    Notes: Abstract Cultural conditions affecting lignin degradation by Phanerochaete chrysosporium in various lignocellulosic materials were studied in comparison to an isolated lignin preparation. With shallow mycelial cultures, the degradation of lignin in wood proceeded more slowly in a 100% O2-atmosphere than in an air atmosphere, indicating that pure oxygen was toxic to the fungus. The organism was able to degrade lignin efficiently even under 30% CO2 and 10% O2 concentrations. Evolution of 14CO2 from labelled lignocellulosic materials was shown not to be representative of total lignin degradation. Addition of glucose to the culture did not affect lignin degradation measured by 14CO2 evolution, whereas lignin degradation measured by Klason lignin method stopped completely (poplar) or slowed considerably (straw). Due to partial depolymerization of lignin to soluble products, measuring only the evolution of 14CO2 results in an underestimation of the total amount of lignin bioaltered. The soluble products from all of the tested lignocellulosic materials and from the isolated lignin had an average molecular weight of about 1,000 and the products could be further fractionated by ion exchange chromatography. The relative amount of these products could be varied from 15 to 45% from the original lignin.

    Notes: Abstract One isoenzyme of malate dehydrogenase with an isoelectric point of 6.4 was found in glucose-repressed cells of Schizosaccharomyces pombe. During respiratory derepression the activity of this isoenzymes decreased rapidly in vivo. In the course of this inactivation two new forms of malate dehydrogenase with isoelectric points of 6.0 and 5.7 appeared. It has been found that these two enzymic forms disappeared 4 h after the exhaustion of glucose; probably they are degradation products of the isoenzyme present in glucose-repressed cells. Fully derepressed cell of this fission yeast contain one isoenzyme of malate dehydrogenase with an isoelectric point of 5.3. The synthesis of this isoenzyme is initiated at glucose concentrations below 1.5 g/l.

    Notes: Abstract Trichosporon cutaneum metabolizes glucose purely oxidatively and cytochrome P450 was not detected in the reduced CO-difference spectrum of whole cells. However, in the isolated microsomal fraction the corresponding monooxygenase was present as shown by the appearence of cytochrome P450, NADPH-cytochrome c (P450) reductase and cytochrome b5. The absorption maximum of the terminal oxidase in the reduced CO-difference spectrum shifted between 447 and 448 nm. Derepression of biosynthesis of all components was achieved by transition of the cells from carbon- to oxygen-limited growth in continuous culture. The monooxygenase exhibited aminopyrine demethylation activity but not ω-hydroxylation activity of lauric acid. With respect to the growth limiting nutrient (carbon and oxygen respectively), mitochondrial cytochrome content showed an analogous behavior as cytochrome P450 and cytochrome b5.

    Notes: Summary Rates of oxygen uptake and the oxygen demand during growth of Candida tropicalis on hexadecane and glucose were determined in batch experiments. Oxygen demand was 2.5 fold higher for the synthesis of one unit of cell mass from hydrocarbon than from glucose. On the other hand specific respiration is of the same order of magnitude for both substrates, e.g. 12 mmoles O2xh-1xg-1 (dry weight) and seems to be a constant of this organism. Higher rates of oxygen supply into the medium had no effect on the specific rates of respiration. Specific growth rates on hexadecane were 2.4 times lower than on glucose. It is concluded, that rates of synthesis of cell components are controlled by the overall capacity of the respiratory pathways.

    Notes: Abstract The surface structure of the hypdrocarbon-utilizing yeast Candida tropicalis was investigated by scanning and transmission electron microscopy (SEM and TEM respectively). The sample preparation technique was based on a rapid cryofixation without any addition of cryoprotectants. In subsequently freeze-dried samples the surface structure was analysed by scanning electron microscopy. Thin sections were prepared from freeze substituted samples. Both techniques revealed hair-like structures at the surface of hydrocarbon-grown cells. The hairy surface structure of the cells was less expressed in glucose-grown cells and it was absent completely after proteolytic digestion of the cells. When cells were incubated with hexadecane prior to cyryofixation a contrast-rich region occured in the hair fringe of thin sections as revealed by TEM. Since these structures were characteristic for hexadecane-grown cells and could not be detected in glucose-grown or proteasetreated cells it was concluded that they originate from hexadecane adhering to the cell surface and are functionally related to hexadecane transport. The structure of the surface and its relation to hydrocarbon transport are discussed in view of earlier results on the chemical composition of the surface layer of the cell wall.

    Notes: Summary The carbohydrate composition and the specific activity of the trehalase of cyclic partially synchronised yeast populations have been investigated. Under glucose limitation and appropriate cultural conditions synchronous growth in a chemostat was achieved. The cells accumulated the reserve carbohydrates during the single cell phase between two buddings. The rapid degradation of part of these reserves began shortly before the swelling of the bud. The importance of the mobilisation of endogenous reserves for the development of the cell is discussed. The specific activity of the trehalase changed during the budding cycle. The result gives rise to the assumption that the synthesis of this enzyme is linked to the growth cycle.

    Notes: Abstract Four major hemoproteins were purified by isoelectric focusing from an extracellular crude enzyme preparation, produced by the white rot fungus Phanerochaete chrysosporium under carbon-limited conditions. Both the crude enzyme and the purified proteins oxidised milled wood lignin, HCl-dioxane-extracted straw lignin and alkali straw lignin in the presence of hydrogen peroxide. The oxidation resulted mainly in further polymerisation of the lignins and was enhanced by addition of veratryl alcohol to the reaction mixture. Alkali straw lignin was also polymerised by horseradish peroxidase, although veratryl alcohol had no influence on this reaction.

    Notes: Abstract Extracellular H2O2-dependent ligninase activity of Phanerochaete chrysosporium was produced in agitated culture conditions when veratryl alcohol or veratraldehyde were added to the cultures. The enzyme production was suppressed by cycloheximide indicating that true protein synthesis occurred. The activated cultures were also able to degrade synthetic lignin. Reduction of veratraldehyde to corresponding alcohol during secondary metabolism was a good indicator of the effect of agitation on cell metabolism. Too high agitation speed led to complete inhibition of both the reduction reaction and the ligninolytic activity.
