    Description: Caffeine (1,3,7-trimethylxanthine) is a biotransformation product of theophylline (1,3-dimethylxanthine) in the human fetus. Liver explants, obtained from human fetuses with gestational ages of 12 to 20 weeks, were incubated with theophylline and produced caffeine and, in lesser amounts, 1,3-dimethyluric acid and 3-methylxanthine. These findings suggest that the predominant pathway in theophylline metabolism in the fetus and newborn infant is the methylation reaction producing caffeine. This may contribute to the neonate's exceedingly slower elimination of caffeine relative to theophylline.     Description: Vascular casts of the pituitary gland have demonstrated a paucity of veins extending from the adenohypophysis to the systemic circulation and have suggested that some adenohypophyseal venous blood returns to the neurohypophysis. The neurohypophyseal capillary bed may function as a vascular switch and in this article a series of 14 questions are proposed regarding the vascular dynamics of the pituitary.     Description: Exposure of L1210 leukemia cells first to 0.1 to 100 micromolar methotrexate and then to 10 micromolar 5-fluorouracil produces a synergistic effect on the number of cells killed in culture. Methotrexate dose-related increases occur in the concentrations of intracellular 5-fluorouracil ribonucleotides and 5-fluoro-2'-deoxyuridylate and in the incorporation of 5-fluorouracil into RNA. These increases are correlated with increased concentrations of intracellular phosphoribosylpyrophosphate.     Description: Photosensitive membrane structures in the retinular cells of the Limulus lateral eye are broken down and renewed daily. The first light onset causes a rapid, synchronous disassembly and buildup of the rhabdom in each photoreceptor cell. The entire process is complete within 30 minutes.     Description: Intraocular recordings from brisk-sustained and brisk-transient ganglion cells in the cat's retina revealed a systematic increase in center size and decrease in spatial cut-off frequency with increasing distance from the area centralis.     Description: A flickering light presented to the eye produces a small alternating voltage at the scalp of a subject. This alternating voltage indicates the following response of the brain to the flicker. If every other flash in the flicker is displaced temporally by as little as 30 microseconds, an asymmetry appears in the brain's alternating voltage.     Description: Micropipettes containing 2 to 50 biological units of beta growth factor (NGF) were placed near growing axons of chick dorsal-root ganglion neurons in tissue culture. The axons turned and grew toward the NGF source within 21 minutes.     Description: A marked reduction of binocular cells in striate cortex is found if 4-week-old kittens are visually stimulated monocularly while anesthetized and held in a stereotaxic apparatus. If the kittens are paralyzed and artificially respirated, changes are not found unless an eye is moved mechanically.     Description: Digitized electromyographic activity of transplanted extensor digitorum longus (EDL) muscles in cats differs from that of control EDL and anterior tibialis muscles lying adjacent to transplanted EDL muscles. In autotransplanted muscles, the cross-sectional area of the fibers shows a negative correlation with mean spike frequency and a positive correlation with mean amplitude.     Description: Iodinated beta H-[2-D-alanine]endorphin exhibits specific binding to cultured human lymphocytes. The binding is inhibited by low concentrations of beta-endorphin and its D-alanine derivative, but is not affected by opiate agonists and antagonists, or by enkephalin analogs, beta-lipotropin, adrenocorticotrophic hormone, or alpha-melanocyte-stimulating hormone; this suggests the existence of a specific, non-opiate binding site (receptor) for beta-endorphin.     Description: Rates of cerebral perfusion were obtained from measurements of the disappearance (wash-out) of oxygen-15 after in situ tissue activation with 45-million-volt x-rays. In an anesthetized cat, typical values were 90 milliliters per minute per 100 grams of tissue, with 55 percent wash-out.     Description: Mouse spinal neurons grown in tissue culture were used to study the membrane effects of the benzodiazepine flurazepam and the naturally occurring purine nucleoside inosine, which competes for benzodiazepine receptor sites in the central nervous system. Application of inosine elicited two types of transmitter-like membrane effects: a rapidly desensitizing excitatory response and a nondesensitizing inhibitory response. Flurazepam produced a similar excitatory response which showed cross-desensitization with the purine excitation. Flurazepam also blocked the inhibitory inosine response.     Description: In cats reared in the dark from birth until 4 months of age, the dorsal lateral geniculate nucleus contained few normal Y cells in either the binocular or monocular segments. Although most of the neurons appeared to be normal X cells unaffected by light deprivation, many cells with abnormal receptive field and response charcteristics were encountered. These effects were permanent, since 1 to 2 years of normal visual experience following initial light deprivation did not lead to any functional recovery.     Description: When placed in a tank of water, aged rats (24 to 27 months old) showed marked impairments in swimming. Compared with young adult rats (3 to 4 months old), the older animals moved their limbs less vigorously and were less successful in keeping their heads above water. The young, but not old, rats maintained a position nearly horizontal to the water surface and planed across it. These movement dysfunctions of aged rats resemble those seen in young adult animals that have sustained injury to brain dopamine-containing neurons. The swimming impairments of the aged rats were reversed by the dopamine receptor stimulant apomorphine and by the biosynthetic precursor of dopamine, L-dopa.     Description: By using two separate electrodes with tips inside a single feline motoneuron, current-voltage characteristics were studied during extracellular iontophoresis of noradrenaline. The usually observed hyperpolarization was accompanied by an increase in membrane resistance and became larger with polarizing and smaller with depolarizing currents. During large depolarizing current injections, the noradrenaline-induced potential reversed its direction, usually at a membrane potential of about -20 millivolts. These data are compatible with the concept that noradrenaline hyperpolarizes nerve cells by decreasing resting membrane conductances to sodium and potassium ions.     Description: When the visual cortex of a newborn kitten is removed, most neurons in the dorsal lateral geniculate nucleus degenerate, but a small population of large cells is spared. Electrophysiological recording revealed that detailed visual topography in the nucleus is abnormal and that single cells have unusually large receptive fields.     Description: Echolocating bats (Eptesicus fuscus) can detect changes as small as 500 nanoseconds in the arrival time of sonar echoes when these changes appear as jitter or alternations in arrival time from one echo to the next. The psychophysical function relating the bat's performance to the magnitude of the jitter corresponds to the half-wave rectified cross-correlation function between the emitted sonar signals and the echoes. The bat perceives the phase or period structure of the sounds, which cover the 25- to 100-kilohertz frequency range, as these are represented in the auditory system after peripheral transformation.     Description: Human speech and animal sounds contain phonemes with prominent and meaningful harmonics. The biosonar signals of the mustache bat also contain up to four harmonics, and each consists of a long constant-frequency component followed by a short frequency-modulated component. Neurons have been found in a large cluster within auditory cortex of this bat whose responses are facilitated by combinations of two or more harmonically related tones. Moreover, the best frequencies for excitation of these neurons are closely associated with the constant-frequency components of the biosonar signals. The properties of these neurons make them well suited for identifying the signals produced by other echolocating mustache bats.     Description: Mouse spinal neurons grown in tissue culture were used to examine the membrane mechanisms of action of the peptide substance P. Two functionally distinct actions were observed, one being a rapidly desensitizing excitation, and the other being a dose-dependent, reversible depression of excitatory responses to the putative amino acid neurotransmitter glutamate.     Description: Administration of a single oral dose of five phenylphosphonothioate esters produced delayed neurotoxicity in hens; their potency was, in descending order, cyanofenphos, EPN, desbromoleptophos, leptophos, and EPBP (Seven). Histological examination showed that in some hens there was marked axonal and myelin degeneration in the spinal cord and peripheral nerves.     Description: The biological activity of recombinant phage and recombinant phage DNA containing monomeric or dimeric polyoma DNA inserts was examined in mice and cultured mouse cells. Recombinant preparations containing a single copy of viral DNA were invariably noninfectious; molecules containing a dimeric polyoma DNA insert were at least seven orders of magnitude less infectious than polyoma virions after parenteral inoculation.     Description: The channels in the junctions of various mammalian cell types--primary cultures and lines--were probed with a series of linear fluorescent amino acid and peptide molecules of different size and charge. Permeability is limited by probe size and electronegativity, these two factors apparently being related reciprocally. In respect to both factors, mammalian junctional channels are more restrictive than insect channels; hence the mammalian channels are narrower, more polar, or both.     Description: Cultures of nonmutant as well as galactokinase-deficient fibroblasts incorporate 20 percent more [35S]sulfate when galactose is substituted for glucose in the medium; galactose-1-phosphate uridyltransferase-deficient cells incorporate 65.5 percent less. In addition to incorporating less [35S]sulfate, the uridyltransferase-deficient cells showed significant accumulation of intracellular galactose-1-phosphate within 4 hours after galactose exposure. Under the same conditions, no difference in [3H]uridine incorporation was observed.     Description: More than 19,000 multisign utterances of an infant chimpanzee (Nim) were analyzed for syntactic and semantic regularities. Lexical regularities were observed in the case of two-sign combinations: particular signs (for example, more) tended to occur in a particular position. These regularities could not be attributed to memorization or to position habits, suggesting that they were structurally constrained. That conclusion, however, was invalidated by videotape analyses, which showed that most of Nim's utterances were prompted by his teacher's prior utterance, and that Nim interrupted his teachers to a much larger extent than a child interrupts an adult's speech.     Description: Damage to DNA appears to be the major cause of most cancer and genetic birth defects and may contribute to aging and heart disease as well. The agents that cause this damage must be identified. Many of these agents are natural chemicals present in the human diet as complex mixtures.     Description: Embryonic chick lens epithelial cells cultured in serum-supplemented medium elongated in the absence of microtubules after treatment with the antimicrotubule drug nocodazole. Colchicine, at concentrations lower than those that dissociate microtubules, blocks cell elongation and the associated increase in cell volume.     Description: Explants of subcutaneous connective tissue from adult BALB/c mice into plastic petri dishes were serially subcultured and tested for tumorigenicity in two ways: by the subcutaneous implantation of cells attached to plastic plates (1 by 5 by 10 millimeters), and by the subcutaneous injection of cells suspended in saline. Cells grown in vitro for 18 or more days before being implanted attached to a plastic plate (2.4 x 10(4) to 3.4 x 10(5) cells per plate) formed tumors after 24 to 79 weeks. The latent period before tumor appearance correlated inversely with the time spent by the cells in tissue culture. Cells inoculated in saline suspension (10 to 100 times the above number per plate) did not form tumors until after 84 days in vitro; plates alone did not induce tumor formation within more than 1 1/2 years of implantation. The tumors arising from the plate-attached cells were transplantable without plates and histologically appeared to be undifferentiated sarcomas. It is well established that smooth-surfaced foreign bodies, regardless of their chemical composition, will produce sarcomas when transplanted subcutaneously in rodents.     Description: Three-day-old rats that were separated from their mothers and deprived of food were found to be capable of feeding either from small puddles of milk or when milk was infused into the front of their mouths. Such feeding was accompanied by a dramatic increase in behavioral activity and only occurred in a warm environment.     Description: The absolute refractory period of neurons projecting from the corticomedial amygdala to the medial preoptic-anterior hypothalamic junction in rats was significantly increased by castration (from 1.01 to 1.61 milliseconds) and decreased again by testosterone (from 1.48 to 0.97 millisecond). Corticomedial amygdala neurons which projected to the capsule of the ventromedial nucleus of the hypothalamus were unaffected.     Description: The interaural phase sensitivity of neurons was studied through the use of binaural beat stimuli. The response of most cells was phase-locked to the beat frequency, which provides a possible neural correlate to the human sensation of binaural beats.     Description: Available evidence suggests that the propensity of digitalis glycosides to produce cardiac arrhythmias is due in part to their neuroexictatory effects. We have performed experiments in cats which support the existence of a neurogenic component in the etiology of digitalis-induced ventricular arrhythmias. Our data further indicate that the locus of this neural effect lies within an area of the medulla 2 millimeters above to 2 millimeters below the obex.     Description: A simple avoidance training procedure during early development produces massive neural traces in visual and somatic cortices of kittens reared in a normal environment. A preponderance of cells in these areas had response preferences for the stimuli used during training. Furthermore, some of these cells exhibited properties never found in normal animals not receiving such training.     Description: Human and mouse chromosomes, stained with either chromomycin A3 or olivomycin, which bind preferentially to G - C-rich DNA (where G is guanosine and C is cytosine), exhibit a Q or a reverse banding pattern, depending on the wavelength used for excitation. The two complementary banding patterns can be observed in the same metaphase simply by changing the combination of excitation filters.     Description: Event-related potentials associated with detected targets in a vigilance task were analyzed in two ways: (i) by sorting the potentials in terms of sequential reaction time bins of 50 milliseconds and (ii) by examining the single trial waveforms. A negative component (N2) covaried in latency with reaction time.     Description: Long-term amphetamine administration to cats (a mean of 8.75 milligrams per kilogram twice daily for 10 days) produced large decreases (40 to 67 percent in serotonin and its major metabolite, 5-hydroxyindoleacetic acid, in all brain regions examined. This treatment also produced several behaviors that are dependent on depressed central serotonergic neurotransmission, and which normally are elicited exclusively by hallucinogenic drugs. Short-term amphetamine administration (15 mg/kg) did not produce these behaviors and resulted in small decreases in brain serotonin and no change in 5-hydroxyindoleacetic acid.
