    Notes: Abstract Research on bias in machine learning algorithms has generally been concerned with the impact of bias on predictive accuracy. We believe that there are other factors that should also play a role in the evaluation of bias. One such factor is the stability of the algorithm; in other words, the repeatability of the results. If we obtain two sets of data from the same phenomenon, with the same underlying probability distribution, then we would like our learning algorithm to induce approximately the same concepts from both sets of data. This paper introduces a method for quantifying stability, based on a measure of the agreement between concepts. We also discuss the relationships among stability, predictive accuracy, and bias.

    Notes: Abstract This paper presents an attempt at the application of catastrophe theory to the stability analysis ofJ-controlled crack growth in three-point bending specimens. By introducing the solutions ofJ- integral in the completely yielding state for the ideal plastic material, the critical condition of losing stability for the crack propagation in the specimen is obtained, based on the cusp catastrophe theory. The process of the crack growth from geometrical sense is described.

    Notes: Abstract For improving chemical and thermal stability of γ -Al2O3 membranes boehmite (AlOOH) sol-particles are coated with Zr4+-species with two techniques. These techniques are heterogeneous precipitation (HP-method) and “surface-reaction-followed-by-polycondensation” (SRPC-method). A continuous coating layer is formed at relative low Zr4+-concentration, about one monolayer coverage of boehmite particles, and with the HP-method. For larger concentrations and for the SRPC-method small particles (order 1 nm) of Zr4+-species are formed on the surface of the boehmite particles. After drying and calcination up to 1000°C no continuous layer of a zirconia phase could be detected for all samples. However the thermal stability of the porous structure is improved. Phase transitions of alumina occur at temperatures of 1100°C and the porous structure of the membrane material is then destabilized.

    Notes: Abstract We investigate the problem of detecting and assessing, by means of static electrical measurements, damage due to corrosion in a structure. Corrosion damage, which is assumed to occur in an inaccessible part of a specimen, is modelled as material loss. The detection device consists of electrodes which inject DC current and measure voltage potentials in the accessible part of the specimen. The topography of the damaged surface is estimated from the measured data. This research is meant to evaluate if a method based on static electrical measurements has the potential to be developed into a useful nondestructive evaluation tool. We propose computational methods that take the measured data and estimate the unknown damaged surface. The methods are studied in order to understand their properties. Several example calculations from synthetic data are presented. Our findings indicate that such a device has limited resolution. However, it offers several advantages that make it worthwhile to pursue further research.

    Notes: Abstract X-ray photoelectron spectroscopy measurements show that exposure of an Fe−Cr−Ni alloy to H2S and O2 under ultrahigh vacuum (UHV) conditions leads to the thermal instability of the Fe oxide without affecting the Cr oxide. An atomically clean metal surface, predominantly Fe in composition and free of internal sulfur, was exposed to H2S under controlled conditions in order to form a sulfided monolayer. Adsorbed S inhibited surface oxidation at temperatures between 325 K and 825 K. The presence of adsorbed S did not, however, inhibit the surface segregation of Cr for temperatures 〉600 K, compared to a S-free sample. Upon annealing of a sulfided sample to 900 K in UHV, the Fe oxide largely disappears, while no change is observed in the Cr oxide. The S-free sample shows no significant change in either the Fe or Cr oxides upon annealing. The results presented in this paper show that sulfidation of an alloy surface prior to oxidation and surface segregation can adversely affect the thermal stability of the Fe oxide without affecting the Cr oxide overlayer resulting from surface segregation and preferential oxidation.

    Notes: Abstract The complex formation reaction between iodine and 1,7-diaza-15-crown-5 (DA15C5) has been studied spectrophotometrically in chloroform at 25°C. The resulting 1:2 (DA15C5:I2) molecular complex was formulated as (DA15C5...;I+)I 3 − . The spectrophotometric results, as well as the conductivity measurements, revealed that the gradual release of triiodide ion from its contact ion paired form in the molecular complex into the solution is the rate determining step of the reaction. The rate constant was calculated ask=(8.8±0.2)×10−3 min−1. The formation constant of the molecular complex was evaluated from the computer fitting of the absorbance-mole ratio data as logK f=6.89±0.09.

    Notes: Abstract The influence of cyclodextrins (CDs) on the chemical stability of the contraceptive steroid progestin, ST1435, in aqueous solution has been studied using reversed phase high performance liquid chromatography. The effects of CD structure, temperature, and CD concentration on the rate of degradation were investigated. It was found that the drug degraded to different extents following a pseudo-first order reaction mechanism. The presence of the host molecules affected the degradation rate as a result of complexation which might result in protection of the labile moiety of the drug molecule against degradation. Hydroxypropyl-β-cyclodextrin (HP-β-CD) and hydroxyethyl-β-cyclodextrin (HE-β-CD) retarded the degradation in contrast to β-cyclodextrin (β-CD) which accelerated the steroid degradation. The stabilizing action of HP-β-CD is larger than that of HE-β-CD. The degradation rate increased upon increasing temperature and the Arrhenius equation is valid. Lineweaver-Burk equation analysis indicated that the steroid included inside the CD cavity degraded three times more slowly than did the free ST1435 in solution. This equation further supported the formation of a 1 : 1 inclusion complex between ST1435 and HP-β-CD with a stability constant of 934.5 M-1 at 65°C.

Tetracoordinated planar structures of the Cu(II) complexes of the Schiff bases and distorted tetrahedral structures of the Cu(II) complexes of compoundsV–VI have been elucidated by ESR and other spectral methods. During the preparation of the complex the hydrolysis of the Schiff base often takes place in the presence of water giving anthranilates and salicylaldehydates of metals to some extent along with the complexes of the Schiff base. The kinetic data for the hydrolysis ofN-salicylideneanthranilic acid (I) in methanol-water solution also are reported.

    Notes: Abstract Sharifnia, Caramanis, and Gershwin [1991] introduced a class of policies for manufacturing systems, called by themlinear corridor policies. They proved that their stability can be discussed by the study of a simpler subset of such policies (cone policies). This paper revisits their work presenting a different description of the dynamics of the systems under study and explores it to device a necessary and sufficient condition for stability, obtained by the strengthening of the assumptions in Sharifnia et al.     Notes: Abstract We investigate the use of splitting methods for the numerical integration of three-dimensional transport-chemistry models. In particular, we investigate various possibilities for the time discretization that can take advantage of the parallelization and vectorization facilities offered by multi-processor vector computers. To suppress wiggles in the numerical solution, we use third-order, upwind-biased discretization of the advection terms, resulting in a five-point coupling in each direction. As an alternative to the usual splitting functions, such as co-ordinate splitting or operator splitting, we consider a splitting function that is based on a three-coloured hopscotch-type splitting in the horizontal direction, whereas full coupling is retained in the vertical direction. Advantages of this splitting function are the easy application of domain decomposition techniques and unconditional stability in the vertical, which is an important property for transport in shallow water. The splitting method is obtained by combining the hopscotch-type splitting function with various second-order splitting formulae from the literature. Although some of the resulting methods are highly accurate, their stability behaviour (due to horizontal advection) is quite poor. Therefore we also discuss several new splitting formulae with the aim to improve the stability characteristics. It turns out that this is possible indeed, but the price to pay is a reduction of the accuracy. Therefore, such methods are to be preferred if accuracy is less crucial than stability; such a situation is frequently encountered in solving transport problems. As part of the project TRUST (Transport and Reactions Unified by Splitting Techniques), preliminary versions of the schemes are implemented on the Cray C98 4256 computer and are available for benchmarking.

    Notes: Abstract Adaptive artificial neural network techniques are introduced and applied to the factorization of 2-D second order polynomials. The proposed neural network is trained using a constrained learning algorithm that achieves minimization of the usual mean square error criterion along with simultaneous satisfaction of multiple equality and inequality constraints between the polynomial coefficients. Using this method, we are able to obtain good approximate solutions for non-factorable polynomials. By incorporating stability constraints into the formalism, our method can be successfully used for the realization of stable 2-D second order IIR filters in cascade form.

    Notes: Abstract We study the continuity properties of the bundle of solutions to a differential inclusion subject to a singular perturbation, i.e., with respect to a scalar parameter ɛ multiplying a part of the derivatives. We give conditions under which every solution of the singularly perturbed inclusion is close, in a certain sense and for a sufficiently small ɛ, to a solution of the degenerate inclusion obtained for ɛ=0. These conditions include both stability and structural requirements (the later having no counterpart in the case of a differential equation). The main result obtained generalizes the well-known Tikhonov theorem for singularly perturbed differential equations.

    Notes: Abstract  The characteristics of poly(oxyethylene) hydrogenated caster oil ether (HCO-10) vesicles were studied for the standpoints of encapsulation efficiency, stability, solubilization and permeability or barrier efficiency. The vesicles of 5% HCO-10 had 6.24% of calcein-entrapment efficiency and 240 nm of mean diameter. The stability of HCO-10 vesicle suspensions was dependent on their concentrations. In the vesicle suspensions of 10% HCO-10 or more, both the size of the vesicles and the fluidity of the suspensions obviously varied with incubation time, indicating that a flocculation occurred; whereas, the vesicle suspension of 5% HCO-10 was relatively stable. The solubilization process of HCO-10 vesicles by SDS was similar to that of EggPC liposomes. The rate constants for permeation of Cl ion and calcein were 2.46×10-3 s-1 and 5.79×10-5 s-1, respectively, suggesting that HCO-10 vesicles possessed some barrier potential for Cl ion and calcein although they were smaller than those of liposomes. Furthermore, the efflux of the solute such as calcein from HCO-10 vesicles was maximum at 37 °C, where the vesicle membrane was presumably destabilized by dehydration of EOs in HCO-10 molecules.

    Notes: Abstract Application of the muramyldipeptide derivative B30-MDP to liposomal vaccines will aid in the development of improved high immunogenicity vaccines. To give full play to the effectiveness of B30-MDP as a liposomal vaccine, it is important to evaluate the effect of cholesterol, dimyristoylphosphatidylcholine (DMPC) or distearoylphosphatidylcholine (DSPC) incorporation on the chemical stability of B30-MDP and physicochemical properties of B30-MDP/lipid mixed vesicles from the view point of pharmaceutics. The observed degradation rate constants of B30-MDP by hydrolysis in B30-MDP/cholesterol mixed vesicles were increased with increasing concentration of cholesterol, however, those in B30-MDP/DMPC and B30-MDP/DSPC mixed vesicles were unchanged with increasing concentration of DMPC and DSPC. The degradation behavior of B30-MDP was then compared with physicochemical properties of B30-MDP/lipid mixed vesicles, such as membrane fluidity and particle size. It was apparent that the degradation of B30-MDP in B30-MDP/cholesterol mixed vesicles was influenced by the particle size, but not by the fluidity of the membranes. In the case of B30-MDP/phospholipid mixed vesicles, MDP/phospholipid mixed vesicles, the degradation of B30-MDP was not influenced by either the membranes' fluidity or the particle size of the mixed vesicles. It is considered that the degradation of B30-MDP in the mixed vesicles is dependent on the membrane state, and the addition of cholesterol to B30-MDP vesicle inhibits the mutual interaction of MDP regions, whereas the addition of phospholipids hardly influences the mutual interaction of MDP regions, possibly owing to phase separation between B30-MDP and phospholipids.

    Notes: Abstract The behavior of a ship encountering large regular waves from astern at low frequency is the object of investigation, with a parallel study of surf-riding and periodic motion paterns. First, the theoretical analysis of surf-riding is extended from purely following to quartering seas. Steady-state continuation is used to identify all possible surf-riding states for one wavelength. Examination of stability indicates the existence of stable and unstable states and predicts a new type of oscillatory surf-riding. Global analysis is also applied to determine the areas of state space which lead to surf-riding for a given ship and wave conditions.     Notes: Abstract The Differential System Method (DSM) permits identification of the physical parameters of finite-difference groundwater flow models in a confined aquifer when piezometric head and source terms are known at each point of the finite-difference lattice for at least two independent flow situations for which the hydraulic gradients are not parallel. Since piezometric head data are usually few and sparse, interpolation of the measured data onto a regular grid can be performed with geostatistical techniques. We apply kriging to the sparse data of a synthetic aquifer to evaluate the stability of the DSM with respect to uncorrelated measurement errors and interpolation errors. The numerical results show that the DSM is stable.

Sufficient conditions placed on the triangles of the triangulation lead to discrete ellipticity. Proof of anO(h 2) error estimate is given for these conditions.

    Notes: Abstract. While learning and development are well characterized in feedforward networks, these features are more difficult to analyze in recurrent networks due to the increased complexity of dual dynamics – the rapid dynamics arising from activation states and the slow dynamics arising from learning or developmental plasticity. We present analytical and numerical results that consider dual dynamics in a recurrent network undergoing Hebbian learning with either constant weight decay or weight normalization. Starting from initially random connections, the recurrent network develops symmetric or near-symmetric connections through Hebbian learning. Reciprocity and modularity arise naturally through correlations in the activation states. Additionally, weight normalization may be better than constant weight decay for the development of multiple attractor states that allow a diverse representation of the inputs. These results suggest a natural mechanism by which synaptic plasticity in recurrent networks such as cortical and brainstem premotor circuits could enhance neural computation and the generation of motor programs.

    Notes: Abstract Purpose. The effects of 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) on the aqueous solubility and stability of two lipophilic bispilocarpine prodrugs were investigated at pH 7.4. Methods. The solubility of prodrugs was studied by phase-solubility method (0–72.5 mM HP-β-CD). The stability of one of the prodrugs was investigated as a function of temperature (40°C–70°C) and HP-β-CD concentration (0–72.5 mM). The apparent rate constants (k 1, k 2) for degradation of prodrug in 1:1 and 1:2 inclusion complexes and apparent stability constants (K 1:1, K l:2) were calculated by the curve-fitting method. Results. The phase-solubility diagrams were classified as Ap-type and the apparent stability constants (K l:l, K l:2) for 1:1- and 1:2-inclusion complexes were calculated to be 143–815 M−l and 29–825 M−1, respectively. The stability of prodrug increased as a function of HP-β-CD concentration over the studied temperature range. The shelf-life (t 90%, calculated by the Arrhenius equation) of the prodrug in 72.5 mM HP-β-CD solution increased 5.1-fold and 6.1-fold at 25°C and 4°C, respectively. Conclusions. The solubility of the prodrugs was shown to increase markedly in phase-solubility studies. The degradation rate of prodrug in stability studies was shown to be slower in the l:2-complex than in the l:l-complex and the relative amounts of complex species were found to be dependent on CD concentration.

    Notes: Abstract Purpose. To study the oxidation of the methionine residue of antiflammin 2 (HDMNKVLDL, AF2) as a function of pH, buffer concentration, ionic strength, and temperature using different concentrations of hydrogen peroxide and to determine the accessibility of methionine residue to oxidation. Methods. Reversed-phase high-performance liquid chromatography (RPHPLC) was used as the main analytical method in determining the oxidation rates of AF2. Calibration curves for AF2 and the oxidation product, methionine sulfoxide of AF2 (Met(O)-3-AF2), were constructed for each measurement using standard materials. Fast Atom Bombardment Mass Spectroscopy (FABMS) was used to characterize the product. Results. Met(O)-3-AF2 was the only oxidation product detected at pH 3.0 to 8.0. The oxidation rates were independent of buffer concentrations, ionic strength, and pH from 3.0 to 7.0. However, there was an acceleration of the rates at basic pHs, and small amounts of degradation products other than Met(O)-3-AF2 were observed in this alkaline region. Conclusions. Oxidation of methionine in AF2 does not cause the biological inactivation reported by other laboratories since this drug is relatively stable under neutral conditions in the absence of oxiding agent.

To study the interaction of the polyene antifungal amphotericin B with phospholipid Langmuir monolayers and to correlate with stability of phospholipid-stabilized drug emulsions. Methods. Pressure—area isotherms of mixed monolayers of amphotericin B (0–20 mol%) and different phospholipid types were recorded using conventional Langmuir trough methods. Emulsion stability of amphotericin B-containing lipid emulsions was measured using dynamic light scattering. Results. Incorporation of amphotericin B into monolayers composed of saturated phospholipids (Lipoid E80-3) had a profound effect on the shape of the isotherm. This effect was directly related to the concentration of amphotericin B in the monolayer. At high drug concentrations, the shape of the isotherms became progressively similar to that of pure DPPC, thus exhibiting regions attributable to phospholipid in different phase states. This effect on isotherm shape was not observed following incorporation of the drug into monolayers composed of the equivalent unsaturated lecithin (Lipoid E80). Conclusions. These results are interpreted as indicating the formation of an amphotericin B-phospholipid complex, resulting in phase separation within the monolayer. The extent and nature of this phase separation was dependent on both the concentration of drug in the system, and the saturation state of the phospholipid component. The relevance of these observations to the stability of amphotericin B drug emulsions stabilised by saturated and unsaturated phospholipid emulsifiers is discussed. These observations may also be relevant to the toxicity of these, and other novel amphotericin B formulations.

    Notes: Abstract Chimeric BR96-doxorubicin conjugate (BR96-DOX) is an immunoconjugate designed to specifically target and kill certain tumor cells. The linker between the chimeric BR96 antibody and DOX is an acid-labile hydrazone group which was designed to undergo lysosomal hydrolysis to release DOX in vivo. Stability studies indicated that acid-catalyzed hydrazone hydrolysis was the major degradation route in vitro. Even under optimal conditions of pH and temperature, the stability of BR96-DOX in solution was not acceptable for long-term storage. Lyophilization of BR96-DOX in the presence of added sugars, such as lactose or sucrose, and subsequent storage of the lyophile under refrigeration significantly improved the stability. Therefore lyophilization appears to be a viable approach for achieving long-term stabilization of BR96-DOX.

    Notes: Abstract Purpose. The development of a sustained release formulation for recombinant human growth hormone (rhGH) as well as other proteins requires that the protein be stable at physiological conditions during its in vivo lifetime. Poly(lactic-co-glycolic acid) (PLGA) microspheres may provide an excellent sustained release formulation for proteins, if protein stability can be maintained. Methods. rhGH was encapsulated in PLGA microspheres using a double emulsion process. Protein released from the microspheres was assessed by several chromatrographic assays, circular dichroism, and a cell-based bioassay. The rates of aggregation, oxidation, diketopiperazine formation, and deamidation were then determined for rhGH released from PLGA microspheres and rhGH in solution (control) during incubation in isotonic buffer, pH 7.4 and 37°C. Results. rhGH PLGA formulations were produced with a low initial burst (〈20%) and a continuous release of rhGH for 30 days. rhGH was released initially from PLGA microspheres in its native form as measured by several assays. In isotonic buffer, pH 7.4 and 37°C, the rates of rhGH oxidation, diketopiperazine formation, and deamidation in the PLGA microspheres were equivalent to the rhGH in solution, but aggregation (dimer formation) occured at a slightly faster rate for protein released from the PLGA microspheres. This difference in aggregation rate was likely due to the high protein concentration used in the encapsulation process. The rhGH released was biologically active throughout the incubation at these conditions which are equivalent to physiological ionic strength and pH. Conclusions. rhGH was successfully encapsulated and released in its fully bioactive form from PLGA microspheres over 30 days. The chemical degradation rates of rhGH were not affected by the PLGA microspheres, indicating that the internal environment of the microspheres was similar to the bulk solution. After administration, the microspheres should become fully hydrated in the subcutaneous space and should experience similar isotonic conditions and pH. Therefore, if a protein formulation provides stability in isotonic buffer, pH 7.4 and 37°C, it should allow for a safe and efficacious sustained release dosage form in PLGA microspheres.

To separate and characterize the different positional isomers of mono-PEGylated salmon calcitonins (mono-PEG-sCTs) and to evaluate the effects of the PEGylation site on the stability of different mono-PEG-sCTs in rat kidney homogenate. Methods. Mono-PEG-sCTs were prepared using succinimidyl carbonate monomethoxy polyethylene glycol (5,000 Da) and separated by gel-filtration HPLC followed by reversed-phase HPLC. To characterize PEGylated sCTs, matrix-assisted laser desorption ionization time of flight mass spectrometry (M ALDI-TOF MS) and reversed-phase HPLC of the trypsin digested samples were performed. Mono-PEG-sCTs and sCT in rat kidney homogenates were measured by column-switching reversed-phase HPLC with on-line detection of the radioiodinated samples using a flow-through radioisotope detector. Results. Three different mono-PEGylated sCTs were separated by reversed-phase gradient HPLC. From the MALDI-TOF MS analysis, the average molecular weight of mono-PEG-sCTs was confirmed as around 8650 Da. The presence of PEG moiety in the mono-PEG-sCTs was also manifested by the fact that the distance between two adjacent mass spectum lines was 44 Da which corresponds to PEG monomer unit. Tryptic digestion analysis demonstrated that these mono-PEG-sCTs are 3 positional isomers of N-terminus, Lys18- and Lys11-residue modified mono-PEGylated sCTs. The degradation half-life of these 3 positional isomers in rat kidney homogenates significantly increased in order of the N-terminus (125.5 min), Lys11- (157.3 min), and Lysl8-residue modified mono-PEGylated sCT (281.5 min) over the native sCT (4.8 min). Conclusions. Three positional isomers of mono-PEGylated sCTs were purified and characterized. Of these, the resistance to proteolytic degradation was highest for the Lysl8-residue modified mono-PEG-sCT. These studies demonstrate that the in vivo stability of PEGylated sCTs is highly dependent on the site of PEG molecule attachment.

    Notes: Abstract The distribution of oleic acid between Salbutamol base drug and the solvent in metered-dose inhalers (MDI's) has been investigated. The equilibrium surfactant concentration in the drug dispersions has been determined using a colorimetric method. The samples examined contained Salbutamol base drug particles and oleic acid dispersed in different propellant blends of freon 11 and 12. The maximum equilibrium concentration observed depended on the propellant blend used. The propellant blend and the distribution of the surfactant affected the dispersion stability. The effect of the surfactant is illustrated by the adsorption isotherm for oleic acid onto the Salbutamol particles. The results are correlated with zeta-potentials and particle size measurements made on similar systems in order to characterize the properties of surfactant stabilized MDI's.

    Notes: Abstract Purpose. The goal of this study was to evaluate the ability of nanoparticles to be used as a targeted delivery system for oligonucleotides. Methods. Pharmacokinetic and tissue distribution were carried out in mice by measuring the radioactivity associated to the model oligothymidylate 33P-pdT16 loaded to poly(isobutylcyanoacryrate) (PIBCA) nanoparticles. In addition, we have used a TLC linear analyzer to measure quantitatively on a polyacrylamide gel electrophoresis, the amount of non degraded pdT16 Results. Organ distribution study has shown that nanoparticles deliver 33P-pdT16 specifically to the liver reducing its distribution in the kidney and in the bone marrow. Nanoparticles could partially protect pdT16 against degradation in the plasma and in the liver 5 min after administration, whereas free oligonucleotide was totally degraded at the same time. Conclusions. Nanoparticles protect oligonucleotides in vivo against degradation and deliver them to the liver.

    Notes: Abstract Purpose. The successful development of controlled release formulations for proteins requires that the protein not be denatured during the manufacturing process. The major objective was to develop formulations that stabilize two recombinant human proteins, human growth hormone (rhGH) and interferon-γ (rhIFN-γ), at high protein concentrations (〉100 mg/mL) in organic solvents commonly used for microencapsulation, methylene chloride and ethyl acetate. Methods. Several excipients were screened to obtain the maximum solubility of each protein. These formulations (aqueous, lyophilized, milled, spray dried, or isoelectric precipitate) were then rapidly screened by emulsification in the organic solvent followed by recovery into excess buffer. Additional screening was performed with solid protein that was suspended in the organic solvent and then recovered with excess buffer. The recovery of native protein was determined by native size exclusion chromatography (SEC-HPLC) and circular dichroism (CD). The selected formulations were encapsulated in poly-lactic-coglycolic acid (PLGA) microspheres by either water-in-oil-in-water (W/O/W) or solid-in-oil-in-water (S/O/W) methods. The initial protein released from the microspheres incubated at physiological conditions was analyzed by SEC-HPLC, CD, and biological assays. Results. The stability of a given formulation in the rapid screening method correlated well with stability during encapsulation in PLGA microspheres. Formulations of rhGH containing Tween 20 or 80 resulted in lower recovery of native protein, while trehalose and mannitol formulations (phosphate buffer, pH 8.0) yielded complete recovery of native rhGH. Other additives such as carboxymethyl cellulose, gelatin, and dextran 70 were not effective stabilizers, and polyethylene glycol provided some stabilization of rhGH. Trehalose/rhGH (1:4 mass ratio) and mannitol/rhGH (1:2 mass ratio) formulations (potassium phosphate buffer, pH 8.0) were lyophilized, reconstituted to 200 and 400 mg/mL rhGH, respectively, and then encapsulated in PLGA micro-spheres. The protein was released from these microspheres in its native state. Lyophilized formulations of rhGH yielded analogous results indicating the ability of trehalose and mannitol to stabilize the protein. Small solid particles of rhGH generated by spray drying (both air and freeze-drying) formulations containing Tween 20 or PEG were stable in ethyl acetate, but not methylene chloride. Similar results were also obtained with rhIFN-γ (137 mg/mL in succinate buffer, pH 5.0), where both mannitol and trehalose were observed to stabilize the protein during exposure to the organic solvents resulting in the release of native rhIFN-γ from PLGA microspheres. Conclusions. The rapid screening method allowed the development of stable concentrated protein solutions or solid protein formulations that could be successfully encapsulated in PLGA microspheres. The excipients observed to stabilize these proteins function by preferential hydration of the protein, and in the dry state (e.g., trehalose) may stabilize the protein via water substitution yielding a protective coating around the protein surface. Studies of other proteins should provide further insight into this mechanism of protein stabilization during encapsulation.

    Notes: Abstract Ultrasonic nebulization of lactate dehydrogenase (LDH) was investigated using a DeVilbiss “Aerosonic” nebulizer. The enzyme (8ml, 0.025mg/ml Na2HPO4, pH 7.0) was completely inactivated after 20 minutes of operation. However, the inactivation profile observed during ultrasonic nebulization was different from that previously observed using air-jet nebulization. At least two mechanisms are involved, one associated with heating and the other with aerosol production. By preventing heating of the nebulizer fluid during operation, the denaturation profile was dramatically altered. By additionally including 0.01% w/v Tween 80 or l%w/v PEG 8000, almost all activity was retained. Similar results were obtained by preventing aerosol production and heating. However, 100% of activity was lost when heating was allowed to occur without aerosol formation. The results demonstrate that cooling in conjunction with a surfactant is one approach that could be used to stabilize proteins to ultrasonic nebulization. However, cooling also significantly reduced solute output from the nebulizer. When operated at 10°C output was negligible. At 50°C the output was 5× greater than that found at room temperature. The median droplet size (µm) was not significantly influenced by the operating temperature of the nebulizer fluid (3.6 ± 0.4, 21°C; 3.9 ± 0.2, 50°C, p = NS (n = 6)) although the size distribution was noted to increase at the higher temperature.

    Notes: Abstract Purpose. The degradation kinetics of thymopentin (RKDVY) and its analogs (RKDVW and RPDVY) in aqueous solution was studied by isothermal and nonisothermal methods. Methods. The isothermal decomposition of thymopentin and its analogs was investigated as a function of pH (2−10), temperature (37, 57, and 80°C) and ionic strength (μ = 0.02 to 1). Nonisothermal decomposition studies were performed using a linear temperature programmer. The temperature increasing rate was set to 0.25°C per hour and the temperature interval varied from 40 to 88°C. Results. The decomposition of thymopentin and its analogs followed first order kinetics. The dependence of the rate constant on temperature followed a linear Arrhenius plot. This indicated that the degradation mechanism of thymopentin and its analogs might be the same within the temperature range studied. The energies of activation were found to be in close agreement for the isothermal and nonisothermal studies, suggesting that the nonisothermal studies may save considerable amount of time in the early stages of drug development. The logK-pH profile of thymopentin suggests that maximum stability is achieved in the pH range of 6−8. Conclusions. These results indicate that the nonisothermal methodology provides an attractive alternative to isothermal methods, as it requires a much lower amount of both material and time, to determine the peptide stability and to estimate the shelf-life for peptide pharmaceutical preparations.

    Notes: Abstract Submicron emulsions of miconazole were stabilized by using a combination of three emulsifiers comprising phospholipids, poloxamer, and deoxycholic acid (DCA). The presence of DCA was vital for prolonged emulsion stability owing to its contribution to the elevated zeta potential of the emulsion. Further, the results by the phospholipid surface labelling colorimetric technique clearly suggested that poloxamer molecules interacted with phospholipid polar-head groups of the mixed DCA-phospholipid interfacial film, resulting in the stabilization of the emulsion by a steric enthalpic entropic mechanism. The plain emulsion vehicle was well tolerated up to a dose of 0.6 ml injected i.v. to BALB/c mice. The maximum tolerated dose of miconazole was 80 and 250 mg/kg in Daktarin® i.v. (a marketed product) and emulsion, respectively, showing an improved safety ratio of 1 to 3 in favor of the emulsion. These results tended to confirm that the adverse effects associated with Daktarin® i.v. injection should be associated with the vehicle rather than with the miconazole itself. In a murine cryptococcosis model, only one mouse out of ten remained alive by day 15 in the infected group treated with Daktarin® i.v., while in the miconazole emulsion treated group, mice began to die from day 16 up to day 25 post inoculation. Thus, the multiple-dose treatment with the miconazole emulsions improved the protection offered to the infected mice. However, the therapeutic levels of miconazole that were reached in the target organ (brain) were lower than those required for complete eradication of Cryptococcus neoformans, which is known to multiply preferentially in the brain.

To identify a suitable nonaqueous, parenterally acceptable suspending vehicle whereby a therapeutic protein is delivered as a stable flowable powder, making it amenable to delivery from sustained delivery systems maintained at body temperature. Methods. Formulations of plasma derived Factor IX (pdFIX) and recombinant human alpha interferon (rhα-lFN) were formulated as dry powders, suspended in various vehicles (perfluorodecalin, perfluorotributylamine, methoxyflurane, polyethylene glycol 400, soybean oil, tetradecane or octanol) and stored at 37°C. Stability was assessed by size exclusion chromatography, reverse phase chromatography, ion exchange chromatography, and bioassay, and was compared to the stability of dry powder formulations stored at 37°C and −80°C. Results. PdFIX was stable when stored at 37°C as a dry powder, or when the dry powder was suspended in the pharmaceutically acceptable vehicles perfluorodecalin or perfluorotributylamine. Suspensions of the powder in other pharmaceutically/parenterally acceptable vehicles such as soybean oil or PEG 400 resulted in aggregation and loss of bioactivity. A dry powder formulation of rhα-lFN suspended in perfluorodecalin was also stable at 37°C. Conclusions. This study shows the potential utility of perfluorinated hydrocarbons as nonaqueous suspending vehicles for long term in-vivo delivery of therapeutic proteins.

Nonisothermal kinetics was studied with the purpose of determining its use in the establishment of the expiration date of pharmaceutical preparations, particularly drugs in solutions and for assessment of stability characteristics of pharmaceutical formulations during the development stage. A comparison of isothermal (55, 70 and 85°C) and nonisothermal kinetics was performed. Aqueous solutions of famotidine were buffered at pH 1.71, 2.24, 2.66, 4.0, 8.5, 9.0 and 10.0 were used. In the nonisothermal studies, the temperature rate of the reaction was continuously varied throughout the experiment. The energies of activation were found to be in close agreement for isothermal and nonisothermal studies, indicating that nonisothermal studies may save considerable amount of time in the early stages of drug development and stability testing. Logk-pH profiles were constructed for 55, 70 and 85°C from the first-order rate constants obtained from isothermal studies at pH values ranging from 1.71 to 10.00. The pH-rate profile indicated that famotidine undergoes specific acid catalysis in the acidic region and general base catalysis in the alkaline region. Hydrolysis in the acidic and alkaline media resulted in the formation of four and five degradation products, respectively. A possible degradation pathway for the acidic and alkaline hydrolysis was discussed.

    Notes: Abstract Purpose. To elucidate the solution conditions that confer stability of aqueous IL-1R using differential scanning calorimetry (DSC). Methods. Optimal pH conditions were determined by monitoring degradation products encountered during accelerated studies (at elevated temperatures) using SDS-PAGE. At the pH optimum, DSC screened for excipients that enhanced thermal stability by shifting the Tm to higher values. Using SEC the relationship between thermal unfolding and stability was investigated by considering if lower Tm's in the presence of preservatives correlated with degradation products at 37°C over time. The degree of aggregation relative to that of a control determined the level of stability achieved. Results. Circular dichroism (CD) measurements confirmed molecular modeling studies showing IL-1R to be about 39% β-sheet. Two major transitions characterized the DSC data with Tm's observed near 47°C and 66°C. Among 21 excipients screened, NaCl exhibited the greatest stabilizing influences based on shifting the low temperature transition to 53°C. The low temperature transition was later found to comprise two transitions, yielding a total of three melting transitions for IL-1R. High Tm's arising from the presence of preservatives correlated with the order of stability (i.e., 0.065% phenol 〉 0.1% m-Cresol 〉 0.9% benzyl alcohol). Conclusions. The three melting transitions are consistent in origin with the cooperative unfolding of three unique immunoglobulin-like domains of IL-1R. Optimal stability was achieved in 20 mM sodium citrate at pH 6 with sufficient NaCl to attain the tonicity of human serum. A correlation between the predicted ranking of stability and the extent of aggregation was demonstrated using DSC.

    Notes: Abstract Purpose. Examination of the dried-state conformation of interleukin-2 (IL-2) was used to determine the pH conditions and stabilizers that provide optimal storage stability for the lyophilized product. Methods. Fourier-transform infrared spectroscopy and accelerated stability studies which examined solubility, aggregate formation, and covalent cross-linking were used. Results. Varying the pH in the absence of excipients resulted in dramatic differences in the dried state conformation of IL-2. At pH 7, IL-2 unfolds extensively upon lyophilization while at pH below 5 it remains essentially native. Additional unfolding was observed upon incubation at elevated temperatures. A strong direct correlation between the retention of the native (aqueous) structure during freeze-drying and enhanced stability is demonstrated. IL-2 prepared at pH 5 is approximately an order of magnitude more stable than at pH 7 with regard to formation of soluble and insoluble aggregates. A similar pH profile was observed in the presence of excipients, although the excipients alter the overall stability profile. Additional accelerated stability studies examined the stabilizers necessary for optimal stability. Conclusions. Excipients with the capacity to substitute for water upon dehydration better preserve the native structure resulting in enhanced stability. Those that have high glass transition temperatures provide the highest level of stability during storage, although they do not prevent dehydration induced unfolding.

The liquid stability of rhG-CSF was investigated after polyethylene glycol (PEG) with an average molecular weight of 6000 daltons was covalently attached to the N-terminal methionine residue. Methods. The conjugation methods chosen for modifying the N-terminal residue were alkylation and acylation. The N-terminally PEGylated rhG-CSF conjugates were purified by cation exchange chromatography. The physical characterization methods of SDS-PAGE, endoproteinase peptide mapping, circular dichroism and in-vivo bioassay were used to test for differences between the PEG-rhG-CSF molecules. Results. Physical characterization indicated no apparent differences in the rhG-CSF molecules that were conjugated with either method. Stability, in liquid at elevated temperatures, of these conjugated molecules indicated that the primary pathway of degradation was aggregation. Conjugation through alkylation offered the distinct advantage of decreasing, by approximately 5 times, the amount of aggregation present as compared to acylation. Conclusions. We suggest, that the increased stability observed for the molecules utilizing the alkylation conjugation method may be due to the preservation of charge on the alpha amino group of rhG-CSF.

    Notes: Abstract The chemistry associated with the process of N,O-acyl migration was explored in both cyclic and linear peptides under aqueous acid conditions. The importance of backbone cyclization and N-methylation of the peptide bond on the kinetics of N,O-acyl migration in a series of linear and cyclic peptides related in structure to cyclosporin A (CsA) were examined. The similarity in the chemical reactivity of the cyclic peptide [MeLeu (3-OH)]1-CsA and the corresponding linear peptide [Val-MeLeu (3-OH)-Abu], suggested that for this series, cyclization of the peptide backbone may not play an important role in controlling the kinetics of N,O-acyl migration. In contrast, the disparity in the chemical reactivity of tripeptides [Val-MeLeu (3-OH)-Abu] and [Val-Leu (3-OH)-Abu], indicated that N-methylation of amide bond significantly impacted the kinetics. Various hypothesis are proposed to account for this observation.

    Notes: Abstract Purpose. The objective was to evaluate the degradation profile of the elastase inhibitor DMP 777 and lay the foundation for formulation development. Methods. The pKa was determined by potentiometric titration in mixed-aqueous solvents. The degradation kinetics were studied as a function of pH, buffer concentration, ionic strength, methanol concentration and temperature using a stability-indicating HPLC assay. The degradation products were identified by LC-MS, NMR, and by comparison with authentic samples. Results. The pKa for the protonated piperazine nitrogen was estimated to be 7.04. The pH-rate profile is described by specific acid-, water-, and specific base-catalyzed pathways. The pH of maximum stability is in the range of 4 to 4.5 where water is the principal catalyst in the reaction. Buffer catalysis, primary salt effects and medium effects were observed. The proposed mechanism for acid catalyzed degradation is the rarely observed AAL1 which involves alkyl-nitrogen heterolysis. The driving force for the reaction appears to lie in the stability of the benzylic carbocation. The proposed mechanism for base catalyzed degradation is BAC2 which involves β-lactam ring opening. The β-lactam ring of DMP 777, a monolactam, appears to be as reactive as that in benzylpenicillin in the k OH controlled region where a similar mechanism of hydrolysis should be operative. A contributing factor to this increased reactivity may lie in the reduced basicity of the β-lactam nitrogen making it a good leaving group. Conclusions. The degradation profile indicates that development of a solution dosage form of DMP 777 with adequate shelf-life stability at room temperature is feasible.

PEGylation of proteins is commonly performed to achieve increased in-vivo circulation half-lives. For rHuMGDF, an additional advantage of PEGylation was enhancedin-vitro shelf-life stability.

    Notes: Abstract Purpose. The solubility and physical stability of human Insulin-like Growth Factor I (hIGF-I) were studied in aqueous solutions with different excipients. Methods. The solubility of hIGF-I was determined by UV-absorption and quantification of light blocking particles. The physical stability of hIGF-I was studied with differential scanning calorimetry (DSC) and circular dichroism (CD) spectroscopy. Results. Human IGF-I precipitated at low temperature in the presence of 140 mM benzyl alcohol and 145 mM sodium chloride. CD data showed that the tertiary structure of hIGF-I during these conditions was perturbed compared to that in 5 mM phosphate buffer. In the presence of benzyl alcohol 290 mM mannitol stabilized hIGF-I. Sodium chloride or mannitol by themselves had no effect on either the solubility or the tertiary structure. Benzyl alcohol was attracted to hIGF-I, whereas sodium chloride was preferentially excluded. The attraction of benzyl alcohol was reinforced by sodium chloride leading to salting-out of hIGF-I. The CD-data indicated interactions of benzyl alcohol with phenylalanine in hIGF-I. Thermal denaturation of hIGF-I occurred in all solutions with sodium chloride, whereas mannitol or benzyl alcohol had no effect on the thermal stability. The thermal stability of hlGF-I was thus decreased in 145 mM sodium chloride although it was excluded from hIGF-I. Conclusions. The self-association and thermal aggregation of hIGF-I is driven by hydrophobic interactions. Benzyl alcohol is attracted to hIGF-I and induces changes in the tertiary structure causing hydrophobic attraction of the protein at low temperatures.

    Notes: Abstract Purpose. Protein stabilization during lyophilization has previously focused on optimization of the formulation as well as the freezing and dehydration process parameters. However, the effect of the reconstitution medium has been largely neglected. We have investigated its effect on aggregate formation using recombinant keratinocyte growth factor (KGF). Methods. The protein was lyophilized under suboptimal conditions to induce aggregation and precipitation upon reconstitution with water. A series of additives were examined by UV spectrophotometry and size exclusion chromatography (SEC-HPLC) for their effects on decreasing the degree of KGF aggregation and precipitation by the increase in recovery of soluble monomer. Results. Several additives resulted in a significant reduction of aggregation, including sulfated polysaccharides, surfactants, polyphosphates, and amino acids. A similar effect was achieved by adjusting the ionic strength of the reconstitution medium. SEC-HPLC indicated that the amount of soluble monomer was also increased by these additives suggesting that the recovery of the soluble protein correlates with the native, monomeric protein. Conclusions. These results suggest that optimization of reconstitution conditions will be a useful methodology for increasing the recovery of soluble, active proteins and that for KGF, the recovery of the soluble protein correlates with the native, monomeric form.

The major products of degradation of PNU-67590A are MP by hydrolysis and methylprednisolone 17-suleptanate (17-E) by 21 → 17 acyl migration. The effect of ionic strength on micelle formation and stability of PNU-67590A in aqueous solution was examined. Methods. PNU-67590A solutions at pH 2 and 8 and ionic strength of 0.05, 0.1, 0.2, and 0.4 M were maintained at 25°C in the dark to measure MP and 17-E levels over time. Results. The rate of degradation of micellar PNU-67590A at pH 8 was less than that of monomeric PNU-67590A, and vice versa at pH 2. Increase in ionic strength decreased both the critical micelle concentration of PNU-67590A and the degradation of micelle PNU-67590A at both pHs, resulting in improved overall stability of PNU-67590A. Conclusions. Formulation of PNU-67590A in a concentrated solution with high ionic strength will maximize stability and shelf-life.

    Notes: Abstract Purpose. The long-term stability and nasal absorption characteristics of a basic nasal formulation of azetirelin, a thyrotropin-releasing hormone analog and its absorption enhancement by incorporation of acylcarnitines in the formulation were investigated. Methods. The long-term stability of basic nasal azetirelin formulations at 25° C was predicted by calculation from the Arrhenius plot of the data on 6 months' storage at 40, 50 and 60° C. Nasal azetirelin absorption characteristics were kinetically examined by intranasal administration to rats, determination of plasma azetirelin level by radioimmunoassay, and fitting the data to a two-compartment model including absorption rate. Results. Basic nasal azetirelin formulations of pH 4.0 and pH 5.1 were predicted to be highly stable. Residual azetirelin after 2 years storage at 25° C was greater than 95%. Nasal absorption characteristics of this formulation in the pH 4.0−6.3 range showed pH-dependency, with pH 4.0 showing the highest absolute bioavailability (Bioav) of 17.1%. This nasal Bioav was 21 times greater than that of oral administration (0.8%). Acylcarnitines with 12 or more carbon atoms in the acyl chain greatly enhanced nasal absorption of azetirelin: Bioavs with lauroylcarnitine chloride (LCC) and palmitoylcarnitine chloride were 96.9% and 72.9%, respectively. This enhancement by LCC plateaued at the low concentration of 0.1%. Conclusions. The basic nasal azetirelin formulation at pH 4.0 is stable and shows adequate absorption, with nasal absorption having greater Bioav than oral absorption. The 12-carbon acylate LCC was the strongest enhancer among acylcarnitines and provided near-total delivery of the administered dose to the blood.

    Notes: Abstract Purpose. To investigate the feasibility of producing freeze-dried poly-(ethylene oxide) (PEO)-surface modified nanoparticles and to study their ability to avoid the mononuclear phagocytic system (MPS), as a function of the PEO chain length and surface density. Methods. The nanoparticles were produced by the salting-out method using blends of poly(D,L-lactic acid) (PLA) and poly(D,L-lactic acid-co-ethylene oxide) (PLA-PEO) copolymers. The nanoparticles were purified by cross-flow filtration and freeze-dried as such or with variable amounts of trehalose as a lyoprotectant. The redispersibility of the particles was determined immediately after freeze-drying and after 12 months of storage at −25° C. The uptake of the nanoparticles by human monocytes was studied in vitro by flow cytometry. Results. PLA-PEO nanoparticles could be produced from all the polymeric blends used. Particle aggregation after freeze-drying was shown to be directly related to the presence of PEO. Whereas this problem could be circumvented by use of trehalose, subsequent aggregation was shown to occur during storage. These phenomena were possibly related to the specific thermal behaviours of PEO and trehalose. In cell studies, a clear relationship between the PEO content and the decrease of uptake was demonstrated. Conclusions. The rational design of freeze-dried PEO-surface modified nanoparticles with potential MPS avoidance ability is feasible by using the polymer blends approach combined with appropriate lyoprotection and optimal storage conditions.

    Notes: Abstract Based on the linear analysis of stability, a dispersion equation is deduced which delineates the evolution of a general 3-dimensional disturbance on the free surface of an incompressible viscous liquid jet injected into a gas with swirl. Here, the dimensionless parameterJ e is again introduced, in the meantime, another dimensionless parameterE called as circulation is also introduced to represent the relative swirling intensity. With respect to the spatial growing disturbance mode, the numerical results obtained from solving the dispersion equation reveal the following facts. First, at the same value ofE, in pace with the changing ofJ e , the variation of disturbance and the critical disturbance mode still keep the same characters. Second, the present results are the same as that of S.P. Lin whenJ e 〉1; but in the range ofJ e 〈1, it's no more the case, the swirl decreases the axisymmetric disturbance, yet increases the asymmetric disturbance, furthermore the swirl may make the character of the most unstable disturbance mode changed (axisymmetric or asymmetric); the above action of the swirl becomes much stronger whenJ e ≪1.

    Notes: Abstract By means of the frozen time approach and the Kronecker product, two criteria of asymptotic stability are derived for the linear, time variant dynamic systems with either short time delays or with weak feedback involving arbitrary time delays, respectively. The stability condition of a periodic impacting motion is given by analyzing the propagation of small, arbitrary perturbation from that motion. In numerical simulations, the periodic impacting motions are classified according to the system states before and after an impact. The numerical results show that there exist many types of vibro-impacts and the bifurcation of periodic vibro-impacts is not smooth.

    Notes: Abstract Global stability properties of dynamic two-country models can be easily studied in the case of perfect international capital flows. With internationally constant relative productivities, balanced-growth path values for factor prices will hold on any path leading to the balanced-growth path unless one country experiences a period of no innovation. Innovation rates converge in the case of perfect international knowledge spillovers but long-run consumption levels and trade patterns are path-dependent. GDP per capita is predicted to converge slowly despite the presence of perfect international capital markets and no explicit inclusion of adjustment costs. We assume that each firm incurs quadratic production-adjustment costs if it changes its output. We present a simple necessary and sufficient condition for stability of the model. Using the condition, we compare the stability of this model with the stability of two related Cournot models in which all firms present are followers. It turns out that the Stackelberg model is “more stable” than these two Cournot models.

    Notes: Abstract Recently it has been shown that the conventional notions of stability in the sense of Lyapunov and asymptotic stability can be used to characterize the stability properties of a class of “logical” discrete event systems (DES). Moreover, it has been shown that stability analysis via the choice of appropriate Lyapunov functions can be used for DES and can be applied to several DES applications including manufacturing systems and computer networks (Passino et al. 1994, Burgess and Passino 1994). In this paper we extend the conventional notions and analysis of uniform boundedness, uniform ultimate boundedness, practical stability, finite time stability, and Lagrange stability so that they apply to the class of logical DES that can be defined on a metric space. Within this stability-theoretic framework we show that the standard Petri net-theoretic notions of boundedness are special cases of Lagrange stability and uniform boundedness. In addition we show that the Petri ent-theoretic approach to boundedness analysis is actually a Lyapunov approach in that the net-theoretic analysis actually produces an appropriate Lyapunov function. Moreover, via the Lyapunov approach we provide a sufficient condition for the uniform ultimate boundedness of General Petri nets. To illustrate the Petri net results, we study the boundedness properties of a rate synchronization network for manufacturing systems. In addition, we provide a detailed analysis of the Lagrange stability of a single-machine manufacturing system that uses a priority-based part servicing policy.

    Notes: Abstract We present some new results which definitively explain thebehavior of the classical, heuristic nonlinear relaxation labelingalgorithm of Rosenfeld, Hummel, and Zucker in terms of theHummel-Zucker consistency theory and dynamical systems theory. Inparticular, it is shown that, when a certain symmetry condition is met,the algorithm possesses a Liapunov function which turns out to be (thenegative of) a well-known consistency measure. This follows almostimmediately from a powerful result of Baum and Eagon developed in thecontext of Markov chain theory. Moreover, it is seen that most of theessential dynamical properties of the algorithm are retained when thesymmetry restriction is relaxed. In this paper, a study of two trunk groups carrying telephony traffic is used to show that instabilities can arise if there is a delay in getting feedback information for a network controller. The network controller seeks to balance the traffic in the two trunk groups, which may represent two paths from a source to a destination. An analysis shows how factors such as holding time, controller gain and feedback delay influence stability.     Notes: Abstract We establish the stability of axial motions (steady motions along the lengthwise direction) of nonlinearly elastic loops of string. A key observation here is that a linear combination of the total energy and the total circulation of the string, both of which are conserved quantities, yields an appropriate Liapunov function. From our previous work [5], we know that there are uncountably many shapes corresponding to a given axial speed. Accordingly, we establish “orbitai” stability (modulo this collection of relative equilibria). For a well-defined class of “soft” materials, there is an upper bound on the axial speed sufficient for stability; “stiff” materials are shown to be orbitally stable at any axial speed.

    Notes: Abstract We present a new simulation code able to simulate the entire propagation of laser pulse, from the amplifiers level up to the focusing stage. This algorithm has some new characteristics that we intend to present. It computes the three-dimensional optical propagation equation using no approximation other than its picosecond expression. The stability has been carefully studied so that it can be applied to any geometry. This is a great improvement since, up to now only cylindrical geometry was accessible for accuracy. In this paper we also present a method using Fast Fourier Transform able to evaluate with a high accuracy, Fresnel’s distribution of a focused laser pulse. The advantages provided by our algorithm are its rapidity and its high physical understanding of the focusing phenomena.

    Notes: Abstract We study thermomechanical deformations of a viscoplastic body deformed in simple shear. The strain gradients are taken as independent kinematic variables and the corresponding higher order stresses are included in the balance laws, and the equation for the yield surface. Three different functional relationships, the power law, and those proposed by Wright and Batra, and Johnson and Cook are used to relate the effective strain rate to the effective stress and temperature. Effects of strain hardening of the material and elastic deformations are neglected. The homogeneous solution of the problem is perturbed and the stability of the problem linear in the perturbation variables is studied. Following Wright and Ockendon's postulate that the wavelength whose initial growth rate is maximum determines the minimum spacing between adjacent shear bands, the shear band spacing is computed. It is found that the minimum shear band spacing is very sensitive to the thermal softening coefficient/exponent, the material characteristic length and the nominal strain-rate. Approximate analytical expressions for the critical wave length for heat conducting nonpolar materials and locally adiabatic deformations of gradient dependent materials are also derived.

    Notes: Summary A relative equilibrium of a Hamiltonian system with symmetry is a point of phase space giving an evolution which is a one-parameter orbit of the action of the symmetry group of the system. The evolutions of sufficiently small perturbations of a formally stable relative equilibrium are arbitrarily confined to that relative equilibrium's orbit under the isotropy subgroup of its momentum. However, interesting evolution along that orbit, here called drift, does occur. In this article, linearizations of relative equilibria are used to construct a first order perturbation theory explaining drift, and also to determine when the set of relative equilibria near a given relative equilibrium is a smooth symplectic submanifold of phase space.

    Notes: Abstract The effect of urea on the stability of α-haematite hydrosols in the presence of various monovalent ions was studied on each side of the pzc (pH 8.2) by measurements of the critical coagulation concentration (CCC) of the hydrosol. It was observed that for pHs lower than the pzc, the effect of urea depends strongly on the nature of coagulating ion and on the pH:The CCC values of IO 3 − increases with urea concentrations but those of ClO 4 − and Cl− decreases. For pHs higher than the pzc, an inversion of the cationic adsorption sequence was observed. This behavior is in a good agreement with the well-known action of urea on the structure of water and also with the general property of the α-haematite water interface.

    Notes: Übersicht  Lineare, periodisch zeitvariante Bewegungsgleichungen treten im Hubschrauber- und Windturbinenbau auf. Der Getriebebau und die Rotordynamik liefern weitere Beispiele. In diesem Bericht wird ein systematischer Weg zu ihrer Lösung dargestellt. Mit dem Ansatz von Hill wird die homogene Lösung über die Lösung eines Eigenwertproblems gewonnen. Nach Anpassung an die Anfangsbedingungen liefert sie die Fundamentalmatrix des Systems. Ähnlich wie bei zeitinvarianten Systemen, existieren auch bei zeitvarianten Systemen Orthogonalitätsbedingungen, die die Eigenvektoren erfüllen. Die Eigenvektoren selbst sind allerdings zeitabhängig. Benutzt man die Eigenvektoren als Ansatzvektor zur Berechnung der erzwungenen Schwingungen (Transformation mit der zeitvarianten Modalmatrix des Systems), so gelingt es, die Bewegungsgleichungen des Systems in entkoppelte, zeitinvariante zu überführen. Sie lassen sich in bekannter Weise lösen. Dieses Vorgehen wird auf eine moderne Windkraftanlage angewandt. Sie wurde zunächst mit 372 Freiheitsgraden modelliert, die aber auf 18 vor der numerischen Weiterbehandlung kondensiert wurden. Das Stabilitätsverhalten und die Antwortspektren auf stochastische Anregung durch den Wind wurden auf dem oben beschriebenen Weg ermittelt.

    Notes: Summary The influence of shear deformation on the buckling behavior of a beam supported laterally by a Winkler elastic foundation is studied. A full investigation of the bifurcation points at which, under axial load, the beam becomes critical with respect to one or two simultaneous buckling modes is made. The configurations and stabilities of the equilibrium paths that bifurcate from the critical points are derived. From the results of theoretical analysis, it becomes evident that shear deformation has a considerable effect upon the equilibriums and stabilities of the post-buckling of the beam. The results for the Bernoulli-Euler beam can be obtained as a limiting case for those of the present beam by letting the shear stiffness tend to infinity.

    Notes: Übersicht Lineare, periodisch zeitvariante Bewegungsgleichungen treten im Hubschrauber- und Windturbinenbau auf. Der Getriebebau und die Rotordynamik liefern weitere Beispiele. In diesem Bericht wird ein systematischer Weg zu ihrer Lösung dargestellt. Mit dem Ansatz von Hill wird die homogene Lösung über die Lösung eines Eigenwertproblems gewonnen. Nach Anpassung an die Anfangsbedingungen liefert sie die Fundamentalmatrix des Systems. Ähnlich wie bei zeitinvarianten Systemen, existieren auch bei zeitvarianten Systemen Orthogonalitätsbedingungen, die die Eigenvektoren erfüllen. Die Eigenvektoren selbst sind allerdings zeitabhängig. Benutzt man die Eigenvektoren als Ansatzvektor zur Berechnung der erzwungenen Schwingungen (Transformation mit der zeitvarianten Modalmatrix des Systems), so gelingt es, die Bewegungsgleichungen des Systems in entkoppelte, zeitinvariante zu überführen. Sie lassen sich in bekannter Weise lösen. Dieses Vorgehen wird auf eine moderne Windkraftanlage angewandt. Sie wurde zunächst mit 372 Freiheitsgraden modelliert, die aber auf 18 vor der numerischen Weiterbehandlung kondensiert wurden. Das Stabilitätsverhalten und die Antwortspektren auf stochastische Anregung durch den Wind wurden auf dem oben beschriebenen Weg ermittelt.

    Notes: Summary In this paper, the global behavior of relative equilibrium states of a three-body satellite with flexible connection under the action of the gravitational torque is studied. With geometric method, the conditions of existence of nontrivial solutions to the relative equilibrium equations are determined. By using reduction method and singularity theory, the conditions of occurrence of bifurcation from trivial solutions are derived, which agree with the existence conditions of nontrivial solutions, and the bifurcation is proved to be pitchfork-bifurcation. The Liapunov stability of each equilibrium state is considered, and a stability diagram in terms of system parameters is presented.

    Notes: Abstract Experiments on stability and quench propagation were performed on a NbTi hollow conductor test module allowing heat conduction only between adjacent layers, while the turns in a layer are thermally insulated (2-D winding). The measurements performed over a wide range of operating conditions were used as the experimental data-base for the comparison with the results of the simulations done using the quench analysis code SARUMAN. In the paper the experimental results are compared with the predictions of a 2-D version of SARUMAN, which show that the simulation is able to reproduce the general features of the quench propagation experiments.

Für eine noch allgemeinere Version des Spiels beweisen wir die Existenz einerε-Gleichgewichtsmenge von Strategien. Wir präsentieren auch ein Ergebnis über die Stabilität der Gleichgewichte, das auf der Grad-Theorie beruht.
    Notes: Abstract In this paper, we introduce a generalization of the polymatrix game (a nonzero sum noncooperativen-person game) considered by Howson and relate the problem of computing an equilibrium set of strategies for such a game to the generalized linear complementarity problem of Cottle and Dantzig. For an even more general version of the game we prove the existence of anε-equilibrium set of strategies. We also present a result on the stability of the equilibria based on degree theory.

    Notes: Abstract Metal clusters arranged on nanostructured oxidized silicon wafers are presented as new model catalyst systems. A photoresist layer spun on top of a wafer was patterned by laser interference exposure. The grid obtained after removing the exposed parts of the resist is used as an etching mask. Hollows with diameters of 300 nm and depths between 50 and 60 nm were etched into the oxide layer using wet chemical methods. Two methods were applied to deposit metal clusters (Pd or Cu) in a defined way within the hollows. The particles ranged from 10 to 50 nm in height and from 80 to 200 nm in diameter. The model catalyst systems were characterized by atomic force microscopy and X-ray photoelectron spectroscopy. The method presented here allows us to produce 4 inch wafers that are covered completely by nanometer-sized structures in a reasonable period of time.

    Notes: Abstract This paper is concerned with the algebraic aspects of the classification of pseudofree, locally linear group actions on a simply connected 4-manifold, particularly with the splitting and stability properties of the associated Hermitian intersection module and its isometry group. Our main result is the proof of stability of the equivariant intersection form for a large class of pseudofree actions. We also prove a topological rigidity theorem stating that two locally linear, pseudofree actions on a closed, oriented, simply connected 4-manifold, with the equivariant intersection forms indefinite and of rank at least 3 at each irreducible character, are topologically conjugate by an orientation preserving homeomorphism if and only if their oriented local representations at the corresponding fixed points are linearly equivalent.

    Notes: Abstract The complexation of protonated hexacyclen and pentacyclen with pyrophosphate, selenite, selenate, molybdate and tungstate anions was studied by differential pulse polarography at 25 °C. The stoichiometry and stability of the resulting anion complexes were evaluated from the pH and concentration-dependence of the peak potentials, respectively. The results established 1 : 1 anion receptor complexation in all cases. In the case of all anions studied, hexacyclen was found to form more stable anion complexes than pentacyclen. The fact that the anion-receptor complexation depends on the structural features of both the anions and the macrocycles used is indicative of a steric controlled interaction.

    Notes: Abstract The paper presents an approach to sequence planning consisting in determining assembly sequences defined in terms of mating and non-mating operations and based on a dynamic expansion of the assembly tree obtained using a knowledge base management system. The planner considers the case of a single-robot assembly workcell. The use of stability and the detailed definition of sequences also by means of several non-mating operations are shown to be powerful instruments in the control of the tree expansion. Forward assembly planning has been chosen, in order to minimize the number of stability checks. Backtracking is avoided by combining precedence relations and stability analysis. Hard and soft constrains are introduced to drive the tree expansion. Hard constraints are precedence relations and stability analysis. All operations are associated to costs, which are used as soft constraints. The operation based approach enables one to manage even non-mating operations and to easily overcome the linearity constraint. Costs enable the planner to manage the association among tools and components. The first section of the paper concerns Stability Analysis that is subdivided into Static and Dynamic Stability Analysis. The former is mainly involved in analyzing gravity effects; the latter is mainly involved in evaluate inertia effects due to manipulation. Stability Analysis is implemented in a simplified form. Fundamental assumptions are: no rotational equilibrium condition is considered; for each reaction force only direction and versus, but not magnitude, are considered; friction is neglected. The second section discusses the structure of the planner and its implementation. The planner is a rule based system. Forward chaining and hypothetical reasoning are the inference strategies used. The knowledge base and the data base of the system are presented and the advantages obtained using a rule based system are discussed. The third section shows two planning examples, showing the performance of the system in a simple case and in an industrial test case, the assembly of a microwave branching filter composed of 26 components.

    Notes: Abstract Sensitivity of the returns to scale (RTS) classifications in data envelopment analysis is studied by means of linear programming problems. The stability region for an observation preserving its current RTS classification (constant, increasing or decreasing returns to scale) can be easily investigated by the optimal values to a set of particular DEA-type formulations. Necessary and sufficient conditions are determined for preserving the RTS classifications when input or output data perturbations are non-proportional. It is shown that the sensitivity analysis method under proportional data perturbations can also be used to estimate the RTS classifications and discover the identical RTS regions yielded by the input-based and the output-based DEA methods. Thus, our approach provides information on both the RTS classifications and the stability of the classifications. This sensitivity analysis method can easily be applied via existing DEA codes.

    Notes: Abstract An indirect adaptive control approach is developed in this paper for robots with unknown nonlinear dynamics using neural networks (NNs). A key property of the proposed approach is that the actual joint angle values in the control law are replaced by the desired joint angles, angle velocities and accelerators, and the bound on the NN reconstruction errors is assumed to be unknown. Main theoretical results for designing such a neuro-controller are given, and the control performance of the proposed controller is verified with simulation studies.
