2008. The purpose of the rovers is to explore and characterize sites on Mars. The 2003 and 2005 missions will select rocks, soil, and atmosphere for return to earth.

    Description: The STS-76 (Shuttle-Mir 3) spaceflight provided an opportunity to test two questions about radiation responses in C. elegans. First, does the absence of gravity modify the dose-response relation for mutation and chromosome aberration and second, what are the features of the mutation spectrum resulting from exposure to cosmic rays? These questions were put to the test in space using the ESA "Biorack" facility which was housed in the Spacehab module aboard shuttle Atlantis. The mission flew in March, 1996 and was a shuttle rendezvous with the Russian space station Mir.

    Description: The acute effects of exposure to microgravity include the development of space motion sickness which usually requires therapeutic intervention. The current drug of choice, promethazine (PMZ), has side effects which include nausea, drowsiness, dizziness, sedation and impaired psychomotor performance. In a ground-based study with commercial airline pilots and shuttle simulator trainers, we measured sleep and psychomotor performance variables, and physiological variables such as blood pressure and heart rate, as a function of circulating drug concentrations in the body. We evaluated a non-invasive sampling method (saliva) as a means of assessing pharmacodynamics following a single intramuscular (IM) dose of PMZ.

    Description: We postulate that centripetal acceleration induced by centrifugation can be used as an inflight sensorimotor countermeasure to retain and/or promote appropriate crewmember responses to sustained changes in gravito-inertial force conditions. Active voluntary motion is required to promote vestibular system conditioning, and both visual and graviceptor sensory feedback are critical for evaluating internal representations of spatial orientation. The goal of our investigation is to use centrifugation to develop an analog to the conflicting visual/gravito-inertial force environment experienced during space flight, and to use voluntary head movements during centrifugation to study mechanisms of adaptation to altered gravity environments. We address the following two hypotheses: (1) Discordant canal-otolith feedback during head movements in a hypergravity tilted environment will cause a reorganization of the spatial processing required for multisensory integration and motor control, resulting in decreased postural stability upon return to normal gravity environment. (2) Adaptation to this "gravito-inertial tilt distortion" will result in a negative after-effect, and readaptation will be expressed by return of postural stability to baseline conditions. During the third year of our grant we concentrated on examining changes in balance control following 90-180 min of centrifugation at 1.4 9. We also began a control study in which we exposed subjects to 90 min of sustained roll tilt in a static (non-rotating) chair. This allowed us to examine adaptation to roll tilt without the hypergravity induced by centrifugation. To these ends, we addressed the question: Is gravity an internal calibration reference for postural control? The remainder of this report is limited to presenting preliminary findings from this study.

    Description: The potential threat of immunosuppression and abnormal inflammatory responses in long-term space travel, leading to unusual predilection for opportunistic infections, malignancy, and death, is of ma or concern to the National Aeronautics and Space Administration (NASA) Program. This application has been devised to seek answers to questions of altered immunity in space travel raised by previous investigations spanning 30-plus years. We propose to do this with the help of knowledge gained by the discovery of the molecular basis of many primary and secondary immunodeficiency diseases and by application of molecular and genetic technology not previously available. Two areas of immunity that previously received little attention in space travel research will be emphasized: specific antibody responses and non-specific inflammation and adhesion. Both of these areas of research will not only add to the growing body of information on the potential effects of space travel on the immune system, but be able to delineate any functional alterations in systems important for antigen presentation, specific immune memory, and cell:cell and cell:endothelium interactions. By more precisely defining molecular dysfunction of components of the immune system, it is hoped that targeted methods of prevention of immune damage in space could be devised.

    Description: Gradient-based optimization requires accurate derivatives of the objective function and constraints. These gradients may have previously been obtained by manual differentiation of analysis codes, symbolic manipulators, finite-difference approximations, or existing automatic differentiation (AD) tools such as ADIFOR (Automatic Differentiation in FORTRAN). Each of these methods has certain deficiencies, particularly when applied to complex, coupled analyses with many design variables. Recently, a new AD tool called ADJIFOR (Automatic Adjoint Generation in FORTRAN), based upon ADIFOR, was developed and demonstrated. Whereas ADIFOR implements forward-mode (direct) differentiation throughout an analysis program to obtain exact derivatives via the chain rule of calculus, ADJIFOR implements the reverse-mode counterpart of the chain rule to obtain exact adjoint form derivatives from FORTRAN code. Automatically-generated adjoint versions of the widely-used CFL3D computational fluid dynamics (CFD) code and an algebraic wing grid generation code were obtained with just a few hours processing time using the ADJIFOR tool. The codes were verified for accuracy and were shown to compute the exact gradient of the wing lift-to-drag ratio, with respect to any number of shape parameters, in about the time required for 7 to 20 function evaluations. The codes have now been executed on various computers with typical memory and disk space for problems with up to 129 x 65 x 33 grid points, and for hundreds to thousands of independent variables. These adjoint codes are now used in a gradient-based aerodynamic shape optimization problem for a swept, tapered wing. For each design iteration, the optimization package constructs an approximate, linear optimization problem, based upon the current objective function, constraints, and gradient values. The optimizer subroutines are called within a design loop employing the approximate linear problem until an optimum shape is found, the design loop limit is reached, or no further design improvement is possible due to active design variable bounds and/or constraints. The resulting shape parameters are then used by the grid generation code to define a new wing surface and computational grid. The lift-to-drag ratio and its gradient are computed for the new design by the automatically-generated adjoint codes. Several optimization iterations may be required to find an optimum wing shape. Results from two sample cases will be discussed. The reader should note that this work primarily represents a demonstration of use of automatically- generated adjoint code within an aerodynamic shape optimization. As such, little significance is placed upon the actual optimization results, relative to the method for obtaining the results.

    Description: We have selected four areas in Valles Marineris as potential landing sites for the 2001 mission. After 20 years of analyses, the formation of the Valles Marineris system of troughs and its associated deposits still has not been sufficiently explained. They could have formed by collapse, as tectonic grabens, or in two stages involving ancestral collapse basins later cut by grabens. Understanding all aspects of the Valles Marineris, in particular the interior layered deposits, would significantly contribute to deciphering the internal and external history of Mars. The deposits have been postulated to be remnants of wall rock, lacustrine deposits, mass wasting deposits, eolian deposits, carbonate deposits, or volcanic deposits. Because an understanding of the formation of Valles Marineris and its interior deposits is so important to deciphering the history of Mars, we have proposed landing sites for the 2001 mission on flat shelves of interior deposits in Melas Chasma.

    Description: Our objective is to propose a landing site that the Mars Surveyor 2001 Lander and Curie Rover could go to on Mars that should meet the safety requirements of the spacecraft landing system and optimize surface operations (chiefly driven by power and communications requirements). This site lies between 1.5-3.5 deg S latitude, 195-198 deg W longitude, along a sharp albedo contact between the low-viscosity flow units of southern Elysium Planitia and the eroded highlands margin east of Aeolis Mensae. A relatively-bright "peninsula-like" protrusion of the eroded highlands into the south Elysium plains in this area reminds us of the head of an Ibis, and so we nickname this site "Ibishead Peninsula". This site is designed to be situated as close to a diversity of geologic units within view of the lander instruments. Based on our experience with the visibility of horizon details from the Mars Pathfinder and Viking landing sites, we stipulate that for horizon features to be resolved suitably for detailed study from the lander, they must be no more than several kilometers distant. This is so that diversity can be placed in a geologic context in a region that we feel has some exciting science potential. This objective is different from the Mars Pathfinder requirement to land at a site with a maximum chance for containing a diversity of rocks within a few tens of meters of the lander, which resulted in the selection of a "grab bag" site.

    Description: One of the greatest unresolved issues concerns the evolution of Mars early in its history; during the time period that accretion was winding down but the frequency of impacting debris was still heavy. Ancient cratered terrain that has only been moderately modified since the period of heavy bombardment covers about a quarter of the planet's surface but the environment during its formation is still uncertain. This terrain was dominantly formed by cratering. But unlike on the airless Moon, the impacting craters were strongly modified by other contemporary surface processes that have produced distinctive features such as 1) dendritic channel networks, 2) rimless, flatfloored craters, 3) obliteration of most craters smaller than a few kilometers in diameter (except for post heavy-bombardment impacts), and 4) smooth intercrater plains. The involvement of water in these modification processes seems unavoidable, but interpretations of the surface conditions on early Mars range from the extremes of 1) the "cold" model which envisions a thin atmosphere and surface temperatures below freezing except for local hydrothermal springs; and 2) the "warm" model, which invokes a thick atmosphere, seasonal temperatures above freezing in temperate and equatorial regions, and at least occasional precipitation as part of an active hydrological cycle. The nature of hydrologic cycles, if they occurred on Mars, would have been critically dependent on the environment. The resolution of where along this spectrum the actual environment of early Mars occurred is clearly a major issue, particularly because the alternate scenarios have much different implications about the possibility that life might have evolved on Mars.

    Description: Regions near Apollinaris Patera are proposed for consideration as Mars Surveyor landing sites. Gulick (1998) proposed this region at the First Mars Surveyor Landing Site workshop; Bulmer and Gregg (1998) provided additional support. Apollinaris Patera is situated on the highlands/lowlands boundary at 8.5S, 186W. The volcano itself has been mapped as Hesperian in age. The regions surrounding Apollinaris show evidence for volcanism, volcano-ice interactions, and erosion by water. Numerous valleys modified by fluvial processes dissect a large fan structure emanating from the southern flank of the volcano. Sapping valleys have formed along the southern terminus of the fan structure. Regions near Apollinaris Patera provide a unique opportunity to sample outcrop lithologies ranging from highland Noachian basement rocks, to Hesperian aged lava flows, channel and flood plain materials, to Amazonian volcanic, ash and channel deposits.

    Description: Over the past year and a half, the Center for Mars Exploration (CMEX) at NASA Ames Research Center (ARC) has been working with the Mars Surveyor Project Office at JPL to promote interactions among the planetary community and to coordinate landing site activities for the Mars Surveyor Project Office. To date, CMEX has been responsible for organizing the first two Mars Surveyor Landing Site workshops, web-archiving resulting information from these workshops, aiding in science evaluations of candidate landing sites, and serving as a liaison between the community and the Project. Most recently, CMEX has also been working with information technologists at Ames to develop a state-of-the-art collaborative web site environment to foster interaction of interested members of the planetary community with the Mars Surveyor Program and the Project Office. The web site will continue to evolve over the next several years as new tools and features are added to support the ongoing Mars Surveyor missions.

    Description: The Isidis basin rim may be key to understanding Mars' past with future lander missions: this area enables the mission objective to explore Mars' climatic and geologic history, including the search for liquid water and evidence of prior or extant life in ancient terrains. While two safe candidate landing sites for Mars Pathfinder were identified in Isidis Planitia, and one is being pursued for the Mars Surveyor 2001 Lander [Crumpler: 3.4 N, 277.8 W], the region around Isidis Planitia, in contrast to Tharsis for example, has only been lightly studied. The advent of new high resolution data sets provides an opportunity to reassess the geologic context of this impact basin and its rim within the Martian geologic sequence as a candidate site for studying Mars' ancient cratered terrain and ancient hydrosphere. This reexamination is warranted by the various hypotheses that Isidis was once filled with ice or water.

    Description: There are many similarities between the Mars Surveyor '01 (MS '01) landing site selection process and that of Mars Pathfinder. The selection process includes two parallel activities in which engineers define and refine the capabilities of the spacecraft through design, testing and modeling and scientists define a set of landing site constraints based on the spacecraft design and landing scenario. As for Pathfinder, the safety of the site is without question the single most important factor, for the simple reason that failure to land safely yields no science and exposes the mission and program to considerable risk. The selection process must be thorough and defensible and capable of surviving multiple withering reviews similar to the Pathfinder decision. On Pathfinder, this was accomplished by attempting to understand the surface properties of sites using available remote sensing data sets and models based on them. Science objectives are factored into the selection process only after the safety of the site is validated. Finally, as for Pathfinder, the selection process is being done in an open environment with multiple opportunities for community involvement including open workshops, with education and outreach opportunities.

    Description: Of the areas that meet the engineering criteria for MSP 01, only two are coincident with magnetic anomalies measured by the MAG/ER instrument on MGS. Area A is centered on about 10 deg S, 202 deg W and extends from about 7.5 deg S to 15 S. This area is associated with three bands of magnetic anomalies, two with positive values surrounding an area with negative values. Area B corresponds with a circular high positive magnetic anomaly and is centered at 13.5 deg S, 166 deg W. In addition to magnetic anomalies, the proposed sites have other attributes that make then attractive from of standpoint of meeting the objectives of the Mars Program. The landing site candidates meet the engineering requirements outlined on the Mars '01 landing site page htip://mars.jpl.nasa.gov/2001/landingsite. These are (source of data in parentheses): latitude between 3 deg N and 12 deg S, rock abundance between 5-10% (IRTM), fine-component thermal inertia 〉 4 cgs units (IRTM), topography 〈 2.5 km (MOLA). There are three exceptions: 1) Area B contains sites that lie up to about 15 deg S, 2) some sites are considered that have rock abundance values of 3-13%. 3) High resolution Viking coverage may not be available.     Description: Computations have been performed on the baseline Reference H wing/body configuration, as well as the Wing 704 configuration, an optimized wing and fuselage combination derived from Ref. H through automated optimization. The parabolized Navier-Stokes solver UPS was employed with viscous terms in two directions in an effort to understand the source and level of potential viscous/inviscid interactions. The paper briefly describes the UPS code and the grids used to obtain the solutions before the discussion of results. Results of these computations indicate that viscous/inviscid interaction can contribute increments to both the pressure- and friction-related drag. Computations were performed for wind tunnel conditions-1.675% scale models at a Reynolds number of 4 million per foot. Turbulent flow results were obtained using the Baldwin-Lomax algebraic turbulence model and were compared with laminar flow results. The laminar flow fields were used to obtain upper bounds on potential interaction effects.

    Description: The NASA-industry team has sponsored several studies in the last two years to address the installed nozzle boattail drag issues. Some early studies suggested that nozzle boattail drag could be as much as 25 to 40 percent of the subsonic cruise. As part of this study tests have been conducted at NASA-Langley to determine the uninstalled drag characteristics of a proposed nozzle. The overall objective was to determine the effects of nozzle external flap curvature and sidewall boattail variations. This test would also provide data for validating CFD predictions of nozzle boattail drag.

    Description: The computational fluid dynamics (CFD) comparisons being presented are compared to each other and to wind tunnel (WT) data on the baseline TCA. Some of the CFD computations were done prior to the tests and others later. Only force data (CL vs CD) from CFD will be presented as part of this report. The WT data presented comes from the testing of the baseline TCA in the Langley Unitary Plan Wind Tunnel (UPWT), Test Section #2. There are 2 sets of wind tunnel data being presented: one from test 1671 of model 2a (flapped wing) and the other from test 1679 of model 2b (solid wing). Most of the plots show only one run from each of the WT tests per configuration. But many repeat runs were taken during the tests. The WT repeat runs showed an uncertainty in the drag of +/- 0.5 count. There were times when the uncertainty in drag was better, +/- 0.25 count. Test 1671 data was of forces and pressures measured from model 2a. The wing had cutouts for installing various leading and trailing edge flaps at lower Mach numbers. The internal duct of the nacelles are not designed and fabricated as defined in the outer mold lines (OML) iges file. The internal duct was fabricated such that a linear transition occurs from the inlet to exhaust. Whereas, the iges definition has a constant area internal duct that quickly transitions from the inlet to exhaust cross sectional shape. The nacelle internal duct was fabricated, the way described, to save time and money. The variation in the cross sectional area is less than 1% from the iges definition. The nacelles were also installed with and without fairings. Fairings are defined as the build up of the nacelles on the upper wing surface so that the nacelles poke through the upper surface as defined in the OML iges file. Test 1679 data was of forces measured from model 2a and 2b. The wing for model 2b was a solid wing. The nacelles were built the same way as for model 2a, except for the nacelle base pressure installation. The nacelles were only tested with the fairings for model 2a and 2b during test 1679.

    Description: Configuration design at Ames was carried out with the SYN87-SB (single block) Euler code using a 193 x 49 x 65 C-H grid. The Euler solver is coupled to the constrained (NPSOL) and the unconstrained (QNMDIF) optimization packages. Since the single block grid is able to model only wing-body configurations, the nacelle/diverter effects were included in the optimization process by SYN87's option to superimpose the nacelle/diverter interference pressures on the wing. These interference pressures were calculated using the AIRPLANE code. AIRPLANE is an Euler solver that uses a unstructured tetrahedral mesh and is capable of computations about arbitrary complete configurations. In addition, the buoyancy effects of the nacelle/diverters were also included in the design process by imposing the pressure field obtained during the design process onto the triangulated surfaces of the nacelle/diverter mesh generated by AIRPLANE. The interference pressures and nacelle buoyancy effects are added to the final forces after each flow field calculation. Full details of the (recently enhanced) ghost nacelle capability are given in a related talk. The pseudo nacelle corrections were greatly improved during this design cycle. During the Ref H and Cycle 1 design activities, the nacelles were only translated and pitched. In the cycle 2 design effort the nacelles can translate vertically, and pitch to accommodate the changes in the lower surface geometry. The diverter heights (between their leading and trailing edges) were modified during design as the shape of the lower wing changed, with the drag of the diverter changing accordingly. Both adjoint and finite difference gradients were used during optimization. The adjoint-based gradients were found to give good direction in the design space for configurations near the starting point, but as the design approached a minimum, the finite difference gradients were found to be more accurate. Use of finite difference gradients was limited by the CPU time limit available on the Cray machines. A typical optimization run using finite difference gradients can use only 30 to 40 design variables and one optimization iteration within the 8 hour queue limit for the chosen grid size and convergence level. The efficiency afforded by the adjoint method allowed for 50-120 design variables and 5-10 optimization iterations in the 8 hour queue. Geometric perturbations to the wing and fuselage were made using the Hicks/Henne (HH) shape functions. The HH functions were distributed uniformly along the chords of the wing defining sections and lofted linearly. During single-surface design, constraints on thickness and volume at selected wing stations were imposed. Both fuselage camber and cross-sectional area distributions were permitted to change during design. The major disadvantage to the use of these functions is the inherent surface waviness produced by repeated use of such functions. Many smoothing operations were required following optimization runs to produce a configuration with reasonable smoothness. Wagner functions were also used on the wing sections but were never used on the fuselage. The Wagner functions are a family of increasingly oscillatory functions that have also been used extensively in airfoil design. The leading and trailing edge regions of the wing were designed by use of polynomial and monomial functions respectively. Twist was attempted but was abandoned because of little performance improvement available from changing the baseline twist.

    Description: The objective of this study is to calibrate a Navier-Stokes code for the TCA (30/10) baseline configuration (partial span leading edge flaps were deflected at 30 degs. and all the trailing edge flaps were deflected at 10 degs). The computational results for several angles of attack are compared with experimental force, moments, and surface pressures. The code used in this study is CFL3D; mesh sequencing and multi-grid were used to full advantage to accelerate convergence. A multi-grid approach was used similar to that used for the Reference H configuration allowing point-to-point matching across all the trailingedge block interfaces. From past experiences with the Reference H (ie, good force, moment, and pressure comparisons were obtained), it was assumed that the mounting system would produce small effects; hence, it was not initially modeled. However, comparisons of lower surface pressures indicated the post mount significantly influenced the lower surface pressures, so the post geometry was inserted into the existing grid using Chimera (overset grids).

    Description: The objective of the present study was to address the questions of: 1) how reliably or consistently the Navier-Stokes methods and processes used by the various organizations can predict integrated skin friction drag, and 2) how well the methods can predict trends within a family of optimized configurations. As a first step, all available skin friction drag predictions were accumulated to obtain a mean and standard deviation for the TCA (Technology Concept Airplane) baseline and each of the optimized configurations. It is observed that the optimization process has had little effect on the predicted skin friction drags. The variation in the mean that is observed is dwarfed by the standard deviations. In order to understand the reasons for the relatively large spreads in the computed results, a number of auxiliary computations have been performed using the UPS and OVERFLOW codes in an effort to identify and quantity potential sources of the variations.

    Description: The paper presents the recent progress made towards developing an efficient and user-friendly parallel environment for routine analysis of large CFD problems. The coarse-grain parallel version of the CFL3D Euler/Navier-Stokes analysis code, CFL3Dhp, has been ported onto most available parallel platforms. The CFL3Dhp solution accuracy on these parallel platforms has been verified with the CFL3D sequential analyses. User-friendly pre- and post-processing tools that enable a seamless transfer from sequential to parallel processing have been written. Static load balancing tool for CFL3Dhp analysis has also been implemented for achieving good parallel efficiency. For large problems, load balancing efficiency as high as 95% can be achieved even when large number of processors are used. Linear scalability of the CFL3Dhp code with increasing number of processors has also been shown using a large installed transonic nozzle boattail analysis. To highlight the fast turn-around time of parallel processing, the TCA full configuration in sideslip Navier-Stokes drag polar at supersonic cruise has been obtained in a day. CFL3Dhp is currently being used as a production analysis tool.

    Description: Automatic Grid Generation Wish List Geometry handling, including CAD clean up and mesh generation, remains a major bottleneck in the application of CFD methods. There is a pressing need for greater automation in several aspects of the geometry preparation in order to reduce set up time and eliminate user intervention as much as possible. Starting from the CAD representation of a configuration, there may be holes or overlapping surfaces which require an intensive effort to establish cleanly abutting surface patches, and collections of many patches may need to be combined for more efficient use of the geometrical representation. Obtaining an accurate and suitable body conforming grid with an adequate distribution of points throughout the flow-field, for the flow conditions of interest, is often the most time consuming task for complex CFD applications. There is a need for a clean unambiguous definition of the CAD geometry. Ideally this would be carried out automatically by smart CAD clean up software. One could also define a standard piece-wise smooth surface representation suitable for use by computational methods and then create software to translate between the various CAD descriptions and the standard representation. Surface meshing remains a time consuming, user intensive procedure. There is a need for automated surface meshing, requiring only minimal user intervention to define the overall density of mesh points. The surface mesher should produce well shaped elements (triangles or quadrilaterals) whose size is determined initially according to the surface curvature with a minimum size for flat pieces, and later refined by the user in other regions if necessary. Present techniques for volume meshing all require some degree of user intervention. There is a need for fully automated and reliable volume mesh generation. In addition, it should be possible to create both surface and volume meshes that meet guaranteed measures of mesh quality (e.g. minimum and maximum angle, stretching ratios, etc.).

    Description: The goal of this project is to better understand the process of spatial orientation and navigation in unfamiliar gravito-inertial environments, and ultimately to use this new information to develop effective countermeasures against the orientation and navigation problems experienced by astronauts. How do we know our location, orientation, and motion of our body with respect to the external environment ? On earth, gravity provides a convenient "down" cue. Large body rotations normally occur only in a horizontal plane. In space, the gravitational down cue is absent. When astronauts roll or pitch upside down, they must recognize where things are around them by a process of mental rotation which involves three dimensions, rather than just one. While working in unfamiliar situations they occasionally misinterpret visual cues and experience striking "visual reorientation illusions" (VRIs), in which the walls, ceiling, and floors of the spacecraft exchange subjective identities. VRIs cause disorientation, reaching errors, trigger attacks of space motion sickness, and potentially complicate emergency escape. MIR crewmembers report that 3D relationships between modules - particularly those with different visual verticals - are difficult to visualize, and so navigating through the node that connects them is not instinctive. Crew members learn routes, but their apparent lack of survey knowledge is a concern should fire, power loss, or depressurization limit visibility. Anecdotally, experience in mockups, parabolic flight, neutral buoyancy and virtual reality (VR) simulators helps. However, no techniques have been developed to quantify individual differences in orientation and navigation abilities, or the effectiveness of preflight visual. orientation training. Our understanding of the underlying physiology - for example how our sense of place and orientation is neurally coded in three dimensions in the limbic system of the brain - is incomplete. During the 16 months that this human and animal research project has been underway, we have obtained several results that are not only of basic research interest, but which have practical implications for the architecture and layout of spacecraft interiors and for the development of astronaut spatial orientation training countermeasures.

    Description: Extended spaceflight under microgravity conditions leads to significant atrophy of weight-bearing muscles. Atrophy and hypertrophy are the extreme outcomes of the high degree of plasticity exhibited by skeletal muscle. Stimuli which control muscle plasticity include neuronal, hormonal, nutritional, and mechanical inputs. The mechanical stimulus for muscle is directly related to the work or exercise against a load performed. Little or no work is performed by weight-bearing muscles under microgravity conditions. A major hypothesis is that focal adhesion kinase (FAK) which is associated with integrin at the adherens junctions and costa meres of all skeletal muscles is an integral part of the major mechanism for molecular signaling upon mechanical stimulation in all muscle fibers. Additionally, we propose that myotonic protein kinase (DMPK) and dystrophin (DYSTR) also participate in distinct mechanically stimulated molecular signaling pathways that are most critical in type I and type II muscle fibers, respectively. To test these hypotheses, we will use the paradigms of hindlimb unloading and overloading in mice as models for microgravity conditions and a potential exercise countermeasure, respectively, in mice. We expect that FAK loss-of-function will impair hypertrophy and enhance atrophy in all skeletal muscle fibers whereas DYSTR and DMPK loss-of-function will have similar but more selective effects on Type IT and Type I fibers, respectively. Gene expression will be monitored by muscle-specific creatine kinase M promoter-reporter construct activity and specific MRNA and protein accumulation in the soleus (type I primarily) and plantaris (type 11 primarily) muscles. With these paradigms and assays, the following Specific Project Aims will be tested in genetically altered mice: 1) identify the roles of DYSTR and its pathway; 2) evaluate the roles of the DMPK and its pathway; 3) characterize the roles of FAK and its pathway and 4) genetically analyze the mechanisms and interactions between the FAK, DYSTR, and DMPK-associated pathways in single and specific combinations of mutants. The identification of potential signaling mechanisms may permit future development of pharmacological countermeasures for amelioration and prevention of the microgravity-induced atrophy in extended spaceflight, and the analysis of both overloading and unloading paradigms may provide further support for development of exercise-based countermeasures. Understanding the basic mechanisms of molecular signaling in muscle plasticity may aid our understanding and treatment of skeletal muscle atrophy not only in spaceflight but in similar problems of the aging population, in prolonged bed rest, and in cachexia associated with chronic disease.

    Description: Crew health is a dominant issue in manned space flight. Microbiological concerns, in particular, have repeatedly emerged as determinants of flight readiness. For example, in at least one case, suspected contamination of the potable water supply nearly forced a launch delay. In another instance, a crew member's urinary tract infection nearly led to early termination of the mission, in part due to the difficulty of accurately diagnosing the nature of the infection in-flight. Microbial problems are an increasing concern with the trend towards longer-duration missions. It is essential to the success of such missions that systems that deliver acceptable quality of air and water during the anticipated lifetime of the spacecraft be available. As mission duration and resupply intervals increase, it will be necessary to rely on advanced life support systems which incorporate both biological and physical-chemical recycling methods for air and water as well as provide food for the crew. It therefore is necessary to develop real-time, robust, in-flight monitoring procedures that are sensitive enough to detect less than 100 CFU (colony forming units) of bacteria per 100 milliliters of water. It would be desirable if the monitoring system could be readily "reprogrammed" to identify specific pathogens if an in-flight incident were to occur. Thus, the monitoring technology must simultaneously detect many organisms of interest, be subject to miniaturization and be highly automated The long range goal of project is to develop such monitoring systems.

    Description: The goal of this project is to develop reliable statistical algorithms for on-line analysis of physiologic and neurobehavioral variables monitored during long-duration space missions. Maintenance of physiologic and neurobehavioral homeostasis during long-duration space missions is crucial for ensuring optimal crew performance. If countermeasures are not applied, alterations in homeostasis will occur in nearly all-physiologic systems. During such missions data from most of these systems will be either continually and/or continuously monitored. Therefore, if these data can be analyzed as they are acquired and the status of these systems can be continually assessed, then once alterations are detected, appropriate countermeasures can be applied to correct them. One of the most important physiologic systems in which to maintain homeostasis during long-duration missions is the circadian system. To detect and treat alterations in circadian physiology during long duration space missions requires development of: 1) a ground-based protocol to assess the status of the circadian system under the light-dark environment in which crews in space will typically work; and 2) appropriate statistical methods to make this assessment. The protocol in Project 1, Circadian Entrainment, Sleep-Wake Regulation and Neurobehavioral will study human volunteers under the simulated light-dark environment of long-duration space missions. Therefore, we propose to develop statistical models to characterize in near real time circadian and neurobehavioral physiology under these conditions. The specific aims of this project are to test the hypotheses that: 1) Dynamic statistical methods based on the Kronauer model of the human circadian system can be developed to estimate circadian phase, period, amplitude from core-temperature data collected under simulated light- dark conditions of long-duration space missions. 2) Analytic formulae and numerical algorithms can be developed to compute the error in the estimates of circadian phase, period and amplitude determined from the data in Specific Aim 1. 3) Statistical models can detect reliably in near real- time (daily) significant alternations in the circadian physiology of individual subjects by analyzing the circadian and neurobehavioral data collected in Project 1. 4) Criteria can be developed using the Kronauer model and the recently developed Jewett model of cognitive -performance and subjective alertness to define altered circadian and neurobehavioral physiology and to set conditions for immediate administration of countermeasures.

    Description: Computational models of the cardiovascular system are powerful adjuncts to ground-based and in-flight experiments. We will provide NSBRI with a model capable of simulating the short-term effects of gravity on cardiovascular function. The model from this project will: (1) provide a rational framework which quantitatively defines interactions among complex cardiovascular parameters and which supports the critical interpretation of experimental results and testing of hypotheses. (2) permit predictions of the impact of specific countermeasures in the context of various hypothetical cardiovascular abnormalities induced by microgravity. Major progress has been made during the first 18 months of the program: (1) We have developed an operational first-order computer model capable of simulating the cardiovascular response to orthostatic stress. The model consists of a lumped parameter hemodynamic model and a complete reflex control system. The latter includes cardiopulmonary and carotid sinus reflex limbs and interactions between the two. (2) We have modeled the physiologic stress of tilt table experiments and lower body negative pressure procedures (LBNP). We have verified our model's predictions by comparing them with experimental findings from the literature. (3) We have established collaborative efforts with leading investigators interested in experimental studies of orthostatic intolerance, cardiovascular control, and physiologic responses to space flight. (4) We have established a standardized method of transferring data to our laboratory from the ongoing NSBRI bedrest studies. We use this data to estimate input parameters to our model and compare our model predictions to actual data to further verify our model. (5) We are in the process of systematically simulating current hypotheses concerning the mechanism underlying orthostatic intolerance by matching our simulations to stand test data from astronauts pre- and post-flight. (6) We are in the process of developing a JAVA version of the simulator which will be distributed amongst the cardiovascular team members. Future work on this project involves modifications of the model to represent a rodent (rat) model, further evaluation of the bedrest astronaut and animal data, and systematic investigation of specific countermeasures.

    Description: Major cardiovascular problems, secondary to cardiovascular deconditioning, may occur on extended space missions. While it is generally assumed that the microgravity state is the primary cause of cardiovascular deconditioning, sleep deprivation and disruption of diurnal rhythms may also play an important role. Factors that could be modified by either or both of these perturbations include: autonomic function and short-term cardiovascular reflexes, vasoreactivity, circadian rhythm of cardiovascular hormones (specifically the renin-angiotensin system) and renal sodium handling and hormonal influences on that process, venous compliance, cardiac mass, and cardiac conduction processes. The purpose of the Human Studies Core is to provide the infrastructure to conduct human experiments which will allow for the assessment of the likely role of such factors in the space travel associated cardiovascular deconditioning process and to develop appropriate countermeasures. The Core takes advantage of a newly-created Intensive Physiologic Monitoring (IPM) Unit at the Brigham and Women's Hospital, Boston, MA, to perform these studies. The Core includes two general experimental protocols. The first protocol involves a head down tilt bed-rest study to simulate microgravity. The second protocol includes the addition of a disruption of circadian rhythms to the simulated microgravity environment. Before and after each of these environmental manipulations, the subjects will undergo acute stressors simulating changes in volume and/or stress, which could occur in space and on return to Earth. The subjects are maintained in a rigidly controlled environment with fixed light/dark cycles, activity pattern, and dietary intake of nutrients, fluids, ions and calories.

    Description: Blood flow to bone has been shown to affect bone mass and presumably bone strength. Preliminary data indicate that blood flow to the rat femur decreases after 14 days of simulated microgravity, using hindlimb suspension (HLS). If adult rats subjected to HLS are given dobutamine, a synthetic catecholamine which can cause peripheral vasodilation and increased blood flow, the loss of cortical bone area usually observed is prevented. Further, mechanisms exist at the molecular level to link changes in bone blood flow to changes in bone cell activity, particularly for vasoactive agents like nitric oxide (NO). The decreases in fluid shear stress created by fluid flow associated with the shifts of plasma volume during microgravity may result in alterations in expression of vasoactive agents such as NO, producing important functional effects on bone cells. The primary aim of this project is to characterize changes in 1) bone blood flow, 2) indices of bone mass, geometry, and strength, and 3) changes in gene expression for modulators of nitric oxide activity (e.g., nitric oxide synthase) and other candidate genes involved in signal transduction of mechanical loading after 3, 7, 14, 21, and 28 days of HLS in the adult rat. Using a rat of at least 5 months of age avoids inadvertently studying effects of simulated microgravity on growing, rather than adult, bone. Utilizing the results of these studies, we will then define how altered blood flow contributes to changes in bone with simulated microgravity by administering a vasodilatory agent (which increases blood flow to tissues) during hindlimb suspension. In all studies, responses in the unloaded hindlimb bones (tibial shaft, femoral neck) will be compared with those in the weightbearing humeral shaft and the non-weightbearing calvarium (skull) from the same animal. Bone volumetric mineral density and geometry will be quantified by peripheral quantitative CT; structural and material properties of the long bones will be determined by 3-point bending (tibia, humerus) or compression (femoral neck) testing to failure. A unique aspect of these studies will be defining the time course of changes in gene expression in bone cell populations with unloading, accomplished with Northern blots, in situ hybridization, and immunohistochemistry. These studies have high relevance for concurrent protocols being proposed by investigators on NSBRI Cardiovascular and Muscle teams, with blood flow data available on a number of tissues other than bone. Further, dobutamine and other Beta-agonists have been tested as countermeasures for altered muscle and cardiovascular function. Results of the intervention tested in our studies have potential relevance for a number of systemic changes seen with prolonged spaceflight.

    Description: Conventional CFD methods and grids do not yield adequate resolution of the complex shock flow pattern generated by a real aircraft geometry. As a result, a unique grid topology and supersonic flow solver was developed at Northrop Grumman based on the characteristic behavior of supersonic wave patterns emanating from the aircraft. Using this approach, it was possible to compute flow fields with adequate resolution several body lengths below the aircraft. In this region, three-dimensional effects are diminished and conventional two-dimensional modified linear theory (MLT) can be applied to estimate ground pressure signatures or sonic booms. To accommodate real aircraft geometries and alleviate the burdensome grid generation task, an implicit marching multi-block, multi-grid finite-volume Euler code was developed as the basis for the sonic boom prediction methodology. The Thomas two-dimensional extrapolation method is built into the Euler code so that ground signatures can be obtained quickly and efficiently with minimum computational effort suitable to the aircraft design environment. The loudness levels of these signatures can then be determined using a NASA generated noise code. Since the Euler code is a three-dimensional flow field solver, the complete circumferential region below the aircraft is computed. The extrapolation of all this field data from a cylinder of constant radius leads to the definition of the entire boom corridor occurring directly below and off to the side of the aircraft's flight path yielding an estimate for the entire noise "annoyance" corridor in miles as well as its magnitude. An automated multidisciplinary sonic boom design optimization software system was developed during the latter part of HSR Phase 1. Using this system, it was found that sonic boom signatures could be reduced through optimization of a variety of geometric aircraft parameters. This system uses a gradient based nonlinear optimizer as the driver in conjunction with a computationally efficient Euler CFD solver (NIIM3DSB) for computing the three-dimensional near-field characteristics of the aircraft. The intent of the design system is to identify and optimize geometric design variables that have a beneficial impact on the ground sonic boom. The system uses a simple wave drag data format to specify the aircraft geometry. The geometry is internally enhanced and analytic methods are used to generate marching grids suitable for the multi-block Euler solver. The Thomas extrapolation method is integrated into this system, and hence, the aircraft's centerline ground sonic boom signature is also automatically computed for a specified cruise altitude and yields the parameters necessary to evaluate the design function. The entire design system has been automated since the gradient based optimization software requires many flow analyses in order to obtain the required sensitivity derivatives for each design variable in order to converge on an optimal solution. Hence, once the problem is defined which includes defining the objective function and geometric and aerodynamic constraints, the system will automatically regenerate the perturbed geometry, the necessary grids, the Euler solution, and finally the ground sonic boom signature at the request of the optimizer.

    Description: The rationale for looking for prokaryote fossils in Martian materials is based on our present understanding of the environmental evolution of that planet in comparison to the history of the terrestrial environments and the development and evolution of life on Earth. On Earth we have clear, albeit indirect, evidence of life in 3.8 b.y.-old rocks from Greenland and the first morphological fossils in 3.3-3.5 b.y.-old cherts from South Africa and Australia. In comparison, Mars, being smaller, probably cooled down after initial aggregation faster than the Earth. Consequently, there could have been liquid water on its surface earlier than on Earth. With a similar exogenous and endogenous input of organics and life-sustaining nutrients as is proposed for the Earth, life could have arisen on that planet, possibly slightly earlier dm it did on Earth. Whereas on Earth liquid water has remained at the surface of the planet since about 4.4 b.y. (with some possible interregnums caused by planet-sterilising impacts before 3.8. b.y. and perhaps a number of periods of a totally frozen Earth, this was not the case with Mars. Although it is not known exactly when surficial water disappeared from the surface, there would have been sufficient time for life to have developed into something similar to the terrestrial prokaryote stage. However, given the earlier environmental deterioration, it is unlikely that it evolved into the eukaryote stage and even evolution of oxygenic photosynthesis may not have been reached. Thus, the impetus of research is on single celled life simnilar to prokaryotes. We are investigating a number of methods of trace element analysis with respect to the Early Archaean microbial fossils. Preliminary neutron activation analysis of carbonaceous layers in the Early Archaean cherts from South Africa and Australia shows some partitioning of elements such as As, Sb, Cr with an especial enrichment of lanthanides in a carbonaceous-rich banded iron sediment . More significantly, preliminary TOF-SIMS investigations of organics in the cherts reveals the presence of a biomarker, which appears to be a derivative of bacterial polymer, in the carbonaceous parts of the rocks. We conclude that a combination of morphological, isotope and biogeochemical methods can be used to successfully identify signs of life in terrestrial material, and that these methods will be useful in searching for signs of life in extraterrestrial materials.

    Description: The 13 samples from Mars identified in the terrestrial meteorite collections vary from dunite to pyroxenite to microgabbro or basalt. All of these rocks appear to have formed from primitive melts with similar major element compositional characteristics; i.e., FeO-rich and Al2O3-Poor melts relative to terrestrial basalt compositions. Although all of the SNC rocks can be derived by melting of the same Al-depleted mantle, contamination of SNC's by a Rb-enriched mantle or crustal source is required to explain the different REE characteristics of SNC rocks. Thus, there are indications of an old crustal rocktype on Mars, and this rock does not appear to have been sampled. This paper focuses primarily on the composition of the SNC basalts, however, and on the compositions of rocks which could be derived from SNC basaltic melt by magmatic processes. In particular, we consider the possible compositions which could be achieved through accumulation of early-formed crystals in the SNC primitive magma. Through a set of experiments we have determined (1) melt (magma) compositions which could be produced by melt evolution as crystals are removed from batches of this magma cooling at depth, and (2) which evolved (Si02enriched, MgO-depleted) rock compositions could be produced from the SNC magma, and how these compare with the Pathfinder andesite composition. Finally, we compare the SNC magma compositions to the Mars soil composition in order to determine whether any source other than SNC is required.

    Description: The Mars Surveyor 2001 Project consists of two missions to Mars, an Orbiter and a Lander, both to be launched in the spring of 2001 for October 2001 (Orbiter) and January 2002 (Lander) arrival at Mars. The Orbiter will support the Lander mission primarily as a communications relay system; the Lander will not have direct-to-Earth communications capability. Science data collected from the Orbiter will also be used to aid in the geologic interpretation of the landing site, along with data from past missions. Combining the Orbiter and Lander missions into a single Project has enabled the streamlining of many activities and an efficient use of personnel and other resources at the Jet Propulsion Laboratory and at the spacecraft contractor, Lockheed Martin Astronautics.

    Description: The MINUTES instrument of the Athena Precursor Experiment (APEX) on the Mars Surveyor 2001 lander mission will perform the first thermal infrared remote sensing observations from the surface of another planet. Experience gained from this experiment will be used to guide observations from identical instruments mounted on the Athena rovers, to be launched in 2003 and 2005. The utility of infrared spectrometers in determining the mineralogic composition of geologic surfaces from airborne and spaceborne platforms has been amply demonstrated. However, relatively little experience exists in using functionally similar instruments on the ground in the context of planetary science. What work has been done on this problem has mostly utilized field spectrometers that are designed to look down on nearby target rocks. While many Mini-TES observations will be made with this type of geometry, it is likely that other observations will be made looking horizontally at the more vertically-oriented facets of rock targets, to avoid spectral contamination from dust mantles. On rover missions, the Mini-TES may also be pointed horizontally at rocks several meters away, to determine if they are worthy of approaching for in situ observations and possible sample cacheing. While these observations will undoubtedly prove useful, there are important, and perhaps unappreciated, differences between horizontal-viewing, surface-based spectroscopy and the more traditional nadir-viewing, orbit or aircraft-based observations. Plans also exist to step the Mini-TES in a rastering motion to build hyperspectral scenes. Horizontal viewing hyperspectral cubes also possess unique qualities that call for innovative analysis techniques. The effect of viewing geometry: In thermal emission spectroscopy, regardless of whether an instrument is looking down on or horizontally at a target, the same basic equation governs the radiance reaching the sensor .

    Description: In a previous publication, we proposed the formation of caves at mega and microscale on Mars and emphasized their potential for the exobiology exploration. The recent MOC images have shown promising indicators that caves are actually existing on Mars. In the first section, we develop the theoretical potential formation of martian caves. Then, we show how MOC is supporting this hypothesis of their formation and the new types of environments it suggests. The existence of caves on Mars from microscale to microscale structures can be predicted according to the Mars geological and climatic history. A first global approach is to consider caves as a result of underground water activity combined with tectonic movement. They can be formed by: (1) diversion of channel courses in underground conduits; (2) fractures of surface drainage patterns; chaotic terrain and collapsed areas in general; (4) seepage face in valley walls and/or headwaters; (5) inactive hydrothermal vents and lava tubes.

    Description: The primary objective of the Thermal Emission Imaging System (THEMIS) on the Mars Surveyor '01 Orbiter is to study the composition of the Martian surface at high spatial resolution. THEMIS will map the surface mineralogy using multi-spectral thermal infrared images in 8 spectral bands from 6.5 to 14.5 microns. In addition, a band centered at 15 microns will be used to map atmospheric temperatures and provide an important aid in separating the surface and atmospheric components. The entire planet will be mapped at 100 m resolution within the available data volume using a multi-spectral, rather than hyperspectral, imaging approach. THEMIS will also acquire 20 m resolution visible images in up to 5 spectral bands using a replica of the Mars 98 Orbiter (MARCI) and Lander (MARDI) cameras. Over 15,000 panchromatic (3,000 5-color), 20 x 20 km images will be acquired for morphology studies and landing site selection. The thermal-infrared spectral region contains the fundamental vibrational absorption bands of most minerals which provide diagnostic information on mineral composition. All geologic materials, including carbonates, hydrothermal silica, sulfates, phosphates, hydroxides, silicates, and oxides have strong absorptions in the 6.5-14.5 micron region. Silica and carbonates, which are key diagnostic minerals in thermal spring deposits, are readily identified using thermal-IR spectra. In addition, the ability to identify all minerals allows the presence of aqueous minerals to be interpreted in the proper geologic context. An extensive suite of studies over the past 35 years has demonstrated the utility of vibrational spectroscopy for the quantitative determination of mineralogy and petrology. The fundamental vibrations within different anion groups, such as C03, S04, P04, and SiO4, produce unique, well separated spectral bands that allow carbonates, sulfates, phosphates, silicates, oxides, and hydroxides to be readily identified. Additional stretching and bending modes involving major cations, such as Mg, Fe, Ca, and Na, allow Further mineral identification, such as the excellent discriminability of minerals within the silicate and carbonate groups.

    Description: Impairment of gaze and head stabilization reflexes can lead to disorientation and reduced performance in sensorimotor tasks such as piloting of spacecraft. Transitions between different gravitoinertial force (gif) environments - as during different phases of space flight - provide an extreme test of the adaptive capabilities of these mechanisms. We wish to determine to what extent the sensorimotor skills acquired in one gravity environment will transfer to others, and to what extent gravity serves as a context cue for inhibiting such transfer. We use the general approach of adapting a response (saccades, vestibuloocular reflex: VOR, or vestibulocollic reflex: VCR) to a particular change in gain or phase in one gif condition, adapting to a different gain or phase in a second gif condition, and then seeing if gif itself - the context cue - can recall the previously-learned adapted responses. Previous evidence indicates that unless there is specific training to induce context-specificity, reflex adaptation is sequential rather than simultaneous. Various experiments in this project investigate the behavioral properties, neurophysiological basis, and anatomical substrate of context-specific learning, using otolith (gravity) signals as a context cue. In the following, we outline the methods for all experiments in this project, and provide details and results on selected experiments.

    Description: Loss of bone mineral during space flight was documented in the 1970's Skylab missions. The USSR space program made similar observations in the 1980's. The Institute of Biomedical Problems in Moscow and NASA JSC in 1989 began to collect pre- and post-flight bone mineral density (BMD) using Hologic QDR 1000 DEXA scanners transferred from JSC to Moscow and Star City. DEXA whole body, hip, and lumbar spine scans were performed prior to and during the first week after return from 4- to 6-month missions (plus one 8-month mission and one 14- month mission) on the Mir space station. These data documented the extent and regional nature of bone loss during long duration space flight. Of the 18 cosmonauts participating in this study between 1990 and 1995, seven flew two missions. BMD scans prior to the second flight compared to the first mission preflight scans indicated that recovery was possibly delayed or incomplete. Because of these findings, NASA and IBMP initiated the study "Bone Mineral Loss and Recovery After Shuttle/Mir Flights" in 1995 to evaluate bone recovery during a 3-year post-flight period. All of the 14 participants thus far evaluated lost bone in at least one region of the spine and lower extremities during flight. Of the 14, only one to date has exhibited full return to baseline BNM values in all regions. The current study will continue until the last participant has reached full bone recovery in all regions, has reached a plateau, or until three years after the flight (2001 for the last mission of the program). Bone mineral density losses in space and difficulty in returning to baseline indicate a need for countermeasure development. In late 1996 NASA JSC and Baylor College of Medicine were approved to conduct two countermeasure studies during 17 weeks of bed rest. In 1997 the studies were begun in the bed rest facility established by NASA, Baylor College of Medicine, and The Methodist Hospital in Houston. To date, three bed rest controls, five resistive exercisers, and four subjects taking alendronate (a bisphoshonate that inhibits osteoclastic resorption of bone) have completed 17 weeks bed rest. In contrast to information currently available from space flight (n=28) and bed rest (n= 12) in which all individuals experienced bone loss in at least one region, one of four subjects taking alendronate and one of five subjects performing heavy resistive exercise at bed rest fully maintained bone density in all regions of the spine and lower extremities. Overall results of both countermeasures which will be presented are encouraging. The study will be completed by mid to late 2000 with 10 subjects in each of three groups.

    Description: High salt diets accelerate bone loss with aging in patients with postmenopausal osteoporosis except when calcium supplementation is provided. We have observed that the decrease in mineral content of growing femurs in juvenile rats, exposed to a space flight model which unloads the hind limbs , is substantially less in animals fed excess salt. To determine whether excess dietary salt has the same effect on the skeleton of the mature animal whose response to unloading is increased resorption and bone loss rather than impaired growth, we carried out a metabolic study in mature rats with hindlimbs unloaded by tailsuspension.

    Description: Promethazine (PMZ) is the antimotion sickness drug of choice in the U.S. Space Shuttle program; however, virtually nothing is known about the bioavailability and performance effects of this drug in the microgravity environment. PMZ has detrimental side effects on human performance on Earth that could affect Shuttle operations. In a recent ground-based study we examined: 1) the effects of promethazine (PMZ) on Shuttle landing performance using the portable inflight landing operations trainer (PILOT), and 2) saliva and urine samples to determine the pharmacokinetics of PMZ. The PILOT performance data is presented here.

    Description: This report considers the effect of canard and horizontal tail vertical position on the aerodynamic characteristics of the PTC configuration without nacelles and diverters. This analysis is followed by three optimization studies using canard and tail incidence as design variables in the first problem followed by an optimization run with canard and tail incidence and wing camber design variables and finally an optimization run with canard incidence and wing camber. The first problem was run at fixed lift while the other two problems were run at fixed angle of attack. The final investigation reported here will show data from a component buildup study using the PTC configuration. This final study will show the aerodynamic interference between the canard, wing and horizontal tail.

The studies include a preliminary assessment of the behavior of the two new 2-equation turbulence models introduced with the latest version of OVERFLOW (v. 1.8f), an investigation into potential improvements in the matrix dissipation scheme currently implemented in OVERFLOW, and an analysis of the observed sensitivity of the code's skin friction predictions to grid stretching at solid surface boundaries.

    Description: The computational results of the optimized complete configurations, including nacelles and diverters, are presented in terms of drag count improvement compared with the TCA baseline configuration at Mach 2.4, C(sub L)=0.1. The three candidate designs are designated by the organization from which they were derived. ARC represents the Ames Research Center 1-03 design, BCAG represents the Boeing Commercial Aircraft Group's design from Seattle, and BLB represents the design from Boeing Long Beach. All CFD methods are in unanimous agreement that the Ames 1-03 configuration has the largest performance improvement, followed closely by the BCAG configuration, with a much smaller improvement attained by Boeing Long Beach. The Ames design was obtained using the single-block wing/body code SYN87-SB with its "pseudo" nacelle option-an elaborate technique for incorporating nacelle/diverter effects into the design optimization process. This technique uses AIRPLANE surface pressure coefficient data with and without the nacelles/diverters. Further details of this method are described. It is reasonable to expect that further improvements could be achieved by including the "real" nacelles directly into the optimization process by use of the newly-developed multiblock optimization code, SYN107-MB, which can handle full configurations.

    Description: The aim of this work is to demonstrate a simple technique which accounts for aeroelastic deformations experienced by HSR wind-tunnel models within CFD computations. With improved correlations, CFD can become a more effective tool for augmenting the post-test understanding of experimental data. The present technique involves the loose coupling of a low-level structural representation within the ELAPS code, to an unstructured Navier-Stokes flow solver, USM3Dns. The ELAPS model is initially calibrated against bending characteristics of the wind-tunnel model. The strength of this method is that, with a single point calibration of a simple structural representation, the static aeroelastic effects can be accounted for in CFD calculations across a range of test conditions. No prior knowledge of the model deformation during the wind-on test is required. This approach has been successfully applied to the high aspect-ratio planforms of subsonic transports. The current challenge is to adapt the procedure to low aspect-ratio planforms typical of HSR configurations.

    Description: LaRC conducted a code validation study for the OVERFLOW code to ascertain its accuracy for boattail drag prediction. The OVERFLOW results compared favorably with the LaRC 16-ft. Transonic Wind Tunnel (TWT) data, and prior CFD solutions from PAB3D and CFL3D. The ultimate goal is to investigate the installation drag of the nacelle boattails with powered nozzles at transonic mach numbers. The OVERFLOW solver was chosen because of its ability to accept volume overlapping structured grid for very complex airframe configurations. Structured grid components for representing the transonic nozzle boattail can be added to the BCAG grid for a TCA airframe with 2D bifurcated inlet and flow through nacelle without alteration. The focus of this research was to determine the suitability of the OVERFLOW solver for accomplishing this ultimate goal. This presentation will first introduce the transonic nozzle boattail wind-tunnel model geometry, followed by an examination of aerodynamic features based on the current OVERFLOW solutions and the solutions obtained previously using PAB3D, comparisons of Cp on the flap surface between the OVERFLOW solutions, wind tunnel data, and solutions from other CFD codes, an assessment of boattail drag count prediction, and a work plan for FY99.

    Description: A concept of system identification applied to high performance aircraft is introduced followed by a discussion on the identification methodology. Special emphasis is given to model postulation using time invariant and time dependent aerodynamic parameters, model structure determination and parameter estimation using ordinary least squares and mixed estimation methods. At the same time problems of data collinearity detection and its assessment are discussed. These parts of methodology are demonstrated in examples using flight data of the X-29A and X-31A aircraft. In the third example wind tunnel oscillatory data of the F-16XL model are used. A strong dependence of these data on frequency led to the development of models with unsteady aerodynamic terms in the form of indicial functions. The paper is completed by concluding remarks.

    Description: This paper reports the predicted M = 2.4 strut-interference effects on a closed aftbody with empennage for the TCA baseline model. The strut mounting technique was needed in order to assess the impact of aft-end shaping, i.e. open for a sting or closed to better represent a flight vehicle. However,this technique can potentially lead to unanticipated effects that are measured on the aft body. Therefore, a set of computations were performed in order to examine the closed aft body with and without strut present, at both zero and non-zero angles of sideslip (AOS). The work was divided into a computational task performed by Javier A. Garriz, using an inviscid (Euler) solver, and a monitoring/reporting task done by John E. Lamar. All this work was performed during FY98 at the NASA Langley Research Center.

    Description: One of the original objectives of the Mars Orbiter Camera (MOC), as proposed in 1985, was to acquire observations to be used in assessing future spacecraft landing sites. Images obtained by the Mars Global Surveyor MOC since March 1999 provide the highest resolution views (1.5-4.5 m/pixel) of the planet ever seen. We have been examining these new data to develop a general view of what Mars is like at meter-scale within the latitudes and elevations that are accessible to the Mars Surveyor 2001 lander. Our goal is to provide guidance to the 2001 landing site selection process, rather than to use MOC images to recommend a specific landing site.

    Description: Purpose. It is generally accepted that DCS symptoms are caused by gas bubbles in tissues. However, current technology of bubble detection only permits monitoring of circulating bubbles, primarily intracardiac. Since the majority of DCS symptoms appear to be caused by extravascular bubbles, it has been suggested that current bubble detection techniques target bubbles that are of importance in only a minority of DCS cases. The purpose of this study is to determine the relationships between measured VGE and DCS symptoms in human subjects exposed to altitude. Methods. The AFRL DCS Research Database contains records on 2044 subject-exposures to simulated altitudes in a hypobaric chamber. VGE monitoring was accomplished using Doppler/Echo Imaging techniques. The Spencer Scale was used to score the VGE. Reporting of DCS symptoms by the subject was the primary end-point of the exposures. Results: The Mantel- Haenzel test indicated a strong correlation between DCS and bubble grade (p-value =0.001). Conclusions. A positive correlation between increasing VGE scores and DCS symptoms, does not imply causatinn. If all non-zero VGE grades are considered, 45.9% of the cases had VGE, but no DCS symptoms. Conversely, almost 1 in 5 subject-exposures resulted in DCS with NO VGE detected. VGE scores are not . good predictors of altitude DCS symptoms and field use of bubble detection for DCS prevention is not supported by this study.

    Description: Elevations measured by the Mars Orbiter Laser Altimeter have yielded a high-accuracy global map of the topography of Mars. Dominant features include the low northern hemisphere, the Tharsis province, and the Hellas impact basin. The northern hemisphere depression is primarily a long-wavelength effect that has been shaped by an internal mechanism. The topography of Tharsis consists of two broad rises. Material excavated from Hellas contributes to the high elevation of the southern hemisphere and to the scarp along the hemispheric boundary. The present topography has three major drainage centers, with the northern lowlands being the largest. The two polar cap volumes yield an upper limit of the present surface water inventory of 3.2 to 4.7 million cubic kilometers.

    Description: Artificial gravity (AG), as a means of preventing physiological deconditioning of astronauts during long-duration space flights, presents certain special challenges to the otolith organs and the adaptive capabilities of the CNS. The key issues regarding the choice of AG acceleration, radius, and rotation rate are reviewed from the viewpoints of physiological requirements and human factors disturbances. Head movements and resultant Coriolis forces on the rotating platform may limit the usefulness of economical short centrifuges for other than brief periods of intermittent stimulation.

    Description: The objective of this study was to determine whether altered intracellular Ca(2+) handling contributes to the specific force loss in the soleus muscle after unloading and/or subsequent reloading of mouse hindlimbs. Three groups of female ICR mice were studied: 1) unloaded mice (n = 11) that were hindlimb suspended for 14 days, 2) reloaded mice (n = 10) that were returned to their cages for 1 day after 14 days of hindlimb suspension, and 3) control mice (n = 10) that had normal cage activity. Maximum isometric tetanic force (P(o)) was determined in the soleus muscle from the left hindlimb, and resting free cytosolic Ca(2+) concentration ([Ca(2+)](i)), tetanic [Ca(2+)](i), and 4-chloro-m-cresol-induced [Ca(2+)](i) were measured in the contralateral soleus muscle by confocal laser scanning microscopy. Unloading and reloading increased resting [Ca(2+)](i) above control by 36% and 24%, respectively. Although unloading reduced P(o) and specific force by 58% and 24%, respectively, compared with control mice, there was no difference in tetanic [Ca(2+)](i). P(o), specific force, and tetanic [Ca(2+)](i) were reduced by 58%, 23%, and 23%, respectively, in the reloaded animals compared with control mice; however, tetanic [Ca(2+)](i) was not different between unloaded and reloaded mice. These data indicate that although hindlimb suspension results in disturbed intracellular Ca(2+) homeostasis, changes in tetanic [Ca(2+)](i) do not contribute to force deficits. Compared with unloading, 24 h of physiological reloading in the mouse do not result in further changes in maximal strength or tetanic [Ca(2+)](i).

    Description: A suite of protein and nonprotein amino acids were detected with high-performance liquid chromatography in the water- and acid-soluble components of an interior fragment of the Martian meteorite Nakhla, which fell in Egypt in 1911. Aspartic and glutamic acids, glycine, alanine, beta-alanine, and gamma-amino-n-butyric acid (gamma-ABA) were the most abundant amino acids detected and were found primarily in the 6 M HCl-hydrolyzed, hot water extract. The concentrations ranged from 20 to 330 parts per billion of bulk meteorite. The amino acid distribution in Nakhla, including the D/L ratios (values range from 〈0.1 to 0.5), is similar to what is found in bacterially degraded organic matter. The amino acids in Nakhla appear to be derived from terrestrial organic matter that infiltrated the meteorite soon after its fall to Earth, although it is possible that some of the amino acids are endogenous to the meteorite. The rapid amino acid contamination of Martian meteorites after direct exposure to the terrestrial environment has important implications for Mars sample-return missions and the curation of the samples from the time of their delivery to Earth.

    Description: In patients with autonomic failure orthostatic hypotension results from an impaired capacity to increase vascular resistance during standing. This fundamental defect leads to increased downward pooling of venous blood and a consequent reduction in stroke volume and cardiac output that exaggerates the orthostatic fall in blood pressure. The location of excessive venous blood pooling has not been established so far, but present data suggest that the abdominal compartment and perhaps leg skin vasculature are the most likely candidates. To improve the orthostatic tolerance in patients with autonomic failure, protective measures that reduce excessive orthostatic blood pooling have been developed and evaluated. These measures include physical counter-manoeuvres and abdominal compression.

    Description: The final decade of the millennium has seen an enormous amount of on-orbit life sciences research, including both short- and long-duration flight research. Life sciences dedicated Space Shuttle flights have made intensive research opportunities available to study on the acute adaptation to weightlessness. The NASA/Mir Science Program combined resources of the USA and Russia to provide the first long-duration flight opportunities for the United States since the Skylab program of the early 1970s. Many of the results of these studies are still being evaluated, and in some cases data are still being collected to assess long-term readaptation to gravity after several months in weightlessness. The surge in life sciences research during this decade serves as a preamble to the opportunities to be provided by the latest addition to the Earth-orbiting structures--the International Space Station.

    Description: As the twentieth century closes, retrospectives cite the Apollo moon missions as one of the important events of the past 100 years. A trip to Mars, however, would be even more challenging and significant. A round-trip Mars journey would require nearly three years away from Earth, a significant leap in complexity compared to the two week long Moon trips or the record-breaking fourteen-month flight on Mir. What would be the physiologic and medical challenges of a Mars flight? Two key areas of physiology present the greatest potential problems--calcium metabolism and radiation exposure. Data from Mir missions show that bone loss continues in space despite an aggressive countermeasure program. Average losses were 0.35% per month, but some load bearing areas lost 〉1% per month. A 1% loss rate, if it continued unabated for 30 months, could produce osteoporosis. Smaller losses could still increase fracture risk. Some bone loss can be well tolerated, particularly if the bone can be regained after the mission. But the effectiveness of post-flight rehabilitation to restore the density and quality of bone after spaceflight is not well known. Bone loss estimates are based on continuous weightlessness exposure, but this is not a requirement for a Mars trip. Most of the time on a Mars trip will be spent in the 1/3 Earth's gravity environment on Mars, and either intermittent or continuous artificial gravity can be provided for the transit between planets (although at an engineering cost). The dosing of the gravity exposure (e.g. the level and duration), however, has not been established. Radiation protection also requires a balance between engineering cost and human health. Excessive shielding could add billions of dollars to the cost of a mission. Trips in interplanetary space, however, expose the crew to heavy high-energy particles from cosmic rays (HZE particles), which have a high linear energy transfer. This high energy leads to significant biological damage (e.g. chromosomal aberrations, cancer induction). A recent report from the Committee on Space Biology and Medicine notes that only one systematic study of cancer induction from high-energy particles has been conducted (using the mouse Harderian gland). Predictions of cancer risk and acceptable radiation exposure in space are extrapolated from minimal data. Other areas of physiology also present problems, such as muscle loss, cardiovascular deconditioning, and vestibular adaptation. Despite all the issues, however, a focussed, aggressive research program that uses the resources of the International Space Station should pave the way for mankind's greatest adventure--a trip to Mars.

    Description: Observations of the gravity field of Mars reveal a planet that has responded differently in its northern and southern hemispheres to major impacts and volcanic processes. The rough, elevated southern hemisphere has a relatively featureless gravitational signature indicating a state of near-isostatic compensation, whereas the smooth, low northern plains display a wider range of gravitational anomalies that indicates a thinner but stronger surface layer than in the south. The northern hemisphere shows evidence for buried impact basins, although none large enough to explain the hemispheric elevation difference. The gravitational potential signature of Tharsis is approximately axisymmetric and contains the Tharsis Montes but not the Olympus Mons or Alba Patera volcanoes. The gravity signature of Valles Marineris extends into Chryse and provides an estimate of material removed by early fluvial activity.

    Description: The effects of spaceflight on mammary metabolism of 10 pregnant rats was measured on Day 20 of pregnancy and after parturition. Rats were flown on the space shuttle from Day 11 through Day 20 of pregnancy. After their return to earth, glucose oxidation to carbon dioxide increased 43% (P 〈 0.05), and incorporation into fatty acids increased 300% (P 〈 0.005) compared to controls. It is unclear whether the enhanced glucose use is due to spaceflight or a response to landing. Casein mRNA and gross histology were not altered at Day 20 of pregnancy. Six rats gave birth (on Day 22 to 23 of pregnancy) and mammary metabolic activity was measured immediately postpartum. The earlier effects of spaceflight were no longer apparent. There was also no difference in expression of beta-casein mRNA. It is clear from these studies that spaceflight does not impair the normal development of the mammary gland, its ability to use glucose, nor the ability to express mRNA for a major milk protein.

    Description: A comparison of laboratory spectra with Galileo data indicates that hydrated sulfuric acid is present and is a major component of Europa's surface. In addition, this moon's visually dark surface material, which spatially correlates with the sulfuric acid concentration, is identified as radiolytically altered sulfur polymers. Radiolysis of the surface by magnetospheric plasma bombardment continuously cycles sulfur between three forms: sulfuric acid, sulfur dioxide, and sulfur polymers, with sulfuric acid being about 50 times as abundant as the other forms. Enhanced sulfuric acid concentrations are found in Europa's geologically young terrains, suggesting that low-temperature, liquid sulfuric acid may influence geological processes.

    Description: The establishment of long-term cultures of functional primary human liver cells (PHLC) is formidable. Developed at NASA, the Rotary Cell Culture System (RCCS) allows the creation of the unique microgravity environment of low shear force, high-mass transfer, and 3-dimensional cell culture of dissimilar cell types. The aim of our study was to establish long-term hepatocyte cultures in simulated microgravity. PHLC were harvested from human livers by collagenase perfusion and were cultured in RCCS. PHLC aggregates were readily formed and increased up to 1 cm long. The expansion of PHLC in bioreactors was further evaluated with microcarriers and biodegradable scaffolds. While microcarriers were not conducive to formation of spheroids, PHLC cultured with biodegradable scaffolds formed aggregates up to 3 cm long. Analyses of PHLC spheroids revealed tissue-like structures composed of hepatocytes, biliary epithelial cells, and/or progenitor liver cells that were arranged as bile duct-like structures along nascent vascular sprouts. Electron microscopy revealed groups of cohesive hepatocytes surrounded by complex stromal structures and reticulin fibers, bile canaliculi with multiple microvilli, and tight cellular junctions. Albumin mRNA was expressed throughout the 60-d culture. A simulated microgravity environment is conducive to maintaining long-term cultures of functional hepatocytes. This model system will assist in developing improved protocols for autologous hepatocyte transplantation, gene therapy, and liver assist devices, and facilitate studies of liver regeneration and cell-to-cell interactions that occur in vivo.

    Description: The ability of living organisms to survive on the smaller bodies in our solar system is examined. The three most significant sterilizing effects include ionizing radiation, prolonged extreme vacuum, and relentless thermal inactivation. Each could be effectively lethal, and even more so in combination, if organisms at some time resided in the surfaces of airless small bodies located near or in the inner solar system. Deep within volatile-rich bodies, certain environments theoretically might provide protection of dormant organisms against these sterilizing factors. Sterility of surface materials to tens or hundreds of centimeters of depth appears inevitable, and to greater depths for bodies which have resided for long periods sunward of about 2 A.U.

    Description: Skeletal unloading leads to decreased bone formation and decreased bone mass. Bone resorption is uncoupled from bone formation, contributing to the bone loss. During spaceflight bone is lost principally from the bones most loaded in the 1-g environment, and some redistribution of bone from the lower extremities to the head appears to take place. Although changes in calcitropic hormones have been demonstrated during skeletal unloading (PTH and 1,25(OH)2D decrease), it remains unclear whether such changes account for or are in response to the changes in bone formation and resorption. Bed rest studies with human volunteers and hindlimb elevation studies with rats have provided useful data to help explain the changes in bone formation during spaceflight. These models of skeletal unloading reproduce a number of the conditions associated with microgravity, and the findings from such studies confirm many of the observations made during spaceflight. Determining the mechanism(s) by which loading of bone is sensed and translated into a signal(s) controlling bone formation remains the holy grail in this field. Such investigations couple biophysics to biochemistry to cell and molecular biology. Although studies with cell cultures have revealed biochemical responses to mechanical loads comparable to that seen in intact bone, it seems likely that matrix-cell interactions underlie much of the mechanocoupling. The role for systemic hormones such as PTH, GH, and 1,25(OH)2D compared to locally produced factors such as IGF-I, PTHrP, BMPs, and TGF-beta in modulating the cellular response to load remains unclear. As the mechanism(s) by which bone responds to mechanical load with increased bone formation are further elucidated, applications of this knowledge to other etiologies of osteoporosis are likely to develop. Skeletal unloading provides a perturbation in bone mineral homeostasis that can be used to understand the mechanisms by which bone mineral homeostasis is maintained, with the expectation that such understanding will lead to effective treatment for disuse osteoporosis.

    Description: PURPOSE: This review is to evaluate the use of biomarkers as an indication of past exposure to radiation or other environmental insults, individual sensitivity and risk for the development of late occurring disease. OVERVIEW: Biomarkers can be subdivided depending on their applications. Markers of exposure and dose can be used to reconstruct and predict past accidental or occupational exposures when limited or no physical measurements were available. Markers of risk or susceptibility can help identify sensitivity individuals that are at increased risk for development of spontaneous disease and may help predict the increased risk in sensitive individuals associated with environmental or therapeutic radiation exposures. Markers of disease represent the initial cellular or molecular changes that occur during disease development. Each of these types of biomarkers serves a unique purpose. OUTLINE: This paper concentrates on biomarkers of dose and exposure and provides a brief review of biomarkers of sensitivity and disease. The review of biomarkers of dose and exposure will demonstrate the usefulness of biomarkers in evaluation of physical factors associated with radiation exposure, such as LET, doserate and dose distribution. It will also evaluate the use of biomarkers to establish relationships that exist between exposure parameters such as energy deposition, environmental concentration of radioactive materials, alpha traversals and dose. In addition, the importance of biological factors on the magnitude of the biomarker response will be reviewed. Some of the factors evaluated will be the influence of species, tissue, cell types and genetic background. The review will demonstrate that markers of sensitivity and disease often have little usefulness in dose-reconstruction and, by the same token, many markers of dose or exposure may not be applicable for prediction of sensitivity or risk.

They were compared with control groups with a similar age distribution. Cataract formation and ocular fundus lesions were scored according to a standardized protocol. Fluorescein angiography and histopathology was performed in selected animals. RESULTS: Cataract formation occurred after a latent period of 3-5 years. Significant cataract induction was observed for photon-doses of 8 and 8.5 Gy and beyond 20 years after proton irradiation. The severity of the lesions represents significant impairment of vision and would require cataract surgery if similar results occurred in human bone marrow transplant patients. Fluorescein angiography demonstrated a normal pattern of retinal vessels in 13 out of 14 animals (93%) from the irradiated group and in eight out of nine animals (89%) from the control group. No additional lesions apart from age-related degenerative changes could be demonstrated. Histological evaluation revealed no radiation-associated vasculopathy. CONCLUSIONS: Radiation alone for doses up to 8.5 Gy of photons does not carry a potential risk for fundus pathology, whereas clinically important cataract induction should be anticipated within 5 years after photon doses of 8.0 and 8.5 Gy and proton doses in excess of 2.5 Gy.

    Description: Diabetes induces pathology throughout the body via nonenzymatic glycation of proteins. Vitreous, which is replete with type 11 collagen, undergoes significant changes in diabetes. The resultant diabetic vitreopathy plays an important role in diabetic retinopathy. Detecting these molecular changes could provide insight into diabetic eye disease as well as molecular effects elsewhere in the body. Human eyes were obtained at autopsy and studied in the fresh, unfixed state. Sclera, choroid, and retina were dissected off the vitreous for dark-field slit microscopy and dynamic light scattering (DLS). For the former, the entire vitreous was exposed. For the latter, only a window at the equator was dissected in some specimens, and the anterior segment was removed leaving the posterior lens capsule intact in others. DLS was performed to determine particle sizes at multiple sites 0.5 mm apart, spanning the globe at the equator (window dissections) and along the antero-posterior axis. Dark-field slit microscopy in diabetic subjects detected findings typical of age-related vitreous degeneration, but at much younger ages than nondiabetic controls. Noninvasive DLS measurements found a greater heterogeneity and larger particle sizes in vitreous of subjects with diabetes as compared to age-matched controls. DLS can detect and quantify the early molecular effects that cause vitreous collagen fibrils to cross-link and aggregate. This could provide valuable insight into ocular and systemic effects of hyperglycemia, because the molecular changes in diabetic vitreopathy could serve as an index of such effects throughout the body. In addition to the diagnostic implications, this methodology could provide a rapid, reproducible way to monitor the response to therapy with novel agents intended to prevent the complications of diabetes on a molecular level.

    Description: Over the course of this award we have: 1) Completed and published the results of a study of the effects of hyperfine particles on reflectance spectra of olivine and quartz, which included the development of scattering codes. Research has also progressed in the analysis of the effects of fine particle sizes on clay spectra. 2) Completed the analysis of the mineralogy of dark regions, showed the insitu compositions are highly correlated to the SNC meteorites, and determined that the martian mantle was depleted in aluminum prior to 2-3 GA ago; Studies of the mineralogic heterogeneity of surficial materials on Mars have also been conducted. and 3) Performed initial work on the study of the physical and chemical processes likely to form and modify duricrust. This includes assessments of erosion rates, solubility and transport of iron in soil environments, and models of pedogenic crust formation.

    Description: The Mars Pathfinder mission illustrated the benefits of including a mobile robotic explorer on a planetary mission. However, for future Mars rover missions, significantly increased autonomy in navigation is required in order to meet demanding mission criteria. To address these requirements, we have developed new path planning and localisation capabilities that allow a rover to navigate robustly to a distant landmark. These algorithms have been implemented on the JPL Rocky 7 prototype microrover and have been tested extensively in the JPL MarsYard, as well as in natural terrain.

    Description: The paper presents two path planners suitable for planetary rovers. The first is based on fuzzy description of the terrain, and genetic algorithm to find a traversable path in a rugged terrain. The second planner uses a global optimization method with a cost function that is the path distance divided by the velocity limit obtained from the consideration of the rover static and dynamic stability. A description of both methods is provided, and the results of paths produced are given which show the effectiveness of the path planners in finding near optimal paths. The features of the methods and their suitability and application for rover path planning are compared

    Description: Future Mars exploration missions will perform two types of experiments: science instrument placement for close-up measurement, and sample acquisition for return to Earth. In this paper we describe algorithms we developed for these tasks, and demonstrate them in field experiments using a self-contained Mars Rover prototype, the Rocky 7 rover. Our algorithms perform visual servoing on an elevation map instead of image features, because the latter are subject to abrupt scale changes during the approach. 'This allows us to compensate for the poor odometry that results from motion on loose terrain. We demonstrate the successful grasp of a 5 cm long rock over 1m away using 103-degree field-of-view stereo cameras, and placement of a flexible mast on a rock outcropping over 5m away using 43 degree FOV stereo cameras.

    Description: The Lockheed Martin spillage study was a substantial effort and is worthy of a separate paper. However, since a paper was not submitted a few of the most pertinent results have been pulled out and included in this paper. The reader is urged to obtain a copy of the complete Boeing Configuration Aerodynamics final 1995 contract report for the complete Lockheed documentation of the spillage work. The supersonic cruise studies presented here focus on the bifurcated - axisymmetric inlet drag delta. In the process of analyzing this delta several test/CFD data correlation problems arose that lead to a correction of the measured drag delta from 4.6 counts to 3.1 counts. This study also lead to much better understanding of the OVERFLOW gridding and solution process, and to increased accuracy of the force and moment data. Detailed observations of the CFD results lead to the conclusion that the 3.1 count difference between the two inlet types could be reduced to approximately 2 counts, with an absolute lower bound of 1.2 counts due to friction drag and the bifurcated lip bevel.
