    Description / Table of Contents: Zusammenfassung Zur Vertiefung des Verständnisses vom Stoffwechsel des Zuckeraustauschstoffes Palatinit® wurden seine zwei Bestandteile D-Glucosyl-α(1→1)-D-mannit und D-Glucosyl-α(1→6)-D-glucit [D-Glucosyl-α(1→6)-D-sorbit] nach dem Verfahren von Karimzadegan et al. auf ihre Glucose-Bioverfügbarkeit an ketotischen Ratten untersucht. Bei Umwandlungsraten in Glucose von 6 bzw. 20 % für Mannit und Glucit (Sorbit) sowie von 39 bzw. 42% für Glucosylmannit und Glucosylglucit erhält demnach der metabolische Glucose-Pool nicht das volle Glucose-Äquivalent aus diesen Verbindungen. Von dem Anteil an präformierter Glucose in den Glucosylhexiten — theoretisches Maximum 50 % — sind nur 36 % aus Glucosylmannit bzw. 32 % aus Glucosylglucit bioverfügbar. Die im Vergleich zur Theorie verminderte Bioverfügbarkeit von Glucose aus Palatinit® wird auf partiellen mikrobiellen Abbau in unteren Darmabschnitten zurückgeführt. Die an Ratten erhaltenen Ergebnisse werden auch für alle anderen Spezies gelten, welche in Caecum und/oder Colon Kohlenhydrate vergären. Die Unterschiede zwischen D-Glucosyl-α(1→1)-D-mannit und D-Glucosyl-α(1→6)-D-glucit werden durch unterschiedliche Verzögerung der Glucoseresorption im Dünndarm, wo auch D-Glucit angreift, bedingt. Die Ermittlung der Glucose-Bioverfügbarkeit gewährt weitgehende Einblicke in das Schicksal von Kohlenhydraten einschließlich der Symbiose zwischen Säugetier und Mikroorganismen im Dickdarm. Da ein ziemlich vollständiger Überblick über die metabolischen Konsequenzen nach ihrer Zufuhr erhalten wird, sollte das Verfahren zur Messung der Bioverfügbarkeit von Glucose daher bei Abschätzungen der Lebensmittelsicherheit anderer Zuckeraustauschstoffe ebenfalls angewandt werden.
    Notes: Summary For the sake of metabolic insight into the fate of the sugar substitute Palatinit®, its two components D-glucosyl-α(1→1)-D-mannitol and D-glucosyl-α(1→6)-D-glucitol [D-glucosyl-α-(1→6)-D-sorbitol] were assayed for glucose bioavailability by the procedure of Karimzadegan et al. using ketotic rats. With conversion rates into glucose of 6 and 20 %, respectively, for free mannitol and glucitol (sorbitol), 39 % for glucosylmannitol and 42 % for glucosylglucitol, the metabolic glucose pool of the rat does not receive the full carbohydrate complement of these compounds. The preformed glucose moiety of the glucosylhexitols is bioavailable by 36 and 32 %, respectively, from glucosylmannitol and glucosylglucitol, with 50 % as theoretical maximum. Less than theoretical bioavailability of glucose from Palatinit® is ascribed to microbial attack in the hindgut. The data on rats are held valid also for other species demonstrating carbohydrate fermentation in the caecum and/or colon. Differences between D-glucosyl-α(1→1)-D-mannitol and D-glucosyl-α(1→6)-D-glucitol are caused by a differential delay of glucose absorption in the small intestine, also exerted by D-glucitol. The deep metabolic insight offered by the glucose bioavailability assay into the fate of carbohydrates includes the mammal-microbial symbiosis in the large bowel. Since a rather complete survey of the metabolic consequences after their intake can be obtained, the assay system should be generally applied in assessments of food safety also of other sugar substitutes.

    Notes: Summary The instrumentation suitable for the acquisition of biomagnetic signals is reviewed. The basic magnetometer components,i.e. SQUIDs, flux transformers and dewars, are studied separately. Possible improvements to the present instrumentation as well as complete set-ups including ferromagnetic or eddy current shields are discussed and their efficiency evaluated.

    Description / Table of Contents: Résumé Après leur naissance, les larves de Cyrtobagous sp. passent de 1 à 4 jours à brouter les racines, et de 0 à 9 jours dans les bourgeons de la plante hôte, Salvinia molesta, avant d'y creuser une galerie dans le rhizome. Bien que les larves aient été capables de survivre jusqu'à 38 jours en broutant les racines, quant elles furent privées de rhizome, elle ne purent achever leur développement. Quand les galeries furent commencées, le développement larvaire, entre 21° et 31°, dépendit de la température et de la teneur en azote de l'hôte. Les larves ne se développèrent pas à 17°. Une haute valeur nutritive de l'aliment consommé par les larves réduisit la durée du développement, mais n'influença pas la durée ultérieure de leur développement nymphal. Les lieux de prise de nourriture et les dégâts produits par des larves isolées furent précisés.
    Notes: Abstract Newly-emerged larvae of a salvinia weevil, Cyrtobagous sp. spent from 1–4 days browsing on the roots and from 0–9 days in the buds of the host plant, Salvinia molesta, prior to tunnelling into the rhizome of this aquatic weed. Although larvae were able to survive up to 38 days browsing on roots when rhizomes were withheld, they were unable to complete development. After tunnelling began, larval development between 21° and 31° was dependent on temperature and nitrogen levels in the host. Larvae failed to develop at 17°. High nutritional intake by the larvae reduced larval development time but did not influence duration of their subsequent pupal development. The feeding sites and plant damage produced by individual larvae were assessed.

    Description / Table of Contents: Résumé Les réponses de Prostephanus truncatus Horn (Coleoptera: Bostrichidae) à deux constituants de la phéromone d'agrégation de Rhyzopertha dominica Fabricius (Coleoptera: Bostrichidae), le (±)-1-méthylbutyl (E)-2-méthyl-2-penténoate et le (±)-1-méthylbutyl (E)-2,4-diméthyl-2-pentéoate, connus comme “Dominicalure 1” et “Dominicalure 2”, ont été étudiées au laboratoire et au champ. Auslaboratoire ces composés ont été essayés individuellement et en mélange 1:2. Une membrane de caoutchouc imprégnée de phéromone a été utilisée comme émetteur lent, et une courbe de réponse en fonction de la dose a été établie. P. truncatus répondait plus vigoureusement à la Dominicalure 2 seule. Tous ces traitements ont été comparés dans un essai aux champs en Tanzanie, dans lesquels des pièges en carton ondulé contenant des diffuseurs de phéromone étaient placés dans des entrepôts de maïs dans les fermes. La Dominicalure 1 et le mélange de 1 et 2 étaient tous deux efficaces pour piéger R. dominica. La Dominicalure 2 a piégé plus de P. truncatus que les autres traitements et était à peu près aussi efficace que le contrôle visuel pour détecter la présence de cet insecte dans les stocks. On peut envisager que la Dominicalure 2 pourrait servir de base à un programme pour lutter contre P. truncatus en Afrique de l'Est.
    Notes: Abstract Laboratory and field studies have been made of the responses of Prostephanus truncatus (Horn) to two components of the aggregation pheromone of Rhyzopertha dominica (Fabricius) — (±)-1-methylbutyl (E)-2-methyl-2-pentenoate and (±)-1-methylbutyl (E)-2,4-dimethyl-2-pentenoate, known as “Dominicalure 1” and “Dominicalure 2” respectively. In the laboratory, these compounds were tested individually and as a 1:2 mixture; P. truncatus responded most strongly to Dominicalure 2 alone. All three treatments were compared in a field trial in Tanzania to monitor both R. dominica and P. truncatus in farm stores. Dominicalure 1 or a mixture of 1 and 2 were both highly effective for trapping R. dominica. In contrast, Dominicalure 2 trapped more P. truncatus than the other treatments and was about as effective as visual inspection at demonstrating the presence of the beetle in stores. Dominicalure 2 could form the basis of a monitoring programme for P. truncatus in East Africa.

Si considerano le condizioni di massimo rapporto segnale-rumore dell’uscita del quantomagnetometro superconduttote (SQM). Si analizza l’influenza dei parametri misurati della sorgente del segnale, del rumore di una sorgente di disturbo esterna e di quello proprio del magnetometro sulla selezione della lunghezza di base del gradiometro.
    Notes: Summary In this paper we deal with an optimization problem of the second-order superconducting gradiometric sensor systems that have been designed for measurement of the weak inhomogeneous magnetic fields in a magnetically disturbed environment. The conditions of maximum signalto-noise ratio of the output of superconducting quantum magnetometers (SQM) are considered. The influence of measured signal source parameters, of an outside disturbing source and of the own noise of the magnetometer on the gradiometer base length selection is analysed.

Si è trovato che la componente antero-posterioreX vettore magnetico del cuore è la piú facile da scoprire. Si suggeriscono conduttori ideali per le altre due componenti. Comunque questi hanno una sensibilità inferiore. Le differenze rispetto ai risultati di precedenti ricercatori sono state spiegate con le differenze tra i modelli.
    Notes: Summary The ideality and sensitivity of several vector magnetocardiographic leads were tested by using an inhomogeneous, physical torso model. The postero-anterior component of the heart’s magnetic vector was found easiest to detect. New, more ideal leads for the other two components are suggested. However, these have lower sensitivity. The differences from the results of former investigators can be explained by the differences of the models.

    Description / Table of Contents: Riassunto Si è studiato lo spettro del rumore di un gradiometro superconduttore a derivata secon da in un agglomerato urbano. I dati sono stati ottenuti dal gradiometro con tre accelerometri ortogonali e un magnetometro a flussometro elettronico triassiale attaccato al dewar. Questi dati sono stati analizzati usando tecniche di elaborazione dei segnali, soprattutto un cancellatore adattativo di rumori, per ridurre i rumori nei dati del gradiometro. I risultati mostrano qui che il rumore dell’esterno e/o il rumore del movimento del dewar possono essere ridotti di 40 dB in potenza di rumore. Si è anche mostrato che, nel caso di basso rapporto segnale-rumore (S/N∼1), può essere estratta un’onda sunusoidale di 10 Hz. Questa tecnica non solo promette riduzione dei rumori, ma ha anche lo scopo d’identificare i segnali di rumore che potrebbero essere veramente considerati parte del responso evocato. L’analisi dei dati che contengono i responsi evocati è ora in corso.
    Notes: Summary The noise spectrum of a superconducting second-derivative gradiometer has been investigated in an urban environment. Data have been acquired from the gradiometer with three orthogonal accelerometers and a triaxial fluxgate magnetometer attached to the dewar. These data have been analyzed by using signal processing techniques, primarily an adaptive noise canceller, to reduce noise in the gradiometer data. Results shown here indicate that the environmental noise and/or the dewar motion noise can be reduced as much as 40 dB in noise power. It is also shown, in the case of poor signal-to-noise ratio (S/N∼1), that a 10 Hz sine wave can be extracted. This technique not only shows promise for noise reduction, but also aids in the identification of noise signals which might be misconstrued as part of the evoked response. Analysis of data containing evoked response is now underway.

    Notes: Summary The instrumentation for biomagnetic applications is described. The small size and weight of the SQUID gradiometer permit easy transport and positioning of the instrument, thus making biomagnetic measurements possible in any noise-free location. The equipment is provided with a battery power supply, and, when filled with 60 cm3 of liquid helium, will operate continuously for 6 h. The device is suitable for magnetocardiography.

Si riesaminano il principio di misurazione e i lavori recenti di altri ricercatori. Si studia l’effetto del movimento legato al cuore della cassa toracica anteriore sul segnale MSPG.I risultati suggeriscono che il movimento del confine cassa toracica-aria può contribuire in modo significativo (∼75%) al segnale MSPG quando è registrato vicino al cuore.
    Notes: Summary Magnetic-susceptibility plethysmography (MSPG) is a new method proposed for monitoring cardiac-volume changes. The measurement principle and recent work by other investigators are reviewed. The effect of the cardiac-related motion of the anterior chest wall on the MSPG signal is studied. The results suggest that the motion of the chest-wall-air boundary may contribute significantly (∼75%) to the MSPG signal when recorded near the heart.

    Notes: Summary Sloping signals were observed in thePR segment of normal magnetocardiograms recorded from five dogs. Larger ramps were observed after first-degree AV nodal blockade was induced pharmacologically, and complete atrial repolarization was observed following isolated atrial contractions in two animals with second-degree block. These ramps, ranging up to 6.7 pT in size and arising from the atria, may complicate magnetic detection of His-Purkinje activity.

    Description / Table of Contents: Riassunto La motivazione fondamentale per lo studio dei fenomeni elettrofisiologici cardiaci tramite il metodo biomagnetico è dovuta al fatto che questa tecnica offre un’alta risoluzione spaziale e sembra fornire informazioni complementari rispetto alle sole registrazioni elettriche. Fino ad oggi l’interesse degli studiosi si è concentrato su due differenti fenomeni cardiaci: 1) lo studio dell’attività del sistema di conduzione atrioventricolare e 2) lo studio della depolarizzazione ventricolare anormalmente ritardata (late potentials). L’interesse clinico per lo studio non invasivo del sistema di His-Purkinje (HPS) è ben noto. Registrazioni cliniche non invasive di late potentials sono particolarmente desiderabili in quanto possono essere preziosi indicatori di rischio di morte improvvisa. In questo lavoro si presenta una rassegna sullo stato delle ricerche in entrambi i campi d’indagine.
    Notes: Summary The fundamental motivation for investigating cardiac electrophysiological phenomena by means of the biomagnetic method has been that this technique should have added spatial resolution and provided complementary information as opposed to electric recordings only. So far the interest of experimentalists has been concentrated on two different kinds of cardiac phenomena: 1) the study of the activity of the cardiac conduction system and 2) of abnormally delayed ventricular depolarization (late potentials). The clinical interest for the noninvasive investigation of the His-Purkinje system (HPS) is well known. Noninvasive clinical recording of late potentials is particularly desirable as they seem to be precious markers for risk of sudden death. A review of the state of research in both fields is presented in this paper.

Allo scopo di risolvere il problema di quanta parte dei segnali misurati sia attribuibile al sistema di His-Purkinje e quanta piuttosto alla ripolarizzazione atriale, è stata recentemente sviluppata una strumentazione appositamente disegnata per misure sul HPS e con una sensibilità di $$20fT/\sqrt {Hz}$$ nella banda di frequenze interessanti ((0.5÷300) Hz). Al contempo si è altresí sviluppato un nuovo modello teorico in stretta corrispondenza con le caratteristiche anatomiche del HPS che fornisce una distribuzione di campo magnetico sul torace in buon accordo qualitativo con i risultati sperimentali.
    Notes: Summary Since 1980, systematic investigations of the activity of the His-Purkinje system (HPS) by the biomagnetic method have been reported on both normal subjects and patients affected by different kinds of conduction disturbances. A first attempt to interpret the experimental results by means of a theoretical model satisfactorily accounted for the measured «ramp» patterns. Moreover, a good temporal coincidence was observed between magnetic signals and invasively recorded His bundle electrograms (HBE). An alternative explanation of the «ramp» pattern, in terms of atrial repolarization, has been recently proposed on the basis of animal experimental investigation. In order to try to solve this problem, we have carried out a twofold approach: a new second-order gradiometer, specifically designed for HPS activity investigation has been successfully set up and tested; it provides a sensitivity of $$20fT/\sqrt {Hz}$$ in the band width of interest ((0.5÷300) Hz). Second, a new model based on a closer correspondence to anatomical features of the HPS has been developed. This model provides well-defined patterns which are in quite good agreement with experimental results.

    Notes: Summary In this paper we present some tentative model considerations, based on the concept of an equivalent current dipole to account for observed auditory evoked responses, with the aim of attempting a comparison of variations in amplitude and latency with calculations based on Rall’s greatly simplified nerve model. A phenomenological analysis of experimental data on the late, auditory evoked magnetic field from the human auditory cortex is presented. The emphasis is here placed on relations between signal properties (e.g. amplitude and latency) and between signal properties and stimulus properties (e.g. amplitude/latency and relative frequency glide Δf/f for a frequency glide stimulus) rather than on questions related to the location of the equivalent current dipole,e.g. the tonotopic organization discussed by Elberlinget al. and by Romaniet al.. Some results of a numerical calculation based on a linear-cable model are presented and compared with experimental results.

Gli stimoli erano impulsi sonori di 1 kHz, lunghi 100 ms con un intervallo modale tra stimoli di 700 ms emessi a livelli di pressione sonora di 40, 60, 80 e 100 dB. Si è trovato che l’ampiezza AEF è correlata all’intensità dello stimolo in modo quadratico, l’ampiezza AEP in modo, lineare. Gli AEF erano di ampiezza maggiore in risposta alla stimolazione contralaterale che in risposta alla stimolazione ipsilaterale. Gli AEP non presentano questa relazione. In un secondo esperimento gli AEF e AEP dell’emisfero destro in risposta al tono di stimolazione nell’orecchio controlaterale in questi 12 soggetti sono stati combinati con dati simili precedenti su 24 soggetti, ottenendo cosí un totale di 36 soggetti, per esaminare la comparabilità della forma dell’onadaP50 di AEP e dell’analogaP50 di AEF. Si è trovato che la latenza dellaP50 decrese in funzione dell’intensità crescente dello stimolo sia per AEF che per AEP, e che la latenza diP50 è decisamente inferiore nelle registrazioni magnetiche rispetto alle registrazioni del potenziale.
    Notes: Summary Auditory evoked magnetic fields (AEFs) and EEG auditory evoked potentials (AEPs) were recorded from left and right auditory cortical regions of 12 normal adult subjects. The magnetic sensor was a figure-eight SQUID gradiometer with a 4 cm baseline oriented so as to be maximally sensitive to a current dipole oriented normal to the Sylvian fissure. Stimuli were 100 ms long 1 kHz tone pips with a modal interstimulus interval of 700 ms delivered at sound pressure levels of 40, 60, 80 and 100 dB. AEF amplitude was found to be related to stimulus intensity in a quadratic fashion, AEP amplitude in a linear fashion. AEFs were of larger amplitude in response to contralateral as compared to ipsilateral stimulation. AEPs did not exhibit such a relationship. In a second experiment right-hemisphere AEFs and AEPs in response to contralateral ear tone stimulation in these 12 subjects were combined with similar previous data from 24 subjects, providing a total of 36 subjects, to examine the comparability of the AEPP50 wave form and the AEFP50 analog. The latency of theP50 was found to decrease as a function of increasing stimulus intensity for both AEFs and AEPs, and theP50 latency was consistently shorter in magnetic compared to potential recordings.

Le mappe dell’ampiezza media dei segnali del ritmo α hanno mostrato 2 massimi dell’attività su 2 regioni simmetriche dello scalpo. Comunque, malgrado una generale simmetria fra gli emisferi, la complessità delle mappe misurate suggerisce una corrispondente complessità dei generatori responsabili dell’attività α.
    Notes: Summary An investigation of the spontaneous brain activity from normal subjects has been carried out by means of the neuromagnetic method. Two different systems were used for magnetic measurements. Both used 2nd-order gradiometers specifically designed for brain studies: 2.9 cm diameter, 5 cm baseline. A r.f. SQUID and a d.c. SQUID were coupled to the gradiometers. The measured noise levels are $$40fT/\sqrt {Hz}$$ and $$25fT/\sqrt {Hz}$$ , respectively. Thorough magnetic mapping of the spontaneous activity was performed on a few subjects. By mapping the average amplitude of the α-rhythm signals two maxima of activity over two symmetric regions of the scalp were clearly observed. Despite a general symmetry between hemispheres, however, the complexity of the measured maps suggests a corresponding complexity of the generators responsible for α-activity.

Si conoscono pochissimi contributi in questo campo. A parte il lavoro del gruppo di biomagnetismo di Roma, il solo contributo clinico finora è stato quello di Hughes e collaboratori che era abbastanza limitato nel numero e nella scelta dei soggetti studiati. La nostra esperienza condotta fin dal 1980 su un gran numero di casi (principalmente epilessie focali) mostra che il MEG può evidenziare attività significative anche se non sono presenti anormalità EEG; esso ha inoltre una risoluzione spaziale maggiore rispetto all’EEG. I foci epilettici mostrano un’area limitata di attività massima. Finora non è stata osservata nessuna evidenza sperimentale di una differente distribuzione delle ampiezze del campo sullo scalpo. La risoluzione spaziale del MEG riguarda anche la possibilità di risolvere la morfologia dei segnali EEG (per esempio onde aguzze EEG risolte in gruppi di spikes dal MEG) o di separare topograficamente le anormalità EEG mescolate e non ben localizzate (per esempio, attività delta ed epilettiche mescolate nell’EEG e discriminate nel MEG). I migliori risultati si osservano quando la localizzazione è nella corteccia esterna. Dopo una descrizione del materiale studiato, si sottolinea l’importanza dei risultati ottenuti per la prima volta in casi di epilessie focali.
    Notes: Summary So far spontaneous magnetoencephalography (MEG) in normal and pathological subjects has not been deeply investigated. Very few contributions in this field are available. Aside from the work of the Rome biomagnetic group in this field, the only clinical contribution up to now has been that of Hughes and co-workers which was rather limited in the number and in the selection of subjects investigated. Our experience, carried out since 1980 on a large number of cases—mainly focal epilepsies— shows that the MEG may evidence significant activities even if EEG abnormalities are not present and has a higher spatial resolution with respect to the EEG. Epileptic foci show a limited area of maximal activity. So far no experimental evidence of a different distribution of field amplitude over the scalp has been observed. The spatial resolution of the MEG also regards the possibility of resolving the morphology of EEG signals (for instance, EEG sharp waves resolved into groups of spikes by MEG) or topographically separate EEG abnormalities mixed up and not well localized (for instance, delta and epileptic activities mixed up in the EEG, but differently in the MEG). The best results are observed when the localization is in the outer cortex. After a description of the material studied, the importance of the results obtained for the first time in cases of focal epilepsies is outlined.

La presente versione del nostro apparato sperimentale permette di effettuare misure MEG con un solo sensore magnetico, impedendo la rilevazione simultanea di attività MEG su punti differenti della testa. Ciò ha condotto gli autori a concentrarsi sulle epilessie focali. Fin dalle prime misure il sensore magnetico ha dimostrato un notevole potere localizzante, rafforzando l’interesse per la localizzazione in casi patologici. In questo lavoro si riportano risultati che sembrano confermare la straordìnaria capacità delle misure MEG nel localizzare sorgenti di attività cerebrali patologiche in casi di epilessie focali.
    Notes: Summary During the last year the authors have been carrying out a systematic investigation on a large number of pathological subjects simultaneously recording the EEG and the MEG. The present version of our experimental apparatus allows MEG measurements with only one magnetic detector, thus preventing the detection of simultaneous MEG activities over different points of the head. This has led the authors to concentrate upon focal epilepsies. Since the first measurements a remarkable localizing power was shown by the magnetic detector, thus strengthening the interest in pathological localization. In this contribution we report results which seem to confirm the extraordinary capability of MEG measurements to localize sources of pathological brain activities in cases of focal epilepsies.

    Notes: Summary In order to evaluate a possible role for tissue phosphate or phosphorylated compounds in mediating the increase in plasma 1,25-(OH)2-D3 levels during dietary phosphate deprivation, measurements of total and acid-soluble renal cortical phosphate content have been made in both intact and hypophysectomized (hypox) rats eating a normal diet and also after four days of dietary phosphate deprivation. Similar measurements were also made in phosphate-deprived hypophysectomized rats replaced with growth hormone (GH). Total and acid-soluble renal cortical phosphate content averaged 81±8 µmol/g and 4.1±0.6 µmol/g, respectively, in intact rats eating the normal diet and were not significantly altered after phosphate deprivation despite a fall in plasma phosphate of about 40% and a fourfold increase in plasma 1,25-(OH)2-D3 levels. Total and acid-soluble renal cortical phosphate content levels were higher in hypox rats, averaging 92±8 µmol/g and 4.9±0.7 µmol/g, respectively, but also did not change after phosphate deprivation. Replacement of phosphate-deprived hypox rats with GH resulted in a further fall in plasma phosphate and a significant increase in plasma 1,25-(OH)2-D3 levels, but there was no change in either total or acid-soluble renal cortical phosphate content. The distribution of organophosphorus compounds in the acid-soluble phosphate fraction in these experiments was also evaluated using31P NMR spectroscopy. Although there appeared to be an increase in the total concentration of organophosphorus compounds after phosphate deprivation, this effect was not altered by hypophysectomy or by replacement of phosphate-deprived hypox rats with GH. These data suggest that unless phosphate deprivation affects only a small or specific cellular phosphate pool, some factor other than renal cell inorganic phosphate content must initiate the increase in renal 1,25-(OH)2-D3 synthesis that occurs during phosphate deprivation.

    Notes: Summary Hypophysectomy abolishes the four- to fivefold increase in plasma 1,25(OH)2D levels that normally accompanies dietary phosphate deprivation in rats despite a smaller but significant decrease in plasma phosphate in these animals. This effect appears within 1 week of hypophysectomy and may be the result of a lack of GH, T3, or some other pituitary hormone. In hypothyroid rats (2 weeks after TPTX) not given replacement T3, plasma 1,25(OH)2D levels rose threefold from 148±57 pmol/l to 402±96 pmol/l (mean±SD) after 4 days of dietary phosphate deprivation. However, in hypophysectomized animals given replacement T3 alone, plasma 1,25(OH)2D levels rose fourfold from 82±13 to 333±230 pmol/l after 4 days of phosphate deprivation. In addition, in hypophysectomized animals replaced with GH alone, plasma 1,25(OH)2D levels rose from 243±86 to 525±85 pmol/l during phosphate deprivation. These results would suggest that both GH and T3 must be absent to prevent enhanced renal 1,25(OH)2D synthesis during phosphate deprivation. GH and T3 appear to play a permissive role since plasma levels of these hormones do not increase when intact rats are deprived of phosphate. Furthermore, bioassayable somatomedin levels are also not increased in intact rats during phosphate deprivation as well as plasma levels of prolactin. As observed previously, plasma 1,25(OH)2D levels were inversely correlated to plasma phosphate concentrations (r=0.46,P〈0.025), despite the inclusion of data points for unreplaced hypophysectomized animals who were hypophosphatemic but showed no increase in plasma 1,25(OH)2D. Thus the possibility remains that GH and T3 may exert their effect by permitting the renal 25OHD-1α-hydroxylase to respond to a change in phosphate concentrations during dietary phosphate deprivation, that, in turn, may ultimately increase renal 1,25(OH)2D synthesis and plasma levels of this hormone.

Two nonparametric methods and their adaptations to bioavailability ratios are reviewed, one based on Wilcoxon's signed rank test (Tukey), and the other on Pitman's permutation test. The necessary assumptions and the merits of these procedures are discussed. The methods are illustrated by an example of a comparative bioavailability study.     Notes: Summary The relative bioavailability and diuretic effect of 2 commercially available tablet preparations of furosemide 40 mg was examined in 10 healthy male volunteers. A close linear relationship between the urinary excretion rate of furosemide and the rate of sodium ion excretion in urine and/or flow rate of urine was demonstrated. There were no significant differences in the urinary excretion of furosemide, sodium and potassium ions or urinary volume following the oral doses. The difference in drug content affected the urinary recovery of furosemide over 24 h but had no effect on the pharmacological response. The analytical power of ANOVA using the various parameters of the responses to furosemide was no lower than when the parameters of urinary excretion of furosemide were used.

TA and OH-TA-ester were determined by densitometric measurement of native fluorescence after thin layer chromatography. Distribution volumes of the central compartment of TA and OH-TA-ester were 1.49 l/kg and 0.11 l/kg, respectively. Terminal half-lives were 255 min for TA and 188 min for OH-TA-ester after i.v. administration. For TA total plasma clearance was 4.5 l/min and renal plasma clearance 0.22 l/kg. The formation of OH-TA-ester was very rapid and the concentration of the metabolite exceeded that of TA at all times. After i.v. administration the urinary recovery of TA and OH-TA-ester was 4.4% and 50.9%, respectively. The bioavailability of TA was 52%, corresponding to absorption of 83%. TA is partly eliminated by a first-pass-effect. The main metabolite of TA is OH-TA-ester, which is pharmacologically active.

Within 24 h, respectively 0.4 and 0.6 mg, canrenone were excreted by the kidney after intravenous and oral administration. The half-life of elimination was 4.9±1.8 h (i.v.) and 3.9±1.2 h (p.o.).

    Notes: Summary The pharmacokinetics and oral biovailability of dexamethasone were studied in 6 patients with neurological disease being treated with high dosages of the drug. A specific high performance liquid chromatographic assay was used to measure dexamethasone concentrations. Unlike the previously published mean figure of 0.78 for the oral bioavailability of the drug given in single doses to healthy volunteers, the mean bioavailability of dexamethasone in the patients studied was 0.53±SD 0.40. It appeared more likely that this incomplete bioavailability was due to presystemic elimination than to poor absorption. The intravenous clearance of the drug was relatively high (0.4902±SD 2291 l kg−1, approximately 65% of expected hepatic plasma flow), the oral clearance higher (2.5804±SD 3.2181 l kg−1 h−1) while the absorption rate constant (4.8729±8.4998 h−1), suggested rapid absorption after oral administration. Prior phenytoin and possibly prior dexamethasone therapy is likely to have contributed to the higher clearance values of the drug in these patients than the values reported in healthy volunteers after single dose studies.

    Notes: Summary Methods for estimating the bioavailability of drugs with long elimination half-lives are examined. Provided both absorption and disposition are linear a simple linear regression method is developed which can be used to calculate bioavailability in situations where only an incomplete estimate of the area under the curve (AUC) is available. The regression method and the traditional method of comparing the AUC following an oral dose to the AUC following an i.v. dose were applied to simulated data. It was found that the AUC ratio method works well as long as absorption is complete within the time over which the AUC is computed. The regression method is less precise than the AUC ratio method but is more accurate for drugs with long absorption half-lives. When applied to published data on a beta blocker the two methods produced comparable results. The bioavailability of amiodarone in three human subjects was calculated to be 0.20, 0.44 and 0.98 using the regression method with similar results from the ratio method. It is not possible to estimate the clearance of amiodarone from single dose data.

The following differences in plasma indomethacin profiles after the 2 treatments were observed: average peak concentrations (Cmax) for the CR regimen were higher and the time to peak (Tmax) was significantly delayed. Trough (pre-morning dose) plasma concentrations (Cmin) on Days 2, 5, 6 and 7 were significantly lower after the CR-formulation. No statistically significant differences between preparations for area under the plasma concentration time curve (AUC0–24h) or for renal clearance were observed. Average steady-state plasma concentrations (C p ss ) on Day 7 of the multiple dose regimens averaged 0.477 and 0.427 µg/ml for the 75 mg CR once daily and the conventional 25 mg t.i.d. treatments, respectively. These results show that the bioavailability of the CR and conventional indomethacin formulations under these multiple-dose conditions was not significantly different.

There was no accumulation of morphine (20–40 mg) when repeatedly administered every 4 to 6 h. The mean steady-state concentration of morphine during the dose interval varied between 5.9 and 68.4 ng/ml (20.7–240 nmol/l), and was linearly related to the daily dose of morphine. There were no significant differences between the tablets and the solution of morphine with regard to relative oral bioavailability or peak concentration. The time-to-maximum plasma concentrations was significantly longer for the tablets. The pain score profile, assessed by a visual analogue scale during a dose interval, showed a similar pattern after the two oral formulations of morphine. No significant linear relationship between the scores and the plasma concentrations of morphine was observed.

Large interindividual differences in the bioavailability of metoprolol were observed in both groups. However, there was no significant difference in AUC, peak plasma concentration or elimination half-life between young and elderly, but time to peak concentration was significantly longer in the elderly. Pretreatment with metoclopramide had no effect on AUC but caused significant increases in peak concentration and decreases in time to peak concentration in both groups. Probanthine pretreatment (only to the young) resulted in a significant decrease in peak concentration of metoprolol and a significant increase in time to peak concentration but had no effect on the AUC. These results suggest that alterations in gastric emptying and gut motility due to ageing or other drugs have no effect on the overall availability of metoprolol to the systemic circulation but may have significant effects on the time to peak plasma concentration and peak concentration achieved after a single oral dose.

    Notes: Summary Serum concentrations of dihydrocodeine and its acid metabolites have been determined in seven human volunteers (6 male) who received the drug orally (30 mg and 60 mg) and intravenously (30 mg) on separate occasions, and in twenty-four patients (12 male) receiving 25 mg or 50 mg of the drug intravenously. The concentrations were estimated by radioimmunoassay on reconstituted extracts from serum after an extraction process which effectively separates dihydrocodeine from its polar acidic metabolites. The intravenous data show that dihydrocodeine kinetics followed a two-compartment distribution model. The concentration curves after oral administration indicated relatively rapid absorption with mean peak concentrations at 1.6h–1.8h. The mean half-lives varied between 3.3h–4.5h. From the AUC, the mean bioavailability of orally administered drug was 21% (range 12–34%). The peak levels of the acidic metabolites occurred between 1.8h–2.0h after oral administration and 2.2h–2.5h after i.v. administration, and they were significantly greater after oral administration. The low bioavailability of dihydrocodeine, together with the earlier and higher plasma levels of the acid metabolites after oral administration is suggestive of substantial first-pass metabolism.

In the present study the pharmacokinetics of oral hydralazine has been evaluated in 7 patients with severe, chronic heart failure, using 8 compensated hypertensives as controls. The pharmacokinetics was evaluated by measuring the plasma concentrations of hydralazine (“apparent” and “real” hydralazine) and hydralazine pyruvate hydrazone, and by assessing acetylator phenotype after a small dose of dapsone. The AUC (area under the plasma concentration curve) following a single, oral 50 mg dose was significantly larger in patients with chronic heart failure NYHA Class III–IV than in patients with essential hypertension without cardiac decompensation. A decreased rate of hepatic elimination of hydralazine is suggested as a major contributory factor to this finding.

    Notes: Summary Endralazine (E), a new hydralazine-like antihypertensive was given intravenously (0.05 mg/kg) to 10 normal volunteers (5 slow and 5 fast acetylators). Plasma levels were fitted to a 3 compartment model and pharmacokinetic parameters (area under curve [AUC 0 ∞ ], clearance, volume of distribution, half-lives) obtained in the usual way. Bioavailabilities of 5 and 10 mg oral doses of E were determined from the AUC 0 ∞ generated in a previous study of oral E given to the same subjects. E had high system bioavailability (73.5–99.1%) suggesting that it was almost completely absorbed without undergoing appreciable first-pass metabolism. Furthermore, dose size and acetylator phenotype did not significantly affect the bioavailability of E. This behaviour contrasts with that of hydralazine where systemic bioavailability was 〈40%, and 2 to 3 times higher in slow acetylators than in fast acetylators. It is concluded that the bioavailability of E is high and not influenced by acetylator phenotype; these properties suggest some clinical advantages for the drug.

    Notes: Summary The absolute oral bioavailability of a sustained release theophylline tablet (Nuelin-SR250), given 12 hourly was determined in 14 asthmatic children aged 5 to 13 years. In 4 of the patients, mean bioavailability of the fourth dose was 38.9±8.4% and that of the sixth dose was 67.9±25.9% (p〈0.05) in the other ten patients. This suggests steady-state had not been achieved after four doses. In the initial study with 9 patients, a significant diurnal variation in predose plasma theophylline concentrations was observed, as the mean morning predose concentrations were 2.9 fold greater than the mean evening predose concentrations (p〈0.005). Dual peak plasma concentrations occurred in 5 out of the 9 patients. The mechanism of this diurnal variation was investigated in a further 5 asthmatic children (10.8 years ±1.6). Morning and night steady-state plasma theophylline concentrations during a continuous intravenous infusion of aminophylline were not different (14.9±5.3 mg/l vs. 15.6±5.9 mg/l), demonstrating that there was no diurnal variation in the plasma clearance of theophylline. The diurnal variation in predose concentrations with Neulin-SR250 was confirmed with the morning concentrations again being 2.6 fold greater than those in the evening. However, bioavailability was not significantly different for day (09.00–21.00) and night (21.00–09.00) dosing intervals after doses 6 and 7 respectively of Nuelin-SR250. The plasma concentration versus time profiles suggested that the diurnal variation in predose concentrations was due to slower absorption of the evening dose.

    Notes: Summary The kinetics of cibenzoline (UP 339.01), a new antiarrhythmic drug, was studied after i.v. and oral administration to 5 healthy subjects. Cibenzoline levels in plasma and urine cibenzoline were measured by a GLC method. After i.v. administration, the total clearance was 826 ml · min−1. The fraction of cibenzoline excreted unchanged in the urine was 0.602 and it was correlated with the creatinine clearance. After i.v. and oral administration, the renal clearances were 499 ml · min−1 and 439 ml · min−1, and the half-lives were 4 h 01 min and 3 h 24 min, respectively. The differences were not significant. Availability by the oral route was 0.92, the maximum plasma concentration being observed at 1 h 36 min. The results were compared with those for other antiarrhythmic drugs.

    Notes: Summary The pharmacokinetics and bioavailability of 3 oral dyphylline preparations, solution (S), regular (R) and sustained release (SR), were studied in 8 healthy subjects (mean age 25 years). A single dose of each preparation, 20 mg·kg−1, was given at one week intervals and multiple serum samples obtained over 24 h. Drug levels were measured by high performance liquid chromatography. No adverse effects were found. The dyphylline half-life for the solution was 2.16±0.18 h and for the tablet 2.59±0.56 h. The mean clearance rate for S was 13.6±1.7 h−1 and volume of distribution 43.0±3.91. Peak concentration (Cmax, µg·ml−1), time of peak (Tmax, h), area under the curve (AUC, µg·ml−1·h) and relative bioavailability (RB, %), were determined for three preparations: The data confirm the short half-life of dyphylline, demonstrate a lack of toxicity for the 20 mg·kg−1 dose and establish bioequivalence for the products studied.

The study was carried out in six healthy subjects who received either i.v. or oral pindolol in random order.     Notes: Summary Using considerably improved analytical methods, the kinetics and effects of clonidine were observed in healthy volunteers over periods of time more than 3 times longer than those previously reported. The high sensitivity and small work load of the newly developed method permitted the performance of low-dose and multipledose trials. 1. The complete bioavailability of clonidine and its elimination half-life (20 to 25.5 h) remained constant after single and multiple doses. 2. Approximately 62% of a given dose was excreted unchanged in the urine, independent of the quantity administered (0.075, 0.15, 0.2, 0.25 or 0.3 mg), the drug formulation (solution, tablet, Perlonget) or of the mode of administration (i.v., p.o.; single or multiple doses). 3. As the pharmacokinetics of the drug were affected by entero-hepatic circulation, it cannot be described by a conventional, open one or two compartment model. 4. The time courses of the plasma clonidine concentration and its drug effects ran asynchronously. 5. On cessation of chronic clonidine administration, blood pressure and plasma catecholamine levels increased to pretreatment levels without exhibiting any “overshoot” reaction.

    Notes: Summary The pharmacokinetics and bioavailability of theophylline following 10 days of multiple doses of a plain uncoated (640 mg, q.i.d.) and a sustained-release tablet formulation (600 mg, b.i.d.) were related to habitual smoking in 11 healthy adult male volunteers, who had previously taken part in a single-dose study of an intravenous preparation of theophylline and of the same oral dosage form. There were significant differences (p〈0.05 to 0.01) in the steady-state mean and minimum theophylline concentration and AUC between the groups (6 smokers versus 5 nonsmokers), but not between other variables. A difference (p〈0.05) in peak time was also found between the dosage forms. The mean elimination t1/2 was significantly (p〈0.05) shorter in smokers than in nonsmokers. The intersubject variability in plasma theophylline concentration observed on the final trial day in the smoking group was larger and diverged more from simulation curves generated from the mean pharmacokinetic parameters of the single-dose study of the same formulations as compared to that of the nonsmoking group. There was no significant difference between the two groups in the mean accumulation ratio and absolute bioavailability of the two dosage forms. The mean morning (7 a.m.) trough theophylline concentrations after both formulations were significantly (p〈0.05 to 0.01) greater than the evening (7 p.m.) values within the same group. The average number of reported side-effects was significantly (p〈0.001) greater during the earlier period (Days 1 to 3) than the later period of the trial. A trend was observed suggesting that the incidence of side-effects was less in smokers than in nonsmokers. The results indicate that smoking is a determinant not only of enhanced elimination of theophylline but that it also produces more variability in the plasma level, irrespective of the dosage form administered or the dosing scheme employed. There may be circadian variation in theophylline kinetics.

Dose dependent pharmacokinetics were found, the VDss and Clt being significantly larger (p〈0.01) after 20 mg i.v. than after 10 mg i.v. The protein binding of prednisolone in all subjects was non-linear, and is the most likely cause of the dose dependent pharmacokinetics, as there was no dose dependent variation in elimination half-time.

    Notes: Summary The pharmacokinetics of orally administered ranitidine were studied in 17 male patients with chronic duodenal ulceration. The patients were divided into 2 groups, 10 responders and 7 nonresponders, on the basis of their endoscopic response to ranitidine treatment. The 10 responders were studied both after a single 150 mg dose (SD) and after multiple dosing (MD) with ranitidine 150 mg twice daily for 4 weeks. The area under the curve (AUC) and maximum concentration (Cmax) were significantly higher (p〈0.01 andp〈0.05, respectively) after MD than after SD, but the half-life (t1/2) and minimum concentration (Cmin) 12 h postdosing did not differ. The non-responders were studied after MD only and their pharmacokinetic characteristics were compared with those of responders. No differences between the 2 groups were found. However, 2 non-responders had particularly low plasma ranitidine levels and high acid output. Such patients may need larger doses of ranitidine for adequate suppression of gastric acid. Five patients (4 responders and 1 non-responder) received ranitidine 20 mg i.v. The drug followed a two-compartment model, with mean values for t1/2β, volume of distribution steady-state and total plasma clearance of 80 min, 701 and 680 ml/min, respectively. The oral bioavailability of ranitidine in these 5 patients showed wide variation (27–76%; mean 51%).

    Notes: Summary The bioavailability of theophylline after oral administration of a new sustained release tablet Theograd®-250 mg was studied in 7 healthy volunteers, under fasting and non-fasting conditions. Whilst fasting the bioavailability was moderate at 64±22% (mean±SD), whereas in the non-fasting state the relatively high bioavailability of 90±13% was found. The drug appeared to be significantly more slowly absorbed when a tablet was taken after a meal, than when it was ingested on an empty stomach. In the former case, the peak level was reached after 6.9±1.0 h, whereas in the fasting state the maximum serum concentration occurred 4.0±1.7 h after administration of the drug. Despite the slow absorption, the peak non-fasting level of 4.4±1.4 mg·l−1 was significantly higher than the 3.1±1.0 mg·l−1 observed in the fasting state. The profiles of the serum concentration-time curves showed that the concentration remained above 75% of Cmax for 8.7±1.3 h in the fasting and 9.0±1.1 h in the non-fasting state. It was concluded that to define the optimal dosage regime for sustained release oral dosage forms of theophylline, the influence of food on absorption from these preparations should be taken into account. Based on the present results, Theograd®-250 mg tablets have predictable absorption and a high (90%) bioavailability if taken after a meal.

    Notes: Summary In four healthy subjects the intestinal absorption of levodopa (l-dopa) was investigated by measuring the plasma concentration of the amino acid following the administration of l-dopa at three different sites in the small intestine. In order to minimize presystemic clearance of l-dopa, the subjects were pretreated with the peripheral decarboxylase inhibitor benserazide 3×50 mg every 8 h on the previous day and 1×50 mg 2 h prior to administration of the l-dopa. L-dopa 100 mg dissolved in 0.05 N HCl and 50 mg benserazide dissolved in 0.05 N HCl were coadministered. Under these conditions no difference in tmax, cmax or AUC of l-dopa was observed between administration of the drug into the proximal or the distal part of duodenum, or into the upper part of jejunum. The results indicate that in healthy subjects, during inhibition of peripheral decarboxylase, the rate and extent of l-dopa absorption does not differ at any site in the upper small intestine.

    Notes: Abstract Ethanol, methanol, acetone, and acetaldehyde—chemicals identified in the inner bark of living trees—were used to bait vane traps placed in crowns of oak trees in Connecticut. Ethanol-baited traps caught more cerambycid, scolytid, and clerid beetles than unbaited traps. Buprestidae were not attracted to ethanol. Acetaldehyde and acetone were not attractive to any family. A mixture of ethanol, methanol, and acetaldehyde was no more attractive than ethanol alone. The vane traps were very effective at catching Cerambycidae and Scolytidae, but ineffective compared to sticky panels at catching Buprestidae.

    Notes: Abstract Antennae of male and femaleIps paraconfusus were equally and highly sensitive to their male-produced, multicomponent aggregation pheromone. Female and male antennae were highly sensitive to the pheromonal component, (S)-(−)-ipsenol, but essentially insensitive to its antipode, (R)-(+)-ipsenol. Further, female and male antennae were more sensitive to the pheromonal component, (S)-cis-verbenol, than to its antipode, (R)-cis-verbenol. Dramatic sexual dimorphism in chiral sensitivity to the ipsdienol enantiortiers was found, with female antennae being more sensitive to the conspecific pheromonal enantiomer, (S)-(+)-ipsdienol, and male antennae being more sensitive to the antipode, (R)-(−)-ipsdienol. Since (R)-(−)-ipsdienol is the principal pheromone of CaliforniaIps pini and interruptsI. paraconfusus aggregation, male antennae appear to be more sensitive to an interspecific allomone than a conspecific pheromone. Of the conspecific pheromonal enantiomers, both male and female antennae were most sensitive to (S)-(+)-ipsdienol, intermediately sensitive to (S)-(−)-ipsenol, and least sensitive to (S)-cis-verbenol. However, when enantiomeric sensitivities were compared to the estimated concentrations of these components in the natural pheromone, (S)-(~)-ipsenol tended to equal or approach the potency of (S)-(+)-ipsdienol as an antennal stimulant, while antennal responsiveness to (S)-cis-verbenol was dramatically less than for the other two pheromonal components. The behavioral implications of such physiological sensitivities are discussed in regard to perception of multicomponent synergistic pheromones and the relative efficacy of each component as an orientation cue.

    Notes: Abstract The antennal sensitivities of both male and femaleIps paraconfusus were found generally to be greatest for conspecific aggregation pheromones (ipsdienol, ipsenol); intermediate for an additional conspecific pheromone (cis-verbenol), an aggregation synergist (2-phenylethanol), and pheromones/allomones of sympatric species (trans-verbenol, verbenone, and frontalin); and lowest for both host terpenes (alpha-pinene and myrcene) and other bark beetle-produced odorants (exo-brevicomin and linalool). Of the enantiomeric compounds tested, antennae of both sexes did not differ in sensitivity between thetrans-verbenol enantiomers at low dosage levels; but at higher dosages, the conspecific-produced enantiomer, (1R,4S,5R)-(+)-trans-verbenol, elicited larger mean EAG responses than its antipode, (1S,4R, 5S)-(−)-trans-verbenol. At the mid-dosage range, female antennae tended to be slightly more responsive to (S)-(−)-verbenone than to (R)-(+)-verbenone, while male antennae were equally responsive to stimulations by either verbenone enantiomer. In field bioassays there was a large and significant reduction in trap catches ofI. paraconfusus on traps where the (S)-(−)- or (R)-(+)-enantiomers of verbenone were evaporated beside logs containing boring conspecific males. Only when the (S)-(−)-enantiomer of verbenone was evaporated beside logs containing boring males did the sex ratio ofI. paraconfusus trapped shift from female-dominated to male-dominated attraction. Thus both physiological and behavioral data suggest a differential chiral sensitivity of female beetles for the verbenone enantiomers. The relative sensitivities between different chiral compounds derived from one or the other of the common precursoral host terpenes, (S)-(−)- and (R)-(+)alpha-pinene or myrcene, are discussed.

    Notes: Abstract At high concentrations, the defensive tergal gland secretion (TGS)3 of the staphylinid beetle,Aleochara curtula, inhibits the male copulatory response (grasping with parameres). Inhibitory chemicals, for which a function as alarm substances is assumed, aren-undecane, 1-undecene,n-dodecanal, toluquinone, and 2-methoxy-3-methyl-1,4-benzoquinone. When emitted in small amounts, however, the TGS releases the male grasping response. The main components with aphrodisiac effect are (Z)-4-tridecene,n-dodecanal, and (Z)-5-tetradecenal. These supplementary mating stimulants, which are not sex specific, work synergistically with the aphrodisiac female sex pheromone from thé epicuticular lipids and are discussed as alerting pheromones of short-term effect. Antennal movements of resting males as an indication of the recognition of a female and the approach to the mate are released at somewhat longer distances, when the TGS is additionally present.

    Notes: Abstract The field responses of English populations of the Dutch elm disease vectors,Scolytus multistriatus andS. scolytus to baits containing 4-methyl-3-heptanol, a host synergist [(−)-α-cubebene or (−)-limonene] and (±)-α-, (+)-β-, (−)-β-, (±)-γ-, or (±)-δ-multistriatin were examined. (±)-α-Multistriatin, released at 5–10 μg/day, enhanced the response ofS. multistriatus to baits containing 4-methyl-3-heptanol and either of the host synergists but had no effect on the capture ofS. scolytus. The release of larger amounts (57 or 365 μg/day) of (±)-α-multistriatin interrupted the response of both species to the 4-methyl-3-heptanol baits. It appears that α-multistriatin has multiple functions as a behavior-modifying substance for the two beetles. The (+)-β-, (−)-β-, (±)-γ-, and (±)-δ-multistriatins were inactive when released at 5–10 μg/day. The results of these field experiments suggest that one bait can be formulated to capture both species.

    Notes: Abstract Insect use of native crucifers may be related to patterns in mustard oil content. Consequently, in 1979 we measured glucosinolate content of Rocky Mountain bittercress,Cardamine cordifolia (Cruciferae), using paper and gas chromatography, in relation to: plant organ, phenology, elevation, habitat, leaf position and weight, and plant consumption by two adapted insect herbivores. Results for each are as follows. (1) The predominant constituent in all vegetative organs was 2-butylglucosinolate; concentration of isothiocyanate-yielding glucosinolates (IYG) was highest in roots (1.11 mg/gfr. wt) and lowest in stems (0.07 mg/g). (2) Concentration of IYG appeared to be higher in plants lacking oxazolidinethione-yielding glucosinolates (OYG) than in those with OYG. (3) Terminal cauline leaves had a higher content of IYG than leaves in other positions on a plant. (4) Heavy leaves had significantly higher concentrations of IYG than did lighter leaves. (5) IYG concentrations were not directly related to elevation. (6) Leaves of plants occurring naturally in the sun had concentrations of IYG similar to those of plants in the usual shaded habitat. However, experimental removal of overhanging willows caused a significant, stress-induced increase in IYG concentrations. Finally, (7) feeding by two adapted herbivores, chrysomelids and psyllids, was associated with lower, rather than higher, IYG concentrations. The results demonstrate significant variation in glucosinolate content in a native crucifer and suggest that some of this variation can be partitioned in relation to the ecological and environmental axes examined.

    Notes: Abstract Two synergistic macrolide aggregation pheromones were isolated from Porapak Q-trapped volatiles obtained from the frass ofCryptolestes ferrugineus (Stephens). These compounds were identified as (E,E)-4,8-dimethyl-4,8-decadien-10-olide (I) and (3Z,11S)-3-dodecen-11-olide (II) and given the trivial names ferrulactone I and II, respectively. Analysis of captured volatiles from separated male and female adults disclosed that the pheromones are male-produced. Additional macrolides were identified in frass volatiles, but were devoid of any pheromonal activity. The structures of I and II were confirmed by comparison with synthetic materials.

    Notes: Abstract Effects of soybean phytoalexins on the feeding of the soybean looper and Mexican bean beetle were investigated to test the hypothesis that phytoalexins might be a defense mechanism of plants against insects as well as against pathogens. Short-term behavioral responses to the phytoalexins were analyzed using dual-choice tests with phytoalexin-rich and phytoalexin-poor (control) tissues. Phytoalexin production was elicited with ultraviolet radiation. Results from the dual-choice tests indicated that 6th instar soybean looper larvae fed equally on the control and phytoalexin-rich tissues. Feeding by adult and 4th instar Mexican bean beetles, however, was strongly deterred by the phytoalexins as evidenced by “single-bite” mandible scars on the phytoalexin-rich cotyledon discs. Nutritional effects of the isoflavonoid phytoalexin glyceollin on early instar soybean looper larvae were tested by incorporating the phytoalexin into an artificial medium at a level of 1% dry weight (0.15% fresh weight). The larvae were reared for 7 days from emergence on diets of control and glyceollin-containing media. Although survival on the glyceollin diets was initially less than on the control diets, under the experimental conditions glyceollin had no significant effect on the growth, development, or subsequent survival of the larvae. Efficiency of food utilization (ECI) was reduced, indicating that the phytoalexins may be a mild digestibility-reducing factor for the loopers. Implications of the results for host-plant resistance are discussed.

    Notes: Summary The response of anther culture to culture temperature was studied in detail using many varieties, F1 hybrids and pollen-derived lines of wheat (Triticum aestivum) as materials. The suitable culture temperature for inducing pollen callus (or embryoids) in wheat anther culture ranged from 26 °C to 30 °C, varying with genotypes. But for the great majority of wheat genotypes the suitable culture temperatures lay between 28 °C and 30°C. The most significant genotypic variation in the response to culture temperature was observed in the comparison between the culture at 33 °C for eight days followed by culture at 25 °C (or 26 °C) and the continuous culture at 25 °C (or 26 °C). This genotypic variation in the response to culture temperature is a heritable character which may be controlled by multiple genes. The effect of culture at 30 °C for eight days followed by culture at 26 °C was similar to, or in some cases, better than that of continuous culture at 28 °C, and the effect of culture at 32 °C for eight days followed by culture at 28 °C was similar to that of continuous culture at 30 °C. In the range from 26 °C to 32 °C, the overwhelming majority of pollen calli emerged before the 40th day after anther inoculation, and the higher the culture temperature, the earlier and more concentrated the emerging period of the pollen callus. The pollen callus obtained at high temperatures above 28 °C should be transferred in time onto the regeneration medium at 25°–27°C to induce shoots.

F1 hybrids of T. aestivum were also produced with A. intermedium (2n=42,E1E1E2E2Z1Z1) and A. junceum (2n=14,JuJu). All wheat-Agropyron hybrids were obtained by embryo rescue technique. Cultivars and reciprocal crosses differed for seed set, seed development and F1 plant production. The F1 hybrids were sterile. Attempts to obtain amphiploids were unsuccessful. However, backcross derivatives were obtained with wheat as the recurrent parent. The level of chromosome pairing in A. trachycaulum x wheat, A. yezoense x wheat and wheat x A. junceum hybrids provided no evidence of homologous or homoeologous pairing. Mean pairing frequencies in A. ciliare x wheat, wheat x A. scirpeum and wheat x A. intermedium hybrids indicated homoeologous or autosyndetic pairing. Ph gene was more effective in regulating homoeologous pairing in A. yezoense x wheat hybrids than in A. ciliare x wheat hybrid. Chromosome pairing data of BC1 derivatives indicated that either some of the wheat chromosomes were eliminated or Agropyron chromosomes caused reduced pairing of wheat homologues.

    Notes: Summary Poor adaptability or functional quality of much germplasm used for breeding high-protein hard red winter wheats prompted mutagenesis as an alternative means of increasing grain protein content. Four hard red winter wheat genotypes — KS644 (‘Triumph// Concho/Triumph’), ‘Kaw’, ‘Parker’, and ‘Shawnee’ — were treated with 0.40 M ethyl methanesulfonate (EMS). Advanced lines (M8-M10) were selected that had a 3-year mean grain protein advantage of 0.7% to 2.0% over controls. Increased grain protein content was generally associated with decreased grain yield and kernel weight, but some high-protein mutant lines had yields or kernel weights similar to those of original genotypes. Changes in height and lodging induced by EMS were generally favorable, most mutants being shorter and lodging less than controls, but blooming date was generally delayed, a deleterious change. One line also changed from resistant to segregating for wheat soil-borne mosaic virus. Mutant lines might be utilized in cross-breeding programs, particularly if negative pleiotropic effects and linkages are absent.

    Notes: Summary Total endosperm protein subunits, extracted from the common wheat cultivar Chinese Spring and from some of its aneuploid lines, were fractionated according to their molecular weight (MW) in an improved high resolution one-dimensional sodium dodecyl sulphate (SDS) polyacrylamide gel electrophoresis (PAGE). The resolution obtained by this method and, in particular, that of the high molecular weight (HMW) glutenin and gliadin subunits approached that of a previous report in which two-dimensional fractionation system based on charge and MW was used. In the cultivar Chinese Spring, 21 discrete protein bands were resolved and the chromosomes controlling many of them were either reconfirmed, or, in some cases, established. The advantages of this high resolution SDS PAGE technique are discussed.

    Notes: Summary Each of the three genomes in hexaploid wheat controls the expression of a specific lectin in the embryo. The chromosomes which control their synthesis were determined using nullisomic-tetrasomic and inter-varietal chromosome substitution lines of ‘Chinese Spring’. All three wheat lectins were shown to be controlled by the homoeologous group 1 chromosomes. Using ditelosomic lines of ‘Chinese Spring’ the lectin genes could be localized on the long arms of chromosomes 1A and 1D. Inter-specific addition and substitution lines of Aegilops umbellulata chromosomes to ‘Chinese Spring’ indicated that chromosome 1U, which is homoeologous to the group 1 chromosomes of wheat, controls lectin synthesis.

    Notes: Summary cDNA clones encoding wheat HMW glutenin subunits have been isolated from a cDNA bank made to poly A+ RNA from developing wheat endosperm var. Chinese Spring. One such clone, pTag 1290, has enabled us to identify the HMW glutenin mRNA species. The DNA sequence of this clone has been partially determined and it contains several tandem DNA repeats. The sequence is discussed in relation to the generation of the HMW glutenin subunit gene family. Analysis of the organization of the HMW glutenin sequences in the wheat genome revealed that the genes encoding HMW glutenin subunits exist in low copy number and are located on the long arm of each of the homoeologous group 1 chromosomes.

    Notes: Summary Studies were conducted to determine the influence of the male sterility-inducing cytoplasm of Triticum timopheevii (Zhuk.) Zhuk. on response of several common winter wheat (T. aestivum L.) nuclear genotypes to photoperiod and vernalization. Comparative studies of cytoplasmic substitution lines provide information on the role of the cytoplasmic genetic mechanism in growth and development. In the case of cytoplasmic male sterility-based hybrid production systems, ubiquity of sterility-inducing cytoplasm in derived hybrids warrants thorough characterization of its influence on plant phenotype. Factorial combinations of cytoplasm (T. timopheevii and T. aestivum), nuclear genotype, and photoperiod or vernalization treatments were evaluated under hydroponic conditions in controlled environment chambers. Interaction of cytoplasm, photoperiod, and nuclear genotype was significant in one or more experiments for days to anthesis and potential spikelet number, and interaction of cytoplasm, vernalization, and nuclear genotype was significant for days to spike emergence. Long day length was associated with increased percentage seed set in one study, but interactions of photoperiod and cytoplasm were not detected for percentage seed set. Interactions involving cytoplasm and photoperiod or vernalization were interpreted as evidence of the existence of genetic factors in cytoplsam of T. timopheevii which alter photoperiod or vernalization responses of alloplasmic plants relative to responses exhibited by euplasmic plants. Since photoperiod and vernalization responses are critical to adaptation, T. timopheevii cytoplasm can alter adaptability of T. aestivum. The specific effect would be nuclear genotype dependent, and does not appear to be of a magnitude greater than that induced by nuclear genetic variability at loci conditioning photoperiod or vernalization responses or other adaptation-determining characteristics. Normal multilocation/year testing of alloplasmic hybrids should therefore adequately identify zones of adaptation.

    Notes: Summary Endosperm protein subunits of 109 primitive and modern lines of hexaploid wheat, Triticum aestivum L. em. Thell., were fractionated by one-dimensional, high resolution, sodium dodecyl sulphate (SDS) polyacrylamide gel electrophoresis (PAGE). A wide range of both qualitative and quantitative variation was observed in the fractions of the high molecular weight (HMW) glutenin and gliadin subunits of the different lines. The qualitative variation was expressed in the number of subunits per fraction and in their molecular weight, as determined by the differential rate of migration. The quantitative variation was expressed in the differential staining intensity of several subunits. The widest variation was detected in the HMW glutenin and gliadin subunits controlled by chromosome 1B while a much smaller variation was observed in those subunits controlled by chromosome 1A and further smaller variation in the subunits controlled by 1D. Only a small number of subunits in both fractions was found to be controlled by chromosome 1A indicating that diploidization of endosperm protein genes in common wheat has been non-random. The genetic and evolutionary implications of these findings are discussed.

    Notes: Summary In this first analysis the protein patterns obtained by two-dimensional gel electrophoresis of 8 day-old leaves from 18 alloplasmic wheat lines are compared. From 440 spots retained on the basis of their reproducibility, 36 proteins were observed to vary in different cytoplasms, allowing us to distinguish the T. aestivum cytoplasm from 5 Aegilops cytoplasms. Twenty-four of the 36 variable proteins could be structurally related to the large subunit of RuBPCase. Nuclear variation between 3 wheat varieties was observed for 14 proteins.

    Notes: Summary Cell proliferation from the mature embryo of wheat occurs on a defined medium in the presence of 2,4-D. Unorganized growth is observed when the 2,4-D concentration is equal to or greater than 2 mg/l, but increasing levels of 2,4-D inhibit cell division. Cell divisions begin after 4 days in culture from parenchyma cells within and near the procambial tissues of the embryo axis. By day 8 continuous meristematic zones are formed in association with vascular tissues, and DNA synthesis and cell divisions are distributed throughout these zones. No morphological evidence exists to show that these zones consist of proliferating root primordia, which are formed only after the level of 2,4-D falls below some critical concentration. When the concentration of 2,4-D is lowered, the meristematic zones first become dissected and then give rise to many root meristems.

    Notes: Abstract This study was conducted to evaluate the influence of route of administration upon the bioavailability and pharmacokinetics of methylprednisolone sodium succinate. Fourteen healthy adult male volunteers received 40 mg doses of methylprednisolone as the following treatments after an overnight fast in a 4-way crossover design: (a) as a 1 ml i.v. bolus;(b) as a 1 ml i.m. injection;(c) administered as an oral solution;and (d) as 5×8 mg oral tablets. Both the ester and free methylprednisolone were determined in plasma and urine. Study results indicate that the ester is rapidly and extensively converted to free methylprednisolone after all routes. The extent of methylprednisolone absorption was equivalent after i.v. and i.m. administration. Both orally administered treatments resulted in a lower extent of absorption attributed to a first-pass effect. Although a slightly lower extent of absorption was demonstrated following the oral administration of the methylprednisolone sodium succinate solution relative to the methylprednisolone oral tablets, this average difference of 9% would probably be of minimal therapeutic importance.
