    Notes: An apparatus is described for the aeration of a series of tissue cultures at rates which may be varied independently for each vessel. The composition of the aerating gas can be adjusted to any desired percentages of CO2 and O2. While designed particularly for study of the aeration requirements of submerged tissue growth, the equipment is equally adaptable for study of submerged growth of bacterial and other systems. The apparatus has been designed for future scale-up and is adaptable to both the overlay and bubble-through types of aeration. Flow rates necessary for producing mixtures of 5.0% CO2 and 95.0% air or 5.6% CO2, 14.0% O2, and 80.4% nitrogen are given. The described system has been operated satisfactorily and has proved to be an inexpensive and useful research tool.

    Notes: The rate of oxygen absorption by aqueous sodium sulphite solutions in tower-type fermentors up to 6 in. in diameter was related both to tower diameter and to the number of sintered Pyrex disks used for dispersing air entering the towers. Maximal rates of absorption were obtained in towers 2.5 in. in diameter. Under comparable conditions, rates of oxygen transfer in towers containing one aeration disk were approximately 30% higher than those in similar towers containing two or more disks. In towers greater than 6 in. in diameter, the absorption rate was independent of vessel diameter and number of aeration disks when more than one disk was used. Bubble coalescence was the main cause of the difference in the rate of oxygen absorption.

    Notes: The authors describe continuous cultivations of Escherichia coli possessing high penicillin-acylase activity in corn-steep liquor, peptone, and ammonium phenylacetate containing nutrient medium. If the cultivation is performed in the absence of ammonium phenylacetate the enzymic activities of cells from the batch as well as the continuous cultures are very low. The enzymic activity of cells in the batch process was considerably increased by the addition of 0.015% ammonium phenylacetate to the nutrient medium. Further increase of ammonium phenylacetate concentration did not result in any further increase of acylase activity. Continuous cultivation of the bacteria at the above ammonium phenyl-acetate concentration was unsuccessful, as the enzymic activity of the bacteria constantly decreased during the process. On increasing the concentration of ammonium phenylacetate in the medium to 0.15% the authors succeeded in maintaining the enzymic activity of the bacteria at the same level as in the batch process performed at 0.015% concentration, throughout the whole continuous cultivation. At the dilution rate D = 0.5 hr.-1 the concentration of cells in the culture effluent from the fermentor at the steady state was equal to cell concentration at the end of batch cultivation. In relation to the cultivation time the output of cells in continuous cultivation is almost seven times higher as compared with the batch process.

    Notes: These experiments show that ABS can be adsorbed and desorbed from microbial cell surfaces. The adsorption equilibria between the free and adsorbed phases are primarily influenced by the pH of the system. The effects of ABS upon the biochemical activities of bacteria depended upon the amount of ABS adsorbed and the type of bacteria present. The behaviour of ABS is essentially the same whether in the presence of pure or mixed bacterial cultures. Two desorption experiments with soil from a reclaimed water spreading basin which has received sewage over a six month period showed that at least 82% of the ABS “loss” was recoverable from the soil. In all the experiments carried out in this study no evidence was obtained for the biochemical degradation of ABS.

    Notes: With the emphasis being given to the development of biologically soft detergents, it is necessary to establish eriteria defining biodegradability of these materials. It is the authors' opinion that a biologically soft detergent should have the following characteristics: (1) Under normal operations the material should be 85% removable by activated sludge. (2) Normally high discharges of the detergent should not influence biological activity. (3) The detergent should ultimately degrade to carbon dioxide and water.

    Notes: Because branched-chain alkyl benzene sulfonate (ABS) is well known to create water pollution problems in surface water and ground water, a study was made on the biodegradation of this surfactant along with other anionic and nonionic surfactants in detergents by water bacteria. Biodegradation was evaluated by the conventional river die-away test employing the methylene-blue method or surface-tension techniques as analytical tools. Under anaerobic conditions, sugar detergents (sucrose esters) seem to be an ultimate solution to water problems created by branched-chain ABS; in aerated water, straight-chain ABS, sulfo tallow methyl esters, and nonionic surfactants are preferable to branched-chain ABS to improve water problems.

    Notes: Shock pressure waves with a frequency of 50/sec. and a maximal pressure of 60-70 kg./cm.2 greatly reduced the viability of E. coli. A similar effect was obtained also on staphylococci, enterococci, Salmonella Zanzibar, and coliphages T2. On the contrary no effect on spores of B. subtilis was found. The effect was practically independent of the maximal pressure within the range from 30-80 kg./cm.2, the highest pressure tested. The effect decreased with increasing cell density of the bacterial suspension. Bacteria surviving shock treatment died considerably more rapidly when stored at 4°C. than bacteria treated in the same way except for exposure to pressure waves. Morphological examinations with the aid of ordinary light microscopy as well as electron microscopy suggest that the effect was due to mechanical action on the bacterial cell walls.

    Notes: Sphaerotilus is usually present in biological waste treatment processes and is considered to be a nuisance organism because it may cause bulking of activated sludge. However, it may play an important role in the decomposition of putrescible organic matter in properly operated biological waste treatment processes. The research described herein is a study of the variety of types of organic compounds which may be decomposed by Sphaerotilus and the effect of some environmental factors upon the rate of decomposition. Although there are some types of compounds which cannot be decomposed by Sphaerotilus, it is shown that this organism is able to assimilate and oxidize a large variety of organic compounds and that it is tolerant of an extremely wide range of environmental conditions. The role of Sphaerotilus in biological waste treatment is discussed in the light of the data presented.

    Notes: It has been found that a bacterium (isolant C12B) grew poorly on 6 and 8 carbon alcohols, well on 10 and 12 carbon alcohols, and slowly but well on the 14 to 18 carbon compounds. Unexpectedly, resting cells oxidized the 6 and 8 carbon compounds most readily and cell-free extracts oxidized 6, 8, and 10 carbon alcohols. Extracts from cells adapted to sodium lauryl sulfate oxidized the 12 carbon alcohol as well, if small concentrations of substrate were used. The bacterium grew on benzene sulfonate and adapted resting cells oxidized that compound. Pure culture degradation studies revealed that isolant C12B used only 11% of tetrapropylene ABS supplied as a sole carbon source, whereas 66% of sodium lauryl sulfate was degraded. Intermediate quantities of kerosene-based alkyl benzene sulfonate and a tallow-derived detergent were utilized. Extracts of another bacterium, TEG-5, grown on tetraethylene glycol, contained enzymes which oxidized polyethylene glycols up to an average molecular weight of 600, but not any larger polymers. This oxidation was visualized by coupling to reduction of ferricyanide.

    Notes: Two sources of oxygen for man in closed-cycle space system environment have been considered in previous studies: (1) photosynthesis using algae, and (2) electrolysis of water. The latter system appears to be the most promising from the standpoints of energy and weight requirements and ability to operate in a zero gravity field. The surplus hydrogen produced by the electrolysis of water may be utilized together with waste carbon dioxide, part of the oxygen, and waste urea by bacteria of the genus Hydrogenomonas to produce cellular protein which might be used as a source of food. A continuous culture system for the propagation of hydrogen-fixing bacteria consists of a baffled borosilicate glass culture vessel provided with an impeller, a reservoir vessel for the culture medium, and an overflow vessel for collecting the bacterial cells removed from the culture vessel. Complete feedback control of all parameters affecting growth can be provided by hydrogen, oxygen, and carbon dioxide sensors, and a pH electrode in the culture medium. In addition, total pressure is monitored. Cell density is controlled in the optimum range by means of a photoelectric cell which dictates the amount of fresh medium to be added and the amount of cells to be removed. Operating data indicate that some of the key parameters are the ratio of hydrogen, oxygen, and carbon dioxide in solution. The harvested bacterial substance is high in protein, which contains all the essential amino acids.

    Notes: Studies were made of oxygen transfer and power in 20-1. novobiocin fermentations with five sizes (23-60% of the tank diameter) of geometrically similar flatblade turbine impellers. The influence of changes in apparent viscosity on oxygen transfer and power was found to be related to impeller diameter. At constant agitator speed the oxygen availability rate (OAR) decreased with increase in apparent viscosity, but the magnitude of the reduction was less with a small impeller than with a large one. At constant power input the small-diameter impeller (23% of tank diameter) gave an OAR about 8 times that of the large one (60% of tank diameter) when tested in viscous beer, whereas with sulfite or yeast all impeller diameters gave the same OAR. Impeller tip velocity correlated with OAR independently of diameter for the five turbine sizes examined. The power required for optimal novobiocin yields increased with increase in turbine diameter, but the yields were independent of diameter at equal OAR.

    Notes: A laboratory bench set-up for the continuous chemical sterilization of microbiological media using β-propiolactone was developed. The apparatus permits the sterilization of up to 4 liters/hr. of liquid media. The parameters of hydrolyzing time and temperature necessary to reduce the β-propiolactone to nontoxic levels were determined using a E. coli-nutrient broth system. The sterilization technique was applied successfully to a variety of media, and its potential usefulness on an industrial scale is discussed.

    Notes: Dissolved oxygen levels in fermentation systems are affected by (a) gas flow rate, (b) degree of agitation, and (c) oxygen partial pressure in the gas. Since the first two variables cannot easily be separated, a method for dissolved oxygen control has been devised based on the variation of oxygen partial pressure in the gas. Oxygen and nitrogen are fed to a special mixing valve capable of varying the proportions of the two gases while keeping the total flow rate constant. A dissolved oxygen electrode and control system adjust the valve setting to automatically maintain a preset oxygen level. Oxygen analyzers, monitoring the inlet and outlet gas streams, provide additional information from which oxygen absorption and uptake rates may be determined. The system provides a means for the quantitative and independent study of aeration and agitation effects. In addition, it offers a basis for routine control of oxygen levels in plant fermentation equipment.

    Notes: A growth model for Claviceps purpurea in submerged batch culture is presented. In developing the model, the basic principles of the growth and the morphological properties of C. purpurea are considered. The growth of C. purpurea is assumed to occur in a three-step manner; the first step involves the assimilation and the growth of cells; the second one involves cell division, and the third one involves transformation of the mature cells to a state where they have no ability to divide but do have the ability to produce ergot alkaloids and then they gradually die. Inorganic phosphate is assumed to be the limiting substrate for the first and the second steps in conditions of carbon source being in excess. The model constants are determined by model simulation and graphical searching techniques to find the minimum value of the absolute difference between the experimental and the simulated curves for biomass, alkaloids, and sucrose.

    Notes: β-Galactosidase and other enzymes were immobilized on p-amino-carbanilated derivatives of cellulose and methylol cellulose using the diazo method and through glutaraldehyde. The optimum conditions for coupling cellulose tri-(p-amino-carbanilate) (CTAC) to β-galactosidase were established. The diazo coupling method with CTAC gave greater activity than with glutaraldehyde when coupled to β-galactosidase (Escherichia coli). The stability of the CTAC-β-galactosidase system was examined. The disubstituted p-amino-carbanilate derivative (CDAC) gave a lower activity, whereas the methylol analog (MCTAC) gave slightly greater activity. The CTAC was also used to immobilize glucose oxidase, trypsin, pepsin, and papain.

    Notes: The hydrophobic interaction of β-galactosidase with Sepharose 4B substituted with 3,3′-diaminodipropylamine was studied in both batch and column experiments. The equilibrium and the binding rate constants were determined for different phosphate buffer concentrations. The equilibrium constants exhibit a hysteresis effect, i.e., desorption constants are less than adsorption constants, and the higher the ionic strength to start the desorption, the larger the effect. The rate data are not satisfactorily described by a simple reversible first-order model. The column chromatographic data are semiquantitatively described by a local equilibrium theory without axial dispersion or intraparticle diffusion.

    Notes: Aspergillus awamori NRRL 4869 was cultured on the solid substrate, wheat bran, in a modified Rollacell apparatus to produce α-galactosidase and invertase. The swivel cap on the elongated bottle permits the introduction of air while the bottle rotates. Parameters of air flow rate (0.05-0.2 liter/kg/min), rpm (0.15-15 rpm), and weight of solids (150 and 300 g) were varied. At low air flow rates (0.05 liter/kg solid/min), α-galactosidase production was minimal independent of the rotation rate. At 0.15 rpm and 0.2 liter/kg solids/min air flow rate, invertase production ceased after five days; whereas α-galactosidase production continued. The modified Rollacell can be a useful apparatus for studying solid-substrate cultures.

    Notes: The hemicellulose fraction of ryegrass straw was extracted with NaOH and used for the production of glucose isomerase by Streptomyces flavogriseus. The level of hemicellulose extracted increased proportionately with increasing NaOH concentration up to about 4%, then the rate of increase slowed down. Hemicellulose extraction was facilitated by the combined application of heat and NaOH. Approximately 15% hemicellulose (12% as pentosan) could be obtained by treating straw with 4% NaOH for either 3 hr at 90°C or 24 hr at room temperature. The highest level (3.04 units/ml culture) of intracellular glucose isomerase was obtained when the organism was grown at 30°C for two days on 2% straw hemicellulose. The organism also produced a high yield of glucose isomerase on xylose or xylan. The NaOH-treated straw residue, after removal of hemicellulose, had approximately 75% higher digestibility and 20% higher feed efficiency for weanling meadow voles than untreated straw. Thus, the residue could be used as animal feed. A process for the production of glucose isomerase and animal feed from ryegrass straw was also proposed.

    Notes: In a system where yeast cells grow on n-alkanes dissolved in oil drops suspended in water, the dispersed oil phase will, in most cases, be fully segregated. This means that each drop has its own history that depends on its degree of saturation with yeast cells. This degree of saturation with yeast cells is determined by a stochastic process depending on adsorption, desorption, and cell production. Although many authors mention segregation as a phenomenon likely to occur, so far this segregation has hardly been taken into account. In this paper the interaction of the population of completely segregated oil drops with the population of yeast cells, which results in growth, is described. The consequences of the model are elucidated by the discussion of some extreme cases. The batch fermentation of hydrocarbons by yeast cell is simulated by means of a Monte Carlo procedure. Adsorption, desorption, and production of yeast cells are considered as chance processes. The history of all individual drops is recorder. The influence of the chance of desorption appears to be much larger than that of the chance of adsorption (at the investigated range). Also the size of the inoculum at the start of the process appears to have a strong influence on the course of fermentation.

    Notes: Whole cells of Bacillus sp., a bacitracin-producing bacteria, were immobilized in polyacrylamide gel. The continuous production of bacitracin by an immobilized whole-cell-containing air-bubbled reactor was examined with 0.5% peptone solution. The bacitracin productivity (28 units/ml/hr) obtained with this system was higher than that with a batch system. The effluent bacitracin concentration increased with increasing aeration rate and reached a steady-state maximum above the aeration rate of 3.0 liter/min. A high bacitracin productivity was retained for at least eight days when the gel was washed with sterilized saline at a flow rate of 250 ml/hr for 2 hr once a day. The half-life of the immobilized whole-cell system was about 10 days. Bacitracin productivity by the immobilized whole-cell reactor was higher than that by a conventional continuous fermentation process at high dilution rates.

    Notes: Choline oxidase (choline: oxygen oxidoreductaserpar; was immobilized on a partially aminated polyacrylonitrile membrane. The enzyme electrode, consisting of an immobilized-enzyme membrane and an oxygen probe, was employed for the determination choline. Dissolved oxygen consumption by the enzymatic reaction was measured amperometrically. The rate assay method was used for the choline determination. The response time of the sensor was 7 sec for choline. The choline assay was done within 1 min. The choline calibration curve was linear from 0 to 0.1mM. The response was reproducible within an average relative error of 2.3% when 0.2mM choline was employed for experiments. The choline in the fermentation media was determined by the sensor. Furthermore, phospholipids in the serum were also determined with native phospholiphase D and the enzyme electrode.

    Notes: A theoretical method for predicting the elution profile of a pulse response in an immobilized-enzyme column is proposed. The method is based on a mass balance model, which is extensively used in gel chromatography. To test the method, a pulse of sucrose solution was applied to a column of spherical acrylamide gel in which was entrapped invertase from yeast, and it was eluted with 0.05M acetate buffer at pH 5.0. The elution curves of the substrate and the product were in fairly good agreement with the theoretically calculated ones. The method was extended to the system of reversible as well consecutive reactions.

    Notes: The shear viscosities of xanthan batch fermentation broths obey a power-law relation τ = K (P)[ẏ]N(P), where the power-law parameters are functions of xanthan concentration P. within the shear rate range of 1.15 to 384.0 sec-1. Cell-free solutions prepared from these samples exhibit the same values of K and N as the corresponding cell-bearing broth. Further, in the course of the fermentation time, t, it was found that K(P) and N(P) were related by an equation of the form ln K[P(t)] = ln τ0 - N[P(t)]·ln ẏ0, leading to a dimensionless power-law equation valid over the entire fermentation: (τ/τ0) = (ẏ/ẏ0)N[P(t)]. The resulting power-low from[(τ/τ0) = [ẏ/ẏ0]N(t)] indicates that all related samples will exhibit power-low plots passing through the common point, (τ0γ0). These equations have previously been observed with other polysaccharide solution. They are also shown here to be applicable to the pullulan fermentation data of Leduy et at. The parameter K is related to polymer concentration (P) with the correlation K = A PB, where B = 1.75 ± 0.25 for xanthan solutions. With X(t), S(t), and P(t) available from paper I, the correlations of this paper (hereafter referred to as II) [K(t) = f(P)(t) and N(t) = g(K(t))] provide a predictive description of the shear viscosity evolution of polysaccharide-producing fermentation broths.

    Notes: A Lactobacillus sp. isolated from soil and capable of growing on xylose-containing medium exhibited high glucose isomerase activity. The enzyme was thermostable, stable toward dialysis, and activated by heat treatment. It did not show the presence of xylose or ribose isomerase activities; the Km for glucose and xylose substrates were 0.48M and 0.513M, respectively. The heat treatment of ultrasonic crude extract gave insoluble fixed active glucose isomerase enzyme. The properties of free and immobilized enzyme in heat-fixed whole cells differed in many respects. The optimum temperature for enzyme activity changed from 70 to 85°C, the optimum substrate concentration changed from 1.0M to 2.4M, and the optimum pH from 7.4 to 6.0. Co2+ and Mg2+ ions activated the enzyme when used singly, but in combination they inhibited the enzyme and Mn2+ had no effect on the enzyme. Free and immobilized enzymes, when used in the used in the conversions of corn and bagasse hydrolysates to fructose, gave 58, 25.6%, and 50, 27.6% conversions, respectively. Immobilized enzyme retained a significant activity for more than 30 hr and was able to operate at higher glucose concentrations showing less products inhibition effect as compared to free enzyme. In the batch process it was able to operate for about eight cycles.

    Notes: Continuous production of L-alanine from L-aspartic acid using immobilized Pseudomonas dacunhae was investigated. Pseudomonas dacunhae cells were immobilized with carrageenan gel. The L-aspartate β-decarboxylase activity of immobilized cells was enhanced by incubating the immobilized cells with a solution of 1M ammonium L-aspartate (pH 5.5) containing 0.1mM pyridoxal-5′-phosphate (PLP) at 37°C over 20 hr. The enzyme activity of immobilized cells was 59% that of intact cells. The pH profile of the enzyme reaction was broader in the immobilized cells than in the free cells. The enzyme activity of immobilized cells was maintained through repeated uses when a substrate solution containing 0.1mM PLP was used. Complete conversion of L-aspartate to L-alanine was attained when a solution of 2M ammonium L-aspartate (pH 6.2) containing 0.1mM PLP was passed upward through the immobilized cell column at a retention time of 8 hr at 37°C. Glutaraldehyde treatment of the immobilized cells resulted in a slight decrease of the enzyme activity but a marked increase of the operational stability. The half-life of enzyme activity was 46 days in glutaraldehyde-treated immobilized cells.

Enzymes prepared by this method was immobilized to 10 different carriers, but only when it was bound to alumina with TiCl4 and to alkylamine porous silica with glutaraldehyde were substantial efficiencies and stabilities achieved. With alumina, the variation of activation procedure, amount of β-xylosidase offered, and activation solution composition yielded maximum activities of over 40 U/g with approximately 70% immobilization efficiency. Variation of binding pH and incubation time led to a maximum immobilized activity of 1.3 U/g with 78% immobilization efficiency on silica.

    Notes: Cells form the yeast Hansenula polymorpha (ATCC 26012) were successfully immobilized by entrapment in a polyacrylamide gel. The resulting gel showed high methanol oxidase activity especially after treatment with a detergent (CTAB). The enzymatic properties of the gel-entrapped cell were not very different from that of the soluble enzyme except that no inhibition was observed at high methanol concentration. In continuous reactors, the gel-entrapped cells showed a much higher stability than other enzyme preparations. The inactivation mechanism was investigated and proved to be the oxidation of essential SH group(s) of the methanol oxidase molecule by hydrogen peroxide. Treatment with β-mercaptoethanol prevented inactivation or regenerated activity.

    Notes: Polymethylglutamate (PMG), a synthetic polypeptide, was used as a new carrier to immobilize urease (EC 3.5.1.5) and uricase (EC 1.7.3.3) by the azide method. The enzymes could be immobilized onto PMG in various forms, such as film, fiber, coating on various beads, and a silicon tube. The retained activities of the immobilized enzymes were excellent (more than 95%), therefore it was possible to immobilized almost all activities of the enzymes added in the coupling mixtures. Heat stabilities of the resulting immobilized enzymes were markedly improved, while the optimal pH and Km values remained almost unchanged. The urease immobilized on the PMG-coated glass beads packed in a column, was found to retain its activity more than 80% of the initial value, even after the occasional use for a year. In view of the improved retained activities and stabilities of the immobilized enzymes, PMG may therefore be a very versatile matrix for the immobilized enzymes.

    Notes: Heat conduction solution enable rapid determination of the heats of aerobic and anaerobic metabolism of substrate by microorganisms. Aliquots of 1.0 ml cell suspension, 5 × 109 cell/ml, were mixed with a few dozen nmol substrate contained in 0.5 ml, under a controlled atmosphere of air, O2, or N2. At these substrate concentration, with adapted microorganisms, metabolism and its heat generation are usually complete within 300 to 600 sec. The raw data yield ΔHapp values. The ΔHapp were determined in the range 0.001 to 0.010% substrate, and extrapolated (limit substrate concentration →0), to yield Δ0H̄, the limiting differential molar heat of metabolism. The Δ0H̄ values express the heat generated when there is rapid metabolism but little new growth, minimal contribution by H+ transfer from metabolites, and maintenance of aerobicity or anaerobicity as specified. Escherichiacoli B/5 was used for aerobic and anaerobic combustion of eight sugars. Pseudomonas multivorans, and an Acinetobacter, strain B-1, were used for aerobic metabolism of benzene, toluene, naphthalene, and a methylnaphthalene. The larger heats of combustion of the hydrocarbons enable the use of aqueous solutions of hydrocarbons well below their solubility limits. The quotient Δ0H̄/n (n = atoms carbon/molecule substrate) varies from (-)36 to (-)67 kcal/mol carbon for the sugars. The most reduced sugar yields the largest exothermic heats. The quotient varies from (-)27 to (-)81 kcal/mol carbon for the aromatic hydrocarbons. Comparison of the calorimetric heats of metabolism of those from total aerobic combustion in aquo (where available) give measure of the efficiencies with which the heat contents of the aqueous substrate are used by the bacteria.

    Notes: A flat-bed hollow-fiber cell culture system has been developed which maximizes the utilization of the large fiber surface while diminishing significantly the problems inherent in a cartridge-type reactor. The reactor core consists of a shallow bed of hollow fibers sandwiched between two stainless-steel microporous filter plates through which the media flow is directed normal to the plane of the fiber bed. Reactors with both 930 and 9300 cm2 of fiber surface have been successfully constructed and operated. A variety of cells has been grown in these reactors including SV3T3 cells, baby hamster kidney cells, Vero cells, and rhesus money kidney cells, and cell products such as plasminogen activator and migration inhibition factor (MIF) were produced. This system offers an excellent prototype for scaleup design.

    Notes: Candida Krusei has a optimum growth temperature of 37°C on SASOL ethanol-isopropanol mixture. The organism was unable to grow on isopropanol, but oxidized it partially to acetone in the presence and absence of ethanol. Growth at 40°C in the alcohol mixture was slightly faster than at 30°C over an ethanol concentration range of 0.43 to 3.6% (v/v), although at both temperatures the growth rate declined continuously with increasing concentration. At an ethanol concentration greater than 3.6% (v/v), the mixture was much more inhibitory to growth at 40 and 30°C. The inhibitory effect was due to the ethanol rather than the isopropanol. Metabolites such as acetate, acetaldehyde, and ethyl acetate accumulated in the medium, but the degree of accumulation depended upon the temperature and alcohol mixture concentration. At 40°C, acetaldehyde and acetate accumulated to a greater extent than 30°C on a 4.0% (v/v) synthetic alcohol mixture and this may also cause the greater inhibition at this temperature. The alcohol mixture is unsuitable for single cell protein (SCP) production in batch culture because of the low cell densities observed at all alcohol concentrations.

    Notes: It has been shown both theoretically and experimentally that interphase nitrogen transport may have a significant influence on the rate of interphase oxygen transport, and thereby also on the value of the volumetric mass transfer coefficient of oxygen, kla, determined in mechanically agitated bubble fermentors using the variants of dynamic method presented in the literature. The experiments were carried out in 1M KCI solution at five stirrer frequencies and two gas inlet levels. The gas interchanges were performed either without interrupting the aeration and agitation of the charge (A) or with the aeration and agitation of the charge turned on at the same time (B). The applied variants of the interchange were N2→ O2→, O2→ N2, N2→ air, air→ N2, O→ O2, and O→ air. In the two last variants the oxygen dissolved in the charge was removed by reacting with sulfite ions. The kla values calculated by allowing for the nitrogen transport for procedure A were approximately equal to the values obtained by disregarding the nitrogen transport, whereas those for procedure B were higher (up to 40%), than the values obtained disregarding the nitrogen transport.

    Notes: The effect of partial recycle of microbial cells on the operation of a chemostat has been investigated for two fermentations. Stable steady states without partial cell recycle were obtained for the conversion of D-sorbitol to L-sorbose by Gluconobacter oxydans subsp. suboxydans 1916B and for the conversion of glucose to 2-ketogluconic acid by Serratia marcescens NRRL B-486. The employment of partial cell recycle dramatically increased product formation rates for both fermentations.

    Notes: Assimilation as well as dissimilation of methanol in yeasts takes place through its oxidative intermediate formaldehyde which is several times more toxic to the growth of microorganisms than methanol itself. Still, the role of formaldehyde, produced during methanol assimilation, upon growth of yeasts is not clear. In the present paper, an attempt has been made to throw some light upon this aspect. Starting with a basic frame work for methanol uptake by yeasts, several models were developed assuming different modes of regulation of key enzymes by methanol and/or formaldehyde. The main feature of the basic framework consists in consideration of two routes for oxidation of formaldehyde to CO2, one associated and the other not associated with production of energy. Further, the rate of energy production form the energy-associated oxidation of formaldehyde is assumed to be controlled by the rate of energy consumption by anabolic reactions. The models were discriminated by subjecting these to biological constraints. As a result, the successful model suggests that in spite of higher inherent toxicity of formaldehyde, methanol exerts the controlling influence upon growth under normal conditions.

    Notes: Selected samples of waste microbial biomass originating from various industrial fermentation processes and biological treatment plants have been screened for biosorbent properties in conjunction with uranium and thorium in aqueous solutions. Biosorption isotherms have been used for the evaluation of biosorptive uptake capacity of the biomass which was also compared to an activated carbon and the ion exchange resin currently used in uranium production processes. Determined uranium and thorium biosorption isotherms were independent of the initial U or Th solution concentration. Solution pH affected the exhibited uptake. In general, lower biosorptive uptake was exhibited at pH 2 than at pH 4. No discernible difference in uptake was observed between pH 4 and pH 5 where the optimum pH for biosorption lies. The biomass of Rhizopus arrhizus at pH 4 exhibited the highest uranium and thorium biosorptive uptake capacity (g) in excess of 180 mg/g. At an equilibrium uranium concentration of 30 mg/liter, R. arrhizus removed approximately 2.5 and 3.3 times more uranium than the ion exchange resin and activated carbon, respectively. Under the same conditions, R. arrhizus removed 20 times more thorium than the ion exchange resin and 2.3 times more than the activated carbon. R. arrhizus also exhibited higher uptake and a generally more favorable isotherm for both uranium and thorium than all other biomass types examined.

    Notes: The effect of immobilization with various glass-forming monomers on the stability of PS II activity of spinach chloroplast was investigated. PS II activity (O2 evolution due to the Hill reaction) was reduced very slightly by the addition of monomers including polyethyleneglycol (PEG). Immobilization of chloroplast was done with hydrophobic monomer as well as hydrophilic monomer and activity of immobilized chloroplast increased with decreasing monomer concentration as far as the polymerization was possible. The activity of immobilized chloroplast was very high and it decayed far more gradually with the storage time in comparison with the decay of unimmobilized chloroplast and was retained more than 30 days. The optimum monomer concentration for immobilization was about 10%. Thermostability of chloroplast also increased greatly by immobilization with these monomers, especially hydrophilic monomers.

    Notes: Results of pilot plant studies using a glass airlift fermentation device (55 liter fermentation volume) have proven the relative merits of such a system in the fermentation of a filamentous mold, Monascus purpureus, on 4% (w/w) starch media. The resultant overall yield of cell mass (Yx/s) of 0.38 was an appreciable increase over the 0.32 obtained with a pilot scale stirred tank fermentor previously studied. Power requirements of the airlift fermentor were approximately 50% of those for the mechanically agitated system. The lack of mechanical shear in the airlift system provides a more gentle environment or the cultivation of organisms than does the high degree of shear prevalent in the mechanically agitated vessels. Mass transfer of oxygen to the aqueous phase of the fermentation volume is improved significantly through use of the airlift device. Mass transfer coefficients in the range of 200 reciprocal hr were obtained to approximately 80 reciprocal hr in the stirred tank fermentor.

    Notes: The theoretical dynamic characteristics of an isothermal continuous flow stirred tank enzyme reactor (CFSTER) operating on two substrates are investigated. Under certain conditions multiple steady states are possible; namely, with an enzyme which binds with the two substrates sequentially. The occurrence of multiple steady states is found to be primarily dictated by three dimensionless parameters which incorporate rate law constants. The global stability of certain steady states is examined by numerically solving the transient material balance on the CFSTER. The effect of recycle on the dynamics of an isothermal plug flow enzyme reactor (PFER) is also studied. A general conclusion indicated by this work is that any open isothermal reaction system wherein the reaction rate law passes through a maximum with increasing substrate concentration and where back mixing occurs with exhibit multiple steady-state behavior in some operating range.

    Notes: The homogeneity of a purified ribonuclease from brewers' yeast was determined by velocity sedimentation and polyacrylamide gel electrophoresis techniques. The velocity sedimentation pattern gave a single peak with a Sapp 3.46 and polyacrylamide gel electrophoresis showed one major band. The absorption spectrum of the enzyme showed maximum absorption at 277-278 nm and minimum at 252 nm. The enzyme was relatively stable to extreme pH values and high temperature. Both NaCl and KCl increased the enzyme activity whereas enzyme was inhibited by divalent metal ions. The inhibition of the enzyme was increased in the order of Ca2+ 〉 Mg2+ 〉 Fe2+Cu2+. Chemical modification studies of the enzyme showed that tryptophan residues and disulfide bonds were required for enzyme activity.

    Notes: Three freshwater and one marine algal species were grown under inorganic carbon limitation in laboratory continuous cultures. Comparisons were made between HCO3- alkalinity and bubbled CO2 as carbon sources. HCO3- alkalinity was an excellent source of inorganic carbon below specific pH levels, but chemical precipitation at high pH placed an upper limit on productivity that was far lower than potential light-limiting levels. With bubbled CO2 it was possible to achieve light limitation. The main factor controlling productivity was the mass flux of inorganic carbon added to the culture, which is the product of gas flow rate and influent PCO2 level. Small bubbles were more efficient than large bubbles at low gas flow rates and PCO2 levels, but led to froth flotation of algal cells and concomitant reductions in productivity at high bubble rates. At 1% CO2 productivity was still dependent on mass fluxes of added carbon, but was independent of bubble size. At high bubble rates with 1% CO2 narcosis was evident. Maximum yields occurred at intermediate dilution rates when inorganic carbon was supplied via bubbled gas.

    Notes: A range of cellulosic raw materials in the form of agricultural crop residue was analyzed for chemical composition and assessed for potential yields of sugars through chemical pretreatment and enzymatic hydrolysis of these materials. Corn stover was used as a representative raw material for a preliminary process design and economic assessment of the production of sugars and ethanol. With the process as presently developed, 24 gal ethanol can be obtained per ton of corn stover at a processing cost of about $1.80/gal exclusive of by-product credits. The analysis shows the cost of ethanol to be highly dependent upon: (1) the cost of the biomass, (2) the extent of conversion to glucose, (3) enzyme recovery and production cost, and (4) potential utilization of xylose. Significant cost reduction appears possible through further research in these directions.

    Notes: The cyanobacterium Anabaena cylindrica B629 was suspended in small glass beds and incubated in a gas-tight glass vessel outdoors under a gas atmosphere comprising carbon monoxide (0.2%), acetylene (5%), oxygen (6.5%), and nitrogen. The solution phase initially contained sodium bicarbonate (10mM) at pH 7. Under these conditions the organism continuously produced hydrogen gas for over three weeks. The temperature of the culture was maintained below 30°C and minimum night temperatures were recorded. The vessel was covered by a shadecloth, which reduced the natural illumination by approximately 70%. The system is an alternative to those requiring the strict absence of oxygen and little nitrogen, and requires virtually no attention during the incubation period.

    Notes: The dynamic oxygen electrode method for measuring KLa requires the use of a dynamic process model. Six models from the literature are described and compared with respect to their accuracy and ease of use. It is shown theoretically that for sufficient accuracy KLa should be less than the inverse electrode response time. Experimental measurements demonstrate their application to viscous and nonviscous systems. The liquid diffusion film is shown to cause an important measurement lag that can be accounted for by a first-order time delay. Investigation on the influence of the experimental starting conditions show the importance of the gas and hold-up dynamics. A new method is proposed to simplify the KLa calculation and to eliminate errors caused by starting conditions. This method, which accounts for gas, film, and electrode dynamical effects, requires only a simple semilog plot of the response data.

    Notes: The determination of growth constant of Aspergillus niger were obtained for semisolid cultures on cassava flour, Manihot esculenta, as a sole carbon source. As a consequence, a technique was developed that consisted of the use of a packed-bed microfermentor with a working volume of 16 cm3. The bed consisted of gelatinized and granulated cassava flour containing mineral nutrients and mold spores. The carbon dioxide produced during the respiration was drawn off with a current of air and then absorbed in a solution of sodium hydroxide. The absorption of CO2, P, was correlated with the specific growth rate μ, by means of the equation P=Keμt, where t is time and K is a constant. Ammonium nitrogen was used as a limiting substrate and its concentration was varied from 0.039 to 2.5% in dry base. The maximum growth rate, μmax, and the saturation constant, Ks, were 0.31 hr-1 and 0.065 mmol (NH4)2SO4/g total dry solids. Substrate inhibition was presented and the constant ki gave a value of 1.5 mmol (NH4)2SO4/g total dry solids. The proposed method is highly recommended for the evaluation of the semisolid fermentation of molds and for strictly aerobic bacteria and yeasts. It can be used especially in the evaluation of the growth of microorganisms on peanut shells, coffee residues, sugar cane bagasse, and other agricultural wastes.

    Notes: An enzyme which catalyzes the synthesis of cephalexin fromD-α phenylglycinemethylester (PGM) and 7-amino-3-desacetoxy-cephalosporanic acid (7-ADCA) was prepared from Xanthomonas citri (IFO 3835) and partially purified 30-fold by ammonium sulfate fractionation, DEAE-cellulose, and Sepharose-4B column chromatography. The Km values for 7-ADCA, PGM, and cephalexin were determined as 11.1, 2.1, and 1.61 mM, respectively. The enzymatic cephalexin synthesis follows the reversible bi-uni reaction kinetics. The equilibrium constant is influenced by the initial mole ratios of 7-ADCA and PGM. The cephalexin hydrolysis is catalyzed by the same cephalexin synthesizing enzyme, but methanol does not participate in the hydrolytic reaction. The amount of enzyme in the reaction mixture affects the initial rate but does not influence the equilibrium product concentration. This cephalexin-synthesizing enzyme was immobilized onto several adsorbents. Among these, Kaolin and bentonite showed a higher retention of enzyme activity and stability for reuse. The immobilized-enzyme reaction kinetics were investigated and compared with those of the soluble enzyme. A rate expression for the enzymatic synthesis of cephalexin was derived. The results of computer simulation showed good agreement with the experimental results.

    Notes: This paper describes the scale-up from 0.1 to 100 liter of the unit process based on 3-mm-diameter glass spheres for the growth of BHK monolayer cells. The production of four strains of FMD virus at the 0.1-, 10-, and 100-liter scales was examined. Cell growth was estimated from measurements of the concentration of glucose in the growth medium, while the release of virus was inferred from measuring the concentration of LDH in the culture supernatant fluid. The yields of virus at 0.1-, 1-, and 10-liter scales were similar but that from the 100-liter version was somewhat lower. The reason for this lower yield and the method used to overcome it over outlined.

    Notes: A culture of R. rubrum cells apparently contaminated with K. pneumoniae were immobilized by entrapment in agar. This system was used as model for hydrogen production by photometabolic means. Observed results indicated that the contaminant exerted a major influence on the observed results. This preparation, when immobilized and used in a specifically designed reactor with glucose substrate, showed operational half-lives of approximately 1000 hr. The feasibility of using this “mixed” culture for producing hydrogen from acid hydrolyzed cellulose and wood sawdust was also examined.

    Notes: Nickel-NiO-BSA-chymotrypsin has been used to hydrolyze casein in both batch and continuous stirred tank reactors. The kinetics of hydrolysis in both types of reactor were considered. Some operational problems encountered using fine powder catalysts in batch stirred tank reactors are discussed. High gradient magnetic separation was found to be a powerful catalysts retention method for magnetic support particles, particularly when using a ferromagnetic collection matrix. Nickel particles of diameter 3-7 μm were easily separated from water at a processing velocity of 39 mm/sec. Nickel powder and precipitated Fe3O4 were also separated satisfactorily using a rotating drum magnetic separator.

    Notes: There is now growing evidence that the production of many secondary metabolic by microorganisms is subjected to carbon-catabolite regulation. Even though the exact mode of this regulation is not yet clear, an engineering analysis of the production process is still possible based upon a suitable hypothesis. By way of simulation of penicillin fermentation data obtained from the literature, a mechanistic model involving a substrate inhibition kinetics of product formation has been verified in this paper. Such a model has been found successful not only in predicting simple sugar-feeding strategy, but also a complicated computer guided strategy based upon controlling biomass growth rates in the tropo and idiophases. Using this model, for strategies for sugar feeding into penicillin fermentation have been investigated. These results show that similar penicillin productivities can be obtained using any of these strategies provided fermentations are carried out under optimal conditions corresponding to the strategy chosen. Effect of maximum oxygen transfer capacity of the fermentor under the conditions of fungal growth has been incorporated using an upper limit of biomass concentration on achievement of which the fermentations must be stopped due to serious oxygen limitations. Results of model simulations with such limits throw light upon the way in which different fermentors may behave with respect to product formation.

    Notes: In order to reduce the severe flux losses encountered during ultrafiltration of protein solutions, proteases were immobilized on Ultrafiltration membranes to hydrolyze the deposited solute molecules. Over a standard 22 hr run 25 to 78% improvement in cumulative permeate yield was obtained when processing 0.5% albumin or hemoglobin. It was also demonstrated that the flux enhancements were due to the biochemical action of the absorbed protease and not to its physical effect as a prefilter coat. with the aid of a model retardation of gel formation mechanism was demonstrated. Economics of the system were shown to be favorable, improving the rate of return on capital investment up to 50% by reduction of the total membrane area of the plant.

A system which produces gramicidin S is taken as an example. The single level technique using the conjugate gradient method is applied to solve the two-stage and the three-stage continuous culture and is compared with the multilevel one. The results show that the application of multilevel techniques is more advantageous and suitable for this system than any other method which has been utilized so far. The advantages of using a microprocessor will be stated.

    Notes: A simple model is developed for the energy transformation in growing microbial systems. The model is based on a linear equation for ATP consumption in the processes of growth and maintenance. A combination of this equation with macroscopic balances for the various components, the systems exchanges with the environment, and application of the concepts of the elementary balance allow the derivation of linear equations for the exchange of substrate, oxygen, and carbon dioxide with the environment. For growth on one sole carbon and energy source the model allows the definition of a critical substrate yield are expected and below which is decreasing substrate yield and energy supply growth limitation are expected. This restriction can be interpreted in a variety of other ways. It supplies a rationale for non-energy-production-coupled transfer of hydrogen to oxygen or wasteful expenditure of ATP in growth on highly reduced substrates. It also allows the formulation of a limit to the maximum yield on oxygen that can never be exceeded in growth on highly reduced substrates.

    Notes: Many changes that occur in a cell during the cell cycle can be demonstrated in synchronous cultures and can reveal dimensions of cell metabolism not attainable by the study of balanced growth of asynchronous populations in batch cultures or the steady state in chemostat cultures. The release of 14CO2 from specifically labeled glucose by phased (continuously synchronized) cultures follows a characteristic pattern (profile) that depends upon the stage in the cell cycle and the period of labeling used. Successive profiles throughout a cycle showed differences that were altered under different nutrient-limiting growth conditions. Profiles obtained with glucose-1-14C, glucose-2-14C, glucose-3,4-14C, and glucose-6-14C and phased cells of Candida utilis under N-, P-, and C-limited growth demonstrated the variable character of the metabolic activity that occurred in the cells while contour changes within the profiles across the cycle indicated possible correlations with activities of the hexose monophosphate, Embden-Meyerhof-Parnas, and tricarboxylic acid cycle pathways during the cell cycle. The basis of these changes and their use as elementary parameters for study of problems of physiological changes in vivo are considered.

    Notes: An enzyme electrode is described for quantitative determination of phenol at micromolar concentrations. Immobilized phenol hydroxylase is attached to the surface of a Clark oxygen electrode. The Maximum rate of oxygen consumption is linearly dependent on phenol concentration over the 0.5-50μM range. The electrode can be used for at least 150 assays without an activity loss. Readout is very rapid - within 30 sec of sample addition. The electrode response is independent of pH between pH 6.5 and 9.5. The response increases linearly with temperature in the interval 10-40°C. It is necessary to incubate the enzyme electrode in a buffer containing NADPH for a few minutes before the addition of sample. This is to make the electrode response independent of the diffusion rate of this cosubstrate. This and other diffusional effects on the performance of the phenol electrode are discussed.

    Notes: Enzyme stability studies have been reinvestigated under the conditions used for cellulose hydrolysis (pH 4.8, 50°C, 24 hr). The cellobiohydrolase (CBH) component as measured on Avicel is less stable than other enzymes of the cellulase complex, and is 60% inactivated by merthiolate (and other Hg compounds) under the above conditions. Endo-β-1,4-glucanase is much more stable, and more resistant to merthiolate and other compounds. Under unshaken conditions the Avicelase of the Rutgers strain C 30 shows greater stability to heat than that of other available strains. Biocides must be selected not only for their ability to prevent contamination, but also for their compatibility with cellulases. Tetracycline and chlortetracycline are inexpensive, effective in very low concentrations, have no harmful effect on the enzymes, and are compatible with the yeasts that subsequently grow on the sugar solutions to produce alcohol. Attempts have been made to stabilize the enzymes by chemical modification in such a way as to maintain their solubility. Glutaraldehyde treatment greatly increased the enzyme size, lowered the pI values, and gave a slight shift in the pH activity curve. There was, unfortunately, no increase in enzyme stability, and the activity of enzymes on solid celluloses was adversely affected. Shaking greatly reduced the hydrolysis of Avicel by Trichoderma reesei C 30 enzyme. The adverse effect was accompanied by a decrease in recoverable enzyme and protein.

    Notes: White-rot fungi, which have the ability to degrade all the wood components including lignin, are of great interest in biotechnological processes based on wood and other lignocellulosic materials. It was demonstrated earlier that enough lignin can be degraded to cause a decrease in the energy demand for production of thermomechanical pulp if wood chips are pretreated by cellulaseless mutants of white-rot fungi. This paper concerns the growth conditions in wood for three white-rot fungi and their cellulaseless mutants in order to determine optimal conditions for such pretreatment processes. The pH and temperature optima have been determined as well as the growth rate in wood. The results show that the growth rate in wood. at least for Cel 44 (a cellulaseless mutant of Sporotrichum pulverulentum), is not the rate-limiting step in delignification. From different mixtures of urea and NH4H2PO4 the optimal nitrogen source was determined for the mutants. The optimal C/N ratio was found to vary between 160/1 and 400/1. It is suggested that the lower the C/N ratio, the faster the growth. It was also demonstrated that both water- and acetone-extractable substances in wood supported the growth of cellulaseless mutants. When some glucose was added to the wood, the weight loss caused by Cel 44 increased. All these observations support earlier findings that lignin in wood cannot be degraded by white-rot fungi unless a more easily metabolizable carbon source is used simultaneously.

    Notes: The partition of n-hexadecane in the spent growth medium of Acinetobacter sp. HOI-N was determined by measuring the increase in the relative aqueous solubility of 3H-hexadecane as compared to controls. The amount of hexadecane partitioned was proportional to the protein concentration. The specific solubility of hexadecane (nmol/mg protein) was analyzed by least-squares fitting yielding an average slope of 0.6 with a standard deviation of 0.3, indicating either nonequilibrium of hexadecane or physical aggregation of protein. The amount of hexadecane partitioned was concentration dependent yielding optically clear microemulsions at hexadecane concentrations of less than 1.4mM and macroemulsions at hexadecane concentrations of 1.4mM or greater. Preliminary results indicated that hexadecane and partitioned by a lipoprotein complex.

    Notes: Crude extract from sweet sorghum supplemented with vetch juice was utilized as the carbohydrate source for fermentative production of lactic acid. Fermentation of media containing 7%(w/v) total sugar was complex completed in 60-80 hr by Lactobacillus plantarum, product yield averaging 85%. Maximum acid production rates were dependent on pH, initial substrate distribution, and concentration, the rates varying from 2 to 5 g(liter·hr.) The lactic acid yield was lowered to 67% under limited medium supplementation. The fermented ammoniated product contained over eight times as much equivalent crude protein (N × 6.25) as the original medium. Unstructured kinetic models were developed for cell growth, lactic acid formation, and substrate consumption in batch fermentation. With the provision of experimentally determined kinetic parameters, the proposed models accurately the fermentation process.
