    Notes: Summary A genetic framework was developed for the interpretation of statistical parameters estimated from a diallel experiment among a fixed set of lines. These included average direct genetic, average maternal genetic, general combining ability, reciprocal, and line and specific direct and maternal heterotic effects. The genetic model is based on direct and maternal additive and dominance genetic effects as would be expected in animal species. The model assumes that dominance is the underlying basis of heterosis. As an example, litter size at birth was analyzed from a 5 × 5 diallel cross with mice.

La procedura è stata applicata per esaminare i MCG di 16 pazienti affetti da pregresso infarto del miocardio.     Notes: Summary A possible clinical application of a high-resolution magnetic isofield contour mapping technique in cardiography is discussed. Preliminary results are presented in applying this technique to patients with recent myocardial infarction. The patients could be divided into two classes; one of them contains patients with electrical late potentials as a subclass.

    Description / Table of Contents: Riassunto Finora la magnetoencefalografia spontanea (MEG) in soggetti normali e patologici non è stata investigata a fondo. Si conoscono pochissimi contributi in questo campo. A parte il lavoro del gruppo di biomagnetismo di Roma, il solo contributo clinico finora è stato quello di Hughes e collaboratori che era abbastanza limitato nel numero e nella scelta dei soggetti studiati. La nostra esperienza condotta fin dal 1980 su un gran numero di casi (principalmente epilessie focali) mostra che il MEG può evidenziare attività significative anche se non sono presenti anormalità EEG; esso ha inoltre una risoluzione spaziale maggiore rispetto all’EEG. I foci epilettici mostrano un’area limitata di attività massima. Finora non è stata osservata nessuna evidenza sperimentale di una differente distribuzione delle ampiezze del campo sullo scalpo. La risoluzione spaziale del MEG riguarda anche la possibilità di risolvere la morfologia dei segnali EEG (per esempio onde aguzze EEG risolte in gruppi di spikes dal MEG) o di separare topograficamente le anormalità EEG mescolate e non ben localizzate (per esempio, attività delta ed epilettiche mescolate nell’EEG e discriminate nel MEG). I migliori risultati si osservano quando la localizzazione è nella corteccia esterna. Dopo una descrizione del materiale studiato, si sottolinea l’importanza dei risultati ottenuti per la prima volta in casi di epilessie focali.
    Notes: Summary So far spontaneous magnetoencephalography (MEG) in normal and pathological subjects has not been deeply investigated. Very few contributions in this field are available. Aside from the work of the Rome biomagnetic group in this field, the only clinical contribution up to now has been that of Hughes and co-workers which was rather limited in the number and in the selection of subjects investigated. Our experience, carried out since 1980 on a large number of cases—mainly focal epilepsies— shows that the MEG may evidence significant activities even if EEG abnormalities are not present and has a higher spatial resolution with respect to the EEG. Epileptic foci show a limited area of maximal activity. So far no experimental evidence of a different distribution of field amplitude over the scalp has been observed. The spatial resolution of the MEG also regards the possibility of resolving the morphology of EEG signals (for instance, EEG sharp waves resolved into groups of spikes by MEG) or topographically separate EEG abnormalities mixed up and not well localized (for instance, delta and epileptic activities mixed up in the EEG, but differently in the MEG). The best results are observed when the localization is in the outer cortex. After a description of the material studied, the importance of the results obtained for the first time in cases of focal epilepsies is outlined.

    Description / Table of Contents: Riassunto Durante l’ultimo anno gli autori hanno condotto una ricerca sistematica su un gran numero di soggetti patologici registrando simultaneamente l’EEG e il MEG. La presente versione del nostro apparato sperimentale permette di effettuare misure MEG con un solo sensore magnetico, impedendo la rilevazione simultanea di attività MEG su punti differenti della testa. Ciò ha condotto gli autori a concentrarsi sulle epilessie focali. Fin dalle prime misure il sensore magnetico ha dimostrato un notevole potere localizzante, rafforzando l’interesse per la localizzazione in casi patologici. In questo lavoro si riportano risultati che sembrano confermare la straordìnaria capacità delle misure MEG nel localizzare sorgenti di attività cerebrali patologiche in casi di epilessie focali.
    Notes: Summary During the last year the authors have been carrying out a systematic investigation on a large number of pathological subjects simultaneously recording the EEG and the MEG. The present version of our experimental apparatus allows MEG measurements with only one magnetic detector, thus preventing the detection of simultaneous MEG activities over different points of the head. This has led the authors to concentrate upon focal epilepsies. Since the first measurements a remarkable localizing power was shown by the magnetic detector, thus strengthening the interest in pathological localization. In this contribution we report results which seem to confirm the extraordinary capability of MEG measurements to localize sources of pathological brain activities in cases of focal epilepsies.

Si è trovato che la componente antero-posterioreX vettore magnetico del cuore è la piú facile da scoprire. Si suggeriscono conduttori ideali per le altre due componenti. Comunque questi hanno una sensibilità inferiore. Le differenze rispetto ai risultati di precedenti ricercatori sono state spiegate con le differenze tra i modelli.
    Notes: Summary The ideality and sensitivity of several vector magnetocardiographic leads were tested by using an inhomogeneous, physical torso model. The postero-anterior component of the heart’s magnetic vector was found easiest to detect. New, more ideal leads for the other two components are suggested. However, these have lower sensitivity. The differences from the results of former investigators can be explained by the differences of the models.

    Description / Table of Contents: Riassunto Si è studiato lo spettro del rumore di un gradiometro superconduttore a derivata secon da in un agglomerato urbano. I dati sono stati ottenuti dal gradiometro con tre accelerometri ortogonali e un magnetometro a flussometro elettronico triassiale attaccato al dewar. Questi dati sono stati analizzati usando tecniche di elaborazione dei segnali, soprattutto un cancellatore adattativo di rumori, per ridurre i rumori nei dati del gradiometro. I risultati mostrano qui che il rumore dell’esterno e/o il rumore del movimento del dewar possono essere ridotti di 40 dB in potenza di rumore. Si è anche mostrato che, nel caso di basso rapporto segnale-rumore (S/N∼1), può essere estratta un’onda sunusoidale di 10 Hz. Questa tecnica non solo promette riduzione dei rumori, ma ha anche lo scopo d’identificare i segnali di rumore che potrebbero essere veramente considerati parte del responso evocato. L’analisi dei dati che contengono i responsi evocati è ora in corso.
    Notes: Summary The noise spectrum of a superconducting second-derivative gradiometer has been investigated in an urban environment. Data have been acquired from the gradiometer with three orthogonal accelerometers and a triaxial fluxgate magnetometer attached to the dewar. These data have been analyzed by using signal processing techniques, primarily an adaptive noise canceller, to reduce noise in the gradiometer data. Results shown here indicate that the environmental noise and/or the dewar motion noise can be reduced as much as 40 dB in noise power. It is also shown, in the case of poor signal-to-noise ratio (S/N∼1), that a 10 Hz sine wave can be extracted. This technique not only shows promise for noise reduction, but also aids in the identification of noise signals which might be misconstrued as part of the evoked response. Analysis of data containing evoked response is now underway.

    Notes: Summary The instrumentation for biomagnetic applications is described. The small size and weight of the SQUID gradiometer permit easy transport and positioning of the instrument, thus making biomagnetic measurements possible in any noise-free location. The equipment is provided with a battery power supply, and, when filled with 60 cm3 of liquid helium, will operate continuously for 6 h. The device is suitable for magnetocardiography.

    Notes: Summary Magnetic-susceptibility plethysmography (MSPG) is a new method proposed for monitoring cardiac-volume changes. The measurement principle and recent work by other investigators are reviewed. The effect of the cardiac-related motion of the anterior chest wall on the MSPG signal is studied. The results suggest that the motion of the chest-wall-air boundary may contribute significantly (∼75%) to the MSPG signal when recorded near the heart.

The influence of measured signal source parameters, of an outside disturbing source and of the own noise of the magnetometer on the gradiometer base length selection is analysed.

These ramps, ranging up to 6.7 pT in size and arising from the atria, may complicate magnetic detection of His-Purkinje activity.

Fino ad oggi l’interesse degli studiosi si è concentrato su due differenti fenomeni cardiaci: 1) lo studio dell’attività del sistema di conduzione atrioventricolare e 2) lo studio della depolarizzazione ventricolare anormalmente ritardata (late potentials). L’interesse clinico per lo studio non invasivo del sistema di His-Purkinje (HPS) è ben noto. Registrazioni cliniche non invasive di late potentials sono particolarmente desiderabili in quanto possono essere preziosi indicatori di rischio di morte improvvisa. In questo lavoro si presenta una rassegna sullo stato delle ricerche in entrambi i campi d’indagine.
    Notes: Summary The fundamental motivation for investigating cardiac electrophysiological phenomena by means of the biomagnetic method has been that this technique should have added spatial resolution and provided complementary information as opposed to electric recordings only. So far the interest of experimentalists has been concentrated on two different kinds of cardiac phenomena: 1) the study of the activity of the cardiac conduction system and 2) of abnormally delayed ventricular depolarization (late potentials). The clinical interest for the noninvasive investigation of the His-Purkinje system (HPS) is well known. Noninvasive clinical recording of late potentials is particularly desirable as they seem to be precious markers for risk of sudden death. A review of the state of research in both fields is presented in this paper.

Allo scopo di risolvere il problema di quanta parte dei segnali misurati sia attribuibile al sistema di His-Purkinje e quanta piuttosto alla ripolarizzazione atriale, è stata recentemente sviluppata una strumentazione appositamente disegnata per misure sul HPS e con una sensibilità di $$20fT/\sqrt {Hz}$$ nella banda di frequenze interessanti ((0.5÷300) Hz). Al contempo si è altresí sviluppato un nuovo modello teorico in stretta corrispondenza con le caratteristiche anatomiche del HPS che fornisce una distribuzione di campo magnetico sul torace in buon accordo qualitativo con i risultati sperimentali.
    Notes: Summary Since 1980, systematic investigations of the activity of the His-Purkinje system (HPS) by the biomagnetic method have been reported on both normal subjects and patients affected by different kinds of conduction disturbances. A first attempt to interpret the experimental results by means of a theoretical model satisfactorily accounted for the measured «ramp» patterns. Moreover, a good temporal coincidence was observed between magnetic signals and invasively recorded His bundle electrograms (HBE). An alternative explanation of the «ramp» pattern, in terms of atrial repolarization, has been recently proposed on the basis of animal experimental investigation. In order to try to solve this problem, we have carried out a twofold approach: a new second-order gradiometer, specifically designed for HPS activity investigation has been successfully set up and tested; it provides a sensitivity of $$20fT/\sqrt {Hz}$$ in the band width of interest ((0.5÷300) Hz). Second, a new model based on a closer correspondence to anatomical features of the HPS has been developed. This model provides well-defined patterns which are in quite good agreement with experimental results.

    Notes: Summary In this paper we present some tentative model considerations, based on the concept of an equivalent current dipole to account for observed auditory evoked responses, with the aim of attempting a comparison of variations in amplitude and latency with calculations based on Rall’s greatly simplified nerve model. A phenomenological analysis of experimental data on the late, auditory evoked magnetic field from the human auditory cortex is presented. The emphasis is here placed on relations between signal properties (e.g. amplitude and latency) and between signal properties and stimulus properties (e.g. amplitude/latency and relative frequency glide Δf/f for a frequency glide stimulus) rather than on questions related to the location of the equivalent current dipole,e.g. the tonotopic organization discussed by Elberlinget al. and by Romaniet al.. Some results of a numerical calculation based on a linear-cable model are presented and compared with experimental results.

Il sensore magnetico era un gradiometro SQUID a forma di otto con una linea di base di 4 cm orientato in modo da essere massimamente sensibile a un dipolo di corrente orientato perpendicolarmente alla fissure di Sylvio. Gli stimoli erano impulsi sonori di 1 kHz, lunghi 100 ms con un intervallo modale tra stimoli di 700 ms emessi a livelli di pressione sonora di 40, 60, 80 e 100 dB. Si è trovato che l’ampiezza AEF è correlata all’intensità dello stimolo in modo quadratico, l’ampiezza AEP in modo, lineare. Gli AEF erano di ampiezza maggiore in risposta alla stimolazione contralaterale che in risposta alla stimolazione ipsilaterale. Gli AEP non presentano questa relazione. In un secondo esperimento gli AEF e AEP dell’emisfero destro in risposta al tono di stimolazione nell’orecchio controlaterale in questi 12 soggetti sono stati combinati con dati simili precedenti su 24 soggetti, ottenendo cosí un totale di 36 soggetti, per esaminare la comparabilità della forma dell’onadaP50 di AEP e dell’analogaP50 di AEF. Si è trovato che la latenza dellaP50 decrese in funzione dell’intensità crescente dello stimolo sia per AEF che per AEP, e che la latenza diP50 è decisamente inferiore nelle registrazioni magnetiche rispetto alle registrazioni del potenziale.
    Notes: Summary Auditory evoked magnetic fields (AEFs) and EEG auditory evoked potentials (AEPs) were recorded from left and right auditory cortical regions of 12 normal adult subjects. The magnetic sensor was a figure-eight SQUID gradiometer with a 4 cm baseline oriented so as to be maximally sensitive to a current dipole oriented normal to the Sylvian fissure. Stimuli were 100 ms long 1 kHz tone pips with a modal interstimulus interval of 700 ms delivered at sound pressure levels of 40, 60, 80 and 100 dB. AEF amplitude was found to be related to stimulus intensity in a quadratic fashion, AEP amplitude in a linear fashion. AEFs were of larger amplitude in response to contralateral as compared to ipsilateral stimulation. AEPs did not exhibit such a relationship. In a second experiment right-hemisphere AEFs and AEPs in response to contralateral ear tone stimulation in these 12 subjects were combined with similar previous data from 24 subjects, providing a total of 36 subjects, to examine the comparability of the AEPP50 wave form and the AEFP50 analog. The latency of theP50 was found to decrease as a function of increasing stimulus intensity for both AEFs and AEPs, and theP50 latency was consistently shorter in magnetic compared to potential recordings.

Ambedue usavano gradiometri di second’ordine disegnati specificatamente per gli studi sul cervello: 2.9 cm di diametro, 5 cm di baseline. Uno SQUID r.f. e uno SQUID d.c. erano accoppiati ai gradiometri. I livelli di rumore misurati sono stati rispettivamente di $$40fT/\sqrt {Hz}$$ e $$25fT/\sqrt {Hz}$$ . In alcuni soggetti sono state effettuate mappe di campi magnetici generati dall’attività spontanea del cervello. Le mappe dell’ampiezza media dei segnali del ritmo α hanno mostrato 2 massimi dell’attività su 2 regioni simmetriche dello scalpo. Comunque, malgrado una generale simmetria fra gli emisferi, la complessità delle mappe misurate suggerisce una corrispondente complessità dei generatori responsabili dell’attività α.
    Notes: Summary An investigation of the spontaneous brain activity from normal subjects has been carried out by means of the neuromagnetic method. Two different systems were used for magnetic measurements. Both used 2nd-order gradiometers specifically designed for brain studies: 2.9 cm diameter, 5 cm baseline. A r.f. SQUID and a d.c. SQUID were coupled to the gradiometers. The measured noise levels are $$40fT/\sqrt {Hz}$$ and $$25fT/\sqrt {Hz}$$ , respectively. Thorough magnetic mapping of the spontaneous activity was performed on a few subjects. By mapping the average amplitude of the α-rhythm signals two maxima of activity over two symmetric regions of the scalp were clearly observed. Despite a general symmetry between hemispheres, however, the complexity of the measured maps suggests a corresponding complexity of the generators responsible for α-activity.

    Notes: Summary This study was undertaken to examine the effects of hydrocortisone injected into male mice on the phenotypic expression and inheritance of the Fused (Fu) gene in their offspring. Data were obtained indicating that there is a hydrocortisone-susceptible period during spermatogenesis. Hydrocortisone injections of males during this period resulted in a statistically significant decrease in the proportion of phenotypically Fu offspring. Genetic analysis with the use of the closely linked recessive marker tufted (tf) demonstrated that the deficit of phenotypically Fu individuals among offspring is not caused by the differential death of gametes, zygotes or embryos. According to genetic data, this deficit is due to a decrease in the penetrance of the Fu gene and partly to its inherited inactivation. The possible mechanisms of the observed phenomenon are discussed.

    Description: When injected into mice, the synthetic double-stranded polynucleotide poly(inosinic) X poly(cytidylic) acid induces high natural killer (NK) cell activity within 4 to 12 hours. Induction of NK activity in mice immunized 2 or 3 days previously, or the addition of NK cells to cultures immunized in vitro 2 or 3 days previously, promotes early termination of the ongoing primary immunoglobulin M antibody response. A target for NK cells is a population of accessory cells that has interacted with antigen and is necessary for sustaining the antibody response.     Description: The human parasite Trypanosoma brucei gambiense grew continuously at 37 degrees C in primary cultures of murine bone marrow. Cultured parasites remained virulent for mice. Rapid parasite growth coincided with the appearance of adherent adipocyte-epitheloid cell aggregates that also promoted hematopoiesis.     Description: Cholesterol-loaded macrophages secrete cholesterol and apolipoprotein E. The current studies show that this secretion occurs by two independent pathways. In the absence of serum, the cells secrete apolipoprotein E, but not cholesterol. In the presence of monensin (an inhibitor of protein secretion), the cells secrete cholesterol, but little apolipoprotein E. After secretion, apolipoprotein E and cholesterol associate with high-density lipoprotein to form a particle that can deliver cholesterol to the liver by receptor-mediated endocytosis.     Description: A diffusible factor produced and secreted by malignant murine cells was capable of inducing plasminogen activator production by normal diploid human fibroblasts. The factor's ability to induce plasminogen activator was insensitive to treatment with nucleases, but its activity was destroyed by digestion with proteases.     Description: Adenosine receptors were made visible on light microscopy by autoradiography with tritiated cyclohexyladenosine. In the cerebellum, adenosine receptors were absent in Weaver mice, which lack granule cells, and were displaced in Reeler mice, which have displacements of granule cells. Thus, adenosine receptors appear to be located on the axon terminals of excitatory granule cells in the cerebellum. Removal of one eye of a rat depleted adenosine receptors in the contralateral superior colliculus, suggesting that the receptors occur on axon terminals of excitatory projections from retinal ganglion cells.     Description: An important question concerning the mechanism of somatic mutation of immunoglobulin variable (V) genes is whether it involves all of the numerous V genes in a differentiated B cell, independent of location, or if it is restricted to a particular chromosomal site. Comparison of the sequence of two alleles of a given V gene shows that the mutations are limited to the rearranged V gene, while the same V gene on the other chromosome has not undergone mutation. This indicates that a V gene sequence alone is not sufficient for somatic mutation to take place.     Description: Isolation of epidermal lamellar bodies has presented a challenge because pressures required to homogenize keratinocytes can destroy these organelles and because the lamellar body readily releases its contents during prolonged isolation procedures. In an attempt to isolate lamellar bodies, sheets of intact stratum corneum and stratum granulosum were obtained from neonatal mice with highly purified staphylococcal epidermolytic toxin, disrupted, and passed through a series of filters. The final filtrate was rich in intact lamellar bodies and contained variable amounts of ribosomes and other vesicular structures. Availability of a highly purified lamellar body preparation from postnatal epidermis should help to clarify the role of this organelle in epidermal function.     Description: Mice infected with reovirus type 1 develop an autoimmune polyendocrine disease. Spleen cells from these mice were fused with myeloma cells and the culture fluids were screened by indirect immunofluorescence for autoantibodies reactive with normal mouse tissues. A large panel of cloned, stable antibody-producing hybridomas has been obtained. Fourteen of the hybridomas make autoantibodies that react with cells in the islets of Langerhans, 24 with cells in the anterior pituitary, 11 with cells in gastric mucosa, and 5 with nuclei. Except for the antibodies to nuclei, the monoclonal autoantibodies are organ-specific. Some, however, show broad cross-species reactivity, recognizing similar antigenic determinants in mouse, rat, pig, and human organs, whereas other recognize determinants only in rodent tissues.     Description: A simple and efficient method of covalently coupling the strong chelator diethylenetriaminepentaacetic acid to proteins was developed for radiolabeling immunoglobulin G antibodies. After being coupled and labeled with indium-111, a monoclonal antibody to carcinoembryonic antigen retained its ability to bind to its antigen in vitro and in vivo.     Description: The distribution of keratin intermediate filaments, previously considered static in organization and imperturbable by conventional drugs used to alter the structure and organization of the cytoskeleton, can be altered significantly by treatment with colchicine and cytochalasin D. The loss of microfilaments and microtubules converts the keratin cytoskeleton from a branching, even distribution to a series of starlike structures whose filaments are maintained by multiple membrane attachment sites.     Description: The metastasis of B16 melanoma cells differed significantly in obese (ob/ob) and lean (+/?) female mice of strain C57BL/6J. When the mice were inoculated subcutaneously with melanoma cells at 10 to 11 months of age, the primary tumor grew more slowly in obese than in lean littermates and the frequency of lung metastasis was greatly reduced. When the mice were injected with the cells at 4 to 7 months, the primary tumor grew at the same rate in obese and lean mice, but the obese mice again showed a significantly reduced frequency of lung metastasis. That this effect was related to an enhanced immunocompetence in obese mice was supported by the finding that splenic lymphocytes of ob/ob mice showed three times the proliferative response to the T-cell mitogen concanavalin A compared with the proliferative response of lean control mice.     Description: Hybridoma technology has made it possible to introduce into continuous culture normal antibody-forming cells and to obtain large amounts of the immunoglobulin produced by each of these cells. Examination of the structure of a number of monoclonal antibodies that react with a single antigen has provided new information on the structural basis of the specificity and affinity of antibodies. Comparisons of families of monoclonal antibodies derived from a single germ line gene revealed the importance of somatic mutation in generating antibody diversity. Monoclonal antibodies that react with variable regions of other monoclonals allow the further dissection and modulation of the immune response.     Description: Arachidonic acid plays a central role in a biological control system where such oxygenated derivatives as prostaglandins, thromboxanes, and leukotrienes are mediators. The leukotrienes are formed by transformation of arachidonic acid into an unstable epoxide intermediate, leukotriene A4, which can be converted enzymatically by hydration to leukotriene B4, and by addition of glutathione to leukotriene C4. This last compound is metabolized to leukotrienes D4 and E4 by successive elimination of a gamma-glutamyl residue and glycine. Slow-reacting substance of anaphylaxis consists of leukotrienes C4, D4, and E4. The cysteinyl-containing leukotrienes are potent bronchoconstrictors, increase vascular permeability in postcapillary venules, and stimulate mucus secretion. Leukotriene B4 causes adhesion and chemotactic movement of leukocytes and stimulates aggregation, enzyme release, and generation of superoxide in neutrophils. Leukotrienes C4, D4, and E4, which are released from the lung tissue of asthmatic subjects exposed to specific allergens, seem to play a pathophysiological role in immediate hypersensitivity reactions.     Description: The structure of the messenger RNA (mRNA) encoding the precursor to mouse submaxillary epidermal growth factor (EGF) was determined from the sequence of a set of overlapping complementary DNA's (cDNA). The mRNA is unexpectedly large, about 4750 nucleotide bases, and predicts the sequence of preproEGF, a protein of 1217 amino acids (133,000 molecular weight). The EGF moiety (53 amino acids) is flanked by polypeptide segments of 976 and 188 amino acids at its amino and carboyxl termini, respectively. If the "sling" is cut at precallosal stages, the would-be callosal fibers whirl into paired neuromas adjacent to the longitudinal cerebral fissure.     Description: Two plasmids containing nonoverlapping deletions of the herpes simplex virus thymidine kinase gene were introduced into thymidine kinase-deficient mouse L cells by DNA-mediated gene transfer. Thymidine kinase-producing transformants were generated by a mixture of the two plasmids at a frequency significantly greater than that generated by either plasmid alone.     Description: Variants of the Dearing strain of reovirus type 3 with antigenically altered hemagglutinin proteins are much less neurovirulent than the parental virus. When injected intracerebrally into mice these variants infected a subset of the brain neurons that were infected by the parental virus. When injected intraperitoneally, the variants did not spread to the brain.     Description: Acetylcholine receptors at innervated neuromuscular junctions are very stable, with half-lives reported to be 6 to 13 days. Their turnover is described as a first-order process, implying a single population of receptors. In this study, two subpopulations of acetylcholine receptors at normally innervated junctions have been identified. One has a rapid turnover rate with a half-life of 18.7 hours, similar to that of extrajunctional receptors, and the other has a slow turnover rate with a half-life of 12.4 days. The rapidly turned over subpopulation represents approximately 20 percent of the total junctional receptors. This finding may account for the discrepancies in previous reports of turnover rates and may explain the rapid reversibility in vivo of agents that "irreversibly" block acetylcholine receptors. This finding also implies that the synthesis rate of junctional acetylcholine receptors may be higher than previous estimates.     Description: Interleukin 2, a lymphokine that acts as a second signal of cellular immune response by way of its action as a T-cell growth factor, was morphologically identified by immunoperoxidase staining. With the use of a monoclonal antibody to interleukin 2 and several complex-forming antisera, the lymphokine was readily distinguished in cytocentrifuge preparations of peripheral blood leukocytes stimulated with a T-cell mitogen. When preparations of cloned interleukin 2 producer and responder cells were stained by the same procedures, discrete patterns of both responder and producer cell phenotypes were revealed. Interleukin 2 producer T cells exhibited a characteristic intense, ringlike cytoplasmic staining, whereas the responder cells (as exemplified by interleukin 2-dependent cell lines) exhibited a less intensive, spotlike membrane staining.     Description: Macrophages isolated from tumor-bearing patients as well as cultured human monocytes express Fc receptors that cross-react strongly with murine immunoglobulins of the G2a but only slightly or not at all with the G1, G2b, or G3 subclasses. Such macrophages in the presence of murine immunoglobulin G2a monoclonal antibodies to tumors mediated the killing of tumor cells in vitro.     Description: Extracts of liver from hemizygous affected mice with the X-linked spfash mutation have 5 to 10 percent of normal ornithine transcarbamylase (OTC) activity, yet the homogeneous enzyme isolated from these extracts is identical to that in controls. The OTC messenger RNA from mutant livers programs the synthesis of two distinct OTC precursor polypeptides--one normal in size, the other distinctly elongated. Both precursors are imported and proteolytically processed by mitochondria, but only the normal one is assembled into active trimer.     Description: Mice infected at birth with replication competent Friend, Moloney, CasBr-M, C2S-M, and 1504-A murine leukemia viruses developed abnormalities of the vibrissae consisting of erratic curvature, shortening, and loss. A number of other virus strains, as well as endogenous AKR-type ecotropic virus and AKR-type, mink cell focus-inducing (MCF) viruses, did not produce these abnormalities. In mice with erythroid and myeloid leukemia, the perivibrissal sinus is the site of extramedullary hematopoiesis, but this did not appear to be the basis of the deformities.     Description: The cloned complementary DNA pMCT-1, which contains an intracisternal A particle long-terminal repeat, is more highly expressed in a mouse colon tumor than in the normal mouse colon.     Description: An unusual isozyme of lactate dehydrogenase, lactate dehydrogenase k, is found in high concentrations in cultured cells transformed by the Kirsten murine sarcoma virus and in many human cancer tissues. In experiments described here high levels of a lactate dehydrogenase k activity were detected in extracts of normal rodent retina. This activity had the same key properties as the human tumor isozyme, namely, a highly cathodic electrophoretic mobility and inhibition of enzymatic activity by oxygen and 5',5'-dipurinenucleoside tetraphosphates.     Description: A model for studying the growth of primary tumors of human endometrium and its regulation by 17 beta-estradiol has been developed in which ovariectomized nude mice are used as recipients. The receptors for sex steroids are maintained during serial transplantation of the tumor in this system. Although the rate of growth of receptor-negative endometrial tumors transplanted into ovariectomized nude mice is unaffected by the sustained presence or absence of estradiol, the growth of receptor-positive tumors is significantly increased by estradiol. Receptor-positive tumors treated with estradiol produced elevated concentrations of progesterone receptor. That the progesterone receptor is functional in this tumor is evident from the induction of estradiol 17 beta-dehydrogenase activity upon progestin administration.     Description: Neonatal mice were reared in an acoustic environment that repetitively entrained activity in a large proportion of primary auditory afferents during the period when the frequency tuning of auditory neurons normally develops. The tuning curves obtained from these mice were significantly broader than those of normally reared mice of the same age.     Description: Tumor ascites fluids from guinea pigs, hamsters, and mice contain activity that rapidly increases microvascular permeability. Similar activity is also secreted by these tumor cells and a variety of other tumor cell lines in vitro.     Description: Aplysiatoxin and debromoaplysiatoxin, which are isolated from the seaweed, Lyngbya gracilis, differ in their chemical structure only by the presence or absence of a bromine residue in the hydrophilic region. The function and the structure-activity relation of the hydrophilic region are not known. Aplysiatoxin increased malignant transformation, stimulated DNA synthesis, and inhibited the binding of phorbol-12,13-dibutyrate and epidermal growth factor to cell receptors. Debromoaplysiatoxin inhibited the binding of these two substances as strongly as aplysiatoxin but did not increase malignant transformation or stimulate DNA synthesis.     Description: Thrombin, the central regulatory enzyme in coagulation, when incubated in nanomolar concentrations with murine neuroblastoma cells produced a rapid and marked increase in tritiated guanosine 3',5'-monophosphate (cyclic GMP) formation that was blocked by hirudin and competitively antagonized by dansylarginine N-(3-ethyl-1,5-pentanediyl)amide. Diisopropylphosphofluoridate-inactivated thrombin as well as the serine protease trypsin were markedly less potent and less effective than alpha-thrombin in producing this effect. Thrombin-stimulated cyclic GMP formation was inhibited by mepacrine and nordihydroguaiaretic acid but unaffected by indomethacin, suggesting that lipoxygenase metabolites of arachidonic acid are involved in the response.     Description: A genomic clone consisting of the Moloney leukemia proviral genome with moderately repetitive mouse sequences was microinjected into the pronucleus of a mouse zygote. An animal was derived that carried multiple copies of proviral DNA in a tandem array. No evidence for homologous recombination was obtained. The viral genome was expressed in this animal and was transmitted as a single unit to its offspring.     Description: A method is described for obtaining antibody-producing hybridomas that are preferentially retained in cultures of fused mouse spleen and myeloma cells. Hybridomas are produced by fusing mouse myeloma cells that are deficient in adenosine phosphoribosyltransferase (APRT) with mouse spleen cells containing Robertsonian 8.12 translocation chromosomes.     Description: The involvement of plasma fibronectin in phagocytosis of bacteria was investigated by testing the binding of fibronectin to several species of bacteria and by evaluating the ability of fibronectin to promote binding and endocytosis of two species of these bacteria by phagocytic cells.     Description: Oral administration of ethanol, n-butanol, or t-butanol to mice 20 minutes before injection of carbon-14-labeled nitrosonornicotine inhibited the localization of radioactivity in bronchial and salivary duct epithelium and in the liver. Localization of radioactivity in the nasal epithelium and esophagus was not significantly reduced.     Description: Murine macrophage and macrophage-depleted splenocyte cultures were incubated under ambient oxygen with a commercially available perfluorocarbon blood substitute. The perfluorocarbon preparation was found to be selectively cytotoxic to macrophages. This finding may be significant in view of the preliminary therapeutic usage of these preparations.     Description: The mouse neuroblastoma-rat glioma hybrid cell line NG108-15 was used to study the acute and chronic interaction of ethanol with intact neural cells. In the short term, ethanol inhibited opiate receptor binding, but after long-term exposure the cells exhibited an apparent adaptive increase in the number of opiate binding sites; this was reversible when ethanol was withdrawn. High concentrations of ethanol (200 mM) increased opiate binding after 18 to 24 hours, whereas lower concentrations (25 to 50 mM) produced similar changes after 2 weeks.     Description: The DNA's from two of four methylcholanthrene-induced mouse fibrosarcomas contained transforming genes that were identical in their pattern of restriction endonuclease resistance to inactivation of biologic activity.     Description: The polymorphism of immune response genes plays a critical role in determining the immune capabilities of a particular individual. The molecular nature of this polymorphism was studied by examining the structure of the coding portions of three alleles of the I-A beta chain gene, an immune response gene whose protein product constitutes a subunit of the I-A molecule. Comparison of the I-A beta chains encoded by these alleles revealed an amino acid sequence divergence of 5 to 8 percent.     Description: Inbred LP/J mice develop abnormal bony lesions that are grossly and histologically similar to the lesions of human otosclerosis. This is the first known occurrence of spontaneous otosclerosis-like lesions in an animal. As in the human disease, these lesions impair audition by immobilizing the ossicles of the middle ear.     Description: The utility of somatic cell genetic analysis for the chromosomal localization of genes in mammals is well established. With the development of recombinant DNA probes and efficient blotting techniques that allow visualization of single-copy cellular genes, somatic cell genetics has been extended from the level of phenotypes expressed by whole cells to the level of the cellular genome itself.
