    Notizen: Permeabilities of several solutes through the composite membranes containing phospholipids have been measured. They were inversely proportional to the content of the phospholipids in the membrane. Both the permeability of solutes and the degree of permeability change around the phase transition temperature of the phospholipids for the hydrophobic solutes such as n-butanol and salicylamide were larger than those for the hydrophilic solutes such as amino acids and pyridoxine. These results suggest thatthe permeation path of hydrophobic solutes is different from that of hydrophilic ones. The addition of phosphatidyl ethanolamine, phosphatidyl serine, or phosphatidic acid to the composite membrane influenced the solute permeability due to the introduced negative charge and/or the change in the molecular packing of phospholipid.

    Notizen: Biosorption of uranium and lead by lyophilized cells of Streptomyces longwoodensis was examined as a function of metal concentration, pH, cell concentration, and culture age. Cells harvested from the stationary growth phase exhibited an exceptionally high capacity for uranium (0.44 g U/g dry weight) at pH 5. Calculated values of the distribution coefficient and separation factor indicated a strong preference of the cell mass for uranyl ions over lead ions. The specific uranium uptake was similar for the cell wall and the cytoplasmic fraction. Uranium uptake was associated with an increase in hydrogen ion concentration, and phosphorus analysis of whole cells indicated a simple stoichiometric ratio between uranium uptake and phosphorus content. It is proposed that metal ions are bound to phosphodiester residues present both in the cell wall and cytoplasmic fractions. Based on this model, it was shown that uranium accumulation exhibits a maximum at pH 4.6 that is supported by experimental data from previous investigations.

    Notizen: A separate role for water activity in the conversion of sugars to ethanol by two strains of yeast is identified. During fermentation of both single and mixed sugar substrates, the water activity was shown to remain constant during the logarithmic growth phase. This is despite the changes in concentration of substrates and product, the constancy reflecting the fact that the greater influence of ethanol on the solution activity is counterbalanced, in the early stages of the fermentation, by its low yield. The end of the log phase of growth coincides with the start of a period of gradually decreasing water activity. For the more ethanol-tolerant strain UQM66Y, growth was found to cease at a constant value of water activity while that for the less tolerant strain UQM70Y depended on both ethanol concentration and water activity. It is argued that water activity is a more appropriate variable than ethanol concentration for describing some of the nonspecific inhibitory effects apparent in ethanol fermentations. A straightforward method for the calculation of water activity during such fermentations based on the use of solution osmolality is presented.

    Notizen: Poly(maleic anhydride styrene) graft copolymers of cellulose, pectin polygalacturonic acid salt, calcium polygalacturonate, and starch were prepared and used to immobilize proteins. The cellulose grafts coupled quite appreciable quantities of acid phosphatase, glucose oxidase, and trypsin. However, the general retention of activity was somewhat disappointing. Further investigation with acid phosphatase showed that the amount of enzyme immobilized increased as the amount of anhydride in the graft copolymer increased but no such relationship existed for the enzymic activity. The cellulose graft copolymers were hydrolyzed and it appeared that the carboxyl group aided adsorption of the enzyme. Attempts to couple acid phosphatase using CMC through the free carboxyl groups, created by hydrolysis, gave only a small increase in the extent of protein coupling. However, the unhydrolyzed system gave a useful degree of immobilization of cells of Bacillus stearothermophilus, as did a poly(maleic anhydride/styrene)-cocellulose system. Attempts to improve the activity by using grafts based on other polysaccharide supports met with mixed success. Pectin products were soluble. Polygalacturonic acid products were partially soluble and extremely high levels of enzymic activity were obtained. This was probably due in part to the hydrophilic nature of the system, which also encouraged absorption of the enzyme. Attempts were made to reduce the solubility by using the calcium pectinate salt. Immobilization of acid phosphatase and trypsin resulted in inceased protein coupling but relatively poor activities were attained. A starch based system gave similar results. Calcium polygalacturonate was used to prepare an insoluble graft copolymeric system containing acrylonitrile-comaleic anhydride. The resulting gels gave excellent coupling with acid phosphatase which had a very good retention of activity.

    Notizen: Urokinase was immobilized by entrapment to fibrocollagenous tubes in order to develop a small-caliber fibrinolytic vascular prosthesis. Several parameters associated with the immobilization process were studied in order to optimize bound urokinase activity and stability. A total of 37% of the absorbing enzyme was attached to the collagen tube and 38% of the attached enzyme retained esterolytic activity, under optimal conditions. In the crosslink step of the entrapment process, the glutaraldehyde concentration was varied from 0.01 to 5.00% (i.e., 0.01, 0.1, 1.0, 3.0, and 5.0%). Urokinase activity was optimized at a 1.0% glutaraldehyde crosslink concentration. Urokinase-bound fibrocollagenous tubes (UK-FCT) prepared at the above glutaraldehyde concentrations were tested for their activity with time. The UK-FCT's with 0.1, 1.0, and 3.0% glutaraldehyde retained constant activity for at least 75 h operation time. The UK-FCT's with 0.1, 1.0, and 3.0% glutaraldehyde retained constant activity for at least 75 h operation time. The UK-FCT's with 5.0 and 0.01% glutaraldehyde remained stable for the first 50 h operation time, but begandeactivating beyond 50 h. UK-FCT'S Crosslinked with 0.01, 0.1, 1.0, and 5.0% glutaraldehyde were recrosslinked with 0.02% glutaraldehyde for 24 h, after they have been operating for 50 h, and the effect of reexposing the crosslink agent on the stability of the UK-FCT's was studied. The results showed that 0.02% glutaraldehyde reexposure had no effect on 0.1, 1.0, and 5.0% glutaraldehyde crosslinked UK-FCT's but exerted an inhibitory effect on a 0.01% crosslink density UK-FCT. Several fibrocollagenous tubes were exposed to various glutaraldehyde concentrations prior to immobilizing urokinase. The subsequent immobilization process occurred under optimal conditions. The effect of the precrosslink step on the activity of the UK-FCT was studied. Results indicated that UK-FCT activity decreases as the precrosslink density increases. The UK-FCT's made under optimal conditions remained stable for at least 75 h operation time, corresponding to ca.1 year of storage time. Ex vivo exposure of UK-FCT's to whole canine blood did not affect catalytic activity. Implantation of a UK-FCT by carotid arterial interposition via an end-to-end anastomosis and subsequent excision after 60 days resulted in an enhanced esterolytic activity which decreased with time to a level close to preoperative levels.

The sensitivity of transient responses to seemingly minor differences in the design of experiments indicated that this type of investigation may be a means to analyze kinetics of microbial cultures as well as the effectiveness of bioreactors.

    Notizen: The kinetics in batch culture of the acetone butanol fermentation by Clostridium acetobutylicum is compared on glucose, xylose, and mixtures of both sugars. The fastest initial growth and transition from an acid to a solvent metabolism occurs on glucose, with a final 62 g/L glucose conversion. On xylose, an initial slower growth rate and a longer metabolic transition result in higher cellular and acids concentration, thus in a level of fermented sugar limited to 47 g/L. Batch fermentations on mixtures of glucose and xylose show that both sugars can be fermented, with a higher rate for glucose. However, xylose fermentation is inducible and inhibited at glucose level above 15 g/L. Mixtures of glucose and xylose yield the highest amount of fermented sugars, up to 68 g/L, as a result of both a fast metabolic transition on glucose and a strong acid reconsumption on xylose. In all cases, solvent production is triggered at a total acid concentration between 4 and 5 g/L, whereas the final inhibition of the fermentation takes place at a total butanol and acid concentration between 18 and 20 g/L.

    Notizen: The present work deals with maltodextrin hydrolysis by glucoamylase immobilized onto corn stover in a fluidized bed reactor. An industrial enzyme preparation was covalently grafted onto corn stover, yielding an activity of up to 372 U/g and 1700 U/g for support particle sizes of 0.8 and 0.2 mm, respectively. A detailed kinetic study, using a differential reactor, allowed the characterization of the influence of mass transfer resistance on the reaction catalyzed by immobilized glucoamylase. A simple and general mathematical model was then developed to describe the experimental conversion data and found to be valid.

    Notizen: The kinetics in fed-batch cultures of acetone butanol fermentation by Clostridium acetobutylicum is compared on glucose, xylose, and mixtures of both sugars. The final conversion yield of sugars into solvents always increases with the sugar feeding rate. At low feeding rates, the sugar concentration in the medium becomes limiting, which results in a slower cellular growth, a slower metabolic transition from an acid to a solvent fermentation and, thus, a higher accumulation of acids. It is only at sufficiently high feeding rates that fed-batch fermentations yield kinetic results comparable to those of batch fermentations. With mixtures of glucose and xylose, because of a maintained low glucose level, both sugars are taken up at the same rate during a first fermentation period. An earlier accumulation of xylose when the fermentation becomes inhibited suggest that xylose utilization is inhibited when the catabolic flux of glucose alone can satisfy the metabolic activity of the cell. Kinetic results with batch and fed-batch fermentations indicate several important features of the regulation of C. acetobutylicum metabolism: an early inhibition by the produced acids; an initiation of solvent formation between 4 and 6 g/L acetic and butyric acid depending on the metabolic activity of the cell; a metabolic transition from acids to solvents production at a rate closely related to the rate of sugar uptake; during solvent production, a reassimilation of acids above a minimal rate of sugar consumption of 0.2 h-1; a final inhibition of the fermentation at a total butanol and acids concentration of ca. 20 g/L.

    Notizen: Method of flow cytometric analysis have recently been developed that make it possible to obtain segregated data on a single cell basis. In particular, it has been previously demonstrated that protein distributions obtained by flow cytometry give information about the law of growth of the cell population and the law of growth of the single cell; thus these distribution show how the microbial population is actually growing at the moment of the analysis and may yield more accurate and predictive information. We have extended the analysis of protein distribution and cell volume distribution to continuous cultures of Saccharomyces cerevisiae growing in a glucose-limited chemostat. We have found that: (1) to each dilution rate corresponds a given protein and volume distribution that does not change with time in steady state cultures; (2) there is a good proportionality between the average cell volume and the average protein content; (3) the protein distribution obtained can be easily analyzed with the model of growth of yeast previously developed in our laboratory; (4) the analysis of perturbed states shows that both protein distribution and volume distribution change very quickly; thus they are very sensitive parameters and can be used for monitoring and controlling industrial fermentation.

    Notizen: This article describes a method for the chemical immobilization of concanavalin A (Con A) on the inside wall of a single hollow cellulose fiber for use in glucose affinity sensor. Periodate oxidation of cellulose fiber followed by a spacer for Con A attachment was deemed to be the most optimal procedure for achieving the highest sensitivity of the sensor without compromising its physical integrity. The effects of variables like the duration of periodate oxidation and its concentration and pH of the spacer coupling step and its duration have been examined. The mechanical strength of the hollow fiber as well as its permeability to the analyte (glucose) have been evaluated prior to and after Con A coupling process.It has been demonstrated that Con A bound hollow fiber prepared according to the procedure outlined here can be successfully used to construct glucose affinity sensor for operation in the physiological range of glucose concentrations.

    Notizen: The concepts of phase separation, anaerobic activated sludge process, and alkali pretreatment have been incorporated in this investigation with the objective of developing rational and cost-effective designs of diphasic anaerobic activated sludge systems, with and without alkali treatment, for methane recovery from water hyacinth (WH). Evaluation of process kinetics and optimization analyses of laboratory data reveal that a diphasic system with alkali treatment could be designed with an alkali pretreatment step (3.6% Na2CO3 + 2.5% Ca(OH)2 (w/w) of WH, 24 h duration) followed by an open acid phase (2.1 days HRT) and closed methane reactor with sludge recycle (5.7 days HRT, 7.7 days MCRT) for gas yield of 50 L/kg WH/d at 35-37°C. Likewise, a diphasic system without alkali treatment could be designed with an open acid phase (2 days HRT) followed by closed methane reactor with sludge recycle (3.2 days HRT, 6 days MCRT) for gas yield of 32.5 L/kg WH/d at 35-37°C. Detailed economic analyses bring forth greater cost-efficacy of the diphasic system without alkali treatment and reveal that the advantage accrued in terms of higher gas yield is overshadowed by the cost of chemicals in the diphasic system with alkali treatment.

    Notizen: Within the framework of a program aiming to improve the existing extractive recovery technology of fermentation products, the state of the art is critically reviewed. The acids under consideration are propionic, lactic, pyruvic, succinic, fumaric, maleic, malic, itaconic, tartaric, citric, and isocitric, all obtained by the aerobic fermentation of glucose via the glycolytic pathway and glyoxylate bypass. With no exception, it is the undissociated monomeric acid that is extracted into carbon-bonded and phosphorus-bonded oxygen donor extractants. In the organic phase, the acids are usually dimerized. The extractive transfer process obeys the Nernst law, and the measured partition coefficients range from about 0.003 for aliphatic hydrocarbons to about 2 to 3 for aliphatic alcohols and ketones to about 10 or more for organophosphates. Equally high distribution ratios are measured when long-chain tertiary amines are employed as extractants, forming bulky salts preferentially soluble in the organic phase.

    Notizen: The kinetics of precipitation of soya protein has been examined in a tubular flow reactor with the precipitants, ammonium sulfate, ethanol, divalent calcium, and sulfuric acid. The precipitate growth profiles obtained in all cases showed the rapid formation of a precipitate phase and the attainment of a final size within 16 s. The final mean particle diameter dm varied with precipitating agent used in the order: sulfuric acid (12.5 μm) 〉 ethanol (7.5 μm) ≈ calcium ion (7.2 μm) 〉 ammonium sulfate (3.1 μm). In the case of ethanol precipitation, changes in the design of the contacting section of the reactor led to differences in the precipitate growth curve. Protein solubility curves are also presented, and with the reactor data, they provide a convenient method for assessing the effect of precipitating agent on the design of protein precipitation reactors.

    Notizen: The biodegradation of lignin by fungi was studied in shake flasks using 14C-labeled kraft lignin and in a deep-tank fermentor using unlabeled kraft lignin. Among the fungi screened, A. fumigatus - isolated in our laboratories - was most potent in lignin biotransformation. Dialysis-type fermentation, designed to study possible accumulation of low MW lignin-derived products, showed no such accumulation. Recalcitrant carbohydrates like mi-crocrystalline cellulose supported higher lignolytic activity than easily metabolized carbohydrates like cellobiose. An assay developed to distinguish between CO2 evolved from lignin and carbohydrate substrates demonstrated no stoichiometric correlation between the metabolism of the two cosubstrates. The submerged fermentations with unlabeled lignin are difficult to monitor since chemical assays do not give accurate and true results. Lignolytic efficiencies that allowed monitoring of such fermentations were defined. Degraded lignins were analyzed for structural modifications. A. fumigatus was clearly superior to C. versicolor in all aspects of lignin degradation; A. fumigatus brought about substantial demethoxylation and dehydroxylation, whereas C. versicolor degraded lignins closely resembled undegraded kraft lignin. There was a good agreement among the different indices of lignin degradation, namely, 14CO evolution, OCH3 loss, OH loss, and monomer and dimer yield after permanganate oxidation.

    Notizen: Mold growth and differentiation are closely related to the formation of secondary products. In solid-substrate fermentations this interrelationship is often more completely realized than in submerged cultures. Solid substrate reactions are used commercially in a limited manner in the western world, but are relatively common in Asia. Basic studies in solid-substrate fermentation should yield results applicable to all types of commercial mold fermentations for the production of a secondary product. This paper presents a relatively simple model for the growth of a mold colony on a solid surface with a defined medium utilizing glucose. Unlike submerged cultures the model must account for both cellular differentiation and the spatial heterogeneity in the system. Model parameters were estimated independently using literature values. The results of the simulation studies suggest that mass transfer limitations are at least partially responsible for the proliferation of differentiated structures on solid substrates as compared to liquid cultures. Since the concentration profile depends on the depth of the substratum, conditions that enhance conidia production can be achieved by controlling the depth of the solid medium.

The enzymes in the xylose catabolic pathway necessary for the conversion of xylose to xylulose are present, although S. cerevisiae cannot grow on xylose as a sole carbon source. Xylose transport is less efficient than glucose transport, and its rate is dependent upon aeration. Xylose reductase appears to be a xylose inducible enzyme and xylitol dehydrogenase activity is constitutive, although both are repressed by glucose. Both xylose reductase and xylitol dehydrogenase activities are five- to tenfold lower in S. cerevisiae as compared to Candida utilis. In vivo conversion of 14C-xylose in S. cerevisiae is demonstrated and xylitol is detected, although no significant levels of any other 14C-labeled metabolites (e. g., ethanol) are observed.

    Notizen: Steam-exploded aspen has been examined as a candidate feedstock for both cellulose production and enzymatic hydrolysis of wood. Batch and fed-batch cultivation methods were evaluated and compared with previous experiments using ball-milled, crystalline cellulose (Solka Floe). Batch cultivation of Trichoderma reesei Rut C-30 on 9 wt% water-washed aspen yielded enzyme productivities and activities comparable to those obtained on Solka Floe (40 FP IU/L-h; 7. 5 FP IU/mL). Fed-batch cultivation of Rut C-30 resulted in higher enzyme productivities and tilers than batch cultivation (50 FP IU/L-h; 15 FP IU/mL). However, the overall enzyme production performance was lower than on Solka Floe at comparable cellulose feeding rates and concentrations. This may be due to the accumulation of steam explosion by-products and lignin in the fermentor.The hydroiysis of water-washed steam-exploded aspen was performed at different enzyme loadings and wood concentrations. Glucose production, using 10 and 15wt% suspension, showed that while glucose concentration increased with wood load, the yield of glucose derived from cellulose decreased. With 10wt% suspensions, it was possible to obtain a cellous conversion to glucose above 95%. Low cellulose levels in the hydrolyzates indicated that the filter paper activity ratios (approximately 1.5), a significant result since the fungus was grown exclusively on wood. mIt also suggested that the observed yield decrease is more likely to be caused by glucose than cellobiose inhibition of the enzymes.

    Notizen: Coalescence frequency has been measured in an air-sparged bubble column by observing the rate with which insoluble tracer gases were mixed within the dispersed gas phase. The effects of gas-sparge rate and salt concentration were examined. Coalescence frequencies increased with increasing gas-sparge rates approximately in proportion to the increase in gas-phase hold up. The coalescence frequency decreased with NaCI concentration to a minimum frequency about half the frequency observed in pure water. Bubble size distributions were also measured in the air-sparged column. The size distribution changed significantly over the same range of NaCI concentration as variations in the coalescence frequency were observed. The gas-sparge rate showed no effect on the bubble size.

    Notizen: A mathematical model for the cell separation technique of centrifugal elutriation is developed. The model simulates both steady and non-steady-state operation of the elutriator. The model can be used to predict the required set of flow rates of elutriating liquid necessary to fractionate a cell culture, the required time of sampling before steady state is achieved, and the range of cell size/cell density combinations contained in any fraction. The model predictions were verified experimentally. Variations in cell density and cell size due to the suspending environment have a significant effect on the accuracy (although not the trends) of the model predictions. Quantification of these variations will lead to significantly more accurate model predictions. An enhanced separation method was developed using the model, to yield finer separation of a cell culture than previously possible. The use of the centrifugal elutriator may now be given a firm theoretical basis, with the quality of separation understood in terms of the basic theory of operation.

They were measured here for the bacterium Pseudomonas aeruginosa, which tends to be predominant in sewage treatment, at a pressure remaining constant or periodically changing in the range of 1-6 atm (abs.). Cellular yields tended to decrease over 1-2 min pressure cycles; in steady operation they decreased considerably with increasing pressure or dissolved oxygen concentration. Maximum specific growth rates were moderately affected by periodic changes in pressure or dissolved oxygen concentration but markedly affected in the higher concentration range under steady operation. Substrate consumption rates on the other hand, were little affected in either case. The values of chemical oxygen demand ranged from 220 to 250 mg/L, and were thus nearly constant.

    Notizen: Growth of a hybridoma culture, along with production of monoclonal antibody, was demonstrated over extended periods in polysulfone hollow fiber membrane modules. The molecular weight cutoffs of the membranes were 70,000, 50,000, and 100,000 daltons. The hybridoma cell line, designated 65/26, produced IgG (2b/κ) directed at mouse thymus cell surface antigen, TL.1. Cell growth occurred in the shell space of the reactor, using supplemented RPMI 1640 (20% fetal bovine serum) supplied from a separate reservoir vessel through the hollow fiber lumen. The reservoir contained 125 mL media, which was changed every 4 days. Concentrations of immunoglobulin were determined by an enzyme immunoassay (using protein A and alkaline phosphatase-labeled antibody conjugate). For the 10K, 50K, and 100K hollow fiber membrane modules, the maximum IgG concentrations detected in the 2.5-mL shell space were 47.5-80, 510, and 740 μg/mL, respectively. In the 125-mL reservoir for the 100K hollow fiber membrane module, the IgG concentration was measured at 260 μg/mL These values compare with an IgG concentration of 1 μg/mL when grown in a standard tissue culture flask and 3.2-7.6 μg/mL when grown in 100 ml media in a spinner flask. In addition, 10K and 50K hollow fiber membrane modules were run in a mode that decreased the fetal bovine serum supplement with time. Differences between these systems suggest that it is possible to obtain high IgG accumulation rates, both during and after the exponential growth phase of the hybridoma population.

    Notizen: Batch and continuous fermentations with Clostridium thermoaceticum (ATCC 39073) using automatic pH control were conducted. The value of μmax obtained from batch fermentation was about 0.14 h-1; acetate yield, which was both growth and non-growth associated, was about 2 mole of acetic acid/mole of glucose, compared with a theoretical maximum value of 3. This low yield, compared with literature data, may be explained by glucose loss through a combination of degradation routes. Continuous fermentation could be sustained for 1600 h or more without contamination problems. Continuous fermentation at high dilution rates indicates that μmax may be well above 0.17 h-1 when fresh feed medium is used. Acetate yields in continuous fermentation were about 77% of theoretical or 2.3 mole of acetic acid/mole of glucose.

    Notizen: Continuous cultures of Saccharomyces cerevisiae are known to exhibit oscillatory behavior in the oxidative region. Important findings of a series of experiments conducted to identify the causes for initiation of and the means for elimination of oscillations in these cultures are reported in this paper. These oscillations are seen to be connected to the growth kinetics of the microorganism and are induced at very low glucose concentrations and at dissolved oxygen (DO) levels that are neither high nor low (DO values between 20 and 78% air saturation at a dilution rate of 0.2 h-1 and pH of 5.5 at 30°C). The oscillatory behavior is encountered over a range of dilution rates (0.09-0.25 h-1 at 30°C for pH = 5.5 and DO = 50% air saturation). The oscillations can be eliminated by raising the DO level above a critical value or by lowering the DO level below a critical value.

    Notizen: A novel mechanistic model for the growth of baker's yeast on glucoseis presented. It is based on the fact that glucose degradation proceeds via two pathways under conditions of aerobic ethanol formation. Part is metabolized oxidatively and part reductively, with ethanol being the end product of reductive energy metabolism. The corresponding metabolic state is designated oxidoreductive. Ethanol can be used oxidatively only. Maximum rates of oxidative glucose and ethanol degradation are governed by the respiratory capacity of the cells. The model is formulated by using the stoichiometric growth equations for pure oxidative and reductive (fermentative) glucose and ethanol metabolism. Together with the experimentally determinable yield coefficients (YX/S) for the respective metabolic pathways, the resulting equation system is sufficiently determined. The superiority of the presented model over hitherto published ones is based on two essential novelities. (1) The model was developed on experimentally easily accessible parameters only. (2) For the modeling of aerobic ethanol formation, the substrate flow was split into two simultaneously operating (i.e., in parallel) metabolic pathways that exhibit different but constant energy-generating efficiencies (respiration and fermentation) and consequently different and constant biomass yields (YX/S). The model allows the prediction of experimental data without parameter adaption in a biologically dubious manner.

    Notizen: Many of the sophisticated sensors desirable for monitoring bioreactors cannot be placed in the bioreactor either because they are not steam sterilizable or because they require nonphysiological operating conditions. Such sensors can be used if they are separated from the bioreactor. Separation of the sensor from the bioreactor causes a time lag in data acquisition. This results in several complexities in the development of an appropriate and stable feedback control system based on a separated sensor. This paper analyzes the optimal control of a bioreactor with a separated sensor without a time lag and analyzes the feedback control (but not necessarily the optimal control) with a time lag. Simulation results indicate that this type of analysis could be extended to more general bioreactor operating conditions.

    Notizen: The effect of the presence of more easily degradable alternative carbon sources on the biodegradation of toxic waste components is of great practical importance. In this work, a mixed phenol/glucose waste was fed to two heterogeneous populations acclimated to different conditions: one was acclimated to phenol as a sole source of carbon and one to a mixed phenol/glucose substrate. Batch substrate utilization experiments were performed under both growth and nonproliferating (no medium nitrogen source) conditions in order to assess substrate removal patterns at the levels of enzyme production and enzyme function. The results indicated that the substrate removal pattern exhibited by the cells was significantly influenced by the acclimation characteristics of the culture. The phenol acclimated cells showed an initial preference for phenol, but the presence of glucose hindered phenol removal rate under both growth and nonproliferating conditions. The cells acclimated to the mixed phenol/glucose waste demonstrated rapid initial glucose removal with a slower concomitant utilization of phenol; acclimation to the mixed waste evidently had a significant impact on the substrate removal pattern for this mixed substrate system.

    Notizen: The time course of the peroxidative bromination of propylene accompanied by in situ generation of hydrogen peroxide by glucose oxidase was examined to improve the productivity of propylene bromohydrin. To prevent the rapid inactivation of lactoperoxidase by excess hydrogen peroxide, it was effective to use lactoperoxidase in large excess as compared with glucose oxidase, and to raise the concentration of bromide ion. However, the rate of glucose consumption was lowered at high concentrations of bromide ion, and at higher mole fraction of oxygen as compared with propylene in the gas mixture. Therefore, it seemed that for the favorable production of bromohydrin there existed the optimal conditions for the concentration of bromide ion and for the composition of oxygen-propylene gas mixture. Such kinetic behaviors of the sequential enzymatic reactions were explained by a mechanism involving free hypobromous acid as a reactive intermediate. Furthermore, the stability of the coimmobilized enzymes with k-carrageenan gels was investigated in continuous operations. The half-life of the enzymes was ca. 60 h for the production of propylene bromohydrin.

Under definite hydrodynamic conditions with low agitation in the reaction medium, the observed behavior is modified compared to the enzyme free in solution: reducing the stirring rate decreases the observed enzymatic activity. But diffusional resistances alone cannot account for these atypical kinetics though mass transfer may certainly play an important role during the transient state of the bioluminescent reaction. After immobilization, the time necessary to reach the steady state increased from 300 ms to 3 min and the two substrates, luciferin and ATP, behave differently with respect to the enzyme: The nature of the saturating substrate first in contact with the bound enzyme is not indifferent suggesting that immobilization can reveal behaviors or mechanisms which are not visualized with the free enzyme.

    Notizen: In relation to the input-output characteristics of enzymatic reactions in the cellular metabolism and biochemical reactors, the validity of the quasi-steady-state and transfer-function representations of reaction velocity has been examined for a basic Michaelis-Menten reaction employing computer simulation, that is, numerical integration of the rate equation. The well-known S-v relationship (relationship between substrate concentration and reaction velocity)derived on the quasi-steady-state assumption is found to be in general a good approximation to the actual velocity throughout the temporal progress of the reaction. The validity of the approximation depends on a ratio of the Michaelis constant to the total enzyme concentration in the reaction system rather than on the individual rate constants. A transfer-function representation is derived on assuming an exponential change in the reaction velocity for the indicial response to the substrate influx rate. The representation has a wider valid region with a decrease in influx rate than with an increase in the influx rate. The validity is most dependent on a ratio of total enzyme concentration to the steady-state concentration of the substrate. The analysis of the linear sensitivity of the reaction velocity to rate constants reveals that the characteristics of these valid representations in systems analysis change according to the phase of the reaction.

    Notizen: This article deals with aspects of the reciprocal interaction between the activity of chloroplast membranes and their microenvironment. The artificial matrices used in the present work to immobilize thylakoids (albumin-glutaraldehyde matrix, polyurethane foam) can be regarded as weak ion exchangers. Thus, the distribution of the solute between the matrix surface and the external solution should, at least in part, be governed by a Donnan equilibrium. The influence of a high ionic strength medium (750mM potassium citrate) on the kinetic parameters (Kp1 Vm) and on the stability of the photosynthetic activity of immobilized chloroplast membranes has been studied. The results show similarities in behavior of the two supports studied in that, for both, a high concentration of salt (citrate) increases the apparent affinity for ferricyanide and allows a better transformation of this electron acceptor in CSTR experiments.

    Notizen: A novel method for the preparation of highly active immobilized enzymes is described. It is based on the binding of enzymes to suitable carriers via monoclonal antibodies, which bind to the enzyme with high affinity without affecting its catalytic activity. The applicability of the method forwarded has been illustrated by the preparation of two samples of highly active immobilized carboxypeptidase A (CPA) preparations as follows: A mouse monoclonal antibody (mAb 100)to CPA that binds to the enzyme with a high-affinity constant without affecting its catalytic activity was prepared, purified, and characterized. Covalent binding of this monoclonal antibody to Eupergit C (EC) or noncovalent binding to Sepharose-protein A (SPA)yielded the conjugated carriers EC-mAb and SPA·mAb, respectively, which reacted specifically with CPA to give the immobilized enzyme preparations EC-mAb·CPA and SPA·mAb·CPA displaying full catalytic activity and improved stability. At pH 7.5 and a temperature range of 4-37°C an apparent binding constant of ∼108M-1 characterizing the interaction of CPA with EC-mAb and SPA·mAb, was obtained. To compare the properties of EC-mAb·CPA and SPA·mAb·CPA with those of immobilized CPA preparations obtained by some representative techniques of covalent binding of the enzyme with a corresponding carrier, the following immobilized CPA preparations were obtained and their properties investigated: EC-CPA (I), a preparation obtained by direct binding of EC with CPA; EC-NH-GA-CPA (II), a derivative obtained by covalent binding of CPA to aminated EC via glutaraldehyde; EC-NH-Su-CPA (III), a CPA derivative obtained by binding the enzyme to aminated EC via a succinyl residue; and EC-HMD-GA-CPA (IV), obtained by binding the enzyme via glutaraldehyde to a hexamethylene diamine derivative of EC. Full enzymic activity for all of the bound enzyme, such as that recorded for the immobilized CPA preparations EC-mAb·CPA and SPA·mAb·CPA, was not detected in any of the insoluble covalently bound enzyme preparations.

    Notizen: Thermostable lipase from Thermomyces lanuginosus was immobilized in untreated microporous membranes. Melted tallow pumped through the membrane did not wash the enzyme out. From 0.4 to 0.9% of the soluble activity remained after immobilization with half-lives of 1-2 months or more at 50°C. Membranes can be acid/base washed and reloaded with enzyme with no adverse effects. Buffer was required for a long half-life, and recycling the buffer improved the mass transfer of glycerol out of the immobilized lipase reactor. Immobilized activity was unaffected when the pH of the aqueous product changed from 5.5 to 6.5.

    Notizen: Mammalian cells grown in culture excrete lactic acid and ammonium ions in quantities that may limit growth and reduce product synthesis. Frequent replenishment of the culture medium is often necessary to prevent waste product accumulation which could inhibit cell growth. Since increased medium replenishment results in increased usage of animal serum, the most expensive raw material, excessive production of waste products lowers the cell and product yield on serum, and hence increases production costs. Strategies for reducing the production of lactic acid and ammonium bymammalian cells via controlled addition of glucose and glutamine will be demonstrated. Mathematical relations coupling ammonium and glutamine kinetics will be described. Additionally, a method for automatic on-line estimation of the cell concentration was developed. This method involves calculating the ATP production rate from the oxygen uptake rate and the lactic acid production rate. Automatic online estimation of the cell concentration is critical if nutrient levels in large-scale mammaliancell cultures are to be accurately maintained via process control.

    Notizen: Sugar-limited batch growth of Dioscorea deltoidea and Catharanthus roseus plant cell cultures was studied in a 14-L stirred tank fermentor. With dissolved oxygen concentration monitored and maintained at nonlimiting levels, growth rates and ratios of dry weight to fresh weight were found to be strongly influenced by sugar concentration. Linear relationships between respiration rate and growth rate were observed, and respiration rate was found to drop to a maintenance level after sugar had been fully depleted from the medium. Diosgenin biosynthesis by D. deltoidea was shown to be independent of growth rate. Ajmalicine biosynthesis in C. roseus was negligible during sugar-limited growth but was induced by inoculation into an 80-g/L glucose solution.

Immobilized cell concentrations of up to 18 g/L volatile suspended solids could be achieved with the s-triazines as sole nitrogen source for growth and carbon sources added at a C—N ratio of about 12. Maximal removal efficiencies of 80% of the s-triazines could be maintained only if (a) the bio-film thickness was limited to avoid oxygen deficiency and (b) the carbon source and complete wastewater (≤50% v/v in the feed) were supplied continuously at a mean hydraulic residence time of ≥20-25 h.

    Notizen: This research is aimed at understanding the reactor-settler interaction. The reactor operating conditions affect the sludge-settling properties due to (a) production of exocellular polymer that serves as a flocculant in the settler and (b) population shift from flocculating bacteria to filamentous microorganisms that do not settle. A structured kinetic model that accounts for stored substrate and production of polymers, biomass, and inerts was constructed and applied to various open and closed reactor configurations. It is based on our observation that, in a batch reactor, the polymer is produced during endogenous respiration. The model describes well the observed temporal variation in substrate, biomass, and polymer concentration. Application of the model to continuous operation predicts increasing polymer concentration with sludge age, which is in good agreement with one study in the literature. Our study in a once-through reactor, as well as others, suggests an inverse dependence. The difference is probably due to different operating modes. Our study confirmed the role of the polymer in the sedimentation process. Under some conditions, however, a slow population shift to filamentous growth becomes the dominant factor.

    Notizen: A comprehensive kinetic study of the acid hydrolysis of concentrated brewing bagasse slurries was performed. The use of the simple series reaction model was found to be suitable when a “heterogeneous correction” (pseudosubstrate-inhibition) is taken into account in slurries with low liquid-to-biomass ratios. Rate constants are shown to be dependent not only on temperature and acid concentration but essentially also on the initial biomass concentration. Actual rate constants, activation energies, and acid and substrate reaction orders are reported for xylan, arabinan, and α-glucan acid saccharification. There is a threshold acid loading necessary to overcome the 80% conversion, but no threshold has been found to overcome the “neutralizing” property of cellulosic materials. Reversible acid capture from brewing bagasse has been postulated. The highest monosccharide concentration into hydrolyzates has been found (65 g/L) after 10 h treatment, but economic considerations led us to treat a mean-concentrated slurry (156 g/L) with 0.3M H2SO4 at 96°C, thus obtaining 45.5 g/L monosaccharides in 5 h with 50% less furfural content. After pH regulation only, growth of Clostridium acetobutylicum has been obtained, although complete sugar comsumption has not been achieved. Experiments are now underway to reach complete digestion and to investigate the increase of enzymic accessibility into residual substrate rich in cellulose.

    Notizen: A mathematical model has been developed for the internal pH control in immobilized enzyme particles. This model describes the kinetics of a coupled system of two enzymes, immobilized in particles of either planar, cylindrical, or spherical shape. The enzyme kinetics are assumed to be of a mixed type, including Michaelis-Menten kinetics, uncompetitive substrate inhibition, and competitive and noncompetitive product inhibition. In a case study we have considered the enzyme combination urease and penicillin acylase, whose kinetics are coupled through the pH dependence of the kinetic parameters. The hydrolysis of urea by urease yields ammonia and carbon dioxide, whereas benzylpenicillin (Pen-G) is converted to 6-amino penicillanic acid and phenyl acetic acid by penicillin acylase. The production of acids by the latter enzyme will cause a decrease in pH. Because of the presence of the ammonia-carbon dioxide system, however, the pH may be kept under control. In order to obtain information about the optimum performance of this enzymatic pH controller, we have computed the effectiveness factor and the conversion in a CSTR at different enzyme loadings. The results of the computer simulations indicate that a high conversion of Pen-G may be achieved (80-90%) at bulk pH values of about 7.5-8.

    Notizen: Growth yield factors, plasmid stability, cellular plasmid content, and cloned gene product activity for Escherichia coli HB101 containing plasmid pDM246 were measured at several dilution rates in continuous culture. Cell mass yield per mass of glucose consumed declined with increasing dilution rate. There was no evidence of plasmid segregational instability in any experiments, none of which employed selective medium. Plasmid content per cell varied with population-specific growth rate as observed in earlier batch experiments with the same strain. Plasmid content declined with increasing specific growth rate following indication of a maximum number of plasmids per cell at specific growth rates of ca. 0.3 h-1. Cloned gene product (β-lactamase) activity exhibited a sharp maximum with respect to dilution rate in continuous culture. Qualitatively different results were observed in previous experiments in batch cultivation in which specific growth rate changes were effected by altering medium composition.

    Notizen: A new, semipermeable hydrogel membrane for encapsulating viable cells has been developed. The encapsulation was performed by consecutively introducing droplets of a suspension of hybridoma cells in a solution of a polyanionic acrylic copolymer into aqueous solutions of three polycationic polymers. As a result of interpolymeric ionic interactions and some chemical reactions, a polyelectrolyte complex membrane was formed at the interface between each droplet and the polycationic polymeric solutions. The hybridoma cells, used as a model system, were derived by fusing spleen cells from immunized BALB/c mice with the NS-1 murine plasmacytoma cell line. The cells divided and gradually filled the microcapsules over a period of 8 days. Prior to encapsulation of the hybridoma cells, the polyanions were tested for toxicity and inhibition of cell growth. A direct relationship was observed between hybridoma cell viability in the acrylic polyanion/RPMI-1640/10% (w/v) fetal calf serum (FCS) solutions and the kinematic viscosities of the solutions of the polyanions. Antihuman IgM was produced by the encapsulated hybridoma cells and immunoassay showed that the antibody concentration was 3 μg/ml of the total culture medium.

    Notizen: Semicontinuous digesters were used to anaerobically treat high-strength whey (70 kg/m3·COD). A maximum loading of 16.1 kg COD/m3·day was obtained with soluble COD removal efficiencies greater than 99%. The use of a chemical flocculant resulted in an increased biomass concentration in the digester compared to a control, thus enabling correspondingly higher space loadings to be applied. With the onset of substantial levels of granulation of the biomass, flocculant dosage was able to be discontinued. This article discusses the performance of the digesters in detail and, briefly, the long-term operational difficulties experienced and the control strategies employed on such systems.

    Notizen: Nickel added in concentrations as low as 10μM significantly increased biogas production in a laboratory poultry waste digester utilizing excreta from laying hens as the organic energy source. It was shown that the initial rate of biogas production increased as early as 4 h after the addition of nickel to the laboratory cultures. Analysis of the excreta for nickel content prior to addition of exogenous NiCl2 showed appreciable amounts of nickel present. The data indicate that nickel naturally present in layer excreta is suboptimal or unavailable to the bacteria for biogas production purposes.

    Notizen: A simple procedure for the entrapment of enzymes in nylon-reinforced cellulose triacetate fibers and the fabrication of reusable enzyme brushes using such fiber bristles for the analytical determination of unknown samples have been described. As an example, enzyme brushes of lactate dehydrogenase were fabricated and were repeatedly employed for the determination of pyruvic acid in clinical samples in place of soluble enzymes. The above brush method compared well with that of wet enzymic analysis of pyruvate in clinical samples in accuracy, sensitivity, and reproducibility. The device also proved to be easy to handle, use, and reprocess for repeated analyses.

    Notizen: By means of stimulus-response experiments an Li+ tracer, models for the fluid flow in a 30-m3 UASB reactor, used for the anaerobic treatment of wastewater, were tested. From the model with the best fit it could be derived that both the sludge bed and the sludge blanket can be described as perfectly mixed tank reactors with short-circuiting flows; the settler volume acts like a plug-flow region.Apart from the volumes of the different flow regions, two parameters are necessary and sufficient to describe the fluid flow in a well functioning UASB reactor, i.e., the short-circuiting flow over the sludge bed and the short-circuiting flow over the sludge blanket. The volumes could be measured accurately.The short-circuiting flow over the sludge bed is a linear function of the sludge bed height. When the optimal height of the sludge bed is defined as the height for which the short-circuiting flows are as small as possible, a bed-height of 3.5-4 m is sufficient (for superficial gas velocities between 1 and 1.5 m/h). This is in contradiction to the results of other authors. The short-circuiting flows over the sludge bed and the sludge blanket were also influenced by the superficial gas velocity.

    Notizen: Tests on acid phosphatase (E.G. 3.1.3.2) deactivation by urea have been performed at two pH values. Two conditions have been used: native enzyme operating batch-wise in dilute solution and stabilized enzyme in continuous flow ultrafiltration membrane reactor. Stabilization is achieved by confining the enzyme within a concentrated solution of a linear chain polymer that forms a polarization layer over the membrane. The results provide significant information on the kinetics and thermodynamics of the complex phenomena taking place during deactivation. Deactivation by urea is also compared with thermal deactivation.

The reactors were monitored at three different temperatures: 10, 20, and 30°C. During the course of digestion, significant solubilization of volatile suspended solids was observed, and its effect on the magnitude of kinetic coefficients was examined. Differences in metabolic activity and sludge stabilization were found between batch and semicontinuous flow patterns. An Arrhenius-type relationship was not found to apply to rate constants for the semicontinuous reactors.

Data concerning the physical characterization of the calorimeter as a fermentor, including stability and sensitivity of the heat signal, are presented. A direct correlation between growth and heat evolution was obtained. Quantitative analysis of the data suggests that the new calorimetric technique could be used for monitoring growth and specific metabolic events, for convenient medium optimization, and as a basis for a novel fermentation process control system.

For comparison, the influence of hydroxyurea on suspended S. cerevisiae has also been investigated. Recovery from DNA synthesis inhibition, indicated by measurements of cell growth rate, DNA content, and light scatter properties, occurred faster in immobilized cells than in the suspended yeast. Morphogenesis in both populations was arrested by hydroxyurea, and there was an accumulation of single immobilized and suspended cells with large buds. Synthesis of protein and RNA was not adversely affected in either cell type. The specific rate of ethanol production by immobilized cells increased by an average of 24%, while, for the suspended cells, specific ethanol productivity was up to three times higher. Glucose consumption rates for both cell types also increased under the influence of hydroxyurea. Immobilized cell ethanol yields were reduced by ca. 16% in the presence of hydroxyurea; suspended cell yields were lower by an average of 50%. Total polysaccharide content was reduced by 65% for suspended cells and increased 30% for immobilized cells after hydroxyurea treatment.     Notizen: A carrier-supported mycelial growth of Penicillium chrysogenum was applied to penicillin fermentation system using celite as a support material. Hyphal growth through the pore matrices of the material showed strong anchorages and provided highly stable biofilm growth. With bioparticles developed in such a manner, both cell growth and penicillin production were observed to increase significantly compared to the conventional dispersed filamentous cultures. Maximum values of specific penicillin production rate were found to be constant regardless of the growth form. A three-phase fluidized-bed fermentor was designed and tested for penicillin production using the bioparticles. Two modes of operation, semicontinuous and repeated fed batch, of the fermentor were tried. It was noted that the overgrowth of free mycelia and the development of fluffy loose bioparticles caused poor mixing and made the fermentor operation quite difficult. Control of the bioparticle size and the extension of production phase were therefore considered important to maintain the reactor productivity at a desired level. From the results of repeated fed-batch operation it was found that the control of bioparticle size could be successfully achieved by phosphate-limiting culture condition. Penicillin production under this condition was also observed to be maintained at a high level (about 80% of the maximum) for at least 1 month.

    Notizen: The effects of pretreating wheat straw with gamma-ray irradiation, ammonium hydroxide, and sodium hydroxide on methane yield, fermentation rate constant, and loss of feedstock constituents were evaluated using laboratory-scale batch fermentors. Results showed that methane yield increased as pretreatment alkali concentration increased, with the highest yield being 37% over untreated straw for the pretreatment consisting of sodium hydroxide dosage of 34 g OH-/kg volatile solids, at 90°C for 1 h. Gamma-ray irradiation had no significant effect on methane yield. Alkaline pretreatment temperatures above 100°C caused a decrease in methane yield. After more than 100 days of fermentation, all of the hemi-cellulose and more than 80% of the cellulose were degraded. The loss in cellulose and hemicellulose accounted for 100% of the volatile solids lost. No consistent effect of pretreatments on batch fermentation rates was noted. Semicontinuous fermentations of straw-manure mixtures confirmed the relative effectiveness of sodium and ammonium-hydroxide pretreatments.

Passage of current through the medium reduced the bacterially generated ferric iron to the ferrous iron substrate. When used in conjunction with an inoculum that had been adapted to the electrolytic growth conditions, this technique increased the protein (cell) concentration by 3.7 times, increased the protein (cell) production rate by 6.5 times, increased the yield coefficient (cellular efficiency) by 8.0 times, and increased the ferrous iron oxidation rate by 1.5 times at 29°C, compared with conventional cultivation techniques. A Monod-type equation with accepted values for the maximum specific growth rate could not account for the increased growth rate under electrolytic conditions.

    Notizen: Model virus inactivation studies with lipid solvents were carried our in antihemophilic factor concentrates. The procoagulant activity obtained was ≥80% recovery with 20% amyl acetate-0.1% deoxycholate. A concurrent reduction of four logs of virus titer was obtained for model viruses provided the viral mass contained significant amounts (≥20%) of lipid. From this preliminary study it appears that further investigations in animal models may be warranted to demonstrate the inactivation of hepatitis B virus, non-A-non-B virus, and AIDS virus with 20% amyl acetate-0.1% deoxycholate in antihemophilic factor concentrates.

    Notizen: Two simultaneous fermentations were performed at 26°C with simultaneous inocula using Clostridium saccharoperbutylacetonicum. Fermentation 1 prevented the gas formed by the biomass from escaping the fermentor while 2 allowed the gas formed to escape. Fermentor 1 provided for the production of butanol, acetone, and ethanol, while when the H2 formed was allowed to escape with fermentor 2, neither butanol nor acetone were produced. Ethanol was also formed in both fermentors and began along with the initial growth of biomass and continued until the fermentations were complete. Butanol and acetone production began after biomass growth had reached a maximum and began to subside. The butanol-acetone-ethanol millimolar yields and ratios were 38:1:14 respectively. The fermentor 2 results show that a yield of 2.1 L H2, 93 or 370 mmol H2/mol glucose, was formed only during the growing stage of growth; neither butanol nor acetone were produced; ethanol was formed throughout the fermentation, reaching a yield of 15.2 mmolar. It appears that hydrogen gas is required for butanol production during the resting stage of growth.

    Notizen: A radial flow hollow fiber bioreactor has been developed that maximizes the utilization of fiber surface for cell growth while eliminating nutrient and metabolic gradients inherent in conventional hollow fiber cartridges. The reactor consists of a central flow distributor tube surrounded by an annular bed of hollow fibers. The central flow distributor tube ensures an axially uniform radial convective flow of nutrients across the fiber bed. Cells attach and proliferate on the outer surface of the fibers. The fibers are pretreated with polylysine to facilitate cell attachment and long-term maintenance of tissuelike densities of cell mass. A mixture of air and CO2 is fed through the tube side of the hollow fibers, ensuring direct oxygenation of the cells and maintenance of pH. Spent medium diffuses across the cell layer into the tube side of the fibers and is convected away along with the spent gas stream. The bioreactor was run as a recycle reactor to permit maximum utilization of nutrient medium. A bioreactor with a membrane surface area of 1150 cm2 was developed and H1 cells were grown to a density of 7.3 × 106 cells/cm2.

    Notizen: Nocardia corallina B276 cells were immobilized by emulsification with liquid paraffin and an antifoam agent at room temperature. The immobilized cells were studied for their ability to carry out the formation of propylene oxide from propylene and oxygen. The evaluations were done with the cells in a bubble-type reactor with a continuous gas feed of 5% propylene and 11.6-95% oxygen, with the balance nitrogen. By using liquid paraffin and antifoam, both the epoxidation activity and the stability were improved, especially for the P-1-200 strain, over that for nonimmobilized cells. The N. corallina cells showed an apparent preference for a hydrophobic, as compared to a hydrophilic, environment. The propylene-oxide-forming activity of the immobilized cells was higher at 40 than at 30°C reactor temperature and with 20% (versus 95%) oxygen in the feed. The stability was markedly better at 30°C and with 20% oxygen. High gas flowrates gave increased apparent activity probably because of less resistance to substrate mass transfer. The effects of pH were minor. The role of glucose as the energy source for regeneration of cofactors for the monooxygenase system also is discussed.

    Notizen: Crystalline insulin was obtained from the pancreas by the method of ultrasound extraction. Extreme changes of pancreas structure were noticed, including rupture of the cells of pancreatic islands and decomposition of pancreatic tissue with the formation of micro-and macro-canals. These changes facilitate the migration of solvent inside the structure and washing out β cell content. Structural changes were maximal in the peripheral parts of the pancreas and in frozen pancreas treated by ultrasound. A rapid filming method recorded factors influencing the extraction process - the dispersing effect of ultrasound, the rapid destruction of pancreatic particles, and the swelling and intensive pulsation of small particles in the ultrasound field. Experimental, control, and standard insulin preparations were nearly identical in Physicochemical properties and ether quality characteristics: activity determined by a biological method and electrophoresis; homogeneity and purity determined by circle chromatography; fractions of insulin determined by starch gel electrophoresis; amino acid composition; solubility; and content of moisture, zinc, and nitrogen.

    Notizen: The ligninolytic activities of four cellulolytic organisms were compared using straw. Only Aspergillus japonicus and Polyporous versicolor appreciably degraded lignin with A. japonicus yielding the most protein. In solid culture, most protein was produced by P. versicolor, closely followed by A. japonicus. Pretreatment of the straw by hot water facilitated biodegradation and protein production. The nutritional value of the residual straw was also increased by some fungal cultures. The greatest amount of degradable polysaccharide in the straw was made available by A. japonicus in liquid media and Pleurotus ostreatus in solid media.

    Notizen: Two continuous stirred tanks reactors (CSTR) and four anaerobic fluidized bed reactors (AFBR) were used to study the treatment of a synthetic meat waste during single-and two-stage anaerobic treatment. Four configurations were investigated; a single-stage CSTR and AFBR and the two-stage systems CSTR-AFBR and AFBR-AFBR. Startup of the anaerobic reactors was achieved within 50 days by use of a regime that included stepped increases in influent COD, methanol substitution of the substrate, and addition of essential trace metals such as cobalt and nickel. Two-stage reactors removed up to 85% of influent COD concentrations of 5000 mg/L, whereas the single-stage AFBR and CSTR removed 76 and 9%, respectively. The proportion of methane in the effluent gases increased as the influent COD concentration was increased. Volumetric production of methane was greatest for the first stage of the AFBR-AFBR system. Solids retention times calculated for the AFBRs ranged from 7 to 12 days, sufficient to support methanogenesis. The AFBRs and two-stage systems were more resistant to an influent pH shock from the operating value of pH 6.8 down to pH 3 than the CSTRs and single-stage reactors. It was concluded that high-rate anaerobic treatment systems were applicable to meat industry wastewaters and that two-stage digestion produced a better quality effluent.

    Notizen: Horseradish peroxidase has been found to vigorously act as a catalyst in a number of water-immiscible organic solvents. The rates of peroxidase-catalyzed oxidation of p-anisidine with H2O2 in toluene, benzene, ethyl and butyl acetates, and ether are in the range of 10-25% of that in water (pH 7.0) at the same reactant concentrations. Per oxidase was coupled with cholesterol oxidase (which was also found to be catalytically active in organic media), and the bienzymic system was successfully used for accurate, reliable, and reproducible determination of cholesterol in toluene.

The device developed was sterilizable by steam and permitted drastic cleaning of the ultrafiltration (UF) membrane without interrupting the continuous fermentation.     Notizen: Perstraction (membrane-aided solvent extraction) was utilized for elimination of ethanol inhibition in continuous ethanol fermentation using high sugar concentrate. Hollow fibers for an artificial kidney were used as a permeable membrane, and their capacity to extract ethanol was examined by using several organic solvents. When tri-n-butylphosphate was used as an extractant, a 500 g/L feed glucose medium was successfully fermented by immobilized yeast cells. During this continuous fermentation a high ehtanol productivity of 48 g/h-L-gel was held, and the solvent requirement per consumed glucose was 6 L-solvent/kg-glucose.

    Notizen: The inhibition of the maximum specific growth and fermentation rate of Zymomonas mobilis by ethanol was studied in turbidostat cultures at constant and stepwise changed ethanol concentrations. Up to 50 g/L ethanol, the inhibition kinetics can be approximated by a linear relationship between the specific growth rate and the ethanol concentration. Above this level, deviations from this linearity are observed. The specific fermentation rates were less inhibited by ethanol than was the specific growth rate. The maximum ethanol concentration achieved was 72 g/L.The response time for the adaptation of a turbidstat culture to step changes in the ethanol concentration was markedly dependent on the concentration level, the response time being large at high ethanol concentrations.

Together with the assumption that the substrate for the anaerobic bacteria is formed by nonionized, volatile fatty acids, the Andrews model is able to describe substrate inhibition and reactor failure due to pH changes.From four batch experiments, with different concentrations of microorganisms, it could be concluded with a reliability of over 95% that the monod model was inadequate and Andrews' model was adequate to describe the measurements. Standard statistical techniques like the X2- and the F-test were used for this purpose.From a parameter sensitivity analysis for the Andrews model it followed that the maximum specific growth rate μAmax of the bacteria and the inhibition constant K1 are the parameters which influence the system most. Thus, these parameter were determined experimentally and most accurately. From calculation of the Thiele modulus for the particles it follows that transport limitation of the substrate in the flocus is not significant. The efficiency η is 0.85 in the worst case.
