    Notes: The Harderian gland of the musk shrew Suncus murinus is elongated anteroposteriorly from in front of the eye to behind the ear. The gland is divided into two portions: an anterior portion (A portion) and a posterior portion (P portion). The single secretory duct of the gland emerges from the anterior end of the P portion, receives several secretory ducts of the A portion during the course along it, runs around the ventral aspect of the eyeball, and finally opens into the anterior corner of conjunctival sacs. The two portions of the gland show a fundamentally similar histological structure, having a poorly developed intraglandular duct system and wide tubular alveoli. The quantity of lipid vacuoles and stromal connective tissue in the A portion is greater than in the P portion. The lipid vacuoles in both portions are surrounded by unit membranes, but their contents appear different.The lacrimal gland of the musk shrew is located along the ventral side of the P portion of the Harderian gland. The lacrimal duct emerges from its anterior end, runs around the ventral and anterior aspects of the ear, crosses the A portion of the Harderian gapos; and, and finally opens at the posterior corner of conjunctival sacs. The lobules of the lacrimal gland comprise a branched duct system and terminal acini with two types of secretory cells: (1) acidic cells positive both for the periodic acid-Schiff reaction (PAS) and for Alcian blue (AB) and (2) neutral cells positive for PAS and negative for AB. Both cell types tend to make separate acini, but when present in the same acinus, the acidic cells occupy relatively peripheral positions in the acinus. Both cell types lack intercellular canaliculi.On the basis of the present study as well as previous descriptions in the literature, the author suggests that the mammalian lacrimal glands can be divided into two sets: (1) a Glandula lacrimalis superior with multiple secretory ducts associated with the upper eyelid and (2) a Glandula lacrimalis inferior with a single secretory duct opening into the lateral corner of the conjunctival sacs. These glands have a fundamentally similar histological structure; but in the rabbit, which possesses both sets of lacrimal glands, they are different. On the other hand, the secretory cells of lacrimal glands generally have no intercellular secretory canaliculi, which are characteristically present between the serous secretory cells of the salivary glands.

    Notes: The leg musculature from 11, 14, and 17 day chick embryos was analyzed histochemically to investigate the temporal and spatial distribution of various types of sulfated glycosaminoglycans present during skeletal muscle development. Types of glycans were identified by selective degradation with specific glycosidases and nitrous acid coupled with Alcian blue staining procedures for sulfated polyanions and with [35S]sulfate autoradiography. On day 11, radiolabeled chondroitin sulfate glycosaminoglycans are localized extracellularly in both the myogenic and connective tissue cell populations. By day 17, incorporation of [35S]sulfate into chondroitin sulfate is substantially reduced, although Alcian blue-stained chondroitin sulfate molecules are still detectable. With increasing age and developmental state of the tissues, radiolabeled and stained dermatan sulfate and heparan sulfate progressively increase in relative quantity compared to chondroitin sulfate both in muscle and in associated connective tissue elements. These changes in glycosaminoglycans correlate well with similar changes previously determined biochemically and further document the alterations in extracellular matrix components during embryonic skeletal myogenesis.

    Notes: The aquiferous system of representatives of the orders Dictyocer-atida, Dendroceratida, and Verongida has been studied to note its relevance to the systematics of the groups. The volume of the choanocyte chamber, the size and shape of the choanocytes, the number of choanocytes per chamber, the relative development of the mesohyl, and the features of endopinacocytes are estimated from scanning and transmission electron microscopic observations of representatives of most families of the three orders. Although the Dysideidae have a reticulate skeleton and were classified in the order Dictyoceratida, they are actually closer to the Aplysillidae (Dendroceratida) than to dictyoceratids. The anatomy and cytology of the Halisarcidae differ profoundly from those of these three orders and are clearly more closely related to nonkeratose sponges. Some changes in classification lead to a pattern with highly homogeneous orders that clearly differ in their anatomic and cytologic features, which does not support the hypothesis of a common origin of the “keratose” sponges.

    Notes: The wall of the asymmetrical saclike lungs of the fishes Polypterus and Erpetoichthys consists of several functionally different tissue layers. Their lumen is lined by a surface epithelium composed of (1) highly attenuated cells, termed pneumocytes I; (2) pneumocytes II with lamellar bodies, presumably indicating surfactant production; (3) mucous cells; and (4) ciliated cells. Underlying the pneumocytes I is a dense capillary net. The thin continuous endothelium of this net, together with the pneumocytes I, constitute the very thin blood-air barrier. The basement membrane of epithelium and endothelium fuse in the area of the blood-air barrier (thickness 210 m̈m). Secretory and ciliary cells form longitudinal rows in the epithelium. Below the zone with a gas-exchanging tissue, a layer of connective tissue containing collagen and special elastic fibers occurs. The blood vessels that give rise to or drain the superficial capillary plexus are located in this connective tissue. The outermost layer of the lung consists of muscle cells, a narrow inner zone with smooth muscle cells, and an outer, broader zone with cross-striated muscle cells. The lung is innervated by myelinated and nonmyelinated nerve fibers. The morphology of the gas-exchange tissue in the lungs of these primitive bony fish is fundamentally very similar to that of the lungs of tetrapod vertebrates. The morphologic observations are in close agreement with physiologic data, disclosing well-developed respiratory capacities. Structural simplicity can be regarded as a model from which the lungs of the higher vertebrates derived. In addition to respiratory function, the lungs seem also to have hydrostatic tasks.

    Notes: Effect of turning of the egg during incubation on development of the area vasculosa of the chick embryo was investigated. The size of the area vasculosa was determined by two methods: direct measurement with calipers and measurement of a template cut from the eggshell by use of an automatic surface area recorder. The effects of turning and additionally the effects of lowered temperature (36°C) on both growth of the area vasculosa by day 7 and embryo growth by day 14 of incubation were investigated. The effects of turning during a critical period for turning, from 3 to 7 days of incubation, were also recorded. Generally, failure to turn eggs retarded growth of the area vasculosa. Turning during the critical period stimulated the extent of growth of the area vasculosa by day 7 of incubation and of subsequent embryonic growth by day 14. Incubation at low temperature resulted both in reduced expansion of the area vasculosa and retarded embryonic growth in a pattern similar to that observed for unturned eggs. It is suggested that turning stimulates development of blood vessels in the area vasculosa via localized increases in blood pressure. The effect of a reduced area vasculosa is considered to retard embryonic development through restricted nutrient uptake from the yolk. The prevailing hypothesis that turning prevents deleterious membrane adhesions is questioned in light of these observations. It is suggested that the physiological basis for the need for turning lies in maximizing the growth rate of the area vasculosa to maximize yolk use and embryonic growth rate.

    Notes: Recessive mutant gene c in axolotls results in a failure of the heart to function because of abnormal embryonic induction processes. The myocardium in this mutant lacks organized sarcomeric myofibrils. The present study was undertaken to determine if developmental abnormalities were evident in other areas of the heart besides the myocardium. A detailed comparative survey of the structure of developing normal and mutant hearts, including the endocardium, its cellular derivatives, and the extracellular matrix, known as cardiac jelly, showed that in the mutant there are fewer than the normal number of endocardial cells lining the heart lumen, the number of mesenchyme cells is reduced, and the cardiac jelly area is greatly enlarged in the posterior part of the truncus adjacent to the ventricle.

    Notes: Plethodontid salamanders have unique nasolabial grooves that may function as “capillary tubes” to convey chemicals to the vomeronasal organ when these animals nose-tap. 3H-proline was placed at the base of these grooves in Plethodon cinereus, and autoradiography revealed large concentrations of radioactive material in the vomeronasal organs. There was no significant accumulation of radioactive material in the main olfactory epithelium. Salamanders with blocked nasolabial grooves lacked significant accumulation of material in their nasolabial grooves or vomeronasal epithelia, although some salamanders had radioactive material in the posterior portion of their vomeronasal organ that had entered through the internal nares. Anteriorly placed vomeronasal organs situated adjacent to the posterior limits of the nasolabial grooves may insure that nose-tapping primarily stimulates the vomeronasal sensory epithelium.

    Notes: A new mechanical model for function of the pharyngeal jaw apparatus in generalized perciform fishes is developed from work with the family Haemulidae. The model is based on anatomical observations, patterns of muscle activity during feeding (electromyography), and the actions of directly stimulated muscles. The primary working stroke of the pharyngeal apparatus involves simultaneous upper jaw depression and retraction against a stabilized and elevating lower jaw. The working stroke is characterized by overlapping activity in most branchial muscles and is resolved into three phases. Four muscles (obliquus dorsalis 3, levator posterior, levator externus 3/4, and obliquus posterior) that act to depress the upper jaws become active in the first phase. Next, the retractor dorsalis, the only upper jaw retracting muscle, becomes active. Finally, there is activity in several muscles (transversus ventrales, pharyngocleithralis externus, pharyngohyoideus, and protractor pectoralis) that attach to the lower jaws. The combined effect of these muscles is to elevate and stabilize the lower jaws against the depressing and retracting upper jaws.The model identifies a novel mechanism of upper jaw depression, here proposed to be the primary component of the perciform pharyngeal jaw bite. The key to this mechanism is the joint between the epibranchial and toothed pharyngobranchial of arches 3 and 4. Dorsal rotation of epibranchials 3 and 4 about the insertion of the obliquus posterior depresses the lateral border of pharyngobranchials 3 and 4 (upper jaw). The obliquus dorsalis 3 muscle crosses the epibranchial-pharyngo-branchial joint in arches 3 and 4, and several additional muscles effect epibranchial rotation. Five upper jaw muscles cause upper jaw depression upon electrical stimulation: the obliquus dorsalis 3, levator posterior, levator externus 3/4, obliquus posterior, and transversus dorsalis. This result directly contradicts previous interpretations of function for the first three muscles. The presence of strong depression of the upper pharyngeal jaws explains the ability of many generalized perciform fishes to crush hard prey in their pharyngeal apparatus.

    Notes: Changes in body morphology during growth and reproduction in the hydromedusa Eleutheria dichotoma are described in terms of variations in eight different characters: umbrella diameter, total surface area, tentacle area, umbrella area, tentacle knob diameter, number of embryos, and diameter and area of buds. Sexually (sex) and vegetatively (veg) reproducing medusae differ significantly in their body morphometrics. Statistically significant allometric relations exist between umbrella diameter and (1) central area (sex and veg); (2) tentacle area (veg); (3) total area (veg); (4) tentacle knob diameter (veg); (5) bud diameter; and (6) number of embryos. A significant correlation between umbrella diameter and area is also found in undetached buds. During sexual reproduction, umbrella area shows positive allometry and loses its correlations to total area, tentacle area, and tentacle knob diameter. Linear and nonlinear bivariate allometric coefficients allow estimation of total body size from only one or two easily measurable attributes, e.g., umbrella and tentacle knob diameter. Curve fitting by the classic allometric equation (y = bxc) is only negligibly worse than that obtained with a “full” equation (y = a + c), and statistical confidence is better.Chemical analyses for carbon and nitrogen content allow estimation of biomass from the projection area of the body surface. The relation factors are 1.06 μgC mm-2 (sex) and 1.14 μgC mm-2 (veg) for carbon and 0.293 μgN mm-2 (sex) and 0.287 μgN mm-2 (veg) for nitrogen. The C:N ratios are 3.6 and 4.0 for sexual and vegetative medusae, respectively. The use of allometric regression formulas to calculate surface areas and to relate these to carbon content provides quick estimations of body size in a microscopic animal.

    Notes: The neuronal organization of the accessory olfactory bulb (AOB), which receives sensory information from the vomeronasal organ, was described in a squamate reptile (Podarcis hispanica) by means of light microscopy. Using the Golgi-impregnation method, seven neuronal types could be distinguished:Periglomerular cells constitute a morphologically heterogeneous population of small neurons located between and around the glomeruli.The mitral cells are diffusely distributed in the AOB. Their cell bodies are usually located within the mitral cell layer, but some of them could be also observed in the plexiform layers. Mitral cells were classified into three subgroups on the basis of their sizes and dendritic tree morphologies. Thus, the “outer mitral cells” have the biggest cell bodies, and their distal secondary dendrites are mainly distributed rostrocaudally in the external plexiform layer. The “inner mitral cells” have large cell bodies, and their secondary dendrites are distributed dorsoventrally and are located deeper than those of the other two subgroups. The third type, the “small mitral cells,” is the smallest one among mitral cells in the AOB, and from their cell bodies, only two main dendritic trunks arise.The granule cells are composed of several categories based on their different cell body locations and dendritic tree morphologies. Thus, the “superficial granule cells” are located exclusively in the external plexiform layer and have small dendritic fields. The “middle granule cells” have fusiform cell bodies - situated in the internal plexiform layer - and present a wide dendritic projection area. Finally, the “deep granule cells” are distributed throughout the granule cell layer and include a great variety of dendritic tree morphologies.The distribution and morphological features of all neuronal types constituting the AOB of Podarcis were compared with those reported on other vertebrates. The results suggest that the lamination pattern and neuronal organization of the AOB in lizards are more similar to that of mammals than to that of the remaining vertebrates.

    Notes: Iridescent butterfly scales are structurally colored, relying upon the interaction of light with detailed architecture to produce their color. In some iridescent scales, the reflective elements are contained within the body of the scale and come in two basic forms, lattices that produce diffraction colors (analogous to those produced by opal), and stacks of laminae that produce thin-film interference colors (analogous to those produced by soap or oil films). Both structures are remarkably complex and precise, yet each is only part of the total edifice built by the cell that makes the scale.To understand better how a cell can produce lattices or thin-film laminae, I studied the development of iridescent scales from two lycaenid butterflies. The presence of diffraction and thin-film scales in the same family (and in some cases on the same individual) suggests that the two types must be developmentally related; yet these results yield no clear explanation as to how. The diffraction lattice appears to be shaped within the boundaries of the scale cell by means of a convoluted series of membranes in which the smooth endoplasmic reticulum plays an important part. The thin-film interference laminae appear to result from the condensation of a network of filaments and tubes secreted outside the boundaries of the cell. This paper outlines the developmental histories of both types of scale and discusses the developmental implications of the mechanisms by which they form.

    Notes: Wing folding spicules, elytral binding patches, and elytral locking devices of adult male and female seed weevils, Smicronyx fulvus LeConte and S. sordidus LeConte, involved in stridulation are described. Sound is produced by both sexes of the two species when the plectrum, paired conical teeth located along the anterior margin of the dorsally elevated seventh sternite, is struck against an elongate file, the pars stridens, on the under surface of the apical portion of each elytron. A second plectrum, on the sixth tergite, is well-developed in males of both species and is used by males to produce sound before and during mating.Sex-specific and species-specific differences in the sound produced is attributed to structural variation in the pars stridens and the elytra. The pars stridens determines frequencies, while the elytra may further modify the sound. The frequency range for male S. fulvus is 1,000 cycles per second (cps) through 13,000 cps and for male S. sordidus is 2,500 cps through 13,000 cps. The frequency range for female S. fulvus is 2,000 cps through 11,500 cps and for female S. sordidus is 900 cps through 11,500 cps.

    Notes: Light and electron microscopic observations of the lateral-line organs of larval Ichthyophis kohtaoensis confirmed earlier reports of the occurrence of two different types of lateral-line organs. One type, the ampullary organ, possesses 15-26 egg-shaped sensory cells. Each sensory cell extends a single kinocilium surrounded by a few microvilli into the ampullary lumen. This is in contrast to the ampullary organs of urodele amphibians that contain only microvilli. The second type of organ, the ordinary neuromast, has 15-24 pear-shaped sensory cells arranged in two to three rows. Each sensory cell shows a kinocilium that is asymmetrically placed with respect to both a basal plate and approximately 60 stereovilli. The sensory cells of ampullary organs are always separated by supporting cells; those of neuromasts are occasionally in contact with one another. Numerous (neuromasts) or few (ampullary organs) mantle cells separate the organs from the epidermal cells. Only afferent synapses are found in the ampullary organs whereas vesicle-filled fibers together with afferent nerve terminals are found in neuromasts. Both organs contain similarly sized presynaptic spheres adjacent to the afferent fibers. It is suggested that the neuromasts have a mechanoreceptive function, whereas the ampullary organs have an electroreceptive one.

    Notes: The cephalic muscles in three species of Entechinus, two species of Opheodrys, and Symphimus mayae display patterns of interspecific variation that are largely congruent with patterns of variation previously described for the skulls of these species. This congruence does not stem from direct correlation between the shapes of associated bones and muscles. In these colubrid snakes, most interspecific variations in muscle form involve changes in the shape or relative position of attachment points that appear unrelated to changes in the gross form of the bony surfaces forming the attachment points and produce no major changes in the architectural array of fibers in the muscle. Data presented here, combined with information from previous comparative studies of colubroid cephalic muscles, support the hypothesis that these muscles are limited in their potential variability by factors favoring parallel arrangements of fibers.

    Notes: Formation of lateral vessels in the esophageal region of Prosorhochmus americanus embryos and coelomogenesis in the pygidial region of larval Magelona sp. are examined and compared. Earliest vessel rudiments of P. americanus are composed of a compact band of mesodermal cells (mesodermal band), lying on a layer of extracellular matrix (ECM) and lacking intercellular junctions. Rudiments are surrounded by presumptive muscle cells. Rudiments at later stages of differentiation possess lumina of differing sizes formed by a separation of apposing cell apices (schizocoely). Aohagrens junctions are apparent between lining cels of vessels following cavitation, and overlying muscle cells exhibit many myofilaments. Mesodermal bands of the recognized coelomate, Magelona sp. consist of glycogen-rich, mesodermal cells resting on ECM and joined by adhaerens junctions. Some of the cells possess a rudimentary cilium. Coelom formation occurs as a splitting of the cell band as is the case for P. americanus. Recognition of an accepted mode of coelomogenesis in P. americanus, correlated with morphological details of adult nemertine vessels, affirms the view that nemertine vessels are coelomic homologues.

    Notes: Eggs of the asteroid Pisaster ochraceus demonstrate cortical granules, a thick vitelline membrane, and a poorly stained jelly coat similar to that seen on the eggs of other echinoderms. When fixed in the presence of alcian blue the jelly coat is seen to be made up of three regions, an inner layer consisting of a meshwork of fibres, a middle layer of thicker fibres, and a dense outer layer. At fertilization the cortical granules release their contents into the potential space between the vitelline layers and a low fertilization membrane consisting of the vitelline layer and a dense component of the corticle granule is formed. Initially the remaining contents of the corticle granules form an amorphous hyaline layer that fills the space between the plasma membrane and the fertilization membrane. At hatching a distinct hyaline layer is present. It persists at least to the bipinnaria stage and consists of four distinct layers. A similar layer is also located over much of the early embryonic endoderm but is lost from the regions involved in the formation of the mesenchyme cells, coelom, and mouth just before these events take place. Numerous large clear vesicles are located in the apex of all cells associated with a hyaline layer. Where the hyaline layer is lacking, only scattered vesicles are present suggesting that the vesicles may be involved in maintenance of the layer. Attempts to identify elements of the hyaline layer by immunofluorescence demonstrated that it appears to bind both antisera and control sera in a nonspecific manner.

    Notes: Six fiber types have been described in the ambiens muscle of red-eared turtles. These include one slow oxidative type, two fast oxidative types, two fast oxidative and glycolytic types, and one fast glycolytic type. Fiber types are non-randomly distributed throughout cross sections of the muscle. There is a decreasing gradient of oxidative staining and an increasing gradient of glycolytic staining along an axis from the superficial to deep regions of the muscle. The slow oxidative fibers are predominantly located within one or two fascicles of the superficial surface of the muscle. The fast glycolytic fibers are predominant in deep fascicles.In contrast to previous reports of histochemically monotypic intrafusal fibers in turtle muscle, ambiens muscle spindles have been observed containing one to eleven intrafusal fibers, including two fiber types. Fiber diameter and area are consistently smaller than observed in most extrafusal fibers. Spindles are predominantly located in superficial and cranial fascicles of the ambiens muscle and are located in regions characterized by extrafusal fibers with high oxidative activity.

    Notes: A study of neuromast ontogeny and lateral line canal formation in Oreochromis aureus and Cichlasoma nigrofasciatum reveals the existence of two classes of neuromasts: those that arise just before hatching (presumptive canal neuromasts, dorsal superficial neuromasts, gap neuromasts, and caudal fin neuromasts) and pairs of neuromasts that arise on each lateral line scale lateral to each canal segment at the same time as canal formation. In the anterior trunk canal segment, each presumptive canal neuromast is accompanied by a dorsoventrally oriented superficial neuromast forming an orthogonal neuromast pair. It is suggested that each of these dorsoventrally oriented superficial neuromasts is homologous to the transverse superficial neuromast row described by Münz (Zoomorphology 93:73-86, '79) in other cichlids. It is further suggested that the longitudinal lines described by Münz (Zoomorphology 93:73-86, '79) are derived from the pair of superficial neuromasts that arise during canal formation. Distinct changes in neuromast topography are documented. Neuromast formation, scale formation, and lateral line canal formation are three distinct and sequential processes. The distribution of neuromasts is correlated with myomere configuration; there is always one presumptive canal neuromast on each myomere. A single scale forms beneath each presumptive canal neuromast. Canal segment formation is initiated with the enclosure of each presumptive canal neuromast by an epithelial bridge which later ossifies. The distinction of these three processes raises questions as to the causal relationships among them.

    Notes: Ultrastructural examination of the head kidney of Periophthalmus koelreuteri (Pallas) (Teleostei, Gobiidae) revealed that the nephronic tubule cells are bound by tight junctions and desmosomes with little intercellular space. The first proximal segment (PI) consists of low columnar cells with well developed brush borders, indented nuclei, and numerous apical endocytic vesicles and lysosomes. A second cell type possessing clusters of apical cilia and lacking brush border and lysosomes is occasionally found between PI cells. The second proximal segment (PII) is formed of high columnar cells with brush border, regular spherical nuclei and numerous mitochondria located between well developed infoldings of the basal membrane. Single ciliary structures protrude into the lumen from PI and PII cells. The distal segment is lined by low columnar epithelium with few microvilli, regular spherical nuclei, numerous scattered mitochondria, and microbodies. The collecting tubule cells are cuboidal with few euchromatic nuclei, some mitochondria, and secondary lysosomes.

    Notes: The feeding mechanism of the South American lungfish, Lepidosiren paradoxa retains many primitive teleostome characteristics. In particular, the process of initial prey capture shares four salient functional features with other primitive vertebrates: (1) prey capture by suction feeding, (2) cranial elevation at the cranio-vertebral joint during the mouth opening phase of the strike, (3) the hyoid apparatus plays a major role in mediating expansion of the oral cavity and is one biomechanical pathway involved in depressing the mandible, and (4) peak hyoid excursion occurs after maximum gape is achieved.Lepidosiren also possesses four key morphological and functional specializations of the feeding mechanism: (1) tooth plates, (2) an enlarged cranial rib serving as a site for the origin of muscles depressing the hyoid apparatus, (3) a depressor mandibulae muscle, apparently not homologous to that of amphibians, and (4) a complex sequence of manipulation and chewing of prey in the oral cavity prior to swallowing. The depressor madibulae is always active during mouth opening, in contrast to some previous suggestions.Chewing cycles include alternating adduction and transport phases. Between each adduction, food may be transported in or out of the buccal cavity to position it between the tooth plates. The depressor mandibulae muscle is active in a double-burst pattern during chewing, with the larger second burst serving to open the mouth during prey transport. Swallowing is characterized by prolonged activity in the hyoid constrictor musculature and the geniothoracicus.Lepidosiren uses hydraulic transport achieved by movements of the hyoid apparatus to position prey within the oral cavity. This function is analagous to that of the tongue in many tetrapods.

    Notes: The cat hindlimb contains several long, biarticular strap muscles composed of parallel muscle fascicles that attach to short tendons. Three of these muscles -sartorius, tenuissimus, and semitendinosus -were studied by dissecting individual gold-stained fibers and determining the surface distribution of acetylcholinesterase-stained end-plate zones. In each muscle, fascicles were composed of muscle fibers that ran only part of the fascicle length and tapered to end as fine strands that interdigitated with other tapering fibers within the muscle mass. Most muscle fibers measured 2-3 cm in length. Fascicles of muscle fibers were crossed by short transverse bands of endplates (1 mm wide by 1-5 mm long) that were spaced at fairly regular intervals from the origin to the insertion of the muscle. The endplate pattern suggested that the fiber fascicles were organized into multiple longitudinal strips. In the sartorius, the temporospatial distribution of electromyographic (EMG) activity evoked by stimulating fine, longitudinal branches of the parent nerve confirmed that each strip was selectively innervated by a small subset of the motor axons. These axons appeared to distribute their endings throughout the entire length of the fascicles, providing for synchronous activation of their in-series fibers.

    Notes: This paper offers a model for the normalized length-tension relation of a muscle fiber based upon sarcomere design. Comparison with measurements published by Gordon et al. (′66) shows an accurate fit as long as the inhomogeneity of sarcomere length in a single muscle fiber is taken into account. Sequential change of filament length and the length of the cross-bridge-free zone leads the model to suggest that most vertebrate sarcomeres tested match the condition of optimal construction for the output of mechanical energy over a full sarcomere contraction movement. Joint optimization of all three morphometric parameters suggests that a slightly better (0.3%) design is theoretically possible. However, this theoretical sarcomere, optimally designed for the conversion of energy, has a low normalized contraction velocity; it provides a poorer match to the combined functional demands of high energy output and high contraction velocity than the real sarcomeres of vertebrates.The sarcomeres in fish myotomes appear to be built suboptimally for isometric contraction, but built optimally for that shortening velocity generating maximum power. During swimming, these muscles do indeed contract concentrically only. The sarcomeres of insect asynchronous flight muscles contract only slightly. They are not built optimally for maximum output of energy across the full range of contraction encountered in vertebrate sarcomeres, but are built almost optimally for the contraction range that they do in fact employ.

    Notes: Development and innervation of the lymph heart musculature of chicken, emu, rhea, and duck was studied by electron microscopy at posthatch ages from 3 days to adulthood. Development of innervation was monitored by acetylcholinesterase staining. Horseradish peroxidase was used to determine the extent of the transverse tubule network. Chickens were unusual among these birds in that lymph heart myocytes had already undergone a definitive differentiation and degeneration by 3 days. In ducks and ratite birds, lymph heart myocytes more slowly but progressively differentiate a cytomorphology that does not conform in all characteristics to cardiac or skeletal muscle and even resembles in some aspects, smooth muscle. Myofibrils become the dominant cytoplasmic structure, transverse tubules form ‘internal couplings’ with agranular reticulum cisternae, and ‘external couplings’ are formed between myocytes at myomyal junctions. The myomyal junctions also contain AChE-positive reaction product and some subplasmalemmal vesicles that lack a dense core. The lymph heart myocardium of ducks of 2 weeks demonstrated mitotic figures. In adult ducks the myosatellite cell numbers diminish and a characteristic pattern of myocyte degeneration appears. In juvenile ducks and ratites some myocytes differentiate to conductile cells, much as the conductile myocytes and myofibers of the blood heart. The lymph heart innervation is described, and the role of nerve in differentiation and maintenance of myocyte morphology in the lymph heart is discussed.

    Notes: Scanning electron micrographs of microcorrosion casts of the renal vascular system of Pseudemys scripta and Testudo hermanni show fairly well-developed, round glomeruli in the former (mean diameter of casts: 83.1 μm) and fewer but bigger, ovoid glomeruli (mean diameter of casts: 111.1 μm/131.6 μm) in the more arid-adapted. T. hermanni. Furthermore, the intrarenal development of the pertiubular capillary system differs in these two species. These relatively minor morphological differences correlate well with the major differences in the ecology of these species, as well as with physiological data on urine composition from the literature.

    Notes: The prominent accessory lobes of Lachi in birds are considered to be marginal nuclei; similar nuclei have been implicated in mechanoreceptive functions in snakes and lampreys. Reptile studies emphasized the involvement of the denticulate ligament with this mechanoreceptive function. This investigation examines the fine structure of the accessory lobes of Lachi in pigeons and their interaction with ligaments for features which might support such a mechanoreceptive function. In the lumbosacral area of the spinal cord, the lateral longitudinal ligaments and the ventral longitudinal ligament are hypertrophied. The ventral transverse ligaments are present only within the lumbosacral segments of the spinal cord and they interconnect with the lateral and ventral longitudinal ligaments. The lateral longitudinal ligament makes intimate contact with the spinal cord, and many glial processes from the spinal cord mingle with and are firmly attached to collagenous fibers of the ligament. The lobes lie dorsal to the lateral longitudinal ligament in the exact area where it interconnects with the transverse ligament. The lobe's multipolar neurons have a number of synaptic contacts but no unusual specializations were noted. Most of each lobe is composed of interdigitating saccular structures filled to varying degrees with flocculent material. The sacs are extensions of the cytoplasm of neuroglial cells, which also give rise to membranes surrounding neuronal processes and the sacs themselves. A possible functional relationship of the lobes and the ligaments of the lumbosacral spinal segments within the vertebral column is described.

    Notes: Several morphological and functional characteristics of the rat gastrocnemius medialis and tibialis anterior muscle were studied in young, adult, and old rats to assess the influence of growth. Antagonist muscles were studied to determine how changes of muscle architecture and functional characteristics are influenced by the demands of increased body weight and by the specific roles of these muscles in locomotion.Both muscles change drastically, for instance, in muscle length, volume, physiological cross-sectional area aponeurosis length, and their muscular geometry changes allometrically for both muscles. The relationships between muscle length, distance between origin and insertion, tendon length, and tibial length also change with growth. Both muscles are rather pennate, so that the increase of physiological cross-sectional area is a major factor in the determination of muscle length.No significant difference could be shown for fundamental physiological characteristics (i.e., functional characteristics normalized for muscular dimensions such as maximal work per unit volume). The changes of morphological and functional variables of both muscles parallel each other as is apparent from the index of antagonist characteristics, which is constant for all variables studied with the exception of muscle volume and tendon length.Consequently, the considerable and similar changes of TA and GM morphology and functional characteristics that take place during growth from approximately four weeks postnatally is not caused by changes of muscular material but by changes of the amount and architectural arrangement of the material involved.

    Notes: Between weaning and adulthood, the length and height of the facial skull of the New Zealand rabbit (Oryctolagus cuniculus) double, whereas much less growth occurs in the width of the face and in the neurocranium. There is a five-fold increase in mass of the masticatory muscles, caused mainly by growth in cross-sectional area. The share of the superficial masseter in the total mass increases at the cost of the jaw openers. There are changes in the direction of the working lines of a few muscles. A 3-dimensional mechanical model was used to predict bite forces at different mandibular positions. It shows that young rabbits are able to generate large bite forces at a wider range of mandibular positions than adults and that the forces are directed more vertically. In young and adult animals, the masticatory muscles differ from each other with respect to the degree of gape at which optimum sarcomere length is reached. Consequently, bite force can be maintained over a range of gapes, larger than predicted on basis of individual length - tension curves. Despite the considerable changes in skull shape and concurrent changes in the jaw muscles, the direction of the resultant force of the closing muscles and its mechanical advantage remain stable during growth. Observed phenomena suggest that during development the possibilities for generation of large bite forces are increased at the cost of a restriction of the range of jaw excursion.

    Notes: The exocrine glandular system of the nymphs and the adults of Dysdercus cingulatus were studied. The D. cingulatus nymphs present 3 dorso-abdominal glands (lying under the 3rd, 4th, and 5th abdominal terga) and a pair of dorso-lateral pygidial glands on the pygidium (tergum 8). Histological and ultrastructural studies show that the upper and lower walls of the dorso-abdominal glands differ in structure; 3 types of cells were described: epidermal cells, unicellular secretory cells, and multicellular secretory units. Each of these exocrine glands plays an important part in the behavior of the nymphs (gregariousness, alarm, defense). The morphology of the various glands is discussed, and the chemistry of their secretions and their biological functions are considered.

    Notes: The fully developed oral disc of the tadpole of Bufo bufo consists of dorsal and ventral labia bearing, respectively, two and three ridges bearing numerous horny denticles, a horny beak provided with jaw sheath serrations, and large lateral papillae that are borne by two cutaneous plicae. As development progresses toward metamorphosis, these structures gradually regress until they disappear. Each cusped clavate labial denticle adheres, by means of a thin peduncle, to a similar labial denticle fixed in the lip and formed by a group of three or four cells that keratinize gradually and thus present remarkable differences in their morphology. Once all the cells of a group have been converted into horny tissue, the denticle sheds and is replaced by the underlying one. The beak serrations also are horny structures; each consists of a columnar band of cells which undergoes a gradual keratinization. The horny cells that detach themselves at intervals, being replaced by those of the underlying anlagen. The labial denticles and the beak serrations keratinize in two distinct ways. In the former, the desmosomal filaments appear to play an important role whereas, in the latter, the keratin seems to be synthesized “ex novo” by the ribosomes.

    Notes: Spermatogenesis of the sea urchin Paracentrotus lividus has been studied at the ultrastructural level after conventional staining of thin sections and after en bloc silver staining. Cytoplasmic dense bodies are present in all steps of spermatogenesis except in late spermatids and spermatozoa. These bodies are closely associated with the development of centrioles and of the acrosomal vesicle during spermiogenesis. After it first appears, acrosomal vesicle is linked to the nuclear envelope by electron-dense material and subsequently acquires a dense core. Later the acrosomal vesicle moves to the apical pole of the cell while maintaining its connection to the nucleus. Although chromatin was highly condensed in the head of spermatozoa, one or more nuclear vacuoles within the nucleus were found to contain uncondensed chromatin fibers. Silver nitrate stains several nuclear and cytoplasmic structures. In the nucleus it stains the nucleolus, specific regions at the periphery of the chromatin, the synaptonemal complexes, the nuclear basal fossa, and the nuclear vacuoles of spermatozoa. In the cytoplasm, silver stains the cytoplasmic dense bodies, the material that connects the acrosomal vesicle to the spermatid nucleus, the spermatozoan subacrosomal and periacrosomal materials, the intercellular bridges of spermatids, and the centrioles. Silver staining is abolished by pretreatment with pronase E, suggesting that silver staining is due to protein.

    Notes: Gonadal differentiation in premetamorphic Bombina orientalis is described and staged. The pattern of events during differentiation in Bombina differs in several respects from that previously described in other anurans. The Bombina gonad initially develops on the ventral surface of the vena cava, where there is no pre-existent somatic genital ridge prior to the arrival of the germ cells. The sexually undifferentiated gonad does not have a distinct cortex and medulla; instead, medullary cells ingress from the mesonephric blastema during sexual differentiation. Formation of a testis or an ovary appears to depend on the ability (or lack of ability) of the medulla to invade the germ cell-containing cortex. In the germ line, sexual differetiation can be recognized by a premeiotic increase in oogonial cell volume.

    Notes: At least five nonporous sensilla with inflexible sockets (npsensilla) occur on each antenna of both sexes of adult Rhodnius prolixus. Externally the sensillum appears as a short, rounded peg set into a pit surrounded by a depression. A very electron-dense material occurs in the peg lumen and the inner aspect of the pit. Filamentous extensions of this material radiate into the overlying outlets.Each sensillum is innervated by three neurons with unbranched dendrites. Two dendrites extend to the peg tip and distally are covered by a dendritic sheath. The portion of these dendrites within the sheath contains a large number of microtubules. The third dendrite terminates near the base of the dentritic sheath and partially wraps around the other two dendrites. Three sheath cells are associated with each sensillum.Based on similarities in structure with sensilla of known function it is probable that the np-sensilla of R. prolixus are thermo-/hygrosensilla responding to cold, dryness and wetness. The sensilla have a number of structural similarities with insect rectal sheath cells known to absorb atmospheric water by electroosmosis. Possibly this process leads to volumetric alterations of cuticular elements associated with the dendrites and ultimately to mechanotransduction.

    Notes: The structures and distribution of encapsulated muscle receptors were examined in serial transverse sections of flexor carpi radialis in the adult cat. Four types of receptors (muscle spindles, Golgi tendon organs, paciniform, and Pacinian corpuscles) were identified. Their structures resembled those encountered in other limb muscles. Pacinian corpuscles were rare and occurred only in the external fascial coat of the muscle near its origin. The other three receptor types were distributed in an uneven but consistent pattern throughout the muscle. As noted previously (Gonyea and Ericson, '77), spindles were largely confined to a deep muscle region comprising less than 20% of the muscle volume, located directly between the long tendon of origin and the tendon of insertion. This region contains the largest proportion of type SO muscle fibers (Gonyea and Ericson, '77). Tendon organs and paciniform corpuscles were concentrated along the tendons that lined the spindle-rich muscle region. This region appeared to be composed of extrafusal fibers that were shorter and of more oblique pinnation than those in other regions. The localization of muscle receptors to the “oxidativex” core of the muscle in its direct line of pull may have functional implications for afferent input to the spinal cord which are discussed. In addition, the possibility is raised that there are more paciniform corpuscles in flexor carpi radialis (and possibly other muscles) than previously thought.

    Notes: The tree sloths, Bradypus and Choloepus, show unusual masticatory specializations, compared to each other and to other mammals. Both have an incomplete zygomatic arch with descending jugal process, a complex superficial masseter, a large temporalis and medial pterygoid musculature, and a lateral pterygoid with two heads. In Choloepus the deep masseter and zygomaticomandibularis are typical when compared to other mammals. However, in Bradypus there is an ascending jugal process from which enlarged and vertically oriented deep masseter and zygomaticomandibularis muscles originate. Although both sloths are folivores, the anterior teeth in Choloepus are caniniform, while those of Bradypus have lost such elongation. In both sloths the glenoid cavity is similarly located; however, in Bradypus the cranioman-dibular joint is raised above the occlusal plane, and the pterygoid flanges are elongated.Prediction of the evolutionary sequence of cranial changes from Choloepus- like (primitive) to Bradypus- like (derived) morphology is based upon the most parsimonious model of masseter-medial pterygoid complex changes for masticatory efficiency improvement. The model proposes that the condylar neck in Bradypus was elongated and that this single change predicated a series of other structural changes.Mandibular movement patterns in both sloths showed anteromedially directed unilateral power strokes as in other mammals. Puncture-crushing, tooth-sharpening, and chewing cycles are distinct in Choloepus, less so in Bradypus. The masticatory rate is slow in sloths compared to other mammals of similar body size, averaging 590 ms per cycle for Choloepus and 510 ms for Bradypus.

    Notes: The objectives of this research were to investigate the morphology of the head skeleton and muscles of the female mosquito, Culiseta inornata (Williston). The skeletal parts were examined after maceration in KOH. The attachments of muscles were determined by dissection. Observations were made with the aid of a dissecting microscope at 70× and lower. Each skeletal part and muscle is illustrated and described. Conclusions regarding the skeleton are as follows: (1) the clypeal area is composed of an anteclypeus and postclypeus, (2) the suture between the anteclypeus and postclypeus is rigid and cannot function as a hinge, (3) the dorsal wall of the labrum terminates at its union with the anteclypeus, (4) the dorsal and epipharyngeal walls of the labrum are united apically, (5) the gena and postclypeus are not separated by a suture, and (6) the labellum is composed of three segments and the furca, of some authors, is absent. Twenty-five muscles were identified, and the origin, insertion, and action of each is described. The tormo-epipharyngalis muscle is attached anterior to the cibarium and fulcral plates. Its origin is on the clypeal apodeme and the insertion is on the epipharynx. This result confirms earlier reports and disagrees with some recent authors. The maxillary teeth are not designed to draw the fascicle into the tissues, but the cervical and leg muscles accomplish the probing process during feeding on a host. An undescribed muscle of the mandible is reported.

    Notes: In addition to the cholinergic innervation described in the sphincter of the efferent filament arteries (Bailly and Dunel-Erb, ′86), an aminergic component has been demonstrated by specific techniques. The Falck fluorescence technique reveals a network of nerve fibers displaying a green fluorescence characteristic of catecholamines. At the ultrastructural level two types of fibers are present, one with clear vesicles and another with densecored vesicles. Axo-axonal synaptic relationships exist between the two types. Results of 5- and 6-OHDA (hydroxydopamine) treatments confirm the presence of an aminergic component.These observations support the notion of a dual innervation: cholinergic and adrenergic of, respectively, parasympathetic and sympathetic origin. The presence of presynaptic modulation is suggested. The aminergic component could inhibit or reduce the release of acetylcholine from cholinergic nerve endings. These results suggest that the sympathetic innervation modulates the vasoconstriction effect of the parasympathetic component.

    Notes: Among the acanthopterygian fishes, the Labridae possess the most highly integrated and specialized pharyngeal jaw apparatus. The integrated feature involves many osteological components and aspects of muscle form, architecture, composition, and function. The upper jaw articulates by means of a true diarthrosis with the pharyngeal process of the parasphenoid, whereas the lower jaw has established physical contact with the cleithrum. Complex muscle fusions have contributed significantly in the development of a double muscle sling operating the lower jaw. The original levator externus 4 fuses with the central head of the obliquus posterior, whereas the original levator posterior combines with the lateral head of the obliquus posterior as well as with the adductor branchialis 5.During the masticatory cycle, both upper and lower jaws undergo complex movement orbits resulting in shearing and crushing functions. Shearing occurs as the forward moving upper jaw collides with the dorsally held lower jaw. Crushing is effected by an extreme posterodorsal movement of the lower jaw against the retracted upper jaw, thereby establishing full occlusion of the teeth.The specialized morphological and functional design of the labrid pharyngeal jaw apparatus is similar to that found in cichlids. In sharp contrast to primitive acanthopterygian fishes, the Labridae and Cichlidae exhibit a spectacular morphological diversity that parallels their ecological diversification. Our combined functional and historical analysis has established a correlation between the complex integration of the pharyngeal jaw apparatus and morphological and ecological diversity in the Labridae and Cichlidae.

    Notes: Previous studies have shown the existence of a sphincter in the efferent filament artery of the teleost gill and its constrictory response to acetylcholine (ACH) and vagal stimulation. This study deals with the muscular organization of this sphincter and the distribution of its innervation as elucidated by degeneration methods and cytochemistry. The sphincter innervation is supplied by the protrematic vagus nerves. Nerve endings filled with cholinergic-type vesicles are located in close association with the adventitial smooth muscle cells and display a strong acetylcholinesterase (ACHE) activity. Section of the protrematic vagus nerve induces a nearly complete degeneration of the sphincter innervation. ACHE-positive nerve cell bodies are present both in the sphincter area and in the protrematic vagus nerve. These results suggest that innervation of the sphincter in the efferent filament artery is cholinergic through the activity of postganglionic axons of the parasympathetic system.

    Notes: The differentiation of fiber type characteristics in the anterior (ALD) and posterior (PLD) latissimus dorsi muscles is examined by the freezefracture technique in 1-, 7- and 30-day-old chicks. Several characteristics of plasma membrane (caveolae, rectilinear arrays, intramembranous particles) and sarcoplasmic reticulum which show fiber type differences in the adult ALD and PLD muscles are compared in the developmental stages. The caveolar density in the ALD fibers is about 20/μm2 at 1 day increasing to about 37/μm2 at 30 days, whereas in the PLD fibers it remains at about 20/μm2 during this period. The distribution of the caveolae in the two muscles is different from the begining; in the ALD fibers the caveolae are distributed throughout the plasma membrane and in PLD fibers they are patterned into clusters overlying the I band regions. The density of intramembranous particles of 1-day ALD and PLD plasma membranes appears similar, but by 7 days the particle counts in the sarcolemma of the ALD muscle are about twice as numerous as those in the PLD muscle. The rectilinear arrays are virtually absent in the ALD muscle, whereas in the PLD muscle their density is about 10/μm2 at 1 day and about 20/μm2 at 7 days. Already at 1 day posthatching the SR in ALD and PLD fibers has the adult configuration, i.e., an open irregular network in ALD fibers and periodically arranged tubules with triadic expansions in the PLD fibers. It is concluded that the membrane structure of ALD and PLD muscles is already different at hatching, and at 1 week the differences are identical to those of slow and fast fibers of the adult stage. The membrane changes, therefore, do not support the view that the ALD muscle undergoes a transitional, fast-type stage in posthatching chicks.

    Notes: Cytodifferentiation and hard tissue formation were studied in Anolis to collect information regarding the phylogenetic history of enamel and the functional significance of the events seen in the mammalian tooth during differentiation. The differentiation of the ameloblasts of Anolis, like that of mammals, shows two phases: In the early phase, the cells are short and rich in free ribosomes, in the late phase the cells elongate, develop an extensive rough endoplasmic reticulum, and the Golgi apparatus moves into that part of the cell next to the basal lamina (the cell apex). The early epithelial-mesenchymal interface resembles that of mammals, suggesting that early mechanisms of induction and epithelial-mesenchymal interaction are similar in Anolis and in mammals.Preameloblast processes and preameloblast-preodontoblast contacts in Anolis are rudimentary compared to those of mammals. While in mammals the preameloblast processes shape the future DEJ (dentin-enamel junction), their involvement in establishing the shape of the DEJ of Anolis is questionable. We suggest that the great development of preameloblast-preodontoblast contacts in mammals may simply increase the efficiency of inductive interactions between these cell types.

    Notes: The spermatozoa of Rhinolophus capensis are stored in the cauda epididymidis for about 10 months, 4 months prior to copulation and 6 months after copulation. Electron microscopy has shown the occurrence of sperm defects (mitochondrial proliferation, bending and coiling of the tail, and Dag defect) throughout the period of sperm storage. However, these defects are more common during the postcopulation period, when excess spermatozoa are being removed, suggesting that they may be associated with sperm degradation.

    Notes: Dramatic depression in granule volume density and size was measured in acinar cells of postnatal rat pancreas following the initiation of feeding. Volume density decreased about threefold from 45% at birth to 16% 2 days thereafter. Mean granule diameter decreased from 1.50 μm to 0.80 μm, an 85% decrease in corresponding granule volume. At the same time, numerical density approximately doubled. At 2 days after birth, cells with smaller granules had lower volume densities, and differences in mean granule volume between cells accounted for most of the differences in volume density. Although the distribution of granule diameter in newborns was lognormal, the distribution at 2 days was heavily skewed to larger sizes. This was the result of skewed distributions within individual cells and not an artifact of sampling. The results corroborate the central role of granule volume in determining changes in the volume density of zymogen granules in the pancreas and suggest that zymogen granules can act as capacitors that can change size as a function of the enzyme contained within.

    Notes: A comparative study of four genera of arvicolid rodents (Clethrionomys, Eothenomys, Alticola, and Synaptomys) presents the first complete description of limb myology for any member of this recently evolved, highly successful family. The study also identifies four forelimb and four hind-limb muscles that exhibit characters of value to systematic analyses within the group. In no instance was postcranial myology sufficient to distinguish among species of Clethrionomys or to distinguish Clethrionomys from Eothenomys. Moreover, in some instances Synaptomys, a genus traditionally presumed primitive within the family, possessed the apomorphic condition for a myological character when compared to the four genera of cricetine rodents that served for outgroup comparisons.

    Notes: Egg capsules of Syndisyrinx franciscanus, an intestinal parasite of sea urchins (Strongylocentrotus spp.), consist of a bulb, which contains the embryos, and a stalk-like filament. The wall of the bulb is about 12 μm thick and is composed of sclerotized proteins. The end of the bulb opposite the attachment of the filament bears a reticulum of hatching sutures. Transmission electron microscopy discloses that hatching sutures traverse the entire thickness of the capsule wall. The inner 9-10 μm of sutures are a uniform 20 nm in width and contain a trilaminar cementum. The outer 2-3 μm of sutures are 15 nm to more than 500 nm in width and contain an electron-lucent cementum. The latter may contain an irregular, median, electron-dense layer or, more commonly, electron-dense granules. The outside of some capsules is partially covered by a thin, electron-dense material.A previous study showed that sutures in intact capsules of Syndisyrinx franciscanus are not affected by host digestive fluids, but are severely weakened immediately prior to hatching owing to activities of the embryos. The hypothesis that the embryos secrete a hatching enzyme is supported by findings that sutures of intact capsules are not affected by externally applied trypsin, but become weakened when capsules are cut open and then incubated in trypsin. Scanning electron microscopy reveals that the outer parts of sutures often remain intact after hatching. We hypothesize that the ability of sutures to resist enzymatic attack from the outside, but not the inside, results from differences in the chemical properties of the cementums in outer and inner parts of sutures.

    Notes: The iris of the toad Bufo marinus is directly photosensitive and will constrict in response to light striking only the iris. This is true even when the iris is isolated from the rest of the eye, and therefore from reflex neuronal influences initiated in the retina. This autonomous response is probably mediated by the sphincter pupillae muscle, since no specialized photoreceptors are present in the iris, nor does the sphincter exhibit any specializations likely to subserve a purely photoreceptive function. The photosensitive sphincter appears typical of smooth muscle and, like mammalian sphincters, possesses many intercellular junctions. The iris possesses a well-developed neuronal plexus with fibers projecting into the sphincter muscle layer. Nerve terminals contain small, agranular (30-70nm) and large, dense-cored (80-120nm) vesicles. No consistent postsynaptic specializations are seen on any cells of the iris, including the cells of the sphincter muscle. The anterior pigment epithelial cells of the iris appear specialized and resemble the myoepithelial dilator muscle described by Kelly and Arnold ('72) for the iris of rats.

    Notes: The cell-lineage of Bithynia tentaculata L. has been studied through the formation of the fourth quartet of micromeres, i.e., the 81-cell stage. Up to the 28-cell stage, the cleavage pattern is radially symmetrical, and the divisions take place synchronously in the quadrants. Bilateral symmetry is established by the formation of the mesentoblast 4d at the future dorsal side. A resting stage preceding the formation of 4d is lacking. The mesentoblast 4d is formed earlier than the other fourth quartet cells, and it divides twice before 4a-4c are formed. Bilateral symmetry in the molluscan cross becomes apparent by different divisions of the tipcell and the basal cell in the dorsal arm. Dorsoventrality is progressively corroborated by the divisions in the second and third quartets of micromeres, which take a different course in the A and B quadrants on the one side and in the C and D quadrants on the other side.

    Notes: Two modes of shedding of rod disc membranes were observed by electron microscopy in bullfrog retinas illuminated for various periods from 10 min to 2 hr. One mode is “autonomous shedding” whereby rods shed disc packets directly into the subretinal space. Most of the discarded disc packets are subsequently brought into contact with villous apical processes of pigment epithelial (PE) cells and are ultimately engulfed by these cells. When some of the shed disc membranes remain in the subretinal space, it appears that these remanants may be phagocytized by ameboid phagocytes. The other mode is “cooperative shedding” whereby rods shed disc packets with the participation of pigment epithelial ensheathment. Shedding of a disc packet from a rod tip, and enclosing of the rod tip by a broad, sleeve-like apical process of a PE cell, take place simultaneously. The separated disc packets may be immediately engulfed by the PE cells without risk of failure. Both villous and sleeve-like types of apical processes of PE cells in the bullfrog lack pigment granules, in contrast to the finger-like apical processes that do contain pigment granules. Villous and sleeve-like apical processes that do contain pigment granules. Villous and sleeve-like processes therefore probably belong to the same category as the leaf-like apical processes of PE cells in mammalian retinas.

    Notes: High-speed cinematography shows that Suncus murinus (Crocidurinae) masticates fast (mean 5.5, 5-10 masticatory cycles per sec). Their grasping behavior is not stereotyped. The unilateral mandibular movements combine vertical, anteroposterior, and lateral displacements; and any masticatory sequence may include crushing, repositioning, shearing, and grinding components. Size and consistency of food influence the duration of individual chewing cycles. As food is transferred to the new working side, the chewing direction reverses, either near maximum closure or near maximum opening. An unfused mandibular symphysis permits tilting movements of the two halves of the mandible. Food may be squeezed between the lower incisors. The working side tilts outward during closing; this may improve shearing or grinding action. The closing phase is posteriorly directed. Thus, the masticatory movements of these shrews differ from those that have been described in many other mammals.

    Notes: The presence of adrenergic innervation was investigated in four different vascular segments of the neotenic tiger salamander, Ambystoma tigrinum, by histofluorescent staining for catecholamines. The segments were the respiratory section of the gill, the branchial shunt vessels, a vascular plexus in the pulmonary artery, and the dorsal aorta. No adrenergic fibers were detected in the respiratory section of the gill or the pulmonary arterial plexus. In contrast, the branchial shunt vessels contained both adrenergic varicosities and catecholamine-containing cell bodies. These cells resemble Type I cells of the mammalian carotid body and amphibian carotid labyrinth. Adrenergic innervation of the dorsal aorta was sparse and restricted to the adventitia. The results suggest that adrenergic nerves may directly regulate blood flow in the gill, and thus gas exchange, by controlling vascular resistance of the branchial shunts. The contractile state of the dorsal aorta may also be under adrenergic control. In addition, it is suggested that the adrenergic cells of the branchial shunts may serve a receptor function in being sensitive to arterial blood gases.

    Notes: Scanning and light microscopy investigations of the anterior alimentary canal of the pear psylla, Psylla pyricola Foerster (Homoptera: Psyllidae), revealed the morphology of the labium and stylets, as well as the presence of sensory structures and a valve in the precibarium. The labium consists of three telescoping segments with an internal labial groove, which surrounds and supports the stylet bundle. Also a part of the labial groove is the internal labial clamp. The stylet bundle is comprised of paired styliform mandibles and maxillae, which interlock to form the food and salivary canals. The stylet bundle proximal to the labium forms a large loop within a membranous crumena. When fully retracted the coiled stylets are under tension. Stylet extension generates increasing tension so that when retracted the stylets readily recoil within the crumena. Penetration of leaf tissues by the stylet bundle is dependent on the interaction between stylet muscles, opening and closing of the labial clamp, the barbed stylet tips, and the ventral position of the labium.Proximal to the crumena the paired stylets separate and diverge at the entrance of the precibarium, which is formed by the interlocking of the epi-and hypopharynges. There are 18 sensory structures in the precibarium, as well as a precibarial valve. These structures appear to be homologous to similar structures observed previously in aphids and leafhoppers. The morphology and the location of the precibarial sensilla suggest that, like the precibarial sensory organs of aphids and leafhoppers, they are gustatory and probably mediate acceptance or rejection of plant fluids, thus playing a major role in locating tissues for feeding.

    Notes: In seven species of lungless salamanders, family Plethodontidae, ranging from medium to very small in body size and from small to very large in cell size, the morphology of the eye and the retina were investigated. Haller's rule was only partially corroborated. While the smallest species had the relatively largest eyes, the largest two species, having the largest cells, showed the third and fourth largest eyes of the series. An effect of cell size was also found with regard to eye morphology. Small species with small cells as well as large species with large cells had relatively small retinae and relatively large lenses. In contrast, small eyes with relatively large cells had absolutely and relatively large retinae and relatively small lenses.The retinae of all investigated plethodontids showed a morphology typical for land vertebrates with two fiber and three nuclear layers. Rods, cones and double cones could clearly be distinguished. A fovea or area centralis was not found. Retina ganglion cell and photoreceptor counts show that the number of these elements was lower than in salamandrids. However, determination of the resolution power of miniaturized eyes based on morphological and behavioral data shows that this does not seem to constitute a functional disadvantage. The morphological and functional properties and constraints of eyes of miniaturized salamanders are discussed.

    Notes: Cells considered to be migratory in the cerebral cortex of adult lizards are ultrastructurally of two types. Nuclei in the first type have highly dispersed chromatin, creating a spongy appearance, whereas in the second type the chromatin is irregularly clumped. Both types of cells are closely associated with processes of radial ependymal glia cells, which perhaps orient their migratory pathways. Cells with spongy chromatin show an increase in cytoplasmic organelles and progressive chromatin condensation as they travel from the ependymal layer to the granular layer. Possibly these cells account for the neuronal increase that takes place in the granular layer during postnatal life. Cells with chromatin clumps are very scarce; ultrastructurally they resemble immature reptilian astroglia cells.

    Notes: The medulla of renculi from kidneys of Ringed seals (Phoca hispida) is completely enclosed by cortex except at the hilum. Within the renculus, the fibromuscular coat of the calyx separates from the transitional epithelium at the level of the corticomedullary junction, where the intrarencular arteries also diverge into the parenchyma. Flat ribbons of this stromal tissue form an arborized framework near the medullary side of the intrarencular arteries and the larger of the arcuate arteries derived from them. The ribbons, which are clearly distinct from periarterial connective tissue, are composed of coarse collagenous fibers, elastic fibers, and smooth muscle cells, all oriented in the direction of the long axes of the ribbons, and myofibroblasts. The proportion of smooth muscle cells decreases and that of myofibroblasts increases with increasing distance from the calyx. At the base of the medullary pyramid, the elements of the framework diminish in width and ultimately blend with the surrounding interstitial tissue. The stromal framework, or basket, is homologous with the Sporta perimedullaris musculosa of cetacean kidneys.

    Notes: A morphological comparison was made of the green livers of male and female lampreys (Petromyzon marinus L.) collected during the upstream (prespawning) migration. Light and electron microscope histochemistry for iron, and both thin sections and freeze-fracture replicas in the electron microscope, revealed some sexual dimorphism in these livers. Ferric iron is much more abundant in the liver of females and is present in the cytoplasmic matrix, in dense bodies, and in vacuoles of hepatocytes. The numerous vacuoles of females may be the deposition site of biliverdin and other bile components that would account for the darker green coloration of the liver compared to males. Hepatocytes in females are also characterized by prominent rough endoplasmic reticulum and Golgi apparatus that reflect the involvement of the cells in vitellogenesis. The presence of numerous lipid droplets in the hepatocytes of males indicates that the liver is an important storage site for fat. The lipid droplets are associated with electron-dense deposits of unknown nature. Large gap junctions typify the parenchymal cells of both male and female livers. Perisinusoidal and sinusoidal cells are similar to those in the nonparenchymal region in other vertebrate livers, namely, endothelial and Kupffer cells, lipocytes (Ito), and some granulated cells. The relationship of lipocytes to fibrous tissue and fibrogenesis is discussed.

    Notes: The fine structure of the hemocytes and nephrocytes in Argas (Persicargas) arboreus is described and compared with that of similar cells in other tick species and insects. The hemocytes are of three types: prohemocytes, with a relatively undifferentiated cytoplasm lacking granular inclusions and probably serving as progenitors of the other hemolymph cell types; plasmatocytes, containing abundant mitochondria, cisternae of rough endoplasmic reticulum (RER), and free ribosomes, as well as some small granular inclusions; granulocytes, the predominant cell type in the hemolymph, containing numerous granules of variable electron density and maturity, and pseudopodia-like processes on the cell surface. Plasmatocytes and granulocytes are phagocytic and possibly also have other functions in the tick body. Cells with intermediate features appear to be in a stage of transition from plasmatocyte to granulocyte.Nephrocytes contain vacuoles enclosing fibrillar material, some electrondense granules, and moderate amounts of the active organelles - mitochondria, RER, and ribosomes. The nephrocyte is surrounded by a basal lamina and its plasma membrane infolds to form many deep invaginations coated by a fine fibrillar material. Openings to these invaginations are closed by membranous diaphragms. Coated tubular elements connect the surface invaginations with large coated vesicles, which appear to be specialized for internalization of proteins from the hemolymph. The dense granules may represent an advanced stage of condensation of ingested protein and thus may be lysosomal residual bodies, or they may develop by accumulation of secretory products.

    Notes: In this study we examined the possible inductive role of the dental papilla from polyphyodont lizard tooth germs. Flank skin sheets of quail ectoderm enzymatically separated from dermal tissue were recombined with lizard tooth papillae and placed on semisolid medium and cultured for 2 days. Subsequently, the recombinants were removed and placed on the chorioallantoic membrane of chick hosts and incubated for 6 days. After this period of 8 days in explant, control tissues differentiated according to their own phenotypes. Lizard dental papilla alone differentiated as fibroblasts. Quail flank skin ectoderm differentiated into epithelial sheets. Intact lizard tooth buds developed into teeth with dentine and incipient enamel. In the best experimental recombinants, advanced and relatively well-constructed teeth were observed, with clear indications of hard tissue deposition in association with quail epithelium. The results show that mesenchyme of the adult lizard dental papilla and embryonic quail ectoderm of heterotopic origin are capable of carrying out the complex sequence of morphogenetic interactions involved in normal odontogenesis.

    Notes: Sperm development in Ornithodoros (Pavlovskyella) erraticus includes the formation of subsurface cisternae in the primary spermatocytes, which divide meiotically to secondary spermatocytes and ultimately to spermatids. During spermiogenesis the spermatid undergo morphological transformation including polarization of the nucleus and subsurface cisternae, formation of a cisternal tube, and modification of the subsurface cisternae to cellular processes surrounded by cisternal vesicles. Further transformation occurs after spermatids are introduced into the female. The spermatid cisternal tube now invaginates to form an inner cord surrounded by an outer sheath. The invaginated inner cord elongates anteriorly as the outer sheath continues to invaginate posteriorly during spermiogenesis. With further elongation, the spermatid membrane ruptures anteriorly, leaving the inner cord exposed as the outer surface of the maturing sperm. Posteriorly, the original plasma membrane invaginates to form an acrosomal canal which becomes surrounded by an acrosome. The hemispherical anterior end of the mature sperm is covered with rows of projections separated from the remainder of the sperm by a row of fringed processes. Except for the posterior end, the rest of the sperm is covered by longitudinally distributed electron-dense cellular processes and an outer mat of more electron-lucent tubular elements. Mitochondria and bundles of microfibrils are found beneath the cellular processes. Microfibrils are suggested to be the principal contractile organelles responsible for sperm motility. Cellular processes appear to be the main external motile structures, while movements of tubular elements and fringed processes may also contribute to sperm motility.

    Notes: Marlins, sailfish, and spearfishes have a heat-producing tissue beneath the brain and adjacent to the eyes. This tissue warms the brain and eyes while the rest of the body remains at water temperature. The heater tissue is derived from the superior rectus eye muscle. Only a portion of this eye muscle contains normal skeletal muscle tissue; the rest consists of the modified muscle tissue that is associated with heat production. The heat-producing portion is supplied with blood through a countercurrent heat exchanger that originates from the carotid artery. The vascular rete prevents the heat being produced by the tissue from being dissipated at the gill. An unusual circulatory supply to the eyes and brain is associated with the presence of the heater tissue in these fishes.

    Notes: The transport of sperm in the cloaca and adjacent regions of the female red-spotted newt was examined. It was found that within 1 min after sperm were introduced into the vent, they progressed in a random pattern past the apertures of the spermatheca (the gladular, sperm storage organ that opens from the anterior roof of the cloaca) forward to the anterior end of the cloaca and on into the posterior regions of the hindgut and bladder. Sperm did not enter the dorsal recess of the cloaca into which the oviducts and ureters open. After 1 day, few sperm remained within the cloaca lumen. Sperm were not transported into the cloacae of artifically inseminated, anesthetized females without prior administration of norepinephrine to their cloacal mounds. Treatment of the cloacal mounds of naturally inseminated females with an antagonist of neuromuscular transmission (lidocaine) decreased the numbers of sperm in the anterior cloaca relative to those of saline-injected control specimens. Neither dead newt sperm nor live rabbit sperm entered the spermatheca. Rabbit sperm, however, entered the oviduct. It is argued that passive and active mechanisms of sperm transport work in concert. Contractions of smooth muscle, which may be initiated during courtship, probably serve to draw sperm passively into the cloaca and up to and beyond the apertures of spermathecal tubules, but sperm, once in the vicinity of those apertures, probably swim actively into them.

    Notes: We recently described basal intracytoplasmic tubules that arise from the basal plasma membrane of rat retinal pigment epithelial cells (RPE). They are a previously undescribed ultrastructural specialization of the basal plasma membrane in addition to the well-known basal folds. This report describes similar tubules in the RPE cells of the rabbit, hamster, and kitten. As in the rat, the tubules in the hamster and kitten ramify singly through the basal cytoplasm and bear no special relationship to any other organelle. In the rabbit, however, stacks of closely apposed tubules frequently abut the large lipid droplets in their RPE cells. Although the function of the tubules is unknown, their occurrence in several different mammals suggests they are involved in the function of RPE cells in general.

    Notes: The keratose demosponge, Dysidea etheria, secretes a skeletal network of interconnecting spongin fibers and incorporates sand grains and other foreign particles into the fibers. Previous studies showed that foreign particles enhanced fiber growth and increased the stiffness of skeletal fibers. The present study uses light and electron microscopy to study the mechanism of particle incorporation. These histological observations show that particles become engulfed into thickened regions of the mesohyl of the dermal membrane, whereupon ameboid cells contact particles and secrete an electron dense layer of extracellular material onto the particle surface. Within several days, particles are moved towards primary skeletal fibers located at cone-shaped protuberances on the sponge surface called conules. Particles accumulate at conules and are consolidated by secretion of spongin at the growing fiber apices. The observations suggest that coordinated migration of groups of mesohyl cells control particle transport to conules and that patterns of cell migration are associated with the structural organization of the dermal membrane. Particle transport may be homologous and analogous to spicule transport in other demosponges.

    Notes: Fossil and extant dipnoans form a well-defined group of osteichthyans. Tooth plates, a feature in common for extant and the majority of fossil dipnoans, are not found in all dipnoans. Nevertheless primitive dipnoans can be defined by 21 characters of the head skeleton: bone arrangement in the posterior part of the skull roof, relation between supraorbital sensory line and bones, five extrascapulars, ossified (soft in post-Devonian dipnoans) upper lip, lack of premaxilla and maxilla, number and arrangement of bones in cheek region, lack of coronoids, the presence of an adsymphysial plate, ossified (soft in post-Devonian dipnoans) lower lip, relationship of oral and mandibular sensory canal to “infradentaries,” course of neurovascular system in lower jaw, symphysial tubuli in lower jaw, gular-shaped submandibulars, anterior naris at the edge and posterior naris within the mouth, median contact of pterygoids back to jaw articulation, posterior position of parasphenoid (buccohypophysial foramen very anterior in parasphenoid), unpaired “vomer,” autostyly, neurocranial support of posterior dermal skull roof, no isolated hypobranchials, and pharyngobranchials reduced or lacking. These 21 features distinguish the dipnoans from all other sarcopterygians. The Lower Devonian genus Diabolepis, which is said by some authors to have a closer relationship to dipnoans than to any other sarcopterygian, is considered to be inadequately known at present for definite statements about its relationship.

    Notes: The dermal skeleton of lungfishes is primitively composed of thick cosmine-covered bones and scales. Many Devonian lungfishes exhibit cosmine, while others have denticles of dentine and enameloid that may be arranged in several superposed layers; still others show denticles early in ontogeny superseded by cosmine later. Cosmine is a set of hard and soft tissues that includes bone, dentine, and enameloid, and a vascular and sensory porecanal network. It is the latter that uniquely distinguishes cosmine from other forms of the dermal skeleton and its presence determines the arrangement of the skeletal tissues. I have proposed that the appearance of denticles or cosmine is dependent on the stage of development of the pore-canal system, which may be fully developed as a network or arrested at the free neuromast stage. Other evolutionary trends in the dipnoan dermal skeleton have resulted from repression of the tissue interactions that produce the skeletal tissues.

    Notes: Remarkably little is known about the biology of the four Protopterus species, apart from certain detailed studies on their nesting behavior and estivating habits. What information we do have indicates that the species are essentially omnivorous carnivores (especially as predators on molluscs) and that they occupy a wide variety of habitats both lentic and lotic. As obligatory air-breathers able to survive temporary and sometimes extended desiccation of a habitat, lungfishes are often permanent residents in areas from which most actinopterygian fishes are excluded.All four species are able to survive prolonged dry periods. The methods they employ in so doing are varied, and include the secretion of subterranean cocoons, lying-up in water-filled subsurface burrows, or simply burrowing into moist regions of the substrate. Some populations of at least two species live in permanent water and so do not estivate, although they apparently retain the ability to do so.Three of the four species spawn in some form of seemingly constructed or prepared nest. The architecture of these nests shows marked inter- and intraspecific variability and is likely to be determined largely by various environmental factors. All three species show some type of parental care. The breeding biology of the fourth species, P. amphibius, is still unknown.Other aspects of the breeding biology and behavior of Protopterus require a great deal more investigation, as does the biology of the young.

    Notes: This paper reviews the cardiorespiratory morphology and physiology of the living lungfishes, in the special context of their highly effective use of both air and water for gas exchange. Particular emphasis is placed on describing those features of the circulatory and respiratory systems that distinguish Neoceratodus from the Lepidosirenidae (Protopterus, Lepidosiren), and which, in turn, distinguish lungfishes from other aquatic vertebrates. Morphological and physiological characters that represent the plesiomorphic condition for the living Dipnoi are indicated (e.g., separate atrial chambers, vertical septum in ventricle, pulmonary veins, conal valves, twisting of bulbus cordis), as are those characters that may be shared derived features of the Lepidosirenidae (e.g., paired lungs, reduced anterior gill arches, well-developed spiral valve in conus). Morphological and physiological comparisons and contrasts with tetrapods are made to elucidate systematic relationships of the Dipnoi with other vertebrates.

    Notes: The phylogenetic affinity of lungfishes has been disputed since their discovery, and they have variously been considered the sister group of actinistians, the sister group of amphibians, or equally related to actinopterygians and crossopterygians. Previous discussions of these hypotheses have considered neural characters, but there has been no general survey of the nervous systems of sarcopterygians that examines the bearing of neural characters on these hypotheses in the context of a cladistic analysis. Such a survey of representatives of all living sarcopterygian groups reveals at least twenty-three characters that are possible apomorphies at some hierarchical level among sarcopterygians. Neural synapomorphies corroborate the phylogenetic hypotheses that actinistians, amphibians, and dipnoans are each monophyletic taxa. The hypothesis that Latimeria is the sister group of amphibians is the least corroborated, as only a single possible synapomorphy, presence of cervical and lumbar enlargements of the spinal cord, supports this hypothesis. The hypothesis that lungfishes are the sister group of amphibians is supported by two possible synapomorphies: loss of a saccus vasculosus and the presence of neurocranial endolymphatic sacs. The hypothesis that actinistians are the sister group of lungfishes is the most corroborated, based on five possible synapomorphies: presence of a superficial isthmal nucleus, a laminated dorsal thalamus with marked protrusion into the third ventricle, olfactory peduncles, evaginated cerebral hemispheres with pronounced septum ependymale, and electroreceptive rostral organs. However, all five characters may be plesiomorphic for bony fishes. The nervous systems of Latimeria and Neoceratodus are very similar to each other, as are the nervous systems of lepidosirenid lungfishes, caecilians, and salamanders. If Neoceratodus is the most plesiomorphic species of living lungfishes, then lepidosirenid apomorphies may have arisen by paedomorphosis. Our inability to examine the neural characters of a relevant outgroup (rhipidistians) may result in many sarcopterygian plesiomorphic characters being interpreted as apomorphic characters, due to the wide distribution of paedomorphic characters among living sarcopterygians and their possible resemblance to plesiomorphic characters present in living outgroups that can be examined.

    Notes: The major anatomical divisions of the cerebellum of the European eel, i.e., corpus cerebelli, lobus vestibulolateralis, and valvula, were studied morphologically and morphometrically. There were differences in cerebellar cytoarchitecture and gross morphology in two stages of the eel life cycle, the trophic stage (yellow eel), and the reproductive stage (silver eel), which are characterized by different degrees of swimming activity. The principal differences between silver and yellow eels in the cytoarchitecture of the corpus cerebelli and the lobus vestibulolateralis were in distribution of Purkinje or Purkinje-like cells in the molecular layer, which is wider in silver eels, in part because of a decreased thickness of the granular cell layer. In the silver eel, the scattering of Purkinje cells was more evident in the lobus vestibulolateralis where the molecular layer is also thicker. The data indicate the transition from the yellow eel to the silver eel is characterized by a migration of granule cells from the ganglionic cell layer to the internal granular layer and by a further development of molecular layer components, e.g., parallel fibers, Purkinje-cell dendrites, etc. In contrast, the thickness of the granular layer and of the Purkinje cell layer, limited to the lower part of the valvula, decreased. There is also a slight increase of cerebellar volume in the silver eel. The volume of the lobus vestibulolateralis was constant. Hypertrophy of the valvula and eminentiae granulares is observed and is due to the migration of cells from the granular layer of the corpus cerebelli whose volume slightly decreases. Perhaps the lobus vestibulolateralis also contributes to the increased volume of eminentiae granulares. Our findings suggest that the cerebellum continues to develop during the passage from the trophic to the reproductive stage of the eel. The appearance of new afferents from the lateral line which becomes more visible in the silver eel probably completes cerebellar ontogeny.

    Notes: Xenopus laevis froglet forelimbs normally respond to amputational injury by forming a heteromorphic cartilaginous rod-shaped outgrowth. However, partial denervation of a forelimb by ablation of the N. radialis or the N. ulnaris, followed in 2 days by amputation through the mid radius-ulna, results in a size deficiency of the regenerative outgrowth 14 and 21 days postamputation. The decreasing quantity of forelimb innervation, as a result of partial denervation by 55 or 45%, apparently has a graded effect on the cell population and on the extent of cartilage development in the outgrowth. As a consequence of amputational injury, a nerve independent response of the periosteum was also found. This response produced considerable thickening in the periosteum and was due to cell proliferation in both the control and denervated cases.

    Notes: Serial transverse paraffin sections of intrafusal muscle fibers of spindles from the extensor pollicis and the extensor digitorum communis of ducks show that only one type of intrafusal muscle fiber exists, based on the mid-equatorial nucleation pattern, diameter, and length. Although the overall range in fiber diameter at the mid-equatorial region is between 4.2-20.0 μm, the average caliber is 10.4 ± 3.18 μm (S.D.) for spindles of the extensor pollicis and 9.3 ± 2.11 μm (S.D.) for spindles of the extensor digitorum communis muscles. The range in spindle length for the extensor pollicis is 290-2,090 μm, average 1,120 ± 569 μm (S.D.), and for the extensor digitorum communis 1,160-2,500 μm, average 1,745 ± 367 μm (S.D.). Therange in number of fibers per spindle for the extensor pollicis muscle is 5-12, average 8.2, and for the extensor digitorum muscle it is 1-11. In the extensor digitorum communis, there appear to be two groups, based on fiber number. Spindles of one group have a range of 5-11 fibers per spindle with an average of 7.2, whereas the second group has a range of 1-4 with an average of 2.7 fibers per spindle. The second group of spindles constitutes 52.5% of the 40 spindles studied, and of these 7.5% were monofibril spindles, 15.0% difibril, 17.5% trifibril, 12.5% quadrifibril spindles.

    Notes: Each of the paired salivary glands of third instar larvae of the humpbacked fly Megaselia scalaris is a bag-like structure with a short neck region from which a single duct emerges. The two ducts form a common duct that empties into the ventral region of the pharynx near the mouthparts. The wall of the glands and ducts consists of a simple squamous epithelium that rests upon a connective tissue layer. Cells in the neck are less flattened than those found elsewhere. The basal surfaces of the cells are infolded most deeply in the neck and the least in the duct. The apical surfaces of the cells possess microvilli except in the duct where the apices of the cells are covered by a complex extracellular layer. This layer displays circularly arranged folds that accommodate a thread-like supportive structure resembling taenidial threads of tracheae. Elaborate junctional complexes are associated with the lateral surfaces of the cells. Elements of these complexes include a zonula adherens, a series of pleated septate desmosomes, and conventional desmosomes. The cytoplasm of the glandular cells is filled with RER and other organelles normally seen in cells that export proteins and mucosubstances. Secretory material found in the lumens of the glands reacts only moderately with the PAS procedure but more strongly with alcian blue and methods that demonstrate proteins. The nuclei of the glandular cells contain single large nucleoli and polytene chromosomes whose banding is rather indistinct. Treatment with EDTA produces detrimental effects on all of the foregoing ultrastructural features of the glands and ducts.

    Notes: The fate and possible roles of the cytoskeleton in the process of conjugation in the hyptrich ciliate Euplotes aediculatus were investigated. Following the coalescence of the plasma membranes of the conjugant cells, a fusion zone or bridge of cytoplasm contributed by both partners is constructed. The sub-alveolar microtubule layers of the vegetative cell cortex remain in place to define the fusion zone boundaries after cell union. The initial fusion zone consists primarily of featureless ground cytoplasm; soon the ground plasm becomes crowded with microtubules and anastomosing smooth endoplasmic reticulum, which become displaced only late in conjugation as the migratory pronuclei are exchanged between partners. Fusion zone microtubules, functioning in some undetermined way, may be involved in the nuclear migration. Resorption of the posterior portion of each partner's buccal apparatus results in the degradation of the component cilia within acid phosphatase-positive autophagic bodies. Silver staining for light microscopy shows that the late fusion zone contracts forward from the posterior border, then constricts to separate the conjugants. In some separating pairs remnants of a microfilamentous assembly are seen at the posterior edge of the fusion zone; the full extent of this system may be masked by partial degradation due to osmium tetroxide fixation. Treatment of conjugants for 6 hours with cytochalasin B prevents separation, possibly through inhibition of the actin-like microfilament assembly in the fusion zone. The observations and experiments favor a model of cell separation following conjugation in which the fusion zone is resorbed by motile or contractile processes occurring within or around the fusion bridge itself.

    Notes: The development of the tentacle, a chemosensory and perhaps tactile structure unique among vertebrates to gymnophione amphibians is described in Dermophis mexicanus and Gymnopis multiplicata. The tentacle is associated with the vomeronasal organ and its glands, and utilizes several structures usually associated with the eye, such as the Harderian gland, the retractor and levator muscles, and their nerves. Innervation of the tentacle itself is from the trigeminal nerve. We present an hypothesis that the tentacle originated from modified eye components.

    Notes: Two types of calcareous spicules occur abundantly in Herdmania momus, a solitary pyurid ascidian with a worldwide warm water distribution. The large spindle-shaped body spicules are 1.5-2.5-mm long and are located primarily in the mantle, siphons, and branchial basket. Each body spicule possesses 100 or more rows of overlapping, unidirectional fringing spines. Numerous body spicules occur regularly spaced within a long common sheath of complex structure, and there are many sheaths per animal. Between neighboring body spicules and overlying the fringing spines are the tightly connected pseudopodial sclerocytes. Spine formation is hypothesized to occur within these cells. The body spicules apparently continue to increase in size throughout the animal's life.The tunic spicules are about one tenth the length of the body spicules. They have 20-40 rows of unidirectional nonoverlapping fringing spines and a mace-shaped spiny base that anchors them at the tunic surface. They form quickly in individual spicular envelopes inside the tunic blood vessels over a 4-5-day period. Each tunic spicule then leaves its surrounding envelope and blood vessel, passes into the tunic, and ultimately protrudes through the outer surface of the tunic. An organic covering inside the envelope closely adheres to the tunic spicules and stains with toluidine blue. Dissolution of the CaCO3 mineral phase by EDTA or EDTA-cetylpyridinium chloride-formaldehyde reveals an intricately patterned organic matrix within or upon which the spicules develop.

    Notes: In all species of phylactolaemates, an individual successively produces daughter buds. Individual daughters are designated, according to the order of appearance, as first bud, second bud, and so on. The first bud appears precociously, while the mother is a pear-shaped vesicle. The idea of regarding a main bud, a duplicate bud, and an adventitious bud as a set is not tenable. In the budding region, the cystidal wall shows a constant wavy movement. In Plumatella colonies, branching occurs only when and where a bud of the second or higher order grows up to a zooid. A branch is composed of longitudinally arranged first zooids with the only exception being the most proximal one. The proportion of first zooids in a colony increases in the order of P. casmiana, P. emarginata, and P. repens. The frequency of branching, therefore, decreases in this order. The ancestrula germinated from the statoblast shows the highest activity of budding. The tendency that successively produced daughters of the ancestrula grow in alternate directions is most conspicuous in P. emarginata and least conspicuous in P. casmiana. Replacement budding occurs in these three species of Plumatella, but only under unfavorable culture conditions. The colony of Gelatinella toanensis is characterized by composite branches, each consisting of an axial branch composed of a series of first zooids and of stunted lateral branchlets. In Hyalinella punctata, multiple budding does not necessarily result in branching; zooids of different budding orders coexist in a branch. The genus Pectinatella comprises two species, P. gelatinosa and P. magnifica. Both produce massive colonies. In P. gelatinosa, the colony proper is sac-like with the convex basal wall, and the polypides can retract with the digestive tracts straight. Each individual (except the ancestrula) of this species produces a pair of daughter buds which are located bilaterally relative to the median sagittal plane of the mother. A left bud produces its first bud to the right, and vice versa. In P. magnifica, the colony proper is very thin and flat. When polypides are retracted, the digestive tracts are folded characteristically. Based on these and other results, phylogenetic relationships among the phylactolaemates are discussed.

    Notes: The gonads from three hermaphrodite species of different invertebrate phyla were studied at the ultastructure level. In the flatworm Dugesia biblica, male germ cells in different stages of development lay in the lumen of testicular follicles surrounded by overlapping parietal cells. The intercellular space formed by cytoplasmic extensions running parallel to the testis wall is occluded by septate junctions and by an electron-dense material. In the leech Placobdella costata, the testis is lined by a unicellular layer of parietal cells surrounded by densely packed connective tissue fibers. No specialized occluding junctions were found between the parietal cells; however, plasmalemma thickenings and electron-dense material in the intercellular space near to the testis lumen were observed. In the lumen, germ cells develop connected to cytoplasmic masses, the cytophore. In the land snail Levantina hierosolyma, male and female germ cells are found together in the same acini; each acinus is surrounded by a thick basement membrane. At the periphery of the acinus is the ovarian layer; centrally to it is the testicular layer. Intercalated between them is a double cellular layer of follicular cells and of Sertoli cells. The inter-Sertoli space is characterized by elaborate septate junctions. In the three species studied male germ cells develop within the lumina of compartments isolated from the somatic tissue. This separation is brought about by specialized septate junctions, and/or by electron-dense material between the cells that form the testis walls, and also by densely packed connective tissue fibers. Our observations strengthen the view that a male germ cell-somatic tissue barrier as described in the literature of the testes of vertebrates and of invertebrates from various phyla is of general occurrence in the animal kingdom.

    Notes: Oocytes from the land hermit crab, Coenobita clypeatus, in various stages of vitellogenesis were examined by light and electron microscopy. Early vitellogenic oocytes are characterized by accumulations of discrete vesicles of endoplasmic reticulum in the perinuclear cytoplasm. As oocytes develop, the endoplasmic reticulum becomes abundant, and numerous Golgi complexes are seen. There is a well developed Golgi-endoplasmic reticulum interaction. Within the confines of the reticulum are discrete intracisternal granules, which can be seen coalescing into electron-dense yolk bodies. Lipid accumulation is seen throughout the cytoplasm. Coincident with the burst of intra-oocytic metabolism are oolemma modifications and micropinocytosis, which provide ultrastructural evidence for extra-oocytic yolk production. The mature oocyte contains numerous yolk and lipid vesicles of varying electron density that comprise both intra- and extra-oocytic substrates.

    Notes: The histochemistry and ultrastructure (SEM and TEM) of the spermatheca of Biomphalaria glabrata was investigated to elucidate the function of this organ and to compare its structure and function to similar organs found in other species. The spermatheca has a debris-filled lumen surrounded by a thin wall of tissue. The cells adjacent to the lumen are of three columnar epithelial cell types. Two cell types have abundant microvilli and mammalian cell-like organelle distribution and morphology. The above cell types differ in the electron density of their cytoplasms, nuclear morphologies, and organelle content. The third cell type differs from the other two in its cytoplasmic makeup. However, the most distinctive difference is the presence of large numbers of cilia at the apical surface with no evidence of microvilli. These columnar cells rest on a basal lamina adjacent to a two to three cell thick muscle layer. The entire organ is surrounded by an adventitia of unusual morphology. Histochemical investigation demonstrated that DNAase, RNAase, and protease are present in the lumen, alkaline phosphatase is associated primarily with the microvilli, small amounts of acid phosphatase are concentrated in the midcell area of the columnar epithelium, and ATPase activity is localized in the muscle cells and just below the absorptive surface of the microvillous cells. The luminal contents and adventitial areas are Sudan Black B positive, all areas of the lumen and organ wall are PAS positive, the cell nuclei and amorphous masses in the lumen showed Feulgen staining, and large vesicles in the columnar cells were Oil Red O positive. Apparently, the spermatheca of B. glabrata is both a digestive and absorptive structure. Although this organ shares functional similarities with those found in opisthobranchs and terrestrial pulmonates, the epithelia of the spermatheca differ dramatically in these groups.

    Notes: In an attempt to investigate the relationships between allometry and locomotory adaptations, we studied the long limb bones of 45 species of insectivores and rodents. Animals ranged from a few grams to about 50 kilograms. Diameter and length of the bones and body mass (when known) were recorded. Regressions of diameter to length, diameter to body mass, and length to body mass were calculated by the least-squares and Model II, or major axis, methods.The results obtained do not agree with the predictions of either the theory of geometric similarity or the theory of elastic similarity. The discrepancies could be due to the fact that animals studied exhibit various modes of locomotion. Moreover, the allometric relationships of the different locomotor patterns are better reflected in insectivores and rodents than in other groups of mammals. The use of a single regression analysis seems to be inadequate when the sample includes a large range of body sizes.

    Notes: Sensilla that line the upper edge of the lip in the leech Hirudo medicinalis and that contain chemoreceptors required for feeding were examined in the scanning and transmission electron microscopes. The sensilla include two size-classes of ciliated button-like mounds - one about 35 μm in diameter and another about 10 μm in diameter. The larger sensilla are at the center of unpigmented patches of skin which are visible in the light microscope, while the smaller sensilla have not been previously described as distinct structures. Electron microscopy, though not light microscopy, shows that the lip sensilla differ markedly from the segmental sensilla of the leech, which have been shown to mediate mechanoreception and photoreception. In particular, the chemosensory lip sensilla contain multiciliated cells with cilia of a uniform length, whereas the segmental sensilla contain uniciliated cells with long, whip-like cilia, as well as multiciliated cells with short, stiff cilia. Thus, the two types of sensilla differ morphologically as well as functionally. In addition to the ciliated sensilla along the upper lip, structures consisting of a short, club-like process surrounded by granular material were observed inside the mouth. These structures may also be chemosensory organs.

    Notes: The architecture and fine structure of the epigastric hematopoietic nodules of the ridgeback prawn, Sicyonia ingentis, are described. The nodules consist of a highly branched series of tubules that contain the maturing hemocytes within a connective tissue stroma. Hemocytes can exit the hematopoietic nodules by penetrating through fenestrations in the endothelial cell layer into the central hemal space or by migrating through the outer later of capsular cells and associated collagen fibrils. Four hemocyte categories were observed: agranular, small granule with cytoplasmic deposits, small granule without cytoplasmic deposits, and large granule hemocytes. This classification was based upon the presence, size, and type of cytoplasmic granules and the presence of cytoplasmic deposits. Only agranular cells and small granule hemocytes without cytoplasmic deposits appeared capable of division. Intermediate stages were observed between agranular hemocytes and small granule hemocytes with deposits and between small granule hemocytes without deposits and large granule hemocytes, suggesting existence of two distinct hemocyte lines.

    Notes: This investigation is a light and electron microscopic description of the submandibular duct salivary bladder of the rat, a dilation of the distal end of the main excretory duct. The wall of the bladder consists of (1) a mucosa with pseudostratified epithelium, (2) a submucosal layer of connective tissue, and (3) an underlying layer of striated muscle. The pseudostratified columnar epithelium lining the bladder is composed of three cell types: (1) light cells, (2) dark cells, and (3) basal cells. The lamina propria contains bundles of collagen, attenuated fibrocytes, capillaries with fenestrated endothelia, and nerve fibers which enter the epithelial layer. The capillaries of the submucosa are not fenestrated. The morphology of the wall of this structure provides evidence that the primary fluid of the submandibular gland is modified in the bladder by transepithelial fluid and ion transport.

    Notes: The innervation and structure of the lung of the Australian snake-necked tortoise, Chelodina longicollis, was examined by using light microscopy including fluorescence histochemical techniques. The anterior lung was divided into a number of compartments with numerous alveolar spaces. The posterior lung was simpler and saclike in structure and alveolar spaces were absent. Smooth muscle fibers occurred in discrete muscle bands and in the walls of the septal bands. Ganglion cells occurred along nerve trunks throughout the lung but were more numerous in the posterior lung. Smooth muscle bands, the extrinsic pulmonary artery, and the arteries within the lung were sparsely innervated by adrenergic fibers. Substance P-containing sensory fibers were not demonstrated. The innervation and structure of the lung are compared to published work on other reptiles.

    Notes: Ultrastuctural changes in the intestinal connective tissue of Xenopus laevis during metamorphosis have been studied. Throughout the larval period to stage 60, the connective tissue consists of a few immature fibroblasts surrounded by a sparse extracellular matrix: few collagen fibrils are visible except close to the thin basal lamina. At the beginning of the transition from larval to adult epithelial form around stage 60, extensive changes are observed in connective tissue. The cells become more numerous and different types appear as the collagen fibrils increase in number and density. Through gaps in the thickened and extensively folded basal lamina, frequent contacts between epithelial and connective tissue cells are established. Thereafter, with the progression of fold formation, the connective tissue cells become oriented according to their position relative to the fold structure. The basal lamina beneath the adult epithelium becomes thin after stage 62, while that beneath the larval epithelium remains thick. Upon the completion of metamorphosis, the connective tissue consists mainly of typical fibroblasts with definite orientation and numerous collagen fibrils. These observations indicate that developmental changes in the connective tissue, especially in the region close to the epithelium, are closely related spatiotemporarily to the transition from the larval to the adult epithelial form. This suggests that tissue interactions between the connective tissue and the epithelium play important roles in controlling the epithelial degeneration, proliferation, and differentiation during metamorphic climax.
