    Early cultivators spread to Cyprus 10,600 y ago [Anthropology] (2012)

    Description: Early Neolithic sedentary villagers started cultivating wild cereals in the Near East 11,500 y ago [Pre-Pottery Neolithic A (PPNA)]. Recent discoveries indicated that Cyprus was frequented by Late PPNA people, but the earliest evidence until now for both the use of cereals and Neolithic villages on the island dates to 10,400 y ago. Here we present the recent archaeological excavation at Klimonas, which demonstrates that established villagers were living on Cyprus between 11,100 and 10,600 y ago. Villagers had stone artifacts and buildings (including a remarkable 10-m diameter communal building) that were similar to those found on Late PPNA sites on the mainland. Cereals were introduced from the Levant, and meat was obtained by hunting the only ungulate living on the island, a small indigenous Cypriot wild boar. Cats and small domestic dogs were brought from the mainland. This colonization suggests well-developed maritime capabilities by the PPNA period, but also that migration from the mainland may have occurred shortly after the beginning of agriculture.

    Stone uses in Reims Cathedral: provenance, physical properties and restoration phases (2013)

    Description: Reims Cathedral is a major monument in the NE of France originally built with local Lutetian limestone. The recent closure of the last quarries has made restoration using the same stone more complicated. The restoration stones used currently are Lutetian limestones from the centre of the Paris Basin (Saint-Pierre-Aigle and Saint-Maximin stones). Mapping of the Cathedral's façades confirmed the data from ancient manuscripts: Courville stone was the original building stone, but several other local stones from various quarries and beds were also used. As a follow-up to this mapping, Lutetian limestones from five disused quarries were sampled for petrophysical characterization tests: thin section analyses, porosimetry, capillary and drying kinetics. The petrophysical properties of the limestones showed differences between the two main local stone types ( Ditrupa limestone and miliolids limestone) and also between the quarries. This study addresses the difficulty of selecting new stone for restoration. Should ancient quarries be re-opened? If so, which ones?

    Concordance of gene expression in human protein complexes reveals tissue specificity and pathology (2013)

    Description: Disease-causing variants in human genes usually lead to phenotypes specific to only a few tissues. Here, we present a method for predicting tissue specificity based on quantitative deregulation of protein complexes. The underlying assumption is that the degree of coordinated expression among proteins in a complex within a given tissue may pinpoint tissues that will be affected by a mutation in the complex and coordinated expression may reveal the complex to be active in the tissue. We identified known disease genes and their protein complex partners in a high-quality human interactome. Each susceptibility gene's tissue involvement was ranked based on coordinated expression with its interaction partners in a non-disease global map of human tissue-specific expression. The approach demonstrated high overall area under the curve (0.78) and was very successfully benchmarked against a random model and an approach not using protein complexes. This was illustrated by correct tissue predictions for three case studies on leptin, insulin-like-growth-factor 2 and the inhibitor of NF-B kinase subunit gamma that show high concordant expression in biologically relevant tissues. Our method identifies novel gene-phenotype associations in human diseases and predicts the tissues where associated phenotypic effects may arise.

    Deep human genealogies reveal a selective advantage to be on an expanding wave front (2011)

    Description: Since their origin, human populations have colonized the whole planet, but the demographic processes governing range expansions are mostly unknown. We analyzed the genealogy of more than one million individuals resulting from a range expansion in Quebec between 1686 and 1960 and reconstructed the spatial dynamics of the expansion. We find that a majority of the present Saguenay Lac-Saint-Jean population can be traced back to ancestors having lived directly on or close to the wave front. Ancestors located on the front contributed significantly more to the current gene pool than those from the range core, likely due to a 20% larger effective fertility of women on the wave front.     X-ray structure of the mouse serotonin 5-HT3 receptor (2014)

    Description: Neurotransmitter-gated ion channels of the Cys-loop receptor family mediate fast neurotransmission throughout the nervous system. The molecular processes of neurotransmitter binding, subsequent opening of the ion channel and ion permeation remain poorly understood. Here we present the X-ray structure of a mammalian Cys-loop receptor, the mouse serotonin 5-HT3 receptor, at 3.5 A resolution. The structure of the proteolysed receptor, made up of two fragments and comprising part of the intracellular domain, was determined in complex with stabilizing nanobodies. The extracellular domain reveals the detailed anatomy of the neurotransmitter binding site capped by a nanobody. The membrane domain delimits an aqueous pore with a 4.6 A constriction. In the intracellular domain, a bundle of five intracellular helices creates a closed vestibule where lateral portals are obstructed by loops.     Reconstructing Native American population history (2012)

    Description: The peopling of the Americas has been the subject of extensive genetic, archaeological and linguistic research; however, central questions remain unresolved. One contentious issue is whether the settlement occurred by means of a single migration or multiple streams of migration from Siberia. The pattern of dispersals within the Americas is also poorly understood. To address these questions at a higher resolution than was previously possible, we assembled data from 52 Native American and 17 Siberian groups genotyped at 364,470 single nucleotide polymorphisms. Here we show that Native Americans descend from at least three streams of Asian gene flow. Most descend entirely from a single ancestral population that we call 'First American'. However, speakers of Eskimo-Aleut languages from the Arctic inherit almost half their ancestry from a second stream of Asian gene flow, and the Na-Dene-speaking Chipewyan from Canada inherit roughly one-tenth of their ancestry from a third stream. We show that the initial peopling followed a southward expansion facilitated by the coast, with sequential population splits and little gene flow after divergence, especially in South America.     Notes: Several species of flatfishes (Pleuronectiformes) have been studied by isoenzyme electrophoresis and DNA/DNA hybridization to establish the evolutionary relationships of this group of fishes. This molecular data-based phylogeny has been compared to previously described phylogenies based on morpho-anatomical criteria. A number of discrepancies are observed and discussed. The high degree of genetic divergence observed within the families of Pleuronectiformes indicates that their origin seems to be much older than suggested by their striking morphological similarity.

    Abstract: Zusammenfassung Aus ESR Messungen wird gefolgert, daß das Radikal CH2=Ċ-CN bei tiefer Temperatur in einer instabilen gebogenen Struktur und bei höherer Temperatur in einer stabilen linearen Struktur vorliegt. Theoretische Berechnungen stehen sowohl für dieses als auch für das Butatrienyl-Radikal in guter Übereinstimmung mit experimentellen Daten.
    Notes: Abstract First, it is shown by ESR that the radical CH2=Ċ-CN at low temperature presents an unstable bent structure and at higher temperature a stable linear structure. A theoretical calculation on this radical and on the butatrienyl one is in good agreement with the experimental results.
