    Notes: Continuous cultures of some heterotrophic Spirilli (Sp. serpens, Sp. curvatum, Sp. sp.) exhibit threshold concentrations of lactate, the growth limiting carbon and energy source, around 15 mg./l. This value corresponds to a threshold population density of 0.92 mg. dry weight/l. at a dilution rate of D = 0.185 hr.-1 (= 0.5 maximum growth rate). The minimum population density, which is regarded as equivalent to the starter population in batch culture, was dependent on the growth rate and the redox potential. The existence of minimum populations and the pronounced effect of population density on growth at low nutrient levels seem to be widespread phenomena closely related to growth under suboptimal conditions.

    Notes: A small fraction, about 0.05%, of the microorganisms in milk are resistant to pressure to at least 150,000 p.s.i. The pressure-resistant microorganisms were shown to be mainly bacterial spores. Some of the pressure-resistant organisms were identified. The lethal effect of pressure is more pronounced in the solid phase than in the liquid phase of water. B. subtilis was found to be capable of surviving solid-phase transitions from Ice II, III, and V to Ice I. The lethal effect of high pressure in the liquid phase of water was found not to be counteracted by an increase in temperature. The pressure resistance of B. subtilis was found to be maximum around neutral or slightly basic pH. The damaging effect of low and high pH are counteracted by sodium chloride and glucose. It is suggested that the primary mechanism of protein denaturation by pressures above 30,000 p.s.i. is ionization and formation of ionic bonds between charged groups on proteins, altering their solubility.

    Notes: The electrochemical activity of the individual chemicals in the nutrient medium for growth of Thiobacillus thiooxidans was studied along with the effect of the gases in equilibrium with their solutions. Several chemicals were active individually; however, the magnitudes, as measured by changes in half-cell potential, were less than that observed when T. thiooxidans was present in their composite mixture. Sterilized Skerman's mineral salts and the American Type Culture Collection (without ferrous sulfate) media were not sensitive electrochemically to changes from pure oxygen to nitrogen atmospheres. When T. thiooxidans was present in these media, the biological half-cell emf became sensitive to changes in the oxygen content of the atmosphere in equilibrium with the organism and nutrient. The ability of T. thiooxidans to make an electrochemical contribution, as registered by a platinum electrode, is substantiated further by these investigations.

    Notes: A substrain of Bacillus cereus 569/H produced above 10,000 units of penicillinase/ ml. when grown in a pilot-plant fermentor using a medium containing Casamino acids techn. (Difco) or N-Z-Amine type B (Sheffield), and salts. Simplified purification and concentration procedures give an overall yield of 50-65% enzyme. The freeze-dried enzyme preparation had a good storage stability in vacuumsealed ampules kept at 4, 30, and 37°C. In vials containing air in the head space, partial inactivation occurred in two months at 30 and 37°C. The freezedried preparation showed satisfactory performance in the production of yoghurt fermented milk.

    Notes: An automatic aeration controller was constructed to assure the optimal amount of oxygen for aerobic biosynthetic processes. A platinum electrode and a KCl bridge for the calomel electrode were inserted hermetically into the fermentor and were sterilized with the culture medium. The electrode potential developed at the surface of the Pt electrode continuously indicated the actual oxygen concentration of the medium. This served as the controlled variable of the system. The concentration of dissolved oxygen resulted from the actual demand and supply of the microorganism. The electrode potential, which was the measured variable of the process, operated a closed-loop automatic aeration system. The controller activated an air inlet valve in order to balance the controlled variable. The automatic controller operated in fermentors of industrial scale for long periods in a satisfactory manner.

    Notes: The kinetics of the gluconic acid fermentation have been studied at pH 5.8 Pseudomonas ovalis B-1486 was used as the fermenting organism. D-Gluconolactone was found as an intermediate in the fermentation and accumulated at times in large amounts. When this occurred, hydrolysis of the lactone was found to be the rate limiting reaction. This can be, under certain conditions, a poor fermentation in which to study oxygen transfer limiting conditions. Further, it is impossible to relate batch and continuous kinetic data for this fermentation unless the lactone hydrolysis reaction is taken into account.

    Notes: The yeast S. cerevisiae was grown on dilute, chemically defined media in continuous culture with either glucose or ammonium sulfate as the growth-limiting ingredient. Changes in dilution rate or glucose concentration induced decaying oscillations in the numbers of yeast growing on ammonium sulfate-limited media. Spot checks indicated that Cell dry weight and Kjeldahl nitrogen followed the cell numbers during these oscillations. With glucose-limited media, there was no response to step changes in ammonium sulfate concentration, and dilution rate step changes gave non-oscillatory transient responses.

    Notes: The foam separation technique of microflotation was studied using A. aerogenes as the test organisms. Gas flow rate, collector and alum (when used) concentrations, and frother dose were held constant. In contrast to E. coli, previously reported, A. aerogenes are removed using both lauric acid and laurylamine as the collectors without prior coagulation with alum. The removals are improved after coagulation, with laurylamine being the more efficient collector. In all cases the removals decrease upon increasing the pH above about 8.

    Notes: The rate of dissolution and dehydration of CO2 in a liquid model system was investigated. Components in the model system established the main conditions which may exist, in the extracellular space of a microbiological culture liquid. The charge in voltage of a glass electrode was measured which indicated the formation of H+ ions in the H2CO3 ⇌ HCO3- H+ reaction. The rate of CO2 hydration increased with the increase of temperature from 0 to 40°C. Likewise the equilibrium of the reaction was shifted towards the forward reaction. Similar results were observed when the tip velocity of the impeller was increased. Data suggest that agitation promotes the dissolution of CO2 in the culture liquid through the reduction of gas-liquid film resistance in the diffusion of this gas. The rate of hydration of CO2 into the bulk of the liquid was independent of pCO2 above the surface of the liquid but depended on pCO2 in the gas bubble within the liquid. The concentration of HCO3- was, furthermore, influenced by the buffer components, buffer capacity, and the viscosity of the system. Since pCO2 and the HCO3- concentration in the extracellular space depend on both physical and chemical factors, the ventilation of a culture liquid necessitates both exhaust of CO2 from the gas bubbles of the culture broth and shift of the H2CO3 ⇌ HCO3- + H+ reaction towards the backward direction.

    Notes: A method for the determination of total heat generation in a fermentation tank and overall heat-transfer coefficient at the cooling surface is outlined. These data, which are essential for the design of cooling systems, are measured during the actual fermentation by dynamic analysis of controlled temperature variations. Each experiment consists of two stages: one in which cooling is cut off, and one in which cooling is constant. The necessary temperature variation is so limited that, the course of the fermentation is not affected.

    Notes: A generalized logistic equation is proposed for the mathematical representation of batch culture kinetic data. Properties of the equation are discussed. A computer program is used to fit the generalized equation to both artificial and actual batch culture data. The equation is shown to be capable of fitting data exhibiting lag, exponential, deceleration, stationary, and death phases, as well as diauxic growth. The fitted equation is useful for differentiation, interpolation, and other manipulations of the data, and it is a convenient means of data storage.

    Notes: Interfacial phenomena are directly involved in the adhesion of a strain of Chlorella, a unicellular alga, to glass surfaces in simple ionic solutions. The principal mechanisms governing the adhesion appear to be electrostatic interaction between electrical double layers and various specific surface interactions resulting from surface heterogeneity and ion adsorption. Under most conditions the algal cells and glass surfaces have negative zeta potentials, and adhesion to glass will not occur; but if, for example, FeCl3 is added to an algal-glass system immersed in 0.05M NaCl, the algal and glass surfaces will possess very different zeta potentials, and adhesion will be strongest under those conditions which produce the greatest, difference in zeta poentials. Prior pretreatment and usage of glass apparatus greatly affect the glass zeta potentials and the adhesion of algal cells to glass. An apparatus for measuring a relative set of numbers representing the force of adhesion of algal cells is described.

    Notes: The growth of batch-cultivated yeast Candida lipolytica on three kinds of gas oil using mineral medium was studied. A linear dependence was found between the production of yeast biomass and the consumption of n-alkanes, while the decrease of freezing point of gas oil during cultivation had a distinct course. This disproportion was explained by different degradation of individual n-alkanes contained in gas oil. The rate of degradation of pentadecane, hexadecane, and heptadecane was the same during the entire cultivation. On the contrary, in the first phase the utilization of shorter chain n-alkanes, nonane to tetradecane, was more rapid while that of longer chain homologs, octadecane to pentacosane, lagged. Rapid utilization of longer chain n-alkanes did not occur before the concentration of the other n-alkanes decreased. Only then the rapid decrease of freezing point appeared.

    Notes: The influence of agitation and aeration on growth and on production of glucose oxidase of Asp. niger has been studied. It was found that both rate of growth and glucose oxidase production was higher at an agitation speed of 700 rpm than at 460 rpm. Further increase in speed of agitation resulted in neither a higher rate of growth nor a higher glucose oxidase activity. Total glucose oxidase activity was highest in a medium containing 5% sugar (at an agitation speed of 700 rpm) and did not get higher when the sugar concentration of the medium was increased to 7%. When pure oxygen was bubbled through the culture the rate of growth of the culture (in the linear phase) was 95 mg. mycelial dry wt./100 ml./hr., and only 61 mg. when air was applied. The glucose oxidase activity of oxygenated culture was double the activity of aerated culture. Viscosity of the homogenized culture became higher with higher concentration of mycelia. The viscosity of oxygenated culture was found to be lower than that of aerated culture.

    Notes: This is the first part of a systematic study on the mechanisms of the Waldhof fermentor. When an agitator is rotating in a liquid, a vortex will develop on the surface. Air is dispersed into the liquid when the vortex is deep enough to reach the agitator. This is the basic mechanism of air dispersion in a Waldhof fermentor. In this work, experimental results, empirical correlations, and theoretical equations were obtained to relate the vortex depth to a number of physical factors including agitator diameter, agitator speed, tank diameter, liquid depth, liquid viscosity, and so on. The vortex depth was found a function of both Reynolds number and Froude Number.

    Notes: A mathematical model is presented for both batch and continuous cultures of microorganisms utilizing inhibitory substrates. The key feature of the model is the use of a inhibition function to relate substrate concentration and specific growth rate. Simulation studies show that the primary result of inhibition by substrate in a batch culture is an increase in the lag time whereas in continuous culture inhibition by substrate may result in process instability. The model should be of value in investigations of the stability of biological processes used for the treatment of certain industrial wastes such as those containing phenols, thiocyanates, nitrates, ammonia, volatile acids, etc., which are known to be inhibitory to many of the organisms metabolizing them.

    Notes: Growth of Saccharomyces cerevisiae LBG H 1022 on ethanol under transient-state conditions was studied. As a cultivation device, an aerated Chemap fermentor combined with continuously working gas analyzers for oxygen and carbon dioxide was used. Yeast cell dry matter, substrate concentration, specific oxygen uptake, specific carbon dioxide release, and respiration quotient were measured during the different transient states. Depending on which range of the dilution rate the initial steady state was found, we obtain different responses to the shift experiment. For the lower range, up to D = 0.07, we deal with damped oscillations ranging above and below the steady-state values. For the higher specific growth rates, the rate of damping is strongly enhanced and the shape of the curves becomes an asymptotic approach to the final steady states.

The apparatus consists of a motor-driven air sparge pipe which may be lowered or raised to give a variable point of entry of the air stream into the culture liquid and hence a variable gas dispersion and gas-liquid contact time. Control of the sparge pipe position is by means of a feedback control loop consisting of a dissolved oxygen probe, an on/off controller, and a reversing electric motor which drives the sparge pipe. The difficulty presented by the relatively slow response of the oxygen probe has been overcome by incorporating an adjustable rate of control action.

The aminoacylase column having the highest activity was obtained when 7500 μmoles/hr. of partially purified aminoacylase was charged into a column packed with 9 ml. of DEAE-Sephadex A-25 (bead type, hydroxy form). By employing a DEAE-Sephadex-aminoacylase column, conditions for continuous optical resolution of acyl-DL-amino acids were investigated. When a solution of 0.2M acetyl-DL-methionine (pH 7.0, containing 5 × 10-4M Co2+) or 0.2M acetyl-DL-phenylalanine (pH 6.0, containing 5 × 10-4M Co2+) was passed through the aminoacylase column at the flow rate of SV = 2.5 or 2.0, respectively, at 50°C., the highest rate of hydrolysis of both substrates was attained. From the column effluents, enzymatically hydrolyzed L-methionine and L-phenylalanine were isolated in a good yield.

    Notes: Toxicological investigations were conducted on two new antibiotics, Muconomycin A and Muconomycin B. These non-nitrogenous antibiotics were found to be highly toxic and capable of inducing profound inflammation in the peritoneal cavity of male albino rats. Either antibiotic produced large volumes (10-20 ml) of inflammatory exudate even when injected intraperitoneally in quantities of 1.6 × 10-10 moles. An extensive profile of the electrolytes and proteins found in inflammatory exudates was developed. Simultaneous assays of the blood serum of treated rats provided a basis for comparing the concentrations of constituents of serum with those of the exudate. The results of these assays showed that the exudate contained lower concentrations of sodium and proteins, and greater amounts of potassium, calcium, and phosphorus than the serum. Chloride ion concentrations were variable. Since previous work showed that one of the manifestations of toxicity of these substances was the production of creatinuria, further studies were carried out with ATP/Creatine Phosphotransferase. These studies show that these antibiotics are potent in vitro inhibitors of the enzyme ATP/Creatine Phosphotransferase.

    Notes: A process was devised for the synthesis of tris(hydroxymethyl)acetic acid by means of bio-oxidation of pentaerythritol. A flavobacterium able to grow on pentaerythritol was isolated from the soil. Mutants were obtained that, were deficient, in their pentaerythritol oxidation system. These mutants could not grow on pentaerythritol, but when grown on an assimilable carbon source; they oxidized pentaerythritol to tris(hydroxymethyl)acetic acid. This conversion readily took place in a medium consisting of 20 g pentaerythritol, 10 g yeast extract, and 2 g acetic acid (neutralized) per liter. Since the mutants were unable to metabolize tris(hydroxymethyl)acetic acid, theoretical conversion yields were attainable. When pentaerythritol was added stepwise and the acid formed was neutralized continuously, 95-100% yields were obtained in concentrations of the order of 60 g/l.

    Notes: In order to increase the availability of the cell bound protein in Scenedesmus algae, mechanical, enzymatic, and chemical methods of degrading the cell wall structure were investigated.Mechanical treatment involved the use of a ball-mill. The algae suspension together with glass beads was milled in a water-cooled chamber equipped with rotating disks. The enzyme tested was a cellulolytic enzyme (Meicelase) and the chemical employed was hydrogen peroxide.In the ball-mill experiments a complete disintegration was achieved ina disintegrator, working with batches. Trails wwere also performed with a continuous disintegrator and the depedence of disintegration on bead size and flow rate was studied. The disintegration determined by microscropic cell count was compared to the increase of the pepsin digestibility.The meicelase treatment caused a slight increase of the pepsin digestibility, as measured after 3 hr pepsin incubation. No increase of the pepsin disgestibility could be detected with hydrogen peroxide treatment.After the ball-mill disintegration 95% of contaminating bacteria were killed and yields of extractable proteins were higher. The capacity of availble continuous ball-mills is such that they could be used on a pilot-plant scale and the energy cost of disintegration would be of the same magnitude as that of separation.

By using a distillation fraction of gas oil distilling between 180-400°C, containing 10-20% of n-alkanes, the optimal condition for biomass production and deparaffination were estimated for various dilution rates and various amounts of gas oil in the medium. The main factor, which influences the yield coefficient by hydrocarbon fermentation is the polyauxie of the hydrocarbon substrate. The penetration of dispersed hydrocarbons into the yeast cell is demonstrated on electron micrographs and the velocity and reversibility of this process is estimated by using tritium-traced hexadecane.

    Notes: Continuous culture in a cascade of vessels with the addition of supplemental nutrients to any stage permits adjustment of the physiological state of the culture in each stage to best achieve a desired performance goal. The yeast Saccharomyces cerevisiae in two-stage continuous cultivation was selected as a model system. With conditions in the first stage held constant- at a selected glucose concentration in the feed stream, dilution rate for the second stage was varied. Cell numbers, dry weight, glucose concentration, respiration coefficient, and titers of several enzymes were determined. The seed rate was defined as the ratio of glucose concentration in the feeds to stage 1 and to stage 2. At low seed rates, the calculated specific growth rate in the second stage was proportional to dilution rate. At higher seed rates, the specific growth rate based on dry weight behaved differently from that based on cell numbers, and the dependence on dilution rate was not linear.

    Notes: The addition of potassium ferrocyanide to the molasses substrate (mash) used in the citric acid fermentation had little or no effect on the total carbon, nitrogen, or phosphorus content of the substrate but reduced the ash content by 1-4% depending on the type and crop year of molasses. Eighteen of the 21 metals identified in beet molasses by spectrographic analysis were precipitated in part by the treatment. The metals known to interfere with citric acid production, particularly manganese and iron, were those most efficiently precipitated. At mash temperatures of 80°C. and approximately 100°C., precipitation was completed within 15 min. after ferrocyanide addition and the reductions in the soluble mineral content in each case were similar. At 25°C. the precipitation reactions occurred but required more than 50 hr. to complete. Significantly more total mineral and more iron, calcium, and copper were precipitated at pHs 4 and 6 than at pH 8; manganese precipitation, however, was not affected by these pH changes.

    Notes: An aflatoxin-producing isolate of Aspergillus flavus was found to be a consistent producer of aflatoxin on all substrates which supported the growth of the mold. In competition with six other selected molds, this isolate was dominant except with one species of Penicillium. Aflatoxin production was parallel to the extent of A. flavus growth whether effected by substrate or competition.

    Notes: A novel and convenient method for the synthesis of guanosine is described. The reaction of AICA-riboside with sodium methylxanthate gave 2-mercaptoinosine in almost quantitative yield. The latter was oxidized with hydrogen peroxide to afford inosine-2-sulfonic acids, which was readily animated to give guanosine in excellent yield. Similarly, the preparation of N2-methylguanosine and N2,N2-dimethylguanosine, minor constituents of transfer RNA, was also accomplished. Furthermore, this procedure was extended to the synthesis of 2′,3′-O-isopropylideneguanosine and the isopropylidene derivatives of various N2-substituted guanosines from 2′,3′-O-isopropylidene-AICA-riboside. Guanosine via 2′,3′-O-isopropylideneguanosine was successfully phosphorylated to give 5′-guanylic acid.

    Notes: A HeLa cell line was propagated in semicontinuous suspension culture, 85 liters final volume, and in continuous flow culture with a volume of 300 ml. or 5 liters in an autoclavable medium to which 8% calf serum had been added. A medium containing 0.1% Methocel and 2% calf serum was also tested. Maximum productivity was obtained at a dilution rate of 0.33 day-1 with a cell density of about 1.0 × 106 cells/ml. The same cell line was also infected with Rubella virus and the production of virus was followed at the 5-liter cultivation level.

    Notes: The purpose of this experimental study was to examine the transient response of a chemostat-type continuous culture of Escherichia coli B to step changes in temperature by following transient limiting substrate concentration and calculating from it the transient growth rate. The transient response to step changes of temperature was tested for four different situations. In the first two cases, temperature was shifted down from 37 to 27°C., and 37 to 32°C. In the last two, it was shifted up from 32 to 37°C., and 27 to 37°C. When the temperature was shifted up, the growth rate increased rather rapidly to its transient maximum value and then decreased slowly until it, settled back into the steady-state value. On the other hand, when the temperature was shifted down, the growth rate decreased relatively rapidly to its transient minimum and then it slowly increased and returned gradually to the steady-state value. The magnitude of the transients was less than would be expected if the transient growth rates followed an Arrhenius function.

    Notes: The inhibitory effect of ethanol concentration p in a medium on the specific rates of growth μ and ethanol production ν of a specific strain of baker's yeast was studied in a chemostat, where except for ethanol as the product, only the concentration of glucose S was controlled to limit the metabolic activity of the yeast. This was designed to supplement the previous findings from the batch experiment, in which ethanol was added artificially and no substrate components were limiting the metabolism of the same yeast, that μ = μ0e-k1p and ν = ν0e-k2p, where k1 and k2 are empirical constants and subscript the 0 denotes respective values at p = 0. The effects of p on the values of μ and ν were confirmed by the Lineweaver-Burk plot to belong to noncompetitive inhibition. The formulas here for μ and ν as affected by p, if extrapolated to the case of no limiting substrates, were in good agreement in respective forms with those derived previously from the batch experiment, though the values of corresponding coefficients in these formulas were different. The differential equations for μ and ν as functions of both p and S and, in addition for the rate of glucose consumption as correlated by the yield factors either with the cell growth rate or the rate of ethanol production, were solved properly with a digital computer. A kinetic, pattern calculated so far was discussed with reference to the data obtained in the batch experiment and those relevant to actual “sake” brewing.

    Notes: A pilot-scale process for the isolation of an aliphatic, amidase from Pseudomonas aeruginosa has been developed. A constitutive, partially irrepressible mutant was employed to give a high initial enzyme concentration. An existing laboratory isolation procedure has been scaled up and modified particularly by substitution of polyethylene glycol for ammonium sulfate precipitation as the first stage in the conversion of the fractionation to continuous operation. Full recovery of activity was achieved with the modification. The recovery of enzyme from a subsequent chromatographic stage was 85% and the maximum overall purification was 28-fold.

    Notes: A growth model for Claviceps purpurea in submerged batch culture is presented. In developing the model, the basic principles of the growth and the morphological properties of C. purpurea are considered. The growth of C. purpurea is assumed to occur in a three-step manner; the first step involves the assimilation and the growth of cells; the second one involves cell division, and the third one involves transformation of the mature cells to a state where they have no ability to divide but do have the ability to produce ergot alkaloids and then they gradually die. Inorganic phosphate is assumed to be the limiting substrate for the first and the second steps in conditions of carbon source being in excess. The model constants are determined by model simulation and graphical searching techniques to find the minimum value of the absolute difference between the experimental and the simulated curves for biomass, alkaloids, and sucrose.

    Notes: β-Galactosidase and other enzymes were immobilized on p-amino-carbanilated derivatives of cellulose and methylol cellulose using the diazo method and through glutaraldehyde. The optimum conditions for coupling cellulose tri-(p-amino-carbanilate) (CTAC) to β-galactosidase were established. The diazo coupling method with CTAC gave greater activity than with glutaraldehyde when coupled to β-galactosidase (Escherichia coli). The stability of the CTAC-β-galactosidase system was examined. The disubstituted p-amino-carbanilate derivative (CDAC) gave a lower activity, whereas the methylol analog (MCTAC) gave slightly greater activity. The CTAC was also used to immobilize glucose oxidase, trypsin, pepsin, and papain.

    Notes: The hydrophobic interaction of β-galactosidase with Sepharose 4B substituted with 3,3′-diaminodipropylamine was studied in both batch and column experiments. The equilibrium and the binding rate constants were determined for different phosphate buffer concentrations. The equilibrium constants exhibit a hysteresis effect, i.e., desorption constants are less than adsorption constants, and the higher the ionic strength to start the desorption, the larger the effect. The rate data are not satisfactorily described by a simple reversible first-order model. The column chromatographic data are semiquantitatively described by a local equilibrium theory without axial dispersion or intraparticle diffusion.

    Notes: Aspergillus awamori NRRL 4869 was cultured on the solid substrate, wheat bran, in a modified Rollacell apparatus to produce α-galactosidase and invertase. The swivel cap on the elongated bottle permits the introduction of air while the bottle rotates. Parameters of air flow rate (0.05-0.2 liter/kg/min), rpm (0.15-15 rpm), and weight of solids (150 and 300 g) were varied. At low air flow rates (0.05 liter/kg solid/min), α-galactosidase production was minimal independent of the rotation rate. At 0.15 rpm and 0.2 liter/kg solids/min air flow rate, invertase production ceased after five days; whereas α-galactosidase production continued. The modified Rollacell can be a useful apparatus for studying solid-substrate cultures.

    Notes: The hemicellulose fraction of ryegrass straw was extracted with NaOH and used for the production of glucose isomerase by Streptomyces flavogriseus. The level of hemicellulose extracted increased proportionately with increasing NaOH concentration up to about 4%, then the rate of increase slowed down. Hemicellulose extraction was facilitated by the combined application of heat and NaOH. Approximately 15% hemicellulose (12% as pentosan) could be obtained by treating straw with 4% NaOH for either 3 hr at 90°C or 24 hr at room temperature. The highest level (3.04 units/ml culture) of intracellular glucose isomerase was obtained when the organism was grown at 30°C for two days on 2% straw hemicellulose. The organism also produced a high yield of glucose isomerase on xylose or xylan. The NaOH-treated straw residue, after removal of hemicellulose, had approximately 75% higher digestibility and 20% higher feed efficiency for weanling meadow voles than untreated straw. Thus, the residue could be used as animal feed. A process for the production of glucose isomerase and animal feed from ryegrass straw was also proposed.

    Notes: In a system where yeast cells grow on n-alkanes dissolved in oil drops suspended in water, the dispersed oil phase will, in most cases, be fully segregated. This means that each drop has its own history that depends on its degree of saturation with yeast cells. This degree of saturation with yeast cells is determined by a stochastic process depending on adsorption, desorption, and cell production. Although many authors mention segregation as a phenomenon likely to occur, so far this segregation has hardly been taken into account. In this paper the interaction of the population of completely segregated oil drops with the population of yeast cells, which results in growth, is described. The consequences of the model are elucidated by the discussion of some extreme cases. The batch fermentation of hydrocarbons by yeast cell is simulated by means of a Monte Carlo procedure. Adsorption, desorption, and production of yeast cells are considered as chance processes. The history of all individual drops is recorder. The influence of the chance of desorption appears to be much larger than that of the chance of adsorption (at the investigated range). Also the size of the inoculum at the start of the process appears to have a strong influence on the course of fermentation.

    Notes: Whole cells of Bacillus sp., a bacitracin-producing bacteria, were immobilized in polyacrylamide gel. The continuous production of bacitracin by an immobilized whole-cell-containing air-bubbled reactor was examined with 0.5% peptone solution. The bacitracin productivity (28 units/ml/hr) obtained with this system was higher than that with a batch system. The effluent bacitracin concentration increased with increasing aeration rate and reached a steady-state maximum above the aeration rate of 3.0 liter/min. A high bacitracin productivity was retained for at least eight days when the gel was washed with sterilized saline at a flow rate of 250 ml/hr for 2 hr once a day. The half-life of the immobilized whole-cell system was about 10 days. Bacitracin productivity by the immobilized whole-cell reactor was higher than that by a conventional continuous fermentation process at high dilution rates.

    Notes: Choline oxidase (choline: oxygen oxidoreductaserpar; was immobilized on a partially aminated polyacrylonitrile membrane. The enzyme electrode, consisting of an immobilized-enzyme membrane and an oxygen probe, was employed for the determination choline. Dissolved oxygen consumption by the enzymatic reaction was measured amperometrically. The rate assay method was used for the choline determination. The response time of the sensor was 7 sec for choline. The choline assay was done within 1 min. The choline calibration curve was linear from 0 to 0.1mM. The response was reproducible within an average relative error of 2.3% when 0.2mM choline was employed for experiments. The choline in the fermentation media was determined by the sensor. Furthermore, phospholipids in the serum were also determined with native phospholiphase D and the enzyme electrode.

    Notes: A theoretical method for predicting the elution profile of a pulse response in an immobilized-enzyme column is proposed. The method is based on a mass balance model, which is extensively used in gel chromatography. To test the method, a pulse of sucrose solution was applied to a column of spherical acrylamide gel in which was entrapped invertase from yeast, and it was eluted with 0.05M acetate buffer at pH 5.0. The elution curves of the substrate and the product were in fairly good agreement with the theoretically calculated ones. The method was extended to the system of reversible as well consecutive reactions.

    Notes: Biomass can be converted to sugars by hydrolysis with enzymes or mineral acids. These sugars can be converted into a number of chemical intermediates in biological reactors. Biological reactions are generally slow and selection of the most efficient reactor is important in these applications. Immobilized-cell reactors allow high cell densities and high throughput by attaching the microorganisms to a fixed support. This paper examines the rate of production of ethanol from glucose by Saccharomyces cerevisia in a packed column. These rates are compared with those for the same reaction in a stirred reactor.

    Notes: The shear viscosities of xanthan batch fermentation broths obey a power-law relation τ = K (P)[ẏ]N(P), where the power-law parameters are functions of xanthan concentration P. within the shear rate range of 1.15 to 384.0 sec-1. Cell-free solutions prepared from these samples exhibit the same values of K and N as the corresponding cell-bearing broth. Further, in the course of the fermentation time, t, it was found that K(P) and N(P) were related by an equation of the form ln K[P(t)] = ln τ0 - N[P(t)]·ln ẏ0, leading to a dimensionless power-law equation valid over the entire fermentation: (τ/τ0) = (ẏ/ẏ0)N[P(t)]. The resulting power-low from[(τ/τ0) = [ẏ/ẏ0]N(t)] indicates that all related samples will exhibit power-low plots passing through the common point, (τ0γ0). These equations have previously been observed with other polysaccharide solution. They are also shown here to be applicable to the pullulan fermentation data of Leduy et at. The parameter K is related to polymer concentration (P) with the correlation K = A PB, where B = 1.75 ± 0.25 for xanthan solutions. With X(t), S(t), and P(t) available from paper I, the correlations of this paper (hereafter referred to as II) [K(t) = f(P)(t) and N(t) = g(K(t))] provide a predictive description of the shear viscosity evolution of polysaccharide-producing fermentation broths.

    Notes: A Lactobacillus sp. isolated from soil and capable of growing on xylose-containing medium exhibited high glucose isomerase activity. The enzyme was thermostable, stable toward dialysis, and activated by heat treatment. It did not show the presence of xylose or ribose isomerase activities; the Km for glucose and xylose substrates were 0.48M and 0.513M, respectively. The heat treatment of ultrasonic crude extract gave insoluble fixed active glucose isomerase enzyme. The properties of free and immobilized enzyme in heat-fixed whole cells differed in many respects. The optimum temperature for enzyme activity changed from 70 to 85°C, the optimum substrate concentration changed from 1.0M to 2.4M, and the optimum pH from 7.4 to 6.0. Co2+ and Mg2+ ions activated the enzyme when used singly, but in combination they inhibited the enzyme and Mn2+ had no effect on the enzyme. Free and immobilized enzymes, when used in the used in the conversions of corn and bagasse hydrolysates to fructose, gave 58, 25.6%, and 50, 27.6% conversions, respectively. Immobilized enzyme retained a significant activity for more than 30 hr and was able to operate at higher glucose concentrations showing less products inhibition effect as compared to free enzyme. In the batch process it was able to operate for about eight cycles.

    Notes: Continuous production of L-alanine from L-aspartic acid using immobilized Pseudomonas dacunhae was investigated. Pseudomonas dacunhae cells were immobilized with carrageenan gel. The L-aspartate β-decarboxylase activity of immobilized cells was enhanced by incubating the immobilized cells with a solution of 1M ammonium L-aspartate (pH 5.5) containing 0.1mM pyridoxal-5′-phosphate (PLP) at 37°C over 20 hr. The enzyme activity of immobilized cells was 59% that of intact cells. The pH profile of the enzyme reaction was broader in the immobilized cells than in the free cells. The enzyme activity of immobilized cells was maintained through repeated uses when a substrate solution containing 0.1mM PLP was used. Complete conversion of L-aspartate to L-alanine was attained when a solution of 2M ammonium L-aspartate (pH 6.2) containing 0.1mM PLP was passed upward through the immobilized cell column at a retention time of 8 hr at 37°C. Glutaraldehyde treatment of the immobilized cells resulted in a slight decrease of the enzyme activity but a marked increase of the operational stability. The half-life of enzyme activity was 46 days in glutaraldehyde-treated immobilized cells.

    Notes: β-Xylosidase from a commercial Aspergillus niger preparation was purified by differential ammonium sulfate precipitation and either gel permeation or cation exchange chromatography, giving 16-fold purification in 32% yield for the first technique or 27-fold purification in 19% yield for the second. The second method in addition almost completely removed interfering β-glucosidase activity. Enzymes prepared by this method was immobilized to 10 different carriers, but only when it was bound to alumina with TiCl4 and to alkylamine porous silica with glutaraldehyde were substantial efficiencies and stabilities achieved. With alumina, the variation of activation procedure, amount of β-xylosidase offered, and activation solution composition yielded maximum activities of over 40 U/g with approximately 70% immobilization efficiency. Variation of binding pH and incubation time led to a maximum immobilized activity of 1.3 U/g with 78% immobilization efficiency on silica.

    Notes: Cells form the yeast Hansenula polymorpha (ATCC 26012) were successfully immobilized by entrapment in a polyacrylamide gel. The resulting gel showed high methanol oxidase activity especially after treatment with a detergent (CTAB). The enzymatic properties of the gel-entrapped cell were not very different from that of the soluble enzyme except that no inhibition was observed at high methanol concentration. In continuous reactors, the gel-entrapped cells showed a much higher stability than other enzyme preparations. The inactivation mechanism was investigated and proved to be the oxidation of essential SH group(s) of the methanol oxidase molecule by hydrogen peroxide. Treatment with β-mercaptoethanol prevented inactivation or regenerated activity.

    Notes: Polymethylglutamate (PMG), a synthetic polypeptide, was used as a new carrier to immobilize urease (EC 3.5.1.5) and uricase (EC 1.7.3.3) by the azide method. The enzymes could be immobilized onto PMG in various forms, such as film, fiber, coating on various beads, and a silicon tube. The retained activities of the immobilized enzymes were excellent (more than 95%), therefore it was possible to immobilized almost all activities of the enzymes added in the coupling mixtures. Heat stabilities of the resulting immobilized enzymes were markedly improved, while the optimal pH and Km values remained almost unchanged. The urease immobilized on the PMG-coated glass beads packed in a column, was found to retain its activity more than 80% of the initial value, even after the occasional use for a year. In view of the improved retained activities and stabilities of the immobilized enzymes, PMG may therefore be a very versatile matrix for the immobilized enzymes.

    Notes: Heat conduction solution enable rapid determination of the heats of aerobic and anaerobic metabolism of substrate by microorganisms. Aliquots of 1.0 ml cell suspension, 5 × 109 cell/ml, were mixed with a few dozen nmol substrate contained in 0.5 ml, under a controlled atmosphere of air, O2, or N2. At these substrate concentration, with adapted microorganisms, metabolism and its heat generation are usually complete within 300 to 600 sec. The raw data yield ΔHapp values. The ΔHapp were determined in the range 0.001 to 0.010% substrate, and extrapolated (limit substrate concentration →0), to yield Δ0H̄, the limiting differential molar heat of metabolism. The Δ0H̄ values express the heat generated when there is rapid metabolism but little new growth, minimal contribution by H+ transfer from metabolites, and maintenance of aerobicity or anaerobicity as specified. Escherichiacoli B/5 was used for aerobic and anaerobic combustion of eight sugars. Pseudomonas multivorans, and an Acinetobacter, strain B-1, were used for aerobic metabolism of benzene, toluene, naphthalene, and a methylnaphthalene. The larger heats of combustion of the hydrocarbons enable the use of aqueous solutions of hydrocarbons well below their solubility limits. The quotient Δ0H̄/n (n = atoms carbon/molecule substrate) varies from (-)36 to (-)67 kcal/mol carbon for the sugars. The most reduced sugar yields the largest exothermic heats. The quotient varies from (-)27 to (-)81 kcal/mol carbon for the aromatic hydrocarbons. Comparison of the calorimetric heats of metabolism of those from total aerobic combustion in aquo (where available) give measure of the efficiencies with which the heat contents of the aqueous substrate are used by the bacteria.

    Notes: A flat-bed hollow-fiber cell culture system has been developed which maximizes the utilization of the large fiber surface while diminishing significantly the problems inherent in a cartridge-type reactor. The reactor core consists of a shallow bed of hollow fibers sandwiched between two stainless-steel microporous filter plates through which the media flow is directed normal to the plane of the fiber bed. Reactors with both 930 and 9300 cm2 of fiber surface have been successfully constructed and operated. A variety of cells has been grown in these reactors including SV3T3 cells, baby hamster kidney cells, Vero cells, and rhesus money kidney cells, and cell products such as plasminogen activator and migration inhibition factor (MIF) were produced. This system offers an excellent prototype for scaleup design.

    Notes: Candida Krusei has a optimum growth temperature of 37°C on SASOL ethanol-isopropanol mixture. The organism was unable to grow on isopropanol, but oxidized it partially to acetone in the presence and absence of ethanol. Growth at 40°C in the alcohol mixture was slightly faster than at 30°C over an ethanol concentration range of 0.43 to 3.6% (v/v), although at both temperatures the growth rate declined continuously with increasing concentration. At an ethanol concentration greater than 3.6% (v/v), the mixture was much more inhibitory to growth at 40 and 30°C. The inhibitory effect was due to the ethanol rather than the isopropanol. Metabolites such as acetate, acetaldehyde, and ethyl acetate accumulated in the medium, but the degree of accumulation depended upon the temperature and alcohol mixture concentration. At 40°C, acetaldehyde and acetate accumulated to a greater extent than 30°C on a 4.0% (v/v) synthetic alcohol mixture and this may also cause the greater inhibition at this temperature. The alcohol mixture is unsuitable for single cell protein (SCP) production in batch culture because of the low cell densities observed at all alcohol concentrations.

    Notes: It has been shown both theoretically and experimentally that interphase nitrogen transport may have a significant influence on the rate of interphase oxygen transport, and thereby also on the value of the volumetric mass transfer coefficient of oxygen, kla, determined in mechanically agitated bubble fermentors using the variants of dynamic method presented in the literature. The experiments were carried out in 1M KCI solution at five stirrer frequencies and two gas inlet levels. The gas interchanges were performed either without interrupting the aeration and agitation of the charge (A) or with the aeration and agitation of the charge turned on at the same time (B). The applied variants of the interchange were N2→ O2→, O2→ N2, N2→ air, air→ N2, O→ O2, and O→ air. In the two last variants the oxygen dissolved in the charge was removed by reacting with sulfite ions. The kla values calculated by allowing for the nitrogen transport for procedure A were approximately equal to the values obtained by disregarding the nitrogen transport, whereas those for procedure B were higher (up to 40%), than the values obtained disregarding the nitrogen transport.

    Notes: The effect of partial recycle of microbial cells on the operation of a chemostat has been investigated for two fermentations. Stable steady states without partial cell recycle were obtained for the conversion of D-sorbitol to L-sorbose by Gluconobacter oxydans subsp. suboxydans 1916B and for the conversion of glucose to 2-ketogluconic acid by Serratia marcescens NRRL B-486. The employment of partial cell recycle dramatically increased product formation rates for both fermentations.

Still, the role of formaldehyde, produced during methanol assimilation, upon growth of yeasts is not clear. In the present paper, an attempt has been made to throw some light upon this aspect. Starting with a basic frame work for methanol uptake by yeasts, several models were developed assuming different modes of regulation of key enzymes by methanol and/or formaldehyde. The main feature of the basic framework consists in consideration of two routes for oxidation of formaldehyde to CO2, one associated and the other not associated with production of energy. Further, the rate of energy production form the energy-associated oxidation of formaldehyde is assumed to be controlled by the rate of energy consumption by anabolic reactions. The models were discriminated by subjecting these to biological constraints. As a result, the successful model suggests that in spite of higher inherent toxicity of formaldehyde, methanol exerts the controlling influence upon growth under normal conditions.

    Notes: A generalized mathematical model, previously developed and experimentally validated, was modified and used to computer-simulate the outcomes of adding two systems for increasing the concentration of cell mass in the fermentor, and thereby the rate and extent of substrate conversion, in a dialysis continuous process for the ammonium lactate fermentation of deproteinized whey. The addition of a nondialysis continuous prefermentor would not improve the process. However, the addition of cell retention time would be reduced ninefold, and the extent of converting substrate to product would be increased 25% and accomplished almost entirely by maintenance metabolism.

    Notes: Selected samples of waste microbial biomass originating from various industrial fermentation processes and biological treatment plants have been screened for biosorbent properties in conjunction with uranium and thorium in aqueous solutions. Biosorption isotherms have been used for the evaluation of biosorptive uptake capacity of the biomass which was also compared to an activated carbon and the ion exchange resin currently used in uranium production processes. Determined uranium and thorium biosorption isotherms were independent of the initial U or Th solution concentration. Solution pH affected the exhibited uptake. In general, lower biosorptive uptake was exhibited at pH 2 than at pH 4. No discernible difference in uptake was observed between pH 4 and pH 5 where the optimum pH for biosorption lies. The biomass of Rhizopus arrhizus at pH 4 exhibited the highest uranium and thorium biosorptive uptake capacity (g) in excess of 180 mg/g. At an equilibrium uranium concentration of 30 mg/liter, R. arrhizus removed approximately 2.5 and 3.3 times more uranium than the ion exchange resin and activated carbon, respectively. Under the same conditions, R. arrhizus removed 20 times more thorium than the ion exchange resin and 2.3 times more than the activated carbon. R. arrhizus also exhibited higher uptake and a generally more favorable isotherm for both uranium and thorium than all other biomass types examined.
