    Beschreibung: The diagnosis of the conservation state of monumental structures from constraints to the spatial distribution of their physical properties on shallow and inner materials represents one of the key objectives in the application of non-invasive techniques. In situ, CRP and 3D ultrasonic tomography can provide an effective coverage of stone materials in space and time. The intrinsic characteristics of the materials that make up a monumental structure and affect the two properties (i.e., reflectivity, longitudinal velocity) through the above methods substantially differ. Consequently, the content of their information is mainly complementary rather than redundant. In this study we present the integrated application of different non-destructive techniques i.e., Close Range Photogrammetry (CRP), and low frequency (24 KHz) ultrasonic tomography complemented by petrographycal analysis based essentially on Optical Microscopy (OM). This integrated methodology has been applied to a Carrara marble column of the Basilica of San Saturnino, in Byzantine-Proto-Romanesque style, which is part of the Paleo Christian complex of the V-VI century. This complex also includes the adjacent Christian necropolis in the square of San Cosimo in the city of Cagliari, Sardinia, Italy. The column under study is made of bare material dating back probably to the first century A.D., it was subjected to various traumas due to disassembly and transport to the site, including damage caused by the close blast of a WWII fragmentation bomb. High resolution 3D modelling of the studied artifact was computed starting from the integration of proximal sensing techniques such as CRP based on Structure from Motion (SfM), with which information about the geometrical anomalies and reflectivity of the investigated marble column surface was obtained. On the other hand, the inner parts of the studied body were successfully inspected in a non-invasive way by computing the velocity pattern of the ultrasonic signal through the investigated materials using 3D ultrasonic tomography. This technique gives information on the elastic properties of the material related with mechanical properties and a number of factors, such as presence of fractures, voids, and flaws. Extracting information on such factors from the elastic wave velocity using 3D tomography provides a non-invasive approach to analyse the property changes of the inner material of the ancient column. The integrated application of in situ CRP and ultrasonic techniques provides a full 3D high resolution model of the investigated artifact. This model enhanced by the knowledge of the petrographic characteristics of the materials, improves the diagnostic process and affords reliable information on the state of conservation of the materials used in the construction processes of the studied monumental structure. The integrated use of the non-destructive techniques described above also provides suitable data for a possible restoration and future preservation.

    Beschreibung: Originating from the boreal forest and often transported over large distances, driftwood characterises many Arctic coastlines. Here we present a combined assessment of radiocarbon (14C) and dendrochronological (ring width) age estimates of driftwood samples to constrain the progradation of two Holocene beach-ridge systems near the Lena Delta in the Siberian Arctic (Laptev Sea). Our data show that the 14C ages obtained on syndepositional driftwood from beach deposits yield surprisingly coherent chronologies for the coastal evolution of the field sites. The dendrochronological analysis of wood from modern driftlines revealed the origin and recent delivery of the wood from the Lena River catchments. This finding suggests that the duration transport lies within the uncertainty of state-of-the-art 14C dating and thus substantiates the validity of age indication obtained from driftwood. This observation will help to better understand changes in similar coastal environments, and to improve our knowledge about the response of coastal systems to past climate and sea-level changes.

    Beschreibung: Earth system and climate modelling involves the simulation of processes on a wide range of scales and within and across various compartments of the Earth system. In practice, component models are often developed independently by different research groups, adapted by others to their special interests and then combined using a dedicated coupling software. This procedure not only leads to a strongly growing number of available versions of model components and coupled setups but also to model- and high-performance computing (HPC)-system-dependent ways of obtaining, configuring, building and operating them. Therefore, implementing these Earth system models (ESMs) can be challenging and extremely time consuming, especially for less experienced modellers or scientists aiming to use different ESMs as in the case of intercomparison projects. To assist researchers and modellers by reducing avoidable complexity, we developed the ESM-Tools software, which provides a standard way for downloading, configuring, compiling, running and monitoring different models on a variety of HPC systems. It should be noted that ESM-Tools is not a coupling software itself but a workflow and infrastructure management tool to provide access to increase usability of already existing components and coupled setups. As coupled ESMs are technically the more challenging tasks, we will focus on coupled setups, always implying that stand-alone models can benefit in the same way. With ESM-Tools, the user is only required to provide a short script consisting of only the experiment-specific definitions, while the software executes all the phases of a simulation in the correct order. The software, which is well documented and easy to install and use, currently supports four ocean models, three atmosphere models, two biogeochemistry models, an ice sheet model, an isostatic adjustment model, a hydrology model and a land-surface model. Compared to previous versions, ESM-Tools has lately been entirely recoded in a high-level programming language (Python) and provides researchers with an even more user-friendly interface for Earth system modelling. ESM-Tools was developed within the framework of the Advanced Earth System Model Capacity project, supported by the Helmholtz Association.

    Beschreibung: Numerous studies have addressed the possible existence of large floating ice sheets in the glacial Arctic Ocean from theoretical, modelling, or seafloor morphology perspectives. Here, we add evidence from the sediment record that support the existence of such freshwater ice caps in certain intervals, and we discuss their implications for possible non-linear and rapid behaviour of such a system in the high latitudes. We present sedimentary activities of 230Th together with 234U/238U ratios, the concentrations of manganese, sulphur and calcium in the context of lithological information and records of microfossils and their isotope composition. New analyses (PS51/038, PS72/396) and a re-analysis of existing marine sediment records (PS1533, PS1235, PS2185, PS2200, amongst others) in view of the naturally occurring radionuclide 230Thex and, where available, 10Be from the Arctic Ocean and the Nordic Seas reveal the widespread occurrence of intervals with a specific geochemical signature. The pattern of these parameters in a pan-Arctic view can best be explained when assuming the repeated presence of freshwater in frozen and liquid form across large parts of the Arctic Ocean and the Nordic Seas. Based on the sedimentary evidence and known environmental constraints at the time, we develop a glacial scenario that explains how these ice sheets, together with eustatic sea-level changes, may have affected the past oceanography of the Arctic Ocean in a fundamental way that must have led to a drastic and non-linear response to external forcing. This concept offers a possibility to explain and to some extent reconcile contrasting age models for the Late Pleistocene in the Arctic Ocean. Our view, if adopted, offers a coherent dating approach across the Arctic Ocean and the Nordic Seas, linked to events outside the Arctic.

    Beschreibung: The modeling of paleoclimate, using physically based tools, is increasingly seen as a strong out-of-sample test of the models that are used for the projection of future climate changes. New to the Coupled Model Intercomparison Project (CMIP6) is the Tier 1 Last Interglacial experiment for 127 000 years ago (lig127k), designed to address the climate responses to stronger orbital forcing than the midHolocene experiment, using the same state-of-the-art models as for the future and following a common experimental protocol. Here we present a first analysis of a multi-model ensemble of 17 climate models, all of which have completed the CMIP6 DECK (Diagnostic, Evaluation and Characterization of Klima) experiments. The equilibrium climate sensitivity (ECS) of these models varies from 1.8 to 5.6 ∘C. The seasonal character of the insolation anomalies results in strong summer warming over the Northern Hemisphere continents in the lig127k ensemble as compared to the CMIP6 piControl and much-reduced minimum sea ice in the Arctic. The multi-model results indicate enhanced summer monsoonal precipitation in the Northern Hemisphere and reductions in the Southern Hemisphere. These responses are greater in the lig127k than the CMIP6 midHolocene simulations as expected from the larger insolation anomalies at 127 than 6 ka. New synthesis for surface temperature and precipitation, targeted for 127 ka, have been developed for comparison to the multi-model ensemble. The lig127k model ensemble and data reconstructions are in good agreement for summer temperature anomalies over Canada, Scandinavia, and the North Atlantic and for precipitation over the Northern Hemisphere continents. The model–data comparisons and mismatches point to further study of the sensitivity of the simulations to uncertainties in the boundary conditions and of the uncertainties and sparse coverage in current proxy reconstructions. The CMIP6–Paleoclimate Modeling Intercomparison Project (PMIP4) lig127k simulations, in combination with the proxy record, improve our confidence in future projections of monsoons, surface temperature, and Arctic sea ice, thus providing a key target for model evaluation and optimization.

    Beschreibung: In order to investigate the impact of spatial resolution on the discrepancy between simulated δ18O and observed δ18O in Greenland ice cores, regional climate simulations are performed with the isotope-enabled regional climate model (RCM) COSMO_iso. For this purpose, isotope-enabled general circulation model (GCM) simulations with the ECHAM5-wiso general circulation model (GCM) under present-day conditions and the MPI-ESM-wiso GCM under mid-Holocene conditions are dynamically downscaled with COSMO_iso for the Arctic region. The capability of COSMO_iso to reproduce observed isotopic ratios in Greenland ice cores for these two periods is investigated by comparing the simulation results to measured δ18O ratios from snow pit samples, Global Network of Isotopes in Precipitation (GNIP) stations and ice cores. To our knowledge, this is the first time that a mid-Holocene isotope-enabled RCM simulation is performed for the Arctic region. Under present-day conditions, a dynamical downscaling of ECHAM5-wiso (1.1◦ × 1.1◦) with COSMO_iso to a spatial resolution of 50km improves the agreement with the measured δ18O ratios for 14 of 19 observational data sets. A further increase in the spatial resolution to 7km does not yield substantial improvements except for the coastal areas with its complex terrain. For the mid-Holocene, a fully coupled MPI-ESM-wiso time slice simulation is downscaled with COSMO_iso to a spatial resolution of 50km. In the mid-Holocene, MPI-ESM-wiso already agrees well with observations in Greenland and a downscaling with COSMO_iso does not further improve the model–data agreement. Despite this lack of improvement in model biases, the study shows that in both periods, observed δ18O values at measurement sites constitute isotope ratios which are mainly within the subgrid-scale variability of the global ECHAM5-wiso and MPI-ESM-wiso simulation results. The correct δ18O ratios are consequently not resolved in the GCM simulation results and need to be extracted by a refinement with an RCM. In this context, the RCM simulations provide a spatial δ18O distribution by which the effects of local uncertainties can be taken into account in the comparison between point measurements and model outputs. Thus, an isotope-enabled GCM–RCM model chain with realistically implemented fractionating processes constitutes a useful supplement to reconstruct regional paleo-climate conditions during the mid-Holocene in Greenland. Such model chains might also be applied to reveal the full potential of GCMs in other regions and climate periods, in which large deviations relative to observed isotope ratios are simulated.

    Beschreibung: Proxy climate records are an invaluable source of information about the earth’s climate prior to the instrumental record. The temporal- and spatial-coverage of records continues to increase, however, these records of past climate are associated with significant uncertainties due to non-climate processes that influence the recorded and measured proxy values. Generally, these uncertainties are timescale-dependent and correlated in time. Accounting for structure in the errors is essential to providing realistic error estimates for smoothed or stacked records, detection of anomalies and identifying trends, but this structure is seldom accounted for. In the first of these companion articles we outlined a theoretical framework for handling proxy uncertainties by deriving the power spectrum of proxy error components from which it is possible to obtain timescale-dependent error estimates. Here in part II, we demonstrate the practical application of this theoretical framework using the example of marine sediment cores. We consider how to obtain estimates for the required parameters and give examples of the application of this approach for typical marine sediment proxy records. Our new approach of estimating and providing timescale-dependent proxy errors overcomes the limitations of simplistic single value error estimates. We aim to provide the conceptual basis for a more quantitative use of paleo-records for applications such as model-data comparison, regional and global synthesis of past climate states and data assimilation.

    Beschreibung: The mid-Pliocene warm period (mPWP; ∼3.2 million years ago) is seen as the most recent time period characterized by a warm climate state, with similar to modern geography and ∼400 ppmv atmospheric CO2 concentration, and is therefore often considered an interesting analogue for near-future climate projections. Paleoenvironmental reconstructions indicate higher surface temperatures, decreasing tropical deserts, and a more humid climate in West Africa characterized by a strengthened West African Monsoon (WAM). Using model results from the second phase of the Pliocene Modelling Intercomparison Project (PlioMIP2) ensemble, we analyse changes of the WAM rainfall during the mPWP by comparing them with the control simulations for the pre-industrial period. The ensemble shows a robust increase in the summer rainfall over West Africa and the Sahara region, with an average increase of 2.5 mm/d, contrasted by a rainfall decrease over the equatorial Atlantic. An anomalous warming of the Sahara and deepening of the Saharan Heat Low, seen in 〉90 % of the models, leads to a strengthening of the WAM and an increased monsoonal flow into the continent. A similar warming of the Sahara is seen in future projections using both phase 3 and 5 of the Coupled Model Intercomparison Project (CMIP3 and CMIP5). Though previous studies of future projections indicate a west–east drying–wetting contrast over the Sahel, PlioMIP2 simulations indicate a uniform rainfall increase in that region in warm climates characterized by increasing greenhouse gas forcing. We note that this effect will further depend on the long-term response of the vegetation to the CO2 forcing.

    Beschreibung: Due to its dryness, the subtropical free troposphere plays a critical role in the radiative balance of the Earth's climate system. But the complex interactions of the dynamical and physical processes controlling the variability in the moisture budget of this sensitive region of the subtropical atmosphere are still not fully understood. Stable water isotopes can provide important information about several of the latter processes, namely subsidence drying, turbulent mixing, and dry and moist convective moistening. In this study, we use high-resolution simulations of the isotope-enabled version of the regional weather and climate prediction model of the Consortium for Small-Scale Modelling (COSMOiso) to investigate predominant moisture transport pathways in the Canary Islands region in the eastern subtropical North Atlantic. Comparison of the simulated isotope signals with multi-platform isotope observations (aircraft, ground- and space-based remote sensing) from a field campaign in summer 2013 shows that COSMOiso can reproduce the observed variability of stable water vapour isotopes on timescales of hours to days, thus allowing us to study the mechanisms that control the subtropical free-tropospheric humidity. Changes in isotopic signals along backward trajectories from the Canary Islands region reveal the physical processes behind the synoptic-scale isotope variability. We identify four predominant moisture transport pathways of mid-tropospheric air, each with distinct isotopic signatures: - air parcels originating from the convective boundary layer of the Saharan heat low (SHL) – these are characterised by a homogeneous isotopic composition with a particularly high δD (median mid-tropospheric δD=−122‰), which results from dry convective mixing of low-level moisture of diverse origin advected into the SHL; - air parcels originating from the free troposphere above the SHL – although experiencing the largest changes in humidity and δD during their subsidence over West Africa, these air parcels typically have lower δD values (median δD=−148‰) than air parcels originating from the boundary layer of the SHL; - air parcels originating from outside the SHL region, typically descending from tropical upper levels south of the SHL, which are often affected by moist convective injections from mesoscale convective systems in the Sahel – their isotopic composition is much less enriched in heavy isotopes (median δD=−175‰) than those from the SHL region; - air parcels subsiding from the upper-level extratropical North Atlantic – this pathway leads to the driest and most depleted conditions (median δD=−255‰) in the middle troposphere near the Canary Islands. The alternation of these transport pathways explains the observed high variability in humidity and δD on synoptic timescales to a large degree. We further show that the four different transport pathways are related to specific large-scale flow conditions. In particular, distinct differences in the location of the North African mid-level anticyclone and of extratropical Rossby wave patterns occur between the four transport pathways. Overall, this study demonstrates that the adopted Lagrangian isotope perspective enhances our understanding of air mass transport and mixing and offers a sound interpretation of the free-tropospheric variability of specific humidity and isotope composition on timescales of hours to days in contrasting atmospheric conditions over the eastern subtropical North Atlantic.

    Beschreibung: Organic and organometallic reactants in aqueous electrolytes, being composed of earth-abundant elements, are promising redox active candidates for cost-effective organic redox flow batteries (ORFBs). Various compounds of ferrocene and methyl viologen have been examined as promising redox actives for this application. Herein, we examined the influence of the electrolyte pH and the salt anion on model redox active organic cations, bis((3-trimethylammonio) propyl)- ferrocene dichloride (BTMAP-Fc) and bis(3-trimethylammonio) propyl viologen tetrachloride (BTMAP-Vi), which have exhibited excellent cycling stability and capacity retention at ≥1.00 M concentration [E. S. Beh, et al. ACS Energy Lett. 2, 639–644 (2017)]. We examined the solvation shell around BTMAP-Fc and BTMAP-Vi at acidic and neutral pH with SO42-, Cl−, and CH3SO3− counterions and elucidated their impact on cation diffusion coefficient, first electron transfer rate constant, and thereby the electrochemical Thiele modulus. The electrochemical Thiele modulus was found to be exponentially correlated with the solvent reorganizational energy (λ) in both neutral and acidic pH. Thus, λ is proposed as a universal descriptor and selection criteria for organic redox flow battery electrolyte compositions. In the specific case of the BTMAP-Fc/BTMAP-Vi ORFB, low pH electrolytes with methanesulfonate or chloride counterions were identified as offering the best balance of transport and kinetic requirements.

    Beschreibung: When displayed on erythrocytes, peptides and proteins can drive antigen-specific immune tolerance. Here, we investigated a straightforward approach based on erythrocyte binding to promote antigen-specific tolerance to both peptides and proteins. We first identified a robust erythrocyte-binding ligand. A pool of one million fully d-chiral peptides was injected into mice, blood cells were isolated, and ligands enriched on these cells were identified using nano-liquid chromatography–tandem mass spectrometry. One round of selection yielded a murine erythrocyte-binding ligand with an 80 nM apparent dissociation constant, Kd. We modified an 83-kDa bacterial protein and a peptide antigen derived from ovalbumin (OVA) with the identified erythrocyte-binding ligand. An administration of the engineered bacterial protein led to decreased protein-specific antibodies in mice. Similarly, mice given the engineered OVA-derived peptide had decreased inflammatory anti-OVA CD8+ T cell responses. These findings suggest that our tolerance-induction strategy is applicable to both peptide and protein antigens and that our in vivo selection strategy can be used for de novo discovery of robust erythrocyte-binding ligands.

    Beschreibung: Platinum group elements (PGE) are considered to be very poorly soluble in aqueous fluids in most natural hydrothermal–magmatic contexts and industrial processes. Here, we combined in situ X-ray absorption spectroscopy and solubility experiments with atomistic and thermodynamic simulations to demonstrate that the trisulfur radical ion S3•− forms very stable and soluble complexes with both PtII and PtIV in sulfur-bearing aqueous solution at elevated temperatures (∼300 °C). These Pt-bearing species enable (re)mobilization, transfer, and focused precipitation of platinum up to 10,000 times more efficiently than any other common inorganic ligand, such as hydroxide, chloride, sulfate, or sulfide. Our results imply a far more important contribution of sulfur-bearing hydrothermal fluids to PGE transfer and accumulation in the Earth’s crust than believed previously. This discovery challenges traditional models of PGE economic concentration from silicate and sulfide melts and provides new possibilities for resource prospecting in hydrothermal shallow crust settings. The exceptionally high capacity of the S3•− ion to bind platinum may also offer new routes for PGE selective extraction from ore and hydrothermal synthesis of noble metal nanomaterials.

    Beschreibung: Gram-negative bacteria take up the essential ion Fe3+ as ferric-siderophore complexes through their outer membrane using TonB-dependent transporters. However, the subsequent route through the inner membrane differs across many bacterial species and siderophore chemistries and is not understood in detail. Here, we report the crystal structure of the inner membrane protein FoxB (from Pseudomonas aeruginosa) that is involved in Fe-siderophore uptake. The structure revealed a fold with two tightly bound heme molecules. In combination with in vitro reduction assays and in vivo iron uptake studies, these results establish FoxB as an inner membrane reductase involved in the release of iron from ferrioxamine during Fe-siderophore uptake.

    Beschreibung: We consider a nonlinear autonomous system of N≫1 degrees of freedom randomly coupled by both relaxational (“gradient”) and nonrelaxational (“solenoidal”) random interactions. We show that with increased interaction strength, such systems generically undergo an abrupt transition from a trivial phase portrait with a single stable equilibrium into a topologically nontrivial regime of “absolute instability” where equilibria are on average exponentially abundant, but typically, all of them are unstable, unless the dynamics is purely gradient. When interactions increase even further, the stable equilibria eventually become on average exponentially abundant unless the interaction is purely solenoidal. We further calculate the mean proportion of equilibria that have a fixed fraction of unstable directions.

    Beschreibung: Humans and nonhuman animals display conformist as well as anticonformist biases in cultural transmission. Whereas many previous mathematical models have incorporated constant conformity coefficients, empirical research suggests that the extent of (anti)conformity in populations can change over time. We incorporate stochastic time-varying conformity coefficients into a widely used conformity model, which assumes a fixed number n of “role models” sampled by each individual. We also allow the number of role models to vary over time (nt). Under anticonformity, nonconvergence can occur in deterministic and stochastic models with different parameter values. Even if strong anticonformity may occur, if conformity or random copying (i.e., neither conformity nor anticonformity) is expected, there is convergence to one of the three equilibria seen in previous deterministic models of conformity. Moreover, this result is robust to stochastic variation in nt. However, dynamic properties of these equilibria may be different from those in deterministic models. For example, with random conformity coefficients, all equilibria can be stochastically locally stable simultaneously. Finally, we study the effect of randomly changing weak selection. Allowing the level of conformity, the number of role models, and selection to vary stochastically may produce a more realistic representation of the wide range of group-level properties that can emerge under (anti)conformist biases. This promises to make interpretation of the effect of conformity on differences between populations, for example those connected by migration, rather difficult. Future research incorporating finite population sizes and migration would contribute added realism to these models.

    Beschreibung: Usutu virus (USUV) is an emerging arbovirus in Europe that has been increasingly identified in asymptomatic humans and donated blood samples and is a cause of increased incidents of neuroinvasive human disease. Treatment or prevention options for USUV disease are currently nonexistent, the result of a lack of understanding of the fundamental elements of USUV pathogenesis. Here, we report two structures of the mature USUV virus, determined at a resolution of 2.4 Å, using single-particle cryogenic electron microscopy. Mature USUV is an icosahedral shell of 180 copies of envelope (E) and membrane (M) proteins arranged in the classic herringbone pattern. However, unlike previous reports of flavivirus structures, we observe virus subpopulations and differences in the fusion loop disulfide bond. Presence of a second, unique E glycosylation site could elucidate host interactions, contributing to the broad USUV tissue tropism. The structures provide a basis for exploring USUV interactions with glycosaminoglycans and lectins, the role of the RGD motif as a receptor, and the inability of West Nile virus therapeutic antibody E16 to neutralize the mature USUV strain SAAR-1776. Finally, we identify three lipid binding sites and predict key residues that likely participate in virus stability and flexibility during membrane fusion. Our findings provide a framework for the development of USUV therapeutics and expand the current knowledge base of flavivirus biology.

    Beschreibung: Regulation systems for fluid-driven soft robots predominantly consist of inflexible and bulky components. These rigid structures considerably limit the adaptability and mobility of these robots. Soft valves in various forms for fluidic actuators have been developed, primarily fluidically or electrically driven. However, fluidic soft valves require external pressure sources that limit robot locomotion. State-of-the-art electrostatic valves are unable to modulate pressure beyond 3.5 kPa with a sufficient flow rate (〉6 mL⋅min−1). In this work, we present an electrically powered soft valve for hydraulic actuators with mesoscale channels based on a different class of ultrahigh-power density dynamic dielectric elastomer actuators. The dynamic dielectric elastomer actuators (DEAs) are actuated at 500 Hz or above. These DEAs generate 300% higher blocked force compared with the dynamic DEAs in previous works and their loaded power density reaches 290 W⋅kg−1 at operating conditions. The soft valves are developed with compact (7 mm tall) and lightweight (0.35 g) dynamic DEAs, and they allow effective control of up to 51 kPa of pressure and a 40 mL⋅min−1 flow rate with a response time less than 0.1 s. The valves can also tune flow rates based on their driving voltages. Using the DEA soft valves, we demonstrate control of hydraulic actuators of different volumes and achieve independent control of multiple actuators powered by a single pressure source. This compact and lightweight DEA valve is capable of unprecedented electrical control of hydraulic actuators, showing the potential for future onboard motion control of soft fluid-driven robots.

    Beschreibung: Metapopulation capacity provides an analytic tool to quantify the impact of landscape configuration on metapopulation persistence, which has proven powerful in biological conservation. Yet surprisingly few efforts have been made to apply this approach to multispecies systems. Here, we extend metapopulation capacity theory to predict the persistence of trophically interacting species. Our results demonstrate that metapopulation capacity could be used to predict the persistence of trophic systems such as prey–predator pairs and food chains in fragmented landscapes. In particular, we derive explicit predictions for food chain length as a function of metapopulation capacity, top-down control, and population dynamical parameters. Under certain assumptions, we show that the fraction of empty patches for the basal species provides a useful indicator to predict the length of food chains that a fragmented landscape can support and confirm this prediction for a host–parasitoid interaction. We further show that the impact of habitat changes on biodiversity can be predicted from changes in metapopulation capacity or approximately by changes in the fraction of empty patches. Our study provides an important step toward a spatially explicit theory of trophic metacommunities and a useful tool for predicting their responses to habitat changes.

    Beschreibung: Hyrrokkin sarcophaga is a parasitic foraminifera that is commonly found in cold-water coral reefs where it infests the file clam Acesta excavata and the scleractinian coral Desmophyllum pertusum (formerly known as Lophelia pertusa). Here, we present measurements of the trace element and isotopic composition of these parasitic foraminifera, analyzed by inductively coupled optical emission spectrometry (ICP-OES), electron probe microanalysis (EPMA) and mass spectrometry (gas-source MS and inductively-coupled-plasma MS). Our results reveal that the geochemical signature of H. sarcophaga depends on the host organism it infests. Sr / Ca ratios are 1.1 mmol mol−1 higher in H. sarcophaga that infest D. pertusum, which could be an indication that dissolved host carbonate material is utilized in shell calcification, given that the aragonite of D. pertusum has a naturally higher Sr concentration compared to the calcite of A. excavata. Similarly, we measure 3.1 ‰ lower δ13C and 0.25 ‰ lower δ18O values in H. sarcophaga that lived on D. pertusum, which might be caused by the direct uptake of the host's carbonate material with a more negative isotopic composition or different pH regimes in these foraminifera (pH can exert a control on the extent of CO2 hydration/hydroxylation) due to the uptake of body fluids of the host. We also observe higher Mn / Ca ratios in foraminifera that lived on A. excavata but did not penetrate the host shell compared to specimen that penetrated the shell, which could be interpreted as a change in food source, changes in the calcification rate, Rayleigh fractionation or changing oxygen conditions. While our measurements provide an interesting insight into the calcification process of this unusual foraminifera, these data also indicate that the geochemistry of this parasitic foraminifera is unlikely to be a reliable indicator of paleoenvironmental conditions using Sr / Ca, Mn / Ca, δ18O or δ13C unless the host organism is known and its geochemical composition can be accounted for.

    Beschreibung: Braiding of topological structures in complex matter fields provides a robust framework for encoding and processing information, and it has been extensively studied in the context of topological quantum computation. In living systems, topological defects are crucial for the localization and organization of biochemical signaling waves, but their braiding dynamics remain unexplored. Here, we show that the spiral wave cores, which organize the Rho-GTP protein signaling dynamics and force generation on the membrane of starfish egg cells, undergo spontaneous braiding dynamics. Experimentally measured world line braiding exponents and topological entropy correlate with cellular activity and agree with predictions from a generic field theory. Our analysis further reveals the creation and annihilation of virtual quasi-particle excitations during defect scattering events, suggesting phenomenological parallels between quantum and living matter.

    Beschreibung: In this work we have performed a detailed study of vectors to ore to a representative volcanic-rock-hosted replacive volcanogenic massive sulfide (VMS) deposit located in the northern Iberian Pyrite Belt (Spain), the Aguas Teñidas deposit. The investigated vectors include the following: (1) mineralogical zoning, (2) host sequence characterization and mineralized unit identification based on whole rock geochemistry discrimination diagrams, (3) study of the characteristics and behaviour of whole rock geochemical anomalies around the ore (e.g. alteration-related compositional changes, characteristics and extent of geochemical halos of indicative elements such as Cu, Zn, Pb, Sb, Tl, and Ba around the deposit), and (4) application of portable X-ray fluorescence (p-XRF) analysis to the detection of the previous vectors. In the footwall, a concentric cone-shaped hydrothermal alteration zone bearing the stockwork passes laterally, from core to edge, from quartz (only local) to chlorite–quartz, sericite–chlorite–quartz, and sericite–quartz alteration zones. The hydrothermal alteration is also found in the hanging wall despite being tectonically allochthonous to the orebody: a proximal sericite alteration zone is followed by a more distal albite-rich one. Whole rock major elements show an increase in alteration indexes (e.g. AI, CCPI) towards the mineralization, a general SiO2 enrichment, and FeO enrichment as well as K2O and Na2O depletion towards the centre of the hydrothermal system, with MgO showing a less systematic behaviour. K2O and Na2O leached from the centre of the system are transported and deposited in more external areas. Copper, Pb, and Zn produce proximal anomalies around mineralized areas, with the more mobile Sb, Tl, and Ba generating wider halos. Whereas Sb and Tl halos form around all mineralized areas, Ba anomalies are restricted to areas around the massive sulfide body. Our results show that proposed vectors, or adaptations designed to overcome p-XRF limitations, can be confidently used by analysing unprepared hand specimens, including the external rough curved surface of drill cores. The data presented in this work are not only applicable to VMS exploration in the Iberian Pyrite Belt, but on a broader scale they will also contribute to improving our general understanding of vectors to ore in replacive-type VMS deposits.

    Beschreibung: We combine satellite data products to provide a first and general overview of the physical sea ice conditions along the drift of the international Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition and a comparison with previous years (2005–2006 to 2018–2019). We find that the MOSAiC drift was around 20 % faster than the climatological mean drift, as a consequence of large-scale low-pressure anomalies prevailing around the Barents–Kara–Laptev sea region between January and March. In winter (October–April), satellite observations show that the sea ice in the vicinity of the Central Observatory (CO; 50 km radius) was rather thin compared to the previous years along the same trajectory. Unlike ice thickness, satellite-derived sea ice concentration, lead frequency and snow thickness during winter months were close to the long-term mean with little variability. With the onset of spring and decreasing distance to the Fram Strait, variability in ice concentration and lead activity increased. In addition, the frequency and strength of deformation events (divergence, convergence and shear) were higher during summer than during winter. Overall, we find that sea ice conditions observed within 5 km distance of the CO are representative for the wider (50 and 100 km) surroundings. An exception is the ice thickness; here we find that sea ice within 50 km radius of the CO was thinner than sea ice within a 100 km radius by a small but consistent factor (4 %) for successive monthly averages. Moreover, satellite acquisitions indicate that the formation of large melt ponds began earlier on the MOSAiC floe than on neighbouring floes.

    Beschreibung: Across the Qinghai–Tibet Plateau (QTP) there is a narrow engineering corridor with widely distributed slopes called the Qinghai–Tibet Engineering Corridor (QTEC), where a variety of important infrastructures are concentrated. These facilities are transportation routes for people, materials, energy, etc. from inland China to the Tibet Autonomous Region. From Golmud to Lhasa, the engineering corridor covers 632 km of permafrost containing the densely developed Qinghai–Tibet Railway and Qinghai–Tibet Highway, as well as power and communication towers. Slope failure in permafrost regions, caused by permafrost degradation, ground ice melting, etc., affects the engineering construction and permafrost environments in the QTEC. We implement a variety of sensors to monitor the hydrological and thermal deformation between permafrost slopes and permafrost engineering projects in the corridor. In addition to soil temperature and moisture sensors, the global navigation satellite system (GNSS), terrestrial laser scanning (TLS), and unmanned aerial vehicles (UAVs) were adopted to monitor the spatial distribution and changes in thermal deformation. An integrated dataset of hydrological and thermal deformation in permafrost engineering and slopes in the QTEC from the 1950s to 2020, including meteorological and ground observations, TLS point cloud data, and RGB and thermal infrared (TIR) images, can be of great value for estimating the hydrological and thermal impact and stability between engineering and slopes under the influence of climate change and engineering disturbance.     Beschreibung: We present the first incorporation of the Common Representative Intermediates version 2.2 tropospheric chemistry mechanism, CRI v2.2, combined with stratospheric chemistry, into the global chemistry–climate United Kingdom Chemistry and Aerosols (UKCA) model to give the CRI-Strat 2 mechanism. A rigorous comparison of CRI-Strat 2 with the earlier version, CRI-Strat, is performed in UKCA in addition to an evaluation of three mechanisms, CRI-Strat 2, CRI-Strat and the standard UKCA chemical mechanism, StratTrop v1.0, against a wide array of surface and airborne chemical data. CRI-Strat 2 comprises a state-of-the-art isoprene scheme, optimized against the Master Chemical Mechanism v3.3.1, which includes isoprene peroxy radical isomerization, HOx recycling through the addition of photolabile hydroperoxy aldehydes (HPALDs), and isoprene epoxy diol (IEPOX) formation. CRI-Strat 2 also features updates to several rate constants for the inorganic chemistry, including the reactions of inorganic nitrogen and O(1D). The update to the isoprene chemistry in CRI-Strat 2 increases OH over the lowest 500 m in tropical forested regions by 30 %–50 % relative to CRI-Strat, leading to an improvement in model–observation comparisons for surface OH and isoprene relative to CRI-Strat and StratTrop. Enhanced oxidants also cause a 25 % reduction in isoprene burden and an increase in oxidation fluxes of isoprene and other biogenic volatile organic compounds (BVOCs) at low altitudes with likely impacts on subsequent aerosol formation, atmospheric lifetime, and climate. By contrast, updates to the rate constants of O(1D) with its main reactants relative to CRI-Strat reduces OH in much of the free troposphere, producing a 2 % increase in the methane lifetime, and increases the tropospheric ozone burden by 8 %, primarily from reduced loss via O(1D)+H2O. The changes to inorganic nitrogen reaction rate constants increase the NOx burden by 4 % and shift the distribution of nitrated species closer to that simulated by StratTrop. CRI-Strat 2 is suitable for multi-decadal model integrations and the improved representation of isoprene chemistry provides an opportunity to explore the consequences of HOx recycling in the United Kingdom Earth System Model (UKESM1). This new mechanism will enable a re-evaluation of the impact of BVOCs on the chemical composition of the atmosphere and further probe the feedback between the biosphere and the climate.

    Beschreibung: The use of mass flow simulations in volcanic hazard zonation and mapping is often limited by model complexity (i.e. uncertainty in correct values of model parameters), a lack of model uncertainty quantification, and limited approaches to incorporate this uncertainty into hazard maps. When quantified, mass flow simulation errors are typically evaluated on a pixel-pair basis, using the difference between simulated and observed (“actual”) map-cell values to evaluate the performance of a model. However, these comparisons conflate location and quantification errors, neglecting possible spatial autocorrelation of evaluated errors. As a result, model performance assessments typically yield moderate accuracy values. In this paper, similarly moderate accuracy values were found in a performance assessment of three depth-averaged numerical models using the 2012 debris avalanche from the Upper Te Maari crater, Tongariro Volcano, as a benchmark. To provide a fairer assessment of performance and evaluate spatial covariance of errors, we use a fuzzy set approach to indicate the proximity of similarly valued map cells. This “fuzzification” of simulated results yields improvements in targeted performance metrics relative to a length scale parameter at the expense of decreases in opposing metrics (e.g. fewer false negatives result in more false positives) and a reduction in resolution. The use of this approach to generate hazard zones incorporating the identified uncertainty and associated trade-offs is demonstrated and indicates a potential use for informed stakeholders by reducing the complexity of uncertainty estimation and supporting decision-making from simulated data.

    Beschreibung: Plant pathogens are responsible for the annual yield loss of crops worldwide and pose a significant threat to global food security. A necessary prelude to many plant disease epidemics is the short-range dispersal of spores, which may generate several disease foci within a field. New information is needed on the mechanisms of plant pathogen spread within and among susceptible plants. Here, we show that self-propelled jumping dew droplets, working synergistically with low wind flow, can propel spores of a fungal plant pathogen (wheat leaf rust) beyond the quiescent boundary layer and disperse them onto neighboring leaves downwind. An array of horizontal water-sensitive papers was used to mimic healthy wheat leaves and showed that up to 25 spores/h may be deposited on a single leaf downwind of the infected leaf during a single dew cycle. These findings reveal that a single dew cycle can disperse copious numbers of fungal spores to other wheat plants, even in the absence of rain splash or strong gusts of wind.

    Beschreibung: Replicability takes on special meaning when researching phenomena that are embedded in space and time, including phenomena distributed on the surface and near surface of the Earth. Two principles, spatial dependence and spatial heterogeneity, are generally characteristic of such phenomena. Various practices have evolved in dealing with spatial heterogeneity, including the use of place-based models. We review the rapidly emerging applications of artificial intelligence to phenomena distributed in space and time and speculate on how the principle of spatial heterogeneity might be addressed. We introduce a concept of weak replicability and discuss possible approaches to its measurement.

    Beschreibung: The inability of adult mammalian cardiomyocytes to proliferate underpins the development of heart failure following myocardial injury. Although the newborn mammalian heart can spontaneously regenerate for a short period of time after birth, this ability is lost within the first week after birth in mice, partly due to increased mitochondrial reactive oxygen species (ROS) production which results in oxidative DNA damage and activation of DNA damage response. This increase in ROS levels coincides with a postnatal switch from anaerobic glycolysis to fatty acid (FA) oxidation by cardiac mitochondria. However, to date, a direct link between mitochondrial substrate utilization and oxidative DNA damage is lacking. Here, we generated ROS-sensitive fluorescent sensors targeted to different subnuclear compartments (chromatin, heterochromatin, telomeres, and nuclear lamin) in neonatal rat ventricular cardiomyocytes, which allowed us to determine the spatial localization of ROS in cardiomyocyte nuclei upon manipulation of mitochondrial respiration. Our results demonstrate that FA utilization by the mitochondria induces a significant increase in ROS detection at the chromatin level compared to other nuclear compartments. These results indicate that mitochondrial metabolic perturbations directly alter the nuclear redox status and that the chromatin appears to be particularly sensitive to the prooxidant effect of FA utilization by the mitochondria.

    Beschreibung: Interactions of electronic and vibrational degrees of freedom are essential for understanding excited-states relaxation pathways of molecular systems at interfaces and surfaces. Here, we present the development of interface-specific two-dimensional electronic–vibrational sum frequency generation (2D-EVSFG) spectroscopy for electronic–vibrational couplings for excited states at interfaces and surfaces. We demonstrate this 2D-EVSFG technique by investigating photoexcited interface-active (E)-4-((4-(dihexylamino) phenyl)diazinyl)-1-methylpyridin-1- lum (AP3) molecules at the air–water interface as an example. Our 2D-EVSFG experiments show strong vibronic couplings of interfacial AP3 molecules upon photoexcitation and subsequent relaxation of a locally excited (LE) state. Time-dependent 2D-EVSFG experiments indicate that the relaxation of the LE state, S2, is strongly coupled with two high-frequency modes of 1,529.1 and 1,568.1 cm−1. Quantum chemistry calculations further verify that the strong vibronic couplings of the two vibrations promote the transition from the S2 state to the lower excited state S1. We believe that this development of 2D-EVSFG opens up an avenue of understanding excited-state dynamics related to interfaces and surfaces.

    Beschreibung: In this study, we analyse the climatology of ionosphere over Nepal based on GPS-derived vertical total electron content (VTEC) observed from four stations as defined in Table 1: KKN4 (27.80∘ N, 85.27∘ E), GRHI (27.95∘ N, 82.49∘ E), JMSM (28.80∘ N, 83.74∘ E) and DLPA (28.98∘ N, 82.81∘ E) during the years 2008 to 2018. The study illustrates the diurnal, monthly, annual, seasonal and solar cycle variations in VTEC during all times of solar cycle 24. The results clearly reveal the presence of equinoctial asymmetry in TEC, which is more pronounced in maximum phases of solar cycle in the year 2014 at KKN4 station, followed by descending, ascending and minimum phases. Diurnal variations in VTEC showed the short-lived day minimum which occurs between 05:00 to 06:00 LT (local time) at all the stations considered, with diurnal peaks between 12:00 and 15:00 LT. The maximum value of TEC is observed more often during the spring equinox than the autumn equinox, with a few asymmetries. Seasonal variation in TEC is observed to be a manifestation of variations in solar flux, particularly regarding the level of solar flux in consecutive solstices.

    Beschreibung: In this study, we use molecular genetic approaches to clarify the role of the Hedgehog (Hh) pathway in regulating the blood–brain/spinal cord barrier (BBB) in the adult mouse central nervous system (CNS). Our work confirms and extends prior studies to demonstrate that astrocytes are the predominant cell type in the adult CNS that transduce Hh signaling, revealed by the expression of Gli1, a target gene of the canonical pathway that is activated in cells receiving Hh, and other key pathway transduction components. Gli1+ (Hh-responsive) astrocytes are distributed in specific regions of the CNS parenchyma, including layers 4/5/6 of the neocortex, hypothalamus, thalamus, and spinal cord, among others. Notably, although BBB properties in endothelial cells are normally regulated by both paracellular and transcellular mechanisms, conditional inactivation of Hh signaling in astrocytes results in transient, region-specific BBB defects that affect transcytosis but not paracellular diffusion. These findings stand in contrast to prior studies that implicated astrocytes as a source of Sonic hedgehog that limited extravasation via both mechanisms [J. I. Alvarez et al., Science 334, 1727–1731 (2011)]. Furthermore, using three distinct Cre driver lines as well as pharmacological approaches to inactivate Hh-pathway transduction globally in CNS astrocytes, we find that these specific BBB defects are only detected in the rostral hypothalamus and spinal cord but not the cortex or other regions where Gli1+ astrocytes are found. Together, our data show that Gli1+ Hh-responsive astrocytes have regionally distinct molecular and functional properties and that the pathway is required to maintain BBB properties in specific regions of the adult mammalian CNS.

    Beschreibung: We report on applications of the ultraviolet-light-emitting-diode-based incoherent broadband cavity-enhanced absorption spectroscopy (UV-LED-IBBCEAS) technique for optical monitoring of HONO, NO2 and CH2O in a simulation chamber. Performance intercomparison of UV-LED-IBBCEAS with a wet chemistry-based NitroMAC sensor and a Fourier transform infrared (FTIR) spectrometer has been carried out on real-time simultaneous measurement of HONO, NO2 and CH2O concentrations during the reaction of NO2 with H2O vapour in CESAM (French acronym for Experimental Multiphasic Atmospheric Simulation Chamber). The 1σ (signal-to-noise ratio (SNR) = 1) detection limits of 112 pptv for NO2, 56 pptv for HONO and 41 ppbv for CH2O over 120 s were found for the UV-LED-IBBCEAS measurement. On the contrary to many set-ups where cavities are installed outside the simulation chamber, we describe here an original in situ permanent installation. The intercomparison results demonstrate that IBBCEAS is a very well suitable technique for in situ simultaneous measurements of multiple chemically reactive species with high sensitivity and high precision even if the absorption bands of these species are overlapped. It offers excellent capacity for non-invasive optical monitoring of chemical reactions without any perturbation. For the application to simulation chambers, it has the advantage to provide a spatially integrated measurement across the reactor and hence to avoid point-sampling-related artefacts.

    Beschreibung: Assimilation of weather radar measurements including radar reflectivity and radial wind data has been operational at the Deutscher Wetterdienst, with a diagonal observation error (OE) covariance matrix. For an implementation of a full OE covariance matrix, the statistics of the OE have to be a priori estimated, for which the Desroziers method has been often used. However, the resulted statistics consists of contributions from different error sources and are difficult to interpret. In this work, we use an approach that is based on samples for truncation error in radar observation space to approximate the representation error due to unresolved scales and processes (RE) and compare its statistics with the OE statistics estimated by the Desroziers method. It is found that the statistics of the RE help the understanding of several important features in the variances and correlation length scales of the OE for both reflectivity and radial wind data and the other error sources from the microphysical scheme, radar observation operator and the superobbing technique may also contribute, for instance, to differences among different elevations and observation types. The statistics presented here can serve as a guideline for selecting which observations are assimilated and for assignment of the OE covariance matrix that can be diagonal or full and correlated.

    Beschreibung: Geometry in materials is a key concept which can determine material behavior in ordering, frustration, and fragmentation. More specifically, the behavior of interacting degrees of freedom subject to arbitrary geometric constraints has the potential to be used for engineering materials with exotic phase behavior. While advances in lithography have allowed for an experimental exploration of geometry on ordering that has no precedent in nature, many of these methods are low throughput or the underlying dynamics remain difficult to observe directly. Here, we introduce an experimental system that enables the study of interacting many-body dynamics by exploiting the physics of multidroplet evaporation subject to two-dimensional spatial constraints. We find that a high-energy initial state of this system settles into frustrated, metastable states with relaxation on two timescales. We understand this process using a minimal dynamical model that simulates the overdamped dynamics of motile droplets by identifying the force exerted on a given droplet as being proportional to the two-dimensional vapor gradients established by its neighbors. Finally, we demonstrate the flexibility of this platform by presenting experimental realizations of droplet−lattice systems representing different spin degrees of freedom and lattice geometries. Our platform enables a rapid and low-cost means to directly visualize dynamics associated with complex many-body systems interacting via long-range interactions. More generally, this platform opens up the rich design space between geometry and interactions for rapid exploration with minimal resources.

    Beschreibung: In most bacteriophages, genome transport across bacterial envelopes is carried out by the tail machinery. In viruses of the Podoviridae family, in which the tail is not long enough to traverse the bacterial wall, it has been postulated that viral core proteins assembled inside the viral head are translocated and reassembled into a tube within the periplasm that extends the tail channel. Bacteriophage T7 infects Escherichia coli, and despite extensive studies, the precise mechanism by which its genome is translocated remains unknown. Using cryo-electron microscopy, we have resolved the structure of two different assemblies of the T7 DNA translocation complex composed of the core proteins gp15 and gp16. Gp15 alone forms a partially folded hexamer, which is further assembled upon interaction with gp16 into a tubular structure, forming a channel that could allow DNA passage. The structure of the gp15–gp16 complex also shows the location within gp16 of a canonical transglycosylase motif involved in the degradation of the bacterial peptidoglycan layer. This complex docks well in the tail extension structure found in the periplasm of T7-infected bacteria and matches the sixfold symmetry of the phage tail. In such cases, gp15 and gp16 that are initially present in the T7 capsid eightfold-symmetric core would change their oligomeric state upon reassembly in the periplasm. Altogether, these results allow us to propose a model for the assembly of the core translocation complex in the periplasm, which furthers understanding of the molecular mechanism involved in the release of T7 viral DNA into the bacterial cytoplasm.

    Beschreibung: Despite widespread yearly vaccination, influenza leads to significant morbidity and mortality across the globe. To make a more broadly protective influenza vaccine, it may be necessary to elicit antibodies that can activate effector functions in immune cells, such as antibody-dependent cellular cytotoxicity (ADCC). There is growing evidence supporting the necessity for ADCC in protection against influenza and herpes simplex virus (HSV), among other infectious diseases. An HSV-2 strain lacking the essential glycoprotein D (gD), was used to create ΔgD-2, which is a highly protective vaccine against lethal HSV-1 and HSV-2 infection in mice. It also elicits high levels of IgG2c antibodies that bind FcγRIV, a receptor that activates ADCC. To make an ADCC-eliciting influenza vaccine, we cloned the hemagglutinin (HA) gene from an H1N1 influenza A strain into the ΔgD-2 HSV vector. Vaccination with ΔgD-2::HAPR8 was protective against homologous influenza challenge and elicited an antibody response against HA that inhibits hemagglutination (HAI+), is predominantly IgG2c, strongly activates FcγRIV, and protects against influenza challenge following passive immunization of naïve mice. Prior exposure of mice to HSV-1, HSV-2, or a replication-defective HSV-2 vaccine (dl5-29) does not reduce protection against influenza by ΔgD-2::HAPR8. This vaccine also continues to elicit protection against both HSV-1 and HSV-2, including high levels of IgG2c antibodies against HSV-2. Mice lacking the interferon-α/β receptor and mice lacking the interferon-γ receptor were also protected against influenza challenge by ΔgD-2::HAPR8. Our results suggest that ΔgD-2 can be used as a vaccine vector against other pathogens, while also eliciting protective anti-HSV immunity.

    Beschreibung: Determining controls on the temperature sensitivity of heterotrophic soil respiration remains critical to incorporating soil–climate feedbacks into climate models. Most information on soil respiratory responses to temperature comes from laboratory incubations of isolated soils and typically subsamples of individual horizons. Inconsistencies between field and laboratory results may be explained by microbial priming supported by cross-horizon exchange of labile C or N. Such exchange is feasible in intact soil profiles but is absent when soils are isolated from surrounding depths. Here we assess the role of soil horizon connectivity, by which we mean the degree to which horizons remain layered and associated with each other as they are in situ, on microbial C and N substrate use and its relationship to the temperature sensitivity of respiration. We accomplished this by exploring changes in C : N, soil organic matter composition (via C : N, amino acid composition and concentration, and nuclear magnetic resonance spectroscopy), and the δ13C of respiratory CO2 during incubations of organic horizons collected across boreal forests in different climate regions where soil C and N compositions differ. The experiments consisted of two treatments: soil incubated (1) with each organic horizon separately and (2) as a whole organic profile, permitting cross-horizon exchange of substrates during the incubation. The soils were incubated at 5 and 15 ∘C for over 430 d. Enhanced microbial use of labile C-rich, but not N-rich, substrates were responsible for enhanced, whole-horizon respiratory responses to temperature relative to individual soil horizons. This impact of a labile C priming mechanism was most emergent in soils from the warmer region, consistent with these soils' lower C bioreactivity relative to soils from the colder region. Specifically, cross-horizon exchange within whole soil profiles prompted increases in mineralization of carbohydrates and more 13C-enriched substrates and increased soil respiratory responses to warming relative to soil horizons incubated in isolation. These findings highlight that soil horizon connectivity can impact microbial substrate use in ways that affect how soil effluxes of CO2 are controlled by temperature. The degree to which this mechanism exerts itself in other soils remains unknown, but these results highlight the importance of understanding mechanisms that operate in intact soil profiles – only rarely studied – in regulating a key soil–climate feedback.

    Beschreibung: Metabolism should drive demography by determining the rates of both biological work and resource demand. Long-standing “rules” for how metabolism should covary with demography permeate biology, from predicting the impacts of climate change to managing fisheries. Evidence for these rules is almost exclusively indirect and in the form of among-species comparisons, while direct evidence is exceptionally rare. In a manipulative field experiment on a sessile marine invertebrate, we created experimental populations that varied in population size (density) and metabolic rate, but not body size. We then tested key theoretical predictions regarding relationships between metabolism and demography by parameterizing population models with lifetime performance data from our field experiment. We found that populations with higher metabolisms had greater intrinsic rates of increase and lower carrying capacities, in qualitative accordance with classic theory. We also found important departures from theory—in particular, carrying capacity declined less steeply than predicted, such that energy use at equilibrium increased with metabolic rate, violating the long-standing axiom of energy equivalence. Theory holds that energy equivalence emerges because resource supply is assumed to be independent of metabolic rate. We find this assumption to be violated under real-world conditions, with potentially far-reaching consequences for the management of biological systems.

    Beschreibung: In the Paleozoic era, more than 400 Ma, a number of insect groups continued molting after forming functional wings. Today, however, flying insects stop molting after metamorphosis when they become fully winged. The only exception is the mayflies (Paleoptera, Ephemeroptera), which molt in the subimago, a flying stage between the nymph and the adult. However, the identity and homology of the subimago still is underexplored. Debate remains regarding whether this stage represents a modified nymph, an adult, or a pupa like that of butterflies. Another relevant question is why mayflies have the subimago stage despite the risk of molting fragile membranous wings. These questions have intrigued numerous authors, but nonetheless, clear answers have not yet been found. By combining morphological studies, hormonal treatments, and molecular analysis in the mayfly Cloeon dipterum, we found answers to these old questions. We observed that treatment with a juvenile hormone analog in the last nymphal instar stimulated the expression of the Kr-h1 gene and reduced that of E93, which suppress and trigger metamorphosis, respectively. The regulation of metamorphosis thus follows the MEKRE93 pathway, as in neopteran insects. Moreover, the treatment prevented the formation of the subimago. These findings suggest that the subimago must be considered an instar of the adult mayfly. We also observed that the forelegs dramatically grow between the last nymphal instar, the subimago, and the adult. This necessary growth spread over the last two stages could explain, at least in part, the adaptive sense of the subimago.

    Beschreibung: Plumes from the boreal spring biomass burning (BB) in northern peninsular Southeast Asia (nPSEA) are lifted into the subtropical jet stream and transported and deposited across nPSEA, South China, Taiwan and even the western North Pacific Ocean. This paper as part of the Seven SouthEast Asian Studies (7-SEAS) project effort attempts to improve the chemical weather prediction capability of the Weather Research and Forecasting coupled with the Community Multiscale for Air Quality (WRF–CMAQ) model over a vast region, from the mountainous near-source burning sites at nPSEA to its downwind region. Several sensitivity analyses of plume rise are compared in the paper, and it is discovered that the initial vertical allocation profile of BB plumes and the plume rise module (PLMRIM) are the main reasons causing the inaccuracies of the WRF–CMAQ simulations. The smoldering emission from the Western Regional Air Partnership (WRAP) empirical algorithm included has improved the accuracies of PM10, O3 and CO at the source. The best performance at the downwind sites is achieved with the inline PLMRIM, which accounts for the atmospheric stratification at the mountainous source region with the FINN burning emission dataset. Such a setup greatly improves not only the BB aerosol concentration prediction over near-source and receptor ground-based measurement sites but also the aerosol vertical distribution and column aerosol optical depth of the BB aerosol along the transport route. The BB aerosols from nPSEA are carried by the subtropical westerlies in the free troposphere to the western North Pacific, while BB aerosol has been found to interact with the local pollutants in the Taiwan region through three conditions: (a) overpassing western Taiwan and entering the central mountain area, (b) mixing down to western Taiwan, (c) transport of local pollutants upwards and mixing with a BB plume on higher ground. The second condition, which involves the prevailing high-pressure system from Asian cold surge, is able to impact most of the population in Taiwan.

    Beschreibung: T helper (Th)17 cells are considered to contribute to inflammatory mechanisms in diseases such as multiple sclerosis (MS). However, the discussion persists regarding their true role in patients. Here, we visualized central nervous system (CNS) inflammatory processes in models of MS live in vivo and in MS brains and discovered that CNS-infiltrating Th17 cells form prolonged stable contact with oligodendrocytes. Strikingly, compared to Th2 cells, direct contact with Th17 worsened experimental demyelination, caused damage to human oligodendrocyte processes, and increased cell death. Importantly, we found that in comparison to Th2 cells, both human and murine Th17 cells express higher levels of the integrin CD29, which is linked to glutamate release pathways. Of note, contact of human Th17 cells with oligodendrocytes triggered release of glutamate, which induced cell stress and changes in biosynthesis of cholesterol and lipids, as revealed by single-cell RNA-sequencing analysis. Finally, exposure to glutamate decreased myelination, whereas blockade of CD29 preserved oligodendrocyte processes from Th17-mediated injury. Our data provide evidence for the direct and deleterious attack of Th17 cells on the myelin compartment and show the potential for therapeutic opportunities in MS.

    Beschreibung: The German Bight located within the central North Sea is a hydro- and morphodynamically highly complex system of estuaries, barrier islands, and part of the world's largest coherent tidal flats, the Wadden Sea. To identify and understand challenges faced by coastal stakeholders, such as harbor operators or governmental agencies, to maintain waterways and employ numerical models for further analyses, it is imperative to have a consistent database for both bathymetry and surface sedimentology. Current commercial and public data products are insufficient in spatial and temporal resolution and coverage for recent analysis methods. Thus, this first part of a two-part publication series of the German joint project EasyGSH-DB describes annual bathymetric digital terrain models at a 10 m gridded resolution for the German North Sea coast and German Bight from 1996 to 2016 (Sievers et al., 2020a, https://doi.org/10.48437/02.2020.K2.7000.0001), as well as surface sedimentological models of discretized cumulative grain size distribution functions for 1996, 2006, and 2016 on 100 m grids (Sievers et al., 2020b, https://doi.org/10.48437/02.2020.K2.7000.0005). Furthermore, basic morphodynamic and sedimentological processing analyses, such as the estimation of, for example, bathymetric stability or surface maps of sedimentological parameters, are provided (Sievers et al., 2020a, b, see respective download links).

    Beschreibung: Natural vision is a dynamic and continuous process. Under natural conditions, visual object recognition typically involves continuous interactions between ocular motion and visual contrasts, resulting in dynamic retinal activations. In order to identify the dynamic variables that participate in this process and are relevant for image recognition, we used a set of images that are just above and below the human recognition threshold and whose recognition typically requires 〉2 s of viewing. We recorded eye movements of participants while attempting to recognize these images within trials lasting 3 s. We then assessed the activation dynamics of retinal ganglion cells resulting from ocular dynamics using a computational model. We found that while the saccadic rate was similar between recognized and unrecognized trials, the fixational ocular speed was significantly larger for unrecognized trials. Interestingly, however, retinal activation level was significantly lower during these unrecognized trials. We used retinal activation patterns and oculomotor parameters of each fixation to train a binary classifier, classifying recognized from unrecognized trials. Only retinal activation patterns could predict recognition, reaching 80% correct classifications on the fourth fixation (on average, ∼2.5 s from trial onset). We thus conclude that the information that is relevant for visual perception is embedded in the dynamic interactions between the oculomotor sequence and the image. Hence, our results suggest that ocular dynamics play an important role in recognition and that understanding the dynamics of retinal activation is crucial for understanding natural vision.

    Beschreibung: Two independent structures of the proton-pumping, respiratory cytochrome bo3 ubiquinol oxidase (cyt bo3) have been determined by cryogenic electron microscopy (cryo-EM) in styrene–maleic acid (SMA) copolymer nanodiscs and in membrane scaffold protein (MSP) nanodiscs to 2.55- and 2.19-Å resolution, respectively. The structures include the metal redox centers (heme b, heme o3, and CuB), the redox-active cross-linked histidine–tyrosine cofactor, and the internal water molecules in the proton-conducting D channel. Each structure also contains one equivalent of ubiquinone-8 (UQ8) in the substrate binding site as well as several phospholipid molecules. The isoprene side chain of UQ8 is clamped within a hydrophobic groove in subunit I by transmembrane helix TM0, which is only present in quinol oxidases and not in the closely related cytochrome c oxidases. Both structures show carbonyl O1 of the UQ8 headgroup hydrogen bonded to D75I and R71I. In both structures, residue H98I occupies two conformations. In conformation 1, H98I forms a hydrogen bond with carbonyl O4 of the UQ8 headgroup, but in conformation 2, the imidazole side chain of H98I has flipped to form a hydrogen bond with E14I at the N-terminal end of TM0. We propose that H98I dynamics facilitate proton transfer from ubiquinol to the periplasmic aqueous phase during oxidation of the substrate. Computational studies show that TM0 creates a channel, allowing access of water to the ubiquinol headgroup and to H98I.

    Beschreibung: Canonical Wnt signaling plays critical roles in development and tissue renewal by regulating β-catenin target genes. Recent evidence showed that β-catenin–independent Wnt signaling is also required for faithful execution of mitosis. However, the targets and specific functions of mitotic Wnt signaling still remain uncharacterized. Using phosphoproteomics, we identified that Wnt signaling regulates the microtubule depolymerase KIF2A during mitosis. We found that Dishevelled recruits KIF2A via its N-terminal and motor domains, which is further promoted upon LRP6 signalosome formation during cell division. We show that Wnt signaling modulates KIF2A interaction with PLK1, which is critical for KIF2A localization at the spindle. Accordingly, inhibition of basal Wnt signaling leads to chromosome misalignment in somatic cells and pluripotent stem cells. We propose that Wnt signaling monitors KIF2A activity at the spindle poles during mitosis to ensure timely chromosome alignment. Our findings highlight a function of Wnt signaling during cell division, which could have important implications for genome maintenance, notably in stem cells.

    Beschreibung: The Phanerozoic tectonothermal evolution of the SW slope of the East European Platform (EEP) in Poland is reconstructed by means of thermal maturity, low-temperature thermochronometry, and thermal modelling. We provide a set of new thermochronometric data and integrate stratigraphic and thermal maturity information to constrain the burial and thermal history of sediments. Apatite fission track (AFT) analysis and zircon (U-Th)/He (ZHe) thermochronology have been carried out on samples of sandstones, bentonites, diabase, and crystalline basement rocks collected from 17 boreholes located in central and NE Poland. They penetrated sedimentary cover of the EEP subdivided from the north to south into the Baltic, Podlasie, and Lublin basins. The average ZHe ages from Proterozoic basement rocks as well as Ordovician to Silurian bentonites and Cambrian to lower Carboniferous sandstones range from 848 ± 81 to 255 ± 22 Ma with a single early Permian age of 288 Ma, corresponding to cooling after a thermal event. The remaining ZHe ages represent partial reset or source ages. The AFT ages of samples are dispersed in the range of 235.8 ± 17.3 Ma (Middle Triassic) to 42.1 ± 11.1 Ma (Paleogene) providing a record of Mesozoic and Cenozoic cooling. The highest frequency of the AFT ages is in the Jurassic and Early Cretaceous prior to Alpine basin inversion. Thermal maturity results are consistent with the SW-ward increase of the Paleozoic and Mesozoic sediments thickness. An important break in a thermal maturity profile exists across the base Permian–Mesozoic unconformity. Thermal modelling showed that significant heating of Ediacaran to Carboniferous sedimentary successions occurred before the Permian with maximum paleotemperatures in the earliest and latest Carboniferous for Baltic–Podlasie and Lublin basins, respectively. The results obtained suggest an important role of early Carboniferous uplift and exhumation at the SW margin of the EEP. The SW slope of the latter was afterward overridden in the Lublin Basin by the Variscan orogenic wedge. Its tectonic loading interrupted Carboniferous uplift and caused resumption of sedimentation in the late Viséan. Consequently, a thermal history of the Lublin Basin is different from that in the Podlasie and Baltic basins but similar to other sections of the Variscan foreland, characterized by maximum burial at the end of Carboniferous. The Mesozoic thermal history was characterized by gradual cooling from peak temperatures at the transition from Triassic to Jurassic due to decreasing heat flow. Burial caused maximum paleotemperatures in the SW part of the study area, where the EEP was covered by an extensive sedimentary pile. However, further NE, due to low temperatures caused by shallow burial, the impact of fluids can be detected by vitrinite reflectance, illite/smectite, and thermochronological data. Our new results emphasize the importance of using multiple low-temperature thermochronometers and thermal modelling in connection with thermal maturity analysis to elucidate the near-surface evolution of platform margins.

    Beschreibung: Snow stands out from materials at the Earth’s surface owing to its unique optical properties. Snow optical properties are sensitive to the snow microstructure, triggering potent climate feedbacks. The impacts of snow microstructure on its optical properties such as reflectance are, to date, only partially understood. However, precise modelling of snow reflectance, particularly bidirectional reflectance, are required in many problems, e.g. to correctly process satellite data over snow-covered areas. This study presents a dataset that combines bidirectional reflectance measurements over 500–2500 nm and the X-ray tomography of the snow microstructure for three snow samples of two different morphological types. The dataset is used to evaluate the stereological approach from Malinka (2014) that relates snow optical properties to the chord length distribution in the snow microstructure. The mean chord length and specific surface area (SSA) retrieved with this approach from the albedo spectrum and those measured by the X-ray tomography are in excellent agreement. The analysis of the 3D images has shown that the random chords of the ice phase obey the gamma distribution with the shape parameter m taking the value approximately equal to or a little greater than 2. For weak and intermediate absorption (high and medium albedo), the simulated bidirectional reflectances reproduce the measured ones accurately but tend to slightly overestimate the anisotropy of the radiation. For such absorptions the use of the exponential law for the ice chord length distribution instead of the one measured with the X-ray tomography does not affect the simulated reflectance. In contrast, under high absorption (albedo of a few percent), snow microstructure and especially facet orientation at the surface play a significant role in the reflectance, particularly at oblique viewing and incidence.

    Beschreibung: Attribution in its general definition aims to quantify drivers of change in a system. According to IPCC Working Group II (WGII) a change in a natural, human or managed system is attributed to climate change by quantifying the difference between the observed state of the system and a counterfactual baseline that characterizes the system's behavior in the absence of climate change, where “climate change refers to any long-term trend in climate, irrespective of its cause” (IPCC, 2014). Impact attribution following this definition remains a challenge because the counterfactual baseline, which characterizes the system behavior in the hypothetical absence of climate change, cannot be observed. Process-based and empirical impact models can fill this gap as they allow us to simulate the counterfactual climate impact baseline. In those simulations, the models are forced by observed direct (human) drivers such as land use changes, changes in water or agricultural management but a counterfactual climate without long-term changes. We here present ATTRICI (ATTRIbuting Climate Impacts), an approach to construct the required counterfactual stationary climate data from observational (factual) climate data. Our method identifies the long-term shifts in the considered daily climate variables that are correlated to global mean temperature change assuming a smooth annual cycle of the associated scaling coefficients for each day of the year. The produced counterfactual climate datasets are used as forcing data within the impact attribution setup of the Inter-Sectoral Impact Model Intercomparison Project (ISIMIP3a). Our method preserves the internal variability of the observed data in the sense that factual and counterfactual data for a given day have the same rank in their respective statistical distributions. The associated impact model simulations allow for quantifying the contribution of climate change to observed long-term changes in impact indicators and for quantifying the contribution of the observed trend in climate to the magnitude of individual impact events. Attribution of climate impacts to anthropogenic forcing would need an additional step separating anthropogenic climate forcing from other sources of climate trends, which is not covered by our method.

    Beschreibung: Plants employ sensor–helper pairs of NLR immune receptors to recognize pathogen effectors and activate immune responses. Yet, the subcellular localization of NLRs pre- and postactivation during pathogen infection remains poorly understood. Here, we show that NRC4, from the “NRC” solanaceous helper NLR family, undergoes dynamic changes in subcellular localization by shuttling to and from the plant–pathogen haustorium interface established during infection by the Irish potato famine pathogen Phytophthora infestans. Specifically, prior to activation, NRC4 accumulates at the extrahaustorial membrane (EHM), presumably to mediate response to perihaustorial effectors that are recognized by NRC4-dependent sensor NLRs. However, not all NLRs accumulate at the EHM, as the closely related helper NRC2 and the distantly related ZAR1 did not accumulate at the EHM. NRC4 required an intact N-terminal coiled-coil domain to accumulate at the EHM, whereas the functionally conserved MADA motif implicated in cell death activation and membrane insertion was dispensable for this process. Strikingly, a constitutively autoactive NRC4 mutant did not accumulate at the EHM and showed punctate distribution that mainly associated with the plasma membrane, suggesting that postactivation, NRC4 may undergo a conformation switch to form clusters that do not preferentially associate with the EHM. When NRC4 is activated by a sensor NLR during infection, however, NRC4 forms puncta mainly at the EHM and, to a lesser extent, at the plasma membrane. We conclude that following activation at the EHM, NRC4 may spread to other cellular membranes from its primary site of activation to trigger immune responses.

    Beschreibung: Sterile α motif domain-containing protein 9-like (SAMD9L) is encoded by a hallmark interferon-induced gene with a role in controlling virus replication that is not well understood. Here, we analyze SAMD9L function from the perspective of human mutations causing neonatal-onset severe autoinflammatory disease. Whole-genome sequencing of two children with leukocytoclastic panniculitis, basal ganglia calcifications, raised blood inflammatory markers, neutrophilia, anemia, thrombocytopaenia, and almost no B cells revealed heterozygous de novo SAMD9L mutations, p.Asn885Thrfs*6 and p.Lys878Serfs*13. These frameshift mutations truncate the SAMD9L protein within a domain a region of homology to the nucleotide-binding and oligomerization domain (NOD) of APAF1, ∼80 amino acids C-terminal to the Walker B motif. Single-cell analysis of human cells expressing green fluorescent protein (GFP)-SAMD9L fusion proteins revealed that enforced expression of wild-type SAMD9L repressed translation of red fluorescent protein messenger RNA and globally repressed endogenous protein translation, cell autonomously and in proportion to the level of GFP-SAMD9L in each cell. The children’s truncating mutations dramatically exaggerated translational repression even at low levels of GFP-SAMD9L per cell, as did a missense Arg986Cys mutation reported recurrently as causing ataxia pancytopenia syndrome. Autoinflammatory disease associated with SAMD9L truncating mutations appears to result from an interferon-induced translational repressor whose activity goes unchecked by the loss of C-terminal domains that may normally sense virus infection.

    Beschreibung: Biocides used in film protection products leaching from facades are known to be a potential threat to the environment. This study identifies individual sources and entry pathways in a small-scale urban area. We investigate emissions of commonly used biocides (terbutryn, diuron, and octylisothiazolinone – OIT) and some of their transformation products (TPs; diuron-desmethyl, terbumeton, terbuthylazine-2-hydroxy, and terbutryn-desethyl) from a 2 ha residential area 13 years after construction has ended. Sampling utilizes existing urban water infrastructure representative for decentralized storm water management in central and northern Europe and applies a two-step approach to (a) determine the occurrence of biocides above water quality limits (i.e., predicted no-effect concentration, PNEC) and (b) identify source areas and characterize entry pathways into surface and groundwater. Monitoring focuses on the analysis of selected biocides and TPs by liquid chromatography–mass spectrometry/mass spectrometry (LC-MS/MS) in water samples taken from facades, rainwater pipes, drainage, and storm water infiltration systems. In standing water in a swale, we found high concentrations of diuron (174 ng L−1) and terbutryn (40 ng L−1) above PNEC for surface water. We confirmed expected sources, i.e., facades. Sampling of rain downpipes from flat roofs identified additional sources of all biocides and two TPs of terbutryn and one TP of diuron. Diuron and terbutryn were found in three drainage pipes representing different entry pathways of biocides. In one drainage pipe collecting road runoff, only diuron-desmethyl and terbutryn-desethyl were detected. In two other drainage pipes collecting infiltrated water through soil, terbuthylazine-2-hydroxy was additionally detected. One of the pipes collecting infiltrated water through soil concentration showed the highest concentrations of terbutryn and two of its TPs (terbutryn-desethyl and terbuthylazine-2-hydroxy). This suggests a high leaching potential of terbutryn. The applied two-step approach determined sources and pathways of biocide and their TPs. This study contributes to expanding knowledge on their entry and distribution and, thus, eventually towards reducing emissions.

    Beschreibung: Natural killer T (NKT) cells detect lipids presented by CD1d. Most studies focus on type I NKT cells that express semi-invariant αβ T cell receptors (TCR) and recognize α-galactosylceramides. However, CD1d also presents structurally distinct lipids to NKT cells expressing diverse TCRs (type II NKT cells), but our knowledge of the antigens for type II NKT cells is limited. An early study identified a nonlipidic NKT cell agonist, phenyl pentamethyldihydrobenzofuransulfonate (PPBF), which is notable for its similarity to common sulfa drugs, but its mechanism of NKT cell activation remained unknown. Here, we demonstrate that a range of pentamethylbenzofuransulfonates (PBFs), including PPBF, activate polyclonal type II NKT cells from human donors. Whereas these sulfa drug–like molecules might have acted pharmacologically on cells, here we demonstrate direct contact between TCRs and PBF-treated CD1d complexes. Further, PBF-treated CD1d tetramers identified type II NKT cell populations expressing αβTCRs and γδTCRs, including those with variable and joining region gene usage (TRAV12-1–TRAJ6) that was conserved across donors. By trapping a CD1d–type II NKT TCR complex for direct mass-spectrometric analysis, we detected molecules that allow the binding of CD1d to TCRs, finding that both selected PBF family members and short-chain sphingomyelin lipids are present in these complexes. Furthermore, the combination of PPBF and short-chain sphingomyelin enhances CD1d tetramer staining of PPBF-reactive T cell lines over either molecule alone. This study demonstrates that nonlipidic small molecules, which resemble sulfa drugs implicated in systemic hypersensitivity and drug allergy reactions, are targeted by a polyclonal population of type II NKT cells in a CD1d-restricted manner.

    Beschreibung: Accurate prediction of fundamental band gaps of crystalline solid-state systems entirely within density functional theory is a long-standing challenge. Here, we present a simple and inexpensive method that achieves this by means of nonempirical optimal tuning of the parameters of a screened range-separated hybrid functional. The tuning involves the enforcement of an ansatz that generalizes the ionization potential theorem to the removal of an electron from an occupied state described by a localized Wannier function in a modestly sized supercell calculation. The method is benchmarked against experiment for a set of systems ranging from narrow band-gap semiconductors to large band-gap insulators, spanning a range of fundamental band gaps from 0.2 to 14.2 electronvolts (eV), and is found to yield quantitative accuracy across the board, with a mean absolute error of ∼0.1 eV and a maximal error of ∼0.2 eV.

    Beschreibung: Regional anomalies of steric sea level are either due to redistribution of heat and freshwater anomalies or due to ocean–atmosphere buoyancy fluxes. Interannual to decadal variability in sea level across the tropical Pacific is mainly due to steric variations driven by wind stress anomalies. The importance of air–sea buoyancy fluxes is less clear. We use a global, eddy-permitting ocean model and a series of sensitivity experiments with quasi-climatological momentum and buoyancy fluxes to identify the contribution of buoyancy fluxes for interannual to decadal sea level variability in the tropical Pacific. We find their contribution on interannual timescales to be strongest in the central tropical Pacific at around a 10∘ latitude in both hemispheres and also relevant in the very east of the tropical domain. Buoyancy-flux-forced anomalies are correlated with variations driven by wind stress changes, but their effect on the prevailing anomalies and the importance of heat and freshwater fluxes vary locally. In the eastern tropical basin, interannual sea level variability is amplified by anomalous heat fluxes, while the importance of freshwater fluxes is small, and neither has any impact on decadal timescales. In the western tropical Pacific, the variability on interannual and decadal timescales is dampened by both heat and freshwater fluxes. The mechanism involves westward-propagating Rossby waves that are triggered during El Niño–Southern Oscillation (ENSO) events by anomalous buoyancy fluxes in the central tropical Pacific and counteract the prevailing sea level anomalies once they reach the western part of the basin.

    Beschreibung: The Nioghalvfjerdsfjorden glacier (also known as the 79∘ North Glacier) drains approximately 8 % of the Greenland Ice Sheet. Supraglacial lakes (SGLs), or surface melt ponds, are a persistent summertime feature and are thought to drain rapidly to the base of the glacier and influence seasonal ice velocity. However, seasonal development and spatial distribution of SGLs in the north-east of Greenland are poorly understood, leaving a substantial error in the estimate of meltwater and its impacts on ice velocity. Using results from an automated detection of melt ponds, atmospheric and surface mass balance modelling, and reanalysis products, we investigate the role of specific climatic conditions in melt onset, extent, and duration from 2016 to 2019. The summers of 2016 and 2019 were characterised by above-average air temperatures, particularly in June, as well as a number of rainfall events, which led to extensive melt ponds to elevations up to 1600 m. Conversely, 2018 was particularly cold, with a large accumulated snowpack, which limited the development of lakes to altitudes less than 800 m. There is evidence of inland expansion and increases in the total area of lakes compared to the early 2000s, as projected by future global warming scenarios.

    Beschreibung: Coincident wide-angle and multi-channel seismic data acquired within the scope of the PAMELA Moz3-5 project allow us to reconsider the formation mechanism of East African margins offshore of southern Mozambique. This study specifically focuses on the sedimentary and deep-crustal architecture of the Limpopo margin (LM) that fringes the eastern edge of the Mozambique’s Coastal Plain (MCP) and its offshore southern prolongation the North Natal Valley (NNV). It relies primarily on the MZ3 profile that runs obliquely from the northeastern NNV towards the Mozambique basin (MB) with additional inputs from a tectonostratigraphy analysis of industrial onshore–offshore seismic lines and nearby or crossing velocity models from companion studies. Over its entire N–S extension the LM appears segmented into (1) a western domain that shows the progressive eastward crustal thinning and termination of the MCP/NNV continental crust and its overlying pre-Neocomian volcano-sedimentary basement and (2) a central corridor of anomalous crust bounded to the east by the Mozambique fracture zone (MFZ) and the oceanic crust of the MB. A prominent basement high marks the boundary between these two domains. Its development was most probably controlled by a steep and deeply rooted fault, i.e., the Limpopo fault. We infer that strike-slip or slightly transtensional rifting occurred along the LM and was accommodated along this Limpopo fault. At depth we propose that ductile shearing was responsible for the thinning of the continental crust and an oceanward flow of lower crustal material. This process was accompanied by intense magmatism that extruded to form the volcanic basement and gave the corridor its peculiar structure and mixed nature. The whole region remained at a relative high level during the rifting period and a shallow marine environment dominated the pre-Neocomian period during the early phase of continent–ocean interaction. It is only some time after break-up in the MB and the initiation of the MFZ that decoupling occurred between the MCP/NNV and the corridor, allowing for the latter to subside and become covered by deep marine sediments. A scenario for the early evolution and formation of the LM is proposed taking into account both recent kinematic and geological constraints. It implies that no or little change in extensional direction occurred between the intra-continental rifting and subsequent phase of continent–ocean interaction.

    Beschreibung: Models are an important tool to predict Earth system dynamics. An accurate prediction of future states of ecosystems depends on not only model structures but also parameterizations. Model parameters can be constrained by data assimilation. However, applications of data assimilation to ecology are restricted by highly technical requirements such as model-dependent coding. To alleviate this technical burden, we developed a model-independent data assimilation (MIDA) module. MIDA works in three steps including data preparation, execution of data assimilation, and visualization. The first step prepares prior ranges of parameter values, a defined number of iterations, and directory paths to access files of observations and models. The execution step calibrates parameter values to best fit the observations and estimates the parameter posterior distributions. The final step automatically visualizes the calibration performance and posterior distributions. MIDA is model independent, and modelers can use MIDA for an accurate and efficient data assimilation in a simple and interactive way without modification of their original models. We applied MIDA to four types of ecological models: the data assimilation linked ecosystem carbon (DALEC) model, a surrogate-based energy exascale earth system model: the land component (ELM), nine phenological models and a stand-alone biome ecological strategy simulator (BiomeE). The applications indicate that MIDA can effectively solve data assimilation problems for different ecological models. Additionally, the easy implementation and model-independent feature of MIDA breaks the technical barrier of applications of data–model fusion in ecology. MIDA facilitates the assimilation of various observations into models for uncertainty reduction in ecological modeling and forecasting.

    Beschreibung: Contact tracing has for decades been a cornerstone of the public health approach to epidemics, including Ebola, severe acute respiratory syndrome, and now COVID-19. It has not yet been possible, however, to causally assess the method’s effectiveness using a randomized controlled trial of the sort familiar throughout other areas of science. This study provides evidence that comes close to that ideal. It exploits a large-scale natural experiment that occurred by accident in England in late September 2020. Because of a coding error involving spreadsheet data used by the health authorities, a total of 15,841 COVID-19 cases (around 20% of all cases) failed to have timely contact tracing. By chance, some areas of England were much more severely affected than others. This study finds that the random breakdown of contact tracing led to more illness and death. Conservative causal estimates imply that, relative to cases that were initially missed by the contact tracing system, cases subject to proper contact tracing were associated with a reduction in subsequent new infections of 63% and a reduction insubsequent COVID-19–related deaths of 66% across the 6 wk following the data glitch.

    Beschreibung: The northern Antarctic Peninsula (NAP) is a highly dynamic transitional zone between the subpolar-polar and oceanic-coastal environments, and it is located in an area affected by intense climate change, including intensification and spatial shifts of the westerlies as well as atmospheric and oceanic warming. In the NAP area, the water masses originate mainly from the Bellingshausen and Weddell seas, which create a marked regional dichotomy thermohaline characteristic. Although the NAP area has relatively easy access when compared to other Southern Ocean environments, our understanding of the water masses' distribution and the dynamical processes affecting the variability of the region is still limited. That limitation is closely linked to the sparse data coverage, as is commonly the case in most Southern Ocean environments. This work provides a novel seasonal three-dimensional high-resolution hydrographic gridded data set for the NAP (version 1), namely the NAPv1.0. Hydrographic measurements from 1990 to 2019 comprising data collected by conductivity, temperature, depth (CTD) casts; sensors from the Marine Mammals Exploring the Oceans Pole to Pole (MEOP) consortium; and Argo floats have been optimally interpolated to produce maps of in situ temperature, practical salinity, and dissolved oxygen at ∼ 10 km spatial resolution and 90 depth levels. The water masses and oceanographic features in this regional gridded product are more accurate than other climatologies and state estimate products currently available. The data sets are available in netCDF format at https://doi.org/10.5281/zenodo.4420006 (Dotto et al., 2021). The novel and comprehensive data sets presented here for the NAPv1.0 product are a valuable tool to be used in studies addressing climatological changes in the unique NAP region since they provide accurate initial conditions for ocean models and improve the end of the 20th- and early 21st-century ocean mean-state representation for that area.

    Beschreibung: Time shapes every single human–environment relationship and is inherent in 21st-century global challenges such as climate change and the urgent move towards global sustainability. Nonetheless, the concept of time is still insufficiently addressed in climate justice debates. This paper aims to help fill this gap by presenting empirical results about experiences of climate change in farming communities in Tamil Nadu, South India, and fishing communities on the Lofoten Islands, Norway. With the help of the five dimensions of affectedness, rhythms and rituals, slow motion, care, and health and well-being, it exemplifies how time matters to issues of climate injustices faced by the communities. The paper promotes a qualitative understanding of time and climate change. Thereby, it may stimulate greater relatability to climate change, as well as discussion likely to lead to conceptual advances.

    Beschreibung: A method is proposed for determining the height of the turbulent mixing layer on the basis of the vertical profiles of the dissipation rate of turbulent energy, which is estimated from lidar measurements of the radial wind velocity using conical scanning by a probe beam around the vertical axis. The accuracy of the proposed method is discussed in detail. It is shown that for the estimation of the mixing layer height (MLH) with the acceptable relative error not exceeding 20 %, the signal-to-noise ratio should be no less than −16 dB, when the relative error of lidar estimation of the dissipation rate does not exceed 30 %. The method was tested in a 6 d experiment in which the wind velocity turbulence was estimated in smog conditions due to forest fires in Siberia in summer 2019. The results of the experiment reveal that the relative error of determination of the MLH time series obtained by this method does not exceed 10 % in the period of turbulence development. The estimates of the turbulent mixing layer height by the proposed method are in a qualitative agreement with the MLH estimated from the distributions of the Richardson number in height and time obtained during the comparison experiment in spring 2020.

    Beschreibung: In the 2019/2020 austral summer, the surface melt duration and extent on the northern George VI Ice Shelf (GVIIS) was exceptional compared to the 31 previous summers of distinctly lower melt. This finding is based on analysis of near-continuous 41-year satellite microwave radiometer and scatterometer data, which are sensitive to meltwater on the ice shelf surface and in the near-surface snow. Using optical satellite imagery from Landsat 8 (2013 to 2020) and Sentinel-2 (2017 to 2020), record volumes of surface meltwater ponding were also observed on the northern GVIIS in 2019/2020, with 23 % of the surface area covered by 0.62 km3 of ponded meltwater on 19 January. These exceptional melt and surface ponding conditions in 2019/2020 were driven by sustained air temperatures ≥0 ∘C for anomalously long periods (55 to 90 h) from late November onwards, which limited meltwater refreezing. The sustained warm periods were likely driven by warm, low-speed (≤7.5 m s−1) northwesterly and northeasterly winds and not by foehn wind conditions, which were only present for 9 h total in the 2019/2020 melt season. Increased surface ponding on ice shelves may threaten their stability through increased potential for hydrofracture initiation; a risk that may increase due to firn air content depletion in response to near-surface melting.

    Beschreibung: In the Pliocene Model Intercomparison Project Phase 2 (PlioMIP2), coupled climate models have been used to simulate an interglacial climate during the mid-Piacenzian warm period (mPWP; 3.264 to 3.025 Ma). Here, we compare the Atlantic Meridional Overturning Circulation (AMOC), poleward ocean heat transport and sea surface warming in the Atlantic simulated with these models. In PlioMIP2, all models simulate an intensified mid-Pliocene AMOC. However, there is no consistent response in the simulated Atlantic ocean heat transport nor in the depth of the Atlantic overturning cell. The models show a large spread in the simulated AMOC maximum, the Atlantic ocean heat transport and the surface warming in the North Atlantic. Although a few models simulate a surface warming of ∼ 8–12 ∘C in the North Atlantic, similar to the reconstruction from Pliocene Research, Interpretation and Synoptic Mapping (PRISM) version 4, most models appear to underestimate this warming. The large model spread and model–data discrepancies in the PlioMIP2 ensemble do not support the hypothesis that an intensification of the AMOC, together with an increase in northward ocean heat transport, is the dominant mechanism for the mid-Pliocene warm climate over the North Atlantic.

    Beschreibung: Many competing criteria are under consideration for prioritizing COVID-19 vaccination. Two criteria based on age are demographic: lives saved and years of future life saved. Vaccinating the very old against COVID-19 saves the most lives, but, since older age is accompanied by falling life expectancy, it is widely supposed that these two goals are in conflict. We show this to be mistaken. The age patterns of COVID-19 mortality are such that vaccinating the oldest first saves the most lives and, surprisingly, also maximizes years of remaining life expectancy. We demonstrate this relationship empirically in the United States, Germany, and South Korea and with mathematical analysis of life tables. Our age-risk results, under usual conditions, also apply to health risks.

    Beschreibung: Electrolyte is very critical to the performance of the high-voltage lithium (Li) metal battery (LMB), which is one of the most attractive candidates for the next-generation high-density energy-storage systems. Electrolyte formulation and structure determine the physical properties of the electrolytes and their interfacial chemistries on the electrode surfaces. Localized high-concentration electrolytes (LHCEs) outperform state-of-the-art carbonate electrolytes in many aspects in LMBs due to their unique solvation structures. Types of fluorinated cosolvents used in LHCEs are investigated here in searching for the most suitable diluent for high-concentration electrolytes (HCEs). Nonsolvating solvents (including fluorinated ethers, fluorinated borate, and fluorinated orthoformate) added in HCEs enable the formation of LHCEs with high-concentration solvation structures. However, low-solvating fluorinated carbonate will coordinate with Li+ ions and form a second solvation shell or a pseudo-LHCE which diminishes the benefits of LHCE. In addition, it is evident that the diluent has significant influence on the electrode/electrolyte interphases (EEIs) beyond retaining the high-concentration solvation structures. Diluent molecules surrounding the high-concentration clusters could accelerate or decelerate the anion decomposition through coparticipation of diluent decomposition in the EEI formation. The varied interphase features lead to significantly different battery performance. This study points out the importance of diluents and their synergetic effects with the conductive salt and the solvating solvent in designing LHCEs. These systematic comparisons and fundamental insights into LHCEs using different types of fluorinated solvents can guide further development of advanced electrolytes for high-voltage LMBs.

    Beschreibung: A ground-based network of more than 1200 Global Navigation Satellite System (GNSS) Continuously Operating Reference Stations (CORS) was analysed using GIPSY-OASIS II software package for the documentation of time and space variations of water vapor in atmosphere during the North Atlantic Waveguide and Downstream impact EXperiment (NAWDEX) during fall 2016. The network extends throughout the North Atlantic, from the Caribbeans to Morocco through Greenland. This paper presents the methodology used for GNSS data processing, screening, and conversion of Zenith Tropospheric Delay (ZTD) estimates to Integrated Water Vapor content (IWV) using surface parameters from reanalysis. The retrieved IWV are used to evaluate the European Centre for Medium-Range Weather Forecasts (ECMWF) reanalyses ERAI and ERA5. ERA5 shows an overall improvement over ERAI in representing the spatial and temporal variability of IWV over the study area. The mean bias is decreased from 0.31±0.63 to 0.19±0.56 kg m−2 (mean ±1σ over all stations) and the standard deviation reduced from 2.17±0.67 to 1.64±0.53 kg m−2 combined with a slight improvement in correlation coefficient from 0.95 to 0.97. At regional scale, both reanalyses show a general wet bias at mid and northern latitudes but a dry bias in the Caribbeans. We hypothesize this results from the different nature of data being assimilated over the tropical oceans. This GNSS IWV data set is intended to be used for a better description of the high impact weather events that occurred during the NAWDEX experiment.

    Beschreibung: The deep chlorophyll maximum (DCM) is a well-known feature of the global ocean. However, its description and the study of its formation are a challenge, especially in the peculiar environment that is the Black Sea. The retrieval of chlorophyll a (chl a) from fluorescence (Fluo) profiles recorded by Biogeochemical Argo (BGC-Argo) floats is not trivial in the Black Sea, due to the very high content of coloured dissolved organic matter (CDOM) which contributes to the fluorescence signal and produces an apparent increase in the chl a concentration with depth. Here, we revised Fluo correction protocols for the Black Sea context using co-located in situ high-performance liquid chromatography (HPLC) and BGC-Argo measurements. The processed set of chl a data (2014–2019) is then used to provide a systematic description of the seasonal DCM dynamics in the Black Sea and to explore different hypotheses concerning the mechanisms underlying its development. Our results show that the corrections applied to the chl a profiles are consistent with HPLC data. In the Black Sea, the DCM begins to form in March, throughout the basin, at a density level set by the previous winter mixed layer. During a first phase (April–May), the DCM remains attached to this particular layer. The spatial homogeneity of this feature suggests a hysteresis mechanism, i.e. that the DCM structure locally influences environmental conditions rather than adapting instantaneously to external factors. In a second phase (July–September), the DCM migrates upward, where there is higher irradiance, which suggests the interplay of biotic factors. Overall, the DCM concentrates around 45 % to 65 % of the total chlorophyll content within a 10 m layer centred around a depth of 30 to 40 m, which stresses the importance of considering DCM dynamics when evaluating phytoplankton productivity at basin scale.

    Beschreibung: A normal contact stiffness model considering 3D topography and elastic–plastic contact of rough surfaces is presented in this paper. The asperities are generated from the measured surfaces using the watershed segmentation and a modified nine-point rectangle. The topography parameters, including the asperity locations, heights, and radii of the summit, are obtained. Asperity shoulder–shoulder contact is considered. The relationship of the contact parameters, such as the contact force, the deformation, and the mean separation of two surfaces, is modelled in the three different contact regimes, namely elastic, elastic–plastic and fully plastic. The asperity contact state is determined, and if the contact occurs, the stiffness of the single asperity pair is calculated and summed as the total normal stiffness of two contact surfaces. The developed model is validated using experimental tests conducted on two types of specimens and is compared with published theoretical models.

    Beschreibung: The contraction of heart cells is controlled by the intermolecular signaling between L-type Ca2+ channels (LCCs) and ryanodine receptors (RyRs), and the nanodistance between them depends on the interaction between junctophilin-2 (JPH2) in the sarcoplasmic reticulum (SR) and caveolin-3 (CAV3) in the transversal tubule (TT). In heart failure, decreased expression of JPH2 compromises LCC–RyR communication leading to deficient blood-pumping power. In the present study, we found that JPH2 and CAV3 transcription was concurrently regulated by serum response factor (SRF) and myocardin. In cardiomyocytes from torpid ground squirrels, compared with those from euthermic counterparts, myocardin expression was up-regulated, which boosted both JPH2 and CAV3 expression. Transmission electron microscopic imaging showed that the physical coupling between TTs and SRs was tightened during hibernation and after myocardin overexpression. Confocal Ca2+ imaging under the whole-cell patch clamp condition revealed that these changes enhanced the efficiency of LCC–RyR intermolecular signaling and fully compensated the adaptive down-regulation of LCCs, maintaining the power of heart contraction while avoiding the risk of calcium overload during hibernation. Our finding not only revealed an essential molecular mechanism underlying the survival of hibernating mammals, but also demonstrated a “reverse model of heart failure” at the molecular level, suggesting a strategy for treating heart diseases.

    Beschreibung: Maintaining internal salt and water balance in response to fluctuating external conditions is essential for animal survival. This is particularly true for insects as their high surface-to-volume ratio makes them highly susceptible to osmotic stress. However, the cellular and hormonal mechanisms that mediate the systemic control of osmotic homeostasis in beetles (Coleoptera), the largest group of insects, remain largely unidentified. Here, we demonstrate that eight neurons in the brain of the red flour beetle Tribolium castaneum respond to internal changes in osmolality by releasing diuretic hormone (DH) 37 and DH47—homologs of vertebrate corticotropin-releasing factor (CRF) hormones—to control systemic water balance. Knockdown of the gene encoding the two hormones (Urinate, Urn8) reduces Malpighian tubule secretion and restricts organismal fluid loss, whereas injection of DH37 or DH47 reverses these phenotypes. We further identify a CRF-like receptor, Urinate receptor (Urn8R), which is exclusively expressed in a functionally unique secondary cell in the beetle tubules, as underlying this response. Activation of Urn8R increases K+ secretion, creating a lumen-positive transepithelial potential that drives fluid secretion. Together, these data show that beetle Malpighian tubules operate by a fundamentally different mechanism than those of other insects. Finally, we adopt a fluorescent labeling strategy to identify the evolutionary origin of this unusual tubule architecture, revealing that it evolved in the last common ancestor of the higher beetle families. Our work thus uncovers an important homeostatic program that is key to maintaining osmotic control in beetles, which evolved parallel to the radiation of the “advanced” beetle lineages.

    Beschreibung: One of the most fundamental questions in the field of Cys-loop receptors (pentameric ligand-gated ion channels, pLGICs) is how the affinity for neurotransmitters and the conductive/nonconductive state of the transmembrane pore are correlated despite the ∼60-Å distance between the corresponding domains. Proposed mechanisms differ, but they all converge into the idea that interactions between wild-type side chains across the extracellular–transmembrane-domain (ECD–TMD) interface are crucial for this phenomenon. Indeed, the successful design of fully functional chimeras that combine intact ECD and TMD modules from different wild-type pLGICs has commonly been ascribed to the residual conservation of sequence that exists at the level of the interfacial loops even between evolutionarily distant parent channels. Here, using mutagenesis, patch-clamp electrophysiology, and radiolabeled-ligand binding experiments, we studied the effect of eliminating this residual conservation of sequence on ion-channel function and cell-surface expression. From our results, we conclude that proper state interconversion (“gating”) does not require conservation of sequence—or even physicochemical properties—across the ECD–TMD interface. Wild-type ECD and TMD side chains undoubtedly interact with their surroundings, but the interactions between them—straddling the interface—do not seem to be more important for gating than those occurring elsewhere in the protein. We propose that gating of pLGICs requires, instead, that the overall structure of the interfacial loops be conserved, and that their relative orientation and distance be the appropriate ones for changes in one side to result in changes in the other, in a phenomenon akin to the nonspecific “bumping” of closely apposed domains.

    Beschreibung: In four study areas within different lithological settings and rockfall activity, lidar data were applied for a morphometric analysis of block sizes, block shapes and talus cone characteristics. This information was used to investigate the dependencies between block size, block shape and lithology on the one hand and runout distances on the other hand. In our study, we were able to show that lithology seems to have an influence on block size and shape and that gravitational sorting did not occur on all of the studied debris cones but that other parameters apparently control the runout length of boulders. Such a parameter seems to be the block shape, as it plays the role of a moderating parameter in two of the four study sites, while we could not confirm this for our other study sites. We also investigated the influence of terrain parameters such as slope inclination, profile curvature and roughness. The derived roughness values show a clear difference between the four study sites and seem to be a good proxy for block size distribution on the talus cones and thus could be used in further studies to analyse a larger sample of block size distribution on talus cones with different lithologies.

    Beschreibung: Understanding the vertical distribution of aerosol helps to reduce the uncertainty in the aerosol life cycle and therefore in the estimation of the direct and indirect aerosol forcing. To improve our understanding, we use measurements from four deployments of the Atmospheric Tomography (ATom) field campaign (ATom1–4) which systematically sampled aerosol and trace gases over the Pacific and Atlantic oceans with near pole-to-pole coverage. We evaluate the UK Earth System Model (UKESM1) against ATom observations in terms of joint biases in the vertical profile of three variables related to new particle formation: total particle number concentration (NTotal), sulfur dioxide (SO2) mixing ratio and the condensation sink. The NTotal, SO2 and condensation sink are interdependent quantities and have a controlling influence on the vertical profile of each other; therefore, analysing them simultaneously helps to avoid getting the right answer for the wrong reasons. The simulated condensation sink in the baseline model is within a factor of 2 of observations, but the NTotal and SO2 show much larger biases mainly in the tropics and high latitudes. We performed a series of model sensitivity tests to identify atmospheric processes that have the strongest influence on overall model performance. The perturbations take the form of global scaling factors or improvements to the representation of atmospheric processes in the model, for example by adding a new boundary layer nucleation scheme. In the boundary layer (below 1 km altitude) and lower troposphere (1–4 km), inclusion of a boundary layer nucleation scheme (Metzger et al., 2010) is critical to obtaining better agreement with observations. However, in the mid (4–8 km) and upper troposphere (〉 8 km), sub-3 nm particle growth, pH of cloud droplets, dimethyl sulfide (DMS) emissions, upper-tropospheric nucleation rate, SO2 gas-scavenging rate and cloud erosion rate play a more dominant role. We find that perturbations to boundary layer nucleation, sub-3 nm growth, cloud droplet pH and DMS emissions reduce the boundary layer and upper tropospheric model bias simultaneously. In a combined simulation with all four perturbations, the SO2 and condensation sink profiles are in much better agreement with observations, but the NTotal profile still shows large deviations, which suggests a possible structural issue with how nucleation or gas/particle transport or aerosol scavenging is handled in the model. These perturbations are well-motivated in that they improve the physical basis of the model and are suitable for implementation in future versions of UKESM.

    Beschreibung: Stratospheric ozone and water vapour are key components of the Earth system, and past and future changes to both have important impacts on global and regional climate. Here, we evaluate long-term changes in these species from the pre-industrial period (1850) to the end of the 21st century in Coupled Model Intercomparison Project phase 6 (CMIP6) models under a range of future emissions scenarios. There is good agreement between the CMIP multi-model mean and observations for total column ozone (TCO), although there is substantial variation between the individual CMIP6 models. For the CMIP6 multi-model mean, global mean TCO has increased from ∼ 300 DU in 1850 to ∼ 305 DU in 1960, before rapidly declining in the 1970s and 1980s following the use and emission of halogenated ozone-depleting substances (ODSs). TCO is projected to return to 1960s values by the middle of the 21st century under the SSP2-4.5, SSP3-7.0, SSP4-3.4, SSP4-6.0, and SSP5-8.5 scenarios, and under the SSP3-7.0 and SSP5-8.5 scenarios TCO values are projected to be ∼ 10 DU higher than the 1960s values by 2100. However, under the SSP1-1.9 and SSP1-1.6 scenarios, TCO is not projected to return to the 1960s values despite reductions in halogenated ODSs due to decreases in tropospheric ozone mixing ratios. This global pattern is similar to regional patterns, except in the tropics where TCO under most scenarios is not projected to return to 1960s values, either through reductions in tropospheric ozone under SSP1-1.9 and SSP1-2.6, or through reductions in lower stratospheric ozone resulting from an acceleration of the Brewer–Dobson circulation under other Shared Socioeconomic Pathways (SSPs). In contrast to TCO, there is poorer agreement between the CMIP6 multi-model mean and observed lower stratospheric water vapour mixing ratios, with the CMIP6 multi-model mean underestimating observed water vapour mixing ratios by ∼ 0.5 ppmv at 70 hPa. CMIP6 multi-model mean stratospheric water vapour mixing ratios in the tropical lower stratosphere have increased by ∼ 0.5 ppmv from the pre-industrial to the present-day period and are projected to increase further by the end of the 21st century. The largest increases (∼ 2 ppmv) are simulated under the future scenarios with the highest assumed forcing pathway (e.g. SSP5-8.5). Tropical lower stratospheric water vapour, and to a lesser extent TCO, shows large variations following explosive volcanic eruptions.

    Beschreibung: High-quality and long-term soil moisture products are significant for hydrologic monitoring and agricultural management. However, the acquired daily Advanced Microwave Scanning Radiometer 2 (AMSR2) soil moisture products are incomplete in global land (just about 30 %–80 % coverage ratio), due to the satellite orbit coverage and the limitations of soil moisture retrieval algorithms. To solve this inevitable problem, we develop a novel spatio-temporal partial convolutional neural network (CNN) for AMSR2 soil moisture product gap-filling. Through the proposed framework, we generate the seamless daily global (SGD) AMSR2 long-term soil moisture products from 2013 to 2019. To further validate the effectiveness of these products, three verification methods are used as follows: (1) in situ validation, (2) time-series validation, and (3) simulated missing-region validation. Results show that the seamless global daily soil moisture products have reliable cooperativity with the selected in situ values. The evaluation indexes of the reconstructed (original) dataset are a correlation coefficient (R) of 0.685 (0.689), root-mean-squared error (RMSE) of 0.097 (0.093), and mean absolute error (MAE) of 0.079 (0.077). The temporal consistency of the reconstructed daily soil moisture products is ensured with the original time-series distribution of valid values. The spatial continuity of the reconstructed regions is in accordance with the spatial information (R: 0.963–0.974, RMSE: 0.065–0.073, and MAE: 0.044–0.052).     Beschreibung: The intertropical convergence zone (ITCZ) is an important component of the tropical rain belt. Climate models continue to struggle to adequately represent the ITCZ and differ substantially in its simulated response to climate change. Here we employ complex network approaches, which extract spatiotemporal variability patterns from climate data, to better understand differences in the dynamics of the ITCZ in state-of-the-art global circulation models (GCMs). For this purpose, we study simulations with 14 GCMs in an idealized slab-ocean aquaplanet setup from TRACMIP – the Tropical Rain belts with an Annual cycle and a Continent Model Intercomparison Project. We construct network representations based on the spatial correlation patterns of monthly surface temperature anomalies and study the zonal-mean patterns of different topological and spatial network characteristics. Specifically, we cluster the GCMs by means of the distributions of their zonal network measures utilizing hierarchical clustering. We find that in the control simulation, the distributions of the zonal network measures are able to pick up model differences in the tropical sea surface temperature (SST) contrast, the ITCZ position, and the strength of the Southern Hemisphere Hadley cell. Although we do not find evidence for consistent modifications in the network structure tracing the response of the ITCZ to global warming in the considered model ensemble, our analysis demonstrates that coherent variations of the global SST field are linked to ITCZ dynamics. This suggests that climate networks can provide a new perspective on ITCZ dynamics and model differences therein.

    Beschreibung: Recent seismicity in Alberta and north-east British Columbia has been attributed to ongoing oil and gas development in the area, due to its temporal and spatial correlation. Prior to such development, the area was seismically quiescent. Here, we show evidence that latent seismicity may occur in areas where previous operations have occurred, even during a shutdown in operations. The global COVID-19 pandemic furnished the unique opportunity to study seismicity during a long period of anthropogenic quiescence. Within the Kiskatinaw area of British Columbia, 389 events were detected from April to August 2020, which encompasses a period with very little hydraulic fracturing operations. This reduction in operations was the result of a government-imposed lockdown severely restricting the movement of people as well as a downturn in the economic market causing industry stock prices to collapse. Except for a reduction in the seismicity rate and a lack of temporal clustering that is often characteristic of hydraulic fracturing induced sequences, the general characteristics of the observed seismicity were similar to the preceding time period of active operations. During the period of relative quiescence, event magnitudes were observed between ML −0.7 and ML 1.2, which is consistent with previous event magnitudes in the area. Hypocentres occurred in a corridor orientated NW–SE, just as seismicity had done in previous years, and were located at depths associated with the target Montney formation or shallower (

    Beschreibung: Satellite observations (International Satellite Cloud Climatology Project (ISCCP), 1983–2009) of linear trends in cloud cover are compared to those in global precipitation (Global Precipitation Climatology Project (GPCP) pentad V2.2, 1983–2009), to investigate possible cause(s) of the linear trends in both cloud cover and precipitation. The spatial distributions of the linear trends in total cloud cover and precipitation are both characterized primarily by a broadening of the major ascending zone of Hadley circulation. Our correlation studies suggest that global warming, Atlantic Multidecadal Oscillation (AMO) and Pacific Decadal Oscillation (PDO) can explain 67 %, 49 % and 38 %, respectively, of the spatial variabilities in the linear trends in cloud cover, but causality is harder to establish. Further analysis of the broadening of the major ascending zone of Hadley circulation shows that the trend in global temperature, rather than those in AMO and PDO, is the primary contributor to the observed linear trends in total cloud cover and precipitation in 1983–2009. The underlying mechanism driving this broadening is proposed to be the moisture–convection–latent-heat feedback cycle under global warming conditions. The global analysis is extended by investigating connections between clouds and precipitation in China, based on a large number of long-running, high-quality surface weather stations in 1957–2005. This reveals a quantitative matching relationship between the reduction in light precipitation and the reduction in total cloud cover. Furthermore, our study suggests that the reduction in cloud cover in China is primarily driven by global temperature; PDO plays a secondary role, while the contribution from AMO and Niño3.4 is insignificant, consistent with the global analysis.

    Beschreibung: This study was performed in the framework of HyMeX (Hydrological cycle in the Mediterranean Experiment), which aimed to study the heavy precipitation that regularly affects the Mediterranean area. A reanalysis with a convective-scale model AROME-WMED (Application of Research to Operations at MEsoscale western Mediterranean) was performed, which assimilated most of the available data for a 2-month period corresponding to the first special observation period of the field campaign (Fourrié et al., 2019). Among them, observations related to the low-level humidity flow were assimilated. Such observations are important for the description of the feeding of the convective mesoscale systems with humidity (Duffourg and Ducrocq, 2011; Bresson et al., 2012; Ricard et al., 2012). Among them there were a dense reprocessed network of high-quality Global Navigation Satellite System (GNSS) zenithal total delay (ZTD) observations, reprocessed data from wind profilers, lidar-derived vertical profiles of humidity (ground and airborne) and Spanish radar data. The aim of the paper is to assess the impact of the assimilation of these four observation types on the analyses and the forecasts from the 3 h forecast range (first guess) up to the 48 h forecast range. In order to assess this impact, several observing system experiments (OSEs) or so-called denial experiments, were carried out by removing one single data set from the observation data set assimilated in the reanalysis. Among the evaluated observations, it is found that the ground-based GNSS ZTD data set provides the largest impact on the analyses and the forecasts, as it represents an evenly spread and frequent data set providing information at each analysis time over the AROME-WMED domain. The impact of the reprocessing of GNSS ZTD data also improves the forecast quality, but this impact is not statistically significant. The assimilation of the Spanish radar data improves the 3 h precipitation forecast quality as well as the short-term (30 h) precipitation forecasts, but this impact remains located over Spain. Moreover, marginal impact from wind profilers was observed on wind background quality. No impacts have been found regarding lidar data, as they represent a very small data set, mainly located over the sea.

    Beschreibung: Systematic measurements of the atmospheric Ar∕N2 ratio have been made at ground-based stations in Japan and Antarctica since 2012. Clear seasonal cycles of the Ar∕N2 ratio with summertime maxima were found at middle- to high-latitude stations, with seasonal amplitudes increasing with increasing latitude. Eight years of the observed Ar∕N2 ratio at Tsukuba (TKB) and Hateruma (HAT), Japan, showed interannual variations in phase with the observed variations in the global ocean heat content (OHC). We calculated secularly increasing trends of 0.75 ± 0.30 and 0.89 ± 0.60 per meg per year from the Ar∕N2 ratio observed at TKB and HAT, respectively, although these trend values are influenced by large interannual variations. In order to examine the possibility of the secular trend in the surface Ar∕N2 ratio being modified significantly by the gravitational separation in the stratosphere, two-dimensional model simulations were carried out by arbitrarily modifying the mass stream function in the model to simulate either a weakening or an enhancement of the Brewer–Dobson circulation (BDC). The secular trend of the Ar∕N2 ratio at TKB, corrected for gravitational separation under the assumption of weakening (enhancement) of BDC simulated by the 2-D model, was 0.60 ± 0.30 (0.88 ± 0.30) per meg per year. By using a conversion factor of 3.5 × 10−23 per meg per joule by assuming a one-box ocean with a temperature of 3.5 ∘C, average OHC increase rates of 17.1 ± 8.6 ZJ yr−1 and 25.1 ± 8.6 ZJ yr−1 for the period 2012–2019 were estimated from the corrected secular trends of the Ar∕N2 ratio for the weakened- and enhanced-BDC conditions, respectively. Both OHC increase rates from the uncorrected- and weakened-BDC secular trends of the Ar∕N2 ratio are consistent with 12.2 ± 1.2 ZJ yr−1 reported by ocean temperature measurements, while that from the enhanced-BDC is outside of the range of the uncertainties. Although the effect of the actual atmospheric circulation on the Ar∕N2 ratio is still unclear and longer-term observations are needed to reduce uncertainty of the secular trend of the surface Ar∕N2 ratio, the analytical results obtained in the present study imply that the surface Ar∕N2 ratio is an important tracer for detecting spatiotemporally integrated changes in OHC and BDC.

    Beschreibung: Understanding the evolution of, and the interactions between, ice sheets and the global climate over geological timescales is important for being able to project their future evolution. However, direct observational evidence of past CO2 concentrations, and the implied radiative forcing, only exists for the past 800 000 years. Records of benthic δ18O date back millions of years but contain signals from both land ice volume and ocean temperature. In recent years, inverse forward modelling has been developed as a method to disentangle these two signals, resulting in mutually consistent reconstructions of ice volume, temperature, and CO2. We use this approach to force a hybrid ice-sheet–climate model with a benthic δ18O stack, reconstructing the evolution of the ice sheets, global mean sea level, and atmospheric CO2 during the late Pliocene and the Pleistocene, from 3.6 million years (Myr) ago to the present day. During the warmer-than-present climates of the late Pliocene, reconstructed CO2 varies widely, from 320–440 ppmv for warm periods to 235–250 ppmv for the early glacial excursion ∼3.3 million years ago. Sea level is relatively stable during this period, with maxima of 6–14 m and minima of 12–26 m during glacial episodes. Both CO2 and sea level are within the wide ranges of values covered by available proxy data for this period. Our results for the Pleistocene agree well with the ice-core CO2 record, as well as with different available sea-level proxy data. For the Early Pleistocene, 2.6–1.2 Myr ago, we simulate 40 kyr glacial cycles, with interglacial CO2 decreasing from 280–300 ppmv at the beginning of the Pleistocene to 250–280 ppmv just before the Mid-Pleistocene Transition (MPT). Peak glacial CO2 decreases from 220–250 to 205–225 ppmv during this period. After the MPT, when the glacial cycles change from 40 to 80 120 kyr cyclicity, the glacial–interglacial contrast increases, with interglacial CO2 varying between 250–320 ppmv and peak glacial values decreasing to 170–210 ppmv.

    Beschreibung: In urban areas, road traffic is a dominant source of nitrogen oxides (NOx=NO+NO2). Although the emissions from individual vehicles are regulated by the European emission standards, real driving emissions often exceed these limits. In this study, two multi-axis differential optical absorption spectroscopy (MAX-DOAS) instruments on opposite sides of the motorway were used to measure the NO2 absorption caused by road traffic at the A60 motorway close to Mainz, Germany. In combination with wind data, the total NOx emissions for the occurring traffic volume can be estimated. Hereto, the ozone-dependent photochemical equilibrium between NO and NO2 is considered. We show that for 10 May 2019 the measured emissions exceed the maximum expected emissions calculated from the European emission standards for standardised test cycles by a factor of 11±7. One major advantage of the method used here is that MAX-DOAS measurements are very sensitive to the integrated NO2 concentration close to the surface. Thus, all emitted NO2 molecules are detected independently from their altitude, and therefore the whole emission plume originating from the nearby motorway is captured, which is a key advantage compared to other approaches such as in situ measurements.

    Beschreibung: Cis-acting RNA elements are crucial for the regulation of polyadenylated RNA stability. The element for nuclear expression (ENE) contains a U-rich internal loop flanked by short helices. An ENE stabilizes RNA by sequestering the poly(A) tail via formation of a triplex structure that inhibits a rapid deadenylation-dependent decay pathway. Structure-based bioinformatic studies identified numerous ENE-like elements in evolutionarily diverse genomes, including a subclass containing two ENE motifs separated by a short double-helical region (double ENEs [dENEs]). Here, the structure of a dENE derived from a rice transposable element (TWIFB1) before and after poly(A) binding (∼24 kDa and ∼33 kDa, respectively) is investigated. We combine biochemical structure probing, small angle X-ray scattering (SAXS), and cryo‐electron microscopy (cryo-EM) to investigate the dENE structure and its local and global structural changes upon poly(A) binding. Our data reveal 1) the directionality of poly(A) binding to the dENE, and 2) that the dENE-poly(A) interaction involves a motif that protects the 3ʹ-most seven adenylates of the poly(A). Furthermore, we demonstrate that the dENE does not undergo a dramatic global conformational change upon poly(A) binding. These findings are consistent with the recently solved crystal structure of a dENE+poly(A) complex [S.-F. Torabi et al., Science 371, eabe6523 (2021)]. Identification of additional modes of poly(A)–RNA interaction opens new venues for better understanding of poly(A) tail biology.

    Beschreibung: Autophagy is a catabolic pathway that provides self-nourishment and maintenance of cellular homeostasis. Autophagy is a fundamental cell protection pathway through metabolic recycling of various intracellular cargos and supplying the breakdown products. Here, we report an autophagy function in governing cell protection during cellular response to energy crisis through cell metabolic rewiring. We observe a role of selective type of autophagy in direct activation of cyclic AMP protein kinase A (PKA) and rejuvenation of mitochondrial function. Mechanistically, autophagy selectively degrades the inhibitory subunit RI of PKA holoenzyme through A-kinase–anchoring protein (AKAP) 11. AKAP11 acts as an autophagy receptor that recruits RI to autophagosomes via LC3. Glucose starvation induces AKAP11-dependent degradation of RI, resulting in PKA activation that potentiates PKA-cAMP response element-binding signaling, mitochondria respiration, and ATP production in accordance with mitochondrial elongation. AKAP11 deficiency inhibits PKA activation and impairs cell survival upon glucose starvation. Our results thus expand the view of autophagy cytoprotection mechanism by demonstrating selective autophagy in RI degradation and PKA activation that fuels the mitochondrial metabolism and confers cell resistance to glucose deprivation implicated in tumor growth.

    Beschreibung: To provide long-term air pollutant exposure estimates for epidemiological studies, it is essential to test the feasibility of developing land-use regression (LUR) models using only routine air quality measurement data and to evaluate the transferability of LUR models between nearby cities. In this study, we developed and evaluated the intercity transferability of annual-average LUR models for ambient respirable suspended particulates (PM10), fine suspended particulates (PM2.5), nitrogen dioxide (NO2) and ozone (O3) in the Taipei–Keelung metropolitan area of northern Taiwan in 2019. Ambient PM10, PM2.5, NO2 and O3 measurements at 30 fixed-site stations were used as the dependent variables, and a total of 156 potential predictor variables in six categories (i.e., population density, road network, land-use type, normalized difference vegetation index, meteorology and elevation) were extracted using buffer spatial analysis. The LUR models were developed using the supervised forward linear regression approach. The LUR models for ambient PM10, PM2.5, NO2 and O3 achieved relatively high prediction performance, with R2 values of 〉 0.72 and leave-one-out cross-validation (LOOCV) R2 values of 〉 0.53. The intercity transferability of LUR models varied among the air pollutants, with transfer-predictive R2 values of 〉 0.62 for NO2 and

    Beschreibung: The species Selenogonus narinoensis was described by Stirton (1947) based on a single specimen which comes from sediments cropping out in the Cocha Verde locality, Nariño Department (Colombia), tentatively referred to the late Pliocene–Pleistocene (MGN 931; IGM p002118, Museo Geológico Nacional, Servicio Geológico Colombiano, Bogotá). However, morphological studies and comparative morphometric observations of the specimen suggest that (1) no diagnostic character supports the validity of the species Selenogonus narinoensis (here considered species inquirenda); (2) a combination of features (e.g., the mandibular condyle located behind the posterior edge of the vertical mandibular ramus, the angular process which projects laterally outwards, a bunolophodont crown morphology, a mesodont crown height, and a simple crown morphology of the third lobe of m3) indicates it belongs to the genus Platygonus; (3) this specimen corresponds to one of the largest South American peccaries; (4) taking into account certain anatomical characters as well as its morphometric range, this specimen is assigned to Platygonus cf. marplatensis. Even though the stratigraphic provenance of the specimen is still doubtful, it can be proposed that (1) it could be one of the most ancient records of tayassuids in South America, as would be expected given its geographical position, and (2) considering the new taxonomic proposal, this specimen represents the first record of Platygonus cf. marplatensis in Colombia and represents one of the northernmost South American records of the genus. This new interpretation would be of great relevance in the Great American Biotic Interchange due to its strategic geographical proximity to the Isthmus of Panama.

    Beschreibung: We experimentally quantified the impact of cloud fraction and cloud type on the heating rate (HR) of black and brown carbon (HRBC and HRBrC). In particular, we examined in more detail the cloud effect on the HR detected in a previous study (Ferrero et al., 2018). High-time-resolution measurements of the aerosol absorption coefficient at multiple wavelengths were coupled with spectral measurements of the direct, diffuse and surface reflected irradiance and with lidar–ceilometer data during a field campaign in Milan, Po Valley (Italy). The experimental set-up allowed for a direct determination of the total HR (and its speciation: HRBC and HRBrC) in all-sky conditions (from clear-sky conditions to cloudy). The highest total HR values were found in the middle of winter (1.43 ± 0.05 K d−1), and the lowest were in spring (0.54 ± 0.02 K d−1). Overall, the HRBrC accounted for 13.7 ± 0.2 % of the total HR, with the BrC being characterized by an absorption Ångström exponent (AAE) of 3.49 ± 0.01. To investigate the role of clouds, sky conditions were classified in terms of cloudiness (fraction of the sky covered by clouds: oktas) and cloud type (stratus, St; cumulus, Cu; stratocumulus, Sc; altostratus, As; altocumulus, Ac; cirrus, Ci; and cirrocumulus–cirrostratus, Cc–Cs). During the campaign, clear-sky conditions were present 23 % of the time, with the remaining time (77 %) being characterized by cloudy conditions. The average cloudiness was 3.58 ± 0.04 oktas (highest in February at 4.56 ± 0.07 oktas and lowest in November at 2.91 ± 0.06 oktas). St clouds were mostly responsible for overcast conditions (7–8 oktas, frequency of 87 % and 96 %); Sc clouds dominated the intermediate cloudiness conditions (5–6 oktas, frequency of 47 % and 66 %); and the transition from Cc–Cs to Sc determined moderate cloudiness (3–4 oktas); finally, low cloudiness (1–2 oktas) was mostly dominated by Ci and Cu (frequency of 59 % and 40 %, respectively). HR measurements showed a constant decrease with increasing cloudiness of the atmosphere, enabling us to quantify for the first time the bias (in %) of the aerosol HR introduced by the simplified assumption of clear-sky conditions in radiative-transfer model calculations. Our results showed that the HR of light-absorbing aerosol was ∼ 20 %–30 % lower in low cloudiness (1–2 oktas) and up to 80 % lower in completely overcast conditions (i.e. 7–8 oktas) compared to clear-sky ones. This means that, in the simplified assumption of clear-sky conditions, the HR of light-absorbing aerosol can be largely overestimated (by 50 % in low cloudiness, 1–2 oktas, and up to 500 % in completely overcast conditions, 7–8 oktas). The impact of different cloud types on the HR was also investigated. Cirrus clouds were found to have a modest impact, decreasing the HRBC and HRBrC by −5 % at most. Cumulus clouds decreased the HRBC and HRBrC by −31 ± 12 % and −26 ± 7 %, respectively; cirrocumulus–cirrostratus clouds decreased the HRBC and HRBrC by −60 ± 8 % and −54 ± 4 %, which was comparable to the impact of altocumulus (−60 ± 6 % and −46 ± 4 %). A higher impact on the HRBC and HRBrC suppression was found for stratocumulus (−63 ± 6 % and −58 ± 4 %, respectively) and altostratus (−78 ± 5 % and −73 ± 4 %, respectively). The highest impact was associated with stratus, suppressing the HRBC and HRBrC by −85 ± 5 % and −83 ± 3 %, respectively. The presence of clouds caused a decrease of both the HRBC and HRBrC (normalized to the absorption coefficient of the respective species) of −11.8 ± 1.2 % and −12.6 ± 1.4 % per okta. This study highlights the need to take into account the role of both cloudiness and different cloud types when estimating the HR caused by both BC and BrC and in turn decrease the uncertainties associated with the quantification of their impact on the climate.

    Beschreibung: The atomic structure of the complete myosin tail within thick filaments isolated from Lethocerus indicus flight muscle is described and compared to crystal structures of recombinant, human cardiac myosin tail segments. Overall, the agreement is good with three exceptions: the proximal S2, in which the filament has heads attached but the crystal structure doesn’t, and skip regions 2 and 4. At the head–tail junction, the tail α-helices are asymmetrically structured encompassing well-defined unfolding of 12 residues for one myosin tail, ∼4 residues of the other, and different degrees of α-helix unwinding for both tail α-helices, thereby providing an atomic resolution description of coiled-coil “uncoiling” at the head–tail junction. Asymmetry is observed in the nonhelical C termini; one C-terminal segment is intercalated between ribbons of myosin tails, the other apparently terminating at Skip 4 of another myosin tail. Between skip residues, crystal and filament structures agree well. Skips 1 and 3 also agree well and show the expected α-helix unwinding and coiled-coil untwisting in response to skip residue insertion. Skips 2 and 4 are different. Skip 2 is accommodated in an unusual manner through an increase in α-helix radius and corresponding reduction in rise/residue. Skip 4 remains helical in one chain, with the other chain unfolded, apparently influenced by the acidic myosin C terminus. The atomic model may shed some light on thick filament mechanosensing and is a step in understanding the complex roles that thick filaments of all species undergo during muscle contraction.

    Beschreibung: Rift Valley fever virus (RVFV), an emerging arboviral and zoonotic bunyavirus, causes severe disease in livestock and humans. Here, we report the isolation of a panel of monoclonal antibodies (mAbs) from the B cells of immune individuals following natural infection in Kenya or immunization with MP-12 vaccine. The B cell responses of individuals who were vaccinated or naturally infected recognized similar epitopes on both Gc and Gn proteins. The Gn-specific mAbs and two mAbs that do not recognize either monomeric Gc or Gn alone but recognized the hetero-oligomer glycoprotein complex (Gc+Gn) when Gc and Gn were coexpressed exhibited potent neutralizing activities in vitro, while Gc-specific mAbs exhibited relatively lower neutralizing capacity. The two Gc+Gn–specific mAbs and the Gn domain A-specific mAbs inhibited RVFV fusion to cells, suggesting that mAbs can inhibit the exposure of the fusion loop in Gc, a class II fusion protein, and thus prevent fusion by an indirect mechanism without direct fusion loop contact. Competition-binding analysis with coexpressed Gc/Gn and mutagenesis library screening indicated that these mAbs recognize four major antigenic sites, with two sites of vulnerability for neutralization on Gn. In experimental models of infection in mice, representative mAbs recognizing three of the antigenic sites reduced morbidity and mortality when used at a low dose in both prophylactic and therapeutic settings. This study identifies multiple candidate mAbs that may be suitable for use in humans against RVFV infection and highlights fusion inhibition against bunyaviruses as a potential contributor to potent antibody-mediated neutralization.

    Beschreibung: Limited water availability, population growth, and climate change have resulted in freshwater crises in many countries. Jordan’s situation is emblematic, compounded by conflict-induced population shocks. Integrating knowledge across hydrology, climatology, agriculture, political science, geography, and economics, we present the Jordan Water Model, a nationwide coupled human–natural-engineered systems model that is used to evaluate Jordan’s freshwater security under climate and socioeconomic changes. The complex systems model simulates the trajectory of Jordan’s water system, representing dynamic interactions between a hierarchy of actors and the natural and engineered water environment. A multiagent modeling approach enables the quantification of impacts at the level of thousands of representative agents across sectors, allowing for the evaluation of both systemwide and distributional outcomes translated into a suite of water-security metrics (vulnerability, equity, shortage duration, and economic well-being). Model results indicate severe, potentially destabilizing, declines in freshwater security. Per capita water availability decreases by approximately 50% by the end of the century.     Beschreibung: What role do ideologically extreme media play in the polarization of society? Here we report results from a randomized longitudinal field experiment embedded in a nationally representative online panel survey (N = 1,037) in which participants were incentivized to change their browser default settings and social media following patterns, boosting the likelihood of encountering news with either a left-leaning (HuffPost) or right-leaning (Fox News) slant during the 2018 US midterm election campaign. Data on ≈ 19 million web visits by respondents indicate that resulting changes in news consumption persisted for at least 8 wk. Greater exposure to partisan news can cause immediate but short-lived increases in website visits and knowledge of recent events. After adjusting for multiple comparisons, however, we find little evidence of a direct impact on opinions or affect. Still, results from later survey waves suggest that both treatments produce a lasting and meaningful decrease in trust in the mainstream media up to 1 y later. Consistent with the minimal-effects tradition, direct consequences of online partisan media are limited, although our findings raise questions about the possibility of subtle, cumulative dynamics. The combination of experimentation and computational social science techniques illustrates a powerful approach for studying the long-term consequences of exposure to partisan news.

    Beschreibung: In nerve cells the genes encoding for α2δ subunits of voltage-gated calcium channels have been linked to synaptic functions and neurological disease. Here we show that α2δ subunits are essential for the formation and organization of glutamatergic synapses. Using a cellular α2δ subunit triple-knockout/knockdown model, we demonstrate a failure in presynaptic differentiation evidenced by defective presynaptic calcium channel clustering and calcium influx, smaller presynaptic active zones, and a strongly reduced accumulation of presynaptic vesicle-associated proteins (synapsin and vGLUT). The presynaptic defect is associated with the downscaling of postsynaptic AMPA receptors and the postsynaptic density. The role of α2δ isoforms as synaptic organizers is highly redundant, as each individual α2δ isoform can rescue presynaptic calcium channel trafficking and expression of synaptic proteins. Moreover, α2δ-2 and α2δ-3 with mutated metal ion-dependent adhesion sites can fully rescue presynaptic synapsin expression but only partially calcium channel trafficking, suggesting that the regulatory role of α2δ subunits is independent from its role as a calcium channel subunit. Our findings influence the current view on excitatory synapse formation. First, our study suggests that postsynaptic differentiation is secondary to presynaptic differentiation. Second, the dependence of presynaptic differentiation on α2δ implicates α2δ subunits as potential nucleation points for the organization of synapses. Finally, our results suggest that α2δ subunits act as transsynaptic organizers of glutamatergic synapses, thereby aligning the synaptic active zone with the postsynaptic density.

    Beschreibung: Extensive and rigorous model intercomparison is of great importance before model application due to the uncertainties in current land surface models (LSMs). Without considering the uncertainties in forcing data and model parameters, this study designed an ensemble of 55 296 experiments to evaluate the Noah LSM with multi-parameterization (Noah-MP) for snow cover events (SCEs), soil temperature (ST) and soil liquid water (SLW) simulation, and investigated the sensitivity of parameterization schemes at a typical permafrost site on the Qinghai–Tibet Plateau (QTP). The results showed that Noah-MP systematically overestimates snow cover, which could be greatly resolved when adopting the sublimation from wind and a semi-implicit snow/soil temperature time scheme. As a result of the overestimated snow, Noah-MP generally underestimates ST, which is mostly influenced by the snow process. A systematic cold bias and large uncertainties in soil temperature remain after eliminating the effects of snow, particularly in the deep layers and during the cold season. The combination of roughness length for heat and under-canopy (below-canopy) aerodynamic resistance contributes to resolving the cold bias in soil temperature. In addition, Noah-MP generally underestimates top SLW. The runoff and groundwater (RUN) process dominates the SLW simulation in comparison to the very limited impacts of all other physical processes. The analysis of the model structural uncertainties and characteristics of each scheme would be constructive to a better understanding of the land surface processes in the permafrost regions of the QTP as well as to further model improvements towards soil hydrothermal regime modeling using LSMs.

    Beschreibung: Organic nitrates are secondary species in the atmosphere. Their fate is related to the chemical transport of pollutants from polluted areas to more distant zones. While their gas-phase chemistry has been studied, their reactivity in condensed phases is far from being understood. However, these compounds represent an important fraction of organic matter in condensed phases. In particular, their partition to the aqueous phase may be especially important for oxidized organic nitrates for which water solubility increases with functionalization. This work has studied for the first time the aqueous-phase ⚫OH-oxidation kinetics of four alkyl nitrates (isopropyl nitrate, isobutyl nitrate, 1-pentyl nitrate, and isopentyl nitrate) and three functionalized organic nitrates (α-nitrooxyacetone, 1-nitrooxy-2-propanol, and isosorbide 5-mononitrate) by developing a novel and accurate competition kinetic method. Low reactivity was observed, with kOH ranging from 8×107 to 3.1×109 L mol−1 s−1 at 296±2 K. Using these results, a previously developed aqueous-phase structure–activity relationship (SAR) was extended, and the resulting parameters confirmed the extreme deactivating effect of the nitrate group, up to two adjacent carbon atoms. The achieved extended SAR was then used to determine the ⚫OH-oxidation rate constants of 49 organic nitrates, including hydroxy nitrates, ketonitrates, aldehyde nitrates, nitrooxy carboxylic acids, and more functionalized organic nitrates such as isoprene and terpene nitrates. Their multiphase atmospheric lifetimes towards ⚫OH oxidation were calculated using these rate constants, and they were compared to their gas-phase lifetimes. Large differences were observed, especially for polyfunctional organic nitrates: for 50 % of the proposed organic nitrates for which the ⚫OH reaction occurs mainly in the aqueous phase (more than 50 % of the overall removal), their ⚫OH-oxidation lifetimes increased by 20 % to 155 % under cloud/fog conditions (liquid water content LWC = 0.35 g m−3). In particular, for 83 % of the proposed terpene nitrates, the reactivity towards ⚫OH occurred mostly (〉98 %) in the aqueous phase, while for 60 % of these terpene nitrates, their lifetimes increased by 25 % to 140 % compared to their gas-phase reactivity. We demonstrate that these effects are of importance under cloud/fog conditions but also under wet aerosol conditions, especially for the terpene nitrates. These results suggest that considering aqueous-phase ⚫OH-oxidation reactivity of biogenic nitrates is necessary to improve the predictions of their atmospheric fate.

    Beschreibung: Scholars have long debated whether animals, which display impressive intelligent behaviors, are consciously aware or not. Yet, because many complex human behaviors and high-level functions can be performed without conscious awareness, it was long considered impossible to untangle whether animals are aware or just conditionally or nonconsciously behaving. Here, we developed an empirical approach to address this question. We harnessed a well-established cross-over double dissociation between nonconscious and conscious processing, in which people perform in completely opposite ways when they are aware of stimuli versus when they are not. To date, no one has explored if similar performance dissociations exist in a nonhuman species. In a series of seven experiments, we first established these signatures in humans using both known and newly developed nonverbal double-dissociation tasks and then identified similar signatures in rhesus monkeys (Macaca mulatta). These results provide robust evidence for two distinct modes of processing in nonhuman primates. This empirical approach makes it feasible to disentangle conscious visual awareness from nonconscious processing in nonhuman species; hence, it can be used to strip away ambiguity when exploring the processes governing intelligent behavior across the animal kingdom. Taken together, these results strongly support the existence of both nonconscious processing as well as functional human-like visual awareness in nonhuman animals.
