    Description: Tweestaarten Diplura behoren met de springstaarten Collembola en beentasters Protura tot de Entognatha. Dat zijn kleine ongevleugelde bodemdieren met zes poten, die zich onderscheiden van de insecten door hun verzonken monddelen. Tot nu toe waren twee soorten voor Nederland gemeld, maar voor België 13. Tijdens een onderzoek naar in mierennesten levende organismen werden twee soorten gevonden die nog niet uit Nederland bekend waren. Verder onderzoek zal zeker nog meer nieuwe tweestaarten kunnen opleveren.

    Description: Larven van kriebelmuggen komen uitsluitend voor in stromend water, waar ze met hun waaiervormige monddelen deeltjes filteren uit het voorbij stromende water. Het zijn goede indicatoren voor zuurstofrijk water. Tot nu toe was niet duidelijk welke soorten precies voorkomen in Nederland en België en de determinatie van kriebelmuggen wordt vaak als lastig beschouwd. Om dit te verhelpen, presenteren we hier determinatietabellen voor alle in Nederland en België voorkomende soorten, zowel voor larven van het laatste stadium als voor poppen.

    Description: De catalogus van de Nederlandse kevers uit 2010 geeft het meest recente overzicht van de Nederlandse kevers. Er staan 372 soorten loopkevers op deze lijst. Per soort wordt een overzicht gegeven van de provincies waaruit waarnemingen bekend zijn. Voor elk zogenaamd provincierecord is een collectie-exemplaar aangeduid als bewijsexemplaar. Sinds 2010 is er veel nieuwe informatie over loopkevers bekend geworden. In dit artikel worden de wijzigingen in de lijst toegelicht.

    Description: Hoe een leek op het gebied van wespen en bijen toch een leuke vondst kan doen. Op zonnige dagen gaat de auteur regelmatig op ‘tuinsafari’. Een tuin waarin rekening wordt gehouden met vogels en insecten, door keuze van beplanting en het ophangen van insectenhotels. De inspanningen leverden op 23 juli 2014 een opvallende wesp op. In een bijna automatische reactie werden snel foto’s gemaakt, in de hoop ze later te kunnen determineren. De wesp vloog daarna weg en werd niet meer gezien. Na determinatie blijkt het reuzenertswesp Leucospis dorsigera te zijn, een nieuwe soort bronswesp voor Nederland.

    Description: De gewone citroenzweefvlieg Xanthogramma pedissequum is een eenvoudig te herkennen zweefvlieg. Tenminste, dat dachten we tot voor kort. Nu blijkt dat de bijna identieke X. dives en X. stackelbergi ook in Nederland rondvliegen. Gelukkig zijn er enkele kenmerken waarmee deze drie soorten citroenzweefvlieg in het veld redelijk herkenbaar blijken. In dit artikel bespreken we deze kenmerken en geven we informatie over verspreiding en vliegtijden van de drie soorten.

    Description: Wespblaaskoppen Leopoldius worden ondanks hun opvallende uiterlijk maar weinig waargenomen. De soorten zijn lastig van elkaar te onderscheiden en bovendien werden tot voor kort variabele kenmerken gebruikt, wat tot veel verwarring heeft geleid. Nu blijkt dat alleen vrouwtjes met zekerheid te herkennen zijn aan de vorm van het klampje. Dit is een lepelvormig uitsteeksel onder het vijfde achterlijfsegment. Het wordt gebruikt bij het afzetten van eieren op een gastheer. Waarschijnlijk zijn limonadewespen de belangrijkste gastheer van Leopoldius. In dit artikel wordt een nieuwe soort voor België en een nieuwe soort voor Luxemburg gemeld. Daarnaast wordt een overzicht geboden van waarnemingen in Nederland, België en Luxemburg en een sleutel tot de Noordwest-Europese soorten.

    Description: Ondanks de fors toegenomen aandacht voor watermacrofauna sinds begin jaren 1980 is de schietmot Molanna albicans maar sporadisch aangetroffen. In dit artikel wordt de eerste larvenvondst van M. albicans buiten Drenthe beschreven. Samen met de vindplaatsen in Drenthe is de soort nu van vijf vennen in Nederland bekend. Aanvullend worden de determinatiekenmerken aan de hand van foto’s geïllustreerd.

    Description: Caddisflies are a species rich insect order. The adults are moth-like, but their wings bear hairs instead of scales. The larvae are aquatic and live in self-constructed cases, made of plant material, sand or debris. The species are used as indicators of water quality, but much is still not known about their biology and ecology. In this paper the preferred substrates and phenology of the larvae of 13 species are described, using the data of a survey of two lowland streams in the east of the Netherlands.

    Description: Slakkendodende vliegen danken hun naam aan hun roofzuchtige, parasitaire larven, die het op slakken gemunt hebben. Uit België en Nederland zijn circa 60 soorten uit deze familie bekend, maar er zijn er zeker meer te ontdekken. Dit artikel meldt een nieuwe soort voor de Nederlandse fauna uit het genus Psacadina, dat in beide landen drie soorten telt. De herkenning en het voorkomen van de drie soorten wordt uitgebreid besproken.

    Description: Zwarte vliegen behoren tot de muggen, maar door hun korte antennen doen ze oppervlakkig aan vliegen denken. De mannetjes zien er met hun grote kop en grote ogen heel anders uit dan de vrouwtjes, die een kleine kop met kleine ogen hebben. In het voorjaar vormen sommige soorten opvallend grote zwermen op windluwe plekken. Aan de 17 soorten die reeds uit Nederland bekend waren, kan er nu één worden toegevoegd: Bibio venosus.

    Description: Astrobunus laevipes is een hooiwagen die via het Rijnbekken recentelijk ons land is binnengekomen. Er waren al vondsten langs de Waal en Nederrijn in Gelderland en het meest oostelijke puntje van de provincie Utrecht. Door gerichte inventarisaties kunnen wij de hooiwagen nu nieuw voor de provincies Zuid-Holland en Noord-Brabant melden. Daarnaast is er van A. laevipes nu ook een waarneming langs de IJssel en enkele westelijker in de provincie Utrecht. Allerlei rivierbegeleidende biotopen langs (uitlopers van) de Nederrijn en Waal worden al door deze opmerkelijke hooiwagen bevolkt, maar de noordelijke uitbreiding via de IJsselvallei lijkt veel trager te verlopen. De Maas lijkt nog niet bereikt.

    Description: Slakkendodende vliegen zijn nog relatief slecht onderzocht in ons land, zeker de kleine soorten. De vertegenwoordigers van het genus Anticheta zijn kleine vliegjes, waarvan slechts twee soorten uit ons land bekend waren. Uitgebreider onderzoek, zowel in collecties als in het veld, heeft twee nieuwe soorten voor de fauna aan het licht gebracht: Anticheta nigra en A. obliviosa. Van de eerste soort waren wereldwijd slechts acht exemplaren bekend.

    Description: Zelfs in een goed onderzochte insectengroep als de zweefvliegen zijn nog altijd nieuwe soorten voor de Nederlandse fauna te ontdekken. Ook in 2014 was het weer raak, dit keer in het fraaie natuurgebied rond de Drentsche Aa. Het lijkt niet toevallig dat deze aanvulling op de Nederlandse fauna juist hier werd gevonden. Deze vindplaats vult ecologische informatie uit het buitenland aan, waardoor langzamerhand een beeld begint te ontstaan van de levenswijze van deze zeldzame zweefvlieg, waarover tot voor kort weinig bekend was.

    Description: De doornkaakzandbij ziet er in het voorjaar een beetje anders uit dan in de zomer. Daarom worden de voorjaarsdieren door sommigen tot een andere soort beschouwd dan de zomerdieren. Uit Nederland zijn slechts drie exemplaren bekend: twee uit het voorjaar en één uit de zomer. Toevallig komen alle exemplaren uit hetzelfde gebied. Of is dit geen toeval?

    Description: Elfjes behoren tot de vroegste zweefvliegen die in het voorjaar uit de pop kruipen. Rond bloeiende wilgen wemelt het vaak van wilgenelfjes Melangyna lasiophthalma, waardoor het zoeken naar de zeldzamere soorten soms lastig is. Toch zijn de meeste soorten relatief makkelijk van elkaar te onderscheiden. Met de vondst van het Sachalinelfje M. pavlovskyi wordt het echter weer iets lastiger.

    Description: Steltmuggen lijken op langpootmuggen en zijn daar nauw aan verwant. Veel soorten zijn groot en karakteristiek gekleurd, maar toch zijn ze in Nederland nog weinig bestudeerd. Dit artikel bewijst dat er nog veel te ontdekken valt: in één jaar tijd zijn drie soorten gevonden die nog niet uit ons land bekend waren: Atypophthalmus inustus, Molophilus niger en Arctoconopa melampodia. Hiermee komt het totaal aantal soorten voor Nederland op 149. De nieuwe meldingen sluiten goed aan op het bekende verspreidingsgebied.

    Description: Jalla dumosa is een in Nederland zeer zeldzame pentatomide, die in 1969 voor het laatst was waargenomen op Terschelling. Na 45 jaren zonder waarnemingen werd ze in 2014 weer in klein aantal waargenomen in de Kooiduinen op Ameland. Rupsen van onder andere de duinparelmoervlinder en de sint-jansvlinder stonden daar op het menu. In deze bijdrage wordt informatie samengevat over de vondsten in Nederland, de biologie en de verspreiding van de soort.

    Description: In August 2014 the exotic jellyfish Blackfordia virginica was captured in the harbour of Amsterdam. This is the first confirmed record of this species in the Netherlands, although in October 2013 a possible specimen was filmed and released. This indicates that the species might be established in the Amsterdam area, although repeated introduction with ballast water cannot be ruled out. In September 2014 hydrozoan polyp colonies closely resembling those of B. virginica were collected near IJmuiden in the Noordzeekanaal, a canal connecting Amsterdam with the North Sea at IJmuiden.

    Description: Wie een steen of boomstronk omkeert en voor het eerst een Trogulus opmerkt, zal dit bizarre beest niet direct associëren met een hooiwagen. Met de korte, stevige poten, het afgeplatte achterlijf dat overdekt is met strooisel en zand, en een trage manier van voortbewegen is het een ongewone verschijning. De soorten van dit geslacht zijn bodembewoners die zich voornamelijk voeden met kleine huisjesslakken. Uit ons land waren tot nu toe twee soorten bekend. De identiteit van een van die twee was lang onzeker, maar door nieuwe inzichten is nu duidelijk om welke soort het werkelijk gaat. In Nederland blijkt bovendien nog een derde soort voor te komen, die hier voor het eerst gemeld wordt.

    Description: Roofvliegen zijn met 40 Nederlandse soorten een vrij kleine groep, die behoorlijk goed is onderzocht. Het gebeurt dan ook niet vaak dat er een nieuwe soort voor de Nederlandse fauna kan worden opgetekend. De meeste roofvliegen zijn grijze vliegen, zo niet de stamjagers van het genus Choerades. Dit zijn juist vrij opvallend gekleurde vliegen. Zo heeft de rode dennenstamjager Choerades gilvus een rood gekleurd achterlijf. Opmerkelijk genoeg blijkt dat in Nederland juist onder deze opvallende verschijning twee soorten schuilgaan.

    Description: Onlangs werd op een braakliggend terrein in Roermond de zaadkever Bruchus brachialis verzameld. Dit is de eerste Nederlandse vondst van deze zuidelijke soort. Een reeks recente waarnemingen uit de ons omringende landen laat zien dat deze soort al enige jaren in een noordwaartse areaaluitbreiding verwikkeld is. Met deze ontdekking komt het aantal Nederlandse Bruchus-soorten op zeven.

    Description: Nieuw gevormd landschap, zoals jonge duinvalleien, opgespoten polders en bedrijventerreinen in ontwikkeling vormen de habitat van gespecialiseerde soorten. Je moet tegen een stootje kunnen om in een dergelijke zandige, schaars begroeide omgeving te overleven. Vooral langdurig warme, droge perioden vormen een probleem voor vochtminnende bodemdieren. De springstaartsoorten die hier overleven zijn warmteminnend en droogteresistent, een bijzondere combinatie voor bodemdieren. Dit geldt ook voor Entomobrya unostrigata, een nieuwe aanwinst voor de fauna van Noordwest-Europa. Er is een aantal populaties van deze soort op de Tweede Maasvlakte gevonden. De dieren zaten hier onder stenen en hout op opgespoten zand.

    Description: De Polychaeta vormen een groep van vrijwel uitsluitend mariene wormen, gekenmerkt door een geleed lichaam met aan de meeste leden een tamelijk groot aantal borstels. Het is een soortenrijke groep; ze spelen een belangrijke rol in de ecologie en diversiteit van vooral zachte, maar ook harde zeebodems. In vergelijking met de kreeftachtigen en weekdieren, eveneens soortenrijke groepen in dezelfde biotoop, zijn borstelwormen minder goed onderzocht. In dit artikel wordt de zeerups Fimbriosthenelais minor nieuw voor ons land gemeld. Met name op stenige en schelprijke bodems zijn nog meer nieuwe borstelwormen voor ons land te verwachten.

    Description: Het kalkdoorntje is een zeldzame sprinkhanensoort, die vooral bekend is uit Zuid-Limburg en van enkele locaties in het rivierengebied. Over het voorkomen van dit kleine sprinkhaantje binnen de Gelderse Poort was lange tijd weinig bekend. Pas vanaf 2007 worden regelmatig waarnemingen gedaan. Toevallige waarnemingen op, voor de auteur, onverwachte locaties vormden de aanleiding voor een grondige inventarisatie in de jaren 2011-2013. Hieruit komt naar voren dat het kalkdoorntje een ruime verspreiding heeft binnen de Gelderse Poort en een meer diverse biotoopkeuze heeft dan voorheen bekend was.

    Description: The relative sensitivities of 309 common invertebrate species in Dutch marine waters are presented for environmental and anthropogenic pressures like organic enrichment, sedimentation and fisheries. The species were furthermore appointed to trophic groups like suspension and deposit feeders. The Dutch Ministry of Infrastructure and the Environment uses these data when calculating the Benthic Ecosystem Quality Index 2 and the Infaunal Trophic Index. These metrics aid in the mandatory monitoring of ecological quality for example for the European Water Framework Directive. The common Dutch species were selected based on their abundance according to, 1. the mwtl dataset including results of on-going monitoring programs issued by the Ministry, 2. the monitoring by volunteer scuba-divers for the anemoon Foundation and 3. the monitoring of fouling plates for the project setl.

    Description: Onze fauna verandert en de mariene fauna is daarop geen uitzondering. Zuidelijke soorten koloniseren onze kustwateren en exotische soorten worden door de mens aangevoerd. Voor sommige soorten is niet duidelijk waarom ze hier opduiken. De sterk veranderende kustbiotopen spelen ongetwijfeld eveneens een rol. Een niet-aflatende stroom onverwachte soorten, soms zelfs behorend tot nieuwe genera en zelfs families maken faunistisch onderzoek in de kustwateren verrassend en boeiend. In het kader van een inventarisatie van kreeftachtigen in het Deltagebied werden in de Oosterschelde bij Zierikzee verscheidene kleine monsters genomen van diverse substraten. De monsters van roodwieren bevatten een aantal opmerkelijke soorten. Dit artikel behandelt Uromunna spec., een zeepissebeddensoort van een familie die in ons land nog niet was aangetroffen.

    Description: This paper describes the discovery of a settled juvenile specimen of the stomatopod Platysquilla eusebia on the Dutch part of the Dogger Bank in the central North Sea. This is the northernmost record in Europe. The species is native to the Mediterranean and to the Atlantic coast from Portugal up to France. Further investigations have to show if the species already forms populations this far north. As the planktonic stages of P. eusebia have already been recorded in prior years, the establishment of the species should not be a problem, providing the circumstances are favourable.

    Description: Met enige regelmaat duiken nieuwe springstaartsoorten op in ons land. Het vinden van een nieuwe soort is soms op toeval gebaseerd. Op een foto van een sprinkhaan was naast de linker voorpoot een klein, donker vlekje te zien. Uitvergroot bleek het om een klein springstaartje te gaan. Het zwart-wit gestreepte patroon liet geen ruimte voor twijfel over de determinatie: Fasciosminthurus quinquefasciatus, een nieuwe springstaart voor de Nederlandse fauna. In dit artikel wordt het genus en de soort voorgesteld, beschrijven we de habitat en geven we informatie over de verspreiding in de rest van Europa.

    Description: De roodbruine heiderouwzwever Exoprosopa capucina is een fraaie en karakteristieke koffiebruine verschijning op de Nederlandse heideterreinen. Zoals veel wolzwevers is deze te herkennen aan de vleugeltekening en de kleur van de lichaamsbeharing. Echter naarmate de dieren ouder worden verliezen ze deze beharing en wordt de determinatie lastiger. Vermoedelijk heeft het verfomfaaide uiterlijk van een vrouwtje Exoprosopa cleomene er voor gezorgd dat deze ruim 50 jaar onopgemerkt tussen de exemplaren van E. capucina heeft gestaan.

    Description: In de afgelopen jaren werd op verschillende plekken in Nederland een houtzwamkever gevonden die tot nu toe niet officieel gemeld werd: Sulcacis bidentulus. Deze soort valt binnen de zeer eenvormige familie Ciidae op door de kleur van de dekschilden, die duidelijk iets lichter bruin zijn dan de rest van het lichaam. De soort zou zich ontwikkelen in de bleke borstelkurkzwam, die voornamelijk op populieren groeit. Oudere vondsten ontbreken en de ontdekking van deze soort op meerdere locaties duidt op een recente vestiging in ons land. Dit sluit aan bij de geconstateerde uitbreiding in Midden-Europa.

    Description: De boomkrekel is een slanke geelbruine krekel, die vooral opvalt door zijn melodieuze zang. Vroeger moest je naar Frankrijk om dit geluid te horen, maar tegenwoordig heerst ook in het rivierengebied bij Nijmegen een mediterrane sfeer. De boomkrekel is in 2004 voor het eerst langs de Waal gevonden en heeft zich daarna uitgebreid. In 2010 en 2011 heeft de Flora- en faunawerkgroep Gelderse Poort met vele vrijwilligers onderzoek gedaan naar de precieze verspreiding. In totaal werd de boomkrekel in 57 kilometerhokken gevonden en de verwachting is dat de soort zich de komende jaren verder zal uitbreiden.

    Description: Hongerwespen behoren tot de grote groep van de sluipwespen. Het zijn slanke wespjes die typisch met hangende pootjes vliegen, waarbij de verbrede achterschenen opvallen. Ze parasiteren bijen en mogelijk ook bepaalde angeldragende wespen en worden daarom vaak bij bijenhotels waargenomen. In het kader van dit artikel zijn de meldingen van Nederlandse hongerwespen kritisch beschouwd. In totaal zijn nu negen soorten uit ons land bekend, waarvan er hier maar liefst zes voor het eerst worden gemeld.

    Description: As most of the sea bottom in the Dutch part of the North Sea consists of sand, marine fauna that live in association with hard substrates are rarely monitored. We report here on the results of a species inventory in June 2011 done by scuba-diving while focusing on a wreck on the Dogger Bank and on rocky bottoms on the Cleaver Bank. This resulted in various new records of species for the Dutch part of the North Sea. This result appeared for a large part linked to the added value of monitoring with scuba-divers. It is therefore concluded that scuba-divers should be used in addition to the more traditional monitoring methods in which dredges and grabs are used, if one aims at getting an accurate view of the biodiversity present in marine regions like the North Sea.

    Description: During an expedition with scuba-divers to the Dutch part of the Brown Ridge in the central North Sea in June 2013, two colonies of the jewel anemone Corynactis viridis were found on the wreck Anna Graebe. With the jewel anemone both a new species and a new animal order, the Corallimorpharia, are added to the autochthonous fauna of the Netherlands. This species typically occurs in the Mediterranean and along the Atlantic coast from Portugal and the west British Isles up to Shetland. As other records of settled colonies of C. viridis in the North Sea were recently reported from Belgian, German and English waters, it is concluded that the jewel anemone, which used to be known as an occasional visitor, should now be considered autochthonous in the North Sea.

    Description: Heleomyzidae zijn kleine tot middelgrote vliegen. Ze krijgen niet veel aandacht, omdat de meeste soorten een nogal verborgen leven leiden en bovendien niet opvallend gekleurd of getekend zijn. De Nederlandse familienaam, afvalvliegen, is voor de meeste soorten niet echt passend, maar veel soorten van de subfamilie Heteromyzinae, waartoe Tephrochlaena oraria behoort, worden dan wel weer vaker rond afval aangetroffen. Tephrochlaena oraria is echter in meerdere opzichten een buitenbeentje. Met deze soort komt de lijst van Nederlandse Heleomyzidae op 49.

    Description: Cucujus cinnaberinus is een recent voor Nederland ontdekte soort. De kever staat op de Habitatrichtlijn, wat onder meer betekent dat hij beschermd is en dat de populatie gemonitord dient te worden. Inventarisatie en monitoring gebeurt in het algemeen door het weghalen van schors van recent gestorven bomen. Hierdoor wordt het leefgebied van de kever echter flink aangetast. Wij voerden een klein onderzoek uit naar de mogelijkheden om C. cinnaberinus te vangen in azijnzuurvallen, zodat de habitat intact gelaten kan worden en de soort toch geïnventariseerd kan worden. Tijdens het voorjaar van 2013 werden vier vallen opgehangen op een locatie waar zich een populatie van deze soort bevindt. Er werden slechts twee individuen verzameld; de vangmethode lijkt dus niet bijzonder geschikt.

    Description: This paper reports on observations made during wreck dive expeditions in 2010-2012, in order to investigate the ecological relevance of shipwrecks on the Dutch Continental Shelf (dcs). Shipwrecks are biodiversity hotspots. The number of species recorded on shipwrecks is similar to the number of species found in soft bottoms of the entire dcs. The soft substrates, however, represent a vastly larger habitat on the dcs than the shipwrecks. Amongst many other taxa, juvenile and large Atlantic cod, linear skeleton shrimp, goldsinny wrasses and leopard spotted gobies were found in the shipwreck habitats. The presence of these important species and their absence from many other habitats, illustrate that shipwrecks function as key habitats, nurseries, and refugia that are rare or absent anywhere else in the Netherlands.

    Description: Sluipwespen van de groep van de Polysphinctini hebben een bijzondere levenswijze. Het vrouwtje van wesp legt een eitje op het achterlijf van een spin. Wanneer het eitje is uitgekomen brengt de larve haar hele leven op het achterlijf van de spin door. Ze maakt kleine wondjes in de huid en voedt zich met lichaamsvloeistof van de spin. Na verloop van tijd ontwikkelt de larve bulten op de rug, een teken dat ze bijna volgroeid is. Ze zuigt dan de spin leeg, de ‘grote slurp’ genaamd, en verpopt zich in het web. In dit artikel wordt de levenswijze van één van deze soorten, Acrodactyla quadrisculpta, voor het eerst beschreven en geïllustreerd.

    Description: De bladwespgenera Phyllocolpa, Tubpontania, Pontania en Euura hebben gemeenschappelijk dat ze gallen vormen op wilg. Recentelijk is er veel vernieuwend taxonomisch werk verricht aan deze groepen. In dit artikel wordt de Nederlandse situatie samengevat. Uit oudere literatuur waren 18 soorten bekend en in het recent gepubliceerde gallenboek worden nog eens 11 soorten voor het eerst voor ons land vermeld. In dit artikel worden nog acht soorten aan de lijst toegevoegd en twee soorten worden van de lijst afgevoerd. In totaal zijn nu 35 soorten galvormende bladwespen van wilg uit Nederland bekend.

    Description: Snuitvliegen zijn rare snuiters onder de zweefvliegen. Ze hebben een verlengd gezicht en een lange tong. Hiermee kunnen ze foerageren op bloemen met diepliggende nectar, in tegenstelling tot de meeste andere zweefvliegen. Als genus zijn de vliegen makkelijk herkenbaar, maar de zeer zeldzame soorten tussen de zeer algemene gewone snuitvlieg uithalen was niet altijd even makkelijk. In dit artikel wordt de herontdekking van de rode snuitvlieg beschreven en een tabel gegeven voor de drie Noordwest-Europese soorten.

    Description: De larven van Trichoptera heten kokerjuffers en leven in het water. Veel soorten bouwen een kokertje van zand, takjes of ander materiaal, vandaar de Nederlandse naam. De volwassen dieren lijken wel wat op nachtvlinders en worden schietmotten genoemd. De laatste jaren heeft het onderzoek naar deze dieren een grote vlucht genomen. Enerzijds spelen ze een rol als indicator voor de waterkwaliteit. De larven worden dan ook veel verzameld tijdens macrofaunabemonsteringen door waterschappen. Anderzijds is er een groeiende groep entomologen die schietmotten bestudeert. In dit artikel wordt Ecclisopteryx dalecarlica voorgesteld, een nieuwe soort voor Nederland.

    Description: New and old records of the robberfly Machimus cowini in the Netherlands (Diptera: Asilidae) Until recently, Machimus cowini (Hobby, 1943) was known from just one record in the Netherlands. An examination of Dutch specimens of the similar M. cingulatus revealed specimens of M. cowini from five new localities. The records are concentrated on the island Ameland and along the rivers Waal and Maas. On Ameland, M. cowini probably occurs in coastal dunes. Near Nijmegen the species was found in grassy vegetation on floodplains and on riverdunes near the river.

    Description: Faunistics and ecology of the syrphid genus Epistrophe in The Netherlands (Diptera: Syrphidae) Ten species of Epistrophe have been found in the Netherlands, two of which, E. cryptica and E. similis, are recorded here for the first time. The distribution, changes in distribution and ecology of the species were investigated and are illustrated by distribution maps and diagrams of the flight period. In all species, except E. eligans, females were observed more than males. Furthermore, females seem to be active later in the season than males. It is discussed that this is probably caused by the differences in behaviour between the males of the species.

    Description: Until recently Boreus hyemalis was considered to be a rare species in The Netherlands. It was only known from a few localities in The Netherlands in the provinces of Noord-Holland, Zuid- Holland, Utrecht, Gelderland and Drenthe. The last few years many new populations have been discovered. In this paper the first records for the provinces of Limburg, Noord-Brabant and Overijssel are presented. The species seems to prefer scarcely vegetated patches in sanddunes, dominated by greyhair grass Corynephorus canescens and the moss Polytrichum piliferum. It proved to be relatively easy to find, when the right places were searched in the right period (October until February).

Die moeilijkheid komt door de variatie, maar vooral ook door gebrek aan bruikbare beschrijvingen. Met dit artikel zijn de negen Nederlandse soorten te determineren. Vanwege het gebrek aan determinatieliteratuur in heel Europa is het in het Engels geschreven. Behalve de maar één keer waargenomen B. domestica, zijn de meeste soorten vrij gewone verschijningen in ons land, zoals uit de kaarten blijkt.

    Description: The occurrence of Digitivalva arnicella in the Netherlands: rediscovery and conservation (Lepidoptera: Plutellidae: Acrolepiinae) Digitivalva arnicella (Heyden, 1863), previously only known from two localities before 1902, has been rediscovered in eight localities in the northern part of the Netherlands (province of Drenthe) on its host Arnica montana. On the basis of leafmines, found in old herbarium collections, it can be concluded that the species was once widespread within the range of its host in the northern and eastern parts of the Netherlands and has apparently been overlooked by entomologists since. D. arnicella is a much endangered species in the Netherlands, because of the dramatic decline of its host. Two of the eight discovered populations have been lost since their discovery in the first half of the 1990’s. Systematics and life history of the species are described and illustrated, the distribution is mapped and advise for management of its sites are given. The current management of some populations of Arnica, by mowing the site completely in August, is disastrous for the young caterpillars, mining the leaves.

    Description: The West Palaearctic species of the subfamily Paxylommatinae are reviewed and the species of the genus Hybrizon Fallén, 1813, from the Palaearctic region are keyed. Hybrizon juncoi (Ceballos, 1957) is recognized as a valid species, a neotype is designated for Hybrizon latebricola Nees, 1834, and a lectotype is designated for Plancus apicalis Curtis, 1833. Paxylomma grandis Rudow, 1883, Ogkosoma schwarzi Haupt, 1913, and Eurypterna arakawae Matsumura, 1918, are new junior synonyms of Eurypterna cremieri (de Romand, 1838).

    Description: Het genus Scrobipalpa in Nederland (Lepidoptera: Gelechiidae) Scrobipalpa is een geslacht van kleine, lastig uit elkaar te houden motjes. In heel Europa zijn ongeveer 70 soorten bekend. Doorgaans zijn de vleugels bruin- of grijsachtig met een tekening van stippels en strepen die bovendien erg kan variëren. Hierdoor kunnen de individuele soorten vaak moeilijk herkend worden. Aan de hand van de genitaliën kunnen soorten wel allemaal gedetermineerd worden. Omdat de literatuur over deze groep niet erg toegankelijk is en bestaande beschrijvingen soms erg onvolledig zijn, wordt dit artikel in het Engels geschreven. Met dit artikel zijn de elf Nederlandse soorten te determineren. De meeste hiervan zijn min of meer gebonden aan de kust. Van alle soorten worden naast beschrijvingen van de vleugeltekening en de genitaliën, ook de verspreiding, de biologie en de ecologie gegeven.

    Description: New records of Cheilosia caerulescens in the Netherlands (Diptera: Syrphidae) Cheilosia caerulescens is a rare hoverfly in The Netherlands. Until 1998 only four records were known, the first in 1986. In 1998 the species was found on three new and one old locality. The record from Heemstede (province of Noord-Holland) is among the northernmost in the European distribution of this species. Three of the four records originate from gardens.

    Description: Interesting records of beetles in The Netherlands (Coleoptera: Staphylinidae, Nitidulidae, Dermestidae) Tachyporus quadriscopulatus Pandellé, 1869 has been recorded for the second time in the Netherlands at Arcen and Velden, province of Limburg. After 27 years, a third record of Carpophilus marginellus Motschulsky, 1858 can be reported from St. Geertruid, in the southern part of the province of Limburg. One specimen was found outdoors, in a wood. One specimen of Anthrenocerus australis Hope, 1843 was also found outdoors, in Oost- Maarland also in southern Limburg, sitting on a flower in a meadow.

    Description: Butterflies in the Netherlands still under pressure (Lepidoptera: Rhopalocera) New information on the Dutch Rhopalocera fauna since the distribution atlas of Tax (1989) is presented. Coenonympha pamphilus was common in large parts of the Netherlands, but has declined dramatically. C. arcania is now formally extinct in our country. An extensive research showed that Maculinea alcon ericae has dissappeared from many sites. Furthermore the smallest of the two remaining populations of Heteropterus morpheus seems to have gone extinct. On the other hand, Callophrys rubi, has colonized new territory. Lampidus boeticus, was observed once as an adult and once a caterpillar was found between snow peas from Egypt. Colias croceus was very abundant in 1998. The reintroductions of Maculinea teleius and M. nausithous in 1990 appear to have been successful.

    Description: The rearing of the botfly Cephenemyia auribarbis (Diptera: Oestridae) After several failures we finally succeeded in rearing botflies Cephenemyia from third-instar larvae. From a red deer shot at 13 March 1998 about one hundred larvae of Cephenemyia auribarbis (Meigen, 1824) were collected. Ten individuals reached the pupal stage. After 22 days, one fullgrown male died in the pupa, and two males emerged and lived for 17 and 18 days respectively. This is one of the few recorded succesful attempts to rear botflies.

    Description: A new record of Chorthippus apricarius in the Netherlands (Orthoptera) Chorthippus apricarius is a very rare grasshopper in The Netherlands. Only three, very widely separated sites are known. In 1998 a small new population has been found in a roadside verge, 2 km southwest of the southernmost site in the south of the province of Limburg.

Major nomenclatorial changes, published elsewhere, have been implemented. The list contributes to the faunistic work of the European Invertebrate Survey - The Netherlands. A separate list of hostplants of the Nitidulidae and Brachypteridae is presented.

    Description: For the first time a gregarious parasitoid belonging to the genus Phaenocarpa Foerster, 1862 (Braconidae: Alysiinae) is reported, and described as P. helophilae spec. nov. from England. The first record of Chloropidae as host for a Phaenocarpa species is given; P. livida (Haliday, 1838) reared from Calamoncosis (Rhaphiopyga) glyceriae Nartshuk, 1958. Sathra debilis Foerster, 1862, is synonymised with P. livida (Haliday). Additionally, Phaenocarpa curticauda spec. nov. from The Netherlands is described and fully illustrated. A lectotype is designated for Alysia pectoralis Zetterstedt, 1838, and Phaenocarpa rufoflava Papp, 1968, is recognised as a valid species.

    Description: The assassin bug genus Rasahus Amyot & Serville (Heteroptera: Reduviidae: Peiratinae) comprises 26 Neotropical species. A cladistic analysis of the genus was carried out using 63 characters from external morphology, body vestiture, and male and female genitalia, with the species considered as terminal taxa. The analysis yielded 149 equally parsimonious cladograms, each with 206 steps, CI = 0.35, and RI = 0.60; the successive weighting procedure resulted in eight cladograms (CI = 0.79 and RI = 0.91). In the strict consensus cladogram, two major clades are delimited: one comprising the species R. rufiventris, R. hamatus, R. arcitenens, R. arcuiger, R. amapaensis, R. thoracicus, R. biguttatus, R. argentinensis, R. limai, R. grandis, and R. angulatus; and the other with R. castaneus, R. aeneus, R. scutellaris, R. maculipennis, R. brasiliensis, R. sulcicollis, R. surinamensis, R. albomaculatus, R. guttatipennis, R. atratus, R. peruensis, R. costarricensis, R. bifurcatus, R. flavovittatus, and R. paraguayensis. A cladistic biogeographic analysis of the provinces of the Neotropical subregion, considering distributional data of the species of Rasahus and three other genera of Peiratinae (Eidmannia, Melanolestes, and Thymbreus) was carried out. Four general area cladograms were obtained applying programs COMPONENT 2.0 and TASS. The most parsimonious general area cladogram (= implying fewer items of error) was obtained with COMPONENT 2.0 minimizing the number of leaves added, and shows the sequence (Desierto, (Caatinga, (Cerrado, (Chacoan, (Caribbean, Amazonian), (Paranaense, Atlantic))))). This sequence of area relationships is congruent with the history previously hypothesized for the subregion, where the development of an open vegetated diagonal (comprising the Chacoan, Cerrado, and Caatinga provinces) due to the aridification induced by the gradual uplift of the Andes, separated the former Amazonian forest in a northwestern part (Caribbean plus Amazonian provinces) and a southeastern part (Paranaense plus Atlantic provinces).

    Notes: Folding type-specific secondary structure propensities of 20 naturally occurring amino acids have been derived from α-helical, β-sheet, α/β, and α+β proteins of known structures. These data show that each residue type of amino acids has intrinsic propensities in different regions of secondary structures for different folding types of proteins. Each of the folding types shows markedly different rank ordering, indicating folding type-specific effects on the secondary structure propensities of amino acids. Rigorous statistical tests have been made to validate the folding type-specific effects. It should be noted that α and β proteins have relatively small α-helices and β-strands forming propensities respectively compared with those of α+β and α/β proteins. This may suggest that, with more complex architectures than α and β proteins, α+β and α/β proteins require larger propensities to distinguish from interacting α-helices and β-strands. Our finding of folding type-specific secondary structure propensities suggests that sequence space accessible to each folding type may have differing features. Differing sequence space features might be constrained by topological requirement for each of the folding types. Almost all strong β-sheet forming residues are hydrophobic in character regardless of folding types, thus suggesting the hydrophobicities of side chains as a key determinant of β-sheet structures. In contrast, conformational entropy of side chains is a major determinant of the helical propensities of amino acids, although other interactions such as hydrophobicities and charged interactions cannot be neglected. These results will be helpful to protein design, class-based secondary structure prediction, and protein folding. © 1998 John Wiley & Sons, Inc. Biopoly 45: 35-49, 1998

    Notes: DNA branched junctions are analogues of Holliday junction recombination intermediates. Partially mobile junctions contain a limited amount of homology flanking the branch point. A partially mobile DNA branched junction has been incorporated into a synthetic double-stranded circular DNA molecule. The junction is flanked by four homologous nucleotide pairs, so that there are five possible locations for the branch point. Two opposite arms of the branched junction are joined to form the circular molecule, which contains 262 nucleotides to the base of the junction. This molecule represents a system whereby torque applied to the circular molecule can have an impact on the junction, by relocating its branch point. Ligation of the molecule produces two topoisomers; about 87% of the product is a relaxed molecule, and the rest is a molecule with one positive supercoil. The position of the branch point is assayed by cleaving the molecule with endonuclease VII. We find that the major site of the branch point in the relaxed topoisomer is at the maximally extruded position in the relaxed molecule.     Notes: The relatively novel combination of flow field-flow fractionation (FFF) and multiangle light scattering (MALS) was employed to study a nondegraded κ-carrageenan in different 0.1M salt solutions. The applicability of the technique was tested, and the effects of salt type and salt composition on the molar mass and radius of gyration were studied. A conformational ordering was induced at room temperature by switching the solvent from 0.1M NaCl (coil form) to 0.1M NaI (helix form). An approximate doubling of the average molar mass and an increase in radius of gyration was then observed, in agreement with results obtained previously using size exclusion chromatography-MALS. This increase in size was attributed to conformational ordering and to the formation of double helices. Severe aggregation was observed above 40% CsI in the 0.1M mixed salt solution of CsI and NaI. This was ascribed to the association of helices into large aggregates. For these large associates, having molar masses of several millions, a reversal of the elution order in flow FFF was detected. © 1998 John Wiley & Sons, Inc. Biopoly 45: 85-96 1998

    Notes: In this article a conformational analysis of the D-alanyl-D-alanine dipeptide, both charged and neutral, has been carried out. The preferred conformations were determined by means of ab initio and semiempirical quantum, together with empirical force field calculations. The AMBER* force field and the 6-31 + G** and 6-31G** ab initio levels give rise to a coincident minimum energy structure, which, on the other hand, differs from that determined by AM1, 3-21 + G, and 3-21G. The solvent effect on the different charged and neutral conformations have been considered through the AMSOL semiempirical method. A quantification regarding the structural similarities between the different dipeptide conformations and the ampicillin has been performed. The results show that the best overlay is attained by the minimum structure energy obtained by using the 6-31 + G** methodology, which presents a planar amidic nitrogen. © 1998 John Wiley & Sons, Inc.     Notes: The conformational behavior of different molecular weight fractions of a synthetic C6-oxidized derivative of cellulose were investigated by means of capillary viscometry, CD, and microcalorimetric measurements. Experiments were carried out in the presence of either monovalent or divalent counterions.The experimental data indicated that C6-oxidized cellulose can assume an ordered extended conformation at low ionic strength, induced by the intrachain repulsions of negative charges. This conformation was suggested to be very similar to the fully extended structure of cellulose. In addition to this, upon increasing the ionic strength, a conformational transition of the order-to-disorder type occurred. In fact, the screening of the electrostatic repulsions introduced a number of conformational kinks into the cellulosic backbone, which enabled the polymer to assume a more coiled conformation hence producing less viscous aqueous solutions. © 1998 John Wiley & Sons, Inc.     Notes: A new method is proposed for the determination of the enthalpy and entropy changes of nonionic origin upon conformational transition of linear biopolyelectrolytes in solution. For all transition midpoints, defined by given temperature and ionic strength, the total free energy change of the system is zero, which means that the nonionic contribution to the free energy change is equal in value and opposite in sign to the polyelectrolytic one. The counterion condensation theory of linear polyelectrolytes provides for the appropriate analytical expression to be used in such calculations. Linear plots of the proper functions of the calculated free energy changes vs the proper functions of temperature allows for the determination of the enthalpic and entropic terms of the nonionic free energy change of transition.The method has been applied to the extensive available data of the ion-induced conformational change of κ-carrageenan, a linear sulfated galactan extracted from seaweeds. The method has proved very successful, with the results showing a remarkable convergency of the enthalpy values for different monovalent counterions. On the other hand, the above approach has made it possible to explain the known effect of counterion specificity on the transition by a small difference in the nonionic entropic contributions. © 1998 John Wiley & Sons, Inc.     Notes: Ultraviolet resonance Raman (UVRR) spectra of H2O and D2O solutions of the nucleoside (dA, dG, dC, dT) and aromatic amino acid (Phe, Trp, Tyr) constituents of DNA viruses have been obtained with laser excitation wavelengths of 257, 244, 238, and 229 nm. Using the 981 cm-1 marker of Na2SO4 as an internal standard, Raman frequencies and scattering cross sections were evaluated for all prominent UVRR bands at each excitation wavelength. The results show that UVRR cross sections of both the nucleosides and amino acids are strongly dependent on excitation wavelength and constitute sensitive and selective probes of the residues.     Notes: The quaternary structure of Lumbricus terrestris hemoglobin was investigated by small-angle x-ray scattering (SAXS). Based on the SAXS data from several independent experiments, a three-dimensional (3D) consensus model was established to simulate the solution structure of this complex protein at low resolution (about 3 nm) and to yield the particle dimensions. The model is built up from a large number of small spheres of different weights, a result of the two-step procedure used to calculate the SAXS model. It accounts for the arrangement of 12 subunits in a hexagonal bilayer structure and for an additional central unit of cylinder-like shape. This model provides an excellent fit of the experimental scattering curve of the protein up to h = 1 nm-1 and a nearly perfect fit of the experimental distance distribution function p(r) in the whole range. Scattering curves and p(r) functions were also calculated for low-resolution models based on 3D reconstructions obtained by cryoelectron microscopy (EM). The calculated functions of these models also provide a very good fit of the experimental scattering curve (even at h 〉 1 nm-1) and p(r) function, if hydration is taken into account and the original model coordinates are slightly rescaled. The comparison of models reveals that both the SAXS-based and the EM-based model lead to a similar simulation of the protein structure and to similar particle dimensions. The essential differences between the models concern the hexagonal bilayer arrangement (eclipsed in the SAXS model, one layer slightly rotated in the EM model), and the mass distribution, mainly on the surface and in the central part of the protein complex. © John Wiley & Sons, Inc.     Notes: Conformations of the α-l-Rhap(1-2)-β-d-Glc1-OMe and β-d-Galp(1-3)-β-d-Glc1-OMe disaccharides and the branched title trisaccharide were examined in DMSO-d6 solution by 1H-nmr. The distance mapping procedure was based on rotating frame nuclear Overhauser effect (NOE) constraints involving C- and O-linked protons, and hydrogen-bond constraints manifested by the splitting of the OH nmr signals for partially deuteriated samples. An “isotopomer-selected NOE” method for the unequivocal identification of mutually hydrogen-bonded hydroxyl groups was suggested. The length of hydrogen bonds thus detected is considered the only one motionally nonaveraged nmr-derived constraint. Molecular mechanics and molecular dynamics methods were used to model the conformational properties of the studied oligosaccharides. Complex conformational search, relying on a regular Φ,Ψ-grid based scanning of the conformational space of the selected glycosidic linkage, combined with simultaneous modeling of different allowed orientations of the pendant groups and the third, neighboring sugar residue, has been carried out. Energy minimizations were performed for each member of the Φ,Ψ grid generated set of conformations. Conformational clustering has been done to group the minimized conformations into families with similar values of glycosidic torsion angles. Several stable syn and anti conformations were found for the 1→2 and 1→3 bonds in the studied disaccharides. Vicinal glycosylation affected strongly the occupancy of conformational states in both branches of the title trisaccharide. The preferred conformational family of the trisaccharide (with average Φ,Ψ values of 38°, 17° for the 1→2 and 48°, 1° for the 1→3 bond, respectively) was shown by nmr to be stabilized by intramolecular hydrogen bonding between the nonbonded Rha and Gal residues. © 1998 John Wiley & Sons, Inc.     Notes: The present study was undertaken to examine the structural features that may be important to explain the immunogenicity of the (110-121) peptide sequence (FWRGDLVFDFQV) of VP3 capsid protein of hepatitis A virus. A conformational analysis of the preferred conformations by CD and molecular mechanics was carried out. Present results suggest that the interaction with liposomes as biomembrane model induces and stabilizes the amphipathic β-structure of the peptide.To study the contribution of amino acid replacements at the RGD tripeptide as well as the influence of the peptide chain length on peptide conformation, solid-phase peptide synthesis of several peptide analogs was carried out and the peptide conformation was studied using CD spectroscopy.     Notes: The critical volume fractions pertaining to the formation of DNA liquid crystals were obtained from polarization microscopy, 31P-nmr, and phase separation experiments. The DNA length (approximately one to two times the persistence length 50 nm), ionic strength, and counterion variety dependencies are reported. The cholesteric-isotropic transition is interpreted in terms of the coexistence equations, which are derived from the solution free energy including orientational entropy and excluded volume effects. With the wormlike chain as reference system, the electrostatic contribution to the free energy is evaluated as a thermodynamic perturbation in the second virial approximation with a Debye-Hückel potential of mean force. The hard core contribution has been evaluated with scaled particle theory and/or a simple generalization of the Carnahan-Starling equation of state for hard spheres. For sufficiently high ionic strengths, the agreement is almost quantitative. At lower amounts of added salt deviations are observed, which are tentatively attributed to counterion screening effects.     Notes: Gelation of gelatin under various conditions has been followed by atomic force microscopy (AFM) with the objective of understanding more fully the structure formed during the gelation process. AFM images were obtained of the structures formed from both the bulk sol and in surface films during the onset of gelation. While gelation occurred in the bulk sol, the extent of helix formation was monitored by measurements of optical rotation, and the molecular aggregation was imaged by AFM. Interfacial gelatin films formed at the air-water interface were also studied. Measurements of surface tension and surface rheology were made periodically and Langmuir-Blodgett films were drawn from the interface to allow AFM imaging of the structure of the interfacial layer as a function of time. Structural studies reveal that at low levels of helical content the gelatin molecules assemble into aggregates containing short segments of dimensions comparable to those expected for gelatin triple helices. With time larger fibrous structures appear whose dimensions suggest that they are bundles of triple helices. As gelation proceeds, the number density of fibers increases at the expense of the smaller aggregates, eventually assembling into a fibrous network. The gel structure appears to be sensitive to the thermal history, and this is particularly important in determining the structure and properties of the interfacial films. © 1998 John Wiley & Sons, Inc.     Notes: In this review I will outline several chemogenetic approaches used to determine the chemical basis of large ribozyme function and structure. The term chemogenetics was first used to describe site-specific functional group modification experiments in the analysis of DNA-protein interactions. Within the past few years equivalent experiments have been performed on large catalytic RNAs using both single-site substitution and interference mapping techniques with nucleotide analogues. While functional group mutagenesis is an important aspect of a chemogenetic approach, chemical correlates to genetic revertants and suppressors must also be realized for the genetic analogy to be intellectually valid and experimentally useful. Several examples of functional group revertants and suppressors have now been obtained within the Tetrahymena group I ribozyme. These experiments define an ensemble of tertiary hydrogen bonds that have made it possible to construct a detailed model of the ribozyme catalytic core. The model includes a functionally important monovalent metal ion binding site, a wobble-wobble receptor motif for helix-helix packing interactions, and a minor groove triple helix. © 1998 John Wiley & Sons, Inc.     Notes: In this review I discuss straightforward and general methods to modify nucleic acid structure with disulfide cross-links. A motivating factor in developing this chemistry was the notion that disulfide bonds would be excellent tools to probe the structure, dynamics, thermodynamics, folding, and function of DNA and RNA, much in the way that cystine cross-links have been used to study proteins. The chemistry described has been used to synthesize disulfide cross-linked hairpins and duplexes, higher order structures like triplexes, nonground-state conformations, and tRNAs. Since the cross-links form quantitatively by mild air oxidation and do not perturb either secondary or tertiary structure, this modification should prove quite useful for the study of nucleic acids. © 1998 John Wiley & Sons, Inc.     Notes: Divalent cations, like magnesium, are crucial for the structural integrity and biological activity of RNA. In this article, we present a picture of how magnesium stabilizes a particular folded form of RNA. The overall stabilization of RNA by Mg2+ is given by the free energy of transferring RNA from a reference univalent salt solution to a mixed salt solution. This term has favorable energetic contributions from two distinct modes of binding: diffuse binding and site binding. In diffuse binding, fully hydrated Mg ions interact with the RNA via nonspecific long-range electrostatic interactions. In site binding, dehydrated Mg2+ interacts with anionic ligands specifically arranged by the RNA fold to act as coordinating ligands for the metal ion. Each of these modes has a strong coulombic contribution to binding; however, site binding is also characterized by substantial changes in ion solvation and other nonelectrostatic contributions. We will show how these energetic differences can be exploited to experimentally distinguish between these two classes of ions using analyses of binding polynomials. We survey a number of specific systems in which Mg2+-RNA interactions have been studied. In well-characterized systems such as certain tRNAs and some rRNA fragments these studies show that site-bound ions can play an important role in RNA stability. However, the crucial role of diffusely bound ions is also evident. We emphasize that diffuse binding can only be described rigorously by a model that accounts for long-range electrostatic forces. To fully understand the role of magnesium ions in RNA stability, theoretical models describing electrostatic forces in systems with complicated structures must be developed. © 1999 John Wiley & Sons, Inc. The main interest in such peptides stems from their ability to interact with phospholipid bilayers and form voltage-dependent transmembrane channels in planar lipid bilayers. In order to provide insights into the lipid-peptide interaction promoting the voltage gating, the conformational study of TA VII in the presence of perdeuterated sodium dodecyl sulfate (SDS-d25) micelles has been carried out. 1H sequential assignments have been performed with the use of two-dimensional homo- and -heteronuclear nmr techniques including double quantum filtered correlated spectroscopy, homonuclear Hartmann-Hahn, nuclear Overhauser effect spectroscopy, 1H-13C heteronuclear single quantum correlation, and heteronuclear multiple bond correlation. Conformational parameters, such as 3JNHCαH coupling constants, temperature coefficients of amide protons (Δδ/ΔTNH) and quantitative nuclear Overhauser enhancement data, lead to detailed structural information. Ninety-eight three-dimensional structures consistent with the nmr data were generated from 231 interproton distances and six Φ dihedral angle restraints, using restrained molecular dynamics and energy minimization calculations. The average rms deviation between the 98 refined structures and the energy-minimized average structure is 0.59 Å for the backbone atoms. The structure of trichorzianin TA VII associated with SDS micelles, as determined by these methods, is characterized by two right-handed helical segments involving residues 1-8 and 11-19, linked by a β-turn that leads to an angle about 90°-100° between the two helix axes; residues 18 and 19 at the end of the C-terminal helix exhibit multiple conformations. © 1998 John Wiley & Sons, Inc.     Notes: The binding of 9-hydroxyellipticine to calf thymus DNA, poly[d(A-T)]2, and poly-[d(G-C)]2 has been studied in detail by means of CD, linear dichroism, resonance light scattering, and molecular dynamics. The transition moment polarizations of 9-hydroxyelliptiycine were determined in polyvinyl alcohol stretched film. Spectroscopic solution studies of the DNA/drug complex are combined with theoretical CD calculations using the final 50 ps of a series of molecular dynamics simulations as input. The spectroscopic data shows 9-hydroxyellipticine to adopt two main binding modes, one intercalative and the other a stacked binding mode involving the formation of drug oligomers in the DNA major groove. Analysis of the intercalated binding mode in poly[d(A-T)]2 suggests the 9-hydroxyellipticine hydroxyl group lies in the minor groove and hydrogen bonds to water with the pyridine ring protruding into the major groove. The stacked binding mode was examined using resonance light scattering and it was concluded that the drug was forming small oligomer stacks rather than extended aggregates. Reduced linear dichroism measurements suggested a binding geometry that precluded a minor groove binding mode where the plane of the drug makes a 45° angle with the plane of the bases. Thus it was concluded that the drug stacks in the major groove. No obvious differences in the mode of binding of 9-hydroxyellipticine were observed between different DNA sequences; however, the stacked binding mode appeared to be more favorable for calf thymus DNA and poly[d(G-C)]2 than for poly[d(A-T)]2, an observation that could be explained by the slightly greater steric hindrance of the poly[d(A-T)]2 major groove. A strong concentration dependence was observed for the two binding modes where intercalation is favored at very low drug load, with stacking interactions becoming more prominent as the drug concentration is increased. Even at DNA : drug mixing ratios of 70:1 the stacked binding mode was still important for GC-rich DNAs. © 1998 John Wiley & Sons, Inc. Biopoly 46: 127-143, 1998

    Notes: The surface properties at the air/water interface and the interaction of branched chain polymeric polypeptides with a general formula poly[Lys-(DL-Alam-X1)], where X = Π (AK), Ser (SAK), or Glu (EAK), with phospholipids were investigated. Polylysine derivatives with polycationic (SAK, AK) or amphoteric (EAK) were capable to spread and form stable monomolecular layers. The stability of monolayers at the air/water interface was dependent on the side-chain terminal amino acid residue of polymers and can be described by SAK 〈 AK 〈 EAK order. The area per amino acid residue values calculated from compression isotherms were in the same range as compared to those of linear poly-α-amino acids and proteins. Moreover, these polymers interact with phospholipid monomolecular layers composed of dipalmitoyl phosphatidyl choline (DPPC) or DPPC/PG (PG: phosphatidyl glycerol; 95/5, mol/mol). Data obtained from compression isotherms of phospholipids spread on aqueous polymer solutions at different initial surface pressure indicated that insertion into lipid monolayers for SAK or AK is more pronounced than for EAK. The interaction between branched polypeptides and phospholipid membranes was further investigated using lipid bilayers with DPPC/PG and fluorescent probes located either at the polar surface [1-(4-trimethylammonium-phenyl)-6-phenyl-1,3,5-hexatriene (TMA-DPH) sodium anilino naphthalene sulfonate (ANS)] or within the hydrophobic core (DPH) of the liposome. Changes in fluorescence intensity and in polarization were observed when TMA-DPH or ANS, but not DPH were used. Comparative data also indicate that all three polymers interact only with the outer surface of the bilayer, but even the most marked penetration of polycationic polypeptide (SAK) did not result in alteration of the ordered state of the alkyl chains in the bilayer. Taken together, data obtained from mono- or bilayer experiments suggest that the interaction between branched polymers and phospholipids are highly dependent on the charge properties (Ser vs Glu) and on the identity (Ser vs Ala) of side-chain terminating amino acids. The binding of polymers to the model membranes could be mainly driven by electrostatic forces, but the significant role of hydrophilic properties in case of SAK cannot be excluded. © 1998 John Wiley & Sons, Inc.     Notes: The relationship between the local backbone conformation and bond angles at Cα of symmetrically substituted Cα,α-dialkylated glycines (Cα,α-dimethylglycine or α-aminoisobutyric acid, Aib; Cα,α-diethylglycine, Deg; Cα,α-di-n-propylglycine, Dpg) has been investigated by molecular dynamics (MD) simulation adopting flat bottom harmonic potentials, instead of the usual harmonic restraints, for the Cα bond angles. The MD simulations show that the Cα bond angles are related to the local backbone conformation, irrespectively of the side-chain length of Aib, Deg, and Dpg residues. Moreover, the N-Cα-C′ (τ) angle is the most sensitive conformational parameter and, in the folded form, is always larger and more flexible than in the extended one. © 1998 John Wiley & Sons, Inc.     Notes: In biological systems, both proteolysis and aminolysis of amide bonds produce activated intermediates through acyl transfer reactions either inter- or intramolecularly. Protein splicing is an illustrative example that proceeds through a series of catalyzed acyl transfer reactions and culminates at an O- or S-acyl intermediate. This intermediate leads to an uncatalyzed acyl migration to form an amide bond in the spliced product. A ligation method mimicking the uncatalyzed final steps in protein splicing has been developed utilizing the acyl transfer amide-bond feature for the blockwise coupling of unprotected, free peptide segments at methionine (Met). The latent thiol moiety of Met can be exploited using homocysteine at the α-amino terminal position of a free peptide for transthioesterification with another free peptide containing an α-thioester to give an S-acyl intermediate. A subsequent, proximity-driven S- to N-acyl migration of this acyl intermediate spontaneously rearranges to form a homocysteinyl amide bond. S-methylation with excess p-nitrobenezensulfonate yields Met at the ligation site. The methionine ligation is selective and orthogonal, and is usually completed within 4 h when performed at slightly basic pH and under strongly reductive conditions. No side reactions due to acylation were observed with any other α-amines of both peptide segments as seen in the synthesis of parathyroid hormone peptides. Furthermore, cyclic peptide can also be obtained through the same strategy by placing both homocysteine at the amino terminus and the thioester at the carboxyl terminus in an unprotected peptide precursor. These biomimetic ligation strategies hold promise for engineering novel peptides and proteins. © 1998 John Wiley & Sons, Inc. Biopoly 46: 319-327, 1998

    Notes: Boundary element methods are used to model the free solution electrophoretic mobility of short DNA fragments. The Stern surfaces of the DNA fragments are modeled as plated cylinders that reproduce translational and rotational diffusion constants. The solvent-accessible and ion-accessible surfaces are taken to be coincident with the Stern surface. The mobilities are computed by solving simultaneously the coupled Navier-Stokes, Poisson, and ion-transport equations. The equilibrium electrostatics are treated at the level of the full Poisson-Boltzmann equation and ion relaxation is included. For polyions as highly charged as short DNA fragments, ion relaxation is substantial. At .11 M KCl, the simulated mobilities of a 20 base pair DNA fragment are in excellent agreement with experiment. At .04 M Tris acetate, pH = 8.0, the simulated mobilities are about 10-15% higher than experimental values and this discrepancy is attributed to the relatively large size of the Tris counterion. The length dependence of the mobility at .11 M KCl is also investigated. Earlier mobility studies on lysozyme are reexamined in view of the present findings. In addition to electrophoretic mobilities, the effective polyion charge measured in steady state electrophoresis and its relationship to the preferential interaction parameter γgG is briefly considered. © 1998 John Wiley & Sons, Inc.
