    Notes: Abstract New experimental data obtained from constant stress rheometers are used to show that the yield stress concept is an idealization, and that, given accurate measurements, no yield stress exists.     Notes: Summary Based on Maron-Pierce's equation, an empirical equation was suggested, which relates the relative viscosity (η r ) of the polymer melt filled with various inorganic filler, such as glass fiber, carbon fiber, talc, precipitated- and natural-calcium carbonate powder, and glassy small sphere, to the volume fraction (φ) of the filler. The equation isη r = (1 −φ/A)−2, whereA is a parameter relating to the packing geometry of the filler, which is similar to the parameterφ 0 in Maron-Pierce's equation. In the equationη r is defined as the ratio of the viscosity of the filledsystem to that of the medium at the same shear stress not the shear rate. The applicability of the equation is above the shear stress about 104 dyne/cm2. The equation has a simple form and is considered to have a practical utility for filled-polymer melt systems.

    Notes: Abstract Thermoreversible gelation of the system 2-propanol/poly (n-butyl methacrylate) — as detected by D'SC or dielectric experiments — does not manifest itself in a straightforward manner in the dynamic-mechanical properties. Its occurrence can, however, be seen in many ways: i) For constant composition of the system and a reference temperature lower than T gel, the storage modulus G′ is larger than the loss modulus G″ in the glass transition zone of the master curve and both vary in an almost parallel manner with the angular frequency ω over almost two decades (whereas this feature is normally found for other gelling systems within the rubber plateau or the flow region). ii) The entanglement molecular weight obtained from G″max is markedly less max than the entanglement molecular weight in the melt divided by ϕ2, the volume fraction of the polymer. iii) The temperature influences change from WLF like to Arrhenius-like behavior as T is lowered in the case of highly concentrated polymer solutions; analogous considerations hold true as ϕ2 is increased at constant T. iv) For sufficiently low temperatures, the activation energy of flow exhibits a maximum in the concentration range where the gelation is — according to DSC experiments — most pronounced. Like with ordinary non-gelling systems it is possible to construct master curves. On the basis of Graessley's theory identical dependencies are obtained for the variation of the entanglement parts of the stationary viscosity with shear rate and for the dependence of the entanglement part of the complex viscosity on the frequency ω of oscillation. Zero shear viscosity and limiting value of the complex viscosity for vanishing ω as a function of ϕ2 match smoothly and exhibit two points of inflection.

    Notes: Abstract The flow behaviour of pure and commercial molten TNT when subjected to very high rates of shear in a rotational viscometer over the temperature range 82.0 to 95.4°C is presented. All data obtained suggest that the behaviour of molten TNT is Newtonian, contrary to the conclusions of some published work, and the temperature dependence of the viscosity of pure TNT is adequately described by an Arrhenius-type relationship of the formη = A e B/T whereA = 0.000 541,B = 3 570 andT is the Kelvin temperature. A similar relationship is obtained for commercial TNT. Under some instrumental conditions discontinuities are seen in the shear stress—shear rate flow curves. An analysis of these discontinuities shows them to be caused by the transition from laminar to turbulent flow. Possible implications for processibility and initiation of explosives are discussed.

    Notes: Abstract The focus of this paper is on the viscoelastic properties of concentrated polymer solutions and polymer melts. Dynamic mechanical measurements were performed on various polystyrene/ethylbenzene solutions with polymer concentrations ranging from 40% up to 100% and temperatures from Tg+30°C up to 70°C (230°C for polymer melts). The basis polymers are two commerical grade polystyrenes (BASF) with M W = 247 kg/mol and 374 kg/mol, respectively. To avoid solvent loss due to evaporating during the measurements, a special sealing technique was used. A phenomenological model which describes quantitatively the relaxation spectrum of concentrated polymer solutions from the flow regime up to the glass transition regime is developed. The relaxation data of the respective polymer melt and the glass transition temperature of the solution are the only input parameters needed. The temperature dependence is described by a universal, concentration invariant WLF-equation. The relaxation spectra are divided into two parts accounting for the entanglement and the segmental relaxation modes, respectively. The relaxation strength related to the flow and entanglement regime scale with c 2.3, whereas the segmental relaxation strength does not alter with concentration. All relaxation times change with concentration proportional to c 3.5. Flow curves can be calculated from these relaxation spectra and thus, our results are useful for engineering applications.

    Notes: Abstract Rheological measurements were used to characterize the behavior of lyocell solutions, i.e., cellulose dissolved in N-methymorpholine-N-oxide. Cellulose sources included dissolving pulp, kraft pulp, sugar cane fibers, and kenaf fibers. The dominance of viscous behavior, G′ values, over elastic behavior, G″ values, is affected by cellulose concentration and molecular weight. At lower concentrations and degrees of polymerization (DP), dissolving pulp solutions show viscous, inelastic behavior at low frequencies. At higher concentration and DP, dissolving pulp solutions are more elastic at higher frequencies. Solutions prepared with kenaf and sugar cane fibers show similar properties to those using pure dissolving pulp, and comparisons suggest the molecular weight and/or the presence of other substances such as lignin in the cellulose from these alternative sources affect the rheology.

    Notes: Abstract The effect of starch granule size on the viscosity of starch-filled poly(hydroxy ester ether) (PHEE) composites was characterized using size-fractionated potato starch, as well as unfractionated starches (rice, corn, wheat, and potato). Potato starch was separated using an air classifier into four particle size fractions: 〈18 μm, 18-24 μm, 24-30 μm, and 〉30 μm. The starch was dried to a moisture content of 0.5% to minimize moisture effects on composite rheology. PHEE and potato starch were extruded with starch volume fractions of 0.46 and 0.66. Stress relaxation, frequency and strain sweep, and temperature-dependence measurements were carried out. Although small variations in viscosity were seen with the different potato starch fractions, differences were not significant at a volume fraction of 0.46. Viscosity differences between the different particle size fractions were more pronounced at a volume fraction of 0.66. The temperature dependence could be described by an Arrhenius relation, with an apparent activation energy of 84 kJ/mole. At a volume fraction of 0.46, the starch/PHEE viscosities increased in the order potato starch 〈 wheat starch ≃ corn starch 〈 rice starch.

    Notes: Summary Enhancement of photovoltage has been found in bone and its two major constituents when placed in a magnetic field in standard Hall geometry. Infrared light has been used for carrier excitation. From data obtained in this manner, values for the light absorption constant, extinction constant, mass absorption constant, and efficiency have been estimated. All these parameters increase with the increase of magnetic field. A possible explanation for the observed phenomena is presented.

    Notes: Summary To test the importance of 24-hydroxylation of vitamin D3 on bone mineralization, rat pups born to vitamin D-deficient females were given either 25-hydroxyvitamin D3 or 24,24-difluoro-25-hydroxyvitamin D3 for 16 days beginning at the time of weaning. Following such treatment analysis of blood samples revealed no detectable 24R,25-(OH)2D3 and 1,25-(OH)2D3 in the rats given the difluoro compound while revealing the expected 24,24-difluoro-25-hydroxyvitamin D3 and 24,24-difluoro-1,25-dihydroxyvitamin D3. The rats given 25-hydroxyvitamin D3 had the expected levels of 25-hydroxyvitamin D3, 24,25-dihydroxyvitamin D3, and 1,25-dihydroxyvitamin D3. Following sacrifice at day 17, postweaning bone mineralization and modeling were studied in long bones using histological methods. Bones taken from vitamin D-deficient rats at the beginning and end of the experimental period had lesions typical of rickets. These included wide growth plates, excessive amounts of osteoid, and metaphyseal fibrosis. Following treatment with either 25-hydroxyvitamin D3 or 24,24-difluoro-25-hydroxyvitamin D3, bone mineralization returned to normal. Growth plate widths and the amount of osteoid on bone surfaces were both substantially reduced and to a similar degree in both treatment groups. Normal cartilage core formation and trabecularization of the metaphyseal primary spongiosa were also restored to a similar degree in both groups. In effect, no difference was observed in any bone parameter studied between the 25-hydroxyvitamin D3- and the 24,24-difluoro-25-hydroxyvitamin D3-treated animals. These results provide strong evidence that 24-hydroxylation of the vitamin D molecule plays little or no role in the modeling and mineralization of bone.

    Notes: Summary The hormonal metabolite of vitamin D3, 1,25-dihydroxyvitamin D3 [1,25(OH)2D3], exerts its biological effects by binding to a cytosolic receptor protein. Such a protein has been demonstrated in vitamin D3 target organs including fetal rat calvariae and more recently in rat osteogenic sarcoma cells. In this study we have compared the binding of 25-hydroxyvitamin D3 [25(OH)D3] and 24,25-dihydroxyvitamin D3 [24,25(OH)2D3] to that of 1,25-(OH)2D3 in fetal rat calvariae and osteogenic sarcoma (OS) cells. Sucrose density sedimentation, DNA-cellulose chromatography, and intracellular uptake studies have been employed to evaluate these interactions. In cytosol preparations from calvariae, [3H]-1,25(OH)2D3 bound to a 3.3S macromolecule and to a much greater extent to a 5.8S macromolecule while both [3H]25(OH)D3 and [3H]24,25(OH)2D3 bound to the 5.8S macromolecule. By incubating intact calvariae and OS cells with labeled metabolites and thus establishing binding intracellularly prior to cell disruption, we have found that the 3.3S protein which has high specificity for 1,25(OH)2D3 occurs inside the cells; the 5.8S protein, however, does not occur inside the cells but is generated after cell disruption. The [3H]-1,25(OH)2D3-receptor complex adsorbed to DNA-cellulose and was eluted from this affinity resin at 0.28M KCl. In contrast, [3H]25(OH)D3 and [3H]-24,25(OH)2D3 binding activity did not adsorb to DNA-cellulose. We conclude that, in contrast to the 3.3S protein, the 5.8S macromolecule does not fulfill receptor criteria but is rather generated by the experimental manipulation of the bone cells. Our data suggest that the vitamin D3 actions on bone are mediated only via the 3.3S receptor, and hence quantitative but not qualitative differences of the effects of the various metabolites are feasible.

    Description / Table of Contents: Résumé La préparation de coupes fines, plano-parallèles d'os non décalcifié, d'épaisseur connue, pour la réalisation de microradiographies et d'autoradiographies quantitatives est décrite. Pour éviter que des fractures de l'os se produisent, le fragment osseux fixé doit être inclus dans un matériel approprié qui ne doit pas se rétracter. Un minimum de force doit être appliqué à l'échantillon pendant la coupe pour éviter des vibrations. La méthode décrite, utilisant une scie circulaire tournant à faible vitesse, est adéquate. Le polissage final des surfaces du specimen est réalisé sur une machine à polir modifiée. On peut préparer un grand nombre de coupes en relativement peu de temps pour des études statistiques.
    Abstract: Zusammenfassung Die Herstellung von dünnen, parallelgestellten Schnitten von unentkalktem Knochen bekannter Dicke zur quantitativen Auto- und Mikroradiographie wird beschrieben. Der Knochen hat die Tendenz, unter Druck zu brechen. Um diese Schwierigkeit zu überwinden, ist es sehr wichtig, daß die fixierte Knochenprobe genügend vom Einbettungsmaterial durchdrungen wird und daß das Einschlußmittel sich vom Knochenrand weg kontrahiert. Beim Schneiden darf nur ein minimaler Druck auf die Probe ausgeübt werden, damit der Knochen nicht zertrümmert wird. Die beschriebene Methode, bei welcher eine spezielle Einrichtung zum Auftropfen des Schmiermittels den Gebrauch einer langsam rotierenden Kreissäge ermöglichte, erwies sich als günstig. Das schließliche Polieren der Oberflächen der Proben wurde auf einer modifizierten Schleifmaschine ausgeführt. Mit dieser Methode kann eine große Anzahl von Schnitten in relativ kurzer Zeit hergestellt werden; dies ist sehr wichtig, um eine angemessene Anzahl von Proben für statistische Analysen zu erhalten.
    Notes: Abstract The preparation of thin, parallel-sided sections of undecalcified bone of known thickness for quantitative auto- and micro-radiography is described. To overcome the tendency of the bone to fracture when stressed, it is essential that the fixed sample of bone should be adequately penetrated by the embedding material and that the mounting material should not contract away from the edge of the bone. Minimum stress must be applied to the sample when cutting or the bone may be shattered. The method described, using a gravity feed to a low speed circular saw, has been found to be suitable. Final polishing of the surfaces of the specimen was carried out on a modified lapping machine. Large numbers of sections can be prepared in a relatively short time by the method and this is essential to obtain an adequate number of samples for statistical analysis.

Cette vitesse traduit la fonction d'ostéoclastes isolés dans le cône coupant et est l'inverse de la vitesse d'apposition (l'épaisseur de l'os déposé par une couche unique d'ostéoblastes par unité de temps). Au niveau de cinquante trois zones de remaniements haversiens, dans plusieurs coupes de quatre côtes différentes de trois chiens adultes, la vitesse longitudinale d'érosion (dans la direction de l'axe longitudinal de l'ostéone) est d'environ 39±14 μ/jour, alors que la vitesse d'érosion radiaire, c'est-á-dire l'élargissement centrifuge du sommet du cône coupant vers son plus large diamètre, est d'environ 7.1±3μ/jour. Etant donné que la dynamique du remaniement osseux lamellaire de l'homme et du chien est très semblable, ces résultats peuvent aussi être appliqués á l'homme.
    Abstract: Zusammenfassung Es wurdenin vivo Tetracylin-Markierung und histologische Analyse von unentkalkten Corticalis-Längsschnitten verwendet, um die durchschnittliche Geschwindigkeit zu messen, mit der die Osteoclastenkegel (cutting cones) der sich entwickelten havers'schen Systeme im Knochen vorrücken. Diese Geschwindigkeit reflektiert die Resorptionstätigkeit eines einzelnen Osteoclasten, im Gegensatz zu Appositionsrate, welche die Intensität der Matrixproduktion durch die Osteoblasten in einer bestimmten Zeitspanne angibt. Bei 53 im Umbau begriffenen Havers'schen Systemen in mehreren Schnitten von vier verschiedenen Rippen dreier erwachsener Hunde war die durchschnittliche Geschwindigkeit des longitudinalen Vorrückens 39±14μ/Tag, während die durchschnittliche Erweiterung des Durchmessers an der Spitze des Resorptionskanales einen Betrat von 7.1±3 μ/Tag ausmachte. Da die Dynamik der Umbauvorgänge im kompakten Lamellenknochen bei Hund und Mensch vergleichbar ist, kann angenommen werden, daß die ermittelten Werte auch für den Menschen zutreffend sind.
    Notes: Abstract Using tetracycline labellingin vivo and histological analysis of undecalcified longitudinal sections of cortical bone, the average speed was measured with which osteoclastic fronts (cutting cones) of the evolving Haversian systems erode bone. This speed reflects the function of the individual osteoclasts within the cutting cone and is opposite to the appositional rate (the thickness of bone deposited by a monolayer of osteoblasts in unit time). In fifty-three Haversian remodeling sites in several sections from four different ribs of three adult dogs, the longitudinal erosion rate (in the direction of long axis of the osteon) averaged 39±14 μ/day, whereas the radial erosion rate, that is the centrifugal enlargement from the tip of the cutting cone to its largest diameter, averaged 7.1±3 μ/day. Since human and canine lamellar bone remodeling dynamics appear closely similar, the estimates in this study may also apply to man.

    Description / Table of Contents: Résumé De la collagénase osseuse de souris, une collagénase tissulaire spécifique, est isolée du milieu de culture tissulaire d'un tibia de souris de 5 jours. En combinant une précipitation de (NH4)2SO4, une filtration moléculaire sur tamis et la chromatographie par échangeurs d'ions, on obtient une fraction d'un rendement d'environ 11%, qui possède une activité enzymatique spécifique 120 fois plus élevée que l'extrait original total. Le poids moléculaire de la fraction, contenant l'activité enzymatique, est d'environ 41000, en se basant sur des études de filtration par tamis moléculaire calibré. Il existe au moins deux fractions principales distinctes et une fraction plus faible, ayant des activités en collagénase et des points isoélectriques distincts. Il n'est pas démontré que ces fractions constituent des isoenzymes de l'os ou de l'os et du cartilage, ou sont dérivés d'une seule enzyme, peu dégradée par l'extraction et la purification. L'activité de la collagénase est inhibée par l'EDTA, la cystéine, le sérum de cheval; mais elle résiste au fluorure de phénylméthyle-sulfonyle, à l'acide epsilon aminocaproique et à l'inhibiteur de trypsine de graine de soja.
    Abstract: Zusammenfassung Mäuseknochen-Kollagenase, eine spezifische Gewebe-Kollagenase, wurde aus Gewebekulturmedien von 5 Tage alten Mäusetibiae isoliert. Durch eine Kombination von (NH4)2SO4-Ausfällung, Gelfiltration und Ionenaustausch-Chromatographie wurde mit ungefähr 11% Ausbeute eine Fraktion erhalten, die eine spezifische Enzymaktivität besaß, welche 120mal größer war als diejenige des ursprünglichen unbehandelten Extraktes. Das Molekulargewicht der die Enzymaktivität enthaltenden Fraktion war ungefähr 41000, was durch kalibrierte Gelfiltrations-Untersuchungen bestimmt wurde. Mindestens zwei verschiedene Hauptfraktionen und eine kleinere Fraktion mit Kollagenaseaktivität wurden erhalten, welche verschiedene isoelektrische Punkte hatten. Es ist unklar, ob diese Fraktionen Isoenzyme aus Knochen oder aus Knochen und Knorpel darstellen, oder ob sie von einem einzelnen Enzym stammen, welches durch die verwendeten Extraktions- und Reinigungstechniken minimal degradiert wurde. Die Kollagenaseaktivität wurde durch EDTA, Cystein und Pferdeserum gehemmt, nicht aber durch Phenylmethylsulfonylfluorid, Epsilon-amino-capronsäure oder Soyabohnen-Trypsin-Hemmer.
    Notes: Abstract Mouse bone collagenase, a specific tissue collagenase, was isolated from the tissue culture media of 5 day old mouse tibiae. By a combination of (NH4)2SO4 precipitation, molecular sieve filtration and ion exchange chromatography a fraction was obtained with a yield of approximately 11% which had a specific enzyme activity 120-fold greater than the original crude extract. The molecular weight of the fraction containing the enzyme activity was approximately 41000 as determined by calibrated molecular sieve filtration studies. There were at least two distinct major fractions and one minor fraction possessing collagenase activity which had distinct isoelectric points. It is not clear whether these fractions represent isoenzymes from bone or from bone and cartilage, or are derived from a single enzyme which has been minimally degraded by the extraction and purification techniques used. Collagenase activity was inhibited by EDTA, cysteine and horse serum, but not by phenylmethylsulfonylfluoride, epsilon amino caproic acid or soybean trypsin inhibitor.

Dans les échantillons de cartilage, le contenu en PPi augmente progressivement dans l'ordre énoncé ci-dessus, de 8.59 μg P/g en poids sec de cartilage au repos à 236 μg P/g en poids sec dans le cartilage calcifié. Cependant, le rapport PPi sur orthophosphate suit une relation inverse: il est plus élevé dans la zone de repos et diminue considérablement lorsque le tissu se calcifie. Le rôle possible du PPi, en inhibant la précipitation de phosphate de calcium amorphe (ACP) et en ralentissant la transformation d'ACP en hydroxyleapatite (HA) dans les tissus calcifiés, est envisagé par rapport à d'autres facteurs, tels que le collagène, le magnésium, les phospholipides et les protéines-polysaccharides, qui peuvent aussi intervenirin vivo.     Abstract: Zusammenfassung Der Gehalt an anorganischem Pyrophosphat (PPi), Orthophosphat und Calcium wurde in ruhendem, proliferierendem, hypertrophischem und verkalktem Knorpel von foetalen Kalbsepiphysen und auch in spongiösem, periostalem und kompaktem Knochen gemessen. Bei den Knorpelproben nahm der Gehalt an PPi progressiv in der oben angegebenen Reihenfolge zu: von 8,59 μg P/g Trockengewicht in ruhendem Knorpel bis zu 236 μg P/g Trockengewicht in verkalktem Knorpel. Das PPi/Orthophosphat-Verhältnis hingegen verlief in umgekehrter Richtung; es war am höchsten in der ruhenden Zone und nahm dramatisch ab, wenn das Gewebe verkalkte. Die mögliche Rolle von PPi bei der Hemmung der Ausfällung von amorphem Calciumphosphat (ACP) und bei der Verlangsamung der Umwandlung von ACP in Hydroxyapatit (HA) in verkalkenden Geweben wird in bezug auf folgende anderen Faktoren, welche den Vorgang in vivo ebenfalls beieinflussen könnten, diskutiert: Collagen, Magnesium, Phospholipide und Proteinpolysaccharide. Bis jetzt war es nicht möglich, einen einzelnen Faktor als sicheren physiologischen Regulator zu identifizieren.
    Notes: Abstract The amounts of inorganic pyrophosphate (PPi), orthophosphate and calcium have been measured in resting, proliferating, hypertrophic and calcified cartilage from foetal calf epiphyses and also in cancellous, periosteal and compact bone. In the cartilage samples, the content of PPi increased progressively in the order named above, from values of 8.59 μg P/g dry weight in resting cartilage to 236 μg P/g dry weight in calcified cartilage. However, the ratio of PPi to orthophosphate followed the reverse relationship and was highest in the resting zone and fell dramatically as the tissue calcified. The possible role of PPi in inhibiting the precipitation of amorphous calcium phosphate (ACP) and in the slowing the transformation of ACP to hydroxyapatite (HA) in calcifying tissues is discussed in relation to other factors, such as collagen, magnesium, phospholipids and proteinpolysaccharides, which might also influence the processin vivo. At present, no single factor can be identified as a proven physiological regulator.

    Abstract: Zusammenfassung Die normale Längenwachstums-Geschwindigkeit der proximalen Wachstumsplatte der Tibia wurde bei Sprague-Dawley-Ratten in einem Alter zwischen 20 und 100 Tagen mittels der Tetracyclinmethode gemessen. Die Wachstumsgeschwindigkeit variierte nur wenig innerhalb der einzelnen Altersgruppen. Die Geschwindigkeit war bei jungen Tieren am höchsten und nahm mit zunehmendem Alter beträchtlich ab. Männliche Ratten wuchsen schneller als weibliche. Diese Arbeit dient als Grundlage, um die experimentelle Beeinflussung des Längenwachstums der Ratte abschätzen zu können.
    Notes: Abstract The rate of normal growth in length from the proximal growth plate of the tibia in the Sprague-Dawley rat was measured between 20 and 100 days of age using the tetracycline method. The growth rate varied only slightly within different age groups. The rate was highest in young animals and decreased considerably with increasing age. Male rats grew faster than female. This study is intended to provide a base for an evaluation of experimental influence on the growth in length of the rat.

Der Streubereich lag in den verschiedenen Gesichtsfeldern zwischen 136 und 177 μm2 per Osteoblast. 400 Zellen wurden einzeln gemessen. Bei diesen war der Durchschnittswert per Osteoblast 143 μm2, mit einer Standard-Abweichung von 33. Die tägliche Anlagerungsrate während einer Periode von 8 Tagen war 3,12 μm (Standard-Abweichung 0,22); sie wurde mittels Tetracyclinmarkierung der Mineralisierungsfront gemessen. The mean territory of 4620 cells in 19 fields was 154 μm2 per osteoblast. The range for the fields was 136 to 177 μm2 per osteoblast. Four hundred cells were measured individually—for these the mean value per osteoblast was 143 μm2 with a standard deviation of 33. The daily rate of apposition over an 8 day period was 3.12 μm (standard deviation 0.22) measured by tetracycline marking of the mineral front. This gave a daily matrix production rate of approximately 470 μm3 per osteoblast.

    Notes: Abstract The piezoelectric constant of mature and immature bone (defined herein) has been measured in an effort to determine whether it varies with age. It was found that the average value of the piezoelectric constantd 14 of femur from three week old calves was 58% of the value of femur from three year old bulls. The results were interpreted to indicate qualitative differences in the corresponding collagen matrices. Mature human tibia from males ranging in age from 21 to 53 years of age showed a small but significant increase ind 14 with age. Some data concerning diseased human bone, and well-preserved human bone excavated in Peru are also presented.

    Abstract: Zusammenfassung Poröse Metall- und Keramikstücke wurden in vivo in Knochen implantiert und nach verschiedenen Intervallen entnommen, um histologisch zu untersuchen, wie weit sie ins Gewebe eingewachsen waren. Die entnommenen Implantate mit dem sie umgebenden Gewebe wurden fixiert, dehydriert und in ein niedrigvisköses Epoxy-Harz eingebettet. Die Proben wurden mit einer Diamantsäge in dicke Schnitte zerteilt, auf Steinscheiben zementiert und von Hand auf etwa 75 μm geschliffen. Sie wurden mit Methylenblau und Alizarin-Rot S schwach gefärbt; dies ergibt einen großen Kontrast zwischen Knochen und Weichgewebe und zeigt die Bildung von verkalktem Knochen innerhalb poröser Metall- und Keramik-Implantate deutlich.
    Notes: Abstract Porous metals and ceramics were implanted in bonein vivo and retrieved at intervals for histological evaluation of tissue ingrowth. Recovered implants with surrounding tissues were fixed, dehydrated and embedded in a low-viscosity epoxy resin embedding medium. The embedded samples were cut into thick sections with a diamond saw, cemented to petrographic slides, and hand-ground to approximately 75 μm. They were lightly stained with methylene blue and Alizarin Red S, which provided great contrast between the bone and soft tissues, and clearly demonstrated the formation of calcified bone within porous metal and ceramic implants.

    Abstract: Zusammenfassung Länge, äußerer Umfang und Gewicht von 45 sauberen, trockenen zweiten Metacarpalknochen wurden bestimmt. Die totale Breite und die Markhöhlenbreite des mittleren Schaftes wurden mit posterior-anterioren Röntgenaufnahmen gemessen. Corticaler und äußerer Umfang und Aschgewicht eines 1 cm langen Stückes vom Schaft jedes Knochens wurden ebenfalls gemessen. Mittels morphometrischer Messungen wurden Schätzungen der geometrischen und densitometrischen Eigenschaften der Mittelschaftregion und des ganzen Knochens berechnet. Einige Schätzungen der geometrischen Eigenschaften wurden abgeleitet unter der Annahme, daß der metacarpale Schaft ein Zylinder ist. Die Beziehungen zwischen den Schätzungen und den geometrischen und densitometrischen Eigenschaften selbst wurden untersucht, und die Gültigkeit des Zylindermodelles wurde geprüft.
    Notes: Abstract The lengths, external volumes and weights of 45 clean, dry, second metacarpals were determined. The midshaft total and medullary widths were measured on postero-anterior radiographs. The cortical volume, external volume and ash weight of a 1 cm length of the shaft of each bone were also measured. From the morphometric measurements estimators of the geometric and densitometric properties of the midshaft section and of the whole bone were computed. Some estimators of the geometric properties were derived on the assumption that the metacarpal shaft is a cylinder. The relationships between the estimators and the geometric and densitometric properties themselves were examined and the validity of the cylinder model was explored.

Le calcium plasmatique s'élève de façon linéaire entre 31 et 250 m Unités d'insuline/100 g de poids corporel. La courbe obtenue n'est guère différente de celle obtenue par extrait parathyroidien. L'administration simultanée d'insuline et d'extrait parathyroidien à des rats thyro-parathyroidectomisés agit sur la calcémie et la phosphorémie par un effet additif. Lorsque des os frontaux d'embryons de poulet sont cultivésin vitro en présence d'insuline cristallisée, le taux de résorption augmente. L'insuline augmente le taux de consommation en glucose des explants et induit une accumulation de citrate dans le milieu de culture. L'insuline stimule donc la résorption osseuse.
    Abstract: Zusammenfassung Der Plasmagehalt an Calcium und Phosphor bei thyroparathyreoidektomierten Ratten wurde durch die Verabreichung von kristallinem Insulin erhöht. Das Plasmacalcium verhielt sich zwischen 31 und 250 m Einheiten Insulin pro 100 g Körpergewicht linear. Die Steigung der erhaltenen Kurve ist nicht signifikant verschieden von derjenigen, die nach Parathyreoidea-Extrakt erhalten wird. Die gleichzeitige Verabreichung von Insulin und Parathyreoidea-Extrakt von thyroparathyreoidektomierten Ratten hatte eine kumulative Wirkung auf den Gehalt von Calcium und Phosphor im Plasma. Wenn Stirnbeine von Kükenembryos mit kristallinem Insulinin vitro kultiviert wurden, erhöhte sich die Resorptionsrate. Das Insulin steigerte den Glucoseverbrauch der Explantate und verursachte eine Anreicherung von Citrat im Kulturmedium. Die Ergebnisse deuten auf eine Stimulation der Knochenresorption durch Insulin.
    Notes: Abstract the administration of crystalline insulin to thyroparathyroidectomized rats raised their calcium and phosphorus plasma levels. The plasma calcium gave a linear response between 31 and 250 mU of insulin/100 g body weight. The slope obtained is not significantly different from that obtained with parathyroid extract. The simultaneous administration of insulin and parathyroid extract to thyroparathyroidectomized rats affected the calcium and phosphorus plasma levels in an additive fashion. When chick embryo frontal bones were cultivatedin vitro with crystalline insulin the rate of resorption increased. Insulin increased the rate of glucose consumption by the explants and induced the accumulation of citrate in the culture medium. It is concluded that insulin stimulates bone resorption.

    Description / Table of Contents: Résumé L'effet de doses quotidiennes, administrées depuis la naissance, de deux types de diphosphonates, à savoir l'éthane-1-hydroxyle-1,1-diphosphonate (EHDP) et le dichlorométhylène diphosphonate (Cl2MDP), sur la croissance et le squelette de souris a été étudié. Les diphosphonates freinent la croissance: les incisives ne font pas leur éruption ou elle est plus tardive. La calcémie est normale. L'administration de Cl2MDP à une dose quotidienne de 10 mg P/kg/jour provoque des modifications squelettiques identiques à celles des souris grises létales atteintes d'ostéopétrose et les animaux meurent après quatre semaines de traitement. Par rapport aux témoins, les souris traitées présentent des os plus étroits, plus denses et plus déformés: les cavités médullaires sont comblées avec de l'os calcifié et du cartilage. La quantité totale de calcium d'un animal n'est pas augmentée par traitement au diphosphonate, par rapport à un témoin de même âge. Chez les souris grises létales et celles traitées aux diphosphonates, la plupart des anomalies est secondaire à une résorption osseuse diminuée. Ces résultats sont commentés en fonction de l'emploi des diphosphonates au cours de remaniements osseux pathologiques augmentés et en fonction du rôle de la résorption osseuse dans le maintien de la calcémie.
    Abstract: Zusammenfassung Mäuse erhielten von der Geburt an tägliche Dosen folgender zwei Diphosphonate: entweder Äthan-1-Hydroxy-1,1-Diphosphonat (EHDP) oder Dichloromethylen-Diphosphonat (Cl2MDP). Es wurde deren Wirkung auf das Wachstum und das Skelet untersucht. Die Diphosphonate verlangsamten das Wachstum, die Schneidezähne brachen nicht oder erst später durch, aber die Höhe des Plasmacalciums blieb normal. Die Verabreichung von Cl2MDP in Dosen von 10 mg P/kg/Tag führt zu Skeletveränderungen, welche denjenigen der „grey-lethal” osteopetrotischen Mäuse gleichen. Die Tiere sterben nach einer Behandlungsdauer von etwa 4 Wochen. Verglichen mit normalen Mäusen von ungefähr gleichem Alter hatten die behandelten Mäuse kleinere, dichtere und mehr keulenförmige Knochen, und die Markhöhlen waren gefüllt mit verkalktem Knochen oder Knorpel. Die Gesamtcalciummenge im Skelet wurde durch die Diphosphonatbehandlung nicht erhöht; dies ergab sich aus einem Vergleich mit der bei normalen Mäusen desselben Alters gefundenen Menge. Es wird vorgeschlagen, daß bei den „grey-lethal” und bei den Diphosphonat-behandelten Mäusen viele der Abnormalitäten als Folge der herabgesetzten Knochenresorption angesehen werden müssen. Die Ergebnisse werden einerseits im Hinblick auf den Gebrauch der Diphosphonate bei pathologischen Bedingungen eines erhöhten Knochenumbaus diskutiert; andererseits werden sie im Zusammenhang mit der Rolle der Knochenresorption bei der Erhaltung des Plasmacalcium-Spiegels besprochen.
    Notes: Abstract The effect of daily doses from birth of two diphosphonates, namely either ethane-1-hydroxy-1,1-diphosphonate (EHDP) or dichloromethylene diphosphonate (Cl2MDP), on the growth and the skeleton of mice has been studied. Diphosphonates slowed growth, the incisors did not erupt or erupted later, but the level of plasma calcium remained normal. The administration of Cl2MDP at a dose rate of 10 mg P/kg/day leads to skeletal changes that are similar to those observed in grey-lethal osteopetrotic mice, and the animals die after about four weeks of treatment. As compared with normal mice of similar age, treated mice had bones that were smaller, denser and more clubshaped, and the marrow cavities were filled with calcified bone or cartilage. The total amount of calcium in the carcass was not increased by diphosphonate treatment, as compared with the amount in normal mice of the same age. It is suggested that both in the grey-lethal and diphosphonate-treated mice many of the abnormalities are secondary to decreased bone resorption. The results are discussed with respect to the use of diphosphonates in pathological conditions of increased bone turnover and with respect to the role of bone resorption in the maintenance of plasma calcium levels.

La masse minérale osseuse ne montre pas de modification significative jusqu'à 60 ans chez l'homme et 50 ans chez la femme, puis on observe une chute. L'épaisseur musculaire décroit, chez l'homme, à partir de 30 ans, alors que chez la femme on n'observe aucune modification significative avant 60 ans. Les corrélations entre ces variables diffèrent selon le sexe. Alors que la masse minérale et l'épaisseur musculaire varient positivement chez l'homme en fonction de l'âge, il n'y a pas de rapport significatif chez la femme, après 60 ans. Cependant, chez ces dernières, la masse osseuse et le tissu graisseux sous-cutané sont en corrélation nette après 60 ans, bien qu'aucun rapport significatif ne soit noté chez l'homme quel que soit le groupe d'âge. Chez des sujets ostéoporotiques, la masse minérale osseuse et l'épaisseur musculaire sont plus faibles chez les hommes par rapport à des sujets normaux du même âge. Chez des femmes ostéoporotiques, cependant, bien que la masse minérale osseuse et le tissu graisseux sous-cutané soient inférieures par rapport à des sujets normaux, l'épaisseur du muscle n'est pas modifiée.
    Abstract: Zusammenfassung Es wurden bei gesunden erwachsenen Männern und Frauen radiologische Messungen der Knochenmineralmasse im proximalen Radius, der Dicke des Muskels und des subkutanen Fettes des Vorderarmes durch die ganze Altersspanne durchgeführt. Die Knochenmineralmasse zeigte keine signifikante Veränderung bis zum 60. Altersjahr bei den Männern und bis zum 50. Altersjahr bei den Frauen; dann jedoch sank sie ab. Die Muskeldicke nahm bei den Männern über 30 Jahren ab, bei den Frauen konnte jedoch eine signifikante Abnahme erst ab 60 Jahren festgestellt werden. Die Korrelation zwischen diesen beiden Werten war bei Männern und Frauen ebenfalls verschieden. Während bei den Männern Knochenmineralmasse und Muskeldicke in jedem Alter meistens eine positive Korrelation zeigte, konnte bei den Frauen nach dem 60. Altersjahr keine signifikante Korrelation gefunden werden. Bei den Frauen zeigten hingegen Knochenmasse und subkutanes Fett eine deutliche Korrelation in der Gruppe nach dem 60. Altersjahr, während bei den Männern in keiner Altersgruppe eine signifikante Korrelation gefunden werden konnte. In einer osteoporotischen Gruppe waren Knochenmineralmasse und Muskeldicke niedriger bei Männern, verglichen mit Kontrollen bei Gesunden desselben Alters. Bei weiblichen osteoporotischen Patienten hingegen zeigte die Muskeldicke keinen Unterschied, während Knochenmineralmasse und subkutanes Fett niedriger waren als bei Gesunden.
    Notes: Abstract Radiologic measurements of bone mineral mass in the proximal radius, muscle width and thickness of the subcutaneous fat of the forearm were studied in normal men and women throughout the adult age range. Bone mineral mass showed no significant change to age 60 in men and age 50 in women, but fell thereafter. Muscle width declined from age 30 in the male population, though no significant reduction was found in women before age 60. The correlations between these variables also differed between males and females. While bone mineral mass and muscle width tended, in males, to be positively correlated at all ages, in females no significant correlation was found after age 60. In females, however, bone mass and subcutaneous fat were distinctly correlated in the over 60 age group, though no significant correlation was found in males in any age group. In an osteoporotic group, bone mineral mass and muscle width were lower in male patients than in normals of similar age. In female osteoporotic patients, however, while bone mineral mass and subcutaneous fat were less than in normals, muscle width showed no difference.

Les résultatsin vitro peuvent être représentés, d'un autre côté, par une série de trois compartiments: celiu du milieu est identique avec le compartiment superficiel de la situationin vivo. Le temps de séjour du calcium dans ce compartiment peut être mis en rapport avec l'énergie de liaison de l'ion clacium, au niveau du côté superficiel d'un cristal d'apatite. Il semble que l'environnement des cristaux d'apatite, 1–4 μ sous la surface osseuse, soit identique à celui d'une solution aqueuse, les rendant susceptibles d'un échange rapide de calcium, mais cette possibilité est tellement réduite dans le volume osseux que le modèle de percolation proposé par Arnold et coll, parait indéfendable.
    Abstract: Zusammenfassung Eine mathematische Analyse von autoradiographischen Daten betreffend Aufnahme und Abgabe von45Ca an Knochenoberflächen beim Kaninchen, beim Hund und beim Menschen zeigte, daß ein Diffusionsmodell die Knochenoberfläche nicht überzeugend beschreibt. Hingegen werdenin vivo-Daten durch ein einziges Oberflächenkompartiment gut erfaßt. Diein vitro. Daten können andererseits durch eine Serie von drei Kompartimenten dargestellt werden; das mittlere dieser Kompartimente ist identisch mit dem Oberflächenkompartiment der Versuchein vivo. Die Verweildauer des Calciums in diesem Kompartiment kann zur Bindungsenergie eines Calciumions an der Oberfläche eines Apatitkristalls in Beziehung gebracht werden. Es wird daraus abgeleitet, daß die Umgebung der Apatitkristalle, die 1–4 μ unter der Knochenoberfläche liegen, derjenigen in wässeriger Lösung gleicht und sie dadurch für einen raschen Calciumaustausch zugänglich macht, da\ jedoch diese Zugänglichkeit innerhalb des Knochenvolumens derart drastisch reduziert wird, daß das Perkolationsmodell von Arnoldet al. nicht brauchbar ist.
    Notes: Abstract A mathematical analysis of autoradiographic data for the uptake and release of45Ca at bone surfaces in rabbit, dog, and man showed that a diffusion model of bone surface is not tenable. Datain vivo are, instead, well represented by a single surface compartment. Thein vitro data, on the other hand, can be represented by a series of three compartments; the middle one of these compartment is identical with the surface compartment of thein vivo situation. The residence time of calcium in this compartment can be related to the binding energy of a calcium ion at a surface site on an apatite crystal. It is concluded that the environment of the apatite crystals 1–4 μ below bone surfaces is similar to that in aqueous solution, making them available for rapid exchange of calcium but that this availability is so drastically reduced within the volume of bone that the percolation model of Arnoldet al. is untenable.

    Description / Table of Contents: Résumé Les effets de l'hydrocortisone, administrée sous deux formes physiques différentes (solubles et cristallines), à une dose identique de 5 mg/kg/jour ont été étudiés sur l'os en croissance chez 52 lapins pendant 6 semaines. Les animaux sont divisés en 1) un groupe témoin, 2) un groupe traité à l'hydrocortisone soluble et 3) un groupe traité à l'hydrocortisone cristalline. Les lapins traités à l'hydrocortisone soluble présentent quelques différences avec ceux traités à l'hydrocortisone cristalline. Par rapport aux témoins, le groupe traité à l'hydrocortisone soluble a augmenté plus faiblement de poids, présente moins de croissance osseuse longitudinale, montre un arrêt de l'ostéogenèse des surfaces et présente une augmentation de la résorption osseuse au niveau des surfaces corticales et de l'endoste. Cependant, le groupe traité à l'hydrocortisone cristalline a perdu de poids, ne présente pas de croissance osseuse longitudinale, ni d'apposition osseuse au niveau de toutes les surfaces, avec cependant une résorption osseuse marquée au niveau des surfaces corticales et de l'endoste, provoquant un amincissement net des corticales. Il semble que le facteur déterminant de cette action différente est l'élévation prolongée du cortisol plasmatique après injection d'hydrocortisone crystalline. Alors que d'autres aufeurs ont montré un rapport étroit médicament-dose, cette étude indique que la durée de l'action du médicament ou la fréquence de son administration par jour peut également être importante.
    Abstract: Zusammenfassung Die Wirkung von Hydrocortison auf den wachsenden Knochen wurde bei 52 Kaninchen untersucht. Das Hydrocortison wurde in zwei Formen (löslich und kristallin), bei gleicher Dosierung von 5 mg/kg/Tag, während 6 Wochen verabreicht. Die Tiere wurden in folgende Gruppen unterteilt: 1. Kontrollen; 2. mit löslichem Hydrocortison behandelte Tiere; 3. mit kristallinem Hydrocortison behandelte Tiere. Die mit löslichem Hydrocortison behandelten Kaninchen wiesen nicht dieselben Veränderungen auf wie die mit kristallinem Hydrocortison behandelten. Im Vergleich mit der Kontrollgruppe zeigte die mit löslichem Hydrocortison behandelte Gruppe folgende Veränderungen: weniger Gewichtszunahme, weniger Wachstum der longitudinalen Knochen, leichte Unterdrücklung der Knochenbildung auf allen Oberflächen und leicht erhöhte Knochenresorption an der corticalen Endostoberfläche. Die mit kristallinem Hydrocortison behandelten Tiere jedoch zeigten: Gewichtsverlust, Stillstand des longitudinalen Knochenwachstums, Stillstand der Knochenbildung auf allen Oberflächen und deutliche Knochenresorption an der corticalen Endostoberfläche, welche zu dramatischer corticaler Verdünnung führte. Es wird vermutet, daß der ausschlaggebende Faktor in dieser unterschiedlichen Wirkung die anhaltende Erhöhung der Cortisolkonzentration im Plasma ist, welche auf die Injektion von kristallinem Hydrocortisol folgt. Während andere Forscher eine starke Dosisabhängigkeit gezeigt haben, deutet diese Untersuchung an, daß die Wirkungsdauer der Substanz oder die Häufigkeit ihrer Verabreichung pro Tag ebenso wichtig sein können.
    Notes: Abstract The effects of hydrocortisone on growing bone administered in two different physical forms (soluble and crystalline) in an identical dosage of 5 mg/kg/day for six weeks were studied in 52 growing rabbits. The animals were divided into (1) controls, (2) soluble hydrocortisone treated and (3) crystalline hydrocortisone treated. The rabbits treated with soluble hydrocortisone showed changes different from those given crystalline hydrocortisone. When compared with the control group, the soluble hydrocortisone treated group gained less weight, had less longitudinal bone growth, had some suppression of bone formation at all surfaces and had some increased bone resorption at the cortical-endosteal surface. However, the crystalline hydrocortisone treated group lost weight, ceased longitudinal bone growth, ceased bone formation at all surfaces and had marked bone resorption at the cortical-endosteal surface resulting in dramatic cortical thinning. It is believed that the determining factor in this differential effect is the prolonged elevation of plasma cortisol folloing injection of the crystalline hydrocortisone. While other investigators have shown a strong drug-dose relationship, this study indicates that the duration of action of the drug or the frequency of its administration per day may be equally important.

    Description / Table of Contents: Résumé Une étude microscopique de la composition du tissu osseux, réalisée initialement au niveau des ostéones, a été étendue à l'os lamellaire interstitiel périosté et aux ≪Mittellinien≫. Le phosphore, calcium, l'azote total, les mucopolysaccharides (sous forme d'hexosamines) et le collagène (sous forme d'hydroxyle-proline) ont été déterminés dans ce tissu au point de vue pondéral et volumétrique. Le calcium a également été déterminé dans les ostéones aux stades initial et final de la calcification. Malgré des rapports Ca/P sensiblement identiques, l'os lamellaire interstitiel périosté et les Mittellinien diffèrent en composition organique. L'os interstitiel a un contenu plus élevé en azote (absolu et par rapport au phosphore) et en hydroxyle-proline et azote non-collagénique que les Mittellinien. En étudiant les résultats sur la composition du tissu ostéoide, les ostéones à divers degrés de calcification et les structures décrites ci-dessus, on se rend compte de la composition variable du tissu osseux d'une plage à l'autre. D'après les résultats analytiques, il semble que dans tout processus de calcification rapide, des protéines non-collagéniques (probablement des protéoglycanes) sont présentes.
    Abstract: Zusammenfassung Eine Untersuchung der Zusammensetzung des Knochengewebes auf der Stufe mikroskopischer Strukturen, welche früher auf die Haversianischen Systeme begrenzt war, ist auf den interstitiellen Lamellenknochen des Periosts und auf die Mittellinien ausgedehnt worden. Phosphor, Calcium, Gesamtstickstoff, Mucopolysaccharide (als Hexosamine) und Kollagen (als Hydroxyprolin) wurden in diesen Materialien in bezug auf Gewicht und Volumen bestimmt. Calcium wurde auch in den Osteonen in der Anfangs- und Endphase der Verkalkung bestimmt. Trotz praktisch identischem Ca/P-Verhältnis unterscheiden sich interstitieller Lamellenknochen des Periosts und Mittellinien in ihrer organischen Zusammensetzung. Interstitieller Knochen hat einen höheren Gesamtstickstoffgehalt (absolut sowie auf Phosphorgehalt bezogen), ebenso mehr Hydroxyprolin und nicht-kollagenen Stockstoff als die Mittellinien. Eine Zusammenstellung von Daten über die Zusammensetzung des Osteoid-Gewebes, der Osteonen in verschiedenen Stadien der Verkalkung und der oben aufgeführten Strukturen zeigt, wie unterschiedlich die Zusammensetzung des Knochengewebes sogar von einer mikroskopischen Struktur zur angrenzenden sein kann. Es wurde versucht, aus den analytischen Daten eine allgemeine Regel betreffend den verkalkungsprozess zu erhalten, nämlich: Wo immer eine Verkalkung rasch stattfindet, sind nicht-kollagene Proteine (möglicherweise Proteoglycane) anwesend.
    Notes: Abstract A study of the composition of bone tissue at the level of microscopic structures, previously limited to the Haversian systems, has been extended to interstitial periosteal lamellar bone and to the “Mittellinien”. Phosphorus, calcium, total nitrogen, mucopolysaccharides (as hexosamines) and collagen (as hydroxyproline) were determined in these materials on both weight and volume basis. Calcium was also been determined in osteones at the initial and final stage of calcification. In spite of virtually identical Ca/P ratios, interstitial periosteal lamellar bone and Mittellinien differ in their organic composition. Interstitial bone has a higher total nitrogen content (both absolute and relative to phosphorus) as well as higher hydroxyproline and non-collagenous nitrogen than the Mittellinien. A compilation of data on the composition of osteoid tissue, osteones at different degrees of calcification and the above structures, shows how variable is the composition of bone tissue even from one microscopic structure to the one adjacent. Tentative indications of a general rule governing the process of calcification were obtained from the analytical data; namely that wherever calcification is taking place rapidly, non-collagenous proteins (possibly proteoglycans) are present.

    Description / Table of Contents: Résumé De la poudre osseuse est décalcifiée et extraite à l'aide de l'E.D.T.A.0.5 M, puis d'un tampon de McIlvaine à pH 7.4. Trois fractions contenant de l'hydroxyproline ont été obtenues à partir de l'extrait de l'E.D.T.A., purifié par filtration sur gel. Ces 3 fractions sont considérées comme des produits de dégradation du collagène, étant donné leur analogie en composition en acides aminés. Le tampon de McIlvaine est fractionné par le sulfate d'ammonium. A partir de la fraction de saturation 0.3 M, après reconstitution avec l'ATP, un fragment de 700 Å de long est observé au microscope électronique. Une comparaison avec du collagène à segment espacé long semble indiquer que ce fragment est un produit de dégradation partielle de collagène et comprend environ un quart de la molécule collagénique de l'extrémité aminée terminale.
    Abstract: Zusammenfassung Knochenpulver wurde mit 0.5 M EDTA entkalkt und anschließen mit McIlvaine-Puffer bei pH 7,4 extrahiert. Aus dem durch Gel-Filtrations-Chromatographie gereinigten EDTA-Extrakt konnten 3 Hydroxyprolin enthaltende Fraktionen gewonnen werden. Diese wurden wegen ihrer kollagenähnlichen Aminosäurenstruktur für Abbauprodukte des Kollagens gehalten. Der McIlvaine-Puffer-Extrakt wurde mittels Ammoniumsulfat fraktioniert. Nach Rekonstitution mit ATP wurde aus der zu 0.3 gesättigten Fraktion ein Fragment in der Länge von 700 Å entnommen und unter dem Elektronenmikroskop geprüft. Vergleiche mit „segment-longspacing collagen” ließen vermuten, daß es sich bei diesem Fragment um teilweise abgebautes Kollagen handelte, das aus ungefähr einem Viertel des Kollagenmoleküls mit der endständigen Aminogruppe besteht.
    Notes: Abstract Bone powder was decalcified and extracted with 0.5 M EDTA and then with McIlvaine buffer at pH 7.4. From the EDTA extract, purified by gel filtration chromatography, three hydroxyproline-containing fractions were obtained which were considered to be degradation products of collagen because of their collagen-like amino-acid composition. The McIlvaine buffer extract was fractionated by ammonium sulfate. From the 0.3 saturation fraction, after reconstitution with ATP, a fragment 700 Å in length was observed with the electron microscope. Comparison with segment-long-spacing collagen suggested that this fragment was a partially-degraded product of collagen and consisted of approximately one quarter of the collagen molecule from the terminal-amino end.

    Description / Table of Contents: Résumé Des biopsies de côte, métacarpe et tibia sont prélevées chez des daims, cinq fois, pendant le cycle de croissance annuelle des andouillers. Les tissus sont étudiés par des méthodes histométriques ou densitométriques, suivies par l'analyse minérale à l'aide de la spectrophotométrie d'absorption atomique. Peu de remaniement s'observe dans le tissu osseux pendant la période de repos, mais pendant la période de croissance maximale des andouillers en juin, le remaniement osseux interne atteint son maximum. La résorption la plus élevée de 22,99% s'observe dans les côtes: elles est de 13,10% dans le métacarpe et 10,37% dans le métatarse. A l'automne, à la fin de la croissance des andouillers, on note une diminution du remaniement qui atteint des valeurs voisines de celle de la période de repos. Au niveau des côtes, les valeurs les plus faibles de poids de cendre, par unité de volume, s'observent au milieu de la période de croissance des andouillers, mais aucune variation cyclique de ces valeurs n'est trouvée dans le métacarpe, le métatarse et le tibia. Le pourcentage de calcium, magnésium et phosphore dans la cendre d'os ne varie pas pendant le cycle de croissance de l'andouiller. Il ne semble donc pas y avoir de changement de composition osseuse pendant les periodes de remaniement élevé.
    Abstract: Zusammenfassung Es wurden beim Wild in 5 Intervallen während des jährlichen Geweihwachstums-Cyclus folgende Biopsie-Proben entnommen: Rippe, Metacarpus, Metatarsus und Tibia. Die Gewebe wurden entweder mit histometrischen Methoden oder mit densitometrischen Messungen untersucht, welche durch eine Mineralanalyse mittels Atom-Absorptions-Spektrophotometrie ergänzt wurden. Während der Ruheperiode des Geweihwachstums erfolgte wenig Knochenneubildung; während der Hauptgeweihwachstumszeit im Juni hingegen erreichte die Knochenneubildung einen Höhepunkt. Die höchste Resorption von 22,99% erfolgte in den Rippen, beim Metacarpus waren es 13,10% und beim Metatarsus 10,37%. Beim Stillstand des Geweihwachstums im Herbst sank auch die Knochenneubildung wieder auf das niedere Niveau ab, das vor dem Geweihwachstum festgestellt wurde. Bei den Rippen zeigten sich die niedrigsten Werte des Aschgewichts per Volumeneinheit in der Mitte des Geweihwachstums; aber bei Metacarpus, Metatarsus und Tibia wurden für diese Werte keine cyclischen Veränderungen festgestellt. Der Prozentsatz von Calcium, Magnesium und Phosphor in der Knochenasche veränderte sich während des Geweihwaschstums-Cyclus nicht, was darauf hindeutet, daß die Zusammensetzung des Knochens während der Perioden erhöhter Neubildung gleich blieb.
    Notes: Abstract Biopsy samples of rib, metacarpus, metatarsus and tibia were obtained from deer at five intervals during the annual antler growth cycle. Tissues were examined by either histometric methods or by densitometric measurements followed by mineral analysis using atomic absorption spectrophotometry. Little remodelling occurred in bones during the period of antler quiescence, but during the peak of antler growth in June, internal bone remodelling reached a peak. The highest resorption of 22.99% occurred in ribs with lower levels of 13.10% for metacarpus and 10.37% for metatarsus. At the cessation of antler growth in the fall, remodelling again decreased to the low level found preceeding antler growth. In ribs lowest values of ash weight per unit volume occurred during the middle of antler growth but no cyclic variations were found for these values in metacarpus, metatarsus and tibia. Percentage of calcium, magnesium and phosphorus in bone ash did not vary during the antler growth cycle indicating no change in bone composition during periods of high remodelling.

    Description / Table of Contents: Résumé Des glycosaminoglycanes de zones au repos, sériées, hypertrophiques et calcifiées de la métaphyse épiphysaire et du cartilage du septum nasal ont été extraits à l'aide d'un solvant, 3 M GuCl, selon Sajdera et Hascall (1969), pour séparer les glycosaminoglycanes en une partie que l'on peut extraire et une partie qui reste liée au tissu. Le cartilage épiphysaire nécessite des temps d'extraction plus prolongés que le cartilage du septum nasal pour extraire des quantités similaires de glycosaminoglycanes acides suggérant une liaison plus étroite de ces derniers dans le tissu. Vers le front de calcification, les glycosaminoglycanes sont extraits plus facilement alors que dans la zone calcifiée pas plus de 30% ne peut être extrait. Des résultats obtenus par le procédé de microfractionnement CPC d'Antonopoulos et coll. (1964) indiquent une distribution similaire selon le poids moléculaire et/ou la densité de charge pour le chondroitine sulfate qui se laisse extraire ou non du cartilage du septum nasal et le chondroitine sulfate que l'on ne peut extraire du cartilage du septum nasal et des zones au repos et sériées de la métaphyse. Les glycosaminoglycanes des zones hypertrophiques et calcifiées que l'on ne peut extraire sont surtout de faible poids moléculaire et/ou leur densité de charge se compare au pool que l'on peut extraire. L'acide hyaluronique ne peut être extrait du septum nasal et des zones au repos, sériées et hypertrophiques avec des concentrations croissantes vers le front de calcification. Dans la zone calcifiée, un changement avec augmentation de l'acide hyaluronique que l'on peut extraire est noté. Ces résultats sont discutés.
    Abstract: Zusammenfassung Die sauren Glycosaminoglycane von ruhenden, säulenförmigen, hypertrophischen und verkalkten Zonen der Epiphysen-Wachstumsplatte und des Nasenseptum-Knorpels vom Kalb wurden mittels eines dissoziierenden Lösungsmittels, 3 M Guanidinchlorid, nach Sajdera und Hascall (1969) extrahiert, um die Glycosaminoglycane in einen extrahierbaren und einen unextrahierbaren Anteil — dieser bleibt ans Gewebe gebunden — zu trennen. Der Epiphysenknorpel beanspruchte längere Extraktionszeiten als der Nasenseptumknorpel, um vergleichbare Mengen saurer Glycosaminoglycane abzugeben. Dies deutet auf eine stärkere Bindung der Proteoglycane im Gewebe. Die Glycosaminoglycane ließen sich leichter aus dem Bereich der Verkalkungsfront extrahieren, während aus der verkalkten Zone nicht mehr als 30% extrahiert werden konnten. Die Resultate, die mit dem Cetylpyridinchlorid-Mikrofraktionierungsverfahren von Antonopouloset al. (1964) erhalten wurden, deuteten auf eine ähnliche Verteilung entsprechend dem Molekulargewicht und/oder der Ladungsdichte für extrahierbares und unextrahierbares Chondroitinsulfat im Nasenseptumknorpel und in ruhenden und säulenförmigen Zonen der Epiphysen-Wachstumsplatte. Die unextrahierbaren Glycosaminoglycane in hypertrophischen und verkalkten Zonen zeigten vorwiegend niederes Molekulargewicht und/oder niedere Ladungsdichte im Vergleich zum extrahierbaren Anteil. Die Hyaluronsäure ließ sich aus Nasenseptum und ruhenden, säulenförmigen und hypertrophischen Zonen nicht extrahieren und zeigte höhere Konzentrationen als in der Verkalkungsfront. In der verkalkten Zone erfolgte eine Verschiebung zu meist extrahierbarer Hyaluronsäure. Die Bedeutung dieser Befunde wird diskutiert.
    Notes: Abstract The acid glycosaminoglycans of resting, columnar, hypertrophic and calcified zones of calf epiphyseal growth plate and of nasal septum cartilage were extracted with a dissociative solvent, 3M GuCl, according to Sajdera and Hascall (1969), to separate the glycosaminoglycans into an extractable pool and an unextractable pool which remains bound within the tissue. Epiphyseal cartilage required longer extraction times than did nasal septum cartilage to extract comparable amounts of acid glycosaminoglycans suggesting a stronger binding of proteoglycans within the tissue. Towards the calcification front the glycosaminoglycans were extracted more easily while in calcified zone not more than 30% could be extracted. Data obtained by the CPC microfractionation procedure of Antonopouloset al. (1964) indicated similar distribution according to molecular weight and/or charge density for extractable and unextractable chondroitin sulphate in nasal septum cartilage and in resting and columnar zones of epiphyseal growth plate. Unextractable glycosaminoglycans in hypertrophic and calcified zones were of predominantly low molecular weight and/or charge density compared to the extractable pool. Hyaluronic acid was unextractable in nasal septum and in resting, columnar and hypertrophic zones with increasing concentrations towards the calcification front. In calcified zone a shift to mainly extractable hyaluronic acid occurred. The significance of these findings is discussed.

Der rachitische Knochen, der einen erhöhten Wassergehalt aufweist, hat einen ähnlichen Natriumaustausch wie der Knochen gleichaltriger Kontrolltiere. Die Behandlung von rachitischen Küken mit Vitamin D3 bewirkte eine Abnahme des Wassergehaltes im Knochen und eine Zunahme des Natriumaustausches.
    Notes: Abstract Sodium exchange in chick bone has been studied on control, rachitic and rachitic treated with vitamin D3 animals. The rachitic bone, which shows increased water content, has similar bone exchange than that of their age control group. Vitamin D3 treatment of rachitic chicks was followed by a decrease in bone water content and increase in bone sodium exchange.

L'augmentation de la force d'animaux, après délivrance, est en rapport avec le poids plus élevé de ces animaux par rapport à des animaux vierges: la surface transversale des os d'animaux âgés, après délivrance, et le poids qu'ils supportent, sont en rapport direct avec le poids de l'animal à sa mort.
    Abstract: Zusammenfassung Die Femora von jüngeren und älteren Ratten wurden mittels mechanischer Tests miteinander in Bezug auf ihre Fähigkeit verglichen, der physiologischen Beanspruchung von Schwangerschaft und Lactation standzuhalten. In beiden Altersgruppen zeigte sich eine Tendenz der Knochen, am Ende der Schwangerschaft stärker zu sein als diejenigen von ungedeckten Tieren desselben Alters. Diese Wirkung war ausgeprägter in den älteren Tieren. Als Folge waren die Knochen der älteren Tiere am Ende der Lactation beinahe so stark wie diejenigen der ungedeckten Tiere deselben Alters, obwohl die Knochen während der Lactation schwächer wurden. Bei den jüngeren Tieren waren die Knochen am Ende der Lactation schwächer als diejenigen der ungedeckten Tiere. Möglicherweise hat die Zunahme der Knochenstärke nach der Schwangerschaft einen Zusammenhang mit dem höheren Gewicht dieser Tiere im Vergleich mit den ungedeckten Tieren; die Querschnittfläche der Knochen älterer postpartaler Tiere und die Last, die sie aushielten, hatten einen engen Zusammenhang mit dem Gewicht des Tieres bei dessen Tod.
    Notes: Abstract The femora of younger and older rats were compared, by means of mechanical tests, for their ability to withstand the physiological stresses of pregnancy and lactation. In both age groups there was a tendency for the bones at the end of pregnancy to be stronger than those of virgins of the same age. This effect was much more clearly marked in the older than in the younger animals. As a result, the bones of older animals were almost as strong, at the end of lactation, as those of virgins of the same, age, even though the bones became weaker during lactation. The bones of younger lactators were weaker than those of virgins of the same age. It is possible that the increase in strength of the postpartum animals is related to the greater weight of such animals compared with virgins; the cross-sectional area of the bones of the older postpartums, and the load they bore, had a high correlation with the weight of the animal at death.

    Description / Table of Contents: Résumé L'effet du fluor, de la parathormone et de la calcitonine sur le métabolisme du calcium osseux a été étudié en culture de tissu en utilisant des demi-calottes crâniennes de souris âgées de cinq jours. Les concentrations de fluor dans les cendres des calottes crâniennes de 0,007% (groupe peu fluoré), 0,041% (groupe moyennement fluoré) et 0,107% (groupe hautement fluoré) ont été obtenues en variant l'ingestion maternelle et post-natale en fluor. Le fluor inhibe la perte de calcium des os cultivés dans le milieu témoin et celui contenant la parathormone et favorise l'enrichissement en laccium des os cultivés dans un milieu contenant de la calcitonine. De l'os mort des groupes fortement et moyennement fluorés s'enrichit plus en calcium à partir du milieu de culture que les os du groupe pauvre en fluor. Le fluor semble agir sur le métabolisme du calcium osseux essentiellement par diminution de la solubilité minérale.
    Abstract: Zusammenfassung Der Einfluß von Fluorid, Parathormon und Calcitonin auf den Calciumstoffwechsel im Knochen wurde in einem Knochenkultur-System untersucht, wobei Halbcalvarien von 5 Tage alten Mäusen verwendet wurden. Fluoridgehalte in der Asche der Halbcalvarien von 0,007% (Gruppe mit niedrigem Fluoridgehalt), 0,041% (Gruppe mit mittlerem Fluoridgehalt) und 0,1075 (Gruppe mit hohem Fluoridgehalt) wurden erhalten, indem die Fluorideinnahme der Mütter oder der Neugeborenen verändert wurde. Fluorid hemmte den Calciumverlust aus Knochen, welche im Kontrollmedium oder in Parathormon-enthaltenden Medien kultiviert worden waren, und es förderte die Calciumaufnahme von Knochen, welche in Calcitoninenthaltendem Medium kultiviert worden waren. Die toten Knochen der Gruppen mit mittlerem und hohem Fluoridgehalt nehmen mehr Calcium aus dem Kulturmedium auf als die Knochen der Gruppe mit niederem Fluorid. Fluorid scheint vor allem durch eine Herabsetzung der Mineral-Löslichkeit auf den Calciumstoffwechsel der Knochen zu wirken.
    Notes: Abstract The influence of fluoride, parathyroid hormone and calcitonin on bone calcium metabolism was investigated in a bone culture system using half-calvaria of five-day old mice. Fluoride levels in the ash of half-calvaria of 0.007% (low fluoride group), 0.041% (moderate fluoride group), and 0.107% (high fluoride group) were achieved by varying the maternal and neonatal intake of fluoride. Fluoride inhibited the loss of calcium from bones cultured in control medium and parathyroid hormone-containing media, and promoted the uptake of calcium by bones cultured in medium containing calcitonin. Dead bones of the moderate and high fluoride groups took up more calcium from the culture medium than bones of the low fluoride group. Fluoride appears to exert its effect on bone calcium metabolism predominantly via a reduction in mineral solubility.

    Description / Table of Contents: Résumé La mithramycine, qui est un antibiotique bloquant le métabolisme de l'ARN, lié à l'ADN, possède des propriétés hypocalcémiquesin vivo. Cette action pourrait être en rapport avec une inhibition de la résorption osseuse. La résorption osseuse de cet antibiotique a été testéein vitro en utilisant des calottes craniennes de souris nouveau-nés. Il semble que la mithramycine, non seulement inhibe la résorption osseusein vitro, mais inhibe aussi la formation osseuse et se révèle cytotoxique à toutes le concentrati ons qui inhibent la résorption osseuse. L'efficacité de l'antibiotique comme inhibiteur de résorption parait liée au degré d'activité de résorption observée lorsqu'on l'introduit dans ce système. Des observations morphologiques posent le problème des anomalies provoquées par cet agent dans le processus normal de différenciation de cellules osseuses au cours de la formation du cartilage.
    Abstract: Zusammenfassung Mithramycin, ein Antibioticum, welches den DNS-abhängigen RNS-Stoffwechsel blockiert, wirktin vivo hypocalcämisch. Es wurde vermutet, daß diese Wirkung durch eine Hemmung der Knochenresorption erzielt wurde. Wir haben dieses Antibioticum auf seine Wirkungen auf die Knochenresorptionin vitro geprüft und verwendeten zu diesem Zwecke Schädeldächer von neugeborenen Mäusen. Die vorliegende Arbeit enthält den Beweis, daß Mithramycin nicht nur die Knochenresorptionin vitro, sondern auch die Knochenbildung hemmt und daß es in allen Konzentrationen, welche die Resorptionshemmung bewirken, cytotoxisch ist. Die Wirksamkeit dieses Antibioticums als Resorptionshemmer scheint im Zusammenhang zu stehen mit dem Ausmaß der Knochenresorptions-Aktivität zum Zeitpunkt, da das Antibioticum in das System eingeführt wird. Morphologische Beobachtungen ließen auch die Frage aufkommen, ob Mithramycin Abnormalitäten im normalen Ablauf der Knochenzell-Verwandlungen, welche zur Bildung von Knorpel führen, verursachen könne oder nicht.
    Notes: Abstract Mithramycin, an antibiotic which blocks DNA-dependent RNA metabolism, has been demonstrated to be hypocalcaemicin vivo. It has been suggested that this effect is obtained by an inhibition of bone resorption. We have tested this antibiotic for its effects on bone resorptionin vitro using new-born mouse calvaria and this report presents evidence that mithramycin not only inhibits bone resorptionin vitro but also inhibits the formation of bone and is cytotoxic at all concentrations which appear to be effective at inhibiting resorption. The effectiveness of this antibiotic at inhibiting resorption appears to be related to the degree of bone resorption activity present when it is introduced to the system. Morphological observations also raised the question as to whether or not this agent may cause abnormalities in the normal processes of bone cell modulation resulting in the formation of cartilage.

    Notes: Abstract Bone matrix demineralized in 0.6 N HCl at 2° for 24 h and implanted in muscle in allogeneic rats possesses consistently reproducible bone morphogenetic activity. Experiments on implants of matrix, obtained from donors injected with3H-tyrosine or3H-tryptophan, or Na35SO4, suggest that bone morphogenetic property is a protein or apart of a protein that is (1) insoluble in buffer solutions, pH 3.6 and 5.0; (2) degraded in buffer solutions at pH 7.4 by an endogenous sulfhydryl-group neutral proteinase; (3) digested by trypsin at 15° within 8 h without solubilization of the helical regions, possibly even without degradation of the nonhelical ends of the bone collagen molecule, and without any loss of the periodic ultrastructure of the collagen fibrils; (4) degraded or removed by 0.1 N NaOH at 2° within 24 h without solubilization of collagen; (5) biologically active even after nitration of tyrosyl groups with tetranitromethane. The release of only one-third of the radioactivity with loss of nearly all yield of new bone by limited tryptic digestion of3H-borohydride-reduced matrix indicates that the bone morphogenetic response is the function of a non-collagenous component. Autoradiographs of implants of matrix with non-collagenous proteins labelled with3H-tryptophan,3H-tyrosine, or both3H-tyrosine and3H-phenyl-alanine demonstrate random dissemination of the radioactive constituents and no evidence of local transfer of labelled proteins or soluble protein derivatives. Hypothetically, the bone morphogenetic response is controlled by an insoluble acidic bone morphogenetic protein or polypeptide (BMP) and a soluble neutral proteinase (BMP-ase) resembling trypsin in activity except functionally more specific for BMP. Firmly bound but separable from bone collagen, BMP is one of many short-lived morphogenetic substances appearing and disappearing throughout embryonic development and persisting in postfetal life. Where the BMP receptor resides and how it activates cell mechanisms of differential repression and derepression of such genes as code for osteogenesis is unknown.

    Notes: Abstract A series of analogues of vitamin D have been tested for their ability to stimulate bone resorption in two test systems used previously to investigate the metabolites of vitamine D. These analogues were tested (a) by directly comparing their action on bone explants of mouse half-calvariain vitro, and (b) by injecting them into young mice and measuring the degree of resorptionin vitro when explants were prepared 18 hours after the injection. It is concluded that the key functional groups concerned with enhancing the activity of vitamin D3 are the 1α- and the 25-hydroxyl,both together; the cis ring structure for ring A appears necessary. 1α-Hydroxycholecalciferol (1α-OHD3) is about as active as 25-OHD3 in the direct test, but its potency is much nearer to that of 1,25-(OH)2D3 when tested by the second (indirect) method; it seems likely that 1α-OHD3 is converted into 1,25-(OH)2D3 in vivo. The results are discussed in relation to the designing of analogues for clinical and experimental use.

    Notes: Abstract The effects of disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) on bone and serum chemistry were investigated in adult rabbits. EHDP was administered by subcutaneous injection at doses of 0.25, 2.5 and 10 mg/kg body weight/day for of 28 days. Blood samples were obtained weekly from each rabbit and serum levels of total calcium, ionized calcium, inorganic phosphate and alkaline phosphatase were determined. At the end of the treatment period all rabbits were sacrificed and the tibiae removed for chemical analysis and histological evaluation. The effect of EHDP administration on serum chemistry was both dose- and time-related. The highest of the three doses, 10 mg/kg/day, resulted in a time-related decrease in total serum calcium. This dose also caused a rapid but transient reduction in serum ionized calcium. The effect of EHDP on serum inorganic phosphate was biphasic. Administration of 2.5 mg/kg/day resulted in a time-related elevation in this parameter, whereas the 10 mg/kg/day dose resulted in a time-related hypophosphatemic response. There were no significant drug-related changes in tibial fat-free dry weight, ash weight, total calcium or total phosphorus values. However, administration of 2.5 and 10 mg/kg/day EHDP resulted in increased osteoid tissue as measured histologically. These results are compared with data from other EHDP studies, and discussed in relation to the maturity and growth-state of the experimental animals.

    Notes: Abstract In order to obtain enough material to analyse the organic matrix of mineralizing tissue a technique for preparative separation according to the degree of mineralization was developed. The method employs ultracentrifugation of powdered material in density gradients made from acetone and bromoform. The density range of the fractions is checked by refractive index measurements of the gradient medium. The amount of the material in the fractions is checked by weighing and their degree of mineralization is estimated by determining the Ca/N and P/N ratios. The homogeneity of the fractions is determined by soft X-ray microscopy. Isolated dissected microscopic bone structure (osteones and lamellar bone fragments) with different degrees of mineralization were fractionated in this way. Chromatography on Sepharose 2B of proteoglycans from costal cartilage exposed to an acetone-bromoform gradient revealed no effect of the gradient medium on the molecular size of the proteoglycans.

    Description / Table of Contents: Résumé Des techniques histochimiques de microscope photonique et électronique montrent que les ostéoclastes métaphysaires chez des rats “sans incisive” présentent des quantités plus importantes de phosphatase acide, d'acryl sulfatase et de trimétaphosphatase acide lysosomiales. L'activité en phosphatase lysosomiale à pH neutre est aussi plus élevée dans les ostéoclastes métaphysaires, sauf dans les cellules situées sous la métaphyse, où l'enzyme est absente. L'absence de résorption de la matrice organique semble en rapport avec l'absence de bordures en brosse et une absence d'enzyme lysosomiale extracellulaire. Malgré cette absence, une dissolution de cristaux inorganiques a été mise en évidence au microscope électronique, suggérant que la dissolution minérale est un processus distinct de la désintégration enzymatique de la matrice organique.
    Abstract: Zusammenfassung Histochemische Techniken für Licht- und Elektronenmikroskopie zeigten, daß metaphysäre Osteoklasten “Schneidezahl-loser” (SL) Ratten erhöhte Mengen von lysosomaler saurer Phosphatase, von Arylsulfatase und von saurer Trimetaphosphatase enthielten. Die Aktivität der lysosomalen Phosphatase bei neutralem pH war in den metaphysären Osteoklasten ebenfalls erhöht, außer in den Zellen direkt unterhalb der Wachstumsplatte, wo dieses Enzym nicht vorkam. Es konnte überhaupt keine Resorption der organischen Matrix festgestellt werden, was übereinzustimmen schien mit der Abwesenheit eines gekräuselten Saumes und einer gleichzeitigen Abwesenheit des extrazellulären lysosomalen Enzyms. Trotzdem wurde im Elektronenmikroskop das Verschwinden anorganischer Kristalle festgestellt, was darauf schließen läßt, daß die Auflösung des Minerals nicht derselbe Vorgang ist wie die enzymatische Auflösung der organischen Matrix.
    Notes: Abstract Histochemical techniques for light and electron microscopy showed that metaphyseal osteoclasts in “incisors absent” rats contained greater than normal amounts of lysosomal acid phosphatase, aryl sulfatase and acid trimetaphosphatase. Lysosomal phosphatase activity at neutral pH was also elevated in the metaphyseal osteoclasts except in those cells immediately beneath the growth plate, where this enzyme was absent. The failure of any discernable resorption of organic matrix appeared to correlate with the absence of a ruffled border and a concomittant absence of extracellular lysosomal enzyme. Despite this failure, electron microscopic evidence of inorganic crystal removal was noted, suggesting that mineral dissolution represents a separate process from the enzymatic breakdown of organic matrix.

    Notes: Abstract The organic-inorganic relationships in bone matrix undergoing osteoclastic resorption have been studied in rat tibial diaphyses using electron microscope techniques in an attempt to identify the steps of the resorption process. Results suggest that bone resorption occurs in two phases: the first, an extracellular phase, leads to bone matrix fragmentation and partial dissolution, and the second, an intracellular phase, to complete digestion of the breakdown products of the bone matrix. The first component of the bone matrix to be attacked by the osteoclast is the ground substance. This induces the release of the crystals lying between, and on, the collagen fibrils; any crystals lying within fibrils are released later, when the fibrils break up. As this stage proceeds, the collagen fibrils retain their normal intrinsic texture, but gradually loose their lateral aggregation, appearing as individual fibrils (some of them uncovered by crystals), mixed with fragments of fibrils and many free crystals. The loosened but otherwise structurally normal collagen fibrils, and their fragments, are strongly argyrophilic. Complete dissolution of the disaggregated fibrils occurs outside the cell, both in the resorption zone and in the initial portion of the channels of the ruffled border. The free crystals present in the resorption zone and those phagocytosed in cytoplasmic vacuoles are organic-inorganic structures, whose organic component (the crystal ghost) is, at least in part, of proteoglycan nature. Dissolution of inorganic material occurs within the cytoplasmic vacuoles of the osteoclasts. Results are viewed in relation to the process of bone resorption and, as far as crystal ghosts are concerned, to that of bone calcification. A tentative summary of the various steps involved in the mechanism of bone resorption is given.

    Notes: Abstract Phenytoin, at a concentration which is known to inhibit parathyroid extract-induced bone resorptionin vitro, has been shown to have no significant effect on either the cyclic AMP content of four-day-old mouse calvaria which had been in tissue culture for 48 h or on the increase in cyclic AMP content occurring after the addition of parathyroid extract to the medium. Phenytoin has also been shown to be an effective inhibitor of bone resorptionin vitro induced by 25-hydroxycholecalciferol. These observations suggest that the inhibitory effect of phenytoin on parathyroid extract and hormone-induced bone resorption takes place beyond the cyclic AMP stage.

    Notes: Abstract An active constituent of the leaves ofSolanum malacoxylon (SM) is shown to promote calcium resorption and citrate production in embryonic chick frontal bone culturedin vitro. When injected into rats, SM reduces the ATP content of liver and kidneys. This phenomenon may be related to the ability of SM to stimulate mitochondrial ATPase activity at pH 9.4. SM significantly reduces the concentration of phosphate necessary to alter the uptake of calcium and respiration of siolated mitochondria. The data suggest that SM influences calcium and phosphate metabolism by affecting ion movements into and out of mitochondria.

Des crîstaux d'apatite synthétique, préparés dans des conditions identiques de T, μ et (HCO 3 − ), ne contiennent que 1/7ème à 1/8ème du CO2 d'arêtes de poissons. Ces résultats indiquent que la composition des liquides dans l'os ne réfléchit pas simplement la composition du sérum circulant, ainsi qu'on l'admet généralement.
    Abstract: Zusammenfassung Es ist in der Literatur berichtet worden, daß Fische, um genügend Sauerstoff zu erhalten, einen Überschuß von Kohlensäure einatmen müssen und folglich einen niedrigen Gehalt an Bicarbonaten im Kreislauf aufweisen. Knochen von Forellen und Karpfen wurden analysiert, um festzustellen, ob ihre Mineralsalzzusammensetzungen die niedrigen Serum-bicarbonate-konzentrationen wiederspiegeln. Die Resultate zeigten keinen wesentlichen Unterschied zwischen dem Carbonatgehalt der Knochen von Fischen und Säugetieren. Synthetische Apatitkristalle, die unter ähnlichen Bedingungen von T, μ und HCO3 bereitet wurden, wiesen nur 1/7–1/8 des Carbonatgehaltes der Fischknochen auf. Diese Ergebnisse deuten zweifellos darauf hin, daß zwischen der Zusammensetzung der Gewebeflüssigkeit der Knochen und des Serums kein einfacher Zusammenhang besteht.
    Notes: Abstract It has been reported in the literature that fish, to get sufficient oxygen, must overventilate with respect to CO2 and therefore exhibit a low circulating level of bicarbonate. Accordingly, the bones of trout and carp were analyzed to learn if the composition of their bone mineral reflects the low serum level of bicarbonate. It was found that the CO2 content of fish bone is not significantly different from that of normal mammalian bone. Synthetic apatite crystals, made under comparable conditions of T, μ, and (HCO 3 − ) were found to contain only 1/7th to 1/8th the CO2 of fish bone. These data strongly suggest that the composition of the fluids in bone does not reflect, in a simple way, the composition of the circulating serum as is generally assumed.

    Description / Table of Contents: Résumé Une hypothèse, destinée à rendre compte de façon semi-quantitative, des grandes variations de temps nécessaire à équilibrer, au niveau du squelette, divers produits radioactifs, introduits dans le torrent circulatoire, est mise au point. Cette hypothèse est basée sur le fait que 3 facteurs interviennent dans les échanges osseux: a) la vitesse de perfusion de l'os, b) la concentration ionique du plasma, et c) la concentration ionique de l'os. Partant de cette hypothèse et de résultats publiés dans la littérature, “des temps de cyclage” sont calculés par Cl−, Na+, K+ et Ca++. Ils varient selon quatre ordres de grandeur (de 10 min pour Cl− à 64 jours Ca++, chez le rat). Ces résultats ont été vérifiés pour des échanges de22Na et de45Ca dans une colonne d'apatite minéralin vitro. Des échanges de36Cl et de42K ont été étudiésin vivo au niveau du fémur de rat. La disparition sanguine du22Na a été suivie chez l'Homme. Enfin des résultats d'échanges de45Ca, au niveau de molaires de rat, publiés dans la littérature, ont été recalculés. Dans chaque cas, les résultats expérimentaux ont confirmé les prédictions de l'hypothèse du cyclage.
    Abstract: Zusammenfassung Es wurde eine Hypothese aufgestellt, um auf semi-quantitative Weise die breite Variabilität der Zeit zu erklären, die benötigt wird, um im Skelett ein Gleichgewicht von verschiedenen radioaktiven in den Kreislauf eingeführten Substanzen herzustellen. Diese Hypothese beruht auf der Annahme, daß der geschwindigkeitsbeschränkende Faktor beim Knochenaustausch durch 3 Variablen definiert ist: a) das Maß der Knochenperfusion, b) die Konzentration der Ionen im Plasma und c) die Konzentration der Ionen im Knochen. Auf Grund dieser Annahme und gestützt auf Angaben aus der Literatur wurden die “cycling times” für Cl−, Na+, K+ und Ca++ berechnet. Diese variieren in 4 Größenordnungen, beispielsweise von 10 min für Cl− bis zu 64 Tagen für Ca++ bei der Ratte. Diese Annahmen wurden für den Austauschin vitro von22Na und45Ca in Apatitmineral in einer Säule geprüft. Der Austausch von36Cl und von42K wurdein vivo am Rattenfemur studiert. Die Abnahme von22Na im menschlichen Blut wurde ebenfalls verfolgt. Schließlich wurden Literaturangaben über den Austausch von45Ca in Rattenbackenzähnen überprüft. Die Versuchsresultate kamen dabei sehr nahe an die auf Grund des “cycling concept” erwarteten. Einige der Zusammenhänge dieser Befunde werden kurz besprochen.
    Notes: Abstract An hypothesis has been developed to explain in a semi-quantitative fashion the wide variations in time required for the equilibration in the skeleton of various radioactive substances when they are introduced into the circulation. The basis for the hypothesis rests on the assumption that three variables define the rate-limiting step in bone exchange: a) the rate of perfusion of bone, b) the concentration of the ion plasma, and c) the concentration of the ion in bone. Using this idea and data from the literature, “cycling times” were calculated for Cl−, Na+, K+, and Ca++. They were found to vary by four orders of magnitude (from 10 min for Cl− to 64 days for Ca++ in the rat). These predictions were tested for22Na- and45Ca-exchange in a column of apatite mineralin vitro. In vivo 36Cl- and42K-exchange in rat femur was studied. The blood disappearance of22Na in man was also examined. Finally, data in the literture of45Ca-exchange in the rat molar was redrawn. In every case, experimental results closely approximated the predictions of the cycling concept. Some of the implications of these findings are briefly discussed.

    Description / Table of Contents: Résumé Les glycosaminoglycanes de l'os compact ont été identifiés, par la méthode de précipitation au chlorure de cetylpyridinium, chez le chien normal et des chiens soumis à 20 ou 30 U.S.P. d'extrait parathyroïdien pendant 5 et 3–4 jours. Les concentrations d'héxosamines totales ainsi que les héxosamines, en rapport avec les glycoaminoglycanes acides, précipités par chlorure de cetylpyridinium, sont déterminées. En outre, la répartition du glycosaminoglycane de la zone d'ossification épiphysaire ainsi que l'incorporation du35S-sulfate dans les glycosaminoglycanes du tissu osseux et du cartilage épiphysaire, après administration de35S-sulfatein vivo, ont été étudiées. Dans l'os compact, les héxosamines, en rapport avec les glycosaminoglycances acides, constitutent environ un tiers de la concentration totale en héxosamine et environ 0,05–0,06% du poids sec total. Le constituant principal des glycosaminoglycances acides osseux est formé par le chondroitine-4-sulfate. Ce dernier est plus riche en sulfate et présente un poids moléculaire plus élevé que le chondroitine sulfate du cartilage épiphysaire, qui, selon des travaux antérieurs, présente des caractéristiques infra-rouges du chondroitine-4-sulfate et du chondroitine-6-sulfate, avec prédominance du premier. Les poids moléculaires du chondroitine sulfate osseux varient surtout d'environ 45000 et 56000. L'acide hyaluronique constitute une faible fraction des glycosaminoglycanes osseux. Des différences marquées de l'activité spécifique des fractions de cohondroitine sulfate de l'os et du cartilage épiphysaire, à poids moléculaires variables, on tété notées de façon répétée. L'administration d'extrait parathyroïdien à des chiens n'a pas d'effet sur les poids moléculaires en chondroitine sulfate de l'os ou du cartilage épiphysaire. Elle n'influence pas non plus les concentrations en héxosamines totales ou en glycosaminoglycances acides. Dans ces tissus, il ne se produit pas d'effect de stimulation ou de dépression concernant l'incorporation de35S-sulfate dans le chondroitine sulfate our sur la séparation moléculaire de molécules transformées en sulfates et/ou de molécules synthétiques de sulfate chondroitine.
    Abstract: Zusammenfassung An normale Hunden und an Hunden die unter 5 respektive 3–4 Tagen 20 oder 30 U.S.P./kg Parathyreoidea-Extract erhalten hatten, wurden im kompakten Knochen die Glykosaminoglykane unter Anwendung der Cetylpyridiniumchlorid-Fällungsmethode identifiziert und die Konzentration des Total-Hexosamins und des Hexosamins, entsprechend den Cetylpyridiniumchlorid fällbaren sauren Glykosaminoglykanen, wurde bestimmt. Außerdem wurden das Glykosaminoglykanmuster der Epiphysenplatte und der Einbau von35S-Sulfat in die Glykosaminoglykane des Knochengewebes und des Epiphysenknorpels nach Zufuhr von35S-Sulfatin vivo studiert. Im kompakten Knochengewebe macht des Hexosamin, entsprechend den sauren Glykosaminoglykanen, ungefähr ein Drittel der totalen Hexosaminkonzentration und ungefähr 0,05–0,06% des totalen Trockengewichtes aus. Der Hauptanteil der sauren Glykosaminoglykane im Knochen war Chondroitin-4-Sulfat. Dieses war in höherem Grad sulfatiert und hatte ein höheres Molekülgewicht als das Chondroitinsulfat der Epiphysenplatte, welches, in Übereinstimmung mit früheren Untersuchungen, Infrarot spektra kennzeichnend für sowohl Chondroitin-4-sulfat als auch für Chondroitin-6-sulfat, das Erstere überwiegend, hatte. Das Molekülgewicht des Hauptanteiles des Knochen-Chondroitinsulfates lag zwischen ungefähr 45000–56000. Ein kleiner Teil der Knochen-Glykosaminoglykane war Hyaluronsäure. Sowohl im Knochengewebe als auch im Epiphysenknorpel wurde in verschiedenen Fraktionen des Chondroitinsulfates, mit unterschiedlichem Molekülgewicht, grße und regelmäßig reproduzierbare Unterschiede in der spezifischen Aktivität gefunden. Behandlung der Hunde mit Parathyreoideaextrakt gab keinen Ausschlag in den Molekülgewichten des Chondroitinsulfates, weder des Knochens noch des Epiphysenknorpels. Ebenso wurde kein eindeutiger Effekt auf die Konzentration des totalen Hexosamins oder der sauren Glykosaminoglykane gefunden. Kein offenbarer, weder anregender noch senkender, Effekt auf den Einbau von35S-Sulfat in das Chondroitinsulfat oder in der molekularen Verteilung der neulich sulfatierten und/oder synthetisierten Moleküle des Chondroitinsulfates dieser Gewebe wurde gefunden.
    Notes: Abstract In normal dogs and in dogs treated with 20 or 30 U.S.P. parathyroid extract for 5 and 3–4 days, respectively, the glycosaminoglycans of compact bone tissue were identified using the cetylpyridinium chloride precipitation method, and the concentrations of total hexosamines and the hexosamines corresponding to cetylpyridinium chloride precipitable acid glycosaminoglycans were determined. Further, the glycosaminoglycan pattern of the epiphyseal plate and the incorporation of35S-sulphate into the glycosaminoglycans of bone tissue and epiphyseal cartilage after administration of35S-sulphatein vivo was studied. In compact bone tissue, the hexosamines corresponding to acid glycosaminoglycans constuted approximately one third of the total hexosamine concentration and approximately 0.05–0.06% of the total dry weight. The main component of the acid glycosaminoglycans in bone was chondroitin-4-sulphate. This was sulphated to a higher degree and also of a higher molecular weight than thechondroitin sulphate of the epiphyseal cartilage, which in accordance with earlier investigations was found to have infrared characteristics of both chondroitin-4-sulphate and chondroitin-6-sulphate, with the former dominating. The molecular weights of the main part of bone chondroitin sulphate ranged from approximately 45,000 to 56,000. A small component of the bone glycosaminoglycans was hyaluronic acid. Large regularly recurring differences in the specific activity of fractions with differences in molecular weight in the condroitin sulphate of bone tissue and epiphyseal cartilage were noted. Treatment of the dogs with parathyroid extract gave no effect on the molecular weights of the chondroitin sulphate of the bone matrix or of the epiphyseal cartilage. Nor was there any unequivocal effect on the concentrations of total hexosamines or on the acid glycosaminoglycans. No evident stimulatory or depressant effect on the incorporation of35S-sulphate into the chondroitin sulphate or in the molecular distribution of newly sulphated and/or synthesized molecules of the condroitin sulphate within these tissues occurd.

    Description / Table of Contents: Résumé Une technique nouvelle, permettant l'analyse de la densité de faibles quantités de tissus calcifiés pulvérisés, est décrite. Cette technique utilise un analyseur électronique de taille de particules et de volume (»Compteur de Coulter«), pour déterminer les volumes relatifs des fractions pulvérisées, isolées par fractionnement densitométrique, après centrifugation dans des mélanges de solvants organiques. Certains des paramètres, responsables de la distribution densitométrique des poudres d'os, sont étudiés et la reproductibilité du fractionnement et de l'analyse est démontrée. L'application de cette méthode à l'os humain est illustrée par des résultats.
    Abstract: Zusammenfassung Es wird eine neue Technik zur Bestimmung der Dichteverteilung von kleinen Mengen pulverisiertem Knochengewebe beschrieben. Für diese Technik wird ein Apparat zur elektronischen Messung von Partikelgröße und-Volumen (Coulter Counter) verwendet; damit wird das totale relative Volumen der pulverisierten Fraktionen bestimmt, die durch Aufteilung der verschiedenen Dichten mittels Zentrifugieren in Mischungen von organischen Lösungsmitteln isoliert werden. Einige der Parameter, welche die Dichteverteilung von Knochenpulver beeinflussen, wurden untersucht und die Reproduzierbarkeit der Fraktionierung und der Analysen aufgezeigt. Die Anwendung dieser Technik auf Proben von Menschenknochen wird veranschaulicht und die Resultate werden besprochen.
    Notes: Abstract A new technique for the density distribution analysis of small quantities of powdered hard tissues is described. The technique uses an electronic particle size and volume analyser (“Coulter Counter”) to determine the total relative volumes of the powder fractions isolated by centrifugal density fractionation in organic solvent mixtures. Some of the parameters controlling the density distributions of bone powders are examined and the reproducibility of the fractionation and analysis demonstrated. The application of this technique to human bone samples is illustrated and the results discussed.

F sous forme de Na2PO3F et Ca sous forme de gluconate de calcium (CaGluc) n'influençaient pas l'un l'autre quant à l'utilisation par le squelette (fémur). Une viscosité élevée, produite par l'addition d'amidon ou de cellulose carboxyméthylée (CMC) à la solution ou dilution ingérée, augmentait l'utilisation du F même si CaGluc était remplacé par citrate de calcium, qui en soi avait un effet réducteur modéré sur l'utilisation du F. Le glycérophosphate de calcium réduisait fortement l'utilisation du F même en présence de CMC. L'utilisation du F comme NaF était fortement réduite par CaGluc, même en présence de CMC. Les concentrations testées de Na2PO3F, NaF ou CMC n'influençaient pas l'utilisation squelettique de Ca comme CaGluc.
    Abstract: Zusammenfassung Es wurden Rattenexperimente mit markierten Substanzen durchgeführt, um die Auswirkung simultaner peroraler Gaben von Fluor (F) und von Calcium (Ca) zu prüfen, und zwar in einem Verhältnis, das für die Behandlung gewisser menschlicher Osteopenien berechnet wurde. Fluor in Form von Na2PO3F und Ca in Form von Calciumgluconat (CaGluc) interferieren gegenseitig nicht bei der Verwertung durch das Skelet (Femur). Eine hohe Viscosität der eingegebenen Lösung oder der Aufschlämmung, die durch Zusatz von Stärke oder Carboxymethylcellulose (CMC) erzielt wurde, erhöhte die Verwertbarkeit von F sogar wenn CaGluc durch Calciumcitrat ersetzt wurde, welches die F-Verwertung leicht reduzierte. Calcium-glyzerophosphat verminderte die Fluoraufnahme in den Knochen stark, sogar in Anwesenheit von CMC. Die Verwertung von F als NaF war stark herabgesetzt durch CaGluc, selbst beim Vorhandensein von CMC. Die untersuchten Konzentrationen von Na2PO3F, NaF oder CMC hatten keinen Einfluß auf die Calciumaufnahme im Skelet in Form von CaGluc.
    Notes: Abstract Rat experiments with labelled compounds were carried out in order to test the possibilities of simultaneous peroral supply of fluorine (F) and calcium (Ca) in proportions calculated for treatment of certain human osteopenias. F in the form of Na2PO3F and Ca in the form of calcium gluconate (CaGluc) did not interfere with each other's utilisation by the skeleton (femur). A high viscosity produced by adding starch or carboxymethyl cellulose (CMC) to the ingested solution or slurry increased the utilisation of F even when CaGluc was replaced by calcium citrate, which moderately reduced F utilisation. Calcium glycerophosphate strongly reduced F utilisation even in the presence of CMC. The utilisation of F as NaF was strongly reduced by CaGluc, even in the presence of CMC. The tested concentrations of Na2PO3F, NaF or CMC did not influence the skeletal utilisation of Ca as CaGluc.

    Notes: Abstract A method is presented for separating osteoclasts from the heterogeneous population of bone and marrow cells. Cell suspensions were prepared from femora of young rabbits by mechanical dispersion. The starting cell suspension typically contained only 1.0%±0.5 osteoclasts. Following an initial 45 min of unit gravity sedimentation in a lucite chamber osteoclasts were primarily distributed in fractions 2–5. A second 45-min sedimentation of these pooled fractions yielded cell suspensions containing greater than 30% osteoclasts (as much as a 50-fold increase over starting percentages). Linear scan analysis, however, revealed that osteoclasts accounted for 73.14%±0.58 of the cell colume. Subsequent in vitro experiments demonstrated linear incorporation of3H-leucine into TCA precipitable protein for cells comprising the osteoclast fraction. Concomitant radiautographs revealed radioactive label in isolated osteoclasts.

    Notes: Summary Male rats weighing 100 g were assigned to groups and injected daily for 10 days with vehicle (control), 0.4, 2.0, 4.0, 10.0, or 20.0 mg/kg/day of ethane-1-hydroxy-1,1-diphosphonate (EHDP) or dichloromethylene diphosphonate (Cl2MDP). The proximal tibial metaphysis and epiphysis were assayed for changes in percentage of hard tissue and bone formation parameters. From the data, information about hard tissue resorption was deduced. All doses of Cl2MDP and doses of 2.0 mg EHDP/kg/day and greater caused significant increases in percentage of hard tissues with Cl2MDP being more effective than similar doses of EHDP in decreasing bone resorption. Osteoclast population parameters were increased with all doses of both Cl2MDP and EHDP with Cl2MDP having a greater effect than similar doses of EHDP. Decreases in the proliferation of the osteoprogenitor pool parallel the decreases in osteoblasts and bone formation parameters. These decreases in osteoprogenitor pool proliferation do not account for the increases with diphosphonates in osteoclast population parameters.

    Notes: Summary The stimulation of bone resorption, assessed by the release of45Ca from prelabeled bones, was associated with an increase in number of osteoclasts per bone section in parathyroid hormone (PTH)-treated bones, but not in lipopolysaccharide (LPS)-treated bones. By contrast the number of nuclei per osteoclast increased following LPS treatment, but was not affected by PTH. LPS-treated bones had more multinucleated cells, some having as many as 27 nuclei per osteoclast. More osteoclasts were adjacent to the bone collar in bones treated with LPS or PTH than in control bones. In LPS-treated bones this area also contained the largest osteoclasts, as determined by the greatest number of nuclei per osteoclast. The results suggest that LPS and PTH stimulate osteoclastic resorption by different mechanisms.

The results show that albumin is more readily extracted from bone pieces than from bone powder which implies that tissue disruption exposes additional protein adsorption sites. It is suggested that incorporation of plasma albumin into calcified matrix during bone formation occurs mainly as a result of its strong interaction with bone mineral.

    Notes: Summary Cortical bone remodeling along the femur diaphysis was determined in normal female rats (Sprague-Dawley) with the tetracycline technique. Three segments on the cortical bone circumference (the anterolateral, the medial, and the posterior) were found to be most suitable for the study of the remodeling process. Oxytetracycline was administered at age 60 and 75 days, and groups of animals were killed at age 75, 85, 95, and 105 days. The accumulated endosteal growth during age 60 to 75 days in the anterolateral segment was found to increase uniformly in the distal direction along the femur diaphysis. A method is described where this accumulated endosteal growth is used. This method eliminates the use of calipers to determine the section level and makes it possible to study comparable sections even after varying periods of time. The proximal part of the diaphysis showed periosteal apposition in all three segments. The periosteal apposition turned into resorption in the distal part of the diaphysis in the anterolateral and medial segments, whereas the periosteal appsition increased in the posterior segment. The endosteal growth increased in the distal direction in the anterolateral and medial segments. Irregular OTC bands made measurements of endosteal remodeling in the posterior segment impossible. The cortical width decreased in the distal direction along the femoral shaft. Comparison between the different age groups is described and also the relation between the accumulated endosteal growth and the diameter of the medullary cavity.

    Notes: Summary The elaboration of bone matrix glycoprotein by osteoblasts of alveolar bone was investigated by radioautography after the intravenous injection of3H-fucose into young rats. At selected times after injection, animals were sacrificed by intracardiac perfusion and demineralized specimens were prepared for light and electron microscope radioautography. At 5 and 10 min after injection, when the blood fucose level was high, silver grains were restricted to the spheroidal and cylindrical saccules of the Golgi apparatus. At 20 min membrane-limited secretory granules were also labeled. By 35 min, the blood fucose level had dropped and silver grains were detected over the apical cortical cytoplasm, in association with secretory granules located therein. Some grains were present over osteoblast processes and the adjacent prebone matrix. By 4 h most of the silver grains had left the cell. At that time they were observed over prebone, adjacent to osteoblast processes, as well as over the prebone-bone junction where a distinct band of label was noted. In demineralized preparations an electron-dense granular material was present at the prebone-bone junction in association with collagen fibrils. These findings provide evidence that osteoblasts in alveolar bone synthesize fucose-containing glycoprotein and indicate that the addition of3H-fucose occurs in the Golgi apparatus. The glycoprotein passes to the apical cortical cytoplasm by way of membrane-limited secretory granules, is exteriorized, and accumulates at the site where prebone transforms into bone (the prebone-bone junction). Since this is also the site of the calcification front, the deposition of labeled glycoprotein may be related to the deposition of bone mineral.

    Notes: Summary The present study is an investigation of the mechanism of hypercalcemia and hyperphosphatemia induced by the intravenous injection of lead acetate (Pb-Ac). A total of 118 male rats were injected with 30 mg/kg of Pb-Ac, or with 16.5 mg/kg of sodium acetate as the control. The levels of serum calcium, phosphorus and lead were then determined at various time periods after the injections. Serum calcium and phosphorus levels increased with time after Pb-Ac injection and the maximum values of calcium (17 mg%) were found after 1 h and of phosphorus (13.5 mg%) after 30 min. Both calcium and phosphorus levels reverted to the normal range after 12 h. The maximum net rates of increase of calcium and phosphorus were found immediately after Pb-Ac injection. At that time, deposition of lead at the calcifying sites of bone and incisor dentin was demonstrated by a histochemical examination. In other experiments the changes in the calcium and phosphorus contents in the medium after shaking bone powder in serum with Pb-Ac in an in vitro system were studied. It was confirmed that the calcium and phosphorus were displaced from the bone mineral, the extent of the displacement being correlated with the concentration of the Pb-Ac added to the medium, and that these displacements were very rapid reactions. These results suggest that hypercalcemia and hyperphosphatemia following Pb-Ac injection results from a direct action of lead on the bone mineral.

    Notes: Summary Recently published bone fluoride values from Iowa are very high compared to earlier reports, suggesting an increase in fluoride intake. Reanalysis of the Iowa specimens shows levels one-fourth those reported by the Iowa laboratory indicating an error in the original report. Seventeen bone specimens, collected from long-term residents of Rochester, New York, drinking 1 ppm F− water, had a mean value of 2085±270 ppm F− on an ashed-weight basis. This value is not significantly different from that predicted by the data of Zipkin et al. in 1958. These data, therefore, do not support the contention that there has been an increase in fluoride intake.

    Notes: Summary The net rate of loss of45Ca from plasma was monitored in normal, thyroparathyroidectomized (TPTX), and nephrectomized rats. Also studied was the effect of a 24 h intravenous infusion of PTH in TPTX rats. The45Ca was injected after an overnight fast and 1 to 4 h prior to the time the first blood sample was obtained. In normal rats, the net rate of loss of45Ca from plasma was slower than in TPTX rats for at least 12 h after radionuclide injection. Nephrectomy did not affect this difference between control and TPTX rats until 8 h after45Ca injection or about 18 h after nephrectomy. At this time a reversal occurred and the net rate of plasma45Ca loss in nephrectomized rats with parathyroids became faster than in TPTX nephrectomized rats. When PTH was infused at a constant rate (3 mU/g body weight/h) plasma calcium concentrations rose gradually for approximately 12 h after which a constant plasma calcium level was maintained with continued PTH infusion. During the period when plasma calcium concentrations were rising, the net rate of loss of45Ca from plasma was slower than in TPTX controls infused only with saline. However, when plasma levels stabilized, plasma45Ca disappearance rates changed such that the rate loss of45Ca from plasma became faster in PTH-infused than in TPTX controls. It is concluded that PTH increases calcium efflux from the same compartment in bone which receives calcium from plasma, thereby returning some recently deposited45Ca to plasma. In the non-equilibrated state, this returns additional45Ca to plasma causing anet decrease in the rate of loss of45Ca from plasma. However, in the equilibrated condition, when both calcium efflux from bone and influx into bone are in balance but at higher flux rates, PTH stimulates the diffusion of45Ca into other bone compartments.

    Notes: Summary A conventional spectrophotometer was used to do radiographic photodensitometry of bone mass. An experiment to produce osteoporosis in guinea pigs required quantification of changes in bone mass. Lack of a densitometer necessitated finding another method for quantification. The method developed involved using industrial film in a closed X-ray system. Optical density of X-rays was then read in a spectrophotometer at nine femur locations. Highly significant correlations between optical density and femur calcium content and femur dry weight per millimeter were obtained. Histological sections from an animal with low optical density measurements confirmed bone loss by showing decreased cortical width and loss of trabecular bone. In addition to using readily available equipment, this method offers the advantage of being able to detect bone loss in specific areas of the bones. In addition, bones are left intact and can be used for other purposes.

    Notes: Summary Carbonate-containing hydroxyapatite, enamel, and bone were irradiated by an X-ray and investigated between 77° and 350°K by means of electron spin resonance (ESR) spectroscopy. The ESR spectrum of enamel irradiated at 77°K in vacuum and observed at the same temperature was almost the same as that of the carbonate-containing hydroxyapatite. The temperature dependence of signal intensities confirms a spin-energy exchange between the mineral and organic constituents in bone, but in enamel no or very little spinenergy exchange between the mineral and organic constituents. Considerable similarity among the ESR spectra of enamel, bone, and carbonate-containing apatite was obtained after X-ray irradiation in air at 300°K with both an X-band and a Q-band ESR spectrometer. The Q-band spectrum can be interpreted in terms of two paramagnetic species. One is identified as a CO 3 3− anion radical which has an axial symmetry withg factors of 2.0029 and 1.9972. The other species is found to be centered atg=2.0019.

    Notes: Abstract Photovoltaic effect (infrared and visible light) is observed in bone and its two major components, collagen and apatite, at room temperature. A dimunition in the magnitude of photovoltage is observed after exposure to ultraviolet light in all the cases. The drift mobility of the charge carriers is obtained by measuring I versus V relationships in sandwich samples and relating them to the permitivity of the medium. Lifetime of the injected carriers is measured in the usual way. The data are consistent with the hypothesis that the effects are due to protonic conduction phenomena.

    Notes: Summary Osteoclastic bone resorption involves the solubilization of the mineral salts and the degradation of noncollagen bone matrix and collagen fibrils. As no recognizable collagen fibrils have ever been reported within cytoplasmic vacuoles in osteoclasts, it is generally assumed that the collagen fibrils are digested extracellularly in the resorption zone. The extent to which lysis occurs extracellularly and whether or not the osteoclasts phagocytose the degradation products remain to be established. In the present communication, a hypothesis is presented suggesting the possibility that osteoclastic resorption of bone involves the participation of two different cell types. According to this hypothesis, osteoclastic bone resorption is initiated by osteoclasts that demineralize areas of bone and degrade noncollagen bone matrix. After the osteoclasts have moved away or become partially detached from the demineralized site, the exposed collagen fibrils are phagocytosed by mononuclear, fibroblast-like or monocyte-derived cells.

    Notes: Summary Plasmaα 2HS-glycoprotein is specifically accumulated in calcified tissues. In the present studies this glycoprotein was isolated from plasma and after iodination with iodine-125 was injected intravenously into young rabbits. The tissue distribution and plasma disappearance rate of this radioactively labeled material were determined. Of the various tissues studied, bone showed the greatest retention of labeled glycoprotein expressed as percentage of the injected dose per gram tissue relative to the plasma content. The rate of loss of iodinatedα 2HS-glycoprotein from plasma was similar to that ofα 2HS-glycoprotein labeled endogenously by using14C-glucosamine or3H-glucosamine. The uptake of exogenously labeled3I-α 2HS-glycoprotein into bone tissue expressed as a percentage of the injected dose was similar to that of endogenously labeled14C-α 2HS-glycoprotein. These results suggest that the125I-labeled material can be used to study further the metabolism ofα 2HS-glycoprotein by bone tissue.

Its effect on the synthesis and release of β-glucuronidase (a referent for lysosomal enzymes) in mouse calvarial explants was studied in an in vitro culture system. A newly described medium (designated as KT medium) was introduced in this specific study. Mithramycin initially inhibited the release of β-glucuronidase into the medium and resulted in an ultimate accumulation of this enzyme in the bone. These results suggest that inhibition of bone resorption by mithramycin may be attributed to interference in release of lysosomal enzymes from bone cells.

In maxillary bones, ribs, and tibia, nuclear concentration of radioactivity was found in osteoprogenitor cells and osteoblasts. Osteocytes and chondrocytes in epiphyseal plates were either unlabeled or weakly labeled. In competition experiments, nuclear concentration of radioactivity was blocked by the injection of a high dose of nonradioactive 1,25-D3 prior to the administration of the labeled hormone, but not by a similar dose of nonradioactive 25-D3. The results are interpreted as indicating that osteoprogenitor cells and osteoblasts are target cells for the direct action of 1,25-D3 on fetal bone.

    Notes: Summary The role of vitamin D in early skeletal development was studied by measuring serum calcium and phosphorus, osseous tissue quantity and mineralization, and endochondral bone elongation in rat fetuses and pups from vitamin D-replete and vitamin D-deficient mothers. At the 20th day of pregnancy there was a slight, yet significant, increase in the amount of osteoid on trabecular bone surfaces in fetuses from vitamin D-deficient mothers. The fetal bones otherwise appeared normal in spite of severe skeletal changes in the vitamin D-deficient mothers. After parturition, the importance of vitamin D in skeletal development becomes progressively more obvious. Serum calcium levels were slightly, yet significantly, lower in vitamin D-deficient than in vitamin D-replete pups and these levels continued to fall in the vitamin D-deficient pups through lactation and after weaning. At 3 days postpartum, there was a small, yet significant, increase in the amount of osteoid on bone surfaces of the vitamin D-deficient pups. The relative amounts of osteoid in the vitamin D-deficient pups continued to increase through lactation and after weaning when compared with vitamin D-replete pups. By the 14th day of lactation and at later periods, there were significant reductions in metaphyseal mineralized tissues in the vitamin D-deficient pups when compared with the vitamin D-replete pups. At weaning and after weaning, there were substantial increases in growth plate thickness and decreases in longitudinal bone growth in the vitamin D-deficient pups when compared with the vitamin D-replete pups. The results from this study indicate that vitamin D does not appear to play a major role in fetal skeletal development. However, after birth, vitamin D becomes progressively more important with age for normal bone development, mineralization, and endochondral growth.

    Notes: Summary When demineralized bone matrix powder is implanted subcutaneously in the rat, the early responses involve the appearance and proliferation of mesenchymal cells at the site of implantation, followed by cartilage and bone formation. The ability of cells to migrate to the implant suggests that chemotaxis may be a critical event in this process. Therefore, using the modified Boyden chamber assay, we tested extracts of demineralized bone matrix for chemotactic activity. We have identified and partially purified, on molecular sieve chromatography, a heat labile and trypsin-sensitive protein (Mr=60,000–70,000) that is a potent chemoattractant for mouse calvaria, osteoblast-like cells (MMB-1), but not for monocytes (putative osteoclast precursors). These findings suggest that chemotactic protein(s) have a significant role in the recruitment of osteoprogenitor cells to a site of bone repair.

    Notes: Summary Uptake of tetracycline by enamel in the short-term was studied at an advanced stage of crown formation and after completion of crown formation in deciduous molars in the cat. Both secretory phase enamel and bands of postsecretory, maturation phase enamel labeled rapidly. The pattern of labeling mimicked that seen in the continuously growing, rootless incisor teeth of the rat, with narrow doublets fusing to form narrow bands with wide unlabeled intervals in the short term. This is a physiological demonstration which indicates that cyclical activity and changes may occurin vivo during the maturation phase of amelogenesis in rooted teeth. It is also noted that dentine did not, and that some circumscribed patches of bone did label in the same animals in the same time interval. Short-term tetracycline labels are lost following conventional histological processing, but are retained after freeze-drying or air-drying.

    Notes: Summary It has long been known that the stress history of bone tissue influences its structure; however, the nature of this relationship remains largely uncharacterized. The objective of this work was to induce a quantifiable change in the stress history ofin vivo bone tissue and examine subsequent changes in structural and material properties that might occur. Continuous compressive loads were applied to the diaphysis of adult mongrel dogs for 2 months. The loads, ranging from 12–130 N, were superposed on the normal activity of the animals by implanting spring loading devices on the diaphysis of the femur. After the animals were sacrificed, mid-diaphysial specimens were subjected to compression testing to determine a structural bulk stiffness. The cross-sectional areas of original bone tissue and new bone deposition were then determined. The ash weights of selected specimens were also determined. The results indicate that a positive correlation between the increase in cross-sectional area and the superposed stress does exist. The new bone apposition was found almost entirely on the periosteal surface. Very little evidence of internal remodeling or endosteal movement was observed. The new tissue was found to have a lower ash weight and appeared to have a disorganized microstructure. Mechanical testing also suggests that the newly deposited tissue is far less stiff than the mature original bone.

Cependant, environ 5% de la charge squelettique présente une vitesse de mobilisation lente, avec une demie-période d'environ 700 jours. Etant donné que cette vitesse est comparable à celle de45Ca, il semble que la faible fraction de Na osseux fasse partie de la structure du cristal. Seuls deux des procédés expérimentaux utilisés ont un effet significatif sur la mobilisation du Na. Il s'agit de la consommation prolongée d'un régime pauvre en calcium et d'un changement de l'activité parathyroidienne. Une augmentation du Na alimentaire affecte l'augmentation de la vitesse de mobilisation du sodium dans son composé rapide, alors que le composé lent est peu ou pas affecté. Les calculs effectués, à partir de nos résultats, indiquent que, contrairement au calcium, le squelette ne sert pas de réservoir significatif pour le maintien du milieu liquide extracellulaire.
    Abstract: Zusammenfassung Ratten erhielten intraperitoneal22Na, und die Retention dieses Isotopes wurde während einer Zeitspanne bis zu 650 Tagen im ganzen Körper und in den Knochen bestimmt. Im Gegensatz zu45Ca, welches sehr rasch von den Knochen aufgenommen und dann nur langsam abgegeben wird, verschwindet der größte Teil des vom Knochen aufgenommenen22Na in einer Halbwertzeit von 3–5,5 Tagen. Jedoch zeigen etwa 5% des Knochengerüstes einen sehr langsamen Turnover mit einer Halbwertzeit von ungefähr 700 Tagen. Da dieser Anteil jenem von45Ca vergleichbar ist, kann daraus geschlossen werden, daß diese kleine Fraktion des Na vom Knochen einen integralen Teil der Kristallstruktur ausmacht. Nur zwei der verschiedenen experimentellen Anordnungen, welche ausprobiert wurden, ergaben eine signifikante Wirkung auf den Na-Turnover im Knochen. Diese bestanden einerseits aus einer langzeitigen Verfütterung von Ca-armer Diät, andererseits aus einer Veränderung der Parathyreoidea-Aktivität. Eine Erhöhung der Na in der Nahrung beeinflußte die schnelle Komponente des Na-Turnover im Knochen; sie zeigte aber wenig bis keine Wirkung auf die langsame Komponente. Berechnungen aus unseren Resultaten lassen vermuten, daß — im Gegensatz zum Ca — das Skelet keine signifikante Reservoir-Funktion zur Erhaltung des Na in der extracellulären Flüssigkeit ausübt.
    Notes: Abstract Rats were given22Na intraperitoneally and the retention of the isotope was determined in whole body and in bone for periods up to 650 days. In contrast to45Ca, which is rapidly taken up by bone and then very slowly released, most of the22Na taken up by bone leaves, with a halftime of 3–5.5 days. However, about 5% of the skeletal burden exhibits a very slow turnover, with a half-time of about 700 days. Since this rate is comparable to that of45Ca, it is concluded that this small fraction of bone Na is an integral part of the crystal structure. Only two of the several experimental procedures which were tried produced a significant effect on bone Na turnover. These were prolonged feeding of a low calcium diet and a change in parathyroid activity. An increase in dietary Na affected the fast component of bone Na turnover, but there was little if any effect on the slow component. Calculations from our data suggest that, in contrast to Ca, the skeleton does not serve a significant reservoir function for the support of extracellular fluid Na.

Au-dessus de 2,6 volts, la vormation des bandes collagéniques (définies dans ce travail) parait inversement proportionelle, dans le temps, au voltage appliqué. La formation des bandes parait liée au processus d'électrolyse. Les auteurs démontrent que les pH élevés se situent au niveau de la cathode et qu'ils sont suffisants pour induire une précipitation du collagène. Les résultats antérieurs, publiés dans la littérature, décrivant l'action de courant électrique implanté, sont interpretés en fonction du mécanisme étudié au cours de ce travail.
    Abstract: Zusammenfassung Die Wirkung eines elektrischen Stromes auf verdünnte essigsaure Lösungen von löslichem Kollagen wurde bei Spannungen zwischen O und 25 Volt untersucht. Über 2,6 Volt wurde die Bildung von Kollagenbanden (in der Arbeit näher beschrieben) beobachtet, und zwar nach Zeiten, die der angewandten Spannung entgegengesetzt proportional verliefen. Die Bandenbildung wird dem Elektrolyseprozeß zugeschrieben. Wir konnten zeigen, daß sich die hohen pH-Werte rund um die Kathode entwickelten und daß diese genügen, um die Kollagenfällung zu veranlassen. Die Natur dieses Vorganges ist solcher Art, daß erin vivo als Antwort auf durch Stress verursachte Biopotentiale nicht vorkommen kann. Der hier beschriebene Mechanismus erlaubt es, Literaturangaben über den Effekt von implantierten Spannungsquellen zu interpretieren.
    Notes: Abstract The effect of electric current on dilute acetic acid solutions of soluble collagen has been studied for impressed voltages of from 0 to 25 volts. Above 2.6 volts the formation of collagen bands (herein defined) were observed at times inversely proportional to the applied voltage. Band formation is attributed to the process of electrolysis. It has been shown that the high pH values are generated in the area of the cathode, and that they are sufficient to induce collagen to precipitate. The nature of the process is such that it cannot occurin vivo as a response to stress induced biopotentials. Reports in the literature describing the effect of implanted voltage sources are interpreted in terms of the mechanism described here.

    Notes: Summary A new semiautomatic technique combining advantages of the manual and fully automatic methods is described for obtaining quantitative static and dynamic histologic data of bone. The hardware consists of a photomicroscope, digitizing platen, digitizer, plotter/printer, floppy disc drive, and computer. The microscope is equipped with a drawing tube through which the image of the digitizing platen is projected over the optical field. The investigator selects and traces all histologic structures to be measured by moving a cursor on the digitizing platen which is visible by its projection over the histologic field. The results on accuracy and static and dynamic precision of this method show that static and dynamic parameters of bone are obtained with a degree of error (〈20%) well within the acceptable range for biologic measurements. Comparison of this method with the grid technique according to Merz and Schenck showed that for almost all micromorphometric parameters comparable absolute data are obtained. Due to the higher precision of our method, however, the number of optical fields evaluated in obtaining these comparable data could be reduced to 25% of the number of fields evaluated by the Merz and Schenck technique. The time requirements for quantitative evaluation of a histologic slide of bone by our technique are 40–50 min; 20–25 min is needed for quantitative evaluation of osteocytes.

    Notes: Summary Quantitative bone histology was done in undecalcified sections of iliac crest bone specimens obtained from 84 normal American individuals. Samples were obtained within 12 h after death in a vertical and horizontal manner from both the right and left iliac crests. In addition to the determination of normal values of micromorphometric parameters of bone in these healthy American subjects, the following studies were carried out: (a) comparison of variance of micromorphometric parameters of bone obtained from the right versus left iliac bone (40 pairs), (b) comparison of micromorphometric parameters of bone obtained in a vertical versus horizontal manner (12 pairs), (c) evaluation of variance with increasing distance from the compact zone in bone samples obtained in a vertical manner (44 pairs), (d) analysis of variation between bone samples obtained more anteriorly versus posteriorly along the iliac crest (N=40), (e) comparison of differences in micromorphometric parameters obtained from age-matched men versus premenopausal women (N=12), and (f) plotting of histograms for assessment of distribution of micromorphometric parameters. The results show that histomorphometric data of bone cannot be easily compared when different techniques are employed for obtaining bone samples. Sampling variations are kept smaller when bone specimens are obtained in a vertical manner. Anterior/posterior variation does not cause major sampling error. If ranges of variation are taken into account, quantitative bone histology is a valuable tool for assessment of bone structure and bone cells.

    Description / Table of Contents: Résumé Un procédé de dissection a été mis au point pour permettre l'analyse zonale du cartilage de l'épiphyse des os de la jambe d'un foetus bovin. Des échantillons de tissu complet et lavé venant des différentes zones ont été analysés pour déterminer leur contenu en électrolyte et en constituants organiques, ainsi que pour leur densité, cendres et humidité. Les résultats ont montré que lorsque la quantité de cendres et la densité augmentaient, l'eau contenu dans le tissu diminuait. Les quantités de cendres dans les zones de cartilage en voie de calcification étaient plus grandes qu'il avait été. Quand elles étaient exprimées comme un pourcentage du poids sec, elles étaient les plus importantes dans le cartilage lavé calcifié que dans le autre zones. Au début de la minéralisation du cartilage, la quantité de Na (m moles/l de tissu frais) diminuait tandis que celles du Ca et du P inorganique augmentaient. Les niveaux de Mg augmentaient pendant que la calcification se poursuivait, mais seulement à une faction du taux du Ca et du P. Les rapports Ca/P inorganique étaient les plus grands dans le cartilage au repos (Cartilage non-différentié hyalin), suggérant un lien initiale entre Ca et les chrondromucoprotéines. Cependant, au début de la calcification, pendant la prolifération du cartilage les rapports Ca/P étaient beaucoup plus petits (ca. 1.50) mais augmentaient graduellement avec l'advancement de la minéralisation. Des changements importants survenaient dans la composition de la phase organique, pendant la calcification endochondrale. Comme il a été déterminé par l'analyse de l'hydroxyproline la quantité de collagéne diminuait progressivement pendant la calcification du cartilage, mais augmentait rapidement pendant la formation d'os. Comme il a été déterminé par l'analyse de l'héxosamine et du sulfute les chrondromucoprotéines étaient aux niveaux les plus éléves pendant la prolifération du cartilage et diminuaient constamment au cours de la calcification. Cependant, bien que la calcification était déja très avancée dans le cartilage hypertrophique, de grandes quantites de mucopolysaccharides étaient encore présentes. Les rapports sulfure/hhéxosamine montraient un léger déclin pendant les premiéres étapes de la calcification, mais augmentaient beaucoup pendant le cours de la minéralisation. Les quantités d'acide sialique étaient plus grandes dans le cartilage de l'épiphyse que dans le cartilage au repos ou dans l'os. Les lipides augmentaient rapidement pendant la calcification du cartilage, mais étaient très réduites dans l'os complètement formé. La signification de ces résultats est discutée.
    Abstract: Zusammenfassung Eine Seziermethode, die eine Schichten-Analyse der Beinepiphysenplatte von Rinderfeten erlaubt, wurde entwickelt. Proben vor und nach Waschen des Gewebes der verschiedenen Schichten werden untersucht in bezug auf Elektrolyte und organische Bestandteile, als auch in bezug auf Dichte, Aschengehalt und Feuchtigkeit. Die Resultate zeigten eine Zunahme des Aschengehaltes und der Dichte, während der Wassergehalt abnahm. Unerwartet hoch waren die Aschenwerte im in Verkalkung begriffenen Knorpel. Ausgedrückt in Prozent Trockengewicht, ergab gewaschener, verkalkter Knorpel den höchsten Wert aller Zonen. In den Frühstadien der Knorpelmineralisation nahm der Natriumgehalt (m Mol/l Frischgewebe) ab, während Ca und anorganischer P zunahmen. Mit fortschreitender Verkalkung erhöhte sich auch der Magnesium-Spiegel, allerdings nur zu einem Bruchteil des Ausmaßes, in welchem Ca und P zunahmen. Die höchsten Ca/P anorg. Verhältnisse wurden im Ruheknorpel (undifferenzierter hyaliner Knorpel) gefunden, was auf eine initiale Bindung von Ca durch Chondromucoproteine hinweist. Die Ca/P-Verhältnisse proliferierenden Knorpels waren jedoch bei Verkalkungsbeginn viel tiefer (ca. 1.50). Diese nahmen allerdings mit fortschreitender Mineralisierung stetig zu. In der endochondralen Verkalkungsphase fanden markante Veränderungen in der Zusammensetzung des organischen Anteils statt. Basierend auf der Hydroxyprolinanalyse nahm der Collagengehalt in der knorpeligen Verkalkungsperiode fortschreitend ab, während er jedoch bei der Knochenbildung rasch zunahm. Die an Hand von Hexosamin- und Schwefelanalysen bestimmten Chondromucoproteingehalte ergaben Höchstwerte im proliferierenden Knorpel und fielen stetig ab mit zunehmender Verkalkung. Trotz der im hypertrophischen Knorpel schon weit fortgeschrittenen Verkalkung waren immer noch große Mengen an Mucopolysacchariden vorhanden. Die Schwefel/Hexosamin-Verhältnisse zeigten eine minimale Abnahme in den frühen Verkalkungsphasen, nahmen jedoch markant zu bei fortschreitender Mineralisation. Der Sialinsäurespiegel war im Epiphysenknorpel, verglichen mit demjenigen des Ruheknorpels oder Knochens, erhöht. In der knorpeligen Verkalkungsphase nahmen die Lipide rasch zu, während jedoch die Werte des vollständig ausgebildeten Knochens stark vermindert waren. Die Bedeutung dieser Ergebnisse wird besprochen.
    Notes: Abstract A dissection procedure has been devised to permit zonal analysis of the epiphyseal plate of fetal calf leg bones. Samples of whole and washed tissue from the various zones were analyzed for their content of electrolyte and organic constituents, as well as for density, ash and moisture. Results showed that as ash content and density increased, water content decreased. Ash levels in calcifying cartilage zones were unexpectedly high. When expressed as a percentage of dry weight, washed calcified cartilage had the highest content of any zone. In the early stages of the mineralization of cartilage, Na content (mmoles/l of fresh tissue) decreased as Ca and inorganic P increased. Magnesium levels increased as calcification proceeded, but only at a fraction of the rate of Ca and P. Ratios of Ca/inorganic P were highest in resting cartilage (non-differentiated hyaline cartilage), suggesting an initial binding of Ca to chondromucoproteins. However, at the onset of calcification in proliferating cartilage, Ca/P ratios were much lower (ca. 1.50), but gradually increased with advancing mineralization. Marked changes occurred in the composition of the organic phase during endochondral calcification. As determined by hydroxyproline analysis, collagen content progressively decreased during cartilaginous calcification, but increased rapidly during bone formation. As determined by hexosamine and sulfur analysis, chondromucoproteins were at highest levels in proliferating cartilage and decreased steadily as calcification increased. However, although calcification was already well advanced in hypertrophic cartilage, large amounts of mucopolysaccharide still were present. Sulfur/hexosamine ratios showed a slight decline during the early stages of calcification, but increased markedly with further mineralization. Sialic acid levels were elevated in epiphyseal cartilage over those in resting cartilage or bone. Lipids increased rapidly during cartilaginous calcification, but were greatly reduced in fully-formed bone. The significance of these findings is discussed.

    Description / Table of Contents: Résumé Une méthode de détermination du contenu total en hydroxyle est décrite au niveau l' élément minéral des tissus calcifés. De l'os bovin anorganique et de l'émail dentaire sont dissouts dans un solvant non-aqueux (du chlorure méthanolique hydrogéné) et l'eau formée à partir des ions hydroxyle et carbonate, par dissolution acide du minéral, est déterminée par titration selon la méthode de Karl Fischer. L'eau, formée par réaction avec le carbonate, est calculée et soustraite du montant total de l'eau pour dèterminer la quantité d'eau, dérivée de l'ion hydroxyle. Les éléments inorganiques des échantillons sont ègalement déterminés. En utilisant des équations simultanées, basées sur l'équivalence électrochimique des ions à la surface et dans la maille de l'hydroxyle-apatite, les résultats des études de composition chimique sont utilisés pour calculer les quantités d'hydroxyle dans chaque substance. Le contenu en ion hydroxyle de l'os anorganique, basé sur ce traitement théorique, tenant compte de 2 hydroxyles par molécule, est de 1.40 moles par gramme, alors que, par analyse directe, on trouve 1.01±0.026 (S.E) m moles par gramme. Pour un échantillon d'émail dentaire, d'origine diverse, les valeurs théoriques et les valeurs expérimentales, relevées pour les contenus en hydroxyle, sont respectivement de 1.64 et 1.62 m moles par gramme.
    Abstract: Zusammenfassung Es wird eine Methode zur Bestimmung des gesamten Hydroxylgehaltes im Mineral von verkalkten Geweben beschrieben. Anorganischer Knochen und Zahnschmelz vom Rind wurden in einem wasserfreien Lösungsmittel (Methanolhydrogenchlorid) gelöst und das aus Hydroxyl- und Carbonat-Ionen sich bildende Wasser beim sauren Auflösen des Minerals mit der Titration nach Karl Fischer bestimmt. Das aus der Carbonatreaktion entstehende Wasser wurde berechnet, von der gesamten Wassermenge abgezogen und so der vom Hydroxylion her stammende Wasseranteil ermittelt. Die Proben wurden auf anorganische Bestandteile hin untersucht. Unter Anwendung simultaner Gleichungen, die sich auf die elektrochemische Äquivalenz der Ionen an der Oberfläche und im Kristallgitter von Hydroxyapatit stützten, konnten die Werte der chemischen Zusammensetzung zur Berechnung der Hydroxylmenge in jeder der Substanzen benützt werden. Auf Grund der theoretischen Berechnung und unter Annahme von 2 Hydroxylgruppen pro Molekül fand sich im anorganischen Knochen ein Hydroxylionengehalt von 1,40 mMol/g; durch die direkte Bestimmung konnte ein solcher von 1,01±0,026 (S.E.) mMol/g nachgewiesen werden. In einer gepoolten Zahnschmelzprobe betrug der Hydroxylgehalt theoretisch berechnet 1,64 mMol/g und praktisch bestimmt 1,62 mMol/g.
    Notes: Abstract A method is described for estimation of the total hydroxyl content of the mineral of calcified tissues. Anorganic bovine bone and dental enamel were dissolved in a nonaqueous solvent (methanolic hydrogen chloride) and the water formed from hydroxyl and carbonate ions by acid dissolution of the mineral was determined by the Karl Fischer titration. Water formed by reaction of carbonate was calculated and subtracted from the total amount of water to give the quantity of water derived from hydroxyl ion. The samples were analyzed for inorganic constituents. Using simultaneous equations, based upon the electrochemical equivalence of ions on the surface and lattice of hydroxyapatite, the data of chemical composition were used to calculate the amounts of hydroxyl in each substance. The content of hydroxyl ion of anorganic bone based on this theoretical treatment, assuming 2 hydroxyls per molecule, was 1.40 m moles per gram, and by direct analysis was 1.01±0.026 (S.E.) m moles per gram. With a pooled dental enamel sample, theoretical and determined values for hydroxyl contents were 1.64 and 1.62 m moles per gram, respectively.

    Notes: Summary Verapamil inhibits the intestinal absorption of calcium (Ca) and increases serum parathyroid hormone in rats. The effects of verapamil on bone tissue after long-term treatment is, however, not well described. Adult female and male Sprague-Dawley rats received verapamil in their drinking water at a dosage of 0.075 mg/ml (low dose) or 0.75 mg/ml (high dose) for 12 weeks; control rats received only drinking water. All rats were fed a diet containing 0.1% Ca and 0.5% P. In female rats, the amount of bone ash per volume was significantly reduced from 0.742 g/ml in controls to 0.713 g/ml after low-dose treatment of verapamil, and to 0.667 g/ml following high-dose treatment (P〈0.01). The tibial length was increased from 39.7 mm in controls to 40.3 mm or to 40.7 mm after low or high doses (P〈0.01). The tibial volume increased from 0.385 ml in controls to 0.397 ml after low doses and to 0.429 ml after high doses (P〈0.01). In contrast, in male rats the amount of bone ash per volume was significantly increased from 0.578 g/ml in controls to 0.580 g/ml after low doses and to 0.620 g/ml after high doses of verapamil (P〈0.01). The tibial bone volume in males as decreased from 0.633 ml in controls to 0.641 ml after low doses and to 0.583 ml after high doses (P〈0.05). The tibial length in the males was not changed by verapamil. The intestinal absorption of Ca was reduced in male rats from 5.28 in controls to 4.03 (serosa/mucosa) after low-dose treatment and to 2.46 after high-dose treatment with verapamil (P〈0.05). In female rats, the intestinal absorption of Ca did not change after verapamil treatment. Thus, chronic treatment with verapamil in female rats induced osteopenia whereas in male rats bone growth was inhibited.

    Notes: Summary The linear attenuation coefficient (μ in cm1) of trabecular bone was modeled for different conditions of bone and marrow composition in order to assess their influence on computed tomography (CT) quantitation. A large relative change (10% of TBV at 15% TBV) of bone concentration resulted in small changes of μ: 2.3% at 60 keV, 3.4% at 44 keV, 5.2% at 29 keV. Relative changes of trabecular bone volume (TBV) on the order of 3% could be detectedin vivo by CT were it not for errors of relocation and for compositional influences on accuracy. The μ (and density) depended critically not only on amounts of bone substance and marrow but on their compositions. Normal variation in the composition of bone substance produced an uncertainty in μ equivalent to 0.5 to 1% TBV. Increases of yellow marrow produced a decrease of μ which could be mistaken for a decrease of bone concentration. The biological variation (90% confidence limit) of marrow composition gives an uncertainty at 15% TBV of about 2.4% TBV at 60 keV, 1.7% at 44 keV, and 1.3% at 29 keV. These correspond to relative uncertainities of 16, 11, and 9% respectively. These factors help explain the large accuracy errors (30%) observed in all studies of trabecular bone where single-energy CT was used. Marrow composition also can affect precision of bone measurement. Systematic shifts of red and yellow marrow could mask biological changes such as those occurring with aging or treatment.

    Notes: Summary Osteopontin, bone sialoprotein, and bone acidic glycoprotein-75 are three acidic phosphoproteins that are isolated from the mineralized phase of bone matrix, are synthesized by osteoblastic cells, and are generally restricted in their distribution to calcified tissues. Although each is a distinct gene product, these proteins share aspartic/glutamic acid contents of 30–36% and each contains multiple phosphoryl and sialyl groups. These properties, plus a strict relationship of acidic macromolecules with cell-controlled mineralization throughout nature, suggest functions in calcium binding and nucleation of calcium hydroxyapatite crystal formation. However, direct proof for such roles is still largely indirect in nature. The purpose of this review is to present two speculative hypotheses regarding acidic phosphoprotein function. The goal was to use new sequence information along with database comparisons to develop a structural rationalization of how these proteins may function in calcium handling by bone. For example, our analysis has identified a conserved polyacidic stretch in all three phosphoproteins which we propose mediates metal binding. Also, conserved motifs were identified that are analogous with those for casein kinase II phosphorylation sites and whose number correlates well with that of phosphoryl groups/protein. A two-state conformational model of calcium binding by bone matrix acidic phosphoproteins is described which incorporates these findings.

    Notes: Summary Young male Sprague-Dawley rats (5–7 weeks old, 80–120 g) were hypophysectomized (HX) and maintained on thyroxin and dexamethasone replacement therapies. Ten days after surgery, some HX rats received a single injection of human growth hormone (hGH), and others five daily injections of hGH. Tartrate-resistant acid phosphatase (TRAP) histochemistry was employed in order to evaluate the number of cells of resorptive potential in the metaphyseal bone of the proximal tibiae of HX rats and was compared with normal rats and HX rats that further received hGH replacement therapy. In normal rats, two populations of TRAP-positive cells were identified: multinuclear cells, which showed histological characteristics of osteoclasts, and small mononuclear cells, the number of which was overwhelming when compared with the number of TRAP-positive multinuclear cells. Both populations were reduced in the HX rat, but more so the mononuclear cells, which were assumed to represent the precursor pool of mature osteoclasts and chondroclasts (P〈0.005). Five daily injections of hGH to HX rats brought about a significant increase in the number of TRAP-positive multinuclear cells, the number of nuclei of these cells, and the number of mononuclear TRAP-positive cells, throughout the metaphyseal bone (P〈0.05). A single injection of hGH increased only the number of TRAP-positive multinuclear cells in the trabecula/bone marrow interface (P〈0.05), indicating a very rapid fusion of precursor cells into mature osteoclasts in that particular location. It was concluded that GH depletion caused a major reduction in the number of cells presenting resorption capacity and that a short hGH replacement regimen resulted in a gradual restoration of these cells n the metaphyseal bone of the proximal tibia of the HX rat.

    Notes: Summary The objective of this study was to identify subjects in whom vitamin K has an effect on markers for calcium and bone metabolism and to detect hitherto-unnoticed correlations between vitamin K-induced changes in these markers. Participants in our studies were apparently healthy women, in whom we measured serum-immunoreactive osteocalcin (irOC) before and after adsorption to hydroxylapatite; total serum alkaline phosphatase (T-AP) and bonespecific alkaline phosphatase (B-AP); and fasting urinary calcium and creatinine. We describe a trial among 145 women who were treated with vitamin K (1 mg/day) for 2 weeks, and a prospective placebo-controlled trial among two groups each of 70 postmenopausal women with a treatment period of 3 months. It turned out that in elderly women vitamin K induced increased levels of serum irOC with a high affinity for hydroxylapatite (irOCbound), whereas that with low affinity (irOCfree) remained unaffected. In placebo-treated women the ratio irOCfree/irOCbound shifted from 0.38 to 0.65 around the 50th year of age. This shift was not found in vitamin K-treated women. After 3 months of treatment the vitamin K-induced changes in irOCbound were correlated with changes in B-AP, whereas irOCfree was correlated to urinary calcium excretion. In fast losers of urinary calcium vitamin K induced a 30% decrease of calcium excretion. The hypothesis is put forward that irOCbound may be a marker for bone formation, that serum irOCfree may be a marker for bone resorption, and that the serum irOCfree/irOCbound ratio may become a marker for skeletal remodeling. It is concluded that vitamin K administration may help to reduce urinary calcium loss in postmenopausal women, notably in the fast losers of calcium. The ratio irOCfree/irOCbound provides more information than total irOC and may become a practical marker for bone metabolism.

L'os est élué progressivement avec du chloroforme, chloroforme/méthanol et chloroforme/méthanol/HCl. Dans chaque éluat, les phospholipides sont identifiés par chromatographie quantitative. Les phosphatides acides et neutres sont présents, dans des proportions variables, dans l'éluat chloroformique de l'os anorganique. Ces lipides constituent plus de 80% des phospholipides totaux liés. L'extraction chloroforme/méthanol permit d'isoler un groupe de lipides essentiellement neutres. La composition de cet éluat d'os anorganique est voisin de l'extrait de solvant neutre d'os compact bovin minéralisé. Au troisième stade d'élution, on a pensé que lorsque l'acide du produit d'extraction agit comme un agent de déminéralisation, on pouvait comparer l'éluat avec l'extrait lipidique, obtenu après déminéralisation de l'os compact. Certes il existe des similitudes entre les phospholipides acides de l'extrait obtenu après déminéralisation et le troisième éluat lipidique de l'os. Il est possible que les phospholipides acides soient liés par des liaisons coordonnées aux ions Ca2+ et Mg2+ de la maille apatitique de l'os. En présence d'acide ou d'agent de déminéralisation, ces ions métalliques sont déplacés. Le rôle de ces composés de liaison, en tant que ponts entre les phases organiques et inorganiques des tissus calcifiés, est discuté.
    Abstract: Zusammenfassung Durch Äquilibration eines Gemisches von Phosphatiden mit anorganischen Knochen wurde die Bindung zwischen Phospholipiden und dem Mineralanteil von Hartgewebe untersucht. Der Knochen wurde sukzessive mit Chloroform, Chloroform/Methanol und Chloroform/Methanol/HCl eluiert. Die Phospholipide wurden in jedem der Eluate chromatographisch identifiziert und anschließend quantitativ bestimmt. Sowohl die sauren als auch die neutralen Phosphatide waren in verschiedenen Mengen im Chloroformeluat des anorganischen Knochens vorhanden. Zusammen machten diese Lipide über 80% der gesamtengebundenen Phospholipide aus. Die Extraktion in Chloroform/Methanol entzog dem anorganischen Knochen eine Gruppe von Lipiden mit vorwiegend neutralem Charakter. Die Zusammensetzung dieses zweiten Eluates aus dem anorganischen Knochen war jener ähnlich, wie sie für den Extrakt aus dem mineralisierten kompakten Rinderknochen mit einem neutralen Lösungsmittel beschrieben ist. Bei der dritten Elution wurde erörtert, daß dieses Eluat mit dem nach Demineralisation erhaltenen Lipidextrakt von kompakten Knochen verglichen werden kann, da die Säure des Extraktionsmittels demineralisierend wirkt. In der Tat wurden, was die sauren Phospholipide anbetrifft, Ähnlichkeiten zwischen dem Extrakt nach Demineralisation und dem dritten Lipideluat des Knochens festgestellt. Als Interpretation dieser Resultate wurde vorgeschlagen, daß die sauren Phospholipide koordinativ an Ca2+- und Mg2+-Ionen des Apatitgitters des Knochens gebunden sind. In einem sauren oder einem demineralisierenden Milieu werden die Bindungen zu Metallionen zerstört. Die Rolle solcher koordinativ wirkender Verbindungen als Brücken zwischen organischen und anorganischen Phasen mineralisierter Gewebe wurde besprochen.
    Notes: Abstract The association of phospholipids with the mineral phase of hard tissues was investigated by equilibrating a mixture of phosphatides with anorganic bone. The bone was eluted sequentially with chloroform, chloroform/methanol and chloroform/methanol/HCl. In each of the eluates, the phospholipids were identified chromatographically and were quantitated. Both acidic and neutral phosphatides were present, in variable proportions, in the chloroform eluate of the anorganic bone. Together, these lipids accounted for over 80% of the total bound phospholipids. Chloroform/methanol extraction removed a group of lipids from the anorganic bone which was predominately neutral in character. The composition of this second eluate of anorganic bone resembled the reported neutral solvent extract of mineralized bovine compact bone. At the third elution step, it was argued that as the acid in the extractant acted as a demineralizing agent, this eluate could be compared with the post-demineralized lipid extract of compact bone. With respect to their acidic phospholipids, similarities were noted between the post-demineralized extract and the third lipid eluate of bone. To interpret this finding, it is speculated that acidic phospholipids were bound through co-ordinating bonds to Ca2+ and Mg2+ ions of the apatite lattice of bone. In the presence of either acid or a demineralizing agent, these metal ions were displaced from the ligands. The role of such co-ordinating compounds as bridges between organic and inorganic phases of mineralized tissues is discussed.

    Notes: Summary Nandrolone decanoate (ND) is an anabolic steroid with a positive effect on bone mass in osteoporotic patients. The mechanism of action, (i.e., reduction of bone resorption and/or Stimulation of bone formation), the ultimate effect on mechanical properties, and the most effective dosage are not yet clear. To address these issues, dose-related effects of the long-term effect of ND on Serum and bone biochemistry, bone mineral content, and bone mechanical properties in ovariectomized (OVX) rats (12 weeks old at the Start of the experiment) were Studied for 6 months. The results were compared with those obtained in agematched, intact, and OVX rats. OVX caused in the femur a significant increase in net periosteal bone formation and net endosteal bone resorption of bone collagen content and torsional strength, and of Serum alkaline phosphatase, osteocalcin, and insulin-like growth factor-I (IGF-I) levels, whereas cortical bone density and calcium/creatinine and phosphorus/creatinine in 24-hour urine were Significantly reduced. Treatment of OVX rats with 1 mg ND/14 days resulted in a Significant increase in periosteal bone formation, femur length, cortical and trabecular bone mineral content and density, torsion stiffness and Strength, and bone IGF-I content, and a decrease in Serum osteocalcin, urinary calcium/creatinine levels, and bone collagen content compared with OVX controls. The higher ND dosage of 2.5 mg/14 days did not improve the results. ND treatment did not reverse all changes induced by OVX to the level of the intact controls. These results indicate that ND acts as an antiresorptive drug and as a bone formation Stimulating drug. Furthermore, the increased bone mass and bone mineral density is associated with improved bone Strength and stiffness and the presence of an increased amount of IGF-I. IGF-I is a growth factor considered to play a role in the maintenance of normal skeletal balance by a paracrine or autocrine mechanism.

    Notes: Summary A simple method for determining histamine and polyamines in various tissues was devised. The method, however, could not be applied to calcified tissues, because the high concentration of Ca2+ in the extract interferes with the chromatographic separation of these amines. By treating the extracts from calcified tissues with K2CO3, we succeeded in removing the Ca2+, and the method could then be applied to determine the amines in bone tissues of mice. By using this method, we examined the contribution of mast cells and histidine decarboxylae (HDC) to the amount of histamine in the bone. The results indicate that (1) the HDC activity in the bone is the highest among the tissues of normal mice, and the histamine produced by the HDC in the bone is metabolized rapidly; (2) a major part of HDC in the bone is present in the bone marrow cells other than mast cells, and most of histamine in the bone is attributable to the histamine pooled in mast cells; (3) mast cells in the diaphysis are located largely along the endosteal lining; and (4) the method devised in this study may be useful for studying the roles of histamine (or mast cells) and polyamines in calcified tissues.

    Notes: Summary Iron overload occurs frequently in thalassemia as a consequence of regular blood transfusions, and iron has been found to accumulate in bone, but skeletal toxicity of iron is not clearly established. In this study, bone biopsies of thalassemic patients were investigated by light (n = 6) and electron microscopy (n = 8) in order to analyze iron distribution and possible iron-associated cellular lesions. Sections (5 μm thick) were used for histomorphometry and iron histochemistry. Ultrathin sections were examined with an energy filtering transmission electron microscope. Iron was identified by electron energy loss spectroscopy (EELS), and iron distribution was visualized by electron spectroscopic imaging (ESI) associated with computer-assisted treatment (two-window method). This study shows that EELS allows the detection of 4500–9000 iron atoms, and that computer-assisted image processing is essential to eliminate background and to obtain the net distribution of an element by ESI. This study shows also that stainable iron was present along trabecular surfaces, mineralizing surfaces, and on cement lines in the biopsies of all patients. Moreover, iron was detected by EELS in small granules (diffusely distributed or condensed in large clusters), in osteoid tissue, and in the cytoplasm of bone cells, but not in the mineralized matrix. The shape and size (9–13 nm) of these granules were similar to those reported for ferritin. As for iron toxicity, all patients had osteoid volume and thickness and osteoblast surface in the normal range. Stainable iron surfaces did not correlate with osteoblast surfaces, plasma ferritin concentrations, or the duration of transfusion therapy. Numerous osteoblasts contained damaged mitochondria, and impaired osteoblast activity can therefore not be excluded.

    Abstract: Zusammenfassung Höfe von schwach mineralisierten Periosteocyten werden in Knochenbiopsien von Patienten mit Vitamin D-resistenter Rachitis häufiger rings um die jungen als um die alten Osteocyten beobachtet. Es scheint somit, daß nach einiger Zeit die unvollständige Mineralisation, die auf Grund der Höfe angenommen werden werden kann, einen “Heilungsprozeß” erfährt und daß deshalb eine verzögerte Mineralisation der perlicunären organischen Matrix erfolgt.
    Notes: Abstract Udermineralized periosteocyte haloes in bone biopsies from patients with vtamin D-resistant rickets occur more frequently around young than around old osteocytes, indicating that in time the incomplete mineralization suggested by the haloes, undergoes a “healing” process and therefore there is a delay in the mineralization of the perilacunar organic matrix.

    Description / Table of Contents: Résumé De l'adjuvant de Freund, constitué deMycobacterium butyricum tués, en suspension dans de l'huile minérale, provoque une arthrite chez les rats qui rappelle certaines formes d'arthrites chez l'Homme. L'arthrite comporte une inflammation de tissu mou, une formation de pannus et deux ostéopathies distinctes, une résorption ossuse accélérée et une formation anormale d'os périarticulaire. L'étidronate disodique, administré à raison de 4 mg/kg/jour par voie cutanée, à partir de l'injection de l'adjuvant, inhibe nettement la résorption osseuse, la formation de pannus, l'érosion inflammatoire de cartilage et la formation d'os pathologique, en rapport avec l'adjuvant. Après arrêt du traitement à l'étidronate disodique, la formationd'os pathologique devient visible radiographiquement deux semaines après l'arrêt du traitement. Le rôle de l'étidronate disodique sur la régulation de la résorption osseuse et sur la concentration du calcium et du phosphate, dans les tissus environnants, ainsi que les rapports des phénomènes arthritiques avec ce type d'expérience sont envisagés. Les résultats suggèrent la possibilité de l'emploi de l'étidronate disodique dans certaines formes d'arthrites humaines.
    Abstract: Zusammenfassung Freund's Adjuvans, das aus abgetötetem und in Mineralöl suspendiertem Mycobacterium butyricum besteht, verursacht bei Ratten eine arthritische Reaktion, welche gewissen Arthritisformen beim Menschen ähnlich ist. Die arthritische Reaktion besteht in einer Entzündung der Weichteile, einer Pannusbildung und zwei verschiedenen Osteopathien, ferner in einer beschleunigten Knochenresorption und einer abnormen periartikulären Knochenbildung. Wird gleichzeitig mit der Verabreichung des Adjuvans Dinatriumetidronat in einer Dosis von 4 mg/kg/Tag subcutan gegeben, so werden sowohl die Knochenresorption, als auch die Pannusbildung, die entzündliche Knorpelerosion und die mit der Adjuvansgabe verbundene pathologische Knochenbildung merklich gehemmt. Wurde die Dinatriumetidronat-Behandlung abgebrochen, so wurde die pathologische Knochenbildung innerhalb 2 Wochen nach Behandlungsabbruch röntgenologisch sichtbar. Die Kontrollfunktion des Dinatriumetidronates bei der Knochenresorption und bei der Calcium- und Phosphatkonzentration in den umliegenden Geweben, sowie die Beziehung dieser Substanz zu den arthritischen Prozessen, wie sie bei diesem Rattenversuch vorliegen, werden besprochen. Auf Grund dieser Resultate ist eine potentielle Anwendung des Dinatriumetidronates bei gewissen Formen der menschlichen Arthritis denkbar.
    Notes: Abstract Freund's Adjuvant, consisting of deadMycobacterium butyricum suspended in mineral oil, produces an arthritic response in rats which resembles certain forms of arthritis in man. The arthritic response consists of soft tissue inflammation, pannus formation and two separate osteopathies; accelerated bone resorption and abnormal periarticular bone formation. Disodium etidronate administered at 4 mg/kg/day subcutaneously from the time of adjuvant injection markedly inhibited bone resorption, pannus formation, inflammatory erosion of cartilage, and the pathologic bone formation associated with the adjuvant model. When the disodium etidronate treatments were discontinued, the pathologic bone formation became radiologically visible within two weeks after cessation of treatment. The role of disodium etidronate in controlling bone resorption and surrounding tissue concentration of calcium and phosphate and the relation to arthritic processes in this rat model are discussed. The data suggest a potential use of disodium etidronate in some forms of human arthritis.

    Notes: Abstract A method of successive approximations is proposed for the evaluation of the time-correlation functions such as those that give the thermal transport coefficients of gases. The method is based on a calculation of the changes in correlations of appropriate functions of the molecular velocity which are a result of collisions in the gas. The decaying rates of the correlations are expressed as integrals of the differential collision cross section. When the first approximation is introduced in the expressions for thermal transport coefficients, results are obtained for the coefficient of binary diffusion and the viscosity and thermal conductivity of single-component systems which are identical with those of the first Chapman-Enskog solutions of the Boltzmann and Enskog equations. For the coefficients of viscosity and thermal conductivity in multicomponent systems, it is shown that the first approximation leads to expressions of the form of the Sutherland and Wassiljewa relations, respectively.

    Notes: Abstract A new irreversible collision rule is introduced for lattice-gas automata. The rule maximizes the flux of momentum in the direction of the local momentum gradient, yielding a negative shear viscosity. Numerical results in 2D show that the negative viscosity leads to the spontaneous ordering of the velocity field, with vorticity resolvable down to one lattice-link length. The new rule may be used in conjunction with previously proposed collision rules to yield a positive shear viscosity lower than the previous rules provide. In particular, Poiseuille flow tests demonstrate a decrease in viscosity by more than a factor of 2.

    Notes: Abstract Numerical calculations have been done of the viscosity of the soft-sphere liquid, using a new molecular dynamics technique. It is based on a formulation of hydrodynamics which is discrete in space and time, and exactly renormalizable. The present data turn out to be sufficient to estimate the viscosity, but determination of the full equations of motion (and therefore renormalization) requires further calculations using a smaller discrete time interval; these are presently under way. The present results indicate that this method is more than 100 times more efficient than previous (Green-Kubo or nonequilibrium molecular dynamics) methods. This suggests that the discrete formulation is the most natural way to approach hydrodynamics.

    Notes: Abstract When nonequilibrium molecular dynamics is used to impose isothermal shear on a two-body periodic system of hard disks or spheres, the equations of motion reduce to those describing a Lorentz gas under shear. In this shearing Lorentz gas a single particle moves, isothermally, through a spatially periodic shearing crystal of infinitely massive scatterers. The curvilinear trajectories are calculated analytically and used to measure the dilute Lorentz gas viscosity at several strain rates. Simulations and solutions of Boltzmann's equation exhibit shear thinning resembling that found inN-body nonequilibrium simulations. For the three-dimensional Lorentz gas we obtained an exact expression for the viscosity which is valid at all strain rates. In two dimensions this is not possible due to the anisotropy of the scattering.

    Notes: Abstract Hydrodynamic excitations in lattice gas cellular automata are described in terms of equilibrium time correlation functions for the local conserved variables. For large space and time scales the linearized hydrodynamic equations are obtained to Navier-Stokes order. Exact expressions for the associated susceptibilities and transport coefficients are identified in terms of correlation functions. The general form of the time correlation functions for conserved densities in the hydrodynamic limit is given and illustrated by some examples suitable for comparison with computer simulation. The transport coefficients are related to time correlation functions for the conserved fluxes in a way analogous to the Green-Kubo expressions for continuous fluids. The general results are applied for a one-component fluid and several types of binary diffusion. Also discussed are the effects of unphysical slow modes such as staggered particle or momentum densities.

    Notes: Abstract Biot's theory is employed to study the reflection and transmission ofSH waves in a sandy layer lying over a fluid-saturated porous solid half-space. The entire medium is considered under constant initial stress. Effects of sandiness, initial stress, anelasticity and viscosity of the interstitial fluid on the partitioning of energy are studied. In the presence of initial stress the incident wave starts attenuating when incider beyond a certain angle (depending upon the amount of initial stress), even if the medium is perfectly clastic. Anelasticity of the solid layer results in the dissipation of energy during transmission. The direction of attenuation vector of incident wave affects the dissipation energy to a large extent. Effect on partitioning of energy reverse at incidence after the critical angle. A complete account of energy returmed back to the underlying half-space and that which is dissipated in the overlying layer has been discussed analytically as well as numerically.

    Notes: Abstract A newly designed cylindrical optical micro-rotator which has slopes for trapping and rotation on its upper surface is proposed. The cylindrical shape is effective in decreasing viscous drag force (damping factor) in the medium. A ray-tracing method considering the beam waist is employed to analyze the radiation pressure exerted on the upper surface of the rotator. We have demonstrated optical trapping and high speed rotation for various optical beam parameters such as the lens numerical aperture and the Gaussian mode profile as well as rotator shape parameters including oblique angle, height and diameter.

    Notes: Summary When amorphous calcium phosphate (ACP) was transformed to crystalline hydroxyapatite (HA) in a series of aqueous slurry concentrations ranging from low to high, the higher slurry concentrations produced more Ca-deficient HA as measured by Ca/P ratio and heat-produced pyrophosphate. We feel that the excess solution phosphate produced in the higher slurry transformations results in lower Ca/P ratio HA. It has been suggested that an ACP is the precursor to bone apatite. Regulation of the in vivo ACP slurry concentration could then control the stoichiometry and, therefore, the metabolic activity of bone apatite. X-ray radial distribution function (RDF) analyses showed that CO 3 2− substitution in HA creates far greater structural distortions than do Ca deficiencies. The latter, however, do produce small, but observable, structural distortions when compared to stoichiometric HA. It now seems clear that the RDF of bone apatite can be modeled by a synthetic, Ca-deficient, CO 3 2− -containing HA.

    Notes: Summary Fractionation of the EDTA-soluble, noncollagenous proteins of the organic matrix of chicken bone by Sephadex G-100 molecular sieving has revealed that the majority of the organic phosphorus is present in two fractions, from one of which a homogeneous phosphoprotein has been isolated. The purified phosphoprotein has an apparent molecular weight of 12,000 and contains bothO-phosphoserine andO-phosphothreonine.31P-NMR spectroscopy demonstrates that all of the organic phosphorus exists in the form of phosphomonoesters which have an average pK2 of 6.8. The phosphoprotein is highly acidic due to its high content of dicarboxylic acids in addition to the presence of organic phosphorus. The characteristic amino acid composition of the phosphoprotein establishes its noncollagenous nature and highlights the differences among bone, dentin, and enamel phosphoproteins. The absence ofγ-carboxyglutamic acid distinguishes it from osteocalcin, the noncollagenousγ-carboxyglutamic acid-containing peptide of bone matrix.

    Notes: Summary The bone mineral status of fifty-one children with diabetes mellitus was studied by single photon absorptiometry. The mean bone mineral content was 13% below values predicted by age, sex, height, and weight. Those children whose diabetes was one year or less in duration were as osteopenic as those whose diabetes was of longer duration. The demineralized children received a higher daily insulin dose than the others. No association was noted between the degree of skeletal demineralization and sex, statural growth, renal function, and serum calcium and phosphorus. No significant changes in bone mineral content were noted longitudinally.
