Heterocyclizations, IXPreparation of Pyrazolo-, Triazolo-, Oxazolo- and Thiazolo-s-triazines with a Bridge Head Nitrogen and of an Isopurine N-Carboxylic EsterIn the absence of a catalyst, aminoazoles (1, 9) generally add ethoxycarbonyl or benzoyl iso(thio)cyanate at the cyclic or exocyclic nitrogen (→2, 4, 6, 10, 13). In contrast, 1, 9 insert in the presence of basic catalysts (2-aminooxazoline and 2-aminothiazoline, 15a, b, in the absence of catalysts) the iso(thio)cyanate between the cyclic and exocyclic N under formation of azolotriazines (3a, 8a, 11a, 16a, b) with a bridge head nitrogen. If the ring contains an ester group in α position to the NH2 group, the insertion occurs between these two groups to give an azolopyrimidine-N-carboxylic ester (28). In contrast, the reaction of 5-amino-tetrazole (17) and 2-aminobenzimidazole (22) with the isothiocyanates of this series results in the elimination of HSCN and formation of the carboxylamino and benzoylamino derivatives (→20, 21, 23), resp.

Aminoazole (1, 9) addieren gewöhnlich Äthoxycarbonyl- oder Benzoyliso(thio)cyanat spontan am Ring-N bzw. an der exocyclischen Aminogruppe (→2, 4, 6, 10, 13). Dagegen lagern 1, 9 in Gegenwart von basischen Katalysatoren, 2-Amino-oxazolin und 2-Amino-thiazolin (15a, b) ohne Katalysator, das Iso(thio)cyanat zwischen den cyclischen und den Aminstickstoff ein, wobei Azolo-triazine (3a, 8a, 11a, 16a, b) mit Brückenkopfstickstoff gebildet werden. Enthält der Ring eine Estergruppe benachbart zu der Aminogruppe, so wird das Isothiocyanat zwischen diese beiden Gruppen eingebaut, und es entsteht ein Azolo-pyrimidin-N-carbonsäureester (28). 5-Amino-tetrazol (17) und 2-Amino-benzimidazol (22) dagegen werden durch die Senföle dieser Reihe unter HSCN-Abspaltung an der Aminogruppe carboxyliert bzw. benzoyliert (→20, 21, 23).
