    Notes: Summary A microsporidian parasite, Nosema sp., has been found in the epithelium of the vas deferens of the spider crab, Libinia dubia. Cells were heavily infected with developing and mature spores. The spore mother cell possesses a dikaryon. The nuclear membrane shows a simple form of nuclear pore. Within the nucleus microtubules of the intercellular mitotic spindle are observed. The sporont undergoes cytokinesis to form the early sporoblast. The sporoblasts possess a dikaryon., and a centriolar plaque may be observed at the nuclear surface. The polar filament develops in association with a modified Golgi apparatus, consisting of numerous cisternae frequently filled with electron dense material, sometimes surrounded by several layers of membrane. The polar filament is formed as two hollow tubes, the inner tube being eccentrically placed within the outer one. Preparations treated with the periodic acid—silver methenamine technique for the detection of complex polysaccharides show a positive reaction in the Golgi, the sperical masses and associated membranes and in the polar filament. The developing spore wall also shows a positive reaction.

    Notes: Tissues of White Leghorn embryos of stages 17-45 and chicks of one day, two days, and three weeks of age were frozen, sectioned in a cryostat and, where appropriate, were fixed in cold calcium formol. Acid phosphatase, non-specific esterase, adenosine triphosphatase, 5-nucleotidase, non-specific glycerophosphatase, nucleotidediphosphatase, and glucose-6-phosphatase were localized in these tissues. Ribonucleic acid, acid mucopolysaccharides, triglycerides, and neutral fats were localized in tissues fixed with FAA and embedded in paraffin.Positive acid phosphatase reactions were obtained in the epithelium of the trachea and esophagus at all stages of development. 5-nucleotidase was found in the muscularis mucosae of the esophagus at all stages. Non-specific esterase appeared with histodifferentiation of the esophageal epithelium. Ribonucleic acid was localized in the basal regions of the epithelium.Mucous glands of the esophagus are rich in ribonucleic acid and acid phosphatase at all stages of development. With histodifferentiation and the onset of secretion of sulfated acid mucopolysaccharides, the glands and their ducts become highly reactive for adenosine triphosphatase and nucleotide-diphosphatase, indicating a role of these enzymes in secretion.

    Notes: Sperm enter the anterior vas deferens individually in the spider crab male. There they become surrounded by secretion products from the cells of the vas deferens, and are compartmentalized into spermatophores of varying size. The anterior vas deferens can be divided into three regions. The epithelium of the anterior vas deferens varies regionally from low to high columnar. The cytoplasm contains vast arrays of rough endoplasmic reticulum and Golgi complexes but few mitochondria. Intercellular spaces contain septate junctions, gap junctions and vesicles.Once the spermatophores have been formed in the anterior vas deferens, they are moved posteriorly to the middle vas deferens where they are stored and surrounded by seminal fluids. The epithelial cells of the middle vas deferens contain large amounts of rough endoplasmic reticulum and Golgi complexes. Numerous micropinocytotic vesicles appear, forming at the cell surface and within the apical cytoplasm. Their suggested function is the resorption of secretion products of the anterior vas deferens which initiated compartmentalization of the spermatozoa into spermatophores.The posterior vas deferens functions primarily as a storage center for spermatophores until they are released at the time of copulation. Seminal fluid surrounding the spermatophores is produced in this region as well as in the middle vas deferens. The cells of this region contain vast arrays of vesicular rough endoplasmic reticulum and Golgi complexes. The cells are multinucleate. Microtubules are numerous throughout the length of the cells and appear to insert on the plasma membrane.

    Notes: Succinic dehydrogenase, NAD and NADP diaphorases, choline oxidase, d-amino acid oxidase, α-glycerophosphate dehydrogenase, glutamic dehydrogenase, lactic dehydrogenase and aldolase were identified histochemically in the esophagus and proventriculus of the developing chick embryo. In general, the deep glands of the proventriculus reacted more strongly than the epithelium and mucous glands of the esophagus to the tests. The intensity of the enzymatic activity seems correlated with the number of mitochondria and metabolic activity of the cells.

    Notes: Ultrastructural study of previtellogenic oocytes found in cystlike clusters scattered throughout the length of the bilobed ovary of the hermit crab Coenobita clypeatus shows a high nuclear:cytoplasm ratio. Large, round nuclei containing synaptinemal complexes serve as good temporal markers for identification of previtellogenic oocytes. The cytoplasm contains many smooth-membraned vesicles filled with granules and probably of nuclear origin. In addition to its complement of Golgi complexes, endoplasmic reticulum, mitochondria, and free ribosomes, the cytoplasm also contains stacks of annulate lamellae, a feature not previously described for decapod oocytes. Typically, the previtellogenic oocyte with its accumulation of ribosomes has the appearance of a nonsynthetic cell preparing to go through a metabolic transition.

Alkaline phosphatase was identified using the azo-dye or Gomori techniques.Alkaline phosphatase activity is found in the ventral mesenchyme of the esophagus surrounding the epithelium to stage 34. At stage 38 activity appears in the basal layers of the epithelium and is subsequently lost in the mesenchymal tissues. This activity persists in the epithelium throughout development and following hatching. At 16 days the mucous glands arise as solid buds of epithelium projecting into the tunica propria. These buds are highly positive for alkaline phosphatase. As differentiation of the glands becomes complete they and their ducts lose their positive phosphatase reaction.On the fourth day of development the trachea is found lying ventral to the esophagus. Mesenchyme has condensed around the tracheal epithelium by the fifth day, at stage 37 (11 days) the first cartilaginous rings appear, and by stage 38 (12 days) all are completely formed. During the third week of development the trachea increases in size. Pseudo-stratified columnar ciliated epithelium begins to appear at 15 days. Alkaline phosphatase is highly reactive in the mesenchyme surrounding the epithelium prior to cartilage formation. After the cartilages have formed the reaction diminishes and in late stages of development phosphatase is found only in the epithelium.

Ribonucleic acid, proteins and acid mucopolysaccharides were identified in tissues fixed in FAA.Acid phosphatase, nucleotide-diphosphatase, adenosine triphosphatase, succinic dehydrogenase, ribonucleic acid and proteins were present in the cells of the deep glands at all stages of development. Alkaline phosphatase and 5-nucleotidase were found only in mesenchymal derivatives of the proventriculus. After the chick begins swallowing and digesting albumen, enzymatic activity increased and non-specific esterase became very reactive.The surface epithelium is covered with a mucous coat. Ribonucleic acid, non-specific esterase, acid phosphatase and nucleotide-diphosphatase were localized in the basal portions of the epithelial cells. The functional significance of these different patterns is discussed.
