Description: The fortieth anniversary of the Strategic and Defence Studies Centre’s founding provided the opportunity to assemble many of Australia’s leading analysts and commentators to review some of the more significant issues that should define Australian defence policy. In the first 20 years after its establishment, SDSC scholars played a prominent role in shaping the ideas and aspirations that eventually found official expression in the 1987 Defence of Australia White Paper. This policy sustained a coherent balance between strategy, force structure and budgets for well over a decade. In recent years, however, the cumulative effects of the end of the Cold War and watershed events like the East Timor experience; the attacks on New York and Washington, D.C., in September 2001; the Bali bombings in October 2002; and the invasion of Iraq in March 2003 have fractured the former consensus on defence policy. These developments have eroded acceptance of the core judgements underpinning defence policy. This has led to a more tenuous connection between some recent major equipment acquisitions and declared policy. The unravelling of the consensus on the ‘defence of Australia’ policy means that we must again undertake a balanced, long-term assessment of the nature of Australia’s strategic interests. Only by doing so can we determine the kinds of armed forces that would contribute most effectively to protecting those interests. The papers collected in this volume are not informed by a common view of where Australia should focus its defence policy, but all address themes that should figure prominently in this difficult but essential task.

Description: We cannot expect in East Asia over the foreseeable future to see the sort of conflation of sovereign states that has occurred in Europe. We must anticipate that, for the foreseeable future, the requirement will be for the sensible management and containment of competitive instincts. The establishment of a multilateral security body in East Asia that includes all the key players, and which the major powers invest with the authority to tackle the shaping of the regional security order, remains a critical piece of unfinished business.

Description: The Hmong are among Australia’s newest immigrant populations. They came as refugees from Laos after the communist revolution of 1975 ended their life there as highland shifting cultivators. The Hmong originate from southern China where many still remain, and others live in Vietnam, Thailand and Burma. Hmong refugees are now also settled in the USA, Canada, France, Germany and French Guyana. Already the beauty and richness of traditional Hmong culture, in particular their shamanism and embroidered costume, has attracted the attention of the Australian public, but little is known about these people, their background or the struggles they have faced to adjust to a new life in Australia. This interdisciplinary collection of articles deals with their music and textiles, gender and language, their social adaptation and their global diaspora. The book aims to bring knowledge of the Hmong to a wider public and contribute to the understanding of these people.

Description: The Representation of Science and Scientists on Postage Stamps examines how the postal authorities of the world have developed unique techniques to portray science and scientists in order to convey a message behind the stamp issue. It is a multi-disciplinary examination that investigates visual representation, semiotics, science, science history and politics, amongst other issues. The author introduces and describes a technique for looking at how and why images may have been chosen. Is the image a mirror reflecting a known reality or is it a lens to challenge, to prompt further thought or action? He also hypothesises that the Internet created a real change to science communication, influencing the way images of science were used. The stamp message changed from that of pursuing a public understanding approach to an awareness of science with its increasing contextual content.

Description: Since September 11, 2001, our newspapers have been filled with the ‘war on terror’; our governments have mobilised their resources for ‘homeland security’; and people everywhere are braced for more terrorist attacks. Yet while the new threat is genuine, we must not lose sight of the continuing security concerns in the Asia-Pacific. Tensions persist on the Korean peninsula, in the Taiwan Straits and the South China Sea, and in Kashmir. The region is well supplied with weapons of mass destruction and may face an arms race, and there are a range of pressing human security issues. Likewise, the strategic realities of the region remain linked with US power, and with the emergence of China as a key regional player. The book examines the developing strategic relationships in the region, and clarifies the dilemmas for Australian policy-makers as they try to balance genuine engagement with the region against a long-standing and valued alliance with the United States. Emerging from discussions between the Shanghai Institute for International Studies and the University of New South Wales at ADFA, Asia-Pacific Security has a particular relevance for foreign-policy professionals and scholars of the region.

Description: In the years between the two world wars of the twentieth century leaders in Western countries worried about a food surplus. The hardships of the Great Depression were intensified by a glut of wheat and consequent low prices on the world market. Yet at the same time nutrition scientists protested that significant proportions of populations, even in affluent countries, were unable to afford a diet ‘adequate for health’. Fresh fruits, vegetables, dairy products and meat were out of reach for the poor. This book traces the work of three men who sought to bring together the interests of farmers and the needs of the hungry: scientist and passionate campaigner for better nutrition, John Boyd Orr; Australian politician and international statesman, Stanley Melbourne Bruce; and Economic Adviser to Bruce at the Australian High Commission in London, Frank Lidgett McDougall. Bruce once said ‘McDougall brings me a new idea every morning’. One of those ideas became the genesis of their work, which helped bring about the formation of the Food and Agriculture Organization of the United Nations in 1945. All three undertook significant roles in the formative years of the organisation. The story of this contribution to the international world order is little known. The cooperation, diplomacy and persistence of these men provides inspiration for tackling the alarming prospect of food shortages in the present century.

Description: "The Australian National University’s Strategic & Defence Studies Centre (SDSC) is Australia’s premier university-based strategic studies think tank. Fifty years after the Centre was founded in 1966, SDSC celebrated its continued research, publications, teaching and government advisory role with a two-day conference entitled ‘New Directions in Strategic Thinking 2.0’. The event saw the podium graced by many of the world’s premier thinkers in the strategic studies field. An evening between those tours to the lectern brought together academics, practitioners and other honoured guests at a commemorative dinner held beneath the widespread wings of the ‘G for George’ bomber in the Australian War Memorial—an event that included SDSC’s own Professor Desmond Ball AO making his last public appearance. Since SDSC’s 25th anniversary, the world has seen the collapse of the Soviet Union and the end of the Cold War. Bipolarity gave way to the emergence of the United States as the world’s sole superpower, a status many now see as under threat. Both the nature of the threats and identity of individual competitors has changed in the interim quarter-century. Non-state actors are presenting rising challenges to national governments. Meanwhile, a diminished Russia and far more wealthy China seek to reassert themselves. Never before has the call for reasoned innovative security studies thinking been more pronounced. Rarely has a group so able to offer that thought come together as was the case in July 2016. This book encapsulates the essence of this cutting-edge thinking and is a must read for those concerned with emerging strategic challenges facing Australia and its security partners."

Description: Paul Dibb AM has had an extraordinary career. He enjoys an international scholarly reputation of the highest order, while at the same time he has done much distinguished public service. He was a pioneer in moving back and forth between posts in government departments, notably the Department of Defence, and academia. He began as a student of Soviet economic geography, and then spent nearly two decades in Australian Defence intelligence, including service as Head of the National Assessments Staff (NAS) in the Joint Intelligence Organisation (JIO) from 1974 to 1978, Deputy Director of JIO in 1978–80, Director of JIO in 1986–88, and Deputy Secretary of Defence (Strategy and Intelligence) in 1988–91, before becoming a Professor in the Strategic and Defence Studies Centre (SDSC) at The Australian National University (where he is now an Emeritus Professor). He has been quite happy to engage in vigorous public debate about important and controversial strategic and defence issues, giving him a high public profile. The contributors include two former Chancellors of ANU, one a former Minister of Defence, and the other a former Secretary of the Department of Defence, a former Chief of the Defence Force (CDF), and other former senior officials, as well as academic specialists in geography, international relations, and strategic and defence studies. ‘This would be a high-quality set of essays for any edited volume, but for a festschrift – a genre that sometimes generates uneven collections – this is an exceptional assembly. The individual pieces are very good; together, they have coherence and power.’ – Professor Ian Hall, Professor of International Relations, Griffith University

Description: The world and China’s place in it have been transformed over the past year. The pressures for change have come from the most severe global financial crisis ever. The crisis has accelerated China’s emergence as a great power. But China and its global partners have yet to think or work through the consequences of its new position for the governance of world affairs. China’s New Place in a World in Crisis discusses and provides in-depth analysis of the following questions. How have China’s growth prospects been affected by the global crisis? How will the crisis and China’s response to it impact China’s major domestic issues, such as industrialisation, urbanisation and the reform of the state-owned sector of the economy? How will the crisis and the international community’s response to it affect the rapidly emerging new international order? What will be China’s, and other major developing countries’, new role? Can China and the world find a way of breaking the nexus between economic growth and environmental sustainability — especially on the issue of climate change?

Description: China’s transformation has been patiently, methodically and very deliberately constructed by a leadership group that has equally carefully protected its monopoly on power. Today’s China is proceeding with great seriousness and determination to become a first-rank state with a balanced portfolio of power and no major vulnerabilities. China takes itself very seriously and is inviting the world to overlook the formidable hard power assets it is determined to acquire in favour of simply enjoying the fruits of its market and trusting in the sincerity of its rhetoric on being determined to become a benign and peaceful new-age major power.

Description: The People’s Republic of China is now over fifty years old. Long considered an outsider, or a club of one, in international relations, China has recently become more active in international institutions. Is China becoming a responsible power in global and regional international relations? How accurate is the traditional perception of China? What factors may be motivating the changes in China’s approach to international institutions and its perceptions of its own role in the world? There is no certainty that China is becoming a more responsible power, recent developments may be just another manifestation of realpolitik. Power and Responsibility in Chinese Foreign Policy provides a vital insight into these issues, analysing the critical issues in China’s international relations– China’s regional and global diplomatic and security problems, the changing role of the People’s Liberation Army, human rights, religious and democratic movements, and the concept of responsibility. Power and Responsibility in Chinese Foreign Policy is an insightful and vital introduction to all sides of the current debate over China’s international relations.

Description: Where the last three decades of the 20th century witnessed a China rising on to the global economic stage, the first three decades of the 21st century are almost certain to bring with them the completion of that rise, not only in economic, but also political and geopolitical terms. China’s integration into the global economy has brought one-fifth of the global population into the world trading system, which has increased global market potential and integration to an unprecedented level. The increased scale and depth of international specialisation propelled by an enlarged world market has offered new opportunities to boost world production, trade and consumption; with the potential for increasing the welfare of all the countries involved. However, China’s integration into the global economy has forced a worldwide reallocation of economic activities. This has increased various kinds of friction in China’s trading and political relations with others, as well as generating several globally significant externalities. Finding ways to accommodate China’s rise in a way that ensures the future stability and prosperity of the world economy and polity is probably the most important task facing the world community in the first half of the 21st century. The book delves into these issues to reflect upon the wide range of opportunities and challenges that have emerged in the context of a rising China.

Description: Asia looks and feels very different now compared to the days of the Cold War. The sense that Asia now works differently can be traced to a single source – the re-emergence of China. China was the dominant power in greater Asia for most of recorded history. This historical norm was interrupted from the early 19th century, too far into the past to be recognisable and readily accommodated by the actors in today’s international arena. A powerful China feels new and unfamiliar. Arriving peacefully at mutually acceptable relationships of power and influence that are very different from those that have prevailed for the past half century will be a demanding process. The world’s track record on challenges of this kind is not terrific. It will call for statesmanship of a consistently high order from all the major players, and building the strongest possible confidence among these players that there are no hidden agendas.

Description: In this monograph, five former secretaries of the Department of Foreign Affairs and Trade (DFAT) reflect on their experiences and the challenges of their times. A far cry from the pukka fantasies of ‘Yes Minister’, their recollections reveal the realpolitik of the policy front line where the secretary must stay ahead of emerging themes and issues in Australia’s international relations while simultaneously exercising governance oversight and providing leadership to a large, professional, diverse and dispersed organisation. From the Cold War to the War on Terror; from the floating of the dollar to GATT and the WTO; managing relations big and small, within our region and without; through relentless administrative reforms, technological change and changes of government; steering DFAT requires ‘steady hands’. This collection of public lectures presented in 2006 to the Australian Institute of International Affairs (AIIA) offers an invaluable resource for those with an interest in recent Australian history, foreign policy and public sector administration

Description: Positive-strand RNA viruses such as poliovirus replicate their genomes on intracellular membranes of their eukaryotic hosts. Electron microscopy has revealed that purified poliovirus RNA-dependent RNA polymerase forms planar and tubular oligomeric arrays. The structural integrity of these arrays correlates with cooperative RNA binding and RNA elongation and is sensitive to mutations that disrupt intermolecular contacts predicted by the polymerase structure. Membranous vesicles isolated from poliovirus-infected cells contain structures consistent with the presence of two-dimensional polymerase arrays on their surfaces during infection. Therefore, host cytoplasmic membranes may function as physical foundations for two-dimensional polymerase arrays, conferring the advantages of surface catalysis to viral RNA replication.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Lyle, John M -- Bullitt, Esther -- Bienz, Kurt -- Kirkegaard, Karla -- AI-42119/AI/NIAID NIH HHS/ -- New York, N.Y. -- Science. 2002 Jun 21;296(5576):2218-22.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Microbiology and Immunology, Stanford University School of Medicine, Stanford, CA 94305, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/12077417" target="_blank"〉PubMed〈/a〉

Description: Despite recent genetic evidence and the promise of individualized medicine, there is a continuing interest in using self-identified categories of race and ethnicity as variables in scientific and medical research. The U.S. Food and Drug Administration recently proposed a standardized approach for the collection of race and ethnicity data in clinical trials. We believe that this move fails to acknowledge new scientific data and recommend that relevant data from individuals be collected and used rather than broad group statistics. We also encourage that increased funding be committed to this important issue.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Haga, Susanne B -- Venter, J Craig -- New York, N.Y. -- Science. 2003 Jul 25;301(5632):466.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Center for the Advancement of Genomics, Rockville, MD 20850, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/12881555" target="_blank"〉PubMed〈/a〉

Description: Direct interaction between platelet receptor glycoprotein Ibalpha (GpIbalpha) and thrombin is required for platelet aggregation and activation at sites of vascular injury. Abnormal GpIbalpha-thrombin binding is associated with many pathological conditions, including occlusive arterial thrombosis and bleeding disorders. The crystal structure of the GpIbalpha-thrombin complex at 2.6 angstrom resolution reveals simultaneous interactions of GpIbalpha with exosite I of one thrombin molecule, and with exosite II of a second thrombin molecule. In the crystal lattice, the periodic arrangement of GpIbalpha-thrombin complexes mirrors a scaffold that could serve as a driving force for tight platelet adhesion. The details of these interactions reconcile GpIbalpha-thrombin binding modes that are presently controversial, highlighting two distinct interfaces that are potential targets for development of novel antithrombotic drugs.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Dumas, John J -- Kumar, Ravindra -- Seehra, Jasbir -- Somers, William S -- Mosyak, Lidia -- New York, N.Y. -- Science. 2003 Jul 11;301(5630):222-6.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Chemical and Screening Sciences, Wyeth, 200 Cambridge Park Drive, Cambridge, MA 02140, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/12855811" target="_blank"〉PubMed〈/a〉

Description: We have used adenosine diphosphate analogs containing electron paramagnetic resonance (EPR) spin moieties and EPR spectroscopy to show that the nucleotide-binding site of kinesin-family motors closes when the motor.diphosphate complex binds to microtubules. Structural analyses demonstrate that a domain movement in the switch 1 region at the nucleotide site, homologous to domain movements in the switch 1 region in the G proteins [heterotrimeric guanine nucleotide-binding proteins], explains the EPR data. The switch movement primes the motor both for the free energy-yielding nucleotide hydrolysis reaction and for subsequent conformational changes that are crucial for the generation of force and directed motion along the microtubule.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Naber, Nariman -- Minehardt, Todd J -- Rice, Sarah -- Chen, Xiaoru -- Grammer, Jean -- Matuska, Marija -- Vale, Ronald D -- Kollman, Peter A -- Car, Roberto -- Yount, Ralph G -- Cooke, Roger -- Pate, Edward -- AR39643/AR/NIAMS NIH HHS/ -- AR42895/AR/NIAMS NIH HHS/ -- DK05915/DK/NIDDK NIH HHS/ -- GM29072/GM/NIGMS NIH HHS/ -- RR1081/RR/NCRR NIH HHS/ -- New York, N.Y. -- Science. 2003 May 2;300(5620):798-801.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Biochemistry, University of California, San Francisco, CA 94143, USA. naber@itsa.ucsf.edu〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/12730601" target="_blank"〉PubMed〈/a〉

Description: The use of Internet references in academic literature is common, and Internet references are frequently inaccessible. The extent of Internet referencing and Internet reference activity in medical or scientific publications was systematically examined in more than 1000 articles published between 2000 and 2003 in the New England Journal of Medicine, The Journal of the American Medical Association, and Science. Internet references accounted for 2.6% of all references (672/25548) and in articles 27 months old, 13% of Internet references were inactive. Publishers, librarians, and readers need to reassess policies, archiving systems, and other resources for addressing Internet reference attrition to prevent further information loss.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Dellavalle, Robert P -- Hester, Eric J -- Heilig, Lauren F -- Drake, Amanda L -- Kuntzman, Jeff W -- Graber, Marla -- Schilling, Lisa M -- K-07 CA92550/CA/NCI NIH HHS/ -- R25 CA49981/CA/NCI NIH HHS/ -- T32 AR07411/AR/NIAMS NIH HHS/ -- New York, N.Y. -- Science. 2003 Oct 31;302(5646):787-8.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Veterans Affairs Medical Center, Denver, CO 80220, USA. robert.dellavalle@uchsc.edu〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/14593153" target="_blank"〉PubMed〈/a〉

Description: Multidrug efflux pumps cause serious problems in cancer chemotherapy and treatment of bacterial infections. Yet high-resolution structures of ligand transporter complexes have previously been unavailable. We obtained x-ray crystallographic structures of the trimeric AcrB pump from Escherichia coli with four structurally diverse ligands. The structures show that three molecules of ligands bind simultaneously to the extremely large central cavity of 5000 cubic angstroms, primarily by hydrophobic, aromatic stacking and van der Waals interactions. Each ligand uses a slightly different subset of AcrB residues for binding. The bound ligand molecules often interact with each other, stabilizing the binding.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Yu, Edward W -- McDermott, Gerry -- Zgurskaya, Helen I -- Nikaido, Hiroshi -- Koshland, Daniel E Jr -- AI 09644/AI/NIAID NIH HHS/ -- New York, N.Y. -- Science. 2003 May 9;300(5621):976-80.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Molecular and Cell Biology, University of California, Berkeley, CA 94720-3202, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/12738864" target="_blank"〉PubMed〈/a〉

Description: The T cell receptor (TCR) inherently has dual specificity. T cells must recognize self-antigens in the thymus during maturation and then discriminate between foreign pathogens in the periphery. A molecular basis for this cross-reactivity is elucidated by the crystal structure of the alloreactive 2C TCR bound to self peptide-major histocompatibility complex (pMHC) antigen H-2Kb-dEV8 refined against anisotropic 3.0 angstrom resolution x-ray data. The interface between peptide and TCR exhibits extremely poor shape complementarity, and the TCR beta chain complementarity-determining region 3 (CDR3) has minimal interaction with the dEV8 peptide. Large conformational changes in three of the TCR CDR loops are induced upon binding, providing a mechanism of structural plasticity to accommodate a variety of different peptide antigens. Extensive TCR interaction with the pMHC alpha helices suggests a generalized orientation that is mediated by the Valpha domain of the TCR and rationalizes how TCRs can effectively "scan" different peptides bound within a large, low-affinity MHC structural framework for those that provide the slight additional kinetic stabilization required for signaling.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Garcia, K C -- Degano, M -- Pease, L R -- Huang, M -- Peterson, P A -- Teyton, L -- Wilson, I A -- AI42266/AI/NIAID NIH HHS/ -- AI42267/AI/NIAID NIH HHS/ -- R01 CA58896/CA/NCI NIH HHS/ -- New York, N.Y. -- Science. 1998 Feb 20;279(5354):1166-72.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Molecular Biology and the Skaggs Institute of Chemical Biology, The Scripps Research Institute, 10550 North Torrey Pines Road, La Jolla, CA 92037, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/9469799" target="_blank"〉PubMed〈/a〉

Description: The 2.5 angstrom resolution x-ray crystal structure of the Escherichia coli RNA polymerase (RNAP) alpha subunit amino-terminal domain (alphaNTD), which is necessary and sufficient to dimerize and assemble the other RNAP subunits into a transcriptionally active enzyme and contains all of the sequence elements conserved among eukaryotic alpha homologs, has been determined. The alphaNTD monomer comprises two distinct, flexibly linked domains, only one of which participates in the dimer interface. In the alphaNTD dimer, a pair of helices from one monomer interact with the cognate helices of the other to form an extensive hydrophobic core. All of the determinants for interactions with the other RNAP subunits lie on one face of the alphaNTD dimer. Sequence alignments, combined with secondary-structure predictions, support proposals that a heterodimer of the eukaryotic RNAP subunits related to Saccharomyces cerevisiae Rpb3 and Rpb11 plays the role of the alphaNTD dimer in prokaryotic RNAP.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Zhang, G -- Darst, S A -- GM19441-01/GM/NIGMS NIH HHS/ -- GM53759/GM/NIGMS NIH HHS/ -- New York, N.Y. -- Science. 1998 Jul 10;281(5374):262-6.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Rockefeller University, 1230 York Avenue, New York, NY 10021, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/9657722" target="_blank"〉PubMed〈/a〉

Description: The three-dimensional structure of a 70-kilodalton amino terminally truncated form of human topoisomerase I in complex with a 22-base pair duplex oligonucleotide, determined to a resolution of 2.8 angstroms, reveals all of the structural elements of the enzyme that contact DNA. The linker region that connects the central core of the enzyme to the carboxyl-terminal domain assumes a coiled-coil configuration and protrudes away from the remainder of the enzyme. The positively charged DNA-proximal surface of the linker makes only a few contacts with the DNA downstream of the cleavage site. In combination with the crystal structures of the reconstituted human topoisomerase I before and after DNA cleavage, this information suggests which amino acid residues are involved in catalyzing phosphodiester bond breakage and religation. The structures also lead to the proposal that the topoisomerization step occurs by a mechanism termed "controlled rotation."〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Stewart, L -- Redinbo, M R -- Qiu, X -- Hol, W G -- Champoux, J J -- CA65656/CA/NCI NIH HHS/ -- GM16713/GM/NIGMS NIH HHS/ -- GM49156/GM/NIGMS NIH HHS/ -- New York, N.Y. -- Science. 1998 Mar 6;279(5356):1534-41.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Biomolecular Structure Center and Department of Biological Structure, School of Medicine, University of Washington, Seattle, WA 98195-7742, USA. emerald_biostructures@rocketmail.com〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/9488652" target="_blank"〉PubMed〈/a〉

Description: Postdoctoral appointments can have different functions and meanings, depending on the field and whether the postdoc is a man or a woman. The Ph.D.'s-Ten Years Later study confirmed that in biochemistry, the postdoc, not the Ph.D., has become the general proving ground for excellence both in academia and industry. Because they spent a longer time in these "mandatory" postdocs, biochemists had the largest proportion of untenured faculty 10 to 13 years after the Ph. D. In mathematics, where substantially fewer postdoctoral positions are available, Ph.D.'s taking postdocs are more likely to obtain faculty positions, but this is true only for men. University administrators should be accountable for monitoring the total time spent in these positions and should provide administrative assistance for skills training, career growth, and the job search. In addition, creative solutions concerning the dual-career couple phenomenon are necessary.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Nerad, M -- Cerny, J -- New York, N.Y. -- Science. 1999 Sep 3;285(5433):1533-5.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Graduate Division, University of California, Berkeley, 424 Sproul Hall, Berkeley, CA 94720-5900, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/10477510" target="_blank"〉PubMed〈/a〉

Description: Photoactive yellow protein (PYP) is a member of the xanthopsin family of eubacterial blue-light photoreceptors. On absorption of light, PYP enters a photocycle that ultimately transduces the energy contained in a light signal into an altered biological response. Nanosecond time-resolved x-ray crystallography was used to determine the structure of the short-lived, red-shifted, intermediate state denoted [pR], which develops within 1 nanosecond after photoelectronic excitation of the chromophore of PYP by absorption of light. The resulting structural model demonstrates that the [pR] state possesses the cis conformation of the 4-hydroxyl cinnamic thioester chromophore, and that the process of trans to cis isomerization is accompanied by the specific formation of new hydrogen bonds that replace those broken upon excitation of the chromophore. Regions of flexibility that compose the chromophore-binding pocket serve to lower the activation energy barrier between the dark state, denoted pG, and [pR], and help initiate entrance into the photocycle. Direct structural evidence is provided for the initial processes of transduction of light energy, which ultimately translate into a physiological signal.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Perman, B -- Srajer, V -- Ren, Z -- Teng, T -- Pradervand, C -- Ursby, T -- Bourgeois, D -- Schotte, F -- Wulff, M -- Kort, R -- Hellingwerf, K -- Moffat, K -- New York, N.Y. -- Science. 1998 Mar 20;279(5358):1946-50.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Biochemistry and Molecular Biology, University of Chicago, Chicago, IL 60637, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/9506946" target="_blank"〉PubMed〈/a〉

Description: Genetic selection was exploited in combination with structure-based design to transform an intimately entwined, dimeric chorismate mutase into a monomeric, four-helix-bundle protein with near native activity. Successful reengineering depended on choosing a thermostable starting protein, introducing point mutations that preferentially destabilize the wild-type dimer, and using directed evolution to optimize an inserted interhelical turn. Contrary to expectations based on studies of other four-helix-bundle proteins, only a small fraction of possible turn sequences (fewer than 0.05 percent) yielded well-behaved, monomeric, and highly active enzymes. Selection for catalytic function thus provides an efficient yet stringent method for rapidly assessing correctly folded polypeptides and may prove generally useful for protein design.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉MacBeath, G -- Kast, P -- Hilvert, D -- New York, N.Y. -- Science. 1998 Mar 20;279(5358):1958-61.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉The Scripps Research Institute, Department of Chemistry, 10550 North Torrey Pines Road, La Jolla, California, 92037, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/9506949" target="_blank"〉PubMed〈/a〉

Description: A "switch" mutant of the Arc repressor homodimer was constructed by interchanging the sequence positions of a hydrophobic core residue, leucine 12, and an adjacent surface polar residue, asparagine 11, in each strand of an intersubunit beta sheet. The mutant protein adopts a fold in which each beta strand is replaced by a right-handed helix and side chains in this region undergo significant repacking. The observed structural changes allow the protein to maintain solvent exposure of polar side chains and optimal burial of hydrophobic side chains. These results suggest that new protein folds can evolve from existing folds without drastic or large-scale mutagenesis.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Cordes, M H -- Walsh, N P -- McKnight, C J -- Sauer, R T -- AI-15706/AI/NIAID NIH HHS/ -- New York, N.Y. -- Science. 1999 Apr 9;284(5412):325-8.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Department of Biology, Massachusetts Institute of Technology, Cambridge, MA 02139, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/10195898" target="_blank"〉PubMed〈/a〉

Description: The Yersinia pseudotuberculosis invasin protein promotes bacterial entry by binding to host cell integrins with higher affinity than natural substrates such as fibronectin. The 2.3 angstrom crystal structure of the invasin extracellular region reveals five domains that form a 180 angstrom rod with structural similarities to tandem fibronectin type III domains. The integrin-binding surfaces of invasin and fibronectin include similarly located key residues, but in the context of different folds and surface shapes. The structures of invasin and fibronectin provide an example of convergent evolution, in which invasin presents an optimized surface for integrin binding, in comparison with host substrates.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Hamburger, Z A -- Brown, M S -- Isberg, R R -- Bjorkman, P J -- New York, N.Y. -- Science. 1999 Oct 8;286(5438):291-5.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Division of Biology 156-29, Howard Hughes Medical Institute, California Institute of Technology, Pasadena, CA 91125, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/10514372" target="_blank"〉PubMed〈/a〉
