Beschreibung: This paper presents a review of the ionospheric scintillation monitoring and modelling by the European groups involved in COST 296. Several of these groups have organized scintillation measurement campaigns at low and high latitudes. Some characteristic results obtained from the measured data are presented. The paper also addresses the modeling activities: four models, based on phase screen techniques, with different options and application domains are detailed. Finally some new trends for research topics are given. This includes the wavelet analysis, the high latitudes analysis, the construction of scintillation maps and the mitigation techniques.

Beschreibung: An electron density profile model with free parameters is introduced. Initially the parameters are calculated on the basis of the ionospheric characteristics automatically obtained from the ionograms by Autoscala and considering the helio- geophysical conditions. The technique used to adjust the free parameters to the particular ionograms recorded is presented.

Beschreibung: New outcomes are proposed for ionospheric absorption starting from the Appleton-Hartree formula, in its complete form. The range of applicability is discussed for the approximate formulae, which are usually employed in the calculation of non-deviative absorption coefficient. These results were achieved by performing a more refined approximation that is valid under quasi-longitudinal (QL) propagation conditions. The more refined QL approximation and the usually employed non-deviative absorption are compared with that derived from a complete formulation. Their expressions, nothing complicated, can usefully be implemented in a software program running on modern computers. Moreover, the importance of considering Booker’s rule is highlighted. A radio link of ground range D = 1000 km was also simulated using ray tracing for a sample daytime ionosphere. Finally, some estimations of the integrated absorption for the radio link considered are provided for different frequencies.

Beschreibung: In this paper we show and discuss the results of gravity and magnetic surveys of Panarea Island and its archipelago. The most recent volcanic manifestation occurred in November 2002 with a shallow submarine gas eruption between the islets of Dattilo, Panarelli, Lisca Bianca, Bottaro and Lisca Nera. Currently, the activity of Panarea is monitored through a multidisciplinary study under the umbrella of the Italian Department of Civil Protection with the goal of defining the hazard of this area. With this aim, in May 2006 the first gravity and magnetic surveys of Panarea Island and its archipelago were performed. The offshore magnetic data were obtained using a marine magnetometer, a Geometrics G880, from the Istituto Idrografico dell Marina (IIM). Onshore and offshore magnetic data were integrated into an unique dataset for complete magnetic coverage of the study area. By using two micro-gravimeters (LaCoste & Romberg), gravity data were collected along tracks every 250 meters. The gravity dataset was processed using the standard method. A Bouguer reduction was applied to the free-air gravity dataset using a detailed digital elevation model of the island and the neighbouring sea after evaluation of the optimal Bouguer density to reduce the topographic effect. The result is a Bouguer anomaly map that shows lateral variations in density distribution and the relationships between the shallow volcanic/crustal features and tectonic lineaments. This evidence is also highlighted by the magnetic pattern, which suggests the importance of the youngest volcanic deposits with respect to the magnetic features of the island.

Beschreibung: The results of this paper demonstrate that the effect of collisions on the group refraction index is small, when the ordinary ray is considered. If, however, in order to improve the performance of a system for automatic interpretation of ionograms, the information contained in ordinary and extraordinary traces is combined, the effect of collisions between the electrons and neutral molecules should be taken into account for the extraordinary ray. The magnitude of these differences is generally very small and must be compared with the resolution in the virtual vertical height of the ionosonde, resolution which is typically of the order of few kilometers.

Beschreibung: The present paper proposes to discuss the ionospheric absorption, assuming a quasi-flat layered ionospheric medium, with small horizontal gradients. A recent complex eikonal model [Settimi et al., 2013b] is applied, useful to calculate the absorption due to the ionospheric D-layer, which can be approximately characterized by a linearized analytical profile of complex refractive index, covering a short range of heights between h1= 50 km and h2= 90 km. Moreover, Settimi et al. [2013c] have already compared the complex eikonal model for the D-layer with the analytical Chapman’s profile of ionospheric electron density; the corresponding absorption coefficient is more accurate than Rawer’s theory [1976] in the range of middle critical frequencies. Finally, in this paper, the simple complex eikonal equations, in quasi-longitudinal (QL) approximation, for calculating the non-deviative absorption coefficient due to the propagation across the D-layer are encoded into a so called COMPLEIK (COMPLex EIKonal) subroutine of the IONORT (IONOspheric Ray-Tracing) program [Azzarone et al., 2012]. The IONORT program, which simulates the three-dimensional (3-D) ray-tracing for high frequencies (HF) waves in the ionosphere, runs on the assimilative ISP (IRI-SIRMUP-P) discrete model over the Mediterranean area [Pezzopane et al., 2011]. As main outcome of the paper, the simple COMPLEIK algorithm is compared to the more elaborate semi-empirical ICEPAC formula [Stewart, undated], which refers to various phenomenological parameters such as the critical frequency of E-layer. COMPLEIK is reliable just like the ICEPAC, with the advantage of being implemented more directly. Indeed, the complex eikonal model depends just on some parameters of the electron density profile, which are numerically calculable, such as the maximum height.

Beschreibung: This paper describes the three-dimensional (3-D) electron density mapping of the ionosphere given as output by the assimilative IRI-SIRMUP-P (ISP) model for three different geomagnetic storms. Results of the 3-D model are shown by comparing the electron density profiles given by the model with the ones measured at two testing ionospheric stations: Roquetes (40.8 °N,0.5 °E), Spain, and San Vito (40.6°N,17.8 °E), Italy. The reference ionospheric stations from which the autoscaled foF2 and M(3000)F2 data as well as the real-time vertical electron density profiles are assimilated by the ISP model are those of El Arenosillo (37.1 °N,353.3° E), Spain, Rome (41.8 °N,12.5 °E), and Gibilmanna (37.9° N,14.0 °E), Italy. Overall, the representation of the ionosphere made by the ISP model is better than the climatological representation made by only the IRI-URSI and the IRI-CCIR models. However, there are few cases for which the assimilation of the autoscaled data from the reference stations causes either a strong underestimation or a strong overestimation of the real conditions of the ionosphere, which is in these cases better represented by only the IRI-URSI model. This ISP misrepresentation is mainly due to the fact that the reference ionospheric stations covering the region mapped by the model turn out to be few, especially for disturbed periods when the ionosphere is very variable both in time and in space and hence a larger number of stations would be required. The inclusion of new additional reference ionospheric stations could surely smooth out this concern.

Beschreibung: Ground-based dual-frequency GPS observations can be used to create images of electron density. This is well established for the Arctic ionosphere; here one of the first results is presented for the Antarctic. In this study, the GPS receivers in the Antarctic are supplemented with another GPS receiver onboard CHAMP. The aim of the study is to demonstrate the technique for investigating geophysical events, for example, an ionospheric disturbance period on 11 February 2004. The images have been validated by in-situ measurements from DMSP and CHAMP satellites, as well as Super Dual Auroral Radar Network (SuperDARN) convection patterns, which are able to confirm the location, presence, and transportation of large-scale plasma patches. This study indicates that although the convection still dominates in the high-latitude ionosphere, soft precipitation within the polar cap may play a role in the evolution of the polar patches. It also illustrates the potential for future multi-instrument studies of the Antarctic.

Beschreibung: We have developed a nanowire nanogenerator that is driven by an ultrasonic wave to produce continuous direct-current output. The nanogenerator was fabricated with vertically aligned zinc oxide nanowire arrays that were placed beneath a zigzag metal electrode with a small gap. The wave drives the electrode up and down to bend and/or vibrate the nanowires. A piezoelectric-semiconducting coupling process converts mechanical energy into electricity. The zigzag electrode acts as an array of parallel integrated metal tips that simultaneously and continuously create, collect, and output electricity from all of the nanowires. The approach presents an adaptable, mobile, and cost-effective technology for harvesting energy from the environment, and it offers a potential solution for powering nanodevices and nanosystems.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Wang, Xudong -- Song, Jinhui -- Liu, Jin -- Wang, Zhong Lin -- New York, N.Y. -- Science. 2007 Apr 6;316(5821):102-5.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉School of Materials Science and Engineering, Georgia Institute of Technology, Atlanta, GA 30332-0245, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/17412957" target="_blank"〉PubMed〈/a〉

Beschreibung: Recent advances in far-field optical nanoscopy have enabled fluorescence imaging with a spatial resolution of 20 to 50 nanometers. Multicolor super-resolution imaging, however, remains a challenging task. Here, we introduce a family of photo-switchable fluorescent probes and demonstrate multicolor stochastic optical reconstruction microscopy (STORM). Each probe consists of a photo-switchable "reporter" fluorophore that can be cycled between fluorescent and dark states, and an "activator" that facilitates photo-activation of the reporter. Combinatorial pairing of reporters and activators allows the creation of probes with many distinct colors. Iterative, color-specific activation of sparse subsets of these probes allows their localization with nanometer accuracy, enabling the construction of a super-resolution STORM image. Using this approach, we demonstrate multicolor imaging of DNA model samples and mammalian cells with 20- to 30-nanometer resolution. This technique will facilitate direct visualization of molecular interactions at the nanometer scale.〈br /〉〈br /〉〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2633025/" target="_blank"〉〈img src="https://static.pubmed.gov/portal/portal3rc.fcgi/4089621/img/3977009" border="0"〉〈/a〉   〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2633025/" target="_blank"〉This paper as free author manuscript - peer-reviewed and accepted for publication〈/a〉〈br /〉〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Bates, Mark -- Huang, Bo -- Dempsey, Graham T -- Zhuang, Xiaowei -- GM 068518/GM/NIGMS NIH HHS/ -- R01 GM068518/GM/NIGMS NIH HHS/ -- R01 GM068518-05/GM/NIGMS NIH HHS/ -- New York, N.Y. -- Science. 2007 Sep 21;317(5845):1749-53. Beschreibung: Responsive behavior, which is intrinsic to natural systems, is becoming a key requirement for advanced artificial materials and devices, presenting a substantial scientific and engineering challenge. We designed dynamic actuation systems by integrating high-aspect-ratio silicon nanocolumns, either free-standing or substrate-attached, with a hydrogel layer. The nanocolumns were put in motion by the "muscle" of the hydrogel, which swells or contracts depending on the humidity level. This actuation resulted in a fast reversible reorientation of the nanocolumns from tilted to perpendicular to the surface. By further controlling the stress field in the hydrogel, the formation of a variety of elaborate reversibly actuated micropatterns was demonstrated. The mechanics of the actuation process have been assessed. Dynamic control over the movement and orientation of surface nanofeatures at the micron and submicron scales may have exciting applications in actuators, microfluidics, or responsive materials.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Sidorenko, Alexander -- Krupenkin, Tom -- Taylor, Ashley -- Fratzl, Peter -- Aizenberg, Joanna -- New York, N.Y. -- Science. 2007 Jan 26;315(5811):487-90.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Bell Laboratories/Alcatel-Lucent, Murray Hill, NJ 07974, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/17255505" target="_blank"〉PubMed〈/a〉

Beschreibung: Most plasmalemmal proteins organize in submicrometer-sized clusters whose architecture and dynamics are still enigmatic. With syntaxin 1 as an example, we applied a combination of far-field optical nanoscopy, biochemistry, fluorescence recovery after photobleaching (FRAP) analysis, and simulations to show that clustering can be explained by self-organization based on simple physical principles. On average, the syntaxin clusters exhibit a diameter of 50 to 60 nanometers and contain 75 densely crowded syntaxins that dynamically exchange with freely diffusing molecules. Self-association depends on weak homophilic protein-protein interactions. Simulations suggest that clustering immobilizes and conformationally constrains the molecules. Moreover, a balance between self-association and crowding-induced steric repulsions is sufficient to explain both the size and dynamics of syntaxin clusters and likely of many oligomerizing membrane proteins that form supramolecular structures.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Sieber, Jochen J -- Willig, Katrin I -- Kutzner, Carsten -- Gerding-Reimers, Claas -- Harke, Benjamin -- Donnert, Gerald -- Rammner, Burkhard -- Eggeling, Christian -- Hell, Stefan W -- Grubmuller, Helmut -- Lang, Thorsten -- New York, N.Y. -- Science. 2007 Aug 24;317(5841):1072-6.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Neurobiology, Max Planck Institute for Biophysical Chemistry, Am Fassberg 11, 37077 Gottingen, Germany.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/17717182" target="_blank"〉PubMed〈/a〉

Beschreibung: Artificial biochemical circuits are likely to play as large a role in biological engineering as electrical circuits have played in the engineering of electromechanical devices. Toward that end, nucleic acids provide a designable substrate for the regulation of biochemical reactions. However, it has been difficult to incorporate signal amplification components. We introduce a design strategy that allows a specified input oligonucleotide to catalyze the release of a specified output oligonucleotide, which in turn can serve as a catalyst for other reactions. This reaction, which is driven forward by the configurational entropy of the released molecule, provides an amplifying circuit element that is simple, fast, modular, composable, and robust. We have constructed and characterized several circuits that amplify nucleic acid signals, including a feedforward cascade with quadratic kinetics and a positive feedback circuit with exponential growth kinetics.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Zhang, David Yu -- Turberfield, Andrew J -- Yurke, Bernard -- Winfree, Erik -- New York, N.Y. -- Science. 2007 Nov 16;318(5853):1121-5.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Computation and Neural Systems, California Institute of Technology, MC 136-93, 1200 East California Boulevard, Pasadena, CA91125, USA. dzhang@dna.caltech.edu〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/18006742" target="_blank"〉PubMed〈/a〉

Beschreibung: The ability to maintain interactions between soluble protein subunits in the gas phase of a mass spectrometer gives critical insight into the stoichiometry and interaction networks of protein complexes. Conversely, for membrane protein complexes in micelles, the transition into the gas phase usually leads to the disruption of interactions, particularly between cytoplasmic and membrane subunits, and a mass spectrum dominated by large aggregates of detergent molecules. We show that by applying nanoelectrospray to a micellar solution of a membrane protein complex, the heteromeric adenosine 5'-triphosphate (ATP)-binding cassette transporter BtuC2D2, we can maintain the complex intact in the gas phase of a mass spectrometer. Dissociation of either transmembrane (BtuC) or cytoplasmic (BtuD) subunits uncovers modifications to the transmembrane subunits and cooperative binding of ATP. By protecting a membrane protein complex within a n-dodecyl-beta-d-maltoside micelle, we demonstrated a powerful strategy that will enable the subunit stoichiometry and ligand-binding properties of membrane complexes to be determined directly, by precise determination of the masses of intact complexes and dissociated subunits.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Barrera, Nelson P -- Di Bartolo, Natalie -- Booth, Paula J -- Robinson, Carol V -- Biotechnology and Biological Sciences Research Council/United Kingdom -- New York, N.Y. -- Science. 2008 Jul 11;321(5886):243-6. doi: 10.1126/science.1159292. Beschreibung: Surface plasmons are electromagnetic waves that can exist at metal interfaces because of coupling between light and free electrons. Restricted to travel along the interface, these waves can be channeled, concentrated, or otherwise manipulated by surface patterning. However, because surface roughness and other inhomogeneities have so far limited surface-plasmon propagation in real plasmonic devices, simple high-throughput methods are needed to fabricate high-quality patterned metals. We combined template stripping with precisely patterned silicon substrates to obtain ultrasmooth pure metal films with grooves, bumps, pyramids, ridges, and holes. Measured surface-plasmon-propagation lengths on the resulting surfaces approach theoretical values for perfectly flat films. With the use of our method, we demonstrated structures that exhibit Raman scattering enhancements above 10(7) for sensing applications and multilayer films for optical metamaterials.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Nagpal, Prashant -- Lindquist, Nathan C -- Oh, Sang-Hyun -- Norris, David J -- New York, N.Y. -- Science. 2009 Jul 31;325(5940):594-7. doi: 10.1126/science.1174655.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Chemical Engineering and Materials Science, University of Minnesota, Minneapolis, MN 55455, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/19644116" target="_blank"〉PubMed〈/a〉

Beschreibung: Toward the realization of nanoscale device control, we report a molecular valve embedded in a membrane that can be opened by illumination with long-wavelength ultraviolet (366 nanometers) light and then resealed by visible irradiation. The valve consists of a channel protein, the mechanosensitive channel of large conductance (MscL) from Escherichia coli, modified by attachment of synthetic compounds that undergo light-induced charge separation to reversibly open and close a 3-nanometer pore. The system is compatible with a classical encapsulation system, the liposome, and external photochemical control over transport through the channel is achieved.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Kocer, Armagan -- Walko, Martin -- Meijberg, Wim -- Feringa, Ben L -- New York, N.Y. -- Science. 2005 Jul 29;309(5735):755-8.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉BiOMaDe Technology Foundation, Nijenborgh 4, 9747 AG Groningen, Netherlands.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/16051792" target="_blank"〉PubMed〈/a〉

Triangular prisms of the polymer epitaxially grow on freshly cleaved mica substrates, and their in-plane and out-of-plane growth rates can be controlled by raster scanning the coated tip across the substrate. Atomic force microscope images were concomitantly recorded, providing a set of photographic images of the process as it spans the nanometer- to micrometer-length scales as a function of environmental conditions.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Liu, Xiaogang -- Zhang, Yi -- Goswami, Dipak K -- Okasinski, John S -- Salaita, Khalid -- Sun, Peng -- Bedzyk, Michael J -- Mirkin, Chad A -- GM62109-02/GM/NIGMS NIH HHS/ -- New York, N.Y. -- Science. 2005 Mar 18;307(5716):1763-6.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Chemistry and Institute for Nanotechnology, Northwestern University, Evanston, IL 60208, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/15774755" target="_blank"〉PubMed〈/a〉

Beschreibung: The application of single molecules as templates for nanodevices is a promising direction for nanotechnology. We used a pair of suspended DNA molecules as templates for superconducting two-nanowire devices. Because the resulting wires are very thin, comparable to the DNA molecules themselves, they are susceptible to thermal fluctuations typical for one-dimensional superconductors and exhibit a nonzero resistance over a broad temperature range. We observed resistance oscillations in these two-nanowire structures that are different from the usual Little-Parks oscillations. Here, we provide a quantitative explanation for the observed quantum interference phenomenon, which takes into account strong phase gradients created in the leads by the applied magnetic field.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Hopkins, David S -- Pekker, David -- Goldbart, Paul M -- Bezryadin, Alexey -- New York, N.Y. -- Science. 2005 Jun 17;308(5729):1762-5.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Physics and Frederick Seitz Materials Research Laboratory, University of Illinois at Urbana-Champaign, Urbana, IL 61801, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/15961664" target="_blank"〉PubMed〈/a〉

Beschreibung: Lateral variations in membrane composition are postulated to play a central role in many cellular events, but it has been difficult to probe membrane composition and organization on length scales of tens to hundreds of nanometers. We present a high-resolution imaging secondary ion mass spectrometry technique to reveal the lipid distribution within a phase-separated membrane with a lateral resolution of approximately 100 nanometers. Quantitative information about the chemical composition within small lipid domains was obtained with the use of isotopic labels to identify each molecular species. Composition variations were detected within some domains.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Kraft, Mary L -- Weber, Peter K -- Longo, Marjorie L -- Hutcheon, Ian D -- Boxer, Steven G -- GM06930/GM/NIGMS NIH HHS/ -- New York, N.Y. -- Science. 2006 Sep 29;313(5795):1948-51.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Chemistry, Stanford University, Stanford, CA 94305, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/17008528" target="_blank"〉PubMed〈/a〉

Beschreibung: A promising approach for detecting biomolecules follows their binding to immobilized probe molecules on microfabricated cantilevers; binding causes surface stresses that bend the cantilever. We measured this deflection, which is on the order of tens of nanometers, by embedding a metal-oxide semiconductor field-effect transistor (MOSFET) into the base of the cantilever and recording decreases in drain current with deflections as small as 5 nanometers. The gate region of the MOSFET responds to surface stresses and thus is embedded in silicon nitride so as to avoid direct contact with the sample solution. This approach, which offers low noise, high sensitivity, and direct readout, was used to detect specific binding events with biotin and antibodies.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Shekhawat, Gajendra -- Tark, Soo-Hyun -- Dravid, Vinayak P -- New York, N.Y. -- Science. 2006 Mar 17;311(5767):1592-5. Beschreibung: We observed highly efficient generation of electron-hole pairs due to impact excitation in single-walled carbon nanotube p-n junction photodiodes. Optical excitation into the second electronic subband E22 leads to striking photocurrent steps in the device I-V(SD) characteristics that occur at voltage intervals of the band-gap energy E(GAP)/e. Spatially and spectrally resolved photocurrent combined with temperature-dependent studies suggest that these steps result from efficient generation of multiple electron-hole pairs from a single hot E22 carrier. This process is both of fundamental interest and relevant for applications in future ultra-efficient photovoltaic devices.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Gabor, Nathaniel M -- Zhong, Zhaohui -- Bosnick, Ken -- Park, Jiwoong -- McEuen, Paul L -- New York, N.Y. -- Science. 2009 Sep 11;325(5946):1367-71. doi: 10.1126/science.1176112.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Laboratory of Atomic and Solid-State Physics, Cornell University, Ithaca, NY 14853, USA. nmg32@cornell.edu〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/19745146" target="_blank"〉PubMed〈/a〉

Beschreibung: We demonstrate the ability to engineer complex shapes that twist and curve at the nanoscale from DNA. Through programmable self-assembly, strands of DNA are directed to form a custom-shaped bundle of tightly cross-linked double helices, arrayed in parallel to their helical axes. Targeted insertions and deletions of base pairs cause the DNA bundles to develop twist of either handedness or to curve. The degree of curvature could be quantitatively controlled, and a radius of curvature as tight as 6 nanometers was achieved. We also combined multiple curved elements to build several different types of intricate nanostructures, such as a wireframe beach ball or square-toothed gears.〈br /〉〈br /〉〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2737683/" target="_blank"〉〈img src="https://static.pubmed.gov/portal/portal3rc.fcgi/4089621/img/3977009" border="0"〉〈/a〉   〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2737683/" target="_blank"〉This paper as free author manuscript - peer-reviewed and accepted for publication〈/a〉〈br /〉〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Dietz, Hendrik -- Douglas, Shawn M -- Shih, William M -- 1DP2OD004641-01/OD/NIH HHS/ -- DP2 OD004641/OD/NIH HHS/ -- DP2 OD004641-01/OD/NIH HHS/ -- New York, N.Y. -- Science. 2009 Aug 7;325(5941):725-30. doi: 10.1126/science.1174251.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Cancer Biology, Dana-Farber Cancer Institute, Boston, MA 02115, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/19661424" target="_blank"〉PubMed〈/a〉

Beschreibung: Genome sequencing of large numbers of individuals promises to advance the understanding, treatment, and prevention of human diseases, among other applications. We describe a genome sequencing platform that achieves efficient imaging and low reagent consumption with combinatorial probe anchor ligation chemistry to independently assay each base from patterned nanoarrays of self-assembling DNA nanoballs. We sequenced three human genomes with this platform, generating an average of 45- to 87-fold coverage per genome and identifying 3.2 to 4.5 million sequence variants per genome. Validation of one genome data set demonstrates a sequence accuracy of about 1 false variant per 100 kilobases. The high accuracy, affordable cost of $4400 for sequencing consumables, and scalability of this platform enable complete human genome sequencing for the detection of rare variants in large-scale genetic studies.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Drmanac, Radoje -- Sparks, Andrew B -- Callow, Matthew J -- Halpern, Aaron L -- Burns, Norman L -- Kermani, Bahram G -- Carnevali, Paolo -- Nazarenko, Igor -- Nilsen, Geoffrey B -- Yeung, George -- Dahl, Fredrik -- Fernandez, Andres -- Staker, Bryan -- Pant, Krishna P -- Baccash, Jonathan -- Borcherding, Adam P -- Brownley, Anushka -- Cedeno, Ryan -- Chen, Linsu -- Chernikoff, Dan -- Cheung, Alex -- Chirita, Razvan -- Curson, Benjamin -- Ebert, Jessica C -- Hacker, Coleen R -- Hartlage, Robert -- Hauser, Brian -- Huang, Steve -- Jiang, Yuan -- Karpinchyk, Vitali -- Koenig, Mark -- Kong, Calvin -- Landers, Tom -- Le, Catherine -- Liu, Jia -- McBride, Celeste E -- Morenzoni, Matt -- Morey, Robert E -- Mutch, Karl -- Perazich, Helena -- Perry, Kimberly -- Peters, Brock A -- Peterson, Joe -- Pethiyagoda, Charit L -- Pothuraju, Kaliprasad -- Richter, Claudia -- Rosenbaum, Abraham M -- Roy, Shaunak -- Shafto, Jay -- Sharanhovich, Uladzislau -- Shannon, Karen W -- Sheppy, Conrad G -- Sun, Michel -- Thakuria, Joseph V -- Tran, Anne -- Vu, Dylan -- Zaranek, Alexander Wait -- Wu, Xiaodi -- Drmanac, Snezana -- Oliphant, Arnold R -- Banyai, William C -- Martin, Bruce -- Ballinger, Dennis G -- Church, George M -- Reid, Clifford A -- New York, N.Y. -- Science. 2010 Jan 1;327(5961):78-81. doi: 10.1126/science.1181498. Beschreibung: We present a strategy to design and construct self-assembling DNA nanostructures that define intricate curved surfaces in three-dimensional (3D) space using the DNA origami folding technique. Double-helical DNA is bent to follow the rounded contours of the target object, and potential strand crossovers are subsequently identified. Concentric rings of DNA are used to generate in-plane curvature, constrained to 2D by rationally designed geometries and crossover networks. Out-of-plane curvature is introduced by adjusting the particular position and pattern of crossovers between adjacent DNA double helices, whose conformation often deviates from the natural, B-form twist density. A series of DNA nanostructures with high curvature--such as 2D arrangements of concentric rings and 3D spherical shells, ellipsoidal shells, and a nanoflask--were assembled.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Han, Dongran -- Pal, Suchetan -- Nangreave, Jeanette -- Deng, Zhengtao -- Liu, Yan -- Yan, Hao -- New York, N.Y. -- Science. 2011 Apr 15;332(6027):342-6. doi: 10.1126/science.1202998.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉The Biodesign Institute, Arizona State University, Tempe, AZ 85287, USA. dongran.han@asu.edu〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/21493857" target="_blank"〉PubMed〈/a〉

Beschreibung: DNA self-assembly has produced diverse synthetic three-dimensional polyhedra. These structures typically have a molecular weight no greater than 5 megadaltons. We report a simple, general strategy for one-step self-assembly of wireframe DNA polyhedra that are more massive than most previous structures. A stiff three-arm-junction DNA origami tile motif with precisely controlled angles and arm lengths was used for hierarchical assembly of polyhedra. We experimentally constructed a tetrahedron (20 megadaltons), a triangular prism (30 megadaltons), a cube (40 megadaltons), a pentagonal prism (50 megadaltons), and a hexagonal prism (60 megadaltons) with edge widths of 100 nanometers. The structures were visualized by means of transmission electron microscopy and three-dimensional DNA-PAINT super-resolution fluorescent microscopy of single molecules in solution.〈br /〉〈br /〉〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC4153385/" target="_blank"〉〈img src="https://static.pubmed.gov/portal/portal3rc.fcgi/4089621/img/3977009" border="0"〉〈/a〉   〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC4153385/" target="_blank"〉This paper as free author manuscript - peer-reviewed and accepted for publication〈/a〉〈br /〉〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Iinuma, Ryosuke -- Ke, Yonggang -- Jungmann, Ralf -- Schlichthaerle, Thomas -- Woehrstein, Johannes B -- Yin, Peng -- 1DP2OD007292/OD/NIH HHS/ -- 1R01EB018659/EB/NIBIB NIH HHS/ -- 5R21HD072481/HD/NICHD NIH HHS/ -- DP2 OD007292/OD/NIH HHS/ -- R01 EB018659/EB/NIBIB NIH HHS/ -- R21 HD072481/HD/NICHD NIH HHS/ -- New York, N.Y. -- Science. 2014 Apr 4;344(6179):65-9. doi: 10.1126/science.1250944.
