    Beschreibung: In a search for genes that regulate circadian rhythms in mammals, the progeny of mice treated with N-ethyl-N-nitrosourea (ENU) were screened for circadian clock mutations. A semidominant mutation, Clock, that lengthens circadian period and abolishes persistence of rhythmicity was identified. Clock segregated as a single gene that mapped to the midportion of mouse chromosome 5, a region syntenic to human chromosome 4.     Beschreibung: The segment of the malarial circumsporozoite (CS) protein designated Region II is highly conserved among different malarial species. A similar sequence is also present in several other proteins, including thrombospondin, properdin, and a blood-stage antigen of Plasmodium falciparum. By means of peptides synthesized from sequences of the Plasmodium vivax CS protein in the vicinity of Region II, it was found that two overlapping 18- to 20-amino acid peptides promoted the adhesion of a variety of human hematopoietic cell lines.     Beschreibung: An assessment of health effects due to ozone and particulate matter (PM10) suggests that each of the 12 million residents of the South Coast Air Basin of California experiences ozone-related symptoms on an average of up to 17 days each year and faces an increased risk of death in any year of 1/10,000 as a result of elevated PM10 exposure. The estimated annual economic value of avoiding these effects is nearly $10 billion.     Beschreibung: When template DNA is saturated with a single-stranded DNA binding protein (SSB), strings of three or four contiguous hexanucleotides (hexamers) can cooperate through base-stacking interactions to prime DNA synthesis specifically from the 3' end of the string. Under the same conditions, priming by individual hexamers is suppressed. Strings of three of four hexamers representing more than 200 of the 4096 possible hexamers primed easily readable sequence ladders at more than 75 different sites in single-stranded or denatured double-stranded templates 6.4 kilobases to 40 kilobase pairs long, with a success rate of 60 to 90 percent. A synthesis of 1 micromole of hexamer supplies enough material for thousands of primings, so multiple libraries of all 4096 hexamers could be distributed at a reasonable cost. Such libraries would allow rapid and economical sequencing.     Beschreibung: The generation of the orientation component of the polarization of matter in an electric field has previously been thought to require interaction of molecules with their neighbors. It is demonstrated that, even in the absence of collisions between neighboring molecules, hot isolated polyatomic molecules can reorient in response to an external field, thereby giving rise to the orientation component of polarization. This reorientation occurs through the interaction of rotation with molecular vibrations, which provides a heat bath to establish thermal rotational equilibrium.     Beschreibung: Seasonal records of tropical sea-surface temperature (SST) over the past 10(5) years can be recovered from high-precision measurements of coral strontium/calcium ratios with the use of thermal ionization mass spectrometry. The temperature dependence of these ratios was calibrated with corals collected at SST recording stations and by (18)O/(16)O thermometry. The results suggest that mean monthly SST may be determined with an apparent accuracy of better than 0.5 degrees C. Measurements on a fossil coral indicate that 10,200 years ago mean annual SSTs near Vanuatu in the southwestern Pacific Ocean were about 5 degrees C colder than today and that seasonal variations in SST were larger.     Beschreibung: Reactive or unstable molecules are key intermediates in many important reactions, but can be difficult to prepare for experimental studies. Species with missing (:CH-OH) or extra (H3) substituents can often be formed conveniently in the gas phase by neutralizing a beam of a more stable ionic counterpart (CH = O+H, H3+). Reionization of the neutral after approximately 10(-6) seconds tests its stability, whereas its unimolecular chemistry can be probed by preparing it with different amounts of internal energy. The resulting neutral products are reionized and mass analyzed. Isomers are then characterized by ion dissociation and a third mass-analysis step.
