    Notes: Summary In the course of studying the plasma adrenocorticotropic hormone (ACTH) and corticosterone responses to synthetic corticotropin releasing factor (CRF), we noted some disparity in the responses. A higher dose (20 μg compared with 5 μg per rat i.a.) produced an equal plasma ACTH but greater plasma corticosterone response in adult male rats. Thus, we examined the possibility that CRF increases adrenocortical responsiveness to ACTH. CRF significantly (p〈0.0005) increased the plasma corticosterone response to ACTH in rats pretreated with dexamethasone. Thus, synthetic CRF increases corticosterone secretion in rats not only by stimulating ACTH secretion, but also by increasing the adrenocortical responsiveness to ACTH.

Le taux de croissance ne varie que légèrement dans les groupes d'âges différents. Il est plus élevé chez les animaux jeunes et décroit considérablement en fonction de l'augmentation de l'âge. Les rats mâles présentent une croissance plus élevée que les femelles. Cette étude a pour but de mettre au point une méthode permettant de déterminer les facteurs expérimentaux, liés à la croissance en longueur du rat.
    Abstract: Zusammenfassung Die normale Längenwachstums-Geschwindigkeit der proximalen Wachstumsplatte der Tibia wurde bei Sprague-Dawley-Ratten in einem Alter zwischen 20 und 100 Tagen mittels der Tetracyclinmethode gemessen. Die Wachstumsgeschwindigkeit variierte nur wenig innerhalb der einzelnen Altersgruppen. Die Geschwindigkeit war bei jungen Tieren am höchsten und nahm mit zunehmendem Alter beträchtlich ab. Männliche Ratten wuchsen schneller als weibliche. Diese Arbeit dient als Grundlage, um die experimentelle Beeinflussung des Längenwachstums der Ratte abschätzen zu können.
    Notes: Abstract The rate of normal growth in length from the proximal growth plate of the tibia in the Sprague-Dawley rat was measured between 20 and 100 days of age using the tetracycline method. The growth rate varied only slightly within different age groups. The rate was highest in young animals and decreased considerably with increasing age. Male rats grew faster than female. This study is intended to provide a base for an evaluation of experimental influence on the growth in length of the rat.

Cet emploi est basé principalement sur les effects ostéosclérotiques bien connus du fluorure de sodium, mais on connait mal son effet sur la force de résistance osseuse. L'influence de diverses concentrations de fluorure de sodium sur la force de résistance osseuse de jeunes rats soumis à des régimes riches et pauvres en calcium a été étudiée. L'administration de fluorure de sodium augmente le diamètre osseux, indiquant une stimulation de la formation de l'os périosté, mais la force de résistance osseuse est réduite ou non modifiée par l'ingestion de fluor.
    Abstract: Zusammenfassung Die therapeutische Verwendung von Natriumfluorid ist für eine Anzahl von Knochenmangel-Krankheiten empfohlen worden. Die Empfehlungen basieren hauptsächlich auf den bekannten osteosklerotischen Wirkungen von Natriumfluorid; über dessen Effekt auf die Knochenstärke ist wenig bekannt. In dieser Arbeit wurde der Einfluß verschiedener Konzentrationen von Natriumfluorid auf die Knochenstärken von wachsenden Ratten mit hoher und niederer Calciumeinnahme untersucht. Die Verabreichung von Natriumfluorid erhöhte den Knochendurchmesser, was auf eine Stimulierung der Periostbildung hinwies, die Knochenstärke wurde jedoch durch Fluorideinnahme herabgesetzt oder nicht beeinflußt.
    Notes: Abstract The therapeutic use of sodium fluoride has been recommended in a variety of osteopenic bone diseases. The recommendations are based mainly on the known osteosclerotic effects of sodium fluoride and little information is available as to its effect on bone strength. The influence of various concentrations of sodium fluoride on bone strength in growing rats on high and low calcium diets was studied. The administration of sodium fluoride increased bone diameter, indicating stimulation of periosteal bone formation, but bone strength was reduced or not affected by fluoride ingestion.

La production cellulaire de la métaphyse est déterminée sur la base de la croissance osseuse longitudinale déterminée par l'oxytétracycline et la taille des cellules en dégénérescence. La diminution de la croissance osseuse longitudinale, en fonction de l'augmentation de l'âge et après hypophysectomie, est due partiellement, à la diminution de production cellulaire et partiellement à une décroissance de la taille des cellules en dégénérescence dans la métaphyse. L'influence de la production cellulaire et de l'activité mitotique prédomine.
    Abstract: Zusammenfassung Die Breite der proximalen Wachstumsplatte der Tibia, deren undifferenzierter und säulenförmiger Zone und die Größe der nahe bei der Metaphyse auftretenden degenerativen Zellen wurden in normalen und hypophysektomierten Sprague-Dawley-Ratten bestimmt. Die Zellproduktion in der Wachstumsplatte wurde aus dem longitudinalen Knochenwachstum berechnet, welches mittels Oxytetracyclin und der Größe der degenerativen Zellen bestimmt wurde. Es wurde festgestellt, daß die Abnahme des longitudinalen Knochenwachstums bei zunehmendem Alter und nach Hypophysektomie zum Teil einem Rückgang in der Zellproduktion, zum Teil einer Verminderung der Größe der degenerativen Zellen in der Wachstumsplatte zuzuschreiben ist. Der Einfluß der Zellproduktion, und somit der mitotischen Aktivität, herrscht vor.
    Notes: Abstract The width of the proximal growth plate of the tibia, its undifferentiated and columnar zone and the size of the degenerative cell close to the metaphysis, were determined in normal and hypophysectomized Sprague-Dawley rats. The cell production in the growth plate was calculated from the longitudinal bone growth determined with oxytetracycline and the degenerative cell size. It was found that the decrease in longitudinal bone growth with increasing age and after hypophysectomy, is due partly to a decrease in cell production, and partly to a decrease in degenerative cell size in the growth plate. The influence of cell production and thus the mitotic activity predominates.

    Notes: Summary This is a study of the fine structure of cells of the 20-day fetal rat calvarium. Special attention is given to identifying and characterizing preosteoclasts. These cells are relatively common and located largely, but not exclusively, at the endocranial bone surface. The preosteoclasts are characterized by abundant mitochondria, an incomplete perinuclear Golgi apparatus, and variable-shaped dense granules. The dense granules are unique in appearance in that they contain an internal dense matrix surrounded by a clear halo. Most granules are circular in shape but some are elongate or tubular in form. Granules with identical appearance are observed in osteoclasts. The preosteoclasts are mononucleate, or occasionally binucleate. It is suggested that because preosteoclasts are morphologically distinctive and relatively abundant, it should be feasible to separate these cells from a heterogeneous cell isolate.

    Notes: Summary Rats were injected with45Ca and horseradish peroxidase to determine the patterns of45Ca incorporation into incisor enamel and the morphological types of the overlying maturation ameloblasts.45Ca autoradiography showed no differences in the patterns of incorporation into enamel between routinely embedded and freeze-dried specimens. Enamel overlaid by ruffle-ended ameloblasts was much more heavily labeled while that overlaid by smooth-ended ameloblasts showed only moderate labeling. The observations lend further support to the hypothesis that the ruffle-ended cells are very active in mineralizing enamel and that the smooth-ended cells are in a passive, restorative phase.

    Notes: Summary The urinary excretion of two connective tissue metabolites was studied in both control and vitamin D deficient rats. Hydroxyproline (HyPRO) excretion was determined after 2, 13 and 22 months (experiment I). It decreased with aging in animals receiving the control diet. On the contrary, this excretion increased as a function of age in vitamin D deficient animals. At the age of 22 months, HyPRO excretion was respectively 31 and 1708 µg a day in control and deficient animals. HyPRO and glycosaminoglycans (GAG) excretion was measured on a group of both control and vitamin D deficient rats at the age of 21 months (experiment II). These results confirm the high excretion of HyPRO in deficient animals. On the contrary, the GAG excretion was higher in control animals than in deficient ones, the mean excretion being respectively 412 and 234 µg a day.

    Notes: Summary A method for quantitative studies of the formation rate of bone has been developed. After vital staining with calcein, the fluorescence of a bone section was measured with a microphotometer controlled by a mini computer. After staining the bone structure with alizarin red S in a second step, the section was measured in transmitted light. The two data sets were combined and the shortest distances between the bone surface and the fluorescence lines were computed. With this information the distance distribution and the bone area between the label and the surface could be calculated in two different ways: with the single labeling and the continuous labeling techniques. The advantages and disadvantages of the two methods are discussed and compared with those of other techniques.

    Notes: Summary The purpose of this work was to analyze the proximal tibial metaphysis of the 170 g rat in a quantitative histologic fashion which would allow some relation to tissue age to be established. Stained 3 µm thick tissue sections were analyzed with the aid of a Merz grid on an eyepiece reticule and a light microscope. Tissue mass and cell distribution were studied in all areas. The rate of change in tissue mass during aging of the metaphysis was calculated. Two regions of the metaphysis were identified. One, corresponding to the primary spongiosa, less than 4.45 days of age, is a region of high turnover of hard tissue and high numbers of osteoblasts and osteoprogenitor cells. The other, corresponding to the secondary spongiosa, is a region of relatively low net tissue turnover and low numbers of osteoblasts and osteoprogenitor cells. Osteoclasts were found relatively more uniformly distributed through the metaphysis than were osteoblasts and osteoprogenitor cells. The rate of bone formation in the primary spongiosa is 50 times that found in the Haversian bone of the rib of 5-year-old humans and about 500 times that found at the cortical-endosteal surface of ribs of 5-year-old humans. It is argued that both cell distribution and tissue distribution in the metaphysis support the concept that osteoblasts and osteoclasts, rather than osteocytes, are responsible for the maturation of the metaphysis. The inhomogeneous distribution of both cells and tissue in the metaphysis has definite meaning for the interpretation of findings concerning the incorporation of radionuclides into the skeleton.

    Notes: Summary The demands of growth are known to exacerbate the effect of phosphorus deprivation (PD). We examined whether changes associated with PD could be prevented in young rats in which growth and growth hormone (GH) were eliminated by hypophysectomy (HPX) and whether PD in normal intact rats (INT) was associated with increased secretion of GH. INT or thyroxine- and ACTH-replaced HPX rats were fed one of the three diets: 0.31% P (NP); 0.027% P (LP), and 0.31% P, pair-fed with LP-mates (NP-PF). The results indicate that HPX did not qualitatively alter several physiologic responses to PD: (a) serum and urinary phosphorus (P) decreased and urinary calcium (Ca) increased; (b) net intestinal Ca retention fell and duodenal sac uptake of45Ca rose; and (c) external P balance was restored and duodenal sac uptake of32P-phosphate increased. Only the hypercalcemia seen in INT, LP rats was prevented by HPX. In INT rats serum immunoassayable GH levels, measured in single samples, were not different between different dietary groups while pituitary bioassayable GH was reduced in both LP and NP-PF rats when compared to the NP rats. Thus, except for hypercalcemia, the physiologic responses associated with PD are not prevented by the elimination of growth and GH, and the development of these responses in INT rats was not associated with a consistent or specific alteration in GH secretion.

    Notes: Summary Protein deficiency was produced by freely feeding young rats a 1% lactalbumin diet for 12 weeks in order to study the effects of protein-calorie malnutrition on skeletal development. During the experiment the food and caloric intake and weight of the experimental animals decreased, while those parameters of the control animals progressively increased. However, when gross caloric intake was expressed as a function of the metabolic size of the animal, the caloric consumption was similar for both groups of animals. The protein-deficient animals exhibited micro-radiographic and histological features of an abnormal pattern of endochondral bone formation. Appositional bone growth, as determined by the daily appositional rate and the percentage of endosteal surfaces undergoing active bone formation, was significantly decreased in these animals, as was the percentage of periosteal surfaces exhibiting resorption. Both chemical analyses of the whole bone and electron probe microanalysis in the specific area of actively calcifying bone revealed no significant differences between the mineral content of control and protein-deficient animals. This study distinguishes the effects of protein deficiency from that of combined protein-calorie deprivation and demonstrates that the abnormal skeletal development observed was the result of a decrease in the quantity of bone formed rather than an altered mineral content.

    Notes: Summary Acid phosphatase was localized in rat incisor ameloblasts without prior decalcification. Whenβ-glycerophosphate was used as the substrate, an intense reaction was observed in the supranuclear region of the secretory ameloblasts. But the reaction was dramatically reduced at the transitional stage and was very weak in the maturation ameloblasts. Whenp-nitrophenylphosphate was the substrate, the reaction product was consistently seen in the Golgi cisternae and the vesicular components of the ameloblasts at all stages of enamel development. These observations suggest that there are two acid phosphatases in ameloblasts. One is in the secretory ameloblasts and the other in the transition and maturation ameloblasts. X-ray micro-analyses for Fe and Pb showed that Fe and acid phosphatase were in the ferritin-containing vesicles at the later stage of enamel maturation. This evidence suggests that ferritin is digested in these vesicles for the release of the Fe pigment to the enamel. An increase in the number of intercellular bridges between ameloblasts was correlated with the dramatic decrease in height of ameloblasts at the pigment release stage. The ameloblast membranes were acid phosphatase positive at the intercellular bridges whenp-nitrophenylphosphate was the substrate. This activity may be involved in the reduction in the surface area of the ameloblast membranes.

    Notes: Summary Preliminary indications of the occurrence of actin and myosin in crude matrix vesicle preparations have been reported previously. In the present study extracellular matrix vesicles from rat alveolar bone were isolated. They were further purified by a sucrose density gradient. SDS-polyacrylamide gel electrophoresis of the purified vesicles revealed the presence of a polypeptide with a molecular weight of 43 K daltons and with electrophoretic mobility identical to that of blood platelet actin. The limited proteolysis of both 43 K dalton vesicular polypeptide and actin byStaphylococcus aureus-V8-protease revealed three fragments with identical electrophoretic mobility. In addition, the vesicular preparations inhibited the activity of DNase I, a property typical of actin monomers. Filamentous material extracted from matrix vesicles showed ultrastructural features of F-actin. Reaction of this material with heavy meromyosin resulted in arrowhead formation, which is characteristic of acto-heavy meromyosin. The occurrence of actin in extracellular matrix vesicles may account for their budding from the osteoblastic plasma membrane, their possible motility in the matrix, and maintenance of the spherical shape.

    Notes: Summary Eighty-eight adult male Sprague-Dawley rats were given a diet with either (a) 0.5% Ca and 0.6% P or (b) 0.01% Ca and 0.6% P. Osteopenia was created by adding prednisolone to the diet. The prophylactic effect of oral 1,25(OH)2D3 on the osteopenia was studied. It was found that prednisolone osteopenia in the rat was associated with defective Ca absorption. By giving an oral dose of 1,25(OH)2D3, it was possible to maintain normal Ca absorption during prednisolone treatment and to prevent the bone loss. No significant hypercalcemia or any kidney calcifications were seen. These results are in contrast to earlier findings, in which subcutaneous administration of 1,25(OH)2D3 failed to prevent prednisolone osteopenia because of its tendency to increase bone resorption.

    Notes: Summary The use of irradiated pollen to bring about limited gene transfer in wheat has been investigated. Doses of X-rays of 2Kr, 3Kr and 5Kr were used to generate M1 progeny between maternal and paternal genotypes differing in quantitative and major gene characters. Cytological studies of M1 plants revealed hybrids with widespread aneuploidy and structural rearrangements in the paternal genome. These effects resulted in phenotypic variation between M1 progeny and complex multivalent formation at meiosis. All M1 plants at the 5Kr and 3Kr doses were sterile and all but 2 plants at the 2Kr dose. Studies of the two M2 families from these plants revealed disturbances in genotype frequencies for some of the marker loci with an excess of maternal homozygotes and a deficit of paternal homozygotes. This was also reflected in a more maternal appearance for quantitative characters. These results are interpreted as showing that irradiation damage to the paternal genome in M1 plants results in the differential transmission of maternal alleles.

    Notes: Summary The effects of the ‘Tom Thumb’ Rht3 dwarfing gene in wheat on plant height, yield components and grain quality in field trials of random inbred lines from two ‘Minister Dwarf’ (Rht3) x rht3 crosses are described. Limited genetic increases in Rht3 plant height, up to 70 cm, were achieved by incorporating genes from the extremely tall variety ‘April Bearded’. Amongst lines from the higher yielding cross, the dwarfing gene did not reduce grain yield but was associated with an increase in grain number of over 25%. Although Rht3 reduced grain protein content by up to 0.8%, breadmaking quality, as measured by SDS sedimentation values and baking tests, was not affected. A controlled environment experiment with near-isogenic Rht3 and rht3 lines indicated that reduced grain size in the dwarfs is a competitive response to the increase in fertility rather than a primary effect of the dwarfing gene.

    Notes: Summary A cytological study has been made of plants regenerated from cultured immature embryos of four wheat cultivars (Triticum aestivum, 2n = 6x = 42). In total, 29% of the 192 plants examined were aneuploid with a range in chromosome numbers of 38–45. Evidence of chromosome structural changes was also found. This variation occurred in regenerants of all four cultivars, but there were large differences in the proportions of aneuploids arising from individual cultures which meant that no significant differences could be demonstrated between cultivars. Chromosome abnormalities were present in plants regenerated both from embryogenic cultures and from cultures in which the origin of shoots could not be distinctly defined.

    Notes: Summary Triticum turgidum L. var. dicoccoides (wild emmer) is an important genetic resource for increasing the protein content of common wheat (Triticum aestivum L.). Many studies have shown that the presence or absence of bands in sodium dodecyl sulfate polyacrylamide (SDS-PAGE) electrophoregrams of wheat storage proteins to be of a purely genetic character. A total protein extraction and SDS-PAGE technique was used to estimate the storage protein genetic variability among 841 accessions of wild emmer collected from various ecological regions in the Middle East. In addition, a computer data bank was developed, recording the onedimension electrophoregram bands for each accession by molecular weight (MW) and relative Coomassie Blue staining intensity as determined from densitometer scans. Analyses of this information are being used to identify specific accessions for further study by two dimension electrofocusing-electrophoresis and breeding and genetic analyses. The computer-assisted analyses indicated that the greatest genetic variability occurs for proteins in the high MW region (above 70,000 MW) followed by those in the medium range (70,000 to 33,300 MW). Comparatively little variability was revealed for protein subunits of below 33,300 MW.

    Notes: Summary Salt-soluble proteins from the endosperms of wheat, barley, and rye have been separated by nonequilibrium electrofocusing x electrophoresis. Genes encoding 14 of the 25 components observed in wheat have been unambiguously assigned to 10 different chromosomes (1B, 3B, 3D, 4A, 4D, 5B, 6B, 6D, 7B, 7D) by analysis of the compensated nulli-tetrasomic series. Five more wheat proteins seem to be controlled by group 2 chromosomes. Analysis of wheat-barley and wheat-rye addition lines has led to the location of genes for 6 out of 20 barley proteins in 4 different chromosomes (1H, 3H, 4H, 6H; 1H is homoeologous to group 7 chromosomes of wheat) and of genes for 5 out of 20 rye proteins in two different chromosomes (2R, 4R). The relationship between the proteins reported here and previously characterized ones is discussed.

    Notes: Summary The study was carried out in the first year on samples of random F5 lines, uniform in height and in heading date, of three crosses between semi dwarf spring wheat cultivars (Triticum aestivum L.), differing in grain weight and in their Rht gene. In the second year only the progenies of the early heading F5 lines were studied. All the material was grown in the absence of lodging. The culm-length genotypes of the different lines were identified by test crosses and by a seedling GA response test. No differences in grain weight were found between the two semi dwarf genotypes (Rht 1Rht1 rht 2rht2 and rht 1rht1 Rht 2Rht2). The tall genotype (rht 1rht1 rht 2rht2) was significantly higher in grain weight than the two semi dwarf genotyes and the grain weight of these genotypes exceeded markedly the grain weight of the dwarf genotype (Rht 1Rht1 Rht 2Rht2). These genotypic effects were independent of differences in plant height, heading date or number of grains per spike.

    Notes: Summary Ten pairs of reciprocal crosses have been made between wheat cultivars which show differences in their glutenin subunit compositions. The F1 seed glutenin subunit composition was studied by means of polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulphate (SDS). The results indicate that all the high molecular weight (HMW) and medium molecular weight (MMW) subunits (from 133,000 to 65,000 daltons) are transmitted to the F1 seed generation from the parental cultivars. In accordance with the triploid nature of the heterozygous endosperm (3n) and with the maternal and paternal gene dosage ratio (2∶1) in the endosperm itself, a significant effect of maternal parent is registered when comparing pairs of reciprocal seeds. Genes coding for the glutenin subunits are expressed whatever their doses are (one, two, or three) in the hybrid endosperm; thus the glutenin subunits inheritance is consistent with the co-dominant type. For one pair of the reciprocal crosses, two MMW parental bands (MW: 71,000 and 66,000) seemed absent in the F1 seed patterns while a new band with an intermediate, apparent MW (68,000) appears. This phenomenon was observed when the glutenins analyzed by electrophoresis were previously separated from other endosperm proteins, and not when they were directly extracted from the ground seed. We assume that the extraction can cause interactions between moieties attached to the subunits and lead to the formation of a complex having an intermediate electrophoretic mobility.

Group 5 chromosome substitution lines from three different cultivar backgrounds indicate that there may be multiple alleles for reduced crossability with rye (Secale cereale L.) and H. bulbosum on both 5A and 5B chromosomes. No reduction in seed set was observed from any of the 5D substitution lines. It was found that the Kr1 locus on chromosome 5B had a more pronounced affect on both rye and H. bulbosum crossability than the Kr2 locus on chromosome 5A and that the effects of both loci were cumulative. The Kr alleles affected both rye and H. bulbosum crossability in a linear, or additive, fashion. The Kr 2 allele was shown to have little effect on H. bulbosum crossability compared to a significant effect on rye crossability. Tests with tetrasomic and nulli-tetrasomic lines of ‘Chinese Spring’ indicated that the kr allele is ‘null’ or inactive in promoting crossability while the Kr allele is active in reducing crossability to both rye and H. bulbosum. Thus, extra doses of the kr allele do not increase rye or H. bulbosum crossability in the presence of the corresponding Kr allele.

    Notes: Summary An analysis of the α-amylase isozymes in GA-treated endosperm of wheat nullisomic-tetrasomics shows that there is more variation at the α-Amy-1 and α-Amy-2 homoeoallelic loci than was previously thought. Among the 16 isozymes produced by genes on the group 7 chromosomes, most could be definitely established as products of a single homoeoallele. Inter-varietal allelic differences would be expected at such loci and clear variation was found in isozymes produced by chromosomes 6B and 7B. The latter allele, α-Amy-B2b carried by the variety ‘Hope’, was used to locate the enzyme structural gene within chromosome 7B relative to the centromere and five other gene markers. The nature of the α-Amy-B2b phenotype and the rare non-parental isozyme patterns found among the recombinant lines indicates that the locus is large and compound, probably involving some degree of intra-locus gene duplication.

    Notes: Summary Gliadins, here defined as those proteins of defatted wheat endosperm which dissolve in 70% (v/v) ethanol at room temperature, were fractionated by gel filtration using Sephadex G-100. The protein which eluted with the void volume of the column, often described as high-molecular-weight (HMW) gliadin, was fractionated by the two different, two dimensional gel electrophoresis procedures of O'Farrell (1975) and O'Farrell et al. (1977). The next two fractions to elute from the gel column, ω-gliadin and α-, β-, γ-gliadin, were analysed similarly. The subunits of HMW gliadin and the classical (i.e. non-aggregated) gliadins map at distinctive positions on the electrophoregrams, the majority of the HMW gliadin subunits being more basic and having a slightly slower electrophoretic mobility than the α-, β-, γ-gliadins. These experiments demonstrate that those gliadins which aggregate to form HMW gliadin are distinct molecular entities and thus coded by different genes to those gliadins which do not aggregate. Glutenin, here prepared by a modification of the pH 6.4 precipitation procedure of Orth and Bushuk (1973), was also analysed by two-dimensional electrophoresis. The low-molecular-weight subunits were found to correspond exactly with the HMW gliadin subunits. Using the nullisomic-tetrasomic lines and the ditelocentric lines of ‘Chinese Spring’, the genes controlling the synthesis of all the major HMW gliadin subunits were shown to be located on the short arms of chromosomes 1A, 1B and 1D, as are the genes coding for the ω-gliadins and the majority of the γ-gliadins.

F1 grains (on maternal spikes) were very similar to the selfed grains on the maternal parent in protein percentage, weight and protein content. These traits were also analyzed in F3 grains developed on F2 spikes of segregating populations derived from reciprocal crosses between the same cultivated parent and another high-protein line of var. ‘dicoccoides’. No significant differences in the mean values of these traits were found between the reciprocal crosses, indicating no cytoplasmic effect. It has been concluded that these grain characteristics are largely determined by the maternal plant.

The population consisted of F2 bulk populations of 42 crosses among 11 genotypes adapted to S. W. Ontario. Heritabilities were: 0.30±0.32 for yield, 0.77±0.15 for height and 0.98±0.08 for test weight. Predicted genetic gain with 10% selection intensity was 0.15 t/ha for yield, 10.1 cm for height and 3.00 kg/hl for test weight. The low heritability for yield indicates that effective selection would require pedigree information and progeny tests, while the high heritabilities for height and test weight indicate that selection for these traits using single plots would be appropriate.

    Notes: Summary Poor adaptability or functional quality of much germplasm used for breeding high-protein hard red winter wheats prompted mutagenesis as an alternative means of increasing grain protein content. Four hard red winter wheat genotypes — KS644 (‘Triumph// Concho/Triumph’), ‘Kaw’, ‘Parker’, and ‘Shawnee’ — were treated with 0.40 M ethyl methanesulfonate (EMS). Advanced lines (M8-M10) were selected that had a 3-year mean grain protein advantage of 0.7% to 2.0% over controls. Increased grain protein content was generally associated with decreased grain yield and kernel weight, but some high-protein mutant lines had yields or kernel weights similar to those of original genotypes. Changes in height and lodging induced by EMS were generally favorable, most mutants being shorter and lodging less than controls, but blooming date was generally delayed, a deleterious change. One line also changed from resistant to segregating for wheat soil-borne mosaic virus. Mutant lines might be utilized in cross-breeding programs, particularly if negative pleiotropic effects and linkages are absent.

These cultivars are genetically related since they originate from the same stock. Moreover, the diversity of sample, containing cultivars with very different French bread-making qualities, makes it possible to investigate the relationship between glutenin subunit composition and bread-making quality. 16 electrophoretic types of glutenin subunits could be distinguished: these were grouped into four classes. Depending on the cultivar, six to eight glutenin subunits with MW more than or equal to 62,000 were detected. Subunits 3 and 5, with an approximate MW of 122,000 and 108,000 respectively, seem to play a prominent role on bread-making quality; they were found in cultivars of good quality and were absent in those unsuitable for making French bread. Two other subunits (9 and 10; MW: 71,000 and 66,000, respectively) have a less defined influence but may be needed in some types of glutenin structure. Aneuploid analysis shows that in ‘Chinese Spring’, subunit 5 is coded by a gene on the long arm of chromosome 1B. The location of genes coding for subunits 3, 9 and 10 could not be determined.

    Notes: Summary Genetic studies of frost resistance were performed on various wheat varieties using diallel, F2 monosomic and substitution analysis. A six-parental cross including reciprocals was carried out, and F1 hybrids and their parents were used for the freezing tests under controlled conditions. Both the general combining ability (GCA) and the specific combining ability (SCA) were significant, indicating additive and non-additive gene action in the inheritance of frost resistance. The high GCA∶SCA ratio revealed a preponderance of additive genetic variance. No significant reciprocal differences were found between the reciprocal crosses. The variance/covariance graphical analysis indicated the partial dominance of frost sensitivity. Frost sensitive varieties had the largest number of dominant genes, while frost resistant varieties had the highest proportion of recessive genes. The magnitude of the additive component of variation was higher than that of the dominance component, and the overall measure of the degree of dominance was smaller than one, so average dominance is incomplete. The increasing and decreasing alleles are not equally frequent at all loci. In this set of wheat varieties the values of narrow and broad heritability are relatively high. F2 monosomic analysis of the winter wheat variety ‘Arthur’ crossed with the monosomics of ‘Chinese Spring’ revealed that the average frost resistance of all the 21 monosomics was lower than that of the disomic. F2 monosomic hybrids 5A, 2B, 4B and 5D proved to be relatively frost resistant, while monosomics 3A, 3B and 6D were the most sensitive. The control of frost resistance in the set of chromosome substitution lines of the variety ‘Cheyenne’ into ‘Chinese Spring’ (with the exception of 2B) indicated that the genes responsible for the frost resistance of ‘Cheyenne’ are localised in chromosomes 5A, 7A, 4B, 5B, 4D and 5D. The genetic basis of frost resistance and problems of analysis are discussed.

Here, principal component analysis has been used to examine the patterns of variability created by those socio-economic factors which may have obscured any underlying relationship which existed between yield and climate. In fact, these analyses revealed the existence of variation peculiar to particular groups of years, a result which could explain why many attempts to relate crop yields directly to climatic factors have proved unsuccessful.     Notes: Summary The major endosperm proteins in a range of genotypes of hexaploid wheat have been fractionated by two-dimensional electrophoresis. The genotypes included nine varieties and forty four intervarietal substitution lines in which chromosomes 1A, 1B, 1D, 6A, 6B or 6D from eight of the varieties have been introduced one at a time into a common genetic background. The appearance of different protein subunits was often correlated with a chromosome substitution. This showed that many of the genes for the high molecular weight protein subunits (molecular weight range 55,000 to 140,000 determined by SDS polyacrylamide gel electrophoresis) are specified by chromosomes 1A, 1B and 1D while many of the lower molecular weight subunits (molecular weight range 30,000 to 45,000) are specified by chromosomes 6A, 6B and 6D. The different protein subunits correlated with chromosome substitution could not always be recognised in the varietal source of the substituted chromosome. The different subunits specified by homologous chromosomes in different wheat varieties may differ in isoelectric point and/or molecular weight.

    Notes: Summary The distribution of three biochemical markers, U-1, CM-4 and Aphv-a, -b, among wheat-Aegilops addition lines carrying Mv chromosomes from Aegilops ventricosa (genomes DvMv) has been investigated. Addition lines which had been previously grouped together on the basis of common non-biochemical characters carried marker U-1, a protein component from the 2M urea extract. The added chromosome, in the appropriate genetic background, seems to confer a high level of resistance to the eyespot disease, caused by the fungus Cercosporella herpotrichoides. The other two markers were concomitantly associated with another similarly formed group of addition lines. Both CM-4, a protein component from the chloroform:methanol extract, and Aphv-a, -b, alkaline phosphate isozymes, have been previously shown to be associated with homoeologous chromosome group 4, which suggests that the added chromosome in the second group of addition lines is 4Mv.

Among the ‘Norin’ gene carriers, dwarf group (D2) was most efficient, followed by very dwarf (D3) and semi-dwarf (D1). Photosynthetic rate and respiratory rate were found to have a positive relationship.

One such clone, pTag 1290, has enabled us to identify the HMW glutenin mRNA species. The DNA sequence of this clone has been partially determined and it contains several tandem DNA repeats. The sequence is discussed in relation to the generation of the HMW glutenin subunit gene family. Analysis of the organization of the HMW glutenin sequences in the wheat genome revealed that the genes encoding HMW glutenin subunits exist in low copy number and are located on the long arm of each of the homoeologous group 1 chromosomes.

    Notes: Summary Studies were conducted to determine the influence of the male sterility-inducing cytoplasm of Triticum timopheevii (Zhuk.) Zhuk. on response of several common winter wheat (T. aestivum L.) nuclear genotypes to photoperiod and vernalization. Comparative studies of cytoplasmic substitution lines provide information on the role of the cytoplasmic genetic mechanism in growth and development. In the case of cytoplasmic male sterility-based hybrid production systems, ubiquity of sterility-inducing cytoplasm in derived hybrids warrants thorough characterization of its influence on plant phenotype. Factorial combinations of cytoplasm (T. timopheevii and T. aestivum), nuclear genotype, and photoperiod or vernalization treatments were evaluated under hydroponic conditions in controlled environment chambers. Interaction of cytoplasm, photoperiod, and nuclear genotype was significant in one or more experiments for days to anthesis and potential spikelet number, and interaction of cytoplasm, vernalization, and nuclear genotype was significant for days to spike emergence. Long day length was associated with increased percentage seed set in one study, but interactions of photoperiod and cytoplasm were not detected for percentage seed set. Interactions involving cytoplasm and photoperiod or vernalization were interpreted as evidence of the existence of genetic factors in cytoplsam of T. timopheevii which alter photoperiod or vernalization responses of alloplasmic plants relative to responses exhibited by euplasmic plants. Since photoperiod and vernalization responses are critical to adaptation, T. timopheevii cytoplasm can alter adaptability of T. aestivum. The specific effect would be nuclear genotype dependent, and does not appear to be of a magnitude greater than that induced by nuclear genetic variability at loci conditioning photoperiod or vernalization responses or other adaptation-determining characteristics. Normal multilocation/year testing of alloplasmic hybrids should therefore adequately identify zones of adaptation.

    Notes: Summary Using in situ hybridization techniques, we have been able to identify the translocated chromosomes resulting from whole arm interchanges between homoeologous chromosomes of wheat and rye. This was possible because radioactive probes are available which recognize specific sites of highly repeated sequence DNA in either rye or wheat chromosomes. The translocated chromosomes analysed in detail were found in plants from a breeding programme designed to substitute chromosome 2R of rye into commercial wheat cultivars. The distribution of rye highly repeated DNA sequences showed modified chromosomes in which (a) most of the telomeric heterochromatin of the short arm and (b) all of the telomeric heterochromatin of the long arm, had disappeared. Subsequent analyses of these chromosomes assaying for wheat highly repeated DNA sequences showed that in type (a), the entire short arm of 2R had been replaced by the short arm of wheat chromosome 2B and in (b), the long arm of 2R had been replaced by the long arm of 2B. The use of these probes has also allowed us to show that rye heterochromatin has little effect on the pairing of the translocated wheat arm to its wheat homologue during meiosis. We have also characterized the chromosomes resulting from a 1B-1R translocation event. From these results, we suggest that the observed loss of telomeric heterochromatin from rye chromosomes in wheat is commonly due to wheat-rye chromosome translocations.

The weights of the F1 grains (on maternal spikes), from intact as well as from defoliated plants, and those of the F2 grains (on f1 spikes), were examined. Grain weight was controlled primarily by the genotype of the maternal tissues (pericarp, testa or other floret or spikelet organs, including the vascular system), with complete dominance of heaviness. No indications suggesting maternal inheritance were obtained. The frequency distribution of the weights of the F2 grains indicated the presence of genotypic effects exerted by the endosperm or embryo. The embryo or endosperm factors for heaviness also seemed to be dominant.

    Notes: Summary The seed proteins of ‘Chinese Spring’ wheat stocks which possess single chromosomes from other plant species related to wheat have been separated by gel electrophoresis in the presence of sodium dodecyl sulphate. Marker protein bands have been detected for both arms of barley chromosome 5, chromosome E (= 1R) and B (= 2R) of rye, chromosomes A,B (= 1Cu) and C (= 5Cu) of Aegilops umbellulata and chromosomes I and III of Agropyron elongatum. These studies, and previous findings, indicate that chromosome 5 of barley, chromosome 1R of rye, chromosome I of Ag. elongatum and possibly chromosome 1Cu of Ae. umbellulata are similar to chromosomes 1A, 1B and 1D in hexaploid wheat in that they carry genes controlling prolamins on their short arms and genes controlling high-molecular-weight (apparent molecular weight greater than 86,000) seed protein species on their long arms. These findings support the idea that all these chromosomes are derived from a common ancestral chromosome and that they have maintained their integrity since their derivation from that ancestral chromosome.

Sr 24 (‘Sear's 3D/Ag’), Sr 24 (TR 380-27 4/3 Ag 14-White seeded recombinant with ‘Agent’ type resistance), Sr 25 (‘Sear's 7D/Ag’), Sr 26 (‘Eagle’), Sr 26 (Knott's 6A/Ag translocation), Sr 27 (WRT 238-5), Combination line (Sr Tt1 + Sr 9b) were observed to be completely effective against all the 19 cultures tested. In addition, a number of lines, such as TAF2d (Sr Agi), Line W(Sr Tt2) and Combination III (Sr Tt1 + Sr 9e), were found to be effective against at least three of the most prevalent races (21, 40A and 117A-1) and a virulent race 122 in Indian natural population. Lines carrying genes other than Sr 2, Sr 9a, Sr 9f (‘Chinese Spring’) and Sr 15 (‘Norka’), and Line E were found to be resistant to one or more cultures of stem rust. The background effect upon the expression of a gene was observed by comparing the range of infection on single gene host lines in either different backgrounds and/or in cultivars with known genes for stem rust resistance against the 12 cultures of stem rust races found in India.

The inheritance of resistance to each race has been determined from the F1 and F2 of the crosses (resistant parents with the susceptible variety, ‘Agra Local’) and F2 progenies from the backcross to ‘Agra Local’. Tests have been performed to see if the two varieties carry common genes/s for resistance. The identity of the genes for resistance has been established from relevant crosses with single gene lines carrying known genes for resistance. A single dominant gene effective to race 15C in E5008 has been demonstrated to be Sr9b. Of the two recessive genes, each producing distinct infection types (0; and 1–3) against race 122, one gene has been inferred to be Sr12 and the second to be a hitherto undesignated gene. The resistance of E6160 against race 15C is controlled by two genes, one dominant and one recessive. The dominant gene has been identified as Sr9b. The recessive gene has been inferred to be a new gene. Similarly, a dominant gene effective against race 122 in E6160 has been observed to be different from those so far designated. In addition, the presence of modifier gene/s in the variety, E6160 has been suggested.

    Notes: Summary The inheritance of the high-molecular-weight (HMW) glutenin protein subunits in hexaploid wheat has been investigated by using sodium dodecyl sulphate-polyacrylamide gel electrophoresis to examine the segregation of these subunits in 496 test-cross seeds. The parents of the f1 hybrid were chosen so that the test-cross seeds segregated for all the HMW glutenin bands. Two glutenin subunits from one parent, believed to be controlled by genes on chromosome 1D, segregated as alternatives to two glutenin subunits from the other parent, a result that supports the assumption that these subunits are controlled by allelic genes at each of two loci that are very closely linked. Similar results were obtained for glutenin subunits believed to be controlled by chromosome IB, which suggests that these subunits are controlled also by allelic genes at each of two loci that are very closely linked. A single glutenin subunit band, believed to be controlled by chromosome 1A, segregated as an alternative to a single glutenin band from the other parent, except that one seed did not possess either band. It was concluded that these bands are controlled either by allelic genes or by nonallelic genes that are very closely linked.

Only ADH-1 is expressed in developing seeds, dry seeds, pollen and germinating seedlings. ADH-2 can be induced in seedling roots or shoots under conditions of partial anaerobiosis or by certain chemicals. Expression of ADH-1 and ADH-2 isoenzymes was investigated in undifferentiated calli from aestivum and durum wheats, rye, triticale and also in in vitro regenerated roots and leaves from aestivum cultures. Wheat callus cultures originating from seed, mature and immature embryos, mesocotyl and root, as well as cultures grown on media containing different supplements did not show any variation in the overall expression of ADH-1 or ADH-2, although differences in the band intensities were observed. The callus isoenzyme pattern was similar to that observed in roots under anaerobic conditions. Both ADH-1 and ADH-2 were expressed in in vitro regenerated roots but were absent in regenerated leaves. Expression of ADH-1 and ADH-2 in wheat calli seems to be related to the type of differentiation.

    Notes: Summary Genetical analysis of the F2 triple test cross design combined with conventional early generations was used to elucidate the genetical control of yield and yield components in two crosses of winter wheat. From estimates of the additive, {d}, and additive X additive, {i}, components of means, together with the additive genetical variance, D, predicted frequencies of recombinant inbred lines that would transgress the parental range were calculated for each cross. The accuracy of predictions was evaluated by comparing expected frequencies with observed numbers in populations of F6 lines previously developed by single seed descent. For both crosses and all characters where an adequate genetical model was found to explain the observed variation between the early generations, good agreement between predicted and observed frequencies of transgressive segregants was obtained. Furthermore, for characters exhibiting significant epistasis, allowance for additive X additive {i} epistasis in the prediction equations was sufficient to allow for skewness of the recombinant inbred population. These results demonstrate that cross performance in wheat can be predicted from genetical analysis of early generations, and the value of this approach in breeding new varieties is discussed.

    Notes: Summary The 6-phosphogluconate dehydrogenase (6-PGD) zymogram phenotypes of wheat, rye and their aneuploid derivatives were determined. Two genes involved in the production of 6-PGD isozymes were located on chromosome arms CRL (4 RL) and FRL (6 RL) of “Imperial” rye. On the basis of differential interactions between wheat and rye chromosomes, evidence was obtained that genes located on chromosomes 6 A, 6 BL and 7 BL control 6-PGD isozyme activities in ‘Chinese Spring’ wheat. The wheat and rye 6-PGD zymogram phenotypes were indicative of homoeologous relationships between rye chromosome 6 RL to wheat chromosomes of group 6, and rye chromosome 4 RL to wheat chromosomes of group 7.

    Notes: Summary Homologous high molecular weight storage prolamins were purified from grain of wheat, rye and barley using combinations of gel filtration, ion-exchange chromatography and preparative isoelectric focusing. Sodium dodecylsulphate polyacrylamide gel electrophoresis showed that the components were single bands with apparent mol.wts. of above 100,000. Molecular weights determined by sedimentation equilibrium ultracentrifugation were considerably lower; 54,700, 67,600 and 69,600 for the components from barley, rye and wheat respectively. Amino acid analysis showed the presence of 13.6 to 16.5 mol% glycine, 29.6 to 34.0 mol% glutamate + glutamine, 11.4 to 13.7 mol% proline and a total of 4.0 to 5.7 mol% basic amino acids. Automated N-terminal amino acid sequencing of the component from wheat showed the presence of cysteine residues at positions 5 and 10, and this is discussed in relation to the possible role of these proteins in the visco-elastic gluten network.

    Notes: Summary The wheat rDNA clone pTA250 was examined in detail to provide a restriction enzyme map and the nucleotide sequence of two of the eleven, 130 bp repeating units found within the spacer region. The 130 bp units showed some sequence heterogeneity. The sequence difference between the two 130 bp units analysed (130.6 and 130.8) was at 7 positions and could be detected as a 4 °C shift in Tm when heterologous and homologous hybrids were compared. This corresponded to a 1.2% change in nucleotide sequence per ΔTm of 1 °C. The sensitivity of the Tm analysis using cloned sequences facilitated the analysis of small sequence variations in the spacer region of different Triticum aestivum cultivars and natural populations of T. turgidum ssp. dicoccoides (referred to as T. dicoccoides). In addition spacer length variation was assayed by restriction enzyme digestion and hybridization with spacer sequence probes. Extensive polymorphism was observed for the spacer region in various cultivars of T. aestivum, although within each cultivar the rDNA clusters were homogeneous and could be assigned to particular chromosomes. Within natural populations of T. dicoccoides polymorphism was also observed but, once again, within any one individual the rDNA clusters appeared to be homogeneous. The polymorphism, at the sequence level (assayed by Tm analysis), was not so great as to prevent the use of spacer sequence variation as a probe for evolutionary relationships. The length variation as assayed by restriction enzyme digestion did not appear to be as useful in this regard, since its range of variation was extensive even within populations of a species.

    Notes: Summary The nature of genome change during polyploid evolution was studied by analysing selected species within the tribe Triticeae. The levels of genome changes examined included structural alterations (translocations, inversions), heterochromatinization, and nucleotide sequence change in the rDNA regions. These analyses provided data for evaluating models of genome evolution in polyploids in the genus Triticum, postulated on the basis of chromosome pairing at metaphase I in interspecies hybrids. The significance of structural chromosome alterations with respect to reduced MI chromosome pairing in interspecific hybrids was assayed by determining the incidence of heterozygosity for translocations and paracentric inversions in the A and B genomes of T. timopheevii ssp. araraticum (referred to as T. araraticum) represented by two lines, 1760 and 2541, and T. aestivum cv. Chinese Spring. Line 1760 differed from Chinese Spring by translocations in chromosomes 1A, 3A, 4A, 6A, 7A, 3B, 4B, 7B and possibly 2B. Line 2541 differed from Chinese Spring by translocations in chromosomes 3A, 6A, 6B and possibly 2B. Line 1760 also differed from Chinese Spring by paracentric inversions in arms 1AL and 4AL whereas line 2541 differed by inversions in 1BL and 4AL (not all chromosomes arms were assayed). The incidence of structural changes in the A and B genomes did not coincide with the more extensive differentiation of the B genomes relative to the A genomes as reflected by chromosome pairing studies. To assay changing degrees of heterochromatinization among species of the genus Triticum, all the diploid and polyploid species were C-banded. No general agreement was observed between the amount of heterochromatin and the ability of the respective chromosomes to pair with chromosomes of the ancestral species. Marked changes in the amount of heterochromatin were found to have occurred during the evolution of some of the polyploids. The analysis of the rDNA region provided evidence for rapid “fixation” of new repeated sequences at two levels, namely, among the 130 bp repeated sequences of the spacer and at the level of the repeated arrays of the 9 kb rDNA units. These occurred both within a given rDNA region and between rDNA regions on nonhomologous chromosomes. The levels of change in the rDNA regions provided good precedent for expecting extensive nucleotide sequence changes associated with differentiation of Triticum genomes and these processes are argued to be the principal cause of genome differentiation as revealed by chromosome pairing studies.

    Notes: Summary This paper deals with the relationship of chloroplast DNA (ctDNA) and cytoplasmic malesterility (CMS) of maize, wheat and rape. Intramolecular heterogeneity and fragment patterns of DNA digested with restriction endonucleases were compared and analyzed. For this purpose methods of agarose gel electrophoresis and two dimensional gel electrophoresis with denaturating solvent concentration gradients were applied. 1) Heat denaturation of ctDNA from a male sterile line in maize yielded a profile with three melting regions. This base sequence heterogeneity was not observed for the ctDNA of its maintainer. 2) Maize ctDNA was digested with EcoR1 and BamH1 restriction endonucleases, wheat and rape ctDNA with EcoR1 only. No significant differences were observed in the comparison of the fragment patterns from the sterile lines and their maintainers with the exception of the sterile line in rape which lacked one fragment present in its maintainer. 3) Two dimensional electrophoresis using a gradient of denaturing agents revealed several differences in number and relative positions of the separated restriction endonuclease fragments between the sterile lines and their respective maintainers in all three tested crops. This raises the possibility that changes in ctDNA may be involved in CMS.

Die beiden Endformationen sind mit ein und derselben Nervenfaser der Gruppe II verbunden und daher als Bestandteile einer sekundären sensorischen Endigung zu betrachten. Die Nervenendformation an einer der beiden „nuclear chain“-Fasern hat anulo-spirale Form. Die Nervenendformation an der anderen „nuclear chain“-Faser weist am Querschnittsbild eine Anzahl von Axonen auf, die zwischen Plasmalemm und Basalmembran der intrafusalen Muskelfaser eng aneinanderliegen. Nicht alle Axonquerschnitte stehen in direktem Kontakt mit der intrafusalen Faser. Das terminale Axon scheint sieh nach Eintritt unter die Basalmembran der intrafusalen Faser mehrfach in relativ dünne Äste unregelmäßigen Verlaufs zu teilen. Diese Form der Endigung könnte ein Korrelat der sog. „flower spray“-Endigung im Sinne Ruffinis (1898) darstellen. Die beiden morphologisch unterschiedlichen Endformationen innerhalb der sekundären Endigung gleichen einander und den Endformationen der primären Endigung bezüglich folgender Ultrastrukturmerkmale: 1. Es besteht synaptischer Kontakt zwischen Axon und intrafusaler Muskelfaser (synaptischer Spalt durchschnittlich 200 Å) ohne Zwischenlagerung von Basalmembranmaterial; 2. die terminalen Axonabschnitte liegen direkt unter der Basalmembran der intrafusalen Muskelfaser und sind nicht von Schwannschen Zellen bedeckt; 3. Mangel an synaptischen Bläschen; 4. desmosomenartige Verhaftungen zwischen Zellmembranen von Axon und intrafusaler Faser; 5. dyadenartige Anlagerungen des sarkoplasmatischen Retikulums an die Zellmembran der intrafusalen Faser im Bereich des synaptischen Spaltes. Nach unseren derzeitigen Vorstellungen sprechen diese Ultrastrukturmerkmale für eine rezeptorische Natur der beschriebenen Nervenendigungen.
    Notes: Summary In a rat muscle spindle transversally cut into a series of alternating semithin and ultrathin sections, two different forms of nerve terminations were found on two neighbouring nuclear chain fibres. The two nerve terminations were connected to the same group II nerve fibre and are consequently constituents of one particular secondary sensory ending. The nerve termination on one of the two nuclear chain fibres is of the anulo-spiral type. The nerve termination on the second nuclear chain fibre shows a number of axons lying closely together between plasma membrane and basement membrane of the intrafusal muscle fibre. Not all of these axons are in direct contact with the intrafusal fibre. The terminating nerve fibre seems to be divided into several branches of rather small diameters and irregular courses. It is suggested that this kind of termination could be a correlate of the so-called “flower spray” type of sensory endings in muscle spindles. The two morphologically different nerve terminations in the secondary ending have the following ultrastructural characteristics in common with those of the primary ending: 1) Synaptic contact between axon and intrafusal muscle fibre (synaptic gap about 200 Å) without interposition of basement membrane material; 2) terminal axons located beneath the basement membrane layer of intrafusal muscle fibres without covering by Schwann cells; 3) lack of synaptic vesicles; 4) desmosome-like structures between plasma membranes of axon and intrafusal muscle fibre, and 5) dyads of the sarcoplasmic reticulum adjacent to the synaptic cleft. According to present knowledge these features indicate that all of these endings are sensory ones.

    Notes: Summary The innervation of the adrenal cortex of the rat and the pig is investigated with the electron microscope. Nerve fibers containing synaptic and two types of dense-cored vesicles come into contact with endocrine cells. There are no specialized pre- and postsynaptic membranes. The synaptic cleft is about 200 Å wide. Generally the basement membrane between nerve and cell is absent. These observations are discussed on the base of more recent experimental findings. Small fibers having an average diameter of about 0.2 to 0.5 μ and containing only tubules and filaments are considered to represent parts of an afferent nervous system.

    Notes: Summary The adrenergic innervation in the submaxillary gland, heart, kidney, small intestine, and accessory male genital organs and the development of the adrenal chromaffin cells and the sympathetic ganglia were studied in the rat from 15 days post coitum to 16 days post partum using the fluorescence histochemical method of Falck and Hillarp. The postnatal development of the noradrenaline concentrations in the heart and vas deferens was followed by fluorometric determinations. At about 15 days post coitum, the anlagen of the sympathetic chains were well visible in the form of two dorsal segmented columns of small branching sympathicoblasts exhibiting an intense catecholamine fluorescence. In the midline, ventrally to these two anlagen, another column of sympathicoblasts developed; this seemed to give rise to the prevertebral ganglia and to the short adrenergic neurons supplying the internal genital organs. At the level of the adrenal anlagen, small intensely fluorescent chromaffin cells were collected in two bilateral groups which became enclosed by adreno-cortical cells. This enclosure was, however, not complete even at two weeks post partum. Bundles of growing sympathetic nerves were visible in the periphery of the various organs studied at 19–21 days post coitum. A terminal innervation of the organs suggestive of a functional transmitter mechanism did not start to establish until at or immediately after birth. The final pattern of innervation was usually reached at about one week post partum, and the following development proceeded largely in the form of a quantitative increase in the number of nerves participating in the innervation apparatus. The adult level of noradrenaline in the heart and vas deferens was reached three to five weeks after birth. The small intestine was an exception in that the final pattern of innervation in the wall was attained immediately after birth. There was no overt difference in the rate of development of the terminal sympathetic innervation in organs supplied by short adrenergic neurons (accessory male genital organs) compared to the innervation of the submaxillary gland, heart and kidney, which receive classical long adrenergic neurons.

    Notes: Summary The granulosa cells of the ovarian follicle of the rat and the domestic fowl have been studied with the light and electron microscope. The nuclei of the granulosa cells were irregular with indentations and large in proportion to the cytoplasm of the cell. The mitochondria had a dense, dark matrix with only few cristac. The Golgi apparatus was moderately developed, located towards the oocyte in a juxtanuclear position. The endoplasmic reticulum was rather sparse. Lipid droplets were only occasionally encountered. Microtubules were regularly observed. The functions of the granulosa cells are discussed. Compared with the steroid-producing cells of the theca interna of the same follicles, the granulosa cells primarily are the nursing cells for the growing oocyte and mainly have the characteristics of protein forming cells.

    Notes: Summary The ultrastructural features of the paraventricular neurones of the non-treated rat are presented comparing them with those of the supraoptic neurones. No striking differences are seen between the general electron microscopic characteristics of the paraventricular and supraoptic neurones. The importance of adequate fixation to obtain good preservation of the neurones is emphasized, since inadequate fixation can cause e.g. artefactual appearance of “dark neurones”. The previously presented classification of the neurosecretory neurones into two categories (e.g. “light and dark neurones”) on the basis of the number of ribosomes is not considered justifiable, since their number can vary to a very great extent even within a single cell. The synthesis of neurosecretory products in the paraventricular neurones obviously follows the general mode of the synthesis of secretory proteins: ribosomes—RER—Golgi complex—secretory vesicles. On the basis of the localization of heavy metal deposits after osmium impregnation and demonstration of acid phosphatase the Golgi complex of the paraventricular neurones is found to be polarized. The direction of the polarity is discussed. The substructures of the dense cores of the neurosecretory granules and of the contents of the lysosomal dense bodies are nearly identical. Therefore it is considered impossible to determine positively the nature of the dark condensed material within the Golgi complex. The characteristics of the immature neurosecretory granules and the possibility of releasing neurosecretory products into the cytoplasm already within the perikarya are speculated.

    Notes: Summary The structure of glycerinated smooth muscle from small intestine of adult rat was investigated by electron microscopy. In the central parts of the tissue blocks a two-filament system was found, consisting of parallel thick and thin filaments with regularly spaced interconnections, closely resembling that of striated muscle. In the peripheral parts of the blocks only thin filaments were found. The thin filaments were identified as actin by the formation of arrowhead complexes after incubation with heavy meromyosin.

    Notes: Summary The localization and configurations of ribosomes in mature white skeletal muscle fibers of the rat were investigated. Differential visualization of ribosomes and glycogen granules was obtained by fixation with glutaraldehyde only and staining of the sections with uranyl acetate. Ribosomes are then electron dense and glycogen granules electron transparent. Their identity was ascertained by selective extractions of ribonucleic acid and polysaccharide. The vast majority of the ribosomes is not membrane-bound. They are located intermyofibrillarly (predominantly at the level of the I-bands), beneath the sarcolemma, and in the paranuclear cones of sarcoplasm. Occasionally short stretches of granular reticulum occur, often as characteristic double walled vesicles with ribosomes on the inner membrane only. Three main types of polysomal configurations are observed: rosettes of 4 to 6 ribosomes, helical arrays, and whorls of up to about 25 probably membrane-bound ribosomes. The average number of ribosomes in the extended helical configurations is estimated to be about 60. It is argued that these helices represent the polysomes instrumental in the synthesis of the large subunits of myosin. It is emphasized that helical polyribosomes are by no means typical of striated muscle, but probably represent a common configuration of large free polysomes.

Nach Immersionsfixation mit Glutaraldehyd oder Osmiumtetroxyd finden sich in der Nebenniere sog. Mischzellen, helle Zellen, syncytiale oder plasmodiale Zellen, die von den Untersuchern als Artefaktbildungen angesehen werden. In allen Proben nach Perfusionsfixation wurden solche Artefaktbildungen niemals beobachtet. Bei Immersionsfixation wurde eine enge Beziehung zwischen Sekretgranula und Cytoplasmamembran beobachtet, was bei Perfusionsfixation selten der Fall war. Die heutigen Theorien über den Sekretionsmechanismus bei Nebennierenmarkzellen werden aufgrund der vorgelegten Ergebnisse diskutiert. Die Arbeit zeigt die wesentlichen Vorteile der Anwendung der Perfusionsfixation für die Untersuchung der Nebenniere.
    Notes: Summary The adrenal medulla of the rat was studied utilizing various methods of fixation. In adrenal medulla specimens after immersion fixation either with glutaraldehyde or osmium tetroxide, elements such as mixed, clear, syncytial, or plasmodial cells, believed to be of artifactual origin, are observed in all of this material examined. These elements are absent in the specimens prepared by perfusion fixation. In specimens prepared by immersion fixation, secretory granules are found in close proximity to the plasma membrane; this localization is infrequent after perfusion fixation. Current theories of the mechanism of secretion of adrenal medullary hormones are discussed on the basis of our results. This investigation demonstrates the advantage and necessity of perfusion fixation in the study of the adrenal medulla.

    Notes: Summary In the enamel organ of rat incisors macrophages are present in the zone of matrix formation, the transitional zone, the enamel maturation and pigmentation zone. The macrophages accumulate adjacent to redifferentiating amelocytes in the transitional zone. The macrophages phagocytize fragments of disintegrating amelocytes. In addition to the well known complement of organelles the macrophages present an elaborated microtubular system, scattered, thick filaments, a cortical feltwork of thin filaments, and spherical nuclear bodies. The microtubules emanate from “attached” and free pericentriolar satellites and radiate aster-like towards the cell surface or into pseudopods or curve along the nuclear surface for long distances, often related to nuclear constrictions. It is suggested that the microtubular system plays a prominent role in directional movement of the macrophages. The cortical filaments, if contractile, may create the cytoplasmic flow necessary for the cell motility.

    Notes: Summary The olfactory tubercle of the rat was studied by electron microscopy both in the normal and after ipsilateral olfactory bulb ablation at survival times of from 14 hours to seven days. Particular emphasis was placed on synaptic structures and their changes following the lesion. Normal synapses are similar to those described in previous studies and presynaptic profiles are of at least three types. Types-A and -B contain round vesicles and form asymmetrical contacts and type-C profiles contain flattened vesicles and form symmetrical contacts. There appear to be two major types of degenerative changes. The electron-lucent type predominates at early survival times and is seen first at 14 hours. These profiles show an early reduction in numbers of vesicles with mitochondrial swelling followed by shrinkage of the profile. These profiles become increasingly electron-dense at later survival times. The second major type of degenerating profile is initially electron-dense. The earliest changes in these profiles are an increased axoplasmic density and increased microtubular density and clumping without apparent loss of vesicles. These profiles also become progressively more electron-dense at longer survival times. The observations are discussed in relation to previous reports.

    Notes: Summary The pattern of intercellular cytoplasmic bridges between rat spermatogonia and between spermatocytes is illustrated from electron microscopy of serial sections. Clones, or syncytia, containing as many as 22 connected spermatogonia and as many as 74 connected spermatocytes were observed. The absence of closed rings of cells agrees with the observation that intercellular bridges are the result of incomplete cell division, rather than cell fusion. The bridges thus are a record of spermatogonial divisions within a clone. In early spermatogonial generations there is a predominantly linear arrangement. The groups of spermatocytes have more side branches. From the presence of synaptonemal complexes it is concluded that the connected spermatocytes of a given clone are in about the same developmental stage. The pattern of intercellular bridges indicates, however, that not all nuclei in a clone undergo mitosis in the same cycle. The connected cells of a clone are therefore not all of the same generation. From unconnected bridges it is assumed that new clones originate from single cells or groups of spermatogonia which separate from an existing clone.

    Notes: Summary Isolated pieces of seminiferous tubules of adult rats were grown in organ culture for up to 8 weeks in Petri dishes on the surface of nutrient agar. The medium consisted of newborn calf serum, Eagle's minimum essential medium, glutamate and antibiotics. This method allowed observation of the contractions of the seminiferous tubules in the culture. Contractility, light and electron microscopic structure and histochemically demonstrable activities of alkaline phosphatase and ATPase of the tubule walls were studied at 1-week intervals. The contractility and alkaline phosphatase activity were maintained in the tubule wall for 3 weeks, and the activity of ATPase was maintained for 5 weeks. The thin filaments of the myoid cells, which are responsible for the contractility, were seen with the electron microscope in tubules cultured for 5 weeks. The organ culture method described in the present paper seems to be valuable for studies concerning the functioning of the myoid cells of the seminiferous tubules and the possibility that this is regulated by hormones.

    Notes: Summary Exocytosis has been demonstrated by electron microscopy in the external zone of the median eminence of the rat. Exocytotic profiles have been observed in nerve fibres characterized by the presence of granular vesicles with median diameters of 90–103 nm and agranular vesicles of about 50 nm. In addition to the small agranular vesicles, coated vesicles of the same size have been found in many nerve fibres, suggesting that at least part of the agranular vesicles in the median eminence originate by micro-pinocytosis. The nature of the fibres showing exocytosis is discussed. Attention is drawn to the possibility of identifying types of fibres in the median eminence by the occurrence of exocytosis.

    Notes: Summary The general ultrastructural features of the hypothalamo-neurohypophysial system in rats with hereditary hypothalamic diabetes insipidus (DI-rats, Brattleboro strain) are described. There is no decisively distinguishing difference between the neurons of the supraoptic and paraventricular nuclei. The neurons of both nuclei show signs of active protein synthesis. The perikarya of the neurons are markedly hypertrophic, the nuclei are large and the nucleoli prominent. In the cytoplasm there are numerous ribosomes, abundant rough-surfaced endoplasmic reticulum and extensive Golgi complexes. However, very few neurosecretory granules are to be seen. The axons of the hypothalamo-neurohypophysial tract are likewise enlarged and the paucity of neurosecretory granules is a striking feature also in the area of the tract. The majority of nerve endings in the posterior pituitary of DI-rats are devoid of neurosecretory granules. Microvesicles are abundant in the nerve endings and there are findings which suggest that microvesicles are involved either in endoor exocytosis. The signs of active protein synthesis and the concomitant paucity of neurosecretory granules are interpreted to imply transportation of the secretory proteins in an extragranular phase. The possible mode of release of the secretory proteins from the nerve endings and the role of microvesicles therein are discussed.

    Notes: Summary The purpose of this study is to investigate the ultrastructural features of perivascular cells as found in the neural lobe of the hypophysis. In particular, an inquiry was made into the nature of the relationship of such cells to neurosecretory fibers and endings. The latter, in fact, are often invaginated within the perivascular cells and enveloped by their processes; furthermore, they often reveal a certain number of empty granules as well as characteristics of degenerative nature. In the course of this study the localization of the perivascular cells has been investigated as well as that of their processes within the extensive interlobular network typical of the hypophysial neural lobe of rodents. Based on the data gathered, the hypothesis is put forward that the perivascular cells play an important role in the turnover of neurosecretory endings, both under physiological and experimental conditions, contributing thereby to the release of post-hypophysial hormones.

Die Gefäßfortsätze von ependymalen und subependymalen Zellen bilden in der subependymalen Zone und in der Zona interna des Ratteninfundibulum mit Nervenfasern kompakte neuro-gliöse Faserbündel, die sich in der Zona externa aufzweigen. 2. In allen Zonen des Infundibulum kommen zwischen den Nervenfasern und den Gefäßfortsätzen zahlreiche neuro-gliöse Synapsen vor. In den Gefäßfortsätzen fällt die hohe Zahl an Mikrotubuli sowie die zahlreichen, vielgestaltigen Einschlüsse auf. 3. In der subependymalen Zone sind die Interzellularspalten an bestimmten Stellen außerordentlich weit. Sie haben eine kanalartige Beschaffenheit und enthalten feine Bündel von Nervenfasern. 4. Von den lateralen Anteilen des Infundibulum her erreichen Dendriten von Ganglienzellen des Nucleus infundibularis die Mitte des Infundibulum. In dieser Region sind axodendritische Synapsen anzutreffen. 5. Morphometrische Analysen der Nervenfaserendigungen der Zona externa von Normaltieren zeigen, daß die prozentuale Verteilung der nach Granulagröße differenzierten Nervenfaserklassen für Mitte und Seite der Zona externa etwa gleich ist. Zwischen der Größe der Elementargranula und der Anschnittfläche der zugehörigen Nervenfasern besteht eine direkte Beziehung. 6. Die Nervenfaserendigungen erreichen die Basalmembran des perikapillären Raumes fast ausschließlich im Bereich von gefäßwärts gerichteten Vorwölbungen der Zona externa. Das Ausmaß, in dem Nervenfasern im Vergleich zu den Gefäßfortsätzen von Ependymund Gliazellen den perivaskulären Raum erreichen, ist medial weitaus größer als lateral. 7. Bei bilateral adrenalektomierten Ratten nimmt in bestimmten, vorwiegend lateral gelegenen Nervenfasern die Zahl und Größe der Elementargranula in Abhängigkeit von der Überlebensdauer zu. Dies dürfte auf eine verstärkte Synthese und Speicherung von Corticotropin-Releasing Factor in diesen Nervenfasern zurückzuführen sein. Gegenüber dem Normalbefund ist die neurohämale Kontaktfläche erheblich vergrößert. Der perivaskuläre Raum enthält zerfallene Nervenfaserteile, die durch Bindegewebszellen phagocytiert werden. Diese Veränderungen dürften durch eine unter Versuchsbedingungen verstärkte Wachstumstendenz der Nervenfasern in Richtung auf die Blutgefäße und durch eine Abschnürung der Nerven-faserendigungen ausgelöst werden.
    Notes: Summary 1. In the subependymal and internal zones of the rat median eminence nerve fibres and vascular processes of ependymal and subependymal cells form neuro-glial bundles. They branch in the external zone. 2. In all these three zones of the infundibulum numerous neuro-glial synapses are found between nerve fibres and vascular processes of glial cells. The vascular processes contain a high number of microtubules as well as polymorphous granular inclusions. 3. In certain regions of the subependymal layer the intercellular spaces are enlarged. They form channel-like spaces containing small bundles of delicate nerve fibres. 4. Nerve cells of the infundibular nucleus located in the lateral parts of the infundibulum send dendrites to the medial parts of the infundibulum. In this area axo-dendritic synapses are found. 5. For morphometric analysis, the nerve fibres of the external zone were classified according to the diameter of their granules. It is shown that in the different regions of the external zone the distribution of the various types of nerve fibre is similar. Moreover it can be seen that a direct correlation exists between the size of the sectional plane of a given nerve fibre and the size of the granules it comprises. 6. Nerve fibre endings abutting on the basement membrane of the pericapillary space are mostly found in bulb-like protrusions of the external zone. The extent to which nerve fibres reach the perivascular space—as compared with the vascular processes of ependymal and glial cells—is higher in the medial than in the lateral parts of the infundibulum. 7. In bilaterally adrenalectomized rats the number and diameter of elementary granules increases in nerve fibres located laterally. This increase is directly related to the survival time and may be due to an enhanced synthesis and storage of Corticotropin-Releasing Factor in these nerve fibres. Compared with the findings in untreated animals the neurohemal contact area is significantly enlarged. The perivascular space contains degenerating nerve fibres which are undergoing phagocytosis by connective tissue cells. It is assumed that these alterations are due to the increased growth of nerve fibres towards the vessels of the “Mantelplexus”, and that, following adrenalectomy, this excessive growth leads to a pinching off of nerve fibres.

    Notes: Summary The adrenergic innervation of the extrarenal blood vessels of the rat left kidney was investigated by fluorescence histochemistry and by electron microscopy. The trunk of the renal artery proximal to the aorta is elastic and appears to be very sparsely innervated. In contrast, near the kidney the renal artery—which divides into 3 to 4 large branches of the muscular type possesses a dense adrenergic innervation. The adrenergic terminal axons are situated in the adventitia close to the external elastic lamella, but only rarely in close contact with smooth muscle cells. In most instances several terminal axons are grouped and enclosed by a Schwann cell, single axons being rare. All terminal axons are able to take up and to store 5-hydroxydopamine which strongly suggests that they are adrenergic. The innervation of the renal vein is more sparse than that of the muscular arteries but somewhat denser than that of the elastic artery. In addition, close to the origin of the renal artery the presence of “small intensively fluorescent” (SIF) cells as well as of some adrenergic ganglion cells is noted. The latter are situated in the adrenergic nonterminal axon bundles, which run parallel to the blood vessels. It is concluded that the uneven adrenergic innervation along the artery as well as individual variations in the branching of the artery are the main causes of the unusually high individual variations of the NA content of this organ such as used in pharmacological experiments.

    Notes: Summary 4-day cyclic adult female Wistar rats were injected subcutaneously with testosterone propionate on diestrus 1 at 16:00 and on diestrus 2 at 10:00 respectively. Non-injected females served as controls. Autopsy was performed on diestrus 2 at 23:00, and on proestrus at 14:00 and 17:00 respectively. The blue Alcian-PAS staining was used to evidence FSH (β) and LH (γ) pituitary cells. In control animals and in diestrus 2 injected females only a small number of FSH cells could be detected on diestrus 2 at 23:00. This number increased markedly on proestrus at 14:00 and decreased on proestrus at 17:00. A similar evolution was observed in diestrus 1 testosterone injected females, but the number of FSH cells appeared higher at any stage of autopsy in these females than in diestrus 2 injected females and in control rats. In control females, numerous LH cells were observed on diestrus 2 at 23:00. The number of these cells was diminished on proestrus at 14:00 and still more at 17:00. On the contrary few LH cells were detected in testosterone injected females on the evening of diestrus 2. An increase of these cells occurred on proestrus at 14:00, followed at 17:00 by only a weak diminution as established by comparison with control animals. An inhibition of FSH release and a suppression of the proestrus surge of LH were therefore supposed to cause, on one hand, the slowing up of follicular growth observed in diestrus 1 injected females and, on the other hand, the blockage of ovulation noted in both diestrus 1 and diestrus 2 treated animals.

    Notes: Summary Neural lobes of adult rats have been explanted and organ-cultured for 5–10 days. While perivascular cells and neurosecretory fibers undergo a progressive degeneration, scattered pituicyte-like cells are observed mostly associated with each other in number of two or three cells for group. Cultured pituicytes are quite similar to in vivo pituicytes except for some particular features as the shape of the nucleus and the number of dense bodies. Furthermore they share with the in vivo pituicytes the phagocytic capacity which appears even increased as far as neurosecretory fibers are concerned. Finally, degenerating pituicytes and free lipid droplets are observed in the intercellular spaces: the significance of these facts is discussed and tentatively interpreted. The organotypic culture of the neural lobe seems to represent an experimental model useful to obtain a relatively pure population of pituicytes.

    Notes: Summary To see whether adenohypophysial tissue has the capacity of self-differentiation, Rathke's pouch together with part of the neurohypophysial primordium was isolated from 12-(Group A), 14- (Group B) and 15-day-old (Group C) rat fetuses and cultivated for 9, 6 and 5 days, respectively. Electron microscopic examination of these explants showed many adenohypophysial cells containing electron dense granules, indicative of cytodifferentiation in all groups. Three cell types were distinguishable on the basis of the size of their granules. Variable amounts of neurohypophysial tissue were seen in some explants of Groups B and C, but not A. This “pars nervosa” contained pituicytes with lipid-like inclusions in their cytoplasm but no neurosecretory axons. The adenohypophysial cells abutting on the pars nervosa were arranged rather regularly like cells of the pars intermedia which contained a few granules of about 200 mμ diameter. From these data it is concluded that in rats granule formation in the adenohypophysial primordial cells starts in the absence of hypothalamic neurosecretory substances. It is uncertain from this study whether these cells actually contain hypophysial hormones.

    Notes: Summary The duct system of the rat exorbital lacrimal gland consists of intercalated ducts, interlobular ducts and excretory ducts. The morphological changes from one type of duct to the next are gradual. At the light microscopical level this consists of a change from a bilaminar epithelium in the intercalated ducts to an epithelium, consisting of approximately three layers — which may be pseudostratified — in the excretory ducts. The basal layer of the intercalated ducts consists of myoepithelial cells, whereas the inner epithelial cells may have both a secretory and an electrolyte transporting function. The interlobular duct epithelium contains many cells with deep infoldings of the basolateral plasma membranes and associated mitochondria, suggesting a similar function to the striated duct epithelium in salivary glands. Numerous basal cells in this epithelium have tentatively been interpreted as unusual myoepithelial cells. Nerve terminals have been observed in the ductal epithelium.

    Notes: Summary Several recent studies suggested that serotonergic neuron-like elements are present in the guinea pig ileum. The present paper reports an extensive study of the digestive tract of the rat with the use of a histofluorescence technique. Administration of the serotonin precursor, tryptophan, associated with a monoamine oxidase inhibitor, did not allow histochemical demonstration of rapidly fading, yellow fluorescent, 6-hydroxydopamine-resistant neurons; conversely such neurons were readily detected in the brain. It is concluded that serotonergic neuron-like elements cannot be detected histochemically in the rat myenteric plexus area after chemical sympathectomy.

    Notes: Summary The response of the Juxtaglomerular apparatus to bilateral adrenalectomy was studied in rats. After the sixth postoperative day, symptoms of severe adrenal insufficiency had developed. In the Juxtaglomerular cells, the volume density of the secretory granules decreased from 32% to 16%. The Golgi apparatus and rough endoplasmic reticulum underwent considerable proliferation, the latter showing strikingly enlarged cisternae.

    Notes: Summary Adrenal glands of adult female rats are heavier than the glands of corresponding male rats. Postpubertal orchiectomy increases the adrenal weight, an effect restored by testosterone replacement. Under the same conditions ovariectomy of 8 weeks duration does not change the adrenal weight while estradiol replacement enhances the relative adrenal weight. Karyometric studies have shown that nuclei in the female zona fasciculata cells are larger (app. 18%) than those of the male. Similar but only slight differences (2%) were observed in the zona reticularis. Orchiectomy results in enlargement of cell nuclei within all zones of the adrenal cortex; testosterone replacement has the opposite effect. Ovariectomy of 8 weeks duration slightly enhances the volume of nuclei of the zona glomerulosa cells, has no effect on the nuclei of the zona fasciculata and reduces the volume of nuclei in the zona reticularis. Estradiol replacement reduces the volume of nuclei of the zona glomerulosa cells but increases nuclear volume in the zona fasciculata and in the zona reticularis. Thus testosterone has an inhibitory effect on the adrenal cortex of the rat while the physiologic effect of estradiol on the morphology of this gland, particularly on the zona fasciculata cells is rather dubious. Neither ovariectomy nor estradiol replacement has an effect on absolute adrenal weight. The stereologic analysis of the volume fractions of particular adrenocortical zones have shown that the volume fraction of the zona glomerulosa in females is smaller as compared with males. On the contrary, the volume fractions of the fasciculata and reticularis zones are higher in female than in male rats. In both sexes gonadectomy and gonadal hormone replacement has an effect on the volume fractions of particular adrenocortical zones, these changes being more pronounced due to orchiectomy or testosterone replacement. The adrenal cortex of all experimental groups contains a similar quantity of parenchymal cells. The cytoplasm volume of a single adrenocortical cell is higher in all zones of the female adrenals as compared with the male. Orchiectomy increases the volume of the cytoplasm in fasciculata and reticularis cells. This effect was reversed by testosterone. Neither ovariectomy nor estradiol replacement has an effect on the volume of the cytoplasm in the fasciculata and reticularis cells. These studies have shown that sex differences with regards to rat adrenal size and weight depend upon the “physiologic atrophy” of the male adrenal cortex, while neither ovariectomy (8 weeks after operation) nor estradiol replacement has a conspicuous effect on the morphologic changes of this gland.

    Notes: Summary In order to characterize different sub-zones in the palisade zone of the rat median eminence, quantitative ultrastructural parameters were applied to brains fixed in aldehyde-osmium tetroxide. The palisade zone has been subdivided in 4 successive sub-zones. Increasing numbers of granular vesicles (GV), especially those smaller than 110 nm, are observed from dorsal to ventral sub-zones. There also are more GV per unit area of nervous tissue in the perivascular than in the more dorsal sub-zones. The individual nerve profiles exhibit a larger size in the perivascular layer than in the more dorsal areas, whereas the number and size of nerve profiles devoid of vesicles diminish from dorsal to ventral. As a consequence more GV occur in the perivascular nerve profiles. In the GV containing nerve profiles the concentration of GV is, however, constant in the different sub-zones. A fluctuating size of the preterminal and terminal parts of the nerve fibres is suggested.

    Notes: Summary Epididymal biopsies from rats that had undergone unilateral or bilateral vasectomies from one to eight months previously were compared with biopsies from their contralateral side or from normal controls to ascertain what ultrastructural changes had occurred. After vasectomy, spermatozoa appeared to dissolve in the lumen of the caput epididymidis and to be absorbed by the principal cells. About 5 weeks after vasectomy, numerous lamellar accumulations became apparent in the supernuclear region. Their resemblance to lysosomes or residual bodies was confirmed by an acid phosphatase reaction. After 10 weeks, similar lamellar and polymorphic accumulations on the contralateral side of animals with unilateral vasectomies indicated that resorption had also increased on the unligated side.

    Notes: Summary The transport of 125I-labeled serum lipoproteins through the aortic endothelium was studied by radioautography. Rat aorta and heart was perfused in vitro with a medium containing human very low density (VLDL), low density (LDL), high density lipoprotein (HDL), delipidated HDL apolipoprotein or rat HDL. In all lipoproteins more than 95% of the radioactivity was TCA precipitable and lipid radioactivity was from 2–4% in HDL, 4–6% in LDL, 7–15% in VLDL. After 18–60 min of perfusion and wash with unlabeled medium, most of the aortic radioactivity was TCA precipitable and the percent of lipid counts was similar to that in the original lipoprotein. Following perfusion with VLDL, LDL, or HDL the radioautographic reaction was seen over the endothelium, the subendothelial space and the inner media, and was separated by an unlabeled zone from the reaction present over the adventitia. Uniform labeling of the entire wall was found after perfusion with HDL apolipoprotein. The presence of silver grains over endothelial cells in regions rich in plasmalemmal vesicles suggested that these organelles participate in the transport of the labeled lipoprotein, as was shown for lactoperoxidase (Stein and Stein, 1972). The present data indicate that cholesterol may enter the aortic wall as a constituent of lipoprotein particles. Since an HDL particle carries less than 1/20 of the cholesterol present in a LDL particle, it seems that the lower susceptibility of the female to atheromatosis might be related to the higher ratio of HDL to LDL particles in the female serum.

    Notes: Summary The response of somatotrophs to large doses of purified bovine growth hormone was examined in uninephrectomized rats drinking 1% sodium chloride in an attempt to prolong the action of the hormone. Growth hormone produced a triphasic response in the ultrastructure of somatotrophs. Uninephrectomy and salt alone induced no changes in somatotrophs as compared to unoperated animals consuming tap water. In the first phase (3 days) the cross sectional area of the cell increased, the size of the largest granule per cell as well as the number of granules per cell increased significantly. All these parameters then reached control level after 7 days of injections during a second phase. Suppression of somatotrophs by exogenous growth hormone was evident by 16 days (third phase) when the area of the cell, the size of the largest secretory granule as well as the number of granules per cell all decreased significantly. Somatotrophs apparently did not react to suppression by a lysosomal mechanism inasmuch as no increase in the number of lysosome-derived structures was seen even at 16 days.

    Description / Table of Contents: Zusammenfassung Radioaktiv markierte Aminosäuren werden bei Säugetieren nach intraokulärer Injektion nicht nur zu den primären optischen Kerngebieten, sondern auch in den Nucleus suprachiasmaticus des Hypothalamus transportiert (Moore und Lenn, 1972; Hendrickson, Wagoner und Cowan, 1972; Moore, 1973). Dieser Befund macht eine direkte Verknüpfung zwischen der Netzhaut und dem sekretorisch aktiven Nucleus suprachiasmaticus wahrscheinlich, obwohl mit Silbertechniken eine solche Verbindung nicht sicher nachgewiesen werden konnte. Karyometrische Studien am Nucleus suprachiasmaticus männlicher Wistar-Ratten zeigen 6 Tage nach beidseitiger Blendung eine signifikante Abnahme der Zellkerndurchmesser. Dieses Ergebnis spricht für eine Aktivitätsänderung der Neurone im Nucleus suprachiasmaticus geblendeter Ratten.
    Notes: Summary Intraocular injection of labeled amino acids results, in different mammals, in an accumulation of radioactive material not only in the primary optic centers but also within the hypothalamic suprachiasmatic nucleus (Moore et al., 1972; Hendrickson et al., 1972; Moore, 1973). This finding argues in favour of a direct connection between the retina and the secretory suprachiasmatic nucleus although silver techniques do not show such a pathway. In male Wistar rats the nuclear diameter of the suprachiasmatic neurons decreases significantly 6 days after bilateral experimental ablation of the retina. These results indicate an alteration in activity of the suprachiasmatic neurons in blinded rats.
