    Description: The proximate composition and amino acids assay of smoked Clarias gariepinus and Oreochromis niloticus were determined using standard procedures. The mean proximate composition of Oreochromis niloticus are 8.9% moisture, 16.00% lipids, 5.6% ash, 44.5% crude protein, 0.42% crude fiber and 24.9% NFE while Clarias gariepinus has 8.19% moisture, 15.55% lipids, 4.725 ash, 40.46% crude protein, 0.50% crude fiber and 30.58% NFE. There is therefore a nutrient variation between the 2 species of fish Oreochromis niloticus has higher crude protein, lipids, moisture and ash content than Clarias gariepinus.The amino acids profile shows that both fishes have 17 amino acids. Apart from Tryptophan, the following essential amino acids Histidine, Methionine, Threonine, Isoleucine, Leucine, Lysine, Valine and Phenylalanine are present in both fish. Oreochromis niloticus however has higher quantities of the essential amino acids except for Histidine than Clarias gariepinus

    Description: Cephalic abnormality in nine mating groups involving Clarias gariepinus with cephalic abnormality, non Cephalic and Maiduguri origin were investigated to determine the level and the genetic basis of the occurrences and its aquaculture implication. The highest mean percentage survival in a Cephalic crossed with non cephalic group was 68.6% whereas the last mean percentage survival was 25.7% in the group of Maiduguri cross with Maiduguri parent. Cephalic abnormality was observed in the crosses with both female and male cephalic with a total frequency ranging from 0 % to 70.8. %. The least number of cephalic abnormality was 18.5% which involved a cross of non cephalic with cephalic, and the highest was 70.8%. This level of cephalic abnormality shows that the defect was hereditary. This result implies that, the genetic factor is a major contributor in the feature of cephalic abnormality in Clarias gariepinus.

    Description: The evaluation of Cr. Cu, Mn, Fe, Ni, Zn,As and Pb concentrations in Clarias gariepinus and Oreochromis niloticus from Lake Geriyo Yola,Adamawa, Nigeria using X - ray fluorescence (XRF) technique is discussed. The analytical samples are irradiated with high energy electrons of 109Cd to produce fluorescent X-rays was produced which passes to the Silicon-lithium detector through Mo target as a source of monochromatic X-rays. The spectrum of energy generated on the detector was processed by a Multi- Channel Analyzer to obtain analytical data. The intensity of the fluorescent X-rays on the detector is proportional to the concentration of the individual element of interest in the sample. This method can identify up to 30 or more elements at the same time. The results showed varying levels of heavy metals in the fishes. The concentrations of Cr, Mn, Cu, and Ni in the fishes were much higher than WHO and PEPA maximum permissible limits, while the concentrations of Zn and Pb were lower than the standards. The results suggest that the lake is polluted with Cr, Mn, Cu and Ni and the consumption of fishes of the lake is life threatening to man.

    Description: The contribution of tilapia aquaculture in Nigeria to world output is negligible due to stunting, poor market value among others. This paper evaluates the aquacultural credentials of tilapia, some genetic improvement technology in cultured tilapia, namely, ploidy, hormonal sex reversal. transgenic, hybridization, and the necessity of Genetic Improvement in accelerating tilapia production in Nigeria. Investigation reveals the presence of O. niloticus with the highest growth perfermancc index (~h1=3.11) for Lake Kanji which indicates high growth potential in suitable culture environment and could serve as a good starting point for genetic development. The presence of ”wesafu”, an ecotype cichlid, endemic to Epe lagoon, Lagos, which grows to 1500g in the wild, appears to be an excellent candidate for genetic improvement of a commercial strain for the growing aquaculture industry. Tilapia Genetic improvement in Nigeria is faced with a number of setbacks. This includes short- term, scattered and disjointed funding, inadequate genetic research facilities, ecological risk, inadequate skilled manpower and poor documentation of tilapia genetic resources among others. Considering the growing importance of tilapia culture, the need to document, conserve, evaluate and utilize tilapia genetic resources is highlighted to enhance the success of food security in Nigeria.

    Description: Tilapia guineensis eggs were spawned and hatched in different salinities (17, 12, 7, 5, 2 and 0) ppt. The study was conducted in eight replicates. The aim of the investigation was to ascertain disparity in hatching size of Tilapia guineensis attributable to salinity. Immediately hatching was observed, body weight of five hatchlings in each replicate was individually weighed in milligramme. The individual total length of the five hatchlings was also measured in millimeter. The height of the yolk sac larva was determined with micro-metre screw gauge graduated in millimeter from the top of the yolk sac to the dorsal region of the larva. Data obtained were used to calculate yolk sac volume. Significant differences (P〈0.01) were observed in body weight and total length of larvae hatched in different salinities. The heaviest and longest larvae were hatched in 12 ppt saline water. The yolk sac volume of larvae hatched in lower salinities were significantly higher (P〈0.01) than the yolk sac volume of larvae hatched in higher salinities. The smallest yolk sac volume was recorded in 12ppt saline water. Our findings show that salinity could be manipulated for economic production of heavier seeds of Tilapia.

    Description: The water analysis of five selected ponds in Ado-Ekiti in Ekiti State, was carried out to determine their suitability for fish culture. Pa rameters analyzed were: Temperature, Turbidity, Carbon (IV) oxide, Alkalinity, Hardness, pH and Dissolved oxygen. Values for temperature ranged between 20.0~'C and 29.0~'C, Turbidity values were between 46.0-50.2cm, Carbon (IV) oxide: 11.5-12.2mg/litre; alkalinity: 100-120mg/liter; hardness: 110-200mg/litre; pH: 6.8-7.8 and dissolved oxygen: 5.8-6.8mg/liter. The values obtained were found to be within acceptable range for fish production.

    Description: Staphylococcus aureus is a common organism on the skin and in the nasal passages of approximately 50% of the population. Heat-treated seafood may become contaminated with this organism by poor handling, then storage at improper temperatures allows the organism to multiply and produce its toxin. This type of food poisoning may be avoided by practicing strict personal hygiene, thorough cleaning and disinfection of equipment, and storage of susceptible food at temperatures below 10°C or above 60°C. Presented in the paper is the methodology of determining S. aureus in fish and fishery products.

    Description: Streptococci are gram-positive cocci, sometimes coccobacilli, arranged in chains. This group of streptococci resides in the intestine of warm-blooded animals. They are bile resistant and capable of growth at 45°C. Fecal streptococci form part of the microflora of many foods without necessarily indicating poor hygiene. They are found in many fermented foods, such as cheese and raw sausage, and often take part in the fermentation process. However, in meat products which have received a severe heat process, the presence of excess numbers of fecal streptococci indicates unhygienic handling and/or faulty storage. Presented in the paper is the methodology of determining the presence of fecal streptococci in fish and fishery products.

    Description: A review is made of studies conducted on cryoprotectants for use in the frozen storage of processed fishery products, such as surimi. Some 150 compounds were screened for cryoprotective effects on fish actomyosin; the findings of experiments investigating the behaviour of carp actomyosin, and fish myosin, actin and other constituent proteins during frozen storage are described. The mechanism of freeze denaturation and of the effects of cryoprotectants is outlined and the application of cryoprotectants in the development of new fish gel products considered.

    Description: This study focused on the assessment of Terminalia suparba (bark, leave and root) based diet to improve the ovulation of Clarias gariepinus brood stocks. Ten female and ten male brood stocks were obtained from a reputable fishfarm in Ogun State, Nigeria and were fed with the diets for four weeks before the breeding exercise. The latency period observed in treatments 1 to 4 was 4hours 30minutes while the ovulation period in treatment 5 was 8 hours. There was no significant different (P〉0.05) among the means. The brood stocks in Treatment 2 (Root) has the highest fecundity count (2001) while those in Treatment 5 (control) has the least fecundity count (1496). The phytochemical analysis of the plant materials showed the presence of Alkaloids, Tannin, Phenol, Glycoside, Flavonoids, Steroids, Phlobatannin, Saponin, Anthraquinones. The physical and chemical parameters of water observed showed that dissolved oxygen value ranges from 3.28 -4.60, pH 6.5 - 8.5, temperature 28.0 - 28.8 and conductivity 8.9 - 9.20. It is therefore concluded that the inclusion of Terminalia suparba in the feed of Clarias gariepinus broodstock will not only improve ovulation but also reduce the latency period.

    Description: Application of genetic manipulation techniques on cultured fish have been employed in developed economies to increase food fish yield. In this study, a simple and safe biotechnology genetic manipulation technique was applied to produce triploid African giant catfish (Clarias gariepiirus Burchell 1822) larvae .Eggs numbering 100~c10 in quadruplicates from C. gariepinus were activated by milt from the same species and were subsequently transferred to a thermoregulated refrigerator maintained at 2degreesC for 20 min to suppress cell division, starting at 4 min after activation. Haploid larvae were produced by activating eggs with UV irradiated milt at 30000 uWcm-2 for 15 min. Fertility, hatchability and survival after one week for triploids were 82.5%, 69.8% and 61.3% against haploid and diploid controls, 100%, 15%, 0% and 100%,93%, 91 % (p 〈 0.05) respectively. Ploidy levels of the embryos cytogenetically were evaluated in day-old posthatched embryos incubated in 0.02% colchicine for 2-4 h. Head and yolk sac were discarded in 0.9% NaCl while remaining fragments were later treated with hypotonic solutions of distilled water for 5 min, and subsequently with diluted catfish serum 1: 4 for 25 min. Cells were fixed with 1:3 acetic acid ethanol mixture. Slide with the chromosome spreads were then stained with 20% Giemsa in phosphate buffered solution. Chromosome numbers obtained were 28~c2, 56~c2, and 75~c2 for haploid control, diploid control and triploid treatments respectively. Using a simple biotechnology technique as above, triploid C. gariepinus larvae were produced in the present study. The potential application on food fish security of triploid catfish in aquaculture especially in Nigeria is discussed.

    Description: One of the priority research projects of the Nigerian Institute for Oceanography and Marine Research (NIOMR) is broodstock development and mass propagation of silver catfish, Chrysichthys nigrodigitatus of which the broodstock is naturally abundant in New Gala River. Selected physical characteristics and water physico-chemical parameters of the upper reach of the New Calabar River at Aluu were investigated. The highest depth value of 9.20 m was recorded in the middle of the river. The width of the river measured during spring high tide and low tide were 174 m and 110 m respectively. The cross-sectional area and average flow velocity of the river were 797.13 m2 and 0.374 m/s respectively. The discharge of the river was computed as 298.13m3/s. The river water was acidic most of the period (Hydrogen ion concentration, pH: 5.12 to 7.43), with low total alkalinity values (4.00 to 12.00 mgCaCO3/L) and low total hardness values (11.5 to 51.2 mgCaCO3/L) which indicate that the river water is black and soft. There was significant (p~,0.05) seasonal variation in temperature (23.1 to 31.7~'C\}, Secchi transparency (9.30 to 19.00 cm), total hardness (11.5 to 51.2 mgCaCO3/L), dissolved oxygen (5.20 to 7.50 mg/L) and dissolved oxygen deficit (0.02 to 2.95mg/L). These results are useful for several stake holders who use the river as a sink for trade wastes, as source of water for fish farming, as source of broodstocks of Chrysichthys nigrodigitatus, as raw water for water treatment plants, and as means of transportation of industrial materials.

    Description: Eight month physicochemical study of Dadin Kowa Dam was carried out forthnightly from July, 2011 to february 2012. Air temperature ranged from 18.25~'C ~c 28~'C ~c 0.50 to 28~'C ~c 2.16. Water tmperature ranged from 18.75~'C ~c 2.21. Transparency ranged from 11.00cm ~c 1.83 to 29.63 cm ~c ~c 2.14. Conductivity ranged from 13.00~Ks/cm ~c 13.33 to 41.38~Ks/cm ~c 5.54. Hydrogen ion (pH) ranged from 6.69 ~c 1.03 to 8.39 ~c 1.17. Dissolved oxygen ranged from 6.39mg/l ~c 1.03 to 10.14mg/l ~c 1.17. Total alkalinity ranged from o.69mg/l ~c 0.38 1.34mb/l ~c 0.02. Total ammonia ranged from 0.20mg/l ~c 0.027 to 0.83mg/l ~c 0.05. There were various levels of correlation between these parameters. There was no significant difference between both sites and months (p〉0.05) of all the parameters except total nitrogen which show a significant difference between the sites (p〈0.05) and transparency also shows a significance between the months (p〈0.05). All physicochemical parameters were almost within the tolerable limits for supporting aquatic life.

    Description: This study looked into the physicochemical and bacteriological investigation of fish and water in Aiba Reservoir. Water and fish samples collected were studied to assess their bacteriological and physicochemical characteristics and their suitability for domestic purposes. Structural questionnaires were administered to the fishermen around the reservoir and experiments were carried out in the laboratory to analyze the samples. The results showed that the total viable bacteria count was high(1.02 x 105 cfu/ml) while few Staphylococcus aureus and E. coli occurred insignificantly. The population of S. aureus and E. coli in the fish gills, liver and tissue were insignificant. The results obtained from the physicochemical analysis of the water samples showed that the reservoir has optimum water temperature of 29degreesC ~c 0.25, a pH of 7.70 ~c 0.20, dissolved Oxygen level of 7.20mgO2/1 ~c 0.20 and total hardness of 45.00 mg/ICaC03 ~c 1.05. The microbial load in the fish sample is higher than that in the water sample. Further strict hygienic measures and surveillance is recommended so as to improve the water quality and to avoid deleterious effects on the health of the Iwo inhabitants and its neighboring communities who also benefit from the reservoir.

    Description: A comparative analysis on biochemistry and Polyacryl Amide Gel Electrophresis was carried out to determine the genetic diversity of diploid and triploid Heterobranchus bidorsalis. Sixteen samples of diploid and triploid farm-raised (mean weight; 512.6g and mean length; 41.6cm) were collected and the electrophoresis analysis was conducted using 5.5% Polyacryl Amide Gel and serum protein obtained from the blood of the live samples. 0.06% Coomassie blue was used for staining the gel while a mixture of ratio 1:2 of glacial acetic acid, meethanol and distilled water was used for de-staining the gel. The diploid and triploid possessed an equal total number of 23 electrophoretic protein bands. The molecular phylogenetics of both samples revealed low genetic variability. Results of this study will serve as a baseline analysis on the current genetic diversity of H. bidorsalis in Nigeria.

    Description: Comparative primary production in Little Stream Farms, near Uyo was investigated in relation to physico-chemical parameters of the adjacent perennial stream and the fish ponds. Except for temperature, which showed the same values for both stations, physico-chemical variables generally showed significant difference (P〈O.05) between the pond water and the adjacent stream. Values of silicates, Turbidity (NTU), Secchi Disc Transparency and Total Suspended Solids (TSS) exhibited significantly variable trends (P〈O.05), especially in the stream water, in response to the commercial sand dredging activities upstream from the fann location. Lowest value of stream transparency (O.1M) recorded was during the peak of the sand dredging, whereas the stream water showed absolute transparency in the absence of the dredging. The water quality parameters also exhibited a general trend of significant seasonal variation (P〈O.05). Five phytoplankton families: Bacillariophyceae, Chlorophyceae, Cyanobacteria, Euglenophyceae and Dinophyceae occurred in the ponds, while three families, Bacillariophyceae, Chlorophyceae and Cyanobacteria occurred in the stream. Bacillariophyceae showed the highest relative abundance in both stations A and B.

    Description: The physico-chemical parameters of Lake Alau were monitored for four months to assess the water quality. Parameters such as temperature, transparency, conductivity, dissolved oxygen, biochemical oxygen demand, Ca, P, etc were studied in five stations (A, B, C, D and E). Water samples were collected monthly from 8:00 to l2:00noon, for four months (July - October 2008, wet season) and analysed. The physico-chemical parameters were within the ranges of unpolluted water bodies. Variation occurs in almost all the months of the study. Significant difference (P〈0.05) occurred in pH, transparency and Ca among the stations, and insignificant difference (P〉0.05) in temperature, conductivity, dissolved oxygen, biochemical oxygen demand, Mg and P. These variations may be due to effects of fertilizer application, herbicides and insecticides in irrigated farms around the Lake.

    Description: Study of some Physico-chemical parameters and fish species composition of Doma Darn was carried out between the month of August 2012. Fishing communities along the dam were selected at sampling stations and collection of fish species. The values of physico-parameters obtained for stations were in the range of 28.10-92 m for depth, 27.70-28.48~'for temperature, 7.03-7.64 for PH, 7.23-7.64mg/l for dissolved oxygen (DO), 30.08-31.75 ntu for turbidity, 94.64-105.17mg/l for alkalinity and 79.17-80.58mg/l for total hardness. The result indicated that there was significant difference (p〈0.5)for depth, temperature turbidity and alkalinity, while there was no significant difference (p〈0.05) for DO, pH and water hardness among the stations. The result of these parameters among the duration shows that there was significant different (p〈0.05) in all the parameters except pH. The study revealed that there were twenty-five (25) fish species belonging to 12 families in the dam. The family Cichlidae and Mochokidae and Bagridae which has three each, While the family, Polypteridae, Channidae, Centropomidae, flepsetidae and Protopteridae has the lowest number of one (1) each. The overall result revealed that the Phvsico-chemical parameters were mostly influenced by duration and not much by stations; and parametersoaram measured were within the recommended water quality requirement for growth and survival of aquatic organisms.

    Description: Cholera is an acute specific infection caused by the organism, Vibrio cholera. Diagnosis may be confirmed by the presence of large numbers of the comma-shaped bacilli on direct microscopic examination of a fecal or vomitus smear, and by the isolation of the organism on culture. Fish and shellfish have been identified as vehicles of cholera. Large numbers of V. cholera must usually be ingested to cause cholera. Thus problems often occur when poor handling and inadequate refrigeration have allowed the organism to multiply. Presented in the paper is the methodology of determining the presence of Vibrio cholera in fish and fishery products.

    Description: Food poisoning due to Vibrio parahaemolyticus is a food-borne infection resulting from the ingestion of a large number of this organism (about 106-109 viable cells). The major symptoms are diarrhea and abdominal pain with headache, fever, and vomiting also occurring. The organisms are excreted during the acute stage of the illness after which they decrease rapidly. The differentiation of V. parahaemolyticus from other pathogenic species of Vibrio is based mainly on salt tolerance, Voges-Proskauer reaction, fermentation of sucrose, and growth at 43°C. Presented in the paper is the methodology of determining the presence of Vibrio cholera in fish and fishery products.

Trapping with methanesulfonyl azide (5a) of 10 in situ or subsequent treatment with trifluoromethanesulfonyl azide (5b) gives mixtures of colourless (15) and intensely yellow N-sulfonylimino-1,4-benzodiazepines 16 along with products due to cleavage of the exocyclic double bond of 10, viz. 11 and 13. The ethylidene compound 10b yields the bicyclic products 18 and 19, apparently by complex sequences of reactions that are triggered by removal of the acidic proton at C-2 of 16b and 16f. The structures of the products are based on spectroscopic evidence and X-ray diffraction analyses performed on 15b, 16d, 16e, and 19.

    Notes: ---The rate constants for the reactions of the 2-azaallenium ion 1b+, the 2-azaallylium ion 2a+ and the iminium ion 3+ with different nucleophiles were determined by 1H NMR spectroscopy. By correlation with the Linear Free Enthalpy Relationship (LFER) lg k20°C = s (E + N), developed by Mayr and Patz, the electrophilicity parameters E(1b+) = -3.7, E(2a+) ≍ -16 and E(3+) = -10.43 were obtained. They show that the relative reactivities of these ions are approximately 1012:1:106. Quantum chemical calculations (ab initio, DFT) of the methyl anion affinities for the ions 1b+,2a+ and3+ are in agreement with the experimental E values. The X-ray structure of 3+·CF3SO3- is reported for the first time; it shows no strong interaction between the cation and the anion.

    Notes: ---For comparison to the recently described 2,3,6,7-tetrakis(dimethylamino)naphthalene (1) the three isomers 2,3, and 4 were synthesized. The basicities of this group of isomers are strongly dependent upon the different mutual orientations of the pairs of dimethylamino substituents: only the isomers 3 and, partially, 4, both with dimethylamino groups in adjacent peri-positions of the naphthalene, are strong “proton sponges”. For the isomers 1 and 2 with the same number and kind of twofold dimethylamino substituents in neighbouring ortho-positions, however, no significant basicity increase is observed. To explain this difference between the two groups of isomers it is suggested that in the ortho-pairs of 1 and 2 the C-N bonds diverge considerably, leading to an increased N···N distance and consequently to less stable [N···H···N]+ hydrogen bonds in contrast to the parallel C-N bonds in the peri-substituted isomers 3 and 4. X-ray crystal structure analyses of the bases and of some of the salts derived therefrom were solved and are discussed. Cyclic voltammetry indicates that 1 to 4 are strong electron donors, reacting easily to radical cations or dications which with suitable acids have been obtained as salts.

The formation of tetrahydrofurans was achieved by an efficient and highly regioselective alkoxyl radical cyclization (5-exo-trig). The 2,3-anti substituted intermediates 9 and 12 cyclize stereoselectively whereas a 2,3-syn-configured O-radical affords both possible diastereomeric addition products in equal amounts. The cyclized tetrahydrofuryl methyl radicals were trapped with the bromine atom donor BrCCl3 to afford the bromomethyl-substituted cyclic ethers 10, 11, 18, and 19 in excellent yields.     Notes: ---Reactions of diaryl thioketones with diazomethane at room temperature afford 4,4,5,5-tetraaryl-1,3-dithiolanes; the scope of this surprising 2:1 interaction has been studied for decades (Schönberg Reaction). The clue to the mechanism was our observation that the stoichiometry is 1:1 at -78 °C, and 2,5-dihydro-2,2-diaryl-1,3,4-thiadiazoles are formed as primary [2+3] cycloadducts. They lose N2 at -45 °C in first-order reactions generating diaryl thioketone S-methylides which can be intercepted by thioketones (→1,3-dithiolanes), multiple CC bonds, or acids HX. In the absence of trapping reagents, the elusive intermediates either dimerize furnishing 2,2,3,3-tetraaryl-1,4-dithianes or give rise to 2,2-diarylthiiranes by electrocyclization. Beyond thiobenzophenone and diazomethane, our main model reaction, the studies involve fluorene-9-thione, 4,4-dimethoxy- and 4,4-dichlorothiobenzophenone. The ring of 2,5-dihydro-2,2-diphenyl-1,3,4-thiadiazole (8) is opened by LDA at -78 °C and derivatives of anion 12 are obtained. - In summa: The Schönberg reaction consists of two 1,3-dipolar cycloadditions, linked by a 1,3-dipolar cycloreversion.

    Notes: ---New electropolymerizable aromatic compounds (i.e. pyrrole, thiophene, aniline) bearing boronic acid and ester substituents have been synthesized and their electrochemical behavior has been investigated. Functionalized polythiophene and polypyrrole films could be anodically generated in acetonitrile, whereas the polyaniline derivative was electroformed in an acidic aqueous solution. The electrochemical responses of some of these materials were changed when fluoride ions were added to the electrolytic solutions. The strongest modifications, caused by binding of fluoride by the immobilized boron, were observed for the polypyrrole derivative in hydroorganic media.

    Notes: ---Organozinc halides derived from Grignard reagents behave differently in their reaction with ethyl (±±)-(2RS,3SR)-tetrahydro-4-methylene-2-phenyl-3-(phenylsulfonyl)furan-3-carboxylate (3) according to the hybridisation of the carbon ligand. During the development of short multi-component reactions for the synthesis of diverse functionalized ethyl 2,5-dihydrofuran-3-carboxylates it was discovered that aryl and vinyl zinc halides undergo clean reaction with 3 in the presence of Pd(PPh3)4. In contrast, when alkyl zinc halides are reacted with 3 in the presence of Pd(PPh3)4, reductive desulfonation of 3 is observed. Remarkably, in the absence of a transition metal catalyst, the allylic substitution of 3 with alkyl zinc halides proceeds cleanly and in moderate to good yield.

    Notes: ---Resorcin[4]arene tetracarboxylic acids 5,6 (A) and resorcin[4]arene tetrapyridines 2,3 (P) self-assemble in chloroform solution to form stable heterotopic AP dimers. Data from NMR titration and dilution experiments, as well as from vapor-pressure osmometry (VPO), indicate that the AP dimer is formed with an association constant greater than 107 M-1. Solid-solution extraction experiments are indicative of the formation of a 2:1 trimer (A2P), while self-associated homotopic species (A2 and A3) can be detected by NMR and VPO. Analysis of the heterotopic noncovalent assembly process over a range of compositions shows that these other species are much less stable than the AP heterodimer, which is the exclusive species at an A/P concentration ratio of 1:1 (〉 99.7% of the total at 10 mM).

    Notes: ---Quinic acid was transformed into phosphitamides 16, 25, and 36, which could be readily linked to 5′-O-unprotected cytidine derivative 17. Ensuing oxidation of the obtained phosphite triesters with tBuO2H and hydrogenolytic de-O-benzylation furnished the corresponding phosphate diesters 18, 26, and 38. Base catalyzed removal of acetyl protecting groups, and methyl ester hydrolysis furnished CMP-Neu5Ac analogues 1d, 1e, and 2. Quinic acid was also transformed into 1,2-unsaturated diallyl α-hydroxymethyl-phosphate derivatives (R)- and (S)-46, which on reaction with cytidine phosphitamide 47 afforded the phosphite triesters. Subsequent oxidation with tBuO2H and then treatment with NEt3 gave phosphate diester derivatives (R)- and (S)-48. Deallylation, acetyl group removal, and methyl ester hydrolysis furnished (R)- and (S)-3, respectively. Treatment of (R)- and (S)-48 with DBU as a base led to acetic acid elimination, thus yielding, after de-O-allylation, acetyl group cleavage, and ester hydrolysis, diene derivative (E)-4. Donor substrate analogues 1d and 1e exhibited good α(2-6)-sialyltransferase inhibition (Ki: 2.0·10-4 and 2.0·10-5 M). However, transition state analogues (R)-, and particularly (S)-3 showed excellent inhibition properties (Ki: 1.6·10-6 and 2.7·10-7 M).

    Notes: ---Three deoxy derivatives 2-4 of the α-mannosidase inhibitor mannostatin A (1) were synthesized, and their inhibition of Jack bean α-mannosidase was evaluated in order to elucidate the roles of each of the three hydroxyl groups of the inhibitor. The 1- and 2-deoxy derivatives 2 and 3 retained some inhibitory activity, although reduced by a factor of about 100 relative to the parent, whereas it was completely lost with the 3-deoxy derivative 4. Structure and activity relationships are discussed in the light of these findings.

Increased reactivities and enantioselectivities in diethylzinc additions to benzaldehyde are obtained from 3 (63% ee R) and 4 (93% ee S), relative to 1 (26% ee S) and 2 (73% ee S). X-ray crystal structures of 3 and of its methylzinc complex 3-Zn reveal out-of-plane bending of the methoxy groups as major geometrical consequences of the trimethylsilyl substitutions. Analyses of QM/MM ONIOM μ-O transition-structure models for 1, 2, 3, and 4 show that trimethylsilyl-induced distortions of methoxy and of dimethylaminomethyl groups explain the observed increased enantioselectivities.

    Notes: ---Operationally simple and environmentally benign procedures have been developed to selectively reduce different α,β-unsaturated ketones, 4,4-dimethylcyclohex-2-ene-1-one (1), isophorone (2), benzylideneacetone (3), chalcone (4) by NaBH4 or by the system NaBH4 + CoCl2. Alternative reaction media to the extensively used MeOH have been explored, and new procedures take advantage of the acceleration and chemoselectivity induced by water or by aqueous micellar solutions. It was possible to selectively and quantitatively afford pure products of 1,2 and of 1,4 reduction as well as the totally reduced compounds (yield and selectivity 〉 90%) by simple changes in the experimental conditions.

    Notes: ---The conformational behavior of the C-glycosyl analogue of sialyl-α-(2→3)-galactose, synthesized as a glycosidase inhibitor, has been studied using a combination of NMR spectroscopy (J and NOE data) and molecular dynamics calculations. The obtained results show that the population distribution of conformers with respect to the orientation about the pseudo-glycosidic linkages is mainly controlled by steric interactions. This is in contrast to findings made for O-glycosides. In these natural compounds, the conformational behavior about the glycosidic linkage Φ is mainly governed by the exo-anomeric effect.

N-Debenzylation led to the corresponding N-unsubstituted azetidines, which were then subjected to palladium-catalysed coupling reactions with aryl bromides to afford chiral N-arylazetidines. (R,R)-N-Benzyl-2,4-dimethylazetidine has been employed in the synthesis of a new cyclopalladated complex, which can be used, for instance, as a chiral recognition agent for phosphorus ligands.

    Notes: ---Syntheses of the spiro nucleosides 2′-deoxyhydantocidin 3a and its 1′-epimer 3b are described. The newly developed route involves a Horner-Wadsworth-Emmons condensation of the phosphonate 16 with the erythrose derivative 15 affording a mixture of six isomers which was fully assigned by NMR spectroscopy. The mixture was directly converted into the final compounds in an efficient base-catalyzed cyclization reaction. A base-catalyzed interconversion between the two isomers was observed.

The type of adduct formed is determined by the molar ratio of the reactants 4 and 5 and by the nature of the substituent in the para position of the propiophenone 5.

    Notes: ---Cyclotrimerisation of benzo-polycyclic bromostannylalkenes 8a-d with Cu(NO3)2·3H2O in THF affords benzotri(benzonorbornadienes) 3a-d as a mixture of the syn and anti isomers. The ratio of syn to anti is close to the 1:3 statistical value in most cases (i.e. in cyclotrimers 3a,b,d), but highly in favour of the anti isomer in 3c, where steric hindrance by the methoxy groups plays an important role in the stereochemistry of the cyclotrimerisation. The substrates for the cyclotrimerisation, i.e. the bromostannyl alkenes 8a-d, were prepared from bromoalkenes 7a-d by treatment with base (LDA) and quenching with trimethyltin chloride. In turn, bromoalkenes 7a-d were prepared from alkenes 5a-d by radical bromination-elimination. The reaction conditions used were designed to minimise Wagner-Meerwein rearrangements that would lead to unwanted bromo isomers. The cup-shaped syn cyclotrimers 3a-d exhibit high electron density within the cavity as determined by AM1 semiempirical calculations of their electrostatic potential surfaces and are valuable substrates for supramolecular chemistry. As an example, it is shown that fullerene C60 is drawn into solution in acetonitrile by complexation with both the syn and anti trimer of benzonorbornadiene 3a.

    Notes: ---This paper describes a liquid chromatographic method using a reversed phase capillary column coupled to an UV detector for the quantitation of thirteen pollutant phenols. Chromatographic separation was carried out with gradient elution at 25.0 ± 0.1°C. The two major anisocratic elution modes (gradient elution and temperature programming) were evaluated. The detection limit range was 10-81 pg (100 nL injected). The chromatographic method combined with liquid-liquid extraction was applied to analysis of these compounds in river water. Recoveries of 75-103% were achieved for most of them.

    Notes: ---The volatile constituents of lulo del Chocó (Solanum topiro) fruit pulp obtained by liquid-liquid extraction were analyzed by capillary GC and capillary GC-MS. In total, 30 components were identified with methyl salicylate, hexadecanoic acid, hexanal, guaiacol, ethyl butanoate, and ethyl acetate being the major components. Chirospecific MDGC analysis revealed the predominance of (R)-ethyl-3-hydroxybutanoate (ee 40%) and the presence of racemic mixtures both of δ-octalactone and of δ-decalactone. For γ-hexalactone, γ-octalactone, and γ-decalactone enantiomeric distributions of 22.4 : 77.6, 22.9 : 77.1, and 20.0 : 80.0, (R) : (S), respectively, were determined. Glycosidically bound aroma compounds were identified by capillary GC and capillary GC-MS after isolation of the glycosidic fraction obtained by Amberlite XAD-2 adsorption and methanol elution followed by hydrolysis with a commercial pectinase enzyme. In total 13 bound aroma compounds (aglycones) were identified. These aglycones mainly consisted of compounds exhibiting aromatic structures. Additionally, with the aid of capillary GC and capillary GC-MS (EI and NCI) of trifluoroacetylated derivatives we identified eight glucosides: the novel 3,6-epoxy-7-megastigmen-5,9-diol β-D-glucopyranoside and the hexyl, benzyl, linalyl oxide (furanic), 2-phenylethyl, vomifolyl (isomer 1), (6S,9R)-vomifolyl, and scopoletin β-D-glucopyranosides.

    Notes: Using a running buffer containing cyclodextrins (CDs) and 2-[N-cyclohexylamino]-ethanesulfonic acid (CHES), positive system peaks were observed in the analysis of a ganglioside mixture by CE-UV. These system peaks were related to CDs in the running buffer because these peaks were also detected when a plug of solution devoid of CDs but having the same CHES concentration and pH as the running buffer was injected. Neutral CDs were separated owing to the formation of inclusion complexes with the anionic CHES ion. One possible explanation for the positive system peaks is that the anionic CD-CHES inclusion complex is displaced by co-ions with higher UV absorptivity.
