    Notes: Summary The influence of the melt temperature (T1) on the crystallization kinetics and the morphology of two semicrystalline polymers: poly(oxymethylene): T m 0 =198° C and poly(ether-ether-ketone) T m 0 =380° C) was investigated using thermal analysis and optical microscopy. For different melt temperatures (T1), isothermal and nonisothermal crystallizations were analysed using the AVRAMI and OZAWA models. Using different analyses, for both polymers we observed a spherulitic growth with the following influence of the melt temperature on the number of spherulites: -T1〈T m 0 produces many small spherulites -T1〉T m 0 gives rise to few large spherulites

    Notes: Abstract Thermal transitions of TDI-based polyurethane elastomers with PTMO as the soft segment were characterized by the depolarization technique in TSC and by using with the thermal windowing technique on selected specimens in the RMA measurements. Results indicate that the broadened thermal transition in the glass transition region as observed in the DSC thermogram is related to the combined Tg transition and the Tglobal transition in the TSC spectrum. This Tglobal transition is associated with the macromolecular property as detected by tan δ in DMA measurement. The increase in the Tg with a high NCO content may be explained by the structural modification found on the urethanic chain with the additional linkage of the hard segment that affects the cooperative motion of the molecular chain. Data measured from DSC, TSC/RMA and DMA with simulated DEA and wide angle X-ray data are presented for the characterization of the polyurethanes. The RMA measurement leads to a compensation search on Tg transition and provides pertinent thermokinetic data that correlates the NCO content with changes in enthalpy and entropy on the relaxation behaviors in the Tg transition of polyurethane elastomers.

    Notes: Abstract Photosynthetic characteristics were investigated in the geographically isolated and restricted mangrove species, P.rhizophoreae. Gas exchange measurements were made on two to seven years old hydroponically grown plants maintained in 10%, 50% and 100% seawater. CO2 exchange in the 50% and 100% seawater treatments was reduced by 10% and 26%, respectively, compared to the 10% seawater treatment. CO2 response curves indicated that carboxylation efficiency was greater in 10% than in 50% seawater, while stomatal limitation increased from 11% to 16% as salinity increased from 10% to 50% seawater. Carbon losses via photorespiration (31% and 41%) and CO2 compensation point (67 and 81 μ11−1) were greater in 50% than in the 10% seawater treatment. Maximal CO2 exchange occurred at 30 °C with no differences among the salinity treatments. The results indicate that P. rhizophoreae exhibits many gas exchange characteristics previously reported for other mangroves.

    Notes: Abstract Isopropylnitrate (IPN) is described as a detonable material used in propellants and explosives. While there is considerable information available on its sensitivity and compatibility with other materials, very little is known about its thermochemical properties. This paper will describe the results obtained from some DSC, heat flux calorimetry (HFC) and accelerating rate calorimetry (ARC) measurements. The ASTM DSC method using a hermetic aluminum pan having a lid with a laser-produced pin hole was used to determine the vapour pressure of IPN1. Results calculated from an Antoine equation are in substantial agreement with those determined from DSC measurements. From the latter measurements, the enthalpy of vaporization was determined to be 35.32±0.62 kJ mol−1. Attempts to determine vapour pressures above about 0.8 MPa resulted in significant decomposition of IPNg. The enthalpy change for decomposition in sealed glass systems was found to be -3.43±0.09 kJ g−1 and -3.85±0.03 kJ g−1, respectively from DSC and HFC measurements on IPN1 samples loaded in air. Slightly larger exotherms were observed for the HFC results in air than those in inert gas, suggesting some oxidation occurs. In contrast, no significant difference in the observed onset temperature of about 150°C was observed for both the HFC and ARC results. From DSC measurements, an Arrhenius activation energy for decomposition of 126±4 kJ mol−1 was found. These measurements were also conducted in sealed glass systems and decomposition appeared to proceed primarily from the liquid phase.

    Notes: Abstract When ethylene-vinyl acetate copolymer, EVA, is heated, a two-stage thermal degradation occurs following its melting. The vinyl acetate content of the copolymer was determined to be 43.8% by using TA 2950 and TA 2050 thermogravimetric instruments. TG/FTIR was used to detect the evolved gas. Acetic acid and trans-1-R-4-R'-cyclohexane were the main products evolved from EVA in the first and second stage, respectively. The apparent activation energies were determined for both stages by differential methods.

    Notes: Abstract A crystallization kinetics analysis of several polypropylene-polyethylene (PP-PE), PP-rich copolymers was made by means of differential scanning calorimetry. The crystallization was studied via calorimetric measurements at different cooling rates. Several additives were added to the base material. Some test samples were subjected to artificial ageing processes. A modified isoconversional method was used to describe the crystallization process under non-isothermal conditions. The value of the Avrami parameter was determined for primary and secondary crystallization.

    Notes: Abstract 14 mixed Co(III) dioximine chelates of the types [Co(Diox·H)2(amine)2]X (X = Br, I, NO3, ClO4) and H[Co(Diox·H)2(N3)2], respectively (Diox·H2-1,2-cyclohexane dione dioxime (nyoxime), 1,2-cycloheptane dione dioxime (heptoxime) 1,2-cyclooctane dione dioxime (octoxime) were obtained and their thermal decompositions were studied in an argon atmosphere. After the dehydration of the crystallohydrates, both types of complexes exhibit 3 decomposition stages. For the [Co(Diox·H)2(amine)2]X type complexes (X = Br, I) the first endothermal stage is the substitution of an amine molecule for the external sphere anion and this process is followed by two exothermal decomposition stages. With H[Co(Diox·H)2(N3)2] type complexes the first and third processes are relatively slow, but the second process is very fast, corresponding to a vertical portion of the TG curves. From the TG curves kinetic parameters were derived for 11 processes and the validity of a non-linear compensation law was observed.

    Notes: Abstract Beside the two well-known minerals cryolite, Na3AlF6, and chiolite, Na5Al3F14, the binary system NaF-AlF3 also contains a third compound, NaAlF4, sodium tetrafluoroaluminate. Solid NaAlF4 has been prepared from its vapour under controlled conditions. The stability of NaAlF4 has been investigated by differential scanning calorimetry. It is shown that the disproportionation of the compound: 5NaAlF4(s)=Na5Al3F14(s)+2AlF3(s) takes place at considerable rate between 700 and 900 K. The enthalpy of this reaction is calculated and found to be -66.9 kJ. Enthalpies of the two solid state transitions α-Na3AlF6 → β-Na3AlF6 and α-AlF3 → β-AlF3 have also been measured and new values are reported. The enthalpy of formation of chiolite, Na5Al3F14, at 900 K has been recalculated from enthalpy increment data obtained by drop calorimetry. A value of ΔH900 o = -7513.6±12.0 kJ mol-1 has been obtained. This value is in disagreement with the recommended value given in JANAF Thermochemical Tables given at 900 K ΔHf o = -7559.2 kJ mol-1.

    Notes: Abstract Two mixtures of pentlandite and the monosulfide solid solution (mss) have been synthesized. The bulk compositions of the samples are Fe6Ni3S8 and Fe3Ni6S8. Differential scanning calorimetry detected exothermic process in the samples under heating. The process takes place in temperature range between phase transition in the mss (near 400 K) and 690 K and is governed by diffusion. X-ray powder diffraction has showed that equilibrium Fe-Ni distribution between pentlandite and the mss is achieved after short-time heating up to 670 K.

    Notes: Abstract Cure kinetics using a differential scanning calorimetry (DSC) technique were analyzed for a thermoplastic modified tetraglycidyl-4,4′-diaminodiphenylmethane (TGDDM) epoxy resin cured with diaminodiphenylsulphone (DDS), an aromatic diamine. The neat resin and its blends with the poly(styrene-co-acrylonitrile) (SAN) of various compositions were studied by applying a phenomenological model proposed by Kamal. Kinetic parameters were determined by fitting experimental data. This model gives a good description of cure kinetics up to the onset of vitrification. Diffusion control was incorporated to describe the cure in the latter stages of cure. The results showed that the addition of SAN did not alter the nature of the reaction, but the reaction rates and final conversions decreased when SAN contents increase, due to reduction of mobility of the reacting species.

    Notes: Abstract Modern thermal analysis, microcalorimetry and new emerging combined techniques which deliver calorimetric, microscopic and spectroscopic data offer a powerful analytical battery for the study of pharmaceuticals. These techniques are very useful in all steps of development of new drug products as well as methods for quality control in production. The characterization of raw materials enables to understand the relationships between polymorphs, solvates and hydrates and to choose the proper development of new drug products with very small amount of material in a very short time. Information on stability, purity is valuable for new entities as well as for marketed drug substances from different suppliers. Excipients which vary from single organic or inorganic entity to complexes matrixes or polymers need to be characterized and properly controlled. The thermodynamic phase-diagrams are the basis of the studies of drug-excipients interactions. They are very useful for the development of new delivery systems. A great number of new formulations need proper knowledge of the behaviour of the glass transition temperature of the components. Semi-liquid systems, interactions in aqueous media are also successfully studied by these techniques.

    Notes: Abstract The polymorphic behaviour of the purine derivative MKS 492 was studied with investigations of suspensions of selected samples in different solvents and of samples obtained by crystallizations. The samples were analyzed by DSC, TG and X-ray diffraction. Six different crystalline modifications called A, B, B’, C, D and E and an amorphous form were identified. Four pure crystalline modifications, A, B, C and D have been manufactured and characterized by DSC, X-ray, IR, solubilities, densities, hygroscopicity and dissolution measurements. The four forms A, C, D and E are monotrop to the form B. The form B is enantiotrop to the form B’, which revealed the highest melting point of all known polymorphs. This form B’ is only stable at high temperature. Temperature resolved X-ray diffraction was very helpful for proper interpretation of the thermal events. The melting peaks of the forms A and C and the endothermic peak corresponding to the enantiotropic transition B into B’ occur in a narrow range of temperature. The form B which is the most stable one at room temperature has been chosen for further development. Quantitative methods to determine the content of the forms A, C and D in samples of form B or to determine the content of form A, B and D in form C have been developed by using X-ray diffraction. Limits of detection are 1 or 2%. For the quantitative determination of the amorphous fraction, X-ray diffraction and microcalorimetry are compared. For high amounts of the amorphous fraction, the X-ray diffraction method is preferred because it is faster. Microcalorimetry is very attractive for levels below 10% amorphous content. The lowest limit of detection is obtained by microcalorimetry, about 1%.

    Notes: Abstract The thermodynamic energy relationship between two crystal modifications of cimetidine was investigated and compared with differences in their processing properties with respect to transformation from one modification to the other. The crystal energies of the two modifications A and D were found to be almost identical and therefore the polymorphs are regarded as virtually isoenergetic crystals. This statement is based on DSC measurements of the melting points and of the enthalpies of fusion for the two crystal forms, which enable the calculation of the Gibbs free energy functions. Furthermore, the statement is supported by measurements of the enthalpies of solution in two different solvents. Both DSC and solution experiments reveal a slightly higher stability of the D modification with respect to the A form. In addition, tribomechanical treatment also indicates modification D to be the more stable one, as well as the higher density of the D form. No transformation during DSC at low heating rate was found which could be used in a stability consideration. As the explicit crystal structures of the two modifications are resolved, it was possible to calculate crystal energies theoretically as well. The theoretical results showed a remarkable difference in the crystal energies at zero degree Kelvin. Furthermore, they were just contradicting experimental findings by stating A being more stable than D. Possible reasons for this discrepancy and the feasibility of today's calculation methods with respect to prediction of stability properties are discussed.

    Notes: Abstract The molecular aggregation of acrylic and methacrylic acid esters containing long-fluorocarbon chains: 2-(perfluoroalkyl)ethyl acrylate (FFnEA) and 2-(perfluoroalkyl)ethyl methacrylate (FFnEMA) (F(CF2)nCH2CH2OCOC(X)=CH2, where X=H, CH3 and n=6, 8, 10) was investigated by differential scanning calorimetry (DSC) and temperature controlled X-ray powder diffraction measurement. These compounds exhibited some characteristic polymorphic behaviors depending on the length of fluorocarbon chain and the α-position methyl group. The solid-state polymerization by γ-ray irradiation was studied for these compounds in the various crystal forms. In the solid-state polymerization, highest polymerizability was observed in the crystal form that exists in the highest temperature region for each compound.

    Notes: Abstract The physical state of benzoic acid (BA) and its interaction with ethyl cellulose (EC) were examined in ethyl cellulose—benzoic acid matrices by Differential Scanning Calorimetry (DSC). The glass transition temperature (Tg) of EC of various matrices having BA in solid solution form (upto 27.7%) was reduced. The BA in matrices containing more than 38.9% drug exhibited distinct melting endotherms due to crystalline form. The peak temperatures of these endotherms were lowered and they broadened as the concentration was lowered. The solubility of BA increased at its melting point as compared to ambient temperature. The melting enthalpy of BA, when plotted as a function of its concentration yielded a straight line with intercept of 330 mg g−1 of matrix. This is the solubility of BA in EC at its melting temperature. Fourier Transform Infra Red Spectroscopy (FTIR) investigations confirmed that hydrogen bonding occurred between EC and BA through hydroxyl groups.

    Notes: Abstract A procedure for measurement of the heat of zeolite dehydration by scanning heating has been designed. Simultaneous data on heat flow (DSC) and mass loss (TG) are required for evaluation. The heating rate depends on the experimental conditions (point-spread function, sample mass, crucible design, and calorimetric reproducibility). Dehydration measurements have three advantages as compared with the sorption procedure: i) one can investigate samples with irreversible dehydration; ii) no approximation model is needed for calculation of the partial molar heat of dehydration; and iii) the procedure is not labor-consuming. The procedure was tested on the natural zeolites heulandite, chabazite and mordenite. The results are close to those measured by the sorption procedure. The partial molar heat of dehydration was found to depend on the water content. It increases from 50 to 87 J mol−1 K−1 for heulandite, from 53 to 81 J mol−1 K−1 for chabazite, and from 51 to 71 J mol−1 K−1 for mordenite. The approximation of the heat of sorption by linear regression was found to be wrong. Detection of a ‘phase transitioN’ after this approximation has no meaning.

    Notes: Abstract The described instrumental method makes it possible that the quasi-static heating technique, well applicable to thermogravimetric measurements, (see Part I of this paper) can be used in the case of DTA and DSC examinations, too. Based on the new type of curves the characteristic transformation temperatures, the whole course of the transformation in dependence of sample temperature, the extent of the enthalpy change caused by the transformation or by its partial processes can accurately be determined. The essentially greater accuracy of the measurements — in comparison to the conventional ones — is due to the quasi-static heating technique which ensures that the transformations should take place under quasi-equilibrium conditions.

    Notes: Abstract The gel to liquid-crystal transition for vesicles in aqueous solution formed by dimethyldi-n-octadecylammonium bromide (DOAB) occurs at 44.7°C. Moreover, the shapes of the scans recorded by a sensitive DSC microcalorimeter are very similar when the vesicular solutions are prepared starting with solid DOAB and comparable amounts of either solid urea or solid alkylureas. Therefore, the DOAB vesicles in aqueous solution accommodate this class of solutes without marked changes in the melting temperature and the enthalpy of the transition. The contrast with effects of added surfactants and simple organic solutes such as THF and ethanol is particularly significant.

    Notes: Abstract Differential scanning calorimetry (DSC) and thermogravimetric analysis (TG) of lithium formate monohydrate (LiHCOO·H2O) were performed in the temperature range 300–700 K. The DSC/TG measurements show that the dehydration process to anhydrous lithium formate (LiHCOO) is complex and occurs in two stages. The data are correlated to the structure and to the arrangement of the molecules in the crystal, including the hydrogen-bonding. Infrared transmittance and Raman spectra of this crystal are reported and commented on.

    Notes: Abstract The thermal decomposition of strontium and barium malonates has been studied isothermally and non-isothermally employing simultaneous TG-DTG-DTA, DSC, XRD and IR spectroscopic techniques. DSC of these malonates has been recorded both in oxygen and nitrogen atmospheres. The decomposition is a single step process and the end product formed is carbonate. The energy of activation and frequency factor values for the decomposition of strontium malonate are 547 kJ mol−1 and 1041 s−1 respectively. The activation energy and frequency factor values for isothermal dehydration of barium malonate sester-hydrate are 57–111 kJ mol−1 and 107–1012 s−1 respectively and the corresponding values for decomposition from DSC are 499.5 kJ mol−1 and 1044 s−1 respectively. The higher thermal stability of strontium malonate as compared to that of barium salt is ascribed to its being anhydrous so that decomposition proceeds without restructuring. Their thermal stabilities have also been compared with that of respective oxalate salts.

    Notes: Abstract Processing thin films for advanced applications, for instance in electronics and optoelectronics, involves several steps starting from precursor synthesis and ending up with the devices. The review will focus on the main chemical deposition methods (CVD, ALE, spray pyrolysis, sol-gel) giving selected examples of problem-solving by thermal analysis. The techniques discussed are TG, DTA/DSC, EGA and their combinations. High-temperature X-ray diffraction (HTXRD) is also a powerful tool for in situ studies of thin films. The examples are taken from solar cell, superconductor and flat panel electroluminescent display technologies.

    Notes: Abstract Nickel sulphide (NiS) was characterised using X-ray diffraction, thermal gravimetric analysis (TG) and differential scanning calorimetry (DSC). The 'as received' Millerite, stoichiometric NiS, observed to be slightly nickel deficient, was found to readily decompose in a nitrogen atmosphere at elevated temperatures (450°C max.) to the sulphur deficient Godlevskite, Ni7S6. DSC and X-ray measurements demonstrated that the high temperature form of the Godlevskite was readily stabilised at room temperature. The kinetics of the α-β re-transformation in Godlevskite were then investigated using DSC and were observed to be first order.

    Notes: Abstract Differential scanning calorimetry (DSC) was used to determine the molar enthalpies of dehydration and decomposition of CoC2O4·2H2O, Co(HCOO)2·2H2O and [Co(NH3)6]2(C2O4)3·4H2O. The first stage of dissociation of each compound is a single-step dehydration both in air and argon atmospheres. The next stages are decomposition processes influenced by experimental parameters. The enthalpies of dehydration and decomposition vary from compound to compound in each atmosphere. The obtained data have been related to the macromechanisms proposed for the thermal decomposition and the parallel-consecutive decomposition-oxidation processes.

    Notes: Abstract This paper investigates the minimum oil content necessary for self-sustained combustion, which is introduced as a criterion for the selection of suitable reservoirs for in-situ combustion processes. Differential scanning calorimetry was used to determine the heat values of oil-limestone mixtures. The minimum temperature required for the total consumption of the fuel was obtained by thermogravimetry (TG/DTG). The minimum amount of oil necessary to sustain combustion was calculated from these two parameters and compared with the oil content of the reservoir. Reservoirs with an oil content greater than or equal to this minimum value were considered feasible. It was seen that the fields examined are generally not suitable for in-situ combustion processes.

    Notes: Abstract Temperature modulated dynamic mechanical analysis (TMDMA) was performed in the same way as temperature modulated DSC (TMDSC) measurements. As in TMDSC TMDMA allows the investigation of reversible and non-reversible phenomena during crystallisation of polymers. The advantage of TMDMA compared to TMDSC is the high sensitivity for small and slow changes in crystallinity, e.g. during re-crystallisation. The combination of TMDMA and TMDSC yields new information about local processes at the surface of polymer crystallites. It is shown that during and after isothermal crystallisation the surface of the individual crystallites is in equilibrium with the surrounding melt.

    Notes: Abstract The influence of heavy ion-irradiation (Ar 5.5 MeV amu-1, 5·1011 ion cm-2) on the melting and crystallization of two PBT films subjected to different modes of thermal treatment was investigated. Differences were observed between the processes occurring in both initial films, due to differences in crystalline phase content. The course of melting and crystallization in heavy ion-irradiated films during first heating, cooling and second heating differs from that in the initial films. The density data and DSC results indicate a decreased crystalline phase content in the PBT films after irradiation.

    Notes: Abstract The terpenoids acetyl sitosterol, lupeol, acetyl diosgenin and stigmasterol were studied. Comparison of the thermogravimetric curves and the activation energies of the terpenoids suggested the following sequence of thermal stability: acetyl sitosterol 〈 acetyl diosgenin 〈 lupeol 〈 stigmasterol. The DSC curves allowed determination of the melting points and the degrees of purity. Comparison of the TG and DSC curves revealed the presence of phase transitions without mass loss that were attributed to rearrangements in the terpenoid molecules.

    Notes: Abstract A narrow, reversible endothermic main transition is found in the aqueous micellar phase of octaethylene glycol tetradecyl ether (C14E8) by DSC, characterized by a transition temperature of 41°C and a ΔH value of 0.5 kcal mol−1, which is not observed by light scattering. This transition is assigned to a cooperative conformational rearrangement of the assembled amphiphilic detergent molecules and not to a micelle aggregation process. It is suggested that the detergent’s polar head group is primarily involved in this rearrangement.

    Notes: Abstract A number of 15 [Fe(Diox#x00B7;H)2L2] type chelates and [Fe(Diox)3(BOR)2] clathrochelates (Diox#x00B7;H2 — dimethylglyoxime, glyoxime, propoxime, nyoxime, furyl-dioxime; L-pyridine, alkyl-pyridine derivatives, diethyl-phenyl-phosphine, diethyl-p-tolyl-phosphine) were obtained and characterized by means of far and middle FTIR and Mössbauer spectroscopic methods. Some structural problems were discussed on the basis of the optical data. The DSC measurements show the higher thermal stability of the clathrochelates without O—H⋯O intramolecular hydrogen bonds (with asymmetric octahedral structure), as compared to the [Fe(Diox#x00B7;H)2L2] trans, symmetric chelates containing O—H⋯O bonds. The kinetic parameters of the thermal decomposition of the complexes have been derived using the nomogram method.

    Notes: Abstract Calorimetry is the method first used by Jackson and McKenna to study the effect of finite-size on the molecular dynamics of glass-formers confined in nano-meter scale pores. It was found that the glass transition is shifted to lower temperature as pore size decreases. Since then, other spectroscopic techniques have corroborated this finding and given more information on the molecular dynamics. These results are used to compare with the predictions of several theories of glass transition, and in particular the coupling model of the author.

    Notes: Abstract A forensic sample consisting of melt-recrystallized polymers that was recovered from the scene of a fire in a factory was identified by differential scanning calorimetry. The factory commonly used two kinds of film sheets, A and B, made by different manufacturers. It was necessary to decide whether the forensic sample related to material A or B. The forensic sample and reference samples of materials A and B were subjected to infrared spectroscopy and pyrolysis gas chromatograph mass spectrometry measurements, which revealed their polyethylene nature. The thermal behaviour of the samples was examined by differential scanning calorimetry (DSC) and they were found to be blends of two kinds of polyethylenes, low-density polyethylene and linear low-density polyethylene. The samples could be identified and distinguished from each other via the DSC measurements.

    Notes: Abstract Two methods for estimating the critical temperature (Tb) of thermal explosion for the highly nitrated nitrocellulose (HNNC) are derived from the Semenov's thermal explosion theory and two non-isothermal kinetic equations, dα/dt=Af(α)e−E/RT and dα/dt=Af(α)[1+E/(RT)(1–To/T)]e−E/RT, using reasonable hypotheses. We can easily obtain the values of the thermal decomposition activation energy (E), the onset temperature (Te) and the initial temperature (To) at which DSC curve deviates from the baseline of the non-isothermal DSC curve of HNNC, and then calculate the critical temperature (Tb) of thermal explosion by the two derived formulae. The results obtained with the two methods for HNNC are in agreement to each other.

    Notes: Abstract The enthalpic relaxation of the title glasses, studied by differential scanning calorimetry, is well described by a mathematical model based on the stretched exponential relaxation function with the relaxation time proportional to the actual viscosity. The dependence of viscosity on temperature and the fictive temperature was expressed by Mazurin's approximation. The relaxation parameters obtained correlated significantly with the glass composition, indicating the changes in the structural of the TiO2 role near a TiO2 content of 3–4 mol%.

Ni(dpt)C4O4·H2O has also been synthesised pyrolytically in the solid state from the corresponding bis complex. All the complexes possess octahedral geometry. The squarate anion takes part in coordination only in monotriamine species. [Ni(medien)2]C4O4 upon heating undergoes phase transition (270–285°C; ΔH=4.9 kJ mol−1) accompanied by colour change pink to grey. Thermal stability decreases with increase in chain length of the triamines.

    Notes: Abstract Amorphous PdZr, PdCuZr and PdCuSi alloy ribbons and powders are characterized by DSC, XRD and XPS in the as-received state and after treatments with oxygen, hydrogen or dilute hydrogen fluoride solution. Zr-containing alloys are shown to undergo substantial structural changes resulting in palladium enrichment on their surface, whereas no apparent changes in the bulk structure are found for PdCuSi.     Notes: Abstract The internal dynamics and the thermal stability of myosin in rabbit psoas muscle fibres in different intermediate states of the ATP hydrolysis cycle were studied by differential scanning calorimetry (DSC) and electron paramagnetic resonance (EPR) spectroscopy. Three overlapping endotherms were detected in rigor, in strongly binding and weakly binding state of myosin to actin. The transition at 58.4°C can be assigned to the nucleotide-binding domain. The transition at highest temperature represents the unfolding of the actin and the contributions arising from the actin-myosin interaction. The transition of 54°C reflects the interaction between the subunits of myosin. Nucleotide binding induced shifts of the melting temperatures and produced variations in the calorimetric enthalpy changes. The changes of the EPR parameters indicated local rearrangements of the internal structure in myosin heads.

    Notes: Abstract The stability and thermal behaviour of chloramphenicol and various of its mixtures were investigated. The thermogravimetric and stability constant results showed that the chloramphenicol base is thermally more stable than the tablet in the studied formulation. The reduction in stability was attributed to the presence of starch in the formulation. The thermal decompositions of the chloramphenicol base and the tablet obey first-order kinetics.

    Notes: Abstract The previously described method involving the use of transient DSC was applied to pharmaceutical powder compacts and to ceramic powder compacts. The samples were prepared by compressing powders of pentaerythritol tetraacetate and two kinds of alumina powder (differing in particle size distribution) up to a pressure of 20 MPa by using a jig. For pentaerythritol tetraacetate, a linear relationship was obtained between the parameter obtained by DSC and the compaction pressure.

    Notes: Abstract Enthalpic relaxation has been used to model the development of the glass transition in polymers, using kinetic parameters determined separately. For this purpose the Kohlrausch-Williams-Watt stretched exponential function, relating the extent of relaxation, Φ(t), to time t and an average relaxation time, τa, i.e. $$1 - \Phi \left( t \right) = \exp \left( { - t/ta} \right)^{\beta }$$ where β is inversely related to the breadth of the relaxation spectrum, has been adopted. The relaxation time dependence on temperature was taken to follow the modified Arrhenius relationship, $$\tau _a = A\exp \left[ {\frac{{X\Delta H}}{{RT}} + \frac{{\left( {1 - X} \right)\Delta H}}{{RT'}}} \right]$$ where T is the storage and T′ the fictive temperature, X is the structure factor and ΔH the activation enthalpy. Both have been found to describe the process of enthalpic relaxation in polymer glasses and a direct comparison has been made with the change in specific heat observed with different cooling rates in DSC experiments.     Notes: Abstract The thermoanalytical behaviour of pyloric caeca during salting and ripening was investigated using a Perkin Elmer DSC 7. Not only the thermal stability of the muscle proteins was influenced by salting but also that of pyloric caeca. It was recognised that the salting itself leads to a remarkable increase of the transition temperature compared with raw herring. An influence of the salt:fish ratio could be observed. The higher the salt content the higher the increase of the denaturation temperature. During ripening the transition temperature remained on a high level or showed only a slight decrease during the investigation period. The dependency from the salt content remained evident. The increase of the transition temperature was accompanied by a decrease of the transition enthalpy. The increase of thermal stability is connected with a decrease of the general proteolytic activity in pyloric caeca. Possibly, the enzymes are diffusing from the pyloric caeca into the muscle and cause there an increase of enzymatic activity observable in North Sea herring accompanied by a decrease of activity in pyloric caeca itself. Simultaneous the thermal stability of pyloric caeca is lowered. The reason for the differences in ripening could be seen in some enzyme-inhibiting factors unknown until now.

    Notes: Abstract Thermal stability of vegetative cells of Listeria monocytogenes, Escherichia coli and Lactobacillus plantarum was studied by counting viable fractions and determining DSC curves of their suspensions. DSC curves in the 5–99°C range showed a series of endothermic transitions between 50 and 60°C, where the heat destruction of cells occurred. Heat denaturation of DNA required a higher temperature than cell killing. Thermal death was strongly influenced by the pH, composition and NaCl content of the suspending buffer. A mathematical model developed by us enabled comparison of DSC peak temperatures and temperatures required for loss of viability.

    Notes: Abstract Differential Scanning Calorimetry combined with Fourier transform infrared spectroscopy, was applied to the study of a number of fulvic and humic acids extracted from soils, peat, river and seawater. The thermal patterns obtained were related to the nature and origin of samples. The low-temperature endotherms were attributed to dehydration and loss of peripheral polysaccharide chains. The endotherm at 250°C observed for soil FA was ascribed to partial decarboxylation of more labile surface COOH groups, whereas the high-temperature exotherms at about 500°C were related to the degree of polycondensation of the aromatic network of the humic molecules.

    Notes: Abstract Photosynthetically active radiation (PAR) and temperature were measured continuously at the surface of estuarine intertidal sediments in the Tagus estuary, Portugal, along two spring-neap tidal cycles. PAR and temperature were strongly conditioned by the periodic tidal inundation, with large and abrupt variations occurring during flooding and ebbing. PAR levels reaching the sediment surface decreased very rapidly to zero or very low values during most of the daytime immersion. Inundation during high tide had the general effect of attenuating the amplitude of daily temperature fluctuation, with the incoming water usually warmer than the sediment during the night or early morning and cooler during the day. The daily progression of tidal emersion resulted in a clear fortnightly variation in total daily PAR reaching the sediment surface, while both daily mean temperature and mean temperature of diurnal low tide periods failed to exhibit a well-defined fortnightly periodicity. The obtained results indicate that the estuarine intertidal environment is dominated, at sub-seasonal time scales, by fortnightly periodicity in irradiance and temperature conditions favourable for benthic photosynthesis.

    Notes: Abstract Microalgal productivity was examined using both a wild type and a phycocyanin-deficient mutant of Synechocystis PCC 6714 (PD-1). The culture was conducted at various light intensities under low and high cell densities in a continuous culture system. At low light intensity, photosynthetic productivity was almost the same for both low and high cell densities. However, at higher light intensities photosynthetic productivity was higher in mutant PD-1 than in the wild type. At 2000 μmol photon m−2 s−1 the productivity was 50% higher in mutant PD-1. This result is consistent with our first report (Nakajima & Ueda, 1997), which showed that photosynthetic productivity can be improved by reducing the light harvesting pigment content in high cell density cultures at high light intensities. It is concluded that the technology for reducing LHP content is a useful method for improving photosynthetic productivity in algal mass production.

    Notes: Abstract The green flagellate Euglena gracilis has been used as a model organism to elucidate the possible large-scale and short-term effects of waste substances from the pulp and paper industry on photosynthetic efficiency (PE). Different concentrations of waste substances before and after treatment in a cleaning system were studied. The uncleaned sample at concentrations up to 1:10 and the cleaned sample at concentrations up to 1:5 showed stimulating effects on the PE after 7 days of incubation compared to the control. The effects of waste substances on the PE of E. gracilis were also studied in combination with short-term studies (20 and 40 min) of ultraviolet-B radiation (UV-B, 280–320 nm). It was shown that increasing concentrations of the uncleaned sample had continuously stimulating effects on the PE and worked protectively against UV-B radiation. The cleaned sample exhibited no effects, or negative effects, on the PE of E. gracilis together with UV-B radiation compared to the experiments with only UV-B radiation. At the concentration 1:1 of the cleaned sample an increase in the PE was detected due to the high concentration of the coloured substances and a decrease in the UV-B penetration. PE revealed itself to be highly sensitive for detecting toxic effects on E. gracilis and is thus very promising for use in regular toxicity tests of waste water from pulp and paper industry.

    Notes: Abstract Photosynthetic (oxygen evolution) and growth (biomass increase) responses to ambient pH and inorganic carbon (Ci) supply were determined for Porphyralinearis grown in 0.5 L glass cylinders in the laboratory, or in 40 L fibreglass outdoor tanks with running seawater. While net photosynthetic rates were uniform at pH 6.0–8.0, dropping only at pH 8.7, growth rates were significantly affected by pH levels other than that of seawater (c. pH 8.3). In glass cylinders, weekly growth rates averaged 76% at external pH 8.0, 13% at pH 8.7 and 26% at pH 7.0. Photosynthetic O2 evolution on a daily basis(i.e. total O2 evolved during day time less total O2 consumed during night time) was similar to the growth responses at all experimental pH levels, apparently due to high dark respiration rates measured at acidic pH. Weekly growth rates averaged 53% in algae grown in fibreglass tanks aerated with regular air (360 mg L-1 CO2) and 28% in algae grown in tanks aerated with CO2-enriched air (750 mg L-1 CO2). The pH of the seawater medium in which P. linear is was grown increased slightly during the day and only rarely reached 9.0. The pH at the boundary layer of algae submerged in seawater increased in response to light reaching, about pH 8.9 within minutes, or remained unchanged for algae submerged in a CO2-free artificial sea water medium. Photosynthesis of P. linearissaturated at Ci concentrations of seawater (K0.5560 μM at pH 8.2) and showed low photosynthetic affinity for CO2(K0.5 61 μM) at pH 6.0. It is therefore concluded that P. linearisuses primarily CO2 with HCO3 - being an alternative source of Ci for photosynthesis. Its fast growth could be related to the enzyme carbonic anhydrase whose activity was detected intra- and extracellularly.

    Notes: Abstract Photosynthesis and cell composition of Porphyraleucosticta discs grown at low (〈 0.0001% in air), current (control) and high (1% CO2 in air)inorganic carbon (Ci) concentrations were analyzed. Carbohydrate content in discs grown at high Ci increased (15.1 mg g-1 FW) with respect to the control (6.4 mg g FW-1), whereas soluble protein content decreased to one-third (5.6 to2.1 mg g-1 FW). Carbohydrate content was unaffected and soluble protein slightly increased in discs grown at low Ci. As a consequence of these changes, a lower C/N molar ratio (8.6) was found in the discs grown at low compared to high Ci(12.4). Nitrate reductase activity increased at high Ci from 0.3 ± 0.2 to 1.7 ± 0.4 μmolNO2 - g-1 FW h-1indicating that reduction and assimilation of nitrate were uncoupled. The response of photosynthesis to increasing irradiance, estimated from O2evolution vs. irradiance curves, was affected by the treatments. Maximum quantum yield (Φ O2°) and effective quantum yield (Φ O2) at 150 μmol photon m-2s-1 decreased by 20% and 50%, respectively, at low Ci. These differences could be due to changes in photosynthetic electron flow between PSII and PSI. Treatments also produced changes in maximal (Fv/Fm) and effective (ΔF/Fm′)quantum yield for photosystem II charge separation.

    Notes: Abstract A study was made of the tolerance to Cu of 11 strains of Cyanophyceae and 7 strains of eukaryotes. These had all been tested within 6 months after isolation for their photosynthetic activity when exposed to Cu (Takamuraet al., 1989) and had repeatedly been subcultured in the medium without Cu for 2 years. Photosynthetic measurements were made in two ways: precultured in medium without Cu or precultured (for one subculture) in medium containing Cu (645 μg 1−1). The results were compared with those obtained within 6 months of isolation. The tolerance of the eukaryotes did not change significantly in any case, but most strains of Cyanophyceae lost their tolerance to Cu within a few subcultures in medium without Cu; however tolerance recovered following one subculture in medium containing an intermediate level of Cu. This rapid adaptation cannot be explained by a constitutive mutation.

    Notes: Abstract The net photosynthesis of the Mediterranean brown seaweedCystoseira barbata f.repens is measured according to irradiance, temperature and salinity. There is not only, a good utilization of low light intensities (light-shade adaptation), but also a specific ability to use a broad range of irradiance, which corresponds in the photosynthesis-irradiance curves to a high initial slope and an extended light saturation level from 300 to 1500 μmol photon m−2 s−1; only very high irradiances induce photoinhibition. Maximum net photosynthesis occurred at temperatures ranging from 20 °C to 30 °C. The alga tolerates not only a low level of salinity, but also a slight increase in salinity; however, at more than 47.5 g 1−1 NaCl, oxygen exchange is significantly reduced. Light, temperature and salinity requirements are discussed, taking into account ecological considerations. Yields and quality of alginic acid are presented according to the irradiance and yearly evolutionin situ in order to aid future cultivation of this species.

The production potential of two strains of P. purpurea and P. umbilicalis from north-east USA was evaluated by measuring rates of photosynthesis (as O2evolution) of material grown at 20 °C. Photosynthetic rates of P. umbilicalis were 80%higher than P. purpurea over the temperature range 5–20 °C, at both sub-saturating andsaturating irradiances (37 and 289 μmol photonm-2 s-1).     Notes: Abstract The effects of nutrients on the photosynthetic recovery of Nostoc flagelliforme during re-hydration were investigated in order to see if their addition was necessary. Net photosynthesis was negligible in distilled water without nutrient-enrichment. Addition of K+ resulted in significant enhancement of net photosynthesis, whereas other nutrients (Fe3+, Mg2+, Na+, NO3 -, PO4 3-, Cl-) and trace-metals (A5) showed little effect. The recovered net photosynthetic activity increased with the increased K+, and reached the maximum at concentrations above 230 μM. Desiccation and re-hydration did not affect the dependence of photosynthetic recovery on K+. It was concluded that dried field populations of N. flagelliforme require exogenous addition of potassium for photosynthetic recovery and that growth may be potassium-limited in nature.

    Notes: Abstract A strain selection procedure using Gracilaria verrucosa gametophytic sporelings was found to be an efficient tool for the improvement of Gracilaria strains. Two strains, C-2 and A-18, which were isolated and grown clonally, showed higher growth rates under high and low temperature conditions, respectively, than the local Gracilaria conferta. Growth rate, photosynthesis and chlorophyll, which were measured under different temperature and photon flux densities, demonstrated an overall advantage of the selected strains over the wild type strains of both G. verrucosa and G. conferta. Growth rates were also generally in positive correlation with the carboxylase activity of Rubisco. The G. verrucosa wild type also had a 40% higher agar content than G. conferta. The selected strains thus showed higher potential for outdoor cultivation than local wild type populations.

    Notes: Abstract Factors controlling the CO2 system parameters, including the partial pressure of CO2 (PCO2) in coral reef waters, were investigated in three mid-oceanic reefs of the Indo-Pacific region. Surface water PCO2 in the lagoons of Majuro Atoll and Palau barrier reef in the Pacific were 25 µatm and 48 µatm higher than those of the offshore waters, respectively, while South Male Atoll lagoon of the Maldives in the Indian Ocean exhibited relatively small difference in PCO2 compared to the offshore water. Observations from Majuro Atoll and Palau barrier reef are consistent with the view that calcium carbonate production predominates in coral reefs. On the other hand, results from South Male Atoll can be attributed to the thorough flushing of the lagoon, which is connected to the open ocean by numerous deep channels. The offshore-lagoon PCO2 difference depends on system-level net organic-to-inorganic carbon production ratio while reef topography, especially residence time of the lagoon, has a secondary effect on the magnitude of the offshore-lagoon difference. A potential for releasing CO2 might be more evident in an enclosed atoll where the reef water has a longer residence time. Oceanic atoll and barrier reef lagoons, which are in the terminal stage of evolutionary history of oceanic volcanic islands, have the potential to release CO2 to the atmosphere.

In vroege stadia van de groei werden stomataire geleiding, transpiratie en netto fotosynthese bij lichtverzadiging (PAR〉300 W m−2) gemeten aan individuele bladeren en met een mobiel instrumentarium met behulp van gewaskappen. Er werden geen significante verschillen gevonden in de waarden van de stomataire geleiding en de gasuitwisslingskarakteristieken als gevolg vanV. dahliae-besmetting tot een maand na opkomst. Daarna leidde infectie metV. dahliae tot een afname van de stomataire geleiding, transpiratie en netto fotosynthese, speciaal bij oudere bladeren en bij planten die meer aan zonlicht waren blootgesteld. Soms vertoondeV. dahliae interactie met droogte en bleken beide effecten minder dan optelbaar. De hoge waarden van de variatiecoëfficiënten maakten een groot aantal metingen per behandeling noodzakelijk; dit was vooral het geval bij metV. dahliae geïnfecteerde planten hetgeen aantoont datV. dahliae vooral in het begin van de groei niet alle bladeren in gelijke mate aantast. Door de matigende invloed van de integratie van alle bladlagen en mogelijk doordat de bovenste bladeren werden gestimuleerd, werd de totale gewasfotosynthese in mindere mate beïnvloed doorV. dahliae dan de individuele bladfotosynthese.     Notes: Abstract Potato (Solanum tuberosum L.) plants cv.Saturna were subjected to infection withVerticillium dahliae and drought stress. At the early stages of growth, stomatal conductance, transpiration and net photosynthesis were measured at light saturation (PAR〉300 m−2) on individual leaves and with mobile field equipment with the aid of field enclosures. No significant changes in stomatal conductance and gas exchange characteristics occurred as a result ofV. dahliae instomatal conductance, transpiration and and photosynthetic rates, especially on older leaves and on plants exposed to direct sunlight for a longer period of time. In combination with drought,V. dahliae only occasionally showed interaction; their effects being less than additive. High values of coefficients of variatoon necessitated a high number of measurements per treatment; the more so in the inoculated plants which shows thatV. dahliae seems to affect certain leaves while not affecting others early in growth. Crop photosynthesis was less reduced byV. dahliae than individual leaf photosynthesis due to the levelling effect of integration over the whole canopy and possibly through a stimulation of the top leaves. The upper non-affected leaves are responsible for the bulk of photosynthetic crop activity. The results indicate that following an infection withV. dahliae photosynthesis is reduced early in growth as a result of drought stress in the leaves.

    Notes: Abstract The growth response of white clover (Trifolium repens L.) to the expected increase in atmospheric partial pressure of CO2 (pCO2) may depend on P availability. A decrease in the rate of transpiration due to increased pCO2 may reduce the amount of P transported to the shoot, thereby causing a change in the partitioning of P between the root and shoot. To test these hypotheses, four concentrations of P in the nutrient solution, combined with two pCO2 treatments, were applied to nodulated white clover plants. Compared to ambient pCO2 (35 Pa), twice ambient pCO2 (70 Pa) reduced the rate of transpiration but did not impair the total P uptake per plant. However, at twice ambient pCO2 and a moderate to high supply of P, concentrations of structural P and soluble P (Pi) were lower in the leaves and higher in the roots. The activity of root acid phosphatase was lower at twice ambient pCO2 than at ambient pCO2; it depended on the Pi concentration in the roots. At the highest P concentration, twice ambient pCO2 stimulated photosynthesis and the growth rate of the plant without affecting the concentration of nonstructural carbohydrates in the leaves. However, at the lower P concentrations, plants at twice ambient pCO2 lost their stimulation of photosynthesis in the afternoon, they accumulated nonstructural carbohydrates in the leaves and their growth rate was not stimulated; indicating C-sink limitation of growth. P nutrition will be crucial to the growth of white clover under the expected future conditions of increased pCO2.

    Notes: Abstract Phosphorus (P) deficiency limits the yield of wheat, particularly by reducing the number of ears per unit of area because of a poor tiller emergence. The objectives of this work were to (i) determine whether tiller emergence under low phosphorus availability is a function of the availability of assimilates for growth or a direct result of low P availability, (ii) attempt to establish a quantitative relation between an index of the availability of P in the plant and the effects of P deficiency on tiller emergence, and (iii) to provide a better understanding of the mechanisms involved in tiller emergence in field-grown wheat. Wheat (Triticum aestivum L., cv. INTA Oasis), was grown in the field under drip irrigation on a typic Argiudol, low in P (5.5 μg P g-1 soil Bray & Kurtz I) in Balcarce, Argentina. Treatments consisted of the combination of three levels of P fertilization 0, 60 and 200 kg P2O5 ha-1, and two levels of assimilate availability, a control (non-shaded) and 65% of reduction in incident irradiance from seedling emergence until the end of tillering (shaded). Phosphorus treatments significantly modified the pattern of growth and development of the plants. Shading reduced the growth and concentration of water-soluble carbohydrates in leaves and stems. Leaf photosynthetic rate at saturating irradiance was reduced by P deficiency, but was not affected by shading. At shoot P concentrations less than 4.2 g P kg-1 the heterogeneity in the plant population increased with respect to the number of plants bearing a certain tiller. At a shoot P concentration of 1.7 g P kg-1 tillering ceased completely. Phosphorus deficiency directly altered the normal pattern of tiller emergence by slowing the emergence of leaves on the main stem (i.e. increasing the phyllochron), and by reducing the maximum rate of tiller emergence for each tiller.

    Notes: Abstract The membrane-bound photooxidizable cytochrome c-554 from Chloroflexus aurantiacus has been purified. The purified protein runs as a single heme staining band on SDS-PAGE with an apparent molecular mass of 43 000 daltons. An extinction coefficient of 28 ± 1 mM−1 cm−1 per heme at 554 nm was found for the dithionite-reduced protein. The potentiometric titration of the hemes takes place over an extended range, showing clearly that the protein does not contain a single heme in a well-defined site. The titration can be fit to a Nernst curve with midpoint potentials at 0, +120, +220 and +300 mV vs the standard hydrogen electrode. Pyridine hemochrome analysis combined with a Lowry protein assay and the SDS-PAGE molecular weight indicates that there are a minimum of three, and probably four hemes per peptide. Amino acid analysis shows 5 histidine residues and 29% hydrophobic residues in the protein. This cytochrome appears to be functionally similar to the bound cytochrome from Rhodopseudomonas viridis. Both cytochrome c-554 from C. aurantiacus and the four-heme cytochrome c-558-553 from R. viridis appear to act as direct electron donors to the special bacteriochlorophyll pair of the photosynthetic reaction center. They have a similar content of hydrophobic amino acids, but differ in isoelectric point, thermodynamic characteristics, spectral properties, and in their ability to be photooxidized at low temperature.

    Notes: Abstract This study investigated the basis of the negative relationship between leaf size and photosynthetic rate per unit of area among five cultivars of soybeans. Exposure of developing mainstem leaves to light, and sizes and light saturated photosynthesis rates of those leaves at maturity were compared in cultivars grown in field plots for two years at Beltsville, Maryland, USA. Plants were grown both in stands at 2.5 cm by 1 m spacing and as isolated plants. While cultivar differences in leaf size were large and consistent in both planting arrangements, significant cultivar differences in light saturated photosynthetic rates were found only in plants grown in stands. Similarly, leaf size was significantly correlated with specific leaf weight only for plants grown in stands. The mainstem apex and developing mainstem leaves experienced more severe shading in large-leaved cultivars than in small-leaved cultivars when plants were grown in stands. Thus, cultivar differences in photosynthetic capacity were probably a consequence of differences in the exposure of developing leaves to light.

The energy storage rate as evaluated by the half-saturation measuring beam intensity (i50) was inhibited by 3-(3,4-dichlorophenyl)-1,1 dimethylurea, by heat inactivation or by artificial electron acceptors specific for photosystem I or photosystem II; and was activated by electron donors to photosystem I. The reactions involving both photosystems were all characterized by a similar maximal energy storage yield of 16±2 percent. The data could be interpreted if we assumed that the energy storage elicited by the photosystems at 35 Hz is detected at the level of the plastoquinone pool.

    Notes: Abstract An autobiographic report of studies on the elucidation of the structure of the chloroplast is presented here. It deals with the discovery of the lamellar structure of chloroplasts by polarization-microscopy, the representation of their layer-like structure with the ultraviolet microscope and the results obtained by the electron-microscope leading to the discovery of the structural elements of the lamellar system. These lamellar structures were in the form of vesicles, and were named thylakoids. Isolation of the chloroplasts made it possible to determine their chemical composition. Amphiphilic lipids, together with water, create bimolecular layers and, therefore, are responsible for the structure of the thylakoid-membranes. The role of membrane proteins was emphasized. The isolation of the individual polypeptides was possible after dissociation in sodium-dodecyl-sulfate. Antisera to these polypeptides were used to localize them in the membrane. These antisera are able to inhibit special steps in the electron transport. Our results of the spectroscopic examination of photosynthetic membrane components are also discussed.

    Notes: Abstract The binding of the herbicide atrazine to thylakoid membranes is often used to quantify Photosystem II reaction centres. Two atrazine binding sites, with high and low affinities, have been observed on the D1 and D2 polypeptides of Photosystem II, respectively (McCarthy S., Jursinic P. and Stemler A. (1988) Plant Physiol. 86S:46). We have observed that the accessibility of the low-affinity binding sites is variable, being limited in freshly isolated thylakoids or in fresh frozen-thawed thylakoids, but increasing during storage of the membranes on ice. In contrast, the accessibility of the high-affinity binding sites, which are titratable at low concentrations (〈 500 nM) of herbicide, is much less variable, although the dissociation constant is greatly influenced by ethanol. We conclude that to quantify Photosystem II reaction centres by atrazine binding, it is sufficient and more reliable to assay only the high-affinity binding sites.

    Notes: Abstract The unicellular photoheterotrophic cyanobacterium Synechococcus sp. PCC 7002 was shown to encode two genes for the Photosystem II reaction center core protein D2 and one gene for the reaction center chlorophyhll-binding protein CP43. These three genes were cloned and their DNA sequences determined along with their flanking DNA sequences. Northern hybridization experiments show that both genes which encode D2, psbD1 and psbD2, are expressed at roughly equivalent levels. For each of the two psbD genes, there are 18 nucleotide differences among the 1059 nucleotides which are translated. The DNA sequences surrounding the coding sequences are nearly 70% divergent. Despite the DNA sequence differences in the genes, the proteins encoded by the two genes are predicted to be identical. The proteins encoded by psbD1 and psbD2 are ∼92% homologous to other sequenced cyanobacterial psbD genes and ∼86% homologous to sequenced chloroplast-encoded psbD genes. The single gene for CP43, psbC, overlaps the 3′ end of psbD1 and is co-transcribed with it. Results from previous sequencing of psbC genes encoded by chloroplasts suggest that the 5′ end of the psbC gene overlaps the 3′ end of the coding sequence of psbD by ∼50 nucleotides. In Synechococcus sp. PCC 7002, the methionine codon previously proposed to be the start codon for psbC is replaced by an ACG (threonine) codon. We propose an alternative start for the psbC gene at a GTG codon 36 nucleotides downstream from the threonine codon. This GTG codon is preceded by a consensus E. coli-like ribosome binding sequence. Both the GTG start codon and its preceding ribosome binding sequence are conserved in all psbC genes sequenced from cyanobacteria and chloroplasts. This suggests that all psbC genes start at this alternative GTG codon. Based on this alternative start codon, the gene product is ∼85% identical to other cyanobacterial psbC gene products and ∼77% identical to eucaryotic chloroplast-encoded psbC gene products.

    Notes: Abstract The effect of photon flux density on inorganic carbon accumulation and photosynthetic CO2 assimilation was determined by CO2 exchange studies at three, limiting CO2 concentrations with a ca-1 mutant of Chlamydomonas reinhardiii. This mutant accumulates a large internal inorganic carbon pool in the light which apparently is unavailable for photosynthetic assimilation. Although steady-state photosynthetic CO2 assimilation did not respond to the varying photon flux densities because of CO2 limitation, components of inorganic-carbon accumulation were not clearly light saturated even at 1100 μmol photons m-2 s-1, indicating a substantial energy requirement for inorganic carbon transport and accumulation. Steady-state photosynthetic CO2 assimilation responded to external CO2 concentrations but not to changing internal inorganic carbon concentrations, confirming that diffusion of CO2 into the cells supplies most of the CO2 for photosynthetic assimilation and that the internal inorganic carbon pool is essentially unavailable for photosynthetic assimilation. The estimated concentration of the internal inorganic carbon pool was found to be relatively insensitive to the external CO2 concentration over the small range tested, as would be expected if the concentration of this pool is limited by the internal to external inorganic carbon gradient. An attempt to use this CO2 exchange method to determine whether inorganic carbon accumulation and photosynthetic CO2 assimilation compete for energy at low photon flux densities proved inconclusive.

    Notes: Abstract The effect of ultraviolet light on thermoluminescence, oxygen evolution and the slow component of delayed light has been investigated in chloroplasts and Pothos leaves. All peaks including peak V (48°C) were inhibited by UV. However, the peak at 48°C which was induced by DCMU was enhanced following UV irradiation of chloroplasts at ambient temperature (23°C) whereas peak II (-12°C) and peak III (10°C) which were also induced by DCMU were inhibited. Chloroplasts treated with DCMU and dark incubated for several minutes at ambient temperature prior to recording of glow curves have also shown enhancement of peak at 48°C. A slow component of delayed light and photosystem II activity of chloroplasts were inhibited by UV whereas photosystem I activity was marginally affected. These results corroborate involvement of photosystem II in generating thermoluminescence and slow components of delayed light in photosynthetic materials.

Williams 82 and A3127] plants were grown in the field under long-term soil moisture deficit and irrigation to determine the effects of severe drought stress on the photosynthetic capacity of soybean leaves. Afternoon leaf water potentials, stomatal conductances, intercellular CO2 concentrations and CO2-assimilation rates for the two soil moisture treatments were compared during the pod elongation and seed enlargement stages of crop development. Leaf CO2-assimilation rates were measured with either ambient (340 μl CO2 l−1) or CO2-enriched (1800 μl CO2 l−1) air. Although seed yield and leaf area per plant were decreased an average of 48 and 31%, respectively, as a result of drought stress, leaf water potentials were reduced only an average of 0.27 MPa during the sampling period. Afternoon leaf CO2-assimilation rates measured with ambient air were decreased an average of 56 and 49% by soil moisture deficit for Williams 82 and A3127, respectively. The reductions in leaf photosynthesis of both cultivars were associated with similar decreases in leaf stomatal conductance and with small increases in leaf intercellular CO2 concentration. When the CO2-enriched air was used, similar afternoon leaf CO2-assimilation rates were found between the soil moisture treatments at each stage of crop development. These results suggest that photosynthetic capacity of soybean leaves is not reduced by severe soil moisture deficit when a stress develops gradually under field conditions.

    Notes: Abstract Simultaneous, non-invasive measurements were made of the rate of photosynthetic CO2 fixation and the state of activation of the chloroplast CF1CF0-ATP synthase (CF) in field-grown sunflower (Helianthus annuus L.) during the dark-to-light transition at sunrise. CO2 fixation showed a linear response with light intensity from zero to about 500–700 μE m-2 s-1. However, at light intensities of only 5–22 μE m-2 s-1, the energetic threshold for activation of the CF was found to be significantly lowered (as compared to the pre-dawn state), presumably through reduction of the regulatory sulfhdryl groups of the γ-subunit of the CF. When these studies were extended to chamber-grown plants, it was found that as little as 5 seconds of illumination at 4 μE m-2 s-1 caused apparently full CF reduction. It is clear, therefore, that the catalytic activation of CF is not rate limiting to the induction of carbon assimilation under field conditions during a natural dark-to-light transition at sunrise. A model, based on the redox properties of the regulatory sulfhydryls, was developed to examine the significance of sulfhydryl midpoint potential in explaining the differences in light sensitivity and oxidation and reduction kinetics, between the CF and other thioredoxin-modulated chloroplast enzymes. Computer simulations of the light-induced regulation of three representative thioredoxin-modulated enzymes are presented.

    Notes: Abstract Short-term studies for comparing some primary metabolic and growth-responses to salt stress in seedlings of two maize genotypes differing in drought resistance were carried out under controlled conditions. Both genotypes revealed high yielding ability in favourable environments. Treatments: Control (Hoagland-Arnon No 1 solution) and salt stress (Hoagland-Arnon solution plus NaCl, Ψs = −0.84MPa). It was found that in both genotypes the activity of the principal metabolic pathway supplying reduced nitrogen (15N) for the synthesis of amino acids and proteins as well as the assimulatory number (14CO2—assimilation relation rate per chlorophyll unit) were decreased under the effect of the stress. These effects were more marked in the resistant genotype. In this genotype the stress induced metabolic activity decline was accompanied by a corresonding reduction of the relative growth rate. Conversely, continuing growth, resulting probably from accumulation of solutes, was observed in the susceptible genotype. On the basis of these and other observations it is assumed that the resistant genotype manifests short-term energy saving stress reactions.

    Notes: Abstract cDNA and the corresponding genomic DNA region encoding Rubisco activase were isolated from the unicellular green alga Chlorococcum littorale. The deduced amino acid sequence encoded by the cDNA was 403 amino acids long and exhibited important homology with those of other known Rubisco activases. Its N-terminal sequence was similar to the chloroplastic transit peptides in Chlamydomonas reinhardtii. The mature protein had a predicted molecular mass of 42 kDa. Five introns were located inside the genomic gene encoding Rubisco activase (rca). Genomic Southern blots indicated that two copies of the rca gene were present in the genome of C. littorale. The level of rca messenger RNA increased when cells of C. littorale were subjected to high-CO2 stress (i.e. grown under at least 20% CO2). Hsp70 heat-shock protein was also induced under high-CO2 conditions and, as expected, was also induced at 35 °C. The rca gene, in contrast, was not induced at 35 °C, indicating that this gene was induced in response to the high CO2 concentration and not to general stress. A search of the promoter-binding proteins by a gel retardation assay showed that, under the high-CO2 conditions, a protein(s) which was probably an activator of the rca transcription was synthesized.

    Notes: Abstract An intrinsic divinyl-chlorophyll a/b antenna and a particular form of phycobiliprotein, phycoerythrin (PE) III, coexist in the marine oxyphotobacterium Prochlorococcus marinus CCMP 1375. The genomic region including the cpeB/A operon of P. marinus was analysed. It encompasses 10 153 nucleotides that encode three structural phycobiliproteins and at least three (possibly five) different polypeptides analogous to cyanobacterial or red algal proteins involved either in the linkage of subunits or the synthesis and attachment of chromophoric groups. This gene cluster is part of the chromosome and is located within a distance of less than 110 kb from a previously characterized region containing the genes aspA-psbA-aroC. Whereas the Prochlorococcus phycobiliproteins are characterized by distinct deletions and amino acid replacements with regard to analogous proteins from other organisms, the gene arrangement resembles the organization of phycobiliprotein genes in some other cyanobacteria, in particular marine Synechococcus strains. The expression of two of the Prochlorococcus polypeptides as recombinant proteins in Escherichia coli allowed the production of individual homologous antisera to the Prochlorococcus α and β PE subunits. Experiments using these sera show that the Prochlorococcus PEs are specifically associated to the thylakoid membrane and that the protein level does not significantly vary as a function of light irradiance or growth phase.

The aim of the investigations presented here was to identify some contrasts between short and tall genotypes in terms of the physiological characteristics associated with grain yield, so as to orientate more efficiently the selection of genotypes, with or without dwarfism genes, for productivity. Various parameters of flag leaf functioning (photosynthesis rate, chlorophyll fluorescence index, leaf area duration) were related to the biological and economic yields and the harvest index for two groups of genotypes that were differentiated by their height. For all genotypes, the relationships between the various traits and the grain yield were difficult to ascertain. For the tall genotypes without dwarfism genes, the classical relationships between grain yield, harvest index, flag leaf area duration and net photosynthesis rate were confirmed. Moreover, the rate of chlorophyll fluorescence decrease (Rfd) during the slow Kautsky kinetics phase, which is representative of the leaf photosynthesis at low light, was found to be an excellent marker of economic yield. Chlorophyll fluorescence decrease was closely related to grain yield and also with other factors that are known to be important in its expression (harvest index, flag leaf area duration). In very short genotypes, the biological yield and directly related factors (leaf area, plant height) were the main parameters associated with economic yield, since the harvest index had approached its upper limit. The selection of short genotypes must therefore maintain the biological yield through an increase in the size of the aerial organs to counterbalance the decrease in height.

    Notes: Abstract In the last few years our knowledge of the structure and function of Photosystem II in oxygen-evolving organisms has increased significantly. The biochemical isolation and characterization of essential protein components and the comparative analysis from purple photosynthetic bacteria (Deisenhofer, Epp, Miki, Huber and Michel (1984) J Mol Biol 180: 385–398) have led to a more concise picture of Photosystem II organization. Thus, it is now generally accepted that the so-called D1 and D2 intrinsic proteins bind the primary reactants and the reducing-side components. Simultaneously, the nature and reaction kinetics of the major electron transfer components have been further clarified. For example, the radicals giving rise to the different forms of EPR Signal II have recently been assigned to oxidized tyrosine residues on the D1 and D2 proteins, while the so-called Q400 component has been assigned to the ferric form of the acceptor-side iron. The primary charge-separation has been meaured to take place in about 3 ps. However, despite all recent major efforts, the location of the manganese ions and the water-oxidation mechanism still remain largely unknown. Other topics which lately have received much attention include the organization of Photosystem II in the thylakoid membrane and the role of lipids and ionic cofactors like bicarbonate, calcium and chloride. This article attempts to give an overall update in this rapidly expanding field.

    Notes: Abstract Thermal emission and photochemical energy storage were examined in photosystem I reaction center/core antenna complexes (about 40 Chl a/P700) using photoacoustic spectroscopy. Satisfactory signals could only be obtained from samples bound to hydroxyapatite and all samples had a low signal-to-noise ratio compared to either PS I or PS II in thylakoid membranes. The energy storage signal was saturated at low intensity (half saturation at 1.5 W m-2) and predicted a photochemical quantum yield of 〉90%. Exogenous donors and acceptors had no effect on the signal amplitudes indicating that energy storage is the result of charge separation between endogenous components. Fe(CN)6 -3 oxidation of P700 and dithionite-induced reduction of acceptors FA-FB inhibited energy storage. These data are compatible with the hypothesis that energy storage in PS I arises from charge separation between P700 and Fe-S centers FA-FB that is stable on the time scale of the photoacoustic modulation. High intensity background light (160 W m-2) caused an irreversible loss of energy storage and correlated with a decrease in oxidizable P700; both are probably the result of high light-induced photoinhibition. By analogy to the low fluorescence yield of PS I, the low signal-to-noise ratio in these preparations is attributed to the short lifetime of Chl singlet excited states in PS I-40 and its indirect effect on the yield of thermal emission.

    Notes: Abstract Photoacoustic detection of oxygen evolution and Emerson enhancement in state 1 and state 2 were compared in a tobacco wild type and mutant (Su/su) deficient in chlorophyll. The mutant shows smaller changes in the distribution of excitation energy between the two photosystems than the wild type. Analysis of Emerson enhancement saturation curves indicates that in the mutant which is deficient in grana partitions and shows less stacking, state 1-state 2 transitions reflect changes in the yield of energy transfer from PS II to PS I (spillover). On the other hand, the wild type containing large grana shows changes in absorption cross-sections of the two photosystems upon state transitions. NaF, a specific phosphatase inhibitor, blocks the transition to state 1, indicating that LHC II phosphorylation has a role in excitation energy regulation in both the mutant as well as the wild type. It is demonstrated that N-ethylmaleimide, a specific sulfhydryl reagent, blocks the transition to state 2, suggesting that a disulfide-sulfhydryl redox couple activates the LHC II kinase in vivo.

    Notes: Abstract A new type of computer controlled spectrophotometer is described which is based on an array of independent, monochromatic pulsed light sources consisting of light emitting diodes (LED) equipped with narrow band interference filters. The LEDs are sequentially pulsed at a high repetition rate. The absorbance information at specific wavelengths is sampled in the μs-time range, using a computer-controlled, highly selective technique of synchronous amplification. A first prototype of this LED Array Spectrophotometer allows simultaneous recording of kinetic changes at 16 different wavelengths in the range from 530 to 600 nm, with a time resolution of 1 ms/point. Special features of the new type of spectrophotometer are: Weak integrated measuring light intensity, high signal/noise ratio even with scattering samples like intact leaves, active baseline adjustment by LED current regulation, computer control of system operation and data analysis. To deconvolute the complex absorbance changes in the cytochrome α-band region, ‘standard spectra’ of the major components are stored in computer memory and used for curve fitting of difference spectra and kinetic changes. As an example of application, the light-induced absorbance changes in a heat-pretreated spinach leaf are analysed. The system effectively separates specific absorbance changes of C550, cyt f, cyt b 559 and cyt b 563 from a large background of non-specific changes.

The instrument utilizes a black and white CCD video camera detector, a commercial video imaging board and a IBM-AT compatible computer. The color output of the imaging board greatly aids in the users ability to visually discriminate areas of interest in the video field. Software development that enables the user to capture kinetic traces in real time from the video images is also described. The system is used to monitor fluorescence from photosynthetic systems. The usefulness of the system in screening for photosynthetic mutants is also demonstrated. The cost of the system can be kept below $12,000.

    Notes: Abstract Wild emmer wheat (Triticum dicoccoides Korn) has shown wide genetic diversity for disease resistance and morpho-physiological traits of economic importance. Our objectives were to test for genetic variation (VG) in photosynthetic characteristics residing within and between native populations sampled from three ecogeographical regions of Israel, and to identify potential sources of high photosynthetic efficiency for future wheat improvement. Accessions sampled in the center of wild emmer distribution (upper Jordan Valley) in a relatively narrow geographical range showed the greatest diversity in CO2-assimilation rate per unit leaf area (A) or per unit chlorophyll (A/Chl). Genetic variation was absent for internal CO2 concentration (Ci) and water-use efficiency (WUE) and generally lacking for stomatal conductance (gs). Leaf area, although quite variable, was not a significant cofactor in assessing genetic potential for photosynthesis. Accessions within a given population showed 10-times more variation in A and A/Chl than populations sampled from different locations in a region. Accessions with the highest photosynthetic efficiency were derived from upland steppic populations located in marginal habitats extending southward into Israel. Some accessions having high photosynthetic capacity (A=32 μmol m-2 s-1) with no significant reduction in leaf size constitute a potentially valuable genetic resource yet untapped for genetic improvement of hexaploid (T. aestivum L.) wheat.

    Notes: Abstract A newly developed photoacoustic system for measurement of photosynthetic reactions in intact leaves is described. The system is based on pulsed light-emitting diodes, the pulse program and pulse response analysis being computer controlled. Separation of various components in the overall photoacoustic signal is achieved by curve fitting analysis of the responses following individual measuring light pulses in the millisecond time domain. This procedure is in distinction to the conventionally used analysis in the frequency domain, with the advantage that various signal components are obtained by on-line deconvolution, yielding simultaneous recordings of photothermal (complement of energy storage) and photobaric (evolution and uptake) signals. The basic components of the new system are described by block diagrams and the principal steps for deconvolution of the overall photoacoustic response are outlined. An example of application with simultaneous recording of chlorophyll fluorescence is given. It is apparent that the photobaric uptake component represents a significant part of the overall signal, particularly during induction of photosynthesis after dark-adaptation. This component probably contains not only O2-uptake but uptake of CO2 as well.

    Notes: Abstract Several leaf anatomical and ultrastructural characteristics usually related with photosynthetic capacity were examined in two Flaveria species with strong differences in anatomy and their F1 hybrid. Flaveria trinervia (Spreng.) Mohr (C4) was the female parent and F. brownii A.M. Powell (C4-like) was the male parent. Quantitative anatomical analysis was made on transverse sections of leaves at both the light and electron microscope level. Four kinds of photosynthetic tissues were considered: bundle sheath (BS), mesophyll adjacent to the BS, mesophyll not adjacent to the BS, and larger spongy mesophyll cells. Flaveria trinvervia partitioned a larger proportion of its photosynthetic cells to BS and the mesophyll layer adjacent to BS and also possessed larger chloroplasts, especially in BS, than did F. brownii. These results suggest that although F. brownii is very C4-like, its anatomy is not as completely C4 as is the case for F. trinervia. In the F1 hybrid the relative contribution of the different tissues to the total photosynthetic tissue volume and area per unit leaf area was quite similar to that of F. trinervia. On the other hand, the chloroplast density and size of the F1 hybrid were fairly similar to those of F. brownii, especially in BS. Thus, there was no evidence of maternal inheritance in the chloroplast characteristics studied. A negative correlation (P〈0.05) between chloroplast size and density was observed among species and relicates within each kind of tissue. This correlation was highest (r=−0.94, P〈0.001) for the BS and when values were plotted on a logarithmic scale. Thus, higher chloroplast numbers for F. brownii and the F1 hybrid were offset by larger chloroplasts in F. trinervia. Less complete C4 photosynthesis in F. brownii may be partially due to incomplete development of Kranz anatomy usually associated with C4 photosynthesis.

    Notes: Abstract The physical and chemical environment, and the phytoplankton primary production of southeastern Brazil were studied in relation to the general oceanographic structure during two research cruises (winter and summer). In each cruise, a total of 91 stations were occupied. Data were collected on the spatial distribution of nutrients, phytoplankton biomass and photosynthetic capacity over the coastal, shelf and oceanic areas off São Paulo, Paraná and Santa Catarina States. During wintertime, the mixing processes between tropical warm waters of the Brazil Current and subantarctic waters of the Malvinas Current formed strong environmental gradients. The drainings of Rio de La Plata and Lagoa dos Patos are transported northwards by coastal currents, enriching the shelf waters off Santa Catarina State with inorganic nutrients and consequently increasing the chlorophyll ‘a’ to the highest concentrations (〉 3.5 mg m −3) measured during the two cruises. In slope waters chlorophyll values were always low (0.05–0.45 mg m −3). The chlorophyll within the euphotic layer varied from 8.8–36.7 and 1.2–18.5 mg m−2 during winter and summer, respectively. The surface photosynthetic rates during winter and summer cruises ranged respectively from 0.21–9.17 and 0.66–19.60 mgC/mgChl.a/h. The mean rates were higher in nearshore waters and decreased seaward. The thermal structure of the water column affected the vertical distribution of chlorophyll a and photosynthesis within the euphotic zone; During unstratified periods (winter) they were uniformly distributed but the occurrence of subsurface peaks of chlorophyll and strong photosynthetic inhibition of low light adapted cells in deeper layers are associated to the seasonal thermocline. Occasionally, upwelling of deep waters from shelf break enriched the deeper euphotic layers in offshore areas. Intensive upwelling was observed off Paranagua Bay (Parana State) and the mechanisms of its formation are discussed.

    Notes: Abstract Photosynthetic rates were compared between Ecklonia cava bladelets with and without zoosporangial sori sampled from the subtidal zone (about 5 m deep) in Nabeta Bay, Shimoda, Japan. Photosynthetic rates of bladelets were lower in the sorus portion than in the non-sorus portion on the basis of area, dry weight and chlorophyll a. Respiration rates were higher in the sorus portion than in the non-sorus portion on the basis of area and chlorophyll a, whereas they were almost the same on a dry weight basis. The differences were mainly due to a large difference in dry weight per unit bladelet area between the sorus and the non-sorus portion. Light compensation points were higher in the sorus portion than in the non-sorus portion.

    Notes: Abstract What follows is a very personal account of my professional life and the early years that preceded it. I have described the social and economic conditions in America and how the nineteen twenties and thirties nurtured our scientific future. The description of the early part of post-World War II research covers my experience in the areas of nutritional biochemistry, biochemical genetics and proceeds to photosynthesis. The latter era lasted around 35 years. For me the most memorable research accomplishments in which I was a participant during this period was the first demonstration of the primary carboxylation enzyme in an in vitro system in algal and higher plants as well to show that it was structurally associated with the chloroplast.Our group while at Oak Ridge and the University of Massachusetts assembled data that described the complete macromolecular assembly of the photosynthetic apparatus of the unusual photosynthetic green bacterium Chloroflexus aurantiacus and created a model of that system which differed greatly from the chomatophore system for the purple bacteria. For the last decade, my scientific journey, with numerous new colleagues has turned to the exciting area of biomaterials.We characterized and modeled the completely new bacterial intracellular inclusions responsible for the synthesis and degredation of biosynthetic, biodegradable and biocompatible bacterial polyesters in the cytoplasm of Pseudomonads.

    Notes: Abstract Unlike the α and β polypeptides of the core light-harvesting complex (LH1) of Rhodobacter (Rb.) sphaeroides, the α and β polypeptides of the peripheral light-harvesting complex (LH2) of this organism will not form a subunit complex by in vitro reconstitution with bacteriochlorophyll. Guided by prior experiments with the LH1 β polypeptides of Rb. sphaeroides and Rhodospirillum rubrum, which defined a set of interactions required to stabilize the subunit complex, a series of mutations to the Rb. sphaeroides LH2 β polypeptide was prepared and studied to determine the minimal changes necessary to enable it to form a subunit-type complex. Three mutants were prepared: Arg at position −10 was changed to Asn (numbering is from the conserved His residue which is known to be coordinated to bacteriochlorophyll); Arg at position −10 and Thr at position +7 were changed to Asn and Arg, respectively; and Arg at position −10 was changed to Trp and the C-terminus from +4 to +10 was replaced with the amino acids found at the corresponding positions in the LH1 β polypeptide of Rb. sphaeroides. Only this last multiple mutant polypeptide formed subunit-type complexes in vitro. Thus, the importance of the C-terminal region, which encompasses conserved residues at positions +4, +6 and +7, is confirmed. Two mutants of the LH1 β polypeptide of Rb. sphaeroides were also constructed to further evaluate the interactions stabilizing the subunit complex and those necessary for oligomerization of subunits to form LH1 complexes. In one of these mutants, Trp at position −10 was changed to Arg, as found in LH2 at this position, and in the other His at position −18 was changed to Val. The results from these mutants allow us to conclude that the residue at the −10 position is unimportant in subunit formation or oligomerization, while the strictly conserved His at −18 is not required for subunit formation but is very important in oligomerization of subunits to form LH1.

    Notes: Abstract A diffusion enrichment technique is presented which allows for chemical enrichment of soft surficial and shallow subsurface sediments and subsequent measurement of O2 production. The sediment is enriched by inserting a perforated tube containing dialysis tubing filled with a nutrient/agar mixture. O2 production by surficial sediment is measured using an inverted, translucent, polyethylene chamber over the sediment. The inside of the chamber contains a collapsible bag connected to the water outside the chamber. When water overlying the sediment is withdrawn from a sampling port, it is displaced with water from outside the chamber, thus preventing contamination of water samples with pore water from below. The technique was tested by enriching near-shore sediments in a large oligotrophic lake with inorganic N and P. NHinf4/p+ additions significantly stimulated benthic primary production as measured by 02 production, whereas enrichment with POinf4/3- had no effect.

    Notes: Abstract The local species Gracilaria conferta and the foreign G. verrucosa were grown together under a wide range of photon flux density and temperature conditions. Gracilaria verrucosa showed a higher growth rate, especially under low temperatures, and higher photosynthetic performances as well as higher ribulose-1,5-bisphosphate carboxylase activity as compared with G. conferta. Gracilaria verrucosa also showed a better quality and yield of agar, suggesting that this species could be more suitable than G. conferta for outdoor cultivation in Israel and may improve winter growth in ponds. Growth rate and agar quality (gel strength) were rated as the most suitable characteristics influencing the preference of strains for outdoor cultivation.

Physiological responses were considered in relation to vertical distribution patterns of these species commonly observed throughout the Azores.     Notes: Abstract The dependence of photosynthesis on light and temperature is modelled through analysis of transition probabilities of photosystems. In the model, two transition probabilities are functions of light, and one transition probability is a function of temperature. The estimated light-saturated photosynthesis of Ecklonia cava blades at 20 °C was 0.037 mg C cm−2 h−1. The value of the activation energy, the standard enthalpy and the standard entropy were estimated to be 56.5 kJ mol−1, 204 kJ mol−1 and 678 J mol−1 K−1, respectively. A production model (an integral photosynthesis model) for an E. cava stand was developed using the photosynthesis model. Production calculated by the model agreed well with observed data during the growing period of an E. cava stand at a field observation site on the west side of Miura Peninsula, Japan. Results of the analysis of the effects of irradiance and temperature on the production of the E. cava community by the model are: 1. Production decreased with irradiance decrease. The estimated compensation irradiance was 26.5 μmol photons m−2 s−1 when the biomass was 3 kg wet mass m−2 (blade:stipe ratio = 2 kg m−2:1 kg m−2) and the temperature was 20 °C. 2. The optimum temperature decreased when irradiance decreased and when biomass increased. The highest estimated value for the optimum temperature was 24.0 °C. The estimated optimum temperature was 18.2 °C when the biomass was 12 kg wet mass m−2 and the photon irradiance was 200 μmol photons m−2 s−1. 3. The amount of biomass that resulted in the maximum production was influenced by irradiance and temperature. At 400 μmol photons m−2 s−1 and 20 °C, the estimated value of the biomass (blade:stipe = 2:1) giving the maximum pr oduction was about 5.3 kg wet mass m−2. However, at 100 μmol photons m−2 s−1 and 24 °C, the estimated value was about 3.0 kg wet mass m−2. The estimated values of the maximum production under the two conditions were 1.05 and 0.30 g C m−2 h−1, respectively.

    Notes: Abstract In illuminated intact spinach chloroplasts, warming to and beyond 40 °C increased the proton permeability of thylakoids before linear electron transport through Photosystem II was inhibited. Simultaneously, antimycin A-sensitive cyclic electron transport around Photosystem II was activated with oxygen or CO2, but not with nitrite as electron acceptors. Between 40 to 42 °C, activation of cyclic electron transport balanced the loss of protons so that a sizeable transthylakoid proton gradient was maintained. When the temperature of darkened spinach leaves was slowly increased to 40°C, reduction of the quinone acceptor of Photosystem II, QA, increased particularly when respiratory CO2 production and autoxidation of plastoquinones was inhibited by decreasing the oxygen content of the atmosphere from 21 to 1%. Simultaneously, Photosystem II activity was partially lost. The enhanced dark QA reduction disappeared after the leaf temperature was decreased to 20 °C. No membrane energization was detected by light-scattering measurements during heating the leaf in the dark. In illuminated spinach leaves, light scattering and nonphotochemical quenching of chlorophyll fluorescence increased during warming to about 40 °C while Photosystem II activity was lost, suggesting extra energization of thylakoid membranes that is unrelated to Photosystem II functioning. After P700 was oxidized by far-red light, its reduction in the dark was biphasic. It was accelerated by factors of up to 10 (fast component) or even 25 (slow component) after short heat exposure of the leaves. Similar acceleration was observed at 20 °C when anaerobiosis or KCN were used to inhibit respiratory oxidation of reductants. Methyl viologen, which accepts electrons from reducing side of Photosystem II, completely abolished heat-induced acceleration of P700+ reduction after far-red light. The data show that increasing the temperature of isolated chloroplasts or intact spinach leaves to about 40 °C not only inhibits linear electron flow through Photosystem II but also activates Photosystem I-driven cyclic electron transport pathways capable of contributing to the transthylakoid proton gradient. Heterogeneity of the kinetics of P700+ reduction after far-red oxidation is discussed in terms of Photosystem I-dependent cyclic electron transport in stroma lamellae and grana margins.

    Notes: Abstract The effect of the light harvesting 1 (LH1) antenna complex on the driving force for light-driven electron transfer in the Rhodobacter sphaeroides reaction center has been examined. Equilibrium redox titrations show that the presence of the LH1 antenna complex influences the free energy change for the primary electron transfer reaction through an effect on the reduction potential of the primary donor. A lowering of the redox potential of the primary donor due to the presence of the core antenna is consistently observed in a series of reaction center mutants in which the reduction potential of the primary donor was varied over a 130 mV range. Estimates of the magnitude of the change in driving force for charge separation from time-resolved delayed fluorescence measurements in the mutant reaction centers suggest that the mutations exert their effect on the driving force largely through an influence on the redox properties of the primary donor. The results demonstrate that the energetics of light-driven electron transfer in reaction centers are sensitive to the environment of the complex, and provide indirect evidence that the kinetics of electron transfer are modulated by the presence of the LH1 antenna complexes that surround the reaction center in the natural membrane.

    Notes: Abstract The kinetic properties of photosynthesis (both transient and steady-state) were monitored using three non-invasive techniques to evaluate limitations on triose-phosphate (triose-P) conversion to carbohydrate in rice. These included analyzing the O2 sensitivity of CO2 fixation and the assimilatory charge (AC) using gas exchange (estimate of the ribulose 1,5- bisphosphate pool) and measuring Photosystem II activity by chlorophyll fluorescence analysis under varying light, temperature and CO2 partial pressures. Photosynthesis was inhibited transiently upon switching from 20 to 2 kPa O2 (reversed O2 sensitivity), the degree of which was correlated with a terminal, steady-state suppression of low O2 enhancement of photosynthesis. Under current ambient levels of CO2 and moderate to high light, the transient pattern was more obvious at 18 °C than at 26 °C while at 34 °C no tra nsient response was observed. The transient inhibition at 18 °C ranged from 15% to 31% depending on the pre-measurement temperature. This pattern, symptomatic of feedback, was observed with increasing light and CO2 partial pressures with the degree of feedback decreasing from moderate (18 °C) up to high temperature (34 °C). Under feedback conditions, the rate of assimilation is shifted from being photorespiration limited to being triose-P utilization limited. Transitory changes in CO2 assimilation rates (A) under low O2 indicative of feedback coincided with a transitory drop in assimilatory charge (AC) and inhibition of electron transport. In contrast to previous studies with many C3 species, our studies indicate that rice shows susceptibility to feedback inhibition under moderate temperatures and current atmospheric levels of CO2.

    Notes: Abstract The photosynthetic characteristics of the Cymbidium plantlet in vitro cultured on Hyponex-agar medium with 2% sucrose were determined based on the measurements of CO2 concentration inside and outside of the culture vessels. The CO2 measurements were made with a gas chromatograph at a PPF (photosynthetic photon flux) of 35, 102 and 226 μmol m-2 s-1, a chamber air temperature of 15, 25 and 35°C and a CO2 concentration outside the vessel of approximately 350, 1100 and 3000 ppm. The net photosynthetic rates were determined on individual plantlets and were expressed on a dry weight basis. The steady-state CO2 concentration during the photoperiod was lower inside the vessel than outside the vessel at any PPF greater than 35 μmol m-2s-1 and at any chamber air temperature. The photosynthetic response curves relating the net photosynthetic rate, PPF, and CO2 concentration in the vessel and chamber air temperature were similar to those for Cymbidium plants grown outside and other C3 plants grown outside under shade. The results indicate that CO2 enrichment for the plantlets in vitro at a relatively high PPF would promote photosynthesis and hence the growth of chlorophyllous shoots/plantlets in vitro and that the plantlets in vitro would make photoautotrophic growth under environmental conditions favorable for photosynthesis.

    Notes: Interspecific ecophysiological differences in response to different light environments are important to consider in regeneration behavior and forest dynamics. The diurnal changes in leaf gas exchange and chlorophyll fluorescence of two dipterocarps, Shorea leprosula (a high light-requiring) and Neobalanocarpus heimii (a low light-requiring), and a pioneer tree species (Macaranga gigantea) growing in open and gap sites were examined. In the open site, the maximum net photosynthetic rate (Pn), photosystem II (PSII) quantum yield (δ; F/Fm′), and relative electron transport rate (r-ETR) through PSII at a given photosynthetic photon flux density (PPFD) was higher in S. leprosula and M. gigantea than in N. heimii, while non-photochemical quenching (NPQ) at a given PPFD was higher in N. heimii. The maximum values of net photosynthetic rate (Pn) in M. gigantea and S. leprosula was higher in the open site (8–11 μmol m−2 s−1) than in the gap site (5 μmol m−2 s−1), whereas that in N. heimii was lower in the open site (2 μmol m−2 s−1) than in the gap site (4 μmol m−2 s−1), indicating that N. heimii was less favorable to the open site. These data provide evidence to support the hypothesis that ecophysiological characteristics link with plant’s regeneration behavior and successional status. Although Pn and stomatal conductance decreased at midday in M. gigantea and S. leprosula in the open site, both r-ETR and leaf temperature remained unchanged. This indicates that stomatal closure rather than reduced photochemical capacity limited Pn in the daytime. Conversely, there was reduced r-ETR under high PPFD conditions in N. heimii in the open site, indicating reduced photochemical capacity. In the gap site, Pn increased in all leaves in the morning before exposure to direct sunlight, suggesting a relatively high use of diffuse light in the morning.

The thermal stability and decomposition patterns for these compounds were compared and interpreted in terms of structural features such as bond character and steric effects. The kinetic parameters and mechanisms of every decomposition stage involved for all these complexes were obtained employing the non-isothermal kinetic analysis method suggested by Malek et al., which showed the kinetics mechanism for pyrolysis of pyridine(or 4-methylpyridine) is an S-B empirical model with lower activation energy, while that of O,O'-dialkyldithiophosphate is a diffusion model. These results are in accord with the fact that two ligands are of different type.

    Notes: Abstract Human serum albumin (HSA) immersed in pyridine-n-hexane mixtures was analyzed using differential scanning calorimetry (DSC). State of the solid HSA in organic solvent mixtures is the non-equilibrium state which is seen as the exothermic peak on the DSC curves. The enthalpy change corresponding to this exothermic peak approaches zero when going from pure pyridine to pure n-hexane. Dependence of the enthalpy change on the pyridine concentration is suggestive that the non-equilibrium state of the immersed HSA results from the HSA-pyridine interactions 'frozen' at the lower temperature. Most likely the temperature-initiated exothermic peak observed on the DSC curves reflects the swelling of HSA by pyridine.

    Notes: Abstract Crystallization kinetics of Al91La5Ni4 amorphous ribbons produced by a melt-spinning method were studied by DSC analysis and X-ray diffraction. The effect of heating rate (from 4 to 200°C min-1) was investigated in the temperature range from 298 to 700 K. Increases the heating rate from 4 to 200°C min-1 resulted in increases of the temperature difference between the two stages of the transformation process: crystallization of Al and crystallization of the Al compounds from 148.9 to 167.4 K. The apparent activation energies for the first step, related to Al crystallization, and to the second step related to crystallization of Al4La and Al3Ni, were found to be 161±9 and 199±10 kJ mol-1, respectively. The results indicate the possibility of tailoring the heating treatment to produce the required fraction of the amorphous phase.
