    Beschreibung: The seismic sequence which occurred in 1998 south of the Island of Ustica (offshore North Sicily) has been interpreted in the frame of historical recent seismicity and the area’s tectonics. This seismic sequence, characterised by shallow-depth and low-magnitude earthquakes (Md max. 4.3), took place in the thick and complex stack of the Sicilian-Maghrebian fold-and-thrust belt. The spatial distribution of the epicentres recorded during the January-August 1998 shows a cluster roughly NW-SE trending. The few shocks which occurred immediately after the Md 4.1 shock of the 14th September were located south-east of Ustica with a rough NE-SW trend. The focal mechanisms of major shocks are of a thrust type with horizontal compressive axes generally N-S trending. In the kinematic evolution of the study area, alternating extensional and contractional events have been recognised as having taken place during the Plio-Pleistocene. The present day seismic activity pointing out a new contractional episode is well framed in this evolutionary trend. The occurrence of pre-existing faults and the large number of earthquakes with low-magnitude support the hypothesis that this seismicity could be related to a frictional (re)activation of faults. Active compression in offshore North Sicily probably reflects the northwards motion of Africa relative to the Eurasian plate.

    Beschreibung: We investigate the background seismicity of Central Italy in the area including southern Latium, Abruzzi and Molise (SLAM project). Within this region, the central Apenninic chain has been historically affected by many strong earthquakes, some of them very destructive such as the 1349 event (Mw ˜6.7) located at the border between southern Latium and western Molise, the 1654 event (Mw ˜6.4) in the southern Latium-Abruzzi area, and the 1805 Boiano earthquake in the northern Matese range (Mw 6.7). The last important seismic sequence occurred in May 1984 in the Comino Valley, southestern Latium (Mw 5.8). The recent activity is characterized by diffuse low-magnitude seismicity, punctuated by localised small sequences during 2009-2012. Our study focuses on the analysis of seismicity recorded in the period 2009-2013. We present earthquake locations and focal mechanism solutions obtained by standard procedures and an optimized regional 1D velocity model based on the Velest algorithm. The waveform data set was collected from the digital recordings of the permanent stations of the Italian national seismic network, the Abruzzi and Molise regional seismic networks, and from a dense seismic survey carried out in the region between November 2011 and May 2013. The temporary network consisted of 17 three-component seismic stations all equipped with Reftek RT130 digitizers and Lennartz 3D/5s sensors. The deployment of this array improved significantly the detection and location of background seismicity. We relocated more than 4300 events with magnitude ML ranging from about 0.5 to 4.2. Earthquakes distribution shows hypocentral depths concentrated within the upper crust, between 2 and 20 km of depth, and is mostly clustered along the Apenninic chain axis. The computed fault-plane solutions generally display normal fault mechanisms, confirming the extensional NE-SW processes active since Pleistocene in the study region.

    Beschreibung: Seismological, soil temperature and hydrological data from Mt. Vesuvius are collected to characterize the present-day activity of the volcanic/hydrothermal system and to detect possible unrest-related phenomena. We present patterns of seismicity and soil temperature in the crater area during the period February 2004-December 2011. The temporal distribution of number and depth of Volcano-Tectonic earthquakes and the energy release are considered. Hourly data of soil temperature have been acquired since January 2004 in different locations along the rim and within the crater. The observed changes of temperature are studied to establish a temporal-based correlation with the volcanic activity and/or with external forcing, as variations of the regional and local stress field acting on the volcano or meteorological phenomena. The comparison between seismic activity and temperature data highlights significant variations possibly related to changes in fluid circulation in the hydrothermal system of the volcano. The common continuous observations start just before a very shallow earthquake occurred in August 2005, which was preceded by a thermal anomaly. This coincidence has been interpreted as related to fluid-driven rock fracturing, as observed in other volcanoes. For the successive temporal patterns, the seismicity rate and energy release are characterized by slight variations accompanied by changes in temperature. This evidence of reactivity of the fumarole thermal field to seismic strain can be used to discriminate between tectonic and volcanic signals at Mt. Vesuvius.

    Beschreibung: Between 1994 and 2010, we completed 16 thermal surveys of Vulcano’s Fossa fumarole field (Aeolian Islands, Italy). In each survey, between 400 and 1,200 vent temperatures were collected using a thermal infrared thermometer from distances of ∼1 m. The results show a general decrease in average vent temperature during 1994–2003, with the average for the entire field falling from ∼220°C in 1994 to ∼150°C by 2003. However, between 2004 and 2010, we witnessed heating, with the average increasing to ∼190°C by 2010. Alongside these annual-scale field-wide trends, we record a spatial re-organisation of the fumarole field, characterised by shut down of vent zones towards the crater floor, matched by rejuvenation of zones located towards the crater rim. Heating may be expected to be associated with deflation because increased amounts of vaporisation will remove volume from the hydrothermal system Gambino and Guglielmino (J Geophys Res 113: B07402, 2008). However, over the 2004–2010 heating period, no ground deformation was observed. Instead, the number of seismic events increased from a typical rate of 37 events per month during 1994–2000 to 195 events per month during 2004–2010. As part of this increase, we noticed a much greater number of high-frequency events associated with rock fracturing. We thus suggest that the heating event of 2004–2010 was the result of changed permeability conditions, rather than change in the heat supply from the deeper magmatic source. Within this scenario, cooling causes shut down of lower sectors and re-establishment of pathways located towards the crater rim, causing fracturing, increased seismicity and heat flow in these regions. This is consistent with the zone of rejuvenation (which lies towards and at the rim) being the most favourable location for fracturing given the stress field of the Fossa cone Schöpa et al. (J Volcanol Geotherm Res 203:133–145, 2011); it is also the most established zone, having been active at least since the early twentieth century. Our data show the value of deploying multi-disciplinary geophysical campaigns at degassing (fumarolic) hydrothermal systems. This allows more complete and constrained understanding of the true heat loss dynamics of the system. In the case study presented here, it allows us to distinguish true heating from apparent heating phases. While the former are triggered from the bottom-up, i.e. they are driven by increases in heat supply from the magmatic source, the latter are triggered from the top-down, i.e. by changing permeability conditions in the uppermost portion of the system to allow more efficient heat flow over zones predisposed to fracturing.

    Beschreibung: In the frame of the NERA project and under the WP 8, an inventory of schools hosting seismic stations for educational purposes has been compiled with the aim, among others, to establish a network for data exchange. Such an inventory reveals that there are more than 600 instruments, most of which in full activity, in schools of the Mediterranean area. The number of stations is somewhat proportional to the date when educational projects began (Zollo et al., in press), so countries like France, United Kingdom or Ireland, all places where a long tradition in “seismology in schools” is established , own most of the existing stations. The make and technical characteristics of these devices widely vary: some schools have assembled their own mechanical seismometer with very simple materials while some others have designed an acquisition system and coupled it to a sensor available on the market. In some cases, stations are bought from semi-professional or professional manufacturers that have devoted a special care to the educational field. These factories have designed cheap instruments the technology of which is based on the more expensive instruments that are instead available at professional level. These recording items are often equipped with an internet connection, have a broad-band like seismometer, offer a real time view of the recording (helicorder) and provide data in SAC format. In a sentence, they are very much similar to the devices currently used in professional (national or regional) networks to monitor seismicity. Moreover, in principle stations installed in a country are very much alike or perfectly identical, representing as a matter of fact a semi-professional seismic network.. It is then straightforward to wonder what is the role of the data recorded and stored by these instruments and especially what is the potential of these information. Are these instruments providing any additional information to the professional seismic networks ? Could they complement a professional database ? In this paper a rough analysis of the data collected by a school network is analysed and compared with “official” data. It is shown that in some cases seismograms recorded from stations in schools can perform very well and their data could, under certain circumstances and with some limitations, be used instead of / in addition to professional data.

    Beschreibung: Multi-beam bathymetry and high-resolution low-penetration seismic reflection profiles of the offshore extensions of the Bétic Internal Zone off Sierra de Cartagena-La Unión margin along its south side and the Mar Menor margin along its east side, the Mazarrón Escarpment forming its southern boundary and the adjacent oceanic Algero-Balearic basin have provided images of the neo-tectonic structures of the region equal to those provided by subaerial photography. For the first time we mapped with unprecedented detail the Mazarrón Escarpment and the Southeast margin of Iberia. The first-order structures of the region are due to the consequence of the collision of the African and Eurasian plates during the Alpine orogeny in late Oligocene-Middle Miocene, the westward migration of the Alborán plate in the Middle Miocene and the desiccation of the Mediterranean in the Messinian (Late Miocene) that led to the deposition of evaporites in the Algero-Balearic basin and erosion of the Mazarrón Escarpment, the Sierra de Cartagena-La Unión shelf, the Mar Menor margin and the adjacent coast. Our data images second order tectonic features (neo-tectonic features) superimposed on the larger structures. These include the deformation of the strata in the Algero-Balearic basin by the gliding of the Plio-Quaternary sediments on Messinian halite on the margins of the basin and sediment loading in its center, the Late Miocene-Quaternary deformation of the area north of the Mazarrón Escarpment resulting from the continuous oblique convergence of the African and Eurasian plates in a NNW-SSE direction, the Miocene to Pleistocene volcanic edifices and pinnacles (dikes), the pockmarks formed by the extrusion of gas/water via faults and the massive gravitational failure of the Mazarrón Escarpment triggered by this plate convergence. The data also show in detail features formed on the Mazarrón Escarpment during the Messinian, Pliocene and Pleistocene regressions and those on the shelf formed during the Pleistocene glacially induced regressions/transgression and sediment drifts generated by modern currents.

    Beschreibung: The technological demand to push the gigahertz (10(9) hertz) switching speed limit of today's magnetic memory and logic devices into the terahertz (10(12) hertz) regime underlies the entire field of spin-electronics and integrated multi-functional devices. This challenge is met by all-optical magnetic switching based on coherent spin manipulation. By analogy to femtosecond chemistry and photosynthetic dynamics--in which photoproducts of chemical and biochemical reactions can be influenced by creating suitable superpositions of molecular states--femtosecond-laser-excited coherence between electronic states can switch magnetic order by 'suddenly' breaking the delicate balance between competing phases of correlated materials: for example, manganites exhibiting colossal magneto-resistance suitable for applications. Here we show femtosecond (10(-15) seconds) photo-induced switching from antiferromagnetic to ferromagnetic ordering in Pr0.7Ca0.3MnO3, by observing the establishment (within about 120 femtoseconds) of a huge temperature-dependent magnetization with photo-excitation threshold behaviour absent in the optical reflectivity. The development of ferromagnetic correlations during the femtosecond laser pulse reveals an initial quantum coherent regime of magnetism, distinguished from the picosecond (10(-12) seconds) lattice-heating regime characterized by phase separation without threshold behaviour. Our simulations reproduce the nonlinear femtosecond spin generation and underpin fast quantum spin-flip fluctuations correlated with coherent superpositions of electronic states to initiate local ferromagnetic correlations.     Beschreibung: The Holocene is the most recent geological epoch spanning from about 11700 years ago to the present day. The most important human civilizations appeared during the Holocene. From the Holocene onwards, environmental changes, and the hazards associated with them, became extremely important for their impact on historical events, in some cases blending with humanity’s vicissitudes and influencing the rise and decline of civilizations. This paper summarises the geological and climatic conditions of Northern Europe during the Holocene and tries to determine whether or not they support the hypothesis formulated by Felice Vinci (Vinci, 2003) about the migration of Baltic populations towards the Mediterranean in the Bronze Age at the end of the “climatic optimum” (Houghton et al., 1990; Rohling & De Rijk, 1999). This study presents data on glacio-eustatic changes and on isostatic uplift together with information on probable tsunamis that occurred in the North Atlantic, North Sea, Scandinavia and the Baltic Sea. Moreover, some data on catastrophic events that affected the Mediterranean region are reported, because these catastrophes could have favoured the settlement of “people coming from the sea” that took advantage of the demographic and socio-economic weakening of indigenous populations (Driessen, 2002). The paper aims to provide geological and palaeogeographic constraints to the hypotheses formulated by Felice Vinci on the migration of Scandinavians towards the Mediterranean. The data analysed have been collected from the available scientific literature (see references). The amount of information available for each geological phenomenon is vast and sometimes theories developed from the same data are in conflict. The comparison between the Mediterranean and the Baltic areas (one of which could have been the theatre of the Homeric events) will be useful to find evidence of geological phenomena within the Homeric texts, giving useful indications to better understand where the poems are set or at least to provide interesting discussion points related to Felice Vinci’s hypothesis (Vinci 2003).

    Beschreibung: Several volcanoes worldwide have shown changes in their stress state as a consequence of the deformation produced by the pressurization of a magmatic body. This study investigates seismic swarms occurring on the western flank of Mt. Etna in January 1997 - January 1998. Integrating seismic observations and geodetic data, we constrained the seismogenic fault system, and on the basis of stress tensor inversion and SHMAX analyses, we infer an inflating pressure source located at 5.5 km b.s.l. beneath the west portion of summit area. Evaluation of Coulomb failure stress (CFS) related to the proposed model, showed how a large part of the seismogenic fault underwent a significant CFS increase (500 kPa). We infer the presence of a sub-vertical faulted region, potentially weak, N50°E oriented beneath the western sector of Mt. Etna. This structure could be brought closer to failure thereby generating seismic swarms as the effect of elastic stress transfer induced by movement and/or overpressure of magmatic masses within the upper crust under the volcano.

    Beschreibung: At Stromboli volcano, Italy, continuous seismic monitoring and periodic, visual observations of volcanic activity for surveillance purposes began in the mid-1980s. Since 1985, two eruptions have occurred, one lasting from December, 1985 until April, 1986, and one in May, 1993. There have also been two small overflows, in 1990 and 1994. Since these episodes of lava effusion, the persistent Strombolian activity of the volcano has had several fluctuations during the past 15 years. Some episodes climaxed in powerful explosions. According to seismic records, these paroxysms consisted of a variable number of explosion quakes in rapid succession (i.e. from tens of seconds to a few minutes), associated with a notable increment in the amplitude of volcanic tremor. Throughout these episodes - which are called explosive sequences - lapilli, fragments of old rock, and bombs of varying dimensions were ejected, affecting an area greater than the crater terrace where the active craters are located. In this article, we describe the explosive sequences recorded at Stromboli between 1985 and 1999. We provide a characterization in terms of reduced displacement and duration for nine episodes occurring in 1998 and 1999. Their reduced displacements range from 15 to 124 cm2; their durations are between 6 and 18 min. We find no change in the frequency content of the seismic signal several minutes before and during the sequences. Considering medium- to long-term behavior, the spectral amplitude of the seismic signal decreases or has low values over several months preceding the occurrence of the paroxysms. This feature is common to 20 of the 22 explosive sequences, and is indicative of internal conditions that periodically characterize the feeder. We surmise that the paroxysms are the result of the partial obstruction of the volcanic conduit when the magma column is low or dropping. The onset of the explosive sequence, causing the sudden removal of the material which forms the obstruction, would trigger a sudden depressurization of the conduit and the rapid rise of magma from depth.

    Beschreibung: Planktonic protozoan grazers have the potential to significantly affect the chemistry of particle-associated trace metals. This is due both to the importance of protists as consumers of bacterial-sized particles, and to the unique low-pH, enzyme-rich microenvironment of the grazer food vacuole. This thesis examines the role of protozoan grazers in the marine geochemistry of strongly hydrolyzed, particle-reactive trace metals, in particular Th and Fe. A series of tracer experiments was carried out in model systems in order to determine the effect of grazer-mediated transformations on the chemical speciation and partitioning of radioisotopes C9Fe, 234Th, 51Cr) associated with prey cells. Results indicate that protozoan grazers are equally able to mobilize intracellular and extracellular trace metals. In some cases, protozoan regeneration of trace metals appears to lead to the formation of metal-organic complexes. Protozoan grazing may generate colloidal material that can scavenge trace metals and, via aggregation, lead to an increase in the metal/organic carbon ratio of aggregated particles. Model system experiments were also conducted in order to determine the effect of grazers on mineral phases, specifically colloidal iron oxide (ferrihydrite). Several independent techniques were employed, including size fractionation ors9Fe-labeled colloids, competitive ligand exchange, and iron-limited diatoms as "probes" for bioavailable Fe. Experimental evidence strongly suggests that protozoan grazing can affect the surface chemistry and increase the dissolution rate of iron oxide phases through phagotrophic ingestion. In further work on protozoan-mediated dissolution of colloidal Fe oxides, a novel tracer technique was developed based on the synthesis of colloidal ferrihydrite impregnated with 133Ba as an inert tracer. This technique was shown to be a sensitive, quantitative indicator for the extent of ferrihydrite dissolution/alteration by a variety of mechanisms, including photochemical reduction and ligand-mediated dissolution. In field experiments using this technique, grazing by naturally occuring protistan assemblages was shown to significantly enhance the dissolution rate of colloidal ferrihydrite over that in non-grazing controls. Laboratory and field results indicate that, when integrated temporally over the entire euphotic zone, protozoan grazing may equal or exceed photoreduction as a pathway for the dissolution of iron oxides.

    Beschreibung: At the oceanic core complex that forms the Atlantis Massif at 30°N on the Mid-Atlantic Ridge, slip along the detachment fault for the last 1.5–2 Ma has brought lower crust and mantle rocks to the seafloor. Hydroacoustic data collected between 1999 and 2003 suggest that seismicity occurred near the top of the Massif, mostly on the southeastern section, while detected seismicity along the adjacent ridge axis was sparse. In 2005, five short-period ocean bottom seismographs (OBS) were deployed on and around the Massif as a pilot experiment to help constrain the distribution of seismicity in this region. Analysis of six months of OBS data indicates that, in contrast to the results of the earlier hydroacoustic study, the vast majority of the seismicity is located within the axial valley. During the OBS deployment, and within the array, seismicity was primarily composed of a relatively constant background rate and two large aftershock sequences that included 5 teleseismic events with magnitudes between 4.0 and 4.5. The aftershock sequences were located on the western side of the axial valley adjacent to the Atlantis Massif and close to the ridge-transform intersection. They follow Omori's law, and constitute more than half of the detected earthquakes. The OBS data also indicate a low but persistent level of seismicity associated with active faulting within the Atlantis Massif in the same region as the hydroacoustically detected seismicity. Within the Massif, the data indicate a north-south striking normal fault, and a left-lateral, strike-slip fault near a prominent, transform-parallel, north-facing scarp. Both features could be explained by changes in the stress field at the inside corner associated with weak coupling on the Atlantis transform. Alternatively, the normal faulting within the Massif might indicate deformation of the detachment surface as it rolls over to near horizontal from an initial dip of about 60° beneath the axis, and the strike-slip events may indicate transform-parallel movement on adjacent detachment surfaces.

    Beschreibung: Daily topical application of the aqueous ethanolic extract of the marine sea grass, Thalassia testudinum, on mice skin exposed to UVB radiation resulted in a dose dependent recovery of the skin macroscopic alterations over a 6-day period. Maximal effect (90%) occurred at a dose of 240 μg/cm2, with no additional effects at higher doses. Bioassay-guided fractionation of the plant extract resulted in the isolation of thalassiolin B (1). Topical application of 1 (240 μg/cm2) markedly reduces skin UVB-induced damage. In addition, thalassiolin B scavenged 2,2-diphenyl-2-picrylhydrazyl radical with an EC50=100 μg/ml. These results suggest that thalassiolin B is responsible for the skin regenerating effects of the crude extract of T. testudinum

    Beschreibung: Bioguided fractionation of Agelas cerebrum crude extract resulted in isolation of four bromopyrrole and four bromopyrrole aminoimidazole alkaloids, identified as 5-bromopyrrole-2-carboxylic acid (1), 4-bromopyrrole-2-carboxylic acid (2), 3,4-bromopyrrole-2-carboxylic acid (3), 4,5-bromopyrrole-2-carboxylic acid (4), oroidin (5), bromoageliferin (6), dibromoageliferin (7) and dibromosceptrin (8) on the basis of spectroscopic data analyses (UV, IR, HRMS, 1D and 2D NMR) and comparison with literature data.     Beschreibung: On June 8, 2008 a Mw = 6.4 earthquake occurred in NW Peloponnese, western Greece. This event is the largest strike-slip earthquake to occur in western Greece during the past 25 years. No surface rupture was observed. Many rock falls, slides, and liquefaction features have been found as is typical for an earthquake of this size. Double-difference relocations of 370 aftershocks show a linear pattern of events and define a clear NE-SW striking mainshock fault plane. The hypocentrer was determined at 18 km depth beneath village Mihoi in SW Achaia. The 24-hr aftershock region extends approximately 30 km in length, and the width of the surface projection of the aftershocks ranges between 5–10 km. The depth of the aftershocks rarely exceeds 22 km. Analysis of high-rate GPS data showed that station RLS (Riolos) which is located 12.8 km N5 W of the epicenter was displaced co-seismically 7 mm to the North in agreement with right-lateral kinematics of the rupture. Static (Coulomb) stress transfer analysis indicates loading of faults near the towns of Patras (north) and Amaliada (south), respectively. The earthquake put more emphasis on the role of strike-slip in the deformation of western Greece also indicating that seismic strain is partitioned between strike-slip and normal-slip events due to obliquity of the Nubia (Africa) – Eurasia convergence.

    Beschreibung: Shear wave splitting is measured at 19 seismic stations of a temporary network deployed in the Val d’Agri area to record low-magnitude seismic activity. The splitting results suggest the presence of an anisotropic layer between the surface and 15 km depth (i.e. above the hypocentres). The dominant fast polarization direction strikes NW–SE parallel to the Apennines orogen and is approximately parallel to the maximum horizontal stress in the region, as well as to major normal faults bordering the Val d’Agri basin. The size of the normalized delay times in the study region is about 0.01 s km−1, suggesting 4.5 per cent shear wave velocity anisotropy (SWVA). On the south-western flank of the basin, where most of the seismicity occurs, we found larger values of normalized delay times, between 0.017 and 0.02 s km−1. These high values suggest a 10 per cent of SWVA. These parameters agree with an interpretation of seismic anisotropy in terms of the Extensive-Dilatancy Anisotropy (EDA) model that considers the rock volume pervaded by fluid-saturated microcracks aligned by the active stress field. Anisotropic parameters are consistent with borehole image logs from deep exploration wells in the Val d’Agri oil field that detect pervasive fluid saturated microcracks striking NW–SE parallel to the maximum horizontal stress in the carbonatic reservoir. However, we cannot rule out the contribution of aligned macroscopic fractures because the main Quaternary normal faults are parallel to the maximum horizontal stress. The strong anisotropy and the seismicity concentration testify for active deformation along the SW flank of the basin.

    Beschreibung: We analyze the seismicity of a small sector of the Northern Apennines merging data from the Italian seismic bulletin with original data collected by temporary seismic networks. Our attention is focused on the region enclosed between the Apenninic watershed and the Adriatic Sea. This portion of belt is interested by the occurrence of diffuse crustal seismicity and small-to-moderate earthquakes. In this paper we study the five small sequences with mainshock having Mw 〈 4.7 that in the past 15 years hit the area. Our interest is addressed to better understand the relationship between these events and the regional seismotectonic setting in terms of seismicity distribution and stress field. Two regions with different behavior in the seismic release can be distinguished: (i) along the watershed where seismicity is clustered at shallow depths (〈 15 km) and where strong earthquakes occurred in the past, (ii) an eastern portion where the seismicity is distributed across all of the crustal volume, locally reaching depths down to 30 km. The focal mechanism of the seismic sequences shows mainly normal fault kinematics coherent with the regional stress field. Detailed stress field analysis suggests a rotation of the principal stress axis moving from the axial part of the chain toward the Adriatic Sea to the east.

    Beschreibung: In this paper we present and discuss an improved picture of the seismicity distribution of the Umbria– Marche–Abruzzi Apennines as obtained through the integration of the national and the regional seismic networks operating from 2002 to 2006. During this period, both the Istituto Nazionale di Geofisica e Vulcanologia (INGV) National Seismic Network and the regional networks have been greatly improved. We compare the results of the integrated catalogue obtained in this study with the Catalogue of the Italian Seismicity between 1981 and 2001 [Castello, B., Selvaggi, G., Chiarabba, C., Amato, A., 2006. CSI Catalogo della sismicità italiana 1981–2002, versione 1.1. INGV-CNT, Roma.http://legacy.ingv.it/CSI )], confirming the basic known features of the seismic activity in the region, but also evidencing some original and interesting results. In particular, the new data set allows us to better define the geometry and kinematics of the crustal seismicity, which is confined to the upper 20 km and shows a clear general deepening from west to east. In the crust, we find additional evidence of extensional seismicity below the central portion of the belt and thrust/reverse faulting mechanisms at the outer fronts of the Apennines. Looking at the seismicity along the belt, it is also possible to observe aseismic regions, which could be due to either locked or creeping portions of the Apenninic fault system. At greater depth, the west-dipping seismicity distribution down to about 70 km confirms the hypothesis of a slab of Adriatic lithosphere subducted below the Apennines, but also suggests that there are strong lateral heterogeneities and possibly tears in the slab.

    Beschreibung: The first comprehensive geochemical data-set of the fluids circulating over a 14,000 km2-wide seismicprone area of the Southern Apennines, Calabria Region (Italy), is presented here. The geochemical investigations were carried out with the twofold aim of constraining the origin and interactions of the circulating fluids and to investigate possible relationships with local faults. Sixty samples of both thermal and cold waters were collected, from which the dissolved gases were extracted. The geochemical features of the water samples display different types and degrees of water–rock interactions, irrespective of the outlet temperature. The calculated equilibrium temperatures of the thermal waters (60–160 C) and the low heat flow of thewhole study area, are consistent with a heating process due to deep water circulation and rapid upflow through lithospheric structures. The composition of the dissolved gases reveals that crustal-originating gases (N2 and CO2-dominated) feed all the groundwaters. The 3He/4He ratios of the dissolved He, in the range of 0.03–0.22Rac for the thermal waters and 0.05–0.63Rac for the cold waters (Rac = He isotope ratio corrected for atmospheric contamination), are mainly the result of a two-component (radiogenic and atmospheric) mixing, although indications of mantle-derived He are found in some cold waters. As the study area had been hit by 18 of the most destructive earthquakes (magnitude ranging from 5.9 to 7.2) occurring over a 280-a time span (1626–1908) in the Southern Apennines, the reported results on the circulating fluids may represent the reference for a better inside knowledge of the fault-fluid relationships and for the development of long-term geochemical monitoring strategies for the area.

    Beschreibung: The goal of this study was to estimate the stress field acting in the Irpinia Region, an area of southern Italy that has been struck in the past by destructive earthquakes and that is now characterized by low to moderate seismicity. The dataset are records of 2,352 aftershocks following the last strong event: the 23 November 1980 earthquake (M 6.9). The earthquakes were recorded at seven seismic stations, on average, and have been located using a three-dimensional (3D) P-wave velocity model and a probabilistic, non-linear, global search technique. The use of a 3D velocity model yielded amore stable estimation of take-off angles, a crucial parameter for focal mechanism computation. The earthquake focal mechanisms were computed from the P-wave first-motion polarity data using the FPFIT algorithm. Fault plane solutions show mostly normal component faulting (pure normal fault and normal fault with a strikeslip component). Only some fault plane solutions show strike-slip and reverse faulting. The stress field is estimated using the method proposed by Michael (J Geophys Res 92:357–368, 1987a) by inverting selected focal mechanisms, and the results show that the Irpinia Region is subjected to a NE–SW extension with horizontal σ3 (plunge 0◦, trend 230◦) and subvertical σ1 (plunge 80◦, trend 320◦), in agreement with the results derived from other stress indicators.

    Beschreibung: The Miano borehole, 1047 m deep, is located close to the river Parma in the Northern Apennines, Italy. A measuring station has been installed to observe the discharge of fluids continuously since November 2004. The upwelling fluid of this artesian well is a mixture of thermal water and CH4 as main components. In non-seismogenic areas, a relatively constant fluid emission would be expected, perhaps overlaid with long term variations from that kind of deep reservoir over time. However, the continuous record of the fluid emission, in particular the water discharge, the gas flow rate and the water temperature, show periods of stable values interrupted by anomalous periods of fluctuations in the recorded parameters. The anomalous variations of these parameters are of low amplitude in comparison to the total values but significant in their long-term trend. Meteorological effects due to rain and barometric pressure were not detected in recorded data probably due to reservoir depth and relatively high reservoir overpressure. Influences due to the ambient temperature after the discharge were evaluated by statistical analysis. Our results suggest that recorded changes in fluid emission parameters can be interpreted as a mixing process of different fluid components at depth by variations in pore pressure as a result of seismogenic stress variation. Local seismicity was analyzed in comparison to the fluid physico-chemical data. The analysis supports the idea that an influence on fluid transport conditions due to geodynamic processes exists. Water temperature data show frequent anomalies probably connected with possible precursory phenomena of local seismic events.

    Beschreibung: The rate of decomposition of the three phenolics, p-hydroxybenzoic acid, protocatechuic acid and gallic acid, whose difference is the amount of OH-groups, was investigated in the soil. With the increase in OH-groups increases the rate of microbial decomposition. Gallic acid decomposes faster than protocatecuic acid and this again faster than p-hydroxybenzoic acid.

    Beschreibung: In April 1991, submarine volcanic eruptions initiated the formation of numerous hydrothermal vents between 9°45′ and 9°52′N along the crest of the East Pacific Rise (EPR). Dramatic changes in biological community structure and vent fluid chemistry have been documented throughout this region since the eruptive event. By April 2004, mussels (Bathymodiolus thermophilus) dominated the faunal assemblages at several of the vent sites formed during of after the 1991 eruptions, whereas other habitats within the region were dominated by the vestimentiferan Riftia pachyptila. In the present paper, we build upon the extensive data sets obtained at these sites over the past decade and describe a manipulative experiment (conducted at 9°49.94′N; 104°14.43′W on the EPR) designed to assess interrelationships between vent fluid chemistry, temperature, biological community structure, and seismic activity. To this end, in situ voltammetric systems and thermal probes were used to measure H2S/HS− and temperature over time in a denuded region of an extensive mussel bed in which an exclusion cage was placed to inhibit the subsequent migration of mussels into the denuded area. Fluid samples were taken from the same locations to characterize the associated microbial constituents. Basalt blocks, which were placed in the cage in April 2004 and subsequently recovered in April 2005, were colonized by more than 25 different species of invertebrates, including numerous vestimentiferans and remarkably few mussels. Recorded temporal changes in vent fluid chemistry and temperature regimes, when coupled with microbiological characterization of the vent fluids and seismic activity data obtained from ocean bottom seismometers, shed considerable light on factors controlling biological community structure in these hydrothermal ecosystems.

    Beschreibung: New high-resolution bathymetric and magnetic data from the western Aeolian sector, southern Tyrrhenian Sea, provide insights into structural and volcanic development of the area, suggesting a strong interaction between volcanism and tectonics. The analysis of these data combined with relocated earthquake distribution, focal plane solutions and strain rate evaluation indicates that the dextral strike-slip Sisifo-Alicudi shear zone is a complex and wide area of active deformation, representing the superficial expression of the deep seated lithospheric tear fault separating the subduction slab below Sicily and Calabria. Most of the observed volcanic features are aligned along a NW–SE trend, such as the Filicudi island-Alicudi North Seamount and Eolo-Enarete alignments, and are dissected by hundred-metre-high scarps along conjugate NNE–SSW trending fault systems. The magnetic field pattern matches the main trends of volcanic features. Spectral analysis and Euler deconvolution of magnetic anomalies show the existence of both deep and shallow sources. High-amplitude, high-frequency anomalies due to shallow sources are dominant close to the volcanic edifices of Alicudi and Filicudi, while the main contribution on the surrounding Eolo, Enarete, Alicudi North and Filicudi North seamounts is given by low-amplitude anomalies and/or deeper magnetic sources. This is probably related to different ages of the volcanic rocks, although hydrothermal processes may have played an important role in blanketing magnetic anomalies, in particular at Enarete and Eolo seamounts. Relative chronology of the eruptive centres and the inferred deformation pattern outline the Quaternary evolution of the western Aeolian Arc: Sisifo, Alicudi North and Filicudi North seamounts might have developed in an early stage, following the Late Pliocene–Early Pleistocene SE-ward migration of arc-related volcanism due to the Ionian subduction hinge retreat; Eolo, Enarete and Filicudi represent later manifestations that led volcanoes to develop duringMid-Late Pleistocene, when the stress regime in the area changed, due to the SSE-ward propagation of the subduction slab tear fault and the consequent reorientation and decrease of trench migration velocity. Finally, volcanic activity occurred in a very short time span at Alicudi, where an almost conical volcanic edifice emerged, suggesting negligible interactions with regional fault systems.

    Notizen: Abstract In the Long Valley caldera, where seismicity has continued essentially uninterrupted since mid-1980 and uplift is documented, samples of water from hot, warm, and cold springs have been collected since September, 1982, and their222Rn concentrations analyzed. Concurrently, rocks encompassing the hydrologic systems feeding the springs were analyzed for their radioelement contents, because their uranium is the ultimate source of the222Rn in the water. The222Rn concentration in the springs varies inversely with their temperature and specific conductance. High concentrations (1500 to 2500 picocuries per liter) occur in dilute cold springs on the margins of the caldera, while low contents (12 to 25 pCi/l) occur in hot to boiling springs. Springwater radon concentrations also correlate slightly with the uranium content of the encompassing rocks. A continuous monitoring system was installed in August, 1983, at a spring issuing from basalt, to provide hourly records of radon concentration. A gamma detector is submerged in a natural pool, and we have observed that the radioactivity measured in this manner is due almost entirely to the222Rn concentration of the water. Initial operation shows diurnal and semidiurnal variations in the222Rn concentration of the springwater that are ascribed to earth tides, suggesting that those variations are responding to small changes in stress in the rocks encompassing the hydrologic system.

    Notizen: The DNA-ligand interactions of a series of phenanthridinium compounds with various combinations of amino, azido, and hydrogen functions at R3 and R8 were examined to determine the contribution of these particular substituents to ligand binding. Spectrophometric titrations using calf-thymus DNA emphasized the importance of amino substituents in conferring a strong interaction and also stabilizing the interaction against reversal by high ionic strength. Although azido groups were not as effective as amino groups, they were more effective than hydrogen functions in enhancing the interaction. Furthermore, an amino substitution at R8 was consistently, though only slightly, more effective than an amino substituent at R3. The results from superhelical titrations using plasmid pBR322 DNA demonstrated that analogs with amino and/or azido functions at both R3 and R8 produced the greatest unwinding, and compounds with an amino or an azido function at R8 proved more effective than those with the corresponding amino or azido substituent at R3.

    Notizen: The magnetic shielding constant of the 31P nucleus of the dimethylphosphate anion is calculated by an ab initio method for different values of the torsion angles about the PO ester bond and different orientations of the methyl groups. The results obtained tend to show that both types of conformational parameters contribute to the value of σ31P. The largest shielding is obtained when the methyl groups are staggered with respect to the PO bond; the smallest, for the eclipsed arrangement. Measurements carried out on the 16 deoxyribodinucleoside monophosphates show that in the majority of cases σ31P is shifted toward lower field for the dimers having large values of 3JPH3′ and 3JPH5′(5″). The theoretical results are discussed in relation to experimental data for polynucleotides and nucleic acids.

    Notizen: The optical modes of DNA that displace one strand against the other are modeled by the motion of an oscillator in an asymmetric quartic double-well potential whose minima represent the A- and B-conformations. Assuming that the variation of the polarizability during vibration derives mainly from the tilting of the base rings relative to the helix axis, the total polarizability tensor is shown to possess approximately ellipsoidal symmetry and to depend nonlinearly on the instantaneous displacement of the two strands. The Raman spectrum of a collection of randomly oriented molecules is calculated. It consists of one or more peaks with characteristic shape. The depolarization ratio is 3/4, independent of molecular conformation and frequency. The results are discussed in the light of existing experimental and theoretical information.

    Notizen: Using the semiempirical potential functions, conformational energies of the model compounds DMP-, d(pCp), d(pGp), and d(pCpGpCp) are calculated, and the B → Z transition is discussed along the pseudorotational path of the sugar ring. For dimethylmonophosphate anion, DMP-, the energy contour map is presented and the stabilities of the phosphodiester conformations discussed. For the sugar ring without the base attached, the minimum energies for each sugar-puckering form are calculated along the pseudorotational path. The energy barrier of the interconversion between the C(3′)-endo form and the C(2′)-endo form is calculated to be about 2.0 kcal/mol. From the conformational energy calculations of the interconversions of mononucleoside diphosphates, d(pCp) and d(pGp), between the C(2′)-endo conformer and the C(3′)-endo conformer, the purine sugar segment is known to be more convertible than the pyrimidine sugar segment. The results also support the finding that the pseudorotational transition occurred with the O(1′)-endo form more easily than with the O(1′)-exo form. Based on the results of conformational studies of DMP-, d(pCp), and d(pGp), a topological transition of the handedness of the model compound, d(pCpGpCp), is studied. The left-handed Z-form is found to be less stable by about 8.5 kcal/mol than is the right-handed B-form. The energy barrier of the Z → B transition is calculated to be about 17.4 kcal/mol. The contributions of the electrostatic and nonbonded energies to the energy barrier are discussed in connection with the conformation changes of the model compound, d(pCpGpCp).

    Notizen: The dielectric properties of aqueous solutions of DNA were measured at frequencies ranging from 0.1 to 12 GHz. The results are analyzed using the Maxwell mixture theory and yield a value for the hydration of the DNA of about 0.4 g/g, which is in the range observed in other investigations. No evidence was found for an additional absorption effect at microwave frequencies, which has been predicted to occur in certain DNA analogs due to the vibrational excitation of the double helix by the applied microwave field.

    Notizen: Self-association of substance P and its C-terminal partial peptide sequences was studied by CD, quasi-electric light scattering, and sedimentation experiments. CD spectra of these peptides are strongly influenced by self-association. They exhibit strong characteristic negative ellipticities, suggesting the formation of a presumably B-type ordered structure. The tendency to form multimers depends on chain length and constitution and has its maximum at the octapeptide (SP 8). The peptide multimers have a broad distribution of sizes in the range of 30- and 800-nm diameter. Subdivision of this distribution into two size classes gives mean diameters of 60-100 nm (predominating)/200-800 nm for substance P and 30-50 nm/200-800 nm for SP 8 multimers.

    Notizen: 13C-nmr chemical shifts of backbone carbonyl and side-chain β-carbons in polypeptides provide structural information. Recent utilization of substituent effects on 13C-nmr chemical shifts (principally γ-effects) has permitted the rationalization of their sequence and conformation dependence when observed in linear, flexible polypeptides. In this report, we apply the γ-effect method to study the 13C-nmr chemical shifts observed in solution and in the solid state for the backbone carbonyl and side-chain β-carbons in conformationally rigid polypeptides, which are usually cyclic. As found previously for flexible, linear polypetides, the relative 13C-nmr chemical shifts observed for the backbone carbonyl and side-chain β-carbons in conformationally rigid polypeptides are predictable from knowledge of their peptide residue sequence (primary structure) and conformation (secondary structure) via the γ-effect method.

    Notizen: The CD spectra of the peptides Boc-X-(Aib-X)n-OMe (n = 1, 2, 3) and Boc-(Aib-X)5-OMe, where X = L-Ala or L-Val have been examined in several solvents. The X = Ala and Val peptides behave similarly in all solvents, suggesting that the Aib residues dominate the folding preferences of these peptides. The decapeptides adopt helical conformations in methanol and trifluoroethanol, with characteristic negative CD bands at 222 and 205 nm. In the heptapeptides, similar spectra with reduced intensities are observed. Comparison with nmr studies suggest that estimates of helical content in oligopeptides by CD methods may lead to erroneous conclusions. The pentapeptides yield solvent-dependent spectra indicative of conformational perturbations. Peptide association in dioxane results in an unusual spectrum with a single negative band at 210 nm for the decapeptides. Disaggregation is induced by the addition of methanol or water to dioxane solutions. Aggregation of the heptapeptides is less pronounced in dioxane, suggesting that a critical helix length may be necessary to promote association stabilized by helix dipole-dipole interactions.

    Notizen: The interaction of poly-5-bromouridylic acid [poly(BU)] with adenosine and 9-methyladenine was studied by equilibrium dialysis, optical melting, and microcalorimetry. The stacking free energy, ω, was estimated as -17.6 kJ/mol for adenosine·2poly(BU) and -18.8 kJ/mol for 9-methyladenine·2poly(BU) from the binding isotherms constructed from equilibrium dialysis results. The binding isotherms constructed from a series of melting curves also gave ω values for adenosine·2poly(BU). The thermal stability of the complex depends on monomer concentration, and the partial molar enthalpies of the complex formation at the midpoint of the transition were evaluated from the Tm coefficients as a function of free monomer concentration. The values of -92.0 and -90.4 kJ/mol were obtained for adenosine·2poly(BU) and 9-methyladenine·2poly(BU) in 0.4M NaCl-0.02M Na-cacodylate-5 × 10-4M EDTA (pH 7.0), respectively. Microcalorimetric measurements provided lower integral heats of reaction values for these complexes, i.e., -73.2 kJ/mol for adenosine·2poly(BU) and -71.5 kJ/mol for 9-methyladenine·2poly(BU). A comparison with a polyribouridylic acid system provided a quantitative understanding of a stabilization by bromination in terms of thermodynamic parameters.

    Notizen: A molecular-dynamics simulation was used to carry out an introductory study of the hydration of a section of a rigid single A- or B-DNA helix with one Na+ counterion per nucleotide. Four Na+ ions and four nucleotides and periodic boundary conditions were used to mimic an infinite helix. The atoms of the helix and the Na+ ions were assumed to be Lennard-Jones spheres that also carried charges. Stillinger four-point charge model water molecules were used. We carried out five calculations, for 26 and 46 water molecules in B-DNA and 20, 32, and 46 in A-DNA fragments. The arrangements of the Na+ ions are found to have some similarities to those obtained by Clementi and Corongiu. In the calculations with 46 water molecules, we found that two Na+ ions can be bridged by about two water molecules and form a hydrated bound pair, which in turn forms a bridge between the guanine N7 and a near phosphate group. These bound pairs may be important in stabilizing the helix structure of DNA molecules.

    Notizen: Ethylurea was used to weaken hydrophobic interactions during collagen fibrillogenesis in vitro. Intact and enzyme-digested type I collagen was studied. In all preparations, ethylurea decreased the extent and rate of fibril formation, inhibition being greatest in the enzyme-digested collagens. With intact collagen (and probably also with carboxypeptidasedigested collagen), there was no evidence the ethylurea altered the mechanism of fibril growth; in pepsin-digested collagen, however, the growth mechanism was altered by ethylurea, possibly reflecting a conformational change of the “hydrophobic cluster” in the C-terminal peptide. Such a structural change could occur in a hydrophobic environment once the distal portion of the C-terminal peptide (presumed to be essential for its structural stability) is removed by pepsin. The results further emphasize the importance of hydrophobic interactions in collagen fibril nucleation and growth in vitro.

    Notizen: We report the crystal-structure analysis of a complex involving the drug proflavine and the two dinucleoside monophosphates cytidylyl-3′,5′-adenosine (CpA) and uridylyl-3′,5′-guanosine (UpG). The planar drug molecule is intercalated between C⃛G and U⃛A Watson-Crick base pairs, in a double-helical fragmentlike arrangement. Sugar conformations at the 3′-ends of the two strands are dissimilar. The backbone conformations fall within the ranges of values noted previously for dinucleoside intercalation complexes, and some correlations involving these are noted. The separation of the two strands and the basic twist angle of 16°, compared to other reported complexes, are indicative of sequence-dependent effects of the drug binding.

    Notizen: Helical hydrophobic moment ratios, 〈h2〉/〈H2〉, have been evaluated for 34 polypeptides under conditions where the helix content is dictated solely by the short-range interactions operative in aqueous media. The mean-square helical hydrophobic moment is denoted by 〈h2〉, and 〈H2〉 is the averaged of the squared hydrophobicites. This ratio would be one in absence of any correlation in the hydrophobicities of amino acid residues in helices. The 〈h2〉/〈H2〉 tend to be substantially larger than values of the analogous ratio formulated for the mean-square dipole moments of typical synthetic polymers. For 24 of the 34 polypeptide chains considered, 〈h2〉/〈H2〉 is found to be greater than one, indicating a tendency to form helices with amphiphilic character. The ratio is exceptionally large in the case of the δ-hemolysins. It is also large for two other surface-active peptides, for two of the four apolipoproteins examined, and for myohemerythrin. A much smaller 〈h2〉/〈H2〉 is found for melittins. If melittins is to form helices with large 〈h2〉/〈H2〉, the configurational statistics must be governed by effects in addition to those short-range interactions that occur when water is the solvent.

    Notizen: Conformational-energy computations have been carried out for the N-acetyl-N′-methylamides of the Pro-Pro, Pro-Gly, and Gly-Pro dipeptides and of the Pro-Pro-Gly-Pro tetrapeptide, serving as models for the conformational analysis of single-stranded poly(Gly-Pro-Pro). The probability of β-bend formation for the Pro-Gly sequence is very high, viz., 0.72 for the terminally blocked Pro-Gly dipeptide, and rises to 0.86 in the tetrapeptide. The β-bend conformations of the Pro-Gly sequence are of low energy in single-chain poly(Gly-Pro-Pro) as well. The β-bend structure had been postulated earlier to be a requirement for post-translational proline hydroxylation during the biosynthesis of collagen. The present results lend strong support to this proposal by demonstrating that the β-bend structure is energetically favorable and hence can be accommodated easily in single-stranded poly(Gly-Pro-Pro).

    Notizen: The interaction of Cu(II) with the protamine scylliorhinine Z3 was studied by means of CD measurements. At a 1:1 molar ratio, three complexes are formed. (1) In the pH range 5-6.5, the results suggest the formation of a five-membered chelate ring through the coordination of two nitrogen atoms, the N-terminal and the contiguous peptide nitrogen. (2) At pH ≥ 6.4, there is involvement of the lateral NH2 group of Arg; at pH 6.5-8, the formation of a 3N cupric complex is strongly suggested. (3) At pH ≥ 8, results indicate the formation of a 4N complex as a major species in Cu(II)-Z3 solution. The transformation from a 2N to a 3N complex, and from a 3N to a 4N complex was followed with the help of the σ(αNH2) → Cu(II) charge-transfer dichroic band transitions. At Cu(II):Z3 molar ratios ≥ 2 and at pH 〉 8, a new dichroic band appears, indicating the involvement of the tyrosine residue side chain in metal-ion complexation.

    Notizen: Studies of the optical birefringence of solutions of acid-soluble collagen from rat-tail tendon at 22°C in the pH range 1.0-6.0 show that collagen exhibits an isotropic to mesophase transition only between pH 2.4 and 3.0 at 10% weight concentration. Such liquid crystalline order is probably essential for the orientation of collagen in a magnetic field. When solutions of neutral salt-soluble collagen were precipitated at pH 7.0 by warming to 37°C (“heat gelling”) in a magnetic field of ca. 20 kG, the resulting fibrils wee oriented perpendicular to the direction of the field. Heat gelling is shown to be a useful technique for maintaining the orientation induced in precursor solutions even after the sample is removed from the magnetic field.

    Notizen: Polyamines are ubiquitous cellular components that interacts strongly with nucleic acids. Although many of the interactions of oligocations with DNA can be rationalized with polyelectrolyte theories that treat counterions as point charges, some structural effects are evident. We have explored the effects of polyamine structure on one important aspect of DNA behavior, its thermal melting transition, by using a series of spermidine analogs NH3(CH2)3NH2(CH2)nNH33+, where n varies from 2 to 8 [Jorstad et al. (1980) J. Bacteriol. 141, 456-463]. For spermidine itself, n = 4. Tm for calf-thymus DNA in the presence of each of these analogs, and the other naturally occurring polyamines putrescine2+ and spermine4+, was measured over a wide range of NaCl concentrations and polyamine:DNA phosphate ratios. There are modest, but significant structural effects. particularly with the shorter n = 2 and 3 derivatives, whose geometry my not allow full electrostatic interaction with DNA. Longer analogs, on the other hand, are not much different than spermidine in their effects on Tm, though a moderate maximum occurs at n = 5. Since polyamines are important in the cellular condensation and packaging of DNA, we have also delineated the critical polyamine and salt concentrations that are required to cause DNA aggregation. Here again, there are significant structural effects, which are not easily rationalized by any simple considerations.

    Notizen: The observed difference between the CD spectrum of B-DNA mononucleosomes and that of free DNA has been attributed to the tertiary structure of the core DNA. This conjecture is tested here. The tertiary structure is modeled as an optical system consisting of two identical, planar, linear retarders rotated with respect to each other. The retarders have the same linear birefringence and linear dichroism as oriented B-DNA. Such an optical system is circularly dichroic. The predicted wavelength dependence and magnitude of the CD are in reasonably close agreement with the experimental results.

    Notizen: The absorption and CD spectra of the complexes of poly(L-arginine) (PLA) and azo dyes have been measured in aqueous solution. On complexation, Blue-shifted additional absorption bands were observed. In the wide pH 4-11 range, induced CD was observed at the visible wavelengths corresponding to the blue-shifted absorption bands. The induced CD arose from the dimeric dye molecules bound to PLA in the α-helical structure. When a modified analysis of induced CE is made by the excition chirality method, the origin of the induced CD can be assigned to the dipole coupling. The PLA-dye complexes showed the counterlockwise (negative, S) chirality of the transition dipole moments of dyes.

    Notizen: Ionization properties of the tyrosyl groups of bovine plasma albumin in various conformational states - the native state (N), the two acid states (F and E), and the state (B) stable at slightly alkaline pH - were studied by means of a stopped-flow-pH-jump technique. The technique allows us to obtain the tyrosyl titration curve in a conformational state that is unstable in the pH region of the titration. The pH jumps from the N and B states to various alkaline pH's, where the tyrosines ionize to bring about a time-dependent increase in absorption at 296 nm, indicating that a number of the tyrosines buried initially become susceptible to ionization as a result of the alkaline transition occurring above pH 10.8. Extrapolation of the observed absorption change to zero time gives a spectrophotometric titration curve in the initial conformational state. Only 30-401% of the 19 tyrosines of the protein can ionize both in the N and the B state at pH 12. The pH jumps from the F and E states, on the other hand, give a decrease in absorption between pH 9 and 11.7, indicating that the tyrosyl groups initially exposed are remarked by refolding after the pH jumps, and the zero-time titration curves show that essentially all the tyrosyl groups ionize normally in these acid states. The results are discussed in relation to the known results of the tyrosyl exposure of the protein measured by other techniques, and the consistency among them demonstrates the effectiveness of the pH-jump titration method. Hydrogen bonding between the abnormal tyrosyl and carboxylate groups as a mechanism to stabilize native albumin is suggested from characteristics of the alkaline transition, which also involves the exposure of the tyrosyl groups, and from the tyrosyl titration curves in the native and acid states.

    Notizen: The dielectric properties of sonicated calf-thymus DNA sodium salt in aqueous solutions have been studied in the frequency range from 40 MHz to 2 GHz by time domain spectroscopy (TDS). A dielectric dispersion not previously reported was found, which has a characteristic frequency of about 150 MHz. All of the dielectric parameters are insensitive to the size of DNA fragments and to helix-to-coil transitions. The study of this dispersion as a function of DNA concentration and temperature allows us to conclude that it may be due to counterion fluctuation on short sections, probably in a direction transverse to the macromolecular axis.

    Notizen: The oxidation of L-adrenaline (epinephrine) in the presence of [Fe(tetpy)(OH)2]+ ions bound to poly(L-glutamate) or to poly(D-glutamate) has been studied at pH 7 (tetpy = 2,2′:6′,2″:6″,2‴-tetrapyridyl). Electron transfer from the substrate to the central metal ion, which is rate-determining, proceeds stereoselectively only when extensive and possibly specific interactions between adrenaline and the peptidic residues of the ordered polymer in the close environment of the active sites occur. This ensures different steric constraints for the two diastereomeric precursor complexes, which are thought to affect the separation and orientation of the redox centers differently, leading to the observed phenomena. Some data on the catalytic oxidation of L-dopa(3,4-dihydroxyphenylalanine) are also presented, showing stereoselective effects similar to those observed with L-adrenaline, despite the diverse distance of the chiral center from the reacting OH groups. A mechanistic interpretation of the results is discussed in the light of a few general considerations concerning the structural features of the catalytic systems. Possible explanations for the finding that stereoselectivity occurs at the expense of the efficiency of catalysis are also considered.

    Notizen: Solution-phase Raman spectra of diastereomeric alanyl dipeptides, D-Ala-L-Ala and L-Ala-L-Ala, and various mono- and dideuterated isotopomers in H2O and D2O, are reported. Spectral differences between the diastereomeric forms are interpreted, using the Raman analog of the coupled oscillator model, in terms of geometric differences between certain vibrations in the diastereomeric forms. Application of the coupled-oscillator formalism allows the determination of a dihedral angle between the coupling vibrations. The results are compared with vibrational coupling employed by other workers in the determination of the vibrational spectra of peptides.

    Notizen: The conformational equilibria of five β-cellobiose conformers have been studied theoretically in 10 solvents. The stability of the conformers in dilute solution has been compared by using the method that has already been tested for 2-methoxytetrahydropyran, β-maltose, and D-glucose. The solvation energy consists of electrostatic, dispersion, and cavity terms which have been determined from the properties of the solute calculated by the PCILO quantum-chemical method and physicochemical properties of the solvents. The calculated abundance of conformers depends on the solvent (e.g., in dioxane C1:C2:C3:C4:C5 = 60.0:34.1:2.9:2.0:1.0; in dimethylsulfoxide, 75.5:22.1:1.8:0.5:0.2; and in water, 82.2:16.2:1.3:0.2:0.1). The results obtained indicate that the preponderant conformer in the aqueous solution is similar to the one adopted by β-cellobiose in the crystalline form. The role of individual contributions to the solvation energy have been analyzed. Based on the determined abundance of conformers, averaged residual optical activity and nmr parameters have been calculated and compared with observable properties. The marked differences observed between solvent-induced conformational changes for β-cellobiose and β-maltose have been discussed from the viewpoint of the solubility of the cellulose.

    Notizen: With a view to understanding the role of hydrogen bonds in the recognition of nucleic acids by proteins, hydrogen bonding between the bases and base pairs of nucleic acids and the amino acids (Asn, Gln, Asp and Glu, and charged residues Arg+, Glu-, and Asp-) has been studied by a second-order perturbation theory. Binding energies have been calculated for all possible configurations involving a pair of hydrogen bonds between the base (or base pair) and the amino acid residue. Our results show that the hydrogen bonding in these cases has a large contribution from electrostatic interaction. In general, the charged amino acids, compared to the uncharged ones, form more stable complexes with bases or base pairs. The hydrogen-bond energies are an order of magnitude smaller than the Coulombic interaction energies between basic amino acids (Lys+, Arg+, and His+) and the phosphate groups of nucleic acids. The stabilities of the complexes of amino acids Asn, Gln, Asp, and Glu with bases are in the order: G-X 〉 C-X 〉 A-X U-X or T-X, and G · C-X 〉 A · T(U)-X, where X is one of these amino acid residues. It has been shown that Glu- and Asp- can recognize guanine in single-stranded nucleic acids; Arg+ can recognize G · C base pairs from A · T base pairs in double-stranded structures.

    Notizen: The effect of histone H1 on the conformation and stability of control and acetylated HeLa high-molecular-weight chromatin that had been stripped of H1 and nonhistone proteins was compared by circular dichroism (CD) and thermal denaturation measurements. Two different preparations of H1, originating from rat thymus and chicken erythrocyte, were used in the reconstitution studies. The control and acetylated stripped chromatin had identical CD and thermal denaturation properties, as did their reconstitutes with rat thymus H1. Reconstitutes of the two chromatins with chicken erythrocyte H1 had similar CD properties, but thermal denaturation studies showed that the acetylated reconstitute was destabilized compared to the control reconstitute. Reconstitutes of both chromatins with chicken erythrocyte H1 had a more condensed and stabilized structure than the reconstitutes with rat thymus H1. Thus, acetylation caused a decrease in the stability of chromatin in the presence of erythrocyte H1, but more marked differences were detected in the structure of stripped chromatin after reassociation with different H1 preparations.

    Notizen: Human fibrinogen was treated with thrombin in the presence of fibrinoligase (Factor XIIIa) and calcium ion at pH 8.5, ionic strength 0.45, and the ensuing polymerization was interrupted at various time intervals (t) both before and after the clotting time (tc) by solubilization with a solution of sodium dodecyl sulfate and urea. Aliquots of the solubilized protein were subjected to gel electrophoresis on polyacrylamide gels after disulfide reduction by dithiothreitol and on agarose gels without reduction. The degree of γ-γ ligation was determined from the former. The latter provided the size distribution of ligated end-to-end sequences produced by splitting the ligated staggered overlapped oligomers down the middle, for degrees of polymerization, x, from 1 to 10. Addition of fibrinoligase (in which the activating thrombin had been inhibited by p-nitrophenyl-p′-guanidinobenzoate, NPGB) to Kabi fibrinogen showed the presence of small amounts of ligatable oligomers. Addition of fibrinoligase to a polymerizing mixture in which the action of thrombin had been stopped before clotting by NPGB produced the same distribution of ligated end-to-end sequences that was obtained when fibrinoligase was originally present, at least for reaction times up to 0.7 of the clotting time. The kinetics of γ-γ ligation by fibrinoligase acting on a polymerized mixture stabilized by NPGB were followed. The reaction was first order in the concentration of ligatable γ-γ junctions and the initial velocity was proportional to the enzyme concentration. The time evolution of size distribution of ligated end-to-end sequences agreed with a theory based on random ligation of ligatable junctions.

    Notizen: A mean-square helical hydrophobic moment, 〈h2〉, is defined for polypeptides in analogy to the mean-square dipole moment, 〈μ2〉, for polymer chains. For a freely jointed polymer chain, 〈μ2〉 is given by Σmi2, where mi denotes the dipole moment associated with bond i. In the absence of any correlations in the hydrophobic moments of individual amino acid residues in the helix, 〈h2〉 is specified by ΣHi2, where Hi denotes the hydrophobicity of residue i. The tendency for correlations in orientations of residue hydrophobic moments in helices therefore dictates the size of 〈h2〉/〈H2〉, where 〈H2〉 denotes the average value of ΣHi2 for all helices. The value of 〈h2〉/〈H2〉 will be greater than one in amphiphilic helices. A necessary prerequisite for this diagnostic usage of 〈h2〉/〈H2〉 is that the residue hydrophobic moment be oriented prependicular to the principal axis of the helix. Matrix-generation schemes are formulated that permit rapid evaluation of 〈h2〉 and 〈H2〉. The behavior of 〈h2〉/〈H2〉 is illustrated by calculations performed for model sequential copolypeptides.

    Notizen: We report a calculation of the distribution of small ions around a charged cylinder representing a polyelectrolyte molecule in solution. The Monte Carlo method of Metropolis, Rosenbluth, and Teller was used to avoid the inaccuracies known to be associated with the Poisson-Boltzmann equation. The systems examined contained a long polyelectrolyte cylinder with charge parameter, χ, equal to 4.2, corresponding approximately to a DNA molecule. In one model, the cylinder had charges on its axis and an exclusion radius to the center of the small ions equal to 10 Å, while the small ions had various radii in the range from 1 to 10 Å and one or two protonic charges. Various systems were studied; some had one species of small ion alone, others had mixtures of different types. The results showed good agreement with the solution of the Poisson-Boltzmann equation when only the species with 1-Å radius was present, but considerable discrepancies appeared with larger ions as a result of excluded volume interactions between the latter. Deviations from the Poisson-Boltzmann equation also appeared when both positive and negative small ions were present; the deviations were in the direction of a higher concentration of both counter- and co-ions, but particularly co-ions, close to the polyelectrolyte. In another model, the charges were arranged along two helices on the surface of the cylinder; the resulting radial distribution of small ions was not much different from that found when the charges were situated on the axis. In all cases there was a striking accumulation of counterions in a layer of concentration exceeding 1 mol/L at the surface of the polyion.

    Notizen: Brillouin scattering was used to determine the longitudinal acoustic velocity along the axis of collagen fibers as the relative humidity decreased from 95 to 0%. Between 0 and 86%, the velocity decreased linearly at a modest rate. Above 86%, the decrease was steep. The changing mass of the absorbed water accounts for most of the velocity change in the lower humidity range. The remaining deviation is probably due to variation of the elastic coupling between molecules. At low humidity, the adsorbed water is known to be attached to the collagen molecules, whereas the additionally adsorbed water at high humidity has the properties of bulk liquid. The high-humidity sonic velocity variation is ascribed to the presence of free water. It is possible to identify the five water regimes of Pineri et al. [(1978) Biopolymers 17, 2799-2815] with the variations of the sonic velocity with water content.

    Notizen: Optical rotation, viscosity, and density studies are reported on solutions of α-gelatin in solvent mixtures of water and various monohydric alcohols. Reversion to the collagen fold by the protein is shown to be order in all cases, but changes in magnitude as a function of concentration of the particular alcohol are observed. The structuring effects of the alcohol on water are seen to be reflected in the extent of helix regeneration by the protein. Shorter chain alcohols appear to influence the initial rate of reversion by direct interaction with the protein.

    Notizen: We have recorded high-resolution 13C-nmr spectra of collagen fibrils in the solid state by the cross-polarization-magic-angle-spinning(CP-MAS)method and analyzed the spectra with reference to those of collagenlike polypeptides. We used two kinds of model polypeptides to obtain reference 13C chemical shifts of major amino acid residues of collagen (Gly, Pro, Ala, and Hyp): the 31-helical polypeptides [(Gly)nII, (Pro)nII, (Hyp)n, and (Ala—Gly—Gly)nII], and the triple-helical polypeptides [(Pro—Gly—Pro)n and (Pro—Ala—Gly)n]. Examination of the 13C chemical shifts of these polypeptides, together with our previous data, showed that the 13C chemical shifts of individual amino acid residues are the same, within experimental error (±0.5 ppm), among different polypeptides with different primary sequences, if the conformations are the same. We found that the 13C chemical shifts of Ala residues of the 31-helical (Ala—Gly—Gly)n and triple-helical (Pro—Ala—Gly)n are significantly displaced, compared with those of the α-helix, β-sheet, and silk I form, and can be utilized as excellent probes to examine conformational features of collagen-like polypeptides. Further, the 13C chemical shifts of Gly and Pro residues in the triple-helical polypeptides are substantially displaced from those found in (Gly)nII and (Pro)nII of the 31-helix, reflecting further conformational change from the 31-helix to the supercoiled triple helix. In particular, the 13C chemical shifts of Gly C = O carbons of the triple-helical polypeptides are substantially displaced upfield (4.1-5.1 ppm), with respect to those of the 31-helical polypeptides. These displacements are interpreted by that Gly C = O of the former is not involved in NH … O = C hydrogen bonds, while this carbon of the latter is linked by these kinds of hydrogen bonds.On the basis of these 13C chemical shifts, as reference data for the collagenlike structure, we were able to assign the 13C-nmr peaks of Gly, Ala, Pro, and Hyp residues of collagen fibrils, which are in good agreement with the values expected from the model polypeptides mentioned above. We also discuss a plausible conformational change of collagen fibrils during denaturation.

Assignments of most protons of dermorphin were facilitated by the study of the N-terminal fragments. Comparison of spectroscopic parameters with relative pharmacological activity is proposed as a possible means of studying flexible agonists in solution.

    Notizen: The magnetic circular dichroism (MCD) of adenine, hypoxanthine, and guanosine 5′-diphosphate reveals that, for each species, the uv-absorption band near 200 nm is composed of at least two electronic transitions. The theory of MCD shows that the dipoles of these transitions are not parallel to one another. The transitions are assigned within the framework of current theories of the electronic structure of the purines. Knowledge of the presence of more than one transition within an absorption envelope is important in interpreting the corresponding natural CD of DNA and RNA.

    Notizen: The aggregation of poly(γ-benzyl-α,L-glutamate) and its enantiomer in toluene has been investigated by following the viscosity as a function of temperature, concentration, molecular weight, molecular-weight distribution, helix chirality, and shear rate. The temperature and concentration data for a 138,000-molecular-weight sample was fitted to an open, reversible end-to-end aggregation model. The aggregation numbers resulting from this fit were consistent with the sudden onset in non-Newtonian flow resulting from only a 0.2-wt% increase in concentration. The association equilibrium constant was then used to predict viscosity for comparison with other data, in particular, the effect of molecular weight and molecular-weight distribution. A mixture of right-and left-handed helices showed the aggregation was not chiral selective. The stiffness of end-to-end aggregated (hydrogen-bonded) molecules differed little from their covalent counterparts, at least below a molecular weight of ∼106. We conclude that polybenzylglutamate aggregation in toluene can be described by an open end-to-end aggregation model.

    Notizen: Quasielastic light-scattering (QELS) studies were performed on a monodisperse preparation of dinucleosomal-sized DNA having an average of 375 base pairs. The QELS data were obtained at several time intervals at each angle examined over the range 30° 〈 θ 〈 60° for the ionic strength solvents 500, 50, 10, and 1 mM in KCl. What results from this study is that two relaxation domains persist over the entire ionic strength range where the “splitting” between these two domains increases as the ionic strength is lowered. The ordinate intercept of Dapp vs Cp/2Cs, where Cp is the polyion concentration and Cs is the added salt concentration, is shown to give the “true” hydrodynamic value for the diffusion coefficient. The “splitting phenomenon” is interpreted in terms of two models currently in vogue: small ion-polyion coupled modes and translational-rotational coupled modes for rigid rods in congested solutions. Although neither model is in perfect agreement with the experimental results, the small ion-polyion coupled mode theory appears to provide a better quantitative description of the observations.
