    Description: Oceanic observations indicate that abyssal mixing is localized in regions of rough topography. How locally mixed fluid interacts with the ambient fluid is an open question. Laboratory experiments explore the interaction of mechanically induced boundary mixing and an interior body of linearly stratified rotating fluid. A single oscillating bar produces a small region of turbulence along the wall at middepth. Mixed fluid quickly reaches a steady state height set by a turbulent-buoyant balance, independent of rotation. Initially, the bar is exposed on three sides. Mixed fluid intrudes directly into the interior rather than forming a boundary current. The circulation patterns suggest a model of unmixed fluid being laterally entrained into the turbulent zone. In accord with the model, observed outflux is constant, independent of stratification and restricted by rotation. Later the bar is laterally confines between two walls, which form a channel opening into the basin. A small percentage of mixed fluid enters a boundary current, which exits the channel. The bulk forms a cyclonic circulation in front of the bar, which blocks the channel and restricts horizontal entrainment. In the confined case, the volume flux of mixed fluid decays with time.

    Description: Experiments are reviewed in which a two-layer salt-stratified tank of water was mixed by turbulence. The density profile began as a single step and evolved to a smooth mixed profile. The turbulence was generated by many excursions of a horizontally moving vertical rod with Richardson number Ri 〉 0.9 and Reynolds Number Re 〉 600. There was almost perfect collapse of all the profiles to one universal profile as a function of a similarity variable. We develop a theoretical model for a simple mixing law with a buoyancy flux that is a function of internal Richardson number Rii. A similarity equation is found. A flux law that increases with small Rii and decreases with large Rii is considered next. Since no analytical solution is known, the similarity concept is tested by numerically integrating the equations in space and time. With buoyancy flux monotonically increasing with internal Richardson number, the similarity approach is valid for a profile starting from a slightly smoothed step. However, a shock forms for a mixing law with higher initial Rii (so that buoyancy flux decreases with Richardson number) and the similarity approach is invalid for those initial conditions.

    Description: Stimulated by new evidence from both "in situ" oceanic observations and results from numerical modelling, a laboratory study of quasigeostrophic flow and turbulence in a rotating homogeneous fluid has been undertaken. Two dimensional turbulence driven by a uniform distribution of sources and sinks which oscillate in time, can be fairly well reproduced in this context. Inertial time scales are about ten times smaller than Ekman spinup time, and typical Reynolds numbers read 2000. The observations emphasize the spectral tendency of the energy containing eddies. The case of no topography is first discussed. In steadily forced turbulence, it is observed that the energy containing scale is significantly larger than the forcing scale. In the decaying stage the red cascade is observed and rates of interaction are measured. Theoretical arguments for both behaviors are presented; the former concerning the forced turbulence case is believed to be new. The forcing is next applied over various large scale topographies, modelling the geophysical beta effect. The polar beta plane geometry preserves the above spectral characteristics but at the same time introduces anisotropy into the flow pattern. A broad westward mean flow develops in the north and is surrounded by a belt of cyclones lying on its southward side. The calculated second-order Eulerian mean flows induced by steadily and uniformly forced Rossby waves in a long zonal channel, exhibit much of the same momentum distribution in the inertial regime. In contrast, the "sliced cylinder" geometry which possesses no closed geostrophic contours drastically modifies the above picture. Both mean flow production and a large scale tendency for the eddies are inhibited. The geographical distribution of the eddy intensities and scales is now wildly inhomogeneous. The second aspect of this work is a study of the interaction of Rossby waves with mean flows. A zonally traveling, forced wave is generated near the southern boundary of a polar beta plane. Due to energy radiation in the free interior and (or) potential vorticity mixing by the finite amplitude waves, a westward zonal flow develops. The effect of the mean flow upon the forced steady waves is to weaken the anticyclones and intensify the cyclones. Pressure time series reveal a growth of harmonics and general spectral broadening as the waves travel freely inwards, suggesting active nonlinear interactions. An experimental test of Rhines' (1977) potential vorticity mixing theory is also presented at free latitudes. The decay period when the driving is suppressed shows that a net transfer from the waves to the mean flow kinetic energy occurs. Connection with hydrodynamic stability theory is discussed. Interaction of Rossby waves with an externally generated westward mean flow allows one to make a controlled study of the critical layer problem. For small amplitude waves, the mean flow is accelerated in the entire region between the forcing and the critical latitude which acts as a wall for mean wave momentum. In nonlinear runs the steady profile of the westward flow indicates that an accelerating force is acting everywhere, revealing the increasing transmission of wave momentum through the critical layer. At the same time, pressure measurements near the critical point show considerable fine structure developing over a long time scale. The third part deals with steady isolated source-sink flows in the sliced cylinder geometry. The response of the fluid to a meridionally oriented steady dipole extends exclusively westward of the forcing. The viscously balanced solutions are discussed and relevance to oceanic abyssal circulation is emphasized. With strong driving, the combination of a cyclone to the north and an anticyclone to the south is absolutely stable although the reverse configuration is not. A connection with a certain class of free, steady, isolated, inertial solutions developed recently by Stern (1976) is made.

    Description: The design and operation of a unique flow measuring instrument for bottom boundary layer studies in the marine environment is documented. The effectiveness of the instrument in acquiring data with which models of near bottom flows in the ocean can be tested is demonstrated by the results of a field experiment in Vineyard Sound. The instrument uses four sensors which measure the mean and fluctuating parts of the three components of the velocity vector at four heights above the sea bed. The sensors employ the acoustic travel time difference technique, and are designed to minimize sensor-induced flow disturbances. BASS, an acronym for Benthic Acoustic Stress Sensor, has a resolution of .033 cm/sec per least bit, a range of ±62 cm/sec, noise of .07 cm/sec in 10 sec, and an estimated accuracy of ±.5 cm/sec, referred to an in situ zero point. A complete set of velocity measurements is made every .750 seconds, each measurement being the vector component averaged over 15 cm. The data is internally recorded on digital cassette tape. Eight hours of continuous data can be recorded. BASS was deployed in a tidal flow in Vineyard Sound at a depth of 10 m where a time series of u, v, and w velocities at 26 cm, 46 cm, 96 cm, and 210 cm above the bottom was recorded. The mean velocity was determined by fitting each 6 hour series with a sixth order polynomial and the deviations from the polynomial, the fluctuating velocity components, were correlated to produce Reynolds stress profiles. The stress series shows very few negative stress events while the dominant positive events have an average duration of 5 seconds and exceed 30 dynes/cm2. Zero offset was removed from the mean by assuming a log profile at maximum ebb. Deviations from a log profile developed when the current dropped below 40% of maximum, i.e., when the flow could no longer be considered steady. A break in the Reynolds stress profile at 1 m suggested a larger length scale than the 1 cm bottom roughness was present in the flow. A value of u* was determined by using the quadratic drag law (u* = 1.56 cm/sec), the log profile method (u* = 1.60 cm/sec), and the eddy correlation method (u* = 1.91 cm/sec). Integral length scales of 5 m cross-stream, and 2.5 m vertically were identified by correlation calculations. Two length scales were present in the downstream direction, 5 m within 1 meter of the wall and 8 m further from the wall.

    Description: Estuarine turbulence is notable in that both the dissipation rate and the buoyancy frequency extend to much higher values than in other natural environments. The high dissipation rates lead to a distinct inertial subrange in the velocity and scalar spectra, which can be exploited for quantifying the turbulence quantities. However, high buoyancy frequencies lead to small Ozmidov scales, which require high sampling rates and small spatial aperture to resolve the turbulent fluxes. A set of observations in a highly stratified estuary demonstrate the effectiveness of a vessel-mounted turbulence array for resolving turbulent processes, and for relating the turbulence to the forcing by the Reynolds-averaged flow. The observations focus on the ebb, when most of the buoyancy flux occurs. Three stages of mixing are observed: (1) intermittent and localized but intense shear instability during the early ebb; (2) continuous and relatively homogeneous shear-induced mixing during the mid-ebb, and weakly stratified, boundary-layer mixing during the late ebb. The mixing efficiency as quantified by the flux Richardson number Rf was frequently observed to be higher than the canonical value of 0.15 from Osborn (J Phys Oceanogr 10:83–89, 1980). The high efficiency may be linked to the temporal–spatial evolution of shear instabilities.

    Description: Nonlinear quasigeostrophic flows in two layers over a topographic slope are considered. The evolution depends on the size of two parameters which indicate the degree of nonlinearity at depth. The first measures the importance of relative vorticity advection and the second of stretching vorticity. Two types of isolated vortex are used to examine the parameter dependence. An initially barotropic vortex remains barotropic only when the first parameter is large, otherwise topographic waves dominate at depth. An Initially surface-trapped vortex larger than deformation scale is baroclinically unstable when the second is large, but is stabilized by the slope otherwise. Both parameters are also relevant to cascading geostrophic turbulence. If the stretching parameter is large, a "barotropic cascade" occurs at the deformation radius (Rhines, 1977) and the cascade "arrests" when the relative vorticity parameter is order unity. If small, layer coupling is hindered and the cascade is arrested at the deformation scale, with the flow dominated by isotropic surface vortices. In both cases, the distinction between vortices and waves is transparent when viewing potential vorticity. It is more difficult to identify waves and vortices from the streamfunction fields, because the waves are present in both layers.

    Description: A series of laboratory experiments were performed to ascertain the extent and manner of turbulence modification induced by low concentration suspended loads of near neutral buoyancy. Hot-wire measurements of the fluid velocity field of free surface flows were obtained in a specially designed flume recirculating a dielectric liquid and 0.5 mm" diameter spherical plastic particles. In the fixed Reynolds number flow (16,800) the data obtained at six different concentration levels, ranging from 0 to 3.5% by volume, indicate that the presence of particles produces substantial turbulence changes. Even at this low level the mean velocity profile shows an increasing gradient near the bed and sharp deviation from a logarithmic profile. The rms level of each of the velocity components u', v' and w' increases, indicating a general rise in turbulence intensity. The Reynolds stress ρ u'v' increases, and its maximum value shifts away from the bed. The overall scale of turbulence appears to remain unchanged. The data indicate that offhand neglect of suspended particle presence is an oversimplification. There is a similarity between these data and those obtained under adverse pressure gradients. Some effort is made to clarify the altered turbulence production meechanism, and some future experimental work is proposed.

Significativement plus de femelles de P. melanarius ont été capturées dans les parcelles traitées pendant cette période d'accroissement après traitement. Une diminution correspondante dans les populations de proies a été observée à la suite des traitements. Chaque femelle de P. melanarius des parcelles traitées avait une fraction significativement moins importance de son intestin remplie d'aliments solides (arthropodes) que celles des parcelles non traitées. Des auteurs précédents avaient observé des taux de capture plus rapides dans les trappes et ne plus grande mobilité des carabes affamés. La réduction des populations proies par l'application d'insecticides affamerait les carabes dont l'activité serait par suite plus élevée. Comme le taux de captures dans les trappes est dû à la fois à la taille de la population et à son activité, on en déduit que la capture par trappes seule ne peut donner une image exacte de l'effet des insecticides sur les populations de carabes dans les champs.
    Notes: Abstract During large-scale field experiments in 1981 and 1982, designed to assess the effects of the insecticides Fenitrothion and Sumicidin (Fenvalerate) on beneficial arthropods in spring barley, significant increases in pitfall-trap catches of Pterostichus melanarius (Illiger) were observed in treated plots compared to untreated controls, several weeks after treatment. Significantly more female P. melanarius were caught in treated plots during these post-treatment increases. Corresponding decreases were observed in prey populations following treatments. Individual female P. melanarius from treated plots had significantly fewer of their gut areas full of solid (arthropod) food when compared to those from untreated control plots. Earlier workers observed both faster recapture rates in pitfalls and higher mobility in hungry carabid beetles. The following hypothesis is suggested: reducing prey populations by the application of insecticides results in hungrier carabid beetles with consequently higher activity. Since pitfall-trap catch is determined not only by population size but also activity, it is argued that pitfall-trap catches alone cannot give a true measure of the effects of insecticides on carabid populations in the field.

Inositversorgte Zellen zeigen eine Verschiebung der Fettsäuren von den Triacylglycerinen in die Phospholipide, im besonderen in Phosphatidylcholin und Phosphatidylinosit. Eine verstärkte Übertragung der Fettsäuren von Triacylglycerinen auf Phospholipide konnte festgestellt werden, wenn vormarkierte Zellen auf ein Nährmedium, welches Cerulenin enthielt, übertragen wurde. Cerulenin inhibiert die Fettsäuresynthese und ruft in wachsenden Zellen Fettsäuremangel hervor. Inositdefiziente Hefezellen, welche einen erhöhten Triacylglycerinspiegel aufweisen, verwenden diese Triacylglycerine unter Fettsäuremangelbedingungen ebenfalls für die Synthese von Phospholipiden, besonders von Phosphatidylcholin, Phosphatidyläthanolamin und Phosphatidylserin. Da aus früheren Arbeiten bekannt ist, daß inSaccharomyces carlsbergensis praktisch keine β-Oxidation existiert, können die Triacylglyerine in diesem Hefestamm als Speicher für Fettsäuren angesehen werden, welche zur Synthese von Phospholipiden dienen.
    Notes: Abstract The turnover of lipids was studied in the yeast,Saccharomyces carlsbergensis ATCC 9080, after prelabeling of the cells with [3H] oleic acid and [14C] palmitic acid. In inositol supplemented cells, a redistribution of fatty acids from triacylglycerols to phospholipids (mainly phosphatidylcholine and phosphatidylinositol) could be demonstrated. An increased transfer of fatty acids from triacylglycerols to phospholipids was observed when prelabeled cells were transferred to a growth medium containing cerulenin, which inhibits fatty acid synthesis and thus induces fatty acid deficiency in the growing cells. Inositol deficient cells contain increased levels of triacylglycerols, which are equally well utilized for phospholipid (mainly phosphatidylcholine, phosphatidylethanolamine and phosphatidylserine) synthesis under conditions of fatty acid deficiency. The present results together with the previous finding that β-oxidation is practically absent inSaccharomyces carlsbergensis suggest that in this yeast triacylglycerols function as storage of fatty acids which can be mobilized for phospholipid biosynthesis.

Qualitative Studien mit HPLC ergaben in frischen Puppen ein Verhältnis von 3:1 Ecdyson zu 20-Hydrooxyecdyson. Pharatstadien enthielten vor allem 20-Hydrooxyecdyson. Das beobachtete einzige Maximum im Titer stimmt überein mit den Resultaten bei andern untersuchten Coleopteren.

Titer declined to 299 pg/mg by the pharate adult stage and to 193 pg/mg body wt just before adult emergence.     Description / Table of Contents: Zusammenfassung Raupen von Malacosoma americanum (F.), einer oligophagen Art, die sich vor allem auf Prunus und andern baumartigen Rosaceen entwickelt, wurden verglichen mit Raupen der polyphageren Verwandten M. disstria Hb. und zwar im Hinblick auf deren Empfindlichkeit auf Unterschiede im Blatt ihrer gemeinsamen Wirtspflanze, Prunus serotina Ehrh. Das Puppengewicht und die Entwicklungszeit bis zur Verpuppung wurden gemessen bei Raupen, welche auf Blättern von freiwachsenden und von beschatteten Jungpflanzen gezüchtet worden waren. Die Blattunterschiede hatten eine ausgesprochene Wirkung, aber es gab keine Unterschiede in der Reaktion der beiden Arten. Dieser Vergleich lässt vermuten, dass die Empfindlichkeit auf intraspezifische Unterschiede der Wirtspflanzenqualität bei wirtsspezifischen und polyphagen Arten gleich ist.     Notes: Abstract Larvae of Malacosoma americanum (F.)(Lepidoptera: Lasiocampidae) an oligophagous species that feeds primarily on Prunus and other rosaceous trees, were compared to larvae of the more highly polyphagous congener M. disstria Hb., with respect to their sensitivity to variation in the foliage of a common host plant, Prunus serotina Ehrh. Pupal weight and time to pupation were measured on larvae reared on foliage from open-grown and from shaded saplings. The difference in foliage had a pronounced effect, but no difference was evident between the species in their response to the variation in foliage. This comparison implies that sensitivity to intraspecific variation in host quality does not differ between host-specific and generalized species. However, results from other species suggest that some species of insects do differ in this respect.

Moreover, the two species did not differ in any measure of efficiency. Our results are compatible with the conclusion that specialized phytophagous insects do not use their host plants with greater physiological efficiency than do generalized insects.

En tenant compte de la mortalité larvaire et des indices de croissance (G.I.), ces plantes ont été divisées en trois catégories principales: 1 celles provoquant une mortalité larvaire de 0–30% et ayant des G.I. ≥60%, constituent les plantes hôtes convenables (Vigna unguiculata seule); 2 les plantes qui provoquent une mortalité larvaire de 30≤50% et ont des G.I. de 30 à 60% de la plante hôte principale (Cajanus cajan, Crotalaria amazonas, C. saltiana, C. mucronata; 3 les plantes qui causent 50–100% de mortalité larvaire et dont les G.I. sont inférieurs à 30% de la plante hôte principale (Crotalaria retusa, C. juncea, C. misereniensis). Les résultats déjà publiés sur les choix du lieu de ponte des femelles et l'utilisation de C. juncea comme plante piège, sont discutés à la lumière de ces données nouvelles.
    Notes: Abstract Flowers of eight plant species were evaluated under laboratory conditions for their suitability as larval growth media for the cowpea pod borer, Maruca testulalis. The plants tested were Crotalaria retusa, C. juncea, C. saltiana, C. misereniensis, C. amazonas, Cajanus cajan and the principal host of the borer, Vigna unguiculata (cowpea), was included for comparison. Based on the data obtained on larval mortality and growth indices (GI) the plants were divided into 3 categories namely: I: Those causing 0–30% mortality and having GI value ≥60% form suitable host plants. This group only included V. unguiculata. — II: Those plant species causing 30≤50% larval mortality and having GI value 30%≤60% of the principal host plant (Cajanus cajan, Crotalaria amazonas, C. saltiana, C. mucronata). This group of species is marginally suitable as hosts. — III: Plants causing 50–100% larval mortality and having GI value ≤30% of principal host plant (C. retusa, C. juncea and C. misereniensis). Previously published data on the oviposition preference of the adult moth are discussed in the light of the present findings and the use of C. juncea as a possible trap crop.

Les distances génétiques entre des sibling species ou des non-sibling species sont très larges. Il en est conclu que de telles estimations ne fournissent pas beaucoup d'informations sur le phénomène de spéciation même. Des coéfficients de reproduction consanguine (F ST) ont été calculés pour des populations d'Y. padellus provenant de six plantes-hôtes. La valeur moyenne F STest .030±.012. Les valuers de F ST des populations recueillies sur Crataegus, (plante-hôte habituelle d'Y. padellus), sont inférieures 2–3.5 fois à celles des populations de l'ensemble des autres plantes-hôtes. L'apparition de races en fonction de l'hôte, mesurée par les différences importantes dans de fréquences des allozymes entre populations sympatriques sur plusieurs plantes-hôtes, a été examinée dans quatre régions. Il apparaît ainsi que la formation de races en fonction de l'hôte se produit chez Y. padellus et que la spéciation sympatrique est un évènement très vraisemblable.
    Notes: Abstract The amount of genetic differentiation at various levels of evolutionary divergence (conspecific populations, sibling species and non-sibling species) in Yponomeuta was determined. Genetic distances between siblings or non-siblings were found to cover a wide range. It is concluded that such estimates do not give much information on the speciation process itself. Inbreeding coefficients were calculated for populations of Y. padellus from a total of six host plants. The grand mean F ST value is 0.030±0.012. F ST values for populations sampled from Crataegus, the common food plant of Y. padellus, are 2–3.5 times smaller than those for populations from the other food plants taken together. Host race formation, as measured by significant differences in allozyme frequencies between sympatric populations on two or more food plants, was investigated in four areas. Host race formation seems to occur in Y. padellus and sympatric speciation is a likely event.

Un récepteur cellulaire additionnel sensible à l'acétate (Z.)-9-tétradécadiényle et à l'acétate (Z.E.)-9,12-tétradécadiényle a été trouvé sur l'antenne du mâle, mais il n'était pas activé par l'extrait de la femelle. Sur le terrain Z,E-9,11–14:Ac, présenté seul, attirait des nombres importants de mâles de D. abietella. L'addition de l'acétate (Z)-9-tétradécényle a inhibé l'attraction des mâles par les pièges.
    Notes: Summary Gas chromatographic analyses coupled with electro-antennographic detection indicated that (Z,E)-9,11-tetradecadienyl acetate (Z,E-9, 11–14:Ac) is a pheromone component of the cone pyralid Dioryctria abietella. Gas chromatographic-mass spectrometric analyses confirmed the presence of a tetradecadienyl acetate with mass spectrum and retention index identical to Z,E-9,11–14:Ac. A receptor cell sensitive to both Z,E-9,11–14:Ac and the female extract was identified on the male antenna. An additional receptor cell sensitive to (Z)-9-tetradecenyl acetate and (Z,E)-9,12-tetradecadienyl acetate was found on the male antenna but was not activated by the female extract. In the field Z,E-9,11–14:Ac presented alone attracted significant numbers of male D. abietella. Addition of (Z)-9-tetradecenyl acetate inhibited the attraction of males to traps.

    Description / Table of Contents: Zusammenfassung Fluormevalosäure (FMev), bekannt als ein spezifischer Inhibitor der Biosynthese des Juvenilhormons (JH) in Insekten, wurde in 0,1–50 μg/Tier Dosis topikal an 3., 4. und 5. Larvenstadien von Hyphantria cunea getestet. Die Raupen wurden auf einem semi-synthetischen, künstlichen Nährboden bei 25° und unter Langtagsbedingungen (18 : 6 St., Licht/Dunkel) gezüchtet. Diese Verbindung rief drei verschiedene Typen spezifischer Reaktionen hervor: 1) verfrühte Metamorphose, 2) gehemmte Häutung und 3) verlängerte Larvenentwicklung. Vor der verfrühten Verpuppung wurde normales Verhalten beobachtet. Die Larven des 3. oder 4. Stadiums häuteten sich meist erst nach einem interkalaren Larvenstadium in verfrühte Puppen. Unter den drei Larvenstadien erwies sich das 5. Stadium gegen die Anti-JH-Verbindung am empfindlichsten. In allen getesteten Entwicklungsphasen wiesen die frisch gehäuteten Larven die höchste Empfindlichkeit gegen FMev auf. Nach der Häutung wurde stufenweises Absinken der FMev-Empfindlichkeit beobachtet, im 5. Larvenstadium verursachte die Verbindung jedoch selbst am letzten Tag des Wachstums zu einen relativ hohen Prozentzahl verfrühte Verpuppung. Eine zweite typische Wirkung von FMev war die Hemmung der Häutungsprozesse. Zwei grundlegende Stufen der Häutungsstörungen unterschieden sich voneinander: 1) Als Folge der Anwendung hoher Dosen von FMev konnte die Mehrzahl der Raupen die alte Larvenkutikula nicht öffnen und ging deswegen in kurzer Zeit zugrunde. 2) Bei Behandlung mit niedrigen Dosen der Anti-JH-Verbindung häuteten sich einige Hyphantria-Larven scheinbar normal; nach der Häutung waren aber alle unfähig, die normalen Bewegungen und die Nahrungsaufnahme fortzusetzen. Auch die vorzeitigen Puppenhäutungen wurden in meisten Fällen durch die Anti-JH-Behandlung gehemmt. Die morphogenetischen Wirkungen von FMev konnten durch eine topikale Behandlung mit Hydroprene, einem hochaktiven JH-Analogen, vollständig oder teilweise verhindert werden.
    Notes: Summary Fluoromevalonate (FMev, ZR-3516) known as an inhibitor of JH biosynthesis was topically applied in 0.1 to 50 μg/specimen doses to the 3rd, 4th, and 5th instar caterpillars of Hyphantria cunea Drury. The anti-JH compound induced 3 main types of specific responses: 1) precocious metamorphosis, 2) inhibition of ecdysis, and 3) prolongation of larval development. Precocious pupation was accompanied by behavioural events typical of normal pupation. Third and 4th instar larvae metamorphosed prematurely mostly with the intervention of an intercalary larval instar. The 5th instar exhibited the highest sensitivity to the anti-JH agent. Within each larval stage the freshly moulted insects proved to be the most susceptible to FMev. Afterwards, the incidence of morphogenetic reaction gradually decreased with age. In another fraction of Hyphantria larvae not responding with precocious pupation, FMev evoked varying degrees of ecdysial disturbance which always resulted in the death of caterpillars. In most cases the anti-JH compound inhibited the premature pupal moult, too, and these affected insects died as tanned pharate pupae. A complete or partial “rescue” from the effects of FMev was elicited, if simultaneously or subsequently, a single topical dose of a JH analogue, hydroprene was also administered.

    Notes: Abstract Newly-emerged larvae of a salvinia weevil, Cyrtobagous sp. spent from 1–4 days browsing on the roots and from 0–9 days in the buds of the host plant, Salvinia molesta, prior to tunnelling into the rhizome of this aquatic weed. Although larvae were able to survive up to 38 days browsing on roots when rhizomes were withheld, they were unable to complete development. After tunnelling began, larval development between 21° and 31° was dependent on temperature and nitrogen levels in the host. Larvae failed to develop at 17°. High nutritional intake by the larvae reduced larval development time but did not influence duration of their subsequent pupal development. The feeding sites and plant damage produced by individual larvae were assessed.

    Description / Table of Contents: Résumé Nous avons examiné quelques aspects des relations entre le parasitoïde Aphidius rhopalosiphi et son hôte Sitobion avenae. La fécondité potentielle à l'émergence (144 œufs) était inférieure à la fécondité “réalisée”: nombre de pucerons momifiés (212). La plupart des œufs étaient pondus dans la première semaine de la vie imaginale (fig. 1). Le super-parasi-tisme s'est produit dans les conditions expérimentales, mais la distribution des larves (fig. 2) s'écartait souvent de la distribution au hasard, ce qui indiquait une certaine discrimination parmi les hôtes. La longévité moyenne était de 13,1 jours et le maximum de 23 jours. Le taux sexuel (fig. 3) pour l'ensemble de la descendance était 1/1, mais changeait dans le temps. Après le premier jour de ponte, plus de femelles que de mâles étaient obtenues; mais après 6 jours, il n'y avait presque plus que des mâles. Le taux de rencontre des aphides et des parasitoïdes augmentait avec la taille (fig. 4a). Le temps de prospection de l'hôte était en moyenne de 2 secondes, mais dans certains cas il atteignait 10 secondes ou plus. La proportion de rencontres efficaces était plus faible au 4ème stade (fig. 4b). Il n'y avait pas de différence significative dans le nombre de larves de parasitoïdes obtenues par rencontre pendant les stades 1 à 3 des pucerons, mais elle était significativement inférieure au 4ème stade par rapport au second. Les taux globaux de succès (taux de rencontre et nombre de larves formées par rencontre) étaient plus élevés aux 2ème stades des pucerons. La réponse fonctionnelle était sigmoïde (fig. 5).
    Notes: Summary Aphidius rhopalosiphi produced 212 offspring on average but in one case 509 deposited eggs were found. Superparasitization occurred but the frequency distribution of parasitoid larvae in aphids differed significantly from random, indicating a certain degree of host discrimination. Average life span of adults was 13.1 days and sex ratio was 1:1. It changed in time among successively produced offspring. Handling time was about 2 sec and was somewhat longer in the fourth instar than in the second. Second and third aphid instars were preferred for oviposition. Functional response was sigmoid and at an aphid density of 100 aphids per cage percentage parasitization decreased.

    Description / Table of Contents: Résumé L'examen en olfactomètre a porté sur les réactions face à d'autres femelles de la même espèce, de femelles vierges ou ayant copulé d'Heliothis armigera Hübner et H. zea Boddie. Le lot comprenait 8 femelles, vierges ou ayant copulé en présence d'une femelle vierge ou ayant copulé. Les 4 combinaisons possibles de femelles vierges et de femelles ayant copulé ont été examinées avec 12 répétitions pour chaque espèce. Un extrait de l'extrémité de l'abdomen de femelles vierges d'une espèce a été présenté aux femelles de l'autre espèce pour examiner les possibilités de réactions interspécifiques aux phéromones. Pour chaque espèce, les réactions interspécifiques de répulsion entre femelles ont été hautement significatives par rapport aux témoins, à l'exception toutefois des réactions de femelle ayant copulé face à des femelles ayant elles aussi copulé. Les répulsions moyennes chez H. armigera et H. zea pour les 8 femelles de chaque expérience ont été: a) vierges en présence d'une vierge: 7,33 et 7,66; b) vierges en présence d'une femelles ayant copulé: 5,76 et 5,58; c) femelles ayant copulé en présence d'une vierge: 4,67 et 4,83. Les différences sont hautement significatives entre chaque paire de moyennes et entre chaque paire et le lot témoin; 3,17; 3,17; 3,42; 4,00 pour H. armigera; 3,17; 3,50; 2,83 et 3,75 pour H. zea. Les femelles vierges des deux espèces, H. armigera et H. zea ont présenté une réaction de répulsion en présence d'un extrait de l'abdomen de l'autre espèce; les répulsions moyennes étant respectivement 5,53 et 5,33 contre 3,83 et 3,58 pour le lot trémoin. On peut en conclure que ces répulsions doivent entraîner une tendance à la répartition uniforme.
    Notes: Abstract An olfactometer was used to determine the effect of pheromones released by females of the bollworms Heliothis armigera (Hübner) and H. zea (Boddie) on females of the same species. Four combinations of virgin and mated females were tested for repellency of one to the other. Evidence is presented that females of the two bollworms were repelled by females of the same species. In addition, extracts of virgin female abdomens of each species repelled virgin females of the other species.

Mehrere Versuche über die Eiablagepräferenz der Parasiten zeigten, dass mehr Eier in Raupen von Baumwolle als in Raupen von Bohnen oder gar von Tomaten abgelegt wurden. Die Wespen unterschieden aber nicht zwischen H. zea und H. virescens. Die Eiablagepräferenzen waren unabhängig von den Kombinationen Wirtsinsekt/Wirtspflanze, in denen sich die Parasiten entwickelt hatten. Die Ergebnisse weisen darauf hin, dass die Pflanze, auf der die Wirtsraupe frisst, wichtig ist für die Wahrscheinlichkeit eines Parasitenangriffs und für den Erfolg der Parasitierung.
    Notes: Summary Microplitis croceipes (Cresson), a parasitoid of Heliothis larvae, was reared on 9 host insect/plant combinations. In general, parasitoid survivorship was (1) higher in host larvae that were reared on cotton than in hosts reared on either bean or tomato, (2) higher in H. zea hosts than in H. virescens and (3) higher in a laboratory strain of H. zea than in a wild strain. A series of parasitoid oviposition preference tests showed that wasps laid more eggs in host larvae on cotton plants than on bean and more on bean than on tomato. However, the wasps did not distinquish between H. zea and H. virescens hosts. Wasp oviposition preferences were not conditioned by the host insect/plant combination in which the parasitoid developed. These results indicate that the plant on which host larvae feed is an important factor in determining both the likelihood of parasitoid attack and the probability of successful parasitism.

Parasitoide schlüpfen in Normalgenerationen aus dem 3., dem 4. und dem 6. Stadium des Wirts; in Ausnahmegenerationen schlüpfen sie aus dem 2. und 5. Stadium. Bis zu 70 Parasitoide können aus einer Altraupe (6. Stadium) schlüpfen und die Dauer dieses Stadiums kann bis auf 2 Wochen verlängert werden. Die Ausnahmegenerationen von A. bignellii aus Zweitlarven dürften aus Eiablagen in frühe Erstlarven stammen. Fünftlarven des Wirts, aus denen Parasitoide schlüpfen, sind ungewöhnlich klein und fressen nicht; sie dürften das Resultat sein eines Uebermasses von Apenteles-Eiern, die in frühe Viertlarven gelegt wurden. Die Synchronisation zwischen dem Parasitoiden und dem Wirt während der Zeit, da E. aurinia im Puppen-, Adult- oder Eistadium ist, wird aufrechterhalten durch ein verlängertes Coconstadium von Apanteles. Die Puppen des Parasitoiden entwickeln sich normal und die Adulten schlüpfen, bleiben aber bis 4 Wochen lang im Cocon, bevor sie sich eine Ausgangsöffnung machen.     Notes: Abstract The gregarious endoparasite, Apanteles bignellii Marshall is specific to the nymphalid butterfly, Euphydryas aurinia (Rottemburg) in the British Isles. The synchronisation between host and parasitoid is described at a site near Oxford, England where both occur. Three regular generations of A. bignellii occur in one generation of the host in the studied population. Relevant features of the biology of A. bignellii and E. aurinia are described, including a method of distinguishing the number of Apanteles larval instars based upon shed cuticle remnants. Mechanisms for host-parasitoid synchronisation are outlined, especially a protracted parasitoid cocoon stage when the host is unavailable for attack during the chrysalis, adult and egg stages. Cool, but sunny weather conditions in spring can influence the degree of parasitisation experienced by final instar host caterpillars. The timing of adult A. bignellii emergence and subsequent attack on early instar hosts can lead to additional, partial, generations of parasitoids from second and fifth instar hosts.

Les aminopeptidases ont été trouvées dans le liquide de la lumière (18%), mais étaient principalement associées avec les fractions insolubles des cellules du ventricule postérieur (82%).     Notes: Abstract Endoproteinase activity was confined to luminal fluid from anterior and posterior ventricular regions of midguts of larvae of the sweetpotato weevil,Cylas formicarius elegantulus (Summers). Aminopeptidase was found in luminal fluid (18%) but was primarily associated with insoluble fractions from cells of the posterior ventriculus (82%). Depending on substrate, carboxypeptidase activity was about equally distributed between luminal fluid and insoluble fractions from posterior ventriculus cells. Amylase was found in luminal fluid in both the anterior and posterior ventriculus. Five bands of amylase activity were detected on starch zymograms following electrophoresis of whole midgut samples. Three of the amylase bands may be plant-derived. At least two α-glucosidases were present, one secreted and one bound to anterior ventriculus cells. β-Glucosidase activity was bound to anterior ventriculus cells whereas α-galactosidase activity was confined to luminal fluid. β-Galactosidase and cellulase activities were found in both luminal fluid and the insoluble cell fraction. Thus, initial digestion of large protein and carbohydrate polymers occurs in luminal fluid in both anterior and posterior ventriculus regions. However, terminal digestion of oligopeptides occurs primarily in the posterior ventriculus while terminal digestion of oligosaccharides occurs in the anterior ventriculus. A 20-fold difference in trypsin-inhibitor concentration was found among five sweetpotato cultivars. Endoproteinases from sweetpotato weevil larvae were inhibited by extracts from the cultivars but cultivars with relatively high concentrations of inhibitor were previously shown to be susceptible to weevil attack in field trials.

Das Wanderverhalten der Larven wird durch die Populationsdichte beeinflußt, wodurch annähernd eine konstante Dichte erhalten bleibt. Das Verhalten der individuellen Larven is dabei nicht statistisch homogen. Es gibt ‘Wanderer’ und ‘Nicht-Wanderer’. 3. Diese Verhaltensunderschiede stehen möglicherweise im Zusammenhang mit physiologischen und morphologischen Faktoren, die den individuellen Fortpflanzungserfolg und das überleben beeinflussen können; ‘Wanderer’ sind dunkler, entwickeln sich schneller und das Gewicht ihrer Puppen ist niedriger als das der ‘Nicht-Wanderer’. 4.     Notes: Abstract The dispersive behaviour of larvae of a polyphagous, wide-spread geometrid (Ectropis excursaria (Guenée)) was studied by examining responses to environmental and endogenous variables. It was found that differences in behaviour can be affected by environmental factors such as light, temperature, density, and plant background as well as some physiological and morphological features. The implications of these relationships are discussed as adaptative strategies to a varying environment.

    Description / Table of Contents: Résumé Vingt-deux espèces de plantes, dont 10 planteshôtes (Solanées), ont été testés comme plantes alimentaires pour des chenilles de ler stade de Manduca sexta. Sur cet ensemble, seulement 12 plantes (dont 9 plantes hôtes) induisaient la prise de nourriture et permettaient la croissance jusqu'au 5ème stade. La diversité des résultats suggère que les plantes pouvaient être classées en hôtes, non-hôtes acceptables et non-hôtes refusés. En utilisant le test du choix alimentaire préférentiel entre deux rondelles de feuilles, les chenilles néonates de ler stade ont préféré nettement les plantes-hôtes aux autres. Cette préférence initiale pour les plantes-hôtes était préservée quand les cheniles étaient élevées sur plantes-hôtes, mais devenait moins nette ou disparaissait pour des chenilles élevées sur d'autres plantes acceptées. Ainsi l'oligophagie ches M. sexta n'est pas induite, mais doit être héritée. Les chenilles néonates, aussi bien que celles de 5ème stade, présentent des préférences hiérarchisées parmi les plantes hôtes ou non. La seule frontiere nette observée était entre espèces de plantes acceptables ou non. Les hiérarchies préférentielles des chenilles du 5ème stade différaient légèrement lors-qu'elles avaient été élevées sur deux plantes-hôtes différentes. La différence essentielle était l'observation d'une préférence accrue pour l'espèce ayant servi à l'élevage, mais deux autres plantes-hôtes changaient aussi de position hiérarchique. La cause de ces changements de préférence a été approfondie, les chenilles étant élevées sur des feuilles de chaque espèce acceptable (hôte ou non). Leurs préférences alimentaires ont été définies en utilisant des combinaisons diverses (hôte x hôte, hôte x non-hôte acceptable, non-hôte acceptable x non-hôte acceptable). L'induction de la préférence alimentaires a été obtenue dans ces trois associations. Ceci montre que l'induction des choix alimentaires chez M. sexta n'est pas limitée aux plantes-hôtes. Le degré d'induction de la préférence alimentaire variait de très fort à indécelable; il dépendait de l'association examinée. La source de la variabilité de cette induction a été examinée en fonction de la relation entre la force de l'induction et les rapports taxonomiques des plantes associées. La relation obervée était inversée pour M. sexta. L'examen des données de la littérature ont révélé une relation du même type pour les autres espèces de Lépidoptères.
    Notes: Abstract Ten host plant (Solanaceae) and twelve non-host plant species were tested as foodplants for first instar larvae of the tobacco hornworm, Manduca sexta. Only nine host and three non-host plant species elicited feeding and supported growth up to fifth instar. The range of acceptability suggested that plants be divided into hosts, acceptable non-hosts, and unacceptable non-hosts. Using the two-choice feeding preference test we found that the initial preference for hosts was preserved when larvae were reared on hosts, but was less strong or absent for larvae reared on acceptable non-hosts. Thus oligophagy in the tobacco hornworm is not induced, but must be inherited. Newly-hatched first instar larvae and fifth instar larvae showed a preference hierarchy among both hosts and non-hosts. Fifth instar larvae reared separately on two different host species showed slightly different preference hierarchies among hosts. The preference for the rearing plant was increased and also two other host species changed positions in hierarchies. Feeding preferences of larvae reared on hosts or acceptable non-hosts were determined using plant combinations of host vs. host, host vs. acceptable non-host, and acceptable non-host vs. acceptable non-host. Induction of feeding preference was found in all three of these categories. This shows that induction of feeding preference in the tobacco hornworm is not restricted to host plant species. The degree to which feeding preferences were induced ranged from very strong to undetectable and dependend on the plant species paired. The strength of induction in the tobacco hornworm was found to correlate inversely with taxonomic relatedness of the plant species paired. Analysis of induction data from the literature revealed a similar correlation for other lepidopteran species.

La méthode gravimétrique est basée sur le rapport poids sec/poids frais de feuilles consommées par les chenilles. Avec la méthode planimétrique, la quantité d'aliment proposée aux chenilles est déterminée par les tracés de la surface des feuilles et le contenu de matière sèche par unité de surface des feuilles. La méthode de planimétrie permet l'utilisation de plus grands rameaux de feuilles et de cages d'élevage extérieures en gaze. Il n'y avait pas de différence appréciable dans les éléments du budget (croissance, ingestion et déjection), ni aucune différence dans la variabilité observée des données attribuable à la méthode utilisée. Cependant, la variabilité attendue d'après la précision des mesures avec la méthode gravimétrique est inférieure à celle de la méthode planimétrique. est inférieure à celle de la méthode planimétrique. Des éléments expérimentaux, inhérents à la méthode gravimétrique, introduisent une variabilité dans les mesures que l'on n'a pas avec la méthode planimétrique. 30–60% de la variabilité dans la consommation ont été attribués à des paramètres internes à la chenille, même quand elles provenaient toutes de la même ooplaque.
    Notes: Abstract Gravimetric and a combination areal-gravimetric methods for determining dry matter budgets for leaf eating Lepidoptera were compared. The gravimetric method is based on dry weight/live weight ratios of the leaves fed to the larvae. In the areal-gravimetric method, the quantity of food offered to the larvae is determined from the area of leaf tracings and the dry matter content per unit area of the leaves. The areal-gravimetric method permits the use of larger leaf sprays and an open, gauze enclosed rearing chamber. There were no consistent differences in budget factors (growth, ingestion or egestion), nor were there any differences in the observed variability of the data attributable to the method used. However, the expected variability based on instrument precision for the gravimetric method is less than for the areal-gravimetric method. Experimental factors inherent in the gravimetric method introduce variability to the measurements that are not present in the areal method. Thirty to 60% of the variability in budget factors was attributed to intrinsic properties of the larvae, even though the larvae were taken from the same egg masses.

    Description / Table of Contents: Résumé Colpoclypeus florus Walk., ectoparasite grégaire de Tortricidae, est capable de provoquer un taux de parasitisme élevé dans plusieurs types de cultures, y compris dans les vergers de pommiers. Dans les vergers aux Pay-Bas, ce parasite n'abonde qu'en été sur la première génération d'Adoxophyes orana. Les autres tortricides échappent à ses attaques, parce qu'elles ne peuvent être attaquées qu'au printemps. Le but de ces études était donc la mise au point d'une technique d'élevage de masse de C. florus, pour relâcher ce parasite en grand nombre dans les vergers au printemps. La technique décrite a permis de produire plusieurs milliers de parasites par semaine. Des lâchers de 16 000–28 000 parasites par ha dans un verger soumis à un programme de lutte intégrée, bien que faisant des victimes parmi toutes les espèces importantes de tortricides présentes, étaient loin d'occasionner le taux de parasitisme escompté. Le nombre de degrés-jours nécessaire au développement des différents stades de C. florus a été déterminé au laboratoire, et son cycle évolutif, inconnu aux Pays-Bas, puisque le parasite ne se trouve qu'en été, a été calculé et comparé aux observations faites en élevages en plein air. C. florus a de 4 à 5 générations par an, et la comparaison de sa phénologie avec celle des différentes tordeuses explique son absence hors de l'été. Au laboratoire, Archips podana était plus parasité, et Pandemis heparana moins parasité qu'A. orana. Le nombre moyen de descendants produits par femelle de C. florus et par chenille-hôte variait selon les hôtes, dépendant probablement de la taille des chenilles. Bien qu'A. orana n'ait pas produit le plus grand nombre de parasites par chenille, elle a été néanmoins préférée aux autres tortricides comme hôte pour les élevages.
    Notes: Abstract Colpoclypeus florus Walk. is a gregarious ectoparasite of tortricids. Details of biology and annual cycle in orchards in the Netherland are reported. It has four to five generations per year, but only females of the second generation, and some of the third, meet with abundant hosts (Adoxophyes orana F.v.R.) of the proper size. Therefore, the parasite is scarce to virtually absent from Duth orchards except in July and August. A technique for mass-rearing of C. florus is described, producing 6 000 wasps per week. 16 000–28 000 wasps/ha were released in spring, in order to control leafrollers. Although all important leafroller species were parasitized, the rate of parasitization was too low to be effective.

    Description / Table of Contents: Résumé Les réponses de Prostephanus truncatus Horn (Coleoptera: Bostrichidae) à deux constituants de la phéromone d'agrégation de Rhyzopertha dominica Fabricius (Coleoptera: Bostrichidae), le (±)-1-méthylbutyl (E)-2-méthyl-2-penténoate et le (±)-1-méthylbutyl (E)-2,4-diméthyl-2-pentéoate, connus comme “Dominicalure 1” et “Dominicalure 2”, ont été étudiées au laboratoire et au champ. Auslaboratoire ces composés ont été essayés individuellement et en mélange 1:2. Une membrane de caoutchouc imprégnée de phéromone a été utilisée comme émetteur lent, et une courbe de réponse en fonction de la dose a été établie. P. truncatus répondait plus vigoureusement à la Dominicalure 2 seule. Tous ces traitements ont été comparés dans un essai aux champs en Tanzanie, dans lesquels des pièges en carton ondulé contenant des diffuseurs de phéromone étaient placés dans des entrepôts de maïs dans les fermes. La Dominicalure 1 et le mélange de 1 et 2 étaient tous deux efficaces pour piéger R. dominica. La Dominicalure 2 a piégé plus de P. truncatus que les autres traitements et était à peu près aussi efficace que le contrôle visuel pour détecter la présence de cet insecte dans les stocks. On peut envisager que la Dominicalure 2 pourrait servir de base à un programme pour lutter contre P. truncatus en Afrique de l'Est.
    Notes: Abstract Laboratory and field studies have been made of the responses of Prostephanus truncatus (Horn) to two components of the aggregation pheromone of Rhyzopertha dominica (Fabricius) — (±)-1-methylbutyl (E)-2-methyl-2-pentenoate and (±)-1-methylbutyl (E)-2,4-dimethyl-2-pentenoate, known as “Dominicalure 1” and “Dominicalure 2” respectively. In the laboratory, these compounds were tested individually and as a 1:2 mixture; P. truncatus responded most strongly to Dominicalure 2 alone. All three treatments were compared in a field trial in Tanzania to monitor both R. dominica and P. truncatus in farm stores. Dominicalure 1 or a mixture of 1 and 2 were both highly effective for trapping R. dominica. In contrast, Dominicalure 2 trapped more P. truncatus than the other treatments and was about as effective as visual inspection at demonstrating the presence of the beetle in stores. Dominicalure 2 could form the basis of a monitoring programme for P. truncatus in East Africa.

Le taux sexuel de la descendance des femelles isolées est favorable aux femelles (t.s.=0,26) quand elles ont à leur disposition des hôtes de grande taille, tandis qu'avec des hôtes petits le taux sexuel est favorable aux mâles (t.s.=0.73). On n'a pas détecté de mortalité différentielle des sexes. Les pourcentage de mâles obtenu le premier jour de ponte des femelles sur les hôtes de grande taille est significativement différent de la moyenne, indépendamment de l'âge de la femelle à ce moment. Cependant, sur des hôtes petits, bien que restant favorable aux mâles dans l'ensemble, une plus grande proportion de femelles à partir des premiers jours de ponte a pu être observée.
    Notes: Abstract The sex ratio of the progeny of single females parasitizing large hosts favoured the females (sex ratio=0.26); but on small hosts favoured the males (0.73). No differences in mortality of the sexes were detected. The sex ratio was independent of female age when large hosts were used. The percentage of males observed in the progeny of the first day of female oviposition was significantly greater than the mean, irrespective of the age at which female oviposition began. When females were exposed to small hosts, a greater percentage of females was observed in the progeny from the last days of oviposition.

    Notes: Summary Recombinational analysis of oxil mutants was performed using a and a mutant strains with the same mitochondrial and nuclear backgrounds, derived from strain 777-3A. In spite of minor inconsistencies the overall map of oxi1 mutations can be constructed on the basis of wild-type recombinant frequencies in the two-point oxi1 − − x oxi1 − crosses. The frequencies of wild-type recombinants varied in a wide range from 0.003% to 16%, reaching the maximal values expected for unlinked mitochondria) markers. No distinct clusters of mutants were observed. The analysis of translation products of oxil mutants showed that all but one of the oxil mutants studied are connected with the conspicuous changes of the polypeptide band corresponding to subunit 11 of cytochrome c oxidase in electrophoresis on polyacrylamide gels. The exceptional G565 mutant showed no conspicuous change in subunit II, but lacked subunit I of cytochrome c oxidase. Various oxi1 mutants seemed to carry premature chain termination mutations. Most of them show a correlation between the length of the putative fragment of subunit II synthesized and the position on the genetic map. The direction of translation is from the V2 to the V60 mutation. The V2 mutation is proximal to cap and V60 proximal to the par locus.

    Notes: Summary We have developed a system for assaying pyrimidine dimers in the 2 ⇐m DNA plasmid of Saccharomyces cerevisiae, using Micrococcus luteus UV endonuclease to nick dimer-containing plasmid molecules and measuring percentages of nicked and covalently closed circles on agarose gels. UV-irradiation induced dimers in plasmid DNA, in vivo, at the same rate as in chromosomal DNA. After a dose of 20 Joules·m−2, approximately 86% of plasmid molecules had. at least one dimer. After 3 h incubation under normal growth conditions only 4% still retained dimers in a wild-type strain. In a rad1 (excision-defective) mutant 81% of plasmid molecules still had dimers after 3 h, suggesting that excision repair operates to remove dimers from plasmid DNA in wild-type yeast. Dimers can be removed from 2 ,um DNA in a rad1 mutant by photoreactivation.

The results demonstrated a case of dimorphism of L8 protein of 60S ribosomal subunit. Of eight strains examined, two strains were one type and six were the other type. The former, which was tentatively designated as altered (A) type, was more acidic than the latter, common (C) type, as shown by mobility difference in pH gradient gel. Heterozygous (A/C) diploid cells contained both types of L8 protein and gave rise to tetrads of 2:2 segregation for A and C types, indicating that the difference of mobility was reflection of the allelic difference of the gene coding for L8 protein.

    Notes: Summary A spontaneous high-level cycloheximide-resistant mutant of the yeast Saccharomyces cerevisiae (strain cy32) is found to have an altered protein of the large subunit (60S) of cytoplasmic ribosomes, namely protein L29. The resistance character segregates together with this biochemical defect and is semidominant in heterozygous diploids. Judged from in vitro susceptibility to inhibition by cycloheximide there are at least 50% resistant ribosomes present in such diploid strains. From these results it is concluded that cycloheximide resistance of mutant cy32 is caused by mutation of a single gene and that it is the structural gene for L29 which is affected. Preliminary genetic mapping data are also reported. They indicate a location of cyhx-32 marker on chromosome 7 near met13.

This plasmid contains the entire sequence of the endogenous 2 μm yeast DNA plasmid in addition to the yeast nuclear LEU2 + gene and the Co1E1 derivative, pMB9. In the [2 μm+] transformants, a new wholly yeast LEU2 + plasmid, pYX, was generated, probably by a recombination event between pJDB219 and 2 μm DNA. The plamid, pYX, in the absence of 2 μm DNA, was found to exist in equimolar amounts of two forms, A and B, which probably arise by intramolecular recombination across the inverted repeat sequences of the 2 μm DNA portion of the plasmid. pJDB219 was found to require the presence of 2 μm DNA to undergo this intramolecular recombination. The results suggest that 2, μm DNA and pYX code for a gene product required in this recombination event which pJDB219 cannot produce.

    Notes: Abstract Changes in the soil phosphorus and potassium status on three farming systems in the Northeast Polder during 25 years are related to the fertilizer regime over this period. The changes in the nutrient status of the soil are reflected in fertilizer policy. High rates of phosphate dressings markedly increased the phosphate status of the soil and built up a reserve of phosphate. In contrast, the potassium status decreased by about half, because only potatoes in the rotation were dressed with potassium. Methods of fertilization are discussed and suggestions are made with regard to possible improvements.

    Notes: Abstract No extensive investigation on the effect of fertilizers on Amazon cocoa variety (Theobroma cacao L.) has been performed in Nigeria. Therefore eight fertilizer treatments involving nitrogen and phosphorus, replicated six times at four locations across southern Nigeria, were established in 1973. The four N levels (N0, N1, N2, N3) involved were 0, 80, 160 and 240 kg ha−1 y−1, and the two P levels (P0, P1) were 0 and 67 kg ha−1 y−1. Results of the first 5 years of fertilizer application are reported. Response to P was observed at all locations, and the response was statistically significant at 2 of the locations. There was no response to the application of nitrogen. The data suggest, however, that there is only a response to phosphorus when nitrogen is applied.

    Notes: Summary Vitellin was purified from eggs of the silkworm,Bombyx mori, by a new method in which vitellin was extracted from isolated yolk granules. The purified vitellin had a molecular weight of 540,000. An antibody against purified vitellin was prepared in rabbits. It reacted with the hemolymph vitellogenin as well as with purified vitellin, but not with other proteins in the hemolymph or in the extract from yolk granules. The anti-vitellin IgG was used to immunocytochemically locate vitellin in theBombyx non-diapause egg during early developmental stages. In the egg, just after oviposition, vitellin was located in internal yolk granules and in small yolk granules of the periplasm. During the early developmental stages studied, vitellin was not metabolized uniformly throughout the egg. The vitellin of the internal yolk granules located at the posterior-dorsal part and of the small peripheral yolk granules was utilized in 16 h and 2 days, respectively, after oviposition. A thin, very vitellin-poor layer was located between the periplasm and the vitellin-rich interior in the newly laid egg. it was always in close contact with the periphery where blastoderm and germ-band cells developed.

    Notes: Abstract Powerful VHF radars are capable of almost continuously monitoring the threedimensional velocity vector and the distribution of turbulence in the middle atmosphere, i.e. the stratosphere and mesosphere. Methods of radar investigations of the middle atmosphere are outlined and the basic parameters, mean and fluctuating velocities as well as reflectivity and persistency of atmospheric structures, are defined. Results of radar investigations are described which show that the tropopause level as well as a criterion on the stability of the lower stratosphere can be deduced. Besides mean wind velocities, VHF radars can measure instantaneous velocities due to acoustic gravity waves. The interaction of gravity waves with the background wind is discussed, and it is shown that cumulus convection is an effective source of gravity waves in the lower stratosphere. The vertical microstructure of the stratosphere, manifesting itself in thin stratified sheets in which temperature steps occur, is investigated by applying knowledge from investigations of the oceanic thermocline. Possible origins, like shear generation and lateral convection of the microstructure of the stratosphere, are discussed. Observations of gravity waves in the mesosphere are reviewed and their connection with turbulence structures is pointed out. Finally, some open questions which could be answered by further VHF radar investigations are summarized.

    Notes: Abstract We have carried out a series of recordings reproducing the small scale structures of temperature and windspeed within the boundary layer. The results obtained have been processed by computer and a direct comparison made between the slope of the spectral density functions and the theoretical slopen=5/3 defining the turbulence in the region of the inertial subrange. The measurements made at various altitudes on thin atmospheric samples demonstrate the variability of the spectral density slope with altitude. The hypotheses put forward by A. M. Obukhov, R. Bolgiano, G. H. Shur, P. Misme, A. Monin or S. Panchev, for example, enable the spectral variations and certain discontinuities in the turbulence observed during the experiments, within and beyond the boundary layer, to be explained.

    Notes: Abstract Dye plumes were generated at three depths in the seasonal thermocline between 7 and 11 m, 22 km south of Key West on 21 August 1980 and photographed at about 10 second intervals with an underwater camera system. Eleven pairs of consecutive pictures are analyzed to determine the mean current vertical shear and the width of the plumes by positioning reference points relative to the rod attached to the camera system. The relative distances of reference points are calibrated with the stereophotogrammetric method for one pair. The eddy diffusivity is calculated by use of a model of turbulent diffusion developed byTaylor (1921). Its values range from 5 to 25 cm2s−1 for the plume widths ranging from 33 to 132 cm. The Richardson number is calculated for each pair of pictures with the vertical density gradient estimated from temperature profiles. Its values are higher than the critical value of 0.25 except for one case. The diffusivity was higher by orders of magnitude than the molecular one and indicates the presence of turbulence together with billow like features of the plumes in spite of high Richardson numbers. This suggests that the billow turbulence might be caused by effects of surface gravity waves and not by the Kelvin-Helmholtz instability.

    Notes: Abstract We have examined the properties and subcellular localization of phytohemagglutinin (PHA), the major lectin of the common bean (Phaseolus vulgaris.), in the axis cells of nearly mature and imbibed mature seeds. On a protein basis the axis contained about 15% as much PHA as the cotyledons. Localization of PHA was done with an indirect immunolabeling method (rabbit antibodies against PHA, followed by colloidal gold particles coated with goat antibodies against rabbit immunoglobulins) on ultra-thin cryosections which were embedded in plastic on the grids after the immunolabeling procedure. The embedding greatly improved the visualization of the subcellular structures. The small (4 nm) collodial gold particles, localized with the electron microscope, were found exclusively over small vacuoles or protein bodies in all the cell types examined (cortical parenchyma cells, vascular-bundle cells, epidermal cells). The matrix of these vacuoles-protein bodies appears considerably less dense than that of the protein bodies in the cotyledons, but the results confirm that in all parts of the embryo PHA is localized in similar structures.

    Notes: Abstract The ultrastructure of the storage parenchyma cells of the cotyledons of developing bean (Phaseolus vulgaris L.) seeds was examined in ultrathin frozen sections of specimens fixed in a mixture of glutaraldehyde, formaldehyde and acrolein, infused with 1 M sucrose, and sectioned at-80° C. Ultrastructural preservation was excellent and the various subcellular organelles could readily be identified in sections which had been stained with uranyl acetate and embedded in Carbowax and methylcellulose. The cells contained large protein bodies, numerous long endoplasmic reticulum cisternae, mitochondria, dictyosomes, and electron-dense vesicles ranging in size from 0.2 to 1.0 μm. Indirect immunolabelling using rabbit immunoglobulin G against purified phaseolin (7S reserve protein), and ferritin-conjugated goat immunoglobulin G against rabbit immunoglobulin G was used to localize phaseolin. With a concentration of 0.1 mg/ml of anti-phaseolin immunoglobin G, heavy labeling with ferritin particles was observed ober the protein bodies, the cisternae of the endoplasmic reticulum, and the vesicles. The same structures were lightly labeled when the concentration of the primary antigen was 0.02 mg/ml. Ferritin particles were also found over the Golgi bodies. The absence of ferritin particles from other organelles such as mitochondria and from areas of cytoplasm devoid of organelles indicated the specificity of the staining, especially at the lower concentration of anti-phaseolin immunoglobulin G.

    Notes: Summary Electric field-induced fusion was induced between Saccharomyces cerevisiae protoplasts from the ρ − heterozygous diploid strain 2114 and the respiratory-competent diploid strain 3441, carrying chromosomal markers. Close membrane contact between the cells of the two different strains (ratio 1:1) was achieved by dielectrophoresis in a weak inhomogeneous alternating field (about 1 kV/cm, 2 MHz). Due to dielectrophoresis pearl chains of two or more cells of the two strains are formed between the electrodes. Cell fusion was induced by application of two single square field pulses sufficiently high to induce reversible electrical breakdown in the membrane contact zone between cells within a pearl chain (about 7 to 8 kV/cm field strength and 40 Ms duration). The two subsequent pulses were applied at an interval of about 10 s. Hybrids could be isolated on selection medium in a high yield (compared with conventional fusion techniques). The hybrids were diploid, respiratory-competent and produced prototrophic spores. Thus, the fused hybrids contained only the chromosomal markers of strain 2114 and the cytoplasmic marker for respiratory competence from strain 3441; electro-fusion thus resulted mainly in plasmogamy.

    Notes: Summary Prototrophic hybrids have been obtained by the fusion of auxotrophic haploid strains of the two yeasts Yarrowia (Saccharomycopsis) lipolytica and Kluyveromyces lactis. The hybrid fusants had a colonial morphology intermediate between that of the two parent strains, were uninucleate, and contained an approximately diploid amount of DNA per cell. The growth rates of all the fusants on a minimal glucose medium were slower than those of the two parents. Two of the fusants studied could utilise a novel range of carbon sources. All of these data suggested that the hybrids contained a diploid nucleus formed by the fusion of the two haploid parental nuclei. However, analytical CsCl density gradient centrifugation demonstrated that the nuclear DNA of the fusants was derived almost entirely from the Y. lipolytica parent. Moreover, an examination of the protein constitution of the fusants by two-dimensional gel electrophoresis showed that their protein patterns were indistinguishable from that of Y. lipolytica. Two possible mechanisms for the formation of a diploid nucleus containing DNA derived almost entirely from one of the haploid parents are discussed.

    Notes: Summary A hybrid plasmid (pLG4) containing pBR325 and the yeast arg4 gene was constructed then used to isolate DNA fragments of Chlamydomonas able to promote high frequency transformation of yeast. Three plasmids containing EcoRI restriction fragments of chloroplast DNA and two plasmids containing Aval fragments of nuclear DNA were shown to support autonomous replication of plasmids in yeast. The three EcoRI fragments correspond to restriction fragments R4, R5 and R11 of native chloroplast DNA. These fragments are clustered in the physical map of chloroplast DNA constructed by Rochaix (1978). All isolated plasmids were shown to transform yeast at high frequency but the yeast transformants were quite unstable mitotically. Potential cloning sites are still available in the new plasmids which could be used as vectors in yeast and possibly in Chlamydomonas itself.

A manganese mutagenesis on such a strain produced chloramphenicol resistant mutants, most of which resulted from mutations in nuclear genes. These mutants may be either dominant or recessive, and they probably do not code for membrane proteins. The few mitochondrial mutants fall into several classes, but all result from mutations in the 21S rRNA gene. The suppressor allele effectively prevents the appearance of the most common group of mitochondrial mutants (those that map at cap1), and thereby enhances the selection of novel mutants in the region.

    Notes: Summary A genetically defined highly suppressive petite yeast strain (ρ −cob+AsEoCoOoPo) was crossed with a grande strain carrying a multiply marked mitochondrial genome (ρ +ArErCrO rpr). Petite diploid progeny, isolated from individual zygotic clones consisting either of wholly petite or mixtures of grande and petite cells, were characterised genetically by crossing to grande haploids. The diploid petites were found to closely resemble the petite parent and in general not to carry mitochondrial markers from the grande parent. In the petites from the mixed clones recombination was detected, but only within the region of homology between the genomes. These observations are inconsistent with models of suppressiveness based on destructive recombination and suggest that the petite genome eliminates the grande genome from zygotic progeny through being preferentially replicated. The most plausible model to explain the observed pattern of zygotic clones postulates a limited number of mDNA replication sites in zygotes, competition for sites between input mDNA molecules and an advantage in this competition for suppressive ρ − mDNA.

    Notes: Summary Genetic and biochemical studies were performed with mutants allocated to the mitochondrial oxi2 gene. Recombinational analysis of 19 oxi2 mutants was performed using α and a mutant strains derived from the same genetic background. The frequencies of wild-type recombinants in oxi2 − × oxi2 − crosses varied from 0.002 to 17%. The map of oxi2 mutations constructed on the basis of these frequencies shows many internal inconsistencies. In the course of rho − deletion mapping five classes of oxi2 mutations were distinguished. The results of deletion analysis are in agreement with those of recombinational mapping. The analysis of mitochondrial translation products by SDS-polyacrylamide electrophoresis of 20 oxi2 mutants shows that 17 of them are connected with conspicuous changes of 22 kd polypeptide band corresponding to subunit III of cytochrome oxidase. At least four of them carried instead of subunit III clearly visible significantly shorter polypeptides (12.8 to 20.1 kd). These were, most likely, shorter fragments of subunit III resulting from chain termination mutations. Colinearity was observed between the lenght of new polypeptides and the positions of the respective mutations on the recombinational map. These data confirm hat oxi2 encodes subunit III of cytochrome oxidase and suggest that translation of the oxi2 gene is in the direction from V303 to V273.

    Notes: Abstract Genes in mitochondria and chloroplasts segregate rapidly during vegetative reproduction. Models to explain this vegetative segregation invoke either random segregation of organelle DNA molecules, or nonrandom segregation with random recombination events. All such models are basically stochastic. To look at vegetative segregation we took heteroplasmic (HET) cells containing mitochondrial mutations at the cap1, eryl and olil loci from several crosses. HETs were repeatedly selected and subcloned. Even after three to five successive subclonings (approximately 60–100 generations) some cells remained heteroplasmic. This confirms and extends previous observations of persistent HETs by Rank and Bech-Hansen (1972) and Forster and Kleese (1975), and by Bolen et al. (1980) for chloroplast genes in Chlamydomonas.

    Notes: Summary The effect of a previously isolated antisuppressor mutation from bakers' yeast, that reduced the efficiency of the tyrosine-inserting ochre suppressor, SUP7-o, on other tyrosine-inserting ochre suppressors has been determined. As expected, the antisuppressor mutation, mod5-1, restricted the capacity of all eight tyrosine-inserting ochre suppressors to suppress nonsense mutations. Based on the suppression of five ochre alleles in the presence of mod5, the eight class I suppressors can be grouped into three subclasses. The most efficient subclass had only one member, SUP4-o. Members of the second group included SUP2-o, SUP3-o, SUP7-o, and SUP8-o. The third and least efficient subclass included SUP5-o, SUP6-o, and SUP1 1-o. These differences in efficiencies are a function of the relative expression of the eight genes encoding tRNATYR.

    Notes: Summary The genetic map of the nuclear genome of the fission yeast Schizosaccharomyces pombe has been extended by mitotic and meiotic mapping data. A total of 158 markers are now assigned to the three linkage groups known in this organism, and 118 of them have been located on the corresponding chromosome map. Chromosome II and III each consist of one linkage group. There is some indication that the two large fragments which define chromosome I are meiotically linked, but the linkage observed is significant at the P = 0.05 level only. The length of the map is at least 1,700 map units, corresponding to an average of about 8 kilobases per map unit. The latter figure is comparable to the one obtained for intragenic recombination in the sup3 gene (Hofer et al. 1979). The basic frequency of gene conversion as measured for 21 genes varies according to a distribution of Poisson (with a modal value of 0.6% conversion per meiosis and per gene), in sharp contrast with Saccharomyces cerevisiae (Fogel et al. 1980) and Ascobolus immersus (Nicolas 1979). This may reflect the rarity of gene or region-specific rec alleles in S. pombe and may be related to the homothallism of this organism.

    Notes: Summary Saccharomyces cerevisiae is normally sensitive to the drug hygromycin B; a hygromycin B concentration of 200 µg/ml in agar plates is sufficient to completely inhibit growth. We constructed yeast-E. coli bifunctional plasmids which confer hygromycin B resistance to Saccharomyces cerevisiae. Promoters and amino terminal coding regions of a heat shock gene, a heat shock cognate gene, and the phosphoglycerate kinase gene from yeast were fused to a bacterial hygromycin B resistance gene. In all three cases, yeast cells containing plasmids with the hybrid hygromycin B resistance gene were resistant to high levels of the drug. Yeast cells containing these plasmids can also be directly selected after transformation by using hygromycin B. The intact bacterial hygromycin B resistance gene and the kanamycin resistance gene from Tn903 were also tested in yeast for their ability to confer resistance to hygromycin B and G418. The intact bacterial genes were not effective in conferring drug resistance to yeast cells.

    Notes: Summary The RAD52 gene of Saccharomyces cerevisiae has previously been shown to be involved in both recombination and DNA repair. Here we report on the cloning of this gene. A plasmid containing a 5.9 kb yeast DNA fragment inserted into the BamH1 site of the YEp13 vector has been isolated and shown to complement the X-ray sensitive phenotype of the rad52-1 mutation. The rad52-1 cells containing the plasmid form larger colonies than similar cells having lost the plasmid. This plasmid has been shown not to complement either the U.V. sensitivity or the recombination defect of the E. coli recA mutation. From the insert various fragments have been subcloned into the YRp7 and YIp5 vectors. Integration events of two of the subclones have been genetically mapped to the chromosomal location of RAD52, indicating that the structural gene has been cloned. A 1.97 kb BamH1 fragment subcloned into YRp7 in one orientation complements the rad52-1 mutation, while the same fragment in the opposite orientation fails to complement. Various other subclones indicate that a BglII site, within the BamH1 fragment, is in the RAD52 gene. This BglII site has been deleted by Sl-nuclease digestion and the resulting deletion inactivates the RAD52 gene. BAL31 deletions from one end of a 1.9 kb Sal1-BamH1 fragment have been isolated; up to 0.9 kb can be deleted without loss of RAD52 activity, indicating that the RAD52 gene is approximately 1 kb or less in length.

Plasmid-encoded DNA polymerase I was synthesised at 2 to 3 times the wild-type E. coli level, and was biochemically indistinguishable from chromosomally-encoded protein. It was able to counteract the radio sensitivity of polA1, polAex1, polAex2 and polA12 mutants, but no complementation of polA107 mutants occurred, even though the plasmid polA+ gene was expressed. S. cerevisiae ars-1 or 2 μ replicative sequences were introduced into the polA+ plasmid. Transformation of yeast with these constructs increased total DNA polymerase levels 2–20 times, depending upon assay conditions. The additional activity was discriminated from yeast DNA polymerases by its ability to use low concentrations of substrate, by its resistance to chemical inhibition, and by co-electrophoresis with pure DNA polymerase I and its proteolytic fragments. The polA+ gene was expressed in yeast without the aid of yeast promotor sequences. However, deletion of cloned DNA more than 99 base pairs in front of the structural gene prevented expression in yeast but not in E. coli, indicating that the two organisms use different sequences for expression of the plasmid polA+ gene.

    Notes: Abstract The budding process of the yeast form of Mucor rouxii was examined by electron microscopy of thin sections with particular reference to wall ontogeny. In most instances the bud wall is seen as a continuation of the inner layers of the parent cell wall. As the bud emerges it ruptures the outer layers of the parent wall. The bud wall is much thinner than the parent wall and remains so while the bud grows into a sphere of about one half the diameter of the parent cell. Then a septum begins to form centripetally, at the neck, by invagination of the plasmalemma. Before the neck canal is completely occuluded, electron-dense wall material is deposited into the septum space. Two separate septum walls are deposited, one on the parent side and one on the bud side of the invaginating plasmalemma. Septum wall formation extends to the surrounding neck walls. In this manner, the parent and bud cytoplasms become fully separated and each is surrounded by a continuous wall. The two cells remain attached to each other by the original neck wall; eventually, the bud abscisses leaving a birth scar on the bud cell and a more pronounced bud scar on the parent cell. In general, the mechanism of budding in this zygomycetous fungus resembles that of an ordinary ascomycetous yeast such as Saccharomyces cerevisiae.

    Notes: Abstract The combination of high-resolution autoradiography and biochemical methods has been used to ascertain the site of mannan synthesis in the yeastSaccharomyces cerevisiae. High-resolution autoradiography has been performed under conditions when addedd-mannose-3H was incorporated exclusively into mannan. Application of “pulse-chase” labelling technique revealed that the radio-active mannose is fixed primarily in the cytoplasmic space from where it is transported into the cell wall. Additional experiments with separated membrane fractions from the same yeast strongly support the hypothesis that the plasmalemma is not directly involved in the biosynthesis of yeast mannan and that the membranes of the endoplasmic reticulum are the sites where the polymerization of mannosyl units takes place.

    Notes: Summary Plasmids that complement the yeast mutations rad50-1, rad51-1, rad54-3 and rad55-3 were obtained by transforming strains that carried a leu2 marker and the particular rad mutation, with YEp13 plasmids containing near random yeast DNA inserts. Integration of these plasmids or of fragments of these plasmids was accomplished. Genetic studies using the integrants established the presence of the genes RAD51, RAD54 and RAD55 in the respective plasmids. However, a BamHI subclone of the rad50-1 complementing plasmid failed to integrate at the RAD50 locus, indicating that no homology exists between this fragment and the RAD50 gene. A BamHI fragment from the RAD54 plasmid was shown to be internal to the RAD54 gene: its integration within a wild type copy of RAD54 causes the cell to become Rad−; its excision is X-ray inducible and restores the Rad+ phenotype. Since cells bearing a disrupted copy of RAD54 are able to survive, we conclude that this gene is not essential.

    Notes: Summary One mutant resistant to carbonylcyanide m-chlorophenylhydrazone (CCCP), an uncoupler of oxidative phosphorylation, was isolated in Saccharomyces cerevisiae. Genetic analysis showed that a single nuclear gene is responsible for increased resistance; this gene was dominant. The mutant showed cross-resistance or collateral sensitivity to several chemically-unrelated inhibitors (cycloheximide, dinitrophenol, tributhyltin chloride, chloramphenicol). The resistance of the mutant is related to a decreased uptake of CCCP which is not expressed in glucose-starved cells. It was shown that glucose induced a CCCP efflux which was more efficient in the mutant than in the wild-type cells. This effect was correlated to a greater acidification of the internal pH by glucose addition in the mutant cells. It was proposed that resistance was not due to a change of permeability of the plasmic membrane itself but to the change of internal pH which determines the extent of accumulation of weak acids or bases.

We now report that a non-Mendelian element, [eta+], causes this inviability. In [eta−] strains the suppressors are not inviable. The [eta+] genetic element segregates to about 70% of the meiotic progeny, although almost all of the spores probably have the [eta+] phenotype for the first few divisions. Growth on 5 mM guanidine hydrochloride efficiently causes [eta+] strains to become [eta−]. The [eta+] factor has many similarities with the previously described [psi+] factor (Cox 1965, 1971). However, [eta+] and [psi+] differ in their patterns of inheritance, and by the fact that [psi+] affects ochre specific and not omnipotent suppressors, while the converse is true of [eta+].

    Notes: Summary Several mit mutants mapping within or near the var1 determinant region have been characterized genetically and biochemically. These mutants were isolated using a new enrichment protocol which simplifies the isolation and identification of rare respiration-deficient mutants of yeast. Two of the mutants, PZ200L and PZ206, map in genome segments which flank the known varl gene reading frame; nevertheless, both belong to the same complementation group, apparently that of the varl gene. A third mutant, PZ200R is closely linked to one of the varl allelic determinants now known to be a short insertion within the gene. All three var1 mutants exhibit decreased levels of mitochondrial protein synthesis and negligible activity of the respiratory enzyme complexes. Another cluster of mutants belonging to a separate complementation group from that defined by PZ200L and PZ206 was also mapped and it contains mutants in the nearby serine tRNA gene. The isolation of these mutants in the varl region shows that the varl locus contains information essential for the maintenance of respiration-competent mitochondria. Because these mutants affect mitochondrial protein synthesis, their existence supports the previous hypothesis that the varl protein is an integral component of mitochondrial ribosomes. Furthermore, the mutant sites are present in a DNA sequence that is highly, rich in A+T residues that also contains a gene. Since approximately 50% of the yeast mitochondrial genome is similarly rich in A+T and since most of those regions have not yet been sequenced it is quite possible that other A+T-rich genes may exist.

    Notes: Summary Contrary to what happens in bacteria, mutations induced by nitrosoguanidine in yeast are not accompanied by an excess of mutations in nearby genes. We have investigated nitrosoguanidine mutagenesis in three regions of the yeast genome: the contiguous DNA segments HIS4A, HIS4B and HIS4C, located on chromosome III; ADE1 and CDC15 separated by about 3 map units on chromosome I; and CAN1, some 50 map units away from the centromere on chromosome V. Revertants at HIS4C never suffered mutations at HIS4A or HIS4B. Reversion at CDC15 did not affect the frequency of mutation at ADE1. No tsm mutations, leading to thermonsensitivity, were found in the immediate vicinity of the locus CAN1 after selecting for canavanine resistant mutants. However, as expected from nitrosoguanidine mutagenesis of replication points and the fixed pattern of chromosome replication, the induced tsm mutations seem not to map randomly over the yeast genome; in fact, two out of the three groups of such tsm mutations studied are located in the same chromosome arm as CAN1, indicating that these two regions are replicated at the same time as CAN1. Replication synchrony is less than perfect, since the tsm mutations of each group affect many different genes.

    Notes: Summary We have found that the application of the amino acid analog canavanine to a culture of yeast cells transformed with chimeric plasmids based on the yeast 2-µm DNA plasmid increases the percentage of cells which have lost the transforming plasmid. This effect is found whether the plasmid carries the CAN1 sensitive allele and the yeast strain carries a can1 mutation confering resistance, or the plasmid contains no CAN1 allele and the yeast strain carries the wild-type CAN1 sensitive allele. Canavanine exerts this effect on yeast strains transformed with chimeric plasmids containing either a portion or the entire 2-µm DNA plasmid, yet canavanine does not appear to effect the maintenance of the native 2-µm DNA plasmid complement within the cell. The effect of canavanine on strains transformed with chimeric plasmids is the same whether or not the yeast strain also contains native 2-µm plasmid DNA. Neither the amino acid analog ethionine, the protein synthesis inhibitor cycloheximide, nor the DNA replication inhibitor hydroxyurea exhibit this effect. Some of the experimental results suggest that canavanine may be a curing agent rather than an agent which selects for spontaneous plasmid loss.

    Notes: Summary The effects of polyamines (spermidine and putrescine) on yeast suppressor tRNA-mediated readthrough of amber and UGA termination codons, in a homologous cell-free system, was examined. The efficiency of readthrough in a [psi+] lysate, mediated by exogenous suppressor tRNA, was significantly increased by polyamines as was the efficiency of endogenous UGA readthrough. The addition of polyamines, in the absence of exogenous suppressor tRNA, did not induce amber or ochre readthrough, nor could polyamines restore efficient termination readthrough in [psi−] lysates. It is concluded that polyamines interact with tRNA to increase the strength and specificity of the codon: anticodon interaction.

From 9.2 kilobase pair BamHI DNA fragment this gene has been localized to a 1.76 kb fragment, the restriction map of which appears different from those reported for the E. coli and the mouse dihydrofolate reductase genes. The enzyme encoded by the chimeric plasmid was established as yeast dihydrofolate reductase by its sensitivity to antifolates in vivo through growth studies and in vitro by enzyme assay. Since, the expression of this gene occurs independent of its orientation within the chimeric plasmid, the 1.76 kb fragment may contain functional regulatory sequences in addition to the structural sequences for yeast dihydrofolate reductase.

    Notes: Summary A three-factor cross of Saccharomyces cerevisiae involving the cap1, ery1, and oli1 loci was done, with partial pedigree analyses of 117 zygotes. First, second, and third buds were removed and the genotypes of their diploid progeny determined, along with those of the residual zygote mother cell. Results were analyzed in terms of frequencies of individual alleles and of recombinant genotypes in the dividing cells. There is a gradual increase in the frequency of homoplasmic cells and in gene frequency variance during these three generations, as would result from stochastic partitioning of mtDNA molecules between mother and bud, probably coupled with random drift of gene frequencies in interphase cells. These phenomena are more pronounced for buds than for mothers, suggesting that buds receive a smaller sample of molecules. End buds are more likely to be homoplasmic and have a lower frequency of recombinant genotypes than do central buds; an end bud is particularly enriched in alleles contributed by the parent that formed that end of the zygote. Zygotes with first central buds produce clones with a higher recombination frequency than do those with first end buds. These results confirm previous studies and suggest that mixing of parental genotypes occurs first in the center of the zygote. If segregation were strictly random, the number of segregating units would have to be much smaller than the number of mtDNA molecules in the zygote. On the other hand, there is no evidence for a region of the molecule (“attachment point”) which segregates deterministically.

    Notes: Summary We have isolated 150 benomyl resistant mutants of the fission yeast Schizosaccharomyces pombe. Seven of these mutants were found to be cold sensitive for mitosis. These mutants were the subject of physiological, cytological and genetical characterisation. Growth and division of the seven mutants were similar to the wild type strain at 35 °C. After shift from the permissive (35 °C) to the restrictive temperature (20 °C) the mutants became blocked in mitosis whilst cellular growth continued. Consequently, elongate cells were formed. Six of the seven benomyl resistant mutants became blocked in mitosis at 20 °C with a single aberrant nucleus. In every case the benomyl resistant and cold sensitive phenotype was due to a mutation in a single nuclear gene. These mutants were found to comprise a single genetic linkage group (ben4) and were unlinked to existing TBZ/MBC resistant mutants of S. pombe. Whilst no cross resistance was found in our mutants to TBZ, six of the seven mutants were super sensitive to the spindle poison CIPC. We believe that the phenotype exhibited by these mutants is consistent with a defective tubulin subunit.
