    Notes: Summary The nucleus intercollicularis is an important site in the control of vocalization in birds. In oscines, a subregion of the nucleus intercollicularis called the dorso-medial intercollicular nucleus appears to play a key role in this process because it receives the majority of the projections from the nucleus robustus archistriatalis and sends most of the projections to the motor nucleus of the hypoglossal nerve. In this paper, we present neurochemical studies of the nucleus intercollicularis in the Japanese quail which suggest the presence of heterogeneity within this structure. One rostral band contains high densities of cholinergic muscarinic receptors identified by quantitative autoradiography using tritiated N-methylscopolamine as the ligand. A caudal dorso-medial region is specifically labeled by estrogen receptors identified using immunocytochemistry and by α2-adrenergic receptors which were quantified by autoradiography using tritiated para-amino-clonidine. This latter sub-region is possibly equivalent to the dorso-medial intercollicular nucleus of oscines. Additional track-tracing studies should be performed to confirm this homology. The coexistence of estrogen and α2-adrenergic receptors within the same structure suggests important functional connections between steroid action and catecholaminergic systems in the brain.

    Notes: Summary Developmental changes of thyrotropin-releasing hormone (TRH)-immunoreactive structures in the brain of mallard embryos were studied by means of immunocytochemistry (PAP technique). The primary antibody was generated against synthetic TRH. Immunoreactive neurons were first detected in the hypothalamus of 14-day-old embryos. By day 20, increasing numbers of immunoreactive perikarya were observed in the paraventricular nucleus, anterior preoptic region and supraoptic region. Immunoreactive fiber projections were seen in the median eminence as early as embryonic day 20; they occurred also in some extrahypothalamic regions (lateral septum, accumbens nucleus). The number and staining intensity of the cell bodies increased up to hatching, and continued to increase during the first week after hatching.

    Notes: Summary In this study electron-microscopic immunocytochemistry was used to localize olfactory marker protein in olfactory epithelia. Rat olfactory-epithelial samples were rapidly frozen, freeze-substituted with acetone, embedded at low temperatures with Lowicryl K11M and labelled on the sections with polyclonal antibodies raised against olfactory marker protein and with protein G conjugated to colloidal gold. Apart from the aforementioned use of acetone, substitution was carried out in the complete absence of chemical fixation, i.e., neither aldehydes nor OsO4 were used. This procedure resulted in localization concurrent with a good ultrastructural preservation. Olfactory-marker protein was present throughout the cytoplasmic compartments of dendrites and dendritic endings of olfactory-receptor cells, but it was not found in organelles such as mitochondria. Olfactory-marker protein was found only in dendriticendings of olfactory-receptor cells mature enough to have given rise to cilia, but these cilia displayed less labelling than dendrites and dendritic endings. Olfactory-marker protein was not found in apices and microvilli of neighboring olfactory-supporting cells.

    Notes: Summary The subcommissural organ (SCO), classified as one of the circumventricular organs, is composed mainly of modified ependymal cells, attributable to a glial lineage. Nevertheless, in the rat, these cells do not possess glial markers such as glial fibrillary acidic protein (GFAP), protein S100, or the enzyme glutamine synthetase (GS). They receive a synaptic 5-HT input and show pharmacological properties for uptake of GABA resembling the uptake mechanism of neurons. In this study, we examine the phenotype of several mammalian SCO (cat, mouse, rabbit) and compare them with the corresponding features of the rat SCO. In all these species, the SCO ependymocytes possess vimentin as an intermediate filament, but never express GFAP or neurofilament proteins. They do not contain GS as do glial cells involved in GABA metabolism, and when they contain protein S100 (rabbit, mouse), its rate is low in comparison to classical glial or ependymal cells. Thus, these ependymocytes display characteristics that differentiate them from other types of glial cells (astrocytes, epithelial ependymocytes and tanycytes). Striking interspecies differences in the capacity of SCO-ependymocytes for uptake of GABA might be related to their innervation and suggest a species-dependent plasticity in their function.

    Notes: Summary On the basis of 1200 Golgi-impregnated brains the structure of the central complex of Drosophila melanogaster was analyzed at the cellular level. The four substructures of the central complex — the ellipsoid body, the fanshaped body, the noduli, and the protocerebral bridge — are composed of (a) columnar small-field elements linking different substructures or regions in the same substructure and (b) tangential large-field neurons forming strata perpendicular to the columns. At least some small-field neurons belong to isomorphic sets, which follow various regular projection patterns. Assuming that the blebs of a neuron are presynaptic and the spines are postsynaptic, the Golgi preparations indicate that small-field neurons projecting to the ventral bodies (accessory area) are the main output from the central complex and that its main input is through the large-field neurons. These in turn are presumed to receive input in various neuropils of the brain including the ventral bodies. Transmitters can be attributed immunocytochemically to some neuron types. For example, GABA is confined to the R1–R4 neurons of the ellipsoid body, whereas these cells are devoid of choline acetyltransferase-like immunore-activity. It is proposed that the central complex is an elaboration of the interhemispheric commissure serving the fast exchange of data between the two brain hemispheres in the control of behavioral activity.

    Notes: Summary Doubts exist as to whether afferent nerve fibers exert a neurotrophic effect on the differentiation of sensory cells in the developing vestibular neuroepithelium. To determine whether innervation of hair cells precedes their differentiation, we have used the L1 adhesion molecule as a marker for axons. The detection of L1 on afferent axons in the otic vesicle of mouse embryos on gestation day 11 shows that nerve fibers penetrate the neuroepithelium before the sensory cells differentiate. L1-immunoreactivity of nerve endings also reveals the considerable fiber ramification on gestation days 14 and 15, i.e., corresponding to the first stages of sensory cell differentiation. The expression of L1 at successive stages of nerve fiber growth in the neuroepithelium, such as fasciculation and ramification, is not consistent with the previous role proposed for L1 as a fascicule-promoting factor and raises the possibility that other mechanisms are involved in L1 mediaded adhesion.

    Notes: Summary The localisation and distribution of 10 vertebrate-derived neuropeptides in the earthworm, Lumbricus terrestris, have been determined by an indirect immunofluorescence technique. The peptides are pancreatic polypeptide (PP), peptide tyrosine tyrosine (PYY), neuropeptide Y (NPY), glucagon (C-terminal), vasoactive intestinal polypeptide (VIP), peptide histidine isoleucine (PHI), gastrinreleasing peptide (GRP), calcitonin gene-related peptide (CGRP), neurotensin (NT), and met-enkephalin. For 6 of the peptides — PYY, NPY, PHI, glucagon, GRP and CGRP — this is the first demonstration of their presence in any annelid, and NT has not previously been described in an oligochaete. Cell bodies and nerve fibres immunoreactive to the 10 peptides occur throughout the CNS. In the PNS, epidermal sensory cells displayed immunoreactivities to PP and PYY, and PP-, PYY-, NPY-, PHI- and GRP-like immunoreactivities occurred in nerve fibres supplying the main body muscles. Nerve fibres immunoreactive to PP and PYY are also associated with the innervation of the gut (pharynx, oesophageal glands, and mid and posterior regions of the intestine). No endocrine cells immunoreactive for any of the antisera tested could be identified in the gut epithelium, suggesting that dual location of peptides in the brain and gut epithelium is a phenomenon that occurred at a later stage in evolution. No immunoreactive elements were detected in any of the organs and ducts of the reproductive and excretory systems.

    Notes: Summary Specific antisera against protein-conjugated γ-aminobutyric acid (GABA) were used in immunocytochemical staining procedures to study the distribution of the putative GABA-like immunoreactive neurons in the optic lobes of Periplaneta. GABA-like immunoreactive structures are evident in all three optic neuropil regions. Six different populations of GABAergic neurons, whose perikarya are grouped around the medulla, are found within the optic lobe. The number of these immunoreactive cells varies greatly and corresponds to the number of ommatidia of the eye. In the proximal part of the lamina, a coarse network of GABA-positive fibres is recognizable. These are the processes of large field tangential cells whose fibres pass through the distal surface of the medulla. A second fibre population of the lamina is made up of the processes of the centrifugal columnar neurons whose perikarya lie proximally to the medulla. The medulla contains 9 layers with GABAergic elements of variable immunoreactivity. Layers 1, 3, 5, 7 and 9 exhibit strong labelling, as a result of partial overlapping of the processes of centrifugal and centripetal columnar neurons, tangential fibres and/or lateral processes of perpendicular fibres and (possibly) processes of amacrines. A strong immunoreactivity is found in the proximal and distal layers of the lobula.

    Notes: Summary The distribution of serotonin (5HT)-containing neurons in the central nervous system of the snail Helix pomatia has been determined in whole-mount preparations by use of immunocytochemical and in vivo 5,6-dihydroxy-tryptamine labelling. 5HT-immunoreactive neuronal somata occur in all but the buccal and pleural ganglia. Immunoreactive fibres are present throughout the central nervous system. The 5HT-immunoreactive neuronal somata characteristically appear in groups, located mainly in the cerebral, pedal, visceral and right parietal ganglia. The majority of 5HT-immunoreactive neurons is located in the pedal ganglia. Additionally a dense network of 5HT-immunoreactive varicose fibres is found in the neural sheath of the central nervous system including all the nerves and ganglia. The number and distribution of 5HT-immunoreactive neurons correlates with that demonstrated by 5,6-dihydroxytryptamine labelling method.

    Notes: Summary An immunocytochemical study by light- and electron microscopy using the antibody against rat hepatic fatty acid-binding protein (FABP) revealed the brush cells in the gastric epithelium of rats to be intensely immunoreactive. The immunoreactive cells were present in a group in the distal wall of the groove between forestomach and glandular stomach, as well as scattered singly in the surface and foveolar epithelia of the glandular stomach. Almost all immunoreactive brush cells had a thin basal process in contact with the basement membrane. No secretory granules with dense cores, similar to those of endocrine cells, were observed in the brush cells. The specific appearance of FABP-immunoreactivity in the brush cell indicates that this cell type is a distinct entity from other epithelial cells in the stomach and that FABP is a useful histochemical marker of the brush cells. FABP may be involved in the specific function(s) of this cell type related to fatty acid metabolism.

    Notes: Summary The mode of expression was investigated for two positive regulatory genes PHO2 and PHO4, whose products are indispensable for the transcriptional control of the structural genes of repressible acid phosphatase and the inorganic phosphate (Pi) transport system in Saccharomyces cerevisiae. Northern analysis of poly(A)+ RNA of the wild-type and the pho regulatory mutants with PHO4 DNA as hybridization probe and expressional analysis of a pho4′-'lacZ fused gene on a YEp plasmid revealed that PHO4 is expressed at a low level, constitutively, and independently of the PHO regulatory system and Pi in the medium. Similar analyses with PHO2 DNA indicated that PHO2 is expressed at an even lower level than PHO4, and is repressed by Pi and by the active PHO2 product, possibly at the translational level, while retaining a substantial level of basal activity.

    Notes: Summary Standard laboratory yeast strains have from four to five genes encoding the methionine initiator tRNA (IMT). Strain S288C has four IMT genes with identical coding sequences that are colinear with the RNA sequence of tRNA I Met . Each of the four IMT genes from strain S288C is located on a different chromosome. A fifth IMT gene with the same coding sequence is present in strain A364A but not in S288C. By making combinations of null alleles in strain S288C, we show that each of the four IMT genes is functional and that tRNA I Met is not limiting in yeast strains with three or more intact genes. Strains containing a single IMT2, 3 or 4 gene grow only after amplification of the remaining IMT gene. Strains with only the IMT1 gene intact are viable but grow extremely slowly; normal growth is restored by the addition of another IMT gene by transformation, providing a direct test for IMT function.

    Notes: Summary We have cloned three distinct nuclear genes, NAM1, NAM7, and NAM8, which alleviate mitochondrial intron mutations of the cytochrome b and COXI (subunit I of cytochrome oxidase) genes when present on multicopy plasmids. These nuclear genes show no sequence homology to each other and are localized on different chromosomes: NAM1 on chromosome IV, NAM7 on chromosome XIII and NAM8 on chromosome VIII. Sequence analysis of the NAM1 gene shows that it encodes a protein of 440 amino acids with a typical presequence that would target the protein to the mitochondrial matrix. Inactivation of the NAM1 gene by gene transplacement leads to a dramatic reduction of the overall synthesis of mitochondrial protein, and a complete absence of the COXI protein which is the result of a specific block in COXI pre-mRNA splicing. The possible mechanisms by which the NAM1 gene product may function are discussed.

    Notes: Summary The leaky expression of the yeast mitochondrial geneoxi1, containing a frameshift mutation (+1), is caused by natural frameshift suppression, as shown previously (Fox and Weiss-Brummer 1980). A drastic decrease in the natural level of frameshifting is found in the presence of thepar r-454 mutation, localized at the 3′ end of the 15 S rRNA gene. This mutation causes resistance to the antibiotic paronomycin in the yeast strains D273-10B and KL14-4A (Li et al. 1982; Tabak et al. 1982). The results of this study imply that in the yeast strain 777-3A this mutation alone is sufficient for restriction of the level of natural frameshifting but is insufficient to confer resistance to paromomycin. A second mutation, arising spontaneously with a frequency of 10−4 leads, in combination with thepar r-454 mutation, to full paromomycin resistance in strain 777-3A.

    Notes: Summary The RAD18 gene of Saccharomyces cerevisiae is involved in mutagenic DNA repair. We describe its isolation from a yeast library introduced into the centromeric YCp50 vector, a low copy number plasmid. The insert was sublconed into YCp50 and into the multicopy YRp7 plasmid. RAD18 is not toxic when present in multiple copies but the UV survival response indicates an heterogeneity in the cell population, a fraction of it being more sensitive. A DNA segment, close to RAD18, is toxic on the multicopy plasmid and may correspond to the tRAN sup61 known to be tightly linked to RAD18. Chromosomal deletions of RAD18 were constructed. The gene is not essential and the deleted strains have the properties of single site mutants. Thus, RAD18 appears to be essentially involved in DNA repair metabolism.

    Notes: Summary The localization of ribulose 1,5-bisphosphate carboxylase/ oxygenase (RuBisCO) in cells ofC. pyrenoidosa grown at varying light intensities was determined by immunoelectron microscopy. Log phase cells grown at photon flux densities of 25 and 75 μEm−2s−1 (light-limiting) and 540 μEm−2s−1 (light-saturating) were fixed in 3% glutaraldehyde and embedded in Lowicryl K4M. Sections were labelled with antiserum to each subunit of RuBisCO followed by protein A-gold. At each light fluence rate, the pyrenoid was heavily labelled by each antibody whereas chloroplast stromal labelling was not above background levels. The apparent absence of stromal RuBisCO at each light level, and hence the lack of enzyme redistribution from pyrenoid to stroma following an increase in light fluence rate, suggests that pyrenoid RuBisCO is functional in vivo.

    Notes: Summary The structural gene, PHO13, for the specific p-nitrophenyl phosphatase of Saccharomyces cerevisiae was cloned and its nucleotide sequence determined. The deduced PHO13 protein consists of 312 amino acids and its molecular weight is 34635. The disruption of the PHO13 gene produced no effect on cell growth, sporulation, or viability of ascospores. The PHO13 locus was mapped at 1.9 centimorgans from the HO locus on the left arm of chromosome IV. By chromosome fragmentation, the PHO13 locus was found to be located about 72 kb from the left-hand telomere of chromosome IV and distal to the HO locus.

    Notes: Summary Isolated nuclei of Saccharomyces cerevisiae were incubated with five restriction nucleases. Out of the twenty-one recognition sequences for these nucleases in the centromere region of chromosome XIV, only five are accessible to cleavage. These sites map 11 by and 74 by to the left and 27 bp, 41 by and 290 by to the right, respectively, of the boundaries of the 118 by functional CEN14 DNA sequence. The distance between the sites accessible to cleavage and closest to CEN14 is 156 bp, suggesting this is the maximal size of DNA protected in CEN14 chromatin. The DNA in CEN14 chromatin protected against cleavage with DNase I and micrococcal nuclease overlaps almost completely with this region. Hypersensitive regions flanking both sides are approximately 60 by long. Analyses of other S. cerevisiae centromeres with footprinting techniques in intact cells or nucleolytic cleavages in isolated nuclei are discussed in relation to our results. We conclude that structural data of chromatin obtained with restriction nucleases are reliable and that the structure of CEN14 chromatin is representative for S. cerevisiae centromeres.

    Notes: Summary Expression of the repressible acid phosphatase (rAPase) gene, PHO5, of Saccharomyces cerevisiae is repressed by a certain level of inorganic phosphate (Pi) in the medium and is derepressed when the Pi concentration is lowered. The Pi signals are conveyed to PHO5 by a regulatory system consisting of proteins coded for by the PHO2, PHO4, PHO80 and PHO81 genes. We have found that the transcription of PHO81 is regulated by Pi through the PHO regulatory system. Increasing the dosage of PHO4 and PHO81 by ligating each gene to YEP13 gives rise to, respectively, considerable and weak synthesis of rAPase by cultivation of the transformants in high-Pi medium; but in low-Pi medium, increased dosage of PHO4 stimulates the rAPase synthesis significantly, whereas PHO81 has no effect. Increased dosage of PHO2 stimulates rAPase synthesis considerably in low-Pi but not in high-Pi. A coordinate increase of PHO80 cancels the dosage effect of PHO4, but not that of PHO81. Coordinate increases of PHO80 and PHO2 give rise to the same phenotype as an increased dosage of PHO80 alone. The level of the PHO4 protein was found to be the limiting factor of the rAPase synthesis and the copy number of the PHO5 gene not to be. These facts accord with the idea that the PHO80 protein transmits the Pi signals to the PHO5 gene via the PHO4 protein, whereas the PHO2 protein does not have a direct function in the signal transmission.

The second gene fusion utilizes a 103 bp yeast invertase NH2-terminal coding sequence at the fusion junction of the hybrid gene described above. The gene fusions, under the control of the α-factor promoter, expressed active β-galactosidase in α haploid yeast cells. The activity could be regulated in a temperature-sensitive sir3 mutant.     Notes: Abstract Reconstitution experiments were performed on lipid bilayer membranes in the presence of purified mitochondrial porin from yeast and of detergent-solubilized mitochondrial outer membranes of a porin-free yeast mutant. The addition of the porin resulted in a strong increase of the membrane conductance, which was caused by the formation of ion-permeable channels in the membranes. Yeast porin has a single-channel conductance of 4.2 nS in 1 M KCl. In the open state it behaves as a general diffusion pore with an effective diameter of 1.7 nm and possesses properties similar to other mitochondrial porins. Surprisingly, the membrane conductance also increased in the presence of detergent extracts of the mitochondrial outer membrane of the mutant. Single-channel recordings of lipid bilayer membranes in the presence of small concentration of the mutant membranes suggested that this membrane also contained a pore. The reconstituted pores had a single-channel conductance of 2.0 nS in 1 M KCl and the characteristics of general diffusion pores with an estimated effective diameter of 1.2 nm. This means that the pores present in the mitochondrial outer membranes of the yeast mutant have a much smaller effective diameter than “normal” mitochondrial porins. Zero-current membrane potential measurements suggested that the second mitochondrial porin is slightly cation-selective, while yeast porin is slightly anion-selective in the open state but highly cation-selective in the closed state. The possible role of these pores in the metabolism of mitochondria is discussed.

    Notes: Abstract The female sex pheromone of the Asian com borer,Ostrinia furnacalis, widespread in Taiwan, was confirmed as (Z)-12-tetradecenyl acetate and its geometric isomer (E)-12-tetradecenyl acetate in a ratio of ca. 3∶1 by gas chromatography and gas chromatography-mass spectrometry in selected ion monitoring mode. Males were attracted by the mixture of these two synthetic components in the field, but the attractiveness was less than by virgin females.     Notes: Abstract Hairpencil secretion ofPseudaletia unipuncta (Haw.) contains acetic acid as well as previously identified benzaldehyde and benzyl alcohol. Age-specific titers of acetic acid were significantly greater than those of benzaldehyde and, at 25 °C, accumulation of both compounds in the hairpencils peaked on the second day after emergence. Excised antennae of males and females perceived both compounds. Antennal response to acetic acid did not vary significantly with age or sex, but male response to benzaldehyde was significantly greater than female response at all ages tested. Antennal response of both males and females to benzaldehyde tended to be greater on the fourth and eighth days after emergence than on the second.

    Notes: Abstract Calling behavior and pheromone emission by virgin femaleE. loftini moths were investigated in laboratory experiments. Calling peaked during the last three hours of the night. Three-day-old moths called more than older or younger moths and began calling earlier in the night than younger moths. Female emissions were collected in flasks without airflow and cylinders with airflow. Control tests indicated that the pheromone components (Z)-11-hexadecenyl acetate (HDA) and (Z)-13-octadecenylacetate (ODA) were 69 and 54% adsorbed on moths, respectively, and the component (Z)-13-octadecenal (ODL) was 92–99% adsorbed depending on its concentration, when put into flasks with noncalling females for 4 hr. Pheromone exposed to moths for less than 4 hr was adsorbed less. After correction for adsorption, the pheromone blend from females calling in flasks was 9:42:49% of HDA/ODL/ODA with an overall emission rate of 58 ng/female/4 hr. Three-day-old females emitted more pheromone than 0- to 2- or 4- to 5-day-old moths, in flasks. Little or no pheromone put into cylinders either downwind or upwind from a male moth was adsorbed by the moth. The pheromone blends from females calling in cylinders, corrected using downwind and upwind control test results, respectively, were 15:35:50 and 13:40:48% of HDA/ODL/ODA with overall emission rates of 32 and 35 ng/female/night.

    Notes: Abstract Ergot alkaloids produced by endophytic fungi in the tribe Balansiae (Clavicipitaceae, Ascomycetes), which infect grasses, may provide plant defense against herbivores. This study examined the effects of six ergot alkaloids on survivorship, feeding, and growth of larvae of the fall armyworm (Spodoptera frugiperda, Lepidoptera: Noctuidae), a generalist herbivore of grasses. Corn leaf disks were soaked in solutions of individual ergot alkaloids at different concentrations and presented to neonate larvae. At the highest concentrations (77–100 mg/liter) of ergonovine, ergotamine, ergocryptine, agroclavine, and elymoclavine, larval weights and/or leaf area consumed after eight days were reduced relative to controls. Lysergol had no effect on larval weights and leaf consumption at any concentration. Although active concentrations were higher than those reported from two host grasses, in vivo levels of ergot alkaloids have not been quantified for most endophyte-infected grasses. The detrimental effects on fall armyworm observed in this study suggest that ergot alkaloids could be responsible, at least in part, for the greater insect resistance of endophyte-infected grasses.

    Notes: Abstract The ability of naturally occurring levels of theFusarium spp. fungal metabolite fusaric acid to synergize the toxicity of the allelochemicals gossypol, a saponin, and 6-methoxy-2-benzoxazolinone to larvae ofHeliothis zea (Boddie) was tested. Levels of fusaric acid comparable to those found near the fungus increased mortality ofH. zea to gossypol, the saponin, and 6-methoxy-2-benzoxazolinone, and decreased the development rate of surviving larvae exposed to gossypol and 6-methoxy-2-benzoxazolinone. Some effect was also noted for levels of fusaric acid found generally distributed throughout infected plants. The chemical properties of fusaric acid suggest that it synergizes the toxicity of the allelochemicals by inhibiting oxidative enzymes responsible for detoxification. Production of the biosynthetically simple fusaric acid may be a fungal strategy for conserving resources as compared to those fungi that produce biosynthetically complex toxins of their own.

    Notes: Abstract Extracts of the intact hairpencil glands and hairs from the hairpencil glands of males ofHeliothis virescens (F.) were analyzed by capillary gas chromatography (GC) and by GC-mass spectroscopy. These analyses indicated that hexadecanyl acetate (212.4 ng/male), hexadecanol (22.3 ng/male), (Z)-11-hexadecenyl acetate (3.5 ng/male), octadecanyl acetate (14.2 ng/male) octadecanol (7.5 ng/male), tetradecanoic acid (2.7 ng/male), hexadecanoic acid (22.3 ng/male), and octadecanoic acid (6.5 ng/male) were present in the extracts. These compounds also were found in volatiles released from the hairpencil glands. In addition, GC analysis using both polar and apolar capillary columns indicated that extracts of the glands and extracts of the hairs from the hairpencil glands contained small amounts of tetradecanyl acetate, (Z)-9-tetradecenyl acetate, tetradecanol, (Z)-7-hexadecenyl acetate, (Z)-9-hexadecenyl acetate, and (Z)-11-hexadecenol. No (Z)-9-tetradecenal was present. Studies indicated that titers of the compounds increased rapidly during the 36 hr after adult eclosion and then leveled off, being maintained at high levels until released when the glandular hairs were exposed. During exposure of the hair pencils substantial amounts of the compounds were released. In vivo application of 500 ng of suspensions of (Z)-11-hexadecenyl acetate, (E)-11-hexadecenyl acetate, (Z)-11-tetradecenyl acetate, or (E)-11-tetradecenyl acetate in dimethyl sulfoxide to the surface of the denuded hairpencil gland showed that biosynthesis proceeds to the alcohol via the acetate.

In model substrate assays, MFOs fromP. polyxenes are twice as active as MFOs fromHeliothis zea, a generalist herbivore not adapted to feeding on either furanocoumarins or furanocoumarin/phytosynergist combinations.P. polyxenes MFOs are 10 and 46 times less sensitive to inhibition by myristicin and safrole, respectively, thanH. zea MFOs and eight times less sensitive to inhibition by safrole than MFOs fromPapilio troilus, a closely related species that does not encounter furanocoumarin/phytosynergist combinations in its diet. Higher MFO activity and decreased sensitivity to MFO inhibitors are important adaptations that allow black swallowtail caterpillars to feed on many umbelliferous plants.

    Notes: Abstract (Z,Z)-7,9-dodecadienyl acetate, a reported male attractant for several New World spp. ofEpinotia, was identified as the primary pheromone of femaleE. tedella (European spruce budmoth) through chemical analysis of pheromone gland washes, the electrophysiological study of male antennal receptor types, and field-trapping tests. The washes contained this compound at a level of approximately 0.4 ng per FE, along with a similar amount of the corresponding alcohol, (Z,Z)-7,9-dodecadien-1-ol. Each compound activated its own specialized type of male receptor cell. No traces of stereoisomers or monoenes were found in the washes. In field-trapping tests conducted in stands of Norway spruce in southern Germany, (Z,Z)-7,9-do-decadienyl acetate as a single chemical proved highly attractive to maleE. tedella over a range of lure doses. The corresponding alcohol or aldehyde did not show attractivity; rather, in binary combinations with the primary pheromone, these compounds, and also the (E,Z)-7,9 stereoisomeric acetate, reduced captures. A 10∶1 blend of (E)-9- and (Z)-9-dodecenyl acetates, reported as anE. tedella male attractant, did not reveal significant captures. No otherEpinotia spp. besidesE. tedella responded to the various 7,9-do-decadienyl test baits during this study.

    Notes: Abstract Pedicularis semibarbata is apparently an obligate hemiparasite of coniferous trees. It is also a facultative parasite ofLupinus fulcratus from which we find that it obtains quinolizidine alkaloids, principally α-isolupanine. As a result, a single population ofP. semibarbata contains both alkaloidrich and alkaloid-free plants. The butterflyEuphydryas editha naturally oviposits on both plant types. This butterfly population, which is the principal herbivore attackingP. semibarbata at this site, is known to contain two morphs. Individuals of a specialist morph discriminate when ovipositing among individualP. semibarbata plants and produce offspring that survive better on accepted than on rejected plants. Those of a generalist morph accept allP. semibarbata plants and produce offspring that survive equally well on plants accepted or rejected by the discriminating morph. Because of the existence of this complex variation among the butterflies, the presence of naturally laid eggs on alkaloid-containing plants still leaves the possibility that the alkaloids may defend the plants against the specialist morph. In experiments on both oviposition preference and larval performance in early instars, we failed to detect any correlation between alkaloid content of a plant and either its acceptability to or suitability for the discriminating morph of the insect. Alkaloid presence in the host-plant population, achieved through root parasitism, is currently neither subject to strong insect-mediated selection nor a major cause of selection on the insects.

    Notes: Abstract Heritability of variation in male pheromone response by pink bollworm moths,Pectinophora gossypiella (Saunders), was examined using a still-air, wing-fanning bioassay. Heritability (±SE) of overall responsiveness, as measured by the mean duration of wing fanning to the blend of pheromone components produced by females [44∶56 ratio of (Z, E)- to (Z, Z)-7,11-hexadecadienyl acetate], was 0.385 ± 0.095. Heritabilities of wingfanning duration to blends with 25 and 65%Z, E isomer were 0.377 ± 0.113 and −0.145 ± 0.103, respectively. These findings indicate an asymmetry in the genetic component of variation in response to pheromone blends with high and low proportions of theZ, E isomer. An index of response specificity for individual males was developed based on the response to an off-blend (either 25 or 65%Z, E isomer) relative to the response to the 44%Z, E blend. Heritabilities of response specificity were 0.117 ± 0.059 and −0.043 ± 0.067 for the 25 and 65%Z, E blends, respectively.

    Notes: Abstract The foliage and fruit of the tomato plantLycopersicon esculentum contains polyphenol oxidases (PPO) and peroxidases (POD) that are compartmentally separated from orthodihydroxyphenolic substrates in situ. However, when leaf tissue is damaged by insect feeding, the enzyme and phenolic substrates come in contact, resulting in the rapid oxidation of phenolics to orthoquinones. When the tomato fruitwormHeliothis zea or the beet army-wormSpodoptera exigua feed on tomato foliage, a substantial amount of the ingested chlorogenic acid is oxidized to chlorogenoquinone by PPO in the insect gut. Additionally, the digestive enzymes of the fruitworm have the potential to further activate foliar oxidase activity in the gut. Chlorogenoquinone is a highly reactive electrophilic molecule that readily binds cova-lently to nucleophilic groups of amino acids and proteins. In particular, the —SH and —NH2 groups of amino acids are susceptible to binding or alkylation. In experiments with tomato foliage, the relative growth rate of the fruitworm was negatively correlated with PPO activity. As the tomato plant matures, foliar PPO activity may increase nearly 10-fold while the growth rate of the fruitworm is severely depressed. In tomato fruit, the levels of PPO are highest in small immature fruit but are essentially negligible in mature fruit. The growth rate of larvae on fruit was also negatively correlated with PPO activity, with the fastest larval growth rate occurring when larvae fed on mature fruit. The reduction in larval growth is proposed to result from the alkylation of amino acids/protein byo-quinones, and the subsequent reduction in the nutritive quality of foliage. This alkylation reduces the digestibility of dietary protein and the bioavailability of amino acids. We believe that this mechanism of digestibility reduction may be extrapolatable to other plant-insect systems because of the ubiquitous cooccurrence of PPO and phenolic substrates among vascular plant species.

    Notes: Abstract The effect of tomatine on larval growth ofHeliothis zea andSpodoptera exigua was assessed by rearing larvae on diets with different concentrations of the chemical added. When reared from neonates, linear dose-response relationships were obtained for both species, withS. exigua being three times more sensitive to tomatine thanH. zea. Tomatine toxicity was completely alleviated inH. zea by the addition of equimolar cholesterol into the diet; however, inS. exigua some toxicity was maintained. Larvae ofS. exigua that were started on control diet were insensitive to tomatine after five days; larvae started on diet with an EC50 of tomatine and then switched to control diet after five days failed to recover from toxicosis. Larval growth ofH. zea, on the other hand, was affected both at the neonate and third-instar stage, but normal growth resumed when the larvae were transferred to control diet. Tomatine had little or no affect on food consumption, assimilation, or dietary utilization of the food by third-instar larvae ofS. exigua, except at a concentration 10 times the EC50. In contrast, the efficiency of food utilization ofH. zea larvae decreased with increasing tomatine concentrations. Assimilation of the food tended to increase, although not significantly, as tomatine levels increased. Food consumption ofH. zea larvae also increased when the tomatine concentration was greater than an EC50. The addition of equimolar cholesterol to diets with an EC50 of tomatine restored weight gain and nutritional indices values to control values. These results are related to the utility of using tomatine in host-plant resistance programs.

    Notes: Abstract Behavioral responses of males from three New York races of European corn borer,Ostrinia nubilalis (Hübner), to various suspected sexpheromone-response antagonists were investigated with a flight tunnel protocol. Males from a bivoltine (Z) race and from a univoltine (Z) race utilizing a natural pheromone blend of 98∶2 (Z)-∶(E)-11-tetradecenyl acetates and males from a bivoltine (E) race utilizing a 1∶99 (Z)-∶(E)-11-tetradecenyl acetate ratio were flown to their natural blends and to blends containing 1 % of one of the following additional compounds: Z9–12∶OAc,E9–12∶OAc,Z9–14∶OAc, andE9–14∶OAc. In each race, the added components lowered significantly the number of individuals completing the behavioral sequence. The only exception was bivoltineE males flown to a blend withE9–12∶OAc added. The number completing the sequence in this case was statistically not significantly lower than the number that completed the sequence to the standard blend. In all three races, theZ9–14∶OAc produced the most dramatic reduction in completed flights, and in the univoltineZ race, this added component was significantly more effective in reducing completed flights than any other added component. The response to the Z9–14∶OAc is understandable given recently published findings of an electrophysiological study of single sensilla in the European corn borer.

    Notes: Abstract Monarch butterfly,Danaus plexippus (L.), larvae were collected during August 1983 from the common milkweed,Asclepias syriaca L., across its extensive North American range from North Dakota, east to Vermont, and south to Virginia. This confirms that the late summer distribution of breeding monarchs in eastern North America coincides with the range of this extremely abundant milkweed resource. Plant cardenolide concentrations, assayed by spectrophotometry in 158 samples from 27 collection sites, were biased towards plants with low cardenolide, and ranged from 4 to 229 μg/ 0.1 g dry weight, with a mean of 50 μg/0.1 g. Monarch larvae reared on these plants stored cardenolides logarithmically, and produced 158 adults with a normally distributed concentration range from 0 to 792 μg/0. l g dry butterfly, with a mean of 234 μg/0.1 g. Thus butterflies increased the mean plant cardenolide concentration by 4.7. The eastern plants and their resultant butterflies had higher cardenolide concentrations than those from the west, and in some areas monarchs sequestered more cardenolide from equivalent plants. Plants growing in small patches had higher cardenolide concentrations than those in larger patches, but this did not influence butterfly concentration. However, younger plants and those at habitat edges had higher cardenolide concentrations than either older, shaded, or open habitat plants, and this did influence butterfly storage. There were no apparent topographical differences reflected in the cardenolides of plants and butterflies. Twenty-eight cardenolides were recognized by thin-layer chromatography, with 27 in plants and 21 in butterflies. Butterflies stored cardenolides within the more polar 46% of the plantR d range, these being sequestered in higher relative concentrations than they occurred in the plants. By comparison with published TLC cardenolide mobilities, spots 3, 4, 9, 16, 24 or 25, 26, and 27, may be the cardenolides syrioside, uzarin, syriobioside, syriogenin, uzarigenin, labriformidin, and labriformin, respectively. Cochromatography with cardenolide standards indicated that desglucosyrioside did not occur in the plants but did occur in 70% of the butterflies, and aspecioside was in 99% of the plants and 100% of the butterflies. The polar aspecioside was the single most concentrated and diagnostic cardenolide in both plants and butterflies. ButterflyR d values were dependent on those of the plant, and both showed remarkable uniformity over the range of areas sampled. Thus contrary to previous reports,A. syriaca has a biogeographically consistent cardenolide fingerprint pattern. The ecological implications of this for understanding the monarch's annual migration cycle are significant.

    Notes: Abstract A model system involving severalNicotiana species containing novel nicotine alkaloids was used to study heritability and expression of alkaloid production in leaf trichomes. The three species that comprise the section Repandae (N. repanda, N. stocktonii, andN. nesophila) were hybridized with eitherN. tabacum orN. sylvestris (neither of which producesN-acylnornicotine). The progeny of the hybrid with sylvestris produced theN-acylnornicotines at a level found in the Repandae parent.Nicotiana repanda was crossed toN. tabacum, and the F2 progeny produced the alkaloid at the same level as the original Repandae parent. Inheritance of the ability to acylate nornicotine in Repandae species is inherited in hybrids in a dominant manner These and other data obtained suggest that theN-acyltransferase that acylates nornicotine in Repandae species inherited in hybrids is in a dominant manner and that the regulatory sequence(s) for the gene is expressed in leaf trichomes when the gene is in a foreignNicotiana background.

    Notes: Abstract Analysis of ovipositor extracts of lab-reared (〉 120 generations) and wildHeliothis zea indicated small but significant differences in the percent composition of the four aldehyde components of the sex pheromone. (Z)-11-hexadecen-1-ol (Z11–16∶OH) was present in both populations and amounted to only 2.8% of the total pheromone. There was no significant difference in four of the six behavioral categories between lab-reared and wild males exposed to the four-component sex pheromone in the flight tunnel. Ninety and 84% males flew to the pheromone source, respectively. However, with the addition ofZ11–16∶OH to the pheromone blend, none of the wild males flew upwind to the stimulus source, whereas 36% of the lab-reared males completed the flight. It is suggested that prolonged inbreeding in closed quarters rendered these males less discriminating to qualitative differences in a pheromone blend.

    Notes: Abstract Behavior of males ofHydraecia micacea (Esper) responding to virgin females and to synthetic pheromone blends were investigated in a laboratory wind tunnel. The synthetic blend consisted of saturated 14∶Ac (68.9%),Z9–14∶Ac (3.4%),E11–14∶Ac (14.6%), andZ11–14∶Ac (13.1%). Virgin females were significantly better lures than the four-component synthetic blend for most behaviors. By deleting components individually from the four-component blend,Z9–14∶Ac,Z11–14∶Ac, and saturated 14∶Ac were found to be necessary for communication butE11–14∶Ac was found to have no effect on typical pheromone-mediated reproductive behaviors. Close-range studies suggested that chemicals of low volatility, released from moths, were important in eliciting copulation attempts. Field studies reinforced laboratory findings regarding the effectiveness of different lures and indicated thatHeliothis traps were the most effective for monitoring.

    Notes: Abstract Ovipositor washings from virgin femaleSpodoptera exempta (Walker) (Lepidoptera: Noctuidae) were analyzed by high-resolution gas chromatography (GC) linked to a male electroantennogram (EAG). GC retention times of the two major EAG responses observed were consistent with their assignment as (Z)-9-tetradecenyl acetate and (Z,E)-9,12-tetradecadienyl acetate, as previously identified. However, three other EAG responses were also noted that had GC retention times consistent with (Z)-9-tetradecenal, (Z)-9-tetradecen-1-o 1, and (Z)-11-hexadecenyl acetate. The components were present in the ratio of 100∶5∶1.5∶3.5∶4, respectively. Further analysis of the ovipositor washings by GC linked to a mass spectrometer (GC-MS) confirmed these findings and indicated the presence of a sixth component consistent with (Z)-11-tetradecenyl acetate present at 2 % of the major component. In field tests carried out in Kenya, (Z)-11-hexadecenyl acetate was the only newly identified component to enhance the catch of the original two-component mixture when presented in their natural ratio. The addition of (Z)-9-tetradecen-1-ol reduced catch, while (Z)-9-tetradecenal and (Z)-11-tetradecenyl acetate had no apparent effect.

    Notes: Abstract Greenhouse-grown plants of five tomato lines varying in their level of 2-tridecanone-mediated resistance toManduca sexta (L.) andLeptinotarsa decemlineata (Say) were found to adversely affect larvae ofCampoletis sonorensis (Cameron), a larval endoparasitoid ofHeliothis zea (Boddie), in a manner directly related to their level of resistance. The parasitoid larvae, which emerge as fifth instars from their host and construct a cocoon on the foliage of their hosts' host plant, suffered extensive mortality during cocoon spinning on highly resistant foliage. Mortality was greatest (82%) on the highly resistant plants ofLycopersicon hirsutum f.glabratum (accession PI 134417) and an F1 backcross [(L. esculentum × PI 134417) × PI 134417] selection. Mortality was intermediate (40 and 28%, respectively) on backcross selections with moderate and low levels of resistance and least (8%) on susceptibleL. esculentum. Removal of the glandular trichomes, which contain 2-tridecanone in their tips, from the foliage eliminated differences in parasitoid mortality among plant lines. Bioassays of 2-tridecanone indicated that it is acutely toxic to fifth instarC. sonorensis larvae at the quantities associated with highly resistant foliage and produces symptoms identical to those observed on resistant foliage. 2-Undecanone, a second methyl ketone present in the glandular trichomes of resistant foliage, was also toxic toC. sonorensis larvae, but significantly less so than 2-tridecanone. The results support the hypothesis that 2-tridecanone is responsible for the observed mortality ofC. sonorensis larvae during cocoon construction on resistant foliage.

Male and female western spruce budworm,Choristoneura occidentalis Freeman, moths exposed to 14F-E11-14: Al had LC50s (CI) of 12.0 (7.4–19.5) µg and 41.7 (22.7–76.2) µg, respectively. Antennectomized moths had lower mortality rates than normal moths. 14F-E11-14:Al elicited slightly lower amplitudes than 97:3 (E/Z)-11-tetradecenal (E/Z11-14:Al) in electroantennogram studies, but the lag or recovery period following excitation by 14F-E11-14:Al was much lower than forE/Z11-14:Al at the same concentrations. 14F-E11-14:Al andE11-14:Al evoked similar responses from male moths in wind-tunnel evaluations but neither compound was as effective as a virgin female in stimulating moths to fly upwind to contact the lure.

(HS) moths 1–7 days old responded positively in a Plexiglas flight tunnel to an attractant extracted with methanol from fresh whole-leaf washes of groundcherry,Physalis angulata L. Response to the groundcherry extract, as indicated by plume-tracking (i.e., upwind flight toward the odor source) and contact with the chemical dispenser did not change significantly during the first 5 hr of scotophase. Overall, ca. 50% of the responding moths also landed on the chemical dispenser; ca. 50% of the moths that landed also deposited eggs. There were no significant differences in the behavioral responses of females mating only once and those that had mated two or more times. Virgin females and male moths were significantly less responsive to the groundcherry attractant than mated females. The flight tunnel bioassay described provides an excellent system for evaluating plant allelochemics associated with host-plant selection.

    Notes: Abstract Two components, (Z)-5- and (Z)-7-tetradecenyl acetates, in the sex pheromone gland of aPlanotortrix excessana sibling species, were found to vary continuously from a ratio of 3∶97 to 71∶29 in individual female moths reared from wild larvae collected around New Zealand. Two laboratory colonies were established from populations representing extremes of these ratios. Although both the ratios and the total quantities of the components in individual female moths from both colonies were significantly different from each other, there was a small number of females that overlapped in both ratio and quantity. The variation was confirmed as intraspecific in a field cage experiment with male moths from both populations mating with tethered female moths from both populations.

    Notes: Abstract The effects of food plant on larval performance and midgut detoxification enzymes were investigated in larvae of the luna moth,Actias luna. Neonate larvae were fed leaves of black cherry, cottonwood, quaking aspen, white willow, red oak, white oak, tulip tree, paper birch, black walnut, butternut, or shagbark hickory. First instar survival, larval duration, and pupal weights were monitored as indices of food quality. Midgut enzyme preparations from fifth instars were assayed for β-glucosidase, quinone reductase, polysubstrate monooxygenase, esterase, and glutathione transferase activities. Larval survival on seven of the 11 plant species, including several recorded host plants, was extremely poor. Larvae performed well, and quite similarly, on birch, walnut, butternut, and hickory. Activities of all enzyme systems except β-glucosidase were significantly influenced by larval host plant. Of the systems assayed, quinone reductase and glutathione transferase activities were especially high. Comparisons of these values with published values for other Lepidoptera support the hypothesis that these enzyme systems are involved in conferring tolerance to juglone and related quinones occurring in members of the plant family Juglandaceae. Results suggest that host plant utilization by luna is more specialized at the individual or population level than at the species level and that biochemical detoxification systems may play a role in such specialization.

Neither species has detectable exopeptidase activity. Studies using enzyme-specific substrates and inhibitors demonstrate that the endopeptidases are serine proteinases (both trypsinlike and chymotrypsinlike) with histidine at the active site. Optimal pH for the tryptic and chymotryptic activity is 8.5 and 8.0, respectively, forT. ni. and 8.0 and 9.0, respectively, forP. rapae. The efficiency of proteolytic digestion (as measured by the rate of in vitro digestion of a standard protein by the midgut luminal fluid) is positively correlated with the larval dietary protein requirement and is significantly influenced by the ratios of tryptic to chymotryptic activity present in the gut lumen of these two species of Lepidoptera.

The active material was extracted inton-butanol and isolated by a series of HPLC separations. TLC of the active fraction and visualization of individual constituents with Kedde's reagent indicated that cardenolides are responsible for deterring oviposition. UV spectra were also characteristic of cardenolides. Bioassays of selected known cardenolides revealed a general lack of activity, except for cymarin, which was as strongly deterrent as the most prominent cardenolide isolated in pure form fromE. cheiranthoides. The results suggest that cardenolides in this plant can explain its escape from cabbage butterflies, but specific structural features of the glycosides are necessary for oviposition-deterring activity.

    Notes: Abstract Hexane and ethanol extracts of seeds from 10 plant species (including neem—Azadirachta indica A. Juss.) of the family Meliaceae were incorporated into artificial diet at various doses and fed to fall armyworm [Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae)] larvae in nochoice tests. All produced significant mortality, reduced larval growth rate, increased time to pupation, or all three, at some concentration. The two highest doses of all of the ethanol extracts caused 100% mortality before pupation, but the hexane extracts tended to be less effective.Aglaia cordata Hiern. ethanol extract was as potent as the comparable neem seed extract at virtually all levels, and its hexane extract was active at much lower concentrations than the neem extract was. The sublethal effects (slower growth and increased time to pupation) were usually detectable at lower doses of extract than mortality was.

The degree of enhancement is increased as the intertrap distance is decreased, and when the low-dose trap is upwind of the high-dose one. The patterns of alteration in trap catch suggest that moths initially attracted by one lure may divert into other traps, which may be either upwind of the original trap (“overshooting”), or downwind (“undershooting”). “Overshooting” can result in up to fivefold increases in catch and may provide a useful method for estimating the attractant zone of particular lure/trap combinations.

The ability to substitute these analogs for the major pheromone component in field tests raises the question: Do the same set of olfactory receptor neurons on the insects' antennae respond to both the major component and the effective structural analogs? To investigate this question, extracellular responses were recorded from single sensilla on the antennae of maleHeliothis zea (Boddie) (Lepidoptera: Noctuidae) to stimulation with increasing doses of (Z)-11-hexadecenal, the major pheromone component, (Z)-9-tetradecen-1-ol formate, and (Z)-1,12-heptadecadiene. Both (Z)-9-tetradecen-1-ol formate and (Z)-1,12-heptadecadiene can substitute for (Z)-11-hexadecenal in trap disruption experiments and affect the sexual behavior of males, although neither has been identified in the volatiles released by calling females. All three compounds elicited responses from the same class of pheromone-sensitive receptor neurons over a range of stimulus concentrations. At equivalent stimulus concentrations, however, (Z)-11-hexadecenal elicited about a 10-fold greater response than (Z)-1,12-heptadecadiene. (Z)-9-Tetradecen-1-ol formate elicited a response slightly higher than, but not significantly different from, the response to (Z)-1,12-heptadecadiene.

The moths use olfactory cues to locate these plants, then feed on leaves, flowers, and roots with the proboscis. To investigate the chemical basis of attraction, sticky traps were baited with roots of a PA-containing plant,Eupatorium maculatum, alkaloids ofE. maculatum, and several derivatives of these alkaloids. Volatile derivatives of the bicyclic pyrrolizidine skeleton attracted all three arctiid species. The dihydropyrrolizines, (S)-(+)-hydroxydanaidal and (R)-(−)-hydroxydanaidal, proved to be the most attractive compounds tested, accounting for over 70% of the moths captured. Different alkaloid derivatives attracted different proportions of male and femaleCisseps. Both (S)-(+)-hydroxydanaidal (52% male) and (R)-(−)-hydroxydanaidal (71% male) attracted a significantly lower percentage ofCisseps males thanE. maculatum roots (87% male).Cisseps males possess eversible scent organs (coremata) that are displayed during courtship. Analysis of corematal extracts revealed the presence of hydroxydanaidal.Cisseps moths thus resemble danaine and ithomiine butterflies, both in their attraction to PA sources and in the presence of PA derivatives in the male scent organs.

    Notes: Abstract 2,4-Dihydroxy-7-methoxy-1,4-benzoxazin-3-one (DIMBOA), the major hydroxamic acid present in corn, and its tritiated derivative, were prepared synthetically for use in the determination of the toxicokinetics of this insect deterrent in the European corn borer (ECB),Ostrinia nubilalis. In growth studies with DIMBOA (0, 0.05, 0.2, and 0.5 mg/g diet), the mean time to pupation and adult emergence were significantly lengthened by an increase in concentration. Pupal and adult weights, for both female and male, decreased with an increase in concentration. Increased larval and pupal mortality occurred at the highest concentration of DIMBOA. DIMBOA, at concentrations of 0.2 and 0.5 mg/g diet, resulted in a decrease in the number of egg masses produced per female, and at 0.5 mg/g diet, in a decrease in the number of eggs per egg mass. Larvae fed from the neonate stage on a diet containing 0.2 mg [3H]- + [1H]DIMBOA/g diet showed an increase in the content of label from fourth to fifth instar, but levels declined at pupation and emergence. A large amount of the labeled compounds was excreted by the insect in the pupal case. In dose-related studies, both uptake and excretion increased with an increase in concentration of DIMBOA (0.05, 0.2, 0.4 mg/g diet), while a body burden (concentration in the tissues/concentration in the frass) of approximately 0.25 was maintained for all concentrations. At the highest dose of DIMBOA (0.4 mg/g), the ECB increased consumption, possibly to compensate for the toxic effects of the compound. In topical application studies, elimination of the labeled compound in the frass was rapid, reaching 65% by 4 hr and 88% by 48 hr. Accumulation of label in tissues other than hemolymph was low. The results show that the ECB does possess adaptive mechanisms to deal with the effects of this host-derived compound.

    Notes: Abstract Xanthotoxin is a phototoxic allomone found in many of the host plants of the black swallowtail,Papilio polyxenes (Lepidoptera: Papilionidae). When added to the diet of final instar larvae, xanthotoxin can induce the cytochrome P450 monooxygenase (P450) activity in midgut microsomes by which it is detoxified. Induction is dose-dependent, increasing sevenfold when larvae feed on parsley treated topically with xanthotoxin at 0.5 or 1.0% fresh weight. Although xanthotoxin exerts much of its toxic effects when photoactivated by ultraviolet light, induction of P450 activity did not differ in the presence or absence of ultraviolet light. Despite a 4.7-fold induction of xanthotoxin-metabolizing P450 activity, total P450 content measured in the same microsomal samples did not increase significantly. These data indicate that multiple forms of P450 exist in the black swallowtail midgut and that they are differentially induced by xanthotoxin.

(E,Z)-8,10-dodecadienyl acetate (E,Z8,10-12:Ac) and (E,E)-8,10-dodecadienyl actetate (E,E8,10-12: Ac) produced both the highest EAG and EAD responses. Only a single antennal stimulatory peak was observed when female abdominal tip extracts were analyzed by GC-FID-EAD, which corresponded exactly with that of authenticE,Z8,10-12: Ac. Field tests confirmedE,Z8,10-12: Ac as the major pheromone component. TheE,Z isomer by itself caught equivalent numbers of male moths as did caged females. The addition of theE,E isomer to theE,Z form increased trap catch, although not significantly (P = 0.05). The addition of (Z,Z)-8,10-dodecadienyl acetate to theE,Z isomer significantly (P=0.05) reduced trap catch, while the addition of (Z,E)-8,10-dodecadienyl acetate to theE,Z form also decreased trap catch, but not significantly (P=0.05).

    Notes: Summary Diuretic factors were studied in the central nervous system of larvae of the tobacco budworm,Heliothis virescens, using [14C]urea as a sensitive indicator for water movement through isolated Malpighian tubules. The assay required Na+ and a pH of 6.0–6.2 for maximum activity. Malpighian tubules had high secretory activity in feeding larvae of the fifth instar, but the activity declined during the burrowing-digging stage that preceded pupation. Malpighian tubules from starved larvae showed a greater response to extracts of nervous tissues than did tubules from feeding larvae, and extracts showed a dose-response relationship with fluid secretion. Diuretic activity was distributed throughout all parts of the central nervous system with the brain having the most activity. Brain extracts increased fluid secretion by in vitro Malpighian tubules by more than 3-fold and doubled the rate of dye clearance from the hemolymph in vivo. Diuretic activity in nervous tissue extracts was unaffected by boiling but sensitive to proteases. Fluid secretion by in vitro tubules was increased by cAMP, dbcAMP, theophylline, octopamine and dopa. These studies provide evidence for the presence of diuretic factors in the central nervous system ofH. virescens larvae and describe a sensitive bioassay for these factors.

The LD50 of purified hypericin was 16 μg/g larval initial fresh weight in constant light (22 W/m2). Reduced irradiance resulted in decreased mortality. Sublethal applications retarded larval growth (body fresh weight) in a dose-dependent manner. Toxicity had an absolute light dependence at the dose levels used, the active wavelengths being 〉500 nm. The phototoxic effect was rapidly lost when larvae were maintained in darkness (〉8 hr) prior to irradiation. However, the potential for light-dependent mortality was retained if larvae were starved prior to irradiation.

Maximum levels of pheromone in Heliothisarmigera were extracted from 2- to 3-day-old females during their 4.5-hr scotophase. On ligation between the head and the thorax of both species, normal pheromone production was inhibited. Homogenates of brains (consisting of brains, subesophageal ganglia, corpora cardiaca, and corpora allata) from both species induced a normal level of pheromone production by ligated females. Moreover, inS. littoralis, female brain hormone induced the production of the two components (Z)-9- and (Z, E)-9,11-tetradecenyl acetate tested by gas chromatography. Partial separation of brain homogenates, using low pressure C18-silica reversed-phase columns, produced a stable and active hormone source that gave a dose-dependent response. This hormone was found in brain extracts of both male and femaleH. armigera andS. littoralis females. In addition,S. littoralis induced pheromone production byH. armigera ligated females.

    Notes: Abstract Several components of an internal kairomone were identified inside eggs of the host,Adoxophyes sp. (Lepidoptere: Tortricidae), that releases egg deposition of the egg-larval parasitoid,Ascogaster reticulatus Watanabe (Hymenoptera: Braconidae). Pupal hemolymph with the same activity as an internal host egg kairomone was used as a convenient test sample. Heat-treated pupal hemolymph was chromatographed on a Sephadex G-25 column. Each fraction was bioassayed and reacted with ninhydrin. The active fractions were ninhydrin-positive. Each fraction was placed onto an araino acid analyzer, which showed that the amino acids were most abundant in active fractions. Among 22 amino acids, alanine, arginine, glycine, histidine, isoleucine, leucine, methionine, proline, serine, tryptophan, and valine were active. The mixture of these active amino acids was as active as the egg-mass homogenate at the same ratio and concentration, suggesting that the most important component as the kairomone in a host egg is the mixture of several amino acids.

    Notes: Abstract Previous experiments demonstrated an oviposition-deterring effect of larval frass in the Egyptian cotton leaf worm,Spodoptera littoralis (Boisd.). In this study, females were shown to perceive the oviposition-deterring substance(s) with their antennae. During dark, airtight, and cold (−10 °C) storage, the deterrent was persistent for at least 395 days. On the other hand, larval frass retained its activity for only two days when applied to cotton leaves. The deterrent activity of frass was independent of larval density. Frass of larvae reared at high densities deterred oviposition as well as frass of larvae feeding separately or in small groups. For significant oviposition deterrence, the minimum amount of frass was in the range of 5–10 mg frass per cotton leaf. An acetone extract of larval frass was highly deterrent, in contrast to extracts prepared with water, ethanol, chloroform, or pentane.

    Notes: Abstract The kinetics of the release of gossyplure, measured in the laboratory at 34.5 ± 0.5 °C, from two liquid flowable formulations are presented and discussed. The data indicate that liquid flowable formulations, defined as those formulations where the active ingredient (in this case gossyplure) is attached to a particulate material, which in turn is suspended in a liquid medium, probably can be successfully used to disseminate pheromones and other behavior-modifying chemicals, although much work remains to be done in the development of these and other liquid flowable formulations.

    Notes: Abstract Cuticular lipid components were examined from fourth-instar larvae ofManduca sexta reared on artificial diet or growing plants. The plants used were potato, tobacco, and tomato grown in pots in a greenhouse. Twenty-eight components made up the bulk of the lipids, but there were significant differences in the proportions of them present in insects reared on the different diets. In the case of some insect cuticular lipid components, there was an obvious relationship with the surface components of the plant, but generally this relationship was weak. Nonetheless, the differences may have ecological relevance, as indicated by preliminary work on predation.

    Notes: Abstract Simulated large aspen tortrix (Choristoneura conflictana) herbivory of quaking aspen (Populus tremuloides) induces significant increases in concentrations of two phenol glycosides, salicortin and tremulacin, in leaves within 24 hr. Crushing of leaf tissue, as must occur when aspen leaves are eaten by chewing insects such as the large aspen tortrix, results in conversion of salicortin and tremulacin to 6-hydroxy-2-cyclohexenone (6-HCH). Salicortin, tremulacin, 6-HCH, and its degradation product, catechol, are all toxic to the large aspen tortrix when fed on an artificial diet. These damage-induced chemical changes provide a plausible mechanism for short-term resistance induced in aspen leaves by insect herbivory.

    Notes: Abstract Seven analogs of aristolochic acids (I, II, III, B, C, D, and E) were isolated from the leaves ofAristolochia debilis and characterized as the larval feeding stimulants of an Aristolochiaceae-feeding swallowtail butterfly,Atrophaneura alcinous. Aristolochic acids showed synergistic activity in combination with the water-soluble components in the leaf extract. Aristolochic acids were detected in the body tissues and specialized organs throughout all life stages ofA. alcinous. Larval osmeterial fluid contained aristolochic acids as high as 2% of the secretions, the compositions of which were similar to that found in the leaf extracts. In contrast, the eggs selectively contained aristolochic acids I and II. These two acids were present both in the egg yolk and egg coating material as well as in the collateral glands (glandulae sebaceae) of the adult females. The cannibalistic activity of the larvae against the eggs and pupae seemed to be triggered by aristolochic acids present on the egg surface and pupal cuticle, suggesting a possible adaptive mechanism in this species. Aristolocic acid I deterred feeding of tree sparrows, which suggested a defensive role against vertebrate predators.

    Notes: Abstract A still-air, wing fanning bioassay measured male pheromone response in the pink bollworm moth,Pectinophora gossypiella (Saunders). As pheromone concentration was increased, the duration of wing fanning increased while its latency decreased. Response duration was longest for the natural pheromone blend [44∶:56 (Z,E)- and (Z,Z)-7,11-hexadecadienyl acetates], although responses to 38% and 50%Z,E blends were not significantly briefer. Preexposure to pheromone had no effect on wing fanning in males retested after 24 hr. Wing fanning was highly correlated with upwind flight and other responses observed in a wind tunnel. Wing fanning duration provides a continuous measure of pheromone response and allows a large number of males to be tested in a short time; these two characteristics of the wingfanning assay are helpful for genetic analyses of male response.

    Notes: Abstract Petrol and ethanolic extracts of six asteraceous weeds were added to artificial diets to screen for growth inhibition and mortality of the variegated cutworm,Peridroma saucia (Hbn). Petrol and ethanolic extracts ofArtemisia tridentata andChamomilla suaveolens and ethanolic extracts ofChrysothamnus nauseosus andCentaurea diffusa severely inhibited larval growth at five times the natural concentrations. The twoC. suaveolens extracts and the ethanol extract ofA. tridentata were active at the natural concentration (100%) and were further examined at 20, 40, 60, and 80% of this level. Inhibition of larval growth was directly related to concentration for each of the three extracts tested. EC50s (effective concentration to inhibit growth by 50% relative to controls) for the three extracts were 36–42% of the naturally occurring level in the plants. Nutritional indices were calculated for secondinstarP. saucia feeding on the activeA. tridentata EtOH extract and the petrol extract fromC. suaveolens. Addition of the activeA. tridentata EtOH or theC. suaveolens petrol extract to the diet resulted in significant reduction in the relative growth rate of larvae, although theA. tridentata extract was much more inhibitory. Dietary utilization was significantly lower for larvae fed theA. tridentata EtOH extract.

    Notes: Abstract Field studies were conducted to determine the effect of (Z)-11-hexadecen-1-ol (Z11-16∶ OH) on capture of tobacco budworm (TBW),Heliothis virescens (F.), males when added to two-component (HV-2) and six-component (HV-6) pheromone blends dispensed from a polyvinyl chloride (PVC) substrate. Numbers of males caught in traps baited with PVC dispensers containing 0.25% Z11-16∶ OH, relative to total pheromone content, were significantly higher than numbers caught with dispensers containing noZ11-16∶ OH. Catches of TBW were also increased significantly in some tests whenZ11-16∶ OH was formulated in the bait at a 1% concentration. Catches were significantly reduced with dispensers containing 5.95%Z11-16∶ OH compared with those containing noZ11-16∶ OH. There were no differences in catch between traps baited with HV-2 vs. HV-6 or HV-2 + 0.25%Z11-16∶ OH vs. HV-6 + 0.25%Z11-16∶ OH when formulated in PVC substrate.

    Notes: Abstract Field populations ofHeliothis spp. were sampled for levels of naturally occurring larval parasitism on six tomato lines varying in levels of 2-tridecanone-mediated resistance toManduca sexta (L.) andLeptinotarsa decemlineata (Say). Second and third instars were parasitized byCampoletis sonorensis (Cameron) (Hymenoptera: Ichneumonidae) andCotesia (=Apantales)marginiventris (Cresson) (Hymenoptera: Braconidae) in 1984 through 1986 and byMicropletis croceipes (Cresson) (Hymenoptera: Braconidae) in 1986. Differences in parasitism by individual and multiple species among host plants were not demonstrated. However, levels of parasitism were low and variable among replicates. Total larval parasitism averaged across all plant lines was less than 6% in 1984 and 1986 and approximately 11% in 1985. In laboratory cage studies,C. sonorensis parasitized fewerH. zea larvae on tomato foliage with high levels of 2-tridecanone than on foliage with low levels. RearingH. zea on diet containing 2-tridecanone and 2-undecanone did not alter incidence of parasitism byC, sonorensis; nor did rearing parasitizedH. zea larvae on chemically treated host diets precondition the parasitoid to higher or lower mortality when transferred to foliage as a substrate for cocoon spinning, regardless of the foliage genotype. However, parasitoid survival during cocoon spinning on foliage varied significantly among plant lines in a manner corresponding to the level of 2-tridecanone-mediated resistance of the foliage. Parasitoid mortality was greatest on highly resistant foliage and lowest on susceptible foliage.

    Notes: Abstract Males of the checkerspot butterfly (Euphydryas chalcedona)patrol and perch near but not on the larval foodplant in search of females. Experiments with tethered butterflies show that searching males chase virgin females for longer times than they do mated females or males. The larvae leave the larval food-plant to pupate. The correspondence between the distance from the larval foodplant to pupation sites and where males search for females suggests that male mate-locating behavior has evolved to maximize the rate of encounters with newly emerged, virgin females. These conclusions are compared to a recent analysis of butterfly mating systems by Odendaal et al. (Am. Nat. 125: 673–678, 1985).

    Notes: Abstract As the ratio of (E)-8-dodecenyl acetate (E8–12∶Ac) to (Z)-8-dodecenyl acetate (Z8–12∶Ac) increased past optimal low levels in the pheromone blend, fewer males were able to fly 2.5 m upwind to the source. The tracks of males that flew in plumes of such high-(E)off-blends were slower and narrower than those of males flying to lower-(E)blends. The tracks were narrower, first of all, because as the proportion of E8–12∶Ac increased, the males steered more into the wind. More of their thrust was directed upwind and therefore their groundspeed to either side of the windline was reduced. In addition, males also reduced their airspeeds to high-(E)blends, which contributed to the decreased groundspeeds and narrower tracks. No significant changes in the frequency of counterturning were found in response to increasing proportions of E8–12∶Ac. The inability to continue upwind flight in a plume of an off ratio was indicated by in-flight arrestment in the plume. Arrestment resulted from changes in the course angles steered by the males and the airspeeds flown.

Les croisements Y. malinellus x Y. rorellus, Y. padellus x Y. rorellus, et Y. rorellus x Y. malinellus n'ont donné aucune descendance, tandis que les chenilles de Y. malinellus x Y. cagnagellus, Y. padellus x Y. cagnagellus et Y. padellus x Y. malinellus sont mortes avant le stade imaginal. Bien que des descendants féconds des deux sexes pouvaient être obtenus à partir de la plupart des croisements, leurs effectifs étaient faibles.     Notes: Abstract Interspecific crosses were made among four closely related members of the Yponomeuta padellus-complex. The results show the potential for gene exchange in the absence of pre-mating barriers. However, mating success was generally lower in the interspecific crosses than in the control crosses. No progeny were obtained from crosses between Y. malinellus Zeller x Y. rorellus (Hübner), Y. padellus (L.) x Y. rorellus and Y. rorellus x Y. malinellus, whereas larvae from Y. malinellus x Y. cagnagellus (Hübner), Y. padellus x Y. cagnagellus and Y. padellus x Y. malinellus did not reach the adult stage. Hybrids of three successful crosses with Y. cagnagellus as maternal species were examined for viability, circadian rhythm of sexual activity, sex-pheromone responses and female host-plant preferences. Developmental problems were found for the crosses cagnagellus x malinellus and cagnagellus x rorellus. Pupal weights and developmental time indicated dominance of Y. cagnagellus, while morphological characters were intermediate to the parents. The age of first calling indicates incomplete dominance of Y. malinellus and Y. padellus, while female moths showed a slight preference for the maternal host plant for calling activities. Hybrid males showed high responsiveness to the corresponding hybrid females. Responses to the parental species indicate that the inheritance of the pheromone-response specificity is governed by a dominance of Y. cagnagellus over Y. malinellus and co-dominance between Y. cagnagellus and Y. padellus.

The photoperiodic induction curve at 10°C resembled the one at 20°C, but with a shift of the critical photoperiod towards the shorter day-length.     Description / Table of Contents: Résumé L'analyse a porté sur les phéromones sexuelles de 3 Yponomeutidae, trouvés en sympatrie sur le fusain. Des expériences sur le comportement de Y. cagnagellus ont montré que la fraction acétate de tétradécyl de la phéromone synergise les fractions connues antérieurement: (E)-11- et (Z)-11-acétates de tétradécényl, lorsque les proportions sont 37/2/100. L'analyse en chromatographie gazeuse d'extraits de glandes à phéromone de Y. irrorellus a mis en évidence, dans les proportions approximatives suivantes 68/56/100/9/6/8/7, de l'acétate de tétradécyl, des (E)-11 et (Z)-11 acétates de tétradécényl, du tétradécanol, des (E)-11 et (Z)-11-tétradécénols, et de l'acétate d'hexadécyl. Un mélange de ces composés a provoqué un vol intégral contre le vent des mâles dans un tunnel à vent et a attiré les mâles dans la nature. L'analyse des extrait de glandes à phéromone de Y. plumbellus a donné de l'acétate de tétradécyl, du (E)-11-acétate de tétradécényl et du (Z)-11-acétate de tétradécényl, dans les proportions: 50/150/100. Une étude par élimination dans la nature a montré que les acétates non saturés étaient les composés fondamentaux de la phéromone. La teneur en (Z)-11-acétate de tétradécényl de la glande à phéromone de Y. plumbellus était d'environ 0.5 ng par femelle, soit approximativement le dixième de celle observée chez les deux autres espèces. Une très faible attraction croisée a été trouvée avec des phéromones totalement synthétisées. Lorsque l'acétate de (E)-11-tétradécényl a été enlevé des phéromones de Y. irrorellus et Y. plumbellus, le reliquat a attiré des nombres significatifs de Y. cagnagellus. Nous en concluons que des mélanges de trois composés ou plus, avec des proportions spécifiques des isomères (E) et (Z), sont indispensables pour une activité complète et pour maintenir un isolement reproductif précopulatoire entre les espèces.
    Notes: Abstract The sex pheromones produced by virgin females of three species of small ermine moths occurring sympatrically on the European spindle tree were analysed by gas chromatography and the synthetic compounds tested against male moths in the laboratory and field. Pheromone gland extracts of Yponomeuta cagnagellus were shown to contain tetradecyl acetate, (E)-11- and (Z)-11-tetradecenyl acetate, tetradecanol, (E)-11- and (Z-11-tetradecenol and hexadecyl acetate in 30/3/100/6/0/7/42) ratio. Wind tunnel bioassays and field tests showed that (E)-11-tetradecenyl acetate primary pheromone components, and that tetradecyl acetate synergised their attractiveness. The alcohols were unimportant in these tests. Analysis of the pheromone gland extracts from Y. irrorellus showed the above components in 68/56/100/9/6/8/17 ratio, and a mixture of these attracted male moths in laboratory and field. Omission of either unsaturated acetate gave unattractive mixtures, and the alcohols were also found to be important for attraction. Pheromone gland extracts from Y. plumbellus were shown to contain the seven components in 46/148/100/20/20/13/25 ratio. Field tests showed that the two monounsaturated acetates are primary pheromone components, and removal of the alcohols had no significant effect. The titre of (Z)-11-tetradecenyl acetate in Y. plumbellus pheromone gland extracts was approximately 0.5 ng per female, one tenth the titre in the other two species. It is concluded that mixtures of three or more pheromone components with specific E/Z ratio are essential for full attraction and contribute towards premating reproductive isolation of the three species. Other factors such as rate of pheromone emission, time of flight and height of flight may also contribute to reproductive isolation.

    Description / Table of Contents: Résumé Les interrelations entre O. nubilalis et le tachinidae P. nigrolineata ont été étudiées dans des conditions abiotiques telles que l'hôte présente un développement avec diapause. La rupture de la diapause a été obtenue par modifications des conditions abiotiques et/ou injection d'ecdystérone. Si le passage au froid a lieu 25 jours après la mue 4/5 et s'il dure 10 semaines, plus de 90% des larves non parasitées lèvent leur diapause. Le temps nécessaire pour obtenir la chrysalidation est corrélé avec l'âge à la mise au froid et surtout avec la durée du froid. Les injections d'ecdystérone, 25 jours après la mue 4/5, chez les larves non parasitées et n'ayant pas séjourné au froid, provoquent la levée de diapause dans un fort pourcentage de cas, mais seulement si les larves ont été transférées en photopériode longue; le temps de chrysalidation est fortement réduit chez les larves injectées par rapport aux témoins placés sous la même photopériode. Chez les larves d'O. nubilalis parasitées au début du 5ème stade larvaire, les larves de P. nigrolineata se développent de la même façon que chez les non diapausantes jusqu'au début du 2ème stade, puis la croissance s'arrête aux environs de 1 mg. Par modification des conditions abiotiques, la larve du parasitoïde ne reprend sa croissance qu'à la levée de diapause de l'hôte et continue ensuite son développement jusqu'à la pupaison qui intervient 25–30 jours après les modifications. Cette synchronisation étroite des développements du parasitoïde et de l'hôte peut être rompue par l'injection d'ecdystérone dans l'hôte; en effet, une faible dose induit la reprise de la croissance du parasitoïde, sans provoquer l'apolyse de l'hôte. Des doses plus élevées provoquent à la fois la reprise immédiate du développement de P. nigrolineata (la formation des pupes intervient alors 6–10 jours après) et l'apolyse de l'hôte. Le parasitoïde augmente la sensibilité de l'hôte: avec la même dose, le pourcentage d'apolyse est plus élevé chez les hôtes parasités que chez les non parasités.
    Notes: Abstract The developmental relationships between Ostrinia nubilalis Hübner (Lepidoptera, Pyralidae) and the tachinid parasitoid Pseudoperichaeta nigrolineata Walker (Diptera, Tachinidae) are described under abiotic conditions which induce development of the host with diapause. Diapause termination was accomplished by modifications of abiotic conditions and/or by ecdysterone injection. Pupation time was correlated with the age at chilling and with the chilling period. Ecdysterone injection in non-parasitized larvae, 25 days after the 4/5 moult, gave a high percentage of diapause termination only under a long day photoperiod and strongly reduced the pupation time. In diapausing larvae parasitized at the beginning of the fifth instar, P. nigrolineata larva develops in the same way as in non-diapausing ones during the first instar, but stops growth in the second, near a weight of 1 mg. When diapause was broken by modifications of obiotic conditions, the parasitoid larva resumes its growth only at host diapause termination (pupariae are obtained 25–30 days after the modifications). The close synchronization between parasitoid and host development is broken by injection of low dose of ecdysterone. Higher doses induce at once parasitoid growth (pupariae are obtained 6–10 days thereafter) and also host apolysis. The parasitoid larvae increase the host sensitivity to exogenous ecdysteroids.

    Description / Table of Contents: Résumé L'avenir des phagodissuadants en protection des cultures dépend, en particulier, de l'aptitude des insectes à devenir insensibles à ces substances. Les phagodissuadants sont classés en relatifs ou absolus, suivant que les insectes consomment les tissus traités ou se laissent mourir de faim. Cette propriété dépend beaucoup de la structure chimique, mais les paramètres biologiques n'ont pas été étudiés à fond. Des expériences ont donc été entreprises pour apprécier les effets combinés de jeûne et d'autres facteurs physiologiques dans l'apparition et l'évolution de la désensibilisation chez une espèce polyphage: Spodoptera frugiperda (Lep. Noctuidae). Des expériences préliminaires ont montré qu'un alcaloïde, l'acide aristolochique, est un puissant phagodissuadant pour S. frugiperda. Les chenilles, réparties en deux groupes expérimentaux, ont toutes été isolées et ont reçu chacune deux feuilles de Phaseolus lunatus: une témoin et une traitée avec 1000 ppm. Dans le groupe 1), les feuilles étaient remplacées dès consommation; dans le groupe 2), elles ne l'étaient pas. Un puissant effet dissuadant a été observé tout au long de l'expérience pour le groupe 1); dans le groupe 2), les chenilles ont finalement consommé les feuilles traitées. L'apparition de la désensibilisation était liée au jeûne. Quelques individus ont commencé à s'alimenter sur la rondelle traitée 3,5 heures aprés avoir totalement consommé la rondelle témoin; le temps médian du début de cette consommation a été de 5,7 h. Une fois la répulsion surmontée, les chenilles ont consommé le feuillage traité de la même façon que le feuillage témoin. Les expériences ont été recommencées avec les mêmes insectes, mais sans que les feuilles témoins aient alors été remplacées dans aucun des 2 groupes. Dans cette seconde expérience, les chenilles, qui avaient précédemment consommé des feuilles traitées, sont à nouveau dissuadées lors de choix binaires; cependant elles ont mangé plus rapidement les feuilles traitées, que ne l'ont fait les chenilles qui avaient disposé d'une source inépuisable de feuilles témoins. Elles se sont alimentées aussi plus facilement et plus généralement au cours de cette seconde expérience qu'au cours de la première. Quoi qu'il en soit, les deux groupes ont présenté finalement une désensibilisation. Un modèle mathématique général est proposé pour évaluer l'apparition et l'évolution de la désensibilisation. Les possibilités d'utilisation des phagodissuadants dans la protection des cultures sont discutées à la lumière de ces résultats.
    Notes: Abstract Fall armyworm larvae, Spodoptera frugiperda (J. G. Smith), were exposed to leaf disks treated with the feeding deterrent aristolochic acid under two types of regimes: In group 1, untreated leaf tissue was replenished as soon as it was consumed; in group 2, the control tissue was not replenished. A strong feeding deterrence was evident during the early stages of the assay in both groups, but eventually declined in group 2. When the experiment was continued through a second 24 h period, the control tissue was not replenished for either group. During this second trial, group 2 larvae, which had previously fed on treated tissue, fed more rapidly on treated tissue than did group 1 larvae. Moreover, group 2 larvae fed more readily and extensively on treated tissue than they had in the first trial. The time for the onset of behavioral desensitization was determined by subtracting the median onset of feeding on untreated tissue from the median interval between when a larva finished all of the control tissue and when it started feeding on treated tissue under no-choice conditions. Once larvae began to feed on treated tissue, their rate of consumption was equal to that of larvae feeding on control tissue. A general model for determining the onset and degree of desensitization was developed. The implications of these results for future experiments aimed at determining the underlying physiological mechanisms and the use of antifeedants in plant protection are discussed.

La réponse à la gravité a été modifiée par la courbure de la surface à escalader et du degré de privation d'aliment des chenilles. Sur une surface d'escalade de 2 cm de diamètre, l'orientation par rapport à la gravité se produit entre 20 et 30° par rapport à l'horizontale. Das run champ de centrifugation, un comportement analogue à une géotaxis négative se produit pour une accélération de 3,5 m s−2. L'analyse des tendances géotactiques des chenilles à tourner sur des plans inclinés suggère qu'elles présentent un comportement métagéotactique pour les inclinaisons de 40 à 90°. Des expériences d'immobilisation laissent penser que les antennes jouent un rôle dans la perception de la gravité. Les chenilles de stades ultérieurs ont présenté une géotaxie négative bien que la réponse à la gravité disparaisse chez les chenilles vagabondes en prénymphe. La prénymphose est aussi caractérisée par une inversion des réponses à la lumière.
    Notes: Abstract The intensity-response curve to light in a vertical plane (where gravity was a conflicting stimulus to light) and a horizontal plane (where gravity was not a conflicting stimulus) suggests that the climbing behaviour of the first instar Ectropis excursaria (Guénée) is consistent with a summation of response to gravity and a response to light. Response to gravity was affected by the curvature of the crawling surface and the level of food deprivation of the caterpillars. On a 2 cm diameter crawling surface orientation to gravity occurred at an inclination between 20° and 30° from the horizontal. In a centrifugal field, a behaviour analogous to negative geotaxis occurred at accelerations of 3.5 ms−2. Analysis of the geotactic turning tendencies of the caterpillar on flat inclined planes suggests that they exhibit meta-geotactic behaviour from inclinations of 40 to 90°. Negative geotaxis was exhibited by the caterpillars of later stadia although a response to gravity was lacking in the prepupal wandering caterpillars. The pre-pupal stage was also characterized by a reversal of the response to light.

Le cultivar sensible ‘Inbred A’ souffre plus des mines de C. partellus que ICZ1-CM, ICZ2-CM et Katumani composite B. Les mines influent significativement sur la croissance des 4 cultivars. Il y a une corrélation négative significative entre le minage des tiges et la production pour les cultivars ‘Inbred A’ et Katumani composite B, mais la relation n'est pas significative pour ICZ1-CM et ICZ2-CM. Les différences entre le minage des tiges et la récolte des 4 cultivars sont dues aux différents degrés de minage des tiges de ces cultivars par C. partellus.
    Notes: Abstract The effects of different degrees of stem tunneling caused by different levels of artificial infestation by the larvae of Chilo partellus (Swinhoe) on growth and yield of 4 maize cultivars were studied. The susceptible ‘Inbred A’ suffered much more tunneling than ICZ1-CM, ICZ2-CM and Katumani Composite B. This tunneling influenced significantly the plant growth of all the four cultivars. There was a significant negative correlation between tunneling and yield for the cultivars ‘Inbred A’ and Katumani Composite B, but such a relationship was not significant for ICZ1-CM and ICZ2-CM. Such differences in the relationships between tunneling and yield among the 4 cultivars are due to different degrees of tunneling caused by C. partellus larvae to the plants of these cultivars.

La présence de la phéromone sexuelle de l'hôte réduit de façon significative le nombre de passages entre les champs odorisés. De mème, les parasites visitent plus fréquemment le champ qui dispense l'odeur de la femelle en appel que le champ témoin placé à l'opposé. Par ailleurs, le temps passé dans le champ qui contient la phéromone sexuelle émise par les femelles vierges en appel est significativement supérieur à la durée de visite des champs témoins. Si l'on utilise comme source d'odeur des femelles vierges qui ne sont pas en appel, la réponse est inversée, les parasites sont alors repoussés par l'odeur de ces papillons et le nombre de visites est distribué de façon aléatoire entre les quatre champs. Ces résultats sont discutés dans le contexte de l'écologie du comportement de recherche chez les parasites oophages.
    Notes: Abstract This paper presents results of olfactometer experiments with the egg parasitoid Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae) and its host the corn earworm moth, Heliothis zea (Boddie) (Lepidoptera: Noctuidae). The sex pheromone of the host significantly reduced the total number of border crossings between odour fields in the olfactometer. Also, female parasitoids made significantly more visits to the calling moth odour field than to the opposite control field in the olfactometer. Further, the wasps spent significantly more time in the olfactometer field containing the sex pheromone released by calling virgin moths, than in control fields. If non-calling virgin moths were used as odour source, the response was reversed and wasps were repelled by the odour of the moths, and the numbers of visits were evenly distributed over the four flow fields. These results are discussed in the context of foraging ecology of egg parasitoids.

    Notes: Abstract The close-range orientation behaviour of male pea moths, Cydia nigricana (F.) (Lepidoptera: Tortricidae), to a substrate contaminated with synthetic sex pheromone was studied in the field. The substrate was a sheet of polypropylene ‘grass’ on a 1 m diam arena, on the upwind edge of which a pheromone trap had been placed previously for 1 h. After removal of the trap, moths continued to be attracted to the arena and to the vacated trap position. Video-recorded tracks showed that the moths behaved anemotactically whilst on the arena. When the arena was revolved so that the vacated trap position was on the downwind edge, moths continued to fly upwind on the arena, even though they were flying away from the original trap position and down a concentration gradient.

    Description / Table of Contents: Résumé Des quantités croissantes de nicotine dans l'alimentation ont prolongé la durée du développement des chenilles saines, bien que l'effet ait chuté aux concentrations les plus élevées. Le sexe de la noctuelle n'a pas eu d'effet sur la durée du développement ou l'action de la nicotine. L'influence de la nicotine sur le poids des chrysalides est inhabituel, en ce sens que les chrysalides les plus lourdes ont été obtenues aux concentrations moyennes. Des hypothèses sont proposés sur l'origine de ce phénomène. La mortalité et la durée de développement de H. annulipes ont augmenté et le poids des adultes a diminué quand la concentration de l'aliment de l'hôte en nicotine s'est élevée. L'effet différente de la nicotine sur des générations successives a pu provenir de modifications de la toxicité de la nicotine en fonction de changements dans la qualité de l'aliment. Quoi qu'il en soit, l'augmentation de la concentration de la nicotine dans l'alimentation de l'hôte a eu un effet négatif cohérent sur la valeur adaptative de H. annulipes.
    Notes: Abstract Experiments were conducted to determine the influence of nicotine (at a range of concentrations) in the food of an herbivorous host on the development, size and survival of its parasitoid. Fall armyworms, Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae) were reared on diets of 0, 0.025, 0.050 and 0.075% nicotine and exposed to parasitism by Hyposoter annulipes (Hymenoptera: Ichneumonidae). As nicotine concentration increased parasitoid mortality and development time increased and adult weight decreased. Development time, pupal weight and survival were recorded for unparasitized armyworms. Unparasitized fall armyworms showed lengthened development and higher mortality but pupal weights were greatest at intermediate nicotine concentrations.

    Description / Table of Contents: Abstract The hexanic extracts from pheronomal glands of T. jacobaeae virgin females have been studied. Gas-chromatographic and mass-spectrometric analyses of the main component (no 1) (98%) in combination with behavioral and electrophysiological bioassays allowed us to determine its structure and absolue configuration: (Z,Z)-3,6-Cis-(S,R)9,10-epoxyheneicosadiene previously described in other Arctiid moths. A minor constituent (no 2) (1.5%) was identified as (Z,Z)-3,6-Cis-(S,R)9,10-epoxyeicosadiene by comparison with synthetic compound. This structure is a new one for a lepidoptera sex pheromone. The third component (no 3) of the same extracts has the same physico-chemical data as those of a previously reported constituent of another Arctiid moth: (Z,Z,Z)-3,6,9 heneicosatriene.

Les analogies entre les spectres du composé no 2 (1,5%) et ceux de l'époxy-9S,10R éicosadiène-3Z,6Z de synthèse nous conduisent à proposer cette structure pour ce constituant minoritaire. Le troisième produit (0,5%) possède les caractéristiques physico-chimiques de l'hénéicosatriène-3Z,6Z,9Z (no 3). Les produits 1 et 3 ont déjà été identifiés comme constituants de phéromones d'Arctiidae tandis que le produit 2 est décrit ici pour la première fois dans une sécrétion phéromonale de Lépidoptère.

    Description / Table of Contents: Résumé La compétition entre les deux ichneumonides endoparasitoïdes solitaires Trieces tricarinatus Grav. et Triclistus yponomeutae Aeschl. a été examinée en élevant et disséquant des Yponomeuta vigintipunctatus Retz. attaqués le même jour par les deux parasitoïdes. Les résultats des dissections des hôtes multiattaqués ont été comparés à ceux des hôtes attaqués par un seul parasite, pour séparer la mortalité ou l'absence de parasitoïde provoquées par la compétition à celles dues à d'autres facteurs. On a constaté qu'un seul parasite était capable d'achever son développement. Dès que l'hôte était nymphosé et que les deux parasitoïdes étaient au premier stade larvaire, une des larves tuait sa compétitrice. Pour une fraction importante des larves éliminées, il a été possible de montrer qu'elles avaient été tuées par une aggression physique, pour les autres ce mode d'élimination était vraisemblable. L'espèce de parasitoïde qui survit dépend du nombre de jours entre la ponte des ichneumonides et la nymphose de l'hôte. Ce délai détermine le degré de développment des parasitoïdes et le moment réel de la compétition. Probablement, T. tricarinatus est le seul à survivre lorsque la nymphose de l'hôte a lieu avant l'éclosion des oeufs de T. yponomeutae.
    Notes: Abstract Competition between the solitary endoparasitoids Trieces tricarinatus (Gravenhorst) and Triclistus yponomeutae Aeschlimann (Hymenoptera: Ichneumonidae) results in the survival of one or the other of them. Once the host, Yponomeuta vigintipunctatus (Retzius) (Lepidoptera: Yponomeutidae) has pupated and both parasitoids have completed their development to first instar larvae, one of the parasitoid larvae kills the other. Which of the parasitoids survives depends on the number of days between parasitization and pupation. As a result of their type of development, this period determines the extent of development of the parasitoids at the moment of actual competition. Apparently, T. tricarinatus survives only if pupation of the host occurs before egg hatching of T. yponomeutae.

Une relation est suggérée entre ces observations et le régime alimentaire de cet insecte; une réduction de l'alimentation, plus importante chez les chenilles de troisième que de cinquième stade de P. brassicae, accompagne le début des altérations du comportement provoquées par le chlordiméforme. La discussion porte sur le mécanisme de contrôle sublétal du chlordiméforme, impliquant des effects immédiats et retardés.
    Notes: Abstract Larvae of the large cabbage white butterfly, Pieris brassicae L. are important pests of cruciferous crops. Third and fifth instars were treated topically with sublethal doses of the formamidine pesticide chlordimeform. A number of excitable actions were induced but they did not persist beyond 24 hours. Following the initial period, however, subsequent development was delayed and mortality increased, particularly when third instars were treated. Dosing fifth-instar larvae caused few further deaths. A link is suggested between these results and the nutritional status of the insects, and they are discussed in terms of how they might contribute towards control.

    Description / Table of Contents: Résumé Un des effets sublétaux du chlordiméforme, insecticide formamidine, est une réduction de la prise de nourriture, par suite de l'altération du comportement. Celle-ci implique une action stimulant l'activité locomotrice du quatrième stade de P. brassicae L., moins sensible aux effets sur l'alimentation. Le but de ce travail est d'examiner comment le chlordiméforme agit sur le comportement des chenilles du second stade. Des chenilles sur des feuilles trempées dans le chlordiméforme (10-6 à 10-2) sont vite excitées, leur déplacements s'accélèrent et les groupements formés sont tous instables, l'alimentation y étant généralement éphémère. L'excitation se réduit au bout de 24 heures, le groupements se reforment, et l'alimentation reprend, bien que pour les concentrations ≥10-4 M, les chenilles aient été encore dispersées entre 24 et 30 heures, au moment où elles commençaient à s'alimenter sur les résidus de chlordiméforme. La perturbation de la concentration des chenilles sur des disques témoins a montré une relation directe entre la taille du groupe et le taux de croissance larvaire. Nous en avons conclu que chez les espèces de lépidoptères qui consomment groupées, une interaction avec l'agrégation larvaire contribue significativement à l'effet du chlordiméforme sur l'alimentation. La discussion porte sur les conséquences pour la croissance et la survie larvaire, particulièrement en ce qui concerne l'installation des chenilles néonates.
    Notes: Abstract The behavioural basis for the reduction in feeding caused by the formamidine pesticide chlordimeform was investigated in second-instar larvae of the large cabbage white butterfly, Pieris brassicae L. Normally, these caterpillars feed communally. On chlordimeform-dipped leaves (10-6 M–10-2 M) they rapidly became excited, locomotion increased and only unstable aggregations formed in which any feeding was usually transient. Over 24 h excitation declined, aggregations re-formed and feeding was resumed. However, between 24 and 30 h, on leaves treated with chlordimeform solutions of 10-4 M or greater, larvae dispersed again. By altering the density of larvae on control discs a direct link was revealed between clump size and larval growth rates. It is concluded that, in species which feed gregariously, interference with larval aggregation contributes significantly to chlordimeform's effects on feeding and its involvement in a sublethal control mechanism in the field is discussed.

    Notes: Summary Polyclonal antibodies raised against ribosomal protein (r-protein) L2 of Schizosaccharomyces pombe were used to check for cross-reaktions with total r-proteins of rat liver. Using this procedure, the rat liver r-proteins, L4 and L24, were identified as being immunologically related to yeast L2. In addtional, homologies between rat liver L4 and L24 were detected. The possible implications for the regulation of r-protein synthesis are discussed.

    Notes: Summary Acentric yeast plasmids are mitotically unstable, apparently because they cannot freely diffuse after replicating and therefore are not included in the daughter nucleus. This behavior could result if plasmids remain attached to structural elements of the nucleus after replicating. Since DNA replication is believed to take place on the nuclear matrix, we tested whether there was a correlation between the mitotic stability of a given plasmid and the extent to which it was found associated with residual nuclear structures. Residual nuclei were prepared from yeast nuclei by extraction with either high salt, 2 M NaCl, or low salt, 10 mM lithium diiodosalicylate (LIS). Hybridization analysis was used to estimate the fraction of plasmid molecules remaining after nuclei were extracted. We examined the extent of matrix association of three ARSI plasmids, Trpl-RI circle (1.45 kb), YRp7 (5.7 kb) and pXBAT (45.1 kb) with mitotic loss rates ranging from 3–25%. In addition we examined the matrix binding of the endogenous 2 μm plasmid and the 2 μm-derived YEp 13 which is relatively stable in the presence of 2 μm and less stable in cir° strains. Among the ARS1 plasmids we observed a negative correlation between stability and matrix association, consistent with models in which binding to the nuclear matrix prevents passive segregation of ARS1 plasmid molecules. No such correlation was observed among the 2 μn plasmids. Among all plasmids examined there is a positive correlation between size and matrix association.

    Notes: Summary A DNA fragment containing the gene encoding orotidine 5′-phosphate decarboxylase (ODC) from the yeast, Schwanniomyces occidentalis (formerly castellii) has been isolated from a genomic library constructed in the S. cerevisiae expression vector, pYcDE8. A recombinant plasmid, p2-lA, containing a 2.47 kb insert was shown to complement the ura3-52 mutation of several strains of S. cerevisiae. This DNA insert was shown to be from Schwanniomyces occidentalis by Southern hybridization analysis. A restriction enzyme cleavage map of the insert has been derived and the ODC gene localized to a 1.1 kb region by deletion analysis. In addition, we have demonstrated that expression of ODC is not dependent on the ADHI promoter carried on pYcDE8. This is the first report of the cloning of a gene from a member of the genus Schwanniomyces.
