    Notes: Abstract The polyphagous predator, Coleomegilla maculata (DeGeer) (Coleoptera: Coccinellidae), commonly oviposits on the native weed, Acalypha ostryaefolia Riddell (Euphorbiaceae), in and around Kentucky sweet corn fields. Cannibalism of eggs by C. maculata adults and larvae is drastically lower on A. ostryaefolia than on nearby sweet corn plants. We examined ovipositional preference of C. maculata for A. ostryaefolia plants or sweet corn plants, dispersal of larvae from A. ostryaefolia plants, capability for dispersal of larvae across bare soil (e.g., to nearby plants), ability of larvae to climb from ground level up A. ostryaefolia plants or sweet corn plants, and effect of A. ostryaefolia borders adjacent to sweet corn plots on C. maculata population density in sweet corn. The ovipositional preference study revealed that C. maculata laid more eggs on A. ostryaefolia than on corn. First-instar C. maculata that hatched from egg clusters on A. ostryaefolia dispersed predominantly by falling, rather than crawling, to the ground. Glandular trichomes on A. ostryaefolia petioles and stems apparently inhibited intraplant movement of first instars, resulting in those larvae falling directly from leaves to the ground. Some first instars were capable of moving at least 8 m across bare soil in 24 h. From the ground, significantly more first instars climbed sweet corn plants than climbed A. ostryaefolia plants. Significantly more larvae were present in sweet corn plots bordered by A. ostryaefolia plants than in sweet corn plots without an A. ostryaefolia border. These findings show that physical attributes of companion plants can significantly influence natural enemy populations on crop plants by affecting interplant dispersal of natural enemies.

    Notes: Abstract The feeding behavior of Diuraphis noxia Mordvilko (Homoptera: Aphididae) on susceptible hosts causes both ultrastructural and tissue level damage which may affect phloem composition. Genetic evidence suggests that endosymbiotic bacteria in most aphids overproduce limiting amino acids to benefit hosts but that D. noxia depends less on endosymbionts for these nutrients, possibly due to an enriched diet. To determine whether D. noxia feeding damage results in higher concentrations of essential amino acids, stylet exudates were analyzed from wheat (Triticum aestivum) damaged to different degrees. Comparison of samples from undamaged and damaged susceptible wheat revealed changes in amino acid composition and an increase in levels of essential amino acids, indicating a nutritionally enhanced ingesta. The changes in stylet exudates paralleled changes in leaf exudates, indicating that the effects are systemic. Feeding damage is not observed on a resistant wheat host, var. Halt, and leaf exudates from infested Halt did not show changes in amino acid composition. Mean relative growth of nymphs was significantly lower on Halt than on susceptible Arapahoe, indicating that Halt is a less suitable host. Both varieties show similar amino acid levels in non-infested samples, suggesting that D. noxia infestation does not enhance the phloem environment in Halt. This study provides evidence that aphid feeding can generate a nutritionally enhanced phloem diet.

    Notes: Abstract Extended sieve element salivation (E1 waveform in the electrical penetration graph) is a characteristic activity during early sieve element punctures, particularly in resistant plants. In order to explore a chemically-mediated mechanism of resistance associated with sieve element salivation, we compared the pattern of feeding behaviour of the aphid, Sitobion fragariae (Walker), on two cultivars of the wheat Triticum aestivum L., with different concentrations of hydroxamic acids (Hx). During 24 h of electronic monitoring, aphids dedicated over 50% of the total time to phloem ingestion from the sieve elements. Total time allocated to E1 in the experiment, time to first E1 within the experiment, time allocated to E1 before a sustained phloem ingestion (E2) and the contribution of sieve element salivation to the phloem phase (E1/[E1+E2]) were significantly higher in the high-Hx cultivar. The increased salivation in plants with higher contents of Hx suggests the existence, at least in this system, of a chemically-mediated sieve element constraint.

    Notes: Abstract  Oxygen and CO2 fluxes were measured in hydroponically grown mycorrhizal and non-mycorrhizal Triticum aestivum L. cv. Hano roots. The NO3 – uptake of the plants was used to estimate the amount of root respiration attributable to ion uptake. Plants were grown at 4 mM N and 10 μM P, where a total and viable mycorrhizal root colonisation of 48% and 18%, respectively, by Glomus mosseae (Nicol. and Gerd.) Gerd. and Trappe (BEG 107) was observed. The O2 consumption and NO3 – uptake rates were similar and the CO2 release was higher in mycorrhizal than in non-mycorrhizal wheat. This resulted in a significantly higher respiratory quotient (RQ, mol CO2 mol–1 O2) in mycorrhizal (1.27±0.13) than in non-mycorrhizal (0.79±0.05) wheat. As the biomass and N and P concentrations in mycorrhizal and non-mycorrhizal wheat were the same, the higher RQ resulted from the mycorrhizal colonisation and not differences in nutrition per se.

    Notes: Abstract  The effect of inoculating wheat (Triticum aestivum L.) with the PO4 3–-solubilizing microorganisms (PSM) Bacillus circulans and Cladosporium herbarum and the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus sp. 88 with or without Mussoorie rock phosphate (MRP) amendment in a nutrient-deficient natural sandy soil was studied. In the sandy soil of low fertility root colonization by VAM fungi was low. Inoculation with Glomus sp. 88 improved root colonization. At maturity, grain and straw yields as well as N and P uptake improved significantly following inoculation with PSM or the VAM fungus. These increases were higher on combined inoculation of PSM and the VAM fungus with MRP amendment. In general, a larger population of PSM was maintained in the rhizosphere of wheat in treatments with VAM fungal inoculation and MRP amendment. The results suggest that combined inoculation with PSM and a VAM fungus along with MRP amendment can improve crop yields in nutrient-deficient soils.

    Notes: Abstract Phosphorus (P) is a major deficiency of soils of south-western Australia (WA). The fertilizer P requirements are not known for grain legumes being evaluated for neutral to alkaline, fine textured soils in WA. To rectify this, glasshouse and field experiments were undertaken to compare the responses of several grain legume species, wheat and canola to applications of single superphosphate and the results are reported in this paper. The glasshouse experiments measured responses of dried tops, harvested at 26 to 42 days after sowing, to P that was freshly-applied (current P) and previously-applied (previous P). Responses in the glasshouse were measured using yield, P concentration and P content (P concentration multiplied by yield) of oven dried tops of the following: wheat (Triticum aestivum), canola (Brassica napus), faba bean (Vicia faba), chickpea (Cicer arietinum), lentil (Lens culinaris), field pea (Pisum sativum), albus lupin (Lupinus albus) and narrow leaf lupin (Lupinus angustifolius). Field experiments in 1994 and 1995 compared seed (grain) yield responses of faba bean, chickpea, lentil, albus lupin and wheat to applications of current P. The P was banded (drilled) with the seed while sowing at 5 cm depth. Canola and wheat produced very large yield responses to increasing applications of current P. Responses were much smaller for albus lupin, faba bean and chickpea. Responses for lentil, narrow leaf lupin and field pea, fell in between responses of the small and large seeded species. Similar trends for responses were obtained as measured using yield, P concentration, or P content. For soils treated with previous P, similar trends were observed as for current P, but differences in yield responses between species were much less marked and the response curves tended to become more sigmoid. In the field experiments, grain yield responses to current P of albus lupin and chickpea were less than that for wheat. Relative to wheat, faba bean was the most responsive grain legume to applications of current P, with lentil producing similar responses to wheat in one experiment at a newly cleared, P deficient site.

    Notes: Abstract The objective of this study was to determine crop and tree productivity in several alley cropping planting patterns. The four- year study was conducted in western Oregon, United States and involved irrigated sweet corn (Zea mays) intercropped between hedgerows of red alder (Alnus rubra) and black locust (Robina pseudoacacia). Three alley cropping planting patterns with variable plant populations and tree-crop proximity were tested with each tree species in a randomized block design. Space available to trees and crops determined yield of both plant components. High crop yield coincided with low pruning yield, and vice versa. Compared to monocropping, yield reductions in traditional alley cropping planting patterns with widely spaced, double tree rows ranged from 5% to 15%. A. rubra and R. pseudoacacia coppiced readily and produced 0.9 to 4.7 tons of dry matter pruning biomass per growing season, depending on planting pattern and year. A. rubra yield continually increased during the trial, while the ability of R. pseudoacacia to produce green manure after frequent coppicing appeared to weaken. After four years, soil organic matter was 4 to 7% higher in the topsoil of an alley cropping system compared to a monocropping system. However, continuous growing of sweet corn resulted in decreased soil organic matter levels in both cropping systems compared to original levels. Crop yield reductions and the need for additional management inputs constrain the implementation of alley cropping in temperate climates. Only if benefits other than minor soil fertility improvements are realized is alley cropping a feasible alternative in temperate climate regions.

    Notes: Abstract Photosynthetic characteristics were investigated in the geographically isolated and restricted mangrove species, P.rhizophoreae. Gas exchange measurements were made on two to seven years old hydroponically grown plants maintained in 10%, 50% and 100% seawater. CO2 exchange in the 50% and 100% seawater treatments was reduced by 10% and 26%, respectively, compared to the 10% seawater treatment. CO2 response curves indicated that carboxylation efficiency was greater in 10% than in 50% seawater, while stomatal limitation increased from 11% to 16% as salinity increased from 10% to 50% seawater. Carbon losses via photorespiration (31% and 41%) and CO2 compensation point (67 and 81 μ11−1) were greater in 50% than in the 10% seawater treatment. Maximal CO2 exchange occurred at 30 °C with no differences among the salinity treatments. The results indicate that P. rhizophoreae exhibits many gas exchange characteristics previously reported for other mangroves.

    Notes: Abstract  Most floral meristem and organ identity genes of dicotyledonous plants belong to the MADS box gene family. Since they are generally transcribed in those tissues and organs whose identity they determine, they are excellent markers for developmental processes. Here we report the cDNA cloning of a pair of MADS box genes, ZMM8 and ZMM14, from the monocotyledonous plant maize. Maize inflorescences are composed of spikelets which contain two florets, an upper and a lower one. Although upper and lower florets develop in a very similar way in male inflorescences, ZMM8 and ZMM14 expression was found in all organs of upper florets, but no transcripts were detected in lower florets. In contrast, two other MADS box genes were found to be expressed in lower florets in the same way as in upper florets. Our observations suggest that during spikelet development ZMM8 and ZMM14 work as selector genes which are involved in distinguishing the upper from the lower floret. Alternatively, these genes may be involved in conferring determinacy to the spikelet or upper floret meristem. Our data suggest that in the phylogenetic lineage that led to maize an ancient type of MADS box gene has been recruited during evolution for the establishment of novel positional information not found within the simple inflorescences of dicotyledonous plants such as Arabidopsis.

    Notes: Abstract  Mitochondria play a critical role in the normal development of the plant male gametophyte and in the disruption of normal gametophyte development associated with cytoplasmically inherited male sterility (CMS). To investigate the role of mitochondria in these processes, the accumulation of mitochondrial gene transcripts and the accumulation of nuclear gene transcripts encoding mitochondrial proteins were investigated through male gametophyte development in normal maize and through the course of pollen abortion in CMS-S maize. Male gametophytes differing in developmental stage were isolated from male-fertile or male-sterile plants by sucrose density gradient centrifugation. Mature pollen was collected from dehiscent anthers of male-fertile plants. Aborted pollen, which collapsed during starch accumulation, was isolated from emergent tassels of CMS-S male-sterile plants. Microspores, developing pollen and mature pollen exhibited striking differences in mitochondrial transcript accumulation. Mature pollen lacked detectable mitochondrial transcripts. Aborted pollen of CMS-S plants contained abundant, intact transcripts of all mitochondrial genes studied, but prematurely degraded transcripts of several nuclear genes. Transcripts of the CMS-S associated mitochondrial open reading frames (orf355 and orf77) were detected from the early stages of microspore development through the aborted pollen stage. The implications of these findings are discussed in terms of the mitochondrial requirements for pollen function and the mechanism of pollen abortion in CMS-S maize.

    Notes: Abstract Embryogenic units of friable maize callus are formed as globular or oblong packets of tightly associated meristematic cells. These units are surrounded by conspicuous cell walls visible in light microscopy after staining with basic fuchsin. Transmission electron microscopy revealed that embryogenic cells are rich in endoplasmic reticulum, polysomes and small protein bodies, and that the outermost layer of their cell walls is composed of fibrillar material. Electron microscopy has also shown that this material covers the surface of embryogenic cells as a distinct layer which we denote as extracellular matrix surface network (ECMSN). Employing histochemical staining with β-glucosyl Yariv phenylglycoside, we localized arabinogalactan-proteins (AGPs) to the outer cell walls of embryogenic units including ECMSN. The most prominent staining was found in cell-cell junction domains. Large non-embryogenic callus cells were not stained with this AGP-specific dye. Immunofluorescence and silver-enhanced immunogold labelling using monoclonal antibody JIM4 has shown that the ECMSN of embryogenic cells is equipped with JIM4 epitope, while non-embryogenic callus cells are devoid of this epitope. We propose that some specific AGPs of the ECMSN might be relevant for cell-cell adhesion and recognition of embryogenic cells during early embryogenic stages, and that the JIM4 antibody can serve as an early marker of embryogenic competence in maize callus culture.

    Notes: Abstract Cytoplasmic male sterility (CMS) is encoded by the plant mitochondrial genome and can be reversed by nuclear restorer-of-fertility(Rf) alleles. In the CMS-S system of maize, reproductive failure and fertility restoration are gametophytic, occurring during the starch-filling stages of pollen development. Transcripts of the CMS-S-associated mitochondrial open reading frames (orf355 and orf77) are present from the early stages of microspore development through the aborted pollen stage. To investigate the molecular basis of fertility restoration, we compared mitochondrial-transcript accumulation in aborting CMS-S pollen and in CMS-S pollen restored to fertility by the Rf3 nuclear allele. In the presence of the Rf3 allele, novel, shorter transcripts of the orf355-orf77, cob and atp6 mitochondrial genes were created, and the relative abundance of larger transcripts was decreased for each of these loci. The altered transcript patterns cosegregated with male fertility conditioned by the Rf3 allele. The novel cob and atp6 transcripts were also observed in leaf-tissues of both normal and S-cytoplasm plants carrying the Rf3 allele. These observations support the hypothesis that the Rf3 allele encodes, or regulates, a modifier of mitochondrial transcript (Mmt) activity that affects both CMS and essential mitochondrial gene transcripts.

    Notes: Abstract  Colonization of Hordeum vulgare L. cv. Salome (barley)and Triticum aestivum L. cv. Caprimus (wheat) roots by the arbuscular mycorrhizal fungus Glomus intraradices Schenck & Smith leads to de novo synthesis of isoprenoid cyclohexenone derivatives with blumenin [9-O-(2′-O-β-glucuronosyl)-β-glucopyranoside of 6-(3-hydroxybutyl)-1,1,5-trimethyl-4-cyclohexen-3-one] as the major constituent and to transient accumulation of hydroxycinnamate amides (4-coumaroylagmatine and -putrescine). Accumulation of these compounds in mycorrhizal wheat roots started 2 weeks after sowing together with the onset of arbuscule formation and proceeded with mycorrhizal progression. Highest levels were reached in 3- to 4-week-old secondary roots (root branches of first and higher order) characterized by the formation of vesicles. In the final developmental stages, the fungus produced massive amounts of spores, enclosing the stele of older root parts (older than 5 weeks) characterized by cortical death. In these root parts, the secondary compounds were detected in trace amounts only, indicating that they were located in the cortical tissues. Some rhizosphere bacteria tested, i.e. Agrobacterium rhizogenes, Pseudomonas fluorescens, and Rhizobium leguminosarum, markedly stimulated both fungal root colonization and blumenin accumulation, thus, acting as mycorrhiza-helper bacteria (MHB). Application of blumenin itself strongly inhibited fungal colonization and arbuscule formation at early stages of mycorrhiza development. This was associated with a markedly reduced accumulation of the hydroxycinnamate amides 4-coumaroylputrescine and -agmatine. The results suggest that both the isoprenoid and the phenylpropanoid metabolism are closely linked to the developmental stage and the extent of fungal colonization. Their possible involvement in the regulation of mycorrhiza development is discussed.

    Notes: Abstract  Chromosome 5A of wheat carries major gene loci for agronomic traits including the vernalization requirement (Vrn-A1) and ear morphology (Q). To determine whether the genetic variation for ear emergence time and plant height is attributable to either of these major genes as pleiotropic effects or independent QTL, we combined a RFLP map constructed from 120 recombinant substitution lines derived from a cross between ‘Chinese Spring’ (Cappelle-Desprez 5A) and CS(Triticum spelta 5A) with data collected from field trials over 3 years. For ear emergence time the main effects on flowering time were by Vrn-A1 and QEet.ocs-5A.1, the latter a QTL in the 28.6-cM Xcdo584/Q interval linked to Q by less than 10 cM. The CS(T. spelta 5A) allele at QEet.ocs-5A.1 contributed to an earlier ear emergence time by 2.7–6.0 days, which was approximately equal to the effects of Vrn-A1. For plant height, three QTLs were identified on the long arm and linked in repulsion. The CS(T. spelta 5A) allele at Vrn-A1 or closely linked to Xfba068 contributed to a height reduction of 3.5–6.1 cm, whereas both the Q allele and Qt.ocs-5A.1 allele within the Xcdo1088/Xbcd9 interval from CS(Cappelle-Desprez 5A) produced a shorter plant. When plant height was partitioned into culm length and ear length, the Vrn-A1 allele and CS(Cappelle-Desprez 5A) allele at QCl.ocs-5A.1 within the Xcd1088/Xbcd9 interval were found to contribute to a shorter culm. CS(T. spelta 5A) allele at q was a major determinant of a long ear, together with minor effects at QEl.ocs-5A.1 within the Xcdo1088/Xbcd9 interval.

    Notes: Abstract  Quantitative trait loci (QTLs) of maize involved in mediating resistance to Peronosclerospora sorghi, the causative agent of sorghum downy mildew (SDM), were detected in a population of recombinant inbred lines (RILs) derived from the Zea mays L. cross between resistant (G62) and susceptible (G58) inbred lines. Field tests of 94 RILs were conducted over two growing seasons using artificial inoculation. Heritability of the disease reaction was high (around 70%). The mapping population of the RILs was also scored for restriction fragment length polymorphic (RFLP) markers. One hundred and six polymorphic RFLP markers were assigned to ten chromosomes covering 1648 cM. Three QTLs were detected that significantly affected resistance to SDM combined across seasons. Two of these mapped quite close together on chromosome 1, while the third one was on chromosome 9. The percentage of phenotypic variance explained by each QTL ranged from 12.4% to 23.8%. Collectively, the three QTLs identified in this study explained 53.6% of the phenotypic variation in susceptibility to the infection. The three resistant QTLs appeared to have additive effects. Increased susceptibility was contributed by the alleles of the susceptible parent. The detection of more than one QTL supports the hypothesis that several qualitative and quantitative genes control resistance to P. sorghi.

    Notes: Abstract  Cultivars with quantitative resistance are widely used to control Setosphaeria turcica (Luttrell) Leonard & Suggs, the causal organism of northern corn leaf blight (NCLB). Here the effectiveness of quantitative trait loci (QTLs) for NCLB resistance was investigated over the course of host plant development in inoculated field trials. A population of 194–256 F2:3 lines derived from a cross between a susceptible Italian (Lo951) and a highly resistant African inbred line (CML202) was tested in three environments in Kenya. The traits assessed were the incubation period (IP), the percentage disease severity (DS 1 to 5, taken biweekly), and the area under the disease progress curve (AUDPC). Considering all resistance traits and environments, a total of 19 putative QTLs were detected by composite interval mapping using a linkage map with 110 RFLP markers. In the combined analysis across environments, nine QTLs were significant (LOD 〉3.0) for DS 3, recorded around flowering time, explaining 71% of the genotypic variance. Four of these nine QTLs displayed significant (P〈0.05) QTL×environment (QTL×E) interaction. Most QTLs were already significant in the juvenile stage (IP) and became less effective after flowering. Across environments, three QTLs conditioned adult-plant resistance, in the sense that they were only significant after flowering. Six QTL alleles on chromosomes 2, 4, 5, 8, and 9 of CML202 should be useful for marker-assisted backcrossing.

    Notes: Abstract  Powdery mildew is one of the major diseases of wheat in regions with a maritime or semi-continental climate and can strongly affect grain yield. The attempt to control powdery mildew with major resistance genes (Pm genes) has not provided a durable resistance. Breeding for quantitative resistance to powdery mildew is more promising, but is difficult to select on a phenotypic basis. In this study, we mapped and characterised quantitative trait loci (QTLs) for adult-plant powdery mildew resistance in a segregating population of 226 recombinant inbred lines derived from the cross of the Swiss wheat variety Forno with the Swiss spelt variety Oberkulmer. Forno possibly contains the Pm5 gene and showed good adult-plant resistance in the field. Oberkulmer does not have any known Pm gene and showed a moderate susceptible reaction. Powdery mildew resistance was assessed in field trials at two locations in 1995 and at three locations in 1996. The high heritability (h2=0.97) for powdery mildew resistance suggests that the environmental influence did not affect the resistance phenotype to a great extent. QTL analysis was based on a genetic map containing 182 loci with 23 linkage groups (2469 cM). With the method of composite interval mapping 18 QTLs for powdery mildew resistance were detected, explaining 77% of the phenotypic variance in a simultaneous fit. Two QTLs with major effects were consistent over all five environments. One of them corresponds to the Pm5 locus derived from Forno on chromosome 7B. The other QTL on 5A, was derived from the spelt variety Oberkulmer and did not correspond to any known Pm gene. In addition, five QTLs were consistent over three environments, and six QTLs over two environments. The QTL at the Pm5 locus showed a large effect, although virulent races for Pm5 were present in the mixture of isolates. Molecular markers linked with QTLs for adult-plant resistance offer the possibility of simultaneous marker-assisted selection for major and minor genes.

    Notes: Abstract  Aegilops tauschii is the diploid D-genome progenitor of bread wheat (Triticum aestivum L. em Thell, 2n=6x=42, AABBDD). A genetic linkage map of the Ae. tauschii genome was constructed, composed of 546 loci. One hundred and thirty two loci (24%) gave distorted segregation ratios. Sixty nine probes (13%) detected multiple copies in the genome. One hundred and twenty three of the 157 markers shared between the Ae. tauschii genetic and T. aestivum physical maps were colinear. The discrepancy in the order of five markers on the Ae. tauschii 3DS genetic map versus the T. aestivum 3D physical map indicated a possible inversion. Further work is needed to verify the discrepancies in the order of markers on the 4D, 5D and 7D Ae. tauschii genetic maps versus the physical and genetic maps of T. aestivum. Using common markers, 164 agronomically important genes were assigned to specific regions on Ae. tauschii linkage, and T. aestivum physical, maps. This information may be useful for map-based cloning and marker-assisted plant breeding.

    Notes: Abstract  Quantitative trait loci (QTLs) for resistance to the fungal pathogen Setosphaeria turcica, the cause of northern corn leaf blight (NCLB), were mapped in a population of 220 F3 families derived from a cross between two moderately resistant European inbred lines, D32 (dent) and D145 (flint). The population was genotyped with 87 RFLP and 7 SSR markers. Trials were conducted in the field in Switzerland, and in the greenhouse with selected F3 families in Germany. The F3 population segregated widely for resistance with transgression of the parents. By composite interval mapping, a total of 13 QTLs were detected with two disease ratings (0 and 3 weeks after flowering). Together these QTLs explained 48% and 62% of the phenotypic variation. Gene action at most QTLs was partially dominant. Eight out of the 13 QTL alleles for resistance were contributed by the more-resistant parent, D145. On chromosomes 3, 5 and 8, QTLs were located in the same chromosomal regions as QTLs in tropical and U.S. Corn Belt germplasm. Some QTLs affected NCLB, head smut and common rust at the same time, with alleles at these loci acting isodirectionally.

    Notes: Abstract  Lodging can strongly affect both the grain yield and the quality of wheat. Lodging represents a quantitative trait and is difficult to assess on a phenotypic basis. Marker-assisted selection (MAS) could therefore become an important tool in breeding for lodging resistance. In this study, we mapped and characterised quantitative trait loci (QTLs) for lodging resistance, as well as morphological traits correlated with lodging, in a segregating population of 226 recombinant inbred lines derived from the cross of the lodging-resistant wheat variety Forno with the susceptible spelt variety Oberkulmer. Lodging, plant height, leaf width, leaf-growth habit, culm stiffness, culm swinging, culm thickness, days to ear emergence and days to flowering were assessed in field trials at two locations in 1996 and at one location in 1997. Additionally, at one location weight and length parameters were also assessed. Plant height and culm stiffness explained 77% of the phenotypic variance of lodging in a multiple regression model over all three environments. QTL analysis of lodging and morphological parameters was based on a genetic map containing 230 loci with 23 linkage groups (2469 cM). With the method of composite interval mapping nine QTLs for lodging resistance were detected, explaining 63% of the phenotypic variance in a simultaneous fit. Seven of these QTLs coincided with QTLs for morphological traits, reflecting the correlations between these traits and lodging. In our population the most efficient way to improve lodging resistance would be by a combination of indirect selection on plant height and culm stiffness together with MAS on the two QTLs for lodging resistance which did not coincide with QTLs for morphological traits.

    Notes: Abstract  We are reporting the successful isolation of wheat chromosome arm 1DS by flow cytometry. A chromosome suspension was prepared for the 1DS ditelosomic line and the normal ‘Chinese Spring’ (CS) by chopping 2-day-old root tip meristems, synchronized by hydroxyurea, in HEPES-magnesium sulfate buffer containing propidium iodide. Chromosomes were analyzed and sorted with a FACS Vantage flow cytometer and cell sorter. An extra peak was observed in the flow karyotype of the ditelosomic line that was absent in ‘CS’. The estimated size of chromosomes from the extra peak matched with the expected size of chromosome 1DS. Chromosomes from the putative 1DS peak were analyzed by both fluorescent microscopy and N-banding analysis. A total of 571 chromosomes from two separate experiments were analyzed, and all were observed to be telosomics except for 2 which were broken. About 82% of these telosomics showed the diagnostic N-band of 1DS, the remaining were unbanded and are probably also 1DS. This strategy can also be used to sort other wheat arms.

    Notes: Abstract  The stability and heritability of three marker genes was investigated in a population of twelve independent transgenic cereal lines (six wheat and six tritordeum). Integration patterns, inheritance of structural transgenes and inheritance of expression were analysed in the T0 and T1 generations for all 12 lines. Transmission and expression were analysed in the T2 generation for 9 lines and in the T3 generation for the six wheat lines. Inheritance of integration patterns was highly stable, and transmission of the transgenes and inheritance of their expression followed Mendelian ratios in the majority of lines. A gradual reduction in uidA expression was observed over three generations, which was not accompanied by a similar reduction in bar expression. Some unexpected phenomena associated with transgene inheritance were also observed and are discussed.

    Notes: Abstract Transgenic wheat plants from specific cultivars can now be routinely engineered in many laboratories. However, our understanding of the factors controlling transgene expression and stability in wheat compared to other cereals is rather limited. Only a few promoters have been tested in transgenic wheat, and relatively little is known of their relative activities and expression parameters. In the present study, the spatial and temporal properties of one heterologous constitutive promoter and one seed‐specific wheat promoter were investigated. We generated constructs with the reporter gene gusA (β‐glucuronidase) driven by: (a) the constitutive maize ubiquitin‐1 (ubi‐1) promoter, and (b) two different‐sized fragments of the seed‐specific low molecular weight glutenin (LMWG1D1) promoter from wheat. The activities of all three promoter constructs were comparable in endosperm tissue. A detailed analysis of spatial and temporal properties of the promoters is described. Heat shock treatment of transgenic plants carrying the ubi‐1: gusA construct resulted in a significant elevation in the levels of GUS activity. The inheritance of transgene expression levels and stability was evaluated over four generations, as a function of transgene integration patterns and copy number.

    Notes: Abstract The heterobasidiomycetes responsible for plant smuts obligatorily require their hosts for the completion of the sexual cycle. Accordingly, the sexual cycle of these fungi could so far be studied only by infecting host plants. We have now induced Ustilago maydis, the causative agent of corn smut, to traverse the whole life cycle by growing mixtures of mating-compatible strains of the fungus on a porous membrane placed on top of embryogenic cell cultures of its host Zea mays. Under these conditions, mating, karyogamy and meiosis take place, and the fungus induces differentiation of the plant cells. These results suggest that embryogenic maize cells produce diffusible compounds needed for completion of the sexual cycle of U. maydis, as the plant does for the pathogen during infection.

    Notes: Abstract Photosynthetically active radiation (PAR) and temperature were measured continuously at the surface of estuarine intertidal sediments in the Tagus estuary, Portugal, along two spring-neap tidal cycles. PAR and temperature were strongly conditioned by the periodic tidal inundation, with large and abrupt variations occurring during flooding and ebbing. PAR levels reaching the sediment surface decreased very rapidly to zero or very low values during most of the daytime immersion. Inundation during high tide had the general effect of attenuating the amplitude of daily temperature fluctuation, with the incoming water usually warmer than the sediment during the night or early morning and cooler during the day. The daily progression of tidal emersion resulted in a clear fortnightly variation in total daily PAR reaching the sediment surface, while both daily mean temperature and mean temperature of diurnal low tide periods failed to exhibit a well-defined fortnightly periodicity. The obtained results indicate that the estuarine intertidal environment is dominated, at sub-seasonal time scales, by fortnightly periodicity in irradiance and temperature conditions favourable for benthic photosynthesis.

    Notes: Abstract Microalgal productivity was examined using both a wild type and a phycocyanin-deficient mutant of Synechocystis PCC 6714 (PD-1). The culture was conducted at various light intensities under low and high cell densities in a continuous culture system. At low light intensity, photosynthetic productivity was almost the same for both low and high cell densities. However, at higher light intensities photosynthetic productivity was higher in mutant PD-1 than in the wild type. At 2000 μmol photon m−2 s−1 the productivity was 50% higher in mutant PD-1. This result is consistent with our first report (Nakajima & Ueda, 1997), which showed that photosynthetic productivity can be improved by reducing the light harvesting pigment content in high cell density cultures at high light intensities. It is concluded that the technology for reducing LHP content is a useful method for improving photosynthetic productivity in algal mass production.

    Notes: Abstract The green flagellate Euglena gracilis has been used as a model organism to elucidate the possible large-scale and short-term effects of waste substances from the pulp and paper industry on photosynthetic efficiency (PE). Different concentrations of waste substances before and after treatment in a cleaning system were studied. The uncleaned sample at concentrations up to 1:10 and the cleaned sample at concentrations up to 1:5 showed stimulating effects on the PE after 7 days of incubation compared to the control. The effects of waste substances on the PE of E. gracilis were also studied in combination with short-term studies (20 and 40 min) of ultraviolet-B radiation (UV-B, 280–320 nm). It was shown that increasing concentrations of the uncleaned sample had continuously stimulating effects on the PE and worked protectively against UV-B radiation. The cleaned sample exhibited no effects, or negative effects, on the PE of E. gracilis together with UV-B radiation compared to the experiments with only UV-B radiation. At the concentration 1:1 of the cleaned sample an increase in the PE was detected due to the high concentration of the coloured substances and a decrease in the UV-B penetration. PE revealed itself to be highly sensitive for detecting toxic effects on E. gracilis and is thus very promising for use in regular toxicity tests of waste water from pulp and paper industry.

    Notes: Abstract The second report ofFusarium globosum is based on strains isolated from wheat in Ishigaki Island, Okinawa, in subtropical Japan. Morphological features of the Japanese isolates are described. These strains show different reactions in conidiogenesis to light conditions. Under continuous BLB light, falcate sporodochial conidia are typically induced, but production of aerial globose conidia is suppressed. In two of the strains, clavate conidia became longer under BLB light. Application of both, complete darkness and continuous BLB light, is recommended as standard light conditions to cultureFusarium isolates.

    Notes: Abstract Photosynthetic (oxygen evolution) and growth (biomass increase) responses to ambient pH and inorganic carbon (Ci) supply were determined for Porphyralinearis grown in 0.5 L glass cylinders in the laboratory, or in 40 L fibreglass outdoor tanks with running seawater. While net photosynthetic rates were uniform at pH 6.0–8.0, dropping only at pH 8.7, growth rates were significantly affected by pH levels other than that of seawater (c. pH 8.3). In glass cylinders, weekly growth rates averaged 76% at external pH 8.0, 13% at pH 8.7 and 26% at pH 7.0. Photosynthetic O2 evolution on a daily basis(i.e. total O2 evolved during day time less total O2 consumed during night time) was similar to the growth responses at all experimental pH levels, apparently due to high dark respiration rates measured at acidic pH. Weekly growth rates averaged 53% in algae grown in fibreglass tanks aerated with regular air (360 mg L-1 CO2) and 28% in algae grown in tanks aerated with CO2-enriched air (750 mg L-1 CO2). The pH of the seawater medium in which P. linear is was grown increased slightly during the day and only rarely reached 9.0. The pH at the boundary layer of algae submerged in seawater increased in response to light reaching, about pH 8.9 within minutes, or remained unchanged for algae submerged in a CO2-free artificial sea water medium. Photosynthesis of P. linearissaturated at Ci concentrations of seawater (K0.5560 μM at pH 8.2) and showed low photosynthetic affinity for CO2(K0.5 61 μM) at pH 6.0. It is therefore concluded that P. linearisuses primarily CO2 with HCO3 - being an alternative source of Ci for photosynthesis. Its fast growth could be related to the enzyme carbonic anhydrase whose activity was detected intra- and extracellularly.

    Notes: Abstract Photosynthesis and cell composition of Porphyraleucosticta discs grown at low (〈 0.0001% in air), current (control) and high (1% CO2 in air)inorganic carbon (Ci) concentrations were analyzed. Carbohydrate content in discs grown at high Ci increased (15.1 mg g-1 FW) with respect to the control (6.4 mg g FW-1), whereas soluble protein content decreased to one-third (5.6 to2.1 mg g-1 FW). Carbohydrate content was unaffected and soluble protein slightly increased in discs grown at low Ci. As a consequence of these changes, a lower C/N molar ratio (8.6) was found in the discs grown at low compared to high Ci(12.4). Nitrate reductase activity increased at high Ci from 0.3 ± 0.2 to 1.7 ± 0.4 μmolNO2 - g-1 FW h-1indicating that reduction and assimilation of nitrate were uncoupled. The response of photosynthesis to increasing irradiance, estimated from O2evolution vs. irradiance curves, was affected by the treatments. Maximum quantum yield (Φ O2°) and effective quantum yield (Φ O2) at 150 μmol photon m-2s-1 decreased by 20% and 50%, respectively, at low Ci. These differences could be due to changes in photosynthetic electron flow between PSII and PSI. Treatments also produced changes in maximal (Fv/Fm) and effective (ΔF/Fm′)quantum yield for photosystem II charge separation.

    Notes: Abstract Due to their rapid growth and nutrient assimilation,Porphyra spp. are good candidates for bioremediation and polyculture. The production potential of two strains of P. purpurea and P. umbilicalis from north-east USA was evaluated by measuring rates of photosynthesis (as O2evolution) of material grown at 20 °C. Photosynthetic rates of P. umbilicalis were 80%higher than P. purpurea over the temperature range 5–20 °C, at both sub-saturating andsaturating irradiances (37 and 289 μmol photonm-2 s-1). Porphyra umbilicalis was more efficient at low irradiances (higher α) and had a higher Pmax (23.0 vs 15.6 μmolO2 g-1 DW min-1) than P.purpurea, suggesting that P. umbilicalis is a better choice for mass culture, where self-shading maybe severe.

    Notes: Abstract The effects of nutrients on the photosynthetic recovery of Nostoc flagelliforme during re-hydration were investigated in order to see if their addition was necessary. Net photosynthesis was negligible in distilled water without nutrient-enrichment. Addition of K+ resulted in significant enhancement of net photosynthesis, whereas other nutrients (Fe3+, Mg2+, Na+, NO3 -, PO4 3-, Cl-) and trace-metals (A5) showed little effect. The recovered net photosynthetic activity increased with the increased K+, and reached the maximum at concentrations above 230 μM. Desiccation and re-hydration did not affect the dependence of photosynthetic recovery on K+. It was concluded that dried field populations of N. flagelliforme require exogenous addition of potassium for photosynthetic recovery and that growth may be potassium-limited in nature.

    Notes: Abstract Factors controlling the CO2 system parameters, including the partial pressure of CO2 (PCO2) in coral reef waters, were investigated in three mid-oceanic reefs of the Indo-Pacific region. Surface water PCO2 in the lagoons of Majuro Atoll and Palau barrier reef in the Pacific were 25 µatm and 48 µatm higher than those of the offshore waters, respectively, while South Male Atoll lagoon of the Maldives in the Indian Ocean exhibited relatively small difference in PCO2 compared to the offshore water. Observations from Majuro Atoll and Palau barrier reef are consistent with the view that calcium carbonate production predominates in coral reefs. On the other hand, results from South Male Atoll can be attributed to the thorough flushing of the lagoon, which is connected to the open ocean by numerous deep channels. The offshore-lagoon PCO2 difference depends on system-level net organic-to-inorganic carbon production ratio while reef topography, especially residence time of the lagoon, has a secondary effect on the magnitude of the offshore-lagoon difference. A potential for releasing CO2 might be more evident in an enclosed atoll where the reef water has a longer residence time. Oceanic atoll and barrier reef lagoons, which are in the terminal stage of evolutionary history of oceanic volcanic islands, have the potential to release CO2 to the atmosphere.

    Notes: Abstract An improved cultivation system for Arabidopsis thaliana was developed, allowing advanced biochemical studies in vitro and in vivo of this important model plant. Highly functional Arabidopsis thylakoids were isolated and used to study both basic and regulatory photosynthetic functions with the aim to create a platform for the characterization of mutants deficient in auxiliary proteins. Light-induced proteolytic degradation of the D1 protein could be followed and shown to be a subsequent event to photoinactivation of electron transport. The phosphorylation and dephosphorylation of thylakoid proteins resembled that seen in spinach leaves although phospho-CP43 revealed an unusual regulatory behavior.

    Notes: Abstract Transformation with a chimeric receptor containing the glucocorticoid transactivation and DNA-binding domains fused to an ecdysteroid receptor ligand-binding domain permits ecdysone agonist-inducible gene expression in monocotyledonous plant cells. The inducible system is based on the specific activation of a chimeric receptor containing the ligand-binding domain of the Heliothis virescens ecdysteroid receptor and the inducer RH5992 (a 20-hydroxyecdysone agonist). RH5992 is an non-steroidal agrochemical with a high specificity for lepidopteran ecdysone receptors. Addition of RH5992 to transformed cells results in high levels of inducible expression in a ligand-specific manner, particularly when the effector receptor is coupled to the strong transactivator VP16. A chimeric construct containing the Drosophila ecdysone ligand-binding domain failed to activate reporter gene activity with RH5992, while activation was observed in the presence of muristeroneA. The system described provides the basis for an inducible gene expression system that is compatible with agricultural use.

    Notes: Summary Abnormally thick cell walls of a clonal maize cell line with the labyrinth wall morphology found in transfer cells were analyzed and compared to the relatively thin and even archetypical walls of a sister cell line. Despite a drastic difference in wall morphology between the transfer and archetypical cell walls, the chemical composition of the walls was essentially the same. There were no major differences in the glycosyl residue composition, in the amount of total lipid, and in the amount of total protein. The amounts of wall material released by chemical extraction of cellulosic, hemicellulosic, and pectic fractions were the same for the two types of walls. There were some differences in the protein profile and in the inorganic ion content between the transfer and archetypical walls. These results indicate that profound changes in wall morphology can be brought about in the absence of gross changes in wall composition and suggest that major changes in time- or place-dependent deposition and/or subtle changes in arrangement of rare wall constituents may be responsible.

    Notes: Summary Protoplasts are frequently isolated from maize coleoptiles with cell-wall-degrading enzymes such as pectolyase (PEC), mazerozyme, and cellulase. Incubation of coleoptiles with these enzymes caused rapid depolarizations of the membrane voltage (V M ). The depolarizing effect of 0.5% (w/v) mazerozyme or 1.5% (w/v) cellulase was unaffected by denaturation of the enzymes. In the case of pectolyase (0.1%, w/v), however, the active enzyme was significantly more potent than the denaturated enzyme in depolarizing coleoptile cells. Exposure to 0.1% active PEC but not to inactive PEC also caused an oxidative burst in coleoptiles and enhanced K+ efflux. Together this suggests that pectic breakdown products of the cell wall act as signal for wounding. Typically addition of 10 μM 1-naphthylene acetic acid (NAA) to coleoptiles causes a transient depolarization followed by a slow hyperpolarization of V M . However, in the presence of PEC, V M only depolarized in NAA. After PEC-treated coleoptiles were washed free of the enzyme, NAA caused only small fluctuations of V M . A similarly small V M response to NAA appeared in coleoptiles pretreated with heatdenaturated supernatant (SUP) from a protoplast isolation buffer, the latter suspected to contain the PEC-generated wounding signal. Comparable pretreatment of coleoptiles with PEC or SUP had no significant effect on the spontaneous and NAA-evoked acidification of the incubation medium. Pretreatment with SUP also had no significant effect on the NAA-stimulated elongation of coleoptile segment. Hence, PEC treatment of coleoptile tissue affects the membrane transport properties of the cells. This effect is partly maintained after removal of the enzyme from the incubation medium, an effect not significant for NAA-generated acidification and cell elongation.

    Notes: Summary Various developmental phases can be distinguished in the definition of the archesporium and the early life of the embryo, takingZea mays (maize) as a model within the family Gramineae, and other families where pertinent: (1) the isolation of the megasporocyte and the functional spore derived from it; (2) the maturation of the specialized walls of the embryo sac, and their reinforcement by ensheathments derived from the contiguous nucellar cells during a sequence of phased genetic ablation; (3) the differentiation of the synergids, the associated flange, and the filiform apparatuses; (4) the blocking of the pollen tube pathway by secondary secretions in the micropylar region and the coagulation of the pollen tube cytoplasm within the filiform apparatuses during the process of fertilization; and finally (5) the development of a compound cutinized envelope of four fused layers (six where the outer integument is also involved) after fertilization. For the nascent haploid generation, the period of maximum vulnerability in respect to both pathogen invasion and the transition from diplophase control occurs during these phases. It is concluded that many of the protective features form a prophylactic shield and are key components of the angiosperms in general, which may have contributed to their evolutionary success as a group. Other physiological or biochemical adaptations or barriers may also supplement the mainly structural features described here.

    Notes: Abstract Effect of zinc (Zn) nutritional status on uptake of inorganic 65Zn was studied in rye (Secale cereale, cv. Aslim), three bread wheat (Triticum aestivum, cvs. Dagdas, Bezostaja, BDME-10) and durum wheat (Triticum durum, cv. Kunduru-1149) cultivars grown for 13 days in nutrient solution under controlled environmental conditions. The cultivars were selected based on their response to Zn deficiency and to Zn fertilization in calcareous soils under field conditions. When grown in Zn-deficient calcareous soil in the field, the rye cultivar had the highest, and the durum wheat the lowest Zn efficiency. Among the bread wheats, BDME-10 showed higher susceptibility to Zn deficiency and Bezostaja and Dagdas were less affected by Zn deficiency. Similarly to field conditions, in nutrient solution visual Zn deficiency symptoms (i.e. necrotic lesions on leaf blade) appeared to be more severe in Kunduru-1149 and BDME-10 and less severe in rye cultivar Aslim. Under Zn deficiency, shoot concentrations of Zn were similar between all cultivars. Cultivars with adequate Zn supply did not differ in uptake and root-to-shoot translocation rate of 65Zn, but under Zn deficiency there were distinct differences; rye showed the highest rate of Zn uptake and the durum wheat the lowest. In the case of bread wheat cultivars, 65Zn uptake rate was about the same and not related to their differential Zn efficiency. Under Zn deficiency, rye had the highest rate of root-to-shoot translocation of 65Zn, while all bread and durum wheat cultivars were similar in their capacity to translocate 65Zn from roots to shoots. When Zn2+ activity in uptake solution ranged between 117 p M and 34550 pM, Zn-efficient and Zn-inefficient bread wheat genotypes were again similar in uptake and root-to-shoot translocation rate of 65Zn. The results indicate that high Zn efficiency of rye can be attributed to its greater Zn uptake capacity from soils. The inability of the durum wheat cultivar Kunduru-1149 to have a high Zn uptake capacity seems to be an important reason for its Zn inefficiency. Differential Zn efficiency between the bread wheat cultivars used in this study is not related to their capacity to take up inorganic Zn.

    Notes: Abstract The growth response of white clover (Trifolium repens L.) to the expected increase in atmospheric partial pressure of CO2 (pCO2) may depend on P availability. A decrease in the rate of transpiration due to increased pCO2 may reduce the amount of P transported to the shoot, thereby causing a change in the partitioning of P between the root and shoot. To test these hypotheses, four concentrations of P in the nutrient solution, combined with two pCO2 treatments, were applied to nodulated white clover plants. Compared to ambient pCO2 (35 Pa), twice ambient pCO2 (70 Pa) reduced the rate of transpiration but did not impair the total P uptake per plant. However, at twice ambient pCO2 and a moderate to high supply of P, concentrations of structural P and soluble P (Pi) were lower in the leaves and higher in the roots. The activity of root acid phosphatase was lower at twice ambient pCO2 than at ambient pCO2; it depended on the Pi concentration in the roots. At the highest P concentration, twice ambient pCO2 stimulated photosynthesis and the growth rate of the plant without affecting the concentration of nonstructural carbohydrates in the leaves. However, at the lower P concentrations, plants at twice ambient pCO2 lost their stimulation of photosynthesis in the afternoon, they accumulated nonstructural carbohydrates in the leaves and their growth rate was not stimulated; indicating C-sink limitation of growth. P nutrition will be crucial to the growth of white clover under the expected future conditions of increased pCO2.

    Notes: Abstract Phosphorus (P) deficiency limits the yield of wheat, particularly by reducing the number of ears per unit of area because of a poor tiller emergence. The objectives of this work were to (i) determine whether tiller emergence under low phosphorus availability is a function of the availability of assimilates for growth or a direct result of low P availability, (ii) attempt to establish a quantitative relation between an index of the availability of P in the plant and the effects of P deficiency on tiller emergence, and (iii) to provide a better understanding of the mechanisms involved in tiller emergence in field-grown wheat. Wheat (Triticum aestivum L., cv. INTA Oasis), was grown in the field under drip irrigation on a typic Argiudol, low in P (5.5 μg P g-1 soil Bray & Kurtz I) in Balcarce, Argentina. Treatments consisted of the combination of three levels of P fertilization 0, 60 and 200 kg P2O5 ha-1, and two levels of assimilate availability, a control (non-shaded) and 65% of reduction in incident irradiance from seedling emergence until the end of tillering (shaded). Phosphorus treatments significantly modified the pattern of growth and development of the plants. Shading reduced the growth and concentration of water-soluble carbohydrates in leaves and stems. Leaf photosynthetic rate at saturating irradiance was reduced by P deficiency, but was not affected by shading. At shoot P concentrations less than 4.2 g P kg-1 the heterogeneity in the plant population increased with respect to the number of plants bearing a certain tiller. At a shoot P concentration of 1.7 g P kg-1 tillering ceased completely. Phosphorus deficiency directly altered the normal pattern of tiller emergence by slowing the emergence of leaves on the main stem (i.e. increasing the phyllochron), and by reducing the maximum rate of tiller emergence for each tiller.

    Notes: Abstract Two methods for measuring proton fluxes along intact maize roots grown with NH 4 + or NO 3 − at pH 6.5 were compared. Videodensitometric measurement of changes in a pH-indicator dye by video camera was used to map pH around roots and determine the amounts of protons released by various root regions. This method was compared with potentiometric determination of the concentration of H+ in the unstirred layer at the root surface using ion-selective microelectrodes. With NH 4 + the roots released large amounts of H+ in preferential regions where the rate of flux can reach 1.4 or even 2.5 nmol m−1 s−1. Videodensitometry indicated a first region of root acidification in the subapical zone, but this was more difficult to localize with microelectrodes. With NO3 − both methods showed that the roots released small amounts of H+ and that the apical region took up H+ in the first 10 mm then sometimes released H+ over the following 10 mm of root. The H+ flux profiles obtained by both methods were in good agreement in terms of both order of magnitude of the fluxes and spatial differences along the root. These results suggest that videodensitometry, which is easier to use than potentiometry, can be used to screen different plant species or cultivars under various experimental conditions. The microelectrode technique is indispensable, however, for studying the underlying mechanisms of net H+ fluxes.

    Notes: Abstract The present work describes an original method to follow rate of 14CO2 and total CO2 production from rhizosphere respiration after plant shoots had been pulse-labelled with 14CO2. We used a radioactivity detector equipped with a plastic cell for flow detection of beta radiation by solid scintillation counting. The radioactivity detector was coupled with an infrared gas analyser. The flow detection of 14CO2 was compared to trapping of 14CO2 in NaOH and counting by liquid scintillation. First, we demonstrated that NaOH (1 M) trapped 95% of the CO2 of a gaseous sample. Then, we determined that the counting efficiency of the radioactivity flow cell was 41% of the activity of gaseous samples as determined by trapping in NaOH (1 M) and by counting by static liquid scintillation. The sensitivity of the 14CO2- flow detection was 0.08 Bq mL−1 air and the precision was 2.9% of the activity measured compared to 0.9% for NaOH trapping method. We presented two applications which illustrate the relevance of 14CO2-flow detection to investigations using 14C to trace photoassimilates within the plant-soil system. First, we examined the kinetics of 14CO2 production when concentrated acid is added to NaH14CO3. This method is the most commonly used to label photoassimilates with 14C. Then, we monitored 14CO2 activity in rhizosphere respiration of 5-week old maize cultivated in soil and whose shoots had been pulse-labelled with 14CO2. We conclude that alkali traps should be used for a cumulative determination of 14CO2 because they are cheap and accurate. On the other hand, we demonstrated that the flow detection of 14CO2 had a finer temporal resolution and was consequently a relevant tool to study C dynamics in the rhizosphere at a short time scale.

    Notes: Abstract The maize P-rr gene conditions floral-specific flavonoid pigmentation, especially in the kernel pericarp and cob. We analyzed the P-rr promoter by transient expression assays, in which segments of the P-rr promoter were fused to the GUS reporter gene and introduced into maize cells by particle bombardment. A basal P-rr promoter fragment (−235 to +326) gave low, but significant, levels of GUS reporter gene expression. Interestingly, two widely spaced segments containing enhancer-like activity were found. When tested individually, both the proximal (−1252 to −236) and distal (−6110 to −4842) segments boosted expression of the basal P-rr promoter::GUS construct about five-fold. A 1.6 kb segment of the P-rr promoter (−1252 to +326) containing the proximal enhancer and the 5′-untranslated leader driving the GUS reporter gene showed preferential expression in BMS and embryogenic suspension cell cultures vs. endosperm-derived suspension cell cultures. These results demonstrate the application of transient assay techniques for the identification of regulatory elements responsible for floral-specific regulation of the complex P-rr gene promoter in maize.

    Notes: Abstract Two cDNAs encoding novel type III maize (Zea mays) GST subunits, ZmGST VI and ZmGST VII, have been cloned in addition to the previously described ZmGST V. Together with the type I GSTs ZmGST I and ZmGST III, these subunits were expressed in Escherichia coli, both individually and in tandem combinations using a customised pET vector. The GST dimers formed were then characterised. When type I GSTs were co-expressed only the respective homodimers were formed rather than the ZmGST I-III heterodimer. The failure to form this heterodimer, together with the negligible herbicide-detoxifying activity associated with recombinant ZmGST III-III, suggests that the identity of herbicide-detoxifying isoenzymes described in maize as being composed of ZmGST III subunits requires re-evaluation. In contrast, co-expression of the type III GSTs ZmGST V and ZmGST VI resulted in the formation of ZmGST V-V, ZmGST VI-VI and ZmGST V-VI dimers in the ratio 1:1:2 as predicted for random subunit association. ZmGST V-VI had kinetic characteristics intermediate between those of the two homodimers, indicating that the subunits were catalytically independent of one another. Co-expression of ZmGST V and ZmGST VII resulted in the formation of ZmGST V-VII and this isoenzyme was subsequently identified in maize plants. Attempts to dimerise type I GST subunits with type III GST subunits proved unsuccessful. These results demonstrate the utility of co-expressing recombinant GSTs to explore the potential of subunit-subunit associations and to help unravel the complexity of homodimeric and heterodimeric GSTs in plants.

    Notes: Abstract cDNA and the corresponding genomic DNA region encoding Rubisco activase were isolated from the unicellular green alga Chlorococcum littorale. The deduced amino acid sequence encoded by the cDNA was 403 amino acids long and exhibited important homology with those of other known Rubisco activases. Its N-terminal sequence was similar to the chloroplastic transit peptides in Chlamydomonas reinhardtii. The mature protein had a predicted molecular mass of 42 kDa. Five introns were located inside the genomic gene encoding Rubisco activase (rca). Genomic Southern blots indicated that two copies of the rca gene were present in the genome of C. littorale. The level of rca messenger RNA increased when cells of C. littorale were subjected to high-CO2 stress (i.e. grown under at least 20% CO2). Hsp70 heat-shock protein was also induced under high-CO2 conditions and, as expected, was also induced at 35 °C. The rca gene, in contrast, was not induced at 35 °C, indicating that this gene was induced in response to the high CO2 concentration and not to general stress. A search of the promoter-binding proteins by a gel retardation assay showed that, under the high-CO2 conditions, a protein(s) which was probably an activator of the rca transcription was synthesized.

    Notes: Abstract The nuclear HMG1 proteins of higher plants are small non-histone proteins that have DNA-bending activity and are considered architectural factors in chromatin. The occurrence of the chromosomal HMG1 proteins, HMGa, HMGc1/2 and HMGd, in various maize tissues was analyzed, and in the course of these studies a novel HMG1 protein, now termed HMGe, was identified. Purification and characterization of HMGe (Mr 13 655) and cloning of the corresponding cDNA revealed that it displays only moderate similarity to other members of the plant HMG1 protein family. The five maize HMG1 proteins could be detected in kernels, leaves, roots and suspension culture cells, indicating that these proteins can be expressed simultaneously and occur relatively ubiquitously. However, the various HMG1 proteins are present in significantly different quantities with HMGa and HMGc1/2 being the most abundant HMG1 proteins in all tissues tested. Furthermore, the relative amounts of the various HMG1 proteins differ among the tissues examined. The HMG1 proteins were found to be relatively stable proteins in vivo, with HMGc1/2, HMGd and HMGe having a half-life of ca. 50 h in cultured cells, while the half-life of the HMGa protein is ca. 65 h. Collectively, these findings are compatible with the concept that the different plant HMG1 proteins might act as general architectural proteins in concert with site-specific factors in the assembly of certain nucleoprotein structures involved in various biological processes.

    Notes: Abstract Responses of a range of barley (Hordeum vulgare L.) genotypes to boron (B) deficiency were studied in two experiments carried out in sand culture and in the field at Chiang Mai, Thailand. In experiment 1, two barley genotypes, Stirling (two-row) and BRB 2 (six-row) and one wheat (Triticum aestivum L.) genotype, SW 41, were evaluated in sand culture with three levels of applied B (0, 0.1 and 1.0 μM B) to the nutrient solution. It was found that B deficiency depressed flag leaf B concentration at booting, grain number and grain yield of all genotypes. In barley Stirling, B deficiency also depressed number of spikes plant-1, spikelets spike-1 and straw yield. However, no significant difference between genotypes in flag leaf B concentration was found under low B treatments. Flag leaf B concentration below 4 mg kg-1 was associated with grain set reduction and could, therefore, be used as a general indicator for B status in barley. In experiment 2, nine barley and two wheat genotypes were evaluated in the field on a low B soil with three levels of B. Boron levels were varied by applying either 2 t of lime ha-1 (BL), no B (B0) or 10 kg Borax ha-1 (B+) to the soil prior to sowing. Genotypes differed in their B response for grain spike-1, grain spikelet-1 and grain set index (GSI). The GSI of the B efficient wheat, Fang 60, exceeded 90% in all B treatments. The B inefficient wheat SW 41 and most of the barley genotypes set grain normally (GSI 〉80%) only at the B+. In B0 GSI of the barley genotypes ranged from 23% to 84%, and in BL from 19% to 65%. Three of the barley with severely depressed GSI in B0 and BL also had a decreased number of spikelets spike-1. In experiment 3, 21 advanced barley lines from the Barley Thailand Yield Nursery 1997/98 (BTYN 1997/98) were screened for B response in sand culture with no added B. Grain Set Index of the Fang 60 and SW 41 checks were 98 and 65%, respectively, and GSI of barley lines ranged between 5 and 90%. One advanced line was identified as B efficient and two as moderately B efficient. The remaining lines ranked between moderately inefficient to inefficient. These experiments have established that there is a range of responses to B in barley genotypes. This variation in the B response was observed in vegetative as well as reproductive growth. Boron efficiency should be considered in breeding and selection of barley in low B soils.

    Notes: Abstract An intrinsic divinyl-chlorophyll a/b antenna and a particular form of phycobiliprotein, phycoerythrin (PE) III, coexist in the marine oxyphotobacterium Prochlorococcus marinus CCMP 1375. The genomic region including the cpeB/A operon of P. marinus was analysed. It encompasses 10 153 nucleotides that encode three structural phycobiliproteins and at least three (possibly five) different polypeptides analogous to cyanobacterial or red algal proteins involved either in the linkage of subunits or the synthesis and attachment of chromophoric groups. This gene cluster is part of the chromosome and is located within a distance of less than 110 kb from a previously characterized region containing the genes aspA-psbA-aroC. Whereas the Prochlorococcus phycobiliproteins are characterized by distinct deletions and amino acid replacements with regard to analogous proteins from other organisms, the gene arrangement resembles the organization of phycobiliprotein genes in some other cyanobacteria, in particular marine Synechococcus strains. The expression of two of the Prochlorococcus polypeptides as recombinant proteins in Escherichia coli allowed the production of individual homologous antisera to the Prochlorococcus α and β PE subunits. Experiments using these sera show that the Prochlorococcus PEs are specifically associated to the thylakoid membrane and that the protein level does not significantly vary as a function of light irradiance or growth phase.

    Notes: Abstract Implication of His-to-Asp and/or Asp-to-His (His-Asp) phosphorelay has been recently reported in signal transduction pathways initiated by ethylene and cytokinin. These signaling systems are generally composed of sensor His-protein kinases, His-containing phosphotransfer (HPt) domains, and response regulator domains. In this study, we isolated maize cDNAs, designated as ZmRR2 and ZmHP2, which encode a response regulator domain and HPt domain, respectively, and we identified their His-to-Asp phosphotransfer activity in vitro. The putative translated product of ZmRR2 was highly similar to that of ZmRR1 (78% identity), a maize response regulator homologue. The putative translated product of ZmHP2 showed similarity to that of HPt domains from Arabidopsis thaliana (AHP1–AHP3: 44 to 47% identity) and Saccharomyces cerevisiae (Ypd1p: 24% identity). In vitro experiments demonstrated that the putative signaling factors can transfer the phosphoryl group from His-80 of ZmHP2 to Asp-90 of ZmRRs. Treating detached leaves with t-zeatin or supplying inorganic nitrogen to the whole plant induced the accumulation of ZmRR1 and ZmRR2 transcripts. On the other hand, the steady-state transcript level of ZmHP2 was not affected by cytokinin or inorganic nitrogen sources. These results indicate that His-Asp phosphotransfer may be involved in the transduction of nitrogen signals mediated by cytokinin, and that multiple response regulators participate in the signaling pathways.

    Notes: Abstract We isolated a full-length cDNA that encodes ATP-dependent phosphoenolpyruvate carboxykinase (EC 4.1.1.49, PCK) from leaves of maize, an NADP-malic enzyme type C4 plant. The mRNA was specifically and rather abundantly expressed in bundle sheath cells in accordance with the recent finding of cell-type-specific localization of PCK protein in maize, which has been detected with antibodies against cucumber PCK protein. The predicted protein had an N-terminal extension, which is characteristic of plant PCKs. The transcript level was much higher in the daytime than at night in 14-day old seedlings. However, in 42-day old plants the extent of diurnal change decreased. The maize PCK was expressed in Escherichia coli with the pET32 plasmid and purified to homogeneity. Through digestion with enterokinase, two types of enzyme were prepared; one with an intact N-terminus and the other lacking its N-terminal 77 amino acid residues due to over-digestion. The truncated protein had about 2-fold higher specific activity than the intact one, and was inhibited by 3-phosphoglycerate (3-PGA) with an I0.5 of 17.5 mM. In contrast, the intact protein was almost insensitive to 3-PGA. These results strongly suggest that the intact N-terminal extension may be involved in the regulation of PCK activity in vivo through some modification such as reversible phosphorylation.

    Notes: Abstract Root length density (RLD) is an important determinant of crop water and nutrient acquisition, but is difficult to measure in the field. On a soil profile, in-situ counts of root impacts per unit surface on soil profiles (NI) can be used to calculate RLD if crop-specific parameters for preferential root orientation (anisotropy) are known. An improved method for field determinations of RLD was developed and validated for maize at sites in Côte d'Ivoire and Burkina Faso. Root anisotropy was measured with cubes of undisturbed soil with 0.1 m sidelength, based on NI observed on three planes oriented perpendicularly to each other. RLD was also measured for the enclosed volume. Repetition of such measurements enabled estimation of the robustness across sites of empirical and geometric models for the relationship between RLD and NI:RLD = NI CO, with CO being the coefficient of root orientation, theoretically equals 2 for an isotropic distribution. Root systems were found to be nearly isotropic, except near the root front (0.3 to 0.5 m), where roots had a preferentially orthotropic orientation. Measured RLD was generally about 50% larger than RLD calculated from observed NI and CO, indicating that at least one of the measurement techniques had a systematic error. The ratio between measured and calculated RLD (CE), which ranged from 0.8 to 2, increased with the age of the plants and decreased with soil depth. CE was therefore introduced as an additional coefficient, resulting in RLD = NI CO CE. The empirical value for CO CE was between 2 and 5. The empirical coefficients CO and CE were the same for the sites in Cote d'Ivoire (oxisol with an iron pan at 0.6 to 0.9 m) and Burkina Faso (alfisol with an iron pan at 0.4 to 0.8 m). The model was validated with independent data sets at both sites, and gave satisfactory predictions of RLD on the basis of NI obtained from single soil planes, which can be easily measured in the field.

    Notes: Abstract The genetic basis of salt tolerance was examined in selected salt tolerant and sensitive material from a sample of accessions previously assessed for variability in salinity tolerance. The North Carolina Model 2 Design and analysis was followed, tolerance being assessed in 10-day-old seedlings grown in salinized solution culture at control (0 mM), 60 mM and 80 mM NaCl concentrations). Salinity tolerance was shown to be under the control of genes with additive and non-additive effects, with broad and narrow sense heritability estimates being approximately 0.7 and 0.4 over all treatments.

    Notes: Abstract The Wx locus controls amylose synthesis in the cereal endosperm. Hexaploid wheat (Triticum aestivum L.) has the three Wx loci on chromosomes 7A ( Wx-A1), 4A (Wx-B1) and 7D (Wx-D1). To verify the effects of null alleles on reducing amylose content and determine the amylose synthesis capacity of each Wx gene independently and accurately, we produced eight possible types of recombinant lines carrying different null alleles at the Wx loci under the ‘Chinese Spring’ genetic background. Amylose content varied from 0% of the waxy ‘Chinese Spring’ to 25% of the ‘Chinese Spring’ normal type. The reducing effect of the single null alleles was the largest in Wx-B1b, and there was no significant difference between Wx-A1b and Wx-D1b. More than 3% reductions in amylose content were detected in the double null types. The results of the double null lines further demonstrated that for the capacity of amylose synthesis, Wx-B1a predominates and produces 21–22% amylose, followed by Wx-D1a (20–21%) and Wx-A1a (15–18%). These significant differences were partly correlated with variation in the amounts of the Wx proteins produced by different Wx genes. However, comparisons of the double null lines with the single null or normal lines indicated that amylose content was not linearly proportional to the number of the Wx genes, suggesting that the Wx genes act in an epistatic manner.

    Notes: Abstract In darkness, dormancy was imposed on seeds of lettuce (Lactuca sativa L. cv. Grand rapids) by high temperature and on seeds of oilseed rape (Brassica napus L. cv. Apex) by osmotic stress using polyethylene glycol (PEG 8000). In both cases, dormancy was broken by incubating the seeds in aqueous extracts of combustion products from Salix viminalis wood chips or Themeda triandra leaves. Dormancy of rapeseed, but not lettuce, was also broken by a solution of smoke from burnt straw of Triticum aestivum. The greatest stimulation from burnt vegetation was achieved with an aqueous extract of pyrolysed willow wood chips, which had been subjected to temperatures of up to 800 °C during combustion in a down-draught gasifier. This suggests that some biologically active substances obtained from combustion of plant tissues are highly heat-stable.

    Notes: Abstract What follows is a very personal account of my professional life and the early years that preceded it. I have described the social and economic conditions in America and how the nineteen twenties and thirties nurtured our scientific future. The description of the early part of post-World War II research covers my experience in the areas of nutritional biochemistry, biochemical genetics and proceeds to photosynthesis. The latter era lasted around 35 years. For me the most memorable research accomplishments in which I was a participant during this period was the first demonstration of the primary carboxylation enzyme in an in vitro system in algal and higher plants as well to show that it was structurally associated with the chloroplast.Our group while at Oak Ridge and the University of Massachusetts assembled data that described the complete macromolecular assembly of the photosynthetic apparatus of the unusual photosynthetic green bacterium Chloroflexus aurantiacus and created a model of that system which differed greatly from the chomatophore system for the purple bacteria. For the last decade, my scientific journey, with numerous new colleagues has turned to the exciting area of biomaterials.We characterized and modeled the completely new bacterial intracellular inclusions responsible for the synthesis and degredation of biosynthetic, biodegradable and biocompatible bacterial polyesters in the cytoplasm of Pseudomonads.

    Notes: Abstract Unlike the α and β polypeptides of the core light-harvesting complex (LH1) of Rhodobacter (Rb.) sphaeroides, the α and β polypeptides of the peripheral light-harvesting complex (LH2) of this organism will not form a subunit complex by in vitro reconstitution with bacteriochlorophyll. Guided by prior experiments with the LH1 β polypeptides of Rb. sphaeroides and Rhodospirillum rubrum, which defined a set of interactions required to stabilize the subunit complex, a series of mutations to the Rb. sphaeroides LH2 β polypeptide was prepared and studied to determine the minimal changes necessary to enable it to form a subunit-type complex. Three mutants were prepared: Arg at position −10 was changed to Asn (numbering is from the conserved His residue which is known to be coordinated to bacteriochlorophyll); Arg at position −10 and Thr at position +7 were changed to Asn and Arg, respectively; and Arg at position −10 was changed to Trp and the C-terminus from +4 to +10 was replaced with the amino acids found at the corresponding positions in the LH1 β polypeptide of Rb. sphaeroides. Only this last multiple mutant polypeptide formed subunit-type complexes in vitro. Thus, the importance of the C-terminal region, which encompasses conserved residues at positions +4, +6 and +7, is confirmed. Two mutants of the LH1 β polypeptide of Rb. sphaeroides were also constructed to further evaluate the interactions stabilizing the subunit complex and those necessary for oligomerization of subunits to form LH1 complexes. In one of these mutants, Trp at position −10 was changed to Arg, as found in LH2 at this position, and in the other His at position −18 was changed to Val. The results from these mutants allow us to conclude that the residue at the −10 position is unimportant in subunit formation or oligomerization, while the strictly conserved His at −18 is not required for subunit formation but is very important in oligomerization of subunits to form LH1.

    Notes: Abstract Maize liquid endosperm extracts contain the enzymes necessary for all of the steps of the plant IAA biosynthetic pathway from tryptophan, and provide a means to assay the pathway in vitro. We have analyzed the reactions in the presence of a series of indole and indole-like analogues in order to evaluate the potential of these compounds to act as inhibitors of IAA biosynthesis. Such inhibitors will be useful to investigate the tryptophan to IAA pathway, to determine the precursors and intermediates involved, and to select for mutants in this process. A number of such compounds were tested using in vitro enzyme assays for both the tryptophan dependent IAA biosynthesis pathway and for tryptophan synthase β activity. Some compounds showed strong inhibition of IAA biosynthesis while having only a slight effect on the reaction rate of tryptophan synthase β. These results: (1) show that IAA biosynthesis can be selectively inhibited relative to tryptophan biosynthesis; (2) suggest potential ways to screen for IAA biosynthetic pathway mutations in plants; and (3) provide additional tools for studies of IAA biosynthesis in plants.

    Notes: Abstract One thousand three hundred and eighty-seven spring wheat germplasm (Triticum aestivum L.) lines belonging to the Indian and CIMMYT wheat programmes were evaluated for their tolerance to foliar blight disease for three consecutive years i.e., from 1994 to 1997. Disease severity at six different growth stages, beginning from tillering to late milk stage, was recorded. None of the genotypes showed immunity to the disease. Of 43 lines showing resistant reaction, a major proportion (25) was represented by CIMMYT material. Comparatively, Indian germplasm lines tended to be more susceptible at more advanced growth stages. Area Under Disease Progress Curve (AUDPC) and Apparent Infection Rate (r) values of resistant lines were much lower than those of susceptible ones, but lower AUDPC in some of the resistant lines did not correspond to a lower 'r' value. Most of the resistant lines were derived from Seri, Myna, Bau, kauz, Hork 's' and Aegilops tauschii Coss.

    Notes: Abstract The dependence of photosynthesis on light and temperature is modelled through analysis of transition probabilities of photosystems. In the model, two transition probabilities are functions of light, and one transition probability is a function of temperature. The estimated light-saturated photosynthesis of Ecklonia cava blades at 20 °C was 0.037 mg C cm−2 h−1. The value of the activation energy, the standard enthalpy and the standard entropy were estimated to be 56.5 kJ mol−1, 204 kJ mol−1 and 678 J mol−1 K−1, respectively. A production model (an integral photosynthesis model) for an E. cava stand was developed using the photosynthesis model. Production calculated by the model agreed well with observed data during the growing period of an E. cava stand at a field observation site on the west side of Miura Peninsula, Japan. Results of the analysis of the effects of irradiance and temperature on the production of the E. cava community by the model are: 1. Production decreased with irradiance decrease. The estimated compensation irradiance was 26.5 μmol photons m−2 s−1 when the biomass was 3 kg wet mass m−2 (blade:stipe ratio = 2 kg m−2:1 kg m−2) and the temperature was 20 °C. 2. The optimum temperature decreased when irradiance decreased and when biomass increased. The highest estimated value for the optimum temperature was 24.0 °C. The estimated optimum temperature was 18.2 °C when the biomass was 12 kg wet mass m−2 and the photon irradiance was 200 μmol photons m−2 s−1. 3. The amount of biomass that resulted in the maximum production was influenced by irradiance and temperature. At 400 μmol photons m−2 s−1 and 20 °C, the estimated value of the biomass (blade:stipe = 2:1) giving the maximum pr oduction was about 5.3 kg wet mass m−2. However, at 100 μmol photons m−2 s−1 and 24 °C, the estimated value was about 3.0 kg wet mass m−2. The estimated values of the maximum production under the two conditions were 1.05 and 0.30 g C m−2 h−1, respectively.

    Notes: Abstract Previous studies have shown that short exposure of plants to high doses of ozone decreases subsequent photosynthesis; initially by reducing carboxylation capacity. This study tests the hypothesis that this is also the primary cause of loss of photosynthetic capacity in leaves affected by development under a low level of ozone. Triticum aestivum and Pisum sativum plants were exposed from germination to ozone in air (80 nmol mol-1 for 7 hours per day, for 18 days. Leaves that had completed lamina expansion at this time were free of visible injury and light absorptance was unaffected. However, some significant changes in photosynthetic gas exchange were evident. Photosynthetic CO2 uptake at light saturation was decreased significantly by 35% in T. aestivum but was unchanged in P. sativum. The reduction in photosynthesis of T. aestivum was accompanied by a 31% decline in the maximum velocity of carboxylation measured in vivo. Decreased stomatal conductance did not contribute to this reduction of photosynthesis because there was no significant change in the stomatal limitation to CO2. Processes directly dependent upon photochemical reactions; that is, the quantum yield of CO2 uptake and capacity for regeneration of ribulose 1,5-bisphosphate were not affected by O3 fumigation in either species. This suggests that for wheat, the quantitative cause of decreased photosynthetic rate in vivo is a decrease in the quantity of active ribulose-1,5- bisphosphate carboxylase-oxygenase.

    Notes: Abstract In illuminated intact spinach chloroplasts, warming to and beyond 40 °C increased the proton permeability of thylakoids before linear electron transport through Photosystem II was inhibited. Simultaneously, antimycin A-sensitive cyclic electron transport around Photosystem II was activated with oxygen or CO2, but not with nitrite as electron acceptors. Between 40 to 42 °C, activation of cyclic electron transport balanced the loss of protons so that a sizeable transthylakoid proton gradient was maintained. When the temperature of darkened spinach leaves was slowly increased to 40°C, reduction of the quinone acceptor of Photosystem II, QA, increased particularly when respiratory CO2 production and autoxidation of plastoquinones was inhibited by decreasing the oxygen content of the atmosphere from 21 to 1%. Simultaneously, Photosystem II activity was partially lost. The enhanced dark QA reduction disappeared after the leaf temperature was decreased to 20 °C. No membrane energization was detected by light-scattering measurements during heating the leaf in the dark. In illuminated spinach leaves, light scattering and nonphotochemical quenching of chlorophyll fluorescence increased during warming to about 40 °C while Photosystem II activity was lost, suggesting extra energization of thylakoid membranes that is unrelated to Photosystem II functioning. After P700 was oxidized by far-red light, its reduction in the dark was biphasic. It was accelerated by factors of up to 10 (fast component) or even 25 (slow component) after short heat exposure of the leaves. Similar acceleration was observed at 20 °C when anaerobiosis or KCN were used to inhibit respiratory oxidation of reductants. Methyl viologen, which accepts electrons from reducing side of Photosystem II, completely abolished heat-induced acceleration of P700+ reduction after far-red light. The data show that increasing the temperature of isolated chloroplasts or intact spinach leaves to about 40 °C not only inhibits linear electron flow through Photosystem II but also activates Photosystem I-driven cyclic electron transport pathways capable of contributing to the transthylakoid proton gradient. Heterogeneity of the kinetics of P700+ reduction after far-red oxidation is discussed in terms of Photosystem I-dependent cyclic electron transport in stroma lamellae and grana margins.

    Notes: Abstract The effect of the light harvesting 1 (LH1) antenna complex on the driving force for light-driven electron transfer in the Rhodobacter sphaeroides reaction center has been examined. Equilibrium redox titrations show that the presence of the LH1 antenna complex influences the free energy change for the primary electron transfer reaction through an effect on the reduction potential of the primary donor. A lowering of the redox potential of the primary donor due to the presence of the core antenna is consistently observed in a series of reaction center mutants in which the reduction potential of the primary donor was varied over a 130 mV range. Estimates of the magnitude of the change in driving force for charge separation from time-resolved delayed fluorescence measurements in the mutant reaction centers suggest that the mutations exert their effect on the driving force largely through an influence on the redox properties of the primary donor. The results demonstrate that the energetics of light-driven electron transfer in reaction centers are sensitive to the environment of the complex, and provide indirect evidence that the kinetics of electron transfer are modulated by the presence of the LH1 antenna complexes that surround the reaction center in the natural membrane.

    Notes: Abstract The kinetic properties of photosynthesis (both transient and steady-state) were monitored using three non-invasive techniques to evaluate limitations on triose-phosphate (triose-P) conversion to carbohydrate in rice. These included analyzing the O2 sensitivity of CO2 fixation and the assimilatory charge (AC) using gas exchange (estimate of the ribulose 1,5- bisphosphate pool) and measuring Photosystem II activity by chlorophyll fluorescence analysis under varying light, temperature and CO2 partial pressures. Photosynthesis was inhibited transiently upon switching from 20 to 2 kPa O2 (reversed O2 sensitivity), the degree of which was correlated with a terminal, steady-state suppression of low O2 enhancement of photosynthesis. Under current ambient levels of CO2 and moderate to high light, the transient pattern was more obvious at 18 °C than at 26 °C while at 34 °C no tra nsient response was observed. The transient inhibition at 18 °C ranged from 15% to 31% depending on the pre-measurement temperature. This pattern, symptomatic of feedback, was observed with increasing light and CO2 partial pressures with the degree of feedback decreasing from moderate (18 °C) up to high temperature (34 °C). Under feedback conditions, the rate of assimilation is shifted from being photorespiration limited to being triose-P utilization limited. Transitory changes in CO2 assimilation rates (A) under low O2 indicative of feedback coincided with a transitory drop in assimilatory charge (AC) and inhibition of electron transport. In contrast to previous studies with many C3 species, our studies indicate that rice shows susceptibility to feedback inhibition under moderate temperatures and current atmospheric levels of CO2.

    Notes: Interspecific ecophysiological differences in response to different light environments are important to consider in regeneration behavior and forest dynamics. The diurnal changes in leaf gas exchange and chlorophyll fluorescence of two dipterocarps, Shorea leprosula (a high light-requiring) and Neobalanocarpus heimii (a low light-requiring), and a pioneer tree species (Macaranga gigantea) growing in open and gap sites were examined. In the open site, the maximum net photosynthetic rate (Pn), photosystem II (PSII) quantum yield (δ; F/Fm′), and relative electron transport rate (r-ETR) through PSII at a given photosynthetic photon flux density (PPFD) was higher in S. leprosula and M. gigantea than in N. heimii, while non-photochemical quenching (NPQ) at a given PPFD was higher in N. heimii. The maximum values of net photosynthetic rate (Pn) in M. gigantea and S. leprosula was higher in the open site (8–11 μmol m−2 s−1) than in the gap site (5 μmol m−2 s−1), whereas that in N. heimii was lower in the open site (2 μmol m−2 s−1) than in the gap site (4 μmol m−2 s−1), indicating that N. heimii was less favorable to the open site. These data provide evidence to support the hypothesis that ecophysiological characteristics link with plant’s regeneration behavior and successional status. Although Pn and stomatal conductance decreased at midday in M. gigantea and S. leprosula in the open site, both r-ETR and leaf temperature remained unchanged. This indicates that stomatal closure rather than reduced photochemical capacity limited Pn in the daytime. Conversely, there was reduced r-ETR under high PPFD conditions in N. heimii in the open site, indicating reduced photochemical capacity. In the gap site, Pn increased in all leaves in the morning before exposure to direct sunlight, suggesting a relatively high use of diffuse light in the morning.

    Notes: Abstract We hypothesized that aphids after previous exposure to hydroxamic acids (Hx), a family of secondary plant compounds deleterious to aphids, are able to reduce their subsequent exposure to them. This hypothesis was tested by evaluating the time to produce salivation into a sieve element (SSE) by the aphid Sitobion fragariae on seedlings of two wheat cultivars of Triticum aestivum differing in their concentration of Hx. The total time to produce a first SSE was significantly longer in the high-Hx cultivar; however, the subsequent, second SSE (first SSE after interruption of probing) in this cultivar was significantly reduced, reaching the level observed in the low-Hx plants. Therefore, a strategy to reduce the exposure to secondary compounds was observed only in the second SSE in high-Hx plants. When the experimental plant was replaced by a new unattacked plant after the first SSE, aphids did not change the behavior described, thus excluding an aphid-induced plant susceptibility. The number of cell punctures and accumulated duration was not affected by previous exposure to Hx, either in low or high Hx cultivars. Total time and pathway time but not cell punctures, seem to be the variables affected by previous exposure to Hx.

    Notes: Abstract One approach to reduce NO3 movement to groundwater is increasing the proportion of N supplied to the crop as NH4–N. Nitrification inhibitors (NI's) can be used to enhance NH4–N supply, but most studies have focused on yield response, with little attention given to environmental impacts. To determine the effect of enhanced NH4 sources on corn grain yield, N uptake and NO3 movement to groundwater, three sidedress materials were compared during three different growing seasons. Application of anhydrous ammonia (AA) and addition of the NI, dicyandiamide (DCD) to urea-ammonium nitrate (UAN) both reduced NO3 leaching losses relative to that incurred with UAN. With AA and UAN + DCD (as compared with UAN) subsoil solution NO3 concentrations were reduced by an average of: 1.1 mg NO3–N kg-1 soil following (fall 1993) a dry growing season; 2.4 mg NO3–N kg-1 soil during (spring and summer 1994) and 1.4 mg NO3–N kg-1 soil after (fall 1994) a wet growing season; and 0.5 mg NO3–N kg-1 soil following (fall 1995) a growing season with intermediate rainfall. Based on average solution NO3 concentrations and approximate drainage after harvest, estimated N losses between harvest and freeze-up were 43, 22 and 19 kg N ha-1 with UAN, UAN + DCD and AA, respectively (average of 3 years). Grain yields and aboveground N uptake were greater with AA and UAN + DCD than with UAN, and residual fertilizer N (applied N less aboveground N uptake) was 18, 6 and -2 kg N ha-1 with UAN, UAN + DCD and AA, respectively (average of 3 years). As is often observed, the trend for greater yield with addition of the NI was not large or consistent enough to meet registration criteria. Data demonstrating reduced NO3 leaching are also relevant, and positive environmental impacts should be a criterion for registration. For growers who are reluctant to use AA, this would provide an alternative source to maximize yield while minimizing NO3 movement to groundwater.

    Notes: Abstract Decreases in Colwell bicarbonate soil test P in the years after applying single (ordinary) superphosphate, and the residual value of superphosphate, was measured in a long-term field experiment on a duplex (texture contrast) soil (sand over lateritic ironstone gravel clay sand at 10–15 cm), at Wongan Hills, Western Australia, typical of many soils used to grow crops in Western Australia. Ten levels of P (0–91 kg P ha-1) were applied once only in late May to different plots in different years from 1988 to 1993. Wheat (Triticum aestivum), or lupin ( Lupinus angustifolius)) were sown in late May of each year, when the P treatments applied that year were banded (drilled) with the seed. Soil samples were collected each June to measure soil test P. Seed (grain) yields of the crops were measured each December. The residual value (RV) of P applied in previous years was calculated relative to P applied in the current year, using grain yields (RVyield) and soil test P (RVsoil). Soil test P measured on soil samples collected in June was related to yields measured in December that year to provide soil P test calibrations. Relative to P applied in the current year, soil test P decreased by between 15 to 30% for P applied one year previously, by 25 to 30% for P applied three years previously, and by 60 to 70% for P applied six years previously. Soil test P was affected by spatial variation, and it also varied in the different years, for P applied in the current year, one year previously, two years previously, etc. Compared with P applied in the current year, mean RVyield determined in the different years decreased by about 40% one year after P application, followed by a further 20% decrease for P applied two years previously, followed by a further 20% decrease for P applied three to five years previously. Relative to current P, RVsoil decreased by about 25% one year after P application, followed by a further 20% for P applied two years previously, followed by a further 10% for P applied three years ago, and followed by a further 6% for P applied four and five years ago. As measured in the different years, the soil P test calibration varied between years for P applied one, two etc. years previously. This was so even when the same cultivar of wheat was grown at the same site in different years.

    Notes: Abstract We describe a protocol for flow cytometry analysis of cell cycle in plants using indirect immunolabelling staining and Vicia faba, Pisum sativum and Zea mays root tip cells as model systems. The protocol is based on simultaneous analysis of two fluorescent signals. The first, obtained after staining with propidium iodide, is used to quantify total nuclear DNA content. The second, obtained after indirect immunofluorescent staining of bromodeoxyuridine (BrdUrd), is used to quantify the amount of BrdUrd incorporated into nuclear DNA. In an attempt to standardize the procedure, the effects of various conditions for partial DNA denaturation using HCl, as well as of BrdUrd concentration and incorporation time on flow cytometry DNA / BrdUrd content analysis have been studied. Maximum BrdUrd-linked fluorescence was observed after a 30 min pulse with 10 μM BrdUrd and after DNA denaturation with 1.5 N HCl (final concentration) for 30 min at 25 °C. Under these conditions, DNA content histograms with relatively small coefficient of variation (〈 4%, full peak) could be obtained. To avoid non-specific staining of cytoplasm and cell walls, the protocol involves the use of nuclei isolated from formaldehyde-fixed tissues. Fixed isolated nuclei are stable and may be stored in hexylene glycol 0.75 M at 4 °C for prolonged periods prior to actual staining and analysis.

    Notes: Abstract Wheat plants, 22d. old, were exposed to wide range of soil water osmotic potential (Ψs = 0 to −1.2 MPa) induced by NaCl and CaCl2 treatments in combination with roots maintained under aerobic (drained at field capacity) or nonaerobic (flooded) conditions in the soil, and sprayed with 10 mg L−1 kinetin solution. In drained plants, not receiving kinetin, increased soil salinity resulted in appreciable inhibition of shoot growth and reduction in chlorophyll (Ch1.), soluble sugars (SS) contents and grain yield. Shoot growth, Ch1. content, soluble sugars and grain yield were significantly lower for flooded plants than unflooded analogues over the entire Ψs range. Both salinity and waterlogging synergize to increase Na+, Ca+ and Cl− accumulation in shoot tissues and to decrease the stability of leaf membranes to either dehydration (40% polyethylene glycol 6000) or heat (51 °C) stress. The ratio of K+/Na+ transported to shoots under aerobic and anaerobic conditions decreased progressively on salinization. The association between the internal mineral element concentrations was largely affected by kinetin treatment. Kinetin application ameliorated the deleterious effects of salinity and oxygen deficiency. It reduced Na+, Ca2+ and Cl− accumulation and improved K+ uptake under salinity and waterlogging stresses. Increased K+/Na+ ratio helped the plants to avoid Na+ toxicity and enhanced shoot growth and grain yield. Kinetin also reduced membrane injury by dehydration and heat stresses and improved the water status of plants under both aerobic and anaerobic conditions. The effects of single factors (Soil salinity ‘Ψs’, soil waterlogging ‘WL’ and Kinetin ‘Kin’) and their interactions (Ψs × WL, Ψs × Kin, WL × Kin and Ψs × WL × Kin) were shown by analysis of variance to be statistically significant for most parameters tested. Calculation of the coefficient of determination (η+) led to three important findings. (1) Salinity (Ψs) was dominant in affecting leaf relative water content (RWC), shoot dry mass, grain yield, stability of leaf membranes to dehydration stress and the contents of Na+, Ca2+, Mg2+ and Cl−. (2) Kinetin (Kin) had a dominant effect on the stability of leaf membranes to heat stress as well as on chlorophyll and soluble sugars contents. (3) The share of waterlogging (WL) was dominant for K+ content. It can be concluded that kinetin application helped wheat plants to grow successfully in the areas subjected to combined effects of salinity and oxygen deficiency, such as in salt marshes.

    Notes: Abstract Inheritance of genes for hairy auricles and hairy leaf sheath of Ae. tauschii in hexaploid wheat backgrounds (synthetic hexaploid wheat and common wheat varieties) was analyzed. The results indicated that hairy auricles and hairy leaf sheath of Ae. tauschii can be transferred and are expressed in hexaploid wheat. In a synthetic hexaploid wheat ('Ae. tauschii' 188) hairy auricles was proved to be controlled by a single dominant gene derived from Ae. tauschii, which was different from the Pa gene located on chromosome 4BS of common wheat. The hairy leaf sheath phenotype of 'Altar 84/Ae. tauschii 188' was also controlled by a single dominant gene derived from Ae. tauschii, which is obviously different from the Hls gene in T. dicoccoides. We suggest to designate the Ae. tauschii genes for hairy auricles and hairy leaf sheath as Pa2 and Hls2, respectively; such genes could be used as useful genetic markers in common wheat.

    Notes: Abstract Grapevine plantlets multiplied in vitro were acclimatized at 40 or 90 μmol m−2 s−1 photon flux density for 12 or 16 h per day, respectively. In the high-light regime a decrease in total chlorophyll and an increase in chlorophyll a/chlorophyll b ratio occurred. However, at high-light intensity lower photosynthetic capacities and higher apparent photosynthesis were measured than at the low-light regime. In leaves expanded during acclimatization, the light compensation point was higher in plantlets under high-light while quantum yield was higher in low-light conditions. High-light also gave rise to an increase in carbohydrate concentration. As a whole, the results suggest that high-light increases carbon assimilation and growth although with a low investment in the photosynthetic apparatus.

    Notes: Abstract Hybrid necrosis in wheat is a problem for gene transfer in wheat breeding. Hybrid necrosis occurs due to dominant complementary interaction of two genes, Ne1 and Ne2. A cross between wheat (Triticum aestivum L.) varieties C306 (drought tolerant, Ne1 carrier) and WL711(high yielding, Ne2 carrier) produced necrotic F1 hybrids, which died before ear emergence and produced no seeds. To overcome the problem of hybrid necrosis, ears enclosed in the leaf sheath were taken and cultured to maturity in liquid medium containing 5% sucrose and 0.04% glutamine. The necrotic hybrids produced only a few seeds per ear compared to parents, but individual grain weight was similar in the hybrid and the parents. The F1 ear culture study has been repeated for three years and F2 seeds obtained. In 1996–97, the cultured ears of F1 hybrids produced 62 seeds, of which only 52 showed germination and were grown under normal field conditions. Out of the 52 seeds, 50% were non-necrotic and showed segregation for various physiological traits. The results reveal that hybrids ears had the potential to form viable seeds. Culturing of wheat ears before ear emergence and production of viable F2 seeds from necrotic hybrids is a simple and efficient method for overcoming the problem of hybrid necrosis.

To overcome the influence of environment on disease expression, the study was conducted by extensive evaluation of advanced generation (F8) recombinant inbred lines (RILs) developed by single seed descent from the cross WL 711 (susceptible) × HD 29 (resistant. The results suggested that HD 29 possesses three major genes for resistance to isolated Ni7 and two genes for resistance to isolate Ni8. One of the two genes controlling resistance to Ni8 is common with one of the genes conferring resistance to Ni7. These observations have important implications in breeding for Karnal bunt resistance.

    Notes: Abstract Elymus tsukushiense Honda (syn. Roegneria kamoji C. Koch) (2n = 6x = 42, StsStsHtsHtsYtsYts) is a hexaploid species, distantly related to bread wheat Triticum aestivum L. em Thell (2n = 6x = 42, AABBDD). Apart from the delineation of evolutionary relationships, this species is a potential source of resistance to scab, a devastating disease of wheat caused by Fusarium graminearum Schw. A standard C-banded karyotype was established identifying all 21 chromosome pairs of E. tsukushiense. By using C-banding and genomic in situ hybridization analyses, three wheat-E. tsukushiense chromosome addition lines, one ditelosomic addition line, and one disomic substitution line were identified in BC2 progenies from wheat × E. tsukushiense hybrids. Twenty DNA markers specific for the seven homoeologous groups of the Triticeae were used to determine the homoeology of the added E. tsukushiense chromosomes. The E. tsukushiense chromosomes in the addition lines NAU702, NAU703, and NAU701 were identified as belonging to homoeologous groups 1, 3, and 5, and thus, were designated as 1Ets#1, 3Ets#1, and 5Ets#1, respectively. NAU751 was identified as a disomic substitution line with chromosome 3A of wheat replaced by chromosome 3Ets#1. Line NAU702 has a high level of resistance to scab and will be used in chromosomal engineering and development of improved wheat germplasm for scab resistance breeding.

    Notes: Abstract The allelic variation of prolamin loci was studied in three F2 progenies from three crosses between the 1BL/1RS cultivar Triana and Yécora Rojo, Pavón and Florence Aurora, cultivars without the translocation. According to the 1:2:1 theoretical proportions observed in the allelic variants of the Glu-B3/Gli-B1 loci of the parent without the translocation, the inheritance as a block of the rye chromosome arm was confirmed. A group of F3-F4 recombinant lines, developed from these crosses was evaluated using the SDS-sedimentation test and the mixograph and alveograph tests. The presence of the 1BL/1RS translocation was not associated with significantly lower grain protein content values or with the optimum mixing time in the mixograph of the genotypes. The effect of the 1BL/1RS translocation on most of the quality parameters was highly dependent on the genetic pool. Significant increases in gluten strength and better mixing properties associated with the presence of some alleles of the Glu-A1, Glu-A3/ Gli-A1 and Gli-D2 loci were detected. The additivity and the interaction of prolamin gene effects with the rye translocation in the 1BL/1RS lines and its possible use in plant breeding are discussed.

    Notes: Abstract A segregating population of doubled-haploid lines issued from the cross between the wheat (Triticum aestivum L. em. Thell) cultivars Courtot, resistant to several isolates of powdery mildew (Blumeria graminis DC. f. sp. tritici Em. Marchal), and Chinese Spring (susceptible) was used to map Mlar, a gene carried by Courtot and conferring resistance to this pathogen. The assignation of Mlar using monosomic lines of Courtot was confirmed by the mapping analysis. Mlar was located on the short arm of the chromosome 1A, in the vicinity of the locus XGli-A5 coding for storage proteins. This result was in accordance with those demonstrating that Mlar was an allele of the Pm3 locus (Pm3g), a gene also involved in the resistance to powdery mildew.

    Notes: Abstract Nineteen cultivars, with large differences in heading date, were evaluated for their response to septoria tritici blotch in two experimental setups in Njoro, Kenya. Due to the more or less constant temperatures during the growing season and the overhead irrigation applied the epidemic conditions were similar over the whole observation period for the early and late cultivars. In experiment 1 the cultivars were assessed for disease severity at the same moment irrespective of the developmental stage, while in experiment 2 the cultivars were assessed at the same developmental stage. Measured at the same time, the disease severity was highest in the early maturing cultivars and lowest in the late maturing cultivars (r = –0.78). When assessed at the same development stage the disease build up was independent of heading date (r = –0.10) but strongly dependent on resistance level. There were no indications that early heading cultivars were more susceptible than late heading cultivars.

    Notes: Abstract Seed storage proteins of 131 Japanese Norin wheat (Triticum aestivum) varieties were fractionated by sodium dodecyl sulfate polyacrylamide gel electrophoresis to determine allelic make-up in varieties at each of three loci that control high-molecular-weight (HMW) glutenin subunits. Three alleles were identified at the Glu-A1 locus, six at the Glu-B1 locus and five at the Glu-D1 locus. Twenty-four different, major glutenin HMW subunits were identified and each contained three to five subunits and seventeen different glutenin subunit patterns were observed for 19 subunits in the 131 Japanese Norin varieties. Fourteen alleles were identified by comparison of subunit mobility with that previously found in hexaploid wheat. Japanese Norin varieties showed a specific pattern of allelic variation in glutenin HMW subunits, different from that of Chinese and other country common wheats in allelic frequency at Glu-1 loci.

    Notes: Abstract Two pot experiments at the Plant Environment Laboratory (PEL), Reading, UK investigated sterility, boron (B) accumulation and B partitioning of wheat cultivars grown with limited B in the growing medium. The first experiment evaluated nine cultivars of spring wheat with diverse field responses to low available soil B, supplied with or without 20 μM B. A second experiment examined the response of a susceptible (SW-41) and a tolerant (Fang-60) cultivar to B-deficiency. These cultivars were supplied with either 20 μM B from sowing to flag leaf emergence and no added B thereafter, or 20 μM B from sowing to maturity. When B was not supplied in the nutrient solution, the number of grains ranged from 4 per ear (cv. BL-1135) to 32 per ear (cv. BL-1249) and sterility of competent florets ranged from 39% to 93%. Boron concentration in the flag leaf at anthesis did not differ greatly when the growing medium contained limited B, but differences between cultivars were evident when B was unlimited. Tolerance of B-deficiency was not related to the B concentration in the flag leaf. Some cultivars produced viable pollen and set grains while others failed to do so at similar B concentrations in the flag leaf. The two contrasting cultivars did not differ much in their pattern of B partitioning when B supply was restricted from flag leaf emergence onwards. Similarly, little evidence was found that the tolerant cultivars translocated B from their leaves, roots or stems when the supply in the growing medium was restricted. The proportion of total B partitioned in different organs was the same irrespective of B supply and cultivar. On average, leaves contained 68% of the total B content in the whole plant compared to 16% in the roots, 10% in the ears and only 6% in the stems. Tolerant or susceptible cultivars of wheat could not be distinguished based on the B concentration and B content of the flag leaf.

    Notes: Abstract Using the PDS-1000/He Biolistic® Particle Delivery System, the microprojectile travel distance, rupture disk pressure and DNA/gold particle concentrations were assessed in order to optimise short and longer-term β-glucuronidase reporter gene expression in microspore-derived embryos of wheat. The effects were also evaluated of using sterile filter paper to support explants and treatment with a high osmoticum medium (0.2 M mannitol/0.2 M sorbitol or 0.4 M maltose). In the optimised procedure, wheat microspore-derived embryos (MDEs), were placed on filter paper and incubated on medium containing 0.4 M maltose, for 4 h pre- and 45 h post-bombardment. Five μl pAHC25 (0.75 mg ml-1 in TE buffer) was precipitated onto 25 μl gold particles (60 mg ml-1 in sterile water), using 20 μl spermidine (0.1 M) and 50 μl CaCl2 (2.5 M). The particles were centrifuged and resuspended in 75 μl absolute ethanol prior to the preparation of 6 macrocarriers. A microprojectile travel distance of 70 mm, a rupture pressure of 1300 p.s.i., and a vacuum of 29′′ Hg were employed. Maltose at 0.4 M in the support medium was the most important factor influencing GUS activity in bombarded tissues. GUS activity, 1 day post-bombardment, reached 52 ± 17 GUS-positive foci/MDE (mean ± s.e.m, n=3), with 17 ± 4 foci/MDE at 15 days, giving a 3.0-fold increase (p〈0.05) compared to expression in MDEs bombarded on medium without a high osmoticum treatment.

    Notes: Abstract Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco; EC 4.1.1.39) has played a central role in our understanding of chloroplast biogenesis and photosynthesis. In particular, its catalysis of the rate-limiting step of CO2 fixation, and the mutual competition of CO2 and O2 at the active site, makes Rubisco a prime focus for genetically engineering an increase in photosynthetic productivity. Although it remains difficult to manipulate the chloroplast-encoded large subunit and nuclear-encoded small subunit of crop plants, much has been learned about the structure/function relationships of Rubisco by expressing prokaryotic genes in Escherichia coli or by exploiting classical genetics and chloroplast transformation of the green alga Chlamydomonas reinhardtii. However, the complexity of chloroplast Rubisco in land plants cannot be completely addressed with the existing model organisms. Two subunits encoded in different genetic compartments have coevolved in the formation of the Rubisco holoenzyme, but the function of the small subunit remains largely unknown. The subunits are posttranslationally modified, assembled via a complex process, and degraded in regulated ways. There is also a second chloroplast protein, Rubisco activase, that is responsible for removing inhibitory molecules from the large-subunit active site. Many of these complex interactions and processes display species specificity. This means that attempts to engineer or discover a better Rubisco may be futile if one cannot transfer the better enzyme to a compatible host. We must frame the questions that address this problem of chloroplast-Rubisco complexity. We must work harder to find the answers.

    Notes: Abstract Absorption and fluorescence spectra of chlorophyll a have been analyzed on the basis of an extended version of Kennard–Stepanov (KS) theory. It is proposed that at least one new electronic state lies just below the normal S1 − S0 transition (Qy), borrowing approximately 2–4% of its strength and contributing to the fluorescence in the tail. The KS anomalies leading to this hypothesis occur in a wide variety of cases, including chlorophyll a in solution and protein-bound chlorophyll a, suggesting that the phenomenon is an intrinsic property of the molecule. Natural candidates for the new state(s) are the second and third triplet states. The relationship of the fluorescence excitation spectrum to KS theory is investigated and applied to explain a red drop in yield.

    Notes: Abstract The specific effects exerted by different heavy metals on both the function and the structure of the photosynthetic apparatus were addressed. The functional analysis performed via the fluorescence induction kinetics revealed that the applied toxic heavy metals can be classified into two groups: Cd and Ni had no significant effect on the photosynthetic electron transport, while Cu, Pb and Zn strongly inhibited the Photosystem II (PS II) activity, as evidenced by the dramatic decreases in both the variable (Fv) and the maximal (Fm) fluorescence. The structural effects of the heavy metal ions on the thylakoid membranes were considered in three relations: (1) lipids, (2) proteins — studied by Fourier transform infrared (FTIR) spectroscopy, and (3) lipid—protein interactions — investigated by electron spin resonance (ESR) spectroscopy using spin-labeled probe molecules. The studied heavy metal ions had only a non-specific rigidifying effect on the thylakoid lipids. As regards proteins, Cd and Ni had no effect on the course of their heat denaturation. The heat denaturation of the proteins was accompanied by a decrease in the α-helix content (1656 cm-1), a parallel increase in the disordered segments (1651 cm-1), a decrease in the intramolecular β-sheet (1636 cm-1) content and the concomitant appearance of an intermolecular β-structure (1621 cm-1). In contrast with Cd and Ni, Cu and Zn blocked the appearance of the intermolecular β-structure. Pb represented an intermediate case. It seems that these heavy metals alter the native membrane structure in such a way that heat-induced aggregation becomes more limited. The ESR data revealed that certain heavy metals also affect the lipid—protein interactions. While Cd and Ni had hardly any effect on the solvation fraction of thylakoid lipids, Cu, Pb and Zn increased the fraction of lipids solvating the proteins. On the basis of the FTIR and ESR data, it seems that Cu, Pb, and Zn increase the surfaces available for lipid—protein interactions by dissociating membrane protein complexes, and that these ‘lipidated’ proteins have a smaller chance to aggregate upon heat denaturation. The data presented here indicate that the damaging effects of poisonous heavy metals are element-specific, Cu, Pb and Zn interact directly with the thylakoid membranes of the photosynthetic apparatus, while Cd and Ni interfere rather with other metabolic processes of plants.

    Notes: Abstract In order to explain the mechanisms of Calvin-cycle regulation, the general properties of metabolic systems under homeostatic flux control are analyzed. It is shown that the main characteristic point for an enzyme in such a system can be the value of a sharp transition from some constant homeostatic flux to a limitation by this enzyme. A special method for the quantitative treatment of the experimental dependence of a metabolic flux such as photosynthesis on enzyme content is developed. It is pointed out that reactions close to a thermodynamic equilibrium under normal conditions can considerably limit the homeostatic fluxes with a decrease of the enzyme content. Calvin-cycle enzymes are classified as non-limiting, near-limiting and limiting. The deduced rules for the regulation of a homeostatic metabolic pathway are used to explain the data obtained for transgenic plants with reduced activities of Calvin-cycle enzymes. The role of compensating mechanisms that maintain the photosynthesis rate constant upon the changes of enzyme contents is analyzed for the Calvin cycle. The developed analysis explains the sharp transitions between limiting and non-limiting conditions that can be seen in transgenic plants with reduced content of some Calvin-cycle enzymes, and the limiting role of such reversible enzymes as aldolase, transketolase and others. The attempt is made to predict the properties of plants with increased enzyme contents in the Calvin cycle.

    Notes: Abstract Marine and terrestrial photosynthetic and chemoautotrophic microorganisms assimilate considerable amounts of carbon dioxide. Like green plastids, the predominant means by which this process occurs is via the Calvin-Benson-Bassham reductive pentose phosphate pathway, where ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) plays a paramount role. Recent findings indicate that this enzyme is subject to diverse means of control, including specific and elaborate means to guarantee its high rate and extent of synthesis. In addition, powerful and specific means to regulate Rubisco activity is a characteristic feature of many microbial systems. In many respects, the diverse properties of microbial Rubisco enzymes suggest interesting strategies to elucidate the molecular basis of CO2/O2 specificity, the ‘holy grail’ of Rubisco biochemistry. These systems thus provide, as the title suggests, ‘different perspectives’ to this fundamental problem. These include vast possibilities for imaginative biological selection using metabolically versatile organisms with well-defined genetic transfer capabilities to solve important issues of Rubisco specificity and molecular control. This review considers the major issues of Rubisco biochemistry and regulation in photosynthetic microoganisms including proteobacteria, cyanobacteria, marine nongreen algae, as well as other interesting prokaryotic and eukaryotic microbial systems recently shown to possess this enzyme.

    Notes: Abstract An in vivo method for the estimation of kinetic parameters of partial reactions of carboxylation of ribulose 1,5-bisphosphate (RuBP) catalyzed by ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) is described. Rubisco in barley, wheat and bean is different in the ability of its active centers to bind RuBP. The rate constant of the formation of the Rubisco-RuBP complex in these plants at 25 °C is 0.414, 0.245 and 0.660 mM-1 s-1, respectively. The rate constant of the reaction of the Rubisco-bound enediol with CO2 does not differ significantly in barley and wheat, and averages 66 mM-1 s-1. Decreased irradiance inhibits Rubisco in two ways: by reducing the concentration of operating catalytic sites and by decreasing the rate constant of binding of RuBP to Rubisco. High concentrations of CO2 inhibit Rubisco by decreasing the concentration of competent carboxylation centers, without any s ignificant influence upon the rate constants of partial reactions.

    Notes: Abstract A site directed mutant of the Photosystem I reaction center of Chlamydomonas reinhardtii has been described previously. [Hallahan et al. (1995) Photosynth Res 46: 257–264]. The mutation, PsaA: D576L, changes the conserved aspartate residue adjacent to one of the cysteine ligands binding the Fe-SX center to PsaA. The mutation, which prevents photosynthetic growth, was observed to change the EPR spectrum of the Fe-SA/B centers bound to the PsaC subunit. We suggested that changes in binding of PsaC to the PsaA/PsaB reaction center prevented efficient electron transfer. Second site suppressors of the mutation have now been isolated which have recovered the ability to grow photosynthetically. DNA analysis of four suppressor strains showed the original D576L mutation is intact, and that no mutations are present elsewhere within the Fe-SX binding region of either PsaA or PsaB, nor within PsaC or PsaJ. Subsequent genetic analysis has indicated that the suppressor mutation(s) is nuclear encoded. The suppressors retain the altered binding of PsaC, indicating that this change is not the cause of failure to grow photosynthetically. Further analysis showed that the rate of electron transfer from the quinone electron carrier A1 to Fe-SX is slowed in the mutant (by a factor of approximately two) and restored to wild type rates in the suppressors. ENDOR spectra of A1 ·– in wild-type and mutant preparations are identical, indicating that the electronic structure of the phyllosemiquinone is not changed. The results suggest that the quinone to Fe-SX center electron transfer is sensitive to the structure of the iron-sulfur center, and may be a critical step in the energy conversion process. They also indicate that the structure of the reaction center may be modified as a result of changes in proteins outside the core of the reaction center.

    Notes: Abstract The effects of CO2 concentration and the effects of growth-light conditions on Ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) deactivation were examined for Spinacea oleracea (spinach). Rubisco deactivation kinetics and the degree that Rubisco activation limited the rise in photosynthesis following an increase in photon flux density (PFD) were determined from gas-exchange time courses. There were no significant differences in the apparent relaxation time for Rubisco deactivation among leaves exposed to high or low CO2 (50 or 1000 μmol mol-1) and low PFD (170 μmol m-2 s-1) or darkness. However, when PFD was increased to 1700 μmol m-2 s-1 following a period of low PFD or darkness, leaves exposed to low CO2 × low PFD showed a lower contribution to the photosynthetic induction process by the activation of Rubisco than leaves exposed to the other treatments. For the growth-light experiments, spinach was grown under high PFD × high red:far-red ratio (R:FR), low PFD × high R:FR, or low PFD × low R:FR light environments. Leaves that matured under the low PFD × low R:FR treatment showed a lower percent change in photosynthesis due to Rubisco activation than leaves exposed to the other growth-light treatments. However, there were no significant differences among the growth-light treatments in the maximum contribution of Rubisco activation to the induction response or in the apparent relaxation time for Rubisco deactivation during shade events.

    Notes: Abstract In order to assess the ecological impacts of the atmospheric CO2 increase on the intertidal macroalgae during emersion, the photosynthesis of Enteromorpha linza (a green alga), Ishige okamurae (a brown alga) and Gloiopeltis furcata (a red alga) was investigated in air as a function of CO2 concentrations and water loss. Their photosynthesis was not saturated at the present atmospheric CO2 level (350 μl l −1 or 15.6 μM), the CO2 compensation point and $$K_{[{\text{mCO}}_{\text{2}} ]} $$ increased with increasing desiccation, showing that desiccation lowers the CO2 affinity of the intertidal macroalgae. It was concluded that E. linza, I. okamurae and G. furcata, while exposed to air, can benefit from atmospheric CO2 rise, especially when the algae have lost some water.

    Notes: Abstract Several samples of the red seaweed, Gelidiella acerosa (Forssk.) Feldmann & Hamel occurring in tidepools, high intertidal rocks, and shallow subtidal areas on a reef flat in Ilocos Norte, northern Philippines were studied in terms of their photosynthetic and respiratory responses (μl O2 gDW−1 h−1) to four salinity (22, 28, 34, 40‰) and three temperature (22, 28, 34 °C) combinations. The upper intertidal plants tolerated low salinities (22–28‰) better than high salinities (34–40‰), while tidepool and subtidal plants were not affected. Temperatures of 22 through 34 °C resulted in a one-fold increase in their photosynthetic rates and insignificant differences in their respiratory rates while tidepool and subtidal plants almost doubled their photosynthetic rates and their respiration rates increased by about 5–50 times. There were no interaction effects. Therefore, intertidal plants appe ared to be more tolerant to wide temperature fluctuations and low salinity levels; while tidepool and subtidal plants were least affected by salinity variations but were quite sensitive to temperature fluctuations. Vegetative and tetrasporic plants had similar photosynthetic and respiratory responses to salinity and temperature variations, although vegetative plants had significantly higher net photosynthesis under the minimum and maximum temperatures tested (22 and 34 °C). Reproductive G. acerosa showed greater tolerance to temperature fluctuations. These responses indicated that physiological changes may have occurred when the species became reproductive. Tolerance of G. acerosa to low salinities suggests that lowering the salinities in culture tanks could be used to eradicate contaminants, i.e., dinoflagellates and filamentous green algae. Temperature of 28 °C appeared to be optimum for all plant types as reflected by their high photosynthetic and low respiratory rates .

    Notes: Summary Ochratoxin A (OTA) is a nephrotoxic and nephrocacinogenic mycotoxin commonly produced by several ubiquitousAspergillus andPenicillium species. OTA is found predominantly in cereals and derived products, and therefore OTA may be transferred into beer from contaminated grain. Thirty-five samples of German beers were analyzed: OTA was extracted with toluene and purified by solid phase extraction using silica cartridges. OTA was determined by high-performance liquid chromatography with fluorescence detection. Since of fluorescence detection is not specific in this instance, the beer samples were also analyzed by high-performance liquid chromatography electrospray ionization-tandem mass spectrometry (HPLC-ESI-MS/MS). OTA levels in the range of 0.1–0.2 μg L−1 were found in 9 beer samples, 21 samples contained trace amounts of OTA (〈0.1 μg L−1) and in 5 samples no OTA was detectable.

    Notes: Summary Doxorubicin is an antineoplastic antibiotic isolated fromStreptomyces peucetius var.cesius clinically used in the treatment of tumors such as lung or breast, Hodgkin's disease and various types of leukemias. The main goal of this study was to develop a simple and sensitive HPLC method with fluorescence detection for the quantitation of doxorubicin in cell culture media collected during an in vitro studies and in human plasma. Solid phase extraction (C2 silica) was applied. The experiment established five-point standard curve (1 ng mL−1 to 100 ng mL−1). The standard curves prepared in blank cell tissue media were linear over the range of doxorubicin assayed and had a mean correlation coefficient of 0.9973±9.43×10−4 and slope 0.02545±1.85×10−3. The standard curves prepared in human plasma were linear and had mean correlation coefficient of 0.997 and slope 0.01885±5.19×10−4. The limit of quantitation for doxorubicin in both specimens was arbitrarily established to be 1 ng mL−1. Intra-day variabilities were determined using 3–4 replicates of control solutions of doxorubicin (3 ng mL−1 and 30 ng mL−1) in blank plasma and cell culture media. Inter-day variabilities were determined over a four day period analyzing replicates of controls. All precision and accuracy values fell within the acceptable range.

    Notes: Summary Human serum albumin (HSA) is one of the most abundant human proteins and has been shown to be heterogeneous. A RP-HPLC method has been developed to separate HSA components in commercially available preparations. Separations were carried out on Aquapore RP-300, C8 columns using gradient elution with a combination of acetonitrile/water mobile phases containing 0.05% trifluoroacetic acid as ion-pairing agent. Optimum resolution was attained on narrow-bore columns using a stepwise, linear gradient that incorporated a shallow intermediate step of 0.20%/min in Mobile Phase B. Under similar elution conditions, separations carried out on standard-size columns showed the expected decrease in resolution due to increased peak widths. A comparative analysis of three commercial products highlighted qualitative and quantitative differences. Capillary zone electrophoresis was used for the analysis of collected RP-HPLC fractions. Results indicated that while the HPLC separation was incomplete, one of the major HPLC peaks was primarily composed of one of the three main components typically separated by CZE. ESI-MS was used to characterize the two major RP-HPLC fractions and also showed that the HPLC separation was incomplete. The MaxEnt transform of the HPLC peaks was consistent with components all being HSA and closely related derivatives.

    Notes: Summary One classical method for quantitation of amino acids in proteins is hydrolysis of the proteins and determination of the free amino acids. Although the drastic experimental conditions necessary for complete hydrolysis always cause degradation of some of the amino acids, if mild hydrolysis conditions are used, a mixture of amino acids and oligopeptides is obtained. If these conditions are adequately tuned, the oligopeptides are almost exclusively dipeptides. For this reason we have initiated a study to find a derivatizing agent suitable for the analysis of amino acids and dipeptides by an absolute method of quantitation already tested for amino acids. FMOC-Cl was found to be a suitable derivatizing agent for this purpose.

    Notes: Summary A rapid method for the simultaneous determination/identification of residual oxytetracycline (OTC) and sulphadimidine (SDD) in meats (beef, pork, chicken) and eggs by high-performance liquid chromatography (HPLC) was developed. The extraction of OTC and SDD was performed using a Sep-Pak® CN cartridge. The extracts contained OTC/SDD analytes when examined by HPLC using a LiChrospher® 100 RP-8 end-capped column and a mobile phase of acetonitrile-acetic acid-water (28:4:68, v/v/v) with a photodiode array detector. The average recoveries from spiked samples (0.1 μg g−1 and 1.0 μg g−1) were in excess of 80.2% with coefficients of variation between 1.5 and 5.0%. The limits of detection for OTC and SDD were 0.05 and 0.02 μg g−1, respectively.

    Notes: Summary Polyethylene glycols (PEGs) of nominal molecular weight (M) 200, 400, 600, 1000, 1500, 3000, 4000 and 6000 were chosen as model compounds and subjected to reversed-phase high performance liquid chromatography (RP-HPLC) on an octadecasilyl silica gel (C18) stationary phase using a binary gradient composed of acetonitrile and water and evaporative light scattering detection (ELSD). Satisfactory resolution of oligomers up to M of 3000 was accomplished; the higher M samples PEG-4000 and PEG-6000 could not be further resolved into the constituent oligomers and therefore, M=4000 marks the upper limit of oligomer resolution. Despite some peak overlapping as a consequence of the more or less broad oligomer distribution, individual types of PEG samples can be distinguished from each other by their characteristic chromatographic fingerprint patterns, as shown with a mixture consisting of PEG-400, PEG-1000, PEG-3000, PEG-4000 and PEG-6000. For this reason, the method is well-suited for characterization of samples containing PEGs widely differing in M. In addition, matrix assisted laser desorption ionization time of flight mass spectroscopy (MALDI-TOF/MS) performed with PEG-600, PEG-1000 and PEG-3000 revealed that the optimum degree of oligomer resolution has been achieved by use of the present method.

    Notes: Summary The enantiomers of primary amino compounds have previously been resolved on a chiral stationary phase (CSP) CSP-18C6I, prepared by immobilizing (+)-18-crown-6 tetracarboxylic acid. In this study related chiral stationary phases were prepared in an effort to broaden the scope of CSP18C6I. CSP-18C6II, synthesized to investigate the effect of spacer length, resolved the enantiomers of 2-amino-1,2-diphenylethanol and 1-(1-naphthyl)ethylamine (1-NEA) (hydrophobic amino compounds) with the largerk values and smaller α values than on CSP-18C6I, probably because of the greater hydrophobicity of CSP-18C6II. Use of CSP-18C6III, synthesized by modification of carboxylic acid functionality of CSP-18C6II by introduction of another chiral moiety,S-1-NEA, resulted in larger α values for 2-amino-1-phenylethanol and 2-amino-1-phenylpropanol than on CSP-18C6II, but the enantiomers of 1-NEA were not resolved, because of steric hindrance between 1-NEA and the chiral moiety. The amide derivativeN-3,5-dinitrobenzoyl-1-(α-naphthyl)ethylamine (DNN) as π-acceptor (3,5-dinitrobenzoyl function) or π-donor (naphthylethylamide function), and no primary amino functionality, was resolved on CSP-18C6III. The mechanism of separation of the enantiomers of DNN was assumed to be the π−π interaction between the 3,5-dinitrobenzoyl function (π-acceptor) of DNN and theS-1-NEA moiety (π-donor) of CSP-18C6III.
