Description: This study was conducted to codify the guideline and indexes of broodstock management in salmonid fish and cryopreservation, selective breeding and sex reversal in hatchery to attain responsible aquaculture development. Cryopreservation includes of sperm and egg in term of economic is important in aquaculture. In this report mentioned the type and name of material. All female and sex reversal fish are produced by steroid hormone which mentioned in this report. Selective breeding include individual selection and family selection which could be used for genetic improvement of fish.

Description: The sex reversal technique in fish is widespread in many countries. The development of these techniques is desirable because rainbow trout males reach their gonad maturity earlier compared to the females. Rainbow trout alevins were treated with 17α-methyltestosterone incorporated in the diet (0.5. 1.5, 3, 6 and 10 mg/kg) for 60 days from the beginning of first feeding. Sex was determined at 180 and 680 dpf by sampling fish (n = 20) from each group and examining gross gonadal morphology under a dissecting microscope. Also sex reversal ratio and growth performance were determined in hormone-treated groups. These sex reversed functional males were reared for brood stock until they attained sexual maturity. At the end of experiment, normal rainbow trout eggs were fertilized with the sperms taken from sex reversed males for producing all-female populations. Examination of the results showed that 17α- methyltestosterone was effective in all treatment. The highest sex reversal ratio with 100% was observed in group treated with 0.5, 1.5 and 3 mg/kg 17α- methyltestosterone. The highest live weight ratios were observed in groups fed with 6, 0.5 17α-methyltestosterone and control group. Female progeny produced from the sex reversed males were 100 % in all males that sired offspring. All female trout stocks produced by this method have advantage in rainbow trout culture since the fish is not meant for direct human consumption and is used to generate brood stock, therefore, difference of growth parameters do not influence the success.

Description: In order to investigate the effect of different temperature of anesthetic (clove tree) and recover (anesthetic-free water) solutions on recovery process, The anesthetic induction times and recovery times of 540 fingerlings of rainbow trout (Oncorhynchus mykiss) weighing 3.2 ± 0.2 g in anesthesia/ recovery solutions with cross-combined of temperature of 5,11 and 17 °C as experimental treatments (9 treatments) were compared. The anesthesia was induced by170 ppm of aqueous clove (Eugenia caryophyllata) solution. Results showed that both induction and recovery times were adversely temperature-dependent. Recovery times of treatments, related to temperature of anesthetic solutions and their temperature, were significantly different (P 〈 0.05). Our results indicated that mean recovery time (three temperature treatments) of the fish exposed to the lower temperature anesthetic solutions generally decreased opposing with higher anesthetic solutions. The lowest recovery time (61±11.1 seconds) in17°C anesthetic-free water occurred in fishes induced to 5°C anesthetic solution. In contrast, the highest recovery time (523±154 seconds) in 5°C water happened in fishes induced to 17°C anesthetic solution. Warm and cold thermal difference in anesthetic and recover solutions could extend recovery time 1 up to 8 minutes. Regarding to aim and demanded time, by selected favorite temperatures can control anesthesia/or recovery time in order to prevent the use of drug over-dosing.

Description: IHNV (Infectious Hematopoietic Necrosis Virus) is one of the most important pathogens in cultured rainbow trout (Oncorhynchus mykiss). The virus causes the disease that affects the hematopoietic tissue and blood vessels of fish, especially in larvae and fingerling stages. The disease is highly contagious and causes mass fish casualty (up to 95%). Therefore, continuous health monitoring of fish farms is necessary. The objectives of the present study were to identify and track the probable presence of the virus in fish farm in Dasht-e-Rome, Kohgiluyeh and Boyer-Ahmad province, Iran. Samples were prepared from the liver, spleen, kidney, egg, sperm and larvae of the adult fish and broodfish during four years from 2012 to 2016. About one percent of the broodstock population was used for sampling (1%×3000 broodfish×4years=120fish). In order to detect the presence of IHN-virus, samples were sent to a laboratory in frozen condition. After preparation of samples, they were inoculated with two types of cell line (BF2 and EPC) and examined by the molecular methods (Reverse Transcriptase-PCR) to detect the presence of IHN virus. The results showed that all samples were free from the IHN virus and the frequency of virus was 0% in all examined samples.

Description: The aim of the present study was to investigate the effect of probioenzyme on the control of yersiniosis as well as to assess the impact of probioenzyme on the expression of immune-related genes in the head kidney of rainbow trout (Oncorhynchus mykiss). Probioenzyme was supplemented at 0.3, and 0.5g/kg diet and fed to fish (average weight: 26 g) in 200l tanks for 60 days. At the end of the feeding period, fish were challenged with yersinia ruckeri. Only the fish fed the diet containing high dose of probioenzyme showed significantly (p〈0.05) improved protection against y. ruckeri compared to the low dose and control groups. Subsequently, real-time PCR was employed to determine the mRNA levels of IL-1β and TNF- α cytokines in the head kidney of the control and probioenzyme groups. High dose of probioenzyme significantly (p〈0.05) up-regulated the TNF- α transcription in the kidney compared with low dose and control groups. Meanwhile, the probioenzyme didn,t change nterleukin-1β mRNA level in the kidney in treatment groups compared with control group (p〉0.05). Only, high dose of probioenzyme noticeably differs IL-1β and TNF-α gene expressions. Based on results, expression levels of studied cytokines were different, dose-dependent and tissue-specific. Results showed that cytokine gene expressions, especially TNF-α, in kidney may affect fish disease resistance. Present study suggests that probioenzyme especially at 0.5g/kg feed may effectively stimulate the immune system and enhance disease resistance in rainbow trout. Meanwhile, it is necessary to study effects of probioenzyme on cytokines in various tissue and different growth period of breeding species.
