    Description: This study was carried out to evaluate the effects of dietary lecithin on growth performance and hematological indices in juveniles of Siberian sturgeon (Acipenser baeri). Fish with initial average weight of 32.9±0.3 grams were fed five isoproteic and isolipidic formulated diets with different soybean lecithin levels including 0, 2.5, 5, 7.5 and 10% in triplicate groups for 8 weeks. Results showed that lecithin supplementation to 7.5% significantly increased some growth indices such as body weight increase (BWI), specific growth rate (SGR), feed conversion ratio (FCR ) final weight (Wf), condition factor (CF) and protein efficiency ratio (PER). Also, increasing dietary lecithin to the level of 10% significantly decreased growth indices. According to the results of hematological assays, hemoglobin (Hb) in treatments of 5% and 7.5% in comparison to other treatments had higher value. Hematocrit (Hct) in fish fed with 7.5% lecithin was higher than those fed with 0 and 2.5% lecithin in the diets. Other hematological indices such as WBC, RBC, MCV, MCH and MCHC showed no significant differences among the treatments. In conclusion, the results of this study indicated that Siberian sturgeon juveniles have a relatively moderate lecithin requirement (between 5 and 7.5 percent of the diet) and these levels of lecithin in diet induces increase of growth performance and improves health status of this species.

    Description: The maintenance of the endangered Caspian brown trout Salmo trutta caspius populations in Iran depends on its artificial breeding. There is no information on how current hatchery breeding protocol influences effective population size of breeders (Ne), which is a critical parameter to maintain genetic diversity in future generations. The current hatchery protocol (CHP) was comparatively evaluated with a balanced hatchery protocol (BHP), where mixtures of equal amounts of gametes per male and female breeder were used to balance parental contributions to progeny. To evaluate putative differences in viability between families, individual crosses were performed and fertilized ova of different families were mixed to constitute balanced family pools. 1440 alevins were totally sampled in the crosses performed from the 11 breeders. An exclusion-based parentage approach using three polymorphic microsatellite markers unambiguously assigned more than 93% of progeny to a single pair of parents. Significantly different contributions of breeders to progeny were observed in CHP (p〈0.05). The primary constraint on Ne in BHP was the unbalanced contribution of males, which seemed a consequence of sperm competition in mixed fertilization caused by differences in sperm quality. Sperm motility duration was positively correlated with the number of sired progeny by each male. The results illustrate the limitations of the BHP in minimizing the loss of genetic diversity observed in CHP. A protocol based on mixture of equal number of fertilized ova from individual male × female crosses emerged as the best alternative for conservation of Caspian brown trout.

    Description: Karyological characteristics of bighead goby (Neogobius kessleri) in the Caspian Sea were studied by examining 30 metaphase chromosome spreads from the kidney tissue of 10 specimens. The chromosome number of this species was found 2n=46 and the arm number as NF=46. The prepared karyotype of this species consisted of 23 pairs acro-telocentric (a-t) chromosomes. The chromosomal formula can be stated as 2n=46 (a-t). Karyological parameters showed that relative length was between 2.34-7.04 and length variation range of chromosomes was between 1.67-5.01 and total length was 71.16µm. It was found that the best chromosomal spread quality were obtained from intraperitonial injection of 40µg/g colchicine for 5 hours, hypotonization of samples in%1 sodium tri-citrate solution in 4ºC and preparation of spreads on cooled slide with flame technique.

    Description: Possibility of producing all-female rainbow trout (Oncorhynchus mykiss) using neomale stock were investigated in Kelardasht hatchery. To do this, we compared the fertilization, hatching rate and growth parameters of progeny in the first year of culture. Histological studies of gonads showed that progenies produced by mating sex-reversed males and normal females were totally female. Eyed-stage egg, hatching and early survival rate in all-female population were 90:69 %, 97.36 %, 91.49 % and in mix sex population were 70.24 %, 98.22 %, 90.73 %, respectively, showing no significant differences (P〉0.05). Lack of maturation in the first year cultured fish led to the identical growth parameters in mix sex and all-female groups (P〉0.05). Results showed that use of sex-reversed males can be one of the best and successful methods of producing all-female populations.

    Description: Caspian trout (Salmo trutta caspius Kessler, 1877) as endemic migratory fish, possessing great ecological economic and biodiversity value. Among the habitat drivers, temperature related effects on the genome were considered in this research. During the embryonic developmental stages, genomic alterations of single cells were assessed by Comet assay. After artificial propagation of wild broodstocks, the batches of eggs, were transferred to incubators with constant temperatures, REDD water recycling, 3 stages water filtration, UV-C application on effluent of each incubator and DO–pH–temperature control. Samples were collected from normal (8°C), cold (4°C) and warm (12°C) treatments on eyed egg, alevin and larval stages. Single Cell Gel Electrophoresis were applied on samples for evaluation of Comet Tail length (TL), DNA in tail (DT) and Tail Moment (TM) indices as DNA damage markers. Comparison between mean values of Comet TL index indicated 85.19 ±9.2, 9.88 ±2.3 and 256.54±14.6 for 4, 8 and 12°C treatments, respectively. DT were 18.07±6.3, 9.72±1.9 and 31.47±2.8 and then TM were measured as 15.27± 4.1, 1.21±0.2 and 95.25±8.6 for 4, 8, and 12°C treatments, respectively. Significant difference between thermal treatments (p〈0.05) observed. Warm treatments demonstrated greater DNA damage in compare with others. TM explained better information in comparison with TL and DT in all analyzed samples. In conclusion, temperatures out of the range of the optimal conditions could significantly affect Caspian trout genome which could be the subject of management considerations pertaining to Caspian trout stocks rehabilitation and captive rearing strategies.

    Description: Out of considerable environmental factors, temperature and its possible effects on life stages of Caspian trout investigated by natural incubation (8 ͦC) condition simulation comparing with cold (4 ͦC) and warm (12 ͦC) constant incubation temperatures in 3 well equipped incubators by water recycling systems. Green eggs triple treatments of wild and F1 cultured brooders were incubated. Incubation implemented in dark by using REDD water and DO–pH – temperature digital monitoring ended to yolk sac absorption and entering larval stage. Numbers and diameters of white fiber muscles measured and significant differences considered between three thermal treatments (P〈0.05) in both wild and cultured stocks. The numbers of white fiber muscles in warm treatment by highest means (72.54) and lowest diameter (8.46 micrometer) compared with Cold treatment white fiber muscles diameter (20.59 micrometer) and numbers (50.72) which were the highest diameter and lowest numbers means between treatments. Hatching success stated considerable mortality for cold treatments and 8°C incubator improved the best temperature in wild treatment. Incubation temperature induced significant effect on white fiber muscles stated considerable index for flesh precursor muscles condition which is subject of natural stocks rehabilitation and domestication projects considerations.

    Description: Six diets supplemented with 0, 100, 200, 400, 800, and 1600mg/kg L-Ascorbyl-2-Polyphosphate (APP) as vitamin C, were fed to juvenile beluga sturgeon in three replicates for 16 weeks. Fifty five fish with an average weight 38.1 plus or minus 0.5g were distributed to each tank after adaptation to the experimental diets. Most of the growth performance indices at week 4 indicated significant differences (P0.05), except in CF and PER (P0.05). The survival was affected by dietary treatment at week 4 (P0.05). No deficiency sign was observed during the trial. Results of this study indicated that vitamin C can have remarkable effects on growth indices in different growth periods. Therefore, regarding the effects of vitamin C on the growth indices and in the fish weight range used in this study, 100-200mg/kg vitamin C (APP) can be considered as the optimum level for rearing the fish.

    Description: Genetic variation of ship sturgeon (Acipenser nudiventris) from the Caspian Sea was investigated using NADH5/6 gene and PCR-RFLP analysis. A total of 80 specimens of the fish were collected from the south Caspian Sea and the Ural River from Kazakhstan. mtDNA ND5/6 gene was amplified by polymerase chain reaction (PCR) digested using 39 Endonucleases Restriction Enzyme. Of the 39 enzymes, five showed polymorphism. Totally, ten composite haplotypes among 80 specimens were detected. Haplotype AAAAA showed maximum frequency (57.5%) whereas haplotypes BBAAA and BABAA showed minimum frequency (1.2%). Haplotype AAAAB was recognized specifically in Ural River specimens. Average haplotype and nucleotide diversity was 0.8516 and 0.007 respectively. Compared to other sturgeon species living in the Caspian Sea, nucleotide diversity of Ship Sturgeon was much lower (0.007). This may be due to smaller population size of this species. Monte-Carol simulation using 1000 interaction did not show any significant differences between haplotype distribution of the fish sampled in the south Caspian Sea (X2=35.48 , P=0.74). However, we detected a significant difference between haplotype of Ship Sturgeon from Ural River and the south Caspian Sea. We conclude that Ship Sturgeon from Ural River is different from the fish in the south Caspian Sea and suggest Cfr131 enzyme as a molecular marker for population differentiation in the Caspian Sea.

    Description: The optimum dose of ultraviolet (UV) irradiation to produce gynogenetic Caspian salmon, Salmo trutta caspius, with emphasis on the "Hertwig effect" and photoreactivation (PR) were investigated. The sperm of Caspian salmon was irradiated with UV at 2010 ± 200 µw.cm-2 in different times including 0, 1, 3, 5, 8, 10, 15, 20, 25, 35 and 45 min and was allowed to fertilize normal ova; the fertilization, eyed and hatching rates were calculated to assess the performance. Using the irradiated sperm decreased the fertilization, eyed as well as hatching rates and the so-called "Hertwig effect" was observed, with the time-dependent decrease in the hatching rates at 0+ to 3 min irradiation, but better hatching rates were observed at more prolonged irradiation times. The best hatching rate was achieved at 25 min of UV irradiation; after that the survival rates rapidly declined to near zero. For PR studies, the semen was irradiated with UV (5, 30 and 120s) and untreated semen (0s) was used as control. Irradiated semen and/or fertilized eggs by treated semen were exposed to visible light (60 W) at a distance of 30 cm for 10 min; the eyed and hatching rates were measured. UV irradiation as low dose as 5s, significantly decreased the hatching rate (P 〈0.05). Semen and/or eggs illumination with visible light could not improve the survival rates (P 〉0.05). So, based on the results of this study, it was impossible to detect any PR mechanism in Caspian salmon.
