Notizen: Abstract. Using several tests for structural stability in regressions with I(1) variables and for the existence of cointegration in models with regime shifts, the empirical evidence on the existence of a structural break in the Spanish long-run demand for broad money (ALP2) is analysed.  The results indicate that shifts affecting the demand for ALP2 in recent years have substantially altered its long-run properties. As to the cause of this structural break, emphasis is placed on the role played by the increasing openness of the Spanish financial system to international markets as obstacles to free capital movements have progressively disappeared.

Notizen: Abstract The Differential System Method (DSM) permits identification of the physical parameters of finite-difference groundwater flow models in a confined aquifer when piezometric head and source terms are known at each point of the finite-difference lattice for at least two independent flow situations for which the hydraulic gradients are not parallel. Since piezometric head data are usually few and sparse, interpolation of the measured data onto a regular grid can be performed with geostatistical techniques. We apply kriging to the sparse data of a synthetic aquifer to evaluate the stability of the DSM with respect to uncorrelated measurement errors and interpolation errors. The numerical results show that the DSM is stable.

Notizen: Abstract Population growth and persistence of Euseius finlandicus (Oudemans), Typhlodromus pyri Scheuten and Kampimodromus aberrans (Oudemans) (Acari: Phytoseiidae) were studied in single-species and two-species systems on apple seedlings primarily infested by Panonychus ulmi Koch (Acari: Tetranychidae) in an environmentally controlled greenhouse. During the experiment, the seedlings developed natural infestations by Tetranychus urticae Koch (Acari: Tetranychidae), Frankliniella occidentalis Pergande (Thysanoptera: Thripidae), and powdery mildew. Several weeks after the start of the experiment a condition of diminishing prey availability was created by use of hexythiazox treatments. Without heterospecific competitors, T. pyri attained a higher population level than E. finlandicus or K. aberrans when similar amounts of food (spider mites) were available to each. Population growth of T. pyri was decisively favoured by the presence of T. urticae. In the single-species systems each predatory species persisted to the end of the experiment in spite of diminishing prey. In two-species systems with T. pyri/E. finlandicus and T. pyri/K. aberrans that were started with the same number of individuals of each species, only T. pyri was left at the end of the experiment. Typhlodromus pyri became more numerous than the other species when prey was abundant (which was in accordance with the results of the single-species groups) and finally displaced E. finlandicus and K. aberrans towards the end of the experiment. The following factors may have contributed to the dominance of T. pyri: (1) the ability of adult females to survive longer without food than those of E. finlandicus and K. aberrans, (2) the ability to complete juvenile development and to sustain reproduction with phytoseiid prey and (3) an advantage in foraging behaviour over K. aberrans and E. finlandicus at low spider mite levels. Euseius finlandicus predominated in the two-species system E. finlandicus/K. aberrans, but both species persisted to the end of the experiment.

Notizen: Abstract A compartmental model is described for the spread of Gambian sleeping sickness in a spatially heterogeneous environment in which vector and human populations migrate between two "patches": the village and the plantations. The number of equilibrium points depends on two "summary parameters": gr the proportion removed among human infectives, and R0, the basic reproduction number. The origin is stable for R0 〈1 and unstable for R0 〉1. Control strategies are assessed by studying the mix of vector control between the two patches that bring R0 below 1. The results demonstrate the importance of vector control in the plantations. For example if 20 percent of flies are in the village and the blood meal rate in the village is 10 percent, then a 20 percent added vector mortality in the village must be combined with a 9 percent added mortality in the plantations in order to bring R0 below 1. The results are quite insentive to the blood meal rate in the village. Optimal strategies (that minimize the total number of flies trapped in both patches) are briefly discussed.

Notizen: Abstract A sol-gel precursor was prepared by the reaction of poly[bis(2-(2-hydroxyethoxy)ethoxy)phosphazene] (1) with 3-isocyanatopropyltriethoxysilane. It was then sol-gel polymerized to produce a covalently interconnected phosphazene-silicate network with urethane functionalities. Isocyanato groups were introduced on the surface of the network through coupling by allophanate formation between hexamethylene diisocyanate and urethane functionalities on the gel surface. Heparin was immobilized on the surface of the network by reacting hydroxyl or amino groups of heparin with the surface isocyanato groups. The activity of the immobilized heparin was 4.0% that of free heparin.

We find conditions on the speed of the waves which imply stability and instability of the solitary waves. The analysis depends on the variational characterization of the ground states rather than information about the linearized operator.

Notizen: Abstract This paper deals with the study of the stability of nonautonomous retarded functional differential equations using the theory of dichotomic maps. After some preliminaries, we prove the theorems on simple and asymptotic stability. Some examples are given to illustrate the application of the method. Main results about asymptotic stability of the equation $$x'(t) = - b(t)x(t - r)$$ and of itsnonlinear generalization $$x'(t) = b(t)f(x(t - r))$$ are established.

Notizen: Abstract We consider a multi‐station queue with a multi‐class input process when any station is available for the service of only some (not all) customer classes. Upon arrival, any customer may choose one of its accessible stations according to some state‐dependent policy. We obtain simple stability criteria for this model in two particular cases when service rates are either station‐ or class‐independent. Then, we study a two‐station queue under general assumptions on service rates. Our proofs are based on the fluid approximation approach.

Notizen: Abstract A closed network is said to be “guaranteed efficient” if the throughput converges under all non-idling policies to the capacity of the bottlenecks in the network, as the number of trapped customers increases to infinity. We obtain a necessary condition for guaranteed efficiency of closed re-entrant lines. For balanced two-station systems, this necessary condition is almost sufficient, differing from it only by the strictness of an inequality. This near characterization is obtained by studying a special type of virtual station called “alternating visit virtual station”. These special virtual stations allow us to relate the necessary condition to certain indices arising in heavy traffic studies using a Brownian network approximation, as well as to certain policies proposed as being extremal with respect to the asymptotic loss in the throughput. Using the near characterization of guaranteed efficiency we also answer the often pondered question of whether an open network or its closed counterpart has greater throughput - the answer is that neither can assure a greater guaranteed throughput.

Notizen: Abstract Motivated by ABR class of service in ATM networks, we study a continuous time queueing system with a feedback control of the arrival rate of some of the sources. The feedback about the queue length or the total workload is provided at regular intervals (variations on it, especially the traffic management specification TM 4.0, are also considered). The propagation delays can be nonnegligible. For a general class of feedback algorithms, we obtain the stability of the system in the presence of one or more bottleneck nodes in the virtual circuit. Our system is general enough that it can be useful to study feedback control in other network protocols. We also obtain rates of convergence to the stationary distributions and finiteness of moments. For the single botterneck case, we provide algorithms to compute the stationary distributions and the moments of the sojourn times in different sets of states. We also show analytically (by showing continuity of stationary distributions and moments) that for small propagation delays, we can provide feedback algorithms which have higher mean throughput, lower probability of overflow and lower delay jitter than any open loop policy. Finally these results are supplemented by some computational results.

Notizen: Abstract The apparent equilibrium constants and enthalpies of complexation of Nd3+, Sm3+, Eu3+, and Gd3+ by xylitol in aqueous solutions containing NaNO3 at an ionic strength of 2.0 mol-kg−1 have been determined by microcalorimetry at 25°C. Since nitrate anion weakly complexes the lanthanide cations, these values are analyzed in terms of competition between xylitol and NO 3 - The method leads to the apparent equilibrium constants and enthalpies of complexation of the lanthanide cations by NO 3 - at this particular ionic strength. Despite the difficulties encountered in characterizing rather weak associations, the results are, whenever comparison is possible, in good agreement with those obtained by direct microcalorimetry. The advantage of this competition method is that it can be used when the enthalpic effects are too weak and insufficiently concentration dependent for direct microcalorimetric determination. In the present case, it allows us to thermodynamically characterize the formation of SmNO 3 2+ and EuNO 3 2+ , processes we have not been able to study directly.

Notizen: Abstract Graphite electrodes comprising silica binder were tested in ethylene carbonate–dimethyl carbonate (EC–DMC), propylene carbonate and tetrahydrofuran solutions. The electrochemical behavior of these electrodes was analyzed using chronopotentiometry, slow-scan rate cyclic voltammetry (SSCV), impedance spectroscopy and potentiostatic intermittent titration technique (PITT). The electrode morphology and integrity were studied by scanning electron microscopy (SEM). Using silica binder, graphite particles are usually embedded in the current collector in an unoriented form. Thus, the electroanalytical study of these electrodes and the comparison of their response with that of highly oriented PVDF based graphite electrodes, provided insight into the effect of particle orientation on the general electrochemical behavior of lithiated graphite anodes. In general, the higher the particle orientation and compact packing in the electrodes' active mass, the better the performance o f the Li–graphite electrodes, as the surface films developed are better passivating and the interparticle electronic contact is also better. The silica binder may have advantages over other binders such as PVDF in its ability to better retain the electrode integrity upon cycling. However, the practical use of such electrodes requires further optimization, especially in connection with particle orientation and compact packing.

Notizen: Abstract We develop a stability and convergence analysis of Galerkin–Petrov schemes based on a general setting of multiresolution generated by several refinable functions for the numerical solution of pseudodifferential equations on smooth closed curves. Particular realizations of such a multiresolution analysis are trial spaces generated by biorthogonal wavelets or by splines with multiple knots. The main result presents necessary and sufficient conditions for the stability of the numerical method in terms of the principal symbol of the pseudodifferential operator and the Fourier transforms of the generating multiscaling functions as well as of the test functionals. Moreover, optimal convergence rates for the approximate solutions in a range of Sobolev spaces are established.

Notizen: Abstract LetK be ad-dimensional star body (with respect to the origino). It is known that the (d−1)-dimensional volume of the intersections ofK with the hyperplanes througho does not uniquely determineK. Uniqueness can only be achieved under additional assumptions, such as central symmetry. Here it is pointed out that if one uses, instead of intersections by hyperplanes, intersections by half-planes that containo on the boundary, then, without any additional assumptions, the volume of these intersections determinesK uniquely. This assertion, and more general results of this kind, together with stability estimates, are obtained from uniqueness results and estimates concerning a particular spherical integral transformation.

Notizen: Abstract The results from ethene hydroformylation at 173°C showed that a Co(acac)3/SiO2 catalyst prepared from Co(acac)3 precursor by gas‐phase deposition was three times as active as a catalyst prepared by impregnation from cobalt nitrate, but oxo‐selectivities were similar. The high propanal selectivities on the Co(acac)3/SiO2 seem to be related to the presence of highly dispersed active sites favouring CO insertion. As dispersion is decreased from 23 to 8% due to increasing metal content (from 5 to 16 wt%), oxo‐selectivity decreased from 39 to 25%. The activity of Co(acac)3/SiO2 remained unchanged during 68 h on stream. The gas‐phase deposition technique described here is a promising method for the preparation of active, selective and stable heterogeneous hydroformylation catalysts.

Notizen: Abstract Metal clusters arranged on nanostructured oxidized silicon wafers are presented as new model catalyst systems. A photoresist layer spun on top of a wafer was patterned by laser interference exposure. The grid obtained after removing the exposed parts of the resist is used as an etching mask. Hollows with diameters of 300 nm and depths between 50 and 60 nm were etched into the oxide layer using wet chemical methods. Two methods were applied to deposit metal clusters (Pd or Cu) in a defined way within the hollows. The particles ranged from 10 to 50 nm in height and from 80 to 200 nm in diameter. The model catalyst systems were characterized by atomic force microscopy and X-ray photoelectron spectroscopy. The method presented here allows us to produce 4 inch wafers that are covered completely by nanometer-sized structures in a reasonable period of time.

Notizen: Abstract  Twenty Pisolithus tinctorius isolates from different geographic locations and different hosts were characterized by the random amplified polymorphic DNA technique. Thirteen arbitrary primers generated 87 DNA fragments, all of them polymorphic. These data were used to calculate genetic distances among the isolates. The pairwise genetic distances ranged from 1 to 100%, with an average of 58.7%. Cluster analysis based on the amplified fragments grouped the isolates according to their host and geographical origins. Group I contained isolates collected in Brazil and group II those collected in the Northern Hemisphere. In addition to the diversity seen at the molecular level, the isolates also showed host specificity. Greenhouse experiments demonstrated that isolates from the Northern Hemisphere colonized mainly Pinus whereas isolates from Brazil colonized only Eucalyptus. The molecular data suggest that the Pisolithus tinctorius isolates analyzed belong to two distinct groups. The data also suggest new guidelines for future investigations on the taxonomy and systematic of this important fungus species. Furthermore, these results support future experiments aimed at the selection and development of improved isolates of P. tinctorius.

Notizen: Abstract Inorganic-organic hybrid materials prepared by the sol-gel process have become an attractive field of study due to the immense versatility associated with this method of composite material preparation. The blending of inorganic precursors with organosilicon reagents enables unique materials to be fabricated with the desired chemical and physical characteristics. The ability to control the interfacial polarity, the degree of porosity, and the chemical functionality in the matrix has been shown to be a powerful tool in the design of materials for sensor, optical, chromatographic, and catalytic applications. In this review, the preparation of the organically modified silicates (ORMOSILs) where the individual precursors are covalently bound to each other is discussed and selected examples of their potential usefulness in analytical applications is presented.

Notizen: Abstract Three amplification protocols were analyzed for error rate and generation of polymorphisms during RAPD analysis. Using a set of 240 primers, the protocols detected similar frequencies of polymorphisms in two inbred sugar beet lines. The error rate was investigated by including a 1:1 mixture of DNA from the two lines in all analyses. Similar error rates, approximately 18%, were detected by the three protocols. Thus, altered amplification conditions did not substantially affect the error rate during RAPD analysis. For each of the three possible pairs of protocols, a positive correlation was obtained for primer and number of polymorphisms. Thus, a set of highly polymorphic RAPD primers can be used effectively, without prior screening, to detect polymorphisms for each protocol.

Notizen: Abstract Accurate identification of parental plants and their hybrids is essential for an effective breeding programme. Traditional classification of cocoa varieties relies on the characterisation of agricultural traits at plant maturity. A rapid and reliable method is described, based on genotypic analysis. An efficient DNA isolation procedure was developed, yielding unsheared DNA of high purity. Two genetic fingerprinting techniques, RAPD and AFLP™, were evaluated for their suitability in distinguishing cocoa varieties. RAPD analysis was unsatisfactory due to the low frequency of polymorphisms and poor reproducibility. AFLP™ was reliable in distinguishing phenotypically identical, known varieties of cocoa. Importantly, AFLP™ also revealed intra- and inter-varietal variation. Notizen: Abstract A simple, efficient and reliable method is described for isolation of total DNA from young leaves of Amaranthus species. This procedure yields a high amount (600–800 µg DNA/g fresh leaf tissue) of good quality DNA free from contaminating proteins, polysaccharides, and coloured pigments. The DNA is suitable for digestion with several restriction endonucleases, preparation of Southern blots, and PCR amplification. The DNA has been successfully used for generating DNA fingerprint profiles and RAPD banding patterns in two species of Amaranthus. The procedure is suitable for processing of a large number of samples simultaneously.

Notizen: Abstract In this paper, we consider the control design problem of vehicle following systems with actuator delays. An upper bound for the time delays is first constructed to guarantee the vehicle stability. Second, sufficient conditions are presented to avoid slinky-effects in the vehicle following. Next, zero steady state achieved by the proposed controller is proven. Finally, simulations are given to examine our claims.

Notizen: Abstract Sixty-three Paracoccidioides brasiliensis isolates obtained from three nine-banded armadillos ( Dasypus novemcinctus), one Amazonian armadillo's and 19 clinical isolates were compared by random amplified polymorphic DNA analysis with the primer OPG-19. The isolates were divided into three major clusters, I, II and III. Coincidences between human and armadillo isolates were observed in clusters I and II. Cluster III consisted only of armadillos' isolates. The results suggested that (I) humans may acquire P. brasiliensis infection by contact with armadillo's environment, (II) there may be P. brasiliensis genotypes peculiar to the animal, and (III) individual armadillos may be infected with P. brasiliensis cells with different genotypes.

Notizen: Abstract Fast and reliable identification of different species of the genus Candida is important to define adequate therapeutic decisions, because the different species have highly variable susceptibilities to antifungal drugs; azoles and amphothericin B. Accurate statistical records on case history and epidemiological studies also depend on effective identification. To address this problem we established a RAPD method that enabled direct identification of five very common species of Candida. Initially, reference band patterns were established for C. albicans, C. tropicalis, C. parapsilosis, C. glabrata and C. krusei. One of the primers, M2, showed remarkably conserved intra-specific patterns of approximately 10 bands each, ranging in size from 2.0 to 0.1 kb. These patterns were significantly different and species-specific. Few bands were conserved between different species of Candida, which was assumed to be consistent with their phylogenetic relatedness. In addition, band patterns were constant and reproducible and DNA isolated from single colonies yielded sufficient DNA for identification. The reference band patterns were then used, in blind experiments, to identify species of Candida in 50 randomly chosen samples, including clinical isolates and ATCC strains. RAPD results were 100% consistent with results obtained by conventional diagnostic methods and were achieved in one day instead of several days taken by conventional methods. Because ideal identification methods should be consistent with phylogeny and taxonomy we tested whether RAPD could be used to calculate genetic distances. Comparison of RAPD phylogenetic trees with 18S rRNA trees showed significant differences in tree topologies which indicated that RAPD data could not accurately measure the relative distances between different species. Also, computer simulations of RAPD random patterns were used to test whether the observed degree of RAPD band pattern similarities could occur at random. These simulations suggested that the level of inter-specific band pattern similarities observed in our data could be obtained at random, while intra-specific pattern similarities could not. RAPD would be helpful to discriminate between isolates but not to quantitate the differences. We suggest that the inaccurate estimate of genetic distances from RAPD is a general limitation of the technique and not a specific problem of our identification method. Because of the repetitive character of the target sequences, genetic distances calculated from RAPD could be affected by paralogy, namely, recombination and duplication events not parallel with speciation events.

Notizen: Abstract The authors discuss the long-run future of decision support systems in marketing. They argue that a growing proportion of marketing decisions can not only be supported but may also be automated. From a standpoint of both efficiency (e.g., management productivity) and effectiveness (e.g., resource allocation decisions), such automation is highly desirable. The authors describe how model-based automated decision-making is likely to penetrate various marketing decision-making environments and how such models can incorporate competitive dynamics. For example, the authors foresee that close to full automation can ultimately take place for many decisions about existing products in stable markets. Partial automation could characterize decision making for new products in stable markets and existing products in unstable markets.

Notizen: Abstract Agroforestry experiments usually include control plots of either pure crop or pure tree stands. A clear distinction should be made between intensively managed biophysical controls and farming system controls with realistic labour input and management regimes. Trying to draw biophysical conclusions from farming system controls (or the reverse) is often not justifiable. The design and management of these elusive control plots is a complicated issue which is often overlooked. Many factors beyond the control of the experiment manager can disturb long term field agroforestry experiments. Some examples from French agroforestry experiments illustrate how uncontrolled factors may bias the results, including the proportion of harvested to planted trees, the weeding regimes, and the use of tree- shelters. The analysis of agroforestry data could be more efficient when considering a continuum of tree – crop mixture management options between the agroforestry plot and the non agroforestry plot. The concept of biophysical control plots becomes then less essential. A relevant modelling approach of interactions between trees and crops should 1) perform correctly for any tree/crop proportion and even for pure stands, when setting the parameters of the other component to zero, 2) provide for the inclusion of new, uncontrolled factors that could emerge through time. The biological efficiency of agroforestry systems may however be a subordinate criterion for agroforestry adoption, as observed at the moment in France. Agroforestry systems with poor biological outcomes can even be very attractive in some ecological or sociological conditions, and only farming system controls may bring this aspect to light.

Notizen: Abstract At sites in the Sahel where windbreaks are used to control wind erosion, management strategies are required to minimise competition for water between trees and crops. Uptake of water by windbreak trees was therefore studied in experiments designed to compare water use among tree species, assess which variables exert most control over transpiration and determine the source of water transpired by windbreak trees. Transpiration and soil-water extraction by Acacia nilotica, Acacia holosericea and Azadirachta indica trees in windbreaks were measured at the ICRISAT Sahelian Centre, Niger. Coupling of windbreaks to the atmosphere was evaluated and a stable isotope technique was used to compare utilisation of groundwater by windbreaks and crops at two sites in Niger with different water table levels. Azadirachta indica used least water, probably as a result of lower stomatal conductances, since windbreaks exhibited good physiological control over transpiration. The potential for competition for water was most severe with Acacia nilotica and Acacia holosericea, as they extracted large quantities of water through lateral roots, and at the location where trees could not access groundwater. At such sites, the effects of competition on crop productivity should be minimised by planting tree species with low water requirements and by using pruning to limit tree transpiration.

Notizen: Abstract In Italy, new agroforestry cultural models could play an important role in the diffusion of plantation forestry timber species. We studied the stem growth rates and leaf water potentials (Ψ) of common walnut (Juglans regia L.), and gravimetric soil moisture (θDW) depletion during the third and fourth growing seasons in an agroforestry trial in central Italy. Since the establishment of experimental plots in 1992, walnut was intercropped with alfalfa (Medicago sativa L.), with or without polyethylene (PE) mulching along tree rows. By the end of the study period, the unmulched-intercropped walnut was almost 68% smaller in stem diameter and height than the sole-unmulched control, demonstrating walnut’s sensitivity to alfalfa competition. The competitive effect of alfalfa on walnut stem growth was effectively controlled/reduced by PE mulching. Stem growth rates of mulched-intercropped walnut were always lower than the control, but much higher (ca. 235% in stem diameter and height) than those of unmulched-intercropped walnut. The higher field performance of mulched-intercropped walnut compared to the un- mulched-intercropped treatment was associated with higher soil moisture and predawn and midday Ψ values during the summer’s driest periods. The presence of PE mulching was also associated with lower levels of soil moisture during the early and late growing season, but this did not negatively affect walnut Ψ and growth rates. These results suggest that in the climatic conditions of central Italy, competition for soil moisture between young common walnut trees and alfalfa can be substantially lowered, but not completely eliminated, by PE mulching.

Notizen: Abstract This paper continues the author's study of procedures for rewriting the well-known Chapman–Enskog expansion used in the kinetic theory of gases. The usual Chapman–Enskog expansion, when used in isothermal fluid motion, will introduce nonlinear instability at super-Burnett order O(ε3) truncation. The procedure given here eliminates the truncation instability and produces the desired dissipation inequality.

Notizen: Abstract An expanded granular sludge bed reactor, inoculated with acclimated sulfidogenic granular sludge, was operated at 33 °C and fed with acetic acid as COD source and sulfate as electron acceptor. The bioreactor had a sulfate conversion efficiency of 80–90% at a high sulfate loading rate of 10.4 g SO4 2--S/l.d after only 60 days of start-up. This was achieved by implementing a dual operational strategy. Firstly acetic acid was dosed near stoichiometry (COD over sulfur ratio = 2.0 to 2.2) which allowed almost complete sulfate removal. Secondly the pH in the bioreactor was kept slightly alkaline (7.9 ± 0.1) which limited the concentration of the inhibitory undissociated hydrogen sulfide H2S (pKa = 7). This allowed the acetotrophic sulfate reducing bacteria to predominate throughout the long term experiment. The limitations of the EGSB technology with respect to the sulfate conversion rate appeared to be related to the biomass wash-out and granule deterioration occurring at superficial upflow velocities above 10 m/h. Increasing the recirculation flow caused a drop in the sulfate reduction rate and efficiency, an increase of the suspended sludge fraction and a considerable loss of biomass into the effluent, yielding bare mainly inorganic granules. Elemental analysis revealed that a considerable amount of the granular sludge dry matter at the end of the experiment, at an upflow velocity of 20 m/h, consisted of calcium (32%), mainly in the form of carbonate deposits, while organic matter only represented 7%.

Notizen: Abstract Protoplasts isolated from `Page' tangelo (Minneola tangelo × clementine) cell suspension cultures were electrically fused with mesophyll protoplasts of orange jessamine [Murraya paniculata (L.) Jack]. Shoots were regenerated after 6 – 10 months of culture, but they were extremely recalcitrant to producing roots in root-induction medium. Complete plantlets were formed via micrografting. Chromosome counting of shoot tips revealed they were tetraploids (2n = 4x = 36). Glutamateoxaloacetate transaminase isozyme and randomly amplified polymorphic DNA analysis confirmed their hybridity. Orange jessamine is immune to citrus huanglongbin, a severe disease of citrus, but sexual incompatibility and limited graft compatibility exist between Citrus and orange jessamine. The cell fusion technique may make it possible to transfer the huanglongbin resistance trait from orange jessamine to Citrus.

Notizen: Abstract Random amplified polymorphic DNA (RAPD) markers were used to determine the genetic stability of long-term (more than 10 years) micropropagated shoots of Japanese black pine (Pinus thunbergii Parl.). Thirty-six shoots consisting of three morphotypes (short, medium, and long needles) were randomly chosen from about 4,000 micropropagated shoots regenerated from the explants of a single nematode-resistant mother plant. Out of 126 primers screened, 30 gave 134 clear reproducible bands. A total of 4,824 bands obtained from these studies exhibited no aberration in RAPD banding patterns among the tested shoots. Our results show that regenerants from our plant micropropagation system are genetically stable.

Notizen: Abstract We developed an efficient plant regeneration system from protoplasts for poplar (Populus alba L.). Protoplasts were isolated from 4-day-old suspension cultures derived from seed-induced calli with a yield of 6.96× 106 cells/g fresh weight cells and then cultured at a concentration of 2.5×105 cells/ml in NH4NO3-free Murashige and Skoog (MS) medium supplemented with 5 µM 2,4-dichlorophenoxyacetic acid (2,4-D), 0.05 µM thidiazuron (TDZ) and 0.5 M glucose as a osmoticum. The plating efficiency of the cultured protoplasts was calculated at 26.5% at day 7 and 31.7% at day 14. Cell colonies were observed after culturing for 4 weeks. Regenerated colonies were propagated through subculture in liquid MS medium supplemented with 5 µM 2,4-D. Buds were induced from regenerated calli on MS medium containing 10 µM kinetin or 1 µM TDZ. Regenerated shoots were rooted on half-strength MS medium, and the plantlets were transplanted in soil. Randomly amplified polymorphic DNA analysis did not detect any DNA polymorphism among the regenerated plants.

Notizen: Abstract Bioactive ceramic coatings have had poor adhesion to substrate. In this study, the bond strength (tensile strength) of titania gel coating to titanium substrate was studied. In the experiments three different pretreatments were used, namely sodium hydroxide corroding, plasma cleaning and titanium nitride coating. Also the effects of heating temperature, heating in vacuum and titanium surface roughness were studied. The sol properties were altered with valeric acid addition. Samples were analysed by SEM-EDX, AES, AFM and tested by bond strength gauge. Those samples in which the titanium surface was precorroded one hour in sodium hydroxide, predeposited by titanium nitride or ground improved the bonding strengths of titania coatings to over 24 MPa. In these samples a fracture occurred at the glue-coating interface.

Notizen: Abstract In a program on the development of metal nanoclusters in sol-gel derived thin films, attempts were made to synthesize pure and mixed metal clusters, control the cluster size and increase the volume fract f the clusters. Thus, Ag, Cu and Ag-Cu nanoclusters were prepared in silica films using dip- and spin-coating techniques. The annealing of Ag/SiO2 films in different atmospheres (air, argon and 5% H2-95% N2 gas) caused modifications of Ag nanoclusters resulting in changes in their surface plasmon resonance (SPR) peak positions. The Cu and Ag-Cu codoped films were annealed in reducing atmosphere (5% H2-95% N2 gas). In order to prepare Cu nanoclusters of different sizes, the concentrations of Cu in Cu/SiO2 composite films were varied from 8 to 30 mol% and annealed at 800°C for different times for growth. The size of the Cu nanoclusters was measured from the half band width of Cu SPR peak (appearing within 570–557 nm range) and X-ray diffraction. In this way Cu-nanoclusters of size ranges from about 3.5 to 10 nm (average diameters) were prepared . The Ag-Cu nanocluster-containing silica films show the existence of both Ag and Cu SPR peaks with some blue shifting in comparison with to their pure analogues depending on the Ag:Cu ratio.

Notizen: Abstract In this work we compared biosorbents obtained by encapsulation of polysaccharides, isolated from waste brewing biomass, in sol-gel derived silicates and an organic polymer. Biosorbents were prepared by mixing cross-linking-agents—organic or siliceous—with dried cells envelopes. Siliceous prepolymers were synthezised via transesterification and hydrolysis from tetraethoxysilane (TEOS) and methanol. Sorption of Cd2+, Cu2+ and Ag+ by biosorbent granules (0.25–0.6 mm) was examined in batch and in a packed column. The biosorbent prepared by interesterification of TEOS showed a 2–3 times higher intensity of sorption than the biosorbent cross-linked with epichlorohydrin (the most effective cross-linking organic agent) while the sorption capacity of both biosorbents was equal. The specific surface area of the silica matrix was 597 m2/g but only traces of metals were sequestered from solution with a concentration of Cd2+ of 50 mg/l. The biosorbent with a silica matrix is a heterogeneous material containing microporous matrix inclusions of thin cell walls. Its high sorption intensity and good mechanical strength will be useful in continuous metal uptake of low concentrations of metals.

Acids were in some cases dissolved in the aqueous phase. Whereas the size of the sol nanoparticles is independent of the micellar composition, the kinetics of the sol-gel transition are not. The gelation time is shorter for the non-ionic surfactant and becomes longer as the acid content in the water increases, and for smaller anions of equal charge.

Thermomechanical analysis showed an increase of the glass transition temperature, due to the stiffness of the polymeric network, as the amount of Eu3+ increased. Europium luminescent properties were used to study structural evolution during the sol-gel transition. For lower doping concentrations dried gels present statistical distributions of Eu3+, typical of an amorphous environment, while for higher concentrations a crystalline-like environment of Eu3+ was observed. A broad emission band was observed in the visible part of the electromagnetic spectrum and assigned to the intrinsic emission from the hybrid polymeric network.

Notizen: Abstract Sol-gel material based mirrors have been produced by forming alternate layers of high refractive index and low refractive index thin films. These mirrors have proven to have a high laser induced damage threshold [LIDT]. Using nitric acid stabilized zirconia derived from zirconium-n-propoxide and base catalyzed silica, a 16 layer mirror with a reflectivity of better than 94% at 351 nm and 45° angle of incidence was fabricated. This had an LIDT of 7.7 J/cm2 at 351 nm with a 0.7 ns pulse width. Crazing prevented further layers being deposited. Both spin and dip coating were attempted with dip coating yielding the best results. The coating structure has been analyzed using XPS depth profiling and AES. The bulk materials have been investigated using transmission electron microscopy (TEM), X-ray diffraction (XRD) and TGA. High refractive index layers using Hafnia with nitric or acetic acid have also been investigated as prospective high LIDT mirrors. Alternative acidic routes to silica have been studied as a possible low index material and a route to preventing crazing.

Notizen: Abstract The formation of a PZT film of 10–20 μm in thickness via an interfacial polymerization of metal-organic precursors has been studied. A commercially available PZT precursor solution developed for dip- or spin-coating was diluted with hexane containing acetylacetone as chelating agent. The solution was poured on water contained in a teflon reaction container of 88 mm inside diameter. A translucent gel film was formed at the interface of the two immiscible liquids. While floating on water it shrank as the evaporation of solvent proceeded until its diameter decreased to about 45% of its initial value. The dried gel film was almost pore free and its thickness was estimated to be about 10 μm.

Notizen: Abstract Thin silica and silica-titania films doped with sulfide nanocrystals of controlled size were fabricated by a method based on the preparation of colloidal particles and their introduction into a glassy matrix through the sol-gel method. Colloidal sols of composition HgxCd1-xS and PbxCd1-xS (with x ranging from 0 to 1) were prepared and used to dope alkoxide solutions for the deposition of thin silica and silica-titania films. Optical absorption spectra were taken on both precursor colloidal sols and derived doped films. X-ray diffraction characterization gave structural information on the nature and size of particles in powders obtained by precipitation from colloidal sols and in doped films. The advantages and limits of the investigated systems are discussed in light of possible applications. The nonlinear properties of the most interesting PbS-doped planar waveguides have been investigated in the near-infrared, at 1.064 μm. A reversible nonlinear effect was measured, with n 2 values ranging from -5 to -20×10-9 cm2/kW.

Notizen: Abstract The sol-gel process has been applied to the preparation of nano-size Sb2S3 crystallite doped silica glasses and thin films. Silica glasses containing 1–1.5 wt% Sb2S3 are prepared by hydrolysis of complex solution of Si(OC2H5)4, SbCl3 and SC(NH2)2, and subsequent heat treatment. The nano-size crystallite in the matrix is observed by means of TEM. The transmission spectra of the films show that the transmission valley shifts toward longer wavelengths with increasing heat treatment time and temperature. Second harmonic generation (SHG) has been observed in the glasses illuminated with intense 1.06 μm and frequency-doubled laser beams from mode-locked Nd:YAG laser.

Notizen: Abstract Organic-inorganic nanocomposite protective coatings are prepared on aluminum substrates by the spinning technique with the concept of incorporating homogeneously nanosized particles (of AlOOH, Al2O3, ZrO2, SiC) into molecular organic-inorganic hybrid matrices. The hybrid matrices are prepared from epoxysilane and bisphenol A with imidazol as catalyst. The AlOOH particles are derived from aluminum isoprooxide and introduced into the hybrid sols directly, and Al2O3, ZrO2, SiC particles are first surface-modified with Si–OH from hydrolyzed TEOS. The coatings are dense, smooth and flexible and inhibit corrosion.

Notizen: Abstract Precursor powders for Pb(Zr1 − xTix) O3 (PZT) thin films were produced by the reaction of zirconium-and titanium-n-propoxides with acetylacetone and lead acetate trihydrate. The subsequent complete removal of volatile components yielded powders that can be handled in air. The powders are indefinitely stable under ambient conditions. High molarity (〉2m) coating sols were prepared by dissolution of the precursor powders in mixtures of 1,3-propanediol, triethanolamine (TEA) and water. Excess amounts of lead to compensate lead loss during firing were easily introduced into these solutions. The deposition of these sols on steel substrates and firing at 600°C yielded PZT films. Many physical parameters like film thickness, morphology and electrical performance could be influenced by choice of the solvent mixture composition and oxide content of the sols. Depending on the preparation dielectric permittivities, εr of up to 840 were measured at 1 kHz. By hysteresis measurements at 50 Hz and a field amplitude of 50 V/μm a remanent polarization of about 40 μC/cm2 and coercivity of about 8 V/μm was obtained. The films were stable against dielectric breakdown up to 70V/μm.

Notizen: Abstract Two polyviologens synthesized by the Menshutkin reaction were incorporated into silica networks using the sol-gel method. The effects of the incorporated polymers on the structure of the resulting organic-inorganic hybrid materials were investigated using X-ray scattering (small and wide angle). This provided evidence that the polyviologens in a lyotropic liquid crystalline state were causing the silica network to become more dense and less branched than sol-gel glasses synthesized without added polyviologen. Additional evidence for the templating effect of these polymers was obtained using FTIR spectroscopy and N2 adsorption-desorption isotherms.

Notizen: Abstract Inorganic oxides have been synthesized successfully under mild reaction conditions using a solvent-free non-hydrolytic sol-gel process based on the condensation reaction of “metal” chlorides with oxygen donors such as alkoxides, aldehydes and ethers. Iron(III) chloride was found to be an effective catalyst for the reactions. The order of reactivity of the halides was generally titanium 〉 aluminium 〉 silicon, but in some cases reaction was halted by premature gelation of intermediate species. Silica, alumina and titania were all prepared and characterized by various methods. Calcination studies on the silicas showed these materials to be amorphous up to at least 700°C, but devitrification occurred at 1000°C. Crystallization was studied by X-ray powder diffraction.

Notizen: Abstract Transparent composite SiO2/organic matrices have been made by the sol-gel method using tetramethoxysilane and either cetyltrimethylammoniumbromide (CTAB) or a polyethylene glycol oligomer (PEG-200) or a cationic polyelectrolyte. We have investigated conditions under which transparent xerogel matrices are possible and we have characterized them mainly by time-resolved fluorescence probing using stretched exponentials. CTAB creates a hydrophobic and PEG-200 a hydrophilic subphase in the matrix. An important property of these composite matrices is that they allow molecular diffusion contrary to matrices made in the absence of surfactants or polymers.

Notizen: Abstract Polysiloxane porous layers doped with TiO- and phenyl-groups were fabricated by the sol-gel method. Starting sols were prepared from alkoxides using catalysis by HCl or HF. Stable SiO-TiO-sols were obtained by using HF. Porous gel layers with thicknesses of 0.3–1.1 μm were coated on fiber surfaces or silicon wafers. Layers with the refractive index of 1.38–1.48 were prepared. The fractional porosity of the layers was estimated to be 0.1–0.35. The interactions of the layers with vapor or liquid chlorinated hydrocarbons, alcohols or aromatic hydrocarbons have been studied in immersing experiments by measuring the output light intensity from the fibers. The observed changes of the output intensity could be correlated to changes of the refractive index of the layer caused by the penetration of the tested chemicals into the layer pores.

Notizen: Abstract GeO2-SiO2 sol-gel planar waveguides doped with Er were deposited by spinning on silica substrates. P2 O5 or Al2O3 were used as co-dopants to improve erbium dissolution in the GeO2-SiO2 matrix. Multilayer amorphous films were obtained at 600 or 700°C. Er ions in the planar waveguide pumped at 980 nm showed fluorescence features around 1530 nm. Narrow fluorescence spectra (∼20 nm) and long lifetimes (∼6 ms) were found in P2O5 co-doped samples, whereas Al2O3 co-doping gave wider spectra (∼50 nm) with slightly lower lifetimes (∼5 ms). The quenching concentration in the Al2O3 co-doped samples was 0.9 mol% Er. Heat treatments in CCl4 improve the active properties and the addition of Yb enhances the pump absorption efficiency.

Notizen: Abstract Er3+ doped-aluminosilicate thin films were prepared on silica and silica/Si substrates by the sol-gel process and dip-coating. The sol-gel aluminosilicate planar waveguides were prepared from silicon and aluminium alkoxides. Their structural characterization has been carried out by Raman spectroscopy, Atomic Force and Scanning Electron Microscopies. The results indicated that these films present an amorphous structure until an annealing temperature of 900°C, while at temperatures higher than 1000°C, crystallization occurs. An estimate of microcrystallite sizes using Raman spectroscopy is given, which agrees with data from scanning electron microscopy. The optical properties have been investigated by Fluorescence spectroscopy in the visible region.

Notizen: Abstract Stable tungsten oxide coating sols suitable for electrochromic applications were prepared by a modified peroxotungstic acid route. Layers up to 250 nm thickness could be deposited on ITO-coated and/or FTO-coated glass substrates in a single dip-coating process. Optoelectrochemical measurements were employed to determine the variation of the electrochromic properties (change in optical density (ΔOD) and switching times) of WO3-layers, investigated as a function of coating parameters, such as chamber humidity and the temperature of heat treatment. High resolution transmission electron microscopy (HR-TEM) has shown that the optimized layers possess a partially crystalline morphology with nanocrystalline regions 2–3 nm in size.

Notizen: Abstract A fiber forming PZT gel containing 58.5 wt% PZT was synthesized by using zirconium-n-propylate, titanium-iso-propylate, lead acetate and butoxyethanol. Unseeded PZT gels and gels containing 0.5 wt% PZT perovskite seeds (Ø = 200–300 nm) could be extruded through a monofilament nozzle (Ø = 100 μm) at pressures between 50 and 150 bar, whereas PZT gels, containing 1 and 2 wt% PZT particles, were pressed through the nozzle at higher pressures (200–300 bar). The microstructure of unseeded and seeded (0.5, 1, 2 wt% PZT) PZT fibers was characterized by SEM. Unseeded fibers had three different shells at 450°C: an external dense shell (approx. 200 nm thick), a middle shell consisting of a porous structure (1.5μm thick) and the center of the fiber, characterized by a matrix containing globular particles. At 700°C, a 200–250 nm thick and dense external shell and a porous fiber interior were be observed. 2 wt% of PZT seeds was necessary to densify the fiber completely. The seeds were located in the center of each PZT perovskite rosette.

Notizen: Abstract The thermal evolution of an alumina gel synthesized by hydrolysis of aluminium alkoxide (sol-gel technique) was studied by thermal analysis (DTA and TGA), X-ray diffraction, FTIR and NMR spectroscopies, and specific surface area measurements. Between 400 and 900°C, γ- and δ-aluminas were formed showing aluminium vacancies preferentially located in tetrahedral sites. The atomic rearrangements produced during α-alumina formation are oriented to the progressive elimination of tetrahedral aluminium in the ultimate phase. The evolution of the specific surface area during heating is explained by changes in structure and microstructure.

Notizen: Abstract The ecological concept of resilience has begun to inform analysis of change in economy-environment systems. The linkages between resilience and the stability of dynamical systems are discussed, along with its role in understanding of the evolution of such systems. Particular linkages discussed include those between resilience, biodiversity and the sustainability of alternative states. Recent developments in modelling the resilience of joint economy-environment systems suggest the advantages of analysing change in the system as a Markov process, the transition probabilities between states offering a natural measure of the resilience of the system in such states. It is argued that this ‘emergent property’ of the collaboration between ecology and economics has far-reaching implications for the way we think about, model and manage the environmental sustainability of economic development.

Notizen: Abstract Amorphous alumina-silicas were prepared from a tetra-alkoxysilane and anhydrous aluminum trichloride or an aluminum alkoxide by a sol-gel process using 2-methyl-2,4-pentanediol, pinacol, 1,2-propanediol, 2,3-butanediol or ethylene glycol as the solvent or complexing agent, and the effect of diols and alkoxy groups on the physical and chemical properties of the alumina-silicas was examined. When the diol or the alkoxy group was bulky, the alumina-silicas had relatively larger micropores, a larger pore volume and higher surface areas. In the conversion of methanol catalyzed by the alumina-silicas, the bulkier diols and alkoxides gave catalysts that produced dimethyl ether in higher yield and hydrocarbons in lower yield. Thus, when ethylene glycol was used as the diol, the best catalyst for the production of hydrocarbons, especially the production of olefins such as ethylene, propylene and butene, was obtained. Furthermore, in comparison with alumina-silica prepared by a traditional kneading process, it was found that the sol-gel alumina-silica could efficiently convert methanol to dimethyl ether and hydrocarbons, but the material prepared by kneading had a very low conversion of methanol to other compounds.

Notizen: Abstract Sb doped sol-gel SnO2 single layers (thickness ≈100 nm) were prepared from alcoholic solution and deposited via a dip coating process on fused silica substrates. The coatings have been sintered at a typical rate of 10–15 cm2/s by CO2 laser irradiation. The laser spot was scanned in one direction at a speed of 15,000 cm/s and the sample was moved in a perpendicular one at a speed up to 250 mm/s. The temperature of the topmost 10 μm layer was monitored by a fast pyrometer (μs resolution). The following properties of the coatings have been determined: the electrical resistivity ρ, the carrier density n, and mobility μ, the structure, the thickness, the mesoscopic and micromorphology and the density. The sintering by CO2 laser radiation is mainly a thermal driven process. At T ≈500°C it allows to obtain coatings with a smaller resistivity (6.8×10−3 Ωcm) than those produced by conventional furnace firing (ρel≈2.9×10−2 Ωcm). The results are discussed in terms of particle size and packing density.

Notizen: Abstract Vanadium dioxide thin films have been deposited from vanadium alkoxides VO(OR)3. An amorphous film is formed that transforms into crystalline VO2 upon heating at 500°C under a reducing atmosphere. Optically transparent VO2 thin films are then obtained that exhibit both electrical and optical switching around 70°C. The switching temperature together with the shape of the hysteresis loop can be modified by doping VO2 films with foreign cations. Doped MxVO2 (M = W6+, Nb5+, Ti4+, Cr3+ or Al3+) thin films have been prepared under the same conditions by mixing the vanadium alkoxide and a metal salt in an alcoholic solution. The switching temperature decreases when the film is doped with high-valent cations (W6+) and increases with low-valent cations (Al3+, Cr3+). The transition temperature first decreases and then increases when TiIV is added to the VO2 film while the width of the hysteresis loop is significantly reduced.

Minimizing shrinkage and enabling relaxation of the drying sol-gel are key factors in the elimination of cracking. Notizen: Abstract Traditionally, magnesium oxide has been considered a typical basic catalyst, catalyst carrier and/or adsorbent. In this study MgO was prepared using Mg-ethoxide dissolved in ethanol and hydrolyzed with various aqueous inorganic acids and bases. We have found that it is possible to induce Brønsted and Lewis acid sites depending on the method of preparation and, more specifically, on the type of inorganic acids used in the hydrolysis stage. In the FTIR spectra of adsorbed pyridine on MgO obtained using aqueous HCl (pH = 3), the bands corresponding to Lewis (1603, 1496 and 1444 cm−1) and Brønsted (1550 cm−1) acid sites are observed. The fact that upon vacuum and high temperature treatment, i.e., at 773 K, the above mentioned bands are clearly distinguishable indicated that the acid sites are very strong. On the other hand, the MgO materials obtained using aqueous acetic acid as a hydrolysis catalyst (pH = 5), and that without any catalyst (pH = 7) showed only Lewis acid sites which decreased markedly upon higher temperature treatment. When aqueous NH4OH (pH = 9) was used as the hydrolysis catalyst, the MgO obtained showed only Lewis acid sites. In all cases the number of Lewis acid sites was greater than that of Brønsted sites.

Notizen: Abstract Bi2O3 thin films were prepared by dipping silica slides in ethanolic solutions of tris(2,2′-6,6′-tetramethylheptane-3, 5-dionato)bismuth(III) [Bi(dpm)3] [1] and heating in air at temperatures ≤500°C. Bi4(SiO4)3 homogeneous thin films were obtained from the reaction of the bismuth oxide coating with the silica glass substrate at temperatures higher than 700°C. For heat treatments at temperatures between 600°C and 700°C, Bi2SiO5 coatings were obtained. The composition and microstructure evolution of the films were determined by Secondary Ion-Mass Spectrometry (SIMS), X-Ray Photoelectron Spectroscopy (XPS) and Glancing Angle X-Ray Diffraction (GA-XRD). The synthesis procedure was reproducible and allowed the control of the Bi2O3 phase composition. Moreover, the thin film annealing parameters were correlated with the formation of bismuth silicates, among which Bi4(SiO4)3 (BSO) is very appealing for the production of fast light-output scintillators [2].

Notizen: Abstract ORMOSILs have been prepared in the series TMOSx·MTMS(100 − x) (where TMOS is tetramethoxysilane; MTMS is methyltrimethoxysilane; x is mol% silane with respect to total silane for 0 ≤ x ≤ 100) by means of acid catalyzed, sol-gel processing. After drying at 60°C, small bulk samples were obtained of excellent optical clarity. Powder X-ray diffraction (XRD) patterns, in the range of 5 to 60°2Θ, were compared with that of fused silica. All the prepared samples were amorphous. Fused silica exhibits one broad peak, d2 centered at d-spacing 4.12 Å. For the TMOS100 silica xerogel, the analogous broad peak had shifted slightly, to be centered at 3.88 Å; and remained in about the same position as x was decreased for the series TMOSx·MTMS(100 − x). In addition, a second, broad peak, d1, was observed for the ORMOSIL series centered at the d-spacing 8.7 Å for MTMS100 (i.e., x = 0) and increasing smoothly as x was increased, reaching 11.3 Å for x = 70, and 〉11.3 Å for x 〉 70. The intensity of d1 was found to have trebled, relative to the intensity of d2, on increasing the organic character of the matrix from TMOS70·MTMS30 to MTMS100. The d2 peak appearing at about 4 Å for both fused silica and the ORMOSILs is assumed to be associated with the spacing between silicon atoms connected by means of an oxygen bridge. The Si–O–Si angle for silica xerogels is known to depend upon the nature of the sol-gel processing and is bigger than that of fused silica. The d1 peak may be associated with the spacing between silicons attached to methyl groups and indicative of channels of methyl groups in the structure. Alternatively, the d1 peak may have its origin in a preferred, discrete structural unit in the matrix for instance cubane based on a octameric silicon arrangement.

Notizen: Abstract Atomic force microscopy (AFM) was used to study the performance of ≈80 nm thick Al2O3-TiO2-SiO2 sol-gel coatings on glass substrates following corrosion in 1 M NaOH solutions at 60°C. The as-prepared coatings were homogeneous on a nanoscale and displayed the “glass pattern” before corrosion. Layers with different compositions behaved differently during the corrosion process. Thus, TiO2 or TiO2-dominated layers had tetragonal-like crystals on their surfaces after corrosion, possibly of anatase composition. On the other hand, layers with a molar ratio Al2O3 : SiO2 near 1 : 2 displayed a pseudo-hexagonal morphology, possibly with a nepheline (Na2O · Al2O3 · 2SiO2) composition. Layers of 5Al2O3-40TiO2-55SiO 2 were corroded is a stepwise fashion and had no special surface morphology.

Notizen: Abstract The development and characterization of channel waveguides using wet-process, low temperature sol-gel chemistry is described. Two structures have been developed. The first one is a one-layer structure, composed of a sol-gel solution which is a mixture of photopolymerizable organosilicate and organozirconate precursors. The other is a multilayer structure with a buffer under the guide and a protective coating. The layers are deposited by the dipping technique. The devices are obtained by UV light exposure of the coating through a predefined mask (channel waveguides). The refractive index increase is sufficient enough to allow the use of waveguides in the 1.55 μm telecommunication window. These waveguides are thick enough to reduce the coupling losses with an optical fiber below 0.5 dB. Also, because of our sandwich structure, the propagation losses are less than 0.1 dB.

Notizen: Abstract Sm2+-doped glasses in the system of Al2O3-SiO2 were prepared by sol-gel processing of metal alkoxides and reacting with H2 gas at 800°C. The hole burning properties of these glasses were investigated. The holes were burned in the 7F0 → 5D0 line of the Sm2+ ions using a DCM dye laser at 77 K. The hole depth increased with increasing the laser irradiation time, reaching up to ∼15% of the total intensity within a few hundred seconds. The hole width was 3 cm−1 full width at half maximum. Fluorescence line narrowing spectra of the 5D0 → 7 F1 transition were analyzed to study the local structure surrounding the Sm2+ ion. It is concluded that the Sm2+ ions are closely coordinated with nine oxygens of the AlO6 group in aluminosilicate glasses and that the addition of Al3+ ions into glass induces an increase in the coordination number of the Sm2+.

Notizen: Abstract The preparation and the optical characteristics of highly oriented PLT thin films are investigated. PLT films fabricated on MgO(100) and c-plane sapphire substrates have highly grown in (100) and (111) orientations, respectively. PLT films with high La content have a near cubic structure and weak anisotropy of refractive indices. The optical propagation losses of PLT films decrease as the La content of the films increases due to a complex interaction of surface roughness reduction and a reduction in the anisotropy refractive index. However, optical scattering in thicker sol-gel PLT thin film waveguides occurs by the internal scattering mechanism from the defects and the interfaces rather than by the surface scattering mechanism.

Glasses heated to 150 to 400°C exhibited room temperature conductivities of ∼10−4 S/cm, larger by ∼2 order of magnitude than that of as-prepared gel, ∼3 × 10−7 S/cm at 30°C. The conductivity increased quadratically with the increasing product of proton and molecular water concentration. These high conductivities were regarded as fast proton transfer accelerated by molecular water bonded with the hydroxyl groups.

Irradiation by an ArF excimer (6.4 eV) and the fourth harmonic generation of a Nd : YAG laser (4.7 eV) was found to be effective in crystallization with a loss of hydroxyl groups and a decrease in the sheet resistance of the sol-gel films. Transparent crystalline indium oxide films were successfully obtained by 6.4 eV laser irradiation at fluences below 20 mJ/(cm2·shot), whereas degradation of the films was induced by a relatively high-fluence beam.

Notizen: Abstract Porous titanate precursor microspheres (20–60 μm in diameter), with a high sorption capacity for radioactive wastes from nuclear reprocessing plants, have been produced on a 50 kg scale by spray-drying precursor sols. Well-dispersed, stable sols were produced by hydrolyzing acetic acid modified tetraisopropyltitanate and peptizing the titania hydrolysate with acidic zirconia sol. The resulting TiO2 /ZrO2 sols were routinely concentrated to 900 g dm−3 (oxide basis) and exhibited excellent stability. These sols were subsequently mixed with dispersible alumina powder and partially aggregated by adding calcium and barium nitrate salts. The resulting sols were spray-dried to produce microspheres with controlled porosity and morphology. The properties of the spray-dried powder were very dependent upon the chemical properties of the precursor sol. In particular, hollow spheres were produced from well-dispersed sols, whereas solid spheres could be produced from partially-aggregated sols.

Notizen: Abstract Nanosized phenyl-capped CdSe particles (quantum dots) were isolated, after preparation inside reversed micelles present in AOT/H2 O/heptane, and then successfully redispersed in amine-functionalized ormosils derived from 3-aminopropyl(trimethoxy)silane (i.e., SA). Doped ormosils were formed into films of a few μm thickness or small bulk samples. Host ormosils were structurally characterized by means of absorption spectroscopy, in the near- and mid-infrared ranges, and by 29Si and 13C solid-state nuclear magnetic resonance (NMR) spectroscopy. Doped ormosils were prepared and investigated via visible spectroscopy. The size of CdSe particles dispersed into the amino-ormosil hosts was calculated using the Effective Mass Model.

Notizen: Abstract Optical cores of preforms for drawing optical fibers doped with Er3+ and Yb3+ were fabricated by the sol-gel method with the aim of increasing the thickness of glass layers coated in a single coating cycle and to determine the relation between the preparation conditions and optical properties of the fibers. Al2O3-P2O5-SiO2 and TiO2-P2O5-SiO2 glasses have been studied as matrices for entrapping the rare-earth elements. Input sols have been prepared from silicon and titanium alkoxides, AlCl3, ErCl3, YbCl3, POCl3, water and a modifier under acidic catalysis of HCl. The sols were coated on the inner wall of a silica substrate tube and the gel layers were sintered at high temperatures up to 2000°C after which the tube was collapsed into the preform. Continuous and homogenous glass films with the maximum thickness of about 8 μm were fabricated. The influence of high-temperature heat treatment of the layers on their composition and optical attenuation was observed. The amplified stimulated emission of Er3+ around 1.55 μm was measured under the excitation of the fibers by an Nd : YAG laser at 1.064 μm.

Notizen: Abstract Metallic phthalocyanines exhibit, among many useful properties, the ability to protect sensors against short, intense optical pulses deleterious to efficient sensor operation. It is possible by means of low temperature sol-gel techniques to obtain porous monolithic materials encapsulating metallic ions, and all sorts of complex organic molecules. We report here the study of water-soluble metallic tetrasulphophthalocyanines (MTSPc) encapsulated in silica obtained by the sol-gel technique. These materials are transparent and stable and exhibit interesting optical properties. Absorption in the visible region of the spectrum reveals aggregation of the MTSPc species to form dimers which modify the optical absorption of excited states and the resulting optical properties. We investigate the relative importance of the various physicochemical parameters affecting dimer formation (dye concentration, pH, presence of species such as dimethylformamide, pyridine, etc.) in order to control and inhibit dimer formation. Two species are chosen as examples of the general behavior: CuTSPc, which shows a strong tendency to dimerize in water and in mixtures of solvents, and (OH)AlTSPc, which does not form dimers in aqueous solutions because of its hindering OH axial group. The experiments performed show that addition of dimethylformamide or pyridine does not inhibit aggregation in CuTSPc, simultaneously causing the blue color to disappear from the prepared gels. In the case of (OH)AlTSPc addition of relatively high concentrations of pyridine brings about the formation of non-monomeric species.

Notizen: Abstract A sol-gel route has been used to produce a nanoporous membrane for the separation of “greenhouse” gases from power plants at elevated temperatures (700–900 K). The membrane has a controlled pore size distribution and is multiphasic, in order to prevent grain growth and pore coarsening. The alumina/titania diphasic ceramic was obtained from alkoxide precursors with additional magnesium or lanthanum doping to stabilize low temperature phases and to provide possible chemisorption of CO2. It was found that both Mg and La stabilized the γ-alumina phase to higher temperature by as much as 200 K, although Mg is less effective.

Notizen: Abstract We report on the effect of organomodified sol-gel materials on fluorosensors for ammonia. The fluorosensors are based on ion pairs composed of an inert fluorophore and a pH-sensitive absorber dye and are embedded in sol-gel glass. Upon exposure to ammonia, deprotonation of the pH-sensitive dye bromophenolblue occurrs, and consequently, energy is transferred from the fluorophore rhodamine B or tetramethylrhodamine ethyl ester to the absorber. The response of the fluorosensors using different ratios of precursors, such as tetramethoxysilane and phenyltrimethoxysilane, is investigated. Detection limits for sol-gel layers composed of 50% tetramethoxysilane and 50% phenyltrimethoxysilane are as low as 0.1 mg/L of aqueous ammonia. Response times are of the order of 3 to 6 min for forward response. The reversibility of the sensor is related to the composition of the organically modified sol-gel glasses and is fastest for sensor layers composed of pure phenyltrimethoxysilane. Conditioning, regeneration, and storage of the layers are shown to be of vital importance for the performance of the sensor layers.

Notizen: Abstract Solanum acaule Bitt. is a disomic tetraploid (4x) wild potato species which is resistant to several potato diseases. Introgression of disease resistance and abiotic stress tolerance to the tetrasomic tetraploid (4x) cultivated potato (S. tuberosum L.) gene pool via crossing has been limited due to the difference in the endosperm balance number. In the present study, protoplast fusion was applied to produce hexaploid (6x) somatic hybrids between the parental lines, tetraploid (4x) S. acaule and two anther-derived dihaploid (2x) lines of S. tuberosum cv. White Lady. One callus (0.4%) of a total of 229 calli obtained regenerated into shoots in the fusion combination S. acaule (+) White Lady 15.dh.8.2.2. All the regenerated shoots were confirmed to be interspecific somatic hybrids using species-specific RAPD markers. In another fusion combination, S. acaule (+) White Lady 7.dh.23.1.1, fifteen calli (5%) regenerated into a total of sixteen shoots from 289 calli. All the analysed somatic hybrids between S. acaule and S. tuberosum were hexaploid. The mean DNA content (2C value) of the combination S. acaule (+) White Lady 15.dh.8.2.2 somatic hybrids (4.55 pg), was approximately the sum (4.69 pg) of the DNA contents of the parental lines, S. acaule (2.95 pg) and S. tuberosum (1.74 pg). In the greenhouse, the two somatic hybrids analysed were normal in their morphological characteristics and more vigorous than their parental lines. Most of the morphological characteristics were closer to the tetraploid S. acaule than to the dihaploid S. tuberosum. The interspecific somatic hybrids are currently being tested for frost tolerance and glycoalkaloid composition.

Notizen: Abstract  The genetic variation of 102 natural populations of wild barley growing in Spain was assessed using RAPDs (random amplified polymorphic DNA). The plant material included the annual species H. marinum subsp. marinum (22 populations) and subsp. gussoneanum (14), H. murinum subsp. murinum (7) and subsp. leporinum (35), and the perennial species H. bulbosum (17) and H. secalinum (7). Ten of the tested 64 arbitrary 10-mer primers amplified polymorphic DNA in all taxonomic units. Analyses was performed within and between populations, species and subspecies. The primers gave a total of 250 RAPD products. The level of polymorphism varied between taxonomic units depending on the primers employed and the plant reproductive system. In general, the most variable were the allogamous species H. secalinum and H. bulbosum and the autogamous H. marinum subsp. marinum. Among the amplified bands, 69 (27%) were shared by at least two different taxonomic units. The remaining bands were specific. The results demonstrate differences in the degree of similarity between taxonomic units. Jaccard’s similarity coefficients for interval measure within and between populations were used to produce a cluster diagram using the unweighted pair-group method (UPGMA). The different populations of the species and subspecies of Hordeum fell into three groups. The first group contained the populations belonging to both subspecies of H. marinum, plus those of H. secalinum. The populations of H. marinum subsp. gussoneanum were very closely associated. Those of H. marinum subsp. marinum were grouped in a broad cluster. The second group, occupying the innermost position of the tree, was very closely associated with the populations of both subspecies of H. murinum. The third branch segregated H. bulbosum. A series of RAPD markers were investigated by cleaving the amplified products of the same size with restriction endonucleases that recognize targets of 4- or 6-bp. The production of equivalent fragments following cleavage by the same enzyme would seem to demonstrate their homology in samples from different individuals, populations or taxonomic units.

Notizen: Abstract  Twenty eight somatic hybrid plants were identified following protoplast fusions between peppermint (Mentha piperita L. cv Black Mitcham), producing high-quality oil, and spearmint (Mentha spicata L. cv Native Spearmint), likewise producing high-quality oil and also possessing resistance to verticillium wilt. Prior to fusion, peppermint protoplasts were subjected to iodoacetic acid to inhibit cell division. Protoplasts of peppermint and spearmint were fused using polyethylene glycol plus DMSO. Fusion products were cultured according to an efficient protoplast-to-plant-cycle protocol developed for peppermint. Using this protocol, iodoacetic acid-treated peppermint protoplasts were not able to divide, whereas untreated spearmint protoplasts had the ability to produce callus but not shoots. Therefore, selection of somatic hybrid calli was based on the presumed capability of hybrid cells to form calli and shoots. Shoots in vitro were initially identified as hybrids using RAPD profiles. Subsequently, observations on morphology, chromosome counts, and Southern-hybridization patterns confirmed their hybrid status. The results of verticillium tests revealed that 18 somatic hybrids were more susceptible than Native Spearmint, while hybrid II-14 had a level of susceptibility intermediate between that of the fusion parents. Oil-analysis of hybrid plants indicated that they all have a GC-profile typical of spearmint oil.

Notizen: Abstract  Alfalfa (Medicago sativa L.) is a forage legume of world-wide importance whose both allogamous and autotetraploid nature maximizes the genetic diversity within natural and cultivated populations. This genetic diversity makes difficult the discrimination between two related populations. We analyzed this genetic diversity by screening DNA from individual plants of eight cultivated and natural populations of M. sativa and M.  falcata using the RAPD method. A high level of genetic variation was found within and between populations. Using five primers, 64 intense bands were scored as present or absent across all populations. Most of the loci were revealed to be highly polymorphic whereas very few population-specific polymorphisms were identified. From these observations, we adopted a method based on the Roger’s genetic distance between populations using the observed frequency of bands to discriminate populations pairwise. Except for one case, the between-population distances were all significantly different from zero. We have also determined the minimal number of bands and individuals required to test for the significance of between-population distances.

In total, 198 random amplified polymorphic DNAs (RAPDs) and 180 restriction fragment length polymorphism (RFLP) loci were scored by using ten primers and 14 enzyme-probe combinations respectively. The polymorphism observed for these markers revealed a high degree of genetic diversity in mangroves at both inter-specific or inter-generic levels. A dendrogram, constructed after pooling both RAPD and RFLP data, using a similarity index was analysed for genome relationships among these species. The dendrogram showed clustering of all the major mangroves, except for Nypa fruticans (Arecaceae), into one group. All species under the tribe Rhizophorae formed a sub-cluster, to which Xylocarpus granatum was found to be the most closesly related species. The clustering pattern implied that Excoecaria agallocha and Acanthus ilicifolius should be considered as true minor mangroves. The present study also provided molecular data favouring the separation of Avicennia spp. from the Verbenaceae to create a monotypic family the Avicenniaceae. The separation of Viscum orientale into the Viscaceae was also favoured.

Notizen: Abstract  We have constructed a sex-averaged genetic linkage map in coastal Douglas-fir (Pseudotsuga menziesii [Mirb.] Franco var ‘menziesii’) using a three-generation outcrossed pedigree and molecular markers. Our research objectives are to learn about genome organization and to identify markers associated with adaptive traits. The map reported here is comprised of 141 markers organized into 17 linkage groups and covers 1,062 centiMorgans (cM). Of the markers positioned on the map, 94 were derived from a Douglas-fir complimentary-DNA (cDNA) library that was constructed from new-growth needle tissue. Other markers include 11 Douglas-fir genomic-DNAs, 20 loblolly pine (Pinus taeda L.) cDNAs, 15 random amplified polymorphic DNAs (RAPDs) and a PCR-amplified phytochrome probe. A high degree of variation was detected in each of the two parents of our mapping population, and many of the restriction fragment length polymorphism (RFLP) and RAPD phenotypes were complex. Marker data were analyzed for linkage using mapping software JOINMAP version 2.0.

Notizen: Abstract  Seven sorghum restorer lines that differentially restore (or maintain) the A1 and A2 cytoplasmic male-sterile (CMS) cytoplasms were studied by RFLP analyses of their mtDNAs and RAPD analyses of their mitochondrial DNA (mtDNA) and total DNA to understand nuclear mitochondrial combinations that are present in these lines. RFLP data from 11 mitochondrial gene probes were inadequate to classify these seven lines. However, the analysis of RAPD profiles of total DNA could distinguish these lines on the basis of their ability to restore completely or partially the fertility in the A1/A2 CMS cytoplasms. Interestingly, RAPD profiles of mtDNAs of these lines also followed the same pattern as that of the total DNA. These results indicate that the different restorer lines possess specific nuclear-cytoplasm combinations. Further, the results also show that the RAPD technique can be used to identify markers for different cytoplasms used in CMS.

Notizen: Abstract  Bulked segregant analysis and comparative mapping were applied to identify molecular markers linked to the Rfo restorer gene used for the Ogu-INRA cytoplasmic male-sterility system in rapeseed. These markers were then used to localise the radish introgression on the B. napus genetic map constructed from the cross ‘Darmor.bzh’ x ’Yudal’. The introgression mapped on the DY15 linkage group. From the comparison of this latter group to the linkage group constructed on a F2 progeny segregating for the radish introgression, it was concluded that the introgression had occurred through homoeologous recombination, that it was not distal and that it had replaced a B. napus region of around 50 cM. A QTL involved in aliphatic seed glucosinolate content was located on the DY15 linkage group at a position corresponding to one end of the introgression. The DNA markers identified in this study are being used in map-based cloning of the Rfo gene and in marker-assisted selection.

Notizen: Abstract  We report the molecular mapping of the py-1 gene for resistance to corky root rot [Pyrenochaeta lycopersici (Schneider and Gerlach)] in tomato using RAPD and RFLP marker analysis. DNA from near-isogenic lines (NILs) of tomato differing in corky root rot resistance was screened with 575 random oligonucleotide primers to detect polymorphic DNAs linked to py-1. Three primers (OPW-04, OPC-02, OPG-19) revealed polymorphisms between the NILs. Twelve resistant and eight susceptible DNA pools derived from segregating F3 families were used to confirm that the RAPD markers were linked to the py-1 gene. Two of the linked amplified fragments, corresponding to OPW-04 and OPC-02, were subsequently cloned and mapped on the tomato molecular linkage map as RFLPs. These clones were located between TG40 and CT31 on the short arm of chromosome 3. Further analysis with selected RFLP markers showed that 7% (8.8 cM) of chromosome 3 of the resistant line ‘Moboglan’ was introgressed from the L. peruvianum donor parent. Three RFLP markers (TG40, TG324, and TG479) from the introgressed part of chromosome 3 were converted to cleaved amplified polymorphism (CAP) markers for use in a polymerase chain reaction (PCR) assay. These PCR markers will allow rapid large-scale screening of tomato populations for corky root rot resistance.

Notizen: Abstract  Random amplified polymorphic DNA (RAPD) markers linked to two morphological markers ( fa and det), three ramosus genes (rms2, rms3 and rms4) and two genes conferring flowering response to photoperiod in pea (sn, dne) were selected by bulk segregant analysis on F2 populations. Two RAPD fragments were cloned and sequenced to generate the two SCAR markers V20 and S2 which are linked to rms3 and dne, respectively. All these genes, except rms2, were previously located on the pea classical linkage map. Rms2 mapped to linkage group IB which contains the afila gene. Precise genetic maps of the regions containing the genes were obtained and compared to the RAPD map generated from the recombinant inbred-lines population of the cross Térèse×K586. This cross was chosen because several mutants were obtained from cultivars Térèse and Torsdag (K586 was derived from Torsdag). This collection of isogenic lines was used for the construction of F2 mapping populations in which polymorphic RAPD markers were already known and mapped. Moreover, the well-known problem in pea of variability in the linkage associations between crosses was avoided. This work contributes to the precise integration between the classical map and the molecular maps existing in pea.

Notizen: Abstract  The potential of PCR-based markers for construction of a genetic linkage map in Einkorn wheat was investigated. From a comparison of polymorphisms between two Einkorn wheats, Triticum monococcum (Mn) and T. boeoticum (Bt), we obtained 49 polymorphic bands produced by 33 primers for inter-simple sequence repeat (ISSR) and 36 polymorphic bands shown by 25 combinations of random amplified polymorphic DNA (RAPD) primers for mapping in 66 individuals in the F2 population. Although 44 ISSR fragments and 29 RAPD fragments statistically showed a 3 : 1 segregation ratio in the F2 population, only 9 markers each of the ISSR and RAPD bands were able to be mapped on the RFLP linkage map of Einkorn wheat. ISSR markers were distributed throughout the chromosomes. The mapped positions of the ISSR markers seemed to be similar to those obtained by the RFLP markers. On the other hand, 4 of the 9 RAPD markers could map the RFLP marker-poor region on the short arm of 3Am, suggesting a potential to map novel regions containing repetitive sequences. Comparisons of the genetic linkage map of Einkorn wheat to the linkage map and cytological map of common wheat revealed that the marker orders between the two maps of Einkorn wheat and common wheat coincided except for 4A, which harbors chromosome rearrangements specific for polyploid wheats, indicating a conservatism between the two genomes. Recombinations in Einkorn wheat chromosomes took place more frequently around the centromere and less at the distal part of chromosomes in comparison to those in common wheat. Nevertheless, recombinations even in Einkorn wheat chromosomes were strongly suppressed around the centromere. In fact, the markers located within 1 cM of the centromere were located almost in the central part of the chromosome arm.

Notizen: Abstract  The variety Vitis vinifera cv Sultanine presents a type of seedlessness in which fertilization occurs but seeds subsequently fail to develop. It has been suggested that this trait might be controlled by three complementary recessive genes regulated by a dominant gene named I. Bulk segregant analysis was used to search for random amplified polymorphic DNA (RAPD) markers linked to the I gene in progeny obtained by crossing two partially seedless genotypes. One hundred and forty decamer primers were screened using bulks obtained by pooling the DNA of extreme individuals from the phenotypic distribution. We identified two RAPD markers which appeared tightly linked to I (at 0.7 and 3.5 cM respectively). The closest marker was used to develop a codominant SCAR (sequence characterized amplified region), named SCC8. This latter marker appeared of great value either to exclude from the progeny potentially seeded individuals or to select for seedless individuals. Indeed, all the seeded individuals of the progeny were found to be homozygous scc8 -/scc8 -, and all the individuals homozygous SCC8 +/SCC8 + were seedless. Moreover, this marker was successfully applied to other natural seedless varieties where codominance persisted. SCC8 was also used to dissect more precisely the genetics of seedlessness. ANOVA analysis indicated that this SCAR marker accounted for at least 64.9% of the phenotypic variation of the seed’s fresh weight and for at least 78.7% of the phenotypic variation of the seed’s dry matter. These results confirmed the presence of a major gene, and also the existence of other complementary recessive genes, controlling the expression of seedlessness.

Notizen: Abstract  Monosomic chromosome addition lines of Brassica oxyrrhina in the background of alloplasmic B. campestris carrying B. oxyrrhina cytoplasm were generated and characterised through morphology, cytology and molecular (RAPD) analysis. Four successive backcrosses of the synthetic alloploid B. oxycamp with B. campestris yielded 24 monosomic addition plants that were grouped into seven different synteny groups based on morphological similarity and RAPD patterns. Each synteny group exhibited morphological features diagnostic for the presence of individual B. oxyrrhina chromosomes including some novel phenotypes. Meiotic studies of the addition lines revealed the homoeology of four B. oxyrrhina chromosomes (synteny groups 1, 3, 5 and 6 ) with B. campestris chromosomes as indicated by trivalent associations, with the highest homoeology (44.23%) in synteny group 1 and the lowest (6.1%) in synteny group 3. Seed fertility of the addition lines ranged from 94.85% (synteny group 1) to 56.98% (synteny group 5). All of the addition lines were male-sterile except synteny group 6 which had 12–16% stainable pollen. Ovule transmission of the B. oxyrrhina chromosomes added to the progenies of addition lines ranged from 23.52% (synteny group 6) to 14% (synteny group 7). RAPD analysis confirmed the validity of synteny grouping based on morphological observations. Approximately 45% of the primers studied were informative, giving B. oxyrrhina-specific RAPD bands unique for each synteny group, except group 6.

Notizen: Abstract  NESTUR (needle-to-stem unit rate) is a stem growth index of conifer seedlings that measures the efficiency of stemwood production per unit of needle growth, and is related to other seedling traits such as height, stem diameter, stem volume and needle volume. Quantitative trait loci (QTLs) affecting the expression of stem growth efficiency in radiata pine seedlings were investigated using a RAPD linkage map constructed from markers scored on haploid, megagametophytic DNA. Four putative QTLs were detected which accounted for 8.5–36.4% of the population variance. A search for evidence of epistasis, using both complete pairwise and conditional interactions, did not yield any statistically significant result. Over a 3-year period, seedlings with high-NESTUR marker alleles showed a superior growth performance of 17–40% for height, diameter and volume over those with low-NESTUR marker alleles.

Notizen: Abstract  A genetic map of Pedunculate oak (Quercus robur) was constructed based on one 5S rDNA, 271 RAPD, ten SCAR, 18 microsatellite, one minisatellite, and six isozyme markers. A total of 94 individuals from a full-sib family was genotyped. Two maps, including 307 markers, were constructed according to the “two-way pseudo-testcross” mapping strategy. Testcross markers segregating in the 1 : 1 ratio were first used to establish separate maternal (893.2 cM, 12 linkage groups) and paternal (921.7 cM, 12 linkage groups) maps. Both maps provided 85–90% genome coverage. Homologies between the male and female linkage groups were then identified based on 74 intercross markers segregating in the 3 : 1, 1 : 2 : 1 and 1 : 1 : 1 : 1 ratios (RAPDs, SCARs, SSRs, 5S rDNA and isozymes) in the hybrid progeny. In each map, approximately 18% of the studied markers showed segregation distortion. More than 60% of the skewed markers were due to an excess of heterozygote genotypes. This map will be used for: (1) studying the molecular organisation of genomic regions involved in inter- and intraspecific differentiation in oaks and (2) identification of QTLs for adaptive traits.

Notizen: Abstract  A genetic linkage map of peach [Prunus persica (L.) Batch] was constructed in order to identify molecular markers linked to economically important agronomic traits that would be particularly useful for long-lived perennial species. An intraspecific F2 population was generated from self-pollinating a single F1 plant from a cross between a flat non-acid peach, ‘Ferjalou Jalousia®’ and an acid round nectarine ‘Fantasia’. Mendelian segregations were observed for 270 markers including four agronomic characters (peach/nectarine, flat/round fruit, acid/non-acid fruit, and pollen sterility) and 1 isoenzyme, 50 RFLP, 92 RAPD, 8 inter-microsatellite amplification (IMA), and 115 amplified fragment length polymorphism (AFLP) markers. Two hundred and forty-nine markers were mapped to 11 linkage groups covering 712 centiMorgans (cM). The average density between pairs of markers is 4.5 cM. For the four agronomic characters studied, molecular markers were identified. This map will be used for the detection of QTL controlling fruit quality in peach and, particularly, the acid and sugar content.

Notizen: Abstract  Neotropical tree crops are affected by a combination of biological and human factors that complicate the study of genetic diversity and crop evolution. Genetic diversity and relationships among southern Mexican populations and horticultural collections of Theobroma cacao (chocolate, cocoa, cacao) are examined in light of the agricultural practices of the Maya. Collections of cacao were obtained from the extremes of its geographic range including archeological sites in southern Mexico where cacao was first domesticated. Genetic diversity was assayed by 57 informative random amplified polymorphic DNA (RAPD) marker loci. A unique sample of the total diversity found in this study exists in the southern Mexican populations. These populations are significantly different from all other cacao with regards to their profile of RAPD bands, including the ‘criollo’ variety, their morphological and geographical group. A population of cacao found in a sinkhole (cenote) in northern Yucatan with genetic affinities to populations in Chiapas suggests the Maya maintained plants far away from their native habitat. This finding concurs with known agroforestry practices of the Maya. Modern efforts to increase germplasm of tropical tree crops such as cacao should carefully examine archeological sites where genetic diversity, either deliberately or by chance, was collected and maintained by ancient cultures.

Notizen: Abstract  Embryogenic nucellar cultures of two polyembryonic mango cultivars, ‘Hindi’ and ‘Carabao’, were selected for resistance to the culture filtrate and phytotoxin of a virulent strain of Colletotrichum gloeosporioides Penz. that was isolated from mango leaves. The cultures were recurrently selected either with progressively increasing concentrations of culture filtrate or by continuous challenge with the same concentration of either culture filtrate phytotoxin. Mycelium growth was inhibited when the pathogen was cocultured with the selected, resistant embryogenic cultures. Conditioned plant growth medium containing macerated resistant embryogenic cultures did not inhibit mycelium growth, confirming that extracellular antifungal compounds were involved in the defense response. Enhanced secretion of chitinase and glucanase was observed in the plant growth medium in which resistant embryogenic cultures and regenerated somatic embryos were grown in comparison with the controls.
