    Description: Determining national carbon stocks is essential in the framework of ongoing climate change mitigation actions. Presently, assessment of carbon stocks in the context of greenhouse gas (GHG)-reporting on a nation-by-nation basis focuses on the terrestrial realm, i.e., carbon held in living plant biomass and soils, and on potential changes in these stocks in response to anthropogenic activities. However, while the ocean and underlying sediments store substantial quantities of carbon, this pool is presently not considered in the context of national inventories. The ongoing disturbances to both terrestrial and marine ecosystems as a consequence of food production, pollution, climate change and other factors, as well as alteration of linkages and C-exchange between continental and oceanic realms, highlight the need for a better understanding of the quantity and vulnerability of carbon stocks in both systems. We present a preliminary comparison of the stocks of organic carbon held in continental margin sediments within the Exclusive Economic Zone of maritime nations with those in their soils. Our study focuses on Namibia, where there is a wealth of marine sediment data, and draws comparisons with sediment data from two other countries with different characteristics, which are Pakistan and the United Kingdom. Results indicate that marine sediment carbon stocks in maritime nations can be similar in magnitude to those of soils. Therefore, if human activities in these areas are managed, carbon stocks in the oceanic realm—particularly over continental margins—could be considered as part of national GHG inventories. This study shows that marine sediment organic carbon stocks can be equal in size or exceed terrestrial carbon stocks of maritime nations. This provides motivation both for improved assessment of sedimentary carbon inventories and for reevaluation of the way that carbon stocks are assessed and valued. The latter carries potential implications for the management of human activities on coastal environments and for their GHG inventories.

    Description: Long-term measurements of ecological effects of warming are often not statistically significant because of annual variability or signal noise. These are reduced in indicators that filter or reduce the noise around the signal and allow effects of climate warming to emerge. In this way, certain indicators act as medium pass filters integrating the signal over years-to-decades. In the Alaskan Arctic, the 25-year record of warming of air temperature revealed no significant trend, yet environmental and ecological changes prove that warming is affecting the ecosystem. The useful indicators are deep permafrost temperatures, vegetation and shrub biomass, satellite measures of canopy reflectance (NDVI), and chemical measures of soil weathering. In contrast, the 18-year record in the Greenland Arctic revealed an extremely high summer air-warming of 1.3°C/decade; the cover of some plant species increased while the cover of others decreased. Useful indicators of change are NDVI and the active layer thickness.

    Notes: Abstract The response of male European corn borer, Ostrinia nubilalis (Hübner) to synthetic pheromone lures containing various isomeric blends of the sex pheromone 11-tetradecenyl acetate was measured in 13 counties in North Carolina. The blends consisted of either 3% Z (‘E strain’), 97% Z (‘Z strain’), or 35% Z (‘hybrid’) 11-tetradecenyl acetate. Response to E strain lures predominated in those counties located in the Coastal Plain (east) of the state, while response to the Z strain pheromone was dominant in the west. A zone of overlap of these broad strain distributions appears to occur in the eastern Piedmont. Within this zone there was substantial response to both E and Z blends. The proportion of these responses changed considerably between generations within years as well as between years. Significantly higher capture rates in hybrid baited traps in parts of the overlap zone may be indicative of increased rates of hybridization between the E and Z strains.

    Notes: Abstract The diel flight periodicity of the nocturnal moth Chilo partellus (Swinhoe) (Lepidoptera;Pyralidae) was measured in the laboratory using an actograph and in the field with suction traps. Females showed almost no flight activity on the night of eclosion. Flight activity of mated females peaked before midnight, the period of peak oviposition activity. Male peak activity occurred after midnight coinciding with female eclosion. Presence or absence of females did not affect when or how long males were active. Data on flight activity and reproductive behaviour are discussed in relation to the use of pheromones to protect maize.

    Notes: Abstract This study examined the impact of irrigation water on certain aspects of an insect-plant relationship in the field including the assessment of plant-mediated water effects on an herbivore's development, survival, and behavior, and plant damage parameters and host tissue water status. Maize (Zea mays L.) plants were arranged in a randomized complete block design in the field over two years in North Carolina (NC). Four blocks were subjected to three different irrigation treatments initiated ca. one week before anthesis: optimal, intermediate, deficit water supply. Each plant was infested with one (1986) or two (1987) black head stage, E-race European corn borer [Ostrinia nubilalis (Hübn.)] (ECB) egg masses at tasselling. ECB development, tunnelling site, and survival as well as plant tissue water status (tissue % water contents [θ] & leaf water potentials [Ψ]) were recorded through July. The irrigation effect on ECB parameters was slight and variable. Internal stalk temperatures of optimal plants were consistently cooler than their deficit counterparts (1 day-degree/day). With degree-days included as an explanatory variable in the analyses, there were no significant irrigation effects on the ECB parameters, except for total proportion of ECB's bored into maize plant parts. More ECB's bored into drier plants than in optimal plants; however, this trend was not significant in 1987. Plant water indices showed that though Ψ responded to irrigation, there were only minor changes in tissue θ, particularly in view of the larger diurnal tissue changes observed and the relatively high, sustained stalk θ levels seen over all treatments. Examination of ECB pupal θ confirmed that dietary water changes were minor or non-limiting to the insects' developmental physiology, because pupal θ was not sensitive to the irrigation treatments. Though water supply changes have drastic developmental and agronomic consequences for the maize plant, little or no changes were seen in the ECB feeding environment. Furthermore, a plant damage model was developed whereby the total % of ECB's tunnelled into maize was related to the mean larval age. The implications of this model on the understanding of ECB tunnelling behavior, damage potential, and pest management is noted.

    Notes: Abstract The relationship between second generation European corn borer (Ostrinia nubilalis Hübner) egg mass numbers and subsequent field corn damage, as measured by stalk cavity numbers, was studied in 79 fields in northeastern North Carolina over three years. A mean of 0.028 egg masses per plant (645 egg masses/23400 plants) was found over the course of the study. Significant differences in oviposition rate were detected between fields and years. Ca. 85% of egg masses were deposited in a five leaf zone surrounding the primary ear; of these, 89% were found on the lower four leaves in this zone. Egg masses appeared to be distributed randomly within fields but at low rates of incidence, and oviposition was relatively uniform between sampling areas within individual fields. Under moderate to high oviposition pressure (mean number of egg masses per plant over the duration of the oviposition period 〉ca. 0.02), eggs laid during the early phases of the oviposition period account for more subsequent stalk damage than eggs laid during the later phases of the oviposition period. Variations in second generation egg mass numbers accounted for ca. 70% of variation in stalk cavity numbers.

    Notes: Abstract The management variables which primarily affect phytoplankton biomass (as chl-a) in Lake Mjøsa, Norway, are total phosphorus loading (TP) and the timing and volume of water through flow (by active storage reservoirs). The response of the lake to changes in these factors is studied using a simulation model of the lake ecosystem. Chl-a responses from both observed data and the simulated results are extracted by multiple regression. Results show that decreasing TP load decreases chl-a, but less at low TP levels (〈 10 mg TP · m−3). There is also a certain time period for peak river flow which gives the least yield of chl-a per unit TP. This time period occurs in early summer (i.e., around June 10) if the total phosphorus load is low, and later if the load is high. Both observations and simulation results show that a high water flow increases chl-a at low epilimnion depths (〈 15 m), but that the same high water flow decreases chl-a when epilmnion is deep.

    Notes: Summary Previous reports suggested that the major cytosolic aldehyde dehydrogenase (ALDH1) was present in fetal and infant livers, but the major mitochondrial isozyme (ALDH2) was absent or severely diminished. Re-examination by means of starch gel electrophoresis followed by enzyme activity staining, and by means of dot blot immuno-hybridization of liver samples with known genotypes of theALDH 2 locus, indicated that bothALDH 1 andALDH 2 genes are expressed in fetal and infant livers. In addition, ALDH4 isozyme was also observed. The results imply that a fetus with the ‘usual’ homozygousALDH 2 1 /ALDH 2 1 genotype, but not one with the atypicalALDH 2 1 /ALDH 2 2 orALDH 2 2 /ALDH 2 2 , is capable of detoxifying acetaldehyde transferred from the mother.

    Notes: Summary An ectomycorrhiza, a specialized root organ, is the result of a complex interaction leading to a finely-tuned symbiosis between a plant and a compatible ectomycorrhizal fungus. Ultrastructural observations combined with cytochemical and biochemical studies reveal that structural and metabolic changes in the symbiont cells lead to the final phenotype of the active ectomycorrhiza. In the present review these changes are interpreted as changes in gene expression and discussed within the context of ectomycorrhiza development. Recent genetic data indicate that the continued vegetative growth of the ectomycorrhizal hyphae and the root tissues, and their ability to switch to symbiotic organ formation, is basically controlled by developmentally critical genes. The activity of these ‘symbiotic genes’ during the differentiation of ectomycorrhizas is associated with extensive changes in the concentration of particular polypeptides and protein biosynthesis. The present state of knowledge about the developmental biology of ectomycorrhizas allows only speculation about the events during their development.

    Notes: Abstract The objective of this study is to discuss the potential impact of a global warming on various aspects of human health. Changes in heat-related mortality are estimated for four countries: the United States, Canada, the People's Republic of China and Egypt. In addition, the potential confounding impact of increased air pollution is considered. Finally, a framework to analyze two vector-borne diseases, onchocerciasis and malaria, which may spread if temperatures increase, is discussed. Our findings suggest that heat-related mortality is estimated to rise significantly in all four countries if the earth warms, with the greatest impacts in China and Egypt. The most sensitive areas are those with intense but irregular heat waves. In the United States, air pollution does not appear to impact daily mortality significantly when severe weather is present, although it seems to have a slight influence when weather conditions are not stressful.

    Notes: Abstract The inherited β-hemoglobinopathies (sickle cell disease and β thalassemia) are the result of a mutation in the adult (β) globin gene. The fetal globin chain, encoded by the γ globin genes, can substitute for the mutated or defective β globin chain, but expression of the γ globin gene is developmentally inactivated prior to birth. Reinducing expression of the normal fetal globin genes is a preferred method of ameliorating sickle cell disease and the β thalassemias. Stimulation of as little as 4–8% fetal globin synthesis in the bone marrow can produce 〉20% fetal hemoglobin in the peripheral circulation, due to enhanced survival of red blood cells containing both sickle and fetal hemoglobin, compared to those containing sickle hemoglobin alone. Butyric acid and butyrate derivatives are generally safe compounds which induce fetal hemoglobin production by stimulating the promoter of the fetal globin genes. An initial trial with the parent compound, delivered as Arginine Butyrate, has demonstrated rapid stimulation of fetal globin expression to levels that have been shown to ameliorate these conditions. Phase 1 trials of an oral butyrate derivative with a long plasma half-life have just begun. These agents now provide a specific new apporach for ameliorating these classic molecular disorders and merit further investigation in larger patient populations.

    Notes: Abstract We compare results of a new model for predicting the short term inter annual changes in chlorophyll-a (chl-a) in lakes after reductions in total phosphorus (TP) to predictions made by least squares regression models. In the new method, slopes of chl-a/TP graphs (both axes in mg · m−3) are depicted in frequency diagrams and used to extract information on the expected, short term chl-a/TP response. The short term response for nine shallow (〈 10 m deep) and nutrient rich lakes to changes in TP was found to be: Chl-a = 0.49 · TP + 17.3, and for nine deep, P-limited lakes: Chl-a = 0.08 · TP + 3.5. If the TP-reduction is known to be greater than 10 mg · m−3, the expected slope increases to 0.58 for shallow lakes and to 0.26 for deep lakes. The slope, 0.58, is 8% lower than the slope for the long term response calculated by regression for the shallow lakes. For deep lakes the slope, 0.26, is 2 to 3 times higher than that calculated by regression, indicating that reductions in TP for deep lakes give greater effects than least squares regression equations suggest. We have also calculated the reduction in TP which will give about 80% probability that a reduction in chl-a will be observed next year. For shallow, P-limited lakes this reduction is about 30 mg · m−3 (5% of average initial in-lake TP concentration), and for deep lakes about 14 mg · m−3 (35% of average initial in-lake TP concentration).

    Notes: Abstract Depth variable vertical eddy diffusion coefficients for heat (K z) were calculated from continuously measured temperature profiles in Überlinger See (western part of Lake Constance). The temperatures were averaged over vertical intervals of 10 m yielding 14 discrete values (maximum depth of Überlinger See: 147 m). A linear fit from 10 June to 29 September 1987 was used to smooth the significant temperature fluctuations caused by internal seiches of Lake Constance. Assuming horizontal homogeneity for the smoothed data the Gradient-Flux-Method was applied to compute vertical diffusion coefficientsK z at different depths using the depth variable volumes and surfaces of the 14 layers. The resulting mean diffusion coefficients for the period from June to September are 0.04 cm2/s near the thermocline and up to 0.8 cm2/s in deeper strata (accuracy: ± 50%). It is shown that horizontal mixing between Überlinger See and Obersee (main lake) alters the computation ofK z by less than 50%. A relationship betweenK z and stability (Brunt-Väisälä) frequencyN is found which corresponds well to the theory of internal wave induced turbulence. Combining the diffusion coefficients with measured phosphorus profiles, a phosphorus flux from the hypolimnion to the epilimnion of (0.7 ± 0.4) mg P m−2 d−1 was calculated, corresponding to about 20% of the average external loading per area of Lake Constance in 1986.

    Notes: Abstract Active oxygen, produced by cultured cells following stimulation with various growth factors, seems to be involved in signal transduction leading to cellular responses such as gene expression and growth modulation. In the present study, the intracellular oxidation state was measured in immortalized human endothelial cells (ECV304) after treatment with tumor necrosis factor (TNF)α, using a fluorescent dye and a laser-scanning confocal microscope. The intracellular oxidation state was increased 60 min after the addition of TNFα, and this increase was abolished by a radical scavenger, N-acetylcysteine (NAC), which is also a precursor of glutathione, and by pyrrolidine dithiocarbamate (PDTC). TNFα increased the steady state level of urokinase-type plasminogen activator (uPA), and NAC inhibited this increase at a dose that also inhibited the increase in the intracellular oxidation state. PDTC, on the other hand, did not affect the induction of the uPA gene by TNFα. These results suggest that intracellular glutathione level rather than the oxidation state is necessary for the induction of the uPA gene by TNFα.

    Notes: Abstract Tumor stroma is a specialized form of tissue that is associated with epithelial neoplasms. Recent evidence indicates that significant changes in proteoglycan content occur in the tumor stroma and that these alterations could support tumor progression and invasion as well as tumor growth. Our main hypothesis is that the generation of tumor stroma is under direct control of the neoplastic cells and that, via a feedback loop, altered proteoglycan gene expression would influence the behavior of tumor cells. In this review, we will focus primarily on the work from our laboratory related to the altered expression of chondroitin sulfate proteoglycan and its role in tumor development and progression. The connective tissue stroma of human colon cancer is enriched in chondroitin sulfate and the stromal cell elements, primarily colon fibroblasts and smooth muscle cells, are responsible for this biosynthetic increase. These changes can be reproduced in vitro by using either tumor metabolites or co-cultures of human colon carcinoma cells and colon mesenchymal cells. The levels of decorin, a leucine-rich proteoglycan involved in the regulation of matrix assembly and cell proliferation, are markedly elevated in the stroma of colon carcinoma. These changes correlate with a marked increase in decorin mRNA levels and a concurrent hypomethylation of decorin gene, a DNA alteration associated with enhanced gene expression. Elucidation of decorin gene structure has revealed an unexpected degree of complexity in the 5′ untranslated region of the gene with two leader exons that are alternatively spliced to the second coding exon. Furthermore, a transforming growth factor beta (TGF-β)-negative element is present in the promoter region of decorin gene. This regulatory domain is likely to be implicated in the silencing of decorin gene by TGF-β and may contribute to the regulation of this matrix gene in the tumor stroma.

Sur les 3 plantes, les mâles se développent en moyenne 1,2 j plus vite que les femelles. Les relations entre vitesse de développement et température ont été déterminées en utilisant à la fois un modèle linéaire et le modèle biophysique à 2 paramètres de Sharpe & DeMichele (1977). Les températures plus basses donnent des adultes des 2 sexes plus gros et plus lourds. Moins de G. nigrifrons se sont développés sur la graminée vivace que sur les 2 graminées annuelles à la température la plus élevée (30°C), tandis qu'à la température la plus basse (18°C) moins de cicadelles se sont développées sur les graminées annuelles. La température semble jouer un rôle significatif en déterminant l'adéquation des plantes comme hôtes convenant au développement de G. nigrifrons. Le potentiel de ponte de cette cicadelle avait été sous-estimé par les étudies précédentes.
    Notes: Abstract Population dynamics of the blackfaced leafhopper, Graminella nigrifrons (Forbes) (Homoptera: Cicadellidae), was studied at five temperatures (18, 21, 24, 27, & 30°C) in the laboratory on seedling maize (Zea mays L.), oats (Avena sativa L.), and the perennial johnsongrass (Sorghum halepense (L.) Pers.). Effects of temperature and host plant on egg to adult mean development time, adult size and weight, and fecundity were determined. Leafhoppers on all three hosts developed fastest at the highest temperature tested (21.3 days), and slowest at the lowest temperature tested (73.2 days). The duration from first to last adult eclosion was shortest at 30°C, (11.5 days) and longest at 18°C (43 days). The sex ratio of males to females did not differ from 1:1, but males developed an average of 1.2 days faster than females on all three hosts. Mean percent development/day ranged from 1.4% at 18°C to 4.7% at 30°C. The relationship of this development rate and temperature was determined using both a linear model and a variable parameter biophysical model. Based on these models, the developmental threshold is estimated at 12–15°C. The lowest temperature yielded larger and heavier adults (312 μg, dry weight) than did the highest temperature (225 μg). Fewer leafhoppers developed on the perennial than the annuals at 30°C and fewer on the annuals than the perennial at 18°C. Our results suggest that early in the season johnsongrass and perhaps other perennials are the superior developmental hosts for this leafhopper, whereas in midsummer when temperatures are highest, annuals are the better hosts.

    Notes: Abstract Oviposition by the African migratory locust, Locusta migratoria migratorioides (Orthoptera: Acrididae), was studied in maize and wheat crops on the Orange Free State Highveld. Maize was shown to be the most important oviposition habitat with peak laying taking place in autumn and early winter when highest pod densities were recorded. Laying was mainly concentrated along the middle of the crop interrows in maize and within clearings in the wheat crop. Despite the uniform layout of these crops, the distribution of egg pods was found to be aggregated. Non-reproductive behaviour, such as locust aggregation, basking and feeding, as well as environmental factors appeared to influence the distribution of egg pods in these crops. Secondary selection for optinum soil moisture and compaction on the laying site enhanced the aggregation of pods.

    Notes: Abstract The toxicity and anticholinesterase activity of tremorgenic fungal metabolites, territrems, to the corn earworm, Helioverpa zea (Boddie) (Lepidoptera, Noctuidae) were examined. In oral toxicity studies, territrem A significantly inhibited growth by 40% at 25 ppm and by 89% at 250 ppm. Territrem B and an epoxy-derivative significantly inhibited growth by ca. 45% at 250 ppm. Piperonyl butoxide administered orally synergized the toxicity of the territrems tested. In topical toxicity studies, the epoxy-derivative caused 26% mortality and 25% growth retardation at 10 mg/gm insects. Territrem A and B were not significantly lethal, but did reduce growth by 15–20% at 10 mg/gm insect. Paraoxon tested in the same way caused 100% mortality at 25 ppm orally and 10 mg/gm topically. However, all territrems tested in vitro as inhibitors of H. zea head acetylcholinesterase were at least comparable to or were more active than paraoxon. Topically administered epoxy-territrem B also inhibited H. zea head acetylcholinesterase. The potential for development of new insecticides from a territrem lead structure is discussed.

    Notes: Abstract Multi-step procedures for the determination of phosphorus, sulfur and chlorine are described and tested against established methods and on reference materials. Phosphorus is separated as hydrogen phosphide, extracted as phosphomolybdic acid, reduced to molydenum blue and measured photometrically (detection limit 0.05 μg/g). Sulfur is separated after reduction as hydrogen sulfide or by means of pyrohydrolysis and measured by ICP-OES (detection limit 0.1 μg/g). Chloride can be measured by ion chromatography after pyrohydrolytic separation (detection limit 0.1 μg/g). The determination of sulfur was also successfully tested on copper and steel samples.

    Notes: Abstract The determination of phosphonate base scale inhibitors in brines by direct current plasma (DCP) and inductively coupled plasma atomic emission spectrometry (ICP-AES) is described. The first method is based on a direct nebulization of the brine samples and plasma using the phosphorus line at 213.618 nm. The second method involves extraction of phosphorus as phospho-antimonyl molybdate complex into methylisobutyl ketone (MIBK) phase and analysis of the extract for molybdenum using the Mo 313.260 nm line. Comparison between the proposed methods and an established recommended method [1] shows excellent agreement between the results in addition to the sensitivity and ease of automation provided by AES.

    Notes: Abstract Sediment cores from the shallow and deep basins of Pyramid Lake, Nevada, revealed variations in composition with depth reflecting changes in lake level, river inflow, and lake productivity. Recent sediments from the period of historical record indicate: (1) CaCO3 and organic content of sediment in the shallow basin decrease at lower lake level, (2) CaCO3 content of deep basin sediments increases when lake level decreases rapidly, and (3) the inorganic P content of sediments increases with decreasing lake volume. Variations in sediment composition also indicate several periods for which productivity in Pyramid Lake may have been elevated over the past 1000 years. Our data provide strong evidence for increased productivity during the first half of the 20th Century, although the typical pattern for cultural eutrophication was not observed. The organic content of sediments also suggests periods of increased productivity in the lake prior to the discovery and development of the region by white settlers. Indeed, a broad peak in organic fractions during the 1800's originates as an increase starting around 1600. However, periods of changing organic content of sediments also correspond to periods when inflow to the lake was probably at extremes (e.g. drought or flood) indicating that fluctuations in river inflow may be an important factor affecting sediment composition in Pyramid Lake.

    Notes: Abstract Forty Fusarium isolates obtained from maize fields were screened for moniliformin production on maize kernels. Twelve isolates, including seven of F. subglutinans, were found to produce moniliformin at levels ranging from 0.4 to 64 ppm. Twenty six isolates were also screened for production of deoxynivalenol, diacetoxyscirpenol, T-2 toxin and zearalenone. Of these, 22, including all 11 isolates of F. graminearum, produced zearalenone at levels ranging from 0.1 to 96.0 ppm, while 13 produced T-2 toxin at low levels, (〈1.1 ppm). Deoxynivalenol and diacetoxyscirpenol were each produced by six isolates, also at low levels (〈1.0 ppm). Three isolates of F. graminearum and one of F. sambucinum produced four toxins simultaneously.

    Notes: Abstract This paper discusses the land types, hydrologic characteristics and processes, and the major modification of these, in relation to mechanisms and magnitude of phosphorus losses to drains and riverine systems which discharge to the Peel-Harvey estuary. About 75% of the coastal plain part of the catchment is cleared of native vegetation and used for dryland, dairy and beef grazing. There are small areas devoted to irrigated pasture and commercial horticulture. Seventy-five percent of the soils of the catchment are sandy surfaced with a poor capacity to retain phosphorus. Though the area is flat, catchment water yields are high because of a large winter rainfall excess and low soil storage capacity. Drainage schemes have been constructed in much of the catchment to remove excess water quickly. This was required initially to allow agricultural expansion and is now important for protecting a growing infrastructure which serves the most populous region of Western Australia.

    Notes: Abstract The European Community is producing annually about 300 × 106 tons of sewage sludges as well as about 150, 950,160 and 200 tons of domestic, agricultural, industrial and other wastes (street litter, dead leaves etc.). About 20–25% of the German sewage sludges, which contain in average about 3.8,1.6, 0.4, 0.6, 5.3% DM−1 N, P, K, Mg and Ca, 202, 5, 131, 349, 53, 3 and 1446 mg kg−1 DM Pb, Cd, Cr, Cu, Ni, Hg, Zn as well as ca. 37 and 5 mg kg−1 Dm polychlorinated hydrocarbons and biphenyls, are recycled annually as fertilizer. In addition environmental impacts on the arable land of Germany may derive from 76,19.2, 64.7, 33.6, 7.8 and 0.1 kg ha−1 a−1 of N, P, K, Ca, Mg and Cu added as animal manures. Besides heavy metals and hazardous organics pathogens are disseminated with organic wastes. Crop production and soil fertility generally profit from the considerable amounts of plant nutrients and carbon in sewage sludges, animal slurries and manures, but the physicochemical soil properties, the composition of microbial, faunal and plant communities as well as the metabolic processes in the soil-, rhizo- and phyllosphere are changed by organic manuring. Consequences for the soil carbon-, nitrogen-and phosphorus-cycle are discussed. Impacts of heavy metals and hazardous organics on the soil biomass and its habitat as well as on transport mechanisms and surival times of disseminated pathogens in soils are reviewed with emphasis on the German situation. A proposal for future strategies (landscape recycling) is made.

    Notes: Abstract Dissolved inorganic nitrogen and phosphorus, their relationship to each other (DIN/DIP) as predisposing (nutrient) factors, as well as prevailing weather as a triggering factor all work together to induce the primary production and hence the eutrophication (hypertrophication) process in surface waters. Sulfate likewise is a decisive predisposing factor influencing the eutrophication process by reducing N availability but increasing P availability and thus acting towards an N limitation of the primary production. This is one of the reasons why marine (coastal) waters and estuaries often exhibit N limitation with respect to primary production, while freshwater ecosystems often tend to exhibit P limitation. Within the N and P balance of agriculture of some countries of Western Europe (Netherlands, Denmark, Switzerland, FRG, UK and Sweden for N, resp. Netherlands, FRG and GDR for P) more the level than the efficiency of the N and P applications indicates the extent of the nutrient surplus. Despite 59–73% N utilization in plant production, the rate of 13–23% for agriculture as a whole equals to the 12–21% efficiency of N use in animal production. The varying N surplus in agriculture in the separate countries of 124 to 465 kg N ha−1 a−1 is determined almost exclusively by the level of the N application and not by its efficiency. The situation is similar for P: In spite of P utilization in plant production of 59–76%, P utilization in total agriculture is only 11–38%, or comparable to the P efficiency within animal production of 10–34%. The differing P excess balance of 55 to 88 kg P2O5 ha−1 a−1 is influenced by the level of the P application. The N and P efficacy of total agriculture hence is determined almost completely by that of animal production, since 83–95% (N basis) and 76–94% (P basis) of the total plant production (on top of the nationally varying levels of N and P use via imported feeds) are fed to animals — with the low N and P utilization cited above. Agriculture's share of the N and P emissions into surface water of several countries/regions in Western Europe (FRG, Netherlands, Italy, Denmark, Switzerland, Norway) ranges from 37 to 82% resp. 27 to 38%. Its share in the flus into the North Sea catchment basin will be about 60% for N and 25% for P related only to the anthropogenic material carried by the rivers. Agriculture's share in the atmospheric N emissions into the North and Baltic Seas can be estimated at about 65% or 55%, resp. while the remaining approx. 35% or 45%, resp. are traceable primarily to anthropogenic burning processes. For agriculture the priority lies in limiting N emissions into surface water caused by leaching, erosion and NH3 emissions, and reducing P emissions mainly through soil conservation (protection against erosion) and water protection. As regards N this means a demand for comprehensive protection of groundwater and atmosphere differentiated according to the potential for losses or the risk of losses on a site, also outside the protection zones. As regards P only those areas can be included in the demand for reduction of emissions that are actually threatened by erosion or surface runoff. Plenty of short-term and long-term measures are available to agriculture to reduce N and P emissions. Especially the long-range measures (such as creating nutrient balances on farms and fields, the integration of animal and plant production, maintaining maximum livestock densities according to the ability of areas to absorb nutrients, altered feeding programs in animal nutrition, changes in livestock keeping (slurry→deep litter), increasing the internal and external recycling of N and P) are capable of bringing about a satisfactory degree of success within the next 20 to 30 years.

    Notes: Abstract With the rapid growth of the poultry industry in Oklahoma, U.S.A., more litter is applied to farm land. Thus, information is required on the impact of applications on regional soil and water resources. The effect of soil and poultry litter management on nitrogen (N) and phosphorus (P) loss in runoff and subsurface flow from four 16 m2 plots (Ruston fine sandy loam, 6 to 8% slope) was investigated under natural rainfall. Plots under Bermudagrass (Cynodon dactylon) received 11 Mg litter ha−1, which amounts to contributions of approximately 410 kg N and 140 kg P ha−1 yr−1. In spring, litter was broadcast on 3 of the plots; the upper half of one and total area of the other two. One of the total-area broadcast plots was tilled to 6 cm, the other remained as no till. The fourth plot served as a control. Relative to the control, litter application increased mean concentrations of total N and total P in runoff during the 16-week study for no-till (15.4 and 5.8 mg L−1) and tilled treatments (16.7 and 6.1 mg L−1). However, values for the half-area application (5.6 and 2.0 mg L−1) were similar to the control (5.7 and 1.3 mg L−1). Interflow (subsurface lateral flow at 70 cm depth) P was not affected by litter application; however, nitrate-N concentrations increased from 0.6 (control) to 2.9 mg L−1 (no till). In all cases, 〈 2 % litter N and P was lost in runoff and interflow, maintaining acceptable water quality concentrations. Although litter increased grass yield (8518 kg ha−1) compared to the control (3501 kg ha−1), yields were not affected by litter management. An 8-fold increase in the plant available P content of surface soil indicates long-term litter management and application rates will be critical to the environmentally sound use of this nutrient resource.

    Notes: Abstract Ensete (Ensete ventricosum W. Cheesm.) is a root crop which stores starch in the root and in the lower part of the stem. It is grown in the southwest of Ethiopia and due to its drought resistance, it is of outstanding importance for the supply of food to the local population. Until now virtually nothing is known about the response of Ensete to fertilizer application. Field trials carried out on three representative soils in Ethiopia showed that Ensete biomass yields were increased significantly on all three soils by nitrogen and phosphorus application. Potassium had only marginal effect on biomass growth but favourably influenced starch production. Sulfate application had no major impact on growth and starch yield. The yield response was well related to the level of available nutrients in the soil, as determined by electroultrafiltration (EUF).     Notes: Abstract In a fertilizer and manure experiment, millet was grown under four treatments (no fertilizer or manure, farmyard manure, chemical fertilizer, and both). Grain yield and total aboveground biomass production of the unfertilized plot were relatively high. The observed differences in total dry matter production must be attributed to differences in nutrient availability, as amount of rainfall and its distribution were favourable. Results show only small differences in distribution of dry matter among the various plant organs between the best and the non-fertilized treatments. Nutrient supply from natural sources, defined as crop content of N, P, and K at maturity without fertilizer application, amounted to 104, 16 and 103 kg ha−1, respectively, which are very high values. Total uptake of calcium and magnesium is related to that of potassium, as the combined content of these three elements is linearly related to total aboveground biomass production. Minimum removal of nitrogen and phosphorus per ton grain dry matter amounts to 29 and 4kg, respectively, and 9 kg potassium per ton total aboveground dry matter. A possible double function of phosphorus as element of structural biomass and for maintenance of electro-neutrality is discussed.

    Notes: Abstract An excess of plant nutrients has caused serious eutrophication in aquatic ecosystems of southwestern Australia manifested by excessive growth and accumulation of green and bluegreen algae. Phosphorus is generally the limiting nutrient for algal growth and phosphatic fertilizers applied to nutrient-deficient, leaching, sandy soils are the main source of P, supplemented by rural industry point sources. Nitrogen is the limiting nutrient in marine embayments with little drainage from the land. Measures to reduce the load of P delivered to drainage include basing fertilizer application rates on soil testing for P and the use of less soluble P fertilizers. Catchment management plans are being implemented with community involvement to reduce P loads and maintain agricultural production. This introductory paper reviews the history of eutrophication in southwestern Australia and of studies into its causes, principally in the large Peel-Harvey estuary. It briefly summarises other papers in this special issue concerned with different aspects of the problem: how to fertilize the land without causing eutrophication.

    Notes: Abstract In order to manage phosphorus (P) losses from soils to waterbodies, knowledge of the mechanisms through which P is retained or released from the soil is essential. Sandy soils of the Peel-Harvey catchment (Western Australia) were subjected to a range of environmental and management factors in the laboratory and field in order to gain an understanding of the mechanisms that affect the magnitude of P losses. Sandy soils accumulated P, despite having little sorption capacity, and this accumulation could be monitored by measuring an acid-extractable fraction. The potential, short-term P loss could be estimated by determination of water-soluble soil solution P prior to winter rains. An annual cycle of the change in arbitrarily defined soil-P pools is discussed in relation to environmental and management factors. Laboratory experiments indicated that P rundown and potential annual P loss in the absence of P fertilizers could be estimated using bicarbonate extractable P. Phosphorus losses were decreased by the application of fertilizers with a low content of water-soluble P. The low ability of sandy soils of the Peel-Harvey coastal catchment to retain P, when compared to other Western Australian soils, is because of low contents of clay minerals and iron and aluminium hydrous oxides.

    Notes: Abstract A three year field study was conducted at five locations in the Nigerian savanna to evaluate the response of early maturing maize variety to varying rates of K and Zn with a view to establishing the K and Zn requirements for maize production in this zone. Treatments consisted of 4 × 3 factorial combinations of 4 levels of K and 3 levels of Zn. Responses to K and Zn fertilization were sporadic and were obtained only in soils of the Southern Guinea savanna and in the soil formed on sedimentary sandstone. There seem to be no problem at present in soils of the Northern Guinea and Sudan savannas where leaching is less intense. It is inferred from this study that K and Zn deficiences are incipient in the high rainfall soils and in the sandstone derived soils. For these soils, 50 kg K/ha and 2–5 kg Zn/ha is suggested as adequate for an early maturing maize crop. Soil data showed that K and Zn responses can be expected when available K and Zn levels fall below 0.1 meq/100 g and 2 ppm respectively.

    Notes: Abstract Sluijsmans and Kolenbrander developed a simple model to describe the availability of animal manure, assuming a readily available, an easily decomposable and a slowly decomposable N fraction. We tested this model on data from an experiment in which farmyard manure had been applied for eleven successive years to silage maize [Zea mays L.] grown on a light sandy soil. The residual effects of this FYM were then measured by growing Italian ryegrass [Lolium multiflorum Lamk.] in the 12th year. The measured uptake of N by the grass of the FYM residues was then compared with the computed values. The measured amounts of N taken up agreed fairly well with the calculated amounts for applications of 50 and 100 t FYM per ha per year. If the rates of manure application are adjusted to crop requirement, the model shows that the potential, long-term release of N from the residual N fraction of FYM will not exceed 20 kg N per ha. For cattle slurry with a smaller residual fraction, the release will be at most 10% of the total annual N application.

    Notes: Abstract It has been suggested that there are several potential benefits of providing nitrogen to cereals via the foliage as urea solution. These include: reduced nitrogen losses through denitrification and leaching compared with nitrogen fertilizer applications to the soil; the ability to provide nitrogen when root activity is impaired e.g., in saline or dry conditions, and uptake late in the season to increase grain nitrogen concentration. Factors that influence the degree of foliar absorption in field conditions have not, however, been clearly defined and losses to the atmosphere and soil can occur. Foliar urea applications may also hinder crop productivity although the explanations for this vary, and include desiccation of leaf cells, aqueous ammonia and urea toxicity, biuret contamination and the disruption of carbohydrate metabolism. It has not yet been determined which one, or combinations, of these mechanisms are most important in field situations. When damage has not been severe, foliar urea applications have increased grain yield, particularly when applied before flag leaf emergence and when nitrogen availability is limiting. Increases in grain nitrogen content are often larger when applications of nitrogen fertilizers to the soil are reduced, and when the urea solution is sprayed either at anthesis or during the following two weeks. It is during this period that foliar urea sprays can be of greater benefit than soil applications with regard to nitrogen utilization by the crop. Increases in wheat grain nitrogen concentration following urea application can improve breadmaking quality. Responses in loaf quality may, however, be variable particularly when increases in grain nitrogen content have been large, and/or when the nitrogen: sulphur ratio in the grain is increased. These circumstances have lead to alterations in the proportions of the different protein fractions which influence breadmaking potential. To exploit the full potential benefits of foliar urea application to cereals, more needs to be known about the mechanisms, and thus how to prevent losses of nitrogen from the foliage, and to reduce the phytotoxic influences of sprays. More information is also required to exploit the reported effects that urea may have on limiting the development of cereal diseases.

    Notes: Abstract A greenhouse experiment, with Okra (Abelmoschus esculentus L.) as the test crop, was conducted on twenty-one soils ranging in Olsen's extractable phosphorus from 1.8 to 15.5µg Pg−1 soil. The experiment was conducted at Punjab Agricultural University, Ludhiana, India. The soils were nonsaline with pH ranging from 7.7 to 8.6. A critical level of 2.55µg Pg−1 soil was predicted by Cate and Nelson's (1971) statistical procedure. Because of a wide range in relative yields, this value did not accurately predict response to applied P. An approach to compute minimum response to applied fertilizer, which is likely to be obtained at a particular Olsen P level, has been presented. It involves calculation of lower 60 percent confidence limits for relative yield and fitting loge-linear regression to the transformed data. The regression was tested on a published data set and was found to hold well.

    Notes: Abstract Two trials inPinus radiata growing on different sites in N.S.W. allowed consideration of fertilizer applications after 2nd or 3rd thinning. The trials included factorial applications of N and P at a single thinning intensity plus a further treatment which allowed assessment of different thinning intensities. The most significant growth responses were obtained by application of N and P in combination. The largest response (additional productivity compared with the unfertilized control) occurred 4 years after application and after 7 years there was no additional absolute response for either of the two sites. The largest fertilizer response was 70 m3 ha−1 over 7 years on one site and 36 m3 ha−1 on the other, indicating differences in absolute responses between sites. It was concluded that in planning treatments the most responsive sites near the end of the rotation should be selected to maximise economic returns. Foliage analyses indicated differences between sites at the commencement of the study. It was concluded that either a single year of foliage analyses at study commencement is of value, or sampling every year of the study should be used to analyse responses, but a single year of analysis during or at the end of the study would not be of value.

Both maximum sorption (Xm) and the binding-energy of sorption as indicated by the binding-energy related constant (k) or the molar free energy (ΔG) of the sorption reaction derived from the Langmuir equation were considerably decreased. On the other hand, phosphate sorption was decreased by increasing concentration of selenite from 0.2µg Se cm−3 to 1.0µg Se cm−3 in the initial solution. The competitive sorption of phosphate with selenite was likely the main mechanism involved in the P-Se interactions. The competitively sorbed selenite exhibited much larger desorption in 0.01M CaCl2 solution, more readily extractable to 0.5M NaHCO3 and significantly higher isotopic exchangeability compared to that sorbed without the competing anion. Results from pot trial using ryegrass indicated that phosphate application increased more efficiently the plant-availability of applied fertilizer Se than that of indegeneous Se in soil.

    Notes: Abstract Alpine glacier retreat resulting from global warming since the close of the Little Ice Age in the 19th and 20th centuries has increased the risk and incidence of some geologic and hydrologic hazards in mountainous alpine regions of North America. Abundant loose debris in recently deglaciated areas at the toe of alpine glaciers provides a ready source of sediment during rainstorms or outburst floods. This sediment can cause debris flows and sedimentation problems in downstream areas. Moraines built during the Little Ice Age can trap and store large volumes of water. These natural dams have no controlled outlets and can fail without warning. Many glacier-dammed lakes have grown in size, while ice dams have shrunk, resulting in greater risks of ice-dam failure. The retreat and thinning of glacier ice has left oversteepened, unstable valley walls and has led to increased incidence of rock and debris avalanches.

    Notes: Abstract Field experiments with silage maize during eight years on a sandy soil in The Netherlands, showed that dicyandiamide (DCD) addition to autumn-applied cattle slurry retarded nitrification, thus reducing nitrate losses during winter. Spring-applied slurry without DCD, however, was on average associated with even lower losses and higher maize dry matter yields. Economically optimum supplies of mineral N in the upper 0.6 m soil layer in spring (EOSMN), amounted to 130–220 kg ha−1. Year to year variation of EOSMN could not be attributed to crop demand only. According to balance sheet calculations on control plots, apparent N mineralization between years varied from 0.36 to 0.94 kg ha−1 d−1. On average, forty percent of the soil mineral N (SMN) supply in spring, was lost during the growing season. Hence, the amounts of residual soil mineral N (RSMN) were lower than expected. Multiple regression with SMN in spring, N crop uptake and cumulative rainfall as explanatory variables, could account for 79 percent of the variation in RSMN. Postponement of slurry applications to spring and limiting N inputs to economically optimum rates, were insufficient measures to keep the nitrate concentration in groundwater below the EC level for drinking water.

    Notes: Abstract Theee trials to evaluat the potential of alley cropping in maize production on the low fertility, acidic soils in Northern Zambia are described. Leucaena leucocephala, Gliricidia sepium, Sesbania sesban, Albizia falcataria, Fleminga congesta, and Cassia spectabilis, were grown in alley crops with hybrid maize and soybean. All trials received recommended rates of P and K fertiliser; N fertiliser was applied at three rates as a subplot treatment. One trial received lime before establishment. Only in the limed trial was there a significant improvement in maize yields through alley cropping; when no N fertiliser was applied, incorporation of Leucaena leucocephala prunings resulted in an increase of up to 95% in yields, with a smaller improvement being produced by Flemingia congesta. There was a significant correlation between the quantity of prunings biomass applied and the proportional increase in maize yields over the control treatment. It is suggested that the lack of effect of most of the tree species on crop yields was due to low biomass production. An economic analysis showed that alley cropping with limed Leucaena was only profitable when fertiliser costs were high in relation to maize prices. However, lime is both expensive and difficult to obtain and transport for most small scale farmers in the region, and is therefore not a practical recommendation. It is suggested that future alley cropping research should focus on screening a wider range of tree species, including other species of Leucaena, for acid tolerance and higher biomass production.

    Notes: Abstract Maize growing next toErythrina hedgerows had 44% lower biomass (p〈0.01) and 35% lower N content (p〈0.1) than maize growing in the middle of the alleys. Maize growing next toGliricidia hedgerows had the same biomass but 56% higher N content (p〈0.1) than maize growing in the middle of the alleys. However these differences did not develop until 2 months after sowing of the maize. Spatial variability in soil nitrogen mineralization and mulch nitrogen release did not explain any of the differences in growth or N uptake of the maize with respect to distance from the trees. It is hypothesized that the slower growth of the maize next to theErythrina trees after 2 months is due to increasing light and/or nutrient competition from the trees as the trees recover from pollarding. The apparent lack of competition fromGlirigidia may be due to different rates of regrowth or different shoot and root architecture. A theoretical model is described demonstrating that if a crop is to take advantage of the higher nutrient availability under alley cropping it must complete the major part of its growth before the trees recover significantly from pollarding, and start competing strongly with the crop.

    Notes: Abstract A detailed study of the soil chemical and physical properties in seven-year-old alley cropping trial containingLeucaena leucocephala andFlemingia congesta in Northern Zambia is described. There was a strong correlation between the maize yield and the total amount of nitrogen applied, both from prunings and fertiliser, suggesting that a major reason for the observed benefit from alley cropping, particularly withLeucaena, was due to an improvement in nitrogen supply.Leucaena produced significantly more biomass, and its leaves had higher concentrations of nitrogen, phosphorous and potassium and lower C/N and C/P ratios than did those ofFlemingia. There was also evidence that the trees had a beneficial effect on other soil chemical properties; under the hedgerows, particularly those ofLeucaena, there were higher levels of organic carbon, Mg, K and ECEC, and pH values were also highest. It is suggested that higher levels of organic carbon in the alley crop treatments were responsible for the improvements observed in soil physical properties. Lower bulk density, lower penetration resistance, and a higher infiltration rate and pore volume fraction were measured in the alley crops, although there was no significant change in the soil water release parameters. A deteriorating effect of constant applications of nitrogen fertiliser on soil fertility was observed; as the level of urea application increased, there were significant decreases in Mg, K and pH, increases in Al and soil acidity, and higher penetrometer resistance. These results highlight the urgent need for further research on biological methods of maintaining soil fertility.

    Notes: Abstract Molecular oxygen is an important regulatory signal in facultative anaerobic bacteria and controles the expression of a great variety of genes positively or negatively. The expression of anaerobic respiration and of related functions of E. coli is controlled by the positive gene regulator FNR, which activates transcription in the absence of O2. The regulated genes carry a FNR consensus sequence upstream of the promoter. Under the same conditions FNR represses some of the genes of aerobic respiration. The binding to the DNA occurs by an α-helix-turn-α-helix DNA-binding domain. FNR contains 5 cysteine residues, four of which are arranged in a cluster close to the N-terminal end. For the function of FNR as a O2-dependent regulator three of the cysteine residues in the cluster and the residue outside the cluster are essential. FNR binds iron as a cofactor which most likely is involved in the O2-sensing by the protein. The experiments indicate that the cysteine residues are responsible for the binding of the iron. From the protein in vivo two functional states can be differentiated, an aerobic or metal-depleted form and an anaerobic form. Only the anaerobic form acts as a gene activator or repressor. Sensing of O2 or of positive redox potentials by the iron ion is thought to cause the conversion of the two functional states. The FNR protein in addition contains a potential nucleotide binding domain. The significance and function of this site is not clear.

This ability has been exploited in genetic approaches to the study of mitochondrial biogenesis. In the last two decades, mitochondrial genetics have made major contributions to the identification of genes on the mitochondrial genome, the mapping of these genes and the establishment of structure-function relationships in the products they encode. Many of the latter are required for post-transcriptional events in mitochondrial gene expression, including the processing of mitochondrial pre-RNAs, the translation of mitochondrial mRNAs, or the assembly of mitochondrial translation products into the membrane. The aim of this review is to describe the genetic approaches used to unravel the intricacies of mitochondrial biogenesis and to summarize recent insights gained from their application.

    Notes: Abstract The Golfe du Lion is mainly bordered by low and narrow sand dunes. Since about four decades, 1/3 of its shoreline has been receding, while 1/3 has been prograding and another 1/3 is stable. Several types of dunes may be described mainly depending on storms, high wind frequencies and sand grain size. Vegetation on dune system is distributed along a primary gradient according to sand stability and soil development, and a secondary gradient along slope of dune according to a seasonal cycle of fresh and salt phreatic water level. Global changes in climate may influence these geomorphological and biological structures mainly through:- Winter minimum temperatures changing the distribution of several plant species, especially in the middle part of the Golfe du Lion. - Frequent high storms which cause damages to the front of the dune systems and disrupt the shore. Changes in dune ecosystems will be cyclic so these tendencies will be obvious only upon a long term period.

    Notes: Abstract Aboveground nitrogen (N) and phosphorus (P) requirement, retranslocation and use efficiency were determined for 28-year-old red oak (Quercus rubra L.), European larch (Larix decidua Miller), white pine (Pinus strobes L.), red pine (Pinus resinosa Ait.) and Norway spruce (Picea abies (L) Karst.) plantations on a similar soil in southwestern Wisconsin. Annual aboveground N and P requirements (kg/ha/yr) totaled 126 and 13 for red oak, 86 and 9 for European larch, 80 and 9 for white pine, 38 and 6 for red pine, and 81 and 13 for Norway spruce, respectively. Nitrogen and P retranslocation from current foliage ranged from 81 and 72%, respectively, for European larch, whereas red pine retranslocated the smallest amount of N (13%) and Norway spruce retranslocated the smallest amount of P (18%). In three evergreen species, uptake accounted for 72 to 74% of annual N requirement whereas for two deciduous species retranslocation accounted for 76 to 77% of the annual N requirement. Nitrogen and P use (ANPP/uptake) was more efficient in deciduous species than evergreen species. The results from this common garden experiment demonstrate that differences in N and P cycling among species may result from intrinsic characteristics (e.g. leaf longevity) rather than environmental conditions.

    Notes: Abstract High quality temperature measurements have been made to depths of 30 to 220 m at 42 sites in 62 observational hydrogeological wells in Alberta. The temperature profiles commonly show near-surface inversions with a minimum temperature at depths of 30 to 50 m. Thermal modelling suggests a surface temperature history with warming reaching 2°C over the past 30 to 60 years. Recent climate warming evident from the analysis of the air temperature data in the region seems to provide at least a partial explanation of the increased ground temperatures. A sudden increase of the surface ground temperature caused by land clearing may be the other explanation, although modelling of such a sudden increase can only explain the observed temperature-depth data if the onset of such warming is 20–30 years old, which is in disagreement with the history of land development in the studied area. The effect of near-surface inversions of the temperature profiles also has been observed in the forested areas. The above support the climate based effect. The superposition of the climatic effect and man-made activity effect upon the ground warming is a very complicated process calling for considerably more research.

    Notes: Abstract We estimate the response of chl-a (mg · m−3) to changes in concentrations of total phosphorus (TP) by calculating the slopeS = Δchl-a/ΔTP in chl-a =f(TP) graphs. Results show that in years where algae are P-limited oligotrophic lakes respond less (median slope 0.21) to changes in nutrient concentrations than eutrophic lakes, (median slope 0.31) and these again less than hypereutrophic lakes, (median slope 1.02). We find no saturation value for the slope within the TP range considered (6–480 mg · m−3). Chl-a in eutrophic lakes responds more frequently to non-nutrient factors than oligotrophic and hypereutrophic lakes. Results obtained by replacing TP with a new nutrient parameter, TP′ = 0.056 · TP · IN0.226, in which inorganic nitrogen, IN, is factored in, suggest that nitrogen has an influence on chl-a in oligotrophic lakes. Blue-green algae respond less to changes in TP than other algal species, e.g., diatoms.

    Notes: Abstract In most lakes eutrophication is linked to an excessive input of phosphorus. Lake restoration by reduction of P-input (external measure) has led to a considerable drop of the P-concentration in all major Swiss lakes as well as in many other lakes. Internal restoration measures such as artificial mixing, drainage of hypolimnetic water, flushing, aeration, biomanipulation and others serve to improve and accelerate the response of a lake to external measures. For the case of Lago di Lugano, a simple two-box model is employed to demonstrate that a reduction of the P-input to about 25% of the present values is necessary to reach the “P-criterion” (P-concentration below 30 µg/l). Internal measures could possibly accelerate the extremely slow response of the northern basin.

It has been observed that pools change little from day to day on the sampling scales usually employed. A simple model is presented where uptake and regeneration rates balance to cause a local steady state concentration for dissolved inorganic nutrients. Enrichment and dilution perturbation experiments with lake water support the idea of steady state nutrient concentrations. Although inorganic nutrient concentrations are often controlled by biota, the absolute concentrations present tell little about the activity of that biota.

    Notes: Abstract Regression results based on data from 46 northern temperate lakes show that total phosphorus (TP) is the best predictor for phytoplankton (as chl-a) at lower trophic levels, TP 〈 200 mg · m−3. A regression including both TP and TN as regressors is the best predictor for lakes with TP 〉 200 mg · m−3. However, the good correlation is probably due to a high correlation between lake average chl-a (all years observed) and lake average TP and TN. Within single hypereutrophic lakes, TN alone is the best predictor. It was not possible to identify a medium trophic domain where TN and TP in combination was the best predictor for chl-a. The ratio TN:TP in the water decreases from about 40 to about 5 with increasing trophic level. Optimum TN:TP ratio for algal species with high abundance during late summer and autumn reflects this decreasing ratio, but within a lesser range, i.e., 20 to 5. In contrast, TN:TP ratios for species abundant during the early vernal period showed no, or an inverse, relation to the TN:TP ratio of the water.

    Notes: Abstract The contribution of nitrogen and phosphorus due to precipitation constitutes the second most important route after superficial runoff. The sampling carried out during a two-year period by means of a precipitation collector allows us to determine the contribution of this route both qualitatively and quantitatively. Nitrogen is mainly supplied in an inorganic form, while phosphorus is principally supplied as orthophosphate. During the period of this study (March 1986–February 1988) it was found that in the Santillana Reservoir Watershed the level of nitrogen supplied by precipitation constitutes an average of 4.87% and the level of phosphorus constitutes 8.01%. The contribution of nitrogen varies in inverse ratio to precipitation and the contribution of phosphorus varies in direct ratio.

After treatment with phorbol esters, THP-1 cells differentiate into macrophage-like cells which mimic native monocyte-derived macrophages in several respects. Compared to other human myeloid cell lines, such as HL-60, U937, KG-1, or HEL cell lines, differentiated THP-1 cells behave more like native monocyte-derived macrophages. Because of these characteristics, the THP-1 cell line provides a valuable model for studying the mechanisms involved in macrophage differentiation, and for exploring the regulation of macrophage-specific genes as they relate to physiological functions displayed by these cells.

    Notes: Summary 150 plants ofD. floribunda representing a single clone were regenerated from a stem tissue culture and regenerants were subjected to cytological, phenotypic and biochemical analysis from the pre-transfer stage to three vegetative growth cycles in the field. The plants could be subdivided into three cytological categories, namely, diploid, mosaic and tetraploid. Diploids, mosaics and the one tetraploid showed diversity amongst themselves with respect to internode length, content of chlorophyll and diosgenin. No marked difference in the length and nature of the leaf or in the type of stoma was recorded. Possible causes of the observed variation are discussed.

    Notes: Abstract There is every reason to expect that it will be possible within the next few years to begin to use farm animals to produce large quantities of some of the human proteins that are needed for the treatment of disease. Revolutionary new opportunities for the production of novel proteins in milk have been created by the development of methods for gene transfer. Exploitation of these opportunities depends upon selection and cloning of milk protein genes and identification of the sequences that govern tissue specific hormonally induced expression in the mammary gland. Studies with three genes, ovine β-lactoglobulin, rat β-casein and whey acidic protein of rat and mouse, suggest that they may all meet this requirement. Fragments of the ovine β-lactoglobulin, murine whey acidic protein and rabbit β-casein genes have directed production of novel proteins in the milk of transgenic mice, sheep, rabbits and pigs. The proteins were biologically active and usually co-migrated with authentic proteins. In early experiments, protein concentration was low, but our recent observations suggest that fusion genes containing genomic clones direct production of concentrations of protein that are suitable for commercial exploitation. In the longer term, two approaches may offer the potential of more reliable expression. Control elements capable of directing expression that is independent of site of insertion of the gene, but dependent on the number of copies of the gene, have been identified for a small number of genes. The availability of such elements for the milk protein genes would increase the reliability of gene expression considerably. Alternatively, targeted mutation of genes may allow the insertion of coding sequences within an existing gene so avoiding position effects.

    Notes: Abstract This chapter is an attempt to summarize some commonly accepted and some more subjective opinions about the regulation of transgene expression in laboratory animals. After a short historical introduction, I present some general notions regarding gene structure/function. The spotlight shifts then to the description of the most popular techniques for gene transfer, including the targeted gene replacement. The different approaches are briefly discussed in terms of intrinsic advantages and limitations regarding gene expression patterns. Furthermore, the role of enhancers, promoters and othercis-acting elements such as silencers and dominant control regions as well as their involvement in the chromatin on-off state are discussed on the basis of a specific example studied in our laboratory. The review concludes by presenting recent results and the new perspectives opening in the field of ‘surrogate’ (also called ‘reversed’) genetics. Some problems which remain to be solved both at the technical as well as at the social-ethical level are also briefly presented.

India Mustard, mediated by Agrobacterium tumefaciens. was developed through the manipulation of the culture medium and the use of the appropriate Agrobacterium strain. High frequency shoot regeneration (90–100%) was obtained from hypocotyl explants grown on medium containing 0.9% agarose, 3.3 mg/L AgNO3 and 0.5–2 mg/L BA in combination with 0.01–0.05 mg/L 2,4-D or 0.1–1 mg/L NAA. Of all the Agrobacterium strains tested, A. tumefaciens A208-SE, carrying the disarmed Ti plasmid and a binary vector pROA93, was the most effective for B. juncea transformation. pROA93 carries the coding sequences of the NPTII and the GUS genes, both driven by a common CaMV 35S promoter in two divergent directions. Inoculated explants grown on the selection medium in the presence of 0.5 mg/L BA and 0.1 mg/L NAA gave rise to transgenic shoots at the highest frequency (9%). All Ro transgenic plants were phenotypically normal, but variation in expression patterns of the GUS gene occurred among the transgenic plants in an organ- and tissue-specific manner. Both the NPTII and the GUS genes were transmitted to the R1 seed progeny and showed co-segregation.

    Notes: Summary Seasonal changes in the hydrogen isotope ratios of xylem waters were measured to determine water sources used for growth in desert plants of southern Utah. While all species used winter-spring recharge precipitation for spring growth, utilization of summer rains was life-form dependent. Annuals and succulent perennials exhibited a complete dependence on summer precipitation. Herbaceous and woody perennial species simultaneously utilized both summer precipitation and remaining winter-spring precipitation, with herbaceous species much more reliant on the summer precipitation component. Several of the woody perennials exhibited no response to summer precipitation. Currently, precipitation in southern Utah is evenly partitioned between winter and summer time periods; however, global circulation models predict that summer precipitation will increase in response to anticipated climate change. Our data indicate that components within the community will differentially responde to the change in precipitation patterns. These results are discussed in relation to impact on competition and possible changes in community structure.

    Notes: Abstract A manipulation experiment was carried out on a field population of the aphid Acyrthosiphon svalbardicum near Ny Ålesund, on the high arctic island of Spitsbergen, using cloches to raise temperature. An average rise in temperature of 2.8 deg. C over the summer season markedly advanced the phenology of both the host plant Dryas octopetala and the aphid. Advanced aphid phenology, with concomitant increases in reproductive output and survival, and successful completion of the life-cycle led to an eleven-fold increase in the number of overwintering eggs. Thermal budget requirements in day degrees above 0°C were calculated for key life-cycle stages of the aphid. Temperature data from Ny Ålesund over the past 23 years were used to calculate thermal budgets for the field site over the same period and these were compared with the requirements of the aphid. Each estimated thermal budget was then adjusted to simulate the effect of a +2, +4, and −2deg. C change in average temperature on aphid performance. This retrospective analysis (i) confirms that the life-cycle of A. svalbardicum is well suited to exploit higher summer temperatures, (ii) indicates that the annual success of local populations are sensitive to small changes in temperature and (iii) suggests that the aphid is living at the limits of its thermal range at Ny Ålesund based on its summer thermal budget requirements.

    Notes: Abstract The larch bud-moth cycle has been observed in the sub-alpine larch-cembran pine forests 16 times since 1850. Infestation is easily recognized by the characteristic red-brown discoloration of the larch crowns due to the wasteful feeding of the bud moth larvae. The heaviest defoliation recurs at intervals of 8.47±0.27 (SE) years, and the larval density per kilogram of larch branches varies more than 10000-fold over four or five generations. The basic regulatory mechanism for this cycle is the induced change in food quality for the two or more subsequent larval generations. Defoliation functions as a negative feedback mechanism acting on larval density. In 1989 local discoloration in the Upper Engadine valley was observed in the usual first focus. In 1990 and 1991, however, instead of the expected widespread defoliation damage, larval densities decreased drastically. Based on extensive field data collected from 1961 to 1991 on the development and the survival of the bud moth (Zeiraphera diniana Gn.) and the phenology of the host, Larix decidua L., this paper shows the effect of weather on survival in the egg stage and on the coincidence of larval hatching with the sprouting of the larch. It is shown that the winter and spring weather conditions in 1989–1991 were conducive to unusually high egg mortality. Since these conditions occurred in three successive generations, population growth was effectively reduced and the cycle collapsed prematurely. Thus the rather persistent cyclicity of the larch-larch bud-moth system was disturbed by weather conditions with a very low probability of occurrence, but due to the inherent high resilience of the system, the next population peak with visible defoliation is expected to occur 1996/1997, provided that the weather conditions return to the climatic standard.

Depending upon fertilizer supply, ecosystem net CO2 uptake per ground area in full sunlight (NCEmax) was 41–81% higher in open-top chambers supplied with doubled ambient partial pressure (p a) of CO2 than in plots receiving ambient CO2. Short-term reversals of the CO2 level suggest that the extent of downward adjustment of canopy photosynthesis under elevated CO2 was 30–40%. ET tended to decline, while water use efficiency (WUE), expressed as the NCEmax:ET ratio, increased more than twofold under elevated CO2. Night-time ecosystem CO2 efflux did not respond to changes in CO2 p a. NCEmax and night-time CO2 efflux were more responsive to mineral fertilizer than the doubling of CO2. This suggests that in these alpine plant communities, atmospheric nutrient input may induce equal or greater effects on gas exchange than increased CO2.

    Notes: Summary Marion Island (47°S, 38°E) has one of the most oceanic climates on earth, with consistently low air temperatures, high precipitation, constantly high humidity, and low incident radiation. Since 1968 mean surface air temperature has increased significantly, by 0.025° C year−1. This was strongly associated with corresponding changes in sea surface temperature but only weakly, or not at all, with variations in radiation and precipitation. We suggest that changing sealevel (atmospheric and oceanic) circulation patterns in the region underlie all of these changes. Sub-Antarctic terrestrial ecosystems are characterized by being species-poor and having a simple trophic structure. Marion Island is no exception and a scenario is presented of the implications of climatic change for the structure and functioning of its ecosystem. Primary production on the island is high and consequently the vegetation has a large annual requirement for nutrients. There are no macroherbivores and even the insects play only a small role as herbivores, so most of the energy and nutrients incorporated in primary production go through a detritus, rather than grazing, cycle. Ameliorating temperatures and increasing CO2 levels are expected to increase productivity and nutrient demand even further. However, most of the plant communities occur on soils which have especially low available levels of nutrients and nutrient mineralization from organic reserves is the main bottleneck in nutrient cycling and primary production. Increasing temperatures will not significantly enhance microbially-mediated mineralization rates since soil microbiological processes on the island are strongly limited by waterlogging, rather than by temperature. The island supports large numbers of soil macro-arthropods, which are responsible for most of the nutrient release from peat and litter. The activities of these animals are strongly temperature dependent and increasing temperature will result in enhanced nutrient availability, allowing the potential for increased primary production due to elevated temperature and CO2 levels to be realized. However, housemice occur on the island and have an important influence on the ecosystem, mainly by feeding on soil invertebrates. The mouse population is strongly temperature-limited and appears to be increasing, possibly as a result of ameliorating temperatures. We suggest that an increasing mouse population, through enhanced predation pressure on soil invertebrates, will decrease overall rates of nutrient cycling and cause imbalances between primary production and decomposition. This, along with more direct effects of mice (e.g. granivory) has important implications for vegetation succession and ecosystem structure and functioning on the island. Some of these are already apparent from comparisons with nearby Prince Edward Island where mice do not occur. Other implications of climatic change for the island are presented which emphasize the very marked influences that invasive organisms have on ecosystem structure and functioning. We suggest that changing sealevel circulation patterns, by allowing opportunities for colonization by new biota, may have an even more important influence on terrestrial sub-Antarctic ecosystems than is suggested merely on the basis of associated changes in temperature or precipitation.

    Notes: Abstract Seasonal net nitrogen (N) and phosphorus (P) mineralization was investigated at Abisko, Swedish Lapland in soils of a subarctic heath and in soils of a colder (by about 4° C), high altitude fellfield by (a) using in situ soil incubation in soils which had been shaded or subjected to two levels of increased temperature, combined with (b) reciprocal transplantation of soils between the two sites. Proportionally large and significant net seasonal mineralization of N, in contrast to non-significant P mineralization, was found in untransplanted and transplanted fellfield soil. In contrast, P was mineralized in proportionally large amounts, in contrast to low N mineralization, in the transplanted and untransplanted heath soil. The differences indicate that P was strongly immobilized in relation to N at the fellfield and that N was more strongly immobilized than P in the heath soil. The immobilization in both soils remained high even after a temperature change of 4–5° C experienced by transplanted soils. Air temperature increases of up to 4–5° C in greenhouses resulted in a soil temperature increase of 1–2° C and did not cause any extra increase of net N and P mineralization. The results suggest that soil temperature increases of up to 2° C, which are likely to occur by the end of the next century as an effect of a predicted 4–5° C rise in air temperature, have only small effects on net mineralization in at least two characteristic tundra soils. These effects are probably smaller than the natural fluctuation of plant available nutrients from site to site, even within the same plant community. A further soil temperature increase of up to 4–5° C may enhance decomposition and gross mineralization, but the rate of net mineralization, and hence the change of nutrient availability to the plants, depends on the extent of microbial immobilization of the extra nutrients released.

    Notes: Abstract Integrative ecophysiological and vegetative responses of Dryas octopetala were measured in response to field perturbations of temperature, precipitation and their interactions in a polar semi-desert in Svalbard, Norway (79°N, 12°E). Leaf carbon isotope discrimination (Δ), total leaf nitrogen concentration and leaf development were determined for photosynthetic leaves collected during the last week of August 1991, after one season of manipulations. Individual leaf weight and the total mass of leaf tissue were significantly lower when water was added, irrespective of temperature regime. Leaf carbon isotope discrimination and estimated long-term c i/c avalues (the ratio of CO2 concentration in leaf intercellular spaces to that in the atmosphere) were significantly higher under all three field manipulation treatments, and Δ was significantly reduced when Dryas was grown under drought conditions in a related greenhouse study. Nitrogen concentrations of plants from the field experiment were significantly lower under warmed conditions regardless of water regime. Our results indicate that changes in environmental conditions in high arctic settings will result in alterations of Dryas leaf gas exchange, as expressed by increases in carbon isotope discrimination, which may be accompanied by shifts in leaf nitrogen content and leaf biomass.

    Notes: Abstract A cytological and molecular analysis was performed to assess the genetic uniformity and true-to-type character of plants regenerated from 20 week-old embryogenic suspension cultures of meadow fescue (Festuca pratensis Huds.), and compared to protoplastderived plants obtained from the same cell suspension. Cytological variation was not observed in a representative sample of plants regenerated directly from the embryogenic suspensions and from protoplasts isolated therefrom. Similarly, no restriction fragment length polymorphisms (RFLPs) were detected in the mitochondrial, plastid and nuclear genomes in the plants analyzed. Randomly amplified polymorphic DNA markers (RAPDs) have been used to characterise molecularly a set of mature meadow fescue plants regenerated from these in vitro cultures. RAPD markers using 18 different short oligonucleotide primers of arbitrary nucleotide sequence in combination with polymerase chain reaction (PCR) allowed the detection of pre-existing polymorphisms in the donor genotypes, but failed to reveal newly generated variation in the protoplast-derived plants compared to their equivalent suspensionculture regenerated materials. The genetic stability of meadow fescue plants regenerated from suspension cultures and protoplasts isolated therefrom and its implications on gene transfer technology for this species are discussed.

    Notes: Abstract A possible response to increasing atmospheric CO2 concentration is to attempt to increase the amount of carbon stored in terrestrial vegetation. One approach to increasing the size of the terrestrial carbon sink is to increase the growth of forests by utilizing intensive forest management practices. This article uses data from the literature and from forest growth and yield models to analyze the impact of three management practices on carbon storage: thinning, fertilization, and control of competing vegetation. Using Douglas-fir (Pseudotsuga menziesii) and loblolly pine (Pinus taeda) as example species, results from experiments with computer simulation models suggest that, for these two species, thinning generally does not increase carbon storage and may actually cause a decrease. The exception is thinning of very dense young stands. Fertilization generally increases carbon storage, although the response can be quite variable. The largest gains in carbon storage are likely to come from fertilizing lower-quality sites and from fertilizing thinned or less dense stands. Forests usually show increased growth in response to fertilization over a wide range of ages. Simulation of the growth of loblolly pine indicates that controlling competing vegetation at an early age helps to maximize stand growth and carbon storage.

    Notes: Abstract A deterministic, one-dimensional, unsteady numerical model has been developed, tested, and applied to simulate mean daily dissolved oxygen (DO) characteristics in 27 lake classes in the state of Minnesota. Reaeration and photosynthesis are the oxygen sources, while respiration, sedimentary, and biochemical water column oxygen demand are the sinks of oxygen in the model. The lake classes are differentiated by surface area (A s), maximum depth (H max), and trophic status expressed as Secchi depth (Z s). Because lake stratification is most important to lake oxygen dynamics, simulated DO characteristics are plotted in terms of a stratification parameterA s/H max 0.25 and Secchi depthZ s. Simulations provide DO profiles on a daily time scale. Specific DO characteristics of ecological and environmental interest are epilimnetic DO, hypolimnetic DO, DO gradient from surface to bottom, and DO minima and maxima. Specific results are as follows: Simulated mean daily and weekly DO values in the epilimnion of all lakes for both past and future climate scenarios are near saturation over the summer season. Hypolimnetic DO values depend strongly on lake morphometry, trophic status, and time throughout the summer season. Future climate conditions are specified as the historical records from 1955 to 1979, adjusted (monthly) by the 2 × CO2 GISS model output to account for doubling of atmospheric CO2. With this climate change, weekly averaged epilimnetic DO is projected to drop by less than 2 mg/liter, and will remain above 7 mg/liter throughout the open water season. The hypolimnetic DO reductions after climate change are on the order of 2–8 mg/liter. Periods of anoxia are longer by as much as 80 days. Those changes would alter water quality dynamics in lakes and have a profound effect on lake ecosystems including indigenous fishes. The results presented are useful for evaluating environmental management options.

    Notes: Abstract Large-scale environmental research efforts are conducted under frameworks that provide a common basis for evaluating research in many diverse fields. Such frameworks should be subjected to critical review to determine if they meet crucial expectations. In the case of the US Global Change Research Program, we perceive that most research follows a framework that we define as the predictive model framework. We believe this framework is insufficient for resolving the unprecedented predicaments posed by global change. We recommend a complementary framework, the system response framework, which directs research toward useful indicators of change rather than precise predictions. We further argue that, even if research is complementary and effective under the two frameworks, conclusive results prior to decisions should not be expected. The burden of proof must itself be a continuing topic of open discourse and inquiry.

    Notes: Summary Cytochrome c oxidase (COX), like other multi-subunit components of the respiratory chain, is controlled by both the nuclear and the mitochondrial genome. In order to find wether there is a close relationship between mRNAs encoded by the nucleus and by the mitochondrion, and between these mRNAs and enzyme activity, we compared six rat tissues (ventricle, liver, m. soleus, m. plantaris, and the white and red portions of m. gastrocnemius). We found a tenfold range for COX activity, a tenfold range for the contents of mRNA III (mitochondrial) and mRNA VIc (nuclear), a threefold range for total [poly(A)+] mRNA content and a sevenfold range for total RNA content in these tissues. The ratio of mRNA III to mRNA VIc was equal in each tissue, indicating the presence of a mechanism that coordinates the two genomes. There was a good correlation between mRNA content and COX activity (r = 0.78 for VIc, r = 0.77 for 111; p 〈 0.0001), demonstrating that the expression of this enzyme is mainly under pretranslational control.

    Notes: Abstract The involvement of a hypocalcemic hormone calcitonin (CT) in the expression of hepatic Ca2+-binding protein regucalcin mRNA was investigated. The change of regucalcin mRNA levels was analyzed by Northern blotting using liver regucalcin complementary DNA (0.9 kb). A single oral administration of calcium chloride (100 mg Ca/100 g body weight) to rats induced a remarkable increase in the serum calcium concentration and a corresponding elevation of the liver calcium content during 120 min after the administration. Thyroparathyroidectomy (TPTX) did not cause a significant increase in the liver calcium content after calcium administration. Hepatic regucalcin mRNA level was markedly elevated by calcium administration; the level was about 180% of controls at 60 min after the administration. This increase was completely abolished by TPTX. A single subcutaneous administration of CT (synthetic eel CT; 25–100 MRC mU/100 g) to TPTX rats received oral administration of calcium (100 mg/100 g) produced a remarkable increase in hepatic regucalcin mRNA levels; the level was about 280% of controls with the dose of 25 MRC mU CT/100 g. The present finding suggests that the expression of hepatic mRNA is stimulated by CT, and that the hormonal effect is mediated through Ca2+ in rat liver.

    Notes: Abstract The effect of phenobarbital on the expression of calcium-binding protein regucalcin mRNA in rat liver was investigated. The change of regucalcin mRNA levels was analyzed by Northern blotting using liver regucalcin cDNA (0.9 kb of open reading frame). Phenobarbital (4, 8 and 12 mg/ 100 g body weight) was intraperitoneally administered to rats 3 times with 24 h intervals, and the animals were sacrificed by bleeding at 24 h after the last administration. The hepatic regucalcin mRNA levels were markedly reduced by phenobarbital administration. This decrease was about 50% of control level with the 12 mg/100 g dose. Moreover, the hepatic regucalcin concentration was significantly decreased by the administration of phenobarbital (12 mg/100 g), although the serum regucalcin concentration was not altered appreciably. Meanwhile, serum transaminases (GOT and GPT) activities were not increased by the administration of phenobarbital (4 and 12 mg/100 g). The present study demonstrates that the expression of hepatic regucalcin mRNA is decreased by phenobarbital administration in rats, suggesting that regucalcin does not have a role in drug metabolism related to phenobarbital.

    Notes: Abstract Transforming Growth Factor-beta1 (TGF-β1) is expressed in the heart by muscle and non-muscle cardiac cells.In vitro, cardiac myocytes and non-muscle cells including cardiac fibroblasts and endothelial cells respond to regulatory effects of TGF-β1. Expression of TGF-β1 in the heart is subject to regulation by hemodynamic stimuli. Increased expression of mRNA transcripts for TGF-β1 has been reported in several models of cardiac hypertrophy. The objective of this study was to determine the effect of TGF-β1 in the myocardium. TGF-β1 was injected intravenously. Expression of mRNA transcripts for functional and structural proteins was determined by Northern hybridization analysis. DNA synthesis was determined by measurement of3H-thymidine incorporation into ventricular DNA. The results showed differential regulation of mRNAs for myocyte- and non-myocyte-specific proteins in the heart of TGF-β1 treated rats. Moderate but statistically significant decrease in DNA synthesis was observed in the heart of TGF-β1 treated rats (37.5%, P〈0.025). Together, these data point to a physiological role for TGF-β1 in the heart. They further suggest that similar to its diversein vitro cell-specific regulatory effects, TGF-β1 may have multicellular targets in the heart. Effect of TGF-β1 alone or combined with those of other cytokines/hormones that come into play, as the result of its administration, may be responsible for altered gene expression and DNA synthesis in the myocardium. We propose that in experimental models of myocardial hypertrophy which are associated with increased expression of TGF-β1 in the heart, the contribution of regulatory effects of this growth factor to the manifestations of ventricular hypertrophy could be significant.

    Notes: Abstract A secondary mutant, derived from an allele of maize alcohol dehydrogenase 1 (Adh1) carrying a Mutator transposable element (Mu1) in its first intron, was reported to exhibit a threefold decrease in ADH enzymatic activity and steady-state RNA levels compared to the original mutant. The original mutant,Adh1-S3034 (abbreviatedS3034), was previously characterized at the molecular level. The derivative, abbreviatedS3034b, has now been cloned; at the DNA sequence level the insertion and surroundingAdh1 sequences are indistinguishable fromS3034. Furthermore, in our lines there is no difference in relative ADH activities between products of the two putative alleles. A comparison of gene expression in heterozygotes obtained by crossing to different tester lines reveals a correlation between the measured decrease in levels of ADH polypeptide produced by the mutant allele and the background in which it is measured; this effect is distinct from any background-related variation in the expression of the progenitor allele. It does not appear to be attributable to alternative patterns of DNA modification. It appears to reflect a background-associated difference in the level of normalAdh1-RNA produced. Thus the previously reported distinction betweenS3034 andS3034b may be due to differences in the extent to which the mutant allele and a given genetic background interact to produce functionalAdh1-RNA.

    Notes: Abstract We have compared the expression of the retinoblastoma (Rb) and p53 genes in normal human fibroblasts, colon carcinoma cell lines, matched pairs of colorectal tumor tissues and adjacent normal mucosa and in synchronized human diploid fibroblast cell line W138. The increased expression of Rb and p53 RNA was observed in a majority of colorectal cancers in comparison to adjacent normal mucosa and is accompanied by proportional increase in the expression of histone H3 gene. The Rb and p53 RNA levels varied significantly between the various colon carcinoma cell lines. However, we found that the expression of Rb and p53 RNA is regulated differently in cell cycle synchronized normal human fibroblasts. The Rb mRNA level did not change with the position in the cell cycle and did not differ significantly whether the cells were serum deprived or in 10% serum. But p53 mRNA expression follows the same pattern as histone H3 mRNA.

    Notes: Abstract The PTP-2 cDNA encoding an intracellular protein tyrosine phosphatase (PTPase-2) was isolated and sequenced from mouse testis and T-cell cDNA libraries. This PTP-2 cDNA was found to be homologous to human PTP-TC and rat PTP-S, and contained 1,551 nucleotides, including 1,146 nucleotides encoding 382 amino acids as well as 5′ (61 nucleotides) and 3′ (344 nucleotides) non-coding regions. Northern blot analysis indicated that PTP-2 mRNA of 1.9 Kb was most abundant in testis and kidney, although it was also present in spleen, muscle, liver, heart and brain.

    Notes: Abstract Calcium has long been known to play a role as a key cytoplasmic second messenger, but until relatively recently its possible involvement in nuclear signal transduction and the regulation of nuclear events has not been extensively studied. Evidence revealing the presence of transmembrane nuclear Ca2+ gradients and a variety of intranuclear Ca2+ binding proteins has fueled renewed interest in this key ion and its involvement in cell-cycle timing and division, gene expression, and protein activation. This review will offer an overview of the current state of knowledge and theory regarding calcium orchestration of nuclear functions and events and discuss possible future directions in this field of study.
