    Description: The CLOCK transcription factor is a key component of the molecular circadian clock within pacemaker neurons of the hypothalamic suprachiasmatic nucleus. We found that homozygous Clock mutant mice have a greatly attenuated diurnal feeding rhythm, are hyperphagic and obese, and develop a metabolic syndrome of hyperleptinemia, hyperlipidemia, hepatic steatosis, hyperglycemia, and hypoinsulinemia. Expression of transcripts encoding selected hypothalamic peptides associated with energy balance was attenuated in the Clock mutant mice.     Description: The length of day, or photoperiod, regulates the annual cycle of reproductive activity in the golden hamster. The inhibitory effects of a short-day photoperiod on testicular function were prevented by nighttime, but not daytime, intraventricular injections of carbachol, a cholinergic agonist. Short pulses of light during the night also block short-day induced testicular regression.     Description: A single daily "surge" in pituitary luteinizing hormone release was observed in ovariectomized-estrogen-treated hamsters expressing an intact circadian rhythm of locomotor activity. In contrast, two luteinizing hormone surges occurred within a single 24-hour period in hamsters whose activity rhythm had dissociated or "split" into two distinct components.     Description: In a search for genes that regulate circadian rhythms in mammals, the progeny of mice treated with N-ethyl-N-nitrosourea (ENU) were screened for circadian clock mutations. A semidominant mutation, Clock, that lengthens circadian period and abolishes persistence of rhythmicity was identified. Clock segregated as a single gene that mapped to the midportion of mouse chromosome 5, a region syntenic to human chromosome 4.     Description: The circadian rhythm of activity in vertebrates often splits into two components after continuous exposure to constant light. This observation suggests that at least two circadian pacemakers underlie the activity rhythm. After unilateral ablation of the hypothalamic suprachiasmatic nuclei in hamsters, the splitting phenomenon was eliminated and a single rhythm of activity was established. The period of the new circadian activity rhythm different from the periods of the split rhythm and that preceding the split.     Notes: Summary Previous studies indicate that steroid hormones alter pineal biochemistry, and it has been suggested that at least part of the negative feedback effect of steroid hormones on pituitary gonadotropin release may be mediated by the pineal gland. In this study, pinealectomy did not alter the inhibitory effect of testosterone on neuroendocrine-gonadal activity in the male rat, suggesting that the pineal gland does not mediate the response of the rat hypothalamic-pituitary axis to testosterone.
