    Beschreibung: This open access book offers the first comprehensive account of the pan-genome concept and its manifold implications. The realization that the genetic repertoire of a biological species always encompasses more than the genome of each individual is one of the earliest examples of big data in biology that opened biology to the unbounded. The study of genetic variation observed within a species challenges existing views and has profound consequences for our understanding of the fundamental mechanisms underpinning bacterial biology and evolution. The underlying rationale extends well beyond the initial prokaryotic focus to all kingdoms of life and evolves into similar concepts for metagenomes, phenomes and epigenomes. The book’s respective chapters address a range of topics, from the serendipitous emergence of the pan-genome concept and its impacts on the fields of microbiology, vaccinology and antimicrobial resistance, to the study of microbial communities, bioinformatic applications and mathematical models that tie in with complex systems and economic theory. Given its scope, the book will appeal to a broad readership interested in population dynamics, evolutionary biology and genomics.

    Beschreibung: Turrialba volcano lies in the southern sector of the Central American Volcanic Front (CAVF) in Costa Rica. The geochemistry of major and trace elements, and Sr and Nd isotopes of a selected suite of volcanic rocks ranging in composition from basaltic andesite to dacite and belonging to the last 10 ka of activity of Turrialba volcano is described, together with the He-, Ne-, and Ar-isotope compositions of fluid inclusions hosted in olivine and pyroxene crystals. Most of the variability in the rock chemistry is consistentwith typical trends of fractional crystallization, but there is an outlying group of andesites that displays an adakite-like composition (with a consistent depletion in high-field-strength elements and a marked enrichment in Sr) and low 3He/4He ratios (7.0–7.2 Ra). The trace-element composition of these rocks is typical of subduction-related magmas influenced by an OIB-like component at the source associated with the subduction of the Galapagos seamounts. The 87Sr/86Sr (0.703612–0.703678) and 143Nd/144Nd (0.512960–0.512984) ratios of the bulk rocks vary within narrowranges, and are among the least-radiogenic isotope signatures of the CAVF volcanoes. The 3He/4He ratios measured in fluid inclusions hosted in olivine crystals (up to 8.1 Ra) are among the highest for the CAVF, and indicate that radiogenic 4He from fluids derived fromthe subducting slab contribute negligibly to the mantle wedge. The difference in He isotopes between most of studied rocks and those showing adakite-like features reasonably reflects two distinct components in the local mantle: (1) a MORB-like component, characterized by the highest He-isotope ratios (7.8–8.1 Ra), and (2) an OIB-like component, characterized by lower He-isotope ratios (7.0–7.2 Ra), coming from the subduction of the Galapagos seamounts. An overview at the regional scale indicates that high He-isotope ratios are peculiar to the two extreme sectors of the CAVF (Costa Rica to the south and Guatemala to the north), whereas in the central sector (Nicaragua) the magma source is probably contaminated by slab fluids. For the past few years Turrialba volcano has been in a volcanic unrest phase that has included a series of explosions, the most recent of which occurred between October 2014 and May 2015. The volcano is subject to an ongoing safety alert due to the possibility of a magmatic eruption. One of the crucial questions to be addressed is the kind of eruption that can be expected, and hence what type of magma is likely to be involved. The high 3He/4He ratios (7.8–8.0 Ra) measured during 2011 at high-temperature fumaroles of Turrialba craters are comparable to those measured in fluid inclusions of basaltic andesites that erupted in 1864–1866, suggesting that the magma currently feeding the shallow plumbing system has similar geochemical characteristics to the most recently erupted magma.

    Beschreibung: Ambrym volcano, in the Vanuatu arc, is one of the most active volcanoes of the Southwest Pacific region, where persistent lava lake and/or Strombolian activity sustains voluminous gas plume emissions. Here we report on the first comprehensive budget for the discharge ofmajor,minor, trace and radioactive volatile species fromAmbrymvolcano, as well as the first data for volatiles dissolved in its basaltic magma (olivine-hosted melt inclusions). In situ MultiGAS analysis of H2O, CO2, SO2 and H2S in crater rim emissions, coupled with filter-pack determination of SO2, halogens, stable and radioactive metals demonstrates a common magmatic source for volcanic gases emitted by its two main active craters, Benbow and Marum. These share a high water content (~93 mol%), similar S/Cl, Cl/ F, Br/Cl molar ratios, similar (210Po/210Pb) and (210Bi/210Pb) activity ratios, as well as comparable proportions in most trace metals. Their difference in CO2/SO2 ratio (1.0 and 5.6–3.0, respectively) is attributed to deeper gasmelt separation at Marum (Strombolian explosions) than Benbow (lava lake degassing) during our measurements in 2007. Airborne UV sensing of the SO2 plume flux (90 kg s−1 or 7800 tons d−1) demonstrates a prevalent degassing contribution (~65%) of Benbow crater in that period and allows us to quantify the total volatile fluxes duringmedium-level eruptive activity of the volcano. Results reveal that Ambrym ranks among the most powerful volcanic gas emitters on Earth, producing between 5% and 9% of current estimates for global subaerial volcanic emissions of H2O, CO2, HCl, Cu, Cr, Cd, Au, Cs and Tl, between 10% and 17% of SO2, HF, HBr, Hg, 210Po and 210Pb, and over 30% of Ag, Se and Sn. Global flux estimates thus need to integrate its contribution and be revised accordingly. Prodigious gas emission from Ambrym does not result from an anomalous volatile enrichment nor a differential excess degassing of its feedingbasalt: this latter contains relativelymodest dissolved amounts ofH2O(≤1.3wt%), CO2 (~0.10 wt%), S (0.075 wt%) and Cl (0.05 wt%), and its degassing under prevalent closed-systemconditions well reproduces the composition of emitted volcanic gases. Instead,we show that the gas discharge is sustained by a very high basalt supply rate of 25m3 s−1, from a large (~ 0.5 km3)magma reservoir probably emplaced at ~3.8 km depth below the summit caldera according to both the H2O-CO2 content of bubble-free melt inclusions and preliminary seismic data. Radioactive disequilibria in the volcanic gases constrain that this reservoir may be entirely renewed in about 240 days. The comparatively low magma extrusion rate requires extensive convective overturn of the basaltic magma column and recycling of the unerupted (denser) degassed magma in the plumbing system, in agreement with textural features of erupted products. Finally, our results suggest that the Indian MORB-type mantle source of Ambrym basalts is modestly enriched in slab-derived water and other volatiles, in agreement with the prevalent volcanoclastic nature of subducted sediments and their lower subduction rate under the central Vanuatu arc due to its collision with the D'Entrecasteaux Ridge.

    Beschreibung: This work presents the results of an extensive geochemical survey aimed at measuring soil CO2 effluxes and soil temperatures over a large portion of Mt. Etna's summit area, coupled with an updated structural survey of the same area. The main goals of this study were i) to find concealed or hidden volcano-tectonic structures in the studied area by detecting anomalous soil gas emissions, ii) to investigate the origin of the emitted gas and the mechanism of gas and heat transport to the surface, iii) to produce a structural model based both on the surface geology and on the soil gas data and, lastly, iv) to contribute to the assessment of hazard from slope failure and crater collapses at Mt. Etna. The results revealed many concealed structural lines that followed the major directions of structural weakness in the summit area of Mt. Etna, mostly due to a combined action of gravitational spreading of the volcano and magma intrusions. Both recent and old volcano-tectonic lines were found to act as pathways for the leakage of magmatic gases to the surface. An important role in driving magmatic gases to the surface is also played by fracturing and faulting due to caldera-forming collapses and smaller crater collapses. Correlation between soil CO2 emissions and soil temperature allowed discriminating areas of active shallow hydrothermal circulation along deep fractures (characterized by high values of both parameters, but mostly soil temperature) from those affected by undeveloped fractures that did not reach the surface (characterized by high CO2 emissions at low temperature). The former corresponded to weak zones of the volcano edifice that were frequently site of past eruptions, indicating that those areas keep a high potential for future opening of eruptive fissures. The latter were likely related to sites where new eruptive fissures may open in the near future due to backward propagation of extensional tectonic stress.

    Beschreibung: The carbon dioxide emissions of volcanoes have been targeted as effective contributors of CO2 to the atmosphere. However, different sources can be effective and active at the same time in the generation and release of CO2 in volcanic zones. Since isotopic fingerprinting of CO2 allows the precise identification of different sources, coupling carbon isotope and CO2 flux measurements enables the evaluation of the mass contribution of each source to the carbon dioxide emissions. This paper accounts for the first extensive spatial analysis of coupled measurements of carbon isotopologues of CO2 in the soil gases and CO2 fluxes discharged by soils on Vulcano Island. An innovative method has been designed, tested and fine-tuned in the laboratory to measure δ13C(CO2) values directly in field using a new type of laser-based isotopologues analyzer, namely a DeltaRay™ (Thermo Fisher Scientific). The method can be used to determine the carbon isotope composition across the full range of CO2 concentrations in the soil gases (0 – 100 vol%). These data have been combined with measurements of the CO2 contents in the soil gases to distinguish CO2 from deep origins from CO2 of biogenic origin in the inhabited area of Vulcano Porto. The method of evaluating the amount of deep-origin CO2 in the soil gases is widely applicable in volcanic and geothermal zones for evaluation and monitoring purposes for both gas and volcanic hazards.

    Beschreibung: We present new real-time measurements of the CO2 concentration and δ13C made from July 16 to 20, 2014 in diluted gases of the active plume emitted by the Central Craters at Mount Etna volcano, Italy. This innovative study involved measuring 13C in plume gases at a very high frequency over 5 days of measurements. The carbon-isotope composition calculated for volcanic CO2 ranged from –1.3‰ to +1.5‰, with uncertainties in the repeated single measurements (i.e., made over periods from 4 to 20 min) that were generally 〈0.7‰, and yet surprisingly varied by larger amounts over the 5-day study period. The range of calculated 13C values mostly overlaps with that indicated for the plume of the Central Craters obtained by discrete sampling and using the isotope-ratio mass spectrometry technique (–2.5‰ 〈 13C 〈 –0.5‰). However, we propose that during particular conditions of volcanic activity, the carbon-isotope composition of CO2 degassed from magma can reach values (up to +1.5‰) that are higher than those reported previously. During this campaign we also made simultaneous measurements of the CO2 and SO2 concentrations using the MultiGAS technique. The volcanic 13C and CO2/SO2 ratios exhibited similar trends over the 5 days of measurements, with the ratios of both tracers peaking on July 16, possibly as a result of the early degassing of CO2 while an eruption was ongoing at Mount Etna. The observed variations and the highest 13C values measured at Mount Etna during this campaign lead to new questions about the variability of this geochemical tracer. The comparisons with the CO2/SO2 ratio also confirm that monitoring 13C in plume gases in real time, coupled to other geochemical tracers, is important for elucidating the magma dynamics at depth.

    Beschreibung: The composition and fluxes of volcanic gases released by persistent open-vent degassing at Bromo Volcano, east Java (Indonesia), were characterised in September 2014 from both in-situ Multi-GAS analysis and remote spectroscopic (dual UVcamera)measurements of volcanic plumeemissions. Our results demonstrate that Bromo volcanic gas is water-rich (H2O/SO2 ratios of 56–160) and has CO2/SO2 (4.1 ± 0.7) and CO2/Stot (3.2 ± 0.7) ratios within the compositional range of other high-temperature magma-derived gases in Indonesia. H2/H2O and H2S/SO2 ratios constrain a magmatic gas source with minimal temperature of ~700 °C and oxygen fugacity of 10-17–10-18 bars. UV camera sensing on September 20 and 21, 2014 indicates a steady daily mean SO2 output of 166 ± 38 t d−1, which is ten times higher than reported from few previous studies. Our results indicate that Bromo ranks amongst the strongest sources of quiescent volcanic SO2 emission measured to date in Indonesia, being comparable to Merapi volcano in central Java. By combining our results for the gas composition with the SO2 plume flux, we assess for the first time the fluxes of H2O (4725 ± 2292 t d−1), CO2 (466 ± 83 t d−1), H2S (25 ± 12 t d−1) and H2 (1.1 ± 0.8) from Bromo. Our study thus contributes a new piece of information to the still limited data base for volcanic gas emissions in Indonesia, and confirms that much remain to be done to fully assess the contribution of this very active arc region to global volcanic gas fluxes.

    Beschreibung: The Principal Cordillera of Central Chile is characterized by two belts of different ages and lithologies: (i) an eastern Mesozoic belt, consisting of limestone- and gypsum-rich sedimentary rocks at the border between Central Chile and Argentina, where the active volcanic arc occurs; and (ii) a western belt of Cenozoic age containing basaltic to andesitic volcanic and volcanoclastic sequences. This distinctive geological setting controls water chemistry of cold and thermal springs in the region, which are fed by meteoric water that circulates through deep regional structures. In the western sector of Principal Cordillera, water–rock interaction processes produce lowTDS, slightly alkaline HCO3 − dominatedwaters, although dissolution of underlyingMesozoic evaporitic rocks occasionally causes SO4 2− and Cl− enrichments. In this area, few Na+–HCO3 − and Na+–SO4 2− waters occurred, being likely produced by a Ca2+–Na+ exchange during water–rock interactions. Differently, the chemical features of Ca2+–Cl−waterswas likely related to an albitization–chloritization process affecting basaltic to andesitic rocks outcropping in this area. Addition of Na+–Cl− brines uprising from the eastern sector through the westverging thrust faults cannot be excluded, as suggested by the occurrence of mantle He (~19%) in dissolved gases. In contrast, in the eastern sector of the study region, mainly characterized by the occurrence of evaporitic sequences and relatively high heat flow,mature Na+–Cl− waters were recognized, the latter being likely related to promising geothermal reservoirs, as supported by the chemical composition of the associated bubbling and fumarolic gases. Their relatively low3He/4He ratios (up to 3.9 Ra)measured in the fumaroles on this area evidenced a significant crustal contamination by radiogenic 4He. The latter was likely due to (i) degassing from 4He-rich magma batches residing in the crust, and/or (ii) addition of fluids interacting with sedimentary rocks. This interpretation is consistent with the measured δ13C-CO2 values (from−13.2 to−5.72‰vs. V-PDB) and the CO2/3He ratios (up to 14.6 × 1010), which suggest that CO2 mostly originates from the limestone-rich basement and recycling of subducted sediments,with an important addition of sedimentary (organic-derived) carbon,whereas mantle degassing contributes at a minor extent. According to geothermometric estimations based on the Na+, K+, Mg2+ and Ca2+ contents, the mature Na+–Cl− rich waters approached a chemical equilibrium with calcite, dolomite, anhydrite, fluorite, albite, K-feldspar and Ca- andMg-saponites at a broad range of temperatures (up to ~300 °C) In the associated gas phase, equilibria of chemical reactions characterized by slowkinetics (e.g. sabatier reaction) suggested significant contributions from hot and oxidizing magmatic gases. This hypothesis is consistent with the δ13C-CO2, Rc/Ra, CO2/3He values of the fumarolic gases. Accordingly, the isotopic signatures of the fumarolic steam is similar to that of fluids discharged from the summit craters of the two active volcanoes in the study area (Tupungatito and Planchón–Peteroa). These results encourage the development of further geochemical and geophysical surveys aimed to provide an exhaustive evaluation of the geothermal potential of these volcanic–hydrothermal systems.

    Beschreibung: This study presents the first geochemical data of fluid discharges collected from February 2010 to March 2015 from the Planchón–Peteroa–Azufre Volcanic Complex (PPAVC), located in the Transitional Southern Volcanic Zone (TSVZ) at the border between Argentina and Chile. During the study period, from January 2010 to July 2011, Peteroa volcano experienced phreatic to phreatomagmatic eruption possibly related to the devastating Maule earthquake occurred on February 27, 2010. The compositional dataset includes low temperature (from 43.2 to 102 °C) gas discharges from (i) the summit of Peteroa volcano and (ii) the SE flank of Azufre volcano, both marked by a significant magmatic fluid contribution, as well as bubbling gases located at the foothill of the Peteroa volcanic edifice, which showed a chemical signature typical of hydrothermal fluids. In 2012, strong compositional changes affected the Peteroa gases fromthe summit area: the acidic gas species, especially SO2, increased, suggesting an input of fluids from magma degassing. Nevertheless, the R/Ra and δ13C–CO2 values decreased, which would imply an enhanced contribution from a meteoric-hydrothermal source. In 2014–2015, the chemical and isotopic compositions of the 2010–2011 gases were partially restored. The anomalous decoupling between the chemical and the isotopic parameters was tentatively interpreted as produced by degassing activity from a small batch of dacitic magma that in 2012 masked the compositional signature of the magmatic fluids released from a basalticmagma that dominated the gas chemistry in 2010–2011. This explanation reliably justifies the observed geochemical data, although the mechanisms leading to the change in time of the dominatingmagmatic fluid source are not clear. At this regard, a geophysical survey able to provide information on the location of the two magma batches could be useful to clarify the possible relationships between the compositional changes that affected the Peteroa fluid discharges and the 2010–2011 eruptive activity.

    Beschreibung: In summer 2013 a toxic and polluting gas blowout (19 tonnes day−1 CO2, 95 kg day−1 CH4) occurred from two shallow boreholes drilled at only 50 m from the International Airport of Rome (Italy), in the town of Fiumicino. Another gas blowout occurred in the same period from a borehole located offshore, 2 km away, also generating sea-water acidification; it lasted only a couple of days. Onshore, CO2was also diffusing fromholes within the soil, particularly toward the airport, generating a soil flux up to 1.8 tonnes day−1. In 3.5 months ~1500 tonnes of CO2 and 5.4 tonnes of CH4 were emitted in the atmosphere. Temporal monitoring of gas geochemistry indicates that in this area a mixing occurs between shallow and pressurized gas pockets, CO2-dominated, but with different chemical (i.e., He/CH4 ratio) and isotopic (3He/4He, δ13C-δDCH4) characteristics. Numerical simulation of CO2 dispersion in the atmosphere showed that dangerous air CO2 concentrations, up to lethal values, were only found near the vents at a height of 0.2 m. Fiumicino is a high blowout risk area, as CO2 rising through deep reaching faults pressurizes the shallowaquifer contained in gravels confined underneath shales of the Tiber delta deposits. The Fiumicino blowout is a typical example of dangerous phenomenon that may occur in urban context lying nearby active or recent volcanoes and requires quick response on hazard assessment by scientists to be addressed to civil protection and administrators.

    Beschreibung: We present a coupled fluid-dynamic and electromagnetic model for volcanic ash plumes. In a forward approach, the model is able to simulate the plume dynamics from prescribed input flow conditions and generate the corresponding synthetic thermal infrared (TIR) image, allowing a comparison with field-based observations. An inversion procedure is then developed to retrieve vent conditions from TIR images, and to independently estimate the mass eruption rate. The adopted fluid-dynamic model is based on a one-dimensional, stationary description of a self-similar turbulent plume, for which an asymptotic analytical solution is obtained. The electromagnetic emission/absorption model is based on Schwarzschild's equation and on Mie's theory for disperse particles, and we assume that particles are coarser than the radiation wavelength (about 10 μm) and that scattering is negligible. In the inversion procedure, model parameter space is sampled to find the optimal set of input conditions which minimizes the difference between the experimental and the synthetic image. Application of the inversion procedure to an ash plume at Santiaguito (Santa Maria volcano, Guatemala) has allowed us to retrieve the main plume input parameters, namely mass flow rate, initial radius, velocity, temperature, gas mass ratio, entrainment coefficient and their related uncertainty. Moreover, by coupling with the electromagnetic model we have been able to obtain a reliable estimate of the equivalent Sauter diameter of the total particle size distribution. The presented method is general and, in principle, can be applied to the spatial distribution of particle concentration and temperature obtained by any fluid-dynamic model, either integral or multidimensional, stationary or time-dependent, single or multiphase. The method discussed here is fast and robust, thus indicating potential for applications to real-time estimation of ash mass flux and particle size distribution, which is crucial for model-based forecasts of the volcanic ash dispersal process.

    Beschreibung: Naples is a large city located between two active volcanic areas: Campi Flegrei to theWand Vesuvius to the SE. The Solfatara crater, inside the caldera of the Campi Flegrei and nearest to the western quarters of the city, is a prodigious source of natural CO2 with a mean emission rate of 1067 ton/d, i.e. seven times higher than that of Vesuvius(151 ton/d). This study shows that the area around the Solfatara and part of the urban area of Naples are affected by the volcanic plume when atmospheric circulatory patterns are dominated by the locally frequent sea breezes. Under these conditions the CO2 content in the air increases above normal values, reaching more than 1000 ppm in proximity to the Solfatara crater to a few tens of ppm several kilometres from the source. Although these values do not indicate a health risk even under the most unfavourable atmospheric conditions, the volcanic source contributes to the total CO2 burden from all urban emissions and hence to overall air quality. An emission rate ten times higher than the present one would lead to an air CO2 concentration in excess of recommended health protection thresholds.

    Beschreibung: We installed a permanent SO2 camera system on Stromboli, Italy, in May 2013, in order to improve our capacity to monitor the SO2 emissions from this volcano. The camera collects images of SO2 concentrations with a period of ~ 10 s, allowing quantification of short-term processes, such as the gas released during the frequent explosions which are synonymous with Stromboli. It also allows quantification of the quiescent gas flux, and therefore comparison with the FLAME network of scanning ultraviolet spectrometers previously installed on the island. Analysis of results from the SO2 camera demonstrated a good agreement with the FLAME network when the plume was blown fully into the field of view of the camera. Permanent volcano monitoring with SO2 cameras is still very much in its infancy, and therefore this finding is a significant step in the use of such cameras for monitoring, whilst also highlighting the requirement of a favourable wind direction and strength. We found that the explosion gas emissions are correlated with seismic events which have a very long period component. There is a variable time lag between event onset time and the increase in gas flux observed by the camera as the explosion gas advects into the field of view of the camera. This variable lag is related to the plume direction, as shown by comparison with the plume location detected with the FLAME network. The correlation between explosion gas emissions and seismic signal amplitude show is consistent with a gas slug-driven mechanism for seismic event production. Comparison of the SO2 camera measurements of the quiescent gas flux shows a fair quantitative agreement with the SO2 flux measured with the FLAME network. Overall, the SO2 camera complements the FLAME network well, as it allows frequent quantification of the explosion gas flux produced by Stromboli, whose signal is in general too brief to be measured with the FLAME network. Further work is required, however, to fully automate the calculation of SO2 flux from the SO2 images captured with the camera, and to adequately account for scattering effects.

    Beschreibung: With this short communication we address the principal issues raised by Delmelle et al. (2014) in relation to the work of Bagnato et al. (2013) concerning the 2010 eruption of Eyjafjallajo¨ kull, Iceland. The principal conclusions of the work of Bagnato et al. (2013) include the observation that protracted gas-aerosol interaction in the plume promotes selective leaching of cation species from ash, with alkalis and Ca (and, among trace elements, Zn and Cu) being more rapidly re-mobilized (and transferred to soluble surface salts) relative to more inert elements (Mg, Ti). They also observed that adsorption onto ash surfaces is a major atmospheric sink of volcanic acidic gases, with 282 tons of elemental sulfur and 605–691 tons of halogens being daily ground deposited via ash over Iceland in early May 2010. Acidic gas adsorption onto ash increases almost linearly with plume aging (e.g., upon increasing in-plume residence times of ash and gases), and is seen to proceed at about 3 time faster rates for HF than for SO2 and HCl. However, Delmelle et al. (2014) criticized our strategies for data acquisition, processing and interpretation. They also raised some objections concerning several key topics explored by Bagnato et al. (2013), with a special focus on the discussion of rates of interaction between ash particles and gases in a volcanic cloud, and the consequent formation of soluble salts on ash surfaces. They also considered incorrect the estimate of depositional fluxes and volatile budget for the Eyjafjallajo¨ kull eruption. While we appreciate the in-depth analysis of Delmelle et al. (2014), we show that most of their criticisms derive from a partial and sometimes incorrect understanding of the work of Bagnato et al. (2013), which overall led to unsupported conclusions and misleading analysis of the original results. Here, we present a detailed response to the comments of Delmelle et al. (2014), accompanied by additional explicative material. The principal conclusions presented in Bagnato et al. (2013) are given additional support by this complementary note.

    Beschreibung: We have analyzed the carbon isotopic composition of CO2, methane, ethane, propane and n-butane, the hydrogen isotopic composition of methane as well as total concentrations of gas constituents contained in theMediterranean volcanic–hydrothermal discharges of Nisyros (Greece), Vesuvio, La Solfatara, Ischia and Pantelleria (all Italy) to determine the origin of the hydrocarbons. Isotopic criteria conventionally used for hydrocarbon classification suggest thermogenic origins, except for Pantelleria, for which an abiogenic origin is indicated. These findings would imply that thermogenic sources can provide methane/(ethane + propane) concentration ratios as high as those usually observed for microbial hydrocarbons. However, additional consideration of gas concentration data challenges the suitability of conventional criteria for the classification of hydrocarbons emanating from hydrothermal environments. Methane seems to be in close equilibrium with co-occurring CO2, whereas its higher chain homologues are not. Therefore, it cannot be excluded that methane on the one hand and ethane, propane and n-butane on the other hand have distinct origins. The carbon isotopic composition of methane might be controlled by the carbon isotopic composition of co-occurring inorganic CO2 and by hydrothermal temperatures whereas the carbon isotopic composition of the higher n-alkanes could correspond to the maturity of organic matter and/or to the residence time of the gasses in the source system

    Beschreibung: Total CO2 output from soil gas and plume, discharged from the Stromboli Island, was estimated. The CO2 emission of the plume emitted from the active crater was estimated on the basis of the SO2 crater output and C/S ratio, while CO2 discharged through diffuse soil emission was quantified on the basis of 419 measurements of CO2 fluxes from the soil of the whole island, performed by using the accumulation chamber method. The results indicate an overall output of ≅416 t day−1 of CO2 from the island. The main contribution to the total CO2 output comes from the summit area (396 t day−1), with 370 t/day from the active crater and 26 t day−1 from the Pizzo sopra La Fossa soil degassing area. The release of CO2 from peripheral areas is ≅20 t day−1 by soil degassing (Scari area mainly). The result of the soil degassing survey confirms the persistence of the highest CO2 degassing areas located on the North-East crater side and Scari area.

    Beschreibung: Here, we report the first continuous data of geochemical parameters acquired directly from the active summit crater of Vulcano. This approach provides a means to better investigate deep geochemical processes associated with the degassing system of Vulcano Island. In particular, we report on soil CO2 fluxes from the upper part of Vulcano, a closed-conduit volcano, from September 2007 to October 2010. Large variations in the soil CO2 and plume SO2 fluxes (order of magnitude), coinciding with other discontinuous geochemical parameters (CO2 concentrations in fumarole gas) and physical parameters (increase of shallow seismic activity and fumarole temperatures) have been recorded. The results from this work suggest new prospects for strengthening geochemical monitoring of volcanic activity and for improving the constraints in the construction of a “geochemical model”, this being a necessary condition to better understand the functioning of volcanic systems.

    Beschreibung: Soil carbonates are key features in soils of arid and semiarid environment, playing an important role from pedogenetic, landscape history, paleoclimatic and environmental points of view. The objectives of this work were (i) to study pathways of pedogenic carbonate (PC) formation, (ii) to distinguish between lithogenic and pedogenic inorganic C by using the natural C isotope abundance, and (iii) to estimate the soil C pools in a gypsiferous semiarid Mediterranean environment (Sicily, Italy). Five soil pedons developed on calcareous and non-calcareous parent materials from Holocene (10,000 years BP) to Upper Tortonian (7.2–5.3 Ma BP) in age were surveyed. During field soil description, the highest stage of carbonate morphology was found in soils developed on non-calcareous Holocene colluvial deposits (youngest deposits in age) which also showed the highest amount of PC. The great amount of PC in soils developed on youngest deposits was ascribed to a soil–landscape relationships. Being located in a doline overhung by gypsum outcrops, precipitation of Ca2+ from gypsum dissolved by rainfall and biogenic CO2 is reliable. The significant positive relationship between soil organic C and pedogenic carbonates δ13C values confirms that PC was formed from biogenic CO2. Organic C pool in the first cubic meter of soil ranged from 17 to 42 kg, whilst pedogenic inorganic C pool from 2.8 to 30.7 kg. The estimated rate of inorganic C accumulation in soils developed on youngest deposits was 2.5 g m−3 y−1, whereas the rate was negligible on older parent material. The hypothesized pathways of PC formation were ex-novo precipitation of gypsum–Ca2+ and biogenic CO2 and dissolution of lithogenic CaCO3 and re-precipitation of Ca2+ with biogenic CO2. From an environmental prospective, investigated soils may act as a sink of C when Ca2+ from gypsum is available for the formation of pedogenic carbonates.

    Beschreibung: Volatile metal(loid)s are known to be emitted from volcanoes worldwide.We tested the suitability of active moss monitoring for tracking volatilemetal(loid)s released fromthe fumarolic field on Vulcano Island, Italy, and differentiated fumaroles from other sources of gaseous and particulate trace elements such as sea spray and soil.Metal(loid) accumulation on the mosses per day did depend neither on the state of the exposed moss (dead or living) nor exposure time (3, 6, or 9 weeks). After collection, mosses were digested with either HNO3/H2O2 or deionized water and analyzed by ICP-MS.While for most elements both extraction methods yielded similar concentrations, higher concentrations were observed e.g. for Pb in the stronger HNO3/H2O2 extracts, indicating the presence of particles, which were not digested and removed by filtration in deionized water extracts. Due to their ubiquitous detection in comparable concentrations at all 23 moss monitoring stations all over the island, Li, Mg and Sr were attributed to sea spray origin. Iron, Co, W, V, Pb, Cr, Mo, and Ba occurred predominantly at the crater, where the soil was not covered by vegetation, and thus likely represent soil-borne particulate transport. Arsenic, Sb, S, Se, Tl, Bi, and I showed a clear concentration maximum within the fumarolic field. Concentrations gradually decreased along a transect in wind direction fromthe fumaroles, which confirms their volcanic origin. Activemossmonitoring thus proved to be an inexpensive and easy-to-apply tool for investigations of volcanic metal(loid) emissions and distributions enabling differentiation of trapped elements by their source of origin.

Most of this heat (57%) is the result of geothermal warming while the remaining 43% is due to gravitational potential energy dissipation. This result indicates that a large area of the central Apennines is very hot with heat flux values 4300 mWm−2. These values are higher than those measured in the magmatic and famously geothermal provinces of Tuscany and Latium and about 1/3 of the total heat discharged at Yellowstone. This finding is surprising because the central Apennines have been thought to be a relatively cold area. Translated by CO2 rich fluids, this heat anomaly suggests the existence of a thermal source such as a large magmatic intrusion at depth. Recent tomographic images of the area support the presence of such an intrusion visible as a broad negative velocity anomaly in seismic waves. Our results indicate that the thermal regime of tectonically active areas of the Earth, where meteoric waters infiltrate and deeply circulate, should be revised on the basis of mass and energy balances of the groundwater systems.

    Beschreibung: Between January 2011 and April 2012, Santorini volcano (Greece) experienced a period of unrest characterised by the onset of detectable seismicity and caldera-wide uplift. This episode of inflation represented the first sizeable intrusion of magma beneath Santorini in the past 50 years. We employ a new approach using 222 Rn– δ 13 C systematics to identify and quantify the source of diffuse degassing at Santorini during the period of renewed activity. Soil CO 2 flux measurements were made across a network of sites on Nea Kameni between September 2010 and January 2012. Gas samples were collected in April and September 2011 for isotopic analysis of CO 2 ( δ 13 C), and radon detectors were deployed during September 2011 to measure ( 222 Rn). Our results reveal a change in the pattern of degassing from the summit of the volcano (Nea Kameni) and suggest an increase in diffuse CO 2 emissions between September 2010 and January 2012. High-CO 2 -flux soil gas samples have δ 13 C ∼ 0 .Using this value and other evidence from the literature we conclude that these CO 2 emissions from Santorini were a mixture between CO 2 sourced from magma, and CO 2 released by the thermal or metamorphic breakdown of crustal limestone. We suggest that this mixing of magmatic and crustal carbonate sources may account more broadly for the typical range of δ 13 CvaluesofCO 2 (from ∼− 4 to ∼+ 1 )in diffuse volcanic and fumarole gas emissions around the Mediterranean, without the need to invoke unusual mantle source compositions. At Santorini a mixing model involving magmatic CO 2 (with δ 13 C of − 3 ± 2 and elevated ( 222 Rn)/CO 2 ratios ∼ 10 5 –10 6 Bqkg − 1 )andCO 2 released from decarbonation of crustal limestone (with ( 222 Rn)/CO 2 ∼ 30–300 Bqkg − 1 ,and δ 13 Cof + 5 ) can account for the δ 13 C and ( 222 Rn)/CO 2 characteristics of the ‘high flux’ gas source. This model suggests ∼ 60% of the carbon in the high flux deep CO 2 end member is of magmatic origin. This combination of δ 13 Cand( 222 Rn) measurements has potential to quantify magmatic and crustal contributions to the diffuse outgassing of CO 2 in volcanic areas, especially those where breakdown of crustal limestone is likely to contribute significantly to the CO 2 flux

    Beschreibung: Copahue volcano is part of the Caviahue–Copahue Volcanic Complex (CCVC),which is located in the southwestern sector of the Caviahue volcano-tectonic depression (Argentina–Chile). This depression is a pull-apart basin accommodating stresses between the southern Liquiñe–Ofqui strike slip and the northern Copahue–Antiñir compressive fault systems, in a back-arc setting with respect to the Southern Andean Volcanic Zone. In this study, we present chemical (inorganic and organic) and isotope compositions (δ13C-CO2, δ15N, 3He/4He, 40Ar/36Ar, δ13C-CH4, δD-CH4, and δD-H2O and δ18O-H2O) of fumaroles and bubbling gases of thermal springs located at the foot of Copahue volcano sampled in 2006, 2007 and 2012. Helium isotope ratios, the highest observed for a Southern American volcano (R/Ra up to 7.94), indicate a non-classic arc-like setting, but rather an extensional regime subdued to asthenospheric thinning. δ13C-CO2 values (from −8.8‰ to −6.8‰ vs. V-PDB), δ15N values (+5.3‰ to +5.5‰ vs. Air) and CO2/3He ratios (from 1.4 to 8.8 × 109) suggest that the magmatic source is significantly affected by contamination of subducted sediments. Gases discharged from the northern sector of the CCVC show contribution of 3He-poor fluids likely permeating through local fault systems. Despite the clear mantle isotope signature in the CCVC gases, the acidic gas species have suffered scrubbing processes by a hydrothermal system mainly recharged by meteoric water. Gas geothermometry in the H2O-CO2-CH4-CO-H2 system suggests that CO and H2 re-equilibrate in a separated vapor phase at 200°–220 °C. On the contrary, rock–fluid interactions controlling CO2, CH4 production from Sabatier reaction and C3H8 dehydrogenation seem to occur within the hydrothermal reservoir at temperatures ranging from 250° to 300 °C. Fumarole gases sampled in 2006–2007 show relatively low N2/He and N2/Ar ratios and high R/Ra values with respect to those measured in 2012. Such compositional and isotope variations were likely related to injection of mafic magma that likely triggered the 2000 eruption. Therefore, changes affecting the magmatic systemhad a delayed effect on the chemistry of the CCVC gases due to the presence of the hydrothermal reservoir. However, geochemical monitoring activities mainly focused on the behavior of inert gas compounds (N2 and He), should be increased to investigate the mechanism at the origin of the unrest started in 2011.

    Beschreibung: Measurements of soil fluxes of hydrothermal gases, with special emphasis on C6H6, as well as chemical composition of mono-aromatic compounds in fumaroles and air, were carried out in April 2012 at the Solfatara crater (Campi Flegrei, Southern Italy) to investigate the distribution and behavior of these species as they migrate through the soil from their deep source to the atmosphere. Soil fluxes of CO2, CH4 and C6H6 exhibit good spatial correlation, suggesting that diffuse degassing is mainly controlled by local fractures. The calculated total output of diffuse C6H6 from Solfatara is 0.10 kg day 1, whereas fluxes of CO2 and CH4 are 79 103 and 1.04 kg day 1, respectively. A comparison between soil gas fluxes and fumarole composition reveals that within the crater soil CH4 is significantly affected by oxidation processes, which are more efficient for low gas fluxes, being dependent on the residence time of the uprising hydrothermal gases at shallow depth. Benzene degradation, mainly proceeding through oxidation via benzoate, seems to be strongly controlled by the presence of a shallow SO2 4 -rich aquifer located in the central and southwestern sectors of the crater, suggesting that the process is particularly efficient when SO2 4 acts as terminal electron acceptor (SO4 reduction). Relatively high C6H6/C7H8 ratios, typical of hydrothermal fluids, were measured in air close to the main fumarolic field of Solfatara crater. Here, C6H6 concentrations, whose detection limit is 0.1 lgm 3, are more than one order of magnitude higher than the limit value for ambient air (5 lgm 3). This suggests that hydrothermal fluids have a strong impact on air quality in the immediate surroundings of the fumarolic vents. Significant concentrations of endogenous mono-aromatics were also detected in air samples collected from the northern and western sides of the crater, where these gas compounds are mostly fed by diffuse degassing through the crater bottom soil.

    Beschreibung: Herein we report on the chemical and isotopic (C, H, O, and He) compositions of the fluids from La Fossa crater fumaroles of Vulcano from 1999 to 2010. Consistent with records obtained since the end of the 1980s, our data show that the geochemical features of the fumarole system have experienced several episodes of remarkable change, each lasting no more than a few months. Typical signatures of these short-term anomalies are large increments in CO2, N2, and He concentrations, coupled to increased 13C/12C isotopic ratios, but their meaning remains widely debated. Within a model of fumarolic fluids based on mixing between hydrothermal and magmatic endmembers, we have developed a novel approach to constrain chemical (He/ CO2 and N2/He) and isotopic (13C/12C, D/H, and 3He/4He) ratios of the magmatic endmember during the short-term anomalies. Although much of the geochemical variability in fumaroles results from changes in mixing proportions, the magmatic fluid unquestionably shows significant variations in time. The magmatic He/CO2, N2/He, 13C/12C, and 3He/4He values throughout 1988–1996 differed from those feeding the anomaly at the end of 2004. Early clues of the new magmatic fluid appeared in 1998–1999, far from any short-term anomaly, whereas new and old magmatic fluids coexisted after 2004. We quantitatively prove that the detected geochemical changes are consistent with the degassing path of a magma having a latitic composition, and suggest the presence of two magma ponding levels at slightly different pressures, where bubble–melt decoupling can occur. The different He-isotope compositions at these levels suggest low hydraulic connectivity typical of a complex reservoir with dike and sill structures. In this framework, the short-term geochemical anomalies are probably due to gas accumulation at the top of magma bodies followed by massive escape, or activation of new degassing levels in the reservoir, for which the stress field almost certainly plays a key role. Such a scenario explains the observed increases in both fumarole output and shallow high-frequency seismicity (due to increased pore pressure) during the anomalies, while being consistent with the concomitant absence of any deep seismicity or ground deformation, eventually related to magma movement.

    Beschreibung: Herein, we present a method for continuous measurement of soil CO2 flux that is completely new and distinct from existing instruments. The foremost difference is that instead of using an infrared gas analyser (IRGA), the new device measures soil CO2 flux by means of a simple pressure sensor, measuring pressure transients inside a closed polymeric tube inserted into the soil. This allows continuous measurements even in soil placed in environments that could potentially damage IRGA. In addition, due to the innovative operating principle, measurements of soil CO2 flux can be effortlessly performed also in strongly harsh weather conditions. Theoretical equations were derived for calculating soil CO2 flux solely using measured transient values. The reliability of the equations was rigorously tested with a variety of experiments. Continuous measurements over four months, acquired in a high-emission area on the Island of Vulcano, compared favourably with the data obtained using an established method.

    Beschreibung: Our study is aimed to develop a 3D physical model of the Campi Flegrei geothermal system, in order to achieve a more accurate and comprehensive representation of the hydrothermal processes occurring in the caldera. The new model, developed by using the TOUGH2 code simulator, accounts for the caldera rocks' physical properties, bathymetry and water table topography. In particular, the computational domain is constrained by density values obtained by tomographic inversion of gravity data collected during several surveys at CF both onshore and offshore the caldera. Empirical relations between density and porosity and between porosity and permeability, derived by published data on samples cored in deep wells or collected in outcrops, allowed us to characterize the main rocks physical parameters controlling the dynamic of the CF geothermal system. We have performed and compared several simulations investigating the effects of the injection at depth, underneath Solfatara crater, of a hot gaseous mixture rich in CO2. We show that, with respect to the available literature on 2D axisymmetric models, the effects of the water table topography together with the bathymetry and the heterogeneous distribution of the rocks' physical properties, lead to important differences in the hydrothermal circulation of fluids at CF. These constraints allow the activation of convective cells with different behaviors, which produce variable patterns of temperature inside the hydrothermal system. As a consequence, the predominant effect is again represented by a central plume below the Solfatara crater, but with a non-axisymmetric structure and a wider extension. Additionally, high temperature zones are present near the coastline and in the middle part of the submerged area of the caldera with a SE–NW alignment. Moreover, our results indicate that, the submerged part of the CF caldera would deserve a more accurate study and survey, being affected by phenomenon of heating and degassing. This information could be very useful in terms of hazard assessment and volcanic risk mitigation in such an active and densely inhabited volcanic and geothermal area.

    Beschreibung: Epidermis micromorphology of in situ Erica arborea L. exposed for generations to long-term effect of volcanic gases in Pisciarelli and Solfatara di Pozzuoli areas have been studied by X-ray analyses, SEM and TEM observations. In particular, the aim of this study is to investigate the effects of volcanic gases on extant and possibly fossil plants. Plants of the same species living in a nearby control site were also studied for comparison. SEM coupled with EDX analysis was used to localize different elements within the leaves (mesophyll, cell wall and cuticle). After conventional and cryo preparation, SEM of mature leaves ascertained that the abaxial side is more serrate in fumigated leaves and hairs, epicuticular wax alterations have also been noted. Leaves experiencing chronic fumigation display stomata more sunken with respect to the epidermal surface. TEM of transverse and longitudinal sections of cuticle showed an outer A2 granular amorphous layer and external to a B1 fibrillous layer. Significant statistical variations of ultrastructural components of the cuticle revealed a response of E. arborea to this extreme environment. At the ultrastructural level, significant variations in thickness of the cell wall plus cuticle, cell wall and A2 layer among fumigated and non-fumigated leaves have been found. In the studied localities a positive correlation between atmospheric CO2 concentration and the thickness of A2 layer also exists. The results are of interest being applicable in the understanding of plant cuticle responses during periods of normal vs. volcanic activity.

    Beschreibung: We present the first chlorine isotope compositional data for gases and lavas from Mount Etna (Italy), and to our knowledge, of active Mediterranean volcanism. We investigated lavas erupted and gases discharged during 2008-2011 from a high-temperature fumarole (HT; 〉300°C) and plume gases from both North East and Central Craters. Most of the samples vary in a narrow range of chlorine isotope composition (37Cl values ≈ 0 ± 0.7‰) with gases partially overlapping with rocks. Only HT gases sampled in 2009 have been clearly affected by secondary processes (37Cl values 〉 15.9‰), resulting in partial removal of chlorine and isotopic fractionation producing a 37Cl enrichment in the residual gaseous HCl. These secondary processes also affect, although to a lesser extent, plume gases from North East Crater (NEC). Although post-magmatic processes are able to modify the chlorine isotope composition, 37Cl values are not affected by magma degassing for residual fractions ≥ 0.3 in the melt, or any effect is within our data variability. Finally, 37Cl values and Cl/K ratios of magmatic chlorine constrain the Etnean source to be compatible with depleted mantle (DMM) contaminated by altered oceanic crust (AOC), in agreement with indications from more common isotopic tracers of mantle processes.

    Beschreibung: Passive samplers were used to measure the atmospheric concentrations of SO2 naturally emitted at three volcanoes in Italy (Etna, Vulcano and Stromboli) and of H2S naturally emitted at three volcanic/geothermal areas in Greece (Milos, Santorini and Nisyros). The measured concentrations and dispersion patterns varied with the strength of the source (open conduits or fumaroles), the meteorological conditions and the area topography. At Etna, Vulcano and Stromboli, SO2 concentrations reach values that are dangerous to people affected by bronchial asthma or lung diseases (〉1000 μg m−3). H2S values measured at Nisyros also exceed the limit considered safe for the same group of people (〉3000 μg m−3). The data obtained using passive samplers represent time-averaged values over periods from a few days up to 1 month, and hence concentrations probably reached much higher peak values that were potentially also dangerous to healthy people. The present study provides evidence of a peculiar volcanic risk associated with tourist exploitation of active volcanic areas. This risk is particularly high at Mt. Etna, where the elderly and people in less-than-perfect health can easily reach areas with dangerous SO2 concentrations via a cableway and off-road vehicles

    Beschreibung: Fluorine adsorption experiments were performed on 28 samples of the first 5 cm of topsoil collected on the flanks of Mt. Etna. The soil samples were equilibrated with F-rich rainwater (3.25 mg/L) at a soil/water weight ratio of 1/25. Aliquots of the supernatant were collected after 1, 7, 72, 720 and 5640 h and analysed for F content. The soil samples could be subdivided into three groups based on their F-adsorption behaviours after 1 h and at the end of the experiment: (1) negative adsorption (F released from the soil to the solution) after 1 h and negative or moderately positive adsorption at the end, (2) from negative after 1 h to strongly positive adsorption at the end, and (3) always strong positive adsorption. The adsorption capacity of the soils was positively correlated with the soil pH, the contents of finer granulometric fractions (clay and silt) and the weathering stage (as quantified by the chemical alteration index). The most F adsorbing soils are found at the periphery of the volcano where aquifers are more vulnerable to contamination due to the shallower depth of the water table. This study further evidences the importance of the Etnean soils in protecting groundwater from an excessive magmatic F input.

    Beschreibung: In this paper, we report four years of soil CO2 emission data measured monthly at 130 sites in two peripheral areas of Mt Etna Volcano that are well known for their high discharge rates of volcanic gas. We remove the influence of atmospheric parameters, and by means of statistical analyses, we (i) demonstrate that variations in CO2 emissions are due mainly to CO2 of a deep origin and (ii) quantify the total amounts of CO2 derived from a deep magma source. Periods of anomalous deep degassing are identified in both areas. A comparison of the timing of these anomalies and geophysical data indicates that the periods of anomalous degassing can be mostly ascribed to intrusions of fresh magma into the Etna plumbing system, which is in agreement with many previous works. Based on the existing literature, we formulate an interpretative framework of magma migration within the plumbing system, consistent with temporal trends in the observed anomalies. Finally, we reconstruct the processes of recent magma ascent at Mt Etna based on our interpretative framework, published geophysical data, and records of volcanic activity.

    Beschreibung: From October 2008 to November 2009, soil CO2, radon and structural field surveys were performed on Mt. Etna, in order to acquire insights into active tectonic structures in a densely populated sector of the south-eastern flank of the volcano, which is involved in the flank dynamics, as highlighted by satellite data (InSAR). The studied area extends about 150 km2, in a sector of the volcano where InSAR results detected several lineaments that were not well-defined from previous geological surveys. In order to validate and better constrain these features with ground data evidences, soil CO2 and soil radon measurements were performed along transects roughly orthogonal to the newly detected faults, with measurement points spaced about 100 m. In each transect, the highest CO2 values were found very close to the lineaments evidenced by InSAR observations. Anomalous soil CO2 and radon values were also measured at old eruptive fractures. In some portions of the investigated area soil gas anomalies were rather broad over transects, probably suggesting a complex structural framework consisting of several parallel volcano-tectonic structures, instead of a single one. Soil gas measurements proved particularly useful in areas at higher altitude on Mt. Etna (i.e. above 900 m asl), where InSAR results are not very informative/ are fairly limited, and allowed recognizing the prolongation of some tectonic lineaments towards the summit of the volcano. At a lower altitude on the volcanic edifice, soil gas anomalies define the active structures indicated by InSAR results prominently, down to almost the coastline and through the northern periphery of the city of Catania. Coupling InSARwith soil gas prospectingmethods has thus proved to be a powerful tool in detecting hidden active structures that do not show significant field evidences.

    Beschreibung: The ordinary, low intensity, activity of Stromboli volcano is sporadically interrupted by more energetic events termed, depending on their intensity, “major explosions” and “paroxysms”. These short-lived energetic episodes represent a potential risk to visitors to the highly accessible summit of Stromboli. Observations made at Stromboli over the last decade have shown that the composition of gas emitted from the summit craters may change prior to such explosions, allowing the possibility that such changes may be used to forecast these potentially dangerous events. In 2008 we installed a novel, remote-controlled, open-path FTIR scanning system called Cerberus at the summit of Stromboli, with the objective of measuring gas compositions from individual vents within the summit crater terrace of the volcano with high temporal resolution and for extended periods. In this work we report the first results from the Cerberus system, collected in August-September 2009, November 2009 and May-June 2010. We find significant, fairly consistent, intra-crater variability for CO2/SO2 and H2O/CO2 ratios, and relatively homogeneous SO2/HCl ratios. In general, the southwest crater is richest in CO2, and the northeast crater poorest, while the central crater is richest in H2O. It thus appears that during the measurement period the southwest crater had a somewhat more direct connection to a primary, deep degassing system; whilst the central and northeast craters reflect a slightly more secondary degassing nature, with a supplementary, shallow H2O source for the central crater, probably related to puffing activity. Such water-rich emissions from the central crater can account for the lower crystal content of its eruption products, and emphasise the role of continual magma supply to the shallowest levels of Stromboli's plumbing system. Our observations of heterogeneous crater gas emissions and high H2O/CO2 ratios do not agree with models of CO2-flushing, and we show that simple depressurisation during magma ascent to the surface is a more likely model for H2O loss at Stromboli. We highlight that alternative explanations other than CO2 flushing are required to explain distributions of H2O and CO2 amounts dissolved in melt inclusions. We detected fairly systematic increases in CO2/SO2 ratio some weeks prior to major explosions, and some evidence of a decrease in this ratio in the days immediately preceding the explosions, with periods of low, stable CO2/SO2 ratios between explosions otherwise. Our measurements, therefore, confirm the medium term (~ weeks) precursory increases previously observed with MultiGas instruments, and, in addition, reveal new, short-term precursory decreases in CO2/SO2 ratios. immediately prior to the major explosions. Such patterns, if shown to be systematic, may be of great utility for hazard management at Stromboli's summit. Our results suggest that intra-crater CO2/SO2 variability may produce short-term peaks and troughs in CO2/SO2 time series measured with in-situ MultiGas instruments, due simply to variations in wind direction.

    Beschreibung: Ambrym is one of the most actively erupting basaltic volcanoes in the Vanuatu island arc. Scoria clasts collected from a fallout deposit in the inner terrace of its Benbow active crater were analyzed through series of synchrotron X-ray computed microtomographic experiments, as well as permeability measurements and simulations. Our goal was to reconstruct and visualize scoria textures in 3D and to quantify vesicularity, permeability, vesicle sizes and distributions in order to understand how gas moves in and out of Ambrym basaltic magma. We find that vesicle size distributions in the volume range between ~ 103 and 1010 μm3 define two scoria classes. Vesicle size distributions in the low-to-moderately (0.44–0.67) vesicular samples can be fit by power laws with an exponent of 1 ± 0.2; distributions in the highly vesicular (0.86–0.88) samples can be fit by power laws with a higher exponent (1.4 to 1.7), as well as by exponential fits. Highly vesicular samples exhibit a very pronounced large vesicle, consisting of networks of smaller, interconnected vesicles, that is more than three orders of magnitude larger in volume than all other vesicles in each distribution. This type of vesicle is not found in the low-to-moderately vesicular samples. In addition, vesicle number density negatively correlates with vesicularity: less vesicular samples have the highest number density and vice versa, and contain far more numerous small-to-medium-sized vesicles than highly vesicular samples. Measured and calculated viscous (Darcian) permeabilities overlap in the range 10− 13 and 10− 9 m2, with higher values in the more vesicular samples. We ascribe these differences in the textural and physical properties of the scoria clasts to their derivation from distinct magma portions in the conduit that were driven by convective overturn and underwent different vesiculation histories and gas transport dynamics. Comparing basaltic scoria clasts from Ambrym to those from mild explosive activity at Stromboli volcano (Italy) reveals that differences in their vesicle size distributions may result from the influence of different crystal contents and shapes on the vesiculation and permeability of the respective magmas. Finally, we highlight how rheological properties have a fundamental role in determining the degassing behaviour of basaltic magma at Ambrym and other volcanoes in general.

    Beschreibung: Seismic activity, ground deformation, and soil and fumarole temperatures acquired during 2004–2007 at Vulcano (Aeolian Islands) are analysed and the time relations among the different time series are discussed. Changes in temperature of fumarolic gases took place during four ‘‘anomalous’’ periods (November 2004–March 2005; October 2005–February 2006; August–October 2006; July–December 2007) at the same time as an increasing number of volcano-seismic events. In particular, the temperatures at high temperature vents and at steam heated soil ranged in time from 180 to 440 C and from 20 to 90 C, respectively. The maximum daily number of volcano-seismic events was 57, reached during the second anomalous period. This seismicity, characterised by focal depth generally lower than 1 km below sea level (b.s.l.) and composed of different kinds of events associated to both resonance and shear failure processes, is related to the shallow dynamics of the hydrothermal system. During the analysed period, very few volcano-tectonic earthquakes took place and tilt recordings showed no sharp or important changes. In light of such observations, the increases in both temperature and volcano-seismic events number were associated to increases in the release of gas from a deep and stable magma body, without magma intrusions within the shallow hydrothermal system. Indeed, a greater release of gas from depth leads to increased fluid circulation, that can promote increases in volcano-seismic events number by both fracturing processes and resonance and vibration in cracks and conduits. The different trends observed in the measured geochemical and geophysical series during the anomalous periods can be due to either time changes in the medium permeability or a changing speed of gas release from a deep magma body. Finally, all the observed variations, together with the changing temporal distribution of the different seismic event kinds, suggest that the hydrothermal system at Vulcano can be considered unsteady and dynamic.

    Beschreibung: A 4-year geochemical survey of some fumaroles at the Voragine summit crater of Mt Etna was performed in combination with synchronous monitoring of peripheral gas emissions at the base of the volcano. This was the first geochemical study at Mt Etna to have included the abundances of Ar, He, and C isotopes. Once the effects of postmagmatic shallow processes were identified and quantitatively removed, the He–Ar–CO2 systematics of the Voragine crater fumaroles and peripheral gas emissions described the same degassing path. Combining the carbon-isotope composition with information about noble gases provided evidence that the crater fumaroles are fed from a two-endmember mixture composed of a deep member coming from pressures between 200 and 400 MPa (depending on time), and a shallower one exsolved at 130 MPa. Similar mixing processes probably also occur in gases from peripheral vents. The simultaneous assessment of d13CCO2 and He/Ar values of crater fumaroles over time has identified simple changes in the mixing proportion between the two endmembers and, moreover, periods during which the exsolution pressure of the deep fluid increased. These periods seem to be linked to pre-eruptive phases of the volcano. The identified open-system degassing processes are indicative of efficient bubble–melt decoupling at depth, whereas the mixing process requires a convective transfer of the deeply exsolved fluids toward shallower levels of magma where further vapor is exsolved. In agreement with the most recent geophysical and petrological data from Mt Etna, these observations allow inferences about a deep portion of the plumbing system (5 to 12 km b.s.l.), comprising sill-like reservoirs connected by small vertical structures, and a main reservoir at 2–3 km b.s.l. that is probably fluxed by magmatic volatiles. 2012 Elsevier Ltd.     Beschreibung: We analyzed crater SO2 fluxes from Mt Etna, together with soil CO2 effluxes from the volcano's flanks, in the period from 2001 to 2005. Between the 2001 and 2002–2003 eruptions, persistently low values of both parameters suggest that no new gas-rich magma was accumulating at shallow depth (b5 km) within Etna's central conduit, whereas very high SO2 sin-eruptive fluxes during the two eruptions indicated sudden decompression of an un-degassed magma rising along newly-formed eccentric conduits. In November 2003, soil CO2 data indicate migration of gas-rich magma from deep (〉10 km) to shallow (b5 km) portions of the feeding conduits, preceded by an increase in crater SO2 fluxes. A similar behavior was observed also during and after the following 2004–2005 eruption. This degassing style matches a period of increased structural instability of the volcanic edifice caused by acceleration of spreading that affected both its eastern and southern flanks. Spreading could have triggered progressively deeper depressurization in the central conduit, inducing release of the more soluble gas (SO2) first, and then of CO2, contrary to what was observed before the 2001 eruption. This suggests that the edifice has depressurized, promoting ascent of fresh-magma and increasing permeability favouring release of CO2 flux. By integrating geochemical and structural data, previous degassing models developed at Mt. Etna have been updated to advance the understanding of eruptive events that occurred in recent years.

    Beschreibung: Cuticle micromorphology together with epidermal and epistomatal wax, in both current- and first-year-old needles of conifer Pinus halepensis (Aleppo pine) trees growing under volcanic gas fumigation was analysed in Pisciarelli area, Campi Flegrei, Southern Italy. As a control, current- and first-year-old needles growing far from volcanic gas emission were also sampled. Using a multidisciplinary approach with SEM, TEM and X-ray, volcanic gases were shown to cause degradation on epicuticular and epistomatal waxes. Significant statistical variations of ultrastructural components of the cuticle, with 30 measurements, including total thickness of the cuticle, and details and proportions of all different layers, and use of confidence interval, revealed a high degree of sensitivity of Aleppo pine to this extreme environment. In the present study, non-significant thickness variations of the cell wall plus cuticle among current- and first-year-old needles of both fumigated and non fumigated trees have been found. However, at the ultrastructural level, significant variations in cell wall and total cuticle thickness, especially within the three zones of B1 fibrillar layer, revealed different equilibria for each of the four types of material. Using energy dispersive X-ray microanalysis, no sulphur was found in either cuticle or epidermal cells, but the presence of H2S in the fumarole gas is suspected to cause indirect and/or direct cuticle alterations of wax structure. Ultrastructural characters of plant cuticles related to emission of volcanic gases during the geological past are also discussed. Among these considerations, an identification key enabling distinction between non fumigated and fumigated materials with 9 characters, provides a good tool detecting the influence of volcanism for extant and fossil plants.

    Beschreibung: The chemistry of Yellowstone fumarole gases shows the existence of two component waters, type MC, influenced by the addition of deep mantle fluid, and type CC, influenced by crustal interactions (CC). MC is high in 3He/4He (22 Ra) and low in 4He/40Ar ( 1), reflecting input of deep mantle components. The other water is characterized by 4He concentrations 3–4 orders of magnitude higher than air-saturated meteoric water (ASW). These high He concentrations originate through circulation in Pleistocene volcanic rocks, as well as outgassing of Tertiary and older (including Archean) basement, some of which could be particularly rich in uranium, a major 4He source. Consideration of CO2–CH4–CO–H2O–H2 gas equilibrium reactions indicates equilibration temperatures from 170 C to 310 C. The estimated temperatures highly correlate with noble-gas variations, suggesting that the two waters differ in temperature. Type CC is 170 C whereas the MC is hotter, at 340 C. This result is similar to models proposed by previous studies of thermal water chemistry. However, instead of mixing the deep hot component simply with cold, meteoric waters we argue that addition of a 4He-rich component, equilibrated at temperatures around 170 C, is necessary to explain the range in fumarole gas chemistry.

    Beschreibung: Gas hazard was evaluated in the three most important cold gas emission zones on the flanks of the quiescent Colli Albani volcano. These zones are located above structural highs of the buried carbonate basement which represents the main regional aquifer and the main reservoir for gas rising from depth. All extensional faults affecting the limestone reservoir represent leaking pathways along which gas rises to the surface and locally accumulates in shallow permeable horizons forming pressurized pockets that may produce gas blowout when reached by wells. The gas, mainly composed by CO2 (〉90 vol.%), contains appreciable quantities of H2S (0.35-6 vol.%), and both represent a potentially high local hazard. Both gases are denser than air and accumulate near ground where they may reach hazardous concentrations, and actually lethal accidents frequently occur to animals watering at local ponds. In order to evaluate the rate of degassing and the related hazard, CO2 and H2S diffuse soil flux surveys have been repeatedly carried out by accumulation chamber. The viscous gas flux of some important discrete emissions has been also evaluated and the CO2 and H2S air concentration measured by portable devises and by Tunable Diode Laser profiles. The minimum potential lethal concentration of the two gases (250 ppm for H2S and 8 vol.% for CO2) is 320 times higher for CO2, whereas the CO2/H2S concentration ratio in the emitted natural gas is significantly lower (15-159). This explains why H2S reaches hazardous, even lethal, concentrations more frequently than CO2. A relevant hazard exists for both gases in the depressed zones (channels, excavations) particularly in the non-windy early hours of the day.

    Beschreibung: Active volcanoes are thought to be important contributors to the atmospheric mercury (Hg) budget, and this chemical element is one of the most harmful atmospheric pollutants, owing to its high toxicity and long residence time in ecosystems. There is, however, considerable uncertainty over the magnitude of the global volcanic Hg flux, since the existing data on volcanogenic Hg emissions are sparse and often ambiguous. In an attempt to extend the currently limited dataset on volcanogenic Hg emissions, we summarize the results of Hg flux measurements at seven active open-conduit volcanoes; Stromboli, Asama, Miyakejima, Montserrat, Ambrym, Yasur, and Nyiragongo.. Data from the domebuilding Soufriere Hills volcano are also reported. Using our determined mercury to SO2 mass ratios in tandem with the simultaneously-determined SO2 emission rates, we estimate that the 7 volcanoes have Hg emission rates ranging from 0.2 to 18 t yr-1 (corresponding to a total Hg flux of ~41 t·yr-1). Based on our dataset and previous work, we propose that a Hg/SO2 plume ratio ~10-5 is bestrepresentative of gas emissions from quiescent degassing volcanoes. Using this ratio, we infer a global volcanic Hg flux from persistent degassing of ~95 t·yr-1

    Beschreibung: During 2007–2008, three CO2 flux surveys were performed on El Chichón volcanic lake, Chiapas, Mexico, with an additional survey in April 2008 covering the entire crater floor (including the lake). The mean CO2 flux calculated by sequential Gaussian simulation from the lake was 1,190 (March 2007), 730 (December 2007) and 1,134 g m−2 day−1 (April 2008) with total emission rates of 164±9.5 (March 2007), 59±2.5 (December 2007) and 109±6.6 t day−1 (April 2008). The mean CO2 flux estimated from the entire crater floor area was 1,102 g m−2 day−1 for April 2008 with a total emission rate of 144±5.9 t day−1. Significant change in CO2 flux was not detected during the period of survey, and the mapping of the CO2 flux highlighted lineaments reflecting the main local and regional tectonic patterns. The 3He/4He ratio (as high as 8.1 RA) for gases in the El Chichón crater is generally higher than those observed at the neighbouring Transmexican Volcanic Belt and the Central American Volcanic Arc. The CO2/3He ratios for the high 3He/4He gases tend to have the MORB-like values (1.41×109), and the CO2/3He ratios for the lower 3He/4He gases fall within the range for the arc-type gases. The high 3He/4He ratios, the MORB-like CO2/3He ratios for the high 3He/4He gases and high proportion of MORB-CO2 (M=25 ±15%) at El Chichón indicate a greater depth for the generation of magma when compared to typical arc volcanoes.

    Beschreibung: On 27 February 2007, a new eruption occurred on Stromboli which lasted until 2 April. It was characterized by effusive activity on the Sciara del Fuoco and by a paroxysmal event (15 March). This crisis represented an opportunity for us to refine the model that had been developed previously (2002-2003 eruption) and to improve our understanding of the relationship between the magmatic dynamics of the volcano and the geochemical variations in the fluids. In particular, the evaluation of the dynamic equilibrium between the volatiles (CO2 and SO2) released from the magma and the corresponding fluids discharged from the summit area allowed us to evaluate the level of criticality of the volcanic activity. One of the major accomplishments of this study is a four-year database of summit soil CO2 flux on the basis of which we define the thresholds (Low-Medium-High) for this parameter that are empirically based on the natural volcanological evolution of Stromboli. The SO2 fluxes of the degassing plume and the CO2 fluxes emitted from the soil at Pizzo Sopra la Fossa are also presented. Noteworthy geochemical signals of volcanic unrest have been clearly identified before, during, and after the effusive activity. These signals were found almost simultaneously in the degassing plume (SO2 flux) and in soil degassing (CO2 flux) at the summit, although the two degassing processes are shown to be clearly different. The interpretation of the results will be useful for future volcanic surveillance at Stromboli.

Data collected at Solfatara and Vulcano, where plumes are fed by fumaroles which are accessible for direct sampling, were first used to validate the technique. For both volcanoes, the plume-derived carbon isotopic compositions are in good agreement with the fumarolic compositions, thus providing confidence on the method, and allowing its application at volcanoes where the volcanic component is inaccessible to direct sampling. As a notable example, we applied the same method to Mount Etna where we derived a δ13C of volcanic CO2 between −0.9±0.27‰ and −1.41± 0.27‰ (Bocca Nuova and Voragine craters). The comparison of our measurements to data reported in previous work values of Etna CO2 from~ −4‰, in the 1970’s and the 1980’s, to~ −1‰ at the present time (2009). This shift toward more positive δ13C values matches a concurrent change in magma composition and an increase in the eruption frequency and energy. We discuss such variations in terms of two possible processes: magma carbonate assimilation and carbon isotopic fractionation due to magma degassing along the Etna plumbing system. Finally, our results highlight potential of systematic measurements of the carbon isotopic composition of the CO2 emitted by volcanic plumes for a better understanding of volcanic processes and for improved surveillance of volcanic activity.

    Beschreibung: Volcanic and geothermal areas are one of the major natural sources of H2S to the atmosphere. Its environmental impact is often the main cause of the opposition to the development of geothermal energy exploitation programs. In this paper we analyze the air concentrations and dispersion pattern of naturally emitted H2S at the geothermal area of Sousaki (Corinthia, Greece). Measurements, made with a network of passive samplers, evidence a rapid decrease of concentration values away from the emission points. The fact that the decrease is more pronounced in the summer with respect to the winter indicates that it is not only due to a dilution effect, but also to redox reactions favoured by higher temperatures and intense sunlight typical of the summer period.

    Beschreibung: This study presents baseline data for future geochemical monitoring of the active Tacaná volcano–hydrothermal system (Mexico–Guatemala). Seven groups of thermal springs, related to a NW/SE-oriented fault scarp cutting the summit area (4,100m a.s.l.), discharge at the northwest foot of the volcano (1,500–2,000m a.s.l.); another one on the southern ends of Tacaná (La Calera). The near-neutral (pH from 5.8 to 6.9) thermal (T from 25.7°C to 63.0°C) HCO3–SO4 waters are thought to have formed by the absorption of a H2S/SO2–CO2-enriched steam into a Cl-rich geothermal aquifer, afterwards mixed by Na/HCO3-enriched meteoric waters originating from the higher elevations of the volcano as stated by the isotopic composition (δD and δ18O) of meteoric and spring waters. Boiling temperature fumaroles (89°C at~3,600m a.s.l. NW of the summit), formed after the May 1986 phreatic explosion, emit isotopically light vapour (δD and δ18O as low as −128 and −19.9‰, respectively) resulting from steam separation from the summit aquifer. Fumarolic as well as bubbling gases at five springs are CO2-dominated. The δ13CCO2 for all gases show typical magmatic values of −3.6 ± 1.3‰ vs V-PDB. The large range in 3He/4He ratios for bubbling, dissolved and fumarolic gases [from 1.3 to 6.9 atmospheric 3He/4He ratio (RA)] is ascribed to a different degree of near-surface boiling processes inside a heterogeneous aquifer at the contact between the volcanic edifice and the crystalline basement (4He source). Tacaná volcano offers a unique opportunity to give insight into shallow hydrothermal and deep magmatic processes affecting the CO2/3He ratio of gases: bubbling springs with lower gas/water ratios show higher 3He/4He ratios and consequently lower CO2/3He ratios (e.g. Zarco spring). Typical Central American CO2/3He and 3He/4He ratios are found for the fumarolic Agua Caliente and Zarco gases (3.1 ± 1.6 × 1010 and 6.0 ± 0.9 RA, respectively). The L/S (5.9 ± 0.5)and (L + S)/M ratios (9.2 ± 0.7) for the same gases are almost identical to the ones calculated for gases in El Salvador, suggesting an enhanced slab contribution as far as the northern extreme of the Central American Volcanic Arc,Tacana

    Beschreibung: Fluorine is one of the many environmental harmful elements released by volcanic activity. The content of total, oxalate extractable and water extractable fluorine was determined in 96 topsoils of three active volcanic systems of southern Italy (Mt. Etna, Stromboli and Vulcano). Total fluorine content (FTOT) ranges from 112 to 7430 mg kg-1, F extracted with oxalate (FOX) ranges from 16 to 2320 mg kg-1 (2 – 93 % of FTOT) and F extracted with distilled water (FH2O) ranges from 1.7 to 159 mg kg-1 (0.2 – 40 % of FTOT). Fluorine in the sampled topsoils derives both from the weathering of volcanic rocks and ashes and from enhanced deposition due to volcanic gas emissions either from open-conduit passive degassing (Mt. Etna and Stromboli) or from a fumarolic field (Vulcano). Fluorine accumulation in the studied soils does generally not present particular environmental issues except for a few anomalous sites at Vulcano where measured contents could be dangerous both for vegetation and for grazing animals.

    Beschreibung: The role of mud volcanoes (MVs) as a source of methane(CH4) flux to the atmosphere and the ocean has been increasingly recognised in the last several years (Milkov 2000; Dimitrov 2002, 2003; Etiope and Klusman 2002; Kopf 2002, 2003; Milkov et al. 2003; Etiope and Milkov 2004). In one of the most recent papers, Kopf (2003) claims to report a reliable estimate of the global CH4 emission from MVs. However, the significance and usefulness of the estimate presented by Kopf (2003) are rather poor. The used dataset is smaller than in previous studies (although the author makes a reverse claim), and some previously published works are misquoted and misinterpreted. Numerous arithmetic mistakes made during simple calculations and data manipulations lead to confusing results and conclusions. In this comment, we highlight some of the most significant problems with the estimates published by Kopf (2003).

    Beschreibung: Abstract: The simultaneous solution of the Planck equation (involving the widely used “dual-band” technique) using two shortwave infrared (SWIR) bands allows for an estimate of the fractional area of the hottest part of an active lava flow (fh), and the background temperature of the cooler crust (Tc). The use of a high spectral and spatial resolution imaging spectrometer with a wide dynamic range of 15 bits (DAIS 7915) in the wavelength range from 0.501 to 12.67 µm resulted in the identification of crustal temperature and fractional areas for an intra-crater hot spot at Mount Etna, Italy. This study indicates the existence of a relationship between these Tc and fh extracted from DAIS and Landsat TM data. When the dual band equation system is performed on a lava flow, a logarithmic distribution is obtained from a plot of the fractional area of the hottest temperature versus the temperature of the cooler crust. An entirely different distribution is obtained over active degassing vents, where increases in Tc occur without any increase in fh. This result indicates that we can use scatter plots of Tc vs. fh to discriminate between different types of volcanic activity, in this case between degassing vents and lava flows, using satellite thermal data.

    Beschreibung: The Monte Nuovo eruption is the most recent event that occurred at Phlegrean Fields (Italy) and lasted from 29 September to 6 October 1538. It was characterized by 2 days of quasi-sustained phreatomagmatic activity generating pumice-bearing pyroclastic density currents and forming a 130-m-high tuff cone (Lower Member deposits). The activity resumed after a pause of 2 days with two discrete Vulcanian explosions that emplaced radially distributed, scoria-bearing pyroclastic flows (Upper Member deposits). The juvenile products of Lower and Upper Members are, respectively, phenocryst-poor, light-coloured pumice and dark scoria fragments with K-phonolitic bulk compositions, identical in terms of both major and trace elements. Groundmass is formed by variable proportions of K-feldspar and glass, along with minor sodalite and Fe-Ti oxide present in the most crystallized samples. Investigations of groundmass compositions and textures were performed to assess the mechanisms of magma ascent, degassing and fragmentation along the conduit and implications for the eruptive dynamics. In pumice of the Lower Member groundmass crystal content increases from 13 to 28 vol% from the base to the top of the sequence. Products of the Upper Member consist of clasts with a groundmass crystal content between 30 and 40 vol% and of totally crystallized fragments. Crystal size distributions of groundmass feldspars shift from a single population at the base of the Lower Member to a double population in the remaining part of the sequence. The average size of both populations regularly increases from the Lower to the Upper Member. Crystal number density increases by two orders of magnitude from the Lower to the Upper Member, suggesting that nucleation dominated during the second phase of the eruption. The overall morphological, compositional and textural data suggest that the juvenile components of the Monte Nuovo eruption are likely to record variations of the magma properties within the conduit. The different textures of pumice clasts from the Lower Member possibly reflect horizontal gradients of the physical properties (P, T) of the ascending magma column, while scoriae from the second phase are thought to result from the disruption of a slowly rising plug crystallizing in response to degassing. In particular, crystal size distribution data point to syn-eruptive degassing-induced crystallization as responsible for the transition in eruptive style from the first to the second phase of the eruption. This mechanism not only has been proved to profoundly affect the dynamics of dome-forming calc-alkaline eruptions, but may also have a strong influence in driving the eruption dynamics of alkaline magmas of intermediate to evolved compositions.

    Notizen: Abstract  Comparison of peptides eluted from human class I and class II major histocompatibility complex (MHC) molecules and the proteins from which they are derived (source proteins) revealed that class I MHC bind peptides derived from proteins that are highly conserved, hydrophilic, and universally expressed, while the peptides themselves are hydrophobic and even more conserved than their source proteins. In contrast, source proteins for class II-bound peptides were not significantly more conserved than a random sample of proteins. Class II-bound peptides were generally more conserved than their source proteins but were significantly less conserved than class I-bound peptides. The characteristics of class I-bound peptides can probably be explained by the selectivity of processing and transport of peptides for binding by class I, while the relative lack of selectivity of peptide binding for class II may explain the high incidence of autoimmune diseases associated with alleles of these molecules.

    Notizen: Abstract  Using an oligonucleotide primer based on a partial goldfish inducible nitric oxide synthase (iNOS) sequence, a complete carp iNOS cDNA was isolated from an activated carp phagocyte cDNA library. Nucleotide and predicted amino acid sequence analysis indicate that carp iNOS encodes a 1127-amino acid protein with 57% sequence identity to human iNOS. Like mammalian NOSs, carp iNOS protein contains putative binding sites for heme, tetrahydrobiopterin, calmodulin, flavine mononucleotide, flavine adenine dinucleotide, and NADPH. Phylogenetic analysis, using neighbor joining, showed that the carp iNOS protein clustered together with the other vertebrate iNOS proteins. Inducibility of carp iNOS was confirmed by reverse transcription-polymerase chain reaction after stimulation of carp phagocytes with lipopolysaccharide or the protozoan blood flagellate Trypanoplasma borreli. These stimulators produced high amounts of nitric oxide that were toxic for T. borreli in vitro. The nuclear transciption factor NF-κB was shown to play a role in the induction of iNOS transcription.

    Notizen: Abstract  Human immunodefiency virus (HIV) poses a major threat to humankind. And though, like humans, chimpanzees are susceptible to HIV infection, they are considered to be resistant to the development of the acquired immune deficiency syndrome (AIDS). Little is known about major histocompatibility complex (MHC) class I diversity in chimpanzee populations and, moreover, whether qualitative aspects of Patr class I molecules may control resistance to AIDS. To address these questions, we assayed MHC class I diversity in a West African chimpanzee population and in some animals from other subspecies of chimpanzee. Application of different techniques allowed the detection of 17 full-length Patr-A, 19 Patr-B, and 10 Patr-C alleles. All Patr-A alleles cluster only into the HLA-A1/A3/A11 family, which supports the idea that chimpanzees have experienced a reduction in their repertoire of A locus alleles. The Patr-B alleles do not cluster in the same lineages as their human equivalents, due to frequent exchange of polymorphic sequence motifs. Furthermore, polymorphic motifs may have been exchanged between Patr-A and Patr-B loci, resulting in convergence. With regard to evolutionary stability, the Patr-C locus is more similar to the Patr-A locus than it is to the Patr-B locus. Despite the relatively low number of animals analyzed, humans and chimpanzees were ascertained as sharing similar degrees of diversity at the contact residues constituting the B and F pockets in the peptide-binding side of MHC class I molecules. Our results indicate that within a small sample of a West African chimpanzee population, a high degree of Patr class I diversity is encountered. This is in agreement with the fact that chimpanzees display more mitochondrial DNA variation than humans. In addition, population analyses demonstrated that particular Patr-B molecules, with the capacity to bind conserved HIV-1 epitopes, are characterized by high gene frequencies. These findings have important implications for evaluating immune responses in HIV vaccine studies and, more importantly, may help in understanding the relative resistance of chimpanzees to AIDS.

    Notizen: Abstract  Polymorphic sequence variation in the peptide-binding domains of MHC class I molecules appears to have been driven largely by the constructive action of natural selection on the specificity of the peptide-binding groove. Similar features are displayed by the variable domains of immunoglobulins generated in the sheep ileal Peyer's patch, but in this case there is evidence that the action of a targeted hypermutator acting on a selected substrate rather than antigen-driven selection is responsible for the pattern of variation in the system. Such a hypermutator acting in the germ line would not only mimic the action of natural selection but also, by convergent mutation, generate similar patterns of variation in unrelated alleles that could be interpreted as evidence for short-tract gene conversion. We analyzed human class I MHC alleles in the light of these data, but failed to find evidence of the action of a similar hypermutator. A search for other mutationally driven patterns of variation also failed, even in hypervariable residues from parsimonious phylogenies. Single-nucleotide variation at these residues is also frequent in recent allelic variants, but the data are as consistent with short-tract gene conversion as with base mutation. We conclude that the patterns of allelic variation in MHC molecules are not driven by mutational pressure, but rather by conventional mutational processes, accompanied by short-tract gene conversion and intense natural selection.

In the pteridophytes, the occurrence of arbuscular mycorrhizas was low in sporophytes of fern-allies and leptosporangiates, whereas in the eusporangiates it was relatively high. From the standpoint of mycotrophism, the evolutionary trend in the Filicineae may be from constantly mycorrhizal to facultative mycorrhizal and finally to nonmycorrhizal plants.

    Notizen: Abstract  Motor patterns of the cardiac sac, the gastric and the pyloric network in the stomatogastric nervous system of the shrimp Penaeus japonicus, the most primitive decapod species, were studied. Single neurons can switch from the gastric or the pyloric pattern to the cardiac sac pattern. Some of the pyloric neurons fire with the gastric pattern. All of the gastric neurons fire with the pyloric pattern, unlike those in reptantians. Proctolin activates and modulates the cardiac sac and the pyloric rhythm, and promotes reconfiguration of the networks. Neurons of the three networks have so many interconnections that they construct a multifunctional neural network like those in Cancer. This network may function in different configurations under the appropriate conditions. Several modes of interactions between the networks found in different reptantian species can apply to the penaeidean shrimp. Such interactions are general features of the stomatogastric nervous system in decapods. Phylogenetic differences among the decapod infraorders are seen in the number and orientation of muscles and the innervation pattern of muscles. The multifunctional networks have existed in the most primitive decapod species, and types of configurations of the networks would have evolved to produce a wide range of motor patterns as the foregut structure has become complex.

    Notizen: Abstract  Pax proteins play a diverse role in early animal development and contain the characteristic paired domain, consisting of two conserved helix-turn-helix motifs. In many Pax proteins the paired domain is fused to a second DNA binding domain of the paired-like homeobox family. By amino acid sequence alignments, secondary structure prediction, 3D-structure comparison, and phylogenetic reconstruction, we analyzed the relationship between Pax proteins and members of the Tc1 family of transposases, which possibly share a common ancestor with Pax proteins. We suggest that the DNA binding domain of an ancestral transposase (proto-Pax transposase) was fused to a homeodomain shortly after the emergence of metazoans about one billion years ago. Using the transposase sequences as an outgroup we reexamined the early evolution of the Pax proteins. Our novel evolutionary scenario features a single homeobox capturing event and an early duplication of Pax genes before the divergence of porifera, indicating a more diverse role of Pax proteins in primitive animals than previously expected.

    Notizen: Abstract  The presence of two sets of paired appendages is one of the defining features of jawed vertebrates. We are interested in identifying genetic systems that could have been responsible for the origin of the first set of such appendages, for their subsequent duplication at a different axial level, and/or for the generation of their distinct identities. It has been hypothesized that four genes of the T-box gene family (Tbx2–Tbx5) played important roles in the course of vertebrate limb evolution. To test this idea, we characterized the orthologs of tetrapod limb-expressed T-box genes from a teleost, Danio rerio. Here we report isolation of three of these genes, tbx2, tbx4, and tbx5. We found that their expression patterns are remarkably similar to those of their tetrapod counterparts. In particular, expression of tbx5 and tbx4 is restricted to pectoral and pelvic fin buds, respectively, while tbx2 can be detected at the anterior and posterior margins of the outgrowing fin buds. This, in combination with conserved expression patterns in other tissues, suggests that the last common ancestor of teleosts and tetrapods possessed all four of these limb-expressed T-box genes (Tbx2–Tbx5), and that these genes had already acquired, and have subsequently maintained, their gene-specific functions. Furthermore, this evidence provides molecular support for the notion that teleost pectoral and pelvic fins and tetrapod fore- and hindlimbs, respectively, are homologous structures, as suggested by comparative morphological analyses.

    Notizen: Abstract  Studies of somitogenesis in vertebrates have identified a number of genes that are regulated by a periodic oscillator that patterns the pre-somitic mesoderm. One of these genes, hairy, is homologous to a Drosophila segmentation gene that also shows periodic spatial expression. This, and the periodic expression of a zebrafish homologue of hairy during somitogenesis, has suggested that insect segmentation and vertebrate somitogenesis may use similar molecular mechanisms and possibly share a common origin. In chicks and mice expression of the lunatic fringe gene also oscillates in the presomitic mesoderm. Fringe encodes an extracellular protein that regulates Notch signalling. This, and the finding that mutations in Notch or its ligands disrupt somite patterning, suggests that Notch signalling plays an important role in vertebrate somitogenesis. Although Notch signalling is not known to play a role in the formation of segments in Drosophila, we reasoned that it might do so in other insects such as the grasshopper, where segment boundaries form between cells, not between syncytial nuclei as they do in Drosophila. Here we report the cloning of a single fringe gene from the grasshopper Schistocerca. We show that it is not detectably expressed in the forming trunk segments of the embryo until after segment boundaries have formed. We conclude that fringe is not part of the mechanism that makes segments in Schistocerca. Thereafter it is expressed in a pattern which shows that it is a downstream target of the segmentation machinery and suggests that it may play a role in segment morphogenesis. Like its Drosophila counterpart, Schistocerca fringe is also expressed in the eye, in rings in the legs, and during oogenesis, in follicle cells.

Since human settlements in southeast Mexico and Central America can be traced as far back as approximately 2500 years BP, most of these events probably affected human activity. In fact, there are reports of pottery shards and other artifacts in deposits from the eruption of 1250 BP. Pottery fragments in deposits of an eruption that took place 2500 BP are also reported in this paper. Thus, the impact of the volcano on human activities has been frequent, with most of the repose intervals lasting between 100 to 600 years. The impact of the eruptions was probably of greater than local extent, because airfall tephra could reach distant sites and possibly even affect weather. The eruptive history of El Chichón also offers clues in the investigation of the Maya civilization. Several researchers have considered the volcano as an important factor in the answer to some intriguing questions such as the extensive use of volcanic ash in Late Classic Maya ceramics or, of greater importance, the causes of the collapse of the Classic Maya civilization.

Natural killer (NK) immunoglobulin-like receptors (KIRs) are a family of polymorphic receptors which interact with specific motifs on HLA class I molecules and modulate NK cytolytic activity. In this study, we analyzed a recently sequenced subgenomic region on chromosome 19q13.4 containing eight members of the KIR receptor repertoire. Six members are clustered within a 100-kb continuous sequence. These genes include a previously unpublished member of the KIR gene family 2DS6, as well as 2DL1, 2DL4, 3DL1, 2DS4, 3DL2, from centromere to telomere. Two additional KIR genes, KIRCI and 2DL3, which may be located centromeric of this cluster were also analyzed. We show that the KIR genes have undergone repeated gene duplications. Diversification between the genes has occurred postduplication primarily as a result of retroelement indels and gene truncation. Using pre- and postduplication Alu sequences identified within these genes as evolutionary molecular clocks, the evolution and duplication of this gene cluster is estimated to have occurred 30–45 million years ago, during primate evolution. A proposed model of the duplication history of the KIR gene family leading to their present organization is presented.

    Notizen: Abstract. The variable region of the immunoglobulin heavy chain is created by a somatic rearrangement of a limited number of germline genes. This mechanism of gene assembly [V(D)J recombination] has been found to take place only in jawed vertebrates (gnathostomes). To understand how this mechanism evolved and diversified it is necessary to study the genomic organization of the heavy-chain gene in different vertebrate lineages. Since there is scant sequence information on the VH locus in fish, shotgun sequencing of a cosmid clone containing part of the VH genomic region of the Japanese pufferfish, Fugu rubripes, was undertaken. Eight full-length VH genes were isolated and characterized. They have higher homology to trout genes, but show the same structural features as VHs found in other vertebrates. Two VH subgroups have been identified whose members are interspersed. The frequency of synonymous and nonsynonymous substitution for VH comparisons between family members was found to be higher in the complementarity-determining regions than in the framework regions. Finally, there are four other genes interspersed with the VH genes, one of which is the first full-length retrotransposon element characterized in vertebrates.

    Notizen: Abstract  The J chain is a component of polymeric immunoglobulin (Ig) molecules and may play an important role in their polymerization and the transport of polymeric Ig across epithelial cells. In this study, the primary structure of the chicken J chain was determined by sequencing cDNA clones. The cDNA had an open reading frame of 476 nucleotides encoding a putative protein of 158 amino acid residues including the signal sequence. The 3′ untranslated region consisted of 1216 nucleotides and a poly(A) tail. The deduced amino acid sequence of the chicken J chain had a high degree of homology to that of human, cow, rabbit, mouse, frog, and earthworm, with eight conserved Cys residues identical to the mammalian J chains. Northern blot hybridization performed with total RNA from various chicken tissues revealed high levels of J-chain mRNA expression in spleen, intestine, Harderian gland, and bursa of Fabricius, and low levels in the thymus. The J chain was expressed in the bursa as early as day 15 of embryogenesis. These data indicated that the chicken J-chain gene displays a high degree of homology with that of other species, and is expressed at an early stage of development of the chicken immune system.

    Notizen: Abstract  Atmospheric CO2 partial pressure (pCO2) was as low as 18 Pa during the Pleistocene and is projected to increase from 36 to 70 Pa CO2 before the end of the 21st century. High pCO2 often increases the growth and reproduction of C3 annuals, whereas low pCO2 decreases growth and may reduce or prevent reproduction. Previous predictions regarding the effects of high and low pCO2 on C3 plants have rarely considered the effects of evolution. Knowledge of the potential for evolution of C3 plants in response to CO2 is important for predicting the degree to which plants may sequester atmospheric CO2 in the future, and for understanding how plants may have functioned in response to low pCO2 during the Pleistocene. Therefore, three studies using Arabidopsis thaliana as a model system for C3 annuals were conducted: (1) a selection experiment to measure responses to selection for high seed number (a major component of fitness) at Pleistocene (20 Pa) and future (70 Pa) pCO2 and to determine changes in development rate and biomass production during selection, (2) a growth experiment to determine if the effects of selection on final biomass were evident prior to reproduction, and (3) a reciprocal transplant experiment to test if pCO2 was a selective agent on Arabidopsis. Arabidopsis showed significant positive responses to selection for high seed number at both 20 and 70 Pa CO2 during the selection process. Furthermore, plants selected at 20 Pa CO2 performed better than plants selected at 70 Pa CO2 under low CO2 conditions, indicating that low CO2 acted as a selective agent on these annuals. However, plants selected at 70 Pa CO2 did not have significantly higher seed production than plants selected at 20 Pa CO2 when grown at high pCO2. Nevertheless, there was some evidence that high CO2 may also be a selective agent because changes in development rate and biomass production during selection occurred in opposite directions at low and high pCO2. Plants selected at high pCO2 showed no change or reductions in biomass relative to control plants due to a decrease in the length of the life cycle, as indicated by earlier initiation of flowering and senescence. In contrast, selection at low CO2 resulted in an average 35% increase in biomass production, due to an increase in the length of the life cycle that resulted in a longer period for biomass accumulation before senescence. From the Arabidopsis model system we conclude that some C3 annuals may have produced greater biomass in response to low pCO2 during the Pleistocene relative to what has been predicted from studies exposing a single generation of C3 plants to low pCO2. Furthermore, C3 annuals may exhibit evolutionary responses to high pCO2 in the future that may result in developmental changes, but these are unlikely to increase biomass production. This series of studies shows that CO2 may potentially act as a selective agent on C3 annuals, producing changes in development rate and carbon accumulation that could not have been predicted from single-generation studies.

    Notizen: Abstract  To identify genes that encode plasma membrane H+-ATPases in the arbuscular mycorrhizal fungus Glomus mosseae two sets of degenerate primers matching highly conserved motifs present in all plant and fungal ATPases were designed. Nested PCR-amplification of G. mosseae genomic DNA using the designed degenerate primers was carried out. Sequence analysis of the cloned PCR products identified five different clones (GmHA1, GmHA2, GmHA3, GmHA4 and GmHA5) encoding putative plasma membrane H+-ATPases. Comparison of the deduced amino-acid sequences of GmHA1–GmHA5 indicate that GmHA1, GmHA3 and GmHA4 are highly identical, while GmHA2 and GmHA5 are more divergent. The evolutionary and functional significance of the divergence found among the different members of the H+-ATPase gene family in G. mosseae is discussed.

    Notizen: Abstract  The conservation of 90 microsatellite-flanking sequences from chickpea in 39 accessions of eight annual and 1 accession of a perennial species of the genus Cicer was investigated. All of the primer sequences successfully amplified microsatellites in related species, indicating the conservation of microsatellite-flanking sequences in chickpea’s relatives. However, the degree of conservation of the primer sites varied between species depending on their known phylogenetic relationship to chickpea, ranging from 92.2% in C. reticulatum, chickpea’s closest relative and potential ancestor, down to 50% for C. cuneatum. A phylogenetic tree revealed that chickpea and the other members of its crossability group were more closely related to the perennial C. anatolicum than to other annual species of the genus. Considerable variation in size and number of amplification products between and within species was observed. Sequence analysis of highly divergent amplification products proved that variation is either due to large differences in the number of microsatellite repeats or to the amplification of a locus unrelated to the one amplified from chickpea. Sequence information and bootstrapping using PAUP suggested that STMSs derived from chickpea may be efficiently and reliably used for synteny studies in chickpea’s crossability group, including C. anatolicum. However, care should be taken when applying these markers to other species of the genus. Considering the data presented here and the known historical record, the age of section Monocicer, including chickpea, is estimated to be about 100,000 years.

    Notizen: Abstract  The seeds of domesticated plants are normally much larger than those of their wild counterparts. This change in seed weight was most likely in response to the selection pressure for yield, uniform germination and seedling vigor which was exerted by humans during domestication. However, despite the evolutionary and agronomic significance of seed weight, very little is know about the genetic and developmental controls of this trait; and, thus far, none of the genes in this pathway have been isolated from any plant species. QTL mapping experiments conducted in tomato during the past decade have allowed the identification of many seed-weight QTLs and have also revealed that only a few loci are responsible for the majority of the seed-weight changes that accompanied the domestication of tomato. This review presents a consensus map for seed weight QTL identified in previously published reports and in unpublished results from our laboratory. This summary of seed-weight QTL data allows for the identification of the major loci controlling this trait in the genus Lycopersicon. It is hoped that this work will allow the elucidation of this important phenotypic transition that occurred during crop-plant domestication and will also provide the starting point for the cloning of a gene responsible for seed-weight variation.

    Notizen: Abstract  Genetic organization of isozyme variation in rice (Oryza sativa L.) was investigated based on 17 polymorphic isozyme loci using a sample of 511 accessions of worldwide origin. The genetic diversity within the species was very high (H=0.36 with 4.82 alleles per locus), as compared with most selfing plant species. Three diversity centers were detected for isozyme variation including South Asia, China and Southeast Asia. The accessions were classified into three well-differentiated cultivar groups corresponding to the indica and japonica subspecies, and a new unnamed group. Variation within the cultivar groups accounted for 80% of the total isozyme variation. Within-country variation accounted for 58% of the total variation while among-region and among-country variation within the cultivar groups accounted for only 14% and 8% of the total variation. Analyses using log-linear models revealed that pronounced non-random associations between and among alleles at many unlinked isozyme loci were organized in a non-hierarchical pattern, and subspecific and macro-geographic differentiation was much more pronounced in multilocus phenotype frequencies than in allelic frequencies at individual loci. These results suggest that selection on multilocus gene complexes was largely responsible for the maintenance of the extensive isozyme variation within the species and the indica-japonica differentiation. Our results further suggest the independent domestication of indica and japonica, the dual origins of the indica rice from China and South Asia (India), and the differentiation of the ecotypes ’javanica’ and the ’temperate japonica’ within the japonica subspecies.

    Notizen: Abstract  Microsatellites have many desirable marker properties. There has been no report of the development and utilization of microsatellite markers in oat. The objectives of the present study were to construct oat microsatellite-enriched libraries, to isolate microsatellite sequences and evaluate their level of polymorphism in Avena species and oat cultivars. One hundred clones were isolated and sequenced from three oat microsatellite-libraries enriched for either (AC/TG) n , (AG/TC) n or (AAG/TTC) n repeats. Seventy eight clones contained microsatellites. A database search showed that 42% of the microsatellite flanking sequences shared significant homology with various repetitive elements. Alu and retrotransposon sequences were the two largest groups associated with the microsatellites. Forty four primer sets were used to amplify the DNA from 12 Avena species and 20 Avena sativa cultivars. Sixty two percent of the primers revealed polymorphism among the Avena species, but only 36% among the cultivars. In the cultivars, the microsatellites associated with repetitive elements were less polymorphic than those not associated with repetitive elements. Only 25% of the microsatellites associated with repetitive elements were polymorphic, while 46% of the microsatellites not associated with repetitive elements showed polymorphism in the cultivars. An average of four alleles with a polymorphism information content (PIC) of 0.57 per primer set was detected among the Avena species, and 3.8 alleles with a PIC of 0.55 among the cultivars. In addition, 54 barley microsatellite primers were tested in Avena species and 26% of the primers amplified microsatellites from oat. Using microsatellite polymorphisms, dendrograms were constructed showing phylogenetic relationships among Avena species and genetic relationships among oat cultivars.

    Notizen: Abstract  The streamside salamander, Ambystoma barbouri, exhibits ineffective antipredator behavior (high emergence rate from refuge, and high activity while out of refuge) and thus suffers heavy predation in stream pools with sunfish. A. barbouri evolved relatively recently from an ancestor that closely resembled a sister species, A. texanum, which breeds in fishless, ephemeral ponds. Sunfish thus represent a relatively new selection pressure for A. barbouri. Phylogenetic inertia predicts that (1) A. texanum should be very poor at coping with fish and (2) because it has only recently been exposed to fish, A. barbouri should still be poor at avoiding fish, but due to its recent exposure to fish, A. barbouri should be better than A. texanum at coping with sunfish. Experimental results provided mixed support for these predictions. As predicted, A. texanum suffered heavy sunfish predation. Compared to A. texanum, A. barbouri showed a greater tendency to initiate alarm moves that enhanced escape success from fish. However, in both the presence and absence of fish, A. barbouri showed higher emergence rates from refuge and higher movement while out of refuge than A. texanum. These behaviors tend to increase exposure to sunfish, i.e., for these key behaviors, A. barbouri apparently evolved in the wrong direction as far as fish predation is concerned. Due to these offsetting effects (increased exposure to fish, increased escape success), A. barbouri is no better at surviving with sunfish than A. texanum. A possible explanation for the high activity of A. barbouri is its use of highly ephemeral habitats (relative to A. texanum) that favor the evolution of higher activity, feeding, and developmental rates for A. barbouri relative to A. texanum.

    Notizen: Abstract The alcohol dehydrogenase genes make up one of the best studied gene families in Drosophila, both in terms of expression and evolution. Moreover, alcohol dehydrogenase genes constitute potential versatile markers in insect transformation experiments. However, due to their rapid evolution, these genes cannot be cloned from other insect genera by DNA hybridization or PCR-based strategies. We have therefore explored an alternative strategy: cloning by functional complementation of appropriate yeast mutants. Here we report that two alcohol dehydrogenase genes from the medfly Ceratitis capitata can functionally replace the yeast enzymes, even though the medfly and yeast genes have evolved independently, acquiring their enzymatic function convergently. Using this method, we have cloned an alcohol dehydrogenase gene from the olive pest Bactrocera oleae. We conclude that functional complementation in yeast can be used to clone alcohol dehydrogenase genes that are unrelated in sequence to those of yeast, thus providing a powerful tool for isolation of dominant insect transformation marker genes.

    Notizen: Abstract We present an overview of the gene content and organization of the mitochondrial genome of Dictyostelium discoideum. The mitochondria genome consists of 55,564 bp with an A + T content of 72.6%. The identified genes include those for two ribosomal RNAs (rnl and rns), 18 tRNAs, ten subunits of the NADH dehydrogenase complex (nad1, 2, 3, 4, 4L, 5, 6, 7, 9 and 11), apocytochrome b (cytb), three subunits of the cytochrome oxidase (cox1/2 and 3), four subunits of the ATP synthase complex (atp1, 6, 8 and 9), 15 ribosomal proteins, and five other ORFs, excluding intronic ORFs. Notable features of D. discoideum mtDNA include the following. (1) All genes are encoded on the same strand of the DNA and a universal genetic code is used. (2) The cox1 gene has no termination codon and is fused to the downstream cox2 gene. The 13 genes for ribosomal proteins and four ORF genes form a cluster 15.4 kb long with several gene overlaps. (3) The number of tRNAs encoded in the genome is not sufficient to support the synthesis of mitochondrial protein. (4) In total, five group I introns reside in rnl and cox1/2, and three of those in cox1/2 contain four free-standing ORFs. We compare the genome to other sequenced mitochondrial genomes, particularly that of Acanthamoeba castellanii.

    Notizen: Abstract This paper modifies the WAVE model for parallel processing in virtual networks to explore and process continuous physical worlds. Expressing distributed activity by cooperative jobs spreading in space and “seeing” each other, rather than as vehicles exchanging messages, the model allows complex missions to be planned in a very flexible manner, with mobile hardware being assigned to the evolving space-conquering programs (waves) dynamically, when required or available. A number of cooperative scenarios in a physical world demonstrate the simplicity and compactness of the wave code. The execution of waves by mobile hardware is discussed, including run-time mapping of waves to vehicles, and supporting multiple distributed jobs in cases of hardware shortages. WAVE can be used efficiently for solving complex problems in space by organized groups of cheap specialized mobile robots, where intelligent behavior is provided by very high level of system organization rather than by the smartness of individual units. In a broader sense, it may also serve as a basic technology for parallel and distributed simulation, and the management of evolution and self-organization of large open systems of different natures.

    Notizen: Abstract The anticodon sequence is a major recognition element for most aminoacyl-tRNA synthetases. We investigated the in vivo effects of changing the anticodon on the aminoacylation specificity in the example of E. coli tRNAPhe. Constructing different anticodon mutants of E. coli tRNAPhe by site-directed mutagenesis, we isolated 22 anticodon mutant tRNAPhe; the anticodons corresponded to 16 amino acids and an opal stop codon. To examine whether the mutant tRNAs had changed their amino acid acceptor specificity in vivo, we tested the viability of E. coli strains containing these tRNAPhe genes in a medium which permitted tRNA induction. Fourteen mutant tRNA genes did not affect host viability. However, eight mutant tRNA genes were toxic to the host and prevented growth, presumably because the anticodon mutants led to translational errors. Many mutant tRNAs which did not affect host viability were not aminoacylated in vivo. Three mutant tRNAs containing anticodon sequences corresponding to lysine (UUU), methionine (CAU) and threonine (UGU) were charged with the amino acid corresponding to their anticodon, but not with phenylalanine. These three tRNAs and tRNAPhe are located in the same cluster in a sequence similarity dendrogram of total E. coli tRNAs. The results support the idea that such tRNAs arising from in vivo evolution are derived by anticodon change from the same ancestor tRNA.

    Notizen: Abstract The ADP-dependent phosphofructokinase (PFK) from Thermococcus zilligii has been purified 950 fold; it had a specific activity of 190 U mg−1. The enzyme required Mg2+ ions for optimal activity and was specific for ADP. The forward reaction kinetics were hyperbolic for both cosubstrates (pH optimum of 6.4), and the apparent K m values for ADP and fructose-6-phosphate were 0.6 mM (apparent V max of 243 U mg−1) and 1.47 mM (apparent V max of 197 U mg−1), respectively. Significantly, the enzyme is indicated to be nonallosteric but was slightly activated by some monovalent cations including Na+ and K+. The protein had a subunit size of 42.2 kDa and an estimated native molecular weight of 66 kDa (gel filtration). Maximal reaction rates for the reverse reaction were attained at pH 7.5–8.0, and the apparent K m values for fructose-1,6-bisphosphate and AMP were 0.56 mM (apparent V max of 2.9 U mg−1) and 12.5 mM, respectively. The biochemical characteristics of this unique ADP-dependent enzymatic activity are compared to ATP and pyrophosphate-dependent phosphofructokinases.

    Notizen: Abstract A protease of a molecular mass of approximately 30 kDa was isolated and purified from the haloalkaliphilic archaeon Natronomonas (formerly Natronobacterium) pharaonis. The enzyme hydrolyzed synthetic peptides, preferentially at the carboxyl terminus of phenylalanine or leucine, as well as large proteins. Hydrolysis occurred over the range of pH from 6 to 12, with an optimum at pH 10. The temperature optimum was 61°C. The enzyme was nearly equally active over the range of salt concentration from 0.5 to 4 M (NaCl or KCl). A strong cross-reaction with a polyclonal antiserum against human chymotrypsin was observed. Enzymatic activity was inhibited by typical serine protease inhibitors. There was significant homology between N-terminal and internal sequences from autolytic fragments and the sequence of bovine chymotrypsinogen B; the overall amino acid composition was similar to that of vertebrate chymotrypsinogens. Evidence for a zymogen-like processing of the protease was obtained. Cell extracts from other halobacteria exhibited similar proteolytic activity and immunoreactivity. The data suggested a widespread distribution of a chymotrypsinogen B-like protease among halo- and haloalkaliphilic Archaea.

    Notizen: Abstract  The mouse Lsh/Ity/Bcg locus regulates natural resistance to intracellular pathogens, and the Nramp1 gene was isolated as its candidate. Nramp is part of a small family of at least two genes, Nramp1 and Nramp2. In the present study, a full-length cDNA for carp NRAMP has been isolated and characterized. Nucleotide and predicted amino acid sequence analysis indicate that the carp NRAMP encodes a 548 amino acid membrane protein with 12 putative transmembrane domains, two N-linked glycosylation sites, and an evolutionarily conserved consensus transport motif. The peptide sequence identity among carp and human NRAMP2 is 78%, and 65% with human NRAMP1. Reverse transcription-polymerase chain reaction revealed that carp NRAMP is ubiquitously expressed. Phylogenetic analysis, using neigbor-joining, showed that the carp NRAMP protein clustered together with mammalian NRAMP2 proteins.

    Notizen: Abstract  The receptor of a T lymphocyte (TCR) recognizes nonself antigens in the company of major histocompatibility complex (MHC) molecules presented to it by the antigen-presenting cell. The variable region of TCR is encoded by either a concatenation of variable region (TCR-V), diversity region (TCR-D), and joining region (TCR-J) genes, or a concatenation of TCR-V and TCR-J genes. The TCR-V genes exist as a multigene family in vertebrate species. Here we study the evolutionary relationships of TCR-V genes from humans, sheep, cattle, rabbits, mice, and chicken. These six species can be classified into two groups according to the frequency of γδ T-cells in their peripheral T-cell populations. The "γδ low" group of species includes humans and mice, in which γδ T-cells constitute very limited portion of the T-cell population. The "γδ high" group includes sheep, cattle, rabbits, and chicken, in which γδ T-cells comprise up to 60% of the T-cell population. Here, we compiled TCR-V sequences from the six species and conducted a phylogenetic analysis. We identified various TCR-V gene subgroups based on the analysis. We found that humans and mice have representatives from nearly all of the subgroups identified, while other species have lost subgroups to different extent. Therefore, the γδ low species have a high degree of diversity of TCR-V genes, while γδ high species all have limited diversity of TCR-V genes. This pattern is similar to that found for immunoglobulin variable region (IGV) genes.

    Notizen: Abstract  A full-length and several partial cDNAs encoding IGK light chains from the marsupial South American opossum, Monodelphis domestica, were isolated and characterized. Using these clones as a starting point, the expressed IGKV repertoire was sampled by anchored polymerase chain reaction using an IGKC-specific primer. Based on nucleotide sequences of twenty unique, expressed IGKV-J combinations, there are at least four IGKV families and two J segments. Southern blot analysis revealed each IGK-V family contains multiple gene segments totaling at least thirty-five IGKV in the opossum genome. No evidence for particular, recurrent IGKV-J combinations in the opossum IGK repertoire was seen, rather the V-J combinations appeared random and diverse. Each of the four IGKV families appear more closely related to V segments from placental mammals than to each other, suggesting the duplication of the IGKV families prior to the separation of marsupials and placental mammals more than one-hundred-million years ago. Overall, the complexity of opossum light chain V segments appears greater than that found in the heavy chain, and light chains are likely to contribute significantly to Ig diversity in this species.With this report, the homologues encoding all three classes of eutherian Ig chains, IGH, IGL, and IGK, have been described in a non-placental mammal.

    Notizen: Abstract  Proper peptide presentation by major histocompatibility complex (MHC)-encoded class II antigens is dependent on the products of the MHC DM loci. We identified the rabbit orthologues (RLA-DMA and -DMB) of human HLA-DMA and -DMB and found that they have 76.9% and 78.8% identity with HLA-DMA and -DMB, respectively. Like classical class II MHC genes, RLA-DM genes are more closely related to human HLA-DM genes than to mouse H2-DM. Among the DM family, there is a high degree of variability at the amino terminus of the DMa chains, and length variability in the cytoplasmic tails of both DMα and DMβ. The rabbit DM genes are coexpressed with class II genes in lymphoid tissues, as are the DM genes of other mammals. The RLA-DM locus maps to the class II region of the rabbit MHC, and is flanked by the DP and DOB loci. Despite having some similarities to class II genes of bony fishes, the DM family represents a separate branch of the MHC class II family.

    Notizen: Abstract The star-nosed mole (Condylura cristata) has a snout surrounded by 22 fleshy and mobile appendages. This unusual structure is not an olfactory organ, as might be assumed from its location, nor is it used to manipulate objects as might be guessed from its appearance. Rather, the star is devoted to the sense of touch, and for this purpose the appendages are covered with thousands of small mechanoreceptive Eimer's organs. Recent behavioral studies find that the star acts much like a tactile eye, having a small behavioral focus, or “fovea” at the center – used for detailed explorations of objects of interest. The peripheral and central nervous systems of the mole reflect these behavioral specializations, such that the small behavioral focus on the nose is more densely innervated in the periphery, and has a greatly enlarged representation in the somatosensory cortex. This somatosensory representation of the tactile fovea is not correlated with anatomical parameters (innervation density) as found in other species, but rather is highly correlated with patterns of behavior. The many surprising parallels between the somatosensory system of the mole, and the visual systems of other mammals, suggest a convergent and perhaps common organization for highly developed sensory systems.

    Notizen: Abstract Peripheral auditory frequency tuning in the ensiferan insect Cyphoderris monstrosa (Orthoptera: Haglidae) was examined by comparing tympanal vibrations and primary auditory receptor responses. In this species there is a mis-match between the frequency of maximal auditory sensitivity and the frequency content of the species' acoustic signals. The mis-match is not a function of the mechanical properties of the tympanum, but is evident at the level of primary receptors. There are two classes of primary receptors: low-tuned and broadly tuned. Differences in the absolute sensitivity of the two receptor types at the male song frequency would allow the auditory system to discriminate intraspecific signals from sounds containing lower frequencies. Comparisons of tympanal and receptor tuning indicated that the sensitivity of the broadly tuned receptors did not differ from that of the tympanum, while low-tuned receptors had significantly narrower frequency tuning. The results suggest that the limited specialization for the encoding of intraspecific signals in the auditory system of C. monstrosa is a primitive rather than a degenerate condition. The limited specialization of C. monstrosa may reflect the evolutionary origin of communication-related hearing from a generalized precursor through the addition of peripheral adaptations (tympana, additional receptors) to enhance frequency sensitivity and discrimination.

    Notizen: Abstract The unusual morphology of a sound-activated plurisegmental ascending interneurone (AN5-AG7) in an insect (Ancistrura nigrovittata, Ensifera, Phaneropteridae) is described. This neurone's soma is located in the penultimate abdominal ganglion. The most prominent arborisations with smooth endings are found in the prothoracic ganglion. The neurone terminates with numerous beaded endings in the brain (protocerebrum). All abdominal ganglia including the penultimate contain only tiny side branches of beaded appearance. The neurone's morphology is compared to the morphology of a `typical' sound-activated plurisegmental neurone of bushcrickets with its soma in the prothorax. In the prothoracic ganglion and in the brain the arborisations of the two cells are very similar. Graded potentials and action potentials are generated in the prothoracic portion of both neurones. Both receive excitation mainly by ultrasound, and inhibition by soma-ipsilateral stimuli. Neither wind, substrate vibration nor touch of the abdomen evoke responses in AN5-AG7. It is assumed that early in evolution this neurone had its dendrites in the ganglion which houses the cell body (like cercal interneurones of this neuromere). Profound evolutionary changes probably have taken place to bring about this neuron's modern morphology.

    Notizen: Abstract Male moths responding to their species-specific sex pheromone, may cease their upwind flight when pheromone components of sympatric species are added to the mixture. The interspecific interaction between the pheromone response of the tortricid moths Cydia pomonella and Adoxophyes orana was investigated in field-trapping and wind-tunnel studies. Addition of the A. orana pheromone [(Z9)-tetradecenylacetate and (Z11)-tetradecenylacetate] to a source containing the C. pomonella pheromone [(E8, E10)-dodecadienol] resulted in a significant inhibition of attraction by male C. pomonella. It is demonstrated that this behavioural antagonist for C. pomonella must be emitted from the same point source to induce this inhibitory effect. A spatial separation of the two interspecific pheromones (at 14 cm, 5 cm and 0.5 cm crosswind) restored the attraction of the conspecific pheromone for male C. pomonella. In contrast to C. pomonella, male A. orana were not inhibited by point sources releasing both the C. pomonella and A. orana pheromone. We suggest that the discrepancy in the interspecific pheromone interaction between these two tortricids can be explained if we consider the evolutionary ecology of interspecific pheromone communication in C. pomonella and A. orana.

A population of agents with various preference types can choose between a safe production technology and a risky production technology.     Notizen: Abstract  Analysis and hydrogeochemical modeling of hydrocarbonate groundwater, including its buffering geochemical properties, have demonstrated that this water has undergone a geochemical transformation almost throughout the whole of one industrial region. It is known that hydrocarbonate groundwater possesses a high protective natural geochemical potential, supporting neutralization of acid atmospheric precipitation. Natural hydrocarbonate water undergoes three stages of anthropogenic transformation caused by acid atmospheric precipitation over more than 50 years. The first stage is transformation of neutral hydrocarbonate water into alkaline water, accompanied by calcite precipitation and sodium carbonate dissolution from the unsaturated zone. The second stage is transformation of alkaline water into neutral hydrocarbonate water; the hydrocarbonate content, being less than the lower limit for background concentrations, showed reduced water buffering properties or protective potential relative to acid precipitation. The third stage is transformation of neutral hydrocarbonate water into acidic water, with a complete loss of protective geochemical potential. This unfavorable ecological situation with natural geochemically hydrocarbonate groundwater shows that natural groundwater less protected from the impact of acid atmospheric precipitation is in a worse ecological condition, which is confirmed by observations in northern and Western Europe and other regions of the world.

    Notizen: Abstract  The genes belonging to the Paired class exert primary developmental functions. They are characterized by six invariant amino acid residues in the homeodomain, while the residue at position 50 can be a serine, glutamine or lysine as in the Pax-type, Q50 Paired-like or the K50 Paired-like homeodomains respectively. Genes in this class emerged early in animal evolution: three distinct Pax genes and two Q50 Paired-like genes have recently been characterised from cnidarians. Phylogenetic molecular reconstructions taking into account homeodomain and paired-domain sequences provide some new perspectives on the evolution of the Paired-class genes. Analysis of 146 Paired-class homeodomains from a wide range of metazoan taxa allowed us to identify 18 families among the three sub-classes from which the aristaless family displays the least diverged position. Both Pax-type and K50 families branch within the Q50 Paired-like sequences implying that these are the most ancestral. Consequently, most Pax genes arose from a Paired-like ancestor, via fusion of a Paired-like homebox gene with a gene encoding only a paired domain; the Cnidaria appear to contain genes representing the ’before’ and ’after’ fusion events.

    Notizen: Abstract  Hairless is a member of the Notch signalling pathway, where it acts as antagonist by binding to Suppressor of Hairless [Su(H)], thereby inhibiting Notch target gene activation. The pathway and its members are highly conserved in metazoans from worms to humans. However, a Hairless orthologue from another species has not yet been identified. The identification of Hairless in largely diverged species by cross-hybridization has failed so far probably due to a low degree of conservation. Therefore, we turned to D. hydei where a Hairless mutation has been described before. The D. hydei Hairless orthologue is reasonably well conserved with regard to gene structure and expression. The prospective Hairless protein orthologues share several highly conserved regions which are separated by quite diverged stretches. As to be expected, the largest region of high conservation corresponds to the Su(H) binding domain. This region is also functionally conserved, since this D. hydei protein domain binds very strongly to the D. melanogaster Su(H) protein. The other conserved regions support our earlier structure-function analysis since they nicely correspond to previously defined, functionally important protein domains. Most notably, the very C-terminal domain which is very sensitive to structural alterations, is nearly identical between the two species. In summary, this evolutionary study improves the knowledge on functionally significant domains of the Hairless protein, and may be helpful for the future identification of homologues in other animals, especially in vertebrates.

    Notizen: Abstract  Evolutionary change in developmental mode in sea urchins is closely tied to an increase in maternal provisioning. We examined the oogenic modifications involved in production of a large egg by comparison of oogenesis in congeneric sea urchins with markedly different sized oocytes and divergent modes of development. Heliocidaris tuberculata has small eggs (95 µm diameter) and the ancestral mode of development through feeding larvae, whereas H. erythrogramma has large eggs (430 µm diameter) and highly modified non-feeding lecithotrophic larvae. Production of a large egg in H. erythrogramma involved both conserved and divergent mechanisms. The pattern and level of vitellogenin gene expression is similar in the two species. Vitellogenin processing is also similar with the gonads of both species incorporating yolk protein from coelomic and hemal stores into nutritive cells with subsequent transfer of this protein into yolk granules in the developing vitellogenic oocyte. Immunocytology of the eggs of both Heliocidaris species indicates they incorporate similar levels of yolk protein. However, H. erythrogramma has evolved a highly divergent second phase of oogenesis characterised by massive deposition of non-vitellogenic material including additional maternal protein and lipid. Maternal provisioning in H. erythrogramma exhibits recapitulation of the ancestral vitellogenic program followed by a novel oogenic phase with hypertrophy of the lipogenic program being a major contributor to the increase in egg size.

    Notizen: Abstract  The amphibian Xenopus laevis is one non-mammalian vertebrate in which the major histocompatibility complex (MHC) has been analyzed extensively. Class IIβ, class Ia, LMP2, LMP7, HSP70, C4, Factor B, and Ring3 genes have been identified and mapped to the MHC. Here, we report the isolation of a transporter associated with antigen processing (TAP) gene, TAP2, and demonstrate its linkage to the MHC. While the ATP-binding region of Xenopus TAP2 is highly conserved in evolution, amino acid identity to other vertebrate TAP proteins was not detected in the N-terminal region. Segregation analysis of 34 individuals from two families showed exact restriction fragment length polymorphism matching between the MHC class Ia gene and the one TAP2 gene demonstrating linkage conservation since the mammalian/amphibian divergence ∼350 million years ago. In addition, one non-MHC-linked TAP2–hybridizing fragment was detected in approximately half of the individuals tested. Interestingly, TAP2 allelic lineages appear to match those of LMP7 and classical class I, which previously were categorized into two highly divergent groups that emerged at least 60 million years ago. Similar to LMP7 and class Ia,TAP2 is expressed ubiquitously with highest levels in intestine and spleen.

    Notizen: Abstract  Two proteasome activators PA28α and β, which have been implicated in antigen processing for loading class I MHC molecules, are synthesized in response to Ifn-γ. The human genes encoding these activators (PSME1 and PSME2, respectively) were analyzed by sequencing. Each gene comprised 11 exons, consistent with gene duplication during vertebrate evolution. The intron/exon organization of both genes was highly conserved, the major difference being the absence of the exon encoding the lysine and glutamic acid-rich 'KEKE' motif in PA28β. Two other genes of relevance to the immune system were located close to those for PA28 at 14q11.2 including ISGF3G, a protein involved in transcription after IFNα signalling. These sequences were also characterized.
