    Notes: Water soluble allergens of grass pollen (Dactylis glomerata) were purified by two successive steps of preparative isoelectric focusing in a flat bed of granulated gel and controlled by analytical isoelectric focusing in agarose gels. In order to separate components cofocusing in the same pH zone but having different electrophoretic mobilities, preparative zone electrophoresis in granulated gels was performed. The homogeneity of the fractions recovered was tested by isoelectric focusing and zone electrophoresis, followed by radioimmunodetection of the allergenic molecules on nitrocellulose prints. The major allergen of Dactylis glomerata pollen was purified with a yield of 7.5 mg from 10 g of pollen grains.

    Notes: Hair proteins have been successfully analysed by high resolution two-dimensional electrophoresis by avoiding S-carboxymethylation which has previously dictated the use of alternative two-dimensional procedures. The high and low-sulfur proteins are simultaneously resolved within the pI range 3-9 and their electrophoretically determined Mr values are consistent with molecular weights obtained by physical methods.

    Notes: Ampholine pH 4-6 and Pharmalyte pH 4.2-4.9, 4.5-5.4 or 5-6 have been used to improve the resolution of the simplified technique of high resolution two-dimensional electrophoresis. All give excellent resolution but Pharmalyte interferes with protein detection methods to an extent which increases with the pH range of the mixture. The effect is characterised by a progressive increase in the intensity and area of background stain associated with the anode and cathode regions of the two-dimensional gels. It is minimal following Serva Blue R (Coomassie Blue) staining but severe following silver staining.

    Notes: Free-flow electrophoresis of proteins in solutions of borax and mannitol was investigated. The borax solution represents an equimolar boric acid-NaOH buffer with a pH of 9.2. On addition of mannitol the pH of the buffer decreases, with increasing polyol concentration, accompanied by a slight decrease in conductivity. Using a 2.5 mM borax solution, buffers with pH from 9.2 to 4.5 can be obtained. Under the influence of the electric field the pH and conductivities of these buffers change in the near-membrane regions of the separation chamber. Despite the changes the buffers are suitable for electrophoresis of proteins in a free-flow apparatus. The separation of bovine hemoglobin as well as a mixture of bovine hemoglobin, human serum albumin and cytochrome C in borax-mannitol solutions is demonstrated at 80-100 mA constant current and a residence time of only 2.0-2.6 min.

    Notes: An improved procedure is described for polypeptide analysis of radiolabeled antigens resolved by crossed immunoelectrophoresis (CIE). The method involves detection of immunoprecipitates by autoradiography of CIE gels dried onto filter paper. This modification allows selected segments of immunoprecipitate arcs to be excised with a high degree of precision. Radiolabeled antigens are extracted from excised precipitates by incubation at 60 °C in Laemmli sample buffer, and polypeptides are visualized by sodium dodecyl sulphate-polyacrylamide gel electrophoresis performed in conjunction with autoradiography or fluorography. Protein antigens of the bacterial outer membrane are shown to retain (in part) their properties of heat-modifiability following drying and extraction, thus facilitating their identification. The procedure is applied to the analysis of fourteen membrane-associated antigens of Escherichia coli and results in the identification of the ompF/C and the ompA gene products, and the resolution of two novel heterooligomeric outer membrane protein antigens. The polypeptide composition of four previously uncharacterized inner membrane antigens is also established. In addition, six antigens which had been characterized by other unrelated methods as common protein antigen, β-galactosidase, adenosine-5′-triphosphatase, dihydrolipoyl dehydrogenase, D-lactate dehydrogenase and the lpp gene product (the Braun lipoprotein) are shown to possess polypeptide profiles which confirm the initial identification. Evidence is also presented to support the thesis that the bound form of the lpp gene product can associate with other proteins of the outer membrane.

    Notes: The method presented in this article was developed specifically to be used for the electrophoretic separation of biological cells. This method uses low cost materials and ordinary clinical laboratory electrophoresis equipment. Results are reasonably fast; time of migration was 20 min using a constant current setting of 6 mA for a single 3 × 4 inch gel tray. The use of SeaPlaque agarose sol at 40 °C allows the cells to migrate freely, while the solidification of the agarose at the end of the migration period fixes the cells in place on the plate. Samples can be taken from any position on the plate while the sol is still liquid. With this method, differences in rate of migration of erythrocytes from selected species of mammals were demonstrated. The method was developed using erythorcytes for samples and without staining of gels or prestaining of samples. Sample sizes of 2000 to 4000 cells gave easily visible bands. Band of cells from some species migrated as much as 40 mm. Separations were done in polystryrene gel tryays. This technique will be tested in the near future for the separation of the various types of blood cells and for separating other cell mixtures, e. g., bacteria.

    Notes: A new approach to isoelectric focusing in polyacrylamide gels is described, based on the use of rehydratable gels which in dry form could be stored for extended periods and which prior to use were rehydrated with solutions of any composition. Ultrathin 60-240 μm polyacrylamide gels, with different composition (5 % T, 3 % C; 3 % T, 4 % C; 3 % T, 20 % C), were polymerized under well-standardized conditions and, after polymerization, washed exhaustively with distilled water to remove any unreacted monomers, catalysts or soluble polymers. The washed gels were impregnated with suitable additives, before drying, to preserve gel functionality on storage. Polyol compounds, such as glycerol, sorbitol and dextran, as well as synthetic polymers like polyethylene glycol and polyvinylpyrrolidone, were the most efficient additives when incorporated into the gel in a concentration of 1-10 %, either as single substances or in different combinations. Prior to isoelectric focusing the dry gels were rehydrated to the original gel volume with a solution of carrier ampholytes, in some experiments with added separators or urea. Kinetic studies have shown rehydration, depending on gel thickness, to be complete within a few minutes. Isoelectric focusing in rehydratable gels was consistently more reproducible than in wet gels by such criteria as regularity of patterns and coalescence of marker proteins, including ferritin, applied at different positions. Rehydratable gels tolerated higher field strengths at the final stage of isoelectric focusing, with typical values of 500-900 V/cm and thus, at a given volt X hour product, equilibrium focusing could be attained in a shorter time, with improved resolution and sharper zones. Ultrathin-layer isoelectric focusing in rehydratable gels proved insensitive to high salt concentrations (up to 0.5 M) of the samples. Rehydratable gels represent a new generation of gels, excelling over the traditional wet gels by better standardized properties, convenient handling and unsurpassed flexibility.

    Notes: Liver proteins of male C57BL/6T mice treated with 0, 50, or 250 mg/kg Aroclor 1254 were analyzed by high-resolution two-dimensional (2-D) electrophoresis. The resulting patterns were processed using a computerized image analysis system and quantitative data selected for a total of 150 protein spots. On the basis of an analysis of liver proteins form five animals in each treatment group, we found 31 proteins that showed quantitative differences attributable to treatment with chlorinated hydrocarbons at a high level of statistical significance. One of the altered proteins appears to be Mitcon:2, a heat-shock sensitive mitochondrial matrix polypeptide; another appears likely to be microsomal cytochrome b5. The results indicate that quantitative 2-D protein mapping may reveal much more detail regarding the in vivo effects of toxic xenobiotics than has previously been available, and thus allow a more informative approach to the testing of toxic compounds.

    Notes: Computer simulations of the concentration profiles of simple biprotic ampholytes with Δ pKs 1, 2 and 3, on immobilized pH gradients (IPG) at extreme pH values (pH 3-4 and pH 10-11) show markedly skewed steady-state profiles with increasing kurtosis at higher Δ pK values. Across neutrality all the peaks are symmetric irrespective of their Δ pK values, but they show very high contribution to the conductivity of the background gel and significant alteration of the local buffering capacity. The problems of skewness, due to the exponential conductivity profiles at low and high pHs, and of gel burning due to a strong electroosmotic flow generated by the net charges in the gel matrix, also at low and high pHs, are solved by incorporating in the IPG gel a strong viscosity gradient. This is generated by a gradient of linear polyacrylamide which is trapped in the gel by the polymerization process.

    Notes: The effcient and rapid technique of ultrathin-layer isoelectric focusing on 50 μm polyacrylamide gels was used for determination of heat-induced changes of whey proteins (β-lactoglobulin A and B as well as α-lactalbumin) and a method for quantitative densitometry was developed. Improved reproducibility could be achieved by (i) introducing an internal standard (carbonic anhydrase), allowing for compensation for minor irregularities in gel composition, sample application or staining, (ii) running each sample in triplicate, and (iii) multiple scanning of each sample at different positions of the focusing track. Coefficients of variation of the integrated peak areas were approximately 3 % for β-lactoglobulin A and B and about 6 % for α-lactalbumin. Skim milk was heated in a pilot heating plant especially designed for reaction kinetic studies. Using the initial content of native whey proteins in each milk sample as a reference value, degrees of denaturation of more than 90 % could be determined for each of the three protein fractions. Results obtained from reaction kinetic studies revealed that denaturation of α-lactalbumin was a first-order reaction.

    Notes: An ultrathin-layer polyacrylamide isoelectric focusing gel is described for obtaining highly resolved, nondistorted, reproducible patterns for group-specific component (Gc) or esterase D (EsD) subtyping. The 1F, 1S, 2 and 1A1 bands of the Gc system were all typeable by isoelectric focusing alone. All common EsD allelic products were easily resolved. Typing Gc or EsD derived from bloodstains was also demonstrated.

    Notes: With the aid of two-dimensional electrophoresis, the immunoglobulins (Ig) in cerebrospinal fluid (CSF) were separated into heavy and light chains and visualized with silver staining. CSF from 36 patients with no abnormal immunologic activity in the central nervous system show a constant pattern of light chain subfractions, however varying strongly in their respective quantities. In 40 patients with inflammatory diseases such as confirmed multiple sclerosis (MS) (n = 10), neurosyphilis (n = 6), lymphocytic meningoradiculitis (n = 3), novel immunoglobulins appear and form a so-called oligoclonal zone in the light chain region. By comparing various oligoclonal zones, differences were found between the arrangement of the novel Ig light chains and the polyclonal light chain background. In the region of the polyclonal light chain background certain protein fractions appear which are non-immunoglobulin and which are not found in serum. The protein fraction at the isoelectric point of 7.8 appears in 9 out of 10 confirmed MS cases, but in none of the 6 neurosyphilis and 3 lymphocytic miningoradicultis cases. In one case of neurosyphilis with an extremely high gamma-globulin content in CSF we found an oligoclonal zone in the heavy chain region.

    Notes: A mathematical approach to the behavior of isotachophoretic and counter ion components in the steady state of isotachophoresis of weak acids was developed. In steady state, the partial time derivative can be replaced by the -ν times χ derivative, where ν is the velocity of isotachophoresis. Application of this relation to mass balance equations of the components gave the expression of concentration gradients as functions of concentrations of the components, conductance and the H+ ion concentration gradient. The results were inserted into the χ derivative of the electric neutrality equation and the electric current equation, including both electrophoretic and diffusional currents. These equations were solved for conductance and H+ ion concentration gradients and the latter quantities could be expressed as functions of concentrations of the components. Thus, concentration gradients of the components were calculable from the concentrations. Given proper initial or boundary conditions for the concentrations of the components, numerical integration gave pH, conductance and concentration profiles of the components. The results were in agreement with those of capillary isotachophoresis experiments.

    Notes: The 21 free amino acids commonly encountered in proteins have been transformed into “carrier ampholyte” species by reacting their primary amino groups with dansyl chloride. These derivatives can thus be focused in an immobilized pH gradient covering the pH 3.1 to 4.1 interval, except for arginine, which still retains a pI of 8.8. Due to their inherent fluorescence, the dansyl derivatives are revealed in UV light, with a sensitivity of the order of 2-4 ng/mm2. All nearest neighbors are separated except for the following couples: Asn-Gln, Gly-Thr, Val-Ile and Cys-Cys2, with a resolving power, in a ΔpI scale, of the order of 0.0018 pH units. Except for a few cases (notably the aromatic amino acids) the order of pI values is well correlated with the pK values of carboxyl groups, suggesting that the latter are not altered by dansylation. From the set of pKCOOH -pI values of the different amino acids, the pK of the tertiary amino group in the dansyl label has been calculated to be 5.11 ± 0.06. Knowing the pK of the amino-dansyl and the pI of the excess, free dansyl label (pI = 3.34) we have derived, for its sulfonic acid group, a pK of 1.57.

    Notes: Comparison between highly repeated DNA fragments of eucaryotic DNA digested by restriction endonucleases and the corresponding pattern of a completely digested DNA can be used as a criterium of complete digestion before the Southern blotting procedure. Results are presented for highly repeated fragments of human DNA, partially and completely restricted by six different endonucleases. Direct visualization of these fragments was obtained by ethidium bromide staining after agarose gel electrophoresis. Fragments corresponding to a complete DNA digestion were retained and can be used as reference.

    Notes: A novel highly sensitive method for the detection and quantification of proteinase activity is presented. The proteinases were forced electrophoretically into the substrate (sodium caseinate) containing agarose gel. The formation of enzyme-substrate complexes retarded the migration of the proteinases in the electric field and decreased the overall mobility of the enzymes. The proteinases were released as a result of the degradation of the substrate, and consequently the height of the obtained rocket shaped zones of hydrolysis were proportional to the concentration of proteinases in the samples. The electrophoretic method accelerated penetration of the substrate-containing gel by the proteinases and subsequently resulted in a 100-fold increase in sensitivity as compared to the radial diffusion method, depending entirely on diffusion.

    Notes: Several isoforms of the barley α-amylase inhibitor were detected in the purified protein fraction and seed extract after isoelectric focusing. Polymorphism not detected by protein staining was revealed by immunoblotting using an antiserum against barley α-amylase inhibitor. The antiserum, previously described as giving cross reactions with the inhibitors of rye and wheat, was used for comparing the electrophoretic patterns of the protein in several lines of rye, in several varieties of wheat and in different triticales. The antiserum was used for immunoblotting after isoelectric focusing at pH 3.5-10. The inhibitor was found in all samples but the degree of polymorphism was low. The same pattern was found for all lines of rye investigated. This pattern differed from the pattern found for wheat, but again, all cultivars of wheat investigated displayed similar patterns. Structural genes for the α-amylase inhibitors were identified in the A and R genomes. Identical genes may exist on the B and D genomes. The wheat and rye structural genes for α-amylase inhibitor were found to be expressed in the octoploid, hexaploid and tetraploid triticales.

    Notes: Using crossed affinoimmunoelectrophoresis with free concanavalin A the glycandependent microheterogeneity of the immunoregulatory glycoprotein orosomucoid was examined in serum from 30 healthy individuals, 28 patients with benign chronic inflammation and 56 patients with cancer of various types and stages and before and after surgery. We found significant differences in glycan structure between the different patient groups. Thus the patients with the acute phase reaction of surgery differed from patients with chronic inflammation, and patients with testicular cancer differed from patients with colorectal cancer in having a greater proportion of biantennary glycan orosomucoid. Surgery was found to result in a temporary change in the relative proportions of the three microheterogeneity types of orosomucoid, lasting about one week.

    Notes: A research method is described for producing ultramicrogradient acrylamide gels in 0.5 μL constant volume capillary tubes (inner diameter 130 μm and 32 mm length) suitable for the determination of protein in kidney tubule fluid. Previous methods have been limited to gels contained in 1 and 5 μL capillary tubes. The acrylamide gel concentration varies from 4 to 27% in a functional gel of approximately 40 nL and a length of 2.0 to 2.9 mm. Relative migration distances in these gels of known proteins of varying molecular weight (6000 to 669 000) are comparable to those measured in gels contained in 5.0 μL capillary tubes. After electrophoresis, the gels are extruded from the capillary tubes, stained with Fast Green, destained and scanned in a Joyce-Loebl microdensitometer with integrator unit. Such gels are capable of retaining and quantitating low molecular weight proteins contained in nanoliter volumes of biological fluids. The reproducibility of this technique for 0.5 μL capillary gels is comparable to that achieved with the routine analysis of proteins in unconcentrated urine using 5.0 μL gels. Examples are given of protein analyses in kidney tubule fluids and urine obtained from normal rats and those with glomerulonephritis.

    Notes: An automated cell-electrophoresis apparatus is described which appears suitable for clinical laboratory applications. A solid state laser is employed; however, the instrument departs from the usual Doppler systems in that coherence is not required and stationary transmission grating optics are used. A dedicated microprocessor performs fast Fourier transforms (FFT), curve fitting for pedestal and low frequency (LF) noise removal, and digital filtering to suppress FFT artifacts. Unique features include fully automated sample handling, and measurements are made at the center of the migration tube in lieu of the “stationary layer” for enhanced signal quality. Experiments show high resolution of spectra from individual and mixed populations of sheep and rabbit erythrocytes. The sample-to-report cycle of 5 or 7.5 min (switchable), high cost/performance ratio and elimination of potential operator errors make the instrument a candidate for clinical use.

    Notes: A procedure for the simultaneous analysis of reduced and oxidized glutathione together with glutathione conjugates of electrophilic organic compounds by capillary isotachophoresis is described. This method can be applied to quality control of such synthetic conjugates, which are commonly employed as affinity ligands in the chromatographic purification of glutathione S-transferases (EC 2.5.1.18). Using this new assay procedure, it has been found that the glutathione conjugate of 1-chloro-2,4-dinitrobenzene is unstable in neutral to alkaline solution, degrading to glutathione and 2,4-dinitrophenol. Anomalies in the electrophoretic mobilities of a homologous series of alkylglutathione derivatives were observed. A possible explanation is provided on the basis of the formation of micelle-like aggregates, this tendency increasing with increasing length of the hyrocarbon chain. A useful and widely applicable aspect of this analytical procedure is as a method of monitoring glutathione S-transferase. The major advantages of this new assay technique can be summarized as follows: (1) the assay does not depend on optical properties of the acceptor, (2) the reaction can be monitored in the presence of unrefined biological material;aliquots can be injected without pretreatment and (3) the spontaneous oxidation of glutathione in the assay mixture can be monitored simultaneously.

    Notes: Isoelectric focusing in mixed carrier ampholyte-immobilized pH gradients (CA-IPG) is an effective way to separate proteins by charge. A method to prepare CA-IPG in capillary tubes and the description of the equipment used are outlined. Two-dimensional gel electrophoresis patterns of human serum and liver biopsies, with isoelectric focusing and CA-IPG as the first dimension, are presented. The comparison with two-dimensional gel electrophoresis patterns obtained by conventional carrier ampholyte pH gradient separation shows the excellent resolution and potential of this technique.

    Notes: Micro two-dimensional electrophoresis in the absence of denaturing agents allowed the analysis of the multiple forms of serum aminopeptidases, including their multimolecular complexes, and the detection of their enzymatic activity towards different substrates. Two forms of alanine aminopeptidase (E.C. 3.4.11.2.), differing in sialic acid content, were detected in sera of healthy subjects. In sera of patients with hepatobiliary diseases a large number of additional spots was observed, some of them identified as multimolecular complexes, probably formed by an amphiphilic form of alanine aminopeptidase. Papain treatment resulted in the dissociation of the complexes and in the appearance of a light form of the enzyme in normal sera, which was otherwise only present in pathological samples. Additional spots corresponding to cysteine aminopeptidase (E.C. 3.4.11.3.) were detected in pregnancy sera.

    Notes: A method for the estimation of amino acid misincorporation into ribosomal proteins is presented. The method is based on the observation that the horizontal mobility of the 52 E. coli ribosomal proteins is proportional to Q/N, where Q is the calculated charge of the protein and N designates the number of residues in the protein. Thus the mobility change caused by a change in charge, such as the replacement of arginine by cysteine, can be predicted and the gel fractions used for scintillation counting cut accordingly. The gel system used in this paper was not the O'Farrell system but a ribosomal system whose first dimension was run at pH 5 in the presence of 8 M urea.

    Notes: A high background found after silver staining of polyacrylamide gels following two-dimensional electrophoresis and generated from the cathode in the second-dimensional separation has been investigated. Material is formed which is electrophoretically mobile and apparently derived from the buffer components. It behaves like a peptide as far as silver staining. The intensity of this background was reduced by treatment of the second-dimensional gel with a photographic fixing agent prior to silver staining. Moreover, spots which previously appeared negatively stained were changed by the treatment and the result was a general improvement in the contrast.

The virus-induced protein Mr 75 000, detected only in lysates of infected cells, revealed a different proteolytic peptide map. It was shown that each of the structural proteins, although demonstrated to be homogeneous in molecular weight, consists of multiple species differing in their isoelectric points. Protein modifications are discussed as possible cause for the charge variations of ADV structural proteins.

    Notes: Two-dimensional electrophoresis with isoelectric focusing in immobilized pH gradients in the first dimension was applied to the fractionation of hydrophobic proteins from the plasma membrane of a Streptococcus strain. The detergents incorporated into the first-dimensional gel slab, especially the zwitterionic ones of the sulfobetaine series, interfere with protein transfer from the first to second dimension by formation of mixed micelles with sodium dodecyl sulfate molecules. In order to reduce their concentration an elution protocol was devised including: (i) fixation for 1 h in 50 % methanol - 12 % acetic acid; (ii) washing with distilled water for 30 min; (iii) equilibration in concentrated Tris buffer; (iv) denaturation in 5 % sodium dodecyl sulfate - 2 % 2-mercaptoethanol. When counting the number of resolved spots in the protein patterns after two-dimensional electrophoresis, the relative solubilizing efficiency of different detergents scored as follows: Nonidet P-40 (NP-40) 〉 (3-[3-cholamidopropyl)dimethylammonio]-1-propananesulfonate (CHAPS) 〉 N-dodecyl-N, N-dimethylammonio-3-propanesulfonate (sulfobetaine SB 12).

Two types of polymorphism were observed: the first, after EcoRI, HindIII and PstI digestion, is due to an intragenic duplication and corresponds to the Hp-α protein polymorphism; the second type, due to point mutation, was represented by two additional restriction sites for EcoRI and PstI. The univocal correspondence between the first type fragment length variants and protein polymorphism of the duplication type permits the establishment of the genotype at the DNA level, and mutational variants of the second type add new markers; the method described here also permits detection of the real genotype of Hp0 alleles.

    Notes: Factors determining the specificity of immunocytochemical (ICC) tissue stainings as well as the various tests to study these factors are discussed. Since every specificity test only deals with particular aspects of the ICC procedure, a practical sequence of known test methods is proposed, which enables the determination of the specificity of the ICC tissue staining and, after possibly needed antiserum purification steps, may result in a monospecific staining. It is made clear that such a sequence has always to include a tissue-spectrum affinity test, in which the spectrum of tissue antigens is controlled for antibody binding. A variety of such tests, consisting of separation of tissue compounds, fixation, and ICC detection, are discussed as well as their pros and cons with respect to their predictability for the actual serum specificity in the tissue section.

    Notes: In order to establish a method for studying myeloid differentiation, light density, non-adherent, T-cell depleted mononuclear cells prepared from 26 normal peripheral blood buffy-coats were cultured in McCoy' 5A medium supplemented with 15 per cent fetal calf serum (FCS) for three weeks at 37°C in a humidified 5 per cent CO2 atmosphere. The total number of viable cells in the cultures on weeks 1 and 2 represented 73 ± 10 per cent and 98 ± 41 per cent of the initial number of viable cells seeded. After one week, blasts represented 26 ± 10 per cent of the initial number of viable cells while all the initially contaminating mature granulocytes had disappeared. After two weeks, granulocytic differentiation was noted in most cultures and viable myelocytes and more mature cells represented 45 ± 26 per cent of the initial number of viable cells. The differentiation was independent on the lot of FCS used. The addition of PHA stimulated leukocyte conditioned medium to the cultures did not enhance granulocytic differentiation. The granulocytic differentiation observed in the absence of exogenous CSF persisted after removing the cells adhering to the bottom of the flasks on day 2 of the culture. An endogenous colony stimulating activity was detected in the cultures on week 3 but its intensity did not clearly correlate with the degree of granulocytic differentiation.

It is suggested that the concomitant changes in metabolism might act as the signal causing the onset of the changes in phosphate handling associated with phosphate deprivation.

    Notes: The pro-drug cortisol succinate is frequently used as a substitute for cortisol in organ cultures. We found, however, that in Dulbecco's modified Eagle's medium the time taken for the ester to undergo 50 per cent hydrolysis (t½) to cortisol was 75 h. When 15 per cent heat-inactivated normal rabbit serum was present t½ decreased to 47 h, but the rate of hydrolysis was not further increased in the presence of porcine articular cartilage or minced synovial tissue. When frozen and thawed synovium was present t½ decreased to 33 h, presumably due to the release of carboxyl-esterases from the disrupted cells. The level of tetrahydrocortisol was low in all of the cultures. The slow hydrolysis of cortisol succinate resulted in the exposure of the tissues to undersirable fluctuations in the concentration of active hormone, which decreased to low levels at each medium change. Thus, in co-cultures of porcine synovium and articular cartilage, cortisol had a greater inhibitory effect than cortisol succinate on the breakdown of cartilage matrix caused by synovial tissue.

    Notes: Effects of the antiarrhythmic drugs (propranolol, perhexiline maleate, lidoflazine and iproveratril) on energy-linked reactions and on membrane potential were studied. Propranolol, perhexiline maleate and lidoflazine inhibit the ATPase activity of undamaged and broken mitochondria, and of submitochondrial particles. All drugs are inhibitors of either ATP-driven or of succinate-driven reduction of NADP+. The antiarrhythmics promote a decrease in the membrane potential upon energization of the mitochondrial membrane by α-ketoglutarate, succinate, or ATP. It was suggested that these drugs have a primary action on the mitochondrial membrane, thus altering the activities of membrane proteins (channels and enzymes).

    Notes: The aim of this study was to investigate the possibility of enhancing the yield of tissue plasminogen activator (tPA) from two epithelial cell lines of normal (non-malignant) derivation grown in tissue culture. The three agents used in this investigation were chosen because of thier proven enhancing effect on analogous cells or products. The anabolic hormone stanozolol was found to have no significant stimulatory effect on these cell lines. A phorbol acetate (12-O-tetradecanoylphorbol 13-acetate) caused a twofold enhancement in tPA yield but the most significant results were obtained with 5-azacytidine. This agent increased the yield by up to fourfold in small stationary cultures and threefold in large-scale microcarier cultures. A combination of azacytidine and phorbol acetate did not have an additive effect on total yield but did alter the kinetics of tPA expression with time. Indications were that the maximum yield with these types of potentiating agents was achieved as it could not be increased by using a combination of two different agents.

    Notes: This report describes a method of in vitro labelling of proteins designed for subsequent analysis of the sample by two-dimensional electrophoresis. As much as 106 cpm can be incorporated in 5 μg of proteins. As the reagent reacts with the amino groups, virtually all the proteins are labelled. Thus, this method is far more sensitive than silver staining and may prove useful when extremely low amounts of sample are to be analysed.

    Notes: A new method for analytical isoelectric focusing of the benzodiazepine receptor in calf brain is described. Brain membranes are photoaffinity labeled with a specific radioligand, solubilized in 9 M urea and separated on 150 μm ultrathin polyacrylamide gels containing 8 M urea. The method combines high resolution with speed and versatility. It may be generally applicable for isoelectric focusing of integral membrane proteins in genetic studies.

    Notes: A method is described for direct analysis of the reaction mixtures of the production of the herbicide glyphosate and the growth regulator glyphosine by capillary isotachophoresis providing simultaneous determination of the products N-phosphonomethyl-glycine and N.N-bis(phosphonomethyl)glycine as well as of all important reaction components - sulfuric, formic, phosphorous, phosphoric, hydroxyacetic, nitrilotriacetic, iminodiacetic and N-phosphonomethyliminodiacetic acids and glycine. The method is suitable for the analyses both of the final products and of mother liquors, where the maximum ratio of the concentrations ranges within 1:100. The time of analysis is dependent on the sample composition and ranges within 5-20 min. relative standard deviation of a single determination being less than 4 rel. %.

    Notes: Sample stream spreading in the transverse direction (direction perpendicular to the chamber center plane) is calculated for two ground-based and one space-based continuous flow electrophoresis chambers. The magnitude of transverse sample stream spreading is shown to be a function of the difference in electrical conductivity between the sample and curtain buffer. Sample/buffer conductivity mismatch distorts the normally uniform electrical field in the vicinity of the sample to produce transverse field components which, in turn, deflect sample material toward the chamber walls. These results indicate that non-gravity effects are important and must be resolved in order for space-based continuous flow electrophoresis to achieve its maximum potential.

    Notes: The reproducibility of protein migration was studied in three electrophoretic systems: sodium dodecyl-sulfate electrophoresis on polyacrylamide gradient, isoelectric focusing on immobilized pH gradients and two-dimensional charge-size separations. The gradient gels were poured either with a two-vessel apparatus or with a set of computer-driven burettes, as described by Altland and Altland (Electrophoresis 1984, 5, 143-147). Standard deviation on band position was cut by 20-50% with the latter approach in comparison with the former. Typical results were: sodium dodecyl sulfate-polyacrylamide gel electrophoresis, 0.6 mm (over a total length of 135 mm); immobilized pH gradients, 0.6 mm (for 179 mm length); two-dimensional separations, 0.8 mm (χ-axis), 0.6 mm (y-axis).

    Notes: Previous studies by chromatography and electrophoresis have demonstrated that myosin is composed of two classes of polypeptide chains: high relative molecular mass (Mr) components (200 000) and a class of lower Mr species, called the “light chains” (LC). It has been shown before by others that the slow-twitching fibers contain 2 types of isoforms: LC1s and LC2s, while the fast-twitching fibers contain 3 types: LC1f, LC3f and LC2f, the latter also existing as phosphorylated forms. In the present investigation, utilizing two-dimensional (2-D) maps generated with narrow immobilized pH gradients in the first dimension, a higher resolution of the myosin LC has been achieved. The LC1s chains are further fractionated into at least four isoforms; LC2s into three; LC2f into four and LC3f into three sub-species in rabbit muscles. Skeletal muscle tropomyosin, which is separated into three forms (αs, αf and β), is here resolved into six components, while actin from skeletal muscle, previously described mainly as a single chain (α), is here sub-fracionated into four isoforms of identical Mr and different charge. The series of isoforms here reported could represent a fine probe for investigating such complex phenomena as the cellular differentiation of human satellite cells from normal and pathological subjects in greater detail than before.

    Notes: The relation between the electrophoretic migration velocity and the concentration of the non-ionic detergent Triton X-100 in the buffer was studied by crossed immunoelectrophoresis of hydrophilic and amphiphilic proteins. The migration velocity of hydrophilic human plasma proteins was not affected by the presence of Triton X-100 in the agarose gels in the concentration range of 1-0 % v/v. In contrast, amphiphilic proteins, like the human erythrocyte proteins glycophorin, band 3 protein and acetylcholinesterase, and the plasma high density lipoprotein (HDL) apolipoprotein, showed increasing migration velocity with decreasing detergent concentration in the gels. An increase of 20-80 % was observed for different amphiphilic proteins when the Triton X-100 concentration was lowered from 1 % to 0.03 % v/v. This probably reflects the influence of the size of the bound detergent micelle on the charge density of the protein. Thus, by performing corssed immunoelectrophoresis with different concentrations of Triton X-100 in the first-dimensional gel it is possible to identify amphiphilic proteins. Presence of 1 % v/v Triton X-100 in the second-dimensional gel ensures reliable identification of the precipitates.

    Notes: Micro-polyacrylamide gel electrophoresis (micro-PAGE) has been shown to be useful for molecular size separation of urinary proteins. In addition to the sodium dodecyl sulfate micro-gradient gel electrophoresis in 10 μL capillaries, which is used routinely for examination of proteinuria in our laboratory, we tried micro-gradient slab gel electrophoresis (micro-slab-PAGE) for molecular weight determination of urinary proteins. It was also employed for monitoring patients with proteinuric diseases. For detailed studied of urinary proteins, micro-two-dimensional-electrophoresis is recommended.

    Notes: Uncrosslinked polyacrylamide, a viscous liquid, provides a molecular sieving effect in electrophoresis qualitatively the same as that of crosslinked gels. This finding confirms previous results by others using uncrosslinked polyacrylamide mixed with various gel matrices. It provides a sieving medium constituted by an entirely and necessarily random association of inert fibers, as stipulated for interpretation of migration by the Ogston theory. Ferguson plots of several proteins in uncrosslinked polyacrylamide exhibit Yo values smaller and less divergent from one another than in 5 % crosslinked gels, suggesting a curvature of the Ferguson plots of crosslinked polyacrylamide gels at low %T. If linearity of these plots at low %T, 0 %C could be demonstrated, a means would be found to determine accurate values of free mobility by their linear extrapolation to 0 gel concentration. Potentially, uncrosslinked polyacrylamide at low concentrations may provide a tool for the analytical sieving of subcellular particles. However, electrophoresis in uncrosslinked polyacrylamide at concentrations of less than 10 % is perturbed by convection, and thus these potential applications will depend on combining it with a suitable anticonvective device.

    Notes: Recently we described a procedure for pouring immobilized pH gradients at ambient temperature in the presence of a neutral buffer (0.1 M Tris-phosphate pH 6.8) (Altland and Rossmann, Electrophoresis 1985, 6, 314-325). After washing in plain water and in 1 % glycerolin water, the gels were dried, sealed in polyethylene bags and stored at -20°C until use. The reproducibility of a batch of 20 gels with a linear pH 6-10 gradient, poured under computer control, was studied by rehydrating the dried gels in a solution containing 8 M urea, 50 mM dithiothreitol and 0.5 % carrier ampholytes (pH 6-10) and by running human red cell lysates as samples. A comparison of the globin patterns obtained at various time intervals within a range of 7 months revealed that there was no detectable shift in the pH gradient within this period and that gels stored for 7 months can be used to clearly identify all human globins from newborns and adults. Since the tested pH gradient was prepared from Immobilines claimed as being the most sensitive to degradation by hydrolysis (e. g. Immobiline pKa 8.5 and 9.3), it is concluded that storage of dry gels, rehydrated prior to use, is a realistic alternative to the preparation of fresh gels from stored stock solutions for achieving reproducible results in long-term comparative studies.

    Notes: When screening natural populations for genetic variation in mitochondrial (mt) DNA it is necessary to screen as many individuals as possible for as large a number of base pairs as possible. This report describes a combination of laboratory techniques - isolation of mtDNA by either density ultracentrifugation or a phenol extraction procedure and DNA fragment visualization by a sensitive silver staining technique. These routines remove the major restriction of tissue sample size and number of individuals that can be screened and allows the investigation of at least 1000 base pairs from 1 g of tissue or 3-400 base pairs from 100 mg of frozen tissue.

    Notes: Renouncing protein fixation with aldehydes, the polyacrylamide gels are incubated in alcoholic solutions of silver nitrate containing acetic acid at 60-70 °C. for a short time and developed with NaOH, also at 60-70 °C. Using various methods of protein fixation, the detection limits of human albumin were determined for 0.75 and 1.5 mm thick polyacrylamide gels with appropriate thickness correction. An overlay technique with silver nitrate containing polyacrylamide gels is described. Dispersing Coomassie Brilliant Blue G-250 in a solution containing HNO3 and NH4NO3 allows rapid silver staining of prestained gels.

    Notes: Gradient (5-30 %) polyacrylamide gel electrophoresis (PAGE) was used for the analysis of serum cholinesterases from the spiny mouse, the mouse and the rat. Enzymatic staining of slab gels was performed using acetylthiocholine as substrate. Eserine, 1.5-bis(4-allyldimethylammoniumphenyl)-pentane-3-one dibromide (BW284C51) and tetraisopropylpyrophosphoramide (iso-OMPA) were used as selective specific inhibitors of total, true and pseudo-cholinesterase, respectively. Five main molecular forms were detected in the three species as sharp, well separated bands. All bands were inhibited completely by eserine while the effects of BW 284C51 and iso-OMPA varied in the three species. This result suggests a common qualitative pattern for the three rodents with a different distribution of true and pseudo-cholinesterase molecular forms for each species. The good resolution shown by the reported densitometric scan recordings confirms the advantages of gradient over homogeneous PAGE in the study of serum cholinesterases.

    Notes: Agarose gel electrophoresis has been shown to give rise to non-linear plots of log (mobility) vs. gel concentration of spherical viruses (Serwer,[1]) and cellular vesicles (Gottlieb et al.[2]). This finding also applies to proteins as shown in this study. Considering that in the non-linear plot, the slope becomes a function of gel concentration, it is possible to determine particle properties and gel parameters by a modification of the conventional method derived from the Ogston theory for long-fiber gels. This treatment shows: (a) In application to data obtained from gel electrophoresis (0.4 to 1.6 % agarose) of viruses (13 to 42 nm radius) and with increasing gel concentration: (i) an increase of apparent total fiber length per g agarose matrix, (ii) a reduction of apparent fiber radius and (iii) a constant fiber volume (per g matrix material) of the agarose fiber. The values of the fiber radii identify the fibers as agarose supercoils (or aggregates of it) with a radius of 20-55 nm. (b) In application to data obtained from gel electrophoresis (1.2 to 8 % agarose) of proteins (1.7 to 5.8 nm radius): a fiber volume indicative of additional sieving by the agarose double helix (of known radius of 0.5-2 nm). This is in agreement with a previous suggestion by Serwer [1] that proteins are able to penetrate into the double-helical network of which the agarose supercoil consists. (c) The likelihood of a continuous transition from case (a) to case (b).

    Notes: In pH 4-10 immobilized pH gradient (IPG) electrophoresis, one observes (a) partial failure of the sample to enter the gel; (b) which halt protein migration; (c) absence of defined transient state band patterns; (d) lateral zone spreading and distorted zone profiles; (d) uneven voltage profiles with a distinct voltage maximum; (f) difficulty in measuring pH profiles with the contact electrode. These problems are remedied through the addition of 0.1 to 1 % carrier ampholytes (CA) to the IPG gel. Problems (a) to (c) disappear in IPG tube gels. IPG in presence of CAs (ICAPG) of pI-range 3.5-9.5 or 6-8 yields a pH gradient, a protein pattern and a pH gradient stability essentially the same as that in IPG gels of the same pH-range 4-10. ICAPG is insensitive to ionic strength of the sample up to 0.1 M. Gel swelling and water exudation increase in ICAPG which make ICAPG in tube gels inapplicable when CA concentrations exceed 0.1 %.

    Notes: Phenol extracted, alkali-treated lipopolysaccharide (aLPS) from vaccine strain (S19) Brucella abortus was demonstrated by two-dimensional gel electrophoresis to consist of at least ten silver staining, polydisperse analogues having different pIs. When tested on nitrocellulose immunoblots, all ten were antigenically reactive with bovine anti-B. abortus polyclonal sera, but only six reacted with anti-B. abortus O-antigen murine monoclonal antibody. Analogues focusing at different pIs were concluded to arise from differences in either core or O-antigen side chain structure or because of covalently bound protein. While not qualitatively different, aLPS from pathogenic B. abortus strain 2308 had lesser amounts of analogues 1, 2, 5, 6, and 8 than did aLPS from strain 19 (vaccine). The 2-D gel electrophoresis method was demonstrated to be of value in the analysis of aLPS from B. abortus and may be useful in the study of lipopolysaccharides from other sources.

    Notes: The electrophoretic behavior of esterase D (E.C. 3.1.1.1.) allozymes 1, 2 and 5 was studied in various buffers and pH ranges, and compared with results obtained on isoelectric focusing. It is shown that the relative mobility of ESD 2 and 5 allozymes is reversed according to the pH of the buffer system. In acidic pH ranges the mobility is: cathode 1〈2〈5 anode; using the same buffer system in basic pH ranges the following order is observed: 1〈5〈2. Technical improvements in agarose gel electrophoresis leading to a considerable economical advantage over the previously described methods are presented.

    Notes: Mathematical simulation of an isotachophoretic boundary between protein and weak acid was made on the assumption of steady state. In order to deal with a protein molecule, a Gaussian model for charge distribution of protein molecule was constructed, based on the assumption that acidic and basic residues on a protein molecule dissociate independently of each other. The results were roughly in agreement with protein stacking experiments with regard to boundary thickness and concentration of the stacked zone.

Resistant high order structures sensitive to the action of urea might cause these variations.     Notes: Development of techniques for silver staining of proteins separated by polyacrylamide gel electrophoresis has increased experimental detection limits. However, the precise basis for the reaction between silver and polypeptides is still unclear and, depending upon the choice of silver reagent, may even differ. We compared protein stain intensity with silver diamine (ammoniacal silver) and silver nitrate reagents based on amino acid composition by determining the protein amount required to generate an arbitrary staining intensity and then calculating moles of each amino acid present in some representative proteins. We compared these values for each amino acid, taken singly, as well as for combinations of two and more. In no case could staining be attributed entirely to specific or classes of amino acids, thus supporting the argument that an inherent difference exists between primary reactive centers for the two staining reagents.

    Notes: A high resolution agarose gel system (Panagel) with silver staining and polyacrylamide disc gel electrophoresis were used to demonstrate oligoclonal bands in cerebrospinal fluid (CSF) from 150 patients identified as having either inflammatory neurological diseases (110 patients) or non-inflammatory neurological diseases (40 patients). The detection of oligoclonal band patterns in high resolution agarose electrophoresis is sensitive: 65% (polyacrylamide gel electrophoresis: 50%) and specific: 95% (polyacrylamide gel electrophoresis: 73%) with respect to inflammatory neurological diseases. This procedure is easier to perform than polyacrylamide disc gel electrophoresis and is recommended for routine examination of CSF especially for the detection of oligoclonal bands.

    Notes: Immunoblotting after sodium dodecyl sulfate polyacrylamide gel electrophoresis of the purified third component of human complement (C3) and its fragments and of unpurified C3 and its degradation fragments in serum or plasma has been examined. The pattern of C3 degradation observed corresponded well with previous observations made with purified proteins. The immunoblotting technique demonstrated a high resolution, and proved to be capable of providing a full analysis of mixtures of C3 fragments in serum with the use of chain-fragment-specific antibody preparations. Additional heterogeneity in C3 and its breakdown products were observed, and covalent adducts between activated C3 and other proteins were detected directly in serum and plasma without any purification of the components. The results further indicated that antibodies raised against denatured C3 reacted quantitatively and qualitatively better with the nitrocellulose-bound C3 than those antibodies raised to native C3, probably reflecting the paratially denatured state of molecules after separation by polyacrylamide gel electrophoresis and blotting onto the nitrocellulose.

The method utilizes low sample volume and can be completed within 5 h. One gel can be used to screen 15 samples, making the procedure ideal for screening erythrocyte galactokinase in newborn screening programs. This method can be ideal for many laboratories interested in screening for galactokinase variants.

    Notes: Progesterone therapy of androgenized rats results in the elimination of histological abnormalities of the uterus associated with constant unopposed oestrogenic stimulation. Subcellular fractions prepared from uteri of normal, androgenized and progesterone treated animals have been analysed by two-dimensional gel electrophoresis. In all fractions, protein changes were observed as a result of androgenization and subsequent progesterone therapy. The identity of some of these proteins has been assigned following peptide mapping. Following progesterone therapy, the relative proportion of cytosolic tubulins, tropomyosins and D-glucose-6-phosphate dehydrogenase were increased. In the abnormal uterus, a protein with similarity to the B subunit of creatine kinase was present in the mitochondrial fraction but could not be detected in the cytosol. Subsequent to progesterone therapy, this protein was no longer detectable in the mitochondrial fraction whilst two molecular forms of the B subunit of creatine kinase appeared in the cytosol. These hormonally-sensitive changes in both structural and enzymatic proteins, which are ubiquitous to most cell types, are discussed in relation to morphological differentiation of the uterus and the molecular mechanisms underlying cell growth.

The apparent hydrolysis rate (derived from the shift of myosin spots in a two-dimensional map) is 20% per year (or 10% if incorporation of acrylic acid, an hydrolysis product, is accounted for). Such reactions in frozen solutions are not uncommon (cf. the spontaneous mutarotation of glucose, Kiovsky, T. E. and Pincock, R. E., J. Amer. Chem. Soc. 1966, 88, 4704-4710).

    Notes: A demountable polyacrylamide gel system is described that uses commercial components and can be adapted to any slab system. The use of GeldBond PAG film produces polyacrylamide gels that can be easily handled and allows separation of the conditions needed for polymerization from those for electrophoresis. Polymerization is performed with one common protocol, and the gel is disassembled. Gels can then be soaked or stored in any desired solutions, followed by reassembly of the equilibrated gel and electrophoresis. This procedure allows for a wide range of electrophoretic conditions starting with a single type of polymerized gel as well as permitting use of systems that have technical difficulties under normal cirumstances such as horizontal gels or those containing the cationic detergent, cetyltrimethylammonium bromide.

    Notes: The theoretical principle and experimental verification of a new separation method for proteins in thin gel slabs called “isotachophoretic focusing” are presented. Isotachophoretic focusing is basically isotachophoresis with two new features: (i) By using adequate spacers a continuous mobility spectrum is generated. Under isotachophoresis conditions, these spacers give rise to a linear electric field gradient in which proteins can be separated in form of focusing bands. The synthesis and isolation of suitable spacers for isotachophoretic focusing are described. (ii) The stack is immobilized or, at least, its migration is retarded by exploiting the counterflow of electroendosmosis resulting from the action of the electric field on charged groups linked to the gel. Immobilization of the stack allows its formation to equilibrium and the protein separation process to continue as long as necessary. Details are given concerning the experimental set-up and factors involved in the optimization of the technique. The separations obtained by isotachophoretic focusing are compared with those obtained by classical electrophoresis.

    Notes: A computer-controlled version of recycling isoelectric focusing has been applied to the purification of recombinant human leukocyte interferons (IFNs) from E. coli. The system is based on continuous recycling between a 12-channel heat-exchange reservoir and a 12-channel focusing cell. It contains ten pH meters with probes in each of the flowing channels and two ultraviolet spectrophotometers with five flow cells per instrument. A minicomputer system acquires real-time data of pH and optical density measurements, controls and continuously monitors voltage and power outputs across the focusing cell, and generates on-line multi-color graphics on an x-y plotter. Recycling isoelectric focusing in the presence of 4 M urea resulted in removal of contaminating bacterial pyrogens from recombinant IFN alpha-2 with complete retention of specific antiviral activity. It also produced a 25-fold purification of crude recombinant IFN alpha-5 in a single step and resolved two biologically active species. These data suggest that recycling isoelectric focusing may be incorporated into the design of simple schemes for the purification of a variety of recombinant proteins.

    Notes: Several human cancer cells possess receptors for 1,25-dihydroxyvitamin D3[1,25-(OH)2D3]. In these cells 1,25-(OH)2D3 has a biphasic concentration-dependent regulatory effect on cell replication and specifically induces its own metabolism. We have studied the effects on these parameters of the native hormone together with those of two analogues fluorinated at the 24-carbon and of 1,24R,25-trihydroxyvitamin D3[1,24R,25-(OH)3D3]. The difluorinated analogue 24,24-difluoro-1,25-(OH)2D3[24,24-F2-1,25-(OH)2D3] is an approximately fivefold more potent inhibitor of cellular replication than the native hormone, while 1,24R,25-(OH)3D3 is about fivefold less potent. This enhanced potency of the fluorinated analogue parallels its enhanced potency in in vivo studies of its effects on calcium and mineral metabolism. However, although the analogue retains replication stimulatory activity, it is clearly no more potent than the native hormone in this activity: 1,24R,25-(OH)3D3 has no significant stimulatory activity. Exposure of the cells to 1,25-(OH)2D3 at 0·05 nM for 6h increases the subsequent conversion of labelled hormone to aqueous phase soluble compounds by 6·7-fold. None of the other compounds had a similar effect at this concentration. At 10nM all 1-hydroxylated compounds increased aqueous phase radioactivity about equally (13 to 17-fold); this effect is still specific since 25-OH D3 had no such effect even at 10nM. Studies on the effects of the fluorinated analogues upon receptor binding of hormone in cell cytosols and uptake of hormone by intact cells clearly demonstrate that the enhanced activity of these analogues is not due to higher receptor affinity or more rapid access to intracellular receptor. These data suggest that this enhanced inhibitory potency, observed in these human cancer cells and by analogy their enhanced potency in vivo, is due to their resistance to metabolism by 24-hydroxylation. In view of our previous observations that 24-hydroxylation is part of the induced metabolic pathway, these data support the hypothesis that 24-hydroxylation is part of an inactivation pathway in these human target cells.

    Notes: Mouse pancreatic islets cultured in vitro were infected with a tissue culture-adapted or a mouse pancreas-adapted strain of Coxsackie B4 (CB4) virus. The effects of the viruses on the islets were assessed by examination of their biochemical functions. It was found that the mouse pancreas-adapted strain of CB4 induced a ‘leakage’ of insulin from islets incubated at a basal (2 mmol l-1) glucose concentration, both at two and four days following infection. However, at a stimulatory concentration of glucose (20 mmol l-1) the rate of insulin secretion appeared to be normal in these islets. At two days the rate of total protein synthesis in islets infected with mouse pancreas-adapted CB4, incubated at high glucose concentration, was reduced; at four days the degree of inhibition was more severe, the rate at basal glucose concentration falling to half that of the control islets and at the stimulatory glucose concentration to a quarter of the control islets. (Pro)insulin biosynthesis was also inhibited, the rate being reduced to less than half the mean control value in islets infected with mouse pancreas-adapted CB4 virus at 20 mmol l-1 glucose at two days; at four days the rate was greatly reduced at both 2 and 20 mmol l-1 glucose. It is concluded from this study that (1) only certain strains of CB4 virus can infect mouse pancreatic islets in vitro and (2) that infection with strains of virus tropic for the islets leads to an impairment of metabolic functions of the B-cells, and is not necessarily lytic.

    Notes: Using frozen liver sections and quantitative cytochemistry it has been established that when cells are allowed to oxidize a cytoplasmic and a mitochondrial substrate simultaneously the resulting oxidative activity is markedly higher than the sum of the oxidation of each substrate measured separately. In the present study this type of synergistic interaction has been confirmed in the kidney, particularly in cells of the pars recta. Our results support the evidence of the influence of cytoplasmic NADPH on the intramitochondrial oxidative process and it is suggested that, in cells of the pars recta, cytosolic NADPH may be involved in intramitochondrial mixed function oxidases such as 1α-hydroxylase: these results could further elucidate the mechanism responsible for the production of the hormonal form of vitamine D3.

    Notes: Insulin receptors have been characterized in rat prostatic epithelial cells by using [125I]insulin and a variety of physicochemical conditions. The binding data at equilibrium (2h at 15°C) could be interpreted in terms of two populations of insulin receptors: a class of receptors with high affinity (Kd = 2·16 nM) and low binding capacity (28·0 fmol mg-1 protein), and another class of receptors with low affinity (Kd = 0·29 μM) and high binding capacity (1·43 pmol mg-1 protein). Proinsulin exhibited a 63-fold lower affinity than insulin for binding sites whereas unrelated peptides were ineffective. The specific binding of insulin increased by about 50 per cent after 96 h of fasting; this increase could be explained by an increase of both the number of the high affinity-low capacity sites and the affinity of the low affinity-high capacity sites. These results together with previous studies on insulin action at the prostatic level strongly suggest that insulin may exert a physiological role on the prostatic epithelium.

    Notes: Arginine vasopressin stimulates Na+-K+-ATPase activity located in the rat thick ascending limb of s'Henle loop. Mammalian hypothalamus appears to produce a factor capable of inhibiting Na+-K+-ATPase activity in a variety of tissues. The effect of a purified rat hypothalamic extract with and without AVP on rat renal Na+-K+-ATPase activity was evaluated by a cytochemical technique. The hypothalamic extract alone failed to affect basal Na+-K+-ATPase activity throughout renal segments after 10 min exposure. Na+-K+-ATPase activity stimulated by AVP (1-10 fmol l-1) for 10 min was inhibited by rat hypothalamic extract over the concentration range 10-7-10-3 U ml-1 in a dose-dependent manner. Complete inhibition of AVP-stimulated Na+-K+-ATPase activity occurred at a hypothalamic extract concentration of 10-3 U ml-1. Only Na+-K+-ATPase activity located in the renal medullary thick ascending limb was influenced by the rat hypothalamic extract.
