    Beschreibung: Long-term oral administration of the long-acting opiate 1-alpha-acetylmethadol (LAAM) to female rats beginning on the day of conception interfered with the dams' ability to carry litters to term. When treatment was initiated 3 weeks prior to mating this effect was not observed. Daily administration of the opiate antagonist naloxone from day 14 of gestation through term, to precipitate withdrawal in utero, resulted in increased stillbirths, decreased pup weight and size, and weight loss 24 hours after birth. These data question the validity of animal experiments which purport to be models for methadone maintenance programs but in which treatment is started immediately prior to or soon after conception.     Beschreibung: The concentration of oxytocin receptors increased in the myometrium of pregnant women and reached maximum levels in early labor. Concentrations of oxytocin receptors were also high in the decidua and reached a maximum at parturition. In vitro, prostaglandin production by the decidua, but not by the myometrium, was increased by the addition of oxytocin. Oxytocin may therefore stimulate uterine contractions by acting both directly on the myometrium and indirectly on decidual prostaglandin production.     Beschreibung: A new process has been developed which is called "Boradeption" to signify boronic acid--dependent phase transfer of water-insoluble agents. Highly fluorescent boronic acid dervatives, FluoroBoras, are solubilized with a physiologically compatible carrier buffer containing a receptor group for boronate adduct formation. The system can be used to stain living cells. In another variation of the Boradeption concept, an insoluble reporter molecule containing a boronate receptor is solubilized with a carrier buffer containing a boronic acid functional group.     Beschreibung: Prolactin administration reportedly increases blood pressure in rats and rabbits. To study the effects of prolactin deficiency on blood pressure, rats were given saline, normal rabbit serum, or rabbit antiserum to rat prolactin on postnatal days 2 to 5. Both males and females given antiserum had significantly lower blood pressure at 14 weeks than rats given saline or normal rabbit serum. Blood pressure differences between females given antiserum and females given saline disappeared during and following pregnancy. The antiserum also lowered the concentration of prolactin in plasma 49 percent in males and decreased the prolactin response to ether stress in both sexes.     Beschreibung: Placental and fetal tissues from 46 human pregnancies were cultured and cytogenetically analyzed in an attempt to document the existence of chromosomal mosaicism confined strictly to tissues of extraembryonic origin. In two gestations in which chromosomal mosaicism was found, it was expressed exclusively in placental chorionic cells and was not detected in cells derived from the embryo proper.     Beschreibung: The immediate induction of the full complement of maternal behavior in nonpregnant ewes primed with estrogen and progesterone has been obtained after 5 minutes of vaginal-cervical stimulation. A similar period of such stimulation given to recently parturient ewes, after the development of selective bonding to their own lambs, reversed their rejection behavior of alien lambs and produced a state of plasticity in maternal behavior, such that ewes receiving vaginal stimulation would accept and adopt alien lambs.     Beschreibung: The scrapie agent causes a degenerative nervous system disease in sheep and goats. Studies with extensively purified preparations demonstrated that the agent contains a protein that is required for infectivity. Chemical modification of the scrapie agent by diethyl pyrocarbonate reduced the titer 1000-fold. Exposure of the inactivated agent to hydroxylamine, a strong nucleophile, resulted in complete restoration of infectivity.     Beschreibung: Female rats are masculinized in utero by male littermates sharing the same uterine horn. Increased anogenital distances in neonatal females and mounting behavior in adult females are related to the presence of males on the caudal side of the females in the uterine horn. Contrary to current beliefs, interamniotic diffusion may not be responsible for the exchange of masculinizing agents among fetuses.     Beschreibung: The incubation of lens proteins with reducing sugars leads to the formation of fluorescent yellow pigments and cross-like similar to those reported in aging and cataractous human lenses. Called nonenzymatic browning or the Maillard reaction, this aging process also occurs in stored foods. Reducing sugars condense with the free amino group of proteins, then rearrange and dehydrate to form unsaturated pigments and cross-linked products. Other areas that require a refocusing of support are engineering science and education, at all levels, in science and engineering. Increasing our strength in these areas is essential to achieve our national economic, social, and political goals.     Beschreibung: When two small doses of ethanol were administered to pregnant mice during the gastrulation stage of embryogenesis, the embryos developed craniofacial malformations closely resembling those seen in the human fetal alcohol syndrome. Striking histological changes appeared in the developing brain (neuroectoderm) within 24 hours of exposure. Decreased development of the neural plate and its derivatives apparently accounts for the craniofacial malformations.     Beschreibung: One of six monoclonal antibodies raised against purified human placental alkaline phosphatase cross-reacts with the adult and fetal forms of intestinal alkaline phosphatase. The placental and intestinal enzymes are nonallelic. A new electrophoretic titration procedure was used to assess the relative reactivities of the different enzymes with the antibody. The placental enzyme was the most reactive. However, the adult intestinal enzyme showed greater reactivity than the fetal enzyme.     Beschreibung: Antisera to human syncytiotrophoblast microvillous cell surface membranes from different placentas are cytotoxic for lymphocytes from some people but not others, demonstrating the presence of allotypic trophoblast-lymphocyte cross-reactive (TLX) antigens. Exploratory principal components factor analysis, performed on limited data consisting of 300 cytotoxic reactions produced by ten separate trophoblast antisera on a panel of lymphocytes from 30 random donors, suggested the presence of three distinct TLX antigen groupings. It is proposed that such TLX alloantigens are central in establishing maternal recognition and protection of the blastocyst, and that lack of recognition results in implantation failure and spontaneous abortion.     Beschreibung: Resonance Raman spectra of photolyzed carbonmonoxyhemoglobin obtained with 10-nanosecond pulses are compared with the spectra of photolyzed carbonmonoxyhemoglobin stabilized at 80 K. In comparing the deoxy with the photodissociated species, the changes in the Raman spectra are the same for these two experimental regimes. These results show that at ambient and cryogenic temperatures the heme pocket in liganded hemoglobin is significantly different from that of deoxyhemoglobin.     Beschreibung: Pregnant rats received 2-[14C]deoxy-D-glucose (2DG) intravenously on the last day of gestation, and their fetuses were delivered 1 hour later by cesarean section. Fetal brains showed high 2DG uptake spread throughout the accessory olfactory bulb and little or no differential uptake in the main olfactory bulb.     Beschreibung: The eye can be visualized ultrasonically in more than 90 percent of fetuses 16 through 42 weeks of gestational age. Slow eye movements are present by 16 weeks. Rapid eye movements begin at 23 weeks and become more frequent between 24 and 35 weeks. Eye inactivity becomes more common after 36 weeks and is associated with sustained diaphragmatic excursions implying a "quiet sleep" state.     Beschreibung: Genetic material has been successfully transferred into the genomes of newborn mice by injection of that material into pronuclei of fertilized eggs. Initial results indicated two patterns of processing the injected DNA: one in which the material was not integrated into the host genome, and another in which the injected genes became associated with high molecular weight DNA. These patterns are maintained through further development to adulthood.     Beschreibung: In ovariectomized rats treated with progesterone, implantation was induced by a minute dose of 17 beta-estradiol. Twenty-four hours later, the concentrations of estradiol receptor in nuclear and cytosol fractions prepared from the endometrium surrounding the blastocyst and the inter-implantation areas remained very low. This indicates that estrogen was not secreted by the blastocyst. The higher receptor content in cytosol from inter-implantation sites may reflect modifications accompanying the decidual reaction since our results show that there is no translocation of the receptor to the nuclei.     Beschreibung: Pregnant rats underfed in the first 2 weeks of gestation had offspring with normal body weight at birth and weaning. However, starting at about 5 weeks of age the male offspring became hyperphagic and gained more weight than the controls. The female offspring did not overeat and did not become obese. Both male and female progeny showed increased fat cell size as adults. In males the epididymal and retroperitoneal fat pads were significantly enlarged nd adipocytes in these pads were hypertrophied.     Beschreibung: Isolated umbilical arteries and veins, obtained from normal women at the end of pregnancy, were incubated in krebs-Ringer bicarbonate solution and exposed to magnesium at concentrations ranging from 0 to 9.6 millimoles per liter. The basal tension of the vessels increased when magnesium was withdrawn and decreased when the concentration of magnesium was raised. Absence of magnesium in the medium significantly potentiated the contractile response of the vessels to bradykinin, angiotensin II, serotonin, and prostaglandin F2 alpha. It appears that magnesium deficiency may be responsible for spasms of umbilical and placental vasculature.     Beschreibung: Infant rat pups, fed through intragastric cannulas from postnatal day 4 through day 18, showed a 19 percent reduction in total brain weight when ethanol was included in their diet on days 4 through 7. This reduction in brain weight occurred even though body growth in the experimental rats was equal to that of their littermate controls. The ethanol-exposed animals were markedly hypoactive during the period of drug administration, then displayed gross body tremors for 3 to 5 days. Throughout the study, the animals treated with ethanol had poor motor coordination and were hyperresponsive.     Beschreibung: Injection of iodine-131-labeled goat immunoglobulin G antibody to human chorionic gonadotropin (hCG) into patients with hCG-secreting trophoblastic and germinal tumors permitted tumor detection and location by external gamma-ray scintigraphy. Excision of one of the metastatic tumors located by this method indicated a tumor/nontumor ration of 39.29.     Beschreibung: Acid extract of human placental tissue contain, by both radioimmunoassay and radioreceptor assay, beta-endrophin-like material. Half of this material will not go through a 5000-dalton filter and on Sephadex G-200 has a molecular size between 25,00 and 50,000 daltons. Of the material going through a 5000-dalton ultrafilter, 80 percent is excluded on Sephadex G-25 and held back, very slightly, on Bio-Rad P6, indicating a molecular size of approximately 4500 to 4800 daltons.     Beschreibung: The synthesis of a placental luteinizing hormone-releasing factor (pLRF), which is immunologically, physiochemically, and biologically indistinguishable from synthetic LRF, was demonstrated. The incorporation of 3H-labeled leucine by human placental tissue in vitro into pLRF was determined by purification on carboxymethyl-cellulose and specific immunoprecipitation of the 3H-labeled pLRF. The specific activity of the pLRF released into the medium increased 100-fold from day 1 to day 2 of culture and attained a concentration of 2.84 microcuries per microgram.     Beschreibung: Mothers among !Kung hunter-gatherers nurse briefly and frequently, with brief intervals between nursing bouts (mean +/- standard error, 13.19 +/- 1.28 minutes). The low levels of 17 beta-estradiol and progesterone in the serum of the mother are correlated with infant's age and with interbout interval, but not with total nursing time. Maternal gonadal function is apparently suppressed by a timing-dependent, prolactin-mediated effect of breast stimulation. Interbout interval may be a key variable in lactation infertility.     Beschreibung: Urine produced by wild female house mice, living in high- and low-density populations and confined to areas within a highway cloverleaf, was tested for its ability to delay puberty in juvenile female mice. Only urine collected from females in the dense population at its maximum density delayed puberty in test females. Urine collected when the population was less dense, or from a population that remained sparse, failed to delay puberty.     Beschreibung: Mice produce litters containing many pups, and the female fetuses that develop between male fetuses have significantly higher concentrations of the male sex steroid testosterone in both their blood and amniotic fluid than do females that develop between other female fetuses. These two types of females differ during later life in many sexually related characteristics.     Beschreibung: Titers of testosterone in plasma were determined by radioimmunoassay in male rat fetuses of stressed and control mothers on days 17, 18, 19, 21, and 23 (the day of birth) after conception. In fetuses of stressed mothers, testosterone concentrations were highest on day 17, declined on days 18 and 19, and then remained unchanged. In the control fetuses, testosterone increased from relatively low concentrations on day 17 to the highest amounts on days 18 and 19, and then declined. However, in those few blastocysts attaching with their antipolar mural trophoblasts, the inner cell mass became subdivided into two parts because of restrictions imposed on its growth by the apically situated polar trophoblasts and the plastic substrate.     Beschreibung: An endotoxin produced by Escherichia coli caused a decrease in prolactin concentrations in the plasma of sows when given at low dosages 2 days postpartum. Five to tenfold increases occurred in the plasma cortisol concentrations. Piglet growth, used as an indicator of milk secretion by the sows, was significantly depressed after the endotoxin administration. Some cases of lactation failure in the periparturient sow may thus be due to endotoxins suppressing prolactin concentrations.     Beschreibung: Metastatic mouse melanoma cells have a high affinity for the basement membrane and the ability to degrade it; these properties may allow tumor cells to invade the membrane and disseminate. In this study it was found that the metastatic potential of mouse melanoma cells varied when the cells were exposed in culture to fibronectin or laminin. After removal of fibronectin or exposure to laminin, the cells had an increased affinity for basement membrane collagen, were more invasive of basement membranes in vitro, and produced more lung colonies in vivo.     Beschreibung: Polypeptide analogs of the known members of the corticotropin-releasing factor (CRF) family were synthesized and tested in vitro and in vivo for enhanced potency or competitive antagonism. Predictive methods and physicochemical measurements had suggested an internal secondary alpha-helical conformation spanning about 25 residues for at least three members of the CRF family. Maximization of alpha-helix-forming potential by amino acid substitutions from the native known sequences (rat/human and ovine CRF, sauvagine, and carp and sucker urotensin 1) led to the synthesis of an analog that was found to be more than twice as potent as either of the parent peptides in vitro. In contrast, certain amino-terminally shortened fragments, such as alpha-helical CRF or ovine CRF residues 8 to 41, 9 to 41, and 10 to 41, were found to be competitive inhibitors in vitro.     Beschreibung: Genetic screening is a systematic search in the population for persons of certain genotypes. The usual purpose is to detect persons who themselves or whose offspring are at risk for genetic diseases or genetically determined susceptibilities to environmental agents. Is genetic screening a marvel about to free us from the scourge of genetic disease or a menace about to invade our privacy and determine who may reproduce? There are three different types of genetic screening. Newborn screening identifies serious genetic disease at birth, permitting prompt treatment to prevent mental and physical retardation. Fetal screening and prenatal diagnosis identify genetic disease in the fetus permitting selective termination of pregnancy and the opportunity to have children free of defects detectable in utero. Carrier screening identifies individuals heterozygous for a gene for a serious recessive disease who may be at risk for affected offspring. The challenge to society is to provide (by way of cost-effective programs) expert services, including genetic counseling and follow-up, to all who may benefit, to ensure confidentiality and freedom of choice, and to avoid misunderstanding and stigmatization. It is recommended that the objective of screening programs should be to maximize the options available to families at risk rather than to reduce the incidence of genetic diseases. Whenever possible, the providers of these services should be the providers of primary health care.     Beschreibung: The effects of marginal malnourishment , infections, and environmental chemicals on growth and reproductive success in Swiss-Webster white mice and wild deer mice were studied with fractional factorial designs. Interaction effects were discovered. For example, malnourished mice were more sensitive to virus exposure and environmental chemicals (a plant growth regulator or polychlorinated biphenyls).     Beschreibung: Inbred BALB/c females were mated and subsequently exposed in a divided cage to "stimulus" males or females whose H-2 type was similar or dissimilar to the stud male's. The incidence of pregnancy blocking was considerably higher when stud and stimulus males differed in H-2 type than when they did not. Similar results were obtained with urine samples of H-2 identical and nonidentical males. Females exposed after mating to other females whose H-2 type differed from the stud male, under the same experimental conditions, also showed an appreciable incidence of pregnancy block.     Notizen: Antioxidants such as bisphenol A (BPA) have long been recommended for retention of physical properties on oven-aging of PVC plasticized with certain plasticizers. We have shown how BPA inhibits oxidative fragmentation of plasticizers such as diisodecyl adipate (DIDA) and thus decreases apparent volatility, and how it improves retention of modulus, elongation, and low temperature flexibility. Low concentrations are more beneficial than either zero or high concentrations, especially on longer aging. At higher concentrations, although the plasticizer is retained in the PVC, it becomes less efficient at low temperature, and low-temperature flex is impaired while elongation is preserved. DIDA exudes during oven-aging unless oxidation occurs to prevent it. When BPA is present, exudation is worst at the most efficient concentrations of antioxidant. This appears to be the normal incompatibility of DIDA at 105°C which is permitted by the antioxidant. On oxidation, the dielectric constant of DIDA rises into the 4-8 range, signifying compatibility with PVC, and the DIDA does not exude.

    Notizen: Measurements of melt viscosity on samples of polytetrafluoroethylene of different molecular weight were carried out at 360°C by means of tensile creep tests in the linear viscoelasticity range. The apparent activation energy for viscous flow in the range between 330° and 380°C was estimated to be 20 kcal/mole. A value of about 7,500 was also determined for the average molecular weight between entanglement points (Me), from the equilibrium compliance (De). Melt viscosity data were compared with zero strength time (ZST) values and a linear correlation was found on a bilogarithmic scale. The dependence of ZST on the applied stress and temperature was also studied and the results are discussed on the basis of Bueche's theory on the creep at rupture above the glass transition temperature.

    Notizen: The mechanism by which TiO2 exerts a phototendering effect on nylon 66 photodegradation has not been previously explained. To aid in the elucidation of this mechanism, the wavelength dependence curves of photodegradation of nylon 66 both with and without TiO2 have been accurately determined. These data, in conjunction with reflectance and phosphorescence measurements, indicate that the phototendering effect of TiO2 proceeds by a chemical rather than an energy transfer mechanism.

    Notizen: It was the aim of this paper to study the influence of the solvent residues in PVC compounds on physical and mechanical properties of samples prepared from them as well as the rheologic properties of their melts. Besides tetrahydrofuran other solvents are also partly retained in PVC even after processing at higher temperatures. The properties of samples prepared from PVC compounds containing solvents depend on the solubility parameter of the solvent used. Solvents with a solubility close to that of PVC have the same effect as plasticizers. Solvents with very high solubility parameters, e.g., water, influence the flow and dynamic properties in just the opposite way than plasticizers and behave rather like fillers. A mechanism for influencing the properties of PVC by the presence of solvent residues is suggested.

    Notizen: Polytetrafluoroethylene surfaces have been treated to reduce thrombogenicity in order to make them suitable for use in prosthetic devices that come in contact with blood. This was done by first etching the surface with potassium in liquid ammonia to produce double bonds and then using these double bonds as sites for grafting on polyacids or as sites for chemical reactions. Tubes so treated were tested for thrombo-genetic activity by implantation in the thoracic aorta or inferior vena cava of dogs. These tests showed that the thrombogenicity of a polytetrafluoroethylene surface can be reduced by attaching negatively charged groups provided the surface concentration of these groups is not too high (order of 1-2 × 10-6 equivalent per cm2 geometric area) and provided the distribution of these groups is uniform. Sulfonic acid groups obtained by chlorosulfonation and carboxyl groups attached by grafting tert-butyl crotonate and hydrolyzing to crotonic acid were effective. Long chains of poly(acrylic acid), poly-(ethylenesulfonic acid), and poly(vinyl alcohol) sulfate were less effective.

    Notizen: The widths at half-value of high-resolution NMR spectra of bound water of cellulose were studied in relation to the temperature change. Two points were made clear: First, the amount of bound water of the cellulose sample can be pointed out with higher accuracy the lower the temperature of the measurement. Second, it is possible to determine in a simple manner the boundary temperature Tc at which water molecules become bound by the cellulose. Tc is characteristic of the cellulose sample of a given water content and corresponds to the glass transition temperature of the water-containing sample. The measurements appear to be readily applicable to other hydrophilic polymers.

    Notizen: Isotactic polypropylene hot-pressed film was subjected to uniaxial and biaxial stretching. The orientation behavior was investigated by means of x-ray pole figure technique, birefringence, electron microscope, and tensile tester. From the x-ray pole figure results it was confirmed that three kinds of crystal orientations, i.e., c-axis along the stretching direction, b-axis normal to the plane of the film, and [110] vector weakly normal to the film, accompany the biaxial stretching of the film. Electron micrographs of the surface of biaxially stretched polypropylene film revealed that, as the elongation increased, fibrillar structures became oriented to the stretching directions. From this orientation behavior a new deformation mechanism based on the woven structure presented by Khoury et al. was proposed. In this mechanism the orientation of the crystals is explained as a phenomenon accompanying the rotation and splitting of the woven structure presumed to be the structural element of polypropylene film.

    Notizen: The reactivity of mercerized, ethanol-washed, never-dried cotton, as indicated by acetylation, is higher than that of mercerized, water-washed, never-dried cotton. Hygroscopicity measurements indicate that the ethanol-washed cotton is not fully amorphous. They also reveal that the accessibility of cotton treated with caustic soda solution of mercerizing strength will be higher than that of cotton treated with ethylamine regardless of whether water or a solvent of lower hydrogen-bonding capacity is used to extract the swelling agent. Based on microscopic examination, acetylation of mercerized, ethanol-washed fibers apparently takes place relatively uniformly. The breaking load of cotton fibers is similar to that of mercerized, ethanol-washed cotton fibers of about 8% acetyl content. However, the crystallinity of these materials, as shown by hygroscopicity studies, is markedly different. This indicates that the fine structure of the cotton fiber can be modified considerably without causing a loss in strength. Yarn tensile tests indicate that mercerized, ethanol-washed cotton yarn with acetyl content between 10% and 30% has a breaking load which is about 12% lower than that of cotton.

    Notizen: Structural turbulence has been detected in dilute aqueous solutions of Polyox Coagulant (also known to be a highly effective drag-reducing agent). The flow line which characterizes structural turbulence from its onset in the laminar region passes well into the fully turbulent region (Reynolds turbulence) with virtually no change in slope, implying that the same molecular oscillations or segmental motions responsible for structural turbulence are now operative in drag reduction. The persistence of structural turbulence at very low concentrations is rationalized on the basis of Busse's explanation of the role of polymer entanglements in viscosity and elastic turbulence.

    Notizen: The effect of grafting styrene onto low-density polyethylene on the diffusion and solubility of benzene and n-hexane in the graft copolymer has been investigated. The diffusion coefficient at zero concentration Dc = 0 for both benzene and n-hexane decreased with the amount of styrene grafting in the polyethylene-styrene graft copolymer membrane. The free volume parameters of the polyethylene-styrene graft copolymer were calculated using benzene and n-hexane as the diffusing species. Results show that there is a large decrease in free volume as grafting proceeds and the effect is more pronounced at low levels of grafting. Solubility was found to be a function of the per cent grafting, there being a 50%-90% increase in benzene solubility in a 26% graft compared to polyethylene. The increase in solubility for n-hexane was considerably lower. The effect of crystallinity on the free volume parameters has also been calculated.

    Notizen: Just how far reactions can go after gelation during the cure of telechelic prepolymers has been a debatable point for some time. Utilizing a recently devised method,7 the curing reactions of some telechelic prepolymers were followed after the gel point. Extents of reaction above 90% occurred only in systems of average functionality close to two, functionality being the average number of reactive groups per molecule. Among systems of higher functionality, maximum extents of reaction of about 70% were most common. The final extent of reaction was only a few per cent above the extent of reaction at the gel point. The maximum extent of reaction varied with the concentration of reactive species and the relationship was a linear one at each functionality of the system. The data were consistent with PA2/r = {0.88/[(h - 1)(j - 1)]} + 0.10 where PA is the fraction of prepolymer reactive groups initially present which have reacted, r is the ratio of the initial number of crosslinking groups to prepolymer reactive groups, and h and j are weighted average functionalities of the two reactants. It is suggested that the limiting factor in defining the final extent of reaction in these systems is the accessibility of reactive groups as determined by solid geometry rather than thermodynamics or reaction kinetics. The final extent of cure is regularly dependent on functionality and one cannot regard functionality and maximum extent of reaction as independent variables.
