    Notes: The cation [Ge,C,H]+ has been generated by the electron ionisation of trichloromethylgermane. Collisional activation experiments were used to establish a Ge-C-H connectivity in this species, for which a significant fraction of the ion population was found to survive neutralisation-reionisation mass spectrometry (NRMS) experiments. Thus, the neutral counterpart [Ge,C,H]0 is stable on a microsecond timescale. Becke's 3 parameter hybrid density functional (B3LYP) was used to map the ion and neutral potential-energy surfaces, in conjunction with double-zeta and triple-zeta basis sets. The computational results obtained using the triple-zeta basis sets suggest that, for the cation, the global minimum is the high spin 3Σ GeCH+, with the first Ge-C-H excited state, 1Σ GeCH+, approximately 39 kcal mol-1 less stable. The lowest energy ion structure with H-Ge-C connectivity is bent (3A′′HGeC+,∠H-Ge-C = 126.3) and 69 kcal mol-1 less stable than the global minimum. For the neutral, a doublet (2π) with Ge-C-H connectivity is predicted to be the global minimum. The classical barrier for the neutral 1,2-hydrogen shift reaction on the doublet surface is negligible (0.1 kcal mol-1), while the smallest barrier for the cation is 13.0 kcal mol-1, corresponding to (3A′′) HGeC+ → (3Σ) GeCH+. Natural bond order analysis has been used to establish the order of the metal-carbon bond for selected states of both the neutral and the ion. Neutral and cationic isomers with Ge-C triple bonds were found to be high-energy excited states, with the metal-carbon bonds in the cation and neutral ground states of order 2.0 and 2.5, respectively. The instability of Ge-C triple bonded species is attributed to the energy required for electronic promotion in the metal in order to achieve a hybrid configuration suitable for the formation of such a bond.

    Notes: The dark blue, air- and moisture-sensitive, thermolabile tristannaallene R*2Sn=Sn=SnR*2 (5) (R* = SitBu3) is prepared by reaction of Sn(OtBu)2 or Sn[N(SiMe3)2]2 with R*Na in pentane/benzene at -25°C. The dark red-brown, air-sensitive, moisture-insensitive, and thermostable cyclotri-stannene R*4Sn3 (6) with a -R*Sn=SnR*- moiety as part of a Sn3 ring is obtained from the reaction of Sn(OtBu)2 or Sn[N(SiMe3)2]2 with R*Na in pentane at 25°C or from the isomerization of 5 at room temperature (τ1/2 = 9.8 h). According to the result of X-ray structural analyses the Sn3 framework of chiral 5 is bent (156°) and the terminal Sn atoms have pyramidal surroundings. The SnSn double bonds in 5 (2.68 Å) are shorter than those found for the hitherto structurally investigated distannenes (2.77-2.91 Å). Even shorter is the double bond in 6 (2.59 Å). The unsaturated Sn atoms here have nearly planar surroundings in perfect analogy to the carbon atoms in CC double bonds. The SnSn double bond in 6 can therefore be considered as the first “true” Sn=Sn bond. The structures of 5 and 6 can be deduced also from 119Sn- and 29Si-NMR studies in solution.

    Notes: The new chiral ligands 2-butyl-8-chloro-1-(4,8-di-tert-butyl-2,10-dimethoxy-5,7-dioxa-6-phosphadibenzo[a,c]cyclohepten-6-yl)-1,2-dihydroquinoline (BIPHENPHOSHQUIN, 3) and 2-butyl-8-chloro-1-(3,5-dioxa-4-phosphacyclohepta[2,1-a;3,4-a′]dinaphthalen-4-yl)-1,2-dihydroquinoline (BINAPHOSHQUIN, 4; 4a: SaRC, 4b: SaSC) have been synthesized starting from the rigid backbone of 8-chloroquinoline. The reactions of 3 and 4 with rhodium(I), palladium(II), and platinum(II) substrates are reported. The reaction of 3 with [Rh(CO)2Cl]2 in a 2:1 molar ratio in hexane afforded a binuclear chloro-bridged rhodium(III) species by intramolecular oxidative addition of the C-Cl bond of ligand 3 across the rhodium(I) centers. The rhodium(III) complex 5, incorporating the enantiomers SaRC-3 and RaSC-3, has been fully characterized by X-ray diffractometry. Reactions of [Pd(PhCN)2Cl2] with the ligands 3 and 4a in 1:2 molar ratio in toluene afforded the products cis-[Pd(3)2Cl2] (9) and cis-[Pd(4a)2Cl2] (10). Similarly, reaction of [Pt(COD)I2] with 4a in a 1:2 molar ratio afforded the complex cis-[Pt(4a)2I2] (11). An X-ray analysis has been carried out to obtain information on the effect of the ligand 4a on the overall structure of 11.

    Notes: The synthesis and characterization of homoleptic zinc(II), cadmium(II), and mercury(II) thiolates derived from 4,4-dimethyl-2-phenyloxazoline (1) are described. The disulfide [Ox2S2 (3), Ox = 2-(4,4-dimethyl-2-oxazolinyl)phenyl] is prepared by the oxidation of lithium arenethiolate OxS-Li+ (2), and structurally characterized. The disulfide 3 is chiral and enantiomerically pure in the solid state. The thiolate derivatives, M[S(Ox)]2 [M = Zn (4), or Cd (5)], are prepared in good yield by the metathesis reactions of MCl2 with lithium arenethiolate 2. The mercury complex 6 is synthesized by treating 3 with elemental mercury. The complexes are quite stable and highly soluble in common nonpolar organic solvents. X-ray diffraction results show that the zinc thiolate 4 and cadmium thiolate 5 are monomeric in the solid state. The geometry around the metal ion in these two complexes is found to be distorted tetrahedral. The zinc complex is chiral and has crystallized in two different crystalline modifications. The single crystal structures have been determined for both the crystalline modifications, a monoclinic modification (4a) and an orthorhombic one (4b). The crystal structure of 4a shows that the complex is “helically” chiral and enantiomerically pure whereas the crystal structure of 4b shows the modification to be racemic. The spontaneous splitting of the racemates, which is responsible for the isolation of optically active compound, indicates that there should be some solubility differences between the racemates and pure enantiomers. 1H- and 13C-NMR measurements indicate that the complex 4 retains its “helical” structure in solution.

    Notes: The chiral 1,2,3,4-tetrahydroisoquinoline intermediates in the Rice and Beyerman routes to morphine, (+)-(R)-1-(3-hydroxy-4-methoxybenzyl)-6-methoxy-1,2,3,4-tetrahydroisoquinoline (6) and (+)-(R)-1-(3,5-dibenzyloxy-4-methoxybenzyl)-6-methoxy-1,2,3,4-tetrahydroisoquinoline (5), were prepared in high ee by ruthenium-catalyzed asymmetric transfer hydrogenation of the corresponding imine precursors (Noyori method). The yield of the key raw material in the Beyerman route, 3,5-dibenzyloxy-4-methoxyphenylacetic acid (1), starting from gallic acid methyl ester (7) was improved by a factor of 5 over previously described syntheses. Key steps in the new procedure are the selective formation of methyl 3,5-dihydroxy-4-methoxybenzoate (9) via the 3,5-diacetate and an improved benzylation of the hydroxyl groups in 9.

    Notes: -Peptide Nucleic Acids (PNAs) are DNA mimics in which the deoxyribose phosphate backbone has been replaced by a pseudo-peptide skeleton composed of N-(2-aminoethyl)glycine units; they bind to complementary DNA strands with high affinity and selectivity. In order to study the effect of stereogenic centers within the backbone on PNA preorganization and DNA binding properties, chiral PNA decamers were synthesized which contained thymine monomers derived from L-Leu and D- or L-Lys inserted either at C-terminus and/or in the middle of an achiral PNA strand. PNAs containing three chiral thymine monomers derived from L-Leu, D- or L-Lys, L-Asp, or D-Glu were also synthesized. CD spectral analyses showed that a charged chiral monomer inserted in the middle of the strand is able to induce a strong preference in the helix handedness of a PNA-PNA duplex. The effect is increased by the presence of three chiral charged monomers. The L-Lys- and L-Asp-PNAs induced a preference for the left-handed and the D-Lys and D-Glu-PNAs for the right-handed conformation. As expected, the PNA-DNA duplexes are dominated by the DNA strand and thus are right-handed with both D- and L-PNAs. However, the D-PNAs, being inherently right-handed, lead to more stable PNA-DNA duplexes than the L-PNAs. The lysine-based PNAs form more stable complexes with the DNA at low ionic strength, due to the electrostatic interactions between the charged lysine side chain and DNA.

    Notes: -The direct cyclization of 2-amino-4-chloro-3,3-dimethylbutanenitrile with potassium tert-butoxide in THF afforded 1-amino-2,2-dimethylcyclopropane-1-carbonitrile and a dimerization product. Various new cis- and trans-1-(tert-butylamino)-2-benzyl-2-methylcyclopropane-carbonitriles and© the corresponding cyclopropanecarboxamides have been synthesized, with focus on the isolation of the pure stereoisomeric cyclopropanecarboxamides. The relative configuration of the stereoisomers was established by X-ray crystallographic analysis of one of the model compounds. A new route to the latter functionalized cyclopropanes was developed by reaction of 1-methoxycyclopropylamines with potassium cyanide. Some remarkable rearrangements of 1-aminocyclopropane-1-carbonitriles into azetidine and oxazine derivatives via Favorskii-derived intermediates are reported. Various aspects of the chemistry of geminally functionalized cyclopropanes are discussed.

    Notes: -The reaction of primary amines bearing tertiary alkyl groups (e.g. R-NH2; R = tBu, 1-adamantyl) with nitrosobenzenes has been found to proceed by oxidative nucleophilic aromatic substitution of hydrogen, thereby affording N-(tert-alkyl)-ortho- and -para-nitrosoanilines. The replacement of hydrogen proceeds more rapidly than the replacement of ortho- or para-nitro or -bromo substituents. With p-nitronitrosobenzene, both ortho-hydrogen atoms are substituted to afford N,N′-di(tert-alkyl)-4-nitro-2-nitroso-1,3-phenylenediamines 8a,b. The addition of oxidizing agents (e.g. MnO2) increases the yield of products. 1H-, 13C-, 14N- and 15N-NMR studies have confirmed the structures of the compounds under investigation. In ortho-nitrosoanilines, the rotamer with the nitroso group syn to the amino group is favored.

    Notes: -Sc(OTf)3 is a new type of a Lewis acid that is different from typical Lewis acids such as AlCl3, BF3, SnCl4, etc. While most Lewis acids are decomposed or deactivated in the presence of water, Sc(OTf)3 is stable and works as a Lewis acid in water solutions. Many nitrogen-containing compounds such as imines and hydrazones are also successfully activated by using a small amount of Sc(OTf)3 in both organic and aqueous solvents. In addition, Sc(OTf)3 can be recovered after reactions are completed and can be reused. While lanthanide triflates [Ln(OTf)3] have similar properties, the catalytic activity of Sc(OTf)3 is higher than that of Ln(OTf)3 in several cases.

    Notes: -The reactions between 1,4-bis(diethylamino)-2,3-dinitro-1,3-butadiene (2), easily obtainable by ring-opening of 3,4-dinitrothiophene with diethylamine, and 2-substituted vinylmagnesium bromides gave good yields of the 4,5-dinitro-1,3,5,7-octatetraene derivatives 3a-e. With 1-phenylmagnesium bromide, conversely, only 3,4-dinitro-1,4-diphenyl-1,3,5-cyclooctatriene (4f) was obtained. From the unsubstituted dinitrooctatetraene 3a both the octatriene cycloisomer 4a and the valence tautomer of the latter (6a) could be isolated and characterized. The results obtained evidence an unfavourable effect of the two nitro groups on the cycloisomerization process of the octatetraene system.

The synthetic strategy is repetitive, utilizes the Suzuki cross-coupling protocol, and involves potentially bifunctional building blocks (modules) which carry one functional group for coupling and one group which serves as place holder. After the coupling, the placeholder is converted into a coupling functionality for the next growth step. The building blocks also carry chloro-substituents which are unaffected by all transformations. For one of the large building blocks it is shown that it actually can be cyclized into an oligophenylene macrocycle with a chloro-substituent at each corner and a pair of hexyl chains at each side.

    Notes: -As part of an ongoing project concerning the synthesis of enantiomerically pure α-hydroxy β-amino acids, we have now developed a general strategy allowing the synthesis of anti-α-alkyl α-hydroxy β-amino acids. Our procedure involves the intermediate formation of trans-oxazolines, which are alkylated at C-5 with good to high diastereoselectivity and then hydrolysed under mildly acidic conditions, affording in quantitative yield the corresponding hydroxy amides. The starting (R)-3-amino-3-phenylpropanoic acid and (S)-3-aminobutanoic acid were obtained in enantiomerically pure form by selective enzymatic hydrolysis of the corresponding phenylacetylamides with penicillin G acylase.

    Notes: -The introduction of alkynyl groups into the 2,3-positions of the 1,4-diazabuta-1,3-diene (DAD) backbone succeeds along two complementary synthetic routes either by condensing triisopropylsilyl-terminated dialkynyl-1,2-diones with primary aromatic amines or by a palladium-mediated alkynylation of bis(imidoyl chlorides). X-ray crystallographic analyses of two dialkynyl DAD derivatives reveals their planar (E-s-trans-E) conformations in the solid state. However, the central CC bond of both DAD backbones investigated has a length of 1.491(2) Å, and is therefore too long to indicate efficient delocalization across the DAD core. The UV/Vis spectra of dialkynyl DADs demonstrate that their absorptions in comparison to those of non-alkynyl DADs are bathochromically shifted by more than 40 nm, thereby demonstrating the suitability of the title compounds for developing NIR chromophores. The electron absorption properties of differentially N,N′-disubstituted dialkynyl DADs gave no indication for a push-pull effect across the DAD skeleton and suggest the ynimine moiety as the active chromophore of dialkynyl DADs. The electrochemical properties of the title compounds were determined by cyclo- and steady state voltammetry and show that introducing alkynyl groups leads to more easily reducible DAD systems. Again, the perception of dialkynyl DADs as covalently linked, but electronically decoupled ynimine units is corroborated by the redox potential of a differentially N,N′-disubstituted dialkynyl DAD. Similar conclusions were drawn from semiempirical MO (PM3) calculations of dialkynyl DADs.

    Notes: In order to develop a generic treatment for infections with Gram-negative bacteria, we developed a synthesis of 2-acylamino-deoxynojirimycin derivatives (17, 18, 19 and 20), which will be used as haptens for raising catalytic antibodies capable of hydrolyzing the interglycosidic bond in the lipid A moiety of endotoxins. A key intermediate in the preparation of compounds 17, 18, 19 and 20 is 3,4,6-tri-O-benzyl-2-[(benzyloxycarbonyl)amino]-2-deoxy-D-glucono-δ-lactam (6), which was prepared from known 3,4,6-tri-O-benzyl-2-[(benzyloxycarbonyl)amino]-2-deoxy-D-glucosamine (1) in four steps in 47% overall yield. Antibodies were generated against 2-[(6-aminohexanoyl)amino]-2-deoxy-D-glucono-δ-lactam (17) coupled to the carrier protein bovine serum albumin.

    Notes: From cultures of Melittangium lichenicola, Archangium gephyra and Myxococcus stipitatus, thirteen new β-methoxyacrylate (MOA) fungicides related to myxothiazols (1) have been isolated. Melithiazols A (2a), D (2b), K (2c), and L (2d) are characterized by a thiazoline-thiazole system, whereas melithiazols B (3a), E (3b), F (3c), G (3d), H (3e), I (3f), M (3g), and N (3h) are bis(thiazoles). Melithiazol C (4), as the first representative of this class of compounds, contains only one thiazole ring. The structures were established on the basis of spectroscopic data, and confirmed in the case of melithiazol E (3b), including its relative configuration, by an X-ray structure analysis. The absolute configuration of melithiazols A (2a) and B (3a) was determined by degradation and CD spectroscopy. Antifungal and cytotoxic activities, inhibition of NADH oxidation, and lipophilicities of melithiazols 2-4, myxothiazols 1, and strobilurin-type compounds are compared.

    Notes: A synthetic route to stable-isotope-substituted L-phenylalanine is presented, which allows the introduction of 13C, 15N, and deuterium labels at any position or combination of positions. For labelling of the aromatic ring, a synthetic route to ethyl benzoate (or benzonitrile) has been developed, based on the electrocyclic ring-closure of a 1,6-disubstituted hexatriene system, with in situ aromatization by elimination of one (amino) substituent. Several important (highly isotopically enriched) synthons have been prepared, namely benzonitrile, benzaldehyde, ethyl benzoate, and ethyl diphenyloxyacetate. Labelled L-phenylalanines have been synthesized from both aromatic precursors by initial conversion into sodium phenylpyruvate and subsequent transformation of this intermediate into the L-α-amino acid by an enzymatic reductive amination reaction. In this manner, highly enriched phenylalanines are obtained on the 10-gram scale and with high enantiomeric purities (≥ 99% ee). The method has been validated by the synthesis of [1′-13C]-L-Phe and [2-D]-L-Phe. In addition, two methods are described for the introduction of isotopes into L-tyrosine starting from the isotopically enriched precursors benzonitrile and ethyl benzoate.

The latter compound was converted into the corresponding trichloroacetimidate donors 8a-b. An alternative approach to trichloroacetimidate 8c commenced with the iodonium ion mediated glycosidation of ethyl 2,3,4-tri-O-benzoyl-1-thio-α-L-rhamnopyranside (15) with 1,2:5,6-diisopropylidene-D-glucofuranose (16) to afford disaccharide 17, which was transformed into 8c in five steps. Condensation of 8a-c with 2-[3,4-di-(tert-butyldimethyl-silyloxy)phenyl]ethanol (12) gave, after deacylation, key intermediate 14. Protecting-group manipulation of 14 and subsequent esterification of resulting 22 with 3,4-di-O-tert-butyldimethylsilylcaffeic acid (27) gave, after deprotection, verbascoside (1).

    Notes: The electrochemical cyanation of N-benzyl-substituted cyclic six-membered α-silylamines including tetrahydroquinoline and piperidine derivatives was studied. The results of these investigations demonstrate that α-silylamines are valuable precursors for the preparation of the corresponding α-cyanoamines. The TMS group α to the N atom not only governs the chemoselectivity of the iminium formation through a preferential desilylation reaction under the experimental conditions (i.e., amine cation radical desilylation versus deprotonation), it also lowers the oxidation potential of tertiary amines compared to their non-TMS counterparts. Both the stereoselectivity and regioselectivity of the cyanide addition were investigated with 3-methylpiperidine as the model compound. The formation of a single cisdiastereoisomer in which the 2-cyano group is axial and the 5-methyl group is equatorial, indicates that the addition of the cyanide anion onto the iminium species is under stereoelectronic control. In addition, the redox reaction involving the intermediate nitrogen-centered cation radical and the cyanide anions played no role, because the α-silyl radical has such a short lifetime.

A catalytic asymmetric reduction of the γ-iodo vinyl ketone 19 with the Corey CBS catalyst gave the ω-side chain 7 with 〉96% ee. Conjugate addition using the reaction with dilithiocyanocuprate followed by mild cleavage of the silyl protective groups and enzymatic hydrolysis of the methyl ester 22 gave (-)-PGE15in high yield.

In the case of 4-substituted benzene-1,2-diols, the ratio of regioisomers is determined by the relative acidity of the two phenolic protons. For 3-substituted benzene-1,2-diols, this ratio is determined only by steric effects in the case of alkyl substituents, although it is determined mainly by the relative stabilities of the corresponding phenates for other substituents; however, for 3-nitrobenzene-1,2-diol, this ratio is determined by the relative leaving-group ability of 2-nitro- or 3-nitrophenate. When the cyclisation was performed in the presence of an optically active phosphane, chiral 2-vinyl-2,3-dihydro-benzo[1,4]dioxin (5) was obtained with enantioselectivity of up to 45% using BINAP as the chiral phosphane.

The key step in this synthesis is the diastereoselective addition of homochiral (2-lithiophenyl)acetaldehyde acetals to the sulfonylimine 25 and to the arylimines 28 and 31. The best diastereoselectivity is obtained by addition of the bis(2-methoxypropan-2-yl)-substituted 1,3-dioxolane 6e to benzylidene-p-anisidine (31) with an HPLC-determined diastereomeric ratio 32c/33c = 92.1:7.9. The N-tosyl and the N-(4-methoxyphenyl) groups of the addition products 26d, 27d, 32c, and 33c are cleaved with sodium in liquid ammonia and ammonium cerium(IV) nitrate, respectively, to yield the primary amines 35 and 36. The acid-catalysed cyclization of the sulfonamides 26d and 27d and the carbamates 37 and 38, prepared from 35 and 36, leads to the enantiomerically pure dihydroisoquinolines 40 and 41, respectively. During the cyclization of the sulfonamides 26d, 27d and the carbamates 37, 38 the chiral auxiliary - the diol 39 - is cleaved unchanged and can be recovered in good yields.

After removal of the hydroxy group, the subunits 26a,b, containing the erythrinane rings A and D, were formed by intramolecular Michael addition. Reduction of the nitro function, followed by cyclization of the resulting amino group with the appended acetate group afforded the bicyclic lactams 29a,b, bearing an angular aryl group. Two-carbon elongation at the nitrogen atoms of 29a and 29b by means of hetero Michael addition of methyl phenyl sulfoxide, followed by Pummerer-type cyclization, gave cis- and trans-11-phenylthioerythrinan-8-ones 35a and 35b, respectively. Reductive desulfurization at C-11 furnished the desired erythrinan-8-ones 39a and 39b in 8 steps from 16.

Under ring enlargement the corresponding cyclic sulfinate esters (δ-sultines) 2 are formed. Only (1R*,2S*)-1b is reactive whereas (1R*,2R*)-1b is not attacked. This observation is explained with the formation of a cyclic intermediate 3.

    Notes: -Two chiral π-electron-rich crown ethers incorporating either a binaphthol or two D-mannitol units have been synthesized and their abilities to bind bipyridinium guests demonstrated. Both crown ethers could be interlocked mechanically with cyclobis(paraquat-p-phenylene) to afford two chiral [2]catenanes. Furthermore, these crown ethers were also mechanically interlocked with a tetracationic cyclophane incorporating a 2,2′-dihydroxy-1,1′-binaphthyl spacer to afford mixtures of diastereoisomeric [2]catenanes. The composition of these mixtures was determined by 1H-NMR-spectroscopic and HPLC analyses which revealed that modest diastereoselection (56:44-67:33) occurs during the kinetically controlled self-assembly of the catenanes. The free energy barriers (12.8-16.8 kcal mol-1) associated with the circumrotation of one macrocyclic component through the cavity of the other and vice versa were determined by variable-temperature 1H-NMR spectroscopy. In addition, another dynamic process involving the “rocking” of the mean planes of the mechanically interlocked macrocycles with respect to each other was also identified and the associated free energy barriers (10.3-10.4 kcal mol-1) determined.

    Notes: -Synthesis of ether-linked octyl 5a′-carba-β-lactosaminide 3 and -isolactosaminide 5 was carried out in seven steps, starting from the coupling products of 1,2-anhydro-5a-carba-β-D-mannopyranose derivative 7, and the oxide anions generated from the octyl N-acetyl-β-D-glucosaminide derivatives 13 and 16, respectively, under basic conditions. The 5a-carba-α-D-mannopyranose residues of the coupling products 17 and 26 were transformed into the β-D-gluco configuration through epimerization of the respective 2′-oxo derivatives 19 and 28, and selective reduction, and then into the β-D-galacto configuration by direct nucleophilic substitution of their 4′,6′-dimesylates 23 and 31 with an acetate ion. Biological assay has shown that 3 is an acceptor substrate for human-milk α-(1→3/4)-fucosyltransferase and, interestingly, 5 is not. In addition, the imino-linked congeners 4 and 6 have been synthesized by coupling of the 4-amino-4-deoxy- and 3-amino-3-deoxy derivatives 37 and 41 of octyl N-acetyl-β-D-glucosaminide, and the carba-sugar epoxide 8, respectively, and subsequent deprotection. Compound 4 is a substrate while 6 is neither a substrate nor an inhibitor for fucosyltransferase. Small-scale enzymatic synthesis was carried out by treatment of 3 and 4 with GDP-fucose and milk fucosyltransferase, which resulted in conversion into the corresponding trisaccharides 47 and 48, respectively.

    Notes: The activation and reaction volumes (ΔV#/ΔV) were determined for the retro Diels-Alder reactions of the parent dihydrobarrelene 1a, its 2-cyano derivative 1b, the exo and endo Diels-Alder adducts of maleic anhydride to naphthalene exo-, endo-4, and the exo and endo Diels-Alder adducts of N-phenylmaleic imide to 6,6-dimethylfulvene exo-, endo-8 from the pressure dependence of the rate constants and the partial molar volumes (V) of reactants and products at various temperatures. The cleavage of exo-4, endo-4, and exo-8 are slightly accelerated by pressure showing negative volumes of activation (ΔV# 〈 0) whereas the others are slightly retarded (ΔV# 〉 0). From the analysis of the volume data including the van der Waals volumes (Vw) one can conclude that the packing coefficients of the pericyclic transition states are equal to or even larger than those of the corresponding Diels-Alder adducts. This finding may be explained with the restriction of the degrees of freedom in the transition states leading to a contraction of the expansion volume.

A standard LAH reduction in the last step of the synthetic sequence transformed the α,β-unsaturated acid 12 into the target molecule 6, a constitutional isomer of nerol (5) with interesting olfactory properties.

    Notes: -For the design of a synthesis of target molecule 1 the retrosynthetic analysis yielded building blocks 2-5, of which ceramide 2-phosphite derivative 2 and aminoethyl phosphite derivative 5 are known. The generation of α-glucosaminyl (1→6)inositol building block 3 was based on pseudodisaccharide 6 which was selectively benzoylated at 6b-O and then selectively benzylated at 3b-O to give 3. The synthesis of tetramannosyl building block 4 started from known ortho ester derivative 8 which was transformed into versatile mannosyl donors 13 and 18 and into acceptor 22. Reaction of 13 with 22 gave α-disaccharide 23, deacetylation and then mannosylation with 18 gave trisaccharide 25; ensuing deacetylation and mannosylation with 13 gave tetrasaccharide 27; deallylation, acetylation, regioselective removal of the anomeric O-acetyl group and treatment with CCl3CN/DBU afforded 4. Glycosylation of 3 with donor 4 led to pseudohexasaccharide 31 in high yield. Replacement of the O-acyl groups by O-benzyl groups and then exchange of the menthyloxycarbonyl group by an O-acetyl group gave 36 which enabled regioselective attachment of 2 and 5. To this end, the 6e-O-silyl group was removed and then the aminoethyl phosphate residue was attached with reagent 5 to give 38 in high yield. 1a-O-Deacetylation and then reaction with 2 afforded 40 as fully protected 1 which was liberated in two steps; treatment with acid removed all acid labile protective groups and finally catalytic hydrogenation afforded the desired GPI anchor 1 which could be fully structurally assigned.

    Notes: -The syntheses of azaMan-β-(1→6)-C-Glc (4), azaGlc-β-(1→6)-C-Glc (5), and azaGal-β-(1→6)-C-Glc (6) based upon double reductive amination of acetylenic carbohydrate-derived diketones is described. The required diketones are obtained by addition of the acetylenic sugar anion derived from dibromoolefin 7 to benzyl-protected mannopyranolactone, glucopyranolactone, or galactopyranolactone, followed by reduction of the ketose and oxidation of the resulting diol. Ensuing double reductive amination and hydrogenolysis affords the target compounds in reasonable to good yields. Enzyme inhibition tests show that neither of the three compounds 4, 5, and 6 inhibit β-glycosidases, while moderate to good inhibitory activities were found on α-glycosidases, the most active being 6 (α-galactosidase: Ki = 0.092 μM).

    Notes: -The crystal structures of methyl 4-methoxycubane-1-carboxylate (1), 1-acetamido-4-fluorocubane (2), methyl 4-acetoxycubane-1-carboxylate (3), 1,4-difluorocubane (4), 1,4-dichlorocubane (5), and N,N-diisopropylcubane-1,4-dicarboxamide (6) have been investigated by means of X-ray diffraction analysis. Fluorine and chlorine substituents cause a shortening of the vicinal bonds, as is seen in the 4-halocubane-1-carboxylates. The cage bonds vicinal to the ester substituent, with a favorable orientation with regard to the π-acceptor influence of this group become longer than the CH-CH bonds. Furthermore, the influence on bond length with respect to the orientation of this group relative to bonds within the cubane skeleton has been investigated experimentally. The effect of the methoxy group has also been found to depend on the orientation. The cage bond antiperiplanar to the methyl group is shortened, while the cage bonds in gauche orientation to this group are lengthened. As seen in the case of the halogen-substituted derivatives, the bonds bearing the acetoxy substituent are shortened due to the σ-acceptor property of this group. Ab initio calculations on compounds 1, 2, 4, and 5 performed at the 6-31G* level confirm the experimental results.

    Notes: -Reaction of 3 with (cyanomethylene)triphenylphosphorane in refluxing dichloromethane or methanol gave mixtures of 4a and 4b in ratios of about 3:1 and 1:3, respectively. The same reaction performed with the furanoid isomer 5 afforded 6a and 6b in (E)/(Z) ratios of approximately 9:1 and 1:2, respectively. Catalytic hydrogenation of the α,β-unsaturated nitriles 4 and 6 with either 10% Pd-C or Raney nickel afforded the saturated nitriles 7 and 9, or the 1-amino-1,2,3-trideoxy-4-octulose derivatives 8 and 10, respectively. In an attempt to transform 6a into the appropriate polyhydroxylated branched-chain pyrrolidine 15, (5R,8S,9R,10S)-8,9,10-trihydroxy-1-aza-6-oxaspiro[4.5]decane, which was identified as its peracylated derivative 16, was isolated. Following an alternative synthetic strategy, partial hydrolysis of compound 9 afforded 17 which was regioselectively transformed into the corresponding derivative 22 via its 8-O-p-toluenesulfonyl derivative 18. Removal of the 4,5-O-isopropylidene group in 22 with aqueous trifluroacetic acid gave the free octulose 23, which was hydrogenated in the presence of 10% Pd-C, to afford the expected (6S,7R,8R,8aR)-6,7,8-trihydroxyindolizidine (1-deoxycasta-nospermine, 11).

    Notes: Two “self-complexing” macrocycles (22ab and 23ab) based on a methylthio-substituted derivative of the electron donor tetrathiafulvalene (TTF) and a cyclic bipyridinium acceptor have been prepared.[1] When “decomplexed” by fractional crystallization, the rigid compound 22b was not able to undergo “recomplexation” to any significant degree, whereas for the more flexible 23b, an equilibrium between “complexed” (23a) and “uncomplexed” (23b) compounds was slowly re-established in solution according to UV/Vis measurements. 23ab was thus seen to behave as a “thermo-switch”, although with a rather slow response. Any complexation between the two separated components, i.e. the cyclic acceptor 3 and the tetrakis(methylthio)tetrathiafulvalene 6, was not observed by 1H-NMR spectroscopy. However, 3 was able to bind unsubstituted TTF (5) in its cavity, albeit with a small association constant of only 60 M-1. Taking advantage of the tetravalency of TTF, we also report the synthesis of a “self-complexing” pyromellitic diimide/TTF macrocycle (33ab). Whereas 22ab and 23ab were prepared employing the concept of template-assisted synthesis,[2] the synthesis of 33ab did not rely on this technique.

The tetrayne units in both molecules deviate considerably from linearity, giving rise to transannular distances of the terminal sp centers of 3.098(2) Å (3) and 4.147(2), 4.196(2) Å (4), and 3.390(2) Å (3) and 4.251(2), 4.252(2) Å (4) for the central sp atoms, respectively.

    Notes: The reactivity of ultrafine zinc powders, produced by a new electrochemical method coupled to the use of ultrasound, has been evaluated for various organometallic reactions such as the Reformatsky reaction, Barbier allylation, alkyne reduction, reductive dehalogenation and p-nitroester cleavage. The results show that zinc powders produced by sonoelectrochemistry are highly reactive. Particle-size effects, reactivity of zinc-copper alloys and metal-waste recycling procedures have also been studied. The production of highly reactive zinc powder by sonoelectrochemical methods appears to be an interesting alternative to conventional methods used in organozinc chemistry and may be especially important in medium- or large-scale processes.

    Notes: The mechanistic reaction pathway for the slow-binding inhibition of rabbit muscle aldolase by D-glycero-tetrulose 1-phosphate (D-erythrulose 1-phosphate) was investigated through the use of its phosphonomethyl isoster 4 which was synthezised for this study. The latter is not a substrate nor a slow-binding inhibitor but interferes in the enzyme-catalyzed reaction with the substrate fructose 1,6-diphosphate in a competitive manner. It was found that phosphonate 4 forms an iminium ion with aldolase and undergoes subsequent α-proton abstraction to form an enamine intermediate. We show from these results that enzyme slow-binding inhibition by D-erythrulose 1-phosphate is consistent with a phosphate β-elimination reaction through the enamine intermediate. This mechanism takes into account the stereochemical features known for aldolase, the parallel between enzyme activity recovery and phosphate release after action of D-erythrulose 1-phosphate, and also the same reaction from dihydroxyacetone phosphate.

    Notes: Protonation of α-sulfinyl carbanions 3, resulting from the selective thiophilic addition of organolithium compounds to aliphatic sulfines 2, provided dithioacetal oxides 4 (63-94% yield). The diastereomeric ratio ranged from 52:48 up to 100:0 with sterically hindered substituents. Thus, (1S*,SS*)-2,2-dimethyl-1-methylsulfanyl-1-methylsulfinylpropane (4ca) was obtained as a single isomer. The origin of the stereoselectivity is rationalized in terms of a thermodynamic equilibrium between the two diastereomers of α-sulfinyl carbanions 3, at which protonation occurs under kinetic control and with retention of configuration with the assistance of the lithium cation. Moreover, the stereochemistry could be totally reversed by converting the lithiated anion intermediate into an aluminium “ate” complex. Thus, (1R*,SS*)-2,2-dimethyl-1-methylsulfanyl-1-methylsulfinylpropane (4ca) was selectively obtained by this method (dr = 0:100).

    Notes: The formal [3+2] cycloaddition of β-amino-substituted α,β-unsaturated Fischer carbenechromium complexes 2, which are easily prepared in a one-pot procedure from terminal alkynes 1 via an initially formed alkynylidene complex by a Michael-type addition of a secondary amine, with a variety of alkynes 3 afford the 1,2,5-trisubstituted 3-alkoxy-5-dialkylamino-1,3-cyclopentadienes 4 with an acetal-protected aldehyde or ketone carbonyl group in either the 5-substituent R1 or the N-substituent R2. Under acidic conditions, both the enol ether moiety in the 5-membered ring as well as the acetals in the side chains are hydrolyzed, and the resulting cyclopentenones 6 with carbonyl-group containing side-chains undergo facile intramolecular aldol reactions to give bicyclo[3.3.0]oct-2-en-4-ones 8 and 8-azabicyclo[3.3.0]oct-2-en-4-ones 12, respectively, in good to very good yields for most cases.Supporting information for this article is available on the WWW under //http://www.wiley-vch.de/contents/jc_2046/1999/99100_s.pdf or from the author.

The X-ray structures of four model compounds (4a, 4d, 4j, 4n) have been determined, each of which shows a short, nonbonded C1-C3 distance in the bicyclo[1.1.1]pentyl subunit of about 1.89 Å.

    Notes: The indium-mediated reaction of Z-protected α-amino aldehydes 1-6 with 2-(bromomethyl)acrylates 8/9 in aqueous solvents has been investigated. The preference for the formation of syn-configured homoallyl alcohols 10-16 (diastereoselectivities ranging from 2:1 to 32:1, yields 68-93%) may be explained by a chelate-controlled reaction. No racemization occurred during the preparation and transformation of the amino aldehydes. Acid-catalyzed esterification (H2SO4 in Et2O) led to α-methylene-γ-butyrolactones 17-22 with yields ranging from 89 to 97%.     Notes: In aprotic media, photoinduced electron transfer from toluene to 1,4-naphthalenedicarbonitrile (DCN) is followed by in-cage proton transfer and radical coupling. However, in the presence of a protic co-solvent, the radical ions diffuse out of the cage and deprotonation of the toluene radical cation takes place from the free solvated species. With tBuOH, this results in reductive benzylation of DCN through coupling of benzyl radicals with DCN-·. With MeOH, secondary hydrogen abstraction by PhCH2· yields hydroxymethyl radicals, which couple with DCN-·. Both benzyl and hydroxymethyl radicals are efficiently trapped by electrophilic alkenes. Thus, in the presence of dimethyl maleate, DCN-photosensitized benzylation - or hydroxymethylation - of the substrate occurs through direct activation of the C-H bond. The same reaction has been accomplished with acrylonitrile, albeit with a lower yield.

The metal-hydrogen exchange promotes immediate 1,4-elimination, yielding an (E)-configured conjugate alkyloxy-functionalized diene. The elimination product can be subjected to a Diels-Alder reaction, which allows further synthetic elaboration. In particular, in the case of farnesal dimethyl acetal the cycloaddition process leads to a derivative that can be transformed into a potential mimic agent of Juvenile hormones.

For 1-bicyclo[3.1.1]heptylcarbene 12, the preferred stabilization is again enlargement of the four-membered ring to give bicyclo[3.2.1]oct-1(7)-ene 13. The further rearrangement of this alkene to 7-bicyclo[3.2.1]octylidene 16is endothermic by 32.2 kcal/mol and should not take place under moderate conditions. (5-Bromobicyclo[3.1.1]heptyl)bromocarbene 32, a derivative of 12 generated from the tetrabromide 23 with MeLi, rearranged by enlargement of the four-membered ring to give alkene 31, which reacted to give the head-to-head 2π+2π dimer 26, whose structure was established by X-ray crystallography.

    Notes: Reaction of fluorenone (4) with a low-valent titanium reagent generated from TiCl4 and Zn in THF in the presence of pyridine gave terfluorenyl (6) in 71% yield, bifluorenylidene (2), the conventional McMurry reaction product, in 2% yield, and additional reduction products. The reductive “trimerization” was rationalized in terms of an attack of the intermediate fluorenone dianion on bifluorenylidene. The molecular structure of a single crystal of 6 indicated an approximately C2 conformation, with a slightly twisted central ring, and two equally folded side moieties with dihedral angles of 58.1° and 58.0° between the central and side five-membered rings. The AM1 and PM3 calculations showed C2 global minima, similar to the conformation in the crystal. The calculated Cs transition state conformations were found to be 21.3 (AM1) and 19.9 (PM3) kcal/mol higher in energy than the global C2 minima. A 2D-NMR NOESY experiment on 6 supported the C2 (+sc,+sc) or C2 (-sc,-sc) conformation in solution.

    Notes: Some neoclerodane (2-4) and labdane (5 and 6) diterpene ketones having a β- or γ-hydroxy function, an α-epoxy group, or an α-olefinic double bond have been irradiated at λ = 313 nm in dry solvents (benzene or methanol). Fruticolone (2) yielded the naturally occurring 5,6-seco-neoclerodan-6,1α-olide derivative fruticolide (1), while hispanolone (5) gave the δ-lactone 8, both transformations involving a Norrish type I photoreaction. The α,β-unsaturated ketone derivative 4 was transformed into 7 by a stereoselective intramolecular [2+2] cycloaddition reaction involving its furanic 13(16)-double bond. Finally, the α-epoxy ketone 6 was rearranged to the 10(9→8)abeo-labda-7,9-dione derivative 10. Apart from the interest in these reactions for synthetic purposes, the photochemical transformation of fruticolone (2) into fruticolide (1) suggests that the latter might originate from the former as an artefact during the extraction and isolation procedure, rather than from any biogenetic pathway.

    Notes: Two simple sea cucumber ganglioside analogs 8(NeuAcα 2→6 Glcβ →1 docosanol) and 9 (NeuAcβ 2→6 Glcβ 1→1 docosanol), containing a long-chain alcohol as a mimic of ceramide, have been synthesized. Coupling of the methyl 2-thioglycoside derivative of N-acetylneuraminic acid 5 with docosyl glucoside derivative 3 afforded the protected ganglioside analogs 6and 7, which were deprotected to give the corresponding ganglioside analogs 8and 9, respectively. The 13C-NMR data obtained for these anomers 8 and 9 should prove useful for the determination of anomeric configurations of sialic acid residues in gangliosides.

    Notes: The discovery of the equilibration of alkynes with ylidenecarbenes at high temperatures has been followed by increasing use of the reactivity of such carbenes in the synthesis of polycyclic aromatic hydrocarbons. This has led to simple and elegant syntheses of fullerene fragments, and of cyclopenta-fused hydrocarbons. The related high-temperature equilibration of arynes with ring-contracted exocyclic carbenes has provided a further pathway for the transformation of polycyclic aromatic systems into less accessible cyclopenta-fused hydrocarbons. Some recent developments in the chemistry of aryl radicals have extended the scope of radical approaches to the synthesis of polycyclic hydrocarbons.

    Notes: Several models for the formation of the 8-membered B-ring of taxanes by an intramolecular nitrile oxide [3+2] cycloaddition were prepared from the monoacetal of 2,2-dimethylcyclohexane-1,3-dione (5). The nitrile oxides were then generated under high-dilution conditions. In most cases only oligomers were obtained. The isoxazoline 19, containing a 9-membered ring, was formed from the nitrile oxide 17, and a bis(isoxazoline) 21was isolated from the reaction of the nitro compound 16c.

    Notes: The lithium α-(dimethylamino)alkoxides resulting from the nucleophilic addition of an organolithium reagent to N,N-dimethylformamide are basic enough to deprotonate alkyltriphenylphosphonium salts suspended in tetrahydrofuran. The aldehydes liberated by the spontaneous decomposition of the resulting α-amino alcohols (hemiaminals) undergo a Wittig reaction with the simultaneously generated phosphorus ylides to afford olefins in excellent overall yields. This in situ method offers the unique advantage in its applicability to labile aldehydes which otherwise would become prey to (Z/E)-isomerization or self-condensation processes.

    Notes: A sulfahydantoin (3-oxo-1,2,5-thiadiazolidine 1,1-dioxides) motif is used as a new type of peptidic constraint to lock two consecutive amide nitrogens by a sulfonyl bridge. The 5-membered heterocyclic motif was prepared starting from proteogenic and synthetic amino acids and chlorosulfonyl isocyanate. Constrained dipeptides were obtained under alkaline conditions (methoxide or tert-butoxide) by cyclization of symmetric and dissymmetric sulfamides. The absolute configuration of the chiral centers for the derivative L-Phe-D-Ala, a congener of the series, was established by X-ray diffraction crystallographic analysis. In addition, the chemo-, regio-, and stereoselectivities of the reactions were studied. In the acylated derivatives, the sulfahydantoin constraint induces a unique backbone conformation with coplanarity of two consecutive peptide bonds.

    Notes: 2-(Dimethylamino)oxazoline-4-carboxylic acids 5 were prepared by condensation of binucleophilic amino acids 4and O-ethyl-N,N-dimethylisourea 3. New heterocyclic N-acetylglucosaminyl amides and chitobiosyl amides 8 were obtained by the coupling of tetraacetylglucosamine 6a or heptaacetylchitobiosylamine 6b with acid chlorides of heterocyclic carboxylic acids 2 or 5a and subsequent deacetylation. Based on their substitution patterns, compounds 8were expected to have inhibitory activity towards chitinases. Enzyme assays showed that glycosyl amides 8 indeed were moderate inhibitors of chitinases, the diacetylchitobiosyl amides 8d-f generally having higher inhibitory activities than the N-acetylglucosaminyl amide derivatives 8a-c. Inhibitory activities depended on the chitinase tested.

    Notes: The structural transformations of flavylium ion in aqueous solutions caused by pH jumps and photoexcitation have been investigated. At pH 〈 1, the stable form is the colored cationic species (AH+). By increasing pH, the concentration of AH+ decreases and, at pH = 5, this form is no longer present. The species obtained immediately after a pH jump undergo transformation processes with pH-dependent rate constants. At pH = 5.2 and 20 °C, the final product is the uncolored trans-chalcone (Ct). This form can be transformed by light excitation into the cis-chalcone (Cc) isomer, which is in equilibrium with the hemiacetal form (B2). This mixture is relatively inert due to the existence of a kinetic barrier that slows down the back thermal isomerization of Cc to the stable Ct form. Such a back reaction to Ct can be totally prevented if the irradiated solution is submitted to a pH jump to pH = 1, which transforms the photoproducts into the stable AH+ species. In basic solution, two more species were detected, namely the anionic forms Cc- and Ct- of the cis and trans chalcone. Ct- is a stable, not photosensitive and luminescent species, whereas Cc- is not stable, being converted into Ct- in the dark. The photochemical and pH-induced transformations of the flavylium cation in the pH range 0-11 can be taken as a basis to design write-lock-read-unlock-erase cycles for an optical molecular-level memory with multiple readout capacity.

    Notes: Calix[4]arenes modified at the upper rim with two pairs of functional groups were synthesized and were used as starting materials for the synthesis of an enzyme model 3-Zn2 that has two ZnII centers and two dimethylamino groups. Under neutral conditions, this dinuclear metallo-enzyme model is catalytically active in the cleavage of the phosphate diester bond of the RNA model substrate 2-hydroxypropyl-p-nitrophenyl phosphate (HPNP). The kinetics of 3-Zn2 were compared with a mononuclear complex 1-Zn and a dinuclear complex 2-Zn2 that lack the dimethylamino groups. A catalysis mechanism is proposed where the ZnII centers of 3-Zn2 cooperate in the activation of the phosphoryl group while one of the dimethylamino groups acts as a general base in the deprotonation of the hydroxyl group of HPNP.

    Notes: The reaction of unprotected sucrose with tert-butyl chloromethyl ketone in dimethylformamide was investigated as a model for the study of the relative reactivity of the various hydroxy groups of sucrose. Besides the monoethers arising from the substitution of the chlorine atom by 2-OH and 1′-OH of sucrose, the major product is a tert-butyl hydroxymethyl 5-membered ring acetal involving 2-OH and 3-OH. The formation of this product illustrates the preeminent reactivity of 2-OH towards the carbonyl group of the α-chloromethyl ketone, leading to an intermediate hemiacetalic anion, which can enter by intramolecular acetal formation through an intermediate epoxide.

    Notes: The partial synthesis of 54 derivatives of thiangazole A (1a), a new polythiazoline antibiotic from Polyangium spec. (myxobacteria), is described. Derivatives with chemical modification of the carboxamide group in the oxazole region were prepared either by N-alkylation to amides 5-14 or by methanolysis to ester 15, and its transformation products 16, 19, 20. Oxidation of the C-5 methyl group of 1a with molecular oxygen led to the hydroxymethyl derivative 21, and two by-products lacking the C-5 methyl group (22), or the entire oxazole ring (23). Key intermediate for analogues with modifications in the styryl region is the aldehyde 27, obtained by direct cleavage of the C-21/C-22 double bond. 27 was transformed into the oximes 37-42 and by Wittig reaction to (21Z)-thiangazole (43) and analogues 44-46 with proton and alkyl residues replacing phenyl. 21,22-Didehydrothiangazole (50) was synthesized in a multi-step reaction from 27 via the 20-alkinyl intermediate 49.     Notes: The asymmetric hydrogenation of methyl 3,5-dioxohexanoate (1) into mixtures of 3,5-dihydroxyesters (2) and 3-hydroxylactones (3) has been reinvestigated with a variety of ruthenium catalysts. Catalysts bearing diphosphanes which possess axial chirality such as (S)-MeO-Biphep give predominantly (3R,5S)-anti-2 in up to 78% de and 95% ee, affording an efficient synthesis of (S)-6-methyl-5,6-dihydro-2-pyrone [(S)-5] in up to 95% enantiomeric excess. On the contrary, some Ru-{amidophosphane-phosphinite} complexes catalyze sluggishly the formation of syn-2 in up to 92% de but with poor enantiomeric excesses. In all cases, methyl (R)-3-hydroxy-5-oxohexanoate [(R)-11] is the exclusive primary hydrogenation product, which can be isolated in high yields and enantiomeric excesses up to 78%. Further hydrogenation of enantiomerically enriched (R)-11 in a separate experiment affords (3R,5R)-syn-2 in high diastereomeric and enantiomeric excesses (up to 80% and 98%, respectively), provided a Ru-(R)-Binap-type catalyst is used.

    Notes: Configurationally unstable lactone-bridged biaryls 4 are cleaved atropo-diastereoselectively using chiral menthol-derived alkali metal alkoxides, to give axially chiral biaryl esters of type 5 in high yields and excellent diastereomeric ratios of up to 〉 99:1. The method permits the optional preparation of each of the two atropisomers from the same lactone precursor (“atropo-divergence”), simply according to the choice of the appropiate mentholate or its enantiomer as the O-nucleophile - or by the use of the mentholate in solution or in suspension. Undesired minor atropisomers of 5 possibly formed (if at all) can be recycled (“axially chiral economy”) by cyclization back to the lactone 4. For the preparation of larger amounts of enantiopure biaryl alcohols 9, an efficient reaction sequence was developed: alcoholysis of 4 → in situ reduction → crystallization. The synthetic value of these alcoholysis reactions for asymmetric biaryl synthesis is illustrated by the transformation of 5 into a broad series of enantiopure biaryls of type 6 with various functional groups ortho to the axis.

The combination of palladium-catalysed coupling of 2 together with 1-alkynes/6-endo-dig annulation reactions in the presence of ammonia leads to functionalised pyrido[3,4-c]thiazoles in satisfactory yields. The utilisation of uncatalysed displacement reactions of the triflate group represents a very simple method for the synthesis of 4-N-,4-O-, and 4-S-substituted thiazoles.
