    Notes: Abstract Recently published results for solution crystallized PE single crystals have shown, that the experimental heat of fusionΔH * is higher, if the solvent is exchanged to silicon oil (oil suspension samples) as compared with dried mats. This has been interpreted by the collapse of the original hollow pyramids during drying, inducing lateral defects within the lamellae. The present investigation does not confirm this unexpected result.ΔH * of dried mats (T c 66 to 91 °C) and of the corresponding oil suspension samples agree within the rather small limits of experimental error. The crystallinities as derived fromΔH *, density or WAXS are in excellent agreement. SEM micrographs of cold fractured dried mats show their spongy macromorphology, but TEM micrographs of stained ultra-thin sections reveal the lamellar morphology of the walls, consisting of curved lamellae and stacked hollow pyramides. If a dried mat is sintered at room temperature, a dense transparent film is obtained with a rather regular stacked morphology of large flat lamellae.ΔH * of these films agrees with that of the original mat.

    Notes: Summary The nucleoside antibiotic nikkomycin has proved to be an effective inhibitor of chitin synthesis in the Mexican bean beetleEpilachna varivestis. Ultrastructural investigations show defects in the procuticular area after nikkomycin application which suggest the complete absence of chitin. A cuticle like this is inflexible and too brittle to satisfy its normal function as an exoskeleton. The individuals are not able to free themselves from the exuvia and finally die. Therefore nikkomycin seems to be a potential insecticide with high specifity.

    Notes: Summary The ultrastructural localization and gradient of activity of alkaline phosphatase were studied with respect to cell differentiation, matrix synthesis, and matrix mineralization in the incisor and molar teeth of 4-day-old Sprague-Dawley rats. The animals were perfused intracardially at room temperature with 2.5% glutaraldehyde in 0.1M sodium cacodylate (pH 7.4) with 3–4% sucrose. The jaws were dissected, immersion-fixed for 24 h, and the incisor and molar tooth germs removed. These were demineralized in 10% EDTA in NaOH (pH 7.4) with 7% sucrose. After reactivation of the enzyme with 0.1M MgCl in Tris-maleate buffer (pH 7.4) at 4°C, the teeth were incubated for alkaline phosphatase in a medium consisting of 6 ml 3% sodiumβ-glycerophosphate, 4 ml 0.2M Tris-HCl buffer (pH 9.2), 3 ml 1.6% MgSO4, 12 ml 0.5% lead citrate (pH⋍12), and 2.1 g sucrose. The pH was adjusted to 9.2 with 0.2M HCl, the volume made up to 30 ml, and the solution centrifuged for 10 min at 5000 rpm. Control teeth were incubated in medium minus the substrate. Finally, the specimens were routinely post-fixed and embedded for sectioning and examination with a Philips 300 electron microscope. A gradient of alkaline phosphatase activity was mapped along the developing teeth in the cells of the stratum intermedium, the proximal borders of the ameloblasts, the early dentine matrix, the predentine-dentine border, matrix vesicles, and the plasma membranes of odontoblasts and subodontoblast cells. The gradient of alkaline phosphatase activity was evident in the forming tooth from the cervical loop to the crown apex and was related to the cellular events, matrix synthesis, and matrix mineralization occurring during odontogenesis.

    Notes: Summary Diaphyseal tibial bone of 12.5 – 13-day and 19-day-old embryos and 20-day-old hatched chicks infected with retrovirus MAV.2-O were examined by transmission electron microscopy. The viruses were associated with lining osteoblasts and osteocytes. Whereas the infection of the osteoblast layer seemed to be a transient stage, virus association with osteocytes was a constant and main ultrastructural feature. The viruses were found either in the osteoid or in the periosteocytic space of the bone lacunae. They arose from dense cytoplasmic areas located near the cell plasmalemma via a budding process. The newly budded virus particles often had a large tail or a fine stalk-like process lost in the extracellular space. The viruses underwent calcification by deposition of inorganic material and were incorporated in the bone trabeculae. No production of virus was observed in typical osteoclasts with well-differentiated ruffled borders. The viral-induced avian osteopetrosis seemed to result from increased bone deposition through stimulation of osteoblast and osteocyte activities, whereas osteoclastic bone resorption seemed to be undisturbed.

    Notes: Abstract The morphology and ultrastructure of the thermophilic cyanobacteriumMastigocladus laminosus were examined by scanning and transmission electron microscopy. Mature cultures consisted of relatively old, wide filaments that branched frequently to form younger, thinner filaments. The cells of the younger filaments had a consistently cylindrical morphology, while those of older filaments were rounded and pleomorphic. The internal ultrastructure of the cells depended somewhat on their age. As young cells became larger and wider, their thylakoids underwent slight rearrangement and spread out toward the center of the cytoplasm. Polyphosphate bodies, carboxysomes (polyhedral bodies), and lipid-body-like structures increased in number as the cells aged, but ribosomes and cyanophycin granules were depleted. Cell division involved septum formation followed by ingrowth of the outer membrane and sheath. Cells in older filaments were separated from each other by a complete layer of sheath material. Septum formation in older cells was also seen to occur parallel to the long axis of the filament, thereby confirming that true branching took place.

    Notes: Abstract Among 12 strains ofChlorella ellipsoidea, C. vulgaris, andC. saccharophila tested, 4 strains (1,C. ellpsoidea; 2,C. vulgaris; 1,C. saccharophila) formed osmotically labile protoplasts after treatment with mixtures of polysaccharide degrading enzymes. The relationship between enzymatical digestibility and structure or composition ofChlorella cell walls were studied by electron microscopy and staining techniques with some specific dyes. The cell wall structures of the 12Chlorella strains were grouped into three types: (1) with a trilaminar outer layer, (2) with a thin outer monolayer, and (3) without an outer layer. Protoplasts were formed only from the strains with a cell wall of Type 2. In the strains with a cell wall of Type 1, the outer layer protected the inner major microfibrillar layer against enzymatic digestion. The cell wall of Type 3 was totally resistant to the enzymes; the chemical composition of the cell wall would be somewhat different from that of other types.

    Notes: Abstract An ultrastructural study of four cyanobacteria (Anabaena cylindrica, Dermocarpa violaceae, Gleocapsa alpicola, Pleurocapsa minor) indicates the presence of previously undescribed thylakoid centers from which photosynthetic membranes (thylakoids) radiate. These peripherally located thylakoid centers are cylinders 30 nm wide by 320 nm long, consisting of globular subunits oriented in nonparallel stacked arrays. Thylakoids are attached to the outer surface of the cylinder along its longitudinal axis. Thylakoid centers appear to be functionally significant due to their structure, location and thylakoid association.

    Notes: Abstract In contrast to former findings lysozyme was able to attack the cell walls ofStaphylococcus aureus under acid conditions. However, experiments with14C-labelled cell walls and ribonuclease indicated that, under these conditions, lysozyme acted less as an muralytic enzyme but more as an activator of pre-existing autolytic wall enzymes. Electron microscopic studies showed that under these acid conditions the cell walls were degraded by a new mechanism (i.e. “attack from the inside”). This attack on the cell wall started asymmetrically within the region of the cross wall and induced the formation of periodically arranged lytic sites between the cytoplasmic membrane and the cell wall proper. Subsequently, a gap between the cell wall and the cytoplasmic membrane resulted and large cell wall segments became detached and suspended in the medium. The sequence of lytic events corresponded to processes known to take place during wall regeneration and wall formation. In the final stage of lysozyme action at pH 5 no cell debris but “stabilized protoplasts” were to be seen without detectable alterations of the primary shape of the cells. At the same time long extended ribbon-like structures appeared outside the bacteria. The origin as well as the chemical nature of this material is discussed. Furthermore, immunological implications are considered.

    Notes: Summary An attempt to produce a set of addition lines of Aegilops sharonensis to the wheat variety ‘Chinese Spring’ produced only one addition line. This was due to preferential transmission of one chromosome from Ae. sharonensis. This chromosome was studied in detail by established cytological methods of chromosome observation and by the newer techniques of C-banding and in situ hybridization of a cloned DNA sequence. The chromosome was found to be partially homologous to an Ae. sharonensis chromosome of similar behaviour in another wheat addition line. The incomplete homology of the two Ae. sharonensis chromosomes was due to the presence of a translocated segment of a wheat chromosome. — Substitution lines of the Ae. sharonensis chromosome for wheat homoeologous group 4 were produced and the Ae. sharonensis chromosome thereby designated 4 S l .

    Notes: Summary Using different stains, wheat chromosomes could be distinguished from rye chromosomes by preferential staining. C-bands of rye chromosomes were preferentially stained with Giemsa while those of wheat chromosomes were preferentially stained with either Leishman or Wright stain. Preferential staining aids the identification of wheat and rye chromosomes and chromosome segments and in particular the recognition of wheat/rye chromosome substitutions and translocations.

    Notes: Summary Two NAD-dependent alcohol dehydrogenases ADH-1 and ADH-2, under independent genetic control of genes designated as Adh-1 and Adh-2 located on chromosomes 4A, 4B and 4D, have been reported in aestivum wheat (Hart 1980). Only ADH-1 is expressed in developing seeds, dry seeds, pollen and germinating seedlings. ADH-2 can be induced in seedling roots or shoots under conditions of partial anaerobiosis or by certain chemicals. Expression of ADH-1 and ADH-2 isoenzymes was investigated in undifferentiated calli from aestivum and durum wheats, rye, triticale and also in in vitro regenerated roots and leaves from aestivum cultures. Wheat callus cultures originating from seed, mature and immature embryos, mesocotyl and root, as well as cultures grown on media containing different supplements did not show any variation in the overall expression of ADH-1 or ADH-2, although differences in the band intensities were observed. The callus isoenzyme pattern was similar to that observed in roots under anaerobic conditions. Both ADH-1 and ADH-2 were expressed in in vitro regenerated roots but were absent in regenerated leaves. Expression of ADH-1 and ADH-2 in wheat calli seems to be related to the type of differentiation.

    Notes: Summary Genetical analysis of the F2 triple test cross design combined with conventional early generations was used to elucidate the genetical control of yield and yield components in two crosses of winter wheat. From estimates of the additive, {d}, and additive X additive, {i}, components of means, together with the additive genetical variance, D, predicted frequencies of recombinant inbred lines that would transgress the parental range were calculated for each cross. The accuracy of predictions was evaluated by comparing expected frequencies with observed numbers in populations of F6 lines previously developed by single seed descent. For both crosses and all characters where an adequate genetical model was found to explain the observed variation between the early generations, good agreement between predicted and observed frequencies of transgressive segregants was obtained. Furthermore, for characters exhibiting significant epistasis, allowance for additive X additive {i} epistasis in the prediction equations was sufficient to allow for skewness of the recombinant inbred population. These results demonstrate that cross performance in wheat can be predicted from genetical analysis of early generations, and the value of this approach in breeding new varieties is discussed.

    Notes: Summary The effects of the Tom Thumb dwarfing gene, Rht3, on the quality and quantity of grain α-amylase produced during germination and by induction with exogenous gibberellic acid are described. In a season conducive to high sprouting damage the gene reduced α-amylase levels in the field by 77%. Selection among random Rht3 genotypes showed that other genetic factors can be combined with the dwarfing gene to further increase sprouting damage resistance.

    Notes: Summary The 6-phosphogluconate dehydrogenase (6-PGD) zymogram phenotypes of wheat, rye and their aneuploid derivatives were determined. Two genes involved in the production of 6-PGD isozymes were located on chromosome arms CRL (4 RL) and FRL (6 RL) of “Imperial” rye. On the basis of differential interactions between wheat and rye chromosomes, evidence was obtained that genes located on chromosomes 6 A, 6 BL and 7 BL control 6-PGD isozyme activities in ‘Chinese Spring’ wheat. The wheat and rye 6-PGD zymogram phenotypes were indicative of homoeologous relationships between rye chromosome 6 RL to wheat chromosomes of group 6, and rye chromosome 4 RL to wheat chromosomes of group 7.

    Notes: Summary Homologous high molecular weight storage prolamins were purified from grain of wheat, rye and barley using combinations of gel filtration, ion-exchange chromatography and preparative isoelectric focusing. Sodium dodecylsulphate polyacrylamide gel electrophoresis showed that the components were single bands with apparent mol.wts. of above 100,000. Molecular weights determined by sedimentation equilibrium ultracentrifugation were considerably lower; 54,700, 67,600 and 69,600 for the components from barley, rye and wheat respectively. Amino acid analysis showed the presence of 13.6 to 16.5 mol% glycine, 29.6 to 34.0 mol% glutamate + glutamine, 11.4 to 13.7 mol% proline and a total of 4.0 to 5.7 mol% basic amino acids. Automated N-terminal amino acid sequencing of the component from wheat showed the presence of cysteine residues at positions 5 and 10, and this is discussed in relation to the possible role of these proteins in the visco-elastic gluten network.

Chromosome pairing behaviour revealed that the alien chromosome in lines TAF-2 and L7 of ‘Vilmorin’-A. intermedium set are homologous to the alien chromosomes in lines P and C of the ‘Caribo’-A. intermedium set respectively. Localization of alcohol dehydrogenase isozyme genes in ‘Vilmorin’-Agropyron addition line L4 and in ‘Caribo’-Agropyron line O indicated relationships with wheat chromosomes of homoeologous group 4.

    Notes: Summary The wheat rDNA clone pTA250 was examined in detail to provide a restriction enzyme map and the nucleotide sequence of two of the eleven, 130 bp repeating units found within the spacer region. The 130 bp units showed some sequence heterogeneity. The sequence difference between the two 130 bp units analysed (130.6 and 130.8) was at 7 positions and could be detected as a 4 °C shift in Tm when heterologous and homologous hybrids were compared. This corresponded to a 1.2% change in nucleotide sequence per ΔTm of 1 °C. The sensitivity of the Tm analysis using cloned sequences facilitated the analysis of small sequence variations in the spacer region of different Triticum aestivum cultivars and natural populations of T. turgidum ssp. dicoccoides (referred to as T. dicoccoides). In addition spacer length variation was assayed by restriction enzyme digestion and hybridization with spacer sequence probes. Extensive polymorphism was observed for the spacer region in various cultivars of T. aestivum, although within each cultivar the rDNA clusters were homogeneous and could be assigned to particular chromosomes. Within natural populations of T. dicoccoides polymorphism was also observed but, once again, within any one individual the rDNA clusters appeared to be homogeneous. The polymorphism, at the sequence level (assayed by Tm analysis), was not so great as to prevent the use of spacer sequence variation as a probe for evolutionary relationships. The length variation as assayed by restriction enzyme digestion did not appear to be as useful in this regard, since its range of variation was extensive even within populations of a species.

    Notes: Summary The nature of genome change during polyploid evolution was studied by analysing selected species within the tribe Triticeae. The levels of genome changes examined included structural alterations (translocations, inversions), heterochromatinization, and nucleotide sequence change in the rDNA regions. These analyses provided data for evaluating models of genome evolution in polyploids in the genus Triticum, postulated on the basis of chromosome pairing at metaphase I in interspecies hybrids. The significance of structural chromosome alterations with respect to reduced MI chromosome pairing in interspecific hybrids was assayed by determining the incidence of heterozygosity for translocations and paracentric inversions in the A and B genomes of T. timopheevii ssp. araraticum (referred to as T. araraticum) represented by two lines, 1760 and 2541, and T. aestivum cv. Chinese Spring. Line 1760 differed from Chinese Spring by translocations in chromosomes 1A, 3A, 4A, 6A, 7A, 3B, 4B, 7B and possibly 2B. Line 2541 differed from Chinese Spring by translocations in chromosomes 3A, 6A, 6B and possibly 2B. Line 1760 also differed from Chinese Spring by paracentric inversions in arms 1AL and 4AL whereas line 2541 differed by inversions in 1BL and 4AL (not all chromosomes arms were assayed). The incidence of structural changes in the A and B genomes did not coincide with the more extensive differentiation of the B genomes relative to the A genomes as reflected by chromosome pairing studies. To assay changing degrees of heterochromatinization among species of the genus Triticum, all the diploid and polyploid species were C-banded. No general agreement was observed between the amount of heterochromatin and the ability of the respective chromosomes to pair with chromosomes of the ancestral species. Marked changes in the amount of heterochromatin were found to have occurred during the evolution of some of the polyploids. The analysis of the rDNA region provided evidence for rapid “fixation” of new repeated sequences at two levels, namely, among the 130 bp repeated sequences of the spacer and at the level of the repeated arrays of the 9 kb rDNA units. These occurred both within a given rDNA region and between rDNA regions on nonhomologous chromosomes. The levels of change in the rDNA regions provided good precedent for expecting extensive nucleotide sequence changes associated with differentiation of Triticum genomes and these processes are argued to be the principal cause of genome differentiation as revealed by chromosome pairing studies.

The presence and distribution of extracellular substances and their source was investigated cytochemically by transmission electron microscopy. Extracellular material, possibly-acid mucopolysaccharide, was observed consistently within the entire length of the raphe of both valves and also as a sheath enveloping the silica frustule. Such quantities of extracellular material are absent in conventionally fixed motile diatoms. Numerous cytoplasmic vesicles, with fibrillar contents, distributed peripherally but concentrated along the raphe and at the cell poles, react strongly with a polysaccharide specific stain; their distribution in the cell and polysaccharide content suggest these may be the source of raphe and sheath material. Results support the most recent theories on the mechanism of locomotion in outline only; the details cannot be clarified. Localization procedures using alcian blue and silver staining of peroxidised sections are discussed briefly.

Differences in thickness and symmetry of membranes and in contents of vesicles were used to distinguish different types of membranes. Mitochondria, vacuoles, plasma membrane, and vesicles with attached ribosomes from homogenized germlings equilibrated at the 1.2/1.4 M interface in discontinuous sucrose gradients. Accelerated flotation in centrifuged Ficol-sucrose gradients resulted in the additional separation of the mixed membranes into three fractions: one contained predominantly intact mitochondria, another was composed of vacuoles and vesicles coated with ribosomes, and a third was enriched in plasma membranes. Based upon morphometric analysis, these fractions contained 92% mitochondria, 53% vacuoles, and 89% plasma membranes, respectively. The source of vesicles coated with ribosomes was investigated since rapidly growing hyphae ofG. persicaria contained little rough endoplasmic reticulum as compared with other classes of membranes. Reconstruction from electron micrographs of mitochondrial fragmentation and vesiculation suggested that most of the ribosome-coated vesicles originated from disrupted mitochondria rather than from rough endoplasmic reticulum. The study demonstrates the utility of ultrastructural markers to identify membranesin vitro independent of, or as an adjunct to, cytochemical and biochemical markers.

This secretion stains positively for protein. Stigmas are of the “wet” type. The cuticle overlying the papillar cells is ridged and at the final stages prior to flowering this cuticle becomes detached from the underlying cellulosic wall. The sub-cuticular space so formed is filled with secretion. InAneilema species detachment of cuticle is at the papillar tip and along the lateral walls. InCommelina species the anticlinal walls of adjacent papillae are strongly attached for much of their length and thus detachment of cuticle is restricted to the papillar tip. The cell wall at the tip in both genera may proliferate forming a rudimentary transfer-cell type wall. The secretion is considered to be produced by the papillar cells. It is PAS positive but fails to stain for protein and in both the light and electron microscopes appears heterogenous. Pollen attachment, hydration, germination and early tube growth are very rapid following self-pollination, the pollen tubes entering the neck of the style within ten minutes of attachment. A unique character combination involving pollen and stigmas in these genera indicates a monophyletic origin.

The effects of a range of cations on tentacle contraction and ultrastructure have been determined. Treatment with 80 mM CaCl2 and 95 mM MgCl2 causes contraction to 28% and 57% of the control length respectively. Re-extension takes over 4 hours in the culture medium, but CaCl2-treated tentacles are re-extended after a 5 minutes treatment with 10−2 M EDTA or 5 × 10−3 M EGTA. CuCl2 causes a significant contraction at 10−5 M (to 77%); LaCl3 at 10−4 M (to 65%); ZnCl2 at 10−2 M (to 65%), but BaCl2, CoCl2, MnCl2, NiCl2, and SrCl2 cause significant changes only at 10−1 M. The cytoplasm of CaCl2-treated cells contains two forms of membraneous structures when viewed in TEM; that of MgCl2-treated cells reveals granular areas of medium electron density. None of these features are seen in control cells. The microtubules of the tentacle canal appear to be intact upon its retraction into the cell with no change occurring in the numbers or relative positions of the microtubules. The tentacle cortex is wrinkled. It is suggested from this and previous work that tentacle contraction may be mediated by a microfilament-based mechanism, and that calcium may be involved.

Zoospores of the aquatic fungi,Chytriomyces aureus andC. hyalinus, contain microbody-lipid globule complexes with an elongate microbody adjacent to the portion of a lipid globule facing the cell's interior and a fenestrated cisterna (the rumposome) opposed to the surface of the lipid globule toward the plasma membrane. Mitochondria are intimately associated with the microbody. Electron microscopy of the microbody-lipid globule complex fixed in glutaraldehyde and osmium tetroxide, with or without tannic acid, reveals cross-linking bridges connecting the rumposome to the plasma membrane, to the microbody, and to microtubules of the rootlet extending from the kinetosome. It is concluded that these bridges are responsible, at least in part, for the consistent location of the microbody-lipid globule complex in the zoospore body. The possible role of the rumposome as a receptor organelle is discussed.

    Notes: Summary Callus-derived suspension cultures of oats dramatically increase the viscosity of the culture media after one month in culture. Colorimetric assays for sugars and protein, as well as measurements of viscosity, suggest that the released material is a long-chain polysaccharide, probably a pectinaceous substance. These cells grow slowly in liquid culture, yet despite their low cell density, they are able to increase the viscosity of the media several fold within seven days after media transfer. Ultrastructural observations show that oat cells have features common to actively-secreting cells; especially evident are numerous dictyosomes with hypertrophied cisternae. Using a combination of filtering and centrifugation techniques we were able to recover large numbers of intact secretory vesicles. The interior of the vesicles stain with periodic acid-silver hexamine, and colormetric analysis of the vesicle pellet for total sugars confirms the presence of polysaccharides in this vesicle fraction. Because of the uniformity of these cells, the high rate of secretion, and the accessability of a large vesicle population, this culture system is'a useful model for studying the secretory process in plant cells.

    Notes: Summary Tissue cultures ofTriticum aestivum L. (wheat) initiated from young inflorescences and immature embryos possessed the potential for regeneration of whole plants. Both a friable and a compact type of callus were produced on Murashige and Skoog's medium with 2 mg/l 2,4-dichlorophenoxyacetic acid. The friable callus contained meristematic centers in which the peripheral cells ceased dividing, elongated, and could be easily separated. Roots were frequently formed in this type of callus. The compact, yellowish, and nodular callus arose from the epithelial and sub-epithelial cells of the embryo scutellum, and the rachis and glumes of the young inflorescence. Such callus had a smooth surface and characteristic chlorophyllous areas. Plants were regenerated only from the compact callus. The first sign of differentiation in the compact callus was the formation of a cleft or notch on the smooth surface, followed by the appearance of trichomes and the direct development of leafy structures which were not associated initially with any shoot meristems. Multiple shoots subsequently arose at the bases of the leafy structures, which are considered modifications of the scutellum, a definitive part of the cereal embryo. Accordingly, we suggest that while typical bipolar embryos are generally not formed, plant regeneration nevertheless takes place through embryogenesis and the precocious germination of the embryoids. Plants regenerated from immature embryo and inflorescence cultures were grown to maturity in soil, and were shown to have the normal chromosome number of 2n=6x=42.

    Notes: Summary Natural senescence of the root cortex was assessed by nuclear staining, for cultivars and chromosome substitution lines of spring wheat known to differ in (1) susceptibility to common root rot, (2) total rhizosphere populations and (3) ability to support growth of a free-living nitrogen-fixing bacterium. Together, three root rot susceptible wheat lines showed significantly more cortical senescence than did three resistant lines; the susceptible lines also support larger rhizosphere populations. The wheat line that supports growth of a nitrogen-fixing bacterium showed significantly less cortical death than did any other line. Substitution of chromosome pairs 5B or 5D between the parent cultivars Rescue and Cadet substantially altered the amount of root cortex death, which is thus genetically determined. It is suggested thatCochliobolus sativus and other weak parasites benefit from early natural senescence of the root cortex, and that the degree of susceptibility or resistance of wheat lines to common root rot is at least partly determined by differences in cortical senescence.

    Notes: Summary We report a study of the mechanism by which the response of plants to waterlogging can be modified by soil temperature. Wheat was grown initially in well-aerated soil in a controlled environment room before the soil was flooded with aerated, deionized water. The soil temperature was maintained constant in the range 6–18°C while the air temperature was at 14°C. Waterlogging damage was greater in plants at the higher soil temperatures when the plants were compared at the same chronological age. However, when compared at the same growth stage, the response to soil temperature was little differenti.e. plants subjected to waterlogging for a long time at low soil temperatures exhibited a similar reduction in growth and other properties as those subjected briefly at higher temperatures. The concentration of dissolved oxygen in the soil solution declined rapidly at all temperatures, being almost zero after 36 h waterlogging. Temperature affected rates of change of the concentrations of dissolved carbon dioxide, ethylene, nitrous oxide, nitrite, nitrate, calcium and potassium. The importance of soil-and plant-determined properties in the waterlogging response of plants at different temperatures are discussed.

    Notes: Summary Nuclear and cytoplasmic staining methods were used to study natural senescence of the root cortex and coleoptile of wheat and barley seedlings grown in glasshouse conditions. Coleoptiles of barley senesced more slowly than those of wheat, paralleling the known difference in rates of root cortex senescence in these cereals. The coleoptiles and root cortices of both cereals senesced more slowly in shaded than in unshaded conditions, but infection of the shoots of barley byErysiphe graminis had little effect on root cortex senescence. The results are discussed in relation to infection by root- and foot-rot fungi. Previous reports on the effects of illumination on take-all infection (Gaeumannomyces graminis) are explained. It is suggested that natural senescence of the coleoptile might affect establishment of infection by the eyespot fungus,Pseudocercosporella herpotrichoides, either directly or through the activities of competing microorganisms.

    Notes: Summary A study conducted in pots to evaluate the effect of different Mg/Ca ratios (2, 4, 8 and 16) and electrolyte concentrations (20 and 80 meq/l) at SAR 10 in irrigation water on the nutrient uptake and yield of wheat crop in two soils revealed that the average grain and dry matter yields of wheat decreased significantly with an increase in Mg/Ca ratio in irrigation water, but the magnitude of decrease was greater at higher electrolyte concentration than at lower electrolyte concentration. The concentration of Na in both straw and grain of wheat increased and that of K decreased with an increase in Mg/Ca ratio and electrolyte concentration of irrigation water, which led to higher Na/Ca and Na/K ratios in the plant. Further, the concentration of Ca and Mg both in straw as well as in grain increased with increasing electrolyte concentration of the irrigation water. An increasing proportion of Mg in saline irrigation water resulted in decreased concentration of Ca and increased concentration of Mg in both straw and grain of wheat crop.     Notes: Abstract Soil samples from a long term field experiment, to study the K depletion pattern, were analysed for K. The values of supply parameters pK−1/2p (Ca+Mg) and free energy δG r 0 were calculated using the data on water soluble and exchangeable cations. Correlation coefficient values between pK−1/2p (Ca+Mg) vs. total K removed, K removal by pearl millet in 1980 and soil available potassium were positive and statistically significant. This indicated that these soils have reached the stage where they would start responding to potassium application. Also, negative but significant correlation of δG r 0 with pK−1/2p (Ca+Mg) and available K, were found. δG r 0 values were positive indicating the affinity of such soils for K and the values, for K fertilized plots were lower in comparison to plots where no K was applied. pK−1/2p (Ca+Mg) parameter was found to be a better measure of K availability over δG r 0 due to either fixation of K or greater contribution of nonexchangeable K fraction towards plant utilized K.

In the greenoouse, barley was grown to maturity in pots on a sandy soil which contained 80 and 120 meq/l of chloride and sulphate dominant salts in its saturation extract, to which 0, 10, 25 and 50 ppm P were added. In the field study, wheat was grown on loamy sand soils having 0, 25, 50 and 75 kg/ha added P levels and irrigated with either Cl- or SO4-dominant saline waters (EC=15−19 mmhos/cm). The results of the greenhouse study indicated that at maturity barley straw and grain yield was significantly increased by 50 ppm of added P both on the non-saline control and the Cl-treatments. However, 25 ppm P was optimal on the SO4-treatments. The Cl content of plants was significantly decreased and S was increased with the increase in the P content of soil. A synergistic relation between the S and P content of barley shoots was observed. In the field study wheat grain yield responded significantly to P applications upto 50 kg/ha level on the Cl-site and there was no response to applied P on the SO4-site, although the former contained more Olsen's P than the latter. The results suggested that P requirement of wheat and barley was greater on Cl- than on SO4-salinity.

The nitrogen treatments did not influence the moisture content of soil. The application of mulch or mulch+Kaolin resulted in significantly higher content of soil moisture in 0–15 cm soil depth as compared to control or Kaolin spray alone which was simultaneously reflected in yield and yield contributing characters.

    Notes: Summary Three wheats and one triticale were grown, up to flowering stage, in pots on calcareous soil adjusted to a range of salinities (S1=3.5, S2=6, S3=8.5, and S4=11 mmhos/cm, 20°C, soilpaste extract) by adding solution consisting of 3∶2∶1 of Na-, Ca- and Mg chlorides in chemical equivalent amounts. Moisture in the pots was kept at 100% (W1), 40% (W2) and 20% (W3) of the available water. The vegetative growth, nitrogen and phosphate were affected by S and W treatments, chloride was affected only by S. The interaction S×W affected only dry weight. Varietal effect was observed between wheat as a group and triticale. Multiple quadratic regression equations of these properties on salinity and water revealed that the higher the available water the wider the range of tolerable salinity. Triticale was relatively more tolerant to water stress. Salinity increases Cl and decreases N, whereas water stress enhances N accumulation to a certain extent. However, in triticale at S3 and S4 the effect of water stress on N was overshadowed by the excessive salinity. This did not occur for the wheat (Florence). P trends were described. R2 for P was low (0.7435–0.3603) which made interpretations rather difficult.

    Notes: Summary Aspects of selection for yield and harvest index were investigated by simulating selection using data from random pedigreed F2, F3, F4 and F5 derived lines from two crosses grown in plots at two sites over two years. Improvement in yield through selection was obtained when the response was measured at the same site and in the same year as the selection. Selecting the best 10 per cent of F2 to F4 derived lines gave F5 derived lines that outyielded random selections by 19 to 53 per cent for one cross and 5 to 23 per cent for the second cross. These lines were 41 to 50 per cent better than the mid-parent in one cross, but were less than the mid-parent in the other cross. However, the response to selection when measured in a different year was little better than random selection. The effect of different sites also reduced the effectiveness of selection. Selection of harvest index in early generations for improvement of yield was ineffective when response was measured at the same site in the same year, or in different years. Contrary to some theoretical proposals, the same improvement in yield was obtained by selecting in early or late generations. While high yielding genotypes may be lost by delaying selection, this is counteracted by the better predictive value of late generations due to their greater homozygosity and homogeneity.

    Notes: Summary The rhinophores of the veliger larva of Rostanga pulchra are located in the intravelar field near the base of the velar lobes. Each rhinophore is a cylindrical structure, tapering distally, and covered with a dense meshwork of microvilli. A conspicuous row of ciliary tufts runs along each side of the rhinophore and several stiffer tufts, composed of fewer cilia, are positioned around the tip or at the base. The rhinophoral epithelium consists of supporting cells, ciliated cells (giving rise to the ciliary rows), dendritic terminals (giving rise to the tufts around the apex), and sinuses containing occasional amebocytes. The lumen of the rhinophore is occupied by the rhinophoral ganglion and muscle cells that are oriented in two perpendicular planes. Cell bodies of the dendritic endings are located within the rhinophoral ganglion, which in turn joins into the optic and cerebral ganglia. Rhinophoral ganglionic neurons do not synapse with each other, but numerous neuromuscular synapses are found in the lumen of the rhinophore. Morphological evidence suggests that the dendritic endings are chemoreceptors and the ciliated cells are possibly mechanoreceptors but are not functional at this stage in development. The functional role of the rhinophores is discussed in relation to larval behavior at settlement and metamorphosis.

    Notes: Summary The presence and differential distribution of substances antigenically related to known vertebrate neuropeptides demonstrated within the corpus cardiacum of the insect Leucophaea are as follows: Of ten mammalian antisera tested, six yielded substantial immunoreactive deposits resembling oxytocin, somatostatin, Substance P, met-enkephalin, bombesin, and neurotensin, respectively. In the remaining four, the reaction was moderate (vasopressin, β-endorphin) or marginal (LH-RF, calcitonin). With regard to their regional distribution, these biochemically distinct reaction products seem to fall into two groups: (1) Materials resembling oxytocin, vasopressin, met-enkephalin, β-endorphin (and presumably also neurotensin and LH-RF) predominate in the central release area of the organ and are considered to be of extrinsic (cerebral) origin. (2) Substances localized primarily in areas rich in intrinsic glandular cells of the corpus cardiacum, and revealed by antisera raised against somatostatin, Substance P, and bombesin, are judged to be synthesized and stored within this organ. In peptidergic fibers entering the adjacent corpora allata, thus far Substance P-, β-endorphin-, and LH-RF-like immunoreactivities have been demonstrated. Some of these “new” neuropeptides may be contained in classical neurosecretory neurons, formerly identified by less specific methods, others must be assigned to additional peptidergic neurons heretofore unknown.

    Notes: Summary With an antiserum to the molluscan cardioactive tetrapeptide FMRF-amide immunoreactive perikarya and nerve fibers were identified in the central and peripheral nervous system of the pond snail Lymnaea stagnalis. Their localization is described. The same antiserum yielded reactive product in particular cells of the epithelium of the alimentary tract. The use of two different fixatives, glutaraldehyde, and a mixture of glutaraldehyde, picric acid, and acetic acid (GPA) showed that certain nerve cells can be identified only in material fixed with either the one or the other of these two fixatives, a result which indicates that in Lymnaea more than one FMRF-amide-like substance may occur. “Positive” axon endings were found in the periphery of various nerves, i.e., in places where neurohormones are released into the blood. Other fibers were found to end, probably synaptically, on other neurons, on epithelial cells in the stomach, and between muscle cells in various parts of the body, e.g., in the heart. In these cases the FMRF-amide-like substance may function as a neurotransmitter or a neuromodulator.

    Notes: Summary By the use of thin sections and freeze-fracture replicas the glomerular and tubular structures of the kidney of the frog (Rana esculenta) were studied with special reference to intercellular junctions. In the glomerulus the filtration barrier is of very variable thickness, and frequent tight and gap junctional contacts occur between podocyte processes. Although structurally less elaborate, the proximal tubule resembles its mammalian counterpart. In the initial part the tight junctions are relatively shallow but become very broad in the mid and distal portions of the proximal tubule. The proximal tubular cells are extensively linked by gap junctions. In some animals the shapes of the cells in the proximal and distal portions of the proximal tubule were markedly different. The distal tubule consists of two segments which differ mainly in the pattern of interdigitations and the structure of the zonulae occludentes. Similarities with the tight junctional morphology of the mammalian distal tubule are striking. In the first part of the distal tubule (diluting segment) a narrow band of parallel tight junctions is found closely resembling that found in the mammalian straight distal tubule; in the more distal part of the distal tubule, however, a broad band of anastomosing tight junctional strands exists, like the zonula occludens of the mammalian convoluted distal tubule. The connecting tubule displays cellular dimorphism: its wall contains a mixture of light and dark (flask) cells. The luminal and basolateral membranes of the flask cells are covered with numerous rod-shaped particles. The tight junctions of the connecting tubule are broad and increase in depth and number of strands along its length; they are typical of a very tight epithelium. In spite of several dissimilarities with phylogenetically younger kidneys our findings suggest that many structural principles of the mammalian kidney are also represented in the kidneys of amphibians. The structural-functional relationships are discussed.

    Notes: Summary The presence of prolactin (PRL)-like material is demonstrated in the brain of rats with the aid of anti-ovine PRL (oPRL) IgG as primary antibody in the unlabeled antibody-enzyme method. Immunoreactive deposits are visualized as an intraneuronal constituent with a widespread distribution in the hypothalamus and neural lobe of the pituitary. Dense networks of reactive nerve terminals derived from two prominent fibre tracts, a ventral (VHT) and a dorsal hypothalamo-neurohypophysial tract (DHT) are seen. The VHT is confined to the median eminence and pars oralis tuberis, the DHT to the pars caudalis tuberis. Both fibre tracts pass through the infundibular stalk into the neural lobe. The origin of the immunoreactive nerve terminals can be elucidated only to some extent. The VHT gives off beaded fibres entering the ependymal and glandular layer of the median eminence. Immunoreactive perikarya are observed in the supraoptic nucleus, the paraventricular nucleus, the anterior hypothalamic nucleus, the anterior commissural nucleus, the preoptic nucleus and the interstitial nucleus of the stria terminalis. A few of the immunoreactive perikarya are observed in close connection with brain vessels and the ependymal cells of the third ventricle. The results indicate that the anti-oPRL has a unique region specificity implying that only a segment of the mammalian PRL molecule is present in these nuclei of the brain. Fragments of PRL may function as neuromodulators or neurotransmitters in the rat brain.

    Notes: Summary The ultrastructure of the pinealocytes of the wild-captured ord kangaroo rat (Dipodomys ordi) was examined. A homogeneous population of pinealocytes was present in the pineal gland of the kangaroo rat. The Golgi apparatus, granular endoplasmic reticulum, mitochondria, lysosomes, dense-core vesicles, vacuoles containing a flocculent material and lipid droplets were consistent components of the pinealocyte cytoplasm, whereas infrequently-observed organelles included centrioles, multivesicular bodies, subsurface cisternae, “synaptic” ribbons and cilia. The number of dense-core vesicles was relatively high and dense-core vesicles and vacuoles containing a flocculent material were present in the same cell. Although it has been recently suggested that two different secretory processes, i.e., neurosecretory-like (Golgi apparatus — dense-core vesicles) and ependymal-like (granular endoplasmic reticulum — vacuoles containing a flocculent material) may be involved in different regulatory mechanisms in the pinealocytes, the definitive answer to this is still far from clear. Therefore, the pineal gland of the kangaroo rat appears to be a good model for the study of the potential relationship between these two secretory processes, especially in respect to seasonal changes.

    Notes: Summary The most frequently occurring cell types in the pars distalis of the pituitary gland of the rainbow trout, (i) the lactotropic, (ii) the gonadotropic, and (iii) the somatotropic cells, were identified in cryosections. Their morphological characteristics were compared with those of Epon-embedded material. Cell location, cell form, position of the nucleus, arrangement of rough endoplasmic reticulum and sizes of secretory granules proved to be useful parameters for identification. The size distribution of secretory granules of corresponding cells in cryosections and Epon sections proved to be similar. Additionally, both the immunoferritin and the unlabeled antibody enzyme method were applied for the immunocytochemical labeling of gonadotropic hormone-producing cells in cryosections. Anti-salmon-GTH as well as anti-carp-GTH serum showed the presence of GTH in both the smaller and the larger granules of the classical GTH cells, but also produced a reaction in TSH cells. Labelling of TSH cells was absent when using anti-β-carp-GTH. Specificity of the reaction depended upon the degree of dilution of the anti-GTH serum. Results with dilutions of 1∶4,000 and 1∶8,000 in the unlabeled antibody enzyme method, and of 1∶8,000 up to 1∶32,000 in the immunoferritin technique were optimal. Acid phosphatase activity in the smaller granules was demonstrated by enzyme cytochemistry in Epon sections. The relationship of the presence of hormone in these granules is discussed. The high sensitivity of the immunocytochemical labeling procedure is discussed with respect to cryo-ultramicrotomy.

    Notes: Summary The prosobranch Fusitriton oregonensis exhibits an unusual form of sperm polymorphism. The viable, eupyrene sperm are attached in groups of about fifty to worm-shaped, apyrene, carrier sperm. There is a second apyrene sperm, which is lancet-shaped and has a different internal organization than the carrier, but does not transport eupyrene sperm. The eupyrene sperm are filiform (185 μm long), with a conical acrosome, elongate nucleus and midpiece. They contain large stores of glycogen in the principal piece, together with an unusually high proportion of protein. The latter is due to a complex interconnecting system of fibres that supports the tail internally. A distinct annulus is located, characteristically, at the junction between midpiece and principal piece. The carrier sperm has a core of about 112 axonemes that arise from basal bodies in the anterior end and extend through its entire length of 36 μm. The basal bodies have unstriated rootlets that are embedded in a granular cap. Large membrane-bound “yolk bodies” are arranged along the length of the carrier sperm, on either side of the median axonemal core. Dense bodies, which may be indigestible residues formed from the degeneration of the nucleus, are excreted by exocytosis. Individual carrier sperm are capable of “corkscrew” propulsion, resembling that of spirochaetes. The lancet sperm is three times as long as the carrier. The sixteen or so axonemes, which are arranged peripherally like a cage enclosing the cytoplasm, originate from a dense centriolar plate in the anterior end. The cytoplasm is filled with secretions including small yolk granules, dense bodies (also excreted), clear vesicles, and a membranated granular secretion that resembles mucus. The possible functions of the lancet and carrier sperm are discussed.

    Notes: Summary Three-dimensional aspects of smooth muscle cells of the microvas-culature were studied ultrastructurally in laboratory rodents by means of serial thin sections and reconstruction of muscle cell models. It was demonstrated that a muscle cell of an arteriole (luminal diameter (LD) 17 μm) in hamster striated muscle was spindle-shaped, 70 μm long, and wound twice round the vessel axis. The volume of the cell was calculated as 750 μm3 and its surface area as 1330 μm2. A muscle cell in an arteriole (LD 6 μm) in the rat retina was irregular in shape, about 22 μm long, and had branched processes. The cell volume was calculated as 139 μm3 and its surface area as 298 μm2.

    Notes: Summary In the gastric mucosa of two teleost species, the perch (Perca fluviatilis) and the catfish (Ameiurus nebulosus) three endocrine cell types were found, located predominantly between the mucoid cells of the gastric mucosa. A fourth cell type is present in the gastric glands of catfish. Each cell type was defined by its characteristic secretory granules. Type-I cells were predominant in both fish. These cells contained round or oval granules with a pleomorphic core. The average diameter of granules was 400 nm for the perch and 270 nm for the catfish. Type-II cells of both species displayed small, highly osmiophilic granules about 100 nm in diameter. The secretory granules of type-III cells (260 nm in the perch and 190 nm in the catfish) were round or slightly oval in shape and were filled with a finely particulate electron-dense material. Type-IV cells of the catfish were found in the gastric glands only. Their cytoplasm was filled with homogeneous, moderately electron-dense granules averaging 340 nm in diameter. The physiological significance of these different morphological types of gastric endocrine cells requires further investigation.

    Notes: Summary The interstitial cells associated with the myenteric plexus of the rabbit colon were studied by scanning and transmission electron microscopy. It was demonstrated that the interstitial cells were stellate or fusiform in shape and located over the ganglia, over nerve bundles and between muscle cells. They were characterized by many slender processes, and resemble fibroblasts. No basal lamina was observed between the interstitial cells and muscle cells. It was concluded that structural features of the interstitial cells are distinctly different from those of neurons, Schwann cells, or of smooth muscle cells, while they show clear similarities to those of fibroblasts. By scanning electron microscopy the shapes and the relations of these cells could be demonstrated in great detail.

    Notes: Summary The noradrenergic terminals in the substantia gelatinosa of the dorsal horn of the cervical spinal cord of the rat were investigated by means of the histofluorescence technique and electron-microscopic cytochemistry using the glyoxylic acid-KMnO4 fixation technique. In accordance with the topographical distribution of fluorescent catecholaminergic fibers, noradrenergic terminals containing small granular vesicles were frequently observed electron microscopically in the outer layer of the substantia gelatinosa. These terminals were most frequently found to appose without showing typical synaptic features, small-caliber dendrites, spine apparatus, and rarely, large caliber dendrites. Only in a few cases, the noradrenergic terminals exhibited typical synaptic contacts with dendritic elements of small size. In addition, noradrenergic terminals apposed non-noradrenergic terminals containing small agranular vesicles. In rats bearing surgical lesions of the dorsal roots, no noradrenergic terminal were found in contact with the degenerated axon terminals in the substantia gelatinosa. These findings suggest that the noradrenergic afferents to the substantia gelatinosa may exert their influence on sensory transmission via dorsal horn cells.

    Notes: Summary Substance-P immunoreactivity has been located in semithin sections of mouse hypothalami and pituitaries and compared with the distribution of other hypothalamic peptides. In the mouse, nerve fibres and terminals reacting with antibodies against substance P (SP) were detected both in the external zone of the median eminence (ME) and in the neural lobe of the pituitary. Immunoreactive SP (ISP) axons of the ME did not react with antibodies against other peptides, i.e. arginine-vasopressin (AVP), oxytocin (OT), somatostatin and enkephalin, and were also negative with an antibody to serotonin. In the neural lobe, SP immunostaining occurred in AVP but not in OT axons. In the hypothalamus, ISP axons were widespread but conspicuously lacking in areas containing AVP neurones, i. e. in the suprachiasmatic nucleus and the clusters of AVP cells in the SO and PV nuclei. In contrast, multiple ISP endings were seen in contact with OT neurones. Immunoreactive cell bodies, only detected after colchicine treatment, belonged to two distinct classes of neurones: 1) single AVP neurones of the SO and PV nuclei; 2) specific (staining only for SP) neurones, scattered or grouped in different areas of the hypothalamus, not showing relationship with any circumscribed nucleus. These results reinforce the opinion that SP can be released as a neurohormone into the vascular portal system and can directly affect the pars distalis. The presence of immunoreactive SP in the neural lobe, which has not been reported in species other than the mouse, may have a different physiological significance.

    Notes: Summary A monoclonal antibody that recognises the C-terminal part of substance P was used to study immunoreactive structures in the substantia nigra by the unlabeled antibody, peroxidase-antiperoxidase procedure. Immunoreactivity was present in nerve fibres in all parts of the substantia nigra, particularly in the pars reticulata and pars lateralis. Electron microscopically two types of bouton immunoreactive for substance P were found: Type 1 contained large electron-lucent vesicles, occasional large granulated vesicles and formed symmetrical synapses with dendrites. Type 2 boutons contained smaller, round electron-lucent vesicles, many large granular vesicles and formed asymmetrical synapses (having prominent postjunctional dense bodies) with dendrites and perikarya. Immunoreactive fibres with varicosities that had been identified light microscopically were studied in serial sections in the electron microscope. Each identified varicosity contained synaptic vesicles and formed a single synapse. An individual fibre formed boutons of only one kind (type 1 or type 2) and could form multiple synapses with the same neuron. Thus, an identified fibre in the pars compacta had eight varicosities, each of which was in synaptic contacts (type 2) with the dendrites or soma of the same neuron. The results are consistent with the concept that substance P is a synaptic transmitter in the substantia nigra and indicate that neurons in this region may receive a significant input from substance P-containing afferents, and that there are at least two types of such afferent fibres.

    Notes: Summary The cellular localization of progesterone-binding protein (PBP) in the guinea-pig placenta was studied by use of immunocytochemical procedures. Within the chorioallantoic placenta, a strong positive reaction was observed in the interlobar and marginal trophoblast from the third week of gestation to term. PBP was localized in the cytoplasm of the syncytiotrophoblast, and the nuclei were never stained. At the ultrastructural level, the immunoreaction was associated with the rough endoplasmic reticulum, the Golgi apparatus and the perinuclear space. No deposits were seen in any other cell organelles. This localization strongly suggests that the interlobar syncytium is related to PBP synthesis. In the labyrinth, a weak immunoreaction was observed by light microscopy around some blood lacunae. At the ultrastructural level the dense deposits were localized in vesicles located near the maternal lacunae. The distribution of PBP was also studied by light microscopy in other tissues from pregnant guinea-pig. No PBP, or PBP-like material, was detected inside cells from liver, muscle, heart, lung, kidney, ovary, and uterus. A weak immunoreaction for PBP was detected in vascularized zones of these organs. These observations strongly suggest that PBP, a protein related to gestation in the guinea-pig, is elaborated by the placental tissue of this hystricomorph rodent. PBP is the first steroid-binding plasma protein shown to be of extrahepatic origin.

    Notes: Summary We describe the ultrastructural localization of isotocin, vasotocin and neurophysin in the magnocellular preoptic nucleus of the goldfish. With the aid of immunocytochemical techniques, we see staining both in classical neurosecretory granules and in diffuse agranular form throughout somata and processes. Signs of cellular and synaptic interactions between chemically identified neurons include axon terminals which contain vasotocin immunoreactivity and membrane specializations (puncta adhaerentia) between adjacent somata. Our investigations provide an anatomical basis for neuroendocrine and neurotransmitter-like functions of peptidergic neurons in the teleost preoptic nucleus.

    Notes: Summary Immunoreactive luteinizing hormone-releasing hormone (LHRH)-like material has been demonstrated in the pineal gland of the adult rat. The objective of the present study was to examine the ontogenetic development of this LHRH-like substance in the rat pineal with the peroxidase-antiperoxidase (PAP) method of Sternberger. LHRH-like immunoreactive material was not observed in pineal glands of newborn rats. The amount of material increased progressively from the 6th–12th day of postnatal development. On day 12, the amount of LHRH-like immunoreactivity was consistent and comparable in all pineal glands of male and female animals examined.

    Notes: Summary Quantitative studies on the population of the gonadotropic cells in the pituitary of adult male rats were performed after trypsic dissociation of the pituitary glands and immunoenzymatic staining with anti-β-LH or anti-β-FSH antisera. Number, area and extinction of labelled cells were measured by use of an image analyser and a cytophotometer. The gonadotrophs represent approximately 14% of the pituitary cells. The mean area of gonadotrophs is significantly larger after staining with anti-β-LH serum than after staining with anti-β-FSH serum. Planimetric measurement of the gonadotrophs reveals their variability in size ranging between 30 and 160 μm2. Moreover, the size distribution depends on the staining serum used: more numerous small-sized cells (〈75 μm2) are stained with anti-β-FSH serum than with anti-β-LH serum, which conversely stains more numerous large-sized cells. Cytophotometric measurements indicate that immunostaining varies greatly among cells of the same size class and that the staining intensity appears to increase according to the cell size. These results emphasize the morphofunctional heterogeneity of the gonadotropic cell population.

    Notes: Summary An ultrastructural and morphometric analysis was made of the nucleolar components in pinealocytes of 40 male Fischer rats sampled at eight times in an LD 12∶12 photoperiod cycle. Comparisons of results from the eight times showed variation in estimated mean volume of the granular component of ±29%, and of the fibrillar component ±11%, in relation to daily means. Peaks in mean volume of total nucleolus and its granular component occurred at 1 h of light. Near maximal and minimal mean volumes of the fibrillar component both occurred during both light and dark. Fibrillar centers (nucleolar organizer regions) of different sizes were found at all sampling times. It is concluded that temporal patterns in 24-h changes in the nucleolar components are most prominent in the granular component, and are more complex than suggested by changes in total nucleolar size or mean dimensions, and than represented by a simple biphasic circadian rhythm. Examples of different stages in the migration of the granular component, and of possible sites of nucleo-cytoplasmic transfer of nucleolar material, are described.

    Notes: Summary Immuno-electron microscopy was performed on live, cultured, early postnatal cerebellar and retinal cells of the mouse to identify A2B5 antigenbearing elements. In cerebellar cultures, granule cells, some immature oligodendroglia, and astroblasts express A2B5 antigen on their cell surfaces. The typical features of astroblasts include large cisternae of the endoplasmic reticulum and a mixed population of intermediate-sized filaments and microtubules. Immature oligodendroglia cells express the antigen on their cell bodies and on procecesses filled with cytoplasm. Cytoplasm-free membranous whorls, however, are devoid of A2B5 antigen, but not of 0 or NS-1 antigens. In retinal cultures, A2B5 antigen is observed on differentiating neurons with the exception of photoreceptor cells as identified by ribbon synapses.

    Notes: Summary The ultrastructure of Pacinian corpuscles located on the crural interosseous membrane was studied in adult rats 6 h to 10 months after transection of the right sciatic nerve. Axon terminals degenerated one day after transection and were engulfed and resorbed by cells of the inner core within one week. The axial space left after removal of the axonal debris was closed by the lamellae of the inner core. The main structural features of the inner core and capsule remained preserved after denervation throughout the period of study. The denervated inner cores, however, became atrophic 10 months after neurotomy, their mean diameter being reduced by 17.5% compared with that of contralateral control corpuscles. The number of capsular lamellae was unaltered, and perineurial pathways of the peripheral nerve stump remained preserved. Schwann cells proliferated and formed Büngner bands during the first month after denervation, but retracted their processes and became atrophic at later stages after neurotomy. Survival of Pacinian corpuscles after long-term denervation in adult rats is in contrast to their rapid degeneration within several days after nerve section in neonates.

    Description / Table of Contents: Résumé L'ultrastructure du tissu lymphoïde associé au tube digestif (GALT) a été étudiée chez l'amphibien urodèle, Pleurodeles waltlii. Les follicules lymphoïdes se présentent comme de vrais infiltrés entre les éléments conjonctifs de la muqueuse. Ils se trouvent principalement constitués par des plasmocytes mûrs et en développement, des macrophages et des granulocytes. Les cellules lymphoïdes migratrices provoquent une invasion massive de l'épithelium intestinal qui présente des modifications notables comme la disparition de la membrane basale et une diminution du nombre de cellules muqueuses. D'après son organisation et ses composants cellulaires, le GALT de P. waltlii semble représenter un précurseur phylogénétique primitif de la “barrière immunologique intestinale” des mammifères.
    Notes: Summary The ultrastructure of gut-associated lymphoid tissue (GALT) has been studied in the salamander, Pleurodeles waltlii. Lymphoid accumulations appear as true infiltrates scattered throughout the lamina propria cell elements. The most important components of these infiltrates are small and medium sized lymphocytes, and, in lesser amounts, developing and mature plasma cells, macrophages and granulocytes. Migrating lymphoid cells massively invade the intestinal epithelium inducing noticeable modifications, such as the disappearance of the basement membrane and decreased numbers of mucous cells. Thus, in its organization and cell composition, the GALT of P. waltlii appears to represent a primitive phylogenetic precursor of the mammalian “intestinalimmunologic ” barrier.

    Notes: Summary In untreated tadpoles of Xenopus laevis, stage 60 (Nieuwkoop and Faber 1956), somatostatin-immunoreactive perikarya and fiber networks are already discernible in the pallium and the septum, as well as in the anterior and posterior hypothalamus. In addition, somatostatin-immunoreactive cerebrospinal fluid (CSF)-contacting neurons were found in the periventricular gray of the most caudal division of the ventral tuber cinereum. The results are discussed with respect to the inhibitory influence of the system of CSF-contacting and other somatostatin-immunoreactive neurons on the anterior pituitary and other target sites, especially during the climax of metamorphosis.

    Notes: Summary Immunoreactive prolactin (IMP) has been localized in the male rat brain using the soluble peroxidase-anti-peroxidase (PAP) technique. In normal untreated animals, reaction product was seen in choroid plexus (CP) and in ependymal cells of the ventricular lining with heaviest concentrations of positively staining cells in the 3rd ventricle near the subcommisural organ (SCO), in the lateral ventricles near the subfornical organ (SFO), and in the 4th ventricle near the area postrema (AP). IMP was also present in numerous ependymal cells resembling tanycytes in the cerebral aqueduct, central canal of the spinal cord at the level of the AP, the organum vasculosum of the lamina terminalis (OVLT) and the floor of the infundibular recess. Immunoreactive cells resembling neurons were localized within the substance of the AP, SCO, and OVLT. IMP was also present in fibers of the zona externa of the median eminence and infundibular stalk; a few cells of the pars tuberalis contained reaction product. Hypophysectomized rats and bromocriptine-treated rats exhibited a similar staining pattern except that bromocriptine treatment eliminated IMP from most CP cells. Hypophysectomy, bromocriptine or estrogen treatment enhanced staining for IMP in cells of the pars tuberalis; estrogen treatment or hypophysectomy produced an increase in the number and distribution of immunoreactive cells as well as increased density of reaction product in cells of the medial habenular nucleus. The functional relevance of prolactin in these locations in the brain, the possible routes of transport of prolactin from the pituitary gland to the central nervous system, and the strong suggestion of extra-pituitary sites of synthesis of a prolactin-like hormone are discussed.

    Notes: Summary In the kidney of the Syrian hamster the descending thin limbs of both the short and long loops of Henle are not spatially separated from each other and descend between the vascular bundles. Ultrastructurally, five different epithelial types are distinguished in the thin limbs of the short and long loops of Henle. Short loops possess only a descending thin limb with a simply organized epithelium (type 1). Long loops comprise an upper and a lower part of the descending thin limb and the ascending thin limb. The upper part of the long descending thin limb is equipped with a complex and highly interdigitating epithelium with shallow junctions (type 2), which gradually transforms into the simple noninterdigitating type-3 epithelium of the lower part. In a minor portion of long descending thin limbs, however, the upper part begins with an even more complexly organized epithelium (type 2a) than type 2. Type-2a epithelium is conspicuously thicker and possesses a more elaborate mode of cellular interdigitation. Along the descent of this tubular part through the inner stripe of the outer medulla, type-2a epithelium transforms into type-2 epithelium. It is suggested that the long descending thin limbs, which start with type-2a epithelium, belong to the longest loops. The type-4 epithelium of the ascending thin limbs is characterized by flat and extensively interdigitating cells with shallow junctions. The unique pattern of the type-2 a epithelium favors the assumption that solute secretion essentially contributes to the increase in concentration of tubular fluid in long descending thin limbs.

Met-enkephalin (100/μg) injected into anesthetized rats, or 20 μg beta-endorphin administered via a previously implanted cannula to unanesthetized animals, will lead to cell degranulation and often to expanded Golgi zones and prominent regions of rough endoplasmic reticulum in secretory cells when tissue is fixed 45–60 min after peptide administration. Treatment of animals with the opiate antagonist naloxone hydrochloride prior to enkephalin injection appeared to prevent the cellular changes elicited with peptide alone. Observations suggest that opiate peptides administered to the cerebrospinal fluid may stimulate release of pro-opiomelanocortin-peptide from pituitary cells.

    Notes: Summary The ovarian surface and associated germ cells have been studied in human fetuses from 12 weeks of age until near term, using light, TEM and SEM techniques. The surface epithelium and related cords proliferate extensively, especially at midterm. The cords in the ovarian cortex appear to be linked with ingrowths from the surface epithelium, and both structures have a common basal lamina. Germ cells are always interspersed among the somatic cells of the surface epithelium and associated cords. These results indicate that both the proliferating cords and surface epithelium may contribute to the formation of early follicles. Furthermore, the occurrence, of elements having some of the features of primitive steroidogenic cells in the regions of cordsurface epithelium continuity, suggests that both structures (surface epithelium and cords) contribute somatic cells, which in addition to becoming granulosa cells, might also contribute to the provision of primitive interstitial cells. Gonocytes tend to migrate through the developing ovarian tissue towards the surface where they become extruded into the peritoneal cavity. This phenomenon might contribute to the reduction in the number of germ cells at birth and parallels the atretic processes within the ovary.

    Notes: Summary The ultrastructural changes induced by the topical application of retinol acetate on hamster cheek pouch epithelium were evaluated using stereological analysis. Electron micrographs were prepared of the basal and superficial regions of the nucleated cell layer of the epithelium obtained from 3 treated and 3 control animals and examined at two levels of magnification. A total of 528 micrographs were analyzed using a coherent double lattice test system. Although the mean thickness of the nucleated cell layer did not change significantly after 10 days of treatment with retinol acetate the formation of keratinized squames was completely inhibited. This was paralleled by significant changes in the volume density of a number of organelles in both the basal and superficial strata. Rough endoplasmic reticulum increased significantly whereas filaments, which maintained a constant diameter of approximately 9 nm, keratohyalin granules and membrane-coating granules decreased in both strata. Desmosomes also showed a significant decrease in numerical area density in the treated tissues. In contrast, no changes were observed in the volume density of the Golgi apparatus, free ribosomes or mitochondria in the treated epithelium. It is concluded that this treatment provides an epithelium lacking all features of keratinization and may be a useful model for examining metabolic activities specifically associated with keratinization.

    Notes: Summary In this study we examine the fine structure of mechanosensory hairs in the antennule of crayfish. The sensory hair is a stiff shaft with feather-like filaments. The hair's base is a large expansion of membrane which allows the hair shaft to deflect. The sensory transducing elements are located far from the hair, but are coupled mechanically with the hair shaft by a fine extracellular chorda. The sensory element is a type of scolopidium which consists of a scolopale cell and three sensory cells with a 9 + 0 type ciliary process. This type of scolopidium is characteristic of the chordotonal organ that has no cuticular structure on the surface of the exoskeleton. In this crustacean hair receptor, the deflection of the cuticular hair is transmitted through the chorda to the scolopidium which is a tension-sensitive transducer. The present study reveals that the mechanosensory hair of decapod crustaceans is a chordotonal organ accompanied by a cuticular hair structure. We also discuss comparative aspects of cuticular and subcuticular chordotonal organs in arthropods.

    Notes: Summary Electron microscopy reveals that sarcolemmata of adjacent muscle cells form pentalaminar junctions by fusion of apposed trilaminar double leaflet membranes. These junctions appear to be candidates for low resistance pathways between muscle fibers. The muscles depolarize slowly when bathed in solutions containing elevated concentrations of KCl, and the sucrose gap method can then be used to measure the potential difference between polarized and depolarized regions. Thus the junctions which we have observed may provide the structural basis for electrical transmission through the sucrose gap.

    Notes: Summary The location of LHRH-containing neuronal elements was investigated in the domestic fowl by means of immunohistochemical techniques. LHRH antisera were raised against synthetic LHRH in the rabbit. The antiserum used in the present study cross-reacted with LHRH of mammalian and avian tissues. LHRH-immunoreactive perikarya are located in the preoptic and in the septal areas, and in the bulbus olfactorius; however, no LHRH-immuno-reactive perikarya were found in the tuberal part of the hypothalamus. LHRH-immunoreactive fibers course from these areas toward the median eminence mainly along the wall of the third ventricle in the form of a periventricular network. Originating from the same cell groups other fibers run caudally immediately above the optic chiasma, forming the median bundle of the tractus preoptico-infundibularis. The third bundle running toward the OVLT is named the tractus preoptico-terminalis. In addition to these structures, LHRH-containing fibers and terminals were also present in different regions of the limbic system, in the dorsal part of the hippocampus, in the tuberculum and bulbus olfactorius, as well as in the optic lobe, nuclei commissurales tectales, organon subcommissurale, periaqueductal area, and pars ventralis mesencephali. The general distribution of the LHRH system in the chicken corresponds principally to that described previously in rodents (Sétáló et al. 1976, 1978). However, some subtle differences were demonstrated between the location of the LHRH system in birds and mammals.

    Notes: Summary The hypothalamo-extrahypophyseal neurophysin pathways (HEH) and the three hypothalamic nuclei secreting neurophysins, the supraoptic (SON), paraventricular (PVN) and suprachiasmatic (SCN) nuclei, of normal and hypophysectomized rats were studied by application of the immunoperoxidase procedure. Eight well-defined HEH pathways were recognized. Their main sites of projection were: lateral septum and subfornical organ (1 and 2); tractus diagonalis (3); medial nucleus of the amygdala and lateral ventricle (4); nucleus periventricularis thalami, nucleus habenulae lateralis and periaqueductal gray (5); periaqueductal gray, pineal organ, collicular recess and subependymal region of the fourth ventricle (6); dorsomedial nucleus and premammillary area (7); perimammillary region, corpus trapezoideum, ventral surface of medulla oblongata, nucleus tractus solitarii, nucleus commissuralis, substantia gelatinosa and formatio reticularis lateralis of the medulla oblongata and spinal cord (8). Neurophysin fibers of unknown origin were found in the frontal cerebral cortex. It was noted that in pathway 5 the amount of immunostainable material undergoes changes with age. The three neurophysin-secreting nuclei reacted differently following hypophysectomy. Among the HEH pathways the only one that seemed to be affected by hypophysectomy was that innervating the lateral septum. It is suggested that the neurons that survive hypophysectomy either do not project to the neural lobe or, alternatively, display axon collaterals projecting outside the neural lobe. Such a neuronal population could be the origin of the HEH pathways.

    Notes: Summary Microinjection of adenine nucleotides and substrates into the cytoplasm of Amoeba proteus followed by EM examination has been used in an attempt to relate alterations in mitochondrial morphology with functional changes. Contracted mitochondria with dark matrix and wide cristae (Type I), and expanded mitochondria with light matrix and narrower cristae (Type II) coexist in normal active amoebae, but their numbers can be varied according to different cell activity states. Following injection of ATP, the mitochondria of the amoebae showed a time-dependent movement towards a predominately Type II form, whilst injections of ADP produced predominately the Type I form. Injection of succinate or deionised water, even in large amounts, had little effect on the numbers of Type I or Type II forms. The change induced by ATP was of long duration; that induced by ADP was influenced by both concentration injected and the cell's substrate levels. With 3 mM solutions of ADP the mitochondrial population was primarily of Type I organelles; higher ADP concentrations or the simultaneous injection of succinate, however, resulted in a switch with time to increased proportions of Type II mitochondria. The results extend the findings of previous in vivo and in vitro mitochondrial studies and are discussed in the light of these.

    Notes: Summary The primordial germ cells (PGCs) of Oryzias latipes in migration to the gonadal anlage have been investigated by light and electron microscopy. The ultrastructure of the PGCs, which occur in the subendodermal space on the syncytial periblast, differ conspicuously from that of the surrounding endodermal cells. After the PGCs move to the cavity between lateral plate and ectoderm, they are taken into the somatomesodermal layer and transferred to the dorsal mesentery where they form gonadal anlage with mesodermal cells. During their translocation to the dorsal mesentery through the somatic mesoderm, apparently without formation of pseudopods, the PGCs are completely surrounded by mesodermal cells. Since these conditions seem unfavorable to the active translocation of the PGCs to the dorsal mesentery, it is more likely that the PGCs are transferred passively by the morphogenic activity of the lateral-plate mesoderm. Counts of the number of the PGCs revealed that they are mitotically dormant during the migratory period. After the completion of the migration, they regain their proliferative activity. The PGCs in the female proliferate more actively than those in the male, which provides the first morphological indication of sex differentiation in this species of fish.

    Notes: Summary The early differentiation of the optic pathway of the trout was studied by means of autoradiography, silver impregnation and electron microscopy. Ingrowth of optic nerve fibers into the optic tectum was consistently shown by tracer application and Golgi studies to occur at stage 28, about one week before hatching. Fibers being arranged in discrete bundles were rapidly growing through the longitudinal axis of tectum and at stage 33 reached its posterior end. Cross sections of these fiber bundles at different positions revealed myelin ensheatment to be initiated at the end of stage 34 at the anterior pole of the tectum. Since in the optic nerve of the trout the onset of myelination occurred even earlier (stage 33), it is assumed that this differentiation process follows a rostro-caudal gradient during development of the optic pathway.

    Notes: Summary Several environmental factors influence the growth of the basommatophoran freshwater snail Lymnaea stagnalis. Growth is hormonally controlled by 4 cerebral clusters of ca 50–75 peptidergic, neuroendocrine Light Green Cells (LGC). The present light, transmission, and scanning electron microscopic study shows that the LGC are synaptically contacted by a tentacle sensory system (TSS). The TSS consists of 2 types of primary sensory neurone, viz. ca 150 S1-cells and ca 50–100 S2-cells. A S1-cell has a non-ciliated dendrite and an axon branch that synaptically contacts the soma of a S2-cell. A S2-cell has a branching, ciliated dendrite. Probably, S1- and S2-cells have different sensory modalities and can integrate sensory information by intersensory interaction. The S2-axons run through the tentacular nerves, the cerebral ganglia, and the intercerebral commissure. In each ganglion S2-axons branch and form synaptic contacts on the axons and somata of the LGC and on glial cells that surround the LGC. In an LGC-cluster, 1–3 LGC-somata are particularly strongly innervated. Probably, the TSS is involved in the environmental control of growth in L. stagnalis.

    Notes: Summary The indirect immunoperoxidase method was used to identify albumin in hepatocytes of rats before and after periods of starvation. All hepatocytes in fed rats contained a relatively large amount of nascent albumin. Overnight fasting reduced the number of hepatocytes with a large amount of albumin to primarily those surrounding terminal hepatic venules. These were estimated to be about 30% of the population. The other cells had only a slight amount of albumin. After 48 h of fasting all hepatocytes contained a low level of albumin.

    Notes: Summary A peritrophic membrane (PM) has been demonstrated in the gut of feeding larvae, nymphs, and adults of the tick Ixodes dammini. This is the first report of a PM in ticks. This temporary structure divides the lumen of the gut into two compartments, an endoperitrophic space, the lumen proper, and an ectoperitrophic space located between the PM and the epithelial cells of the gut wall. The PM is a mechanical barrier and even such small particles as ribosomes derived from ingested reticulocytes are retained in the lumen proper; they are never found in the ectoperitrophic compartment. In Ixodes dammini fed on hamsters infected with Babesia microti some of the parasites are found in the ectoperitrophic space. This passage is accomplished by a highly specialized organelle, the arrowhead, which develops in some Babesia during their metamorphosis in the gut of the vector. The arrowhead, while passing through the PM, changes its fine structure and loses its internal organization as if releasing some of its contents. Its disintegration continues and it disappears shortly after the Babesia have entered the epithelial cells. Only Babesia equipped with the arrowhead structure are able to cross the PM. This is the first documented case of a parasite traversing a solidified PM.

    Notes: Summary In order to test the premise that non-adrenergic, non-cholinergic (NANC) autonomic nerves have a distinctive ultrastructural appearance, clearly different from that of cholinergic nerves, a detailed quantitative ultrastructural analysis has been made of the non-adrenergic innervation of 15 tissues thought from pharmacological evidence to be innervated by NANC nerves (rat and rabbit anococcygeus muscles; rabbit hepatic portal vein; extrinsically denervated toad lung); cholinergic nerves (atria of rat, rabbit, guinea-pig and toad); or both (guinea-pig cervical and thoracic trachealis muscle; rabbit rectococcygeus muscle; urinary bladder of rat, rabbit, guinea-pig and toad) in addition to their adrenergic supply. Following fixation with a modified chromaffin procedure allowing identification of adrenergic nerves, large, randomly selected samples of non-adrenergic nerve profiles from each tissue were analysed with respect to numbers, relative proportions, and size frequency distributions of different vesicle classes within the profiles. The neuromuscular relationships within each tissue were also analysed. On the basis of these analyses, it is clear that there are no consistent ultrastructural differences between cholinergic and NANC autonomic nerves: neither proportions nor sizes of the vesicles provide any clue as to the transmitter used by a particular nerve. The great majority of nerve profiles, whether cholinergic or NANC, contain predominantly small clear “synaptic” vesicles. Large filled “peptidergic” vesicles are no more common in most NANC nerves than in most cholinergic ones. It is concluded, on ultrastructural grounds, that the primary transmitter in these NANC autonomie nerves is most likely to be stored in and released from the small clear vesicles.

    Notes: Summary The distribution of luteinizing hormone-releasing hormone (LHRH)-immunoreactive perikarya and processes was examined, in the untreated rat, with the unlabeled antibody enzyme method of immunocytochemistry on thick 50 μm vibratome sections. LHRH neurons were primarily observed in the preoptico-anterior hypothalamic and septal areas. Projections from these cell bodies to the median eminence form three distinct pathways, one laterally along the course of the optic tracts, one medially through the periventricular stratum of the third ventricle, and one through the tractus infundibularis. In addition, some of these cell bodies project to the organum vasculosum of the lamina terminalis (OVLT) and the subfornical organ (SFO). LHRH immunoreactive neurons were also noted in the anterior olfactory regions; they project along the medial olfactory tract to the olfactory bulb.

    Notes: Summary Pigment of tail-fin melanophores in periodic albino Xenopus laevis tadpoles is dispersed in response to darkness and to α-MSH in a manner similar to wild-type melanophores. However, periodic albino tadpoles lack the response to different background conditions and the melatonin-induced aggregation in darkness. The tyrosinase activity in cells of the latter type tadpoles is weak compared to the wild-type cells. Ultrastructural examination of melanophores from periodic albino mutants and cells from wild-type tadpoles shows similar organelles at corresponding sites. A morphological difference can be observed in the fine structure of the melanosomes, which in albinos resembles an earlier stage of development. It is postulated that periodic albino Xenopus laevis possess the cellular mechanism to disperse pigment in the melanophores, but that under physiological conditions the release of α-MSH appears to be absent or scarce.

    Notes: Summary Scanning electron microscopy of various regions of the body of the marine gastropod Pleurobranchaea californica (McFarland) has revealed a characteristic cell type that bears cilia with dilated discoid-shaped tips. The tips of the cilia consist of an expansion of the ciliary membrane around a looped distal extension of the axoneme. These kinocilia have been observed in numerous other marine invertebrates and are generally referred to as paddle cilia (Tamarin et al. 1974) or discocilia (Heimler 1978). Although many functions have been proposed for paddle cilia, little empirical evidence supports any of the proposals. In Pleurobranchaea we have found that the distribution of this ciliated cell type corresponds exactly to areas of the body known from behavioral studies (Lee et al. 1974; Davis and Matera 1981) to mediate chemoreception. Transmission electron microscopy of the epithelium lining the rhinophores and tentacles of Pleurobranchaea revealed details of the ultrastructure of these ciliated cells and showed that they are primary receptors. These ciliated receptors lie in a yellow-brown pseudostratified columnar epithelium that superficially resembles the olfactory mucosa of vertebrates.

The PT forms a cortical layer especially conspicuous around the anterior tip of the PD in some species (Trionyx, Kinosternon, Sternotherus), or it forms a thick dorsal layer of tissue irregularly extending onto the sides of the PD in others (Pseudemys, Chrysemys, Lepidochelys, Chelonia). Immunocytochemical studies using unlabelled second antibody and peroxidase-antiperoxidase reveal that in turtles of all ages, the PT tissue allied with the PD (the PTinterna) is composed primarily of cells containing glycoprotein hormones (FSH, LH and TSH), especially the gonadotropins. The juxPT, however, consists mainly of secretory cells unstained by the antisera tested and includes only a small number of gonadotropes and thyrotropes. Although usually widely distributed in the testudinate adenohypophysis, the great majority of gonadotropes and thyrotropes present in the hatchling are in the PTinterna. It is probable that a concentration of these cells in the PTinterna is widespread among vertebrates. In all turtles examined, lactotropes occur principally in the anterior and ventral part of the PD proper; somatotropes are posterior and dorsal. Corticotropes are concentrated as the lactotropes in the anterior PD, but some are also scattered throughout the posterior half of the gland. Lactotropes, corticotropes, and with a few exceptions, somatotropes, do not occur in PT tissue in the turtle.

    Notes: Summary The ultrastructure of the seminiferous tubules was studied in rats that had been subjected to whole body irradiation on the 19th day of gestation. The seminiferous tubules from 3 months-old irradiated animals are devoid of germ cells and contain only Sertoli cells. Compared with controls of the same age, the seminiferous tubule basal membrane is thickened and multilayered and several alterations are observed in the Sertoli cells. The most characteristic of these alterations are: (a) an abnormal number of nuclear heterochromatin clumps, (b) the presence of numerous cytoplasmic vacuoles and various sized lipid droplets, (c) elaborate interdigitations and junctions between adjacent cells, and (d) the presence of anomalous ectoplasmic specializations disposed perpendicularly to the Sertoli cell membrane.

    Notes: Summary The possible biogenesis of two pigment granule types present in the monochromatic, brown chromatosomes enveloping the ventral nerve chord of the freshwater palaemonid shrimps Macrobrachium acanthurus, M. heterochirus and M. olfersii is examined by transmission electron microscopy in thin section and freeze fracture replicas. Prominent, membrane limited granules are suggested to have their origin in a complex, juxtanuclear, smooth endoplasmic reticulum labyrinth, continuous with the nuclear envelope. Amembranous, lipocarotenoid granules possibly derive from the external surface of the smooth endoplasmic reticulum. Nuclear envelope and SER membranes contain numerous 11 nm diameter intramembranous particles while pigment granule membranes exhibit fewer particles. A dictyosomal origin for the lipocarotenoid granules is discounted. Granulogenesis is suggested to be a continuous process in crustacean chromatophores.

    Notes: Summary Ultrastructural and stereological assessment of the mature avian anterior latissimus dorsi (ALD) muscle showed that it contains two kinds of extrafusal fibers. This fine structural dichotomy of fiber types in the ALD correlated well with their previously reported histochemical duality. Distinct differences occur in sarcomere banding, myofibrillar area, sarcotubular and mitochondrial density, and in morphology of motor-nerve terminals. Both myofiber types in this muscle were interpreted as representing varieties of “slow” or tonic muscle fibers. Both fibers contain myofibrils that, despite differences in cross-sectional area, were large, irregular, and ribbon-shaped, typical of the “Felderstruktur” appearance of true “slow” fibers. Whereas the majority of fibers (type-1) are devoid of well-defined M-bands, the minor fiber population (type-2) exhibit prominent M-bands in the center of each sarcomere. In addition, type-1 tonic fibers contain a significantly lower mitochondrial and sarcotubular volume than the tonic fibers of type-2. While both fiber types exhibit motor-nerve terminals that are small, smooth and punctate in appearance, those on the type2 fibers often had a number of shallow postjunctional folds. Whether or not these two classes of extrafusal fiber in this muscle represent two separate and distinct types of motor units remains to be determined functionally.

    Notes: Summary The ultrastructure of the pineal parenchymal cells, pinealocytes and glia-like cells, of the Chinese hamster (Cricetulus griseus) is described. Pinealocyte nuclei contain spindle-shaped inclusions consisting of bundles of closely packed parallel filaments of about 8 nm in thickness. Mitochondria contain lamellar and tubular cristae. Smaller and larger mitochondria are usually localized separately in individual pinealocytes. Flattened mitochondria contain two to four closely arranged, unusual lamellar cristae, which lie parallel to the surface of the mitochondria. These cristae exhibit particular structures. Rough endoplasmic reticulum is frequently found in close association with such flattened mitochondria. Tubular expansions of the outer membrane of mitochondria occur frequently. The pinealocyte cytoplasm contains stacks of flattened cisternae, which are continuous with both smooth and rough endoplasmic reticulum. Pinealocyte cell bodies and processes contain granulated vesicles. Although larger granulated vesicles measuring up to 300 nm in diameter occasionally occur, smaller granulated vesicles (about 100 nm in diameter) and larger ones are usually localized separately in individual cells. Vacuoles containing flocculent material are found in groups in close association with rough endoplasmic reticulum. Glia-like cells with darker cytoplasm and nuclei lie surrounding pericapillary or intercellular spaces. The glia-like cells are characterized by the presence of abundant lipid droplets and occasional pigment granules. Bundles of filaments run parallel to the long axis of the processes of the glia-like cells.

    Notes: Summary Substance P-immunoreactive nerve terminals were found in several locations in the anterior segment of the rabbit eye. In the iris they occurred in the sphincter muscle and were randomly distributed in the iris stroma with some fibres running close to the dilator muscle. In the ciliary body these immunoreactive elements were few and occurred within bundles of nerve fibres, while in the ciliary processes they were more numerous with a predominantly subepithelial location. Blood vessels in the anterior uvea were often surrounded by substance P-immunoreactive fibres. No substance P-fibres were found in the cornea, while the sclera contained very few such elements. Using conventional in vitro techniques it was found that the sphincter pupillae muscle of the iris responded to electrical stimulation with a contraction that was resistant to cholinergic and adrenergic blockade, but was inhibited by the neuronal blocker tetrodotoxin. This indicates the existence of a non-cholinergic, non-adrenergic neuronal mediator of the contractile response. Exogenously applied substance P produced a long-lasting contraction of the spincter muscle, an observation compatible with the view that substance P is the noncholinergic, non-adrenergic neurotransmitter involved.

    Notes: Summary The ultrastructure of gastrin cells in the rat antrum was analyzed with standardized and quantitative planimetric methods. Resting and active cells were compared. The gastrin cells were activated by removal of the acidproducing part of the stomach (fundectomy). As a result the serum gastrin concentrations were greatly elevated. Compared with gastrin cells in fasted control rats the gastrin cells in fundectomized rats were increased in number, contained fewer cytoplasmic granules, increased amount of endoplasmic reticulum, and an enlarged Golgi area. Generally, the secretory granules of the gastrin cell displayed a wide range of electron density from highly electron-dense to electron-lucent. They exhibited certain characteristic features: 1) Electron-dense granules made up a greater proportion of the total granule population in active gastrin cells than in resting cells. 2) Electron-dense granules were more frequent near the Golgi stacks than in the periphery of the cell. 3) Electron-dense granules were smaller in size than the electron-lucent granules; hence, small electron-dense granules probably represent young granules (progranules), while large, electron-lucent granules represent mature (old) granules. 4) Electron-dense granules invariably displayed a more intense immunoreactivity than electron-lucent granules. The gastrins are generated from a large precursor molecule. The posttranslational processing of this precursor is reflected in the gastrin-component pattern. The gastrin-component pattern in antral extracts of fundectomized and normal fasting rats differed in that the proportion of the gastrin-4-like component was reduced, whereas the gastrin-34-like component was increased in the fundectomized rats. The results suggest a greater proportion of small gastrin components in the mature granules than in the newly formed ones, presumably due to more extensive conversion of larger forms into smaller forms with a longer granule half-life. As a result gastrin-17-and gastrin-34-like components make up a larger proportion of total gastrin in active gastrin cells than in resting gastrin cells.

    Notes: Summary Normal diploid cells require serum to proliferate in culture. Platelet-derived growth factor has been identified as the main serum component responsible for this effect. Here, smooth muscle cells were isolated enzymatically from the aorta of 5-day-old rats and cultured in the presence of 10% whole blood serum (WBS) or plasma-derived serum (PDS), i.e. with or without platelet factor, and studied by transmission electron microscopy. The cells proliferated actively in WBS-medium but remained quiescent in PDS-medium. Fine structurally, cells from WBS-cultures demonstrated numerous mitochondria, an extensive rough endoplasmic reticulum (RER), a large Golgi complex, a few lysosomes, and microfilaments arranged in parallel bundles. After transfer to PDS-medium, the RER- and Golgi cisternae were markedly dilated and the number of membrane-associated ribosomes decreased. Segregation of fragments of cytoplasm within autophagosomes was frequently observed and the number of lysosomes increased. Lipid droplets were more abundant and often gathered in the Golgi area. Moreover, the cells had become more irregular in shape and showed many bleb-like processes at their surface. Microfilament bundles had also become more prominent and crossed each other in different directions. These observations show that the removal of platelet factor from the medium clearly modifies the fine structure of cultured smooth muscle cells. The findings are in good agreement with the concept that platelet factor not only supports the proliferation of cultured cells but also stimulates their secretory activity.

    Notes: Summary A new isolation and purification procedure for endothelial cells of the rat liver and the conditions for large scale survival of these cells in maintenance culture are reported. Cells isolated by this new method and cultured with homologous rat serum on a collagen matrix show the restoration of several ultrastructural characteristics typical of rat liver endothelial cells in situ, including the broad cytoplasmic extensions that contain the sieve plates. These fenestrated cytoplasmic projections, which cover the liver sinusoids in vivo, are well preserved and are reformed in a manner reminiscent of the situation in situ. Reformation of specific membrane receptors is indicated by the reappearance of the capacity to take up horseradish peroxidase by adsorptive endocytosis, a characteristic that is lost during the cell isolation procedure. From the results obtained in this study, maintenance culture of rat liver endothelial cells seems to be a promising system for studying the regulation of pore size of the fenestrated sieve plates by alcohol and certain hormones, for studying the interaction of endothelial cells with other liver cells and tumor cells, and for studying the mechanisms of adsorptive endocytosis.

    Notes: Summary In embryos of the domestic mallard, domestic fowl, and Japanese quail vasotocin-, mesotocin-, luliberin (LHRH)-, met-enkephalin-, cortico- tropin-, and somatostatin-immunoreactive perikarya and fiber formations were visualized at different incubation stages by means of the PAP technique (Sternberger 1979). The most striking results were: (1) Vasotocin-, mesotocin-, and luliberin-immunoreactive systems display, up to the late embryonic period, morphological features most probably related to a neurohormonal function. (2) Met-enkephalin immunoreactivity appears very late during embryonic life; it is restricted to fiber networks and not found in perikarya. (3) Corticotropin immunoreactivity is observed in the tuberal region temporarily at the end of the second and the beginning of the last third of the incubation period. (4) Somatostatin-immunoreactive material is present (i) at the end of the first third of incubation, in association with the olfactory system; (ii) during the same period, adjacent to thin-layered portions of the roof of the brain; (iii) shortly thereafter, in cells of both pancreatic primordia and thyroid gland; and (iv) onward from the middle of the incubation period, in a mesencephalic cell group. The striking difference, in the early embryo, between the mature somatostatin system and the immature character of the surrounding tissues may indicate that somatostatin plays a role in the development of the brain, as well as the pancreas, and the thyroid gland.
